本発明の肥満の予測方法の一つは、動物の体内又は動物から単離された試料中に存在するレンサ球菌科(Streptococcaceae)、ルミノコッカス科(Ruminococcaceae)、ビフィドバクテリウム科(Bifidobacteriaceae)、エンテロコッカス科(Enterococcaceae)、エリュシペロトリクス科(Erysipelotrichaceae)、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)及びパラプレボテラ科(Paraprevotellaceae)からなる群から選ばれる1つ以上の科について、当該科に属する菌種数を測定する測定工程を備えることを特徴とするものである。
対象となる動物は特に限定されず、好ましくは哺乳類であり、例えば、犬、猫、ウサギ、フェレット等が挙げられ、特に犬が好ましい。
動物の年齢は5歳以下が好ましく、3~5歳であることが特に好ましい。
動物の体内又は動物から単離された試料中に存在する菌の種類やその数を検査、測定する方法は公知の方法をいずれも用いることができる。菌としては、腸内細菌が好ましく、試料としては、動物の糞便が好ましい。すなわち、好適な態様としては、動物の糞便を試料として、動物の腸内細菌叢を調べることが挙げられる。
菌種数の測定は、NGSなどのシーケンサーを用いたアンプリコンシーケンスなどの公知のメタゲノム解析法や細菌叢の解析方法を用いることができる。例えば、試料中に含まれるあらゆる生物のDNAやRNAの塩基配列情報を次世代シーケンサーを用いて解析することによって、当該試料中に含まれる生物を同定する方法が挙げられる。好ましくは、試料中に含まれる16SrRNA遺伝子の全部又は一部を、必要に応じて増幅して、シーケンスを行い、得られた配列をソフトウェアを用いて解析し、試料中の細菌の組成データを得る方法が挙げられる。試料中の細菌の組成データを、ソフトウェアで処理すること、あるいは、Genbank、Greengenes、SILVA databaseといった遺伝子データベースを参照することによって、試料中に含まれる細菌の菌種の帰属を決定し、特定の科に属する細菌の菌種数を測定することができる。
本発明の肥満の予測方法は、動物の体内又は動物から単離された試料中に存在する特定の科に属する菌(細菌)の菌種数を測定する測定工程を備えることを特徴とするものであり、当該菌種数が少ない場合に、将来、当該動物が肥満となることを予測するものである。ここでいう将来とは、所定期間内にという意味であり、期間は特に限定されないが、好ましくは3年以内、より好ましくは2年以内、さらに好ましくは1年以内である。また、肥満の予測であるから、本発明の方法の使用後、当該動物の食生活や生活習慣の改善により、肥満とならない事態も充分に想定される。むしろ、当該動物の飼育者に対して、当該動物が肥満になる危険性があることを伝えることによって、当該動物の食生活や生活習慣を改善することを促すという使い方もできる。例えば、本発明の肥満の予測方法による予測結果に応じて、肥満を防止するための食事、肥満になりにくい細菌を含むサプリ、低カロリーの食事、低糖質の食事、ダイエットメニュー等を提案したり、推奨することができる。
また、本発明の肥満の予測方法による予測結果に応じて、肥満を防止するための飲料、食事、サプリメントを製造あるいはカスタマイズすることもできる。肥満防止に関するサービスとして、本発明の肥満の予測方法による予測、予測結果の提供、予測結果に応じた飲料、食事、サプリメントを製造あるいはカスタマイズ、当該飲料、食事、サプリメントの提案、推奨という形態もとり得る。また、このようなサービスを提供した後に、さらに本発明の肥満の予測方法を実施し、肥満傾向が改善したのかどうかを提示するといった方法も可能である。上記飲料、食事、サプリメントには、食餌療法用飲料、ダイエット食品、栄養補助用添加物等が含まれる。
また、上記菌種数の測定結果に、さらに、遺伝子検査の結果(肥満に影響しやすい遺伝子の有無)と組み合わせて肥満を予測することが好ましい。遺伝子検査の結果を組み合わせることにより、高い精度で肥満の予測をすることができる。
菌種数の減少や菌種数が少ないことは、肥満ではないあるいは肥満傾向にない同種の動物の菌種数との比較に基づいて決定してもよいし、同一の動物について経時的に複数回菌種数の測定を行い、当該動物における菌種数の変化を見ることによって決定してもよい。
また、動物種ごとに、上記科に属する菌種数のデータベースを構築しておき、当該データベース中の菌種数との比較によって、菌種数が減っているあるいは少ないことを確認することもできる。
