본원 발명의 일 목적을 달성하기 위하여 본원 발명에 따른 스페로이드 또는 오가노이드 체외 모델을 3차원으로 모사하기 위한 미세 유체 칩은, 공급부, 상기 공급부의 하부에 형성되는 형성부, 상기 형성부의 하부에 형성되며, 상기 공급부로부터 배양용 세포 또는 혈관 조성용 물질을 공급받는 배양부를 포함할 수 있으며, 상기 공급부를 통해 공급된 유체가 상기 형성부 내부에 응축되어 맺혀 상기 배양부의 하부로부터 이격된 상태로 유지될 수 있다.

인간의 질병 치료를 위해 수많은 약물이 개발되고 평가되고 있으며, 약물 개발을 위한 체외 인간 조직 및 장기 모사 모델 연구가 진행되고 있다. 모델 중 세포들을 모아서 3차원으로 형성하는 스페로이드(Spheroid) 및 오가노이드(Organoid)가 있는데, 이는 2차원으로 배양된 세포보다 복잡하고 다채로운 미세환경을 제공할 수 있다는 장점이 있다. 하지만 일반적인 스페로이드 및 오가노이드는 관류 가능한 3차원 혈관이 부재하여 산소 및 영양분 공급이 원활하게 되지 않고, 모델 내 체내 환경을 조성하기 어렵다는 문제점을 지니고 있다.
이러한 한계점을 극복하고자 미세 유체 칩 내에서 스페로이드 및 오가노이드와 혈관 네트워크를 공배양한 모델들이 개발 되었다. 그러나 선행 공배양 모델들의 경우 공통적으로 칩 내 스페로이드 및 오가노이드를 주입하는 과정이 상당히 복잡하고 어렵다는 단점이 존재한다.
구체적으로, 선행 공배양 모델들은 먼저 다른 플랫폼(U-shaped 96 well plate)에서 스페로이드 및 오가노이드를 미리 형성하여야 하며, 형성된 스페로이드 및 오가노이드를 웰 플레이트(well plate) 내에서 파이펫을 사용하여 추출한 후 칩 내에 주입하는 과정에서 상당한 숙련도를 필요로 한다. 또 실험자의 개별 능력에 따라 실험 결과에 차이가 생길 수 있어 재현성이 확보되지 않으며 편리성과 효율성이 낮은 실정이다.
본원 발명의 일 목적은 하나의 칩에서 3차원 혈관화된 스페로이드 또는 오가노이드 체외 모델을 제작할 수 있는 미세 유체 칩을 제공하기 위한 것이다.
본원 발명의 다른 일 목적은 행잉드랍(Hanging drop) 및 3D 바이오프린팅이 적용 가능한 미세 유체 칩을 제공하기 위한 것이다.
본원의 기술적 과제는 이에 제한되지 않으며 통상의 기술자가 이해할 수 있는 범위의 모든 과제를 포함하는 것으로 해석되어야 할 것이다.
본원 발명의 일 목적을 달성하기 위하여 본원 발명에 따른 스페로이드 또는 오가노이드 체외 모델을 3차원으로 모사하기 위한 미세 유체 칩은, 공급부, 상기 공급부의 하부에 형성되는 형성부, 상기 형성부의 하부에 형성되며, 상기 공급부로부터 배양용 세포 또는 혈관 조성용 물질을 공급받는 배양부를 포함할 수 있으며, 상기 공급부를 통해 공급된 유체가 상기 형성부 내부에 응축되어 맺혀 상기 배양부의 하부로부터 이격된 상태로 유지될 수 있다.
일 실시예에서, 공급부는 하부를 향할수록 좁아지는 테이퍼링 형상으로 형성될 수 있다.

본원 발명에 따른 미세 유체 칩은 파이펫 팁 규격에 맞는 주입구를 구비하여 다양한 3D 바이오프린팅 및 행잉드랍의 방법으로 스페로이드 또는 오가노이드 형성을 가능하게 한다. 이를 통해 기존에 외부에서 스페로이드 또는 오가노이드를 형성하고 혈관과의 공배양을 통해 미세 유체 칩으로 옮기는 과정이 통합되어 하나의 칩에서 간편하고 신속하게 수행될 수 있다.
