Abstract
La présente invention concerne l'utilisation d'une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l'eau à un pH compris entre 5 et 10, comme filtre anti-UV. Elle se rapporte également à une composition cosmétique comprenant une telle lignine.
Description
TITRE : UTILISATION DE LIGNINE HYDROSOLUBLE COMME FILTRE ANTI-UV
La présente invention concerne l’utilisation d’une lignine hydrosoluble obtenue par traitement enzymatique notamment avec au moins une enzyme bilirubine oxydase, comme filtre anti-UV. Elle se rapporte également à une composition cosmétique comprenant une telle lignine.
La protection solaire est un enjeu majeur du domaine cosmétique. En particulier, parmi les différents filtres chimiques ou organiques utilisés, la stabilité de ces produits est un impératif à respecter pour préserver leur efficacité et leur innocuité.
A titre d’illustration, une étude très récente publiée dans Chemical Research in Toxicology (Downs et al, Benzophenone Accumulates over Time from the Degradation of Octocrylene in Commercial Sunscreen Products, 2021 , 34, 4, 1046-1054) laisse entrevoir un effet nocif de l’octocrylène lors de son vieillissement, ce qui n’avait pas été mis en avant jusqu’à présent. Cette étude montre qu’au fil du temps, l’octocrylène se transforme en benzophénone, un composé dont les auteurs expliquent qu’il est génotoxique, cancérogène et perturbateur endocrinien.
Il existe donc un réel besoin de remplacer ce type de filtres chimiques anti-UV, ou du moins d’en diminuer la quantité dans les produits cosmétiques.
La lignine est un polymère naturel disponible en grande quantité, en particulier de par les usines de transformation en pâte à papier ou cellulose. À la différence des polysaccharides, ce polymère phénolique a fait l’objet de peu de valorisation industrielle en dehors des applications « lignosulfonates » et de valorisation énergétique. Or, les applications potentielles de la lignine sont nombreuses. Les lignines aujourd’hui commerciales sont essentiellement solubles dans des solvants organiques ou des solutions aqueuses alcalines très concentrées en base (pH >10). Certaines lignines sont modifiées chimiquement et fonctionnalisées pour donner des lignosulfonates qui sont solubles dans des solvants aqueux de pH neutre, acide ou légèrement basique, mais elles contiennent toujours de nombreuses impuretés.
Le brevet US 10,729,624 décrit l’utilisation de microcapsules mixtes à base de lignines solubles dans des solvants organiques et de filtres chimiques comme filtres UV. La
lignine présente dans un solvant est dissoute dans une solution aqueuse de pH égal à 12, puis ajustée à un pH de 7-10. Le fait que cette lignine ne soit soluble qu’en milieu organique (liposoluble) limite fortement la quantité qui peut être incorporée dans des crèmes solaires, et rend plus difficile leur formulation. En effet, la grande majorité des produits de protection solaire sont des émulsions directes huile-dans-eau (H/E) dans lesquelles la phase dispersée (phase grasse) est minoritaire.
Cependant, il n’existe aucune lignine utilisable en tant que telle, soluble dans un milieu aqueux de pH neutre, qui soit efficace pour protéger les surfaces (par exemple les matières kératiniques telles que la peau mais aussi les supports de revêtement de surface) des IIV, notamment des UVA et/ou des UVB.
La présente invention répond à ce problème.
De manière surprenante, comme montré en exemples, les inventeurs montrent que la lignine hydrosoluble selon l’invention, soluble dans l’eau à un pH compris entre 5 et 10, et solubilisée dans la phase aqueuse continue, améliore clairement les performances anti- UV par rapport à une lignine commerciale liposoluble : cette lignine hydrosoluble présente une protection plus grande dans les UVB (SPF), mais également une absorbance plus importante dans les UVA. De plus, le caractère hydrosoluble permet d’utiliser cette lignine en plus grande quantité, sans compromettre la stabilité de la formule. Les actifs anti-UV sont alors mieux distribués sur les matières kératiniques, après leur étalement. Enfin, cette lignine est biosourcée, et présente un intérêt certain par rapport aux filtres anti-UV chimiques classiques petrosourcés.
La présente invention a donc pour objet l’utilisation d’une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, comme filtre anti-UV. De préférence, le traitement enzymatique est effectué avec au moins une enzyme bilirubine oxydase.
La lignine hydrosoluble obtenue par traitement enzymatique, de préférence avec au moins une enzyme bilirubine oxydase, de préférence ne comprenant pas ou peu de groupes sulfonate, est appelée « lignine selon l’invention » ou « lignine hydrosoluble selon l’invention » dans la présente demande.
La présente invention a également pour objet une composition cosmétique comprenant :
une phase aqueuse comprenant au moins une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, et une phase huileuse et/ou au moins un polymère épaississant hydrophile choisi parmi la cellulose et ses dérivés, les gommes naturelles, les scléroglucanes, les dérivés de chitine et de chitosane, les carraghénanes et les gellanes.
