Abstract
본 발명은 안구건조증 또는 안구건조증 연관 안과 질환의 예방, 개선 또는 치료용 조성물에 관한 것으로서, 본 발명에서는 PKA(protein kinase A) 세포신호전달체계를 활성화하여 마이봄샘 기능 장애에서 발생될 수 있는 지질 합성 저하를 효과적으로 개선하여, 마이봄샘에서 분비된 지질이 안구 표면 수분 증발을 줄여 안구건조증 또는 안구건조증 연관 안과 질환을 예방, 개선 또는 치료할 수 있다.
Description
본 발명은 안구건조증 또는 안구건조증 연관 안과 질환의 예방, 개선 또는 치료용 조성물에 관한 것이다.
직장 및 작업환경 변화와 미세먼지 등과 같은 기후 악화로 인하여 전 세계적으로 안구건조증 환자가 증가하고 있다. 나아가, 전자기기 사용이 늘고 시력교정시술이 확대되면서 안구건조증 치료제에 대한 요구가 증가되고 있다.
상기 안구건조증은 눈부심, 눈의 자극감, 침침함, 이물감, 작열감, 국소 소양감 등의 증상을 나타내는 증상으로서 눈의 눈물 분비량이 감소하거나 성분의 변화로 인해 나타나는 증상이다.
안구건조증의 가장 흔한 원인은 노화로서 정상적인 노화 현상에 의해 눈물 분비량이 감소하거나 눈물의 상태가 변화함으로 나타날 수 있다. 또한, 류마티스성 관절염, 쇼그렌 증후군, 루프스, 공피증, 당뇨병, 비타민 A 결핍증 등의 질병이 있으면 눈물 분비가 감소하여 안구건조 증상이 나타나기도 하며, 만성결막염, 갑상선 질환, 갱년기 여성호르몬 감소, 약물복용 등에 의해 나타날 수도 있다.
뿐만 아니라, 눈의 보호막 역할을 하는 지방층, 수성층, 점액층 3가지 성분으로 이루어지는 눈물막이 불안정하게 되면 눈이 자주 자극을 받게되어 눈물 생산 감소, 눈꺼풀의 감기는 기능 불량 등으로 인해 안구건조증이 발생될 수 있다.
그 외에 환경적인 요인으로서 주위 환경이 건조하거나, 연기, 먼지 자극, 햇빛, 바람 등으로 눈이 자극되거나, 독서나 컴퓨터를 하면서 무의식 중에 눈을 깜박이는 횟수가 줄어들면 안구 건조증이 생기기 쉽다.
안구건조증의 가장 보편적인 대처는 인공누액을 이용하여 모자라는 눈물 성분을 공급해 주는 것으로, 용액, 연고, 묽은 젤리 형태 등 다양한 종류가 있다. 그러나, 인공누액을 이용하는 방법은 안구건조증의 증상을 일시적으로 개선하는 효능이 있을 뿐, 안구건조증의 진행을 억제하여 증상을 근원적으로 완화하기에는 역부족하다.
한편, 마이봄샘(meibomian gland, MG)은 상부 및 하부족 근판에 위치한 변형된 피지선으로서, 수성층을 코팅하고, 안구 표면을 매끄럽게 할 뿐만 아니라, 외인성 물질로부터 눈을 보호하는 등의 역할을 수행한다. 특히, 마이봄샘이 제대로 기능하게 되면, 마이봄샘에서 분비된 지질이 안구 표면 수분 증발을 줄이고, 안구 건조를 예방할 수 있다. 이와 같은 마이봄샘의 기능 장애가 유발되는 경우 환자의 삶의 질에 영향을 미칠 수 있는 안구 불편감, 통증, 결막 충혈, 시력 저하 및 안구건조 등의 증상이 발생될 수 있다.
이에, 마이봄샘 기능 장애를 개선하여 안구건조증을 예방 또는 치료할 수 있는 약물의 개발이 필요한 시점이다.
본 발명의 일 목적은 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 치료용 약학 조성물을 제공하는 것이다.
본 발명의 다른 목적은 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 건강기능식품을 제공하는 것이다.
본 발명의 또 다른 목적은 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 점안제를 제공하는 것이다.
본 발명의 일 구현 예에서는 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 치료용 약학 조성물을 제공한다.
본 발명의 상기 약학 조성물은 PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함한다.
