Abstract
본 발명은 Diaportheone A1, Diaportheone A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물에 관한 것이다. 보다 상세하게는, 본 발명의 약제학적 조성물을 사용하는 경우, 아밀로이드 베타(amyloid beta, Aβ)의 응집을 억제하고, 활성산소종(reactive oxygen species, ROS)을 감소시키며, 미토콘드리아 막 전위의 손실을 감소시킴으로써, 산화 스트레스에 대한 신경보호 효과가 있어, 알츠하이머병의 예방 또는 치료에 사용될 수 있다.
Description
본 발명은 대한민국 과학기술정보통신부의 지원 하에서 과제고유번호 1711115820에 의해 이루어진 것으로서, 상기 과제의 연구관리전문기관은 한국연구재단, 연구사업명은 "개인기초연구(과기정통부)(R&D)", 연구과제명은 "한국형 알츠하이머병 관련 돌연변이의 병인적 메커니즘 및 바이오마커의 multi variate 분석과 신약 개발 위한 한국형 세포와 동물 모델 개발 연구”, 주관기관은 가천대학교, 연구기간은 2020.03.01.-2023.02.28.이다.
본 특허출원은 2021년 02월 16일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2021-0020749호에 대하여 우선권을 주장하며, 상기 특허출원의 개시사항은 본원에 참조로서 삽입된다.
본 발명은 천연물에서의 신약 발견에 관한 것으로, 보다 상세하게는 Diaportheone A1, Diaportheone A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물에 관한 것이며, 또한 상기 약제학적 조성물은 알츠하이머병의 예방 또는 치료에 사용된다.
크로몬(Chromone)은 벤조아닐화된 γ-피론 헤테로고리 화합물(benzoannelated γ-pyrone heterocyclic compound)로 자연에 널리 분포되어 있다. 천연물과 크로몬 구조를 갖는 합성물 모두에서 상당수의 화합물이 항염증성, 항균제, 항종양, 항당뇨, 항알레르기, 이뇨제, 저혈당, 지질 저하, 항산화 등 다양한 약리학적 특성을 보인다. 이들의 구조적 다양성과 합성 접근성 덕분에 크로몬 스캐폴드(scaffold)를 가진 화합물은 유기 합성에서 탁월한 표적이며 약물 발견과 의약 화학에서 중요한 역할을 한다.
알츠하이머병(Alzheimer's disease, AD)은 노인들 사이에서 가장 흔한 신경 퇴행성 질환으로 기억력 상실, 치매 및 지속적인 인지 저하를 특징으로 한다. 전 세계적으로 4,700만 명의 노인이 알츠하이머병을 앓고 있으며, 2050년에는 1억 1,300만 명으로 늘어날 것으로 추정된다. 알츠하이머병의 발병 기전은 아직 완전히 밝혀지지 않은 유전적 및 생화학적 요인의 복잡하고 상호 연결된 네트워크를 포함한다. 비정상적인 아밀로이드 베타(Amyloid beta, Aβ), 산화 스트레스 및 신경 염증은 AD와 관련된 병리학적 특성 중 일부이다. 알츠하이머병에 대한 치료법은 아직 없지만, 발명의 조기 단계에서의 전략적 치료가 유익한 것으로 입증되었다. 현재 미국 FDA가 알츠하이머병과 관련된 증상의 진행을 최소화하기 위해 승인한 약은 5개뿐이다. 이들은 아세틸 콜린에스테라제(acetyl cholinesterase) 억제제인 갈란타민(galantamine), 리바스티그민(rivastigmine), 도네페질(donepezil), 타크린(tacrine), NMDA 수용체 길항제 메만틴(memantine)을 포함하는 천연 제품 기반 화합물이다. 이러한 약물은 알츠하이머병의 증상에 대한 증상 완화를 제공하지만 질병 진행 및 예방에 유의한 효과를 보이진 않는다. 따라서 대체 치료 또는 치료법을 찾는 데에는 어려움이 있다.
