(1) 바실러스 및 유산균 균주의 배양(1) Cultivation of Bacillus and Lactobacillus strains
바실러스 4종은 20 ㎖의 NB(nutrient broth) 배지에 각각 접종하고 180 rpm, 35℃에서 48시간 배양하여 OD값을 0.5로 조정하여 준비하였으며, 유산균 4종은 20 ㎖의 MRS 액체배지에 각각 접종하고 37℃에서 48시간 배양하여 OD값을 0.5로 조정하여 준비하였다. 상기 8종의 균주를 5 ℓ의 JBT 최적 배지(표 2)에 0.2%(v/v)씩 각각 접종하고 33℃에서 통기량 0.4 vvm, 교반속도 250 rpm으로 24시간 동안 배양하여 준비하였다. (2) 스트렙토마이세스 속 균주의 배양(2) Cultivation of strains of the genus Streptomyces
스트렙토마이세스 속 균주는 TSA(tryptic soy agar) 배지에 도말하여 30℃에서 3~4주 동안 배양한 후, 상기 배양된 균주가 포자를 형성하면 150 ㎖의 GSS 액체배지(glucose 20 g, soybean meal 25 g, soluble starch 10 g, beef extract 1 g, yeast extract 4 g, sodium chloride 2 g, dipotassium phosphate 0.25 g, calcium carbonate 2 g, 증류수 1 ℓ)에 접종하여 30℃, 170 rpm으로 10일간 배양하였다. 이후, 45,000 rpm으로 30분간 원심분리하여 상층액과 침전물을 분리하였다. 상기 상층액은 0.45 μm의 필터로 여과하여 준비하였고(시료 1), 상기 침전물에 아세톤 또는 에탄올을 100 ㎖ 첨가하고 170 rpm에서 10시간 동안 교반한 후, 45,000 rpm으로 30분간 원심분리하여 수득한 상층액을 0.45 μm의 필터로 여과하여 준비하였다(시료 2). 트리메소프림(trimethoprim)은 염증 치료에 사용되는 항균물질 중 하나로, WHO 필수 의약품 목록에 등재되어 있는 효과적이고 안전한 의약품으로 사용되고 있다. 이에, 상기 시료 1과 시료 2를 동일한 양으로 혼합한 후(시료 3), 하기 표 3과 같은 조건으로 HPLC 분석을 통해 트리메소프림의 함량을 측정하였다. 아스퍼질러스 시도위(Aspergillus sydowii) 및 아스퍼질러스 플라부스(Aspergillus flavus)를 이용한 디스크 확산법을 통해 항균 활성을 분석하였다. 각 균주의 배양액 150 ㎕를 TSA 배지에 도말하고 10~20분 동안 건조시킨 후, 멸균된 8 ㎜ 페이퍼 디스크를 TSA 배지 위에 올려놓았다. 이후, 본 발명의 항균분해제 50 ㎕를 상기 페이퍼 디스크에 점적하고 37℃, 24시간 동안 배양한 다음 페이퍼 디스크 주변의 억제환을 관찰하였다. 증류수, 99.9%(v/v) 에탄올 또는 30%(v/v) 에탄올 처리군을 대조군으로 이용하였다.
