본 발명은 나라트립탄 또는 그의 약학적으로 허용가능한 염을 포함하는 구강용해 필름 제형에 관한 것으로, 구체적으로, 나라트립탄 또는 그의 약학적으로 허용가능한 염 및 완충제를 포함하는 것을 특징으로 하는 구강용해 필름 제형에 관한 것이다. [대표도] 도 1
Description
본 발명은 나라트립탄 또는 그의 약학적으로 허용가능한 염을 포함하는 구강용해 필름 제형에 관한 것으로, 구체적으로, 나라트립탄 또는 그의 약학적으로 허용가능한 염 및 완충제를 포함하는 것을 특징으로 하는 구강용해 필름 제형에 관한 것이다.
편두통은 뇌, 뇌신경 및 뇌혈관의 기능 이상으로 인하여 발생하는 두통의 일종으로, 어느 연령에서나 발생하지만 10-20대에 처음으로 발생하여 40-50대가 가장 흔하며, 우리나라의 경우 여성의 9%, 남성의 3%가 앓고 있는 매우 흔한 질환이다.
편두통이 발생하는 기전은 아직 명확하게 밝혀지지는 않았으나, 세로토닌, 도파민, 글루타메이트와 같은 신경전달물질이 중요한 역할을 하는 것으로 생각되고 있다. 따라서, 세로토닌 수용체에 특이적으로 작용하는 트립탄 계열의 화합물이 주목받고 있다.
트립탄계 화합물은 선택적으로 두개혈관의 세로토닌 5-HT1B 및 5-HT1D 수용체에 결합하여 뇌혈관을 수축시키는 작용을 한다. 후속적으로 선염증성(pro-inflammatory) 신경펩티드 방출을 억제하는 작용을 하기도 한다.
통상의 트립탄계 화합물은 경구용 정제로서, 물과 함께 복용되는데, 환자나 어린이 등이 약물을 삼키기 어려운 문제가 있다. 이러한 문제점을 극복하기 위해서 최근에는 물 없이도 구강에서 신속하게 붕해되어 삼킬 수 있는 구강속붕해정(Orally Disintegrating Tablet, ODT)이 개발되었다 (한국등록특허 제10-1626873호, 한국공개특허 제10-2001-0107754호). 하지만, 구강속붕해정의 경우 복용은 정제보다 간편하지만 경도가 낮아 쉽게 깨지며, 부형제로 인한 부피 증가로 휴대가 용이하지 않다. 또한 다량의 부형제 사용은 구강 내에서 이물감을 줄 수 있다.
이러한 구강속붕해정의 단점을 보완하여 최근에는 부피와 부형제의 용량을 줄인 필름형 제제가 개발되고 있다. 구강용해 필름(Orally Disintegrating Film, ODF)은 두께가 얇고 유연한 성상으로 휴대가 용이하며 쉽게 깨지지 않는다. 또한, 필름형으로 표면적이 넓어 붕해시간이 짧으므로 구강 내에서의 이물감과 약물의 쓴맛을 차폐한다.
그러나, 구강용해 필름의 경우, 약물, 필름형성기제 및 기타 첨가물의 혼합물이 일정한 점도를 유지해야 하며, 점도를 유지하기 위해서는 물 또는 유기 용매의 양이 제한적이므로, 일부 난용성 약물이나 불수용성 약물은 구강용해 필름에 적합하지 않다. 특히, 트립탄과 같은 유효성분은 온도 및 습도에 불안정하여 구강용해 필름 제형으로 제조하기 어렵다.
이에 본 발명자들은 트립탄과 같은 유효성분이 안정하게 유지되고, 구강용해시 유효성분이 신속하게 용출되어 약효를 발휘할 수 있고, 필름 제형의 보관 중에서도 석출 등의 부작용이 발생하지 않는 우수한 필름 제형을 개발하여 본 발명을 완성하였다.

