Abstract
La présente invention a pour objet un produit de combinaison comprenant à titre de substances actives, au moins de la vitamine A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng, pour aider à retrouver la forme.
Description
La présente invention a pour objet un produit de combinaison comprenant à titre de substances actives, au moins de la vitamine A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng, pour aider à retrouver la forme.
Certaines périodes, comme la croissance, les phases de convalescence, le froid sollicitent et fatiguent l’organisme. Parfois, une alimentation déséquilibrée ou un manque d’appétit ne couvrent pas à 100% les besoins journaliers. Les enfants ou les adultes peuvent alors avoir besoin d’un apport en vitamines et oligo-éléments pour retrouver forme et vitalité.
Afin de palier au déficit en certaines vitamines et minéraux, il existe différents produits sur le marché. Parmi les produits commercialisés, on peut citer à titre d’exemple les produits Vitascorbol Multi commercialisés par la Cooper ou Azinc Forme Vitalité ou encore Azinc Energie Max commercialisés par les Laboratoires Arkopharma. Ces produits bien que comprenant une association de vitamines et minéraux ont des quantités de vitamines du groupe B, de vitamine C et de ginseng très inférieures à la composition selon l’invention. Ainsi, la Demanderesse a mis au point, et c’est l’objet de la présente invention, une formulation plus complète permettant une action revitalisante plus prononcée.
Aucun des produits proposés ne contient l’association objet de la présente invention.
La Demanderesse a donc souhaité développer un produit qui puisse être utilisé par une population large, qui ne contient pas de produits d’origine animale, pas de conservateurs et sans lactose.
La Demanderesse a mis au point, et c’est l’objet de la présente invention, un produit de combinaison comprenant à titre de substances actives, au moins vitamine A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng, pour aider à retrouver la forme.
Par « aider à retrouver la forme », on entend dans le cadre de la présente invention, aider à retrouver l’énergie, ne plus avoir de sensation de fatigue.
Un tel produit permet donc de pallier un déficit d’apport en vitamines et minéraux avec une composition qui ne contient ni produit d’origine animale ni lactose ni conservateurs.
Le produit de la présente invention présente une galénique adaptée à l’adulte.
De manière avantageuse, la composition selon l’invention est associée à une composition comprenant un complexe de desmodium, pavot de Californie, magnésium et vitamine B6, qui en stimulant le fonctionnement des cellules hépatiques et facilitant l’endormissement permettant d’optimiser l’action desdites vitamines et/ou minéraux sur l’organisme.
La présente invention se rapporte à un produit de combinaison, comprenant à titre de substances actives, au moins de la vitamine A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng pour aider à retrouver la forme.
Dans certains modes de réalisation, le produit de combinaison selon l'invention comprend, en plus des vitamines A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng, à titre de substances actives :
- Des plantes ou extraits de plantes
- Des vitamines et minéraux, tels que par exemple les vitamines K, le calcium ou l’iode
Dans certains modes de réalisation, le produit de combinaison de l'invention est caractérisé en ce que les vitamines A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, le zinc, le fer, le molybdène, le sélénium, le manganèse, le chrome, le cuivre et le ginseng sont présents dans une composition unique comprenant :

Selon un mode de réalisation particulier de l'invention, le produit de combinaison est caractérisé en ce que les ingrédients actifs, c’est-à-dire les vitamines A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, le zinc, le fer, le molybdène, le sélénium, le manganèse, le chrome, le cuivre et le ginseng sont dans un milieu adapté à l'administration par voie orale. De préférence, le milieu adapté est un milieu solide. De manière préférentielle, le produit de combinaison se présente sous la forme de comprimé.
Dans la présente invention, la demanderesse a mis en évidence que l'association des vitamines A, B1, B2, B3, B5, B8, B9, B12, C, D3, E, du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng présentait un effet synergique pour aider à retrouver la forme.
La vitamine A
La vitamine A est une vitamine liposoluble.
Dans l'organisme, elle existe sous forme de rétinol, de rétinal, d'acide rétinoïque (trétinoïne) et de phosphate de rétinyle. Ces molécules sont altérées par l'oxygène de l'air, altérations accélérées par la lumière et la chaleur.
Les besoins quotidiens en vitamine A pour un adulte sont estimés à 2 400 UI (femme, femme enceinte exceptée) ou à 3 400 UI (homme) soit de 0,7 à 1 mg environ.
Une unité internationale de vitamine A correspond à 0,3 μg de rétinol. Le rétinol est considéré comme l'unité de base, le rétinol-équivalent, ce qui permet de comparer l'activité vitaminique des différents dérivés de la vitamine A.
La vitamine A est impliquée dans la croissance des os et la synthèse de pigments de l'œil. Le rétinol est parfois utilisé dans le traitement des acnés sévères.
Un sévère déficit en vitamine A peut provoquer une cécité ; il se manifeste par une peau pâle et sèche. Si elle est prolongée, la carence en vitamine A peut être mortelle, tout comme une surdose.
La vitamine A est une vitamine importante pour la vision.
L'absorption se fait par l'intestin et nécessite la présence de graisses et de sels biliaires. La thyroxine la favorise.
Le stockage s'effectue presque uniquement dans les cellules stellaires (anciennement cellules de Ito) du foie, beaucoup plus accessoirement dans les capsules surrénales, les testicules, les ovaires et les reins.
La carence d'apport est, le plus souvent, responsable de l'hypovitaminose A, en particulier chez le nourrisson soumis à un régime sans lait. Les adultes peuvent également souffrir d'un apport insuffisant lorsqu'ils se soumettent à des régimes stricts, éliminant de leur diététique : laitages, œufs, graisses, légumes verts ou autres légumes (carottes, pommes de terre douces, poivrons, citrouille...) et de fruits (melons cantaloups, mangues et abricots).
