Abstract
本願発明の課題はホワイトスポットシンドロームウイルス(WSSV)感染に対する防御効果のあるワクチンを提供することである。本発明のホワイトスポットシンドロームウイルス(WSSV)の感染を予防又は治療するためのワクチンは、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列を含む抗原性ペプチドであって、8~30個のアミノ酸残基からなる抗原性ペプチドを含む。
Description
本発明は、ホワイトスポットシンドロームウイルス(WSSV)の感染を予防又は治療するためのワクチンに関する。
ホワイトスポット病(以下、WSD)は、水生甲殻類が罹るウイルス性の感染症であり、外殻に斑点が生じ、発症から2週間以内にほぼ100%が死滅するため、養殖エビの最も壊滅的な病気の1つとして知られている。WSDによる世界的な損失は約150億ドルと推定され、年間10億ドルの増加率で被害が拡大していると言われている。したがって、世界的にWSDの有効な対策に対する需要は高い。
WSDの原因ウイルスは、ニマウイルス科(Nimaviridae)Whispovirus属の唯一の種であるホワイトスポットシンドロームウイルス(WSSV)である。WSSVは、スーパーコイル状の環状2本鎖DNA(dsDNA)を含む大きな桿菌型エンベロープウイルスであり、ビリオンのサイズは約80~100×250~350nmであり、棒状のヌクレオカプシドが三層膜で覆われており、ゲノムの長さは285~305kbpで、9つのタンデムリピート保存領域と180の推定オープンリーディングフレーム(ORF)があるが、ORFの多くはデータベースの既知のタンパク質と相同性がない。またWSSVは、少なくとも6つのビリオンタンパク質、すなわち、エンベロープタンパク質由来のVP19及びVP28、テグメントタンパク質由来のVP24及びVP26、並びに、ヌクレオカプシドタンパク質由来のVP15及びVP664を含むことが知られている(非特許文献1、2)。
WSSVに感染したエビには、嗜眠、食欲不振、食物摂取の減少、修復活性の低下、甘皮の弛緩、赤みがかった変色、外骨格上の白い石灰化スポット(直径0.5~3mm)等の症候が見られ、一週間以内に90%の致死率をもたらす。
WSSVワクチンの開発において、ウイルスタンパク質サブユニット、弱毒化されたWSSV及びDNA/RNAベースの免疫剤が知られている(非特許文献3、4)。その大部分は、VP19、VP24、VP26、VP28、VP292及びVP466等の組換えサブユニットワクチンで、その中ではVP28の利用が報告されている(非特許文献5~9)。これらのWSSVサブユニットワクチンは、大腸菌で発現したものである。
本発明者はこれまでに、5つの組換えWSSVタンパク質VP15、VP19、VP24、VP26及びVP28の抗WSSV効果(WSSV感染防御効果)を調べた。これらのタンパク質をカイコ及び大腸菌を用いて発現及び精製を行い、クルマエビを用いたin vivo抗ウイルスアッセイにより、ヌクレオカプシドタンパク質VP15がWSSV感染に対する高い免疫原性を示すことを確認した(非特許文献10)。

本発明は、ホワイトスポットシンドロームウイルス(WSSV)感染に対する防御効果のあるワクチンを提供することを目的とする。
本発明者らは、鋭意研究を重ねた結果、WSSVタンパク質VP15の断片ペプチドであるSR-11(配列番号7)が、高いWSSV感染防御効果を有することを見出し、本発明を完成させた。
すなわち、本発明は、以下のとおりである。

配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列を含む抗原性ペプチドであって、8~30個のアミノ酸残基からなる抗原性ペプチドを含む、ホワイトスポットシンドロームウイルス(WSSV)の感染を予防又は治療するためのワクチン。

本発明のワクチンによれば、甲殻類動物においてホワイトスポット病を予防又は治療することができる。また、本発明における抗原性ペプチドが短く(8~50個のアミノ酸残基)、かつ、大腸菌又はカイコにおいて簡単に大量に生産できるため、養殖エビへの適用が期待される。
〔ワクチン〕
本実施形態のワクチンは、ホワイトスポットシンドロームウイルス(WSSV)の感染を予防又は治療するためのワクチンであり、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列を含む抗原性ペプチドであって、8~30個のアミノ酸残基からなる抗原性ペプチドを含む。ここで、抗原性ペプチドとは、抗原性(免疫原性)を有するペプチド、特にエビなどの水生甲殻類動物において、抗原性、特にWSSV抗原性を有するペプチドを意味する。
本実施形態のワクチンは、ホワイトスポットシンドロームウイルス(WSSV)の感染を予防又は治療するためのワクチンであり、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列を含む抗原性ペプチドであって、8~30個のアミノ酸残基からなる抗原性ペプチドを含む。ここで、抗原性ペプチドとは、抗原性(免疫原性)を有するペプチド、特にエビなどの水生甲殻類動物において、抗原性、特にWSSV抗原性を有するペプチドを意味する。
WSSVのゲノム配列(WSSV CN01、NCBI、アクセッション番号:NC_003225)は知られており、また、5つのタンパク質:VP15(wsv 214)、VP19(wsv 414)、VP24(wsv 002)、VP26(wsv 311)及びVP28(wsv 421)の存在も知られている。VP15(配列番号1)は、80アミノ酸からなるタンパク質であり、極めて高いpI値を有し、真核生物起源のDNA結合タンパク質やバキュロウイルスp6.9との相同性を示している。VP15は、セリン、アルギニン及びリジンをそれぞれ20~21%含む。VP15は、ウイルスゲノムのキャプシドへのパッケージングに関与している主要なヌクレオカプシドタンパク質であるにもかかわらず、その結晶構造は未だに知られていない。