형성된 스페로이드 또는 오가노이드와 함께 혈관 조성용 물질이 공배양되어 혈관화된 스페로이드 또는 오가노이드 모델을 확보할 수 있다. 이는 기존 모델에 비해 높은 체내 유사성을 확보한 체외 모델로서, 약물 개발을 위한 모델로 활용 가능하다.
또한 3D프린터 및 사출 성형을 통해 본 발명의 미세 유체 칩을 제작할 수 있어 높은 효율성을 확보할 수 있다.

본원 명세서 전체에서 사용되는 용어 '스페로이드(spheroid)'는 자연적인 자가 조립이나 스캐폴드의 존재 없이 외부 힘에 의한 세포 간 접합으로 형성된 세포 덩어리를 말한다. 일반적으로 스페로이드는 외부에서 ECM(세포외기질, extracellular matrix)의 역할을 하는 물질을 따로 넣어줄 필요 없이 스스로 ECM을 분비하여 자연적인 ECM의 조성을 유지한다.
스페로이드는 뇌 조직의 세포 구성의 다양성, 전기 생리학적 특성, ECM 분비 및 물성과 같은 기초적인 특성을 재현하기 위해 이용되어왔다. Dingle은 신생 랫트의 대뇌 피질 세포를 네 개의 다른 밀도(1K, 2K, 4K, 8K cells/spheroid)로 스페로이드를 만들었다. 14 일 후에 안정한 크기에 도달하여 생체 밀도와 유사하게 조립되었다(2 ~ 4 x 105 cells/mm3). 면역 조직 화학 분석 결과 뉴런뿐만 아니라 성상 세포, 희소 돌기 아교 세포 및 심지어 소교세포까지 포함하는 여러 개의 신경교세포가 그 안에 존재함이 확인되었다. 패치 클램프 실험으로 스페로이드 내의 뉴런이 전기적으로 활성을 드러내고, 연접후(postsynaptic) 전류(PSCs)의 발생을 검출해 시냅스 네트워크가 형성되었음을 보여주었다. 또한, 라미닌을 분비하고 신생 쥐 피질과 유사한 탄성 계수를 나타내었다 (Pei et al., 2017).
본원 명세서 전체에서 사용되는 용어 '오가노이드(organoid)'는 실제 장기의 세포 구성, 구조 및 기능을 닮은 큰 세포 집합체이다. PSCs와 성체줄기세포(ASCs)를 이용하여 오가노이드는 인간 조직의 기초적인 기능 중 일부를 나타낼 수 있다. 여러 가지 면에서 유기체는 진보된 스페로이드라고 할 수 있다. 왜냐하면 오가노이드에서 줄기 세포는 공간적으로 구분된 분화와 세포 정렬을 스스로 구성함에 반해 스페로이드 모델에서는 줄기세포 또는 선구 세포(progenitor cell)가 부족하여 자가 재생 및 분화가 미진하게 일어나기 때문이다. 결과적으로 오가노이드는 스페로이드보다 더 오랜 기간 동안 유지될 수 있는 보다 안정한 3D 세포 구조체를 만들어낸다.