Diaportheone A1 및 Diaportheone A2는 천연물 Diaportheone A의 합성 유사체이다. 이러한 거울상 이성질체 화합물은 Diaportheone A의 전체 합성 과정에서의 중간 화합물이다. Diaportheone A1 및 Diaportheone A2는 99% ee(enantiomeric excess, 거울상 이성질체 과잉) 이상에서 얻어져 분리되었으며 X선 결정학에 의해 절대 배열이 결정되었다(Tan et al., 2019). 문헌 검색에 따르면 Diaportheone A1 및 A2의 구조와 관련된 분자 표적은 없다.
본 발명자들은 알츠하이머병의 증상 완화, 질병 진행 억제, 예방 및 치료에 유의한 효과를 제공하는 신약을 천연물에서 탐색 및 발굴하여 이를 포함하는 약제학적 조성물을 개발하고자 예의 연구 노력하였다. 그 결과, 크로몬(chromone) 구조를 갖는 두 이성질체인 Diaportheone A1 및 Diaportheone A2가 활성산소종(ROS) 감소 및 미토콘드리아 막 전위 손실의 감소와 같은 신경보호 효과와 아밀로이드 베타(Aβ)의 응집을 억제하는 항-아밀로이드 생성 효과를 가지는 화합물임을 규명함으로써, 본 발명을 완성하게 되었다.
본 발명의 목적은 Diaportheone A1, Diaportheone A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 Diaportheone A1, Diaportheone A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 신경보호에 의한 알츠하이머병의 예방 또는 치료용 약제학적 조성물을 제공하는 것이다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 본 발명은 Diaportheone A1, Diaportheone A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 신경보호에 의한 퇴행성 신경질환의 예방 또는 치료용 약제학적 조성물로서, 상기 Diaportheone A1은 하기 화학식 1로 표기되고, 상기 Diaportheone A2는 하기 화학식 2로 표기되는 것인, 약제학적 조성물을 제공한다:

본 발명자들은 알츠하이머병의 증상 완화, 질병 진행 억제, 예방 및 치료에 유의한 효과를 제공하는 신약을 천연물에서 탐색 및 발굴하여 이를 포함하는 약제학적 조성물을 개발하고자 예의 연구 노력하였다. 그 결과, 크로몬(chromone) 구조를 갖는 두 이성질체인 Diaportheone A1 및 Diaportheone A2가 활성산소종(ROS) 감소 및 미토콘드리아 막 전위 손실의 감소와 같은 신경보호 효과와 아밀로이드 베타(Aβ)의 응집을 억제하는 항-아밀로이드 생성 효과를 가지는 화합물임을 규명하였다.

본 명세서에서 Diaportheone A 및 B는 열대 식물인 판다누스 아마릴리폴리우스(Pandanus amaryllifolius)의 내생 진균(endophytic fungi)인 Diaporthe sp. P133으로부터 분리되거나 본 발명이 속하는 분야에 공지된 방법(Tan et al., 2019; 및 Bungihan et al., 2011 참조)에 의해 합성될 수 있다. 또한, 본 명세서의 Diaportheone A 및 B는 포유동물에서 일반적으로 결핵을 일으키는 병원성 균주, 마이코박테리움 튜버쿨로시스(Mycobacterium tuberculosis) H37Rv의 성장을 각각 최소 억제 농도(minimum inhibitory concentration, MIC) 100.9 μM 및 3.5 μM에서 억제하는 항-결핵 활성을 갖는다고 공지되어 있다(Bungihan et al., 2011 참조).
