Abstract
COVID-19に対して強い抗ウイルス活性を有する、医薬品、食品又は衛生用品に利用可能な組成物を提供する。 下記式(I)で表される化合物からなる群から選択される、少なくとも1つの化合物又はその誘導体を有効成分として含む、抗ウイルス活性を有する組成物を提供する。(式中、R16は、R13と共にエーテル結合又はチオエーテル結合を形成して架橋しているか、あるいは、水素、ハロゲン 、水酸基、チオール基、置換若しくは非置換の炭素数1~3のアシルオキシ基、置換若しくは非置換の炭素数1~4の飽和若しくは不飽和炭化水素基、又は、置換若しくは非置換の炭素数1~4のアルコキシ基である;R1~R7、R15及びR17~R20は、それぞれ、水素、ハロゲン、水酸基、チオール基、置換若しくは非置換の炭素数1~3のアシルオキシ基、置換若しくは非置換の炭素数1~4の飽和若しくは不飽和炭化水素基、又は、置換若しくは非置換の炭素数1~4のアルコキシ基である;R14は、R8と共にジスルフィド結合を形成して架橋しているか、水素である;R13は、R16と共にエーテル結合又はチオエーテル結合を形成して架橋しているか、R8と共にジスルフィド結合を形成して架橋している;R9及びR12は、それぞれ水素又は置換若しくは非置換の炭素数1~3のアルコキシ基である;R10及びR11は、それぞれ酸素又は硫黄である;R8はR13又はR14と共にジスルフィド結合を形成して架橋している)。
Description
本発明は、抗ウイルス活性を有する化合物を含む、医薬品、食品等に使用可能な組成物に関する。
コロナウイルスは、ニドウイルス目コロナウイルス科オルソコロナウイルス亜科よりなるRNAウイルスの一群である。コロナウイルスは、ポジティブセンス一本鎖RNAゲノムと、らせん対称のヌクレオカプシドを持つエンベロープウイルスであり、カプシド表面からこん棒型の突起が突き出た特徴的な形状を有する。コロナウイルスは、RNAウイルスの中でも大型のウイルスであり、そのゲノムサイズは約26~32キロベースである。
コロナウイルスは、哺乳類、鳥類等に感染性疾患を引き起こすRNAウイルスとして知られ、特に、軽度~重篤の気道感染症を引き起こすことが知られる。ヒトにおいては、コロナウイルスは、軽症の場合は一般的な風邪の原因ウイルス(風邪の原因ウイルスは、コロナウイルス以外にも多数知られる)となるが、SARS、MERS、およびCOVID-19等の一部のコロナウイルスは、より致命的な症状を引き起こすことが知られる。特に、2019年に中国で発見された新型コロナウイルスであるCOVID-19は、短期間に全世界に蔓延し、有効な治療薬、ワクチンが存在しなかったという背景もあり、多数の感染者及び死者を出すこととなった。現在も、その数は増え続けており、早急にCOVID-19の治療薬及びワクチンの開発と普及がなされることが望まれる。
COVID-19感染症を特異的に治療する医薬品(承認薬)は、現在のところ存在せず、一部の既存の抗ウイルス剤、特にコロナウイルスと同じRNAウイルスである、エボラウイルス、インフルエンザウイルス等の抗ウイルス剤、例えばレムデシビル、ファビピラビル等がCOVID-19感染症の治療に使用されている。また、COVID-19は、ヒトにおいて過剰な免疫反応を引き起こし、これにより、ヒトが呼吸不全等の重篤な状態に陥ることが知られる。そのため、上記の既存の抗ウイルス剤に加えて、またはこれに代えて、ステロイド等の免疫抑制剤が使用されている。
RNAウイルスに対する抗ウイルス剤は、その作用機序によって、RNAポリメラーゼ阻害剤(例えば、レムデシビル、ファビピラビル)、逆転写酵素阻害剤(例えば、アバカビル)、プロテアーゼ阻害剤(例えば、リトナビル)等に分類される。これらの阻害剤は、ヒトには存在せず、RNAウイルスにのみ存在する酵素を阻害する薬剤であり、ヒトへの副作用が生じにくいことが期待され、抗ウイルス剤として開発されたものである。
ニホンコウジカビ(Aspergillus oryzae)に含まれるアスピロクロリン(aspirochlorine)は、当初は「A30641」という名称で、抗真菌剤として報告された化合物である(非特許文献1、2)。アスピロクロリンは、真菌のタンパク質合成に対して、高選択的かつ強力な阻害活性を有することが報告される一方で(非特許文献3)、抗ウイルス活性は低いことが報告されている(非特許文献1)。非特許文献4~6には、アスピロクロリンの推定生合成経路が報告されている。

上記の通り、COVID-19の治療において、COVID-19を治療標的として開発された治療薬は、現在のところ使用されていない。COVID-19感染症に対する既存の抗ウイルス剤の有効性が報告されているものの、治療効率を考慮すると、より効率よくCOVID-19の増殖等を阻害し得る抗ウイルス活性を有する薬剤が望まれる。
