Abstract
본 발명은 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체 및 이의 용도에 관한 것으로, 본 발명의 목적은 hACE2에 결합하는 항체를 제조하고, 각 항체가 SARS-CoV2 바이러스의 세포 진입을 억제하는 것을 입증하여 상기 항체가 SARS-CoV2에 의한 항 바이러스 효과를 나타냄을 확인함으로써, 상기 항체를 포함하는 조성물을 SARS-CoV2에 의한 감염증의 예방 또는 치료용 약학적 조성물로 제공한다.
Description
본 발명은 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체 및 이의 용도에 관한 것이다.
코로나바이러스 (Coronavirus)는 약 27-32 kb 정도의 (+) strand RNA genome 단일 조각으로 구성된 바이러스로서 사람과 다른 포유동물들에 분포한다. 코로나바이러스는 복제와 전사과정을 거쳐서 게놈-RNA (genome-RNA)와 3' 말단에 공통의 mRNA를 가지는 서브게놈 RNA (subgenomic RNA) 6~8개를 생산한다고 알려져 있다. 대부분의 사람에서 코로나바이러스 감염은 가벼운 증상을 나타내나 감염력이 높아 지난 20여 년간 10,000명 이상의 사람에 SARS (중증호흡기증후군, 치사율 10%) 코로나바이러스 및 MERS (중동호흡기증후군, 치사율 37%) 코로나바이러스가 감염되었다.
최근에 발견된 신종 코로나바이러스 (SARS-CoV-2, Severe acute respiratory syndrome coronavirus 2) 감염증인 COVID-19는 2019년 12월 1일 중국에서 발견되어 2019년 12월 12일 최초 보고된 급성 호흡기 증후군으로, 발열과 기침, 호흡곤란, 비정형 폐렴 등의 증세를 보인다. 2020년 1월부터는 중국 국외로도 광범위하게 전파되었으며, 중국내 춘절 연휴를 전후하여 빠른 전염으로 감염자 급증, 우한시 도시 기능 전체가 마비되는 등 우려할 사태로 발전하고 있다.
SARS-CoV2는 인간 폐 상피세포에 주로 존재하는 안지오텐신 변환 효소 2(Angiotensin-converting enzyme 2; hACE2) 단백질에 결합하여 세포 내로 침입하게 되며 바이러스의 유전물질을 인간 세포 내부에서 복제시켜 바이러스를 재생산한다. 코로나 바이러스가 인간 세포로 침입하는 것을 방지하는 전략을 찾는 것이 COVID-19 감염병을 치료하는 근본적인 방법이다. 코로나 바이러스의 표면에 존재하는 스파이크(Spike) 단백질 S1-RBD(Receptor Binding Domain)은 hACE2 단백질에 결합하며, 이때 결합된 3차원 구조에 대해서는 최근 연구들에 의하여 밝혀져 있다.
SARS-CoV2와 hACE2의 결합을 차단할 수 있으면 근본적으로 코로나 바이러스의 세포 침입을 막을 수 있을 것으로 기대되고 있으며, COVID-19인 SARS-CoV2에 의한 감염증을 예방 또는 치료하기 위해 SARS-CoV2와 hACE2의 결합을 차단할 수 있는 새로운 수단에 대한 연구가 지속되고 있다.
상기와 같은 문제점을 해결하기 위해, 본 발명의 목적은 hACE2에 결합하는 항체를 제조하고, 각 항체가 SARS-CoV2 바이러스의 세포 진입을 억제하는 것을 입증하여 상기 항체가 SARS-CoV2에 의한 항 바이러스 효과를 나타냄을 확인함으로써, 상기 항체를 포함하는 조성물을 SARS-CoV2에 의한 감염증의 예방 또는 치료용 약학적 조성물로 제공하는 것이다.
본 발명은 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체를 제공한다.
