Abstract
本発明は、アデノ随伴ウイルスの産生量を向上させることができる、アデノ随伴ウイルスの製造方法を提供すること、並びに上記したアデノ随伴ウイルスの製造方法において使用するための細胞および発現ベクターを提供することを課題とする。本発明によれば、E4ORF6遺伝子を外来プロモーターによって発現させることを含むアデノ随伴ウイルスの製造方法であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、アデノ随伴ウイルスの製造方法が提供される。
Description
本発明は、アデノ随伴ウイルスの製造方法に関する。さらに本発明は、上記したアデノ随伴ウイルスの製造方法において使用するための細胞および発現ベクターに関する。
アデノ随伴ウイルス(adeno-associated virus:AAVともいう)は、パルボウイルス科に属する直鎖一本鎖DNAウイルスである。野生型AAVゲノムは、複製の調節遺伝子(Rep遺伝子)とカプシドの構造遺伝子(Cap遺伝子)を含み、ウイルスの複製とパッケージングのために逆方向末端反復配列(ITR)が隣接している。AAVベクターは増殖又は非増殖のいずれの細胞にも遺伝子導入が可能であり、特に非分裂細胞においては長期間の発現が可能である。また、AAVは、非病原性であると考えられており、免疫原性が低い。上記のことから、AAVベクターは、遺伝子治療用ベクターとして臨床応用が進んでいる。
AAVは、アデノウイルスやヘルペスウイルスなどのヘルパーウイルスの存在下で増殖する非エンベロープウイルスである。遺伝子治療もしくは核酸導入に用いるAAVを作製する際には、古典的にはアデノウイルスを宿主細胞に共感染させて、AAV複製が行なわれていた。また、アデノウイルスのヘルパー作用を担う遺伝子が明らかにされ、この遺伝子を搭載したプラスミドも使用されている。例えば、Rep遺伝子およびCap遺伝子を含むプラスミド、アデノウイルスヘルパープラスミド、および導入遺伝子を含むプラスミドを同時にHEK293細胞にトランスフェクションすることで組換え型AAV(rAAV)へとパッケージングすることができる。
特許文献1には、(a)AAVのREPタンパク質をコードする塩基配列、(b)AAVのカプシドの構造タンパク質をコードする塩基配列、(c)第一のAAVの逆方向末端反復(ITR)を含む塩基配列、(d)第二のAAVの逆方向末端反復(ITR)を含む塩基配列、(e)第一のITRと第二のITRとの間に位置する、外来のタンパク質をコードする塩基配列を挿入するための塩基配列又は/及び外来のタンパク質をコードする塩基配列、(f)アデノウイルスのE2A領域を含む塩基配列、(g)アデノウイルスのE4領域を含む塩基配列,及び(h)アデノウイルスのVA-RNA領域を含む塩基配列を含む核酸分子が記載されている。
特許文献2には、少なくとも4つの別個の組換え標的部位(RTS)と、E1A、E1B又はそれらの組み合わせを含むアデノウイルス(Ad)遺伝子と、Ad遺伝子に作動可能に連結されるプロモーターとを含み、RTS、Ad遺伝子及びプロモーターが染色体に組み込まれている哺乳動物細胞、組換えアデノ随伴ウイルス(rAAV)プロデューサー宿主細胞を生成するために上記細胞を使用する方法、並びにAAVプロデューサー宿主細胞を使用してrAAVを生産、パッケージング及び精製する方法が記載されている。

特許文献4には、(i)アデノウイルスE2、E4ORF6、およびVA-RNA遺伝子を含むヘルパープラスミドを含むアクセサリー構築物、(ii)1以上の調節配列に作動可能に連結されたAAVRepおよびAAVCapを含むトランスプラスミドを含むAAVヘルパー構築物、及び(iii)1以上の調節配列に作動可能に連結された目的の異種遺伝子に隣接するAAV逆方向末端反復配列を含むプラスミド(pCis)を含むAAVベクターを含むトランスフェクション培地中で細胞をインキュベートすることを含む、哺乳動物宿主細胞において組換えAAVビリオンを産生するインビトロ法が記載されている。
上記した先行技術においては、ヘルパープラスミド中にE4遺伝子領域がコードされているが、E4遺伝子の発現量の最適化は検討されていない。本発明は、アデノ随伴ウイルスの産生量を向上させることができる、アデノ随伴ウイルスの製造方法を提供することを課題とする。本発明はさらに、上記したアデノ随伴ウイルスの製造方法において使用するための細胞および発現ベクターを提供することを課題とする。
本発明者らは上記課題を解決するために鋭意検討した結果、アデノ随伴ウイルスの産生に必要な遺伝子であるE4ORF6の発現量を低下させることによって、またはE4ORF6遺伝子の細胞への導入量を低下させることによって、アデノ随伴ウイルスの産生量を向上できることを見出した。本発明は、上記知見に基づいて完成したものである。
本発明によれば、以下の発明が提供される。
<1> E4ORF6遺伝子を外来プロモーターによって発現させることを含むアデノ随伴ウイルスの製造方法であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、アデノ随伴ウイルスの製造方法。
<2> E4ORF6の発現の低下が、E4ORF6のmRNA発現の低下及び/又はE4ORF6のタンパク質発現の低下である、<1>に記載のアデノ随伴ウイルスの製造方法。
<3> エンハンサー、サイトメガロウイルス由来プロモーター、E4ORF6遺伝子及びポリA配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現の量が1/2以下に低下している、<1>又は<2>に記載のアデノ随伴ウイルスの製造方法。
<4> 以下の(a)~(e)の何れか一以上によりE4ORF6の発現が低下している、<1>から<3>の何れか一に記載のアデノ随伴ウイルスの製造方法。
(a)外来プロモーターが、サイトメガロウイルス由来プロモーターよりも転写活性が弱いプロモーターである;
(b)E4ORF6遺伝子のKozak配列が、改変されたKozak配列であるか、Kozak配列を使用しない;
(c)エンハンサー配列が、改変されたエンハンサー配列であるか、エンハンサー配列を使用しない;

