Abstract
本発明の課題は、配合変化を起こさず、澄明であり、さらに十分な保存効力を有するジクアホソル点眼液を調製するにあたって、取り扱いが容易なジクアホソルと防腐剤との組合せを探索することである。 本発明は、ジクアホソルまたはその塩を有効成分として含有し、およびアルキルジアミノエチルグリシンまたはその塩を含有する水性点眼液である。
Description
本発明は、ジクアホソルまたはその塩を有効成分として含有する水性点眼液(以下、ジクアホソル点眼液ともいう)であって、アルキルジアミノエチルグリシンまたはその塩を含有する水性点眼液に関する。
ジクアホソルはP1,P4-ジ(ウリジン-5’)四リン酸またはUp4Uとも呼ばれるプリン受容体アゴニストであり、特許第3652707号公報(特許文献1)に開示されているように、涙液分泌促進作用を有することが知られている。また、Cornea,23(8),784-792(2004)(非特許文献1)には、ジクアホソル四ナトリウム塩を含有する点眼液を点眼投与することにより、ドライアイ患者の角膜上皮障害が改善したことが記載されている。実際、我が国においては、3%(w/v)の濃度のジクアホソル四ナトリウム塩を含有する点眼液がドライアイ治療薬として使用されている(製品名:ジクアス(登録商標)点眼液3%)。
他方、一般に、点眼液には、開封後一定期間にわたり何回も使用するタイプ(マルチドーズ型点眼液)と1回使い切りタイプ(ユニットドーズ型点眼液)がある。特に、マルチドーズ型点眼液には、使用時の微生物汚染等による製品の腐敗を防止し、点眼液の保存安定性を確保するために、防腐剤が含まれることが一般的である。
しかしながら、いかなる防腐剤であっても点眼液に好適に使用できるわけではなく、例えば、主薬と防腐剤との組み合わせによっては、濃度依存的に配合変化(沈殿・懸濁)を起こしてしまい取扱いが困難になることがある。例えば、処方の開発の際に、より高い保存効力を付与するために配合量を増やしたい場合などは問題となる。

したがって、配合変化を起こさず、澄明であり、さらに十分な保存効力を有するジクアホソル点眼液を調製するにあたって、取り扱いが容易なジクアホソルと防腐剤との組合せを探索することは興味深い課題である。
本発明者らは、上記課題を解決するため鋭意研究を行った結果、ジクアホソル点眼液に、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を用いた場合、広い濃度範囲での使用においても配合変化が生じず安定であり、また適量の配合により十分な保存効力を付与できることを見出した。すなわち、本発明者らは、ジクアホソル点眼液を調製するにあたり、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を用いることで、配合変化を起こさず、澄明であり、さらに十分な保存効力を有する点眼液を得ることができることを見出して、本発明を完成させた。
すなわち、本発明は、以下を提供する。
(1)ジクアホソルまたはその塩を有効成分として含有し、およびアルキルジアミノエチルグリシンまたはその塩を含有する水性点眼液。
(2)アルキルジアミノエチルグリシンまたはその塩の濃度が0.1%(w/v)以下である、(1)に記載の水性点眼液。
(3)アルキルジアミノエチルグリシンまたはその塩の濃度が0.01%(w/v)以下である、(1)に記載の水性点眼液。
(4)アルキルジアミノエチルグリシンまたはその塩の濃度が0.0001~0.01%(w/v)である、(1)に記載の水性点眼液。
(5)アルキルジアミノエチルグリシンまたはその塩の濃度が0.001%(w/v)超0.01%(w/v)以下である、(1)に記載の水性点眼液。
(6)アルキルジアミノエチルグリシンまたはその塩の濃度が0.001%(w/v)超0.005%(w/v)以下である、(1)に記載の水性点眼液。
(7)アルキルジアミノエチルグリシンまたはその塩がアルキルジアミノエチルグリシン塩酸塩である、(1)~(6)のいずれかに記載の水性点眼液。
(8)ジクアホソルまたはその塩の濃度が1~10%(w/v)である、(1)~(7)のいずれかに記載の水性点眼液。
(9)ジクアホソルまたはその塩の濃度が3%(w/v)である、(1)~(7)のいずれかに記載の水性点眼液。