본 발명은 일반적인 경구투여용 제형과 비교하여 물 없이도 복용 가능하여 복약의 편의성을 높인 구강용해 필름 제형에 관한 것으로, 더욱이 기존에 알려진 구강용해 제형과 비교하여 안정성 및 용출율 등의 제제학적 특성이 개선된 구강용해 필름 제형을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여, 본 발명은 유효성분으로 나라트립탄, 수마트립탄, 엘레트립탄, 프로바트립탄, 알모트립탄, 졸미트립탄, 리자트립탄, 또는 이의 염, 및 완충제를 포함하는 구강용해 필름 제형을 제공한다.
일 실시 태양에서, 상기 유효성분은 나라트립탄인 것을 특징으로 하는 구강용해 필름 제형일 수 있다.
일 실시 태양에서, 상기 제형은 용출시험시 상기 유효성분이 5분 내에 85% 이상, 더 바람직하게는 90% 이상, 가장 바람직하게는 95% 이상 용출되는 것을 특징으로 하는 구강용해 필름 제형일 수 있다.
상기 완충제는 시트르산, 시트르산나트륨, 시트르산 칼륨, 아세트산, 아세트산나트륨, 말산, 글루탐산나트륨, 글라이신, 탄산나트륨, 말레산, 젖산나트륨, 인산이수소나트륨, 인산일수소나트륨, 인산이수소칼륨, 인산일수소칼륨, 또는 이의 혼합물일 수 있으나, 이에 제한되지 않는다.
일 실시 태양에서, 상기 완충제는 시트르산 및 시트르산 나트륨의 혼합물을 포함하고, 필름 고형분의 총 중량에 대하여 상기 완충제 0.1 내지 2.0 중량%를 포함할 수 있다.
일 실시 태양에서, 상기 완충제로 필름 고형분의 총 중량에 대하여 시트르산나트륨 0.1 내지 2.0 중량% 및 시트르산은 0.01 내지 0.1 중량%를 포함할 수 있다.
상기 "필름 고형분 총 중량"은 상기 필름 제형에서 용제를 제외한 계면활성제, 결합제, 가소제, 붕해제, 감미제, 착향제, 착색제 등의 고형성분 중량의 총 합을 말한다.
본 발명은 가속안정성 시험시 6개월후 총 유연물질의 함량이 3% 미만, 더욱 바람직하게는 2% 미만, 가장 바람직하게는 1% 미만인 구강용해 필름 제형을 제공한다.
일 실시 태양에서, 상기 제형은 필름형성제를 추가로 포함할 수 있다. 상기 필름형성제는 플루란, 알긴산나트륨, 하이드록시프로필 셀룰로스, 하이드록시프로필메틸셀룰로스 또는 이의 혼합물일 수 있으며, 바람직하게는 하이드록시프로필메틸셀룰로스일 수 있으나, 이에 제한되지 않는다.
또한, 본 발명은 (A) 용매에 완충제를 넣어 혼합액의 pH를 pH5.4 내지 pH7.4로 조정하는 단계; (B) 상기 pH가 조정된 혼합액에 유효성분, 필름형성제 및 첨가제를 넣고 교반하여 최종 혼합액을 제조하는 단계; 및 (C) 상기 최종 혼합액에 열을 가해 건조하여 박막형태의 필름을 제조하는 단계를 포함하는, 나라트립탄, 수마트립탄, 엘레트립탄, 프로바트립탄, 알모트립탄, 졸미트립탄, 리자트립탄 또는 이의 염을 유효성분으로 포함하는 구강용해 필름 제형의 제조 방법을 제공한다.
일 실시 태양에서, 상기 용매는 물, C1-C6 알코올 및 이의 혼합용매일 수있으며, 바람직하게 30% 내지 90%의 에탄올 수용액일 수 있고, 더욱 바람직하게는 50 내지 80% 에탄올 수용액일 수 있으나, 이에 제한되지 않는다.