本発明は、COVID-19に対して強い抗ウイルス活性を有する、医薬品、食品又は衛生用品に使用可能な組成物を提供することを目的とする。
本発明者らは、鋭意検討を重ねた結果、抗ウイルス活性が低いとされてきたアスピロクロリンについて、その前駆体に、COVID-19のメインプロテアーゼ(Mpro)を阻害する活性を有する化合物が存在することを見出し、本発明を完成させるに至った。すなわち、本発明は、以下の通りである。
(1)下記式(I)で表される化合物からなる群から選択される、少なくとも1つの化合物又はその誘導体を有効成分として含む、抗ウイルス活性を有する組成物。

本発明によれば、COVID-19に対して強い抗ウイルス活性を有する、医薬品、食品又は衛生用品に使用可能な組成物を提供することが可能である。
本明細書に示される化学式は、特に記載がない限り、あらゆる幾何学異性体及び光学的異性体を含むものとする。本明細書において「含有量」又は「量」とは、特に記載のない限り、モル量を指すものとする。また「モル濃度(M)」は、特に記載のない限り、溶液単位体積当たりのモル量(モル/dm3)を指すものとする。
1.抗ウイルス活性を有する組成物
1-1.有効成分
本発明の組成物は、下記式(I)で表される化合物からなる群から選択される、少なくとも1つの化合物又はその誘導体を有効成分として含み、抗ウイルス活性を有する組成物である。

1-1.有効成分
本発明の組成物は、下記式(I)で表される化合物からなる群から選択される、少なくとも1つの化合物又はその誘導体を有効成分として含み、抗ウイルス活性を有する組成物である。
本明細書において「置換若しくは非置換の炭素数1~3のアシルオキシ基」とは、メタノイルオキシ基、エタノイルオキシ基(アセトキシ基)又はプロパノイルオキシ基の基本骨格を有し、水素がハロゲン、水酸基、チオール基又は炭素数1~4のアルコキシ基で置換されているか、又は置換されていないアシルオキシ基をいう。また、本明細書において「置換若しくは非置換の炭素数1~4の飽和若しくは不飽和炭化水素基」とは、メチル基、エチル基、プロニル基、イソプロピル基、ブチル基、イソブチル基、tert-ブチル基、2-プロペニル基、2-ブテニル基、3-ブテニル基、2-プロピニル基、2-ブチニル基、3-ブチニル基を基本骨格として、水素がハロゲン、水酸基、チオール基又は炭素数1~4のアルコキシ基で置換されているか、又は置換されていない炭化水素基をいう。
本明細書において「置換若しくは非置換の炭素数1~4のアルコキシ基」とは、メトキシ基、エトキシ基、プロポキシ基、イソプロポキシ基、ブトキシ基、イソブトキシ基、tert-ブトキシ基を基本骨格として、水素がハロゲン、水酸基、チオール基又は炭素数1~4の飽和若しくは不飽和炭化水素で置換されているか、又は置換されていないアルコキシ基をいう。アルコキシ基は、不飽和結合を有していてもよい。
本明細書において、「ハロゲン」とは、周期表17族に属する元素を指し、フッ素(F)、塩素(Cl)、臭素(Br)、ヨウ素(I)、アスタチン(At)が含まれる。本発明の化合物に使用されるハロゲンは、好適にはフッ素又は塩素であり、より好適にはフッ素である。
本明細書において、「水酸基」及び「チオール基」は、イオン化した状態、すなわち、「-O-」及び「-S-」で示される状態であってもよい。また、Na+、K+,Ca2+、NH4
+等の陽イオンと塩を形成してもよい。また、水素がハロゲン、好適にはフッ素に置換されたもの、すなわち、「-O-F」及び「-S-F」で示される構造であってもよい。
上記式(I)において、「R16は、R13と共にエーテル結合又はチオエーテル結合を形成して架橋している」との記載は、R16とR13の2つの置換基を有することを示すものではなく、式中のaの位置の炭素とbの位置の炭素とが、エーテル結合又はチオエーテル結合で連結されていることをいう。また、上記式(I)において、「R8はR13又はR14と共にジスルフィド結合を形成して架橋している」との記載は、上記式(I)において、cの位置の炭素と、aの位置の炭素又はdの位置の炭素とが、ジスルフィド結合で連結されていることをいう。なお、aの位置の炭素とbの位置の炭素とがエーテル結合で連結されている場合、ジスルフィド結合はcの位置の炭素とdの位置の炭素とを連結する。
本発明の組成物は、上記式(I)で表される化合物のうち、好ましくは、下記式(II)又は(III)で示される化合物を有効成分として含む。

本明細書において「誘導体」とは、ある化合物の所望の活性をより高くするため、副作用(例えば、細胞毒性等)を低減するため、水溶性を高めるため、あるいは安定性を高めるために、その所望の活性が失われない程度に当該化合物の構造に修飾を加えたものをいう。適用可能な修飾は、特に限定されないが、例えば、PEG修飾、アミノ酸修飾、ペプチド修飾、ビオチン修飾、メチル化等が挙げられる。