<6> 少なくともE4ORF6遺伝子を含むE4領域を細胞に導入することを含む、アデノ随伴ウイルスの製造方法であって、Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量と比較して、E4ORF6遺伝子の細胞への導入量が1/2以下である、アデノ随伴ウイルスの製造方法。
<7> E4領域の細胞への導入量が、Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量の1/10以下である、<6>に記載のアデノ随伴ウイルスの製造方法。

本発明のアデノ随伴ウイルスの製造方法、細胞および発現ベクターによれば、アデノ随伴ウイルスの産生量を向上させることができる。
以下、本開示の実施形態の一例について説明する。但し、本開示は、以下の実施形態に何ら限定されるものではなく、本開示の目的の範囲内において、適宜、変更を加えて実施することができる。本明細書において「~」を用いて示された数値範囲は、「~」の前後に記載される数値をそれぞれ最小値及び最大値として含む範囲を意味する。
<用語の説明>
プロモーターとは、RNAポリメラーゼを結合させることができ、下流のコード配列又は非コード配列の転写開始に関与するDNA制御領域/配列を意味する。
プロモーターとは、RNAポリメラーゼを結合させることができ、下流のコード配列又は非コード配列の転写開始に関与するDNA制御領域/配列を意味する。
ベクターとは、外来遺伝子を別の細胞に人為的に運ぶために利用されるDNAまたはRNA分子である。遺伝子を発現するためのベクターを発現ベクターという。導入したい外来遺伝子を含むベクターが細胞内に導入されると、細胞内において外来遺伝子の複製及び/又は発現が行われる。ベクターとしては、エピソーム性(例えばプラスミド)ベクター、及び非エピソーム性ベクターが挙げられる。ベクターは、トランスフェクション、トランスダクション、細胞融合及びリポフェクションなどの方法により、宿主細胞に導入することができる。
搭載とは、染色体や核酸配列に他の核酸配列が組み込まれることを意味する。
遺伝子は、ポリペプチドをコードするヌクレオチドを意味し、例えば、cDNAやゲノムDNAといった核酸分子が挙げられる。
本発明において、Kozak配列とは、E4ORF6の開始コドン(ATG)の前の配列を意味し、GCCACCである。
本発明において、IRES配列とは、internal ribosome entry site のことである。mRNA の5’側のcap構造なしにリボソームに認識されて翻訳を開始する機能をもつ配列であれば、限定されない。好ましくはseq16に開示されている配列と90%以上の同一性を持つ配列である。より好ましくは、seq16に開示されている配列と95%以上の同一性を持つ配列であり、最も好ましくは、seq16と同一の配列である。
<アデノ随伴ウイルスの製造方法>
本発明のアデノ随伴ウイルスの製造方法の第一の態様は、E4ORF6遺伝子を外来プロモーターによって発現させることを含むアデノ随伴ウイルスの製造方法であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、アデノ随伴ウイルスの製造方法である。
本発明のアデノ随伴ウイルスの製造方法の第一の態様は、E4ORF6遺伝子を外来プロモーターによって発現させることを含むアデノ随伴ウイルスの製造方法であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、アデノ随伴ウイルスの製造方法である。
本発明のアデノ随伴ウイルスの製造方法の第二の態様は、少なくともE4ORF6遺伝子を含むE4領域を細胞に導入することを含む、アデノ随伴ウイルスの製造方法であって、Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量と比較して、E4ORF6遺伝子の細胞への導入量が1/2以下である、アデノ随伴ウイルスの製造方法である。
上記の通り、本発明においては、アデノ随伴ウイルス産生に必要な遺伝子であるE4ORF6の発現量を低下させることにより、アデノ随伴ウイルスを高い量で生産することができる。また、本発明においては、E4ORF6による細胞障害を低下させ、遺伝子導入後の細胞生存率を高めることが可能である。さらに本発明の第二の態様においては、プラスミドの導入比率が低下するので、コストを低下することができる。
本発明においては、E4ORF6遺伝子を外来プロモーターによって発現させる。
外来プロモーターとは、野生型が含まないプロモーターのことを意味し、E4遺伝子の場合は、野生型アデノのE4領域に含まれないプロモーターを意味する。E4ORF6遺伝を発現するための外来プロモーターとしては、サイトメガロウイルス由来プロモーター(CMVプロモーター)を使用してもよいし、またはCMVプロモーターよりも転写活性が弱いプロモーターを使用してもよい。CMVプロモーターよりも転写活性が弱いプロモーターについては後記する。
外来プロモーターとは、野生型が含まないプロモーターのことを意味し、E4遺伝子の場合は、野生型アデノのE4領域に含まれないプロモーターを意味する。E4ORF6遺伝を発現するための外来プロモーターとしては、サイトメガロウイルス由来プロモーター(CMVプロモーター)を使用してもよいし、またはCMVプロモーターよりも転写活性が弱いプロモーターを使用してもよい。CMVプロモーターよりも転写活性が弱いプロモーターについては後記する。
エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現を含む発現ユニットを用いてE4ORF6を発現させた場合とは、当業者であれば、強くE4ORF6を発現させたことを意味することは理解できる。一態様においては、seq4でE4ORF6を発現させた場合を意味する。即ち、本発明における「エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している」とは、好ましくは、後記する実施例のseq4(配列番号4の塩基配列を有するプラスミド)を用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下していることを意味する。また、本発明においては、5’Cap構造を介してE4ORF6遺伝子を発現させる方法よりも、5’Cap構造を介さずにE4ORF6遺伝子を発現させる方法(例えば、IRES配列を用いてE4ORF6遺伝子を発現させる方法)が好ましい。
本発明のアデノ随伴ウイルスの製造方法の第一の態様においては、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現は低下していればよいが、好ましくは1/2以下、より好ましくは1/4以下、より好ましくは1/10以下、より好ましくは1/50以下、より好ましくは1/250以下、より好ましくは1/1250以下に低下している。
E4ORF6の発現の低下は、E4ORF6のmRNA発現の低下及び/又はE4ORF6のタンパク質発現の低下である。
E4ORF6遺伝子のmRNA発現量としては、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、1/2以下であることが好ましく、1/4以下であることがより好ましく、1/10以下であることがより好ましく、1/50であることがより好ましく、1/250以下であることがより好ましく、1/1250以下であることがより好ましい。
E4ORF6遺伝子のmRNA発現量としては、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、1/2以下であることが好ましく、1/4以下であることがより好ましく、1/10以下であることがより好ましく、1/50であることがより好ましく、1/250以下であることがより好ましく、1/1250以下であることがより好ましい。
E4ORF6遺伝子により発現されるタンパク質の発現量としては、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、1/2以下であることが好ましく、1/4以下であることがより好ましく、1/10以下であることがより好ましく、1/50であることがより好ましく、1/250以下であることがより好ましく、1/1250以下であることがより好ましい。
E4ORF6の発現を低下させる手段は、好ましくは、以下の(a)~(e)の何れか一以上である。本発明によれば、外来プロモーターに連結されたE4ORF6を含む発現ベクターであって、以下の(a)~(e)の何れか一以上を満たす発現ベクターが提供される。
(a)外来プロモーターが、サイトメガロウイルス由来プロモーターよりも転写活性が弱いプロモーターである;
(b)E4ORF6遺伝子のKozak配列が、改変されたKozak配列であるか、Kozak配列を使用しない;