À côté de cette carence d'apport, il existe également une carence d'utilisation, car le métabolisme de la vitamine A exige l'intégrité des fonctions hépatiques et intestinales. De plus, les troubles du transit intestinal inhibent la transformation du bêta-carotène en vitamine A.
Les premiers signes de la carence sont la photophobie et la cécité crépusculaire. Il s'ajoute rapidement des troubles divers portant sur la croissance, entraînant la dédifférenciation des épithéliums avec diminution de la résistance aux infections et affectant certaines glandes endocrines et les divers métabolismes.
L'administration de la vitamine A est indiquée non seulement au cours des symptômes de carence mais aussi contre certains troubles métaboliques et en cas de prédisposition aux infections.
La vitamine A est également utile à titre prophylactique lors de circonstances physiologiques qui en accroissent les besoins (grossesse car risque d'effet tératogène, allaitement, syndrome prémenstruel) mais aussi dans d'autres circonstances telles que : retard de croissance, diminution de la résistance aux infections, photophobie, héméralopie, xérophtalmie, kératomalacie, ichtyose, kératose pilaire, hyperkératose, acné, cheveux secs, ternes et cassants. Cependant, elle possède des mises en garde, par exemple lors de grossesses, du fait de malformations de l'enfant (effet tératogène) constatées sur des femmes ayant pris trop de vitamine A.
La supplémentation systématique chez l'enfant, dans les pays où le déficit est endémique, montre une réduction de la mortalité. En revanche, elle semble être inefficace, et peut-être même délétère chez les filles, lorsque la vitamine A est administrée chez le nouveau-né.
Des recherches menées conjointement par des chercheurs chinois (Université de Chongqing) et canadiens (Université de la Colombie-Britannique) ont montré, sur l'animal, qu’en cas de carence en vitamine A, les rongeurs sont davantage affectés par la formation de plaques amyloïdes, caractéristiques de la maladie d’Alzheimer, et par des troubles cognitifs, concernant en particulier la mémoire.
Selon un mode de réalisation préférée de la présente invention, la vitamine A est présente dans la composition selon l’invention en une teneur de 200 à 2000 µg de préférence 400 à 1500 µg par composition unique.
La vitamine B1
La thiamine ou vitamine B1 (ou encore aneurine) est un précurseur métabolique de la thiamine pyrophosphate (TPP), une coenzyme essentielle à certaines décarboxylases. Chez les animaux, la thiamine est une vitamine hydrosoluble de la famille des vitamines B qu'ils doivent trouver dans leur alimentation. Elle est en revanche synthétisée par les bactéries, les plantes et les champignons. Elle est indispensable à la transformation des glucides en énergie par le cycle de Krebs et est nécessaire au bon fonctionnement du système nerveux et des muscles. Elle est en effet indispensable à la transformation du pyruvate produit par la glycolyse et toxique pour le système nerveux.
Chez l'homme, une carence alimentaire en vitamine B1 cause le béribéri et peut aussi causer une encéphalopathie de Gayet-Wernicke.
Chez l'homme, les besoins estimés sont de 1,3 mg à 1,5 mg par jour. Chez l'adulte en bonne santé, l'absorption digestive est de 4,5 % de la dose ingérée, et baisse à 1,5 % en cas d'alcoolo-dépendance.
Selon un mode de réalisation préférée de la présente invention, la vitamine B1 est présente dans la composition selon l’invention en une teneur de 1,5 à 20 mg de préférence 2 à 10 mg par composition unique.
La vitamine B2
La vitamine B2, correspondant à la riboflavine, ou lactoflavine, est une vitamine hydrosoluble nécessaire à la synthèse de la flavine adénine dinucléotide (FAD) et de la flavine mononucléotide (FMN), deux cofacteurs essentiels aux flavoprotéines.
La vitamine B2 joue un rôle important dans la transformation des aliments simples (glucides, lipides et protéines) en énergie. Elle intervient dans le métabolisme de réparation des muscles.
La riboflavine se trouve naturellement dans les asperges, le maïs soufflé, les bananes, les kakis, le gombo, les blettes à carde, le fromage, le lait, le yaourt, la viande, les œufs, le poisson et les haricots verts, dont chacun contient au moins 0,1 mg de vitamine B par 85-300g. D'autres sources sont les légumes verts feuillus, le foie, les reins, les légumineuses, les tomates, les levures, les champignons et les amandes.
Elle est également utilisée en tant que traitement de fond de la migraine.
Selon un mode de réalisation préférée de la présente invention, la vitamine B2 est présente dans la composition selon l’invention en une teneur de 2 à 15 mg, de préférence 3 à 8 mg par composition unique.
La vitamine B3
La vitamine B3 est une vitamine hydrosoluble qui correspond à deux molécules : la niacine (acide nicotinique) et son amide, la nicotinamide, parfois appelée niacinamide. Elle est aussi appelée vitamine PP pour pellagra preventive car une carence en cette vitamine est responsable de la pellagre.
La vitamine B3, précurseur du NAD+ (nicotinamide adénine dinucléotide) et du NADP+ (nicotinamide adénine dinucléotide phosphate), est nécessaire comme cofacteur d'oxydoréduction au métabolisme des glucides, des lipides et des protéines.
La vitamine B3 se trouve principalement dans les volailles, le foie de veau et les céréales.