본 발명의 상기 "PKA 세포신호전달체계"란, cAMP(cyclic AMP)에 의존적인 PKA 단백질이 관여하는 신호전달체계를 의미한다. 이와 같은 세포신호전달체계는 다양한 대사 효소를 인산화 하여 세포 내 대사를 조절할 수 있다.
본 발명의 목적상 상기 PKA 세포신호전달체계 활성화제는 하기 화학식 1로 표시되는 화합물, 또는 이의 유도체일 수 있다:

본 발명의 상기 화학식 1로 표시되는 화합물((3R,4aR,5S,6S,6aS,10S,10aR,10bS)-6,10,10b-trihydroxy-3,4a,7,7,10a-pentamethyl-1-oxo-3-vinyldodecahydro-1H-benzo[f]chromen-5-yl acetate)은 열대성 식물 콜레우스 포스콜리 뿌리에 들어있는 성분으로서, 아데닐산 고리화효소를 활성화하는 저분자 화합물이며, 세포 내 cAMP를 증가시킨다. 이와 같은 화합물을 마이봄샘에 처리하는 경우 PKA 세포신호전달체계가 활성화되어 마이봄샘에서 지질합성과 관련되는 PPARγ 및 SREBP-1 단백질 및 이를 암호화하는 유전자의 발현 수준이 현저하게 증가됨으로써, 마이봄샘의 기능부전을 개선시키고 안구건조증을 예방, 개선 또는 치료할 수 있다.
본 발명의 상기 “유도체”란, 어떤 유기 화합물을 모체로 했을 때 작용기의 도입, 산화, 환원, 원자의 치환 등 모체의 구조와 성질을 대폭 변화시키지 않는 한도 내에서 변화된 화합물을 의미한다. 예를 들면, 상기 화학식 1로 표시되는 화합물의 유도체는 하기 화학식 2로 표시되는 화합물([(3R,4aS,5S,6S,6aS,10aS,10bR)-3-ethenyl-6-hydroxy-3,4a,7,7,10a-pentamethyl-1-oxo-2,5,6,6a,8,9,10,10b-octahydrobenzo[f]chromen-5-yl] acetate)과 같이 일부 작용기가 치환된 화합물일 수 있으나, 이에 제한되는 것은 아니다:

본 발명의 상기 "안구건조증"이란, 눈물샘의 염증 및 각막의 탈신경화에 의하여 눈물 분비가 억제되거나 마이봄샘의 기능부전(Meibomian gland dysfunction, MGD) 또는 눈꺼풀의 기능 이상에 따른 비정상적인 눈물 증발 등에 의하여 발생하는 질환일 수 있고, 상기 안구건조증에 의해 눈물이 과다 건조되어 각막의 상처로 나타나는 각결막 상피장애일 수 있다. 본 발명의 목적상, 상기 안구건조증은 마이봄샘의 기능부전에 의해 발생되는 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 "안구건조증 연관 안과 질환"이란, 안구건조증으로 인하여 발생될 수 있는 질환으로서, 건성 각결막염, 건성으로 인한 각막 궤양, 건성안으로 인한 안건염 및 건성으로 인한 눈의 충혈로 이루어진 군으로부터 선택되는 어느 하나 이상인 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 "마이봄샘의 기능부전"이란, 증발형 안구건조증의 주요한 원인이며, 안구건조증 치료 측면에서도 가장 큰 비중을 차지하고 있다. 마이봄샘 기능부전은 마이봄샘의 배출구가 막히거나, 마이봄샘 분비의 양적 또는 질적인 변화로 발생하는 만성 질환이다. 이러한 마이봄샘 기능부전은 과도한 눈물의 증발로 인해 눈물의 삼투압 증가 및 염증의 증가로 이어지고 심한 경우 마이봄샘이 파괴될 수 있다. 따라서 마이봄샘 기능부전의 치료는 마이봄의 질적인 향상과 마이봄샘의 배출을 증가시키고, 염증을 감소시켜 수분층과 지방층의 균형을 정상화하여 안정성을 회복하는 것을 목표로 하고 있다. 본 발명의 목적상 상기 PKA 세포신호전달체계 활성화제는 마이봄샘의 기능, 예를 들면 지질을 생성하는 과정을 촉진하여 균형된 지방층을 가질 수 있도록 함으로써 안구건조증을 매우 효과적으로 예방, 개선 또는 치료할 수 있다.