本発明においては、E4領域と、E2A遺伝子及びVA-RNA遺伝子とは、同一のプラスミドベクターにより細胞に導入してもよいし、異なるプラスミドベクターにより細胞に導入してもよい。
本発明においては、E4領域は、E4ORF6遺伝子と、E4ORF6遺伝子以外の遺伝子とを含むもの、またはE4ORF6遺伝子のみからなるもののいずれでもよい。好ましくは、E4領域はE4ORF6遺伝子のみからなるものである。好ましくは、E4領域がE4ORF遺伝子のみからなり、ポリAシグナル配列を含まない。また、E4領域は発現調節可能なプロモーターの下流に存在することが好ましい。

本発明のアデノ随伴ウイルスの製造方法においては、ウイルスヘルパー遺伝子を細胞に導入する。
ウイルスヘルパー遺伝子は、プロモーターの制御下にあることが好ましく、発現調節可能なプロモーターの制御下にあってもよい。
ウイルスヘルパー遺伝子は、プロモーターの制御下にあることが好ましく、発現調節可能なプロモーターの制御下にあってもよい。
発現調節可能なプロモーターとしては、刺激の有無により発現のオンとオフを調節できるプロモーターを使用することができる。刺激としては、化学的刺激(内在性ホルモン・ストレス応答、ラクトース、テトラサイクリン又はその誘導体(例えば、ドキシサイクリン)、クミン酸、タンパク質(ラパマイシン、FKCsA、アブシシン酸(ABA)など)、タモキシフェン/Cre-loxP(タモキシフェンにより活性化を誘導できるように改変したCreプロモーターを使用する系)、リボスイッチ)、物理的刺激(青色光、熱)などを挙げることができるが、特に限定されない。
発現調節可能なプロモーターは、薬剤により発現調節可能なプロモーターでもよい。薬剤としては、好ましくはテトラサイクリン又はその誘導体(例えば、ドキシサイクリン)である。
発現調節可能なプロモーターとしては、テトラサイクリン応答性プロモーター、RU486誘導性プロモーター、エクジソン誘導性プロモーター、ラパマイシン誘導性プロモーター、及びメタロチオネインプロモーターなどが挙げられる。具体的なテトラサイクリン応答性プロモーターには、Tetオン/オフシステム(TET systems社製)が挙げられる。
発現調節可能なプロモーターは、テトラサイクリンにより発現調節可能なプロモーターであるTetオン/オフシステムでもよく、Tetオンシステムでもよい。
Tetオンシステムにおいては、プロモーターは、少なくとも一つのTetオペロンを付加的に含む。Tetオペロン(テトラサイクリン制御転写活性化)を使用することにより、抗生物質であるテトラサイクリン又はその誘導体のうちの一つ(例えば、ドキシサイクリン)の存在下において転写のオン又はオフを可逆的に切り替えることができる。細胞内に存在するTetリプレッサータンパク質は、プロモーターへと導入されるTet operator配列に結合することによって発現を遮断する。したがって、TetリプレッサーがTet operator配列と結合している場合には、遺伝子発現は見られない。テトラサイクリン又はドキシサイクリンを添加すると、Tetリプレッサーは隔離されてプロモーター活性を可能とし、遺伝子発現がオンにされる。Tetオペロン系は、Invitrogeから入手可能なpcDNA(商標)4/TO哺乳動物発現ベクターにおいて使用されるTetオペロンなど、広く入手可能である。
Tetオンシステムにおいて、Tet operator配列は、発現を調節したいプロモーターの上流にあっても下流にあってもよい。発現をオフにしたときの発現漏れを減らすという点において、Tet operator配列は、発現調節可能なプロモーターの下流にあることが好ましい。