또한, 본 발명은 상기 항체를 유효성분으로 포함하는 SARS-CoV2에 의한 감염증의 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명에 따르면, 본원발명의 항체들은 hACE2에 결합하여 SARS-CoV2 바이러스의 세포 진입을 억제함으로써, SARS-CoV2 바이러스에 의한 감염증을 예방 또는 치료하는 효과를 나타낼 수 있다.

상기 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸 히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다.
본 발명에 있어서, 상기 약학적 조성물에 포함되는 첨가제의 함량은 특별히 한정되는 것은 아니며 통상의 제제화에 사용되는 함량 범위 내에서 적절하게 조절될 수 있다.
상기 약학적 조성물은 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립, 정제, 크림, 젤, 패취, 분무제, 연고제, 경고제, 로션제, 리니멘트제, 파스타제 및 카타플라스마제로 이루어진 군으로부터 선택되는 하나 이상의 외용제 형태로 제형화될 수 있다.
본 발명의 약학적 조성물은 제형화를 위해 추가로 있는 약학적으로 허용가능한 담체 및 희석제를 포함할 수 있다. 상기 약학적으로 허용가능한 담체 및 희석제는 전분, 당, 및 만니톨과 같은 부형제, 칼슘 포스페이트 등과 같은 충전제 및 증량제, 카르복시메틸셀룰로오스, 히드록시프로필셀룰로오스 등과 같은 셀룰로오스 유도체, 젤라틴, 알긴산염, 및 폴리비닐 피롤리돈 등과 같은 결합제, 활석, 스테아린산 칼슘, 수소화 피마자유 및 폴리에틸렌 글리콜과 같은 윤활제, 포비돈, 크로스포비돈과 같은 붕해제, 폴리소르베이트, 세틸알코올, 및 글리세롤 등과 같은 계면활성제를 포함하나, 이에 한정되지 않는다. 상기 약학적으로 허용가능한 담체 및 희석제는 대상체에게 생물학적 및 생리학적으로 친화적인 것일 수 있다. 희석제의 예로는 염수, 수용성 완충액, 용매 및/또는 분산제(dispersion media)를 들 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여(예를 들어, 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있다. 경구 투여일 경우, 정제, 트로키제 (troches), 로젠지 (lozenge), 수용성 현탁액, 유성 현탁액, 조제 분말, 과립, 에멀젼, 하드 캡슐, 소프트 캡슐, 시럽 또는 엘릭시르제 등으로 제형화될 수 있다. 비경구 투여일 경우, 주사액, 좌제, 호흡기 흡입용 분말, 스프레이용 에어로졸제, 연고, 도포용 파우더, 오일, 크림 등으로 제형화 될 수 있다.
본 발명의 약학적 조성물의 투여량은 환자의 상태 및 체중, 연령, 성별, 건강상태, 식이 체질 특이성, 제제의 성질, 질병의 정도, 조성물의 투여시간, 투여방법, 투여기간 또는 간격, 배설율, 및 약물 형태에 따라 그 범위가 다양할 수 있으며, 이 분야 통상의 기술자에 의해 적절하게 선택될 수 있다. 예컨대, 약 0.1 내지 10,000 mg/kg의 범위일 수 있으나 이제 제한되지 않으며, 하루 일회 내지 수회에 나누어 투여될 수 있다.
상기 약학적 조성물은 목적하는 방법에 따라 경구 투여되거나 비경구 투여(예를 들면, 정맥 내, 피하 내, 복강 내 또는 국소에 적용)될 수 있다. 본 발명의 약학적 조성물의 약학적 유효량, 유효 투여량은 약학적 조성물의 제제화 방법, 투여 방식, 투여 시간 및/또는 투여 경로 등에 의해 다양해질 수 있으며, 당해 기술 분야에서 통상의 지식을 가진 자는 목적하는 치료에 효과적인 투여량을 용이하게 결정하고 처방할 수 있다. 본 발명의 약학적 조성물의 투여는 하루에 1회 투여될 수 있고, 수회에 나누어 투여될 수도 있다.