ニホンコウジカビ(Aspergillus oryzae(A.oryzae))におけるアスピロクロリンの推定生合成経路の概略図を図1に示す。図中「1」で示される化合物がアスピロクロリンである。アスピロクロリンは、2分子のフェニルアラニンを開始物質として合成される。まず、2分子のフェニルアラニンが、非リボソームペプチド構成酵素(NRPS)AclPにより、フェニルアラニンの環状二量体(cyclo-Phe-Phe)に変換される。次いで、各種アスピロクロリン合成酵素の働き(AclGによるグルタチオン付加、AclIJKによる段階的なチオール分解、及びAclTによるジチオール酸化)により、特徴的なジスルフィド結合を有する化合物16が生成される。化合物16は、チオレドキシンオキシドレダクターゼ様酵素AclRの働きにより、架橋エーテル結合が導入されて化合物15に変換され、さらに、シトクロムP450系酵素AclLの働きにより、水酸基が導入されて化合物3が生成される。化合物3のフェノール基が切り離されて化合物4が生成され、次いで、シトクロムP450系酵素AclO及びメチルトランスフェラーゼAclUの働きにより、複素環のNにメトキシ基が導入されて化合物2が生成される。さらに、ハロゲナーゼAclHの働きにより塩素が導入されてアスピロクロリン(化合物1)が合成される。詳細な生合成の機序や、アスピロクロリン合成酵素の構造等については、非特許文献4~6の記載を参照されたい。
本発明者らは、アスピロクロリン(化合物1)、並びに、その前駆体と推定される図1に示す化合物2、3、15及び16を調製し、COVID-19のメインプロテアーゼ活性阻害試験を行った。アスピロクロリン、及び化合物2、3、15及び16の調製方法は、特に限定されないが、A.oryzaeの培養液から、公知の方法、例えば非特許文献5及び6に記載の方法で抽出、精製することで調製することができる。アスピロクロリン及び化合物2は、例えば、A.oryzaeのAclR強制発現株の培養液から、メタノール抽出、水-酢酸エチル分配及びカラム精製を経て得ることができる。化合物3は、例えば、A.oryzaeのAclO欠損、AclR強制発現株の培養液から、アセトン抽出、水-酢酸エチル分配及びカラム精製を経て得ることができる。化合物15は、例えば、A.oryzaeのAclL欠損、AclR強制発現株の培養液から、アセトン抽出、水-酢酸エチル分配及びカラム精製を経て得ることができる。化合物16は、例えば、A.oryzaeのAclR欠損、AclP強制発現株の培養液から、エタノール抽出、水-酢酸エチル分配及びカラム精製を経て得ることができる。いずれの化合物も、1H-NMR及び13C-NMRを用いて分子構造を確認することが可能である。
化合物1、2、3、15及び16による阻害試験の結果、これらすべての化合物にCOVID-19のメインプロテアーゼ阻害活性があることが確認されたが、化合物1及び2においては阻害活性が低いのに対して、化合物3、15及び16において、阻害活性が顕著に高いことを発見した。
1-2.組成物の構成
本発明の組成物は、有効成分として、「1-1.有効成分」の項で説明した化合物のうち、少なくとも1つの化合物を含む、抗ウイルス活性を有する組成物である。本発明の組成物には、上記の化合物が単体で含まれていてもよく、また、複数種の化合物が混合されて含まれていてもよい。
本発明の組成物は、有効成分として、「1-1.有効成分」の項で説明した化合物のうち、少なくとも1つの化合物を含む、抗ウイルス活性を有する組成物である。本発明の組成物には、上記の化合物が単体で含まれていてもよく、また、複数種の化合物が混合されて含まれていてもよい。
本明細書において「抗ウイルス活性」とは、ウイルスの感染能力及び/又は増殖能力を部分的に又は完全に阻害して、結果的にウイルスの数を減じる作用をいう。特に限定されるものではないが、本発明において、「抗ウイルス活性」は、COVID-19の増殖阻害活性であることが好ましい。特に、COVID-19の増殖にはメインプロテアーゼの働きが必須とされていることから、本発明において、「抗ウイルス活性」は、COVID-19のメインプロテアーゼ阻害活性であることがより好ましい。
