心筋細胞の分化の促進に高い機能を発揮し、心疾患後の心機能改善に効果を有する、心筋分化を促進するための組成物および心筋分化を促進するための医薬組成物を提供する。miR-132を有効成分として含有する組成物および医薬組成物である。
Description
本発明は、心疾患の予防および/または治療に用いる、心筋分化を促進するための組成物および医薬組成物に関する。
本願は、2021年3月23日に米国に仮出願された米国特許出願番号63/164,581号に基づき優先権を主張し、その内容をここに援用する。

現在、日本では心筋梗塞の症例が年間10万例弱認められる。心臓の治療において、ヒトの心臓組織が傷害後も修復可能となれば、心筋梗塞による死を激減させうる可能性がある。しかしながら、ヒトの心臓は原則として再生しない臓器とされている。心臓の再生能力の有無は、心筋細胞の増殖能の有無によって決定されているのではないかとの説に則って、心筋細胞を増殖可能とすることにより、心臓の再生が可能になるのでは、と期待され、種々の研究がなされている。
心臓再生医療は、現在多能性幹細胞から誘導した心筋細胞や心臓様組織の移植(細胞移植や組織移植)による新規治療開発が中心である。その一方で、より材料の調製が簡易で患者への侵襲性が低い治療法の開発へとシフトしていくことは必定である。心筋細胞の増殖を促進するような成分、またはその成分を含む培養物等の混合物等が見つかれば、その成分を投与することで、培養した心筋細胞や心臓様組織を移植するよりも侵襲性が低い治療法に用いることが期待できる。
一方、治療に用いることができる可能性のある培養物として、細胞や細胞外小胞の培養物、培養上清等が治療用材料として研究が進められている。例えば、その中でエクソソームを含む細胞外小胞(EV)などが、現在注目されている大きな研究ターゲットの一つである。
非特許文献1では、Cardiosphereと呼ばれる心臓組織から培養してSphere状にした細胞の塊を、小児拡張型心筋症患者に移植する研究内容を開示している。この文献では、機能的にこの疾患への関与のある可能性のある成分として、マイクロRNA-146(miR-146)が挙げられている。

非特許文献1では、小児拡張型心筋症患者に対する機能に関与している可能性のある成分としてmiR-146が挙げられているが、この成分の心臓組織に対する影響、例えば心筋細胞自体の分化、増殖に関与しているかの機序は充分に明らかになっていない。
組織から培養した細胞塊、細胞外小胞(EV)などについては、内容物はそれが由来する細胞種や培養条件等々により大きく異なることが知られてきており、治療材料として製造しようとした際に、品質等を一定に保ちにくい側面がある。そのため、これらを単純に直接の治療材料とするには難がある。そこで、これらのEV等の内容物のうち、治療効果のある成分が何であるかを特定し、また、その成分をEV等の構成を模した手段により輸送して、治療材料として用いる手法を開発することが考えられる。
本発明者らは、多能性幹細胞(PSC)の分化機構の研究から、分化途上のPSC由来の細胞外小胞(EV)がPSCや胎仔における心筋細胞分化を促進することを見出した。そこで、心筋細胞分化を促進する成分を、心疾患への処置に役立てるべく、さらに同EVの成分、主にEVの内容物において、同心筋分化に寄与する分子を探索し、輸送手段に応用できるようさらに研究を進めていった。
本発明は、上記事情に鑑みてなされたものであって、心筋細胞の分化の促進に高い機能を発揮し、心疾患後の心機能改善に効果を有する、心筋分化を促進するための組成物および心筋分化を促進するための医薬組成物を提供することを目的とする。
本発明の実施態様は、以下の側面を有する。

本発明によれば、心筋細胞の分化の促進に高い機能を発揮し、心疾患後の心機能改善に効果を有する、組成物および医薬組成物が得られる。
以下、本発明に係る組成物および医薬組成物について、実施形態を示して説明する。ただし、本発明は以下の実施形態に限定されるものではない。
(心筋分化を促進するための組成物)
本実施形態の組成物は、心筋分化を促進するための組成物で、miR-132を有効成分として含有する。

miR-132は、マイクロRNA(miRNA)であり、タンパク質をコードしない短鎖RNA分子の一種である。マウス等のげっ歯類やヒトをはじめ複数の哺乳類のゲノム上にmiR-132のファミリーがみられる。本実施形態では、各種のmiR-132ファミリーの分子から選択することができる。また、miR-132分子は市販されているものを用いることができる。マイクロRNAとしての機能を有するものであれば、miR-132分子の各種の変異体を使用することもできる。
例えば、心筋分化を促進する対象がヒトであれば、ヒトmiR-132(RNA配列は配列番号:1)(ゲノム配列はNCBI RefSeq:NC_000017.11)、マウスであればマウスmiR-132(RNA配列は配列番号:2)(ゲノム配列はNCBI RefSeq:NC_000077.7)などから選ぶことができる。