L'acide nicotinique est appelé vitamine B3 parce qu'elle est la troisième vitamine B à avoir été découverte. Pour éviter de suggérer que la nicotine -ou la cigarette- contient une vitamine et pour dissocier de la nicotine cette vitamine et les additifs alimentaires en contenant, le nom plus abstrait de niacine a été forgé, mot-valise de nicotinic acid vitamin.
Les vitamines B facilitent la conversion de la nourriture (glucides) en énergie (glucose). La niacine est utile dans le processus de régulation des hormones liées au stress et améliore la circulation sanguine. Ces vitamines sont hydrosolubles et le corps ne les stocke pas.
Les symptômes d'une carence légère sont l'indigestion, la fatigue, le vomissement, ou la dépression. Elle peut être due à une grossesse.
La niacine – mais pas la nicotinamide – baisse la concentration de cholestérol dans le sang en diminuant légèrement chez le patient ayant déjà fait un infarctus du myocarde le risque de récidive ainsi que la mortalité. Ces données datent cependant d'avant l'utilisation des statines.
La niacine prévient la progression de l'athérosclérose et minimise le risque d'attaque cardiaque.
Selon un mode de réalisation préférée de la présente invention, la vitamine B3 est présente dans la composition selon l’invention en une teneur de 1 à 40 mg, de préférence 10 à 30 mg par composition unique.
La vitamine B5
La vitamine B5, correspondant à l’acide pantothénique, est une vitamine hydrosoluble. C'est un précurseur métabolique de la coenzyme A, essentielle à la synthèse et au métabolisme des protéines, des glucides et des lipides.
La racine ethnique provient de p a ntothen qui, en grec, signifie « partout » : cette vitamine se trouve en effet dans presque tous les aliments. Cependant, on la trouve en proportions particulièrement concentrées dans les germes et céréales complètes et dans les variétés d'ortie piquante. La gelée royale est l'un des produits naturels connu le plus riche qui soit en vitamine B5.
L'acide pantothénique est constitué d'une molécule d'acide pantoïque et d'une molécule de β-alanine, liées par une fonction amide.
Précurseur et constituant du coenzyme A, la vitamine B5 favorise la croissance et la résistance de la peau et des muqueuses. Elle est nécessaire au métabolisme des glucides, lipides et protéines et participe à la synthèse de certaines hormones. L'acide pantothénique est détruit par la chaleur en solution aqueuse.
Les besoins du corps humain sont estimés à 5 mg par jour à partir de 16 ans, à 2 mg chez les nourrissons et à 7 mg pour les allaitantes mais généralement seule une sous-alimentation peut entraîner une réelle carence.
Selon un mode de réalisation préférée de la présente invention, la vitamine B5 est présente dans la composition selon l’invention en une teneur de 8 à 40 mg de préférence 10 à 30 mg par composition unique.
La vitamine B8 ou biotine
La vitamine B8, correspondant à la biotine, est une vitamine hydrosoluble encore souvent appelée coenzyme R, vitamine H, ou encore vitamine B7 dans de nombreux pays, notamment en Allemagne ou dans les pays anglo-saxons (chez ces derniers, la vitamine B8 est une appellation erronée pour l'AMP, tandis qu'en France la vitamine B7 désigne improprement l'inositol, ces deux composés n'ayant aucun rapport entre eux ni avec la biotine).
La biotine est une coenzyme qui participe au métabolisme des acides gras, des glucides et des acides aminés, ainsi qu'à la biosynthèse des vitamines B9 et B12. On la retrouve donc, d'un point de vue biologique, dans toutes les espèces vivantes. Au plan diététique, on la trouve notamment dans les céréales complètes, le foie, les œufs et le lait, mais aussi le soja, les noisettes, les levures et en moindres quantités dans les autres aliments.
La biotine est composée d'un noyau de 2-imidazolidinone fusionné avec un noyau thiolane auquel est rattaché un groupe acide pentanoïque. Bien qu'existant sous 8 formes différentes, une seule, la D-Biotine est présente naturellement et a une pleine activité vitaminique. De nombreux analogues sont utilisés en chimie et biochimie, comme la 2-Iminobiotine (affinité pour l'avidine moindre et dépendante du pH) ou les biotines pégylées.
La biotine est nécessaire à la croissance cellulaire, à la production d'acides gras, et au métabolisme des graisses et des acides aminés. Elle joue un rôle dans le cycle de Krebs qui est le processus biochimique par lequel l'énergie est produite au cours de la respiration aérobie. La biotine participe non seulement à diverses réactions métaboliques, mais elle aide aussi au transfert du dioxyde de carbone. La biotine est également utile dans le maintien d'un niveau constant de sucre dans le sang (glycémie). La biotine est souvent recommandée pour renforcer les cheveux et les ongles. En conséquence, c'est un composant de nombreux cosmétiques et produits de santé pour les cheveux et la peau.
La déficience en biotine est extrêmement rare puisque les bactéries intestinales en produisent généralement plus que les besoins quotidiens de l'organisme.
La biotine est largement présente dans une variété d'aliments, mais le plus souvent à de faibles concentrations. Il y a seulement deux types d'aliments qui contiennent de grandes quantités de biotine, incluant la gelée royale et la levure de bière. Les meilleures sources naturelles de biotine dans l'alimentation humaine sont le foie, les légumineuses, le soja, la bette carde, les tomates, la laitue romaine, et les carottes. La liste des aliments comprenant de la biotine comprend les amandes, les œufs, les oignons, le chou, le concombre, le chou-fleur, le lait de chèvre, le lait de vache, les framboises, les fraises, le flétan, l'avoine et les noix. Les plus importantes sources naturelles dans l'alimentation des animaux non ruminants sont les oléagineux, la luzerne et les levures sèches. La quantité de biotine contenue dans la nourriture varie et peut être influencée par des facteurs tels que les variétés végétales, la saison et le rendement.
