Abstract
본 발명은 암의 항암 효과 증진용 약학적 조성물에 관한 것으로서, 보다 구체적으로 탈유비퀴틴화 효소인 USP14를 억제하는 것으로 알려진 IU1 계열 화합물을 포함하는 조성물을 프로탁과 병용하여 투여하는 경우, 프로탁이 가진 암의 예방 또는 치료 효과를 현저하게 증진시키는 것을 확인하였다. 또한, 본 발명의 약학적 조성물을 활용하는 경우, 기존 보다 저용량의 프로탁을 사용하는 경우에도 암을 효과적으로 치료할 수 있어, 독성 및 부작용의 문제에서 자유로울 수 있고, 프로탁의 단독 사용의 경우 발생하는 응용적 한계와 문제점을 개선하여 항암 치료 분야에서 폭넓게 활용될 수 있다.
Description
본 발명은 항암 효과 증진용 약학적 조성물에 관한 것으로서, 보다 구체적으로 탈유비퀴틴화 효소(deubiquitinase, DUB) 중 하나인 유비퀴틴 특이적 프로테아제 14 (ubiquitin-specific protease 14, 이하 USP14로 지칭) 억제제 중 아이유원 (이하 IU1으로 지칭) 계열 화합물을 포함하는, 프로탁(PROTAC)의 암의 예방 및/또는 치료 효과 증진용 조성물에 관한 것이다.
기존의 신약은 질병표적단백질의 활성 부위나 리간드 결합 부위를 저해하는 원리로 작용하며, 현재 대부분의 소분자 억제제(small-molecule inhibitor)와 치료 항체(therapeutic antibody)가 이러한 방식에 기반해 개발되었다. 상기 약물이 암질환을 포함하는 몇몇 인간 질병에 효과적인 치료제로 사용되고 있으나, 여전히 압도적으로 많은 수의 질병과 질병표적단백질들에 대한 적절한 치료제는 개발되어 있지 않은 상황이다. 약 2만 개의 인간 단백질 중에서 오직 3%만이 현재 FDA 승인을 받은 약물 타겟으로 알려져 있고, 약 7%의 단백질들은 원칙적으로 약물 개발이 가능(druggable)하나 이제까지 성공하지 못한 이유로 난해하거나 도전적인(challenging) 타겟으로 분류할 수 있다. 무엇보다도 대부분의 질병표적단백질에 해당하는 약 3100 여개(16%)의 타겟들은 현재의 신약개발 기술의 적용 자체가 불가능하기 때문에 "undruggable"하다고 간주되고 있다(Nature Reviews Drug Discovery, 16, 19-34). 또한 기존의 활성부위지향성(active site-directed) 신약의 경우 종종 약물 내성의 문제로 인해 결국에는 무력화되기도 한다.
프로탁(PROTAC)은 이러한 전통적인 신약기술개발의 한계를 극복하기 위한 새로운 패러다임으로서, 세포가 단백질을 분해하기 위해서 사용하는 내재적인 단백질 분해기작인 유비퀴틴-프로테아좀 시스템(ubiquitin-proteasome system, UPS)을 사용함으로써 기존 기술로 분해가 불가능한 단백질의 분해 유도(induced proteolysis)가 가능하다. 이러한 "표적단백질 분해 기법(targeted protein degradation, TDP)"은 2001년에 처음 보고되었고, 현재 큰 주목을 받으며 관련 기술의 활발한 개발이 이루어지고 있다(Cell Res, 26, 484-498). 프로탁의 가장 획기적인 특성은 기존에 난해하거나 undruggable 하다고 여겨져 온 다양한 병리단백질이라 하더라도 이러한 분해유도기술의 성질을 이용하여 "제거하는 방식으로 제어"가 가능하다는 점이다. 프로탁의 응용 범위는 원칙적으로 분해가 가능한 모든 단백질과 유비퀴틴화(ubiquitination) 능력이 있는 모든 E3 리가아제(ligase)에 적용되며, 단순히 특정 질병이나 특정 표적단백질에만 국한되지 않기 때문에 플랫폼 기술로 발전할 무궁무진한 잠재력을 지닌다. 실제로 KRas(G12C), AR(AR-V7), STAT3, Tau, CDK9, BET family, BCR-ABL, BTK, HDAC family 등의 여러 중요한 질병 타겟을 분해시키는 프로탁의 개발이 현재 활발히 진행되고 있다.
그러나, 프로탁 기법의 혁신성과 큰 잠재적 가치에도 불구하고, 이 화합물에 의한 표적단백질 분해의 정확한 분자적 기전은 여전히 불명확하다. 프로탁은 E3 리가아제와 표적단백질을 연결하여 삼원 구조(ternary structure)를 형성하는 방식으로 작용하는 것은 분명하나(Drug Discovery Today: Technologies, 31, 15-27), 프로탁에 의한 유비퀴틴화 양상, 형태, 종류 및 조절에 대한 연구는 전혀 이루어져 있지 않다. 다만, 프로탁에 의한 유비퀴틴화 촉진 과정에서 형성되는 폴리유비퀴틴사슬(poly-Ub chains)이나 다중유비퀴틴사슬(multi-Ub chains)이 탈유비퀴틴화 효소(deubiquitinase, DUB)에 의해 잘리게 된다면, 프로탁이 타겟으로 하는 단백질 기질의 분해가 발생하지 않는다. 따라서, 이러한 관점에서 프로탁의 타겟 단백질 분해 기능을 개선하는 연구가 필요한 실정이다.
