本発明の検出用センサは、前述のように、SARS-CoV-2の検出用センサであって、前記本発明の核酸分子を含むことを特徴とする。本発明の検出用センサは、前記本発明の核酸分子を含んでいればよく、その他の構成は、特に制限されない。本発明の検出用センサを使用すれば、例えば、前記核酸分子と前記SARS-CoV-2スパイク糖タンパク質とを結合させることで、前述のように、前記SARS-CoV-2を検出できる。
本発明の検出試薬は、前記本発明のSARS-CoV-2スパイク糖タンパク質結合核酸分子を含むことを特徴とする。本発明の検出試薬は、前記本発明の核酸分子を含んでいればよく、その他の構成は何ら制限されない。本発明の検出試薬を使用すれば、前述のように、例えば、前記SARS-CoV-2の検出等を行うことができる。
本発明の検出方法は、前述のように、試料と核酸分子とを接触させ、前記試料中のSARS-CoV-2スパイク糖タンパク質を検出する工程を含み、前記核酸分子が、前記本発明のSARS-CoV-2スパイク糖タンパク質結合核酸分子であり、前記検出工程において、前記試料中のSARS-CoV-2スパイク糖タンパク質と前記核酸分子とを結合させて、前記結合により、前記試料中のSARS-CoV-2を検出することを特徴とする。本発明の検出方法は、前記本発明の核酸分子を使用することが特徴であって、その他の工程および条件等は、特に制限されない。また、本発明の検出方法は、前記本発明の核酸分子として、前記本発明のSARS-CoV-2検出用センサまたは本発明の検出試薬もしくは検出キットを使用してもよい。
本発明のSARS-CoV-2ウイルスの不活性化剤は、前述のように、前記本発明のSARS-CoV-2スパイク糖タンパク質結合核酸分子を含むことを特徴とする。本発明の不活性化剤は、前記本発明の核酸分子を使用することが特徴であって、その他の構成は、特に制限されない。
本発明のSARS-CoV-2スパイク糖タンパク質結合核酸分子について、SARS-CoV-2スパイク糖タンパク質に対する結合能を、SPRにより確認した。
前記配列番号1~7の塩基配列からなるポリヌクレオチド(SARS-CoV-2スパイク糖タンパク質結合核酸分子1~7(以下、それぞれ、「結合核酸分子1~7」ともいう))を合成し、実施例のDNAアプタマーとした。SARS-CoV-2スパイク糖タンパク質結合核酸分子1~7は、前記配列番号1~7の塩基配列における下線部のチミンヌクレオチド残基を、前記式(1)で表されるNG7とした。
下記表1に示す4種類の市販の試薬を用い、以下の試験に使用した。なお、表中、ターゲットは、それぞれ、「RBDドメイン」が、SARS-CoV-2スパイク糖タンパク質を構成するRBDドメイン(以下、RBDともいう。)、「S1+S2ドメイン」が、SARS-CoV-2スパイク糖タンパク質を構成するS1+S2ドメイン(以下、S1S2ともいう。)、「S1ドメイン」が、SARS-CoV-2スパイク糖タンパク質を構成するS1ドメイン(以下、S1ともいう。)、「三量体」が、前記S1+S2ドメインの三量体(以下、三量体ともいう。)を示す。なお、各ターゲットのアミノ酸配列は、以下のNCBIアクセッション番号に登録されたアミノ酸配列に基づき調製されており、前述の配列番号15~18のアミノ酸配列を有する。

結合能の解析には、ProteOn XPR36(BioRad社)を、その使用説明書にしたがって使用した。
本発明のSARS-CoV-2スパイク糖タンパク質結合核酸分子について、核酸の非特異的結合を防ぐ効果のあるデキストランを加えた溶液条件で、SARS-CoV-2スパイク糖タンパク質に対する結合能を、SPRにより確認した。また、動態パラメータを求めた。
前記配列番号1~7の塩基配列からなるポリヌクレオチドに加え、前記配列番号8~13の塩基配列からなるポリヌクレオチド(SARS-CoV-2スパイク糖タンパク質結合核酸分子8~13(以下、それぞれ、結合核酸分子8~13ともいう))を、前記実施例1と同様にして合成した。

試料として、前記実施例1の表1に示す前記RBD、および前記三量体を用いた。
SPRによる結合能の解析は、前記実施例1と同様にして行った。前記各試料の濃度は、それぞれ、前記RBDの場合、400、200、100、50、25nmol/Lとし、前記三量体の場合、200、100、50、25、12.5nmol/Lとした。
本発明のSARS-CoV-2スパイク糖タンパク質結合核酸分子について、SARS-CoV-2スパイク糖タンパク質に対する結合能を、ELAA(Enzyme Linked Aptamer Assay)によって評価した。
前記配列番号1および8の塩基配列からなるポリヌクレオチド(結合核酸分子1および結合核酸分子8)を、前記実施例1と同様にして合成した。
試料として、前記実施例1の表1に示す前記RBDを用いた。
ELAAによる結合能の解析は、非特許文献2(I. Shiratori et al, Biochem Biophys Res Commun 443 (2014) 37-41.)に記載の方法と同様に行った。具体的には、前記RBDを50mmol/l 炭酸バッファー(pH9.6)で1μg/100μlになるように希釈した。前記希釈後、前記RBDは、 Maxisorpプレート(Thermo Scientific/Nunc, Waltham, MA)のウェルに1μg/ウェルになるように加え、4℃、一晩の条件下で、固相化した。前記固相化後、ウェルを200μlのSB1Tバッファー(40mmol/l HEPES(pH7.4)、125mmol/l NaCl、1mmol/l MgCl2、5mmol/l KCl、0.01% Tween20)で1回洗浄し、前記洗浄後、200μlのTBS blocking buffer(Cat No.: 37570、Pierce Biotechnology社製)で、室温(約25℃、以下、同様)、1時間の条件下で、ブロッキングを行った。つぎに、実施例1(3)と同様にして、アプタマーの5’末端を、ビオチン化した。前記ビオチン化後、前記アプタマーは、SB1Tバッファーで1μmol/lに調整し、95℃、5分の条件下で熱変性を行った。前記変性後、4℃に急冷してフォールディングを行った。前記アプタマーは、RBDを固相化した前記ウェルに50μl/ウェルになるように加え、25℃、1時間の条件下で、インキュベートした。前記インキュベート後、前記ウェルを200μlのSB1Tバッファーで3回洗浄した。前記洗浄後、SB1Tバッファーで1000倍に希釈したstreptavidin-horseradish peroxidase(SA-HRP)(Citiva社製)を100μl加え、25℃、30分の条件下で、インキュベートした。前記インキュベート後、前記ウェルをSB1Tバッファーで3回洗浄した。前記洗浄後、TMB solution (MOSS社製)を100μl加え、室温、10分の条件下で、インキュベートした。前記インキュベート後、0.5Nの硫酸を加え、反応を停止させた。その後、infinite M1000Pro(Tecan社製)を用いて、前記ウェルについて、490nmと620nm(バックグラウンド)との吸光度を測定した。そして、各結合核酸分子について、490nmと620nmとの吸光度の差分(ABS(490nm-620nm))を算出した。コントロールは、前記結合核酸分子1および8を未添加とした以外は同様にして、吸光度の差分を算出した。