본 발명의 상기 "예방"은 질병 또는 병증의 발병을 억제하거나 지연시키는 모든 행위를 의미한다. 본 발명의 목적상 상기 조성물은 마이봄샘의 지질 생성을 촉진하여 안구건조증 또는 안구건조증 연관 안과 질환의 발병 시기를 지연시키거나, 발병을 억제하는 것을 의미한다.
본 발명의 상기 "치료"는 질병 또는 병증의 진행을 지연, 중단 또는 역전시키는 모든 행위를 의미하는 것으로서, 본 발명의 목적상 상기 조성물은 마이봄샘의 지질 생성을 촉진하여 안구건조증 또는 안구건조증 연관 안과 질환의 진행을 중단, 경감, 완화 또는 없애거나 역전시키는 것을 의미한다.
본 발명의 상기 약학 조성물은 캡슐, 정제, 과립, 주사제, 연고제, 분말 또는 음료 형태임을 특징으로 할 수 있으며, 상기 약학 조성물은 인간을 대상으로 하는 것을 특징으로 할 수 있다.
본 발명의 상기 약학 조성물은 이들로 한정되는 것은 아니지만, 각각 통상의 방법에 따라 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사 용액의 형태로 제형화하여 사용될 수 있다.
본 발명의 상기 약학 조성물은 약학적으로 허용 가능한 담체를 포함할 수 있다. 상기 약학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있고, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다.
본 발명의 상기 약학 조성물의 제형은 상술한 바와 같은 약학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(Elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형화 할 수 있다.
본 발명의 상기 조성물은 현탁액, 시럽제, 에멀젼, 리포좀, 산제, 분말제, 과립제, 정제, 서방형 제제, 점안제(eye drop), 캡슐제, 콘택트렌즈 세정제 및 콘택트렌즈 윤활제로 구성된 군에서 선택되는 어느 하나의 제형으로 제조될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명의 상기 약학 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다. 본 발명에서 상기 "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 설하, 직장, 병소내, 안구(예를 들어 점안 등) 및 두개골내 주사 또는 주입 기술을 포함한다.
본 발명의 상기 약학 조성물은 사용된 특정 화합물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여 시간, 투여 경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약무 형태, 투여 경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 50 mg/kg 또는 0.001 내지 50 mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 다른 구현 예에서는 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 건강기능식품을 제공한다.
본 발명의 상기 건강기능식품은 PKA 세포신호전달체계 활성화제, 또는 이의 유도체를 유효성분으로 포함한다.
본 발명의 상기 건강기능식품에서, 안구 건조증, PKA 세포신호전달체계 활성화제, 유도체, 화학식 1로 표시되는 화합물, 안구 건조증 연관 안과 질환, 예방 등은 앞서 약학 조성물에 기재된 바와 동일하여 명세서의 과도한 복잡성을 피하기 위해 그 기재를 생략한다.
본 발명의 상기 "건강기능식품"이란, "기능성 식품" 및 "건강 식품"의 의미를 모두 포함한다. 상기 기능성 식품은 특정보건용 식품(food for special health use, FoSHU)와 동일한 용어로, 영양 공급 외에도 생체조절기능이 효율적으로 나타나도록 가공된 의학, 의료효과가 높은 식품을 의미한다. 또한, 상기 건강 식품 은 일반식품에 비해 적극적인 건강유지나 증진 효과를 가지는 식품을 의미하고, 건강보조식품(health supplement food)은 건강 보조 목적의 식품을 의미한다. 경우에 따라, 기능성식품, 건강식품, 건강보조식품의 용어는 혼용되어 사용될 수 있다.
본 발명의 상기 건강기능식품은 안구건조증의 개선 및 회복에 유용한 효과를 얻기 위하여 정제, 캅셀, 분말, 과립, 액상, 환 등의 다양한 형태로 제조될 수 있다.
본 발명의 상기 건강기능식품은 영양 보조제, 식품 첨가제 및 사료 등의 형태로 제조될 수 있으며, 인간 또는 가축을 비롯한 동물을 취식 대상으로 할 수 있다.