配列番号7に示すアミノ酸配列からなるペプチド(全長:11アミノ酸残基)は、VP15タンパク質の37番目~47番目のアミノ酸残基からなるVP15の断片ペプチドであり、本明細書において「SR-11」と呼ばれる場合がある。SR-11は、エビに投与することによって、免疫を惹起し、WSSVに対する感染予防効果をもたらす。すなわち、SR-11はWSSV抗原性(WSSV免疫原性)を持ち、WSSV感染防御を可能にする、WSSVワクチンとして機能する。
本実施形態における抗原性ペプチドは、上述のとおり、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列(本明細書において「SR-11変異体」と呼ぶ場合がある)を含む。すなわち、本実施形態における抗原性ペプチドは、少なくともその一部として、SR-11又はSR-11変異体を含む。SR-11変異体は、WSSV抗原性を持っていれば特に限定されないが、SR-11のアミノ酸配列中の任意の位置における、1個、2個若しくは3個のアミノ酸残基の欠失、置換若しくは挿入を有してよい。2個若しくは3個のアミノ酸残基の場合、連続したアミノ酸残基であっても、不連続のアミノ酸残基であってもよい。
置換又は挿入付加されるアミノ酸は天然型と非天然型を問わない。天然型アミノ酸としては、L-アラニン、L-アスパラギン、L-アスパラギン酸、L-グルタミン、L-グルタミン酸、グリシン、L-ヒスチジン、L-イソロイシン、L-ロイシン、L-リジン、L-アルギニン、L-メチオニン、L-フェニルアラニン、L-プロリン、L-セリン、L-スレオニン、L-トリプトファン、L-チロシン、L-バリン、L-システイン等が挙げられる。
以下に、相互に置換可能なアミノ酸の例を示す。同一群に含まれるアミノ酸は相互に置換可能である。

本実施形態における抗原性ペプチドは、上述のSR-11又はSR-11変異体からなるか、又は上述のSR-11又はSR-11変異体を一部として含み、WSSV抗原性を持っていればその長さは特に限定されないが、好ましくは8~30個のアミノ酸残基からなり、より好ましくは、8~25個、8~20個、9~20個、10~20個、9~15個、10~15個、9~13個、又は10~12個のアミノ酸残基からなる。抗原性ペプチドがWSSV抗原性を有する限り、抗原性ペプチドにおけるSR-11又はSR-11変異体以外の部分のアミノ酸残基は特に限定されず、SR-11又はSR-11変異体のWSSV抗原性に影響を与えないか、WSSV抗原性を向上させるアミノ酸残基であることが好ましい。
本実施形態における抗原性ペプチドはまた、当該抗原性ペプチドの精製に利用されるタンパク質タグと融合タンパク質を形成した形態であってもよい。タグとしては、例えばHisタグ、Flagタグ、mycタグ、グルタチオン-S-トランスフェラーゼ(GST)、マルトース結合タンパク質(MBP)、ストレップ-タグ等が挙げられ、これらのタグは短いアミノ酸配列を有しており、WSSV抗原性に影響を与えないため、抗原性ペプチドとの融合タンパク質のままにワクチンとして使用されてもよい。または、精製後抗原性ペプチドとタンパク質タグとを切り離してもよい。
本実施形態における抗原性ペプチドは、SR-11又はSR-11変異体からなることが好ましい。すなわち、抗原性ペプチドが、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個、好ましくは2個、より好ましくは1個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列からなることが好ましく、配列番号7に示すアミノ酸配列からなることが特に好ましい。
本実施形態における、SR-11又はSR-11変異体を含む抗原性ペプチドは、配列番号1に示すアミノ酸配列の連続した8~30個のアミノ残基からなるペプチドであるか、又は、配列番号1に示すアミノ酸配列の連続した複数のアミノ残基からなるペプチドと80%以上、好ましくは85%以上、90%以上、又は95%以上の配列同一性を有する、8~30個のアミノ残基からなるペプチドであってもよい。配列番号1に示すアミノ酸配列からなるタンパク質は、VP15タンパク質であり、VP15タンパク質の37番目~47番目のアミノ酸残基からなるペプチドはSR-11である。上述したように、本実施形態における抗原性ペプチドは、SR-11又はSR-11変異体を配列の全部又は一部として含む。すなわち、本実施形態における抗原性ペプチドは、VP15タンパク質の断片ペプチド又は断片ペプチド変異体であって、SR-11又はSR-11変異体を配列の全部又は一部として含むものである。
本実施形態における抗原性ペプチドは、人工的に合成してもよいが、後述の形質転換体や組換えカイコにて発現させることによって大量生産することができる。後述の形質転換体又は組換えカイコにて発現させたペプチドを用いる場合、目的の抗原性ペプチドを精製しても精製しなくてもよい。特に経口投与の場合は、組換えカイコをそのまま乾燥させて、餌に混ぜて使用してもよい。
本実施形態のワクチンは、有効量の抗原性ペプチドのみからなるものであってもよく、有効量の抗原性ペプチド以外の他の成分を含んでいてもよい。
本実施形態のワクチンの剤形は、例えば、液状、粉末状(凍結乾燥粉末、乾燥粉末)、顆粒状等であってもよい。抗原性ペプチド以外の他の成分としては、ワクチン製造に通常用いられる担体であってよく、例えば、食塩水、緩衝食塩水、デキストロース、水、グリセロール、等張水性緩衝液及びそれらの組み合わせが挙げられる。他の成分としてはさらに、乳化剤、保存剤(例えば、チメロサール)、等張化剤、pH調整剤、不活化剤(例えば、ホルマリン)等が、更に適宜配合されてもよい。
本実施形態のワクチンは、抗原性ペプチドと、必要に応じて、担体、抗原性を増強する成分等の他の成分と混合することにより得ることができる。