本発明の組成物に含まれる有効成分の含有量は、モル比において、アスピロクロリンの含有量よりも高いことが好ましい。アスピロクロリン及び上記有効成分は、A.oryzae等の天然物から得られうる化合物であるものの、通常、A.oryzae等の天然物においては、前駆体である上記有効成分よりもアスピロクロリンの含有量の方が高い。一方で、本発明の好適な組成物は、有効成分の相対的含有量において、明らかに通常の天然物等から得られる組成物とは異なるものである。有効成分の含有量は、モル比で、アスピロクロリンの5~100倍であることが好ましく、10~100倍であることがより好ましい。このように、アスピロクロリンに対して有効成分の含有量の高い組成物は、例えば、A.oryzaeの培養液から抽出精製した有効成分を直接組成物に添加する方法や、アスピロクロリン合成酵素の一部(例えば、シトクロムP450系酵素AclO)を阻害する物質を添加してA.oryzaeを培養した培養液から組成物を調製する方法により取得可能である。あるいは、アスピロクロリン生合成酵素遺伝子を高発現化し、並びに転写因子を高発現化あるいは欠損させたA.oryzaeの培養液より取得可能である。
本発明の組成物の性状は、特に限定されず、水溶液等の液状であってもよく、また、粉末状、顆粒状であってもよい。本発明の組成物は、ウイルスの数、特にCOVID-19の数を減らす目的であれば、その用途は特に限定されないが、例えば、医薬品、食品及び衛生用品に使用することができる。本発明の組成物における、好ましい有効成分の含有量、有効成分以外に含まれる成分、組成物の性状等は、用途によって異なるため、以下、用途別に説明する。
2.医薬品(医薬組成物)
本発明の組成物は、医薬組成物として使用することができる。本項において、本発明の組成物を、医薬組成物又は医薬品とも称する。

本明細書において、「医薬組成物」及び「医薬品」は、疾患の治療及び/又は予防のために対象に投与される物を指し、本発明においては、ウイルス感染症、好ましくはCOVID-19感染症の治療及び/または予防のために投与される物を指す。治療及び/又は予防の対象は、ヒト、チンパンジーを含む霊長類、イヌ、ネコなどのペット動物、ウシ、ウマ、ヒツジ、ヤギなどの家畜動物、マウス、ラットなどの齧歯類、動物園で飼育される動物などの哺乳動物であり、好ましくは、ヒトである。
本発明の医薬組成物において、有効成分である化合物(以下、「薬剤」とも称する)の含有量は、薬剤の作用効果及び安定性、医薬組成物の剤形、使用する担体の種類、及び投与方法、投与する対象の状態等の様々な条件によって異なる。これらは、当該分野の公知技術に基づいて適宜選択すればよいが、例えば、対象への投与量が、1~100nmol/kg体重/日、特に10~100nmol/kg体重/日となるような含有量とすることができる。
本発明の医薬組成物は、必要に応じてさらに製薬上許容可能な担体を含むことができる。「製薬上許容可能な担体」とは、製剤技術分野において通常使用する添加剤をいう。例えば、賦形剤、結合剤、崩壊剤、充填剤、乳化剤、流動添加調節剤、滑沢剤等が挙げられる。
賦形剤としては、単糖、二糖類、シクロデキストリン及び多糖類のような糖(より具体的には、限定はしないが、グルコース、スクロース、ラクトース、ラフィノース、マンニトール、ソルビトール、イノシトール、デキストリン、マルトデキストリン、デンプン及びセルロースを含む)、金属塩(例えば、塩化ナトリウム、リン酸ナトリウム若しくはリン酸カルシウム、硫酸カルシウム、硫酸マグネシウム、炭酸カルシウム)、クエン酸、酒石酸、グリシン、低、中、高分子量のポリエチレングリコール(PEG)、プルロニック(登録商標)、カオリン、ケイ酸、あるいはそれらの組み合わせが例として挙げられる。
結合剤としては、トウモロコシ、コムギ、コメ、若しくはジャガイモのデンプンを用いたデンプン糊、単シロップ、グルコース液、ゼラチン、トラガカント、メチルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロースナトリウム、セラック及び/又はポリビニルピロリドン等が例として挙げられる。
崩壊剤としては、前記デンプンや、乳糖、カルボキシメチルデンプン、架橋ポリビニルピロリドン、アガー、ラミナラン末、炭酸水素ナトリウム、炭酸カルシウム、アルギン酸若しくはアルギン酸ナトリウム、ポリオキシエチレンソルビタン脂肪酸エステル、ラウリル硫酸ナトリウム、ステアリン酸モノグリセリド又はそれらの塩が例として挙げられる。
充填剤としては、前記糖及び/又はリン酸カルシウム(例えば、リン酸三カルシウム、若しくはリン酸水素カルシウム)が例として挙げられる。
乳化剤としては、ソルビタン脂肪酸エステル、グリセリン脂肪酸エステル、ショ糖脂肪酸エステル、プロピレングリコール脂肪酸エステルが例として挙げられる。
流動添加調節剤及び滑沢剤としては、ケイ酸塩、タルク、ステアリン酸塩又はポリエチレングリコールが例として挙げられる。
このような担体は、主として前記剤形形成を容易にし、また剤形及び薬理効果を維持するために用いられるものであり、必要に応じて適宜使用すればよい。上記の添加剤の他、必要であれば矯味矯臭剤、可溶化剤、懸濁剤、希釈剤、界面活性剤、安定剤、吸収促進剤、増量剤、付湿剤、保湿剤、吸着剤、崩壊抑制剤、コーティング剤、着色剤、保存剤、抗酸化剤、香料、風味剤、甘味剤、緩衝剤等を含むこともできる。