プロモーターの具体例としては、特に限定されないが、サイトメガロウイルス由来プロモーター(CMVプロモーター)(所望によりエンハンサーを含む)、SV40初期プロモーター、ヒト伸長因子-1α(EF-1α)プロモーター、ヒトユビキチンCプロモーター、レトロウイルスのラウス肉腫ウイルス(RSV)LTRプロモーター、ジヒドロ葉酸還元酵素プロモーター、β-アクチンプロモーター、及びホスホグリセリン酸キナーゼ(PGK)プロモーターなどを挙げることができる。
ウイルスヘルパー遺伝子とは、アデノ随伴ウイルスの複製及びパッケージングを可能にするための非アデノ随伴ウイルス遺伝子である。ウイルスヘルパー遺伝子としては、アデノ随伴ウイルス以外の他種ウイルスに由来する遺伝子が使用される。ウイルスヘルパー遺伝子の具体例としては、アデノウイルス又はヘルペスウイルスに由来するウイルスヘルパー遺伝子を挙げることができ、好ましくは、ウイルスヘルパー遺伝子は、アデノウイルス由来である。
アデノウイルスとは、アデノウイルス科のファミリーの、二本鎖DNAを含む二十面体ヌクレオカプシドを有する非エンベロープ型ウイルスを指す。50超のアデノウイルスのサブタイプがヒトから単離され、多くの更なるサブタイプが他の哺乳動物及び鳥類から単離されている。これらのサブタイプは、アデノウイルス科のファミリーに属し、2つの属(すなわちマストアデノウイルス属及びアビアデノウイルス属)に分類されている。これらのアデノウイルスは、形態学的及び構造的に類似している。しかしながら、ヒトにおいて、アデノウイルスは、異なる免疫学的特性を示し、すなわち血清型に分類される。アデノウイルスの2つのヒト血清型(すなわちAd2及びAd5)が重点的に研究されている。
アデノウイルス由来のウイルスヘルパー遺伝子は、アデノ随伴ウイルスの複製及びパッケージングに関与する遺伝子である。
アデノウイルス由来のウイルスヘルパー遺伝子としては、E1A、E1B、E2A、E4、及びVA-RNAなどを挙げることができる。E1領域の全部又はその一部を有する宿主細胞において、AAVのゲノムが複製されてカプシドにパッケージングされて、ウイルスビリオンを形成するために必要なアデノウイルスゲノムの領域は、E2A領域、E4領域、及びVA-RNA領域である。E4領域による機能について、E4領域のオープンリーディングフレーム6(E4ORF6)によりコードされるE4 34kDa蛋白質がAAVの複製に必要とされる。好ましくは、ウイルスヘルパー遺伝子は、E2遺伝子、E4遺伝子(E4ORF6)、およびVA-RNA遺伝子である。VA-RNA遺伝子は、好ましくは、VA-RNAI遺伝子である。
アデノウイルス由来のウイルスヘルパー遺伝子(E1A、E1B、E2A、E4、及びVA-RNAなど)は、野生型の遺伝子でもよいが、本来有する機能を発揮するものである限り,野生型の遺伝子に、塩基の置換、欠失、挿入または付加等の改変がなされた遺伝子を使用してもよい。
野生型のウイルスヘルパー遺伝子に、塩基の置換、欠失、挿入または付加等の改変を行う場合、改変される塩基の個数は,好ましくは1~20個,より好ましくは1~10個,更に好ましくは1~3個である。改変を加えたウイルスヘルパー遺伝子の塩基配列は,野生型のウイルスヘルパー遺伝子の塩基配列と,好ましくは85%以上の配列同一性を示し,より好ましくは90%以上の配列同一性を示し,更に好ましくは,95%以上の配列同一性を示し,更により好ましくは98%以上の配列同一性を示す。
ウイルスヘルパー遺伝子を細胞に導入する場合、ウイルスヘルパー遺伝子を含むベクターを細胞に導入することができる。
ウイルスヘルパー遺伝子を含むベクターとしては、例えば、プラスミド、ウイルス由来の配列、人工的に設計した核酸などを使用することができ、好ましくは、プラスミドである。
本発明のアデノ随伴ウイルスの製造方法においては、アデノ随伴ウイルス遺伝子を細胞に導入する。