本発明者らは、多能性幹細胞(PSC)の分化機構の研究から、分化途上のPSC由来の細胞外小胞(EV)がPSCや胎仔における心筋細胞分化を促進することを見出した。さらに、同EVの内容物において同心筋分化に寄与する分子を探索し、miR-132を含むマイクロRNAを同定した。
すなわち、多能性状態から3種の胚葉すべてに分化できる胚性幹細胞(ESC)について、プロテインキナーゼA(PKA)が活性化したESC(PKA-ESC)は、3種の胚葉のうち、中胚葉細胞の出現が加速されている。分化の遅い細胞と分化の速い細胞の表現型を同期させて、同じ分化段階に到達する細胞の表現型のシンクロニーを研究した。細胞の表現型のシンクロニーを調節している分子機構の存在を調査し、細胞外小胞(EV)とそのマイクロRNA(miR)の含有量が原因であると特定した。PKA-ESCに由来するEVは、中胚葉分化や、さらに心筋細胞の分化を促進していることを見出した。
さらに、PKA-ESCからEVで特異的に増加した分子の中で、PKA活性化を再構成し、中胚葉系統の分化を促進できる機能性分子を探索したところ、miR-132が同定された。さらに、本発明者らは、miR-132を含む人工ナノ粒子をexvivoマウス胚培養に添加することによって、同様の心筋細胞誘導が再現されることも見出した。
本発明者らは、これらの結果から、miR-132が心筋細胞誘導を直接促進する効果を有することができる分子であることを見出した。miR-132は、心筋細胞誘導を有効に促進するので、miR-132を含む組成物は、心筋細胞の増殖、心疾患の治療などの対処、または心疾患の予防等の分野に広く応用することができる。
また、miR-132は、マイクロRNAすなわち十数~数十核酸残基の小分子の一種である。そのため、他の分子に封入する、または混入するなどの手段を介した各種のデリバリーシステムを介して輸送することが容易である。EVなどを模したデリバリーシステム、例えばナノ粒子を介して輸送することで、患者への侵襲性が低い治療に用いることができる。また、miR-132は小分子であるため調整も容易である。
本実施形態の組成物は、粒子状であることも好ましい。ここで粒子状とは、一般に形状や大きさによる影響が少ない充分に径の小さい物体を指すが、本実施形態では、例えば核酸鎖のような長鎖状ではなく、小型で、かつ最も大きい短径と最も大きい長径の差が比較的小さい塊状の物体を指すものとする。具体的には、最も大きい径がナノサイズであること、1~1000nmであることが好ましい。また、前記粒子状は、径が50~500nmであることも好ましい。このサイズであることで、マイクロRNAを粒子の中に含有することが容易であり、また大きすぎないことで投与が容易である。
組成物が粒子状であるときは、前記粒子を構成する成分が、miR-132の他の成分(以下、粒子成分)を含むことが好ましい。この粒子成分は、粒子の形状および/または大きさをある程度規定する。この粒子成分は、生体適合性が高い化合物であることが好ましい。粒子成分としては、従来知られたドラッグデリバリーに用いられる化合物等を用いることができる。
本実施形態の組成物は、前記粒子の粒子成分が、乳酸・グリコール酸共重合体(PLGA)ナノ粒子またはリポソームであることも好ましい。PLGAナノ粒子およびリポソームは、調整および大きさ等のデザインが容易であり、生体適合性が高く、治療材料として用いる組成物に好適に応用できる。
PLGA化合物は、共重合の割合、製造方法および使用する分子の大きさなどは特に限定されないが、例えば乳酸とグリコールの共重合割合が1:5~5:1、重合度は2000~20000Daなどから選択できる。
リポソームは、製造方法および使用する分子の大きさなどは特に限定されず、従来知られた方法から適宜選択できる。例えば、リン脂質やコレステロールなどの脂肪酸を用いてLNP(脂質ナノ粒子)を調整してもよい。
本実施形態の組成物は、粒子であるときは、前述の粒子にmiR-132を含む。
粒子がmiR-132を含む形態としては、前記粒子成分にmiR-132が混合または複合されていてもよい。例えば、混合または複合の形態としては、粒子成分の分子間にmiR-132が配置され、分子間力で固着していてもよい。また、粒子成分に対してmiR-132が固定化されていてもよい。なお、粒子成分とmiR-132との固着または固定化が強固なものである場合、粒子成分は一定条件で分解するものを選択すると、分子の輸送において高度な選択制を持たせることができる。
粒子がmiR-132を含む形態としては、前記粒子成分にmiR-132が混合または複合されていてもよい。例えば、混合または複合の形態としては、粒子成分の分子間にmiR-132が配置され、分子間力で固着していてもよい。また、粒子成分に対してmiR-132が固定化されていてもよい。なお、粒子成分とmiR-132との固着または固定化が強固なものである場合、粒子成分は一定条件で分解するものを選択すると、分子の輸送において高度な選択制を持たせることができる。
具体的には、前記粒子にmiR-132が導入されている。導入の手段は、従来知られたものを用いてもよい。例えば、乳酸・グリコール酸共重合体(PLGA)ナノ粒子にマイクロRNAを導入(インプット)する手段としては、水相(W相)にマイクロRNA、有機相(O相)にビオチン-PLGAを加え、混合してW/O相を作り、さらにこのW/O相を別の水系のW相に対して滴下し、溶媒を留去することでマイクロRNAを導入されたPLGAナノ粒子が得られる。
LNPにmiR-132を導入する手段としては、リポソームの調整と共に行ってもよい。例えば、上述の脂質を含む脂質溶液を攪拌しながら、miR-132を含む水系の核酸溶液を滴下して脂質/核酸溶液を調整し、この脂質/核酸溶液を緩衝液内にさらに滴下することによって得られる。
本実施形態の組成物は、心筋分化を促進するためにはin vitroの細胞に対しても用いることもでき、また生物の体内の心筋組織に対して用いることもできる。上述したように、本実施形態の組成物は、輸送や投与が容易なので、生物の体内に用いることが好適である。
(心疾患を治療または予防するための医薬組成物)
本実施形態の医薬組成物は、心疾患を治療または予防するための医薬組成物で、miR-132を有効成分として含有する。医薬組成物は、粒子状であってもよい。粒子状の医薬組成物は、乳酸・グリコール酸共重合体(PLGA)ナノ粒子またはリポソームであってもよい。
上記構成は、上述の心筋分化を促進するための組成物について記載したものと同様の構成から選択することができる。
本実施形態の医薬組成物は、心疾患を治療または予防するための医薬組成物で、miR-132を有効成分として含有する。医薬組成物は、粒子状であってもよい。粒子状の医薬組成物は、乳酸・グリコール酸共重合体(PLGA)ナノ粒子またはリポソームであってもよい。
上記構成は、上述の心筋分化を促進するための組成物について記載したものと同様の構成から選択することができる。
本実施形態の医薬組成物は、ヒトの心臓の疾患(心疾患)の治療に用いることができる。ここで心臓の疾患とは、心臓における疾患、または心臓の処置の必要がある疾患を広く含む。本実施形態の心筋細胞を治療に用いるには、得られた心筋細胞を患者に投与する、または得られた心筋細胞を心臓の疾患の治療剤として用いてもよい。前記心筋細胞の投与の方法としては、細胞を液状に懸濁して心筋層に投与する、または細胞をシートやバンデージ等を介して添付するといった手段を用いることができる。前記心臓の疾患としては、心不全、虚血性心疾患、心筋梗塞、心筋症、心筋炎、肥大型心筋症、拡張相肥大型心筋症、拡張型心筋症などの疾患、または障害による欠損等を広く含み、心臓に影響のある疾患であれば限定されない。
本実施形態の医薬組成物は、前記心疾患が心筋梗塞であることも好ましい。心筋梗塞は現在症例が多数認められ、深刻な問題となっている。上述のように、本実施形態の医薬組成物は心筋分化を促進するための成分を含むので、傷害の生じた心臓組織を補うことができる。また、上述したように手術等によらずとも医薬組成物の投与により治療を行うことができるので、患者への負担が少なく、深刻な症状の生じた患者への処置に特に有効である。
本実施形態の医薬組成物の有効量の目安は、心疾患の患者に対して、miRNA-132の量として0.4-40μg/kgが好ましい。また、前述の粒子成分としてリポソームを用いた場合の粒子を含む医薬組成物の場合は、0.04-4.0mg/kgが好ましい。前述の粒子成分としてPLGA-NPを用いた場合の粒子を含む医薬組成物の場合は、0.004-0.4mg/kgが好ましい。
このとき、心筋細胞増殖剤は、適宜他の成分を含んでいてもよい。例えば、心臓その他の循環器の疾患の処置に用いられる成分として従来知られているものを含んでいてもよい。

本発明者らは、EVを介した細胞間コミュニケーションによる分化の過程で、細胞の表現型が同期するという新しい生物学的現象「細胞の表現型同期(PSyC)」について研究を進める過程で、EVの1つの潜在的な候補分子である、miR-132を特定した。本発明者らは、この分子が、PSyCのレシピエント細胞の分子リンクを再構成し、EVのin vivo効果を再現して中胚葉誘導体を誘導することができることを見出した。
本発明者らの知見により、これまで中胚葉を誘導することが報告されていなかったmiR-132が、PKA活性化の再構成を通じてEVレシピエント細胞に中胚葉誘導を引き起こすことが見出された。EVは、ホルモンに類似した遠隔効果を介して作用する新しい細胞間コミュニケーションモダリティとして注目されている。EVは、細胞由来の膜構造の不均一なグループであり、それぞれエンドソーム系に由来する、または原形質膜から脱落する、エクソソームおよびマイクロベシクルを含むと考えられている。例えば、TSG101のサイレンシングは、エクソソーム分泌を減少させることが報告されており、一貫して、TSG101のノックアウトは細胞の増殖と分化を阻害し、TSG101KOマウスは着床段階の前後で胚の致死性を示すことが報告されている。一方で、エクソソーム産生に関与する他の因子nSMase2をノックアウトしたマウスは、すべての組織で形成不全と成長遅延を示すが、胎児の致死性は示さない。発明者らの研究では、PSyCはnSMase2阻害剤によって有意にブロックされたが、完全にはブロックせず、他のEV生成メカニズム、エクソソームおよび/またはマイクロベシクルの関与も示唆していると考えられた。
本発明者らは、近接した細胞間の新しい細胞間コミュニケーションメカニズムを見出した。すなわち、PSyCは、細胞の運命決定と分化の段階を厳密に調整する同期メカニズムを通じて、正常組織を構築するための細胞サブセットを確立することから、さらに、PSyCは組織の発達と恒常性を調節するための重要なメカニズムであると推測された。組織および器官は、隣接する細胞間のEV交換を介して、より活発で大量の分子輸送によって確立および維持される可能性が考えられた。
本発明者らは、miR-132含有ナノ粒子の投与により、心筋梗塞後の心機能低下が抑制される傾向が認められることを見出した。心筋梗塞モデルは、虚血-再灌流傷害(I/R)モデルであり、心筋梗塞後にカテーテル治療により再開通した臨床状況に即している。またナノ粒子の投与は経静脈的であり、臨床における通常の静脈注射や点滴治療に相当し、侵襲性が少なく臨床的有用性が高いと思われる。カテーテルを用いて経冠動脈的に投与する形でも十分な治療応用が期待できる。
本実施形態の組成物および医薬組成物は、心筋細胞分化促進、さらには心筋梗塞後の心機能改善にmiR-132が効果を有すること、及び、その生体へのデリバリーシステムとして、ナノ粒子が有効であることを示し、miR-132含有ナノ粒子が、心臓再生治療薬として応用できることが期待される。また、静脈注射や点滴などの形で臨床応用が可能であれば、心臓カテーテルが施行できない小規模医療施設でも心臓再生治療が可能となり、その応用範囲は非常に大きい。
以下、実施例によって本発明を具体的に説明するが、本発明は以下の記載によって限定されない。
[使用した材料、方法]

miR-132のゲノム領域を最小プロモーターとHiBitを含む一本鎖オリゴデオキシヌクレアーゼ(ssODN)で置き換えることによって調整した。ssODN(Integrated DNA Technologies Coralville、USAに注文)およびCas9を発現するpX330プラスミドとmiR-132遺伝子座を標的とするsgRNAを、リポフェクタミンLTX試薬を使用してPKA-ESCに導入した。Nano Glo HiBiT Lytic Detection System(N3030、Promega)を使用してHiBitタンパク質を検出することにより、細胞を選択した。pX330は、Feng Zhang博士(マサチューセッツ工科大学)から提供を受けた。

