In particular, the Nanoparticle based composition comprising deionized water, copper nanoparticles, sodium carbonate, calcium chloride, ammonium chloride, and sodium citrate. A COPPER NANOPARTICLE COMPOSITION AND METHOD OF
PRODUCING THE SAME
FIELD OF THE INVENTION
5 [0001] The present disclosure generally relates to a nanoparticle composition. The present disclosure, in particular, relates to a nanocopper composition. Specifically, the present disclosure relates to a composition that aids to prevent and control infectious diseases, preferably coronavirus or norovims. The present disclosure also relates to a method of preparing the nanoparticle composition. 0
BACKGROUND
[0002] Coronavirus disease 2019 or COVID-19 is an infectious disease which is spreading globally at an unprecedented rate. Experts are encouraging social distance and use of face masks in order to prevent further spread of the COVID-5 19 and other pathogenic diseases. Hygienic practices such as washing hands regularly will protect from many pathogenic diseases.
[0003] Typically, disinfectants utilize 60 to 95 percent of alcohol with combination of isopropanol, ethanol, and n-propanol. These alcohol-based0 sanitizers destroy cell wall of microbes or interferes with their metabolism thereby killing the microbes in the process. However, the alcohol-based hand sanitizers will kill both beneficial and non-beneficial microbes which causes various problems in humans. However, it is still not known, whether available hand- sanitizer are effective against killing viruses such as novel coronavirus. 5
[0004] Conventionally, alcohol-based hand sanitizers are used to wash hands before intake of any food products. This prevents the microbes to enter into the human body along with the food. However, alcohol will also be ingested along with the food products thereby resulting in alcohol poisoning in the human body. 0 The endocrine dismptors which are available in the alcohol is a major factor for early puberty in children. The most common types of hostile health effects for
both alcohol and non-alcohol-based hand sanitizers include ocular irritation, vomiting, conjunctivitis, oral infection, cough, abdominal pain, etc.
[0005] There are no preventive composition are available in the market that is effective against the deadly and fatal infectious diseases like coronavirus. Surprisingly, it was observed that the nanoparticles is a potential antiviral agent due to unique attributes of nanomaterials including intrinsic anti-viral property. However, there is no data or study available on any nanoparticle composition that can kill coronavirus or norovirus.
[0006] Therefore, there exists a need to provide a nanoparticle composition that requires less amount of alcohol and also aids in preventing and controlling infectious diseases, preferably coronavirus or norovirus and method of preparing the same.
OBJECT OF THE INVENTION
[0007] Some of the objects of the present disclosure, with at least one embodiment herein satisfy, are listed herein below:
[0008] It is one of the primary object of the present disclosure to provide a nanoparticle composition that aids to prevent and control infectious diseases, preferably coronavirus or norovirus. [0009] It is one of the object of the present disclosure to provide a nanoparticle composition having minimal amount of alcohol.
[0010] It is another object of the present disclosure to provide a nanoparticle composition that is environmentally benign and easily available.
[0011] It is another object of the present disclosure to explore the antimicrobial or antiviral properties activities of nanocopper, which is safe and abundantly available. [0012] It is another object of the present disclosure to provide an eco-friendly nanocopper composition, which is cost efficient.
[0013] It is another object of the present disclosure to provide safe and effective composition against air infection and harmless to human body.
[0014] It is another object of the present disclosure to provide a method of preparing a nanoparticle composition that kills coronavims or norovims.
SUMMARY OF THE INVENTION
[0015] The present disclosure discloses a nanoparticle composition comprising, by volume: deionized water in an amount having a range of 88%-98% of the total composition; metal nanoparticles in an amount having a range of 0.4%-0.6 % of the total composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm; one or more pH modifiers having an amount in the range of 0.2% - 0.6% of the total composition; one or more surfactant having an amount in the range of 0.1%-0.3% of the total composition; one or more humectant having an amount in the range of 0.2%- 0.4% of the total composition; and one or more essential oils having an amount in the range of 0.1% - 0.2% of the total composition.
[0016] The present disclosure also relates to a method of preparing a nanoparticle composition, comprising the steps of: mixing copper chloride dihydrate and sodium hydroxide in water for a time period of 55 mins -65 mins to form solution A; mixing one or more pH modifiers in deionized water to form solution B; and heating solution B for a time period of 50 mins-70 mins till the
temperature reaches 55 C-65 C; mixing one or more surfactants and humectants in deionized water for a time period of 10 hours -12 hours to form solution C; mixing essential oils in the deionized water to form solution D for a time period of 25 mins-35 mins; and mixing the solutions A-D together to form the nanoparticles composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm.

possible media applications of air, water surface, and soil. It proves to be powerful antibacterial, fungicide, vimcide that kills away wide range of microorganisms. In particular, it was found effective in killing Coronavims (Covid-19). Tea Tree Oil: The tea tree oil is one of the essential oil that is be effective against killing micro-organism.