De préférence, la composition selon l’invention est substantiellement exempte de microcapsules. Par « substantiellement exempte », on entend que la composition comprend moins de 1 % en poids par rapport au poids total de composition, de préférence moins de 0,5% en poids, de préférence moins de 0,3% en poids, de préférence moins de 0,1 % en poids de microcapsules. De préférence, la composition selon l’invention est totalement exempte de microcapsules.
La présente invention a également pour objet un procédé cosmétique non- thérapeutique de soin des matières kératiniques, de préférence la peau, comprenant l’application sur lesdites matières kératiniques, de préférence la peau, d’une composition cosmétique telle que décrite ci-avant.
LIGNINE HYDROSOLUBLE SELON L’INVENTION
La lignine hydrosoluble selon l’invention est obtenue par traitement enzymatique. Cette modification par voie enzymatique est spécifique. En effet, la lignine obtenue diffère d'une lignine soluble obtenue par voie chimique, par la faible présence ou l’absence d'impuretés de synthèse par voie chimique et l’absence de groupes sulfate ou sulfonate notamment. De préférence, le traitement enzymatique est effectué avec au moins une enzyme bilirubine oxydase.
La lignine selon l’invention est hydrosoluble. Par « hydrosoluble », on entend que la lignine selon l’invention est soluble dans une solution aqueuse, de préférence dans l’eau, à une concentration allant jusqu’à 90% en poids par rapport au poids total de solution. La solubilité de la lignine est appréciée à température ambiante, de préférence dans une solution aqueuse, typiquement de l’eau, par microscopie et par re-dissolution spontanée de la lignine lyophilisée dans un solvant aqueux. On n’observe aucun précipité.
En outre, la lignine hydrosoluble selon l’invention est soluble dans l’eau à un pH compris entre 5 et 10, de préférence compris entre 6 et 8.
De préférence, la lignine hydrosoluble selon l’invention est obtenue par un procédé comprenant les étapes suivantes : a- mélange de la lignine avec de l’eau comprenant de préférence un agent tampon, b- de préférence, traitement aux ultrasons du mélange obtenu en fin d’étape a, c- traitement enzymatique de la lignine obtenue en a ou en b, avec au moins une enzyme bilirubine oxydase, en présence ou absence d’un médiateur rédox, pour obtenir une lignine soluble en milieu aqueux de pH supérieur ou égal à 5 et inférieur ou égal à 10, de préférence compris entre 6 et 8, et d- séparation de la lignine hydrosoluble des sels du mélange et d’éventuels composés insolubles obtenus en c, de préférence par dialyse ou centrifugation.
Typiquement, un procédé permettant d’obtenir la lignine hydrosoluble selon l’invention comprend la solubilisation de lignine dans une solution comprenant de l’eau, et de préférence également un agent tampon (étape a). Selon un mode de réalisation préféré, la solubilisation de lignine a lieu dans l’eau comme unique solvant, et de préférence avec un agent tampon.
Selon un mode de réalisation, la lignine est du type kraft (ou lignine alcaline). On parle de lignine kraft lorsqu’elle est obtenue par un procédé kraft (aussi connu comme « kraft pulping » ou processus au sulfate) qui est un processus de conversion du bois en pâte à papier. Le procédé kraft comprend le traitement de copeaux de bois avec un mélange chaud d'eau, d'hydroxyde de sodium, et de sulfure de sodium, cassant les fibres de bois et séparant la lignine et l'hémicellulose de la cellulose. Cette technique comprend des étapes mécaniques et chimiques, notamment dits d’imprégnation et de cuisson.
Selon un mode de réalisation, la lignine est du type organosolv.
Par exemple, un agent tampon est choisi parmi un tampon borate, un tampon phosphate de sodium ou tout autre tampon permettant de maintenir un pH neutre à basique.
La lignine dispersée dans la solution subit de préférence un traitement aux ultrasons (étape b), de manière à fragmenter le matériau en de petites particules qui pourront mieux réagir avec les enzymes.
Ensuite, le traitement enzymatique de la lignine est effectué (étape c). Ce traitement enzymatique de la lignine selon l’invention comprend la mise en contact de la solution aqueuse de lignine dispersée avec au moins une enzyme bilirubine oxydase (BOD), en présence ou absence d’un médiateur rédox, et l’obtention d’une lignine soluble en milieu de pH supérieur ou égal à 6, de préférence supérieur ou égal à 7. En effet, des enzymes du type bilirubine oxydase, qui sont stables et actives en milieu acide mais aussi en milieu
neutre et basique, permettent de modifier la lignine de manière à la rendre soluble à pH neutre ou basique (i.e. à pH 5 à 10). De plus, ces bilirubines oxydases sont plus actives et plus stables que les laccases à des températures plus élevées (jusqu’à 70°C).
Selon une variante, la bilirubine oxydase (BOD) est d’origine fongique. Selon un mode de réalisation, la BOD utilisée dans le cadre de la présente invention est une BOD décrite dans la demande internationale WO 2012/160517 (BOD E.C. 1.3.3.5. d’origine Magnaporthe oryzae).