Selon un mode de réalisation préférée de la présente invention, la vitamine B8 est présente dans la composition selon l’invention en une teneur de 50 à 500 µg de préférence 100 à 300 µg par composition unique.
La vitamine B9
La vitamine B9, autre nom de l'acide folique (folate, folacine ou vitamine M, acide pteroyl-L-glutamique, pteroyl-L-glutamate et acide pteroylmonoglutamique), est une vitamine hydrosoluble.
L'acide folique est le précurseur métabolique d'une coenzyme, le tétrahydrofolate (FH4 ou THF4), impliquée notamment dans la synthèse des bases nucléiques, purines et pyrimidines, constituant les acides nucléiques (ADN et ARN) du matériel génétique. Le THF intervient également dans la synthèse d'acides aminés tels que la méthionine, l'histidine et la sérine.
Chez l'humain, les besoins en acide folique et substances apparentées sont de l'ordre de 300 μg par jour.
C'est l'une des carences vitaminiques les plus communes, surtout dans les pays et milieux défavorisés.
Une carence peut survenir dans les situations suivantes :
- apports insuffisants en aliments riches en folates (légumes verts, fruits et abats) ou besoins accrus (femmes enceintes ou allaitantes) ;
- pathologies caractérisées par un renouvellement cellulaire important (anémies, cancers...) ou par une malabsorption intestinale (maladies digestives, hépatiques...) ;
- prise de médicaments anti-folates (méthotrexate, sulfasalazine, anti-épileptique...).
- beaucoup plus rarement : troubles génétiques du métabolisme des folates.
Une carence en folate entraîne principalement des troubles hématologiques, voisins de ceux d'une carence en vitamine B12, et des troubles neurologiques progressifs. Ceux-ci débutent par une anxiété ou un syndrome dépressif jusqu'à la démence ou à la psychose.
Un déficit en folates entraîne essentiellement une anémie qui se caractérise par une taille augmentée des hématies (anémie macrocytaire) et l’apparition de mégaloblastes dans la moelle osseuse. Le terme anémie mégaloblastique est habituellement restreint aux carences en vitamine B12 ou en folates, mais toute perturbation de la synthèse des purines, pyrimidines et protéines peut provoquer une anémie mégaloblastique.
Dans le traitement des anémies mégaloblastiques, l'acide folique et la vitamine B12 ont tous deux leur place. Mais l'acide folique, à lui seul, ne peut ni prévenir, ni guérir les complications neurologiques (syndrome neuro-anémique).
Il est admis d'autre part que la vitamine B12 catalyse la synthèse de la thymidine, substance importante pour le métabolisme des nucléotides. La thymidine est formée à partir de la thymine, dont la synthèse est catalysée par l'acide folique. C'est pour cette raison que l'acide folique est combiné à la vitamine B12 pour le traitement des anémies normochromes macrocytaires de type mégaloblastique.
Un taux d’acide folique bas serait lié à une augmentation du risque de dépression de 55 %. L'association d'acide folique à un antidépresseur pourrait être utile.
L'effet inhibiteur sur les déficits cognitifs de la personne âgée est beaucoup plus controversé. Il semble cependant qu'une supplémentation artificielle en acide folique pendant une longue durée sur une population âgée dont la nutrition est carencée en cette vitamine pourrait sensiblement en améliorer l'état cognitif. Les folates permettraient également de diminuer les problèmes d'audition chez la personne âgée.
Les folates sont nécessaires à la division et au maintien cellulaire. Un apport est donc crucial pendant les périodes de croissance accélérée, comme l'enfance et la grossesse. La supplémentation en folates durant le premier trimestre de la grossesse diminuerait significativement le risque de certaines malformations. Les recommandations actuelles sont une supplémentation de 0,4 mg en folates avant la conception (au moins un mois avant et deux mois après) pour la prévention des anomalies de fermeture du tube neural. Le dosage peut être réévalué à la hausse (5 mg) pour les femmes ayant des antécédents d'AFTN (fente labiale, anomalies de fermeture du tube neural).
Cette supplémentation pourrait également diminuer, de manière faible, le risque de survenue d'une malformation cardiaque grave ainsi que celui de la survenue de troubles du spectre autistique.

La vitamine B12
La vitamine B12, également appelée cobalamine, est une vitamine hydrosoluble essentielle au fonctionnement normal du cerveau (elle participe à la synthèse des neurotransmetteurs), du système nerveux (elle est indispensable au maintien de l'intégrité du système nerveux et tout particulièrement de la gaine de myéline qui protège les nerfs et optimise leur fonctionnement) et à la formation du sang. C'est l'une des huit vitamines B. Elle est normalement impliquée comme cofacteur dans le métabolisme de chacune des cellules du corps humain, plus particulièrement dans la synthèse de l'ADN et sa régulation ainsi que dans la synthèse des acides gras et dans la production d'énergie.
Elle existe sous plusieurs formes appartenant à la famille des cobalamines : cyanocobalamine, hydroxocobalamine, méthylcobalamine et adénosylcobalamine, les deux premières étant ses formes stables. Les cobalamines ont une structure chimique proche de l’hème mais l’atome central de fer y est remplacé par un atome de cobalt, d’où leur nom.
Un déficit en vitamine B12 entraîne une forme d'anémie dont l'une des caractéristiques est la présence de globules rouges fortement augmentés en taille (macrocytose). Relativement rare, cette carence peut notamment résulter d'apports insuffisants chez des personnes suivant un régime végétalien strict non complémenté, ou de problèmes d'absorption.