本発明の医薬組成物は、薬剤の薬理効果を失わない範囲において、他の薬剤を含有することもできる。例えば、注射剤の場合であれば、他の抗生物質を所定量含有していても良い。
本発明の医薬組成物の剤形は、有効成分である薬剤、及び他の付加的な有効成分を不活化させない形態であれば特に限定しない。例えば、液体、固体又は半固体のいずれであってもよい。具体的な剤形としては、例えば、液剤、散剤、顆粒剤、錠剤、カプセル剤、舌下剤、トローチ剤等の経口剤形又は注射剤、懸濁剤、乳剤、点眼剤、点鼻剤、クリーム剤、軟膏剤、硬膏剤、シップ剤及び座剤等の非経口剤形が挙げられる。
本発明の医薬組成物は、含有される有効成分が失活しない、あらゆる適当な方法で投与することができる。例えば、経口又は非経口(例えば、注射、エアロゾル、塗布、点眼、点鼻)のいずれであってもよい。
本発明の医薬組成物の投与頻度は、3日に1回~1日5回、特に1日1回~1日3回とすることが好ましい。本発明の医薬組成物の投与期間は特に限定されないが、例えば、3日間~1ヶ月、特に1週間~2週間とすることができる。
3.食品用組成物
本発明の組成物は、食品用組成物として使用してもよい。以下、本項において、本発明の組成物を食品用組成物とも称する。本発明の食品用組成物は、既存の食品に添加するための組成物であってもよく、また、組成物自体を食品の主要構成物としてもよい。なお、本発明の食品用組成物は、利用者の健康増進を目的とするものであり、製造者の利便のために使用される、食品衛生法上の「食品添加物」とは異なるものである。
本発明の組成物は、食品用組成物として使用してもよい。以下、本項において、本発明の組成物を食品用組成物とも称する。本発明の食品用組成物は、既存の食品に添加するための組成物であってもよく、また、組成物自体を食品の主要構成物としてもよい。なお、本発明の食品用組成物は、利用者の健康増進を目的とするものであり、製造者の利便のために使用される、食品衛生法上の「食品添加物」とは異なるものである。
本明細書において「食品」とは、対象が日常的な栄養摂取及び/又は健康維持のために摂取する物を指す。ここでいう食品を摂取する対象は、ヒト、チンパンジーを含む霊長類、イヌ、ネコなどのペット動物、ウシ、ウマ、ヒツジ、ヤギなどの家畜動物、マウス、ラットなどの齧歯類、動物園で飼育される動物などの哺乳動物であり、好ましくは、ヒトである。本発明の食品用組成物は、ウイルス感染症、特にCOVID-19感染症の予防を目的として摂取されてもよい。
本発明の食品用組成物を既存の食品に添加する場合、特に限定されないが、有効成分を効率よく分散させることから、例えば、飲料、スープ、シチュー等の流動性のある食品や、ジャム、味噌、ヨーグルト等のペースト状の食品に添加することができる。また、有効成分の有効性の維持を容易にするため、例えば、-18℃以下の冷凍食品や、2~10℃の冷蔵食品に添加することができる。いずれの場合も、食品用組成物の添加量は、有効成分の効果が維持され、かつ、食品の嗜好性を損なわない量とすることが好ましい。組成物の添加量は、添加する対象となる食品によって異なるが、適宜調整することが可能である。
本発明の食品用組成物は、それ自体が食品の主要構成物であってもよい。この場合、食品用組成物の性状は、対象が無理なく摂取できるものであれば特に限定されないが、例えば、液体(飲料)、粉末、顆粒、錠剤又はカプセル剤等とすることができる。この場合、本発明の組成物は、必要に応じて担体を含むことができる。担体としては、賦形剤、結合剤、崩壊剤、充填剤、乳化剤、流動添加調節剤、滑沢剤等が挙げられる。使用可能な担体は、必要に応じて、例えば「2.医薬組成物」に記載のものを適用可能である。
本発明の食品用組成物において、有効成分の含有量は、対象の摂取量が1~100nmol/kg体重/日、特に10~100nmol/kg体重/日となるような含有量とすることができる。摂取頻度は、3日に1回~1日5回、特に1日1回~1日3回とすることが好ましい。本発明の食品用組成物の好適な摂取頻度及び期間は特に限定されない。
4.衛生用品
本発明の衛生用品は、「1-1.有効成分」の項に記載した化合物のうち、少なくとも1つの化合物を含む組成物を含むことを特徴とする。

本明細書において、「衛生用品」とは、健康者であるヒトが、日常生活において、身体の清浄、室内の衛生維持、病気の予防等をすることにより、健康を保持するため使用する機械器具等でないものをいう。本明細書においては、薬機法(医薬品、医療機器等の品質、有効性及び安全性の確保等に関する法律)上の「医薬部外品」も「衛生用品」に含まれるものとする。例えば、衛生用品には、ガーゼ、シート、マスク、綿棒等が含まれる。本発明の衛生用品は、特に限定されないが、例えば、ヒトやペットの身体を清拭する清拭材や、室内の拭き掃除等に使用される清掃シート等として好適に使用可能である。本発明の衛生用品は、抗ウイルス活性を有する組成物を含むことから、ウイルス感染症、特にCOVID-19感染症の予防効果が期待できる。