본 발명의 구강용해 필름 제형은 편두통의 치료 또는 예방에 사용될 수 있다.
본 발명의 구강용해 필름 제형은 토피라메이트를 유효성분으로 포함하는 다른 제형과 동시에 또는 시간 간격을 두고 투여될 수 있다.
또한, 2.5mg의 나라트립탄을 유효성분으로 포함하는 본 발명의 구강용해 필름을 비글견에 투여시, AUC가 150 ng·hr/mL 내지 250 ng·hr/mL이고, Cmax가 30 ng/mL내지 60 ng/mL일 수 있다.
본 발명의 나라트립탄을 포함하는 구강용해 필름 제형은 구강용해시 신속하게 용해되어, 신속한 약효 발현을 가능하게 하며, 장기간(6개월 이상)의 가속시험에도 안정성을 유지한다. 따라서, 편두통을 갖는 환자가 물 없이도 복용이 가능하여 복용의 편의성을 증진시키고, 증상의 발생시 즉시 복용할 수 있도록 휴대가 간편하여, 환자들에게 유용하게 사용될 수 있다.

상기 유효성분으로 나라트립탄, 수마트립탄, 엘레트립탄, 프로바트립탄, 알모트립탄, 졸미트립탄, 리자트립탄은 트립탄계 화합물이며, 트립탄계 화합물 그 자체뿐만 아니라 트립탄의 용매화물 또는 수화물 모두 본 발명의 범위에 속한다. 트립탄계 화합물은 세레토닌에 선택적인 아고니스트로서, 5-HT1B1D에 아고니스트로서 사용될 수 있는데, 편두통, 급발성 편두통 또는 군발성 편두통에 사용되고, 약물의 복용 용량이 적어 필름 제형으로 적합한 약물이 될 수 있다.
상기 "염"은 환자에게 비독성이고 무해하며, 상기 염에 기인한 부작용이 본 발명의 화합물의 이로운 효능을 저하시키지 않는 임의의 산부가염 또는 염기부가염일 수 있다. 적합한 염을 형성하는 무기산은 염산, 인산, 황산, 질산, 주석산, 브롬화수소산, 요오드화수소산, 아질산, 또는 아인산 등이 있고, 적합한 염을 형성하는 유기산은 글리콜산, 락트산, 피루브산, 말론산, 숙신산, 푸마르산, 말산, 타르타르산, 시트르산, 아스코르브산, 말레산, 벤조산, 페닐아세트산, 신남산, 살리실산, 니코틴산, 토실산, 캄포술폰산, 나프토산, 아세트산, 트리플루오로아세트산, 옥살산, 만데르산, 프로피온산, 구연산, 젖산, 글루콘산, 갈락투론산, 글루탐산, 글루타르산, 글루쿠론산, 아스파르트산, 카본산, 바닐릭산, 벤젠설폰산, p-톨루엔설폰산, 또는 메탄설폰산 등이 있으나, 이에 제한되는 것은 아니다.
상기 "구강용해 필름 (orally dissolving film, ODF)"은 구강붕해 필름 (oral disintegrating film) 또는 스트립 (strip)이라고도 불리며, 구강(oral cavity) 내에서 용해, 분산 등 붕해시켜 복용할 수 있는 필름 제형을 말한다. 이러한 필름 제형은 혀 위에 놓아 녹이게 되나, 입천장, 설하, 입안 안뜰 (buccal) 등에 부착하여 투여될 수 있다. 본 발명에 따른 필름 제형은 물 없이 복용 가능한 장점을 갖는다.
상기 "약동학적 파라미터(pharmacokinetics parameter)"는 약물의 흡수, 분포, 대사, 배설을 시간에 따른 체내 약물 농도의 변화를 통해 판별하는 기준을 말한다. 이는 투여한 약물의 혈중농도와 시간의 관계를 나타낸 그래프로, 그려진 곡선과 가로축에 둘러싸인 부분의 면적을 의미하는 곡선하면적(area under the curve, AUC) 및 약물을 투여한 후와 두 번째 투여 전에 신체의 특정 구획이나 시험 구역에서 약물이 달성하는 최대 농도 또는 최대치를 의미하는 최고혈장농도(maximum plasma concentration, Cmax)를 포함한다.