Chez les humains, la vitamine B12 provient exclusivement des aliments d'origine animale, de compléments alimentaires ou d'aliments enrichis en vitamine B12. Ces compléments et aliments enrichis sont la seule source significative de vitamine B12 pour les végétaliens.
Selon un mode de réalisation préférée de la présente invention, la vitamine B12 est présente dans la composition selon l’invention en une teneur de 1,1 à 10 µg de préférence 2 à 5 µg par composition unique.
La vitamine C
La vitamine C est une vitamine hydrosoluble sensible à la chaleur et à la lumière jouant un rôle important dans le métabolisme de l'être humain et de nombreux autres mammifères. Chimiquement parlant, il s'agit de l'acide L-ascorbique, un des stéréoisomères de l'acide ascorbique, et de ses sels, les ascorbates. Les plus courants sont l'ascorbate de sodium et l'ascorbate de calcium.
La vitamine C est un cofacteur enzymatique impliqué dans un certain nombre de réactions physiologiques (hydroxylation). Elle est requise dans la synthèse du collagène et des globules rouges et contribue au système immunitaire. Elle joue également un rôle dans le métabolisme du fer en tant que promoteur de son absorption; son utilisation est donc déconseillée chez les patients porteurs d'une surcharge en fer et particulièrement d'une hémochromatose. Sous forme oxydée (acide déshydroascorbique), elle traverse la barrière hémato-encéphalique pour accéder au cerveau et à plusieurs organes à forte concentration en vitamine C.
Selon un mode de réalisation préférée de la présente invention, la vitamine C est présente dans la composition selon l’invention en une teneur de 130 à 400 mg de préférence 150 à 300 mg par composition unique.
La vitamine D3
Le cholécalciférol est une forme de vitamine D, également appelée vitamine D3. La vitamine D3 est un sécostéroïde plutôt parent de la testostérone, du cholestérol et du cortisol.
La vitamine D possède une double origine : elle est apportée par l'alimentation et synthétisée par l'organisme au niveau de la peau sous l'action des rayons UVB du soleil.
Les sujets s'exposant très peu au soleil peuvent présenter des carences plus ou moins importantes en vitamine D. Les personnes à peau claire synthétisent la vitamine D plus vite que celles à peau foncée, et ont donc besoin d'une durée d'exposition moins longue pour transformer les précurseurs de la vitamine D produits par le foie en vitamine D. La quantité de vitamine D que la peau peut produire est limitée ; une exposition prolongée à la lumière solaire (plus de 30 minutes pour une personne à peau claire, jusqu'à trois heures pour les personnes à peau sombre) n'augmente pas la quantité de cholécalciférol produite par l'organisme. Pour une personne à peau claire, une exposition de 10 à 15 minutes quelques fois par semaine suffit à couvrir les besoins en vitamine D3. La vitamine D fabriquée par la peau est mise en réserve au niveau du foie, du muscle et du tissu gras et utilisée à la demande au cours des périodes hivernales.
Deux formes de vitamine D existent : la vitamine D2 ou ergocalciférol produite par les végétaux et la vitamine D3 ou cholécalciférol produite par les animaux, ainsi que par certains lichens. Chez l'homme, ces deux formes sont converties en 1,25-dihydroxyvitamine D (communément appelé vitamine D), leur principal métabolite actif.
La fonction essentielle de la vitamine D est d'augmenter la capacité d'absorption de l'intestin pour le calcium et le phosphore ce qui permet :
- 1. d'assurer une minéralisation optimale des tissus minéralisés, notamment os, cartilage et dents ;
- 2. de contribuer au maintien des concentrations plasmatiques du calcium et du phosphore.
Les signes cliniques de carence en vitamine D sont : l'ostéomalacie et le rachitisme au niveau osseux, la baisse de tonus musculaire, les crises de tétanie et les convulsions (en relation avec une hypocalcémie) et parfois l'anémie. Chez les personnes âgées, la carence en vitamine D constitue un terrain favorable à la perte osseuse (donc à l'ostéoporose), et à la dépression (comme l'a confirmé une étude néerlandaise évoquée dans Archives of General Psychiatry (mai 2008)).
Plusieurs tranches d'âge constituent des populations à risque de carence en vitamine D : ce sont les nouveau-nés, les nourrissons, les femmes enceintes et les personnes âgées, particulièrement celles placées en institution, qui s'exposent peu au soleil et/ou ont des besoins accrus. D'autres facteurs peuvent aggraver ces risques de carence, tels que la forte pigmentation cutanée, les régimes alimentaires spécifiques (végétarien, macrobiotique), ou bien les pathologies induisant une malabsorption intestinale.
Les apports nutritionnels conseillés en vitamine D ont été définis en considérant que la production endogène couvre 50 à 70 % des besoins quotidiens en cette vitamine.
Un nombre limité d'aliments contient des quantités significatives de vitamine D. En effet, ces teneurs sont étroitement liées à la présence de matières grasses dans les aliments. Les meilleures sources de vitamine D sont les poissons de mer gras, quel que soit leur mode de préparation et de conservation (cuisson à l'eau, fumage, appertisation, grillade) : harengs, saumons, sardines, maquereaux par exemple. Le jaune d’œuf, les produits laitiers, le soja et ses dérivés (tofu et lait de soja par exemple) et certains champignons (avec de grandes différences selon les variétés) sont aussi des sources de vitamine D. De faibles quantités de vitamine D sont également apportées par les viandes, les abats et les pâtés. Les poissons maigres (raie, sole, colin…) en sont dépourvus. Certains lichens produisent également du cholécalciférol (vitamine D3), et sont utilisés pour produire des compléments alimentaires végétaux en cette vitamine.