일반적으로 오가노이드는 다양한 종류의 세포들의 내재적인 능력을 보존하고 발달과정에서의 세부적인 부분들을 재현하는 데에 다른 방법들보다 더 적합하다. 특히 실제 조직의 기본적인 구조와 생리학적 특징을 발견하는데 많은 성과가 있었다. 그럼에도 불구하고 이러한 자체 조립을 통해 만들어지는 모델에는 몇 가지 한계가 있다. 첫째, 크기와 모양이 매우 다양하다. 또한, PSCs가 감지되어 자라는 니치(niche, 미세주변환경)의 역할을 마트리겔(Matrigel)이 할 수 있다는 것이 밝혀지면서 신경 오가노이드를 형성할 때는 대부분 마트리겔이 이용되는데, 마트리겔의 조성이 완전히 분석되지 않았기 때문에, 마트리겔의 사용을 포함하는 많은 오가노이드들은 배치-투-배치 불일치(batch to batch inconsistency), 즉 생산된 마트리겔 조성의 불균질로 인한 문제를 피할 수 없으며, 이 때문에 오가노이드를 고효율 스크리닝(high throughput screening)에 이용하기 힘들다. 또한 일반적으로 오가노이드 모델은 스페로이드 모델보다 사이즈가 크다. 이 때문에 중심부에서 양분과 산소의 결핍으로 인한 세포의 괴사는 불가피하다. 따라서 기체 및 영양소 교환을 촉진하기 위해서는 모세 혈관 네트워크를 오가노이드 구조 안에 형성해야 할 필요가 있다 (Pei et al., 2017).
본원 명세서 전체에서 사용되는 용어 '행잉드랍(hanging drop)'은, 예를들어 10~50ul의 배양액을 배양 용기의 뚜껑에 배양 용기의 뚜껑에 매달아 고정하고, 중력으로 인해 물방울의 아래 부분에 모인 세포를 하나의 덩어리 형태(스페로이드)로 3차원 세포배양하는 모델을 의미한다.
본원 명세서 전체에서 사용되는 용어 '3D 바이오프린팅(3D bio printing)'은 3D 프린팅 기술과 생명공학이 융합된 개념을 의미한다. 이를 이용하여 살아있는 세포를 원하는 형상으로 쌓아 올려 조직, 장기를 제작할 수 있다. 기본적인 출력 방식은 일반 3D 프린팅과 동일하나, 3D 바이오프린팅은 살아있는 세포를 출력하기 위해 생체적합성을 갖는 고분자, 생체 재료 등을 기본 소재로 사용한다는 차이점이 있다. 3D 바이오프린팅 기법으로는 작은 크기의 방울로 재료를 분사하여 출력하는 잉크젯(Inkjet) 방식, 일정 점도 이상의 재료를 공압이나 피스톤으로 밀어내는 미세 압출(Micro-extrusion) 방식, 그리고 광 경화성 수지 표면에 광원을 조사하는 광 조형 방식이 있다. 잉크젯 방식은 출력 속도가 빠르고, 적은 비용이 장점이 될 수 있겠지만, 열응력에 의한 세포가 손상될 위험이 있다. 미세 압출 방식은 세포를 고밀도로 적층할 수 있어 다양한 형상 제작이 가능하다는 장점이 있어 가장 널리 사용되고 있지만, 프린팅 정밀도와 속도를 보다 향상시키는 것이 필요하다. 광 조형 방식은 노즐에서의 세포 막힘 현상이 없을 뿐만 아니라 정교한 프린팅이 가능하다는 장점을 가지고 있지만, 느린 출력 속도를 개선하고, 광 조형을 위한 소재와 세포간의 적합성을 향상시키는 것이 필요하다(김동립, 전예일. 2018).

공급부(200)는 하부를 향할수록 좁아지는 테이퍼링 형상으로 형성되어 공급부재를 통해 제1공급부(210)로 공급된 배양용 세포가 공급부(200)의 측면을 따라 흐르도록 할 수 있다. 구체적으로, 공급부재의 팁을 제1공급부(210)의 측면에 접하여 배양용 세포를 공급할 경우, 배양용 세포 내 공기의 유입을 최소화하는 효과를 얻을 수 있다.

도 3a를 참조하면 형성부(320)의 하부에 배양부(340)가 형성될 수 있다. 배양부(340)에는 상기한 스페로이드 및 오가노이드 중 어느 하나 이상과 공급된 혈관 조성용 세포가 위치하여 3차원 혈관화된 스페로이드 및 오가노이드 중 하나 이상이 형성될 수 있다. 또한 배양부(340)에는 3차원 혈관화 된 스페로이드 및 오가노이드 중 하나 이상이 형성되고, 그 이후의 실험을 위하여 형성부(320)보다 크게 형성될 수 있다.