Selon une autre variante, la BOD est d’origine bactérienne. Selon un mode de réalisation, la BOD utilisée dans le cadre de la présente invention est une BOD décrite dans la demande internationale WO 2011/117839 (BOD E.C. 1.3.3.5. d’origine Bacillus pumilus).
Typiquement, la bilirubine oxydase est ajoutée à la solution aqueuse (de préférence avec un agent tampon) contenant la lignine, en présence ou absence d’un médiateur rédox, puis placée dans des conditions permettant son action enzymatique sur la lignine afin de la solubiliser. Typiquement, la bilirubine oxydase est incubée entre 20°C et 80°C, de préférence sous agitation et apport d’oxygène. Typiquement, l’action enzymatique se déroule pendant 2 à 48 heures, de préférence pendant 10 à 24 heures.
Parmi les médiateurs rédox, on peut citer par exemple le 2,2-azino-bis-[3- ethylbenzthiazoline-6-sulfonicacid] (ABTS), le 2,6-dimethoxyphenol (2,6-DMP), Syringaldazine, la bilirubine conjuguée ou non conjuguée, ou les composés d’osmiums.
La lignine obtenue après traitement enzymatique en solvant aqueux est séparée des sels et d’éventuels composés insolubles, par exemple par dialyse ou centrifugation (étape d).
Selon un mode de réalisation préféré, on sépare, par exemple par centrifugation, les insolubles de la lignine hydrosoluble. La lignine hydrosoluble est présente dans le surnageant lorsqu’on utilise la centrifugation.
Selon un mode de réalisation, la lignine hydrosoluble est lyophilisée.
La lignine hydrosoluble selon l’invention est soluble dans l’eau à un pH supérieur ou égal à 5, de préférence supérieur ou égal à 6, de préférence supérieur ou égal à 7, et inférieur à 10, de préférence inférieur à 9, de préférence inférieur à 8.
De préférence, la lignine est soluble sur une plage de pH allant de 6 à 8.
Selon une variante préférée, la lignine hydrosoluble selon la présente invention ne comprend pas ou peu de groupes sulfonate. Par « peu de groupes sulfonate », on entend une teneur massique maximale de 1 % en poids de groupes sulfonate par rapport au poids total de lignine. Une faible quantité de groupes sulfonate permet avantageusement d’apporter à la lignine hydrosoluble des propriétés uniques d’intérêt, en particulier une très faible transmittance.
UTILISATION DE LA LIGNINE COMME FILTRE ANTI-UV
La présente invention a pour objet l’utilisation d’une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, comme filtre anti-UV. La présente invention a également pour objet l’utilisation d’une composition comprenant une phase aqueuse, ladite phase aqueuse comprenant une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, comme filtre anti-UV. De préférence, le traitement enzymatique est effectué avec au moins une enzyme bilirubine oxydase. De préférence, la lignine hydrosoluble ne comprend pas ou peu de groupes sulfonate.
En effet, de préférence, la lignine hydrosoluble selon l’invention est solubilisée dans la phase aqueuse d’une composition.
Cette utilisation est applicable dans le domaine cosmétique, notamment pour formuler des crèmes solaires mais aussi des compositions de maquillage ou de soin ; et dans d’autres domaines. Par exemple, la lignine selon l’invention peut être utilisée dans différents matériaux, dans une composition de peinture ou de vernis, dans une composition pesticide, dans des textiles, dans des compositions de bétons ou de ciments, dans des films alimentaires ou différents revêtements ou substrats. En effet, lorsqu’elle est incorporée dans une composition ou un matériau ou un revêtement ou un substrat, elle permet, après application ou utilisation, d’apporter à ladite composition ou audit matériau ou audit revêtement ou audit substrat des propriétés anti-UV.
De préférence, la présente invention a pour objet l’utilisation d’une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, comme filtre anti-UV dans des compositions cosmétiques, par exemple des compositions de maquillage ou de soin.
De préférence, la présente invention a également pour objet l’utilisation d’une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à
un pH compris entre 5 et 10, comme filtre anti-UV dans une composition de peinture ou de vernis, dans une composition pesticide, dans des textiles, dans des compositions de bétons ou de ciments, dans des films alimentaires ou dans différents revêtements ou substrats.
COMPOSITION COSMETIQUE
La composition cosmétique selon l’invention comprend au moins une phase aqueuse comprenant au moins une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10. De préférence, le traitement enzymatique est effectué avec au moins une enzyme bilirubine oxydase. De préférence, la lignine hydrosoluble ne comprend pas ou peu de groupes sulfonate.
De préférence, cette composition comprend une phase aqueuse dans laquelle la lignine hydrosoluble selon l’invention est solubilisée, et une phase huileuse.