例えば、菌種数が、対象動物や対象動物の平均値と比較して、好ましくは1%以上、より好ましくは3%以上、さらに好ましくは5%以上少ない場合、肥満になる可能性があると予測することができる。
また、別の例では、菌種数が、同一の動物において、前回の測定時と比較して、好ましくは1%以上、より好ましくは3%以上、さらに好ましくは5%以上減少している場合、肥満になる可能性があると予測することができる。
菌の菌種数に関するデータとしては、例えば、菌種数、菌種数に応じて付与されたスコア、菌種数が所定値以上であるか所定値未満であるかに応じて付与されたスコア等が挙げられる。各菌科のスコアを合計して算出した合計スコアを用いてもよい。
菌の占有率に関するデータとしては、例えば、占有率、占有率に応じて付与されたスコア、占有率が所定値以上であるか所定値未満であるかに応じて付与されたスコア等が挙げられる。各菌科のスコアを合計して算出した合計スコアを用いてもよい。
測定対象となる科は、レンサ球菌科(Streptococcaceae)、ルミノコッカス科(Ruminococcaceae)、ビフィドバクテリウム科(Bifidobacteriaceae)、エンテロコッカス科(Enterococcaceae)、エリュシペロトリクス科(Erysipelotrichaceae)、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)及びパラプレボテラ科(Paraprevotellaceae)からなる群から選ばれる1種以上である。特に、コリオバクテリウム科(Coriobacteriaceae)及び/又はプレボテラ科(Prevotellaceae)に属する菌の菌種数を測定することが好ましい。これらの科に属する菌は、肥満となっている動物及び肥満に関連する疾病に罹患している動物の双方において、健常動物との比較において菌種数が減少していることが本発明者らによって確認されたものである。
さらに、上記に加えて、或いは、上記に代えて、アルカリゲネス科(Alcaligenaceae)、フソバクテリウム科(Fusobacteriaceae)及びベイロネラ科(Veillonellaceae)からなる群から選ばれる1つ以上の科に属する菌の占有率を測定してもよい。占有率とは、腸内細菌叢などの細菌叢に占める各菌種の存在比(検出比率)のことをいう。
レンサ球菌科は、ラクトバシラス目の中に置かれ、ラクトコッカス属、ラクトウム属、レンサ球菌属を含む。
ルミノコッカス科は、クロストリジウム綱クロストリジウム目に属する細菌の科である。ルミノコッカス属のほか、属として、Acetanaerobacterium、Acutalibacter、Anaerobacterium、Anaerofilum、Anaerotruncus、Dysosmobacter、Ercella、Ethanoligenens、Faecalibacterium、Fastidiosipila、Hydrogenoanaerobacterium、Oscillibacter、Oscillospira、Papillibacter、Pseudobacteroides、Sporobacter、Subdoligranulum、Youngiibacterが含まれる。
ビフィドバクテリウム科は、ビフィドバクテリウム目に含まれる科であり、ビフィドバクテリウム属を含む。
エンテロコッカス科は、ラクトバシラス目に属するグラム陽性の真正細菌の科である。代表的な属として、エンテロコッカス属(Enterococcus)、Melissococcus、Pilibacter、テトラジェノコッカス属(Tetragenococcus)及びVagococcusが含まれる。
エリュシペロトリクス科は、エリュシペロトリクス目に属する科であり、属として、Allobaculum、Breznakia、Bulleidia、Catenibacterium、Catenisphaera、 Coprobacillus、Dielma、Dubosiella、Eggerthia、Erysipelothrix、Faecalibaculum、Holdemania、Ileibacterium、Kandleria、Longibaculum、Longicatena、Solobacterium Turicibacterが含まれる。
コリオバクテリウム科は、コリオバクテリウム目に属する科であり、属として、Atopobium属、Olsenella属が含まれる。
プレボテラ科は、バクテロイデス目に属する科であり、プレボテラ属を含む。
コプロバチルス科は、エリュシペロトリクス目に属する科であり、Coprobacillus属を含む。