도 4a를 참조하면 웰(100)은 공급부(200) 외에도 배양부(340)의 상부에 접하고 공급부(200) 및 형성부(320)와 구획되어 형성되며 배양부(340)에 배양액 또는 실험 물질을 공급할 수 있는 리저버(420)를 포함할 수 있다. 구체적으로 리저버(420)와 형성부(320) 및 공급부(200) 사이에는 격벽이 존재하며, 리저버(420)의 하부 일부는 배양부(340)의 상부 일부와 이어지도록 형성될 수 있다. 리저버(420)에는 3차원 혈관화된 스페로이드 및 오가노이드 중 어느 하나 이상이 형성된 미세 유체 칩(1000)을 이용하여 실험을 진행할 때 배양액 또는 실험 물질이 공급될 수 있다.
도 4a 및 4b를 참조하면 웰(100)은 주입부(440) 및 주입부(440)-배양부(340)를 연결하는 채널(460)을 포함할 수 있다. 주입부(440)는 배양부(340)의 가장자리에 위치할 혈관 내피세포를 공급할 수 있다. 공급된 혈관 내피세포는 채널(460)을 따라 배양부(340)로 이동할 수 있다. 구체적으로, 주입부(440)는 공급부(200), 형성부(320), 또는 리저버(420)와 연걸되지 않으며, 오직 채널(460)을 통해 배양부(340)와 연결된다. 이에 따라 채널(460)은 미세 유체 칩 내부에서 'L' 모양으로 형성될 수 있다.
주입부(440)에 공급된 혈관 내피 세포는 채널(460)을 통해 배양부(340)로 이동하여 배양부(340)에 형성된 3차원 혈관화된 스페로이드 및 오가노이드 중 어느 하나 이상의 가장자리에 위치할 수 있다. 이후 혈관 내피 세포는 혈관 내피 조직을 형성하며 3차원 혈관화된 스페로이드 및 오가노이드 중 어느 하나 이상의 내외로 물질 교환이 원활하게 이루어지도록 하거나, 배양부(340) 내에서 세포가 이동할 수 있는 통로 역할을 수행할 수 있다.
도 4c를 참조하면 배양부(340)의 하부에 베이스부(480)를 추가로 구비할 수 있다. 본원 발명의 미세 유체 칩(1000)은 베이스부(480)가 없는 상태로 제작 또는 판매될 수 있으나, 사용 시에는 베이스부(480)를 추가로 구비함이 바람직하다. 베이스부(480)로 사용할 수 있는 재료는 폴리카보네이트 필름(Polycarbonate film; PC flim) 또는 유리(Glass)가 있으나 이에 제한되는 것은 아니다.
실시예 1. 3차원 혈관화 된 스페로이드 또는 오가노이드 체외 모델 형성
도 5를 참조하여 실시예 1에 따른 체외 모델 형성과정을 설명한다.
아래는 체외 모델을 형성하는 하나의 과정에 불과하며, 본 발명의 이해를 돕기 위해 제시했을 뿐 본 발명이 아래 실시예로 제한되어 해석된다는 것을 의미하지는 않는다.
Step 1. 행잉드랍 및 3D 바이오프린팅(Day 0)
파이펫을 제1공급부(210)에 삽입해 배양용 세포를 제1공급부(210)의 측면에 공급한다. 제1공급부(210)에 형성된 단차(230)를 이용하면 더 편리하다. 공급된 배양용 세포는 공급부(200)의 측면을 따라 흘러 형성부(320) 내부에 응축되어 맺힌다. 맺힌 세포는 행잉드랍 및 3D 바이오프린팅을 활용하여 스페로이드 및 오가노이드 중 어느 하나 이상을 형성하도록 한다.