Selon un mode de réalisation préférée de la présente invention, la vitamine D3 est présente dans la composition selon l’invention en une teneur de 1 à 15 µg de préférence 3 à 10 µg par composition unique.
Le zinc
Le zinc est un métal blanc bleuâtre (symbole Zn) que l’on trouve à l’état naturel dans le monde minéral sous forme de blende (sulfure de zinc) ou de calamine (carbonate de zinc). L’oligothérapie l’utilise sous deux formes de sels : oxyde de zinc et permanganate de zinc. Dans le cadre de la présente invention, on préfèrera l’oxyde de zinc.
Il est vrai que le zinc a gagné depuis peu l’un des premiers rangs de la hiérarchie des oligo-éléments tant son importance apparaît vitale au sein de l’organisme. Le corps humain en contient environ 2,5 g (soit 1 à 2 g de moins seulement que le fer, pour donner un ordre de grandeur). Stocké dans le foie, le pancréas, la prostate (chez l’homme) et les glandes surrénales, le zinc est un co-facteur enzymatique majeur puisqu’il entre dans la composition de plus de 100 enzymes différents. Pour donner un seul exemple de ce rôle enzymatique, le zinc est l’agent clé de l’enzyme Super Oxyde Dismutase (SOD) qui a des propriétés anti-oxydantes, s’opposant à la prolifération des radicaux libres, indispensables pour freiner les maladies dégénératives liées au vieillissement. Les tissus humains les plus sensibles à l’oxydation sont les cellules de la peau, ce qui explique l’intérêt d’une supplémentation en zinc afin de traiter les dermatoses, l’acné en particulier. Mais le zinc a d’autres indications. Il régule en particulier les fonctions hypophysaires. Il contribue également au métabolisme de l’insuline pancréatique et joue un rôle dans l’équilibre acido-basique.

Selon un mode de réalisation préférée de la présente invention, le zinc est présent dans la composition selon l’invention en une teneur de 1 à 40 mg de préférence 10 à 30 mg par composition unique.
Le fer
Le fer est un oligo-élément et fait partie des sels minéraux indispensables qu'on retrouve dans les aliments, mais peut être toxique sous certaines formes. Une carence en fer est source d'anémie et peut affecter le développement cognitif et socio-émotionnel du cerveau de l'enfant ou exacerber les effets de certaines intoxications (saturnisme par exemple).
Le fer est essentiel au transport de l'oxygène et à la formation des globules rouges dans le sang. Il est un constituant essentiel des mitochondries, puisqu'il entre dans la composition de l'hème du cytochrome c. Il joue aussi un rôle dans la fabrication de nouvelles cellules, d'hormones et de neurotransmetteurs. Le fer contenu dans les végétaux (fer dit « non héminique ») Fe3+ ou fer ferrique est moins bien absorbé par l'organisme que celui contenu dans les aliments crus d'origine animale (fer « héminique ») Fe2+ ou fer ferreux. La cuisson des viandes transforme une partie du fer héminique en fer non héminique, moins biodisponible.
En revanche son absorption est inhibée par la consommation de thé et/ou de café car les tanins (polyphénols) sont des chélateurs de fer. C'est pourquoi il est recommandé aux personnes à risques (adolescentes, femmes enceintes, femmes en âge de procréer, végétariens) et buveuses de thé ou de café d'en boire plutôt une heure avant le repas ou deux heures après.
Pour la femme en ménopause et l’homme adulte, les apports journaliers recommandés de fer sont de 10 mg ; ce besoin nutritionnel est de 16 à 18 mg pour la femme de sa puberté à la ménopause.

Le molybdène
Le molybdène est un oligo-élément essentiel à l'organisme. On le retrouve en très faible quantité dans l'organisme, surtout stocké dans le foie et les reins. Cet oligo-élément permet notamment la synthèse de protéines, d'enzymes et des purines qui entrent dans la composition de l'ADN.
Selon un mode de réalisation préférée de la présente invention, le molybdène est présent dans la composition selon l’invention en une teneur de 10 à 100 µg de préférence 15 à 70 µg par composition unique.
Le sélénium
Le sélénium est un oligo-élément constituant des sélénoprotéines dont fait partie le principal antioxydant intracellulaire, la glutathion péroxydase. On en trouve dans les rognons de porc ou de bœuf, l’ail, le poisson et les mollusques. La nutrition occidentale satisfait largement les besoins quotidiens en cet élément mais il est impossible de prédire les concentrations corporelles de sélénium à partir de l'apport nutritionnel parce que son utilisation et sa rétention sont dépendantes de la présence d'acide folique, de vitamine B12 et négativement affectées par la présence d'homocystéine.
Selon un mode de réalisation préférée de la présente invention, le sélénium est présent dans la composition selon l’invention en une teneur de 10 à 100 µg de préférence 15 à 70 µg par composition unique.
Le manganèse
Le manganèse est un oligo-élément (nécessaire à l'homme pour survivre) ; la carence en manganèse (moins de 2 à 3 mg/jour pour un adulte moyen), conduit à des troubles de la reproduction pour les deux sexes, des malformations osseuses, des dépigmentations, une ataxie et une altération du système nerveux central.