본 발명의 상기 건강기능식품은 당업계에 공지된 통상적인 방법에 따라 다양한 형태로 제조할 수 있다. 일반 식품으로는 이에 한정되지 않지만 음료(알콜성 음료 포함), 과실 및 그의 가공식품(예: 과일 통조림, 병조림, 잼, 마아말레이드 등), 어류, 육류 및 그 가공식품(예: 햄, 소시지 콘비이프 등), 빵 류 및 면류(예: 우동, 메밀국수, 라면, 스파게티, 마카로니 등), 과즙, 각종 드링크, 쿠키, 엿, 유제품(예: 버터, 치즈 등), 식용식물 유지, 마아가린, 식물성 단백질, 레토르트 식품, 냉동식품, 각종 조미료(예: 된장, 간장, 소스 등) 등에 안토시아닌 올리고머를 첨가하여 제조할 수 있다.
본 발명의 상기 건강기능식품은 영양보조제로 제조되는 경우 캡슐, 타블렛, 환 등의 제형에 안토시아닌 올리고머가 첨가될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 건강기능식품은 차, 주스 및 드링크의 형태로 제조하여 음용할 수 있도록 액상화, 과립화, 캡슐화 및 분말화하여 제조될 수 있다. 여기서, 상기 건강기능식품이 드링크 형태로 제조되는 경우, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드; 말토스, 수크로스와 같은 디사카라이드; 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드; 자일리톨, 소르비톨, 에리트리톨 등의 당알콜일 수 있다. 감미제는 타우마틴, 스테비아 추출물과 같은 천연 감미제; 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 mL 당 일반적으로 약 0.01 ~ 0.04 g, 바람직하게는 약 0.02 ~ 0.03 g이다.
본 발명의 상기 건강기능식품은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산, 펙트산의 염, 알긴산, 알긴산의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 또는 탄산화제 등을 함유할 수 있다. 그 밖에도 드링크 형태로 제조되는 경우에는 과일주스, 과일주스 음료, 또는 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 첨가제의 성분은 독립적으로 또는 혼합하여 사용할 수 있고, 그 비율은 크게 중요하진 않지만 본 발명의 식품조성물의 유효성분 100 중량부당 0.01 ~ 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
본 발명의 상기 "개선"이란, 질병 또는 병증 상태를 호전 또는 이롭게 변경하는 모든 행위를 의미하는 것으로, 본 발명의 목적상 상기 조성물은 마이봄샘의 지질 생성을 촉진하여 안구건조증 또는 안구건조증 연관 안과 질환 증상을 호전시키는 것을 의미한다.
본 발명의 또 다른 구현 예에서는 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 점안제를 제공한다.
본 발명의 상기 점안제는 PKA 세포신호전달체계 활성화제, 또는 이의 유도체를 유효성분으로 포함한다.
본 발명의 상기 점안제에서, 안구 건조증, PKA 세포신호전달체계 활성화제, 유도체, 화학식 1로 표시되는 화합물, 안구 건조증 연관 안과 질환, 예방, 개선 등은 앞서 약학 조성물 및 건강기능식품에 기재된 바와 동일하여 명세서의 과도한 복잡성을 피하기 위해 그 기재를 생략한다.
본 발명의 상기 점안제는 점증제를 추가로 포함할 수 있다. 상기 점증제는 당업계에서 일반적으로 사용되는 의약용 또는 식용의 점증제를 의미하며, 키토산, 히알루로닉산, 히드록시프로필메칠셀룰로즈(HPMC), 히드록시프로필셀룰로즈(HPC), 히드록시에칠셀룰로즈(HEC), 폴리비닐피롤리돈(PVP), 카르복시메칠셀룰로즈(CMC), 카보머(carbomer), 글리세린(glycerin), 폴리에칠렌옥사이드(PEO), 히알루론산 (Hyaluronic acid) 등 일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 점안제는 수용액을 포함하는 담체를 추가적으로 포함하며, 액상인 것을 특징으로 한다. 상기 수용액을 포함하는 담체는 증류수, 인산완충액, 균형염 용액 및 생리식염수로 이루어진 군에서 선택된 하나 이상의 약학적으로 허용되는 담체인 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 약학적으로 허용가능한 담체는 생물체를 상당히 자극하지 않고 투여 유효성분의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 말한다. 상기 약제학적으로 허용가능한 담체의 예로는 등장화제, 완충제, 안정제, pH 조절제 및 용제 등이 있다. 상기 용제는 증류수, 인산완충액(phosphate buffered saline), 균형염 용액(balanced salt solution), 생리식염수(saline) 또는 이들의 혼합물 등을 포함할 수 있다. 이때 사용되는 함량은 제조하고자 하는 점안액의 총 용량에 필요한 양에 맞추어 사용량을 조절할 필요가 있다. 즉, 점안액의 경우에는 전체 점안액 중 상기 점안제 외에 용제로 사용량을 조절한다.