本実施形態のワクチンは、甲殻類動物、例えば養殖用甲殻類動物、特にホワイトスポット病に罹患する甲殻類動物、好ましくはエビ、ロブスター等の水生甲殻類動物に投与されるためのものであり、エビとしては、特にクルマエビ(Marsupenaeus japonicus)、バナメイエビ(Litopenaeus vannamei)、ウシエビ(Penaeus monodon)、及びインドエビ(Fenneropenaeus indicus)などが挙げられる。本実施形態のワクチンを甲殻類動物に投与することによって、ホワイトスポット病を有効に予防又は治療できる。
本実施形態のワクチンの投与経路は特に限定されず、例えば、筋肉内注射であってもよく、経口投与であってもよい。投与回数は1回でも効果があるが、複数回の投与、例えば、2回、3回、4回若しくは5回投与するとよい高い感染防御効果が得られる。また、定期的に投与すること、例えば年に1~25回、特に年に3回、年に4回、年に6回、年に12回又は月に1~4回投与することが好ましい。複数回投与の場合、投与間隔は特に限定されず、例えば1日~3ヶ月、1週間~2か月、10~30日間又は15~25日間あけて投与してもよい。また、経口投与の場合、食餌に混ぜて、一定の間隔で(例えば、3か月に1回、2か月に1回、1か月に1回、10日に1回、5日に1回、又は毎日)給餌することによって投与してもよい。
本実施形態のワクチンの投与量は、投与頻度及び投与経路によって異なり得るが、例えば、1~100μg抗原性ペプチド/g体重であればよく、5~50μg抗原性ペプチド/g体重、10~45μg抗原性ペプチド/g体重、又は、10~20μg抗原性ペプチド/g体重であることが好ましい。
〔ペプチド〕
本実施形態のペプチドは、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換、若しくは挿入を有するアミノ酸配列を含み、かつ、8~30個のアミノ酸残基を有する、単離されたペプチドである。本実施形態のペプチドは、上述の抗原性ペプチドであってよい。
本実施形態のペプチドは、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換、若しくは挿入を有するアミノ酸配列を含み、かつ、8~30個のアミノ酸残基を有する、単離されたペプチドである。本実施形態のペプチドは、上述の抗原性ペプチドであってよい。
〔食餌〕
本実施形態の食餌は、本実施形態のワクチン又はペプチドを有効量で含む。食餌は、甲殻類動物のための食餌であることが好ましく、ワクチン又はペプチドの有効量としては、1~100μg抗原性ペプチド/日/g体重であればよく、5~50μg抗原性ペプチド/日/g体重、10~45μg抗原性ペプチド/g体重、又は、10~20μg抗原性ペプチド/日/g体重であることが好ましい。有効量が投与間隔によって異なり得、適宜に調整することができる。例えば、5日に1回の投与の有効量は、毎日投与の有効量が1.5~3倍、好ましくは2倍であってよい。
本実施形態の食餌は、本実施形態のワクチン又はペプチドを有効量で含む。食餌は、甲殻類動物のための食餌であることが好ましく、ワクチン又はペプチドの有効量としては、1~100μg抗原性ペプチド/日/g体重であればよく、5~50μg抗原性ペプチド/日/g体重、10~45μg抗原性ペプチド/g体重、又は、10~20μg抗原性ペプチド/日/g体重であることが好ましい。有効量が投与間隔によって異なり得、適宜に調整することができる。例えば、5日に1回の投与の有効量は、毎日投与の有効量が1.5~3倍、好ましくは2倍であってよい。
本実施形態の食餌は、本実施形態のワクチン又はペプチドの他に、甲殻類動物に適した食餌成分を含むものであればよい。本実施形態の食餌は、具体的には、例えばタンパク質、脂質、炭水化物、ビタミン及びミネラル類を含んでいてもよく、より具体的には、フィッシュミール、オキアミミール、小麦粉、とうもろこし、アルファルファミール、ビール酵母、小麦胚芽、イカミール、海藻粉末、濃縮アルファルファ、魚油、スピルリナ、キトサン、ケール、海苔、アミノ酸、カロチノイド、ガーリック、ビタミン類(塩化コリン、ビタミンE、ビタミンC、イノシトール、ビタミンB5、ビタミンB2、ビタミンA、ビタミンB1、ビタミンB6、ビタミンB3、葉酸、ビタミンD3、ビオチン)、ミネラル類(Ca、Fe、Mg、Zn、Mn、Cu、I)を含んでもよい。
食餌としては、市販の飼料に本実施形態のワクチン又はペプチドを添加したものであってもよく、ここで、本実施形態のワクチン又はペプチドとして、後述の形質転換体又は組換えカイコにて発現させたペプチドを用いる場合、目的の抗原性ペプチドを精製することなく、形質転換体又は組換えカイコの乾燥粉末をそのまま用いてもよい。一実施形態の食餌は、本実施形態のワクチン又はペプチドを含む健康飼料であってよい。
本実施形態の食餌を、甲殻類動物に一定の間隔で(例えば、3か月に1回、2か月に1回、1か月に1回、10日に1回、5日に1回、又は毎日)投与することによって、ホワイトスポット病を有効に予防又は治療できる。
〔核酸分子〕
本実施形態の核酸分子は、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列からなるペプチドを含み、8~30個のアミノ酸残基を有する抗原性ペプチドをコードする。
本実施形態の核酸分子は、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列からなるペプチドを含み、8~30個のアミノ酸残基を有する抗原性ペプチドをコードする。
本実施形態の核酸分子は、例えば、配列番号1に示すアミノ酸配列をコードするワイルドタイプVP15遺伝子を鋳型としてエラープローンPCR等に供することにより取得することができる。または、部位特異的変異を導入することによっても得ることができる。本実施形態の核酸分子は、また配列番号1又は7のアミノ酸配列に基づき設計したアミノ酸配列をコードする核酸分子を人工合成によって得ることができる。
上記のように取得した核酸分子は、そのまま、又は適当な制限酵素等で切断し、常法によりベクターに組み込み、得られた組換え発現ベクターを宿主細胞に導入した後、通常用いられる塩基配列解析方法等の塩基配列分析装置を用いて分析することにより、該核酸分子の塩基配列を決定することができる。