Le manganèse est cofacteur de nombreuses enzymes (glycosyltransférase, pyruvate carboxylase, GTP oxaloacétate carboxylase, isocitrate déshydrogénase, malique déshydrogénase, arginine synthétase, glutamine synthétase) intervenant dans des processus métaboliques variés. Il est particulièrement présent dans le métabolisme des hydrates de carbone et la synthèse des mucopolysaccharides. C'est aussi un métal essentiel pour la synthèse d'enzymes (Mn-SOD) participant à la lutte contre le stress oxydant et qui préviennent des dommages causés par les radicaux libres. Il participe aussi à la synthèse de la vitamine E et à l'efficacité de la vitamine B1 (thiamine). Cet activateur de l'oxygène moléculaire joue un rôle dans le fonctionnement de métalloprotéines telles que la superoxyde dismutase.
De nombreux systèmes enzymatiques qui utilisent le magnésium peuvent fonctionner avec le manganèse, mais avec des caractéristiques enzymatiques (Km, Vmax) modifiées. Il peut parfois aussi remplacer le zinc dans d'autres enzymes.
Selon un mode de réalisation préférée de la présente invention, le manganèse est présent dans la composition selon l’invention en une teneur de 0,01 à 5 mg de préférence 1 à 3 mg par composition unique.
Le chrome
Le chrome trivalent à faible dose est un oligo-élément, vital pour le corps humain, essentiel pour le métabolisme du sucre chez l'être humain. Une déficience en chrome peut affecter le potentiel de l'insuline à réguler le niveau de sucre dans l'organisme.
Le chrome n'a pas, comme les autres oligo-éléments, été trouvé dans une protéine avec une activité biologique, et donc son mécanisme d'action dans la régulation du sucre reste inexpliqué.
En fait, les composés organiques du chrome (III) sont plutôt stables, plus que ceux des métaux de transition de la même période (Mn, Fe, Co, Ni, Cu...) et sont donc peu susceptibles de participer à des réactions biologiques, par définition réversibles.
Selon un mode de réalisation préférée de la présente invention, le chrome est présent dans la composition selon l’invention en une teneur de 10 à 60 µg de préférence 15 à 40 µg par composition unique.
Le cuivre
Le cuivre est un oligo-élément indispensable à la vie (hommes, plantes, animaux, et micro-organismes). Le corps humain contient normalement du cuivre à une concentration d’environ 1,4 à 2,1 mg par kg. On trouve du cuivre dans le foie, les muscles et les os. Le cuivre est transporté par la circulation sanguine au moyen d’une protéine nommée céruléoplasmine. Après absorption du cuivre au niveau de l’intestin, il est acheminé vers le foie, lié à l’albumine. Le métabolisme et l’excrétion du cuivre sont contrôlés par la fourniture au foie de céruléoplasmine, et le cuivre est excrété dans la bile. Au niveau cellulaire, le cuivre est présent dans nombre d’enzymes et de protéines, y compris le cytochrome C oxydase et certaines superoxydes dismutases (SOD). Le cuivre sert au transport biologique d’électrons, e.g. les protéines « bleu cuivre », azurine et plastocyanine. La dénomination « bleu cuivre » vient de leur intense couleur bleue due à une bande d’absorption (autour de 600 nm) par transfert de charge coordinat / métal (LMCT).

Le ginseng
Le ginseng (Panax ginseng) est une plante vivace qui appartient au genre Panax de la famille des Araliaceae. Environ dix espèces de ginseng sont cultivées à travers le monde, les espèces les plus connues sont Panax ginseng C.A. Meyer, originaire d'Asie du Nord-Est, et Panax quinquefolium (ginseng américain). La racine du ginseng est réputée pour ses propriétés pharmaceutiques, mais est aussi utilisée comme aliment classique (légume). Le nom générique « Panax » vient des mots grecs Pan, qui signifie « tout », et Akos, qui veut dire « remède ». Panax (en français : panacée) désigne donc le remède universel. Le ginseng est une base essentielle de la pharmacopée asiatique et sa renommée est proverbiale en Asie.
Le ginseng pourrait augmenter la synthèse d’oxyde nitrique par la paroi des vaisseaux, contribuant à un relâchement des fibres musculaires.
Ses principaux constituants bioactifs sont les ginsénosides, dont les multiples effets bénéfiques ont été mis en évidence expérimentalement, notamment leurs effets anti-inflammatoires et antioxydants.
Le ginseng est utilisé en cas de fatigue générale, physique ou intellectuelle. Il est aussi utilisé dans des boissons énergisantes comme stimulant. Depuis des millénaires, les médecines traditionnelles chinoises, japonaises et coréennes l'utilisent pour ses effets toniques et aphrodisiaques.
Selon un mode de réalisation préférée de la présente invention, le ginseng est présent dans la composition selon l’invention en une teneur de 45 à 200 mg de préférence 50 à 100 mg par composition unique.
Milieu physiologiquement acceptable
Un milieu physiologiquement acceptable est un milieu n'ayant aucun effet toxique ou préjudiciable dans les conditions d'utilisation et inerte vis-à-vis des ingrédients actifs du produit de combinaison. Un tel milieu peut par exemple comprendre différents adjuvants selon sa finalité thérapeutique ou cosmétique, comme de façon non limitative, des arômes, des colorants, des charges, des conservateurs, des diluants, des agents mouillants ou de suspension, etc. De tels milieux peuvent également permettre une libération immédiate, modifiée ou retardée de l'un ou l'autre des ingrédients actifs.
Le milieu physiologiquement acceptable peut être composé de texturants, de colorants, d’arômes, de sucre (s).
Par « texturants », on entend les composés qui modifient la texture des produits dans lesquels ils sont incorporés. Des exemples de texturants sont : les farines de guar et de caroube ; les gommes arabiques, de xanthane et de gellane ; les carraghénanes ; les amidons (natifs ou modifiés) ; la cellulose microcristalline ; la pectine ; les alginates (E400 à E405) ; l'agar-agar ; et leurs mélanges. Dans le cadre de la présente invention, le texturant sera de préférence de la pectine.
