Production of lipid particles> siRNA was added to 1 mM citrate buffer (pH 4.0) to prepare an acidic siRNA aqueous solution (25°C, siRNA concentration: 71.4 nM). On the other hand, lipids ((lipid 1) DOP-DEDA, DOP-PPZ, or DOP-MPPZ, (lipid 2) dipalmitoylphosphatidylcholine (DPPC), and (lipid 3) cholesterol (Chol)) were mixed at a ratio of 45:10:45.
- RNA quantification reagent (RiboGreen reagent, manufactured by Thermo Fisher Scientific) was used. Specifically, it was carried out as follows. 2% Triton-X 100 or RNase free water was added to the lipid particle solution. The resulting solution, RNase free water, and RiboGreen reagent were mixed in wells of a 96-well black plate. After shaking the plate for 5 minutes, the fluorescence intensity of each well was measured.

- ⁇ Test Example 3-2 Toxicity Evaluation Test> MDA-MB-231 human breast cancer cells were seeded in a 96-well plate (7 ⁇ 10 3 cells/well) and cultured at 37° C. for 24 hours. Lipid particle solution (containing 0.6/2/6 pmol of siRNA) or lipid complex solution (complex prepared using Lipofectamine® 2000 (manufactured by Thermo Fisher Scientific), containing 0.6/2/6 pmol of siRNA) was added dropwise to the wells and cultured at 37°C for 96 hours. Cytotoxicity of lipid particles and lipid complexes was evaluated using a Viability/Cytotoxicity Multiplex Assay Kit (Dojindo Laboratories).

The amount of PLK1 mRNA was quantified by real-time PCR using cDNA as a template and TB Green (registered trademark) Premix Ex Taq (trademark) II (manufactured by Takara Bio Inc.).

