近年、トンネル状の分離流路に複数の柱状の障害物(ピラー)が配置されたピラーアレイ構造を有するカラムを固定相として備えた液体クロマトグラフィーが開発され、研究が進められている。粒子充填型カラム又はモノリス型カラムの場合、粒子のサイズ及び配置、並びに三次元網目構造を構成する網目のサイズ及び配置を精密に制御することが難しいという問題がある一方で、ピラーアレイカラムの場合、半導体微細加工等の技術を用いてピラーを製造し配置することができるため、設計図通りの構造を精密に作製することができ、高い分離効率を達成できるという利点がある。
光学異性体は幅広い技術分野で用いられており、特に医薬や農薬等の分野においては、用いられる物質に光学活性な化合物が多く、光学異性体に関する研究開発が活発に行われてきた。よって、液体クロマトグラフィー等のデバイスに用いられる分離カラムにおいて、複数の成分が混合された試料から光学異性体を分離できることは大きな利点となる。すなわち、光学異性体の分離を行うことができない上記の従来のデバイス、特にマイクロ流体デバイスには、改善の余地が残されていた。

本実施形態に係るマイクロ流体デバイスでは、ピラーの表面に光学活性ポリマーが担持されているため、トンネル状の分離流路に通液された流体から光学異性体を分離することができる。
本開示においては、マイクロ流体デバイスに備えられる「トンネル状の分離流路と、前記分離流路内に設けられた柱状の障害物と、前記障害物の表面に担持されたリガンドと、を備える要素」をピラーアレイカラムとも称し、以下、該ピラーアレイカラムについて説明する。
本開示においては、特段の断りがない限り、複数の対象が存在し得る数値条件は、その平均値を示すものとする。具体的には、例えば、ピラーの高さは、複数のピラーの高さの各々の数値を平均した数値とする。
以下、ピラーアレイカラム10の各構成要素について詳述する。

分離流路を構成する部材の材料、つまり、流路壁の材料は、特段制限されないが、半導体加工がしやすい観点から、シリコンであることが特に好ましい。
ピラーアレイカラム10は、柱状の障害物(ピラー)12を備える。ピラー12の形態は特段制限されず、ピラー12の底面の形状は、三角形状、四角形状(特には矩形状)、五角形状、もしくは六角形状等の多角形状、又は円柱形状等であってよいが、半導体加工がしやすい観点から、四角形状であることが好ましく、矩形状であることが特に好ましい。図1のピラーアレイカラム10におけるピラー12の底面の形状は、矩形状である。
ピラーの配置は特段制限されないが、ピークのブロード化やカラムの圧力上昇を防ぐ観点から、均一に配列されていることが好ましい。

光学活性ポリマーの種類は特段制限されず、多糖又はその誘導体、ポリ(メタ)アクリル酸アミド、ポリアミノ酸、ポリアミド、等が挙げられるが、分離性能が高い観点から、多糖又はその誘導体が好ましい。

エステル誘導体やカルバメート誘導体の具体例として、例えば、特公平4-42371号公報には、フェニルカルバメートの芳香族環の水素の一部がハロゲン(フッ素または塩素)で置換された置換基により、セルロースの水酸基が修飾されてなるセルロース誘導体や、特開2005-315668号公報に記載されている、フェニルカルバメートの芳香族環の水素の一部が、フッ素、アルキル基、またはアルコキシ基で置換された置換基により、セルロースまたはアミロースの水酸基が修飾されたセルロース誘導体及びアミロース誘導体についても本開示のリガンドとして使用可能である。
多糖誘導体については、上記のものに限られず、適宜所望の種類を用いることが可能である。
前記多糖又はその誘導体をピラーに担持させる方法は特段制限されず、物理的に吸着させて担持させてもよく、化学的に結合させて担持させてもよい。物理的に吸着させる方法としては、例えば、光学活性ポリマーをピラーにコーティングする方法が挙げられ、また、化学的に結合させる方法としては、ピラーと多糖誘導体の間での化学結合、第三成分を使用した化学結合、又は担体上の多糖誘導体への光照射、γ線などの放射線照射、もしくはマイクロ波などの電磁波照射などによって引き起こされる反応、もしくはラジカル開始剤などを用いるラジカル反応により化学結合を生じさせる方法が挙げられる。