이하, 본 발명의 이해를 돕기 위하여 실시예를 들어 상세하게 설명하기로 한다. 다만 하기의 실시예는 본 발명의 내용을 예시하는 것일 뿐 본 발명의 범위가 하기 실시예에 한정되는 것은 아니다. 본 발명의 실시예는 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해 제공되는 것이다.

제조예
>
각 항체는 human antibody phage library에서 Solution panning을 통해 도출하였다. Panning 진행 하루 전에, Epoxy bead(Dynabeads M270 epoxy, Thermo #14301) 에 rhACE2(ACRO, Human ACE2 / ACEH Protein, His Tag, # AC2-H52H8)를 결합시켰다. 이후 rhACE2가 결합된 epoxy bead에 phage library 추가하여 incubation 하였고, 여러 번의 세척 및 용출을 통해 rhACE2에 결합하는 phage library pool을 도출하였다. 이후 도출된 phage library를 single clone으로 구분하여 single clone phage ELISA를 진행하였다. rhACE2를 0.2 ug/well의 농도로 96 웰에 코팅한 이후 각 single clone phage들을 각 well에 처리하였고, 2차 항체(Sino biological, 11973-MM05T-H)를 사용하여 ELISA 실험을 수행하였다. 이 실험에서 rhACE2에 결합하는 phage clone의 서열을 확인하였고, Sc-Fv-Linker-Fc form으로의 정제를 수행하였다.
제조된 항체의 아미노산 서열은 vbase2.org를 이용한 서열 분석을 통해 확인하였다. 제조된 항체는 Sc-Fv-linker-Fc form 및 Expi293 system을 이용하여 정제되었다. pFUSE vector에 상기 항체 클론들의 DNA 서열을 삽입하였고, 이렇게 제작된 pFUSE vector는 Sc-Fv-Linker-Fc의 정보를 coding하고 있다. pFUSE vector를 이용한 단백질의 발현은 Expi293™ Expression System Kit(Gibco, A14635)를 사용하여 진행되었다. 2~3x106의 density인 Expi293 세포에서 vector transfection이 진행되었고, Expifectamin 293 reagent를 이용하여 transfection 하였다. Transfection 된 세포는 4일간 shaking incubator에서 incubation 되었고 이후 단백질을 harvest하였다.
단백질의 정제는 HPLC(high-performance liquid chromatography) 기기(Akta prime, cytiva)를 이용하여 진행되었다. 정제에는 HiTrap Protein G HP antibody purification columns(cytiva, # 17040501)이 사용되었다. Harvest된 soup을 기기를 통해 칼럼에 로딩하였고, 이후 wash와 elution을 통해 항체를 정제하였다.

실시예 1. ACE2에 대한 결합능 평가
상기 제조된 항체가 ACE2에 결합하는지 여부를 평가하였다. rhACE2가 0.2 ug/well의 농도로 코팅된 96 웰에 각 항체들을 0.02ug/ml, 0.1ug/ml, 0.5ug/ml, 및 2.5ug/ml 농도로 처리하고, 2차 항체로 anti-human-Fc-HRP 항체를 사용하여 ELISA 실험을 수행하였다. 결합 정도는 450 nm 에서의 흡수 파장으로 측정하였다. BSA(Bovine Serum Albumin)는 대조군으로 사용되었다. 도 1에 나타난 바와 같이, 제조된 항체들이 모두 rhACE2에 결합하는 것으로 확인되었다. C3 및 C21의 경우 농도가 증가함에 따라 다른 항체에 비해 결합 정도가 높게 나타났다.
실시예 2. 항바이러스 효과 평가
상기 제조된 항체가 ACE2에 결합하여 SARS-CoV2에 대한 항바이러스 효과를 나타내는지 확인하기 위해, 가상의 바이러스인 SARS-2-S를 제조하고 이의 세포 진입 여부를 차단하는지 평가하였다.