パラプレボテラ科は、バクテロイデス目に属する科であり、パラプレボテラ属を含む。また、パラプレボラ科を独立した科とせずに、パラプレボテラ属をプレボテラ科に含める分類方法もある。
アルカリゲネス科は、ブルクホルデリア目に属する科であり、アルカリゲネス属を含む。
フソバクテリウム科は、フソバクテリウム目に属する科であり、フソバクテリウム属を含む。
ベイロネラ科は、Clostridiales目に属する科であり、属として、Acetonema、
Acidaminococcus、Allisonella、Anaeroarcus、Anaeroglobus、Anaeromusa、

本発明の動物の肥満の判定方法は、動物の体内又は動物から単離された試料中に存在するレンサ球菌科Streptococcaceae、ルミノコッカス科Ruminococcaceae、ビフィドバクテリウム科Bifidobacteriaceae、エンテロコッカス科Enterococcaceae、エリュシペロトリクス科Erysipelotrichaceae、コリオバクテリウム科Coriobacteriaceae、プレボテラ科Prevotellaceae、コプロバチルス科Coprobacillaceae、及び、パラプレボテラ科Paraprevotellaceaeからなる群から選ばれる1つ以上の科について、当該科に属する菌種数を測定する測定工程を備えるものである。
上記1つ以上の科について、当該科に属する菌種数が減少している、あるいは、少ない場合に、当該動物が肥満であると判定することができる。
菌種数の減少や菌種数が少ないことは、肥満ではないあるいは肥満傾向にない同種の動物の菌種数との比較に基づいて決定してもよいし、同一の動物について経時的に複数回菌種数の測定を行い、当該動物における菌種数の変化を見ることによって決定してもよい。
また、動物種ごとに、上記科に属する菌種数のデータベースを構築しておき、当該データベース中の菌種数との比較によって、菌種数が減っているあるいは少ないことを確認することもできる。
例えば、菌種数が、標準体型である対照動物や対照動物の平均値(体型が「ふつう」であると飼育者が回答した動物や標準体型である動物等の平均値)と比較して、好ましくは1%以上、より好ましくは3%以上、さらに好ましくは5%以上少ない場合、肥満になる可能性があると予測することができる。
また、別の例では、菌種数が、同一の動物において、前回の測定時と比較して、好ましくは1%以上、より好ましくは3%以上、さらに好ましくは5%以上減少している場合、肥満になる可能性があると予測することができる。
本発明の疾患罹患の予測方法は、動物の体内又は動物から単離された試料中に存在するコリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、パラプレボテラ科(Paraprevotellaceae)、アルカリゲネス科(Alcaligenaceae)、フソバクテリウム科(Fusobacteriaceae)及びベイロネラ科(Veillonellaceae)からなる群から選ばれる1つ以上の科について、当該科に属する菌の占有率を測定する測定工程を備えることを特徴とするものである。
本発明者らの検討により、肥満群の動物で罹患割合が高い特定の疾患(内分泌疾患、呼吸器疾患、血液疾患、筋骨格疾患、皮膚疾患、耳の疾患、全身性疾患、肝臓・胆道・膵臓疾患)に罹患している動物では、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、パラプレボテラ科(Paraprevotellaceae)、アルカリゲネス科(Alcaligenaceae)、フソバクテリウム科(Fusobacteriaceae)又はベイロネラ科(Veillonellaceae)に属する菌の腸内細菌叢中の占有率や保有率が低いことが明らかとなった。したがって、これらの科に属する菌は、肥満と関連する特定疾患への罹患率との関連性があることが分かる。
上記1つ以上の科について、当該科に属する菌種について、菌種数や占有率が減少している、あるいは、少ない場合に、当該動物が疾患に罹患する可能性がある(高い)と判定することができる。
菌種数の減少や菌種数が少ないことは、同種の動物の菌種数との比較に基づいて決定してもよいし、同一の動物について経時的に複数回菌種数の測定を行い、当該動物における菌種数の変化を見ることによって決定してもよい。
また、動物種ごとに、上記科に属する菌種数のデータベースを構築しておき、当該データベース中の菌種数との比較によって、菌種数が減っているあるいは少ないことを確認することもできる。