本発明の衛生用品は、例えば、紙、不織布、布、脱脂綿等に、有効成分を含む組成物を水溶液等の状態で含浸させた、湿潤物品(例えば、湿潤シート)とすることができる。あるいは、本発明の衛生用品は、液状の組成物そのものであり(衛生組成物)、使用直前に、使用者が紙、不織布、布、脱脂綿等に当該組成物を噴霧するなどして含浸させてから使用してもよい。衛生用品に含浸させて使用する場合、本発明の組成物における有効成分の含有量は、ウイルス感染症予防効果が得られ、かつ、衛生用品をヒトの皮膚等に接触させても発赤等の異常が生じにくい含有量とすることが好ましい。組成物における有効成分の含有量は、使用する有効成分、具体的な衛生用品の素材、形状、使用形態によって適宜調整できるが、例えば、1~100mM、特に10~100mMとすることができる。
本発明の衛生用品には、前記有効成分に加えて、エタノール、イソプロパノール、次亜塩素酸ナトリウム、塩化セチルピリジニウム(CPC)、ポピヨンヨード、乳化ナトリウム等の公知の抗菌剤、殺菌剤又は除菌剤が含まれていてもよい。
以下、本発明の実施例により、本発明の内容をさらに詳細に説明するが、本発明を実施例の範囲に限定することを意図したものではない。
<実施例1:アスピロクロリンによるCOVID-19メインプロテアーゼ阻害試験>
(1)試料調製
COVID-19メインプロテアーゼ(Mpro)(R&D Systems Recombinant SARS-CoV-2 3CL Protease,キャリアフリー,No.E-720(Bio-Techne社製))を、20mM Tris-HCl(pH7.5)で終濃度125nMとなるように希釈した。
(1)試料調製
COVID-19メインプロテアーゼ(Mpro)(R&D Systems Recombinant SARS-CoV-2 3CL Protease,キャリアフリー,No.E-720(Bio-Techne社製))を、20mM Tris-HCl(pH7.5)で終濃度125nMとなるように希釈した。
(2)アスピロクロリンの調製
アスピロクロリン(下記式(VII))は、以下の方法で調製した。
A.oryzae AOKW8株(aclR遺伝子強制発現株(非特許文献5参照))の胞子をCDSTUU培地20mLに植菌し、30℃にて振盪培養を24時間行った(前培養)。本前培養物を5.0LのCDSTUU培地に全量植菌し、30℃にて振盪培養をさらに62時間行った。得られた培養物5.0Lをメタノール5.0Lで抽出し、濾過にて菌体を除去した後に減圧濃縮を行い、発酵生産物エキスを得た。本エキスに対し、水-酢酸エチル系での分配操作を行い、有機層を回収した。本有機層を、計2度のシリカゲルカラムクロマトグラフィー、続いて計2度の逆相HPLCによる精製を通じ、アスピロクロリン14.9mgを得た。得られた化合物の分子構造は、1H-NMR及び13C-NMRを用いて確認した。
アスピロクロリン(下記式(VII))は、以下の方法で調製した。
(3)アスピロクロリンによるCOVID-19 Mpro阻害試験
96ウェルマイクロタイタープレートに、(1)で調製したMpro希釈液を49μL添加し、ここに、各濃度のアスピロクロリン溶液1μLを加えた。アスピロクロリンの終濃度は、0、5、10、50、100、500及び1000μMとした。室温で5分間静置した後、10μg/mLのCovidyte TF670(AAT Bioquest社製)溶液(基質溶液)を50μL添加し、2時間反応させた。基質添加後、5分毎にマイクロプレート蛍光光度計(SpectraMax Gemini EM(Molecular Devices社製))で640/680nmの蛍光強度を測定した。
96ウェルマイクロタイタープレートに、(1)で調製したMpro希釈液を49μL添加し、ここに、各濃度のアスピロクロリン溶液1μLを加えた。アスピロクロリンの終濃度は、0、5、10、50、100、500及び1000μMとした。室温で5分間静置した後、10μg/mLのCovidyte TF670(AAT Bioquest社製)溶液(基質溶液)を50μL添加し、2時間反応させた。基質添加後、5分毎にマイクロプレート蛍光光度計(SpectraMax Gemini EM(Molecular Devices社製))で640/680nmの蛍光強度を測定した。
図2は、各濃度のアスピロクロリンを添加した際のMproの酵素活性の推移を示す。アスピロクロリンの終濃度を500μM以上とした場合に、Mproの活性がわずかに下がることが確認できた。