ESC系統は、10%ノックアウト血清代替品(KSR;10828-028、Thermo Fisher Scientific)、1%ウシ胎児血清(FBS;SAFC Biosciences、USA)を含むグラスゴー最小必須培地(GMEM;11710-035、Thermo Fisher Scientific)、2,000ユニット/ mL白血病抑制因子(LIF;Merck Millipore)、1mMピルビン酸ナトリウム(Sigma-Aldrich)、1%MEM非必須アミノ酸(NEAA;11140-050、Thermo Fisher Scientific)、50U/mLペニシリン(Meiji Seika Pharma)、50μg/mLストレプトマイシン(Meiji Seika Pharma)および0.1mM 2-メルカプトエタノール(2-ME;21985-023、Thermo Fisher Scientific)内にて培養した。また、PKA-ESCの維持のために、1μg/mLのドキシサイクリン(Dox)を追加した。

また、EVの分泌を阻害するために、凝集体をマヌマイシンA(BVT-0091、BioViotica)またはGW4869(13127、Cayman Chemical)で処理した。

ESC系統は、低接着ディッシュ、PrimeSurface Dish 35 mm(MS-9035X、住友ベークライト)、またはU底円錐ウェル(ウェルあたり9,000細胞、100mL;MS)を備えた低細胞接着96ウェルプレートに播種した。培養は9096U、Sumilon PrimeSurfaceプレート、住友ベークライト)分化培地を使用した。24時間後、浮遊凝集体を収集し、マルチウェルプレート(Falcon)にプレーティングした。キメラ細胞凝集体を作製するために、ESC株を示された比率で混合した。

培養細胞は、分化中のD1.5、D2.5、D3.5、およびD4.5(D=日)に採取され、アロフィコシアニン(APC)結合抗Flk1 mAb(AVAS12)とフィコエリトリン(PE)の組み合わせで染色した。結合した抗血小板由来成長因子受容体(PDGFRα mAb(12-1401-81、eBioscience))を、FACS Aria(Becton Dickinson)またはCytoFLEX S(Beckman Coulter)で分析した。
(免疫組織化学)
組織染色は以下で行った。培養細胞を氷冷した4%パラホルムアルデヒド溶液で1時間固定し、2%スキムミルク(232100、Becton Dickinson)で4℃で24時間ブロックし、一次抗体と4℃で24時間インキュベートした。ここで一次抗体としては、Flk1(AVAS12、自社製、1:500,000)、PDGFRα(3174S、Cell Signaling Technology、1:500)、Oct4(sc-5279、Santa Cruz Biotechnology、1:200)およびNanog(RCAB002P、ReproCELL、1:300)を使用した。
一次抗体のインキュベーション後、細胞をリン酸緩衝生理食塩水(PBS)で3回洗浄し、Alexa Fluor(Thermo Fisher Scientific、1:500)に結合した二次抗体と4℃で24時間インキュベートした。
核はDAPI(4,6ジアミジノ-2-フェニルインドール;D3571、Thermo FisherScientific)で染色した。

分離したEVを、2-MEを含まない2×サンプルバッファー溶液(ナカライテスク、日本)で溶解し、70℃で5分間インキュベートした。サンプルは、勾配ゲル(E-T1020L、Atto)を使用してドデシル硫酸ナトリウム-ポリアクリルアミドゲル電気泳動(SDS-PAGE)で泳動し、続いてニトロセルロース膜に電気泳動で転写した。ブロット後、メンブレンをブロッキング剤Blocking One(03953-95、Nacalai Tesque)で1時間インキュベートし、次に一次抗体とともに4℃で24時間インキュベートした。二次抗体としては、西洋ワサビペルオキシダーゼ(HRP)と結合した抗マウスまたはラットIgG抗体を使用した(1:50,000)。シグナルエンハンサーとして、Can Get Signal Immunoreaction Enhancer Solution Kit(NKB-101、東洋紡)を使用した。免疫反応性は、強化された化学発光キット、Immobilon Western(WBKLS0500、Merck Millipore)を使用して検出した。信号強度は、ImageQuant TLソフトウェア(GE Healthcare Life Sciences)を使用して計算した。
分離したEVを、2-MEを含まない2×サンプルバッファー溶液(ナカライテスク、日本)で溶解し、70℃で5分間インキュベートした。サンプルは、勾配ゲル(E-T1020L、Atto)を使用してドデシル硫酸ナトリウム-ポリアクリルアミドゲル電気泳動(SDS-PAGE)で泳動し、続いてニトロセルロース膜に電気泳動で転写した。ブロット後、メンブレンをブロッキング剤Blocking One(03953-95、Nacalai Tesque)で1時間インキュベートし、次に一次抗体とともに4℃で24時間インキュベートした。二次抗体としては、西洋ワサビペルオキシダーゼ(HRP)と結合した抗マウスまたはラットIgG抗体を使用した(1:50,000)。シグナルエンハンサーとして、Can Get Signal Immunoreaction Enhancer Solution Kit(NKB-101、東洋紡)を使用した。免疫反応性は、強化された化学発光キット、Immobilon Western(WBKLS0500、Merck Millipore)を使用して検出した。信号強度は、ImageQuant TLソフトウェア(GE Healthcare Life Sciences)を使用して計算した。

ついで、上清を回収し、12,000gで30分間室温で遠心分離した。細胞の破片と細胞外マトリックスの塊を取り除くために、上清を0.2μmポアフィルター(ザルトリウス)でろ過し、次に上清を100,000gで60分間、4℃で超遠心分離した。この操作のペレットとして、EVを分離した。
1.0×105 PKA-ESCを、分化培地を含む10cmディッシュに播種した。2日後、培地を新しいものに交換した。さらに1.5~2.5日間培養した後、分化培地を回収し、2,000gで20分間室温で遠心分離した。

EVをPBS(30μg/mL)に懸濁し、炭素安定化コロジオンコーティンググリッド(400メッシュ;Nisshin-EM)の膜側に滴下(20μL/液滴)し、室温で10分間放置した。溶液を濾紙で除去し、蒸留水ですすいだ。蒸留水に溶解した酢酸ウラニルの1%溶液をグリッドに適用し、次に室温で1分間放置した。ついで、試薬を濾紙で除去し、乾燥させた。EVは、透過型電子顕微鏡(TEM;H-7650、日立ハイテクノロジーズ、日本)で画像化した。

(胚のex vivo培養)
E6.5の胚は、5%FBS(SAFC Biosciences、USA)を含むダルベッコ改変イーグル培地(DMEM)/F12(1:1)(11039-012、Thermo Fisher Scientific)で子宮と脱落膜を解剖することによって分離した。単離された胚は、50%ラット血清、0.1mM NEAA、1mMピルビン酸ナトリウムおよび0.5mMの2-MEを含む500μL DMEM(21063-029、Thermo Fisher Scientific)を使用して、0.4 μmポア(3460、Corning)(低接着用)の12mmトランスウェルで培養した。
E6.5の胚は、5%FBS(SAFC Biosciences、USA)を含むダルベッコ改変イーグル培地(DMEM)/F12(1:1)(11039-012、Thermo Fisher Scientific)で子宮と脱落膜を解剖することによって分離した。単離された胚は、50%ラット血清、0.1mM NEAA、1mMピルビン酸ナトリウムおよび0.5mMの2-MEを含む500μL DMEM(21063-029、Thermo Fisher Scientific)を使用して、0.4 μmポア(3460、Corning)(低接着用)の12mmトランスウェルで培養した。
EVの有無にかかわらず2日間培養した後、胚を0.25%トリプシン/エチレンジアミン四酢酸(EDTA)で37℃で15~25分間解離させた。細胞をPE結合抗PDGFRαmAb(12-1401-81、eBioscience)で染色し、FACS Aria(Becton Dickinson)を使用して分析した。
E3.5の胚は、子宮をKSOM培地(社内で調製)で洗浄して回収した。胚をM2培地(M7167、Sigma-Aldrich)の液滴に収集した。透明帯は、酸性タイロードの溶液滴(T1788、Sigma-Aldrich)に短時間さらすことで除去した。
M2ミディアムドロップとタイロードの溶液ドロップを鉱油(HiGROW OIL、扶桑薬品工業)で覆い、37℃、5%CO2で事前に平衡化した。