例えば、菌種数が、対象動物や対象動物の平均値と比較して、好ましくは1%以上、より好ましくは3%以上、さらに好ましくは5%以上少ない場合、疾患に罹患する可能性があると予測することができる。
また、別の例では、菌種数が、同一の動物において、前回の測定時と比較して、好ましくは1%以上、より好ましくは3%以上、さらに好ましくは5%以上減少している場合、疾患に罹患する可能性があると予測することができる。
本発明の保険料の算出方法は、上記の動物の肥満の予測方法による予測結果に基づいて保険料を算出することを特徴とするものである。肥満になることが予測される動物、あるいは、肥満になりやすい動物、肥満傾向のある動物は、将来、疾患、特に肥満に関連する疾患に罹患する可能性が、通常体型の同種動物と比較して高いことが想定されるからその分保険料を高く算定することができる。また、例えば、肥満傾向になることが予想されない動物、肥満になりにくい動物、又は肥満傾向にない動物については、保険料を安く算定することができる。
本発明の肥満の予測方法は、動物の腸内細菌叢の多様度指数を測定する工程を備える動物の肥満の予測方法である。多様度指数としては、シャノン・ウィーナーの多様度指数(Shannon Index)、シンプソンの多様度指数等が挙げられるが、シャノン・ウィーナーの多様度指数が好ましい。
動物の腸内細菌叢の多様度指数が高いほど、当該動物が肥満になりにくいと予測することができる。
腸内細菌叢の多様度指数は公知の方法によって測定することができる。その他の点は上記と同様である。
本発明の動物の肥満予測システムは、動物の体内又は動物から単離された試料中に存在する菌について、所定の菌科に属する菌の菌種数又は占有率に関するデータを受け付ける受付手段と、動物の体内又は動物から単離された試料中に存在する菌について、所定の菌科に属する菌の菌種数又は占有率に関するデータから当該動物が肥満になるか否かを予測する判定手段と、を備えるものである。
本発明の受付手段は、肥満になる可能性を予測したい動物由来の試料における所定の菌科に属する菌の菌種数又は占有率に関するデータの入力を受け付ける手段である。動物としては、犬、猫、鳥、ウサギ、フェレット等が挙げられる。また、対象となる動物の年齢は限定されないが、好ましくは0歳以上、より好ましくは0歳~12歳、さらに好ましくは1歳~10歳である。菌種数又は占有率に関するデータの受付方法は、端末へのデータの入力、送信などいずれの方法であってもよい。
ラベルを用いる場合は、細菌叢中の占有率を測定し、入力手段に数値を入力する前に、当該測定された占有率に応じて、予め定めておいた対応表から特定のラベルを割当てて、そのラベルを受付手段に入力することができる。
また、菌種数に基づいて設定されたラベルとは、菌種数に応じて適宜設定されたラベルである。例えば、菌種数が所定値を超える場合は「1」、超えない場合は「0」といったラベルを付すことができる。
また、菌種数に基づいて設定されたスコアとは、菌種数に応じて適宜設定されたスコアである。例えば、特定の科について、その科に属する菌の菌種数が所定値以上であれば「+1」、所定値以下であれば「0」や「-1」というスコアが与えられる。
このようなスコアを菌科ごとに算出し、受付手段に入力してもよい。また、受付手段に菌科に属する菌の菌種数や占有率を入力し、当該入力された菌の菌種数についてのデータや占有率に基づいて予め設定されたスコア付与基準に基づいて各菌科ごとにスコアを算出するという構成でもよい。
本発明の肥満予測システムは、判定手段が、菌科ごとに算出又は入力されたスコアを合計し、得られた合計スコアに基づいて肥満になる可能性を予測判定するという構成でもよい。
本発明の判定手段は、受付手段に入力された動物由来の試料中の細菌叢における所定の菌科に属する菌の菌種数又は占有率に関するデータからその動物が所定期間内に肥満になるか否かを予測判定する手段である。予測判定方法は特に限定されない。例えば、プロセッサが、予め設定されたプログラムを用いて動物由来の試料中の細菌叢における所定の菌科に属する菌の菌種数又は占有率に関するデータからその動物が所定期間内に肥満になるか否かを予測判定する。また、上記のように、菌科ごとの菌の菌種数や占有率から、予め設定された基準に従ってスコアを算出し、当該スコアを合計した合計スコアに基づいて、肥満リスクを判定するといった構成でもよい。すなわち、合計スコアが所定値以上なら、肥満リスクが高く(或いは肥満リスクが低く)、合計スコアが所定値未満なら肥満リスクが低い(或いは肥満リスクが高い)といった具合である。