상기 "완충제"는 pH 조절제로서, 산, 염기, 염류 등을 포함하며, 예를 들어 시트르산, 시트르산나트륨, 시트르산 칼륨, 아세트산, 아세트산나트륨, 말산, 글루탐산나트륨, 글라이신, 탄산나트륨, 말레산, 젖산나트륨, 인산이수소나트륨, 인산일수소나트륨, 인산이수소칼륨, 인산일수소칼륨, 또는 이의 혼합물 등을 포함하나, 이에 제한되는 것은 아니다. 본 발명의 바람직한 완충제는 시트르산, 시트르산나트륨 또는 이의 혼합물일 수 있다.

이하 실시예를 통하여 본 발명을 더욱 상세하게 설명하고자 하나, 하기의 실시예는 단지 설명의 목적을 위한 것이며 본원 발명의 범위를 한정하고자 하는 것은 아니다.
[실시예 1 내지 6]
실시예 필름 제형의 제조
하기 표 1의 조성으로 실시예 1 내지 6의 필름 제형을 다음과 같이 제조하였다.
조제용기에 정제수 및 완충제를 넣고 주성분인 나라트립탄염산염을 추가한 후 교반하여 용액의 pH를 5.4에서 7.4 범위로 조정하였다. 조제용기에 나머지 첨가제들을 투입하여 균질한 액이 될 때까지 교반하고, 이를 필름제조액으로 사용하였다.
상기 제조된 필름제조액을 OHP 필름에 붓고, 필름 어플리케이터로 캐스팅한 뒤, 70℃ 이상의 온도에서 건조시키고, OHP필름과 분리시켜 나라트립탄염산염이 함유된 필름을 얻었다. 필름을 1매 크기로 재단 후, 알루미늄 포장지로 포장하였다.

상기 실시예 1 내지 6과 동일한 공정을 통하여 하기 표 3의 조성으로 비교예 6 내지 8의 필름 제형을 제조하였다.

[실험예 1]
나라트립탄 필름 4액의 용출 시험
실시예 1의 필름 제형으로 하기의 조건 및 방법으로 용출 시험을 수행하였다. 구체적인 용출률의 측정방법과 조건은 대한민국 식품의약품안전처에서 인정한 시험방법에 따라 하기와 같이 설정하였다.
(1) 검액의 조제
실시예 1에서 제조한 필름 1매 및 시험액으로 pH1.2, pH4.0, pH6.8, DW 900mL를 사용하여 대한약전 제11개정 용출시험법 제4법에 따라 37.0 ± 0.5℃에서 매분 50 회전으로 디스크를 사용하여 시험하였다.
용출시험 시작 5분, 10분, 15분, 30분 후에 용출액 6mL를 각각 취하여 0.45μm 멤브레인 필터로 여과한 액을 검액으로 하였다.
(2) 조작조건
검출기: 고성능 액체 크로마토그래피 (측정파장: 224nm)
칼럼 : C18 (4.6mm x 15cm, 5㎛)
이동상: 이소프로필 알코올: 용액A* (1:9)
*용액A: 0.6mL의 인산을 물 900mL에 희석후, 이 액에 트리에틸아민을 넣어 pH2.5로 조정한다.
유량: 1.0mL/min
상기 용출시험 결과를 하기 표 4 및 도 1에 나타내었다.

상기 표 4 및 도 1에 나타낸 바와 같이, 5분 이내에 나라트립탄 95% 이상이 용출되는 것을 확인하였다. 구강용해필름은 신속하게 방출되는 것이 중요한데, 실시예 1의 제형은 모든 pH에서 상기 조건을 만족하는 우수한 효과를 갖는 것을 알 수 있다.