<実施例2:アスピロクロリン前駆体によるCOVID-19メインプロテアーゼ阻害試験>
(1)アスピロクロリン前駆体(化合物2)の調製
下記式(VIII)で表される化合物2は、以下の方法で調製した。
A.oryzae AOKW8株(aclR遺伝子強制発現株(非特許文献5参照))の胞子をCDSTUU培地20mLに植菌し、30℃にて振盪培養を24時間行った(前培養)。本前培養物を5.0LのCDSTUU培地に全量植菌し、30℃にて、振盪培養をさらに62時間行った。得られた培養物5.0Lをメタノール5.0Lにて抽出し、濾過にて菌体を除去した後に減圧濃縮を行い、発酵生産物エキスを得た。本エキスに対し、水-酢酸エチル系での分配操作を行い、有機層を回収した。本有機層を、計2度のシリカゲルカラムクロマトグラフィー、続いて計2度の逆相HPLCによる精製を通じ、アスピロクロリン前駆体(化合物2)2.2mgを得た。得られた化合物の分子構造は、1H-NMR及び13C-NMRを用いて確認した。
(1)アスピロクロリン前駆体(化合物2)の調製
下記式(VIII)で表される化合物2は、以下の方法で調製した。
(2)アスピロクロリン前駆体(化合物3)の調製
下記式(IV)で表される化合物3は、以下の方法で調製した。
A.oryzae ΔaclO/AOKW8株(aclO欠損型aclR遺伝子強制発現株(非特許文献5参照))の胞子をCDST培地20mLに植菌し、30℃にて振盪培養を24時間行った。本前培養物を5.0LのCDST培地に全量植菌し、30℃にて振盪培養をさらに70時間行った。得られた培養物5.0Lをアセトン5.0Lにて抽出し、濾過にて菌体を除去した後に減圧濃縮を行い、発酵生産物エキスを得た。本エキスに対し、水-酢酸エチル系での分配操作を行い、有機層を回収した。本有機層を、シリカゲルカラムクロマトグラフィー、続いて計2度の逆相HPLCによる精製を通じ、アスピロクロリン前駆体(化合物3)2.6mgを得た。得られた化合物の分子構造は、1H-NMR及び13C-NMRを用いて確認した。
下記式(IV)で表される化合物3は、以下の方法で調製した。
(3)アスピロクロリン前駆体(化合物15)の調製
下記式(V)で表される化合物15は、以下の方法で調製した。
A.oryzae ΔaclL/AOKW8株(aclL欠損型aclR遺伝子強制発現株(非特許文献5参照))の胞子をCDST培地20mLに植菌し、30℃にて振盪培養を23時間行った(前培養)。本前培養物を5.0LのCDST培地に全量植菌し、30℃にて振盪培養をさらに70時間行った。得られた培養物5.0Lをアセトン2.0Lにて抽出し、濾過にて菌体を譲与した後に減圧濃縮を行い、発酵生産物エキスを得た。本エキスに対し、水-酢酸エチル系での分配操作を行い、有機層を回収した。本有機層を、シリカゲルカラムクロマトグラフィー、続いて計2度の逆相HPLCによる精製を通じ、アスピロクロリン前駆体(化合物15)5.0mgを得た。得られた化合物の分子構造は、1H-NMR及び13C-NMRを用いて確認した。
下記式(V)で表される化合物15は、以下の方法で調製した。
(4)アスピロクロリン前駆体(化合物16)の調製
下記式(VI)で表される化合物16は、以下の方法で調製した。
A.oryzae ΔaclR/AOKW7-OE::aclP株(aclR欠損型aclP遺伝子強制発現株(非特許文献6参照))の胞子をmodified zeolite培地20mLに植菌し、30℃にて振盪培養を23時間行った(前培養)。本前培養物を5.0Lの同培地に全量植菌し、30℃にて振盪培養をさらに50時間行った。得られた培養物5.0Lをエタノール8.0Lにて抽出し、濾過にて菌体を除去した後に減圧濃縮を行い、発酵生産物エキスを得た。本エキスに対し、水-酢酸エチル系での分配操作を行い、有機層を回収した。本有機層を、シリカゲルカラムクロマトグラフィー、続いて計2度の逆相HPLCによる精製を通じ、アスピロクロリン前駆体(化合物16)0.9mgを得た。得られた化合物の分子構造は、1H-NMR及び13C-NMRを用いて確認した。
下記式(VI)で表される化合物16は、以下の方法で調製した。
(5)各化合物によるCOVID-19 Mpro阻害試験
COVID-19 Mpro希釈液は、実施例1と同様の方法で調製した。96ウェルマイクロタイタープレートに、Mpro希釈液を49μL添加し、ここに、化合物2、3、15及び16の溶液1μLをそれぞれ加えた。各化合物の終濃度は、200μMとした。室温で5分間静置した後、10μg/mLのCovidyte TF670(AAT Bioquest社製)溶液(基質溶液)を50μL添加し、2時間反応させた。基質添加後、5分毎にマイクロプレート蛍光光度計(SpectraMax Gemini EM(Molecular Devices社製))で640/680nmの蛍光強度を測定した。陰性対照として、化合物溶液に代えてDMSOを1μL添加したものについても同様の試験を行った。