본 발명의 화합물, "Diaportheone A1"은 본 명세서의 천연물 Diaportheone A의 합성 유사체이고 Diaportheone A의 8번 탄소에 연결된 하이드록시기(hydroxyl group)가 메톡시기(methoxy group)로 치환된 구조를 가지며 1번 탄소의 절대 배열이 S인 화합물로서, Diaportheone A2와 거울상 이성질체 관계에 있는, 다음의 화학식 1로 표시되는 화합물을 의미한다:

도 7은 Aβ1-42-유도 SH-SY5Y 세포에서의 Diaportheone A1 및 Diaportheone A2의 미토콘드리아 막 전위(mitochondrial membrane potential, ΔΨm) 분석 결과를 나타낸다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실시예 1. Diaportheone A1 및 Diaportheone A2의 합성
이전에 보고된 방법(Tan, M.A. et al., 2019)을 참조하여 본 발명의 두 거울상 이성질체인 Diaportheone A1 및 Diaportheone A2를 합성하였다. 보다 상세하게는 Diaportheone A1 및 A2는 Diaportheone A의 전체 합성에서 라세미 혼합물인 합성 중간체로 제조되었다(4 단계, 전체 수율 56%). 합성은 97% 수율로 BCl3를 사용하여 상업적으로 입수 가능한 메틸-2,6-디메톡시벤조에이트(methyl-2,6-dimethoxybenzoate)의 모노-디메틸화로부터 개시되었다. 모노-디메틸화된 벤조에이트 생성물은 소듐 메틸설파닐메타이드(sodium methylsulfinylmethide)를 사용하여 황색 결정으로서 67.2% 수율의 β-케토설폭사이드(β-ketosulfoxide)로 전환되었다(5당량 NaH/25당량 DMSO, 70℃, 1시간). 37% 숙신알데히드(succinaldehyde) 및 촉매량의 메틸 피페리딘(methyl piperidine)과 함께 β-케토설폭사이드를 처리함으로써 원-팟 고리화(one-pot cyclization) 및 in situ 열 신-제거(thermal syn-elimination)를 통해 86% 수율로 diaportheone A1 및 A2의 라세미 혼합물 얻었다. 라세미 혼합물은 Chiralpak IG 컬럼을 이용하여 분취(preparative) SFC 방법으로 분리되었다. 두 거울상 이성질체 모두 99% ee 초과를 나타내었고 각각의 절대 배열은 X-선 결정학에 의해 분명하게 결정되었다(Tan MA et al., 2019).
상기에서 얻은 Diaportheone A1 및 A2를 EtOH(에탄올), MeOH(메탄올), EtOAc(에틸 아세테이트), CHCl3, CH2Cl2, 또는 DMSO(dimethyl sulfoxide) 용매에 용해하여 저장 용액(stock solution)을 제조하였고, 최종 농도가 1 μM, 10 μM, 20 μM 및 50 μM이 되도록 DMEM(Dulbecco's Modified Eagle Medium) 용액으로 희석하여 이하의 실시예에서 시험물질로 사용하였다. 용매만을 포함하는 조성물을 대조군으로 사용하였다.
실시예 2. SH-SY5Y 세포 배양(cell culture) 방법
신경모세포종 SH-SY5Y 세포(neuroblastoma, ATCC CRL-2266)는 10% FBS, 1% 카나마이신(kanamycin) 및 1% 항생제-항균제(antibiotic-antimycotic)가 보충된 DMEM에서 37℃ 및 5% CO2에서 배양되었고, 일주일에 2회 계대 배양되었다. 이하의 실시예에서는 실시예 2의 방법으로 배양된 SH-SY5Y 세포가 80% 내지 90%의 세포밀집도(cell confluency)에 도달한 시점에서 실험을 수행하였다.
실시예 3. 본 발명의 시험물질 2종 자체의 세포독성 확인
신경보호 실험에 적용될 수 있는 적절한 농도를 결정하기 위해 ATP 발광 분석을 사용하여 SH-SY5Y 세포에서 본 발명의 시험물질 2종 자체의 세포독성을 확인하였다.
세포 생존율(cell viability)은 다음과 같은 ATP 발광 분석(ATP luminescence assay)을 통해 측정되고 분석되었다. 2 Х 104 세포/well의 SH-SY5Y 세포를 96-웰 플레이트에서 계대 배양한 후, 24시간 동안 배양하였다. 이후, 세포에 24시간 동안 상술한 실시예 1의 시험물질을 처리하였다. 배지(media)를 제거하고, 세포를 PBS로 세척(washing)한 뒤, 새로운 배지를 첨가하고, 추가로 30분 동안 배양하였다. 그런 다음, CellTiter-Glo® luminescent 시약을 첨가하고 발광을 다중-플레이트 판독기(multi-plate reader)에서 확인하였다. 데이터를 분석하고 세포 생존율 퍼센트(%)를 대조군에 비교하였다. 세포 생존율 퍼센트(%)는 3번의 실험을 반복한 값의 평균 ± SD(standard deviation)로 표현되었고, 대조 세포의 값을 사용하여 정규화하였다. 결과는 도 1에 나타내었다.