透明帯を含まない胚盤胞を、20%FBSを含み2 mM L-グルタミン(25030-081、Thermo Fisher Scientific)を添加したin vitro培地(IVC1)(Advanced DMEM/F12(12634-010、Thermo Fisher Scientific)で洗浄した。ペニシリン(25ユニット/mL)/ストレプトマイシン(25μg/mL)、1×インスリン-トランスフェリン-セレニウム-エタノールアミン(ITS-X)(51500-056、Thermo Fisher Scientific)、8nM-エストラジオール(E8875、Sigma-Aldrich)、200ng/mLプロゲステロン(160-24511、和光、日本)および25μM N-アセチル-L-システイン(A7250、Sigma-Aldrich))を使用して、ミネラルオイルとM2培地を除去し、μ-スライド8ウェル(80826、イビディ)のプレートに播種した。前記プレートのウェルは、前述のように事前に平衡化されたIVC1培地で満たされていた。
胚がプレートの底に付着してから2、3日後、培地を化学合成されたIVC2培地(30%KSRを含み、2mM L-グルタミン、ペニシリン(25ユニット/mL)を添加したAdvanced DMEM/F12)に交換した。ストレプトマイシン(25μg/mL)、1×ITS-X、8nM-エストラジオール、200ng/mLプロゲステロンおよび25μM N-アセチル-L-システイン)、5%CO2中37℃で胚培養を行った。胚は、0日目から8日目まで2日ごとにEVまたはポリマーポリ(DL-ラクチド-co-グリコリド)(PLGA)ナノ粒子で処理した。

(PLGAナノ粒子の調製)
平均分子量が20,000Daで、ラクチドとグリコリドの共重合体比が75:25のビオチン化PLGA(Nanosoft Polymers、NC)をナノ粒子のマトリックスとして使用した。ポリビニルアルコール(PVA-403、Kuraray)は分散剤として使用した。
mirVana miRNA mimic hsa-mir-132-3p(4464066、Thermo Fisher Science)を組み込んだPLGAナノ粒子は、前述のように、RNAaseを含まない水中でエマルジョン溶媒拡散法を使用して調製した(Kawashima et al.,(1998),Eur. J. Pharm. Biopharm.45,41-48)。ヒトmiR-132-3pとマウスmiR-132-3pの配列は同一であった。
miR-132-PLGAには11.6%(w/w)のmiR-132が含まれていた。蒸留水中のナノ粒子懸濁液のサンプルを使用して、粒子サイズを決定した。動的光散乱に基づくmiR-132-PLGAの直径の中央値は262nmであった。

本発明者らは、胚性幹細胞(ESC)を用いて多能性幹細胞(PSC)の分化機構の研究を行った。ESCは、初期胚盤胞の内部細胞塊(ICM)に由来し、多能性状態からin vitroで3つの胚葉すべてに分化できるため、細胞分化プロセスを研究するためのツールとして用いることができる。
(心筋細胞分化促進に関与する因子の探索)
(胚性幹細胞の分化段階の検討)
本発明者らは、胚性幹細胞(ESC)を用いて多能性幹細胞(PSC)の分化機構の研究を行った。ESCは、初期胚盤胞の内部細胞塊(ICM)に由来し、多能性状態からin vitroで3つの胚葉すべてに分化できるため、細胞分化プロセスを研究するためのツールとして用いることができる。
本発明者らは以前に、プロテインキナーゼA(PKA)がドキシサイクリンの枯渇(Dox-OFF)によって構成的に活性化されるマウスESCラインPKA-ESCを生成している。図1はPKA活性化の実験システムを示す。PKA-ESCは、ドキシサイクリン調節発現システム(Dox-Off)を介して、構成的に活性な形態のPKA(CA-PKA)を発現する。未分化状態を維持するために、PKA-ESCをLIFおよびDoxの存在下で培養した。LIFは分化誘導のために除外されている。PKA-ESCは、Dox+よりもDox-の方が速く分化する。図38に骨材培養の概略図を示す。マウスESCを低接着ディッシュに播種して骨材を作成し、LIFの枯渇によって分化誘導を開始し、24時間後、骨材を通常のプレートに再プレートし、約12時間後に細胞接着している。Flk1+細胞において、PKA-ESC(Dox-)においてDox+より早く分化が起こることを示している。
図2はPKA-ESCの分化中のFlk1の免疫染色を示す。図3はPKA-ESCでのFlk1の出現に関するFACS分析を示す。図4はFACS分析によるFlk1を発現するPKA-ESCのパーセンテージで、マンホイットニーU検定を使用してデータを分析したものを示す。Dox-条件下では、PKA-ESCはより早くそして有意に多くのFlk1+細胞を示している(*PKA-ESC(Dox+)に対してP<0.05)。これ非活性化したPKA-ESCでは活性化は1%未満で、活性化したPKA-ESCでは40%以上であった。
また、図39はPDGFRαの純粋なPKA-ESC凝集体のFACS分析、図40はPDGFRα+PKA-ESCのパーセンテージを、マンホイットニーU検定を使用して分析したデータ(*PKA-ESC(Dox+)に対してP<0.05)、図41は純粋なPKA-ESC凝集体の分化中のPDGFRαの免疫染色を示す。他の中胚葉マーカーであるPDGFRαもPKA活性化により向上が見られた。
図42~45はFlk1、およびPDGFRαの純粋なControl-ESC凝集体のFACS分析でFlk1+コントロール-ESCおよびPDGFRα+コントロール-ESCのパーセンテージを示す。図46と47は純粋なControl-ESC凝集体の分化中のFlk1(図46)およびPDGFRα(図47)の免疫染色である。これらの結果から、分化の速度はDox曝露によっては変化はなかった。Dox+、Dox-の両方でコントロールESC内でのFlk1+細胞量は10%以下であった(図42)。
また、図39はPDGFRαの純粋なPKA-ESC凝集体のFACS分析、図40はPDGFRα+PKA-ESCのパーセンテージを、マンホイットニーU検定を使用して分析したデータ(*PKA-ESC(Dox+)に対してP<0.05)、図41は純粋なPKA-ESC凝集体の分化中のPDGFRαの免疫染色を示す。他の中胚葉マーカーであるPDGFRαもPKA活性化により向上が見られた。
図42~45はFlk1、およびPDGFRαの純粋なControl-ESC凝集体のFACS分析でFlk1+コントロール-ESCおよびPDGFRα+コントロール-ESCのパーセンテージを示す。図46と47は純粋なControl-ESC凝集体の分化中のFlk1(図46)およびPDGFRα(図47)の免疫染色である。これらの結果から、分化の速度はDox曝露によっては変化はなかった。Dox+、Dox-の両方でコントロールESC内でのFlk1+細胞量は10%以下であった(図42)。
ついで、PKA-ESCの細胞凝集体とコントロールESCを作成した。図5は、キメラ骨材共培養分化システムの概略図を示す。PKA-ESCとControl-ESCを低接着ディッシュに3:1の比率で播種してキメラ凝集体を作成し、LIFの枯渇によって分化誘導を開始し、24時間後、骨材を通常のプレートに再プレートし、約12時間後に細胞接着した。この共培養システムでは、PKAの活性化により2つの細胞株間では異なる分化速度が見られた。すなわち、分化は、Dox枯渇を伴うPKA活性化後のPKA-ESCでのみ加速されると予想された。
図6は、PKA-ESCおよびControl-ESCキメラ凝集体の分化中のFlk1の免疫染色を示す。キメラ骨材(白)中のFlk1+Control-ESCの数は、Dox-条件で増加した。図7は、キメラ凝集体上のFlk1外観のFACS分析を示す。Dox-条件下では、PKA-ESC集団(青)のFlk1+細胞が早く現れたが、D3.5までに、Control-ESC集団(緑)のFlk1+細胞の割合はPKA-ESC集団と同じになった。図8は分化中にFlk1を発現するキメラ凝集体のパーセンテージで、データは平均±S.Dを表し、マンホイットニーU検定を使用してデータを分析した(*Control-ESCと比較してP<0.05)。これらの結果より、キメラ凝集体では、PKA-ESCでのPKA活性化により、PKA-ESCだけでなく、D3.5のControl-ESCでも同等のレベルで中胚葉細胞の早期出現が誘導されるという結果が得られた。