Dans ce cas, typiquement, la composition selon l’invention est une émulsion. De préférence, la phase huileuse est dispersée dans la phase aqueuse, et on obtient alors une émulsion directe (huile-dans-eau ou H/E). La phase aqueuse peut également être dispersée dans la phase huileuse, et on obtient alors une émulsion inverse (eau-dans-huile ou E/H).
La composition cosmétique selon l’invention peut également comprendre un polymère épaississant, pour obtenir une composition gélifiée, par exemple un gel.
Phase aqueuse
La phase aqueuse selon l’invention comprend au moins de l’eau et la lignine hydrosoluble selon l’invention. La lignine hydrosoluble selon l’invention est solubilisée dans l’eau.
De préférence, la composition selon l’invention comprend au moins 50% en poids d’eau par rapport au poids total de composition, de préférence au moins 60% en poids, de préférence au moins 70% en poids.
De préférence, la composition selon l’invention comprend de 50% à 99% en poids d’eau par rapport au poids total de composition, de préférence de 65% à 95% en poids, de préférence de 70% à 90% en poids.
La phase aqueuse peut également comprendre au moins un additif choisi parmi les conservateurs et les humectants. Typiquement, le conservateur peut être choisi parmi les parabènes et l’alcool benzylique. Typiquement, l’humectant peut être choisi parmi les polyols, de préférence la glycérine ou le sorbitol.
De préférence, la composition selon l’invention comprend de 55% à 95% en poids de phase aqueuse par rapport au poids total de composition, de préférence de 65% à 90% en poids, de préférence de 75% à 90% en poids.
De préférence, si la composition selon l’invention se présente sous forme d’émulsion inverse, elle comprend de 1 % à 50% en poids d’eau par rapport au poids total de composition, de préférence de 5% à 45% en poids.
Lignine hydrosoluble selon l’invention
La phase aqueuse comprend au moins une lignine hydrosoluble selon l’invention.
La lignine hydrosoluble selon l’invention est telle que définie précédemment.
De préférence, la composition selon l’invention comprend au moins 1% en poids de lignine hydrosoluble selon l’invention par rapport au poids total de composition, de préférence au moins 1 ,5% en poids, de préférence au moins 2% en poids.
De préférence, la composition selon l’invention comprend de 1% à 30% en poids de lignine hydrosoluble selon l’invention par rapport au poids total de composition, de préférence de 1 ,5% à 25% en poids, de préférence de 2% à 20% en poids.
Polymère épaississant hydrophile
La phase aqueuse selon l’invention peut comprendre au moins un polymère épaississant hydrophile choisi parmi la cellulose et ses dérivés, les gommes naturelles, les scléroglucanes, les dérivés de chitine et de chitosane, les carraghénanes et les gellanes.
Par polymère épaississant hydrophile, on entend un polymère épaississant soluble ou dispersible dans l'eau.
Ledit polymère épaississant hydrophile est de préférence choisi parmi :
- la cellulose et ses dérivés, tels que la carboxyméthylcellulose, l'hydroxyméthylcellullose, l'hydroéthylcellulose, l'hydroxypropylcellulose ou l'hydroxypropylméthylcellulose ;
- les gommes naturelles, telles que la gomme de xanthane, la gomme de guar, la gomme arabique ou la gomme de caroube ;
- les scléroglucanes ;
- les dérivés de chitine et de chitosane ;
- les carraghénanes ; et
- les gellanes.
A titre complémentaire, la composition selon l’invention peut comprendre, outre l’épaississant ci-dessus, un épaississant supplémentaire de type carbomère.
De préférence, la composition selon l’invention comprend une phase aqueuse dans laquelle la lignine hydrosoluble selon l’invention est solubilisée, et au moins un polymère épaississant hydrophile choisi parmi la cellulose et ses dérivés, les gommes naturelles, les scléroglucanes, les dérivés de chitine et de chitosane, les carraghénanes et les gellanes.
Une telle composition est un gel aqueux.
De préférence, le polymère épaississant hydrophile est présent en une quantité allant de 0,1% à 5% en poids par rapport au poids total de composition, de préférence de 0,2% à 3% en poids.
Phase huileuse
La composition selon l’invention peut comprendre une phase huileuse. La phase huileuse comprend au moins un composé lipophile (i.e. non soluble dans l’eau), de préférence choisi parmi les huiles, les cires, les beurres et leurs mélanges.
Par « huile », on entend un composé lipophile liquide à température ambiante.
Par « cire », on entend un composé lipophile solide à température ambiante.
Par « beurre », on entend un composé lipophile semi-liquide ou semi-solide à température ambiante.
Les huiles, cires et beurres sont bien connus de l’art antérieur. A titre d’exemples non limitatifs, on peut notamment citer les huiles végétales, la lanoline, le dicaprylyl
carbonate, les beurres d’origine végétale comme le beurre de karité ou le beurre de cacao, ou les cires d’origine végétale ou d’origine animale, telle que par exemple la cire d’abeille.
De préférence, la composition selon l’invention comprend de 5% à 50% en poids de phase huileuse par rapport au poids total de composition, de préférence de 10% à 40% en poids, de préférence de 15% à 30% en poids.