도 1에 나타낸 바와 같이, 실시예 1의 시험물질 2종, Diaportheone A1 및 Diaportheone A2는 SH-SY5Y 세포에서 50 μM 농도까지 독성이 없는 것으로 나타났다. 따라서, 이하의 실시예에서는 1 μM, 10 μM 및 20 μM 농도를 적용하여 신경보호 실험을 수행하였다.
실시예 4. 아밀로이드 베타(Aβ) 응집의 억제 효과
본 발명의 시험물질 2종이 아밀로이드 베타 1-42의 응집을 억제할 수 있는지 여부를 평가하기 위해 티오플라빈 T(Thioflavin T, ThT) 분석을 수행하였다.
티오플라빈 T 분석은 다음과 같이 수행하였다. PBS(phosphate buffered saline)에 용해한 아밀로이드 베타(Aβ1-42, AggreSureTM)를 384-웰 플레이트(384-well plate)에서 37℃로 24시간 동안 상술한 실시예 1의 시험물질 또는 페놀 레드(phenol red, 양성 대조군)와 함께 혹은 함께하지 않고 배양하였다. ThT 용액(Sigma Aldrich)을 첨가하고 추가로 15분 동안 배양하였다. 형광 신호(excitation wavelength(Ex, 여기 파장): 450 nm; emission wavelength(Em, 방사 파장): 510 nm)는 마이크로-플레이트 판독기(PerkinElmer Victor-3® micro-plate reader)를 사용하여 측정되었다. 실험의 결과로 diaportheone A1 및 A2의 아밀로이드-베타 응집 억제율(%)을 아래 표 1에 나타내었다.
표 1에 나타낸 바와 같이, ThT 분석 결과, 양성 대조군인 페놀 레드의 아밀로이드 베타 응집 억제율 65.78% ± 2.97(50 μM)과 비교하여 실시예 1의 diaportheone A1은 80.41% ± 1.4(50 μM), diaportheone A2는 73.68% ± 1.7(50 μM)로 아밀로이드 베타 응집 억제율을 나타내 유의한 차이(p <0.05)가 있었다.

상기 결과로부터, 본 발명자들은 본 발명의 시험물질 2종이 아밀로이드 베타의 응집을 억제하는 효과를 확인하였다.
실시예 5. 아밀로이드 베타(Aβ) 올리고머 생성의 저해 효과
본 발명의 시험물질 2종이 아밀로이드 베타 1-42 올리고머의 생성을 저해할 수 있는지 여부를 평가하기 위해 MDS(Multimer Detection System) 분석을 수행하였다.
MDS 분석 방법은 다음과 같다. 모노머(monomer) 상태의 Aβ와 실시예 1에 따른 시험물질 2종을 마이크로플레이트(microplate)에 첨가하여 상온에서 인큐베이션(incubation)하였다. 시간에 따른 Aβ 올리고머(oligomer)의 생성 증가를 모니터링하기 위하여 0시간, 2시간 및 4시간에 혼합물을 옮겨서 -80℃에 보관하였다. Aβ를 타겟으로 하는 6E10 항체를 플레이트에 코팅(coating)한 뒤에 BSA(bovine serum albumin)가 포함된 블로킹 버퍼(blocking buffer)를 처리하였다. 시간별로 얼려 두었던 Aβ와 약물 혼합물을 Aβ 올리고머를 검출하기 위하여, 6E10 항체와 중복되는 Aβ 에피톱(epitope)을 인식하여 이에 부착하는 HRP 컨쥬게이션 된 WO2 항체(horseradish peroxidase conjugated WO2 antibody)를 처리하였다. TMB(tetramethylbenzidine) Solution을 첨가하여 30분 동안 반응한 뒤 Stop Solution을 넣어 반응을 종결시켰다. 마이크로플레이트 판독기(microplate reader)를 통해 흡광도를 측정하여 Aβ 올리고머의 값을 비교하였다. MDS 데이터는 2번의 실험을 반복한 값의 평균으로 나타내었다. 결과는 도 2에 나타내었다.
도 2에 나타낸 바와 같이, MDS 분석 결과, 아밀로이드 베타 1-42만 처리한 음성 대조군은 4시간대에서 1.963 ± 0.219의 올리고머 신호(oligomer signal)를 나타냈다. 실시예 1에 따른 Diaportheone A1의 올리고머 신호는 1.697 ± 0.026이었으며, Diaportheone A2의 올리고머 신호는 1.433 ± 0.067로 나타났다.