본 발명의 상기 점안제는 국소 투여 또는 점안 투여되는 것을 특징으로 한다.
본 발명의 또 다른 구현 예에서는 PKA(Protein kinase A) 세포신호전달체계 활성화제를 환자에게 투여하는 것을 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 치료방법을 제공한다.
본 발명의 상기 PKA 세포신호전달체계 활성화제는 하기 화학식 1로 표시되는 약물, 또는 이의 유도체인 것인, 안구건조증 또는 안구건조증 연관 안과 질환의 치료방법을 제공한다.

도 5 및 6은 본 발명의 일 실시예에 따른 HMGEC 세포주에서 포스콜린을 처리한 지 0일(D0), 2일(D2) 및 4일(D4)에, PPARγ(peroxisome proliferator-activated receptor gamma) 및 SREBP-1(Sterol regulatory element-binding protein-1) 단백질의 발현 수준을 면역 블롯팅을 통해 확인한 결과(도 5)와, 이를 정량화하여 그래프(도 6)로 나타낸 것이다.
도 7은 본 발명의 일 실시예에 따른 HMGEC 세포주에서 포스콜린을 처리한 지 0일(D0), 2일(D2) 및 4일(D4)에, PPARγ 및 SREBP-1 단백질을 암호화하는 mRNA의 발현 수준을 정량 실시간 중합효소 연쇄 반응 분석을 통해 확인한 결과를 나타낸 것이다.
본 발명은 PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 치료용 약학 조성물을 제공한다.
또한 본 발명은 PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 건강기능식품을 제공한다.
또한 본 발명은 PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 점안제를 제공한다.
이하, 본 발명을 하기의 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 의해 한정되는 것은 아니다.
실시예
[실험 방법 1]
세포 배양, 분화 및 약물 처리
선행문헌(Jun, I., et al., Interleukin-4 stimulates lipogenesis in meibocytes by activating the STAT6/PPARγ signaling pathway. Ocul Surf, 2020. 18(4): p. 575-582.)에 기재된 것과 동일하게, 인간 유래 마이봄샘 상피 세포 주 (Human meibomigan gland epithelial cell; HMGEC, 이하, 'HMGEC 세포주'라 함)를 케라티노사이트 혈청-프리 미디아 (Keratinocyte Serum-Free media, KSFM; Gibco, Grand Island, NY, USA)을 이용하여 배양하였다. 10%의 우태아혈청, 10 ng/ml의 상피 성장 인자(Epidermal growth factor, EGF; R&D System, Minneapolis, MN, USA)를 상기 미디아에 넣어 DMEM-F12 배양액으로 변경되도록 함으로써, 상기 세포의 분화를 유도하였다. 분화 후 0일(D0)은 세포의 분화를 위하여 배양액을 교체해 준 날로 정의하였으며, 분화를 위하여 4일(D4)동안 교체된 배양액에서 세포를 배양하였다.
상기 분화를 유도하는 과정과 동시에 포스콜린(Forskolin, Sigma, St. Louis, MO, USA)을 처리하여 PKA(Protein kinase A) 신호전달체계를 활성화시켰다. 이때, 세포 배양액 및 약물은 2일마다 신선한 것으로 교체하였다.
[실험 방법 2]
정량 실시간 중합효소 연쇄 반응 분석(Quantitative, real time polymerase chain reaction, qPCR)
12 웰 플레이트에 HMGEC 세포주를 분주한 뒤에 상기 실험 방법 1에 기재된 방법으로 세포를 배양하였다. Tri-RNA 시약을 이용하여 배양된 세포의 전체 RNA를 분리하고, EcoDryTM 프리믹스(TaKaRa, Shigan, Japan)를 이용하여 분리된 전체 RNA 1 ㎍을 주형으로 하는 cDNA를 합성하였다. 여기서, SYBR 그린 PCR 마스터 믹스(SYBR green PCR Master Mix, Applied Biosystems, Foster City, CA, USA)를 통해 ViiA7(Applied Biosystems)으로 상대적인 mRNA의 양을 측정하였다. 측정 대상 유전자의 발현 수준은 하우스키핑(Housekeeping) 유전자에 해당하는 GAPDH를 기준으로 보정하여 Ct 값의 차이(deltaCt, dCt)를 계산하였다. 측정 대상 유전자의 발현 수준은 실험군과 대조군 사이의 dCt 값의 차이(delta delta Ct, ddCt)를 통해 확인하였다. 이와 같은 과정은 적어도 3번 이상 반복하여 결과를 도출하였다.