상기와 같은 문제를 해결하기 위해, 본 발명자들은 프로탁의 타겟 단백질 분해 기능을 증진시키기 위한 물질을 발굴하려 연구한 결과, 탈유비퀴틴화 효소 중 USP14를 억제하는 IU1 계열 화합물을 프로탁과 병용하여 처리하는 경우, 프로탁의 단백질 분해 기능이 현저히 증진되는 것을 확인함으로써 본 발명을 완성하였다.
이에, 본 발명은 USP14(ubiquitin-specific protease 14) 억제제 및 프로탁(proteolysis-targeting chimera, PROTAC)을 포함하는, 암의 예방 또는 치료용 약학적 조성물을 제공하는 것을 목적으로 한다.
또한, 본 발명은 프로탁과 병용되는 암의 예방 또는 치료 효과 증진용 약학적 조성물로서, 상기 조성물은 USP14 억제제를 유효성분으로 포함하는 것을 특징으로 하는, 약학적 조성물을 제공하는 것을 다른 목적으로 한다.
또한, 본 발명은 USP14 억제제 및 프로탁을 포함하는 암의 예방 또는 개선용 건강기능식품을 제공하는 것을 또 다른 목적으로 한다.
또한, 본 발명은 USP14 억제제 및 프로탁을 개체에 투여하는 단계를 포함하는 암의 예방 또는 치료 방법을 제공하는 것을 또 다른 목적으로 한다.
또한, 본 발명은 암의 예방 또는 치료용 약제의 제조를 위한 USP14 억제제 및 프로탁의 용도를 제공하는 것을 또 다른 목적으로 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기 목적을 달성하기 위해, 본 발명은 USP14(ubiquitin-specific protease 14) 억제제 및 프로탁(proteolysis-targeting chimera, PROTAC)을 포함하는, 암의 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명의 일 구현예에 있어서, 상기 USP14 억제제는 IU1 계열 화합물일 수 있다.

본 발명에 따라 USP14의 억제제인 IU1 계열 화합물을 프로탁과 병용하여 투여하는 경우, 프로탁의 타겟 단백질 분해 효과 및 암 치료 효과가 증진됨을 확인함으로써, 향후 암의 예방 및 치료 효과 증진용으로 본 발명의 IU1 계열 화합물이 유용하게 활용될 수 있을 것으로 기대된다.
단, 본 발명의 효과는 상기 효과로 한정되는 것은 아니며, 본 발명의 상세한 설명 또는 청구범위에 기재된 발명의 구성으로부터 추론 가능한 모든 효과를 포함하는 것으로 이해되어야 한다.

도 5a 및 도 5b는 HeLa 세포에서 IU1 화합물을 서로 다른 농도 조건으로 설정한 다음, IU1 화합물의 프로탁의 단백질 분해능 증진 효과를 CDK9 프로탁 농도 의존적으로 확인한 결과를 나타낸 도이다.
도 6a 및 도 6b는 HeLa 세포에서 CDK9 프로탁을 서로 다른 농도 조건으로 설정한 다음, IU1 화합물의 프로탁의 단백질 분해능 증진 효과를 IU1 농도 의존적으로 확인한 결과를 나타낸 도이다.

발명자들은 프로탁의 단백질 분해 기능을 저해하는 탈유비퀴틴화 효소 중 USP14를 억제하는 IU1 계열 화합물을 프로탁과 병용하여 처리하는 경우, 프로탁의 단백질 분해 기능이 증진되는 것을 확인하였는바, 이로써 본 발명을 완성하게 되었다.
이에, 본 발명은 USP14(ubiquitin-specific protease 14) 억제제 및 프로탁(PROTAC)을 포함하는, 암의 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명에서 사용되는 용어 "USP14"는 인간 단백체에 약 100여개 존재하며, 유비퀴틴화를 가역적으로 되돌릴 수 있는 유일한 효소 그룹인 탈유비퀴틴화 효소(deubiquitinating enzyme, DUB) 중 하나로서, 프로테아좀(proteasome) 상에 존재하며, 다중유비퀴틴 사슬에 대해서 특이적으로 작용하는 탈유비퀴틴화 효소이다.

상기 프로테아좀은 세포내 단백질을 분해하는 시스템인 유비퀴틴-프로테아좀 시스템(ubiquitin-proteasome system, UPS)의 구성중 하나로서, 유비퀴틴이 표지된 기질 단백질의 펩타이드 결합을 파괴함으로써 단백질을 분해하는 역할을 수행한다.
본 발명에 있어서, 용어 "단백질"은 "폴리펩타이드" 또는 "폴리펩티드"와 동일한 의미로 사용될 수 있으며, 아미노산 잔기의 폴리머를 지칭한다. 단백질은 하나 이상의 아미노산 잔기가 상응하는 자연 발생한 아미노산의 인공적인 화학적 모방체인 아미노산 폴리머, 뿐만 아니라 자연 발생한 아미노산 폴리머, 변형된 잔기를 함유하는 것들, 및 비-자연 발생한 아미노산 폴리머에 적용된다.