상기 결과로부터, 본 발명자들은 본 발명의 시험물질 2종이 아밀로이드 베타 1-42 올리고머의 생성을 직접적으로 저해함을 확인하였다.
실시예 6. Aβ 또는 H2O2에 대한 신경세포 보호 효과 - 세포 생존율
본 발명의 시험물질 2종의 신경보호 가능성은 Aβ 또는 H2O2로 SH-SY5Y 세포를 손상시킴으로써 확인하였다. 실시예 2와 같은 방법으로 신경아세포종 SH-SY5Y 세포를 준비하여, 준비된 신경모세포종 SH-SY5Y 세포(neuroblastoma SH-SY5Y cell, ATCC CRL-2266)를 2 Х 104 세포/well로 계대 배양하고 24시간 동안 배양하였다. 안정화시킨 후, 세포는 아밀로이드 베타 1-42(5 μM) 또는 H2O2(100 μM)와 함께 배양하기 전에 6시간 동안 본 발명의 화합물로 전처리하였다. 용매 대조군(본 발명의 시험물질을 처리하지 않은 대조군 세포), 아밀로이드 베타 또는 H2O2 단독, 및 본 발명의 시험물질만 처리한 실험군 또한 실험하였다. 세포생존율은 실시예 3에 기재된 ATP 발광 분석 방법으로 측정하였고, 세포 생존율 퍼센트(%)는 세 번의 실험을 통해 결정되었다. 결과는 도 3 및 도 4에 나타내었다.
1) H2O2-유도 세포 손상에 대한 신경 보호 효과
도 3에 나타낸 바와 같이, 본 발명의 시험물질 2종, Diaportheone A1 및 Diaportheone A2 모두 H2O2-유도 SH-SY5Y 세포에서 산화 스트레스에 대한 신경보호 효과를 보였다. H2O2 단독 처리군과 비교할 때, H2O2를 처리한 세포에 Diaportheone A1을 전처리한 경우 20 μM 농도에서, Diaportheone A2를 전처리한 경우 20 μM 및 10 μM 농도에서 세포 생존율(%)에 대한 통계적으로 유의한 차이(*: p <0.05)를 나타내 이들의 신경보호 효과를 확인하였다.
2) Aβ-유도 세포 손상에 대한 신경 보호 효과
도 4에 나타낸 바와 같이, 실시예 1의 시험물질 2종, Diaportheone A1 및 Diaportheone A2는 아밀로이드 베타-유도 SH-SY5Y 세포에서 Diaportheone A1를 전처리한 경우 20 μM 및 10 μM 농도에서, Diaportheone A2를 전처리한 경우 20 μM 농도에서 아밀로이드 베타만 처리된 세포에 비해 세포 생존율(%)에 대한 통계적으로 유의한 차이(*: p <0.05)를 나타내 이들의 신경보호 효과를 확인하였다.
실시예 7. 신경세포 보호 기작의 확인 - 활성 산소종의 생산 저해
실시예 1에 따른 시험물질 2종이 보이는 신경보호 효과의 가능한 작용 기작을 결정하기 위해 활성산소종(Reactive Oxygen Species, ROS)의 수준을 측정하였다. ROS 수준은 산화 스트레스에 대한 생화학적 마커로서 메커니즘적인 고려사항이다. ROS 수준은 아래에 기재한 바와 같이 ROS-민감성 H2DCFDA(2',7'-dichlorodihydrofluorescein diacetate) 염색법을 사용하여 측정하였다.
활성산소종(Reactive Oxygen Species, ROS)의 측정은 이전에 보고된 대로 H2DCFDA(2',7'-dichlorodihydrofluorescein diacetate) 염색 방법을 사용하여 수행되었다(Gonzalez-Sarrias et al., 2016; 및 Peρalver et al., 2020 참조).