本発明の判定手段による予測判定の出力の形式は特に限定されず、例えば、パソコンやスマートフォンなどの端末の画面上において、「今後1年以内に肥満になる可能性あり」、「今後1年以内に肥満になる可能性は高い」あるいは、「今後1年以内に肥満になる可能性は○%」、「肥満傾向があります。」といった表示をすることで予測判定を出力することができる。好ましくは、肥満になりやすいかどうかを、利用者の端末の画面上でアイコンで表示する。アイコンは、例えば、図7のように、利用者に分かりやすいように、肥満のなりやすさを数段階で、図形により表すことが好ましい。
本発明の動物の肥満予測システムは、判定手段から判定結果を受信し、判定結果を出力する出力手段を別途有していてもよい。
上記のように、本発明の動物の肥満予測システムは、所定の菌科に属する菌の菌種数又は占有率に関するデータを用いるものである。所定の菌科としては、特に限定されないが、例えば、上記した菌科が挙げられる。また、菌科として、前記所定の菌科が、レンサ球菌科(Streptococcaceae)、ルミノコッカス科(Ruminococcaceae)、ビフィドバクテリウム科(Bifidobacteriaceae)、エンテロコッカス科(Enterococcaceae)、エリュシペロトリクス科(Erysipelotrichaceae)、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、パラプレボテラ科(Paraprevotellaceae)、アルカリゲネス科(Alcaligenaceae)、フソバクテリウム科(Fusobacteriaceae)及びベイロネラ科(Veillonellaceae)からなる群から選ばれる1つ以上の科が好ましい。
図9中、端末40は、肥満予測システムを利用したい者(ユーザ)が利用する端末である。端末40は、例えばパーソナルコンピュータ、スマートフォンやタブレット端末などが挙げられる。端末40は、CPUなどの処理部、ハードディスク、ROMあるいはRAMなどの記憶部、液晶パネルなどの表示部、マウス、キーボード、タッチパネルなどの入力部、ネットワークアダプタなどの通信部などを含んで構成される。
ユーザーは、端末40から、サーバにアクセスし、対象となる動物の試料中の細菌叢、好ましくは腸内細菌叢(以下、腸内細菌叢の例で説明する。)の所定の菌科に属する菌の菌種数又は占有率に関するデータ、及び、必要に応じて、顔画像(写真)、当該動物の種類、品種、年齢、体重、既往歴などの情報を入力、送信する。
また、ユーザーは、端末40がサーバにアクセスすることによって、サーバにおける肥満予測結果を受信することができる。
記憶部10は、例えばROM、RAMあるいはハードディスクなどから構成される。記憶部10には、サーバの各部を動作させるための情報処理プログラムが記憶され、特に、判定手段11のためのソフトウェアなどが記憶される。
本実施形態では、判定手段や受付手段がサーバに格納され、ユーザーの端末とインターネットやLAN等の接続手段で接続される態様を説明したが、本発明はこれに限定されず、判定手段、受付手段、インターフェース部が一つのサーバや装置内に格納される態様や、利用者が利用する端末を別途必要としない態様等であってもよい。
提案手段12と判定手段11は一つのソフトウェアであってもよい。
(糞便試料からのDNA抽出)
以下のようにして、各犬(トイプードル)から糞便試料を採取し、DNAを抽出した。
犬(トイプードル)の飼育者が糞便の採取キットを用いて、犬の糞便試料を採取した。当該糞便試料を受領し、水に懸濁した。
次に、糞便懸濁液200 μLとLysis buffer(224μg/mLのProtenaseKを含む)810 μLをビーズチューブに添加し、ビーズ式ホモジナイザーにてビーズ破砕(6,000 rpm、破砕20秒、インターバル30秒、破砕20秒)を行った。その後、検体を70℃のヒートブロック上にて10分間静置することでProtenase Kによる処理を行い、続いて95℃のヒートブロック上にて5分間静置することでProtenase Kを不活化した。溶菌処理を行った検体はchemagic 360(PerkinElmer)を用い、chemagicキットstool用プロトコルにてDNAの自動抽出を行い、100 μLのDNA抽出液を得た。