De préférence, si la composition selon l’invention se présente sous forme d’émulsion inverse, elle comprend de 50% à 95% en poids de phase huileuse par rapport au poids total de composition.
Une telle composition sous forme d’émulsion peut également comprendre au moins un tensioactif, au moins un filtre UV lipophile et/ou au moins une matière colorante.
Tensioactif
La composition selon l’invention peut comprendre au moins un tensioactif. De préférence, le tensioactif est choisi parmi les tensioactifs anioniques, les tensioactifs non ioniques, les tensioactifs amphotères et leurs mélanges.
De préférence, la composition selon l’invention comprend au moins un tensioactif non ionique.
Filtre UV lipophile
La composition selon l’invention peut comprendre au moins un filtre UV lipophile. Ledit filtre est solubilisé dans la phase huileuse.
De préférence, ledit filtre UV est choisi parmi les filtres UV minéraux, les filtres UV organiques et leurs mélanges.
Comme filtre UV minéral, on peut citer l’oxyde de cérium, le dioxyde de titane (TiO2), l’oxyde de zinc ou leurs mélanges, de préférence le dioxyde de titane (TiO2), l’oxyde de zinc ou leurs mélanges. Le filtre UV minéral peut être fonctionnalisé par un traitement de surface, par exemple avec un tensioactif ou bien avec un oxyde métallique (notamment silice ou alumine), pour lui conférer différentes propriétés.
Comme filtre UV organique, on peut citer tout filtre UV organique autorisé. De préférence, les filtres UV organiques utilisables selon la présente invention sont choisis parmi les dérivés du benzylidène camphre, les dérivés de la benzophénone, l’acide para- aminobenzoïque et ses dérivés, les dérivés de triazine, les dérivés du dibenzoylméthane, les dérivés salicyliques, les dérivés cinnamiques, les dérivés de p,p-diphénylacrylate, les dérivés du phényl benzimidazole, les dérivés du phényl benzotriazole et les dérivés du benzalmalonate.
De préférence, le filtre UV organique est choisi parmi le camphor benzalkonium methosulfate, l’éthylhexyl méthoxycinnamate, l'homosalate, l’acide para-aminobenzoïque, l’isoamyl p-méthoxycinnamate, le PEG-25 PABA, l’éthylhexyl dimethyl PABA (ou padimate O), l’octocrylène, l’anisotriazine, le polysilicone-15, le drométrizole trisiloxane, l’oxybenzone (ou benzophenone-3), l’acide sulfonique téréphthalylidène dicamphre (ou ecamsule), l’acide sulfonique benzylidène dicamphre, le polyacrylamidométhyl benzylidène dicamphre, l’octyl triazone (ou ethylhexyl triazone), le butyl methoxydibenzoylméthane (BMDBM ou avobenzone), le 4-méthylbenzylidene camphre, le diéthylhexyl butamido triazone, le diéthylamino hydroxybenzoyl hexyl benzoate, le phenyl-salicylate, l’éthylhexyl salicylate, le disodium phenyl dibenzimidazole tetrasulfonate, méthylène bis-benzotriazolyl tetramethylbutylphenol, l’acide phénylbenzimidazole sulfonique (ou ensulizole), la benzophenone-4 ou 5 et le bis-ethylhexyloxyphenol methoxyphenyl triazine.
De préférence, la composition selon l’invention comprend au moins un filtre UV lipophile choisi parmi les filtres UV organiques. De préférence, la composition selon l’invention comprend au moins un filtre UV lipophile choisi parmi les filtres UV minéraux, de préférence parmi le TiO2 et l’oxyde de cérium, avantageusement le TiO2.
Sans vouloir être lié par une quelconque théorie, comme montré en exemples, la lignine hydrosoluble selon l’invention présente un effet synergique avec un filtre anti-UV minéral de type TiO2 ou oxyde de cérium, de préférence avec un filtre anti-UV minéral de type TiO2.
De préférence, quelle que soit la nature de la composition, le(s) f iltre(s) UV est(sont) présent(s) en une quantité allant de 1 % à 20% en poids par rapport au poids total de composition, de préférence de 2% à 15% en poids, de préférence de 5% à 12% en poids.
Matière colorante
La composition selon l’invention peut comprendre au moins une matière colorante.
De préférence, la matière colorante est choisie parmi les pigments, les nacres et les colorants liposolubles.
Les pigments sont de préférence choisis parmi les oxydes de zirconium ou de cérium, les oxydes de fer ou de chrome, notamment l'oxyde de fer rouge, l'oxyde de fer jaune, l'oxyde de fer brun, l'oxyde de fer noir, le violet de manganèse, le bleu outremer, la curcumine, le béta-carotène, la chlorophylle, l'hydrate de chrome et le bleu ferrique, et leurs mélanges.