本開示の別の実施形態であるマイクロ流体デバイスの製造方法(以下、単に「マイクロ流体デバイスの製造方法」とも称する。)は、トンネル状の分離流路と、前記分離流路内に設けられた柱状の障害物とを備える基材を準備する基材準備工程と、前記分離流路にリガンドを含む溶液を通液することで前記障害物の表面にリガンドを担持させる担持工程と、を有し、前記リガンドが光学活性ポリマーであることを特徴とする、マイクロ流体デバイスの製造方法である。上述のマイクロ流体デバイスの説明における用語や条件は、特段の断りがない限り、製造方法に係る本実施形態においても同様に適用することができる。
本開示では、ピラーにリガンドが担持されていないピラーアレイカラムを備える基材をリガンド未担持基材とも称する。
本実施形態に係るマイクロ流体デバイスの製造方法は、トンネル状の分離流路と、前記分離流路内に設けられた柱状の障害物とを備える基材(リガンド未担持基材)を準備する基材準備工程を有する。
上記基材を製造する方法は特段制限されず、公知の方法により、又は公知の方法を組み合わせて製造することができるが、設計図通りに製造することができる観点から、半導体加工技術を用いる工程、具体的には、半導体加工技術を用いて基材をパターニングする工程を有する方法が好ましい。該基材の形状は特段制限されないが、扱いやすさおよび製造容易性の観点から、チップ形状であることが好ましい。本開示において、チップ形状の基材(単に「チップ」と称する場合もある。)とは、一枚の板状の部材を意味する。
基材の態様はピラーアレイカラムを設けることができるものであれば特段制限されず、市販品を用いることができる。チップ形状の基材を用いる場合、例えば、シリコン等からなるバルク状の材料から切り出して形成してもよく、シリコン等の材料を含む組成物を硬化させて形成してもよく、市販品をそのまま用いてもよい。
具体的なリガンド未担持基材の製造方法の一例を、図3に沿って説明する。
上記の露光、現像の操作、パターン形成、掘り下げ、及び陽極接合等の処理は、公知の方法により行うことができる。
本実施形態に係るマイクロ流体デバイスの製造方法は、前記分離流路にリガンドを含む溶液を通液することで前記障害物(ピラー)の表面にリガンドを担持させる担持工程を有する。
流路にリガンドの溶液を通液する方法は特段制限されず、公知の方法により行うことができ、例えば、図1のF(i)からF(o)にリガンドを含む溶液を通液させることができる。通液する際の流量は、特段制限されず、例えば、0.1cm3/s以上、10.0cm3/s以下であってよく、1.0cm3/s以上、5.0cm3/s以下であってよい。
通液後、ピラーアレイカラムを自然乾燥、減圧乾燥又は加熱処理等により分離流路中のピラー上に存在する(塗布されている)リガンドの溶液を乾燥させることにより、ピラーにリガンドを担持させる(コーティングさせる)ことができる。加熱処理を実施する場合、加熱温度は特段制限されず、用いる溶液に応じて設定することができ、例えば、10℃以上、100℃以下であってよく、30℃以上、80℃以下であってよい。
ピラーへ光学活性ポリマー付着させるために通液させる溶液中のリガンド(特に光学活性ポリマー)の含有量は、本開示の効果が得られる範囲であれば特段制限されないが、光学異性体を安定して分離することができる観点から、通常0.01mmоl/L以上であり、0.1mmоl/L以上であることが好ましく、0.5mmоl/L以上であることがより好ましく、また、通常2.0mmоl/L以下であり、1.5mmоl/L以下であることが好ましく、1.0mmоl/L以下であることがより好ましい。
溶媒は、1種を単独で用いても、2種以上を任意の種類及び組み合わせで併用することができる。
本実施形態に係るマイクロ流体デバイスの製造方法は、上記の基材準備工程及び担持工程以外の工程(その他の工程)を有していてよく、例えば、リガンド(光学活性ポリマー)を合成する合成工程、リガンドを溶媒に溶解させてリガンドを含む溶液を製造する溶液準備工程等を有していてよい。
また、後述のマイクロ流体デバイスに備え得るミキサー等を備える場合、ミキサーの形成工程、誘導体化試薬を導入するためのインレットの形成工程、及び誘導体化試薬流路の形成工程も備えることができる。