核酸分子の塩基配列を決定する際に用いることができるベクターとしては、pFastBac、又はpGEX-6P-1等が挙げられる。
本実施形態の核酸分子は、本実施形態の抗原性ペプチドをコードする核酸の他に、上述のタンパク質タグをコードする核酸を含んでもよい。この場合、本実施形態の抗原性ペプチドとタンパク質タグとが融合タンパク質として発現できるようにすることが好ましい。
〔形質転換体・組換えカイコ〕
本実施形態の形質転換体は、本実施形態の核酸分子で宿主細胞を形質転換することによって得ることができる。
本実施形態の形質転換体は、本実施形態の核酸分子で宿主細胞を形質転換することによって得ることができる。
宿主細胞としては、本実施形態の抗原性ペプチドを発現できるものであれば特に限定されないが、例えば、大腸菌等の原核生物、酵母、昆虫細胞、植物細胞、動物細胞等が挙げられる。好ましくは大腸菌、酵母、昆虫細胞又は哺乳類動物培養細胞である。これらの宿主細胞で抗原性ペプチドを発現させるために、本実施形態の核酸分子をそのまま、又は本実施形態の核酸分子をベクターに組み込んで得られた組換え発現ベクターを用いて、宿主細胞を形質転換することが好ましい。
組換え発現ベクターに利用できるベクターは特に限定されず、宿主細胞によって適宜に選択することができる。例えば、宿主細胞が大腸菌の場合、ベクターとしては、pGEX-6P-1等が挙げられる。また、酵母細胞の場合、ベクターとしては、pYES2等が挙げられる。動物細胞の場合は、ベクターとしては、pcDNA3.1(+)等が挙げられる。
形質転換体は、上記核酸分子又は組換え発現ベクターを宿主細胞に導入することによって得ることができる。導入は常法によって行えばよい。
形質転換体を適切な培地にて培養し、必要に応じて発現誘導することによって、本実施形態の抗原性ペプチドを発現させ、それを回収し、必要に応じて精製することで、本実施形態の抗原性ペプチドを得ることができる。
本実施形態の組換えカイコは、本実施形態の核酸分子を組み込むことによって得ることができる。
本実施形態の核酸分子はそのまま、又は、本実施形態の核酸分子をベクターに組み込んで得られた組換え発現ベクターを用いてカイコに組み込むことができる。カイコに組み込むためのベクターとしては、例えばpFastBac等が挙げられる。
本実施形態の抗原性ペプチドの産生に組換えカイコが好ましく用いられる。カイコはある程度温度制御が可能な空間があれば飼育でき、また、桑の葉又は人工飼料等を餌とするため、バイオリアクター、培地、発酵槽等の制御装置は必要とせず生産コストも低い。さらにカイコ一頭あたりのタンパク質発現量は、大腸菌培養液30mlの発現量に匹敵するといわれ、立体構造、修飾等の品質が高いタンパク質を得ることもできる。
カイコとしては、幼虫、好ましくは5齢幼虫を用いてもよく、サナギを用いてもよい。カイコの場合、精製コストが高いが、経口投与の場合は、抗原性ペプチドを精製せず、組換えカイコをそのまま飼料として投与できる観点から、サナギであってよい。また、タンパク質及び脂質が豊富であり栄養的に優れている観点からも、サナギであってよい。
本実施形態の核酸分子をカイコに組み込む方法としては、mNPV Bacmid発現方法が挙げられ、抗原性ペプチドを発現させることがでる。また、発現させた後に、抗原性ペプチドをGSTタグ、又はFLAGタグによってアフィニティー回収できる。その後、必要に応じて精製することができる。
以下に実施例を挙げて本発明を詳細に説明するが、本発明は何らこれらに限定されるものではない。
実施例1 抗WSSV効果を示すWSSV抗原のスクリーニング
(1)WSSV-VP15の断片ペプチドの発現
図1に示すように、WSSV-VP15タンパク質(配列番号1)を、1~25番目アミノ酸残基からなるペプチド(VP15(aa1-25);配列番号2)、26~57番目アミノ酸残基からなるペプチド(VP15(aa26-57);配列番号3)、58~80番目アミノ酸残基からなるペプチド(VP15(aa58-80);配列番号4)、及び、1~25番目アミノ酸残基と58~80番目アミノ酸残基とを直接連結したペプチド(VP15(aa1-25,58-80);配列番号5)の4つに断片化した。
(1)WSSV-VP15の断片ペプチドの発現
図1に示すように、WSSV-VP15タンパク質(配列番号1)を、1~25番目アミノ酸残基からなるペプチド(VP15(aa1-25);配列番号2)、26~57番目アミノ酸残基からなるペプチド(VP15(aa26-57);配列番号3)、58~80番目アミノ酸残基からなるペプチド(VP15(aa58-80);配列番号4)、及び、1~25番目アミノ酸残基と58~80番目アミノ酸残基とを直接連結したペプチド(VP15(aa1-25,58-80);配列番号5)の4つに断片化した。
発明者らの以前の研究では、WSSV-VP15(wsv 214)のC末端にFLAGタグ(DYKDDDDK;配列番号10)を融合した融合タンパク質をコードするDNAを作製し、発現ベクターpGEX-6P-1(GEヘルスケア、米国)に、GST(グルタチオン-S-トランスフェラーゼ、26kDa)のC末端にGSTと融合タンパク質となるように、クローニングした(非特許文献10)。この構築されたプラスミドを「pGEX-VP15」と名付けた。
pGEX-VP15を鋳型とし、VP-15及びその上記4つの断片ペプチドのそれぞれを発現するプラスミドを作製した。具体的には、KOD-PLUS-NEO(東洋紡、日本)を用いて、表1に示すプライマーセットを用いて、逆PCRを行った。表1において、各プライマーセットはその左側に記載される断片ペプチドを発現するプラスミド断片を増幅するためのプライマーセットであり、FWはフォーワードプライマーであり、RVはリバースプライマーである。逆PCRは、94℃で2分間の変性の後、98℃で10秒間、57℃で30秒間、68℃で2分30秒間の30サイクルを行い、最後に68℃で7分間伸長した。次に、増幅産物をT4DNAポリメラーゼ(NEB、日本)で処理した後に、自己連結(セルフライゲーション)させた。得られた各ペプチド断片を含むプラスミドを熱ショック法により大腸菌DH5αに形質転換した。形質転換した大腸菌をアンピシリン50μg/mlを含む寒天培地に接種し、アンピシリン陽性コロニーからプラスミドを抽出し、それぞれの断片の配列をサンガーDNA配列決定により確認した。