본 발명에서 사용되는 용어 "프로탁(proteolysis-targeting chimera, PROTAC)"은 E3 ligase 결합 모듈-연결체-표적 단백질 결합 모듈로 구성된 플랫폼 기술로서, 질병 유발 표적단백질의 유비퀴틴화를 통해 생체 내 분해를 유도할 수 있으며, 기존의 약물 개발 방식으로부터 유래된 활성지향성(active site-directed) 약물로는 저해하기 어려운 대부분의 질병 유발 단백질을 분해함으로써 저해할 수 있다.

본 발명에서 사용되는 용어 "암"이란 세포가 정상적인 성장 한계를 무시하고 분열 및 성장하는 공격적(aggressive) 특성, 주위 조직에 침투하는 침투적(invasive) 특성 및 체내의 다른 부위로 퍼지는 전이적(metastatic) 특성을 갖는 세포에 의한 질병을 총칭한다.
본 발명에 있어서, 상기 암은 그 종류에 있어서 특별히 한정된 것은 아니며, 뇌종양, 두경부암, 유방암, 폐암, 식도암, 위암, 십이지장암, 충수암, 대장암, 직장암, 간암, 췌장암, 담낭암, 담관암, 항문암, 신암, 수뇨관암, 방광암, 전립선암, 음경암, 정소암, 자궁암, 난소암, 외음암, 질암 또는 피부암일 수 있으며, 본 발명의 일 실시예에서는 자궁암, 전립선암 세포주에서 IU1 계열 화합물 및 프로탁의 병용투여에 의한 항암 효과를 확인하였으나, 이에 한정하는 것은 아니다.
본 발명에서 사용되는 용어, "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 암을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어 "치료"란 본 발명에 따른 약학적 조성물의 투여에 의해 암에 의한 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명자들은 구체적인 실시예를 통해 본 발명에 따른 IU1 계열 화합물이 프로탁의 항암 효과를 증진시킬 수 있음을 확인하였다.
본 발명의 일 실시예에서는, IU1 계열 화합물인 IU1과 CDK9를 타겟으로 하는 프로탁을 병용하여 처리하는 경우, IU1만 단독으로 처리하는 경우 CDK9 단백질을 분해하는 효과가 전혀 확인되지 않았음에도 불구하고, 프로탁의 CDK9 단백질 분해 효과를 현저하게 증진시키는 것을 확인할 수 있었으며, 이러한 효과는 IU1 및 프로탁의 농도와 처리 시간에 의존적임을 확인하였다(실시예 2 및 7 참조).
본 발명의 다른 실시예에서는, 실시예 2에서 확인한 IU1 및 CDK9를 타겟으로 하는 프로탁의 CDK9 단백질 분해 효과의 증진이 단순히 단백질 분해 효과 증진에 그치는 것이 아닌, 실제로 암세포의 생장을 억제하는 것을 확인하였다(실시예 3 참조).
본 발명의 또 다른 실시예에서는, 본 발명의 IU1 및 프로탁 병용에 의한 타겟 단백질 분해 및 항암 효과가 자궁암 세포주인 HeLa 세포에만 효과가 있는 것이 아닌, 일반적인 암세포에도 적용될 수 있다는 사실을 확인하기 위해, 전립선암 세포주인 LNCaP 세포에 IU1과 프로탁을 병용으로 처리했을 때에도, HeLa 세포에 처리했을 때와 같이 프로탁의 타겟 단백질(CDK9)의 분해능이 현저하게 증진됨을 확인하였다(실시예 4 참조).
본 발명의 또 다른 실시예에서는, CDK9를 타겟 단백질로 하는 프로탁 외의 프로탁에 대해서도 단백질 분해능을 증진시킬 수 있는지 여부를 확인하기 위해, E3 리가아제 VHL 기반 프로탁인 BRD4 프로탁에 대하여 BRD4 단백질의 분해 효과 증진 여부를 확인하였다. 그 결과, IU1을 BRD4 프로탁과 병용하여 처리하는 경우, BRD4 프로탁의 농도 의존적으로 프로탁의 BRD4 단백질 분해 효과가 증진됨을 확인하였다(실시예 5 참조).
또한, BRD4 프로탁 이외의 VHL 기반 프로탁인 SHP-2 프로탁에 대해서 SHP-2 단백질의 분해 효과 증진 여부를 확인한 결과, IU1을 SHP-2 프로탁과 병용처리 하였을 때, SHP-2 프로탁의 농도 의존적으로 SHP-2 단백질 분해 효과가 증진되었음을 확인하였다(실시예 5 참조).
본 발명의 또 다른 실시예에서는, 본 발명의 IU1 계열 화합물 중 하나인 IU1-47을 활용하는 경우에도 IU1을 프로탁과 병용하여 처리하는 경우와 유사하게 프로탁의 타겟 단백질 분해능이 증진되고, 오히려 IU1-47을 처리했을 때, IU1을 프로탁과 병용하여 처리했을 때보다 프로탁의 타겟 단백질 분해능이 현저하게 증가했음을 확인하였다(실시예 6 참조).