[실험 방법 3]
면역 블랏팅(Immune Blotting)
12 웰 플레이트에 HMGEC 세포주를 분주한 뒤에 상기 실험 방법 1에 기재된 방법으로 세포를 배양하였다. 이렇게 배양된 세포를 4 ℃의 PBS(Phosphate buffered saline)을 이용하여 두 번 세척하였다. 1 μM PMSF(Roche, Basel, Switzerland)를 함유한 리파 완충액(RIPA buffer, Biosesang, Seongnam, South Korea)을 이용하여 상기 세척된 세포를 용해시켜 세포 용해액을 얻었다. BCA 단백질 분석 시약(Thermo Fisher Scientific, Waltham, MA, USA)을 이용하여 상기 세포 용해액을 정량화 하였다. 그 뒤에 동일한 양의 세포 용해액을 SDS(Sodium dodecyl sulfate) 샘플 완충액에 혼합하고, 10% SDS-PAGS 겔에 로딩하고 전기영동을 수행하였다. 전기영동을 통해 겔 내에서 분리된 단백질을 PVDF 멤브레인에 이동시키고, 목적하는 단백질에 특이적으로 결합하는 일차 항체 및 상기 일차 항체에 특이적으로 결합하는 이차 항체와 반응시키는 과정을 통해 목적하는 단백질을 검출하였다. 이와 같은 과정은 적어도 3번 이상 반복하여 결과를 도출하였다.
[실험 방법 4]
아디포어(Adipored) 분석
HMGEC 세포주를 96웰 플레이트에 상기 실험 방법 1에 기재된 방법대로 배양한 뒤, PBS를 이용하여 2번 세척하였다. 그런 다음, AdipoRedTM (Lonza) 시약을 넣어 상기 세포주를 염색하였다. Fluorimeter(Varioskan Flash, Thermo Fisher Scientific)를 이용하여 상기 염색된 세포주를 485 nm의 파장의 빛으로 자극하고, 572 nm의 파장에서 배출되는 형광을 측정하였다. 이와 같은 과정은 적어도 3번 이상 반복하여 결과를 도출하였다.
[실험 방법 5]
공초점 형광면역세포화학 염색
Nunc쪠 Lab-Tek쪠 II Chamber Slide쪠 system(Thermo Fisher Scientific)에서 상기 실험 방법 1의 과정을 통해 분화를 유도하는 HMGEC 세포주를 4% 파라포름알데히드(paraformaldehyde, Biosesan)를 이용하여 상온에서 30분 동안 고정하였다. 이 후 세포 내 지질 방울(droplet)을 확인하기 위해 리피드톡스(LipidTOX, Invitrogen) 시약을 30분간 상온에서 처리하여 염색하고, 슬라이드유리에 마운팅 한 뒤에, Zeiss LSM 700 공초점현미경 (Carl Zeiss, Oberkochen, Germany)으로 형광을 관찰하였다.
[실험 결과 1]
HMGEC 세포주에서 포스콜린에 의한 PKA(Protein kinase A) 신호전달체계 활성화
상기 실험 방법 1에 기재된 바에 의해 분화를 유도하는 HMGEC 세포주에 포스콜린을 처리하고, 면역 블롯팅을 통해 5분, 15분, 30분, 60 분 및 120분에 PKA 신호전달체계 하위 신호인 CREB 단백질의 인산화를 확인하여, 그 결과를 도 1에 나타내고, 이를 정량화하여 그래프로 도 2에 나타내었다.
도 1 및 2에서 보는 바와 같이, 대조군과 비교하여 포스콜린을 처리한지 15분부터 CREB 단백질의 인산화가 통계적으로 유의하게 증가되었으며, CREB 단백질의 인산화는 60분까지 지속되는 것을 확인하였다.
상기 결과를 통해, HMGEC 세포주에서 PKA 신호전달체계가 세포 내에서 생리적인 역할을 담당하고 있음을 알 수 있다.