なお、本開示において、マイクロ流体デバイスとは、μmオーダーの流路や構造を有するデバイスを意味し、デバイス全体のサイズがμmオーダーである必要はない。
マイクロ流体デバイスに試料用流路を形成させる方法は特段制限されないが、設計図通りに製造することができる観点から、上述した基材に分離流路を形成させるための半導体加工技術を用いる方法、具体的には、半導体加工技術を用いて基板をパターニングする工程を有する方法を適用することが好ましい。
なお、図4における分離流路32の前後の移動相及び試料用流路の態様は、特段制限されず、移動相及び試料を通液できるものであればよく、例えば、分離流路からピラーを除いたトンネル状の空洞であってよい。また、移動相のみが通液される移動相流路(移動相と試料とが混合される前の移動相の通液流路)についても、移動相を通液出来る態様であれば特段制限されない。
図4において、マイクロ流体デバイスは、移動相インレット34を備え、水、有機溶媒又は緩衝液等の移動相が装置に導入され、また、試料インレット35を備え、試料が装置に導入される。試料インレット35から導入された試料は、試料注入部36で移動相インレット34から導入された移動相と混合され、移動相と共に分離流路32に導入される。移動相と共に導入された試料に含まれる各成分は、分離流路32で流れ方向に分離される。なお、分離流路32に導入された試料及び移動相は、アウトレット37から排出される。この場合、試料及び移動相は、図1に示されるF(i)に沿って分離流路に流入する。また、試料インレット35から試料処理装置に導入され、試料注入部36で移動相に混合されなかった試料は、試料アウトレット38から排出される。なお、試料インレット35から試料アウトレット38までは、チップ33に形成した溝である試料流路31で接続されている。なお、マイクロ流体デバイス30を構成する分離流路32、移動相インレット34、試料インレット35、及びアウトレット37等は、シリコン等の基板上に溝として形成され、必要に応じてミキサー等も形成され、溝の上部(開口部分)を図4に示すようにガラスで覆った構造とすることができる。
基材のピラーアレイカラムが備わる側の面の面積は、特段制限されないが、扱いやすさの観点から、通常100mm2以上であり、400mm2以上であることが好ましく、また、コスト抑制の観点から、通常10,000mm2以下であり、5,000mm2以下であることが好ましい。
基材の厚さは、特段制限されないが、扱いやすさの観点から、通常100μm以上であり、150μm以上であることが好ましく、また、コスト抑制の観点から、通常1,000μm以下であり、500μm以下であることが好ましい。
また、基材のピラーアレイカラムが備わる側の面の形状が矩形状である場合、各辺の長さは、特段制限されないが、扱いやすさの観点から、通常10mm以上であり、20mm以上であることが好ましく、また、コスト抑制の観点から、通常100mm以下であり、70mm以下であることが好ましい。
なお、図5及び6に示す分離流路のターン構造は、平面視した場合における内側壁面の形状が外周側拡開状のテーパー状に形成された構造となっているが、この形状に限定されず、任意のターン構造であってよい。
[リガンド未担持基材の製造]
マイクロ流体デバイスとして、図6に示される構成(ただし、分離流路のターン構造の数は12とした。)において、厚さ500μm、1辺20mmの正方形マイクロチップ上に、試料流路31として深さ60μmの溝を形成し、移動相及び試料が流れるその他の流路を深さ30μmの溝として形成した。また、分離流路32は、幅400μmとした(ターン構造の曲線部分の最も細い部分の幅は110μm)。
また、分離流路の幅(流れ方向に直交する方向の長さ)が400μm、流れ方向の長さが110mmであり、分離流路32におけるピラーは、辺の長さが3μm、面同士の間隔が2μmで整列した構造とした。
以上のマイクロ流体デバイスは、シリコン基板に従来公知のフォトリソグラフィ法及び深掘り反応性イオンエッチングによって各構造を形成した後、従来公知の陽極接合法によってガラスと貼り合わせることにより、リガンド未担持基材を製造した。