본 발명의 또 다른 실시예에서는, IU1 계열 외 탈유비퀴틴화 효소 억제제의 프로탁의 단백질 분해능 증진 효과 여부 확인하기 위해, HeLa 세포에서 비특이적 탈유비퀴틴화 효소 저해제(pan DUB inhibitor) PR-619 화합물의 CDK9 프로탁 단백질 분해능 증진 효과를 IU1-47과 비교하여 확인하였다. 그 결과, IU1-47은 프로탁의 분해 효과를 현저하게 증진시키는 반면, PR-619는 처리한 모든 농도에서 프로탁의 분해 효과를 나타내지 않았다(실시예 8 참조).
또한, HeLa 세포에서 특정 탈유비퀴틴화 효소 저해제인 USP7/47 dual inhibitor(이하 P50429라 지칭)를 CDK9 프로탁과 병용처리하여 프로탁의 단백질 분해능 증진 효과를 IU1-47과 비교한 결과, IU1-47 (20 μM)은 프로탁의 분해 효과를 현저하게 증진시키는 반면, P50429는 프로탁의 분해 효과를 증진시키지 않음을 확인하였다(실시예 8 참조).
본 발명자들은 상기와 같은 결과를 통해 본 발명의 USP14 억제제인 IU1 계열 화합물을 프로탁과 병용하여 투여하는 경우, 프로탁의 타겟 단백질에 대한 분해 효과 및 항암 효과를 현저하게 증진시킬 수 있기에, 암 치료 분야에서 폭넓게 활용될 수 있다는 사실을 확인하였다.
본 발명의 다른 양태로서, 본 발명은 프로탁과 병용되는 암의 예방 또는 치료 효과 증진용 약학적 조성물로서, 상기 조성물은 USP14 억제제를 유효성분으로 포함하는 것을 특징으로 하는, 약학적 조성물을 제공한다.
상기 "프로탁", "암", "예방", "치료", "USP14 억제제" 등은 전술한 범위 내일 수 있다.
본 발명에 있어서, 상기 "증진"이란, 본 발명에 따른 암의 예방 또는 치료 효과 증진용 약학적 조성물이 프로탁과 병용 투여될 경우, 프로탁이 단독 투여되는 경우 보다 우수하게 암을 억제시키거나 발병을 지연시키거나 암에 의한 증세가 현저하게 호전되는 것을 의미한다.
상기 병용은 프로탁과 동시에(simultaneous), 별도로(separate) 또는 순차적(sequential)으로 투여되는 것일 수 있다.
본 발명에 있어서, 상기 암의 예방 또는 치료 효과 증진용 약학적 조성물이 프로탁과 병용투여되는 경우, 프로탁이 단독 투여되는 경우 보다 프로탁의 타겟 단백질에 대한 분해 효과 및 항암 효과를 현저하게 증진시킬 수 있다.
본 발명에서 용어, "투여"는 어떠한 적절한 방법으로 대상에게 본 발명의 약학적 조성물을 도입하는 것을 말하며, 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있다. 비경구투여 경로의 예로는 피하, 복강 내, 폐 내, 비강 내, 근육 내, 정맥 내 또는 동맥 내가 포함된다.
본 발명의 약학적 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 또는 희석제를 추가로 포함할 수 있다. 약학적으로 허용 가능한 담체를 포함하는 조성물은 경구 또는 비경구의 여러 가지 제형일 수 있다. 제제화 할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 제제에는 정제, 환제, 산제, 과립제, 캡슐제, 현탁제, 내용액제, 유제, 시럽제, 멸균된 수용액, 비수성용제, 동결건조제제 등이 포함될 수 있으며, 이러한 제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제, 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 혼합하여 조제될 수 있다.
또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용될 수 있다. 경구투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함될 수 있다. 비수성용제, 현탁용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
또한, 본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에서의 용어 "약학적으로 유효한 양"은 의학적 용도에 적용 가능한 합리적인 수혜/위험 비율로 암을 억제 또는 완화하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 체중, 식이, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
본 발명의 약학적 조성물의 투여는 하루에 한 번 투여되는 것일 수도 있고, 수 회 나누어 투여되는 것일 수 있고, 격일에 한 번 투여되는 것일 수도 있으며, 일주일에 한 번 투여되는 것일 수도 있다. 구체적으로, 상기 약학적 조성물은 0.001 내지 1000 mg/kg/day로, 보다 구체적으로 0.1 내지 100 ㎎/kg/day로 투여될 수 있다.
본 발명의 약학적 조성물은 암의 예방 또는 치료 효과를 가지는 공지의 조성물 또는 다른 약학적 조성물과 병행하여 투여하는 것일 수 있고, 동시에, 별도로, 또는 순차적으로 투여하는 것일 수 있으며, 단일 또는 다중 투여하는 것일 수 있다. 상기 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.

실시예 1. 실험준비
1-1. 시약 및 항체
본 발명에서 활용된 시약 및 항체는 시판되어 용이하게 구할 수 있는 것을 활용하였으며, 구체적으로 본 발명에 활용된 화합물은 하기 표 1에 나타낸 것을 활용하였고, 항체는 하기 표 2에 나타낸 것을 활용하였다.