図6に示すFlk1の免疫染色によると、Dox-条件下で、GFP-細胞(PKA-ESC)だけでなくGFP+細胞(コントロールESC)でもFlk1+細胞の早期出現が観察されたことを示しましている。GFP+Flk1+細胞の数は、図6に示すように、Dox-,Flk1/GFPで明色でみられる。この細胞数はD2.5から現れ、この条件でのControl-ESCのより速い分化を示唆している。
フローサイトメトリー分析(図7および図8)は、Dox枯渇後のFlk1+細胞の早期の同期した出現をより明確に示した。コントロール条件(Dox+)では、PKA-ESCとコントロールESCの両方が、D4.5まで同等のレベルでFlk1+細胞に同様に分化していた(図8)。また、PKA-ESCでのみPKAを活性化するDox-条件では、Flk1+細胞の出現はD2.5からのPKA-ESCで誘発され、D3.5-4.5で対照条件よりもはるかに高いレベルに達していた。
Doxの枯渇がPKAを活性化しなかったとしても、PKA-ESCの初期の分化に続いているかのように、Control-ESC集団のFlk1+細胞が現れ始め、最終的にはDKA-ESC集団の細胞とパーセンテージで追いつくという結果が見られた(コントロール-ESC、図7)。Control-ESC集団のFlk1+細胞の割合は、DKA-ESC集団のそれよりもD3.5まで遅れていたが、その後、それらの分化レベルは同様となっていた(図8)。
図48は、キメラ凝集体の分化中のPDGFRαの免疫染色を示す。キメラ凝集体中のPDGFRα+ Control-ESCの数(明色)は、Dox-条件で増加した。図49は、キメラ凝集体でのPDGFRα発現のFACS分析を示す。それぞれのグラフにおいて左側がPKA-ESC、右側がコントロール-ESCである。図50は、PDGFRαを発現する分化中のキメラ凝集体のパーセンテージを示し、データは平均±S.Dを表す。
これらの結果から、Control-ESC集団におけるPDGFRα+細胞の同様の初期の同期した出現は、PKA活性化後のキメラ細胞凝集体で見られた。これらの結果は、異なる細胞集団(すなわち、PSyC)の細胞表現型を同期させる新しい細胞メカニズムが存在することを示している。
これらの結果から、Control-ESC集団におけるPDGFRα+細胞の同様の初期の同期した出現は、PKA活性化後のキメラ細胞凝集体で見られた。これらの結果は、異なる細胞集団(すなわち、PSyC)の細胞表現型を同期させる新しい細胞メカニズムが存在することを示している。
(PSyCのメカニズムの検討)
ついで、本発明者らは、PSyCのメカニズムを検討した。体液性因子、ギャップ結合、EVなどのPSyCを行うことができる細胞間通信システムを検索し、EVを有力な候補として発見した。ここでEVには、エクソソームとマイクロベシクル(微小胞、エクトソームとも呼ばれる)が含まれる。エクソソームは、エンドソームの内向きの出芽によって生成される50~150nmサイズのEVである。マイクロベシクルは、原形質膜の直接出芽によって生成される100~1000nmサイズのEVである。細胞は、EVを介して、mRNA、miR、およびタンパク質など、いくつかの種類の生体分子を伝達する。

図9は、EV分泌が阻害されたキメラ凝集体共培養分化システムの概略図を示すものである。PKA-ESCとControl-ESCを低接着ディッシュに3:1の比率で播種してキメラ凝集体を作成し、LIFの枯渇によって分化誘導を開始した。24時間後、骨材を通常のプレートに再播種し、EV分泌阻害剤とともに培養した。
図10は、マヌマイシンA、GW4869の処理によるFlk1細胞の数を示し、Dox条件下でのD3.5上のPKA-ESCおよびControl-ESCキメラ凝集体をFACSによって分析した。ここで、マヌマイシンAとGW4869は、エクソソーム分泌を調節する酵素中性スフィンゴミエリナーゼ2(nSMase2)の2つの阻害剤である。Flk1+細胞の数は、EV分泌阻害剤であるマヌマイシンA(10μM)またはGW4869(5μM)の処理により減少した。図11は、Dox-条件下でFlk1を発現するキメラ凝集体におけるPKA-ESCおよびControl-ESCのパーセンテージを示す。Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。これらの結果によると、マヌマイシンAまたはGW4869処理のいずれかは、D3.5でのPKA活性化下のキメラ凝集体のPKA-ESCに影響を与えることなく、Control-ESC集団でのみFlk1+細胞の発現を特異的かつ有意に減少させた。

図12は、マヌマイシンA(10μM)、GW4869(5μM)、またはDMSO(コントロール)を使用したキメラ凝集体におけるFlk1+コントロール-ESC(明色)の免疫染色を示す。Control-ESCによるFlk1の発現が阻害剤処理によって減少したことが明らかになった。これらの結果は、PSyCにEVが関与していることを示唆している。
ついで、PKA-ESCから分泌されたEVの影響を分析するために、EVの収集を行った。図13に、EVの収集と処理の概略図を示す。コントロール-ESC凝集体はLIFなしでプレーティングし、EVはPKA-ESCの馴化培地からペレットとして収集し、Control-ESCの骨材に添加した。PKA-ESCから分泌されたEVの影響を分析するために、非アクティブ(EV(Dox+))およびアクティブ(EV(Dox-))PKA条件下でPKA-ESCの馴化培地でEVを分離した。
図14に、10mLの馴化培地からのエクソソームタンパク質溶解物中のCD9、CD63、およびCD81のイムノブロットを示す。EVマーカーであるこれらは、主にエクソソームで、場合によってはマイクロベシクルで発現している。図の結果より、EVの分離が確認された。

ここで、図51には馴化培地中のEV粒子、図52にはEVに含まれるタンパク質の濃度を示す。これらの結果からは、EV(Dox+)とEV(Dox-)の粒子数とタンパク質濃度に有意差は認められなかった。
図17にはD3.5上のEV処理されたControl-ESCにおけるFlk1 +細胞のFACS分析を示す。図18にはFlk1を発現するコントロールESCのパーセンテージを示し、Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。また、図53には分化3.5日目およびPKA-ESCの培地(Dox+またはDox-)からのEVで処理されたControl-ESCでのPDGFRα発現のFACS分析を示した。図54にはPDGFRα+細胞のパーセンテージを示し、Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。データは平均±S.Dを表す。
純粋なControl-ESC凝集体へのEV(Dox-)の添加は、EVまたはEV(Dox+)がない場合よりもはるかに高い割合のFlk1+およびPDGFRα+細胞を誘導することが示された。
図19には、D3.5のEV処理コントロール-ESCにおける中胚葉マーカー(Flk1、PDGFRα)の免疫染色を示す。データは平均±S.Dを表す。一貫して、免疫染色は、EV(Dox-)処理後にFlk1およびPDGFRαの発現の劇的な増加を示した。これらの結果は、活性化されたPKA-ESCからのEVが、Control-ESCから中胚葉への分化を促進して分化レベルを同期させることを示している。
ここで、図51には馴化培地中のEV粒子、図52にはEVに含まれるタンパク質の濃度を示す。これらの結果からは、EV(Dox+)とEV(Dox-)の粒子数とタンパク質濃度に有意差は認められなかった。

(EV由来のmiRによるPSyCの制御の検討)
発明者らはついで、次に、PSyCを誘発するEV内の分子を特定しようと試みた。
まず、EV(Dox+)とEV(Dox-)に含まれるグローバルmiR発現プロファイルをmiR-seqで比較した。
図20は、活性化PKA-ESC(Dox-)からのEVと不活性化PKA-ESC(Dox+)からのEVで差次的に発現するmiRNAをまとめたMAプロットを示す。これらの結果からは、活性化したDox-においては、miR-126、miR-132、miR-184、miR-193a、miR-212、およびmiR-483に富んでいることが判明した。
発明者らはついで、次に、PSyCを誘発するEV内の分子を特定しようと試みた。

図22は、1:3の比率での凝集体におけるFlk1+レシピエント細胞のパーセンテージを示し、Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析したものを示す。
これらの結果によると、レシピエント細胞(GFP-)におけるFlk1+中胚葉分化は、miR-132またはマルチmiR細胞株と混合した場合にのみ有意に促進された。