Par « colorants », on entend des composés d'origine naturelle ou synthétique susceptibles de conférer à eux seuls une coloration à la composition. Des exemples de colorants alimentaires sont connus en Europe sous les codes E100 à E180. Dans le cadre de la présente invention, on utilisera de préférence les colorants d’origine naturelle tels que : E140 (ou chlorophylles), E 163 (ou anthocyanes), E 160c (ou extrait de paprika) ou encore E100 (ou curcumine).
Par « arômes », on entend des composés d'origine naturelle ou synthétique susceptibles à eux seuls de modifier le goût de la composition. Des exemples d'arômes sont l’abricot, l’ananas, la banane, le cassis, la cerise, le citron, le lime, la fraise, la framboise, les fruits de la passion, la mandarine, la menthe, l’orange, la pistache, la poire, le réglisse, la rose, l’anis, la myrtille, la cannelle, le gingembre, le pamplemousse, la pomme, le kiwi, le lychee, le melon, la noix de coco, la pêche, la groseille, la goyave, la grenadine, la mangue, la mûre, la papaye, la vanilline, l'extrait naturel de vanille, les huiles essentielles et leurs mélanges.
Par sucre ou sucres, on entend, au sens de la présente invention, tout hydrate de carbone édulcorant, de préférence du sirop de glucose, du saccharose, glucose, fructose, sucre inverti, miel, maltose, polyol ou leurs mélanges.
Actif(s) additionnel(s)
Tout actif additionnel présentant un intérêt nutritionnel et/ou aidant à retrouver la forme peut également être ajouté aux produits de combinaison de l'invention sous forme pharmaceutique ou cosmétique. Le produit de combinaison pourra comprendre, à titre d'exemple, des vitamines, des sels minéraux, des acides aminés essentiels, des acides gras essentiels, des oligo-éléments, divers extraits végétaux, des fibres, des antioxydants, des flavonoïdes.
Avantageusement, un tel actif additionnel peut être destiné à renforcer l'effet souhaité tel que décrit précédemment.
Bien entendu, l'homme du métier veillera à choisir les actifs additionnels ainsi que leur quantité de telle manière que les propriétés avantageuses de la composition conforme à l'invention ne soient pas, ou substantiellement pas, altérées par l'adjonction envisagée.
A titre d'actif additionnel utilisable, on peut citer :
- les vitamines hydrosolubles et les vitamines liposolubles. Des exemples de vitamines hydrosolubles présentes dans le comprimé préparé selon l’invention sont notamment : la vitamine B6 (pyridoxine). Des exemples de vitamines liposolubles présentes dans les comprimés préparés selon l’invention sont la vitamine K (phylloquinone).
- les antioxydants, tels que les curcuminoïdes ; les caroténoïdes, notamment choisis parmi le beta.-carotène, l'astaxanthine, la zéaxanthine, la lutéine ou les composés en contenant comme les baies de goji ; des composés polyphénols, les flavonoïdes tels que les catéchines ; les proanthocyanidines, les anthocyanines, les OPC (oligomères procyanidoliques) ; les ubiquinones ; les extraits de café contenant des polyphénols et/ou des diterpènes ; les extraits de chicorés ; les extraits de ginkgo biloba ; les extraits de raisins riches en proanthocyanidines; les extraits de piment ; les extraits de soja ; le cacao ; la grenade ; l'Emblica ; le CoenzymeQ10 ; le sélénium,
- les minéraux, tels que le calcium ou l'iode,
- les acides aminés, notamment les acides aminés soufrés, tels que des précurseurs de glutathion, les acides aminés du sélénium, la citrulline,
- les phytostérols, le resvératrol, l'hespéridine, la néohesperidine, l'acide orthosilicique, le monométhylsilanetriol, et leurs mélanges.
De façon préférée, le produit de combinaison selon l'invention est administré par voie orale. Les formes appropriées pour la voie orale sont les comprimés. Dans le cas de la préparation d'une composition solide sous forme de comprimés, on pourra par exemple mélanger les ingrédients actifs principaux avec un véhicule physiologiquement acceptable tel que les dérivés cellulosique (cellulose microcristalline, hydroxypropylméthyl cellulose), le magnésium stéarate, l'amidon, et le saccharose.

Claims (3)
- Produit de combinaison comprenant, à titre de substances actives, au moins de la vitamine A, B1, B2, B3, B5, B8, B9, B12, C, D3, E du zinc, du fer, du molybdène, du sélénium, du manganèse, du chrome, du cuivre et du ginseng pour aider à retrouver la forme dans une composition unique comprenant :
de 200 à 2000 µg de préférence 400 à 1500 µg de vitamine A
de 1,5 à 20 mg de préférence 2 à 10 mg de vitamine B1
de 2 à 15 mg, de préférence 2 à 8 mg de vitamine B2
de 1 à 40 mg, de préférence 10 à 30 mg de vitamine B3
de 8 à 40 mg de préférence 10 à 30 mg de vitamine B5
de 50 à 500 µg de préférence 100 à 300 µg de vitamine B8
de 50 à 500 µg de préférence 100 à 300 µg de vitamine B9
de 1,1 à 10 µg, de préférence 2 à 5 µg de vitamine B12
de 130 à 400 mg de préférence 150 à 300 mg de vitamine C
de 1 à 15 µg de préférence 3 à 10 µg de vitamine D3
de 1 à 40 mg de préférence 10 à 30 mg de zinc
de 1 à 15 mg de préférence 3 à 10 mg de fer
de 10 à 100 µg de préférence 15 à 70 µg de molybdène