(2)組換えタンパク質の発現及び精製
(1)で得られた各プラスミドを、エレクトロポレーション法で大腸菌Rosetta gami-B(Novagen,Inc.、日本)に形質転換した。形質転換された大腸菌Rosetta gami-B細胞を、50μg/mlアンピシリンを添加したLB培地中で37℃、一晩増殖させた。次いで、得られた培養物を、250mlのアンピシリン含有LB培地が入ったフラスコに移し、150rpmで振盪しながら37℃でインキュベートした。OD600が0.5に達したとき、培養物を氷上で30分間冷却し、イソプロピル-β-チオガラクトピラノシド(IPTG)を培養物に最終濃度0.5mMになるまで添加することによってタンパク質発現を誘導し、16℃で18時間インキュベーションを継続した。18時間後、大腸菌を遠心分離(6,000×g、4℃、15分)により回収し、リン酸緩衝食塩水(PBS、pH7.3)で2回洗浄し、使用されるまで-80℃で保存した。
(1)で得られた各プラスミドを、エレクトロポレーション法で大腸菌Rosetta gami-B(Novagen,Inc.、日本)に形質転換した。形質転換された大腸菌Rosetta gami-B細胞を、50μg/mlアンピシリンを添加したLB培地中で37℃、一晩増殖させた。次いで、得られた培養物を、250mlのアンピシリン含有LB培地が入ったフラスコに移し、150rpmで振盪しながら37℃でインキュベートした。OD600が0.5に達したとき、培養物を氷上で30分間冷却し、イソプロピル-β-チオガラクトピラノシド(IPTG)を培養物に最終濃度0.5mMになるまで添加することによってタンパク質発現を誘導し、16℃で18時間インキュベーションを継続した。18時間後、大腸菌を遠心分離(6,000×g、4℃、15分)により回収し、リン酸緩衝食塩水(PBS、pH7.3)で2回洗浄し、使用されるまで-80℃で保存した。
大腸菌を、1×プロテイナーゼ阻害剤(cOmplete(商標)、Mini、EDTA-free Protease Inhibitor Cocktail、Sigma-Aldrich、日本)、及び、10μg/mlリゾチームを含有するPBS中に再懸濁した。懸濁液を氷上で超音波処理し(70%振幅、30秒オン/オフ、15サイクル)、遠心分離し(10,000×g、4℃、10分)、0.2μm酢酸セルロース膜(Minisart(登録商標)NML、Sartorius、日本)を通して濾過した。
得られた各VP-15断片ペプチドとGSTの融合タンパク質を含む上澄み及び沈殿物をそれぞれ、SDS-PAGE(15%SDSゲル)、及び抗FLAGタグ抗体(1:10000、MBL、日本)を用いたウエスタンブロット法で解析した。図2(a)はSDS-PAGEの結果を、図2(b)はウエスタンブロットの結果をそれぞれ示す。
VP-15とGSTとの融合タンパク質の分子量は37.0kDaであり、そして、各VP-15断片ペプチドとGSTの各融合タンパク質の分子量は、それぞれ30.8kDa(VP15(aa1-25))、31.6kDa(VP15(aa26-57))、30.5kDa(VP15(aa58-80))及び33.5kDa(VP15(aa1-25, 58-80))であると推測される。図2に示す各融合タンパク質の分子量は推測分子量と一致したことから、各融合タンパク質の発現が確認された。
各GST融合組換えタンパク質を含む上澄みはさらに、GST親和性クロマトグラフィー(Glutathione Sepharose4Fast Flow、GEヘルスケア、日本)によって精製した。タンパク質濃度は、Amicon Ultra-15 30K遠心フィルターユニット(メルク、日本)を用いてPBSに対して透析した後、Pierce BCA Protein Assay Kit(Thermo Fisher Scientific、日本)によって測定した。精製後の融合タンパク質のSDS-PAGE(12%SDSゲル)結果を図3に示す。
(3)各VP-15断片ペプチドのWSSV感染に対する防御効果
クルマエビ(平均体重3.1~6.8g、n=20/群)は、7つの群(5つの実験群と2つの対照群)に分けた。実験群の、VP15群、VP15(aa1-25)群、VP15(aa26-57)群、VP15(aa58-80)群、又はVP15(aa1-25、58-80)群のクルマエビには、それぞれの断片ペプチドを10μg/gエビ体重の量で筋肉内注射(IM)によって接種し(初回免疫)、さらに20日後に2回目の接種(追加免疫)を行い、さらに10日後、WSSV(2.69×104DNAコピー/尾)でチャレンジ(感染)を行った。対照群のエビにも、実験群と同じ要領で、PBS群には10μL/尾のPBSを、対照群のGST群には10μg/gエビ体重のGST-VP15(aa26-57)をそれぞれ接種した。対照群のエビにも、実験群と同じ要領でPBS又はGSTを筋肉内注射した。

なお、WSSVは以下のように入手した。天然のWSSVに感染した稚エビから調製したウイルスの10-3希釈液をクルマエビ成体(平均体重78g)に筋肉内注射し、血リンパは感染3日後に採取し、-80℃で保存した。各実験の前に、保存されたウイルスを解凍し、4℃にて10分間1500×gで遠心分離し、得られた上清をPBSで希釈してチャレンジ試験を行った。
クルマエビは、砂床のダブルボトムタンクを使用して、24±1.8°Cの脱塩素電気分解海水(33.05±0.13ppt)で飼育した。1日あたり体重の3%の市販のエビの餌(Juveniles P-2、マルハ、日本)を与えた。
PBS対照群及びGST対照群は、4日目から7日目までの生存率が劇的に低下し、チャレンジ後20日目で生存率がそれぞれ20%及び33.3%に達した。VP15(aa1-25)投与群の場合、GST群と同様の傾向を示した。VP15、VP15(aa58-80)、又はVP15(aa1-25、58-80)投与群は、20日目でそれぞれ42.1%、47.6%、及び38.9%の生存率を示した。一方、VP15(aa26-57)は、20日目で生存率が57.9%を示し、有意差を示す唯一の実験群であった(χ2テスト、p<0:05)(図4)。