Lemon Grass Oil: The lemongrass oil is found to be effective against Acinetobacter baumanii (A. baumanii), Aeromonas veronii (A. veronii), Enterococcus faecalis (E. faecalis), Escherichia coli (E. coli), Klebsiella pneumonia (K. pneumonia), Salmonella enterica (S. enterica) serotype typhimurium, Serratia marcesens (S. marcesens), Proteus vulgaris (P. vulgaris), Enterobacter aerogenes (E. aerogenes), Corynebacterium equii (C. equii) and Staphylococcus aureus (S. aureus).
Peppermint Oil: Peppermint oil possesses antibacterial activity in vitro and has moderate antibacterial effects against both Gram-positive and Gram-negative bacteria. Peppermint is also found to possess antiviral and fungicidal activities. [0073] The invention is further illustrated by the following example, which is provided to be exemplary of the invention and does not limit the scope of the invention. While the present invention has been described in terms of its specific embodiments, certain modifications and equivalents will be apparent to those skilled in the art and are intended to be included within the scope of the present invention.
EXAMPLES
Method of preparing a water-based composition 1: Copper chloride dihydrate and Sodium hydroxide was mixed in water for approximately 1 hour to prepare copper nanoparticles, named as Solution A.
Further, sodium carbonate, calcium chloride, and ammonium chloride is mixed in deionized water to prepare a solution B and heating heat it till 60°C for approximately 1 hour. Sodium citrate, glycerin, and coco glucoside are mixed in deionized water to form solution C for 12 hours. Further, tea tree oil, lemon grass oil, and peppermint oil are mixed in deionized water to form solution D for 30 mins. Then, final solution was prepared by mixing all the solutions (A, B, C, & D) for 24 hours to prepare a disinfectant composition.
Method of preparing a water-based composition 2:
Copper chloride dihydrate, propanol, and Sodium hydroxide were mixed in water for approximately 1 hour to form copper nanoparticles, named as Solution A. Further, sodium carbonate, calcium chloride, and ammonium chloride were mixed in deionized water to form solution B and heating it till 60°C for approximately 1 hour. Sodium citrate, glycerin, and coco glucoside in deionized water were mixed to form solution C for 12 hours. Further, tea tree oil, lemon grass oil, and
peppermint oil were mixed in deionized water to form solution D for 30 mins. Then, final solution was prepared by mixing all the solutions (A, B, C, & D) for 24 hours to prepare a disinfectant composition.
Antimicrobial activity of the composition Ir
The analysis of the antimicrobial activity was carried out for composition 1, under dirty conditions (bovine serum albumin 3 g/L and erythrocytes 3mL/L), at 100% concentration (pure), and during 30 seconds of exposure. The composition 1 5 shows virucidal activity against the mandatory viruses, Adenovirus type 5 and Murine Norovirus. The activity is evaluated according to NF EN 16777:2018 guideline. Analysis is based on the method ASTM E2315 (Assessment of Antimicrobial Activity Using a Time-Kill Procedure).
10 The composition 1, batch not indicated, under dirty conditions, pure (100%) and diluted at 50%, and during 30 seconds of exposure, shows virucidal activity against Coronavirus 229E, with a mean reduction of 5.29 ± 0.40 TCID50, for both concentrations and two replicates, compared with the virus control after 30 seconds of exposure (T* - mean = 5.79 ± 0.40 TCID50) when the activity is 15 assayed according with the NF EN 16777: 2018 guideline.
Table 4: Effect of composition 1 on spraying it on normal mask
The composition- 1 was sprayed on the normal mask, no growth of bacteria colony occurred for around 6 hours. Please refer to figure no. 4 which shows that no growth colony was observed in comparison with the normal ask. Further, in case of spraying only propanol on normal mask the bacteria colonies increased exponentially. 5 Furthermore, the anti-allergic and irritation test was conducted as per the guidelines of Drugs and Cosmetics Act, 1940 and the said tests came out negative.
Antimicrobial activity of the composition 2:-
The analysis of the antimicrobial activity was carried out for composition 2, under dirty conditions (bovine serum albumin 3 g/L and erythrocytes 3mL/L), at 100% concentration (pure), and during 30 seconds of exposure.
15 Transmission electron microscope
Transmission electron microscopy (TEM) has been carried out to find out the size of CuO-NPs (Fig. 5) formed by the process of the present application. The TEM research was carried out on a 200 kV operating HITACHI H-800. Most of the CuO-NPs were sphere-shaped and suspended well in the solution within the size range of 50-100 nm and are shown in (Fig. 6). The standard particle size was around 75 nm. 90% of particles were under 100 nm of size.
SPECIFIC EMBODIMENTS OF THE INVENTION [0074] The present disclosure relates to a nanoparticle composition comprising, by volume: deionized water in an amount having a range of 88%-98% of the total composition; metal nanoparticles in an amount having a range of 0.4%-0.6 % of the total composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm; one or more pH modifiers having an amount in the range of 0.2% - 0.6% of the total composition; one or more surfactant having an amount in the range of 0.1%-0.3% of the total composition; one or more humectant having an amount in the range of 0.2%- 0.4% of the total composition; and one or more essential oils having an amount in the range of 0.1% - 0.2% of the total composition.