Step 2. 혈관 패터닝(Day 1)
파이펫을 제2공급부(220)에 삽입해 혈관 조성용 물질을 제2공급부(220)에 공급한다. 이때 파이펫의 팁을 형성부(320)의 계면(322)에 빈틈없이 맞닿게 한다. 이에 따라 혈관 조성용 물질은 형성부 내부에 맺히지 않으며, 형성부(320)을 지나 배양부(340)의 바닥면으로 이동하게 된다. 이와 동시에 이미 형성부(320)에 형성된 스페로이드 및 오가노이드 중 어느 하나 이상은 공급된 혈관 조성용 물질과 함께 배양부(340)로 이동한다.
Step 3. 3차원 혈관화(Day 7~)
배양부(340)에 3차원 혈관화 된 스페로이드 및 오가노이드 중 어느 하나 이상의 체외 모델이 형성된다. 추가적으로, 주입부(440) 및 채널(460)을 통해 모델 가장자리에 혈관 내피 세포를 공급하여 배양액과 모델 간의 물질 교환이 원활히 이루어질수록 있도록 할 수 있다. 모델이 완성된 후에는 리저버(420)를 통해 배양액과 실험 물질을 공급할 수 있다.
실시예 2. 행잉드랍 및 3D바이오프린팅 최적조건
본 발명의 일 실시예로서 행잉드랍의 최적조건을 설명한다.
도 6a를 참조하면, 공급부(200) 및 형성부(320)에 수용된 유체 물질의 체적을 V(μl), 형성부(320) 하부의 경계면의 지름을 D(mm), 경계면으로부터 배양부(340) 바닥면 사이의 거리를 H(mm), 형성부(320) 상부의 경계면과 테이퍼링 형상 사이의 각도를 α라고 한다.
이때, 세포가 형성부(320)의 계면(322)에 효과적으로 맺히기 위한 체적과 경계면 지름간의 조건은 도 6b와 같다. (아래에서부터 α = 30°, 45°, 60°, 64°, 75°, 90°)
도 6b를 참조하면, 각 α값에 대해 직선으로 표현된 일차 함수 이하의 영역에서 하기의 수학식 조건을 만족할 때 안정적으로 배양용 세포의 행잉드랍 및 3D 바이오프린팅이 가능한 것을 확인할 수 있었으며(pinning; 도 7a 및7b 참조), 그 외의 경우 배양용 세포 방울이 터지는 현상(bulging; 도 7a 참조)이 일어날 수 있다.
또한, 상기한 구현예에 대해, 형성부(320) 하부의 높이(H)와 형성부의 상부와 하부의 경계면 지름간의 조건은 다음과 같다. (도 6c 참조).
[수학식] H > = 0.35D (α값과 무관함)
별개로, D가 1mm 미만일 때 스페로이드 또는 오가노이드가 제대로 형성되지 않음을 확인한 바, D는 최소 1mm이상인 것이 바람직하다.
또한, 도 8은 스페로이드의 지름(μm)과 세포 수 간의 상관관계를 나타낸 도면이다.
실시예 3. 스페로이드 또는 오가노이드 체외 모델 제작
본원 발명의 미세 유체 칩(1000)의 일 구현예를 사용하여 일반 세포주 또는 암환자 세포의 스페로이드 또는 오가노이드 체외 모델을 제작하였다.
도 9는 체외 모델을 제작하는 과정을 나타낸 도면으로, 이를 참조하면 (1) 모델 내부에 세포 수가 많을수록(위), (2) 배양이 진행될수록(아래) 세포간의 응집이 잘 이루어진다는 것을 알 수 있다.
도 10은 일반 세포주(HCT116, U87MG, HEPG2, SW620)와 암환자 세포(환자#1, 환자#2)를 이용하여 스페로이드 또는 오가노이드를 패터닝한 사진이다. 구체적으로 암세포와 섬유아세포를 각각 10,000개씩 총합 20,000개의 세포를 12ul의 배양액에 희석한 후 미세 유체 칩(1000)에 주입하여 패터닝을 진행하였다.