実施例2 WSSV-VP15における抗原性ドメインの同定
(1)ペプチドの合成
実施例1により、VP15(aa26-57)は他のペプチド断片よりも高い抗原性を有することが分かった。そこで、VP15(aa26-57)をさらに断片化し、抗原性ドメインを同定した。

図5に示すように、VP15(aa26-57)をさらに4つのペプチド断片:KR-11(KTKSRRGSKKR、配列番号6)、SR-11(STTAGRISKRR、配列番号7)、KK-13(KRRSPSMKKRAGK、配列番号8)、及び、SK-10(SPSMKKRAGK、配列番号9)に断片化し、それぞれ商業的に合成した(GL Biochem Ltd.,中国)。
ペプチドの特性をそれぞれ高速液体クロマトグラフィー(HPLC)及びエレクトロスプレイイオン化質量分析(ESI-MS)により分析した。HPLCを、Inertsil ODS-SPカラム(純度>95%)を用いて各ペプチドの精製のために使用した。精製した合成ペプチドの純度及び分子量を、液体クロマトグラフィー-質量分析と組み合わせたエレクトロスプレイイオン化(LC-MS/ESI、Agilent-6125B)を用いて分析した。
(2)VP15(aa26-57)の各断片ペプチドのWSSV感染に対する防御効果
実施例1の(3)と同様な方法で、各ペプチド断片のクルマエビのWSSV感染に対する防御効果を確認した。実験群の、GST-VP15(aa26-57)群(n=11)、KR-11群(n=13)、SR-11群(n=11)、SK-10群(n=11)、KK-13群(n=13)のクルマエビには、それぞれの断片ペプチドを10μg/gエビ体重の量で筋肉内注射(IM)によって接種し(初回免疫)、さらに20日後に2回目の接種(追加免疫)を行い、さらに10日後、WSSV(2.69×104DNAコピー/尾)でチャレンジを行った。PBS対照群(n=10)のエビにも、実験群と同じ要領で、10μL/尾エビのPBSを接種した。
実施例1の(3)と同様な方法で、各ペプチド断片のクルマエビのWSSV感染に対する防御効果を確認した。実験群の、GST-VP15(aa26-57)群(n=11)、KR-11群(n=13)、SR-11群(n=11)、SK-10群(n=11)、KK-13群(n=13)のクルマエビには、それぞれの断片ペプチドを10μg/gエビ体重の量で筋肉内注射(IM)によって接種し(初回免疫)、さらに20日後に2回目の接種(追加免疫)を行い、さらに10日後、WSSV(2.69×104DNAコピー/尾)でチャレンジを行った。PBS対照群(n=10)のエビにも、実験群と同じ要領で、10μL/尾エビのPBSを接種した。
各群のエビを14日間観察し、生存率(%)を算出した(図6(a))。KR-11、SK-10、及びKK-13を接種した実験群のエビは、チャレンジ2週間後の生存率は約54%であり、PBS対照群の53%と同程度であった。一方、SR-11の場合、チャレンジ2週間後の生存率が約80%であり、陽性対照のGST-VP15(aa26-57)群と同程度であった。このことは、SR-11がWSSV感染に対して高い防御効果を有することを示唆しており、SR-11が有効なWSSV抗原性ペプチドであると考えられる。
(3)SR-11によるWSSV感染に対する防御効果の再現性
(2)の結果の再現性を確認するために、エビ尾数を25尾に増やして同様な実験を行った。対照群としてPBS群、実験群としてSR-11群とGST-VP15(aa26-57)群を用いた。WSSVチャレンジ後20日まで観察を行った(図6(b))。その結果、PBS群では、チャレンジ6日間後から生存率が急速に低下し、20日目には50%まで低下した。一方、SR-11群及びGST-VP15(aa26-57)群の場合、チャレンジ20日後の生存率はそれぞれ77%と73%であり、SR-11による高いWSSV感染防御効果が認められた。

実施例3 カイコにおけるVP15及びSR-11の発現
pGEX-VP15を鋳型とし、表1に記載されたプライマーセットを用いて、逆PCRによって、VP15(配列番号1)又はVP15の断片ペプチドであるSR-11(配列番号7)が、GSTの下流になり、かつ、Flagタグの上流となるように、GST及びFlagタグと融合したGST-VP15又はSR-11をコードする核酸分子を組み込んだpFastBacベクターを作製した。さらに、該pFastBacベクターを、BmNPVバクミド(特許第4288389号)をもつ大腸菌BmDH10Bacに形質転換することによって、カイコ発現用組換えBmNPVバクミドを作製した。なお、得られた組換えBmNPVバクミドをカイコ幼虫及びサナギに注射して発現させた。注射は、BmNPVバクミドDNAを0.2μgBmNPVバクミド/1μLの濃度でDMRIE-C(Thermo Fisher Scientific、日本)に混合し、得られた溶液を針26ゲージの注射器で20μL/匹でカイコ5齢幼虫又はサナギに注射した。)

バクミドを注射して5日後、カイコ幼虫又はサナギの脂肪体を回収し、TBS(1.37mol/l NaCl、26.8mmol/l KCl、250mmol/l Tris-HCl、0.1% Triton-X、pH7.6)に懸濁し、超音波破砕後、8000g×15分間遠心分離し、上清部と沈殿部とに分け、ウエスタンブロットを行った。上清部サンプル、沈殿部サンプルのそれぞれに、SDS-PAGE用Sample Buffer Solution(0.125M Tris-HCl、4(w/v)% SDSゲル、20(v/v)% Glycerol、0.01(w/v)% 2-メルカプトエタノール)を加え、100℃で10分間熱処理した。熱処理後のサンプルを3μL、SDSアクリドアミドゲル(12%)にアプライし、25mA×65分で電気泳動を行い、ウエスタンブロットに用いた。ウエスタンブロットの一次抗体にマウス抗Flagタグ抗体(1:10000、MBL、日本)、二次抗体に抗マウス抗体(1:10000)を用い、タンパク質を検出した(MBL、日本)。カイコ幼虫では、上清部に多くのVP15とVP15ペプチドの発現が確認されたが、サナギの場合、上清部と沈殿部にほぼ同量発現されたことが確認された(図7)。