セルロース誘導体(Cellulose tris(3,5-dimethylphenylcarbamate))500mgをアセトン1Lに溶解させた。続いて、この溶液をシリンジポンプ(YSP-101「標準タイプ」、株式会社ワイエムシィ製)を使用し、室温(25℃)、流速2μL/minで60分間、上記のリガンド未担持基材へ通液させた。その後、チップを温度60℃で6時間真空乾燥(減圧度<10mmHg)(角形真空定温乾燥器DP300、ヤマト科学株式会社製)させてリガンドの担持を行い、マイクロ流体デバイスを製造した。
上記マイクロ流体デバイスについて、移動相インレット34からは水/アセトニトリル(富士フイルム和光純薬株式会社製)=40/60(v/v)を、試薬インレット35からは下記の式(1)で表される4-ブロモメチル-7-メトキシクマリン(Br-Mmc)で蛍光誘導体化したフルルビプロフェン(富士フイルム和光純薬株式会社製)を、流速0.5μL/min、試料濃度100μM、検出位置near Outlet(カラム長110mm付近)で送液した。
試料の分離中は、蛍光顕微鏡を用いて固定視野にて動画を撮影した。この動画から、流路上の一部分の蛍光強度の時間変化をAndor SOLIS(ver. 4.28.30001.0: Andor Technologies, South Windsor, CT, USA)を用いてプロットすることでクロマトグラムを得た。また送液時の背圧はマイクロポンプのモニターに表示される圧力値に従い記録した。評価結果を図7に示す。
[リガンド未担持基材の製造]
実施例1と同様の方法でリガンド未担持基材を製造した。
セルロース誘導体(Cellulose tris(3,5-dimethylphenylcarbamate))の量を500mgから10mgに変更したこと以外は実施例1と同様の方法でリガンドの担持を行い、マイクロ流体デバイスを製造した。
実施例1と同様の方法で分離評価を行った。評価結果を図8に示す。
[リガンド未担持基材の製造]
実施例1と同様の方法でリガンド未担持基材を製造した。
セルロース誘導体(Cellulose tris(3,5-dimethylphenylcarbamate))の量を500mgから100mgに変更したこと以外は実施例1と同様の方法でリガンドの担持を行い、マイクロ流体デバイスを製造した。
移動相インレット34からは水/アセトニトリル(富士フイルム和光純薬製)の比率を40/60(v/v)から50/50(v/v)に変更したこと以外は実施例1と同様の方法で分離評価を行った。評価結果を図9に示す。

Claims (11)
- トンネル状の分離流路と、前記分離流路内に設けられた柱状の障害物と、
前記障害物の表面に担持されたリガンドと、を備え、
前記リガンドが光学活性ポリマーであることを特徴とする、マイクロ流体デバイス。 - 前記光学活性ポリマーが、多糖又はその誘導体であることを特徴とする、請求項1に記載のマイクロ流体デバイス。
- 前記分離流路がターン構造を有し、該ターン構造が、平面視した場合における内側壁面の形状が外周側拡開状のテーパー状に形成された構造である、請求項1又は2に記載のマイクロ流体デバイス。
- 前記ターン構造において、平面視した場合における外周の長さと内周の長さが同一である、請求項3に記載のマイクロ流体デバイス。
- 前記分離流路がターン構造を有し、該ターン構造が、ピラーの個数の密度が内周側から外周側にかけて小さくなるような勾配を有する構造である、請求項1又は2に記載のマイクロ流体デバイス。
- 試料を導入するための試料用流路をさらに備える、請求項1~5のいずれか1項に記載のマイクロ流体デバイス。
- 前記分離流路と前記試料用流路が、同一の基材上に備えられている、請求項6に記載のマイクロ流体デバイス。
- 前記基材の形状がチップ形状である、請求項7に記載のマイクロ流体デバイス。
- トンネル状の分離流路と、前記分離流路内に設けられた柱状の障害物とを備える基材を準備する基材準備工程と、
前記分離流路にリガンドを含む溶液を通液することで前記障害物の表面にリガンドを担持させる担持工程と、
を有し、前記リガンドが光学活性ポリマーであることを特徴とする、マイクロ流体デバイスの製造方法。 - 前記基材の形状がチップ形状である、請求項9に記載のマイクロ流体デバイスの製造方法。