(10)水性点眼液のpHが6~8の範囲である、(1)~(9)のいずれかに記載の水性点眼液。
(11)ジクアホソルまたはその塩が、ジクアホソルナトリウムである、(1)~(10)のいずれかに記載の水性点眼液。
(12)ジクアホソルまたはその塩を有効成分として含有する水性点眼液に保存効力を付与する方法であって、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする、方法。
(13)防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする、ジクアホソルまたはその塩を有効成分として含有する澄明な水性点眼液の製造方法。

(2)アルキルジアミノエチルグリシンまたはその塩の濃度が0.1%(w/v)以下である、(1)に記載の水性点眼液。
(3)アルキルジアミノエチルグリシンまたはその塩の濃度が0.01%(w/v)以下である、(1)に記載の水性点眼液。
(4)アルキルジアミノエチルグリシンまたはその塩の濃度が0.0001~0.01%(w/v)である、(1)に記載の水性点眼液。
(5)アルキルジアミノエチルグリシンまたはその塩の濃度が0.001%(w/v)超0.01%(w/v)以下である、(1)に記載の水性点眼液。
(6)アルキルジアミノエチルグリシンまたはその塩の濃度が0.001%(w/v)超0.005%(w/v)以下である、(1)に記載の水性点眼液。
(7)アルキルジアミノエチルグリシンまたはその塩がアルキルジアミノエチルグリシン塩酸塩である、(1)~(6)のいずれかに記載の水性点眼液。

なお、前記(1)~(13)の各構成は、任意に2つ以上を選択して組み合わせることができる。
本発明によれば、配合変化を起こさず、澄明であり、さらに十分な保存効力を有するジクアホソル点眼液を得ることができる。
以下に、本発明に関してさらに詳しく説明する。
本発明において、「水性点眼液」とは水を溶媒(基剤)とする点眼液を意味する。
本明細書において、「%(w/v)」は、本発明の水性点眼液100mL中に含まれる対象成分の質量(g)を意味する。
ジクアホソルは、下記化学構造式で示される化合物である。
「ジクアホソルの塩」としては、医薬として許容される塩であれば特に制限はなく、リチウム、ナトリウム、カリウム、カルシウム、マグネシウム、亜鉛などとの金属塩;塩酸、臭化水素酸、ヨウ化水素酸、硝酸、硫酸、リン酸などの無機酸との塩;酢酸、フマル酸、マレイン酸、コハク酸、クエン酸、酒石酸、アジピン酸、グルコン酸、グルコヘプト酸、グルクロン酸、テレフタル酸、メタンスルホン酸、乳酸、馬尿酸、1,2-エタンジスルホン酸、イセチオン酸、ラクトビオン酸、オレイン酸、パモ酸、ポリガラクツロン酸、ステアリン酸、タンニン酸、トリフルオロメタンスルホン酸、ベンゼンスルホン酸、p-トルエンスルホン酸、硫酸ラウリルエステル、硫酸メチル、ナフタレンスルホン酸、スルホサリチル酸などの有機酸との塩;臭化メチル、ヨウ化メチルなどとの四級アンモニウム塩;臭素イオン、塩素イオン、ヨウ素イオンなどのハロゲンイオンとの塩;アンモニアとの塩;トリエチレンジアミン、2-アミノエタノール、2,2-イミノビス(エタノール)、1-デオキシ-1-(メチルアミノ)-2-D-ソルビトール、2-アミノ-2-(ヒドロキシメチル)-1,3-プロパンジオール、プロカイン、N,N-ビス(フェニルメチル)-1,2-エタンジアミンなどの有機アミンとの塩などが挙げられる。
本発明において、「ジクアホソルまたはその塩」には、ジクアホソル(フリー体)またはその塩の水和物および有機溶媒和物も含まれる。
ジクアホソルまたはその塩に、結晶多形および結晶多形群(結晶多形システム)が存在する場合には、それらの結晶多形体および結晶多形群(結晶多形システム)も本発明の範囲に含まれる。ここで、結晶多形群(結晶多形システム)とは、それらの結晶の製造、晶出、保存などの条件および状態により、結晶形が変化する場合の各段階における個々の結晶形およびその過程全体を意味する。
本発明の「ジクアホソルまたはその塩」として好ましいのはジクアホソルのナトリウム塩であり、下記化学構造式で示されるジクアホソル四ナトリウム塩(以下、単に「ジクアホソルナトリウム」ともいう)が特に好ましい。
ジクアホソルまたはその塩については、特表2001-510484号公報に開示された方法などにより製造することができる。