1-2. 화합물 용해, 보관 및 분주
본 발명의 실험을 위해 활용된 화합물은 파우더 형태로 합성된 것을 무수성(anhydrous) DMSO(Sigma)를 용매로 하여 50 내지 100 mM의 농도로 용해하였다. 용해된 화합물은 연속희석(serial dilution)을 통해 바로 실험에 이용하거나, 실험에 이용하기 전에는 -80℃에 소량의 단위로 나누어 보관함으로써 동결해동과정을 최소화하였다. 극소량의 화합물의 다중 분주(multi-dispensing)가 필요한 경우에는, 맞춤형으로 제작된 고밀도 반복 스크리닝 자동화 워크스테이션(customized Biomek i5 HDR system, Beckman Coulter) 기기와 핀툴(pin tool)을 이용하여 정밀 분주하였다.
1-3. 세포 배양
본 발명의 실험을 위해 활용된 HeLa 세포(자궁경부암 세포) 및 LNCaP 세포(전립선암 세포)는 ATCC(American Type Culture Collection)에서 얻었으며, 각각 10% FBS, 1% Penicillin/Streptomycin 및 1% L- glutamine을 함유하는 Dulbecco's Modified Eagle's Medium (DMEM, 1x) 또는 RPMI-1640 (1x) 배지를 이용하여, 37℃, 5% CO2 세포 인큐베이터에서 배양하였다. 프로탁(PROTAC)과 IU1 및 IU1-47 등의 탈유비퀴틴화 효소(Deubiquitinating enzyme, DUB) 억제제 실험을 위해, 억제제 처리 하루 전에 5x105(HeLa) 또는 1x106(LNCaP)개의 세포를 12-웰 플레이트(Corning, #3513)의 각 웰에 시딩(seeding)한 다음, 인큐베이터에서 배양하였다. 배양 24 시간 후, 각 웰을 연속희석(serial dilution)한 화합물이 포함된 배지로 교체하고, 동일한 조건에서 24 시간 이내로 배양하였다. 모든 대조군의 경우 실험군으로 처리한 화합물 대비 용매(vehicle)인 DMSO를 동량으로 처리하여 비교하였다.
1-4. 웨스턴 블로팅(Western blotting)
웨스턴 블로팅을 위한 샘플을 준비하기 위해, HeLa 및 LNCaP 세포를 ice-cold PBS(1x)로 1회 세척한 후, protease inhibitor cocktail(Roche), 1 mM PMSF 및 1 mM DTT를 추가한 RIPA buffer(ROCKLAND, MB-030-050) 또는 1% SDS lysis buffer [1%(w/v) SDS, 10 mM Tris-HCl, 1 mM EDTA, pH 8.0]를 사용하여 용해(lysis)하였다. 모든 용해물(lysates)은 BCA protein assay(Thermo Fisher Scientific)로 정량한 다음, 각각의 용해물 15 내지 30 ㎍을 SDS-PAGE 방법으로 running하였다. 이후 PVDF 막(membrane)에 transfer, blocking solution(5% BSA 또는 5% non-fat dry milk in 1x TBST) 처리, 지정된 일차 항체 처리, HRP-linked 이차 항체 처리 순서대로 실험을 수행하였다. 각 처리 과정 사이에는 과량의 1x TBST로 여러 번 세척하였다. 막 각각은 케미닥(iBright750, Invitrogen)을 이용하여 이미지 분석 및 정량하였다.
1-5. 세포 생존율 측정
본 발명의 화합물 처리에 의한 세포 생존율(Cell viability)을 측정하기 위해, HeLa 및 LNCaP 세포를 96-웰 플레이트(SPL, #33396)에 500 cells/well(HeLa) 또는 3000 cells/well(LNCaP)로 시딩 하였고, 37℃, 5% CO2의 인큐베이터에서 배양하였다. 24 시간 후 화합물 함유 배지로 교체한 다음, 48 시간 동안 배양기에서 같은 조건으로 배양하였다. 이후 ATP-lite 1 step assay kit(Perkin Elmer)을 사용하여 Envision microplate reader (Perkin Elmer)에서 발광도 분석을 통해 세포 생존율을 측정하였다.
1-6. 데이터 정량화 및 분석
웨스턴 블로팅을 통해 얻어낸 데이터는 iBright에서 제공하는 분석 소프트웨어를 활용하여 정량화를 수행하였으며, 모든 표적단백질의 수준은 GAPDH로 정규화 하였다. 정량화 및 정규화를 수행한 다음, GraphPad Prism software를 사용한 비선형 회귀분석(nonlinear regression)을 통해 본 발명의 실험에서 활용한 화합물들의 DC50 값을 도출하였다.
세포 생존율 확인을 위한 실험에서 측정된 결과는 엑셀로 초기 data processing한 다음, Prism software를 사용한 비선형 회귀분석을 통해 IC50 값을 도출하였다.
실시예 2. IU1의 프로탁의 단백질 분해능 증진 효과 확인
2-1. 프로탁의 단백질 분해능 확인
프로탁(PROTAC)은 세포의 내재적인 단백질 분해기작인 유비퀴틴-프로테아좀 시스템(ubiquitin-proteasome system, UPS)을 이용하는 표적단백질 분해 기법(targeted protein degradation, TDP)으로서, 구체적인 매커니즘은 도 1에 나타낸 바와 같다.