[실험예 2]
안정성 비교 실험 1
대조약으로 나라믹정 및 실시예 1 및 2의 필름 제형으로 하기의 방법으로 안정성 실험을 수행하였다.
가속챔버(온도 40℃, 습도 75%, 알루미늄 4중지 포장)에 2개월 동안 보관한 샘플에서 발생된 유연물질을 측정하였고 그 결과를 하기 표 5 및 도 2에 나타내었다. 조작 조건은 다음과 같다.
검출기: 고성능 액체 크로마토그래피 (측정파장: 225nm)
칼럼 : 페닐그룹 (4.6mm x 15cm, 5㎛)
이동상:
**용액A: monobasic ammosium phosphate 5.75g을 물에 녹여 1L로 하고, 인산을 넣어 pH3.00±0.05로 조정한다.
유량: 1.3mL/min

상기 표 5 및 도 2에 나타낸 바와 같이, 대조약은 초기 유연물질의 양이 0.31%이고, 실시예 1의 필름은 0.07%로 대조약 대비 4배 더 적은 유연물질의 양을 나타내었다. 또한, 2개월 가속 실험 후에도 실시예 1 및 실시예 2 필름은 0.2%로 대조약 대비 더 적은 양의 유연물질을 나타내었다. 결국, 실시예 1 및 실시예 2의 필름 제형이 월등하게 우수한 안정성을 갖는 것을 알 수 있다.
[실험예 4]
안정성 비교 실험 2
비교예 1의 필름 제형과 실시예 1의 필름 제형에 대한 안정성 시험을 하기와 같이 수행하였다.
가속챔버(온도 40℃, 습도 75%, 알루미늄 4중지 포장)에 1개월 동안 보관한 샘플에서 발생된 유연물질을 측정하였고 그 결과를 하기 표 6 및 도 3에 나타내었다. 조작 조건은 실험예 3과 같다.

상기 표 6 및 도 4에 나타낸 바와 같이, 비교예 1의 필름은 초기 유연물질의 양이 0.07%이고, 실시예 1의 필름은 0.30%로 비교예 1의 필름과 비교하여 4배 더 적은 유연물질의 양을 나타내었다. 또한, 1개월 가속 실험 후, 비교예 1의 필름은 0.57이고, 실시예 1의 필름은 0.12%로 비교예 1의 필름과 비교하여 4-5배 더 적은 유연물질의 양을 나타내었다.
결국, 완충제 시스템을 갖고 있는 실시예 1의 필름 제형이 월등하게 우수한 안정성을 갖는 것을 알 수 있다.
[실험예 5]
안정성 비교 실험 3
비교예 2의 필름 (산성), 비교예 3의 필름 (염기성), 및 실시예 1의 필름 (시트르산 완충제)을 대상으로 6개월 가속 시험을 수행하였다.
그 결과를 하기 표 7에 나타내었다.

상기 표 7에 나타낸 바와 같이, 실시예 1의 필름은 비교예 2 및 3의 필름과 비교하여 유연물질의 양이 매우 작으며 월등하게 우수한 안정성을 갖는 것을 확인하였다. 특히, 가속 6개월째, 실시예 1의 필름은 유연물질의 양이 0.39%이나, 비교예 2 및 3의 필름은 각각 유연물질의 양이 3.58% 및 4.24%로 실시예 1의 필름과 비교하여 10배정도 많은 것을 알 수 있다.
결국, 중성의 완충제를 사용하는 실시예 1의 필름이 우수한 안정성을 갖는 것을 확인하였다.
[실험예 6]
안정성 비교 실험 4
상기 비교예 4 (인산이수소나트륨(NaH2PO4) 및 인산일수소나트륨(Na2HPO4)) 및 비교예 5 (인산이수소칼륨(KH2PO4) 및 인산일수소칼륨(K2HPO4)) 필름, 및 실시예 2 (시트르산 완충제) 필름을 대상으로 가속 시험을 수행하였다.