さらに、miR-132 KO PKA-ESCを生成することにより、レシピエント細胞の中胚葉誘導におけるEV転送miR-132の役割を確認した。
図55は、分化2.5日目のコントロール-ESCでのPDGFRα発現のFACS分析を示した。PKA-ESC、miR-132-KOPKA-ESCまたはコントロール(EVなし)の培地(Dox-)からのEVで処理したものである。
図56は、PKA-ESC、miR-132-KO PKA-ESCまたはコントロール(EVなし)の培地(Dox-)からのEVで処理された3.5日目のコントロール-ESCにおけるFlk1およびPDGFRαのRNA発現レベルを示した。値はコントロールに対して正規化されている。Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。
図57は、miR-132-PLGAの有無にかかわらず処理された3.5日目のコントロール-ESCにおけるFlk1のmRNA発現レベルを示した。値はコントロールに対して正規化されている。マンホイットニーU検定を使用してデータを分析した。
EVは、PKA活性化(Dox-)後のPKA-ESCまたはmiR-132 KO PKA-ESCの上清から調製し、分化中のレシピエント細胞に添加した。
これらの結果によると、Dox-(EV(Dox-))を含むPKA-ESCからのEVは、レシピエント細胞の中胚葉分化を促進したが(図17で示した結果と同様である)、miR-132 KO PKA-ESC(Dox-)に由来するEVは中胚葉分化を促進することはなかった。これらの結果は、EV(Dox-)のmiR-132が、レシピエント細胞の中胚葉分化の増強における主要なメッセンジャー分子であり、PSyCに寄与することを示している。
図55は、分化2.5日目のコントロール-ESCでのPDGFRα発現のFACS分析を示した。PKA-ESC、miR-132-KOPKA-ESCまたはコントロール(EVなし)の培地(Dox-)からのEVで処理したものである。
図56は、PKA-ESC、miR-132-KO PKA-ESCまたはコントロール(EVなし)の培地(Dox-)からのEVで処理された3.5日目のコントロール-ESCにおけるFlk1およびPDGFRαのRNA発現レベルを示した。値はコントロールに対して正規化されている。Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。
図57は、miR-132-PLGAの有無にかかわらず処理された3.5日目のコントロール-ESCにおけるFlk1のmRNA発現レベルを示した。値はコントロールに対して正規化されている。マンホイットニーU検定を使用してデータを分析した。
EVは、PKA活性化(Dox-)後のPKA-ESCまたはmiR-132 KO PKA-ESCの上清から調製し、分化中のレシピエント細胞に添加した。
これらの結果によると、Dox-(EV(Dox-))を含むPKA-ESCからのEVは、レシピエント細胞の中胚葉分化を促進したが(図17で示した結果と同様である)、miR-132 KO PKA-ESC(Dox-)に由来するEVは中胚葉分化を促進することはなかった。これらの結果は、EV(Dox-)のmiR-132が、レシピエント細胞の中胚葉分化の増強における主要なメッセンジャー分子であり、PSyCに寄与することを示している。
さらに、miR-132を搭載したEVを受け取った後のレシピエント細胞におけるPSyC誘導の細胞内分子機構を調査した。
図23は、キャピラリーウエスタンブロットにおけるSpry1、Rasa1およびpCREBの検出結果を示す。核画分のタンパク質を分析してSpry1とpCREBの発現レベルを測定し、Lamin A/Cで正規化した。細胞質画分のタンパク質を分析してRasa1の発現レベルを測定し、β-アクチンで正規化し、Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。
線維芽細胞成長因子経路のアンタゴニストであるSpry1、およびRasGAPアクチベーターであるRasa1は、miR-132の標的であることが知られる。図の結果からは、Control-ESCをEV(Dox-)で処理すると、Spry1およびRasa1タンパク質レベルが大幅に低下し、miR-132がEVを介した伝達を通じてControl-ESCで作用することを示している。
図23は、キャピラリーウエスタンブロットにおけるSpry1、Rasa1およびpCREBの検出結果を示す。核画分のタンパク質を分析してSpry1とpCREBの発現レベルを測定し、Lamin A/Cで正規化した。細胞質画分のタンパク質を分析してRasa1の発現レベルを測定し、β-アクチンで正規化し、Kruskal-Wallis検定とそれに続くSteel-Dwass検定を使用してデータを分析した。

総括すると、これらの結果は、PKA活性化後のPsyCにおける分子的関連を次のように示唆している。まず、PKA-ESCで開始されたPKAの活性化は、CREBリン酸化を介してFlk1+中胚葉細胞の分化とmiR-132の発現を促進する。ついで、活性化されたPKA-ESCは、miR-132の含有量が高いEVを分泌します。miR-132は、EVを介した転送を介してレシピエント細胞に送達され、Spry1とRasa1を抑制する。これにより、Control-ESCのCREBリン酸化を介してPKA経路が活性化される。したがって、元のPKA-ESCで開始されたPKAの活性化は、PKA-ESCから派生したEVを介してControl-ESCで再構成し、PSyCを呼び起こすことができる。これらの発見に続いて、PSyCは、ドナー由来のEVを介して近接したドナー細胞とレシピエント細胞の同様の細胞内環境を再構成する新しい生物学的メカニズムであると結論付けることができる。

ついで発明者らは、分化中のESCからのEVが胚発生中の細胞運命に影響を与えるかどうかを調べた。まず、EV(Dox-)が中胚葉期胚に及ぼす影響を確認した。
図27は、マウス胚培養の概略図を示す。E6.5マウス胚を収集し、EV(Dox-)処理を行ってex vivoで2日間培養した。胚を分離し、FACSによるex vivo培養の2日後に胚のPDGFRα+細胞を調た。

さらに、初期段階からの細胞運命決定に対するEV(Dox-)の影響を調べた。着床前から着床後まで連続的に胚を観察するために、Bedzhov,I.et al.,(2014) Nat.Protoc.,9,2732-9で開示されたex vivo培養システムを使用した。
図30に、マウス胚培養の概略図を示す。E3.5マウス胚を収集し、EVを使用してex vivoで培養した。子宮から分離されたE3.5マウス胚は、IVC1培地のマイクロスライド上で培養した。 2~3日目にマウス胚をスライドに付着させた後、培地をIVC2培地に交換し、胚を8日目または10日目まで培養した。EVは2日に1回添加した。
図31に、Dox-条件下でUDControl-ESCまたは分化PKA-ESCから単離されたEVで8日間培養されたE3.5マウス胚でのcTnTの免疫染色を示した。図32に、E3.5マウス胚を未分化ESC由来EV(EV(UD))または早期分化PKA-ESC由来EV(EV(Dox-))とともに10日間培養した際の、cTnT+胚のパーセンテージを示した。図33に、同様の培養での拍動胚のパーセンテージを示した。

これらの結果からは、予想困難なことであるが、初期の胚をEV(Dox-)で培養すると、胚の発生が劇的に変化した。心臓トロポニン-T陽性細胞を伴う心筋細胞の出現は、EV(Dox-)で処理された胚(17個の胚のうち5個)で増強されたが、未分化のControl-ESCから収集されたEV(EV(UD))で処理された胚では増強されず(19個中0個の胚)、未処理の胚でも僅かしか増強されなかった(21個中1個の胚)。
さらに、わずかなEV(UD)処理および未処理の胚(それぞれ40個中2個の胚と21個中0個の胚)でわずかな拍動またはけいれんが検出されたのに対し、明白かつ典型的な自発的拍動を伴う別個の細胞クラスターが、時には全胚で、EV(Dox-)処理胚で観察された(85個の胚のうち11個)。したがって、EV(Dox-)は、発生中の胚において、細胞を心筋細胞を含む中胚葉誘導体に変える可能性があることが示された。
さらに、わずかなEV(UD)処理および未処理の胚(それぞれ40個中2個の胚と21個中0個の胚)でわずかな拍動またはけいれんが検出されたのに対し、明白かつ典型的な自発的拍動を伴う別個の細胞クラスターが、時には全胚で、EV(Dox-)処理胚で観察された(85個の胚のうち11個)。したがって、EV(Dox-)は、発生中の胚において、細胞を心筋細胞を含む中胚葉誘導体に変える可能性があることが示された。
さらに、miR-132が同様に細胞運命決定に作用するかどうかを確認しました。この目的のために、生分解性ポリマーポリ(DL-ラクチド-co-グリコリド)(PLGA)から処方され、核酸を含むさまざまな分子を捕捉し、細胞膜に浸透し、カプセル化された内容物を細胞内、細胞質に送達できる人工高分子ナノ粒子を採用した。miR-132(miR-132-PLGA)を含むPLGAナノ粒子を配合した。前述の図57の結果より、分化中のマウスESCに追加されたmiR-132-PLGAは、中胚葉マーカーFlk1 mRNA発現のアップレギュレーションに成功し、miR-132-PLGAがEV(Dox-)の機能を模倣できることを示している。
図34は、miR-132-PLGAを用いてE3.5から10日間培養した鼓動するマウス胚を示し、図35は、8日目と10日目の胚の拍動のパーセンテージを示した。図36は、miR-132-PLGAの有無それぞれにおいて10日間培養されたE3.5マウス胚でのcTnTの免疫染色を示した。図37は、10日目のcTnT+胚のパーセンテージを示し、データは平均±S.Dを表す。
EV(Dox-)を添加した場合と同様に、miR-132-PLGAをE3.5マウス胚に添加した場合、8日目から心筋細胞の拍動が観察され、10日目まで数が増加した。コントロール(N=28)では3%である一方、miR-132-PLGAグループ(N=20)では25%に達していた。心筋細胞が拍動しているすべての胚は、心臓トロポニン-Tに対して陽性であった。したがって、miR-132は、in vivoで中胚葉および心筋細胞への細胞運命を変化させることに関して、EV(Dox-)と同じ可能性を示すことが示された。
EV(Dox-)を添加した場合と同様に、miR-132-PLGAをE3.5マウス胚に添加した場合、8日目から心筋細胞の拍動が観察され、10日目まで数が増加した。コントロール(N=28)では3%である一方、miR-132-PLGAグループ(N=20)では25%に達していた。心筋細胞が拍動しているすべての胚は、心臓トロポニン-Tに対して陽性であった。したがって、miR-132は、in vivoで中胚葉および心筋細胞への細胞運命を変化させることに関して、EV(Dox-)と同じ可能性を示すことが示された。