メタ16Sシーケンス解析はillumina 16SMetagenomic Sequencing Library Preparation(バージョン15044223B)を改変して行った。まず、16S rRNA遺伝子の可変領域V3-V4を含む460 bpの領域をユニバーサルプライマー(Illumina_16S_341FおよびIllumina_16S_805RPCR)を用いたPCRで増幅した。PCR反応液は10 μLのDNA抽出液、0.05 μLの各プライマー(100 μM)、12.5 μLの2x KAPA HiFi Hot-Start ReadyMix(F. Hoffmann-La Roche、Switzerland)、2.4 μLのPCR gradewaterを混合して調製した。PCRには95℃ 3分間の熱変性後、95℃ 30秒、55℃ 30秒、72℃ 30秒のサイクルを30回繰り返し、最後に72℃ 5分の伸長反応を行った。増幅産物は磁気ビーズを用いて精製し、50 μLのBuffer EB(QIAGEN、Germany)で溶出した。精製後の増幅産物はNextera XT Index Kitv2(illumina、CA、US)を用いてPCRを行い、インデックスを付加した。PCR反応液は2.5 μLの増幅産物、2.5 μLの各プライマー、12.5 μLの2x KAPA HiFi Hot-Start ReadyMix、5 uLのPCR grade waterを混合して調製した。PCRには95℃ 3分間の熱変性後、95℃ 30秒、55℃ 30秒、72℃ 30秒のサイクルを12回繰り返し、最後に72℃ 5分の伸長反応を行った。インデックス付加を行った増幅産物は磁気ビーズを用いて精製し、80-105 μLのBuffer EBで溶出した。各増幅産物の濃度はNanoPhotometer(Implen、CA、US)で測定し、1.4 nMに調製した後、等量ずつ混合し、これをシーケンス用ライブラリーとした。シーケンス用ライブラリーのDNA濃度および増幅産物のサイズを電気泳動にて確認し、これをMiSeqにより解析した。解析にはMiSeq Reagent Kit V3を用い、2×300 bpのペアエンドシーケンスを行った。得られた配列はMiSeq Reporterにて解析し、細菌の組成データを得た。
上記で用いたユニバーサルプライマーの配列は以下のとおりである。このユニバーサルプライマーは市販されているものを購入することができる。

菌種数の測定についての結果を図1に示す。図1において、棒グラフが示す菌種数はふつう群、肥満群それぞれの、各菌科の検出された菌種数の平均値である。当該菌種が検出されない個体もいるため、平均値が1を下回る場合もある。
図1から明らかなように、肥満群に属するトイプードルについて、レンサ球菌科(Streptococcaceae)、ルミノコッカス科(Ruminococcaceae)、ビフィドバクテリウム科(Bifidobacteriaceae)、エンテロコッカス科(Enterococcaceae)、エリュシペロトリクス科(Erysipelotrichaceae)、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、及び、パラプレボテラ科(Paraprevotellaceae)のそれぞれについて、ふつう群に属するトイプードルと比較して、菌種数が5%以上少ないことが明らかとなった。なお、菌種数の減少割合「%」については、「肥満群の平均菌種数/ふつう群の平均菌種数」で算出した。
得られた細菌の組成データ(FASTQファイル)から、QIIME2を用いてShanon Index(シャノン・ウィナーの多様度指数)を算出した。そして、年齢が3~5歳の群と、6~11歳の群に分けて、Shanon Indexと肥満割合との相関を見た。
結果を図2Aおよび図2Bに示す。
図2Aおよび図2Bから明らかなように、3~5歳の群において、多様度指数(Shanon Index)と、肥満割合との相関性が高いことが分かった。なお、ここでいう「肥満割合」とは、同じ多様度指数を示す個体群中における肥満個体の割合である。その際、多様度指数は小数点第二位で四捨五入し、第一位までで揃えている。例えば、多様度指数が「3.2」と検出された個体が120おり、その内肥満個体が12いた場合、多様度指数3.2の肥満割合は10%となる。
肥満群に属するトイプードルと、ふつう群に属するトイプードルについて、それぞれ回答の前後180日以内に疾患に罹患したかをグラフにした。結果を図3に示す。疾患への罹患の有無は、ペット保険の保険金請求記録を用い、糞便採取を行った各個体の糞便採取日の前後180日以内に保険金支払請求があったか否かで調査した。
図3から明らかなように、肥満群に属するトイプードルでは、内分泌疾患、呼吸器疾患、血液疾患、筋骨格疾患、皮膚疾患、耳の疾患、全身性疾患、肝臓・胆道・膵臓疾患といった疾患(以下「特定疾患」という。)への罹患確率が高いことが分かる。