특정 시간이 지나게 되면 특정한 지름을 가진 스페로이드 또는 오가노이드가 형성되는데, 실험 개시로부터 24시간 이후 200μm에서 300μm사이의 지름을 가진 스페로이드 또는 오가노이드가 형성된 것을 확인할 수 있었다.
또한 도 11은 본원 발명에 따른 미세 유체 칩(1000)을 활용하여 제작한, 3차원화 혈관화된 스페로이드 또는 오가노이드의 형광 현미경 사진을 나타낸다.

Claims (12)

  1. 미세 유체 칩에 있어서,
    공급부;
    상기 공급부의 하부에 형성되는 형성부; 및
    상기 형성부의 하부에 형성되며, 상기 공급부로부터 배양용 세포 또는 혈관 조성용 물질을 공급받는 배양부를 포함하고,
    상기 공급부를 통해 공급된 유체가 상기 형성부 내부에 응축되어 맺혀 상기 배양부의 하부로부터 이격된 상태로 유지되는 것인, 미세 유체 칩.
  2. 제1항에 있어서,
    상기 공급부는
    하부를 향할수록 좁아지는 테이퍼링 형상으로 형성된 것인,
    미세 유체 칩.
  3. 제1항에 있어서,
    상기 공급부는 상기 배양용 세포를 공급하기 위한 제1공급부; 및
    상기 혈관 조성용 물질을 공급하기 위한 제2공급부
    를 포함하는, 미세 유체 칩.
  4. 제3항에 있어서,
    상기 제1공급부는 상기 제2공급부의 상부에 형성되어 상기 배양용 세포가 공급부의 측면을 따라 공급되게 하고,
    상기 제2공급부는 상기 형성부 상부에 형성되어 상기 혈관 조성용 물질이 상기 형성부에 직접 공급되게 하는,
    미세 유체 칩.
  5. 제3항에 있어서,
    상기 제1공급부에는 단차가 형성되는 것인,
    미세 유체 칩.
  6. 제1항에 있어서,
    상기 형성부는 상부를 향할수록 좁아지는 테이퍼링 형상을 포함하는 것인,
    미세 유체 칩.
  7. 제1항에 있어서,
    상기 배양부는
    내부에 3차원 혈관화된 스페로이드 및 오가노이드 중 하나 이상이 형성 가능하도록 구성된 것인,
    미세 유체 칩.
  8. 제1항에 있어서,
    상기 배양부의 상부에 접하고,
    상기 공급부 및 상기 형성부와 구획되어 형성되며,
    상기 배양부에 배양액 또는 실험 물질을 공급할 수 있는 리저버를 추가로 포함하는,
    미세 유체 칩.
  9. 제1항에 있어서,
    상기 배양부의 가장자리에 혈관 내피세포를 공급할 수 있는 주입부; 및
    상기 주입부와 상기 배양부를 연결하는 채널을 추가로 포함하는,
    미세 유체 칩.
  10. 제1항에 있어서,
    상기 배양부의 하부에 형성된 베이스부를 추가로 포함하는,
    미세 유체 칩.
  11. 미세 유체 칩을 이용하여 스페로이드 또는 오가노이드 체외 모델을 형성하는 방법에 있어서,
    상기 미세 유체 칩의 공급부에 배양용 세포를 공급하는 단계;
    상기 공급된 배양용 세포가 상기 공급부와 형성부 사이에 응축되어 맺혀 스페로이드 또는 오가노이드를 형성하는 단계;
    상기 공급부에 혈관 조성용 물질을 공급하는 단계; 및
    배양부에 3차원 혈관화된 스페로이드 또는 오가노이드를 형성하는 단계
    를 포함하는,
    스페로이드 또는 오가노이드 체외 모델 형성 방법.
  12. 제11항에 있어서,
    상기 배양부에 배양액, 실험 물질, 또는 혈관 내피 세포를 공급하는 단계를 더 포함하는,
    스페로이드 또는 오가노이드 체외 모델 형성 방법.