가상의 바이러스인 SARS-2-S은 SARS-CoV2와 동일하게 spike 단백질을 발현하는 렌티 바이러스로 내부에 GFP 및 Luciferase 유전자를 보유하고 있어, 세포의 감염 여부를 쉽게 판별할 수 있다. SARS-2-S는 가상의 바이러스를 제조하는 Kit로 Wuhan-Hu-1 Spike-Pseudotyped Lentiviral Kit (NR-52948, BEI resources)를 사용하였다. 이 Kit는 spike protein을 포함하는 vector(NR-52514, viral entry protein w/ Spike protein), Luciferase와 GFP를 발현하게 하는 backbone vector(NR-52516, lentiviral backbone w/ Luc2; ZsGreen), 세 종류의 helper plasmid(NR-52517, NR-52518, NR-52519)로 구성되어 있다. 다섯 종류의 plasmid를 제품과 함께 제공된 protocol(Crawford KHD. Protocol and Reagents for Pseudotyping Lentiviral Particles with SARS-CoV-2 Spike Protein for Neutralization Assays. Viruses. 2020 May 6;12(5):513. doi: 10.3390/v12050513. PMID: 32384820; PMCID: PMC7291041.)에 따라 HEK293FT 세포에 transfection 진행하여 incubation하였고, 이후 가상바이러스를 수확하였다.
HEK-293T-hACE2 세포(NR-52511, BEI resources)에 상기 제조된 항체를 0 ug/ml, 0.1 ug/ml, 1 ug/ml, 또는 10 ug/ml 농도로 37℃, 5% CO2조건으로 1 시간 동안 전처리 하였다. 이후, 항체가 존재하는 조건에서 가상의 바이러스인 SARS-2-S(SARS-Related Coronavirus 2)를 Wuhan-Hu-1 Spike-Pseudotyped Lentiviral Kit (NR-52948, BEI resources)를 이용하여 감염시켰다. 세포를 48hpi에서 용해하고 제조사의 지침에 따라 Bright-Glo™ (E2610, Promega, Madison, WI, USA)로 Luciferase 활성을 측정하고, Perkinelmer EnVision microplate reader를 사용하여 Luciferase 활성의 정량화 하였다.
도 2에 나타난 바와 같이, 상기 제조된 항체의 전처리 농도가 증가할수록 SARS-2-S의 감염 정도가 감소하는 것으로 나타났다. 상기 결과는 상기 항체들이 ACE2의 결합을 통해 SARS-CoV2의 감염을 방지하는 항바이러스 효과를 나타내는 것을 입증한다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 즉, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다.

- 서열번호 1로 표시되는 아미노산 서열을 갖는 경쇄; 및 서열번호 2로 표시되는 아미노산 서열을 갖는 중쇄를 포함하는 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체.
- 서열번호 3으로 표시되는 아미노산 서열을 갖는 경쇄; 및 서열번호 4로 표시되는 아미노산 서열을 갖는 중쇄를 포함하는 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체.
- 서열번호 5로 표시되는 아미노산 서열을 갖는 경쇄; 및 서열번호 6으로 표시되는 아미노산 서열을 갖는 중쇄를 포함하는 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체.
- 서열번호 7로 표시되는 아미노산 서열을 갖는 경쇄; 및 서열번호 8로 표시되는 아미노산 서열을 갖는 중쇄를 포함하는 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체.
- 서열번호 9로 표시되는 아미노산 서열을 갖는 경쇄; 및 서열번호 10으로 표시되는 아미노산 서열을 갖는 중쇄를 포함하는 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체.
- 서열번호 11로 표시되는 아미노산 서열을 갖는 경쇄; 및 서열번호 12로 표시되는 아미노산 서열을 갖는 중쇄를 포함하는 ACE2에 결합하는 SARS-CoV2에 대한 항 바이러스 항체.
- 제1항 내지 제6항 중 어느 한 항의 항체를 유효성분으로 포함하는 SARS-CoV2에 의한 감염증의 예방 또는 치료용 약학적 조성물.