본 발명자들은 프로탁을 활용한 기질의 유비퀴틴화는 다양한 종류와 형태의 폴리유비퀴틴사슬(poly-Ub chains) 혹은 다중유비퀴틴사슬(multi-Ub chains)을 형성할 것이며, 이 중 상당 부분이 USP14을 비롯한 탈유비퀴틴화 효소 반응에 의해 분해가 저해될 것이고, 그렇기에 탈유비퀴틴화 효소를 억제하였을 때, 프로탁에 의한 단백질 분해효과가 증진될 것으로 도 2와 같이 예상하고 구체적인 실험을 수행하였다.
우선, 도 3a와 같은 형태로 작용하여 CDK9 단백질을 분해하는 것으로 보고되어 있는 프로탁(이하 CDK9 프로탁)의 실제 단백질 분해효과를 확인하기 위해, 유방암(MB231), 전립선암(LNCaP) 및 자궁경부암(HeLa) 세포주를 활용한 단백질 분해능 확인 실험을 수행하였다.
그 결과, 도 3b에 나타낸 바와 같이, CDK9 단백질을 분해하는 것으로 알려진 CDK9 프로탁이 3종류의 암세포주 모두에서 기존의 보고된 결과와 일치하는 CDK9 단백질 분해능을 보여주는 것을 확인하였다.
2-2. IU1의 프로탁의 단백질 분해능 증진 효과 확인
상기 실시예 2-1과 동일한 조건으로 탈유비퀴틴화 효소 USP14의 억제제인 IU1에 의한 CDK9 프로탁의 단백질 분해능 증진 효과를 확인하였다.
구체적으로 IU1과 CDK9 프로탁을 HeLa 세포에 같이 처리한 결과, 도 4a에 나타낸 바와 같이, 처리 농도(프로탁: 1, 10, 25, 50 및 100 nM, IU1: 50 μM)에 따라 CDK9 단백질의 분해효과가 현저하게 증가됨을 확인할 수 있었다. 이러한 효과는 도 4b에 나타낸 바와 같이, IU1 단독으로 처리(IU1 최대 처리 농도: 50 μM)했을 때, CDK9 단백질의 분해가 발생하지 않음을 확인할 수 있었다.
또한, 도 4c에 나타낸 바와 같이, IU1, 프로테아좀(proteasome) 억제제인 MG132 및 오토파지(autophagy) 억제제인 바필로마이신(bafilomycin) A1(Baf)의 처리 유무를 달리하면서 CDK9 프로탁을 처리하였다.
그 결과, 프로테아좀 억제제인 MG132를 프로탁 및 IU1과 같이 처리한 경우, CDK9 단백질의 수준이 대조군과 유사하게 복구되었음을 확인한 반면, 바필로마이신(bafilomycin) A1을 처리한 경우, 프로탁 및 IU1의 처리에 의한 CDK9 단백질의 분해가 여전히 유지되고 있음을 확인할 수 있었다.
이에, 본 발명자들은 프로탁 및 IU1을 동시에 처리하는 경우, 리소좀(lysosome)이 아닌, 프로테아좀 의존적으로 표적 단백질의 분해가 일어남을 확인할 수 있었다.
2-3. IU1 및 프로탁의 농도 의존적 단백질 분해효과 확인
탈유비퀴틴화 효소 억제제인 IU1의 CDK9 프로탁의 단백질 분해효과 증진 수준을 농도에 따라 구체적으로 확인하였다.
우선, IU1의 농도(10, 25, 75 및 100 μM)를 고정한 다음, CDK9 프로탁의 농도 의존적인 조건(1, 10 25, 50 및 100 nM)에서 프로탁의 단백질 분해효과를 관찰한 결과, 도 5a 및 도 5b에 나타낸 바와 같이, 10 μM의 IU1 농도를 제외한 모든 농도에서 프로탁 농도 의존적으로 IU1이 CDK9 프로탁에 의한 CDK9 분해효과를 증진시킴을 확인하였다.
또한, CDK9 프로탁의 농도(5, 10, 20 및 25 nM)를 고정한 다음, IU1의 농도 의존적인 조건(10, 25, 50, 75 및 100 μM)에서 프로탁의 단백질 분해효과를 관찰한 결과, 도 6a 및 도 6b에 나타낸 바와 같이, 모든 농도에서 IU1 농도 의존적으로 단백질 분해효과가 증진됨을 확인하였다.
2-4. IU1 및 프로탁의 병용처리에 의한 단백질의 DC
50
값 측정
HeLa 세포에서 CDK9 프로탁을 Vehicle(DMSO)로 처리한 대조군과 IU1을 병용처리한 그룹에서 프로탁의 농도를 높여가며(0.1, 1, 5, 10, 20, 30, 50, 75, 100 및 250 nM) 웨스턴 블롯을 수행하여 CDK9 단백질의 수준을 확인하였다.
그 결과, 도 7a 및 도 7b에 나타낸 바와 같이, 대조군에 비해 IU1을 처리한 그룹, 그 중에서도 높은 농도의 IU1을 처리한 그룹에서 높은 CDK9 단백질 분해능을 보여줌을 확인하였다.
또한, 이러한 결과를 정량화하여, CDK9 단백질분해 DC50(50%의 표적단백질분해가 일어나는 프로탁 약물 농도)를 측정하였다.
그 결과, 도 7c에 나타낸 바와 같이, IU1 농도가 증가함에 따라 CDK9 프로탁의 DC50값이 현저하게 감소하는 것을 확인하여, IU1을 CDK9 프로탁과 병용하여 처리하는 경우 CDK9 프로탁의 CDK9 단백질 분해능이 현저하게 상승됨을 확인할 수 있었다.