실시예 2의 방법으로 배양한 SH-SY5Y 세포의 24시간 안정화 후, 산화 스트레스를 세포에 가한 경우(산화 스트레스 있음, with oxidative stress), SH-SY5Y 세포(2 Х 104 세포/well)를 2시간 동안 실시예 1의 시험물질로 전처리한 후 100 μM의 H2O2로 추가 4시간 동안 배양하였다. 산화 스트레스를 세포에 가하지 않은 대조 세포의 경우(산화 스트레스 없음, no oxidative stress), 본 발명의 실시예 1의 시험물질을 처리하여 세포를 6시간 동안 배양하였다. 이후 상기 세포에 25 μM의 H2DCFDA로 처리하고 37℃에서 빛을 차단시킨 채 2시간 동안 추가 배양하였다. 형광 강도(Ex: 495 nm; Em: 520 nm)는 마이크로플레이트 판독기(microplate reader)에서 측정되었다. ROS 수준은 3회 반복 측정되었으며, 본 발명의 시험물질을 처리하지 않은 대조군 세포의 백분율(100%)로 계산되었다. 그래프의 데이터는 세 번 반복 시행한 값의 평균 ± SD를 나타내며, * 표시는 H2O2만을 처리한 세포와의 유의미한 차이(p <0.05)를 보이는 것을 의미한다. 결과는 도 5 및 도 6에 나타내었다.
도 5 및 도 6에 각각 나타낸 바와 같이, 본 발명의 시험물질 2종, Diaportheone A1 및 Diaportheone A2를 처리한 실험군에서 H2O2만을 처리한 세포 대비 20 μM 농도에서 H2O2-유도 SH-SY5Y 세포에서의 세포 내 활성산소종(ROS) 수준이 감소하였음을 확인하였다.
상기 결과로부터 본 발명의 시험물질 2종은 H2O2-처리 SH-SY5Y 세포에서 ROS 생성을 감소시킴으로써 신경세포 보호 효능을 나타냄을 알 수 있었다.
실시예 8. 신경세포 보호 기작의 확인 - 미토콘드리아의 막 전위 손실의 억제
실시예 1에 따른 시험물질 2종이 보이는 신경보호 효과의 가능한 작용 기작을 결정하기 위해 미토콘드리아 막 전위를 측정하였다. 미토콘드리아 막 전위(mitochondrial membrane potential, MMP, ΔΨm)의 분석은 이전에 공지된 바(Alvariρo et al., 2019)와 같이 제조업체의 프로토콜에 따라 TMRE(tetramethylrhodamine, methyl ester) 염색 방법을 사용하여 수행되었다. SH-SY5Y 세포(2 Х 104 세포/well)를 24시간 동안 순응화(acclimatization) 한 후, 세포를 2시간 동안 실시예 1의 시험물질 2종으로 전처리하여 24시간 동안 5 μM의 아밀로이드 베타 1-42와 함께 인큐베이션 하였다. 이후 1 μM의 TMRE 염색 용액을 첨가하고 30분 동안 37℃에서 배양하였다. 형광 강도(Ex: 549 nm; Em: 575 nm)를 마이크로플레이트 판독기(microplate reader)에서 확인하였다. ΔΨm은 3회 반복 측정하였으며 본 발명의 화합물을 처리하지 않은 대조군 세포의 백분율(100%)로 계산되었다. 그래프의 * 표시는 Aβ가 처리된 세포와 비교할 때, p <0.05인 경우를 의미하며, 데이터는 세 번 반복 시행한 값의 평균 ± SD를 나타낸다. 결과는 도 7에 나타내었다.
도 7에 나타낸 바와 같이, TMRE(tetramethylrhodamine, methyl ester) 염색 키트를 사용하여 미토콘드리아 막 전위를 분석한 결과, Aβ만을 처리한 세포 대비 실시예 1의 시험물질 2종이 20 μM 농도에서 아밀로이드 베타-유도 SH-SY5Y 세포의 미토콘드리아 막 전위를 유의하게 증가시켰음(p <0.05)을 확인하였다.
본 발명자들은 상기 결과로부터 본 발명의 Diaportheone A1 및 Diaportheone A2가 미토콘드리아 막 전위의 손실을 억제함으로써 미토콘드리아의 기능장애를 예방하여 신경세포를 보호하는 효과가 있음을 확인할 수 있었다.