アデノ随伴ウイルス遺伝子とは、一つ又は複数のアデノ随伴ウイルスの血清型に由来する一つ又は複数の核酸配列から構成される遺伝子を指す。アデノ随伴ウイルス遺伝子は、好ましくは、AAVの複製及びパッケージングに関与する遺伝子、及びAAV構成タンパク質をコードする遺伝子である。
好ましくは、アデノ随伴ウイルス遺伝子としては、Rep遺伝子およびCap遺伝子を使用する。Rep遺伝子およびCap遺伝子は、ビリオンの複製とパッケージングに関与するタンパク質をコードしている。Rep遺伝子は野生型では、P5プロモーターおよびp19プロモーターから発現される。Cap領域は、VP1、VP2、およびVP3を発現させる。Cap遺伝子が天然に保持するプロモーターとしてはp40プロモーターを挙げることができる。
アデノ随伴ウイルス遺伝子(Rep遺伝子およびCap遺伝子など)は、野生型の遺伝子でもよいが、本来有する機能を発揮するものである限り、野生型の遺伝子に、塩基の置換、欠失、挿入または付加等の改変がなされた遺伝子を使用してもよい。
野生型のアデノ随伴ウイルス遺伝子(Rep遺伝子およびCap遺伝子など)に、塩基の置換、欠失、挿入または付加等の改変を行う場合、改変される塩基の個数は、好ましくは1~20個、より好ましくは1~10個、更に好ましくは1~3個である。改変を加えたアデノ随伴ウイルス遺伝子の塩基配列は、野生型のアデノ随伴ウイルス遺伝子の塩基配列と、好ましくは85%以上の配列同一性を示し、より好ましくは90%以上の配列同一性を示し、更に好ましくは、95%以上の配列同一性を示し、更により好ましくは98%以上の配列同一性を示す。
Rep遺伝子およびCap遺伝子を細胞に導入する場合、Rep遺伝子およびCap遺伝子を含むベクターを細胞に導入することができる。ベクターにおけるRep遺伝子およびCap遺伝子の配置は特に限定されず、Rep遺伝子は、Cap遺伝子の上流に位置してもよいし、Cap遺伝子の下流に位置してもよいが、好ましくは、Rep遺伝子は、Cap遺伝子の上流に位置する。
Rep遺伝子は、当業者に公知の、ウイルスゲノムの複製に集合的に必要とされるウイルスの複製タンパク質、又は、例えばヒトヘルペスウイルス6(HHV-6)Rep遺伝子など(AAV-2 DNA複製を媒介することが公知)の、その機能的ホモログをコードするAAVゲノムの領域を意味する。したがって、Rep遺伝子のコード領域は少なくとも、AAVのREP78及びREP68(「長い形態のREPタンパク質」)並びにREP52及びREP40(「短い形態のREPタンパク質」)をコードする遺伝子又はその機能的ホモログを含む。本発明で用いられるRep遺伝子のコード領域は、いかなるAAV血清型に由来するものでもよいが、AAV2に由来するものが好ましい。AAV2に由来するものとしては、REP78及びREP68並びにREP52及びREP40、ITRが挙げられる。
Cap遺伝子は、当業者に公知のウイルスのカプシドタンパク質をコードするAAVゲノム中の領域を意味する。これらのカプシドタンパク質の例は、AAVカプシドタンパク質VP1、VP2及びVP3である。本発明において使用するCap遺伝子は、いかなるAAV血清型に由来するものでもよいが、AAV2もしくはAAV5に由来するものが好ましい。特に好ましくはAAV5である。
Rep遺伝子およびCap遺伝子を含むベクターとしては、例えば、プラスミド、ウイルス由来の核酸配列、人工的に設計した核酸などを使用することができ、好ましくは、プラスミドである。
本発明のアデノ随伴ウイルスの製造方法においては、治療又は予防用遺伝子を細胞に導入してもよい。
治療又は予防用遺伝子としては、標的細胞のゲノムにおいては不完全であるか又は失われている遺伝子、または、所望の生物学的又は治療的効果(例えば抗ウイルス機能)を有する非天然タンパク質をコードする遺伝子などを使用することができるが、特に限定されない。治療又は予防用遺伝子の具体例としては、炎症性疾患、自己免疫、慢性及び伝染性疾患(AIDS、癌、神経系疾患、心血管疾患、過剰コレスタ血症などの障害を含む)、貧血及び血友病などの各種の血液疾患、遺伝子欠損(例えば嚢胞性線維形成、ゴーシェ疾患、アデノシンデアミナーゼ(ADA)欠損、気腫など)の治療又は予防のために使用される遺伝子が挙げられる。
治療又は予防用遺伝子としては、がんに対する、及びウイルス疾患に対するアンチセンス療法において有用であるいくつかのアンチセンスオリゴヌクレオチド(例えばmRNAの翻訳開始部位(AUGコドン)周辺の配列に対する相補的短鎖オリゴヌクレオチド)でもよい。
治療又は予防用遺伝子は、治療又は予防用遺伝子を発現させるためのプロモーターに連結されていてもよい。治療又は予防用遺伝子を発現させるためのプロモーターは特に限定されないが、サイトメガロウイルス由来のプロモーター(所望によりエンハンサーを含む)、SV40初期プロモーター、ヒト伸長因子-1α(EF-1α)プロモーター、ヒトユビキチンCプロモーター、レトロウイルスのラウス肉腫ウイルスLTRプロモーター、ジヒドロ葉酸還元酵素プロモーター、β-アクチンプロモーター、及びホスホグリセリン酸キナーゼ(PGK)プロモーターなどを挙げることができる。治療又は予防用遺伝子および遺伝子を発現させるためのプロモーターはITR配列に挟まれていることが好ましい。
本発明におけるベクターの組み合わせの好ましい態様としては、以下の態様を挙げることができる。
(1)Rep遺伝子およびCap遺伝子を含むベクター、E2A遺伝子およびVA-RNA遺伝子を含むベクター、E4遺伝子を含むベクター、並びに所望の治療又は予防用遺伝子を含むベクターを使用し、E4遺伝子を有するベクターの導入比率を小さくする態様:
(2)Rep遺伝子およびCap遺伝子を含むベクター、E2A遺伝子およびVA-RNA遺伝子を含むベクター、E4ORF6遺伝子を含むベクター、並びに所望の治療又は予防用遺伝子を含むベクターを使用し、E4ORF6遺伝子を有するベクターの導入比率を小さくする態様:

本発明によるアデノ随伴ウイルスの製造方法においては、ウイルスヘルパー遺伝子と、アデノ随伴ウイルス遺伝子と、必要な場合には、所望の治療又は予防用遺伝子を含む細胞を培養することによって、アデノ随伴ウイルスを製造することができる。
アデノ随伴ウイルスの製造方法としては、TT(Triple Transfection)法、パッケージングセル法、またはProducer cell法のいずれでもよい。E4遺伝子の代わりにE4ORF6を用いる場合には、特にTT法が好ましい。
TT法とは、Rep遺伝子およびCap遺伝子を含むプラスミド、アデノウイルスヘルパープラスミド、および導入遺伝子(例えば、所望の治療又は予防用遺伝子)を含むプラスミドを同時にHEK293細胞にトランスフェクションすることで組換え型AAV(rAAV)へとパッケージングする方法である。
パッケージングセル法とは、TT法における一部遺伝子をあらかじめ染色体に組み込んだ細胞に対して、残りの遺伝子をトランスフェクションにより導入することで、AAVを産生する方法である。
Producer cell法とは、TT法における全遺伝子を染色体に組み込んだ細胞を用いて、トランスフェクション操作なしでAAVを産生する方法である。
本発明における特に好ましい態様としては、以下の態様を挙げることができる。
Rep遺伝子およびCap遺伝子を搭載したプラスミドと、E2A遺伝子、VA-RNA遺伝子およびE4ORF6を搭載したプラスミドと、GOI遺伝子を搭載したプラスミドとを遺伝子導入する工程を含み、上記E4ORF6を搭載したプラスミドはE4ORF6を発現させるためのenhancer及びPolyAシグナル配列を有さないことを含む、AAV遺伝子の製造方法。
Rep遺伝子およびCap遺伝子を搭載したプラスミドと、E2A遺伝子、VA-RNA遺伝子およびE4ORF6を搭載したプラスミドと、GOI遺伝子を搭載したプラスミドとを遺伝子導入する工程を含み、上記E4ORF6を搭載したプラスミドはE4ORF6を発現させるためのenhancer及びPolyAシグナル配列を有さないことを含む、AAV遺伝子の製造方法。
Rep遺伝子およびCap遺伝子を搭載したプラスミドと、E2A遺伝子、VA-RNA遺伝子およびE4ORF6を搭載したプラスミドと、GOI遺伝子を搭載したプラスミドとを遺伝子導入する工程を含み、上記E4ORF6を搭載したプラスミドのE4ORF6を発現させるプロモーターがCAGプロモーターである、AAV遺伝子の製造方法。
本発明によるアデノ随伴ウイルスの製造方法における細胞の培養は、細胞の培養のための通常の条件において行うことができる。使用する培地、及び培養条件は、当業者であれば適宜選択することができる。培地としては、Expi29 Expression Medium(Thermo Fisher Scientific,A1435101)、10%(vol/vol)ウシ胎仔血清(FBS)を含有するダルベッコ改変イーグル培地(DMEM)、無血清UltraCULTURE(商標)培地(Lonza)などを使用することができるが、特に限定されない。
培養温度は、一般的には25℃~45℃であり、好ましくは30℃~42℃であり、より好ましくは35℃~40℃であり、一例としては37℃である。
CO2濃度は、一般的には3~10%CO2であり、好ましくは5~10%CO2であり、一例としては8%CO2である。

細胞は、任意の体積の培地において培養することができ、例えば、1mLから2000Lの培地において細胞を培養することができ、好ましくは1L~2000Lであり、50L~2000Lがより好ましく、500L~2000Lが特に好ましい。
培養は振盪攪拌しながら行ってもよい。振盪攪拌する場合の攪拌速度は、一般的には50rpm~200rpmであり、好ましくは80~150rpmである。

製造されたアデノ随伴ウイルスの力価は、当業者に公知の通常の方法により測定することができる。例えば、培養後の細胞培養液を回収し、凍結融解にて細胞を破砕した後、遠心分離によって上清を回収する。回収した上清にMgCl2及びBenzonaseを添加して反応を行うことにより、gDNA(ゲノムDNA)や残存プラスミドを消化することができる。上記により得られたアデノ随伴ウイルスを含むサンプルを、ddPCR(Droplet Digital PCR)サンプルとして使用し、ddPCRを行うことによりAAVの力価を測定することが可能である。
<細胞>
本発明の細胞の第一の態様は、外来プロモーターによって発現されるE4ORF6遺伝子を少なくとも有する細胞であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、細胞である。

本発明の細胞は、外来プロモーターによって発現されるE4ORF6遺伝子を少なくとも有している。好ましくは、上記のE4ORF6遺伝子を含むベクターを染色体に有している。染色体中に有するとは、ベクターの全長もしくは一部が染色体に組み込まれていることを意味する。ベクターが染色体に組み込まれていることにより、アデノ随伴ウイルスの産生に必要な遺伝子は細胞内に恒常的に維持されることになり、アデノ随伴ウイルスを任意のタイミングで産生することが可能となる。
E4ORF6遺伝子を含むベクターは、発現されるように導入されていればよいが、永続的な発現となることがより好ましい。永続的な発現とは、細胞が分裂した場合に、E4ORF6遺伝子を含むベクターも複製され、分裂後の細胞もベクターを有することになることをいう。永続的な発現は、ベクターが細胞の染色体の中に組み込まれることや細胞質内で複製する能力をもつベクターを用いることによって行うことができる。
E4ORF6遺伝子を含むベクターは、トランスフェクション、トランスダクション、リポフェクションなどの方法により、細胞に導入することができる。
トランスフェクションとは、化学的手段(例えば、リン酸カルシウム仲介沈殿)、機械的手段(例えば、エレクトロポレーション)または物理的手段(例えば、生体弾(bioballistic)による送達)によって真核細胞の膜を横断して核酸を細胞に導入することをいう。