Les nacres sont de préférence choisies parmi les borosilicates ou le mica, naturel ou synthétique, et éventuellement recouvert d'oxyde de titane (TiC>2) , d'oxyde de fer (Fe20s) (rouge, jaune ou noir), d'oxyde de chrome (Cr20s) ou d'un mélange de ces oxydes.
Les colorants liposolubles sont de préférence choisis parmi les quinoléines, les anthraquinones, les triarylméthanes, les xanthènes, les cyanines, les phtalocyanines, les colorants nitroso, azo et indigoïdes.
L’invention est maintenant illustrée par les exemples qui suivent.
Dans les exemples, tous les pourcentages sont donnés en poids par rapport au poids total de composition, sauf indication contraire ; la température est celle ambiante (20- 25°C) et exprimée en degré Celsius sauf indication contraire, la pression est la pression atmosphérique (101325 Pa), sauf indication contraire.
Exemples
Exemple 1 - Préparation de lignine hydrosoluble selon l’invention
Le procédé est décrit dans la demande WO2019/076893 :
2g de lignine kraft, achetée chez Sigma-Aldrich sous la référence (370959) sont repris dans 100mL de tampon 50mM Borate pH9 (étape a).
5,2mg de BOD de Bacillus pumilus sont ensuite ajoutés à cette solution, puis cette dernière est placée à 60°C sous agitation avec un barreau aimanté à 100 tr/m et un bullage d’air comprimé à 0,1 L.min-1, pendant 16 heures (étape c). La solution est ensuite dialysée dans un boudin de dialyse de 100 mL de 10 kDa, dans trois bains de 4 L d’eau mQ, le
premier de 2 heures, le deuxième de 4 heures et le troisième pour la nuit (étape d). La solution est centrifugée 5 minutes à 1500 tr/mn.
Le surnageant contenant la lignine hydrosoluble selon l’invention est lyophilisé pendant la nuit et conservé à température ambiante.
Exemple 2 - Evaluation des propriétés anti-UV de la lignine hydrosoluble selon l’invention
Une crème solaire de référence (P2) utilisée pour l’étalonnage des tests in vivo de mesure des facteurs de protection solaire (FPS ou Sun Protection Factors - SPF) est utilisée ici comme base de formule pour tester l’efficacité de la lignine (cf tableau 1 ).
Composition de la crème solaire de référence P2
2% ou 4% en poids de la lignine hydrosoluble selon l’invention, par rapport au poids total de composition, sont incorporés dans la phase aqueuse continue de P2.
Séparément, 2% ou 4% en poids d’une lignine commerciale lipophile (Lignine Kraft, Sigma Aldrich- ref 370959) sont dissouts dans la phase grasse dispersée de P2 afin de comparer les deux procédés et les deux types de lignine.
Des mesures de SPF ont été réalisées in vitro sur ces quatre crèmes avec la lignine commerciale (tableau 2) et la lignine hydrosoluble de l’invention (tableau 3).
La mesure du SPF est réalisée in vitro à l’aide de substrats en PMMA moulés de rugosité Ra~4pm (HelioScreen Labs, HELIOPLATE HD6). Les différentes formulations sont alors étalées sur ces supports polymère en respectant le protocole de la norme ISO 24443 {Détermination in vitro de la photoprotection UVA). 1 ,3 mg/cm2 de produit est alors déposé sous forme de gouttes uniformes et réparties de façon symétrique sur le substrat. L’étalement se poursuit alors par de petits mouvements circulaires avec l’index pendant moins de 30 secondes avant de reproduire des va et vient horizontaux. Les échantillons sont ensuite mis dans l’obscurité et laissés à sécher pendant 2 heures. Leur transmittance est ensuite mesurée à l’aide d’un spectrophotomètre muni d’une sphère d’intégration. Le SPF est finalement calculé à l’aide de l’équation suivante :

Le tableau 3 montre que l’ajout de 4% en poids de lignine hydrosoluble selon l’invention permet de plus que doubler la valeur du SPF (20,9 à 42,2).
Comme le montrent ces résultats, le SPF augmente plus avec l’utilisation de lignine hydrosoluble selon l’invention dans la phase aqueuse continue qu’avec un ajout de lignine lipophile commerciale dans la phase grasse dispersée, ce qui souligne l’intérêt de la lignine hydrosoluble.
La comparaison des spectres de transmittance montre de manière plus globale l’intérêt d’utiliser de la lignine hydrosoluble, dont l’absorbance est nettement plus importante que la lignine commerciale dans les UVA.
Des formulations additionnelles de P2, comprenant 3% en poids de TiO2 à la place des deux filtres organiques (acide para-amino benzoïque et oxybenzone) de la P2 (cf Tableau 1 ), et 2%, 4%, 6% ou 8% en poids de lignine hydrosoluble dans la phase aqueuse, ont été réalisées. Elles comprennent le filtre minéral de dioxyde de titane (TiO2).
