該連結構造は、カチオンの状態で、下記式(C1)~(C7)からなる群より選択されるカチオン性基を有し、前記カチオン性人工核酸は、他のヌクレオチドのリン酸基と前記カチオン性基との静電相互作用により会合しうることを特徴とする核酸アナログ。
該中空部分に医薬が包接されていることを特徴とする〔13〕に記載の核酸送達用構造体。
前記核酸アナログの前記カチオン性基のpKaよりも低く、前記核酸のリン酸のpKaよりも高いpH条件で前記核酸アナログと前記核酸とを静電相互作用により会合させて会合核酸構造を形成させる会合工程と、を有することを特徴とする核酸送達用構造体の製造方法。

以下、本発明の核酸アナログについて説明する。本発明の核酸アナログは、カチオン性人工核酸と、このカチオン性人工核酸に結合した親水性重合体と、から構成される。核酸アナログは、送達する対象となる核酸(以下、「送達対象核酸」という場合がある)を標的部位に送達するための担体として好ましく使用される。以下、核酸アナログについて詳細に説明する。
カチオン性人工核酸は、リボース及びデオキシリボースから選択される環構造に塩基が結合した構成単位と、2つの構成単位の間を連結する連結構造と、からなる骨格構造を有している。連結構造は、カチオンの状態で、下記式(C1)~(C7)からなる群より選択されるカチオン性基を有している。
親水性重合体は、核酸アナログの用途等に応じて、種々の重合体を使用することができる。特に、核酸を標的部位に送達するための担体として核酸アナログを使用する場合、親水性重合体としては、生体適合性を有するものが好ましい。また、核酸アナログがカチオン性であることから、親水性重合体としては、核酸アナログと電気的に相互作用(吸引、反発)しにくい中性のものが好ましい。
また、A1は、核酸塩基や、リン酸エステル誘導体(-PO(OH)-,-PS(OH)-,PO(SH)-)をモノマー単位とした重合体として用いることもできる。モノマー単位のA1のエチレングリコールの繰り返し回数qは1~20、好ましくは3~10であり、モノマーの構造は同一でも異なっていてもよく、モノマー単位の重合度は1~10、好ましくは2~10、さらに好ましくは2~8である。そのようなリン酸エステル誘導体モノマー単位としては、エチレングリコールリン酸エステル誘導体単位、ジエチレングリコールリン酸エステル誘導体単位、トレエチレングリコールリン酸エステル誘導体単位、ヘキサエチレングリコールリン酸エステル誘導体単位などを挙げることができる。これらの誘導体単位は、さらに重合体のモノマーとして用いることもできる。リン酸エステル誘導体単位の重合度は、人工核酸の大きさにもよるが、人工核酸の塩基数が10~30であれば、1~10、好ましくは2~10、さらに好ましくは2~8である。モノマー単位はトリエチレングリコールリン酸エステル誘導体、ヘキサエチレングリコールリン酸エステル誘導体であることが最も好ましい上記で述べた親水性重合体のモノマーの種類や重合度、特にエチレングリコールリン酸誘導体モノマーの重合度は、後述する構造体の形成がしやすさ、生体内で異物として認識されにくさ、分解安定性や血中滞留性の低くなる、などの観点から、選択することができる。
本発明の核酸アナログは、送達対象核酸を標的部位に送達するための担体として特に好ましく用いることができる。図1は、核酸アナログを担体として使用した場合について説明する模式図である。この図に示すように、核酸アナログは、一次構造として、カチオン性基と親水性重合体とを備えており、天然核酸などの送達対象核酸はアニオン性を有している(図の「合成」参照)。このアニオン性の送達対象核酸と核酸アナログのカチオン性基との間で静電相互作用により会合した会合構造が形成され、担体である核酸アナログと送達対象核酸とで構成される複合体(イオンコンプレックス)となる(図の「ナノ粒子形成」の「二本鎖形成」参照)。

(1)核酸アナログの製造方法
次に、核酸アナログの製造方法について説明する。核酸アナログは、種々の方法で製造することができるが、カチオン性人工核酸と親水性重合体とを個別に合成し、これらを結合する方法を挙げることができる。すなわち、核酸アナログの製造方法は、カチオン性人工核酸を合成するカチオン性核酸合成工程と、親水性重合体を合成する親水性重合体工程と、カチオン性人工核酸と親水性重合体とを結合する結合工程とを有する。
カチオン性人工核酸は、2段階の反応で合成することができる(2段階合成法)。2段階合成法としては、主に3種類を挙げることができる。以下、順次説明する。
この方法では、核酸自動合成により核酸の硫黄化(S化)を行い、これに親水性重合体ホスホロアミダイト(例えば、エチレングリコールホスホロアミダイト)を連結させてオリゴPS体―親水性重合体を合成したのち、更にBr化合物を反応させて核酸にカチオン性基を導入する(後述する実施例のTEG-PSオリゴ、HEG-PSオリゴの系)。
カチオン性人工核酸は、ヌクレオチド骨格の場合、後述する実施例に記載のように、連結構造にチオリン酸エステルを導入する工程と、カチオン性基を有するブロモ化合物とチオリン酸エステルとを反応させて連結構造にカチオン性基を導入する工程を含む方法で合成することができる。
(a-1-2)チオリン酸エステルの導入
チオリン酸エステルは、公知のホスホロアミダイト法によって合成することができる。ホスホロアミダイト法の概要としては、5’が4,4’-ジメトキシトリチル(DMTr)基で保護されたヌクレオシド又はヌクレオチドを固相に担持させる(担持工程)。つぎに、ジクロロ酢酸などの脱保護試薬でDMTr基を脱保護し(脱保護工程)、4,5-ジシアノイミダゾールなどの活性化剤の存在下でホスホロアミダイトヌクレオチドとカップリングさせる(カップリング工程)。その後、(N,N-ジメチルアミノメチリデン)アミノ]-3H-1,2,4-ジチアゾリン-3-チオン(DDTT)などの硫化剤によって亜リン酸エステルをチオリン酸エステルに変換する(硫化工程)。あるいはヨウ素、ピリジンなどを含む酸化剤によって亜リン酸エステルをリン酸ジエステルに変換する(酸化工程)。この工程を繰り返すことで、連結構造にチオリン酸エステルを含む合成核酸を製造することができる。ホスホロアミダイトの塩基の種類を変えることで、所望の配列を有する合成核酸を製造することができる。なお、酸化工程に替えて硫化工程を行うことで、ヌクレオチド骨格を構成する連結構造の所望の位置にのみチオリン酸エステルを導入することも可能である。
次に、得られた合成核酸に、アミン又はアンモニウム基を有するブロモ化合物を反応させる。ブロモ化合物としては、例えば、3-ブロモ-1-プロピルアミン臭化水素酸塩、2-ブロモ-N,N-ジエチルエチルアミン臭化水素酸塩、(3-ブロモプロピル)トリメチルアンモニウムブロミドなどを例示することができる。合成核酸とブロモ化合物との反応は、リン酸緩衝液などで行うことができ、反応条件は適宜設定できるが、例えばpHは5~7の範囲内、反応温度は30~60℃、反応時間10~50時間で行うことができる。
この方法では、核酸自動合成により核酸のボラノホスフェート化(B化)を行い、これに親水性重合体ホスホロアミダイト(例えば、エチレングリコールホスホロアミダイト)を連結させてオリゴB化―親水性重合体を合成(図2)したのち、更にヨウ素酸化によってアミノ基化合物を反応させてカチオン性基を導入する(図3の戦略2)。この方法では、後述のカチオンホスホロアミダイトを用いることで1つのリン酸基に2つのカチオン性基を導入することも可能である(ダブルカチオン導入)。
この方法では、核酸自動合成によるカチオン性人工核酸を合成ののち、親水部をクリック反応で導入する(後述する実施例のPEG-PMOの系)。すなわち、骨格にカチオン性基を有するカチオン性人工核酸を合成したのち、これにアジドポリエチレングリコールなどの親水性アジド化合物をクリック反応により連結させる方法である。
先に説明した方法は、チオリン酸エステルを含む合成核酸を製造し、これにブロモ化合物を反応させる2段階の合成方法であったが、1段階でカチオン性人工核酸を合成することもできる。図2はこの方法のスキームを示している。この方法の概略としては、カチオンホスホロアミダイトとエチレングリコールホスホロアミダイトの核酸自動合成によるカチオン-親水性重合体の1段階合成である(図2、図3の戦略1)。この方法も、基本的には2段階合成法で示したホスホロアミダイト法と類似したスキームであるが、一部に相違がある。
次に、核酸送達用構造体の製造方法について説明する。核酸送達用構造体は、送達対象核酸と核酸アナログとを会合させて複合体を形成させる(会合工程)。核酸アナログのカチオン性基と送達対象核酸のリン酸基との間に相補性がある場合は、アニーリングによって両者を結合して二本鎖を形成させる。アニーリングの際の核酸アナログと送達対象核酸の比率としては特に制限はないが、核酸アナログ:送達対象核酸=1:1~1:10の範囲内が好ましい。アニーリングは所定温度への昇温と、その後の降温によって行う。アニーリングとしては、80℃以上への昇温が好ましく、90℃以上への昇温より好ましい。また、昇温した温度の保持時間としては、5分以上が好ましく、10分以上がより好ましい。その後、50℃以下、好ましくは30℃以下に降温し、10分以上、好ましくは30分以上その温度を保持する。
本発明の核酸送達方法は、上記の核酸送達用構造体を投与して標的部位内に核酸送達用構造体を取り込ませる投与工程と、標的部位内で核酸を放出させる放出工程と、を有する。投与工程では、送達対象核酸と結合した状態の上記の構造体を、人やその他の哺乳動物などに投与する。構造体を含む薬剤は、血液中に投与することが好ましいが、治療対象となる疾患や親水性重合体の種類などに応じて適宜決定することができる。投与された構造体は、細胞が標的部位である場合はエンドソーム経由で細胞質に取り込まれる。放出工程では、標的部位である細胞内に取り込まれた構造体から送達対象核酸が放出される。カチオン性基が四級アンモニウムなどpH応答性の高い構造体では、エンドソーム内の酸性環境下で構造崩壊して送達対象核酸を放出しやすくなる。また、上記のように、構造体の中空部分に低分子の医薬を包接させて評定部位内で放出させることも可能である。このような方法で、標的部位に送達対象核酸や低分子医薬を送達することができる。
(1―1)オリゴヌクレオチド合成
オリゴヌクレオチド(SEQID1-4,8,12-15,19-21,25,33,37)は、Gene Design Inc.(Osaka,Japan)より購入した。なお、親水性重合体が結合したオリゴヌクレオチドは、エチレングリコールのホスホロアミダイト体を核酸配列合成と同様にGene Desing Inc.にて自動核酸合成機で連結した(連結部位はホスホジエステル結合)。
モルフォリノ核酸(SEQID29)は、Gene Tools,LCC(Philomath, Pregon,US)より購入した。オリゴヌクレオチド(SEQID30-32,34-36,38-40)は、それぞれSEQID29、24、28を出発物質とし、アジドポリエチレングリコール化合物(10当量)を用いて、硫酸銅(II)(10当量)とアスコルビン酸ナトリウム(10当量)を加え、PBS(pH7.4)中で、室温で15分反応し親水性重合体を連結した。反応後、HPLC精製と脱塩、濃縮を経て生成物を得た。アジド化合物のPoly(ethylene glycol)methyl ether azide, average Mn 1000(10K), 5000(50K), 10000(100K)は、Merck(Darmstadt,Germany)より購入した。
合成したオリゴヌクレオチドの配列を表2及び表3に示す。SEQID1-2はマイクロRNA-143のアンチセンス鎖(AS-1,2)、SEQID3-11はマイクロRNA-143のセンス鎖(S-1-9)、SEQID12-13はレニーラルシフェラーゼに対するsiRNAのアンチセンス鎖、SEQID14-18レニーラルシフェラーゼに対するsiRNAのセンス鎖を示す(以下、アンチセンス鎖を「AS」、センス鎖を「S」と略す場合がある。)。この表において、「N」(大文字)はRNAを、「dN」はDNAを、「Nf」は2’-FRNAを、「Nm」は2’-OMeRNAを、「X」は-(OCH2CH2)3O-、「Y」は-(OCH2CH2)6O-を示す。「*」は-P(O)OH-を、「^」は-P(S)OH-を、「1」は-P(S-(CH2)3-NH2)OH-を、「2」は-P(S-(CH2)2-N(CH2CH3)2)OH-を、「3」は-P(-S(CH2)3-N(CH3)3)OH-を、「pmo」はモルフォリノ核酸を、それぞれ意味する(これらの一部については下記の化学式を参照。なお、「^」については2つの構造が共役した構造を示す。)。「E」はエチニル基を、「L」はリガンド化合物(グルコース、ガラクトース、フコース、アミノ糖)を、「S」はジスルフィドを示す。

(1)アニーリング
各ヌクレオチドを滅菌水に溶解した後、AS:S=1:1-10の重量比で混合した。その後、98℃で15分間、25℃で50分間、45℃で50分間インキュベートし、アニーリング混合物(下記表のAN1-24)を得た。
得られたアニーリング混合物は、20wt%アクリルアミドゲル電気泳動(PAGE)、透過型電子顕微鏡(TEM: Transmission electron microscope)、原子間力顕微鏡(AFM: Atomic force microscope)、動的光散乱法(DLS: Dynamic light scattering)、円偏光二色性(CD: Circular dichroism)などを用いて物性を評価した。
10μMに調整したアニーリング混合物は、7.5又は20wt%アクリルアミドゲルに5-10pmolアプライし、25-40分間泳動した。泳動後、Alliance Q9(UVITEC, Cambridge, UK)でCy3の蛍光を検出した。撮影後、SYBrGoldで5分間核酸を染色し、SYBR Gold Nucleic Acid Gel Stain(Thermo Fisher Scientific, Waltham, MA)の蛍光を検出した。なお、マーカーは100bpDNA Ladder(Takara Bio Inc., Shiga, Japan)と20bpDNA Ladder(Takara Bio Inc., Shiga, Japan)をサンプルの両端にアプライした。
グリッド基盤上に10μMアニーリング混合物を5μL滴下し、自然乾燥させた後、リンタングステン酸で染色し、JEM-2010(JEOL Ltd.,Tokyo, Japan)で観察した。
マイカ基盤上に1.5μMアニーリング混合物を5μL滴下し、一晩自然乾燥させた後、10分間真空乾燥させて、Nanocute(Hitachi High-Tech Corp., Tokyo, Japan)で観察した。
DLSは、DelsaMax PRO(Beckman Coulter, Brea, CA, USA)を用いて37℃で測定した。アニーリング混合物の濃度は20-50μMとした。自己相関関数より流体力学的半径(RH)を算出した。pHによる構造変化は1N HClをpH5になるように調整した。血清耐性評価は、50%FBS中で、37℃でインキュベートして経時的な構造変化を評価した。
CDは、円二色性分散計(J-820KS; JASCO Corporation, Tokyo, Japan)を用いて波長240-320nm,温度20-80℃の範囲で光路長1cmの石英セルを用いて37℃で測定を行った。アニーリング混合物の濃度は1.5μMとした。
図4(A)はAN4(S-4(実施例2)とAS-1)、図の(B)はAN5(S-5(実施例3)とAS-1)のアニーリング混合物の泳動結果を示した。図の(A)の左図のレーン5から7ではCy3(シアニン色素)に由来するSEQID1’のバンドが薄くなった。ここで、SEQID1’は、SEQID1とSEQID2を9:1で混合したものであり、Cy3はSEQID2に由来する。これはSEQID6の混合比が1から5と増えると、SEQID1’が構造形成した可能性が示唆された。この現象は、Cy3検出後にRNA染色後の右図のレーン5から7も同様の傾向が観察された。図の(B)の左図のレーン5から7はCy3に由来するSEQID1’のバンドが検出されなかった。これは、SEQID7のアニーリング混合ではSEQID7の混合比が1から10の増加に関係なく、1の時点からの構造形成が示唆された。図の(C)は、AN4をTEMで観察した結果で、直径60nm程度の構造体が観察された。図の(D)は、AN5をAFMで観察した結果で、直径75nm程度の構造体が観察された。図の(E、F)より、AN4とAF5の流体力学的半径(RH)はそれぞれ、35nmと45nmだった。これらの結果より、アニーリング混合物はナノ構造体を形成することが明らかとなった。他のアニーリング混合物のRHを下記の表に示す。

図5に、構造体のpH応答による構造変化と構造体の血清中での安定性を評価した結果を示す。図の(A、B)より、pH7の条件下では、AN4とAN5ともに、AN2に観測されたA型らせん構造に特徴的な260nmの正のピークは長波長側にシフトし、210nmの激しいネガティブの落ち込みは観測されなかった。pHを5に変化させた時、AN4は変化が見られなかったのに対し、AN5では260nmの正のピークの強度が低下した。この時のRHを測定した結果を図の(C、D)に示す。AN4は、RHが35nmから85nmと増大した。一方、AN5では、pH5の時に、自己相関関数の強度が大幅に低下し、RHが算出できなかった。これらの結果から、AN4は、カチオン性基が三級アミンであるが、pHが7から5に変化することで、構造体のキラリティは大きく変化しないが、RHが2倍程度変化することが明らかとなった。AN5は、カチオン性基が四級アンモニウムであるが、pHが7から5に変化すると、構造体のキラリティもRHも大きく変化し、特に、RHが算出できなかったことからpH低下に伴い構造体が崩壊することが示唆された。図の(E)では、AN4の経時的な血清耐性をRHから評価した。24時間と、48時間において血清とインキュベートする前のRHと比較して変化は見られず、48時間と長時間において血清耐性を示すことが明らかとなった。
図6では、構造体による核酸送達の評価として、DLD-1細胞株を用いたレニーラルシフェラーゼ(RL)活性阻害評価(図の(A))と、マイクロRNA-143の送達評価(図の(B、C))した結果を示す。AN12(S-12(実施例7)とAS-3)、AN13(S-13(実施例8)とAS-3)、AN14(S-14(実施例9)とAS-3)は、添加濃度に依存してRLの発現阻害の傾向が評価され、構造体を形成するレニーラルシフェラーゼ配列による阻害活性が確認された(図の(A))。図の(B)では、AN4、AN5が添加濃度依存的にDLD-1の生存率を大きく低下させた。DLD-1からタンパクを抽出し、マイクロRNA-143の標的タンパクであるK-RASの存在を評価した(図の(C))。その結果、AN4やAN5でK-RASの発現が抑えられていることが明らかとなり、AN4やAN5は特にマイクロRNA-143を送達できることが示された。他のアニーリング混合物の結果を下記の表に示す。
それぞれの構造体のDLD-1細胞への取り込みは、共焦点レーザー顕微鏡で観察された。図7(C、D、E、G)はRNAに由来するCy3の赤色蛍光像が観察され、リソソームの緑色像とは独立して存在することが重ね合わせ像より示された。特に図の(D)のAN4は強い細胞取り込みが観察された。図の(I)はAN4の取り込みを120倍率で観察した像を示す。結果より構造体は細胞核やリソソームと交わることなく細胞質に取り込まれていることが推測された。
図8は、モルフォリノ核酸を用いたアニーリング混合物(AN28(S-25とAS-7)、AN29(S-28とAS-7))のpH7と4における電気泳動の結果を示す。泳動後、ゲルをRNA染色し、撮影した。電気泳動の条件は、7.5wt% acrylamide gel、Loading Volume:10pmol/well、Time:40min、Dye:Sybrgold、Detection:UV→Ebtrfillterである。レーン1と4は、アニーリング前のSEQID34とSEQID38である。それぞれSEQID30とアニーリングし、pH7とpH4に調整した。その結果から、AN28とAN29はpHが7から4に変化するとバンドが高分子側へシフトした。DLSによる粒子系の測定より、AN29(pH7)は2nm(レーン5)、AN29(pH4)は150nm(レーン6)であった。モルフォリノ核酸を用いたアニーリング混合物はpH変化によって構造体を形成することが示唆された。
非相補鎖のアニーリング混合物(AN25-27)の泳動結果を図9(A)に示す。レーン3のAN25(S-17(実施例10)とAS-3)は、SEQID12(ルシフェラーゼのAS鎖)のバンドが薄くなり、ウェル部分に新たな蛍光が観察された。レーン4のAN26(S-18(実施例18)とAS-1)は、新たな位置にバンドが観察された。レーン5のAN27(S-19(実施例12)とAS-3)では非相補鎖の相互作用は特に観察されていない。続いて、共焦点顕微鏡でAN26を観察した(図の(B),(C))。その結果、約250nmの蛍光が観察され構造体が存在することが示唆された。AN26を用いて、MCF―7細胞株(ヒト乳腺がん)を用いたレニーラルシフェラーゼ(RL)活性阻害評価を行なった(図の(D))。AN26の添加濃度に依存してRLの発現が阻害された。この時、AN26はMCF―7に取り込まれることを観測している(図の(E))。
(1)細胞培養
ヒト大腸がん細胞株DLD-1(K-RAS mutant; G13D)とヒト乳腺細胞株MCF―7は、Japanese Collection Research Bioresouses Cell Bankより購入した。DLD-1細胞は、10%非働化FBS含有RPMI-1640(Waco Inc., Osaka, Japan)を用いて5%CO2、37℃の条件下で培養した。MCF7細胞は、10%非働化FBS、非必須アミノ酸、ピルビン酸ナトリウム含有MEMα(Waco Inc., Osaka, Japan)を用いて5%CO2、37℃の条件下で培養した。
DLD-1、MCF―7は、5×104cell/wellで24ウェルプレートに播種し、一晩培養した。その後、psiCheck2(Promega Corporation, Madison, WI, USA)100ngをTransfast(Waco Inc., Osaka, Japan)を用いて添加した。翌日、細胞を回収したのち、5×104cell/wellで96ウェルプレートに播種し、一晩培養した。次に、最終濃度が1―1000nMとなるように各アニーリング混合物を添加し、48時間培養した。その後、上清液を除去しプレートを-200℃で6時間凍結した。その後、レニーラルシフェラーゼの阻害活性をDual-Glo(登録商標) Luciferase Assay System(Promega Corporation, Madison, WI, USA)を用いて発光シグナルとして検出した。その結果を図6(A)、図9(D)、11に示す。
(1)構造体によるDLD-1、MCF―7増殖抑制効果
DLD-1、MCF―7は1×103cell/wellで96ウェルプレートに播種し、一晩培養した。その後、最終濃度が1―100nMになるように各アニーリング混合物を添加し、72時間培養した後、残存する生細胞を細胞増殖/細胞毒性アッセイキット(Cell counting WST-8; Waco Inc., Osaka, Japan)を用いて吸光シグナルとして検出した。なお、NT(non treatment)は、miRNAを導入していないコントロールで、AN2はリポフェクション試薬を使用していないコントロール(Naked)である。その結果を図6(B)、図12に示す。
(1)ウェスタンブロット解析
(a)タンパク質サンプル調整:DLD-1、MCF―7は1×105cell/wellで6ウェルプレートに播種し、一晩培養した。その後、最終濃度が50nMになるように各アニーリング混合物を添加し、48時間培養した後、スクレーパーで細胞を剥がし、15mLチューブに細胞懸濁液を回収した。遠心して細胞のペレットを得た後、タンパク抽出を行った。タンパク抽出液には、protein lysis buffer(10mM Tris-HCl、 0.1%SDS、 1%NP-40、 0.1%デオキシコール酸ナトリウム、 150mM NaCl、 1mM EDTA)に1% Protease inhibitor cocktail(Nakarai, Kyoto, Japan)、Phosphatase inhibitor cocktail I, II(Merck Millipore, Burlington, MA, US)、及び、IIIを混合して用いた。タンパク質抽出液に細胞のペレットを懸濁させ、20分間氷中に静置した。その後、15,000rpm、4℃、20分間遠心分離した。遠心分離した上清を回収し、タンパク質サンプルとした。タンパク質サンプルは、SDS buffer(62.5mM Tris-HCl、2%SDS、10%グリセロール、50mM DTT、0.01%ブロモフェノールブルー)と混和して20μg/μLに調整し、98℃で5分間煮沸処理した後、氷上で5分間静置した。
電気泳動には、イージーセパレーター(Wako Inc., Osaka, Japan)及びSuper Sep Ace(Wako Inc., Osaka, Japan)を用いた。泳動後、ゲルをBlotting buffer(25mM Tris、 0.2M グリシン、20%エタノール)に5分間浸した。PVDFメンブレン(Perkin Elmer Life Sciences)はメタノールに10分間浸し、超純水に10分間浸した。その後、Blotting Bufferに5分間浸した。陽極側から、Blotting bufferに浸したろ紙、PVDFメンブレン、ゲル、ろ紙の順に重ね、15V、370mAで40分間転写した。転写後、0.1% Tween20含有50 mM Tris-HCl buffer(TBST)で洗浄し、Brocking buffer(PVDF;)に1時間浸した。TBSTで洗浄し、抗体希釈液(2% BSA、0.01%アジ化ナトリウム、TBST)で希釈した一次抗体に浸して4℃で一晩反応させた。TBSTで洗浄した後、二次抗体に浸して室温で1時間静置した。その後、TBSTで洗浄し、Luminata Forte Western HRP Substrate(WBLUF0500; Merck Millipore, Burlington, MA, US)で発光させた後に、GloMax Navigator(Promega Corporation, Madison, WI, USA)を用いて検出した。コントロールにはanti-GAPDH(Cell Signaling Technology, Danvers, MA, US)を使用した。K-RASに対する抗体はAbcam(Cambridge, UK)から購入した。その結果を図6(C)、図13に示す。
(1)DLD-1、MCF―7への構造の取り込み観察
DLD-1、MCF―7は、5×103cell/wellで96ウェルプレートに播種し、一晩培養した。その後、最終濃度が50nMになるように各アニーリング混合物を添加し、24-72時間培養した後、共焦点レーザー顕微鏡(FV3000; OLYMPUS, Tokyo, Japan)を用いて蛍光像を観察した。なお、核は、Cellstain Hoechst 33342(Wako Inc., Osaka, Japan)で染色し、リソソームはCellLight Lysosomes-GFP(Thermo Fisher Scientific, Waltham, MA)で染色した。
(1)オリゴヌクレオチド合成の実験データ
オリゴヌクレオチド(SEQID41,45)はGene Design Inc.(Osaka,Japan)より購入した。オリゴヌクレオチド(SEQID41(E=Glu)、SEQID45(E=Glu))は、それぞれSEQID41,45を出発物質とし、アジド化合物(10当量)を用いて、アスコルビン酸、銅イオンを添加しPBS(pH7.4)中で、30分間室温で反応した(反応の参考文献:DOI:https://doi.org/10.1039/C0CC04979D)。ここで「SEQID41(E=Glu)」は、SEQID41の親水性基にリガンドとしてグルコースが結合したリガンド人工核酸を意味する。反応に用いる溶液は全て脱気したものを用いた。反応後、高速液体クロマトグラフィーにて反応を確認し分取した。分取後、脱塩処理し凍結乾燥機で濃縮した。その後、ブロモ化合物(100-1000当量)を用いてPBS中45℃で24時間反応した。反応後、Float-A-Lyzer G2,CE,1ml,3.5-5KDa(Repligen,Waltham,MA,US)を用いて5-7日間蒸留水で透析した。透析後、凍結乾燥機で濃縮し、化合物(SEQID43(L=Glu)、SEQID47(L=Glu))を得た。ブロモ化合物(2-ブロモ-N,N-ジエチルエチルアミン臭化水素酸塩)はMerck(Darmstadt,Germany)より購入した。新たに合成したオリゴヌクレオチドの配列を表7に、アニーリング混合物を表8に示す。
実験に供したマウスは、広島大学の動物実験ガイドラインに基づいて飼育した。担がんマウスはマウス背中皮下へハンクス液に懸濁した大腸がん細胞(DLD-1細胞、8.0×105cells)の移植により作製した。固形がん生着後(移植7日後)より3日に1回(計4回)マウスの尾静脈からサンプルを投与した。投与したサンプルは、生理食塩水、AN1とAN4(容量100μL,核酸(ガイド鎖)375μg/kg)である。3日に1回マウスの腫瘍の長径(Lmm)と短径(Smm)をノギスで計測し、腫瘍堆積(Vmm3)を体積の式:V=(L×S2)×0.5より算出した。担がんマウスの腫瘍の体積変化より抗腫瘍効果を評価した。
担がんマウスはマウス背中皮下へハンクス液に懸濁した大腸がん細胞(DLD-1細胞8.0×105cells)の移植により作製した。固形がん生着後(移植7日後)より3日に1回(計4回)生理食塩水、AN1、AN4を尾静脈投与(容量100μL,核酸(ガイド鎖)375μg/kg)した。3日に1回マウスの腫瘍の長径(Lmm)と短径(Smm)をノギスで計測し、腫瘍堆積(Vmm3)を体積の式:V=(L×S2)×0.5より算出した。担がんマウスの腫瘍の体積変化より抗腫瘍効果を評価した。
担がんマウスにAN1、AN4を尾静脈投与(容量100μL,核酸(ガイド鎖)375μg/kg)し、尾静脈から経時的(1,2,4,8,24時間)に採血し、Cy3の蛍光強度を測定し、投与した核酸の血中における生物学的半減期を求めたところ約2.4時間であった。また、24時間後にそれぞれの臓器を回収し、Cy3の蛍光強度を測定したところ、肝臓においてはCy3の蛍光は観測されず、投与した核酸の肝臓への集積は検出されなかったものの、腫瘍細胞においてはCy3の蛍光が観察され集積していることが確認された。なお、LNPによりRNAsを投与した先行研究(Chen,S.,et al,Journal of Controlled Release,235,236-244(2016))によると、投与した核酸の血中における半減期は0.25時間であり、肝臓への集積率は91.4%。
In vitroで有効性が実証されたAN4をin vivoへ展開し、大腸がん担持マウスにおける抗腫瘍効果と組織動態を評価した。その結果を図14に示す。がん細胞を移植して7日後より、マウスの尾静脈にサンプルを投与し、腫瘍の体積変化を観察した。AN4を投与したグループは、生理食塩水やAN1(Naked143)を投与したグループと比較して腫瘍体積の増加が抑制され、抗腫瘍効果の誘導が観察された。動態評価では、担がんモデルマウスの尾静脈より蛍光標識したAN4を投与し24時間後に各臓器を回収し、蛍光強度を測定することで、臓器への取り込みを測定した。その結果、AN4は、腫瘍に高く取り込まれており、肝臓への蓄積はごくわずかであった。本結果は、核酸の運搬体として広く利用されている脂質ナノ粒子(LNP)の課題である肝臓への蓄積を回避する他の運搬技術として展開が期待される。
全身投与におけるDDSは、“パッシブ・ターゲティング”と“アクティブ・ターゲティング”がある。パッシブ・ターゲティングでは、腫瘍周辺の血管内皮の特徴である、Enhanced Permeability and Retention effect(EPR効果)の利用が知られている。アクティブ・ターゲティングは、標的細胞や組織だけに特異的送達することによって、より少ない投与量でがんに効率よく核酸を送達することが期待される。そこで、アクティブ・ターゲティングとしてリガンド導入構造体(AN30)について検討した。
Claims (18)
- カチオン性人工核酸と、該カチオン性人工核酸に結合した親水性重合体と、からなる核酸アナログであって、
前記カチオン性人工核酸は、リボース及びデオキシリボースから選択される環構造に塩基が結合した構成単位と、2つの前記構成単位の間を連結する連結構造と、からなる骨格構造を有し、
該連結構造は、カチオンの状態で、下記式(C1)~(C7)からなる群より選択されるカチオン性基を有し、前記カチオン性人工核酸は、他のヌクレオチドのリン酸基と前記カチオン性基との静電相互作用により会合しうることを特徴とする核酸アナログ。
- 前記カチオン性基のpKaが6~9の範囲であることを特徴とする請求項1に記載の核酸アナログ。
- 前記親水性重合体が、ポリエチレングリコール、ポリビニルアルコール、ポリグルタミン酸、ポリビニルピロリドン、ポリアクリルアミド、ポリエチレンイミン、ポリアルキルアクリレート、ポリオキサゾリン、ポリアクリルアミド、ポリ(カルボキシベタインメタクリレート)、ポリ(スルホベタインメタクリレート)、ポリ(2-メタアクリロイルオキシエチルホスホコリン)、ヒアルロン酸、キトサン及びデキストラン並びにこれらの誘導体から選択されることを特徴とする請求項1に記載の核酸アナログ。
- 送達対象である核酸を標的部位に送達するための担体であることを特徴とする請求項1に記載の核酸アナログ。
- 請求項8に記載の核酸アナログと送達対象である核酸とからなり該核酸を標的部位に送達するための核酸送達用構造体であって、前記カチオン性人工核酸と前記核酸とが静電相互作用により会合した会合構造を有することを特徴とする核酸送達用構造体。
- 複数の前記核酸送達用構造体が会合したナノスケールの構造体であることを特徴とする請求項9に記載の核酸送達用構造体。
- 前記会合構造が内側に、前記親水性重合体が外側に位置するベシクル又はミセルであることを特徴とする請求項10に記載の核酸送達用構造体。
- リガンドで修飾されていることを特徴とする請求項11に記載の核酸送達用構造体。
- 医薬を更に有することを特徴する請求項11に記載の核酸送達用構造体。
- 前記核酸送達用構造体が、中心部に中空部分を有する球状構造体であり、
該中空部分に医薬が包接されていることを特徴とする請求項13に記載の核酸送達用構造体。 - 請求項1に記載の核酸アナログの製造方法であって、前記カチオン性人工核酸を合成するカチオン性核酸合成工程と、該カチオン性人工核酸と前記親水性重合体とを結合する結合工程とを備え、
前記カチオン性核酸合成工程は、前記連結構造にチオリン酸エステルを導入する工程と、該チオリン酸エステルとブロモ化合物とを反応させる工程と、を備えることを特徴とする核酸アナログの製造方法。 - 請求項9に記載の核酸送達用構造体の製造方法であって、
前記核酸アナログの前記カチオン性基のpKaよりも低く、前記核酸のリン酸のpKaよりも高いpH条件で前記核酸アナログと前記核酸とを静電相互作用により会合させて会合構造を形成させる会合工程と、を有することを特徴とする核酸送達用構造体の製造方法。 - 複数の前記核酸送達用構造体を会合させて、前記会合構造が内側に、前記親水性重合体が外側に位置するベシクル又はミセルを水性溶媒中で形成する会合体形成工程と、を更に備えることを特徴とする請求項16に記載の核酸送達用構造体の製造方法。
- 請求項9に記載の核酸送達用構造体を用いて標的部位に前記核酸を送達するための核酸送達方法であって、
前記核酸送達用構造体を投与して前記標的部位内に前記核酸送達用構造体を取り込ませる投与工程と、
前記標的部位内で前記核酸を放出させる放出工程と、を有することを特徴とする核酸送達方法。