実施例4 VP15、VP15(aa26-57)、及びSR-11と受容体との相互作用
VP15、VP15(aa26-57)、及びSR-11と、受容体であるMjgCqR-Hisとの相互作用をウエスタンブロットで確認した。

VP15、VP15(aa26-57)、及びSR-11は、それぞれが、GSTの下流になり、かつ、Flagタグの上流となるように、GST及びFlagタグと融合した融合タンパク質をコードする核酸分子を作製して、大腸菌にて発現させた。対照として、GSTのみを用いた。一方、受容体MjgCqR-Hisは、上述と同様な方法でカイコで発現させた。
精製したそれぞれのタンパク質又はペプチドを用いてウエスタンブロットを行った。レーン1及び2にはGST、レーン3及び4にはGST-VP15、レーン5及び6にはGST-VP15(aa26-57)、レーン7及び8にはGST-SR11をそれぞれロードし、ウエスタンブロットによって、受容体MjgCqRとの結合を調べた(図8)。(A)及び(B)は抗Hisタグ(MBL、日本)を用いMjgCqR-Hisを検出し、(C)は抗GSTタグを用いGSTを検出し、(D)は抗Flagタグを用いGST-VP15、GST-VP15(aa26-57)及びGST-SR11GSTを検出した。
その結果、VP15、VP15(aa26-57)、及びSR-11と、受容体MjgCqR-Hisとの結合が確認できた(図8(B))。一方、GSTのみでは受容体との結合が見られなかった(図8(A))。このことから、VP15、VP15(aa26-57)、及びSR-11は明確に受容体と相互作用を行っていることが確認できた。
以上より、VP15の断片ペプチドのうち、26~57番目アミノ酸残基からなるペプチド(VP15(aa26-57);配列番号3)が最も高いWSSV感染防御効果を示し、特にVP15(aa26-57)の断片であるSR-11(STTAGRISKRR、配列番号7)が最も高いWSSV感染防御効果を示すことを、初めて発見した。
実施例5 各種ワクチンの経口投与による感染防御効果の評価
(1)各種ワクチン含有飼料の調製
経口投与による感染防御効果を評価するため、表2に示す各種タンパク質/ペプチドを含有する精製液1及び粉末2~6を用いた。GSTタンパク質は、実施例1と同様にして大腸菌で発現させ、GSTアフィニティカラムで精製することによって、精製液1(陰性対照、2mgタンパク質/mL)を得た。GSTタンパク質は、実施例3と同様にしてカイコサナギで発現させ、発現後に精製せず、凍結乾燥させ、乳鉢ですりつぶして粉末2(陰性対照)を得た。各種目的の抗原性ペプチド(VP15、VP15(aa26-57)、又はSR-11)とGSTとの融合タンパク質は、同様にカイコサナギで発現させ、発現後に精製せず、凍結乾燥させ、乳鉢ですりつぶして粉末3~5を得た。SR-11は、実施例2と同様に人工合成し、粉末6を得た。得られた粉末3~5における目的の抗原性ペプチド(VP15、VP15(aa26-57)、又はSR-11)の含有量を定量し、表2に示す。
(1)各種ワクチン含有飼料の調製
経口投与による感染防御効果を評価するため、表2に示す各種タンパク質/ペプチドを含有する精製液1及び粉末2~6を用いた。GSTタンパク質は、実施例1と同様にして大腸菌で発現させ、GSTアフィニティカラムで精製することによって、精製液1(陰性対照、2mgタンパク質/mL)を得た。GSTタンパク質は、実施例3と同様にしてカイコサナギで発現させ、発現後に精製せず、凍結乾燥させ、乳鉢ですりつぶして粉末2(陰性対照)を得た。各種目的の抗原性ペプチド(VP15、VP15(aa26-57)、又はSR-11)とGSTとの融合タンパク質は、同様にカイコサナギで発現させ、発現後に精製せず、凍結乾燥させ、乳鉢ですりつぶして粉末3~5を得た。SR-11は、実施例2と同様に人工合成し、粉末6を得た。得られた粉末3~5における目的の抗原性ペプチド(VP15、VP15(aa26-57)、又はSR-11)の含有量を定量し、表2に示す。
精製液1及び粉末2~6を、表3に示す組成となるように、市販の餌(Juveniles P-2、マルハ、日本)に混合して、合計29.8gの混合餌を調製した。この混合餌に、展着剤(Shering-Plough Animal Health、USA)を0.5%添加した。その後、混合餌の6%に当たるPBS緩衝液1788μLを混合し、ワクチン含有飼料1~6を得た。ワクチン含有飼料1及び2を陰性対照とした。また、各種タンパク質/ペプチドを添加せず、同様に調製した飼料(PBS飼料)も陰性対照とした。
(2)ワクチン含有飼料の投与による感染防御効果
表4に示すように、クルマエビ(体重0.65g、n=15~25/群)を7つの群(3つの対照群と4つの実験群)に分けた。実験群の、GST-VP15群、GST-VP15(aa26-57)群、GST-SR-11群、SR-11群のクルマエビには、それぞれワクチン含有飼料3~6を1.3g/日の量で投与し、23日間飼育した。その後、通常の飼料に戻し、6日間飼育を続け、30日目にWSSV感染試験を行った。対照群のエビにも、実験群と同じ要領で、GST(大腸菌由来)群、GST(カイコサナギ由来)群及びPBS群には、それぞれワクチン含有飼料1~2、PBS飼料を投与した。
表4に示すように、クルマエビ(体重0.65g、n=15~25/群)を7つの群(3つの対照群と4つの実験群)に分けた。実験群の、GST-VP15群、GST-VP15(aa26-57)群、GST-SR-11群、SR-11群のクルマエビには、それぞれワクチン含有飼料3~6を1.3g/日の量で投与し、23日間飼育した。その後、通常の飼料に戻し、6日間飼育を続け、30日目にWSSV感染試験を行った。対照群のエビにも、実験群と同じ要領で、GST(大腸菌由来)群、GST(カイコサナギ由来)群及びPBS群には、それぞれワクチン含有飼料1~2、PBS飼料を投与した。