特定疾患に罹患したトイプードル(3~13歳。合計3199匹)と、そうではないトイプードル(3~13歳。無事故群。5088匹)について、腸内細菌叢における特定の科に属する菌種の占有率及び保有率を測定した。
結果を図4及び図5に示す。図4は、特定の科に属する菌種の占有率(占有率とは、腸内細菌叢に占める各菌種の存在比(検出比率)であり、検出結果にある「hit rate」と同義である。)を示すグラフであり、図5は、保有率(保有率とは、ふつう群、肥満群等特定のカテゴリーで群全体を見た際に、その細菌を持つ個体の割合である。保有率は、各個体が当該細菌を持つか、持たないかのみに関する値であり、「hit rate」等の「どの程度もつか」といった量的な大小関係は含まない値である。)を示すグラフである。
図4及び図5から、特定疾患への罹患と、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、パラプレボテラ科(Paraprevotellaceae)、アルカリゲネス科(Alcaligenaceae)、フソバクテリウム科(Fusobacteriaceae)又はベイロネラ科(Veillonellaceae)に属する菌の菌種の保有の有無が関連することが分かる。
図1のデータを算出するにあたって調査対象となった3~13歳の9390個体(うち、肥満の個体は1029個体)のトイプードルについて、レンサ球菌科(Streptococcaceae)、ルミノコッカス科(Ruminococcaceae)、ビフィドバクテリウム科(Bifidobacteriaceae)、エンテロコッカス科(Enterococcaceae)、エリュシペロトリクス科(Erysipelotrichaceae)、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、及び、パラプレボテラ科(Paraprevotellaceae)の合計の菌種数と肥満割合をプロットし、グラフにした。なお、ここでいう「肥満割合」とは、同じ菌種数を示す個体群中における肥満個体の割合である。
結果を図6に示す。
図6から、上記科に属する菌の菌種数と肥満とは強い相関があることが分かる。
実施例1と同様にして、犬の糞便試料を採取し、0~18歳の3,610個体の犬について、腸内細菌叢中の科ごとの菌種数の測定を行った。なお、肥満かどうかの分類は飼育者からの犬の体格に関する問診データに従い、肥満、ふつう、痩せているの3群に分けた。肥満群が677個体、ふつう群が2,671個体、痩せている群が262個体であった。

- 動物の体内又は動物から単離された試料中に存在するレンサ球菌科(Streptococcaceae)、ルミノコッカス科(Ruminococcaceae)、ビフィドバクテリウム科(Bifidobacteriaceae)、エンテロコッカス科(Enterococcaceae)、エリュシペロトリクス科(Erysipelotrichaceae)、コリオバクテリウム科(Coriobacteriaceae)、プレボテラ科(Prevotellaceae)、コプロバチルス科(Coprobacillaceae)、及び、パラプレボテラ科(Paraprevotellaceae)からなる群から選ばれる1つ以上の科について、当該科に属する菌種数を測定する測定工程を備える動物の肥満の予測方法。
- 前記動物の体内が腸である請求項1記載の動物の肥満の予測方法。
- 前記動物から単離された試料が動物の糞便である請求項1記載の動物の肥満の予測方法。
- 前記測定工程が、コリオバクテリウム科(Coriobacteriaceae)及び/又はプレボテラ科(Prevotellaceae)に属する菌の菌種数を測定するものである請求項1~3のいずれか一項記載の動物の肥満の予測方法。
- さらに、アルカリゲネス科(Alcaligenaceae)、フソバクテリウム科(Fusobacteriaceae)及びベイロネラ科(Veillonellaceae)からなる群から選ばれる1つ以上の科について、当該科に属する菌の占有率を測定する測定工程を備える請求項1~4のいずれか一項記載の動物の肥満の予測方法。

- 前記判定手段による予測結果とともに、所定の菌科に属する所定の菌科に属する菌の菌種数又は占有率の大小を、アイコンで表示する表示部を備える請求項17記載の動物の肥満予測システム。
- 前記判定手段による予測結果に応じて肥満を改善するためのアドバイスを提供するアドバイス手段を備える請求項16~18のいずれか一項記載の動物の肥満予測システム。
- 前記判定手段による予測結果に応じてフードをレコメンドする提案手段を備える請求項16~19のいずれか一項記載の動物の肥満予測システム。