項13. 項11又は12に記載のアルコール溶液と酸性水溶液とを混合する工程を含む、脂質粒子の製造方法。
本発明は、その一態様において、一般式(1):
で表されるリン脂質(本明細書において、「本発明のリン脂質」と示すこともある。)に関する。以下に、これについて説明する。
に従って又は準じて合成することができる。
本発明は、その一態様として、本発明のリン脂質(本明細書において、「リン脂質A」と示すこともある。)を含有する、脂質粒子(本明細書において、「本発明の脂質粒子」と示すこともある。)に関する。以下に、これについて説明する。
本発明は、その一態様として、本発明の脂質粒子を含有する、医薬(本明細書において、「本発明の医薬」と示すこともある。)に関する。また、本発明の脂質粒子は、試薬としても利用することができる。
1,2-di-(9Z-octadecenoyl)-sn-glycero-3-phosphoethylpiperazine)(DOP-PPZ)、及び1,2-di-(9Z-octadecenoyl)-sn-glycero-3-phosphomethylpiperazine(DOP-MPPZ)を、以下のスキームに従って合成した。
3.01 g(3.82 mmol)のDOPCを酢酸エチルに溶解させ、その溶液に6.90 gの1-(2-Hydroxyethyl) piperazine(53 mmol)を溶解させたpH5.5の0.5 M 酢酸酸緩衝液を加えて、40℃に加温した。加温後、PLDP(旭化成ファーマ社製、ホスホリパーゼD)(1440 U)を加え、40℃で撹拌した。19時間後、TLCにてDOPCの消失を確認した。なお、1ユニットは、至適条件下(温度30℃で、最も化学反応が進む酸性度)で毎分1マイクロモル(μmol)の基質を変化されることができる酵素量(1マイクロモル毎分)と定義される。
1.00 g(1.27 mmol)のDOPCを酢酸エチルに溶解させ、その溶液に2.55 gの4-Methylpiperazine-1-ethanol(17.7 mmol)を溶解させたpH5.5の0.5 M 酢酸酸緩衝液を加えて、40℃に加温した。加温後、PLDP(旭化成ファーマ社製、ホスホリパーゼD)(480 U)を加え、40℃で撹拌した。19時間後、TLCにてDOPCの消失を確認した。なお、1ユニットは、至適条件下(温度30℃で、最も化学反応が進む酸性度)で毎分1マイクロモル(μmol)の基質を変化されることができる酵素量(1マイクロモル毎分)と定義される。
DOP-DD(DOP-DEDA)を以下のスキームに従って合成した。具体的には、特許文献1に記載の方法に従って合成した。
日本薬局方通則に基づいてエタノール溶解性試験を実施した。
<試験例2-1.脂質粒子の製造>
siRNAを1 mMクエン酸緩衝液(pH 4.0)に添加して、siRNA酸性水溶液を調製した(25℃、siRNA濃度:71.4 nM)。一方で、脂質((脂質1)DOP-DEDA、DOP-PPZ、又はDOP-MPPZ、(脂質2)ジパルミトイルホスファチジルコリン(DPPC)、及び(脂質3)コレステロール(Chol))を45:10:45のモル比(脂質1:脂質2:脂質3)でエタノールに添加して、リン脂質のアルコール溶液を調製した(25℃、脂質濃度:2.5 mM)。リン脂質アルコール溶液に対して5倍容量のsiRNA酸性水溶液を使用し(siRNA/脂質モル比=1/7000)、マイクロ流路(KeyChem-Basic、ワイエムシィ社製)を用いて、脂質粒子を得た。最後にエタノールを透析によって除去した。 <試験例2-2.各種物性値の測定>
脂質粒子をRNase free waterで50倍希釈した後、ゼータサイザーナノZS(Malvern社製)を用いて粒子径と多分散指数(PDI)を測定した。また、脂質粒子を緩衝液(pH=4.0、5.0、6.0、又は7.0)で50倍希釈した後、ζ-Potentialを測定した。
<試験例3-1.脂質粒子の製造>
実施例2-1と同様にして脂質粒子を製造した。
MDA-MB-231ヒト乳がん細胞を96ウェルプレートに播種(7×103 cells/ウェル)し、37℃で24時間培養した。脂質粒子溶液(siRNA 0.6/2/6 pmolを含む)又は脂質複合体溶液(Lipofectamine(登録商標) 2000(Thermo Fisher Scientific製)を用いて調製した複合体、siRNA 0.6/2/6 pmolを含む)をウェルに滴下し、37℃で96時間培養した。脂質粒子及び脂質複合体の細胞毒性は、Viability/Cytotoxicity Multiplex Assay Kit(同仁化学研究所)を用いて評価した。陽性対照群用のウェルにはLysis buffer 20 μLを滴下し、37℃で30分間インキュベートした。各ウェルの上清100 μLを96ウェルクリアプレートに移した後、上清にキットのLDH assay reagent液を100 μL添加し、室温で30分間インキュベートした。その後Stop solution 50 μLを加え、吸光度(absorbance 450nm)を測定した。また、細胞が付着したプレートから培地を除去し、各ウェルにWST-8 assay reagent液(Cell Counting Kit: 培地 = 1:9)を120 μL添加した。37℃で5時間インキュベートした後、吸光度(absorbance 450nm)を測定した。
<試験例4-1.脂質粒子の製造>
脂質1としてDOP-PPZを用いて且つsiRNAとしてPLK1遺伝子を標的とするsiRNA又は非特異的siRNA(si Control)を使用する以外は試験例2-1と同様にして、脂質粒子を製造した。
MDA-MB-231ヒト乳がん細胞を6ウェルプレートに播種(2×105 cells/ウェル)し、37℃で24時間培養した。脂質粒子溶液(siRNA 2/18 pmol含む)又はRnase Free Water(Control)を各ウェルに滴下し、37℃で12時間培養した。培地交換し、さらに37℃で60時間培養した。QIAshredder及びRNeasy Mini Kit(QIAGEN社製)を用いて、細胞からtotal RNAを抽出した。First-Strand cDNA Synthesis Kit(GE Healthcare Life Sciences社製)を用いて、total RNAからcDNAを合成した。cDNAを鋳型として、TB Green(登録商標) Premix Ex Taq(商標) II(タカラバイオ社製)を用いて、real-time PCRによりPLK1 mRNA量を定量した。

- 前記鎖式炭化水素基が不飽和鎖式炭化水素基である、請求項1に記載のリン脂質。
- 前記鎖式炭化水素基の炭素数が11~23である、請求項1又は2に記載のリン脂質。
- 前記mが2である、請求項1~3のいずれかに記載のリン脂質。
- 前記R3が水素原子又はアルキル基である、請求項1~4のいずれかに記載のリン脂質。
- 請求項1~5のいずれかに記載のリン脂質(リン脂質A)を含有する、脂質粒子。
- 薬物を内包する、請求項6に記載の脂質粒子。
- 前記薬物がポリヌクレオチドである、請求項7に記載の脂質粒子。
- ステロールを含有する、請求項6~8のいずれかに記載の脂質粒子。
- 前記リン脂質Aが不飽和鎖式炭化水素基を有するリン脂質であり、且つ、さらに、前記リン脂質A以外のリン脂質(リン脂質B)を含有する、請求項6~9のいずれかに記載の脂質粒子。
- 請求項1~5のいずれかに記載のリン脂質を含有する、アルコール溶液。
- 前記アルコールがエタノールである、請求項11に記載のアルコール溶液
- 請求項11又は12に記載のアルコール溶液と酸性水溶液とを混合する工程を含む、脂質粒子の製造方法。
- 請求項6~10のいずれかに記載の脂質粒子を含有する、医薬。