WSSV感染試験は、浸漬攻撃試験を採用した。WSSV感染によって死亡したエビを体重の9倍量のPBSを添加し磨砕した後、4℃にて15分間1500×gで遠心分離し、得られた上清を所定のDNAコピー数となるようにPBSで希釈し、WSSV磨砕液とした。浸漬攻撃試験では、例えば、攻撃エビ個体の総重量30g/水量(L)として、1.27×106DNAコピー/gエビとなるように、WSSV磨砕液を水量の10-2.8希釈で添加し、2時間浸漬した。浸漬攻撃が終了後、試験に供したエビをネットに取り出し、清浄な海水により3分間の流水洗浄を行い、飼育水槽に収容した。
陰性対照として用いた大腸菌又はカイコサナギ由来のGST群では、WSSV感染試験20日後の相対生存率が、それぞれ60.3%及び68.8%であった。GST-VP15群では、相対生存率は81.1%であったが、GST-VP15(aa26-57)群では相対生存率が100%であった。このことから、VP15(aa26-57)のWSSV感染に対する防御効果が高いことが明らかになった。また、GST-SR-11群の場合、相対生存率が72.2%であり、大腸菌及びカイコサナギ由来のGST群の相対生存率より高かったが、GST-VP15群の相対生存率よりは低かった。SR-11群も、90.5%と高い相対生存率を示した。GST-SR-11群の相対生存率がGST-VP15群よりも低かったが、SR-11群の相対生存率はGST-VP15群よりも高いことから、SR-11の場合、融合タンパク質に存在する形態よりもSR-11のみの方が、WSSV感染に対する防御効果が高いことが示された。GST-VP15群及びGST-VP15(aa26-57)群のいずれも相対生存率が80%以上であることから、実験群の各種タンパク質(ワクチン)は、経口投与によるエビのWSSV感染防御にも効果的であると考えられる。
実施例6 ワクチン含有飼料の投与による遺伝子発現量の変化
ワクチン含有飼料の投与によるエビの免疫系に対する影響を調べるため、各種遺伝子、すなわち、NF-κB免疫反応において中心的役割を果たす転写因子であるDorsal遺伝子及びRelish遺伝子、抗ウイルス応答におけるシグナルを誘導し、AMPの発現を向上するSTAT遺伝子、並びに、病原体の侵入を阻止するタンパク質の発現を誘導するProPO遺伝子の、投与前後の発現量の変化を評価した。
ワクチン含有飼料の投与によるエビの免疫系に対する影響を調べるため、各種遺伝子、すなわち、NF-κB免疫反応において中心的役割を果たす転写因子であるDorsal遺伝子及びRelish遺伝子、抗ウイルス応答におけるシグナルを誘導し、AMPの発現を向上するSTAT遺伝子、並びに、病原体の侵入を阻止するタンパク質の発現を誘導するProPO遺伝子の、投与前後の発現量の変化を評価した。
実施例5の(2)と同様に、ワクチン含有飼料を投与してから0、1、3及び4日後、クルマエビの鰓からトータルRNAを、NucleoSpin RNA(Macherey-Nagel、ドイツ)を用いて抽出した。抽出したトータルRNAを、ReverTra Ace qPCR RT Master Mix(東洋紡、日本)を用いて逆転写し、ファーストストランドcDNA合成した。ファーストストランドcDNAを鋳型として、表5に示す各プライマーセットを使用して定量的リアルタイムPCR(qPCR)によって、Dorsal、Relish、STAT及びProPO遺伝子のmRNAの発現量を評価した。対照としてβ-アクチンを用いた。qPCRは、Thunderbird SYBR qPCR Mix(Mx3000P、Agilent、日本)を用いて、95℃で1分間、95℃で15秒間、55℃で30秒間、72℃で1分間を40サイクル行った。その結果を図10及び11に示す。なお、各遺伝子のmRNAの相対発現量は、β-アクチン遺伝子の発現量に対する相対値である。また、β-アクチン遺伝子の発現量を「1」とした。
図10及び11から、Dorsal、Relish、STAT及びProPOの遺伝子発現はいずれも14日目に最高値を示したことが分かった。Dorsal、Relishについては、初期には、SR-11群の遺伝子発現が著しく高かったが、14日目にはGST-VP15(aa26-57)群、GST-VP15群及びGST-SR-11群の遺伝子発現が高く、いずれもカイコサナギで発現させたものであった(図10)。STATとProPOの場合も、初期にはSR-11群の遺伝子発現が高かったが、14日目にはSTATはGST-SR-11群が、ProPOはGST-VP15群及びGST-VP15(aa26-57)群の遺伝子発現が最高であった(図11)。したがって、STAT又はProPOの遺伝子発現におけるSR-11とVP15との機能が若干異なると考えられる。人工合成したSR-11の場合、初期のいずれの遺伝子発現量も高い値であったが、時間と共に低下傾向であった。一方で、カイコサナギで発現させたVP15やSR-11の場合、効果が持続的であることが確認された。

- 配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列を含む抗原性ペプチドであって、8~30個のアミノ酸残基からなる抗原性ペプチドを含む、ホワイトスポットシンドロームウイルス(WSSV)の感染を予防又は治療するためのワクチン。
- 前記抗原性ペプチドが、8~20個のアミノ酸残基からなる、請求項1に記載のワクチン。
- 前記抗原性ペプチドが、配列番号7に示すアミノ酸配列、又は、配列番号7に示すアミノ酸配列において、1~3個のアミノ酸残基の欠失、置換若しくは挿入を有するアミノ酸配列からなる、請求項1又は2に記載のワクチン。
- 前記抗原性ペプチドが、配列番号1に示すアミノ酸配列の連続した8~30個のアミノ残基を含むペプチドであるか、又は、配列番号1に示すアミノ酸配列の連続した8~30個のアミノ残基からなるペプチドと80%以上の配列同一性を有するペプチドである、請求項1に記載のワクチン。