Ces formulations montrent l’intérêt d’associer la lignine hydrosoluble à d’autres filtres, afin d’améliorer les propriétés anti-UV des produits solaires. Les résultats obtenus laissent supposer l’existence d’une synergie entre les filtres minéraux et la lignine hydrosoluble. En effet, une crème de référence P2 contenant, comme seuls filtres, 3% en poids de TiO2 et 6% en poids de lignine hydrosoluble, permet d’obtenir un SPF in vitro de plus de 34.
La lignine hydrosoluble selon l’invention présente plusieurs avantages incontestables vis-à-vis notamment de la lignine commerciale liposoluble:
1 - à quantité égale dans les formules, elle apporte toujours un SPF significativement plus élevé que la lignine commerciale ;
2- les spectres d’absorbance des crèmes formulées avec la lignine hydrosoluble présentent toujours une absorbance plus élevée sur l’ensemble du spectre UVB et UVA que ceux des crèmes formulées avec la lignine commerciale ;
3- les spectres d’absorbances des crèmes formulées avec la lignine hydrosoluble présentent également une forme différente, avec une absorbance beaucoup plus importante dans les UVA. La lignine hydrosoluble de l’invention présente donc une meilleure protection dans les UVA ;
4- l’utilisation de la lignine hydrosoluble facilite la formulation des compositions solaires, car elle permet l’incorporation d’une quantité plus élevée de lignine. En particulier, il est possible de dépasser 10% en poids, ce qui semble impossible avec la lignine liposoluble commerciale incorporée dans la phase grasse dispersée (car cela engendre des problèmes de stabilité et de texture) ; et
5- le fait d’incorporer la lignine hydrosoluble dans la phase aqueuse continue permet une meilleure distribution des filtres, une fois la crème étalée sur la peau et la fraction volatile évaporée.
Exemple 3 - Comparaison de la transmittance de la lignine hydrosoluble selon l’invention avec une lignine hydrosoluble traitée chimiquement
La composition P2 selon le tableau 1 de l’exemple 2, sans les deux filtres organiques, a été mélangée avec 4% en poids de lignine hydrosoluble selon l’invention par rapport au poids total de composition. Ladite lignine hydrosoluble selon l’invention a été obtenue selon l’exemple 1. On obtient ainsi une composition selon l’invention.
La composition P2 selon le tableau 1 de l’exemple 2, sans les deux filtres organiques, a également été mélangée avec 4% en poids par rapport au poids total de composition de lignine hydrosoluble obtenue par traitement chimique. Ladite lignine hydrosoluble a été obtenue par un procédé au sulfite. Elle comprend des groupes sulfonate. On obtient ainsi une composition comparative.
[Fig 1 ] La figure 1 illustre les spectres de transmittance obtenus avec la composition P2 telle quelle sans les deux filtres organiques (« P2 sans filtre », comparatif), la composition comparative avec lignine traitée chimiquement (« P2 sans filtre + 4% lignine
comparative sulfonatée ») et la composition selon l’invention avec lignine traitée enzymatiquement (« P2 sans filtre + 4% lignine selon l’invention »).
Les résultats montrent que seule la composition selon l’invention présente la plus faible transmittance T et donc une absorbance A (A = -log (T)) satisfaisante à la fois dans les UVA et les UVB.
Le traitement enzymatique, au contraire d’un traitement chimique, apporte ainsi des propriétés uniques d’intérêt à la lignine hydrosoluble.
Exemple 4 - Etude de la combinaison d’oxyde de cérium et de lignine hydrosoluble selon l’invention
Des mesures d'absorbance UV ont été réalisées sur une dispersion de nanoparticules de CeC>2, une solution de lignine enzymatique et un mélange des deux afin d'évaluer la synergie éventuelle du mélange. Pour cela, 20 ml d’une dispersion de nanoparticules de CeC>2 à 0,05g/l et pH=1 ,5 a été mélangée à 20ml d’une solution de lignine enzymatique à la même concentration massique (0,05 g/l) donnant un mélange à 0,05g/l d'actifs au total. Le pH du mélange a ensuite été ajusté à pH=8 (NaOH) où la nouvelle dispersion était stable et transparente. 20ml d’une dispersion de nanoparticules de CeC>2 à 0,05 g/l et pH=1 ,5 a été ajustée à pH=8 (NaOH). 20ml d’une solution de lignine enzymatique à 0,05g/l a été ajustée à pH=8 (HCl).
La figure 2 est un graphique représentant l’évolution de l’absorbance de ces trois solutions en fonction de la longueur d’onde. Ce graphique montre que le mélange des deux actifs (complexes CeO2-lignine) présente une absorbance plus élevée que les deux actifs pris séparément pour une même concentration totale d’actifs de 0,05g/l, illustrant donc un effet de synergie entre la lignine enzymatique et l’oxyde de cérium. Cela permet de réduire la concentration massique de l'un des deux constituants.
Claims
1. Utilisation d’une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, comme filtre anti-UV, la lignine hydrosoluble ne comprenant pas de groupes sulfonate ou comprenant une teneur massique maximale de 1% en poids de groupes sulfonate par rapport au poids total de lignine hydrosoluble.