[材料及び方法]
<細胞培養>
浮遊化HEK293細胞(Thermo Fisher Scientific, R79007)を12mLのExpi29 Expression Medium(Thermo Fisher Scientific,A1435101)で1×107cells/mLとなるように懸濁し、125mL振とうフラスコで24時間培養した。培養液は120rpmで一定に攪拌しながら、37℃、8%CO2条件下でインキュベートを行った。
<細胞培養>

<Real-time PCR>
遺伝子導入後72時間、37℃、8%CO2条件下で培養を行ったHEK細胞を300xgの遠心により回収した。RNeasy Plus Mini Kit(Qiagen)を用いてmRNAを精製し、High Capacity RNA to cDNA Kitを用いた逆転写によりcDNAを作製した。作製したcDNAはTaqMan Gene Expression Master Mix(Applied Biosystems)を用いて、E4ORF6に対するプライマー(seq8、seq9)およびプローブ(seq10)でE4ORF6を検出し、GAPDH(Thermo Fisher Scientific, Hs02786624_g1)に対するTaqMan Gene Expression Assayを用いた規格化によって定量評価した。Seq8~Seq10を、配列表の配列番号8~10に示す。
遺伝子導入後72時間、37℃、8%CO2条件下で培養を行ったHEK細胞を300xgの遠心により回収した。RNeasy Plus Mini Kit(Qiagen)を用いてmRNAを精製し、High Capacity RNA to cDNA Kitを用いた逆転写によりcDNAを作製した。作製したcDNAはTaqMan Gene Expression Master Mix(Applied Biosystems)を用いて、E4ORF6に対するプライマー(seq8、seq9)およびプローブ(seq10)でE4ORF6を検出し、GAPDH(Thermo Fisher Scientific, Hs02786624_g1)に対するTaqMan Gene Expression Assayを用いた規格化によって定量評価した。Seq8~Seq10を、配列表の配列番号8~10に示す。
<AAV力価測定>
遺伝子導入後72時間、37℃、8%CO2条件下で培養を行ったHEK細胞培養液を回収し、-80℃での凍結融解にて細胞を破砕した。その後、13,800×gの遠心分離によって上清を回収し、終濃度2mmol/L MgCl2、25U/mL Benzonaseを添加し、37℃で2時間反応を行い、gDNA(ゲノムDNA)や残存プラスミドを消化した。反応後のサンプルは、95℃で15分、70℃で3分、40℃で3分、20℃で3分の順にインキュベーションし、Benzonaseを失活させ、これをddPCR(Droplet Digital PCR)サンプルとした。ddPCRサンプルはTE buffer(pH8.0)に対して0.05%Pluronic F-68 および10μg/mLウシ胸腺DNAを混合した溶液で50倍に希釈した。氷上でチューブにddPCRtm Supermix for Probes(no dUTP)(BIORAD社) 10μL、希釈済みのddPCRサンプル2μL、プライマー(seq11、seq12、最終濃度900nmol/L)およびプローブ(seq13、最終濃度250nmol/L)、ヌクレアーゼフリーwater(Thermo Fisher Scientific,10977015)を混合して、総液量22μLに調整し、ddPCRを行った。seq11、seq12及びseq13の配列を、配列表の配列番号11~13に示す。

ddPCRにおいては、ドロップレット形成機器(QX200 Droplet Generator、BIORAD)にて調製液からドロップレット形成し、続いて95℃で10分、94℃で30秒および55℃を40サイクル、98℃で10分の順でインキュベーションした。ドロップレットリーダー(QX200 Droplet Reader、BIO RAD)により測定を行い、AAVゲノム力価(vg/mL)を算出した。
<細胞障害性評価>
遺伝子導入細胞の障害性は、遺伝子導入から72時間後trypan blue染色を行い、染色陽性細胞のカウントから障害細胞率(%)を算出した。

E4ORF6を搭載したプラスミドの導入量を変えた時のAAV産生量の変化を評価した。HEK293細胞に以下の組合せで遺伝子を導入し、72時間後のE4ORF6発現量を定量評価した結果を図1に示し、ddPCRによりAAVの力価を測定した結果を図2に示し、Trypanble染色による細胞の障害率を図3に示す。

図7の結果から、E4ORF6遺伝子をコードしたプラスミドのenhancerおよびpolyAシグナル配列を除去することで、E4ORF6の発現量が低下することがわかった。図8の結果から、配列変更によりAAV産生量が改善することがわかった。
E4ORF6遺伝子をTet operator配列を組み込んだ発現調節可能なプロモーターの下にあるRFP(赤色蛍光タンパク質)遺伝子のさらに下流にIRESを介してつないだ設計(Seq14:TET-RFP―IRES―E4ORF6)および、E4ORF6遺伝子下流のpolyAシグナル配列を除去したプラスミド設計(Seq15:TET-RFP―IRES―E4ORF6ΔPolyA)のプラスミドを用いて、それぞれHEK細胞に導入しテトラサイクリン非存在下における増殖による細胞数増加を測定した。同様の条件で、E4領域(seq5)導入細胞における細胞数増加を測定し、比較した結果を図9に示す。
図9の結果から、Seq14、Seq15導入細胞は、E4領域導入に比べて細胞数の増加が大きいことがわかった。