예를 들어, IU1을 75 μM 만큼 병용하여 처리하는 경우 CDK9 프로탁의 단백질 분해능이 Vehicle을 처리한 대조군에 비해 약 5배가량 증진됨을 DC50 값을 통해 확인할 수 있었다.
실시예 3. IU1 및 프로탁의 병용처리에 의한 암세포의 생존율 확인
상기 실시예 2에서 확인한 IU1 및 CDK9 프로탁의 병용에 의해 증진된 CDK9 단백질의 분해 효과에 의해 실질적으로 프로탁의 암세포에 대한 항암 효과가 향상되는지 여부를 확인하기 위해, HeLa 세포에 IU1 및 CKD9 프로탁을 처리한 다음, 암세포의 생장율을 확인하였다.
그 결과, 도 8에 나타낸 바와 같이, IC50(50%의 세포 생장 저해가 일어나는 프로탁 약물 농도)를 측정하였을 때, IU1을 50 μM 만큼 병용하여 처리하는 경우, CDK9 프로탁의 항암 효과가 현저하게 증진되는 것을 확인할 수 있었다.
실시예 4. IU1의 LNCaP 세포에서 프로탁의 단백질 분해능 증진 확인
본 발명의 IU1의 프로탁 단백질 분해효과 증진이 자궁경부암 세포인 HeLa 세포주 뿐만 아니라, 다른 암종에도 효과가 있다는 사실을 확인하기 위해, 전립선암 세포주인 LNCaP 세포주에 IU1과 CDK9 프로탁을 함께 처리한 다음, 단백질 분해효과를 확인하였다.
그 결과, 도 9에 나타낸 바와 같이, CDK9 프로탁만을 단독으로 처리한 경우에 비해, IU1과 CDK9 프로탁을 함께 처리한 경우, CDK9 단백질의 분해효과가 현저하게 증가됨을 확인하였다.
또한, 상기와 같은 효과는 상기 실시예 2-3에서 수행한 바와 같이, IU1의 농도(10, 25, 75 및 100 μM)를 고정한 다음, CDK9 프로탁의 농도 의존적인 조건(1, 5, 10, 25 및 50 nM)에서 프로탁의 단백질 분해효과를 관찰한 결과, 도 10a 및 도 10b에 나타낸 바와 같이, 모든 농도에서 프로탁의 농도 의존적으로 IU1가 CDK9 프로탁에 의한 CDK9 분해효과를 증진시킴을 확인하였다.
상기와 같은 결과를 통해 본 발명자들은 본 발명의 USP14 억제제의 프로탁의 단백질 분해능 증진 효과가 특정 암종에 국한되는 것이 아닌, 일반적인 암종 모두에 적용될 수 있음을 확인하였다.
실시예 5. IU1의 CDK9 프로탁 외의 프로탁의 단백질 분해능 증진 효과 확인
본 발명의 USP14 억제제가 상기 실시예 2 및 실시예 4에서 실험을 통해 확인한 CDK9 프로탁 외의 프로탁에 대해서도 단백질 분해능을 증진시킬 수 있는지 여부를 확인하기 위해, E3 리가아제 VHL 기반 프로탁인 BRD4 프로탁에 대하여 BRD4 단백질의 분해효과 증진 여부를 확인하였다. 이를 위해 1% SDS lysis buffer 및 RIPA buffer에서 BRD4 프로탁만 처리한 대조군과 IU1과 BRD4 프로탁을 병용하여 처리한 실험군을 비교한 결과, 도 11에 나타낸 바와 같이, 두 가지의 버퍼 모두에서 50 μM의 IU1을 BRD4 프로탁과 병용하여 처리하는 경우, BRD4 프로탁의 농도 의존적으로 프로탁의 BRD4 단백질 분해효과가 증진됨을 확인할 수 있었다.
또한, BRD4 프로탁 이외의 VHL 기반 프로탁인 SHP-2 프로탁에 대해서 도 7에서의 실험과 유사한 실험 조건 하에서 SHP-2 단백질의 분해효과 증진 여부를 확인하였다. 그 결과, 도 12a 및 도 12b에서 나타낸 바와 같이, SHP-2 프로탁에 대해서도 IU1을 병용처리 하였을 때, SHP-2 프로탁의 농도 의존적으로 SHP-2 단백질 분해효과가 증진되었음을 확인할 수 있었다.
또한, 상기와 같은 결과를 통해 본 발명자들은 본 발명의 USP14 억제제의 프로탁의 단백질 분해능 증진 효과가 CDK9 프로탁과 같이 CRBN 기반 E3 리가아제 프로탁에만 국한되는 것이 아닌, VHL 기반 E3 리가아제 프로탁에도 적용될 수 있음을 확인하였다.