[실험 결과 2]
포스콜린에 의한 HMGEC 세포주의 지질 생성 촉진 여부 확인
분화를 유도하는 HMGEC 세포주에 포스콜린을 4일 동안 처리하고, 2일(D2) 및 4일(D4)에 상기 실험 방법 4 및 5에 기재된 방법에 의해 지질 생성의 정도를 측정하여, 그 결과를 도 3 및 4에 나타내었다.
도 3에서 보는 바와 같이, 포스콜린을 처리한지 2일(D2) 및 4일(D4)에 모두에서 대조군과 비교하여 세포 내 지질의 양이 모두 현저하게 증가되었다.
또한, 도 4에서 보는 바와 같이, 대조군과 비교하여 포스콜린을 처리한지 4일(D4)에 대조군과 비교하여 생성된 지질의 양이 2배 이상 증가되었다.
상기 결과를 통해 PKA 신호전달체계를 활성화시키는 약물을 사용하는 경우 HMGEC 세포주에서 지질의 생성이 현저하게 증가되는 것을 알 수 있다.
[실험 결과 3]
포스콜린에 의한 HMGEC 세포주의 지질 생성 관련 중요 단백질의 발현 수준 변화 확인
분화를 유도하는 HMGEC 세포주에 포스콜린을 처리하고, 0일(D0), 2일(D2) 및 4일(D4)에 지질 생성 관련 중요 단백질인 PPARγ(peroxisome proliferator-activated receptor gamma) 및 SREBP-1(Sterol regulatory element-binding protein-1) 단백질 및 이를 암호하는 mRNA의 발현 수준을 상기 실험 방법 2 및 3에 기재된 방법에 의해 확인하여, 그 결과를 도 5 내지 7에 나타내었다.
도 5 내지 7에서 보는 바와 같이, HMGEC 세포주에 포스콜린을 처리하였을 때, 대조군과 비교하여 2일(D2) 및 4일(D4)에서 PPARγ 및 SREBP-1 단백질 및 이를 암호화하는 mRNA의 발현 수준이 증가되었다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
본 발명에서는 PKA(protein kinase A) 세포신호전달체계를 활성화하여 마이봄샘 기능 장애에서 발생될 수 있는 지질 합성 저하를 효과적으로 개선하여, 마이봄샘에서 분비된 지질이 안구 표면 수분 증발을 줄여 안구건조증 또는 안구건조증 연관 안과 질환을 예방, 개선 또는 치료할 수 있으므로 산업상 이용가능성이 인정된다.
Claims (15)
- PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서,상기 안구건조증은 마이봄샘 기능장애로 유발된 것인, 약학 조성물.
- 제1항에 있어서,상기 안구건조증 연관 안과 질환은 건성 각결막염, 건성으로 인한 각막 궤양, 건성안으로 인한 안건염 및 건성으로 인한 눈의 충혈로 이루어진 군으로부터 선택되는 어느 하나 이상인 것인, 약학 조성물.
- 제1항에 있어서, 상기 조성물은 현탁액, 시럽제, 에멀젼, 리포좀, 산제, 분말제, 과립제, 정제, 서방형 제제, 점안제(eye drop), 캡슐제, 콘택트렌즈 세정제 및 콘택트렌즈 윤활제로 구성된 군에서 선택되는 어느 하나의 제형인 것인, 약학 조성물.
- PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 건강기능식품.
- 제6항에 있어서,상기 안구건조증은 마이봄샘 기능장애로 유발된 것인, 건강기능식품.
- PKA(Protein kinase A) 세포신호전달체계 활성화제를 유효성분으로 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 예방 또는 개선용 점안제.
- 제9항에 있어서,상기 안구건조증은 마이봄샘 기능장애로 유발된 것인, 점안제.
- PKA(Protein kinase A) 세포신호전달체계 활성화제를 환자에게 투여하는 것을 포함하는, 안구건조증 또는 안구건조증 연관 안과 질환의 치료방법.
- 제12항에 있어서,상기 안구건조증은 마이봄샘 기능장애로 유발된 것인, 안구건조증 또는 안구건조증 연관 안과 질환의 치료방법.
- 제12항에 있어서,상기 안구건조증 연관 안과 질환은 건성 각결막염, 건성으로 인한 각막 궤양, 건성안으로 인한 안건염 및 건성으로 인한 눈의 충혈로 이루어진 군으로부터 선택되는 어느 하나 이상인 것인, 안구건조증 또는 안구건조증 연관 안과 질환의 치료방법.