TET-RFP―IRES―E4ORF6および、TET-RFP―IRES―E4ORF6ΔPolyAをHEK細胞に導入し、G418薬剤に処理によってプラスミドが組み込まれた安定発現細胞を作製した。安定発現細胞に対して、Rep、Cap、E2、VA-RNA遺伝子を導入し、AAV産生力価を測定した結果を図10に示す。
図10の結果から、作製したTET-RFP―IRES―E4ORF6および、TET-RFP―IRES―E4ORF6ΔPolyAの安定発現細胞を用いてAAVが産生可能であることを確認した。また、PolyAシグナル配列を除去することで力価が向上することがわかった。
Claims (16)
- E4ORF6遺伝子を外来プロモーターによって発現させることを含むアデノ随伴ウイルスの製造方法であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、アデノ随伴ウイルスの製造方法。
- E4ORF6の発現の低下が、E4ORF6のmRNA発現の低下及び/又はE4ORF6のタンパク質発現の低下である、請求項1に記載のアデノ随伴ウイルスの製造方法。
- エンハンサー、サイトメガロウイルス由来プロモーター、E4ORF6遺伝子及びポリA配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現の量が1/2以下に低下している、請求項1又は2に記載のアデノ随伴ウイルスの製造方法。
- 以下の(a)~(e)の何れか一以上によりE4ORF6の発現が低下している、請求項1から3の何れか一項に記載のアデノ随伴ウイルスの製造方法。
(a)外来プロモーターが、サイトメガロウイルス由来プロモーターよりも転写活性が弱いプロモーターである;
(b)E4ORF6遺伝子のKozak配列が、改変されたKozak配列であるか、Kozak配列を使用しない;
(c)エンハンサー配列が、改変されたエンハンサー配列であるか、エンハンサー配列を使用しない;
(d)ポリAシグナル配列が、改変されたポリAシグナル配列であるか、ポリAシグナル配列を使用しない;
(e)IRES配列を用いてE4ORF6遺伝子を発現させる: - Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量と比較して、E4ORF6遺伝子の細胞への導入量が1/2以下である、請求項1から4の何れか一項に記載のアデノ随伴ウイルスの製造方法。
- 少なくともE4ORF6遺伝子を含むE4領域を細胞に導入することを含む、アデノ随伴ウイルスの製造方法であって、Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量と比較して、E4ORF6遺伝子の細胞への導入量が1/2以下である、アデノ随伴ウイルスの製造方法。
- E4領域の細胞への導入量が、Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量の1/10以下である、請求項6に記載のアデノ随伴ウイルスの製造方法。
- E4領域と、E2A遺伝子及びVA-RNA遺伝子とが、異なるプラスミドベクターにより細胞に導入される、請求項6又は7に記載のアデノ随伴ウイルスの製造方法。
- E4領域がE4ORF6遺伝子のみからなる、請求項6から8の何れか一項に記載のアデノ随伴ウイルスの製造方法。
- 外来プロモーターによって発現されるE4ORF6遺伝子を少なくとも有する細胞であって、エンハンサー、サイトメガロウイルス由来プロモーター、Kozak配列、E4ORF6遺伝子及びポリAシグナル配列を含む発現ユニットを用いてE4ORF6を発現させた場合と比較して、E4ORF6の発現が低下している、細胞。
- 外来プロモーターによって発現されるE4ORF6遺伝子が、以下の(a)~(e)の何れか一以上を満たす発現ベクターにより細胞に導入されている、請求項10に記載の細胞。
(a)外来プロモーターが、サイトメガロウイルス由来プロモーターよりも転写活性が弱いプロモーターである;
(b)E4ORF6遺伝子のKozak配列が、改変されたKozak配列であるか、Kozak配列を有さない;
(c)エンハンサー配列が、改変されたエンハンサー配列であるか、エンハンサー配列を有さない;
(d)ポリAシグナル配列が、改変されたポリAシグナル配列であるか、ポリAシグナル配列を有さない;
(e)IRES配列を用いてE4ORF6遺伝子を発現させる: - 外来プロモーターによって発現されるE4ORF6遺伝子を少なくとも有する細胞であって、Rep遺伝子、Cap遺伝子、E2A遺伝子、VA-RNA遺伝子、所望の治療又は予防用遺伝子のうちのいずれか一つ以上の遺伝子の細胞への導入量よりも、E4ORF6遺伝子の細胞への導入量の方が少ない、細胞。
- さらに、E2A遺伝子及びVA-RNA遺伝子を有する、請求項10~12の何れか一項に記載の細胞。
- さらに、Rep遺伝子を有する、請求項10~13の何れか一項に記載の細胞。
- さらにCap遺伝子、及び/又は所望の治療又は予防用遺伝子を有する、請求項10~14の何れか一項に記載の細胞。
- 外来プロモーターに連結されたE4ORF6を含む発現ベクターであって、以下の(a)~(e)の何れか一以上を満たす発現ベクター。
(a)外来プロモーターが、サイトメガロウイルス由来プロモーターよりも転写活性が弱いプロモーターである;
(b)E4ORF6遺伝子のKozak配列が、改変されたKozak配列であるか、Kozak配列を有さない;
(c)エンハンサー配列が、改変されたエンハンサー配列であるか、エンハンサー配列を有さない;
(d)ポリAシグナル配列が、改変されたポリAシグナル配列であるか、ポリAシグナル配列を有さない;
(e)IRES配列を用いてE4ORF6遺伝子を発現させる:
