Various studies have shown that in vitro digestion of ingredients by enzymes from the target shrimp or fish species is associated with apparent protein digestibility (APD). For almost half a century, the pH-stat method has been used to monitor the effects of heat treatment on the initial rates of trypsin proteolysis of soy protein. Previous research has also demonstrated the relationship between apparent protein digestibility (APD) and in vitro DH in juvenile Pacific white shrimp.

The pH of a one- liter solution was adjusted to 8.6 and heated to boiling with frequent stirring for 2 minutes. The solution was then placed on sterile plates until the agar solidified and stored at 2-8°C until use.

  • TSA Agar Teryptone Soy Agar: for isolation, purification and growth tests with probiotic strains, Difco TSA medium was used. Powdered agar (40g) was dissolved in 1 liter of distilled water. The solution was homogenized for 2 minutes and subsequently plated and autoclaved at 121 °C for 15 minutes.
  • TSB Stepticase Soy Broth: Difco TBS medium was used for the isolation, purification and growth tests with strains of V. parahaemolyticus , V. vulnificus and several commercial probiotics.
  • Powered agar (30g) was dissolved in 1 liter of distilled water. The solution was homogenized for 2 minutes and subsequently plated and autoclaved at 121°C for 15 minutes.
  • HQPC was added to all the agars and liquid media at three concentrations: 5%, 10% and 15%. All inclusions into the culture media at the indicated concentrations were used to measure the effect against V. parahaemolyticus and commercial probiotics from three companies.
  • Agar Inoculations shrimp post-larva (0. lg) were weighed, macerated and resuspended in 200 pi of sterile distilled water. A 50 m ⁇ sample was inoculated in TCBS agar and ChromAgar. The results showed that HQPC at 1% and 2% at time zero had an inhibitory effect of 27%.
  • the growth of V. parahaemolyticus is compromised after the first 6 hours of culture for both dosages with a 40% reduction in growth. At 12 hours there is a slight recovery, and at 18 hours, the reactivation bacteria growth is back to normal (FIG. 20).

  • V. vulnificus were isolated from larval samples with disease signs and used in this example. In Ecuador, the association of V. parahaemolyticus and V. vulnificus at levels of 10 4 CFU has been determined to be the cause of high mortalities.
  • Strains of V. parahaemolyticus and V. vulnificus were resuspended and incubated to reactivate for 1 hr at 32°C in TSB culture medium with the inclusion of HQPC at 5%, 10% and 15%. Incubations were carried out immediately in ChromAgar Vibrio to determine the effect on bacteria growth. The results showed that HQPC at 10% inclusion level can strongly affect V. vulnificus growth. Rate with a 72% reduction in bacteria population at 18 hrs. At the 15% inclusion level the growth rate of V. parahaemolyticus is reduced significantly up to 63% (FIG. 21).

  • Example 14 Evaluation of the inhibitory activity of HQPC at inclusion levels of 1% and 2% against three (3) strains of commercial probiotics [00245] Three (3) commercial probiotics were used for this test. The strains were reactivated in TSB culture medium with the inclusion of 1% and 2% HQPC and incubated at 30°C for 24 hrs. Subsequently, samples were incubated every six (6) hrs in TSA medium without salt. The results shown are from CFU readings and bacteria levels counted in the agar plates versus CFU counts obtained from the controls. This ratio was used to find a relationship of HQPC inhibitions vs. probiotic activity. The commercial probiotics show an inhibition of 71%, 38% and 6% with HQPC at 1% and 72%, 44% and 8% with HQPC at 2% (FIG. 23).
  • Example 15 Effects of HQPC on Shrimp at High Stocking Density
  • Pathogen free shrimp were used in this Example (as determined by PCR). Tanks with a capacity of 350 L were used, and all tanks were outfitted with an activated coral filter, aerated and covered with a plastic film cap to reduce the risk of cross contamination. Brackish water with a salinity of 20 parts per thousand (ppt) was used.