2. Composition cosmétique comprenant : une phase aqueuse comprenant au moins une lignine hydrosoluble obtenue par traitement enzymatique, ladite lignine étant soluble dans l’eau à un pH compris entre 5 et 10, la lignine hydrosoluble ne comprenant pas de groupes sulfonate ou comprenant une teneur massique maximale de 1 % en poids de groupes sulfonate par rapport au poids total de lignine hydrosoluble, et une phase huileuse et/ou au moins un polymère épaississant hydrophile choisi parmi la cellulose et ses dérivés, les gommes naturelles, les scléroglucanes, les dérivés de chitine et de chitosane, les carraghénanes et les gellanes.
3. Utilisation selon la revendication 1 ou composition cosmétique selon la revendication 2, dans laquelle le traitement enzymatique est effectué avec au moins une enzyme bilirubine oxydase.
4. Utilisation selon la revendication 1 ou 3, ou composition cosmétique selon la revendication 2 ou 3, dans laquelle la lignine hydrosoluble est obtenue par un procédé comprenant les étapes suivantes : a- mélange de la lignine avec de l’eau comprenant de préférence un agent tampon, b- de préférence, traitement aux ultrasons du mélange obtenu en fin d’étape a, c- traitement enzymatique de la lignine obtenue en a ou en b, avec au moins une enzyme bilirubine oxydase, en présence ou absence d’un médiateur rédox, pour obtenir une lignine soluble en milieu aqueux de pH supérieur ou égal à 5 et inférieur ou égal à 10, de préférence compris entre 6 et 8, d- séparation de la lignine hydrosoluble des sels du mélange et d’éventuels composés insolubles obtenus en c, de préférence par dialyse ou centrifugation.
5. Utilisation selon la revendication 3 ou 4, ou composition cosmétique selon la revendication 3 ou 4, dans laquelle la bilirubine oxydase est d’origine fongique, de
préférence est une bilirubine oxydase d’origine Magnaporthe oryzae ; ou bien la bilirubine oxydase est d’origine bactérienne, de préférence est une bilirubine oxydase d’origine Bacillus pumilus.
6. Composition cosmétique selon l’une des revendications 2 à 5, qui comprend une phase aqueuse dans laquelle la lignine hydrosoluble est solubilisée, et une phase huileuse.
7. Composition cosmétique selon la revendication 6, dans laquelle la phase huileuse est dispersée dans la phase aqueuse, ou bien la phase aqueuse est dispersée dans la phase huileuse.
8. Composition cosmétique selon la revendication 6 ou 7, qui comprend au moins un tensioactif, au moins un filtre LIV lipophile et/ou au moins une matière colorante.
9. Composition cosmétique selon l’une des revendications 6 à 8, qui comprend au moins un filtre LIV lipophile choisi parmi les filtres LIV minéraux, les filtres LIV organiques et leurs mélanges, de préférence parmi l’oxyde de cérium, le dioxyde de titane, l’oxyde de zinc, les dérivés du benzylidène camphre, les dérivés de la benzophénone, l’acide para-aminobenzoïque et ses dérivés, les dérivés de triazine, les dérivés du dibenzoylméthane, les dérivés salicyliques, les dérivés cinnamiques, les dérivés de |3,p- diphénylacrylate, les dérivés du phényl benzimidazole, les dérivés du phényl benzotriazole et les dérivés du benzalmalonate, plus préférentiellement parmi le dioxyde de titane, l’oxyde de zinc, le camphor benzalkonium methosulfate, l’éthylhexyl méthoxycinnamate, l'homosalate, l’acide para-aminobenzoïque, l’isoamyl p-méthoxycinnamate, le PEG-25 PABA, l’éthylhexyl dimethyl PABA, l’octocrylène, l’anisotriazine, le polysilicone-15, le drométrizole trisiloxane, l’oxybenzone, l’acide sulfonique téréphthalylidène dicamphre, l’acide sulfonique benzylidène dicamphre, le polyacrylamidométhyl benzylidène dicamphre, l’octyl triazone, le butyl methoxydibenzoylméthane, le 4-méthylbenzylidene camphre, le diéthylhexyl butamido triazone, le diéthylamino hydroxybenzoyl hexyl benzoate, le phenylsalicylate, l’éthylhexyl salicylate, le disodium phenyl dibenzimidazole tetrasulfonate, méthylène bis-benzotriazolyl, tetramethylbutylphenol, l’acide phénylbenzimidazole sulfonique, la benzophenone-4 ou 5 et le bis-ethylhexyloxyphenol methoxyphenyl triazine.
10. Composition cosmétique selon l’une des revendications 2 à 5, qui comprend une phase aqueuse dans laquelle la lignine hydrosoluble est solubilisée, et au moins un polymère épaississant hydrophile choisi parmi la cellulose et ses dérivés, les
gommes naturelles, les scléroglucanes, les dérivés de chitine et de chitosane, les carraghénanes et les gellanes.