COVID-19 Mpro希釈液は、実施例1と同様の方法で調製した。96ウェルマイクロタイタープレートに、Mpro希釈液を49μL添加し、ここに、化合物2、3、15及び16の溶液1μLをそれぞれ加えた。各化合物の終濃度は、200μMとした。室温で5分間静置した後、10μg/mLのCovidyte TF670(AAT Bioquest社製)溶液(基質溶液)を50μL添加し、2時間反応させた。基質添加後、5分毎にマイクロプレート蛍光光度計(SpectraMax Gemini EM(Molecular Devices社製))で640/680nmの蛍光強度を測定した。陰性対照として、化合物溶液に代えてDMSOを1μL添加したものについても同様の試験を行った。
図3は、各化合物溶液を添加した際のMproの酵素活性の推移を示す。いずれの化合物も、DMSOと比較してMproの活性を下げることが確認できたが、化合物3、15及び16、特に化合物3及び16を添加した場合にMproの活性が顕著に下がることが確認できた。
<実施例3:アスピロクロリン前駆体のCOVID-19 Mpro50%阻害濃度(IC50)の測定
COVID-19 Mpro(R&D Systems Recombinant SARS-CoV-2 3CL Protease,キャリアフリー,No.E-720(Bio-Techne社製))を、20mM Tris-HCl(pH7.5)で終濃度30nMとなるように希釈してMpro希釈液を調製した。96ウェルマイクロタイタープレートに、Mpro希釈液を49μL添加し、ここに、実施例2で調製した化合物3、15及び16の溶液1μLをそれぞれ加えた。化合物3、15又は16の終濃度は、0.3μM~200μMとした。各ウェルに10μg/mLのCovidyte TF670(AAT Bioquest社製)溶液(基質溶液)を50μL添加し、2時間反応させ、マイクロプレート蛍光光度計(SpectraMax Gemini EM(Molecular Devices社製))で640/680nmの蛍光強度を測定した。化合物を添加しない条件の蛍光強度を1.0として、各化合物溶液添加時の蛍光強度を算出した。
COVID-19 Mpro(R&D Systems Recombinant SARS-CoV-2 3CL Protease,キャリアフリー,No.E-720(Bio-Techne社製))を、20mM Tris-HCl(pH7.5)で終濃度30nMとなるように希釈してMpro希釈液を調製した。96ウェルマイクロタイタープレートに、Mpro希釈液を49μL添加し、ここに、実施例2で調製した化合物3、15及び16の溶液1μLをそれぞれ加えた。化合物3、15又は16の終濃度は、0.3μM~200μMとした。各ウェルに10μg/mLのCovidyte TF670(AAT Bioquest社製)溶液(基質溶液)を50μL添加し、2時間反応させ、マイクロプレート蛍光光度計(SpectraMax Gemini EM(Molecular Devices社製))で640/680nmの蛍光強度を測定した。化合物を添加しない条件の蛍光強度を1.0として、各化合物溶液添加時の蛍光強度を算出した。
図4は、各化合物溶液とMproの活性との関係を示す。表1に、図4の結果より算出した、各化合物のMpro50%阻害濃度(IC50)を示す。化合物3及び16において、Mproに対する特に高い阻害活性が観察された。
本発明は、製薬業、食品製造業、衛生用品製造業、飲食業等の幅広い分野で利用可能な発明である。

Claims (11)
- 下記式(I)で表される化合物からなる群から選択される、少なくとも1つの化合物又はその誘導体を有効成分として含む、抗ウイルス活性を有する組成物。
- 前記化合物が、下記式(II)及び(III)で表される構造を有する化合物からなる群より選択される、請求項1に記載の組成物。
- 前記抗ウイルス活性が、COVID-19の増殖阻害活性である、請求項1~5のいずれか1項に記載の組成物。
- 前記抗ウイルス活性が、COVID-19のメインプロテアーゼ阻害活性である、請求項6に記載の組成物。
- 前記有効成分の含有量が、アスピロクロリンの含有量よりも高い、請求項1~7のいずれか1項に記載の組成物。
- 前記組成物が医薬組成物である、請求項1~8のいずれか1項に記載の組成物。
- 前記組成物が食品用組成物である、請求項1~8のいずれか1項に記載の組成物。
- 請求項1~8のいずれか1項に記載の組成物を含む、衛生用品。