그 결과, 도 4 및 도 5에 나타낸 바와 같이, 실시예 1의 필름이 비교예 4 및 5의 필름과 비교하여 우수한 안정성을 나타내는 것을 확인하였다.
[실험예 7]
완충제 농도에 따른 성상 비교
실시예 1의 필름 제형을, 완충제의 농도 0.5, 1, 2, 3 및 3.6배로 제조한 후, 각각 80도 및 90도에서 12분간 건조시 필름의 형상을 관찰하였다 (1배 = 0.05M). 그 결과를 하기 표 8, 9 및 도 6, 7에 나타내었다.

상기 표에서 나타낸 바와 같이, 완충제 농도가 2배 이상부터 결정이 석출되는 것을 확인하였다.
추가로, 실시예 1의 필름 제형을, 완충제의 농도 1.25, 1.5 및 1.75배로 제조한 후, 각각 80도 및 90도에서 12분간 건조시 필름의 형상을 관찰하였다.
그 결과를 하기 표 10, 11 및 도 8, 9에 나타내었다.

상기 표에 나타낸 바와 같이, 완충제 농도가 1.25배까지는 결정이 석출되지 않는 것을 확인였다.
[실험예 8]
완충제 농도에 따른 안정성 비교 실험
실시예 1의 필름 제형을, 완충제의 농도 0.75, 1 및 1.25배로 제조한 후, 안정성 시험을 수행하였다.
그 결과, 도 10에 나타낸 바와 같이, 농도별로 안정성의 차이가 없는 것을 확인하여, 완충제 농도가 1.25배까지는 안정성이 우수한 것을 알 수 있다.
[실험예 9]
나라트립탄 필름에 대한 동물실험
대조약으로 나라믹정, 병용투여약으로 토파맥스정, 실시예 1의 필름 제형으로 하기의 방법으로 동물실험을 수행하였다.
G1은 비글견에게 대조약(나라믹정)을 1정 투여하였고, G2는 비글견에게 실시예 1의 필름을 1매 투여하였으며, G3는 나라트립탄과 토피라메이트 약물의 상호작용을 확인하기 위하여 비글견에게 실시예1의 필름 1매와, 토파맥스정을 1정 투여하였다.
비글견에게 투여개시 후 0, 15분, 30분, 1시간, 2시간, 3시간, 4시간, 5시간, 6시간, 8시간, 12시간, 24시간에 혈액을 채취하여 약물의 혈장 농도를 측정하였다.
약물의 혈장농도를 측정하고 약물동태학적 파라미터를 산출하여 그 결과를 하기 표 12에 나타내었다.

상기 표 12에 나타낸 바와 같이, G1군과 G2군의 AUC, Tmax, 및 Cmax는 동등한 것으로 평가되었다. 따라서, 대조약과 실시예 1의 두 제형의 약물효과 발현시간, 최대 농도, 및 생체이용률은 동등한 것을 알 수 있다. 또한, G1군(실시예 1)과 G3군(실시예1+병용투여약)의 경우에도 AUC, Tmax, 및 Cmax는 동등한 것으로 평가되었다. 구강용해 필름의 병용투여시 단독투여시와 동일하게 신뢰구간 80%~125% 내에서 적합하여 동등한것으로 평가되며 나라트립탄과 토피라메이트 약물의 상호작용이 없는 것을 확인하였다.
Claims (17)
- a) 유효성분으로 나라트립탄, 수마트립탄, 엘레트립탄, 프로바트립탄, 알모트립탄, 졸미트립탄, 리자트립탄, 또는 이의 염; 및b) 완충제를 포함하는 구강용해 필름 제형.
- 제1항에 있어서, 상기 유효성분은 나라트립탄인 것을 특징으로 하는 구강용해 필름 제형.