통계 분석
달리 언급되지 않는 한, 본 발명에 따른 실시예의 데이터는 최소 3회 반복 실험값의 평균 ± SD(standard deviation)로 계산되었다. 통계 분석은 Levene의 테스트와 Tukey의 HSD(honestly significant difference) 테스트를 사용한 일원분산분석(one-way ANOVA)에 의해 결정되었다. * p <0.05에서 통계적 유의성을 갖는 것으로 나타냈다.
결론
본 발명자들은 상기 실시예에서 살펴본 바와 같이 본 발명의 두 화합물 Diaportheone A1 및 A2가 신경독성을 나타내지 않으면서도 신경보호 효능을 나타내어, 알츠하이머병 등의 퇴행성 신경질환에 대한 예방 및 치료 효과를 가지고 있다는 것을 확인하였다. 구체적으로 본 발명의 실시예 1에서 제조한 시험물질 2종, 즉 Diaportheone A1 및 A2는 실시예 4의 티오플라빈 T(ThT) 분석과 실시예 5의 MDS 분석을 통해 아밀로이드 베타(Aβ)의 응집을 억제하고 아밀로이드 베타 올리고머의 생성을 직접적으로 저해하는 것으로 확인되었다. H2O2 및 Aβ 유발 신경모세포종 SH-SY5Y 세포에서 모두 세포 생존율 증가를 보였으며, 이는 실시예 1의 시험물질 2종이 신경보호 효과를 가진다는 것을 입증하는 결과이다(실시예 6 참조). 실시예 1의 시험물질 2종 모두 손상된 SH-SY5Y 세포에서 산화 스트레스에 대한 생화학적 마커인 ROS 수준과 미토콘드리아 기능장애를 감소시켰다(실시예 7 및 8 참조). 따라서, 상기 결과들로부터 본 발명의 Diaportheone A1, A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 약제학적 조성물은 Aβ, ROS, 산화적 손상에 의한 미토콘드리아의 막 전위 저하로 인한 신경세포 손상으로부터 신경세포를 보호함으로써 알츠하이머병과 같은 퇴행성 신경질환의 예방 또는 치료를 위해 유용하게 사용될 수 있다.
Claims (8)
- 제 1 항에 있어서, 상기 조성물은 아밀로이드 베타(amyloid beta, Aβ)의 응집을 억제하는 것인, 약제학적 조성물.
- 제 1 항에 있어서, 상기 조성물은 아밀로이드 베타(amyloid beta, Aβ)의 올리고머 생성을 저해하는 것인, 약제학적 조성물.
- 제 1 항에 있어서, 상기 조성물은 활성산소종(reactive oxygen species, ROS)을 감소시키는 것인, 약제학적 조성물.
- 제 4 항에 있어서, 상기 활성산소종은 과산화수소(hydrogen peroxide, H2O2), 과산화물(peroxide), 초과산화물(superoxide, O2 -), 하이드록실 라디칼(hydroxyl radical, ·OH), 알콕시 라디칼(alkoxy radical, RO·), 퍼옥시 라디칼(peroxy radical, ROO·), 유기 하이드로퍼옥사이드(organic hydroperoxide, ROOH), 일중항산소(singlet oxygen, 1O2) 및 알파-산소(alpha-oxygen, α-O)로 이루어진 군으로부터 선택되는 1종 이상인 것인, 약제학적 조성물.
- 제 1 항에 있어서, 상기 조성물은 아밀로이드 베타 또는 산화 스트레스에 의한 미토콘드리아 막 전위(mitochondrial membrane potential, ΔΨm) 손실을 감소시키는 것인, 약제학적 조성물.
- 제 1 항 내지 제 6 항 중 어느 한 항에 있어서, 상기 퇴행성 신경질환은 알츠하이머병, 파킨슨병, 헌팅턴병, 근위축성 측삭경화증, 다발성 경화증, 허혈성 뇌손상, 뇌염, 뇌수막염, 길랑-바레 증후군 및 외상성 뇌손상으로 이루어진 군으로부터 선택된 것인, 약제학적 조성물.
- Diaportheone A1, Diaportheone A2, 이들의 약제학적으로 허용 가능한 염, 또는 이들의 조합을 유효성분으로 포함하는 약제학적 조성물을 이를 필요로 하는 대상체(subject)에게 투여하는 단계를 포함하는 퇴행성 신경질환의 치료 방법.