실시예 6. IU1 외 USP14 억제제의 프로탁의 단백질 분해능 증진 효과 확인
본 발명의 상기 실시예 2 내지 4에서 프로탁의 단백질 분해 증진 효과를 확인한 USP14 탈유비퀴틴화 효소 억제제인 IU1 외에도 IU1-47을 비롯한 IU1 계열 억제제가 USP14를 억제하는 것으로 연구를 통해 확인되었기에, IU1 외의 IU1 계열 USP14 억제제로 알려진 IU1-47도 IU1과 마찬가지로 프로탁의 단백질 분해능을 증진시킬 수 있는지 여부를 확인하였다. 이를 위해 Vehicle(DMSO) 군과 IU1-47의 각 농도(10, 20 및 30 μM)를 고정하고 CDK9 프로탁의 처리 농도(0.1, 1, 5, 10, 20, 30, 50, 75, 100 및 250 nM)를 달리하면서 CDK9 프로탁을 처리하고 CDK9 단백질 분해수준을 확인하였다.
그 결과, 도 13a 및 도 13b에 나타낸 바와 같이, Vehicle만을 처리한 대조군에 비해, 모든 농도에서 현저한 수준으로 프로탁의 단백질을 분해능을 증진시키는 효과가 있다는 사실을 확인하였다.
또한, 도 13c에서 상기 도 13a 및 도 13b에서 얻어낸 데이터를 정량화한 다음, DC50 값을 구체적으로 확인했을 때, IU1 계열 탈유비퀴틴화 효소 억제제인 IU1-47을 CDK9 프로탁과 병용하여 처리하는 경우, IU1-47을 20 μM으로 처리한 조건에서 CDK9 프로탁만 단독으로 처리하는 것에 비해 약 6배 수준으로 프로탁의 단백질 분해능을 증진시킴을 확인할 수 있었다.
또한, 상기와 같은 IU1-47의 프로탁에 대한 단백질 분해능 증진 효과가 IU1과 유사하게 농도 의존적인지 여부를 확인하기 위해, 프로탁의 농도를 각각 고정한 다음(5, 10 및 20 nM), IU1-47의 농도를 바꾸어 가면서 IU1-47과 프로탁의 병용처리시의 단백질 분해 수준을 확인하였다.
그 결과, 도 14에 나타낸 바와 같이, 단순히 IU1-47만을 처리했을 때에는 CDK9을 전혀 분해하지 못하는 것을 확인하였으나, CDK9 프로탁과 처리했을 때, 농도 의존적으로 프로탁의 분해효과를 현저하게 증진시키는 것을 확인할 수 있었다.
실시예 7. IU1의 시간경과에 따른 프로탁의 단백질 분해능 증진 확인
HeLa 세포에서 CDK9 프로탁(100 nM) 단독 혹은 IU1(50 μM)과 병용처리 조건에서, IU1 화합물의 CDK9 프로탁 단백질 분해능 증진 효과를 시간경과 의존적(0, 1, 3, 6, 12 및 24 h)으로 확인하였다.
그 결과, 도 15에 나타낸 바와 같이 IU1을 병용처리한 경우, 프로탁의 분해효과를 더 빠르게 증진시키는 것을 확인할 수 있었다.
실시예 8. IU1 계열 외 탈유비퀴틴화 효소 억제제의 프로탁의 단백질 분해능 증진 효과 여부 확인
HeLa 세포에서 비특이적 탈유비퀴틴화 효소 저해제(pan DUB inhibitor) PR-619 화합물을 서로 다른 농도 조건(0, 1, 5, 10 및 20 μM)으로 설정한 다음, PR-619 화합물의 CDK9 프로탁 단백질 분해능 증진 효과를 IU1-47(10 μM)과 비교하여 확인하였다.
그 결과, 도 16에 나타낸 바와 같이 IU1-47은 프로탁의 분해효과를 현저하게 증진시키는 반면, PR-619는 처리한 모든 농도에서 프로탁의 분해효과를 나타내지 않았다.
또한, HeLa 세포에서 특정 탈유비퀴틴화 효소 저해제인 USP7/47 dual inhibitor(이하 P50429라 지칭)를 1 및 10 μM 로 설정하고 CDK9 프로탁과 병용처리하여 프로탁의 단백질 분해능 증진 효과를 IU1-47과 비교하였다.
그 결과, 도 17에 나타낸 바와 같이 IU1-47 (20 μM)은 프로탁의 분해효과를 현저하게 증진시키는 반면, P50429는 프로탁의 분해효과를 증진시키지 않았다.
이에, 본 발명자들은 탈유비퀴틴화 효소 저해제에 의한 프로탁의 분해 증진 효과는 비특이적인 탈유비퀴틴화 효소 억제제나 USP7/47 탈유비퀴틴화 효소 억제제를 통해서가 아닌, IU1 계열의 저해제에 의한 특이적인 방식을 통해 일어남을 확인할 수 있었다.
본 발명자들은 상기와 같은 결과를 통해 탈유비퀴틴화 효소인 USP14 억제제 중 IU1 계열은 IU1 뿐만 아니라, IU1-47 또한 프로탁의 단백질 분해능을 증진시키는 효과가 있으며, 이를 통해 본 발명의 화합물들을 프로탁과 병용하여 처리하는 경우 프로탁의 항암효과를 현저하게 증진시킬 수 있다는 사실을 확인하였다.
상기 진술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.

- USP14(ubiquitin-specific protease 14) 억제제 및 프로탁(proteolysis-targeting chimera, PROTAC)을 포함하는, 암의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,상기 USP14 억제제는 IU1 계열 화합물인 것을 특징으로 하는, 조성물.