5mLチューブに1.0mLリンゴ酸緩衝液、15mL遠沈管に3.0mL PBS(-)を添加した。1mg/mLmiRNA溶液を10μLとリンゴ酸緩衝液114μLを1.5mLチューブに溶解した。
下記の脂質溶液を新しい5mLチューブに添加(ピペッティング)した。10mM SS-OP 75μL、10mM cholesterol 75μL(パラフィルムで密閉)1mM PEG-DMG 45μL(3mMの場合は15μLに調整可能)、EtOH 205μL(total 400μLになるように調整した)。
この脂質溶液をvortexミキサーで攪拌しながら(チューブの上を持つ)、核酸溶液を徐々に添加(滴下に10秒ほどかける)、終わっても3秒ほど攪拌し続けた。ついで、前記リンゴ酸緩衝液を攪拌しながら滴下した。
PBS(-)の入った1の15mlチューブをvortexミキサーで攪拌しながら、5の脂質/核酸溶液を徐々に滴下した(滴下に10秒ほどかけた)。全量をAmiconに移し、500μLのPBSで容器を洗い、洗浄液もロスが少ないようAmiconに移した。
ついで、1000g、室温、5minで延伸分離した。様子を見ながら何度か遠心、懸濁し、今回は×4回=20分を行った。
ついで、Amiconを取り出して下層を捨てた。4mL PBSを上層に足して、再度遠心する(20分ほど遠心し、最終溶液を100μL以下とした)。Amicon内のフィルターをPBSで何回か洗浄しながら、天秤を用いて質量(=容積)を測定し、1.0gにした(新しい1.5mlチューブを使用)。調整した脂質ナノ粒子(LNP)は4℃で保存した。
下記の脂質溶液を新しい5mLチューブに添加(ピペッティング)した。10mM SS-OP 75μL、10mM cholesterol 75μL(パラフィルムで密閉)1mM PEG-DMG 45μL(3mMの場合は15μLに調整可能)、EtOH 205μL(total 400μLになるように調整した)。

を準備した。
溶液を500mlナス型試料フラスコへ入れ、PVAが泡立つので分量外のMilli-Qを三角フラスコに入れ、泡がなくなるまでナスフラスコに入れていった。フラスコ+溶液の重量は243.64gであった。エバポレーターによる留去の操作を行い、終了後重量を測定(フラスコ+溶液)はすると240.88gであった。
使用機器として
エバポレーター:N-1200B, NVC-2200, CCA-1111, OSB-2100(回転数:150rpm, 減圧:120hPa 60min 、ついで 60hPa 20min)

(3:粗濾過)
減圧濾過用フィルターホルダー:KG-47、ADVANTEC、吸引濾過ビン:アズワンを用いて、5.0μmフィルター:1枚を介して減圧濾過した。

(4:洗浄)
濾過物を50mLチューブに分注し、12000G、時間:60min、温度:4℃で遠心分離した。上清をデカントで捨て、チューブにMilliQ水を10mL加えた。5min超音波にかけ、沈殿をMilliQに懸濁させた。遠心分離から懸濁までの操作をもう一度繰り返した。ついで、上清を捨て、Milli-Q水を5mL加えた。
濾過物を50mLチューブに分注し、12000G、時間:60min、温度:4℃で遠心分離した。上清をデカントで捨て、チューブにMilliQ水を10mL加えた。5min超音波にかけ、沈殿をMilliQに懸濁させた。遠心分離から懸濁までの操作をもう一度繰り返した。ついで、上清を捨て、Milli-Q水を5mL加えた。
(5:凍結乾燥)
凍結乾燥機:FDU-1200, EYELAを用い、試料フラスコとしてナス型 $29:300mL, 500mL, 1000mL EYELAを用いて、減圧:~10Pa,温度:-48℃,時間:over nightの条件で凍結乾燥を行った。
具体的な操作としては、50mlチューブを液体窒素につけ、回転させながら、予備凍結試料の凍結を確認し、凍結乾燥機に装着させたあと、Over nightで凍結乾燥した。
凍結乾燥機:FDU-1200, EYELAを用い、試料フラスコとしてナス型 $29:300mL, 500mL, 1000mL EYELAを用いて、減圧:~10Pa,温度:-48℃,時間:over nightの条件で凍結乾燥を行った。
具体的な操作としては、50mlチューブを液体窒素につけ、回転させながら、予備凍結試料の凍結を確認し、凍結乾燥機に装着させたあと、Over nightで凍結乾燥した。
得られたmiRNA-PLGA-NPの品質確認は、miRNA分子量を14,000として、

試薬として、以下のものを用いた。
・SS-OP, cholesterol, PEG-DMG・・・(前述の脂質ナノ粒子の作成方法にて作成)
・Biotin化PLGA・・・(前述のPLGA-NPの作成方法にて作成)

まず、前述のmiRNA封入脂質ナノ粒子(LNP)およびPLGAナノ粒子を作成した。
また、培養細胞を準備した。細胞はRAW264.7(P3),1.35×106cells/well(24wellプレート)、培地;DMEM(FBS10%,P/S1%)で培養した。
前述の作成したナノ粒子を、miRNAの濃度を揃えるため調整した。PLGAナノ粒子:1mg(粉末)をPBS60mlに希釈し、脂質ナノ粒子は原液そのままを使用した。
下記濃度に応じてmiRNA封入ナノ粒子またはmiRNA(naked)を培地にアプライした。例えば、70nM(miRNA濃度として)としたい場合、miRNAの分子量を14000、回収率を100%と仮定し、調整したLNP(またはPLGAナノ粒子)を培地500μLあたり50μL添加した。Naked miRNAの場合は1mg/mlに調整したものを培地500μLあたり0.5μL添加した。
24時間後にPBSで2回washし、細胞を回収した。
この細胞を、miRNA抽出、逆転写、リアルタイムPCRにて細胞内に導入されたmiR-132-3pを、naked miRNA, mir-LNP, mir-PLGA間で比較検討した(内部標準遺伝子:U6)。
また、培養細胞を準備した。細胞はRAW264.7(P3),1.35×106cells/well(24wellプレート)、培地;DMEM(FBS10%,P/S1%)で培養した。
前述の作成したナノ粒子を、miRNAの濃度を揃えるため調整した。PLGAナノ粒子:1mg(粉末)をPBS60mlに希釈し、脂質ナノ粒子は原液そのままを使用した。
下記濃度に応じてmiRNA封入ナノ粒子またはmiRNA(naked)を培地にアプライした。例えば、70nM(miRNA濃度として)としたい場合、miRNAの分子量を14000、回収率を100%と仮定し、調整したLNP(またはPLGAナノ粒子)を培地500μLあたり50μL添加した。Naked miRNAの場合は1mg/mlに調整したものを培地500μLあたり0.5μL添加した。
24時間後にPBSで2回washし、細胞を回収した。
この細胞を、miRNA抽出、逆転写、リアルタイムPCRにて細胞内に導入されたmiR-132-3pを、naked miRNA, mir-LNP, mir-PLGA間で比較検討した(内部標準遺伝子:U6)。
図59に、各サンプルごとのmiR-132-3p量の比較を示した。miRNA単独(naked)での投与ではトランスフェクションは成立しないが、ナノDDSによって用量依存的にmiRNAのトランスフェクションが可能であった。
[試験例3]
(心筋虚血再灌流マウスモデルに対するmiR-132-3p-LNP(Liposome)の投与)
マウスの心筋梗塞モデルを用い、miR-132-3p-LNPを投与する試験を行った。心筋梗塞モデルとしては、虚血-再灌流モデルを用いた。すなわち、いったん冠動脈をしばらく縛って虚血にし、その後血流を再開すると、再開後に活性酸素等様々な作用で心筋細胞死が起こるモデルである。臨床でいえば、心筋梗塞になって血管が詰まった後に病院に運ばれ、カテーテルで血流再開された、という状況に似せたモデルであり、心疾患の状況に対する投与の効果を検討することができる。再灌流時に対照のリポソームのみ、及びmiR-132含有リポソームを冠動脈から投与した。
(心筋虚血再灌流マウスモデルに対するmiR-132-3p-LNP(Liposome)の投与)
マウスの心筋梗塞モデルを用い、miR-132-3p-LNPを投与する試験を行った。心筋梗塞モデルとしては、虚血-再灌流モデルを用いた。すなわち、いったん冠動脈をしばらく縛って虚血にし、その後血流を再開すると、再開後に活性酸素等様々な作用で心筋細胞死が起こるモデルである。臨床でいえば、心筋梗塞になって血管が詰まった後に病院に運ばれ、カテーテルで血流再開された、という状況に似せたモデルであり、心疾患の状況に対する投与の効果を検討することができる。再灌流時に対照のリポソームのみ、及びmiR-132含有リポソームを冠動脈から投与した。
研究プロトコールは、九州大学医学部の動物実験倫理委員会によってレビューおよび承認され、米国生理学会のガイドラインおよび実験動物の管理と使用に関するNIHガイド第8版に従って実施された。
成体のオスのC57BL/6Jマウス(8~12週齢)(CLEA Japan、Inc、東京、日本)を使用した。動物は、通常の餌と水を自由に摂取させて、12時間の明暗サイクルの下で維持されていた。血圧と心拍数はテールカフ法(MK-2000、室町機械株式会社、東京、日本)で測定した。マウスに挿管し、2L/分の酸素中の2%イソフルランで麻酔した。心臓は、左開胸術および心膜切除術によって露出された。