- 제1항에 있어서, 용출시험시 상기 유효성분이 5분 내에 85% 이상 용출되는 것을 특징으로 하는 구강용해 필름 제형.
- 제3항에 있어서, 용출시험시 상기 유효성분이 5분 내에 90% 이상 용출되는 것을 특징으로 하는 구강용해 필름 제형.
- 제1항에 있어서, 상기 완충제는 시트르산, 시트르산나트륨, 시트르산 칼륨, 아세트산, 아세트산나트륨, 말산, 글루탐산나트륨, 글라이신, 탄산나트륨, 말레산, 젖산나트륨, 인산이수소나트륨, 인산일수소나트륨, 인산이수소칼륨, 인산일수소칼륨, 또는 이의 혼합물인 것을 특징으로 하는 구강용해 필름 제형.
- 제5항에 있어서, 상기 완충제는 시트르산 및 시트르산 나트륨의 혼합물을포함하고, 필름 고형분의 총 중량에 대하여 상기 완충제 0.1 내지 2.0 중량%를 포함하는 것을 특징으로 하는 구강용해 필름 제형.
- 제6항에 있어서, 완충제로 필름 고형분의 총 중량에 대하여 시트르산나트륨 0.1 내지 2.0 중량% 및 시트르산은 0.01 내지 0.1 중량%를 포함하는 것을 특징으로 하는 구강용해 필름 제형
- 제1항에 있어서, 가속안정성 시험시 6개월후 총 유연물질의 함량이 3% 미만인 것을 특징으로 하는 구강용해 필름 제형.
- 제8항에 있어서, 가속안정성 시험시 6개월후 총 유연물질의 함량이 2% 미만인 것을 특징으로 하는 구강용해 필름 제형.
- 제1항에 있어서, 상기 제형은 필름형성제를 추가로 포함하는 것을 특징으로 하는 구강용해 필름 제형.
- 제10항에 있어서, 상기 필름형성제는 플루란, 알긴산나트륨, 하이드록시프로필 셀룰로스, 하이드록시프로필메틸셀룰로스 또는 이의 혼합물인 것을 특징으로 하는 구강용해 필름 제형.
- 제11항에 있어서, 상기 필름형성제는 하이드록시프로필메틸셀룰로스인 것을 특징으로 하는 구강용해 필름 제형.
- 하기 단계를 포함하는 나라트립탄, 수마트립탄, 엘레트립탄, 프로바트립탄, 알모트립탄, 졸미트립탄, 리자트립탄 또는 이의 염을 유효성분으로 포함하는 구강용해 필름 제형의 제조 방법:(A) 용매에 완충제를 넣어 혼합액의 pH를 pH 5.4 내지 pH 7.4로 조정하는 단계;(B) 상기 pH가 조정된 혼합액에 유효성분, 필름형성제 및 첨가제를 넣고 교반하여 최종 혼합액을 제조하는 단계; 및(C) 상기 최종 혼합액에 열을 가해 건조하여 박막형태의 필름을 제조하는단계.
- 제13항에 있어서, 상기 용매는 물, C1-C6 알코올 및 이의 혼합용매인 것을 특징으로 하는 구강용해 필름 제형의 제조 방법.
- 제1항에 있어서, 상기 구강용해 필름 제형은 편두통의 치료 또는 예방을 특징으로 하는, 구강용해 필름 제형.
- 제1항에 있어서, 토피라메이트를 유효성분으로 포함하는 다른 제형과 동시에 또는 시간 간격을 두고 투여될 수 있는 것을 특징으로 하는, 구강용해 필름 제형.
- 제2항에 있어서, 상기 2.5mg의 나라트립탄을 유효성분으로 포함하는 구강용해 필름을 비글견에 투여시, AUC가 150 ng·hr/mL 내지 250 ng·hr/mL이고, Cmax가 30 ng/mL내지 60 ng/mL인 것을 특징으로 하는 구강용해 필름 제형.