c) heating solution B for a time period of 50 mins-70 mins till the temperature reaches 55 C-65 C; d) mixing one or more surfactants and humectants in deionized water for a time period of 10 hours -12 hours to form solution C; e) mixing essential oils in the deionized water to form solution D for a time period of 25 mins-35 mins; and f) mixing the solutions A-D together to form the nanoparticle composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm.

INDUSTRIAL APPLICABILITY
The nanoparticle composition of the present disclosure kills coronavirus (COVID- 19) or norovirus. It has anti-fungal and anti-microbial activities. The nanoparticle composition can be used in the composition that are selected from the group consisting of paints, disinfectant, hand sanitizer, surface cleaner, or a coating formulation. It is environmentally benign and easily available. It is a herbal composition which is safe and abundantly available and is cost efficient.
Claims
1. A nanoparticle composition comprising, by volume: deionized water in an amount having a range of 88%-98% of the total composition; metal nanoparticles in an amount having a range of 0.4%-0.6 % of the total composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm; one or more pH modifiers having an amount in the range of 0.2% - 0.6% of the total composition; one or more surfactant having an amount in the range of 0.1%-0.3% of the total composition; one or more humectant having an amount in the range of 0.2%- 0.4% of the total composition; and one or more essential oils having an amount in the range of 0.1% - 0.2% of the total composition.
2. The nanoparticle composition as claimed in claim 1, wherein the one or more pH modifiers are selected from the group consisting of sodium carbonate, calcium chloride, ammonium chloride, sodium citrate, or a combination thereof.
3. The nanoparticle composition as claimed in claim 1, wherein the surfactant is coco-glucoside and wherein the humectant is glycerin.
4. The nanoparticle composition as claimed in claim 1, wherein the one or more essential oils are selected from the group of lemon grass oil, peppermint oil, tea tree oil, or a combination thereof.
5. The nanoparticle composition as claimed in claim 1, further comprising an alcohol in an amount of 0-12 vol. % of the total composition, wherein the alcohol is propanol.
6. The nanoparticle composition as claimed in claim 1, wherein the nanoparticle composition is used against a microorganism selected from the group consisting of a bacterium, a fungus, a mold, and a virus.
7. The nanoparticle composition as claimed in claim 6, wherein the vims is coronavims (COVID-19) or norovims.
8. The nanoparticle composition as claimed in claim 1, wherein the nanoparticle composition is used in the composition that are selected from the group consisting of paints, disinfectant, hand sanitizer, surface cleaner, or a coating formulation.
9. The nanoparticle composition as claimed in claim 1, wherein the nanoparticle composition is in the form of gel, solution, lotion, foam or cream.
10. The nanoparticle composition as claimed in claim 8, wherein the hand sanitizer composition has pH in the range of 6 to 8.
11. A method of preparing a nanoparticle composition, comprising the steps of: a) mixing copper chloride dihydrate and sodium hydroxide in water for a time period of 55 mins-65 mins to form solution A; b) mixing one or more pH modifiers in deionized water to form solution B; and c) heating solution B for a time period of 50 mins-70 mins till the temperature reaches 55 C-65 C; d) mixing one or more surfactants and humectants in deionized water for a time period of 10 hours -12 hours to form solution C;
e) mixing essential oils in the deionized water to form solution D for a time period of 25 mins-35 mins; and f) mixing the solutions A-D together to form the nanoparticle composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm.
12. The method as claimed in claim 11, wherein the components of the nanoparticle composition comprising, by volume: deionized water in an amount having a range of 88%-98% of the total composition; metal nanoparticles in an amount having a range of 0.4%-0.6 % of the total composition, wherein the metal nanoparticles are copper nanoparticles, and wherein the particle size of the nanoparticles is within the range of 50 nm-100 nm; one or more pH modifiers having an amount in the range of 0.2% - 0.6% of the total composition; one or more surfactant having an amount in the range of 0.1%-0.3% of the total composition; one or more humectant having an amount in the range of 0.2% - 0.4% of the total composition; and one or more essential oils having an amount in the range of 0.1% - 0.2% of the total composition.
13. The method as claimed in claim 12, wherein the one or more pH modifiers are selected from the group consisting of sodium carbonate, calcium chloride, ammonium chloride, sodium citrate, or a combination thereof.
14. The method as claimed in claim 11, wherein the surfactant is coco-glucoside and wherein the humectant is glycerin.
15. The method as claimed in claim 11, wherein the one or more essential oils are selected from the group of lemon grass oil, peppermint oil, tea tree oil, or a combination thereof.
16. The method as claimed in claim 11, wherein mixing in step f) is carried out for a time period of 22 hours-25 hours to form the nanoparticle composition.
17. The method as claimed in claim 11, wherein the nanoparticle composition further comprises propanol in the amount of 0-12 vol.%
18. The method as claimed in claim 11, wherein the solution A is a solution of copper nanoparticles that forms from a chemical precipitation of the copper chloride dihydrate and the sodium hydroxide.