心筋虚血は、左前下行枝(LAD)を、LADの横に配置されたシリコンチューブを備えた8-0ナイロン縫合糸を使用して結紮することによって誘発した。閉塞の30分後、血流を回復するためにシリコンチューブを取り外した(再灌流)。再灌流時に、100ngのmiRNAを含むmiRNA-LNP(mir132-LNPまたはmiRNA-negative control-LNP)をPBS(合計100microL)で希釈して尾静脈から注入した。処置後、マウスは完全に回復するまで加熱ボード上に置かれた。
マウスを2L/分の酸素中の2%イソフルランで麻酔し、再灌流の24時間後に挿管した。左前下行枝を結紮し、2%エバンスブルー染料(Sigma-Aldrich、セントルイス、ミズーリ州、米国)を下大静脈から注射した。次に、心臓を切除し、生理食塩水で灌流し、1mmの厚さで連続した断面に切断した。切片を1%2,3,5-トリフェニルテトラゾリウムクロリド(TTC、Sigma-Aldrich、セントルイス、ミズーリ州、米国)と37℃で10分間インキュベートした後、ホルムアルデヒドに入れ、実体顕微鏡(Nikon、HC-2500)で観察した。
TTCによる非染色LV領域を、梗塞領域と見なした。MI領域(TTCネガティブ、白)、AAR内の非MI領域(TTCポジティブ/エバンスブルーネガティブ、赤)、非虚血領域(TTCポジティブ/エバンスブルーポジティブ、紫)、およびAAR(エバンスブルーネガティブ)とし、これらをImageJソフトウェア(バージョン1.51h)を使用して定量化した。
図60に、心筋虚血再灌流マウスモデルに対するmiR-132-3p-LNP投与の顕微鏡写真図を示す。図中の左(a)がコントロールのLNP(すなわち、リポソームのみ)、右(b)がmiR132-LNP、すなわちmiR-132を100ng/100μLの割合で含有するものを投与した図である。左(a)は非染色(図中の白色、明色)部位が広く、梗塞領域が多いのに対して、右(b)は非染色領域が少ないことを示す。すなわち、マウスの心筋梗塞のモデルにおいて、miR-132を含有するリポソーム粒子を投与したものは心臓の梗塞・虚血領域が減少している結果が得られた。
図61に、染色領域の定量化の結果を示した。miR132-LNPでは、Control LNPに対して、Infarct Size(虚血の領域の大きさ)/Area at riskが大きく減少している結果が得られた。
図62に、染色領域の定量化についてN=6とした場合の結果を示した。N=6において、miR132-LNPではInfarct Size/Area at riskは有意に減少した。
[試験例4]
(心筋梗塞マウスモデルに対するmiR-132-3p-LNP(Liposome)の投与)
上述の心筋虚血再灌流マウスモデルと同様で、完全に冠動脈を縛り再灌流しないことで、心臓の異常がさらに重い状態(心筋梗塞に近い)を想定したモデルを用いて検討を行った。
(心筋梗塞マウスモデルに対するmiR-132-3p-LNP(Liposome)の投与)
上述の心筋虚血再灌流マウスモデルと同様で、完全に冠動脈を縛り再灌流しないことで、心臓の異常がさらに重い状態(心筋梗塞に近い)を想定したモデルを用いて検討を行った。
マウス10週齢 (C57BL/6J)について、冠動脈結紮後LNP静脈内投与、24時間後に心臓組織(虚血部位、非虚血部位、各30mg)を回収した。RNA later内で急速冷凍し、Tissue lyserでRNAを回収した。その他の操作については、試験例3と同様に行った。

内因性(マウスの心臓で発現される)miR132がどれくらいかを検討した。U6遺伝子(マウスに恒常的に発現しているmiRの一つ)の発現に比べて、miR132の発現がどれくらいか相対的な量を検討した。
図63にRT-PCRによるmiR-132発現量の比較を示した。U6遺伝子を1とした場合の発現量比で、n=3である。心筋梗塞後24時間で、非虚血部位(Non-ischemic area)がおよそ0.5前後であるのに対して、虚血部位(Ischemic area)ではmiR132はおよそ6倍に増加しているという結果が得られた。miR-132非投与(Control LNP投与)マウス冠動脈結紮24時間後の虚血領域で、内因性のmiR-132の発現亢進している。急性心筋梗塞の病態生理において、内因性のmiR-132が保護的な役割を果たしている可能性が示唆された。
図63にRT-PCRによるmiR-132発現量の比較を示した。U6遺伝子を1とした場合の発現量比で、n=3である。心筋梗塞後24時間で、非虚血部位(Non-ischemic area)がおよそ0.5前後であるのに対して、虚血部位(Ischemic area)ではmiR132はおよそ6倍に増加しているという結果が得られた。miR-132非投与(Control LNP投与)マウス冠動脈結紮24時間後の虚血領域で、内因性のmiR-132の発現亢進している。急性心筋梗塞の病態生理において、内因性のmiR-132が保護的な役割を果たしている可能性が示唆された。
ついで、リポソームを経静脈的に投与したときにリポソームが梗塞部位に送達できるか検討した。心筋梗塞モデルにmiR-132-Liposome静脈内投与し、24時間後の心臓組織において非虚血領域および虚血領域でmiR-132の発現量を調べることで、送達が行われるか確認した。
図64にリポソームを経静脈的に投与したときのRT-PCRによるmiR-132発現量の比較を示した。U6遺伝子を1とした場合の発現量比で、n=3である。miR-132-Liposomeと、コントロール(LNPのみ)を投与したもの、非虚血領域および虚血領域で合計4種のサンプルについて行っている。
Controlを投与した場合の発現量が、先のスライドの内因性miR132発現に相当すると考えられる。
非虚血(健常)部位にも虚血部位にも、内因性miR132の10-20倍ほどの量のmiR132が認められ、経静脈的投与でmiR132を心筋梗塞部位に送達できることが示された。
図64にリポソームを経静脈的に投与したときのRT-PCRによるmiR-132発現量の比較を示した。U6遺伝子を1とした場合の発現量比で、n=3である。miR-132-Liposomeと、コントロール(LNPのみ)を投与したもの、非虚血領域および虚血領域で合計4種のサンプルについて行っている。
Controlを投与した場合の発現量が、先のスライドの内因性miR132発現に相当すると考えられる。
非虚血(健常)部位にも虚血部位にも、内因性miR132の10-20倍ほどの量のmiR132が認められ、経静脈的投与でmiR132を心筋梗塞部位に送達できることが示された。
これらの結果から、心筋梗塞後、miR-132-Liposomeを静脈内投与し、24時間後の心臓組織において非虚血領域および虚血領域でmiR-132の送達を確認できた。Lipid Nanoparticleに封入して投与されたmiR-132が心臓組織に送達され、心筋梗塞の縮小に寄与したことが示された。
本発明は、心筋細胞の分化の促進に高い機能を発揮し、心疾患後の心機能改善に効果を有する、心筋分化を促進するための組成物および心筋分化を促進するための医薬組成物を提供することができる。
Claims (7)
- miR-132を有効成分として含有する、心筋分化を促進するための組成物。
- 粒子状である、請求項1に記載の組成物。
- 乳酸・グリコール酸共重合体(PLGA)ナノ粒子またはリポソームである、請求項2に記載の組成物。
- miR-132を有効成分として含有する、心疾患を治療または予防するための医薬組成物。
- 粒子状である、請求項4に記載の医薬組成物。
- 乳酸・グリコール酸共重合体(PLGA)ナノ粒子またはリポソームである、請求項5に記載の医薬組成物。
- 前記心疾患が心筋梗塞である、請求項4~6のいずれか1項に記載の医薬組成物。