本点眼液はジクアホソルまたはその塩以外の有効成分を含有することもできるが、好ましくは、ジクアホソルまたはその塩を唯一の有効成分として含有する。
本点眼液中のジクアホソルまたはその塩の濃度は、例えば、0.1~10%(w/v)であり、1~10%(w/v)であることが好ましく、1~5%(w/v)であることがより好ましく、3%(w/v)であることが特に好ましい。
本発明の水性点眼液は、防腐剤としてアルキルジアミノエチルグリシンまたはその塩、好ましくはアルキルジアミノエチルグリシン塩酸塩が配合される。アルキルジアミノエチルグリシン塩酸塩は、一般に使用されるアルキルジアミノエチルグリシン塩酸塩(示性式:[RNHCH2CH2NHCH2CH2NHCH2COOH]・HCl(RはC8H17~C16H33、主としてC12H25およびC14H29からなる))を使用できる。
アルキルジアミノエチルグリシンまたはその塩は、公知の方法により合成してもよく、市販品として入手することもできる。
本発明の水性点眼液において、アルキルジアミノエチルグリシンまたはその塩の濃度は、例えば、0.1%(w/v)以下、0.05%(w/v)以下、0.02%(w/v)以下、0.01%(w/v)以下で使用でき、好ましくは、0.008%(w/v)以下、0.007%(w/v)以下、0.006%(w/v)以下で使用でき、より好ましくは、0.005%(w/v)以下で使用できる。また、アルキルジアミノエチルグリシンまたはその塩の濃度の下限は、例えば0.0001%(w/v)以上で使用でき、好ましくは0.001%(w/v)超で使用でき、より好ましくは0.002%(w/v)以上、0.003%(w/v)以上、0.004%(w/v)以上で使用でき、さらに好ましくは0.005%(w/v)以上で使用できる。また、上記いずれの上限および下限をそれぞれ独立に選択し組み合わせて範囲とすることができる。具体的に、アルキルジアミノエチルグリシンまたはその塩の濃度の範囲としては、例えば、0.0001~0.1%(w/v)が好ましく、0.0001~0.05%(w/v)がより好ましく、0.0001~0.01%(w/v)がさらに好ましく、0.001%(w/v)超0.01%(w/v)以下がさらにより好ましく、0.001%(w/v)超0.005%(w/v)以下が特に好ましい。
本点眼液には、汎用されている技術を用い、必要に応じて製薬学的に許容される添加剤を添加することができ、例えば、リン酸ナトリウム、リン酸水素ナトリウム、リン酸水素ナトリウム水和物、リン酸二水素ナトリウム、酢酸ナトリウム、イプシロン-アミノカプロン酸などの緩衝化剤;塩化ナトリウム、塩化カリウム、濃グリセリン、マンニトールなどの等張化剤;ポリオキシエチレンソルビタンモノオレート、ステアリン酸ポリオキシル40、ポリオキシエチレン硬化ヒマシ油などの界面活性剤;クエン酸ナトリウム、エデト酸ナトリウム、エデト酸ナトリウム水和物などの安定化剤などを必要に応じて選択し、添加することができる。
本点眼液のpHは眼科製剤に許容される範囲内にあればよいが、通常pH4~8の範囲内が好ましく、pH6~8の範囲がより好ましく、pH7付近が特に好ましい。本点眼液には、適宜、塩酸や水酸化ナトリウムなどのpH調節剤を添加することができる。
本点眼液は、ジクアホソルまたはその塩を有効成分として含有し、「ドライアイの予防および/または治療」に使用することができる。
ドライアイは「様々な要因による涙液および角結膜上皮の慢性疾患であり、眼不快感や視覚異常を伴う疾患」と定義づけられ、乾性角結膜炎(KCS)はドライアイに含まれる。また、本発明においては、ソフトコンタクトレンズ装用を原因とするドライアイ症状の発生もドライアイに含まれるものとする。
ドライアイ症状には、眼乾燥感、眼不快感、眼疲労感、鈍重感、羞明感、眼痛、霧視(かすみ目)などの自覚症状の他、充血、角結膜上皮障害などの他覚所見も含まれる。
ドライアイの病因については不明点も多いが、シェーグレン症候群;先天性無涙腺症;サルコイドーシス;骨髄移植による移植片対宿主病(GVHD:Graft Versus Host Disease);眼類天疱瘡;スティーブンス・ジョンソン症候群;トラコーマなどを原因とする涙器閉塞;糖尿病;角膜屈折矯正手術(LASIK:Laser(-assisted) in Situ Keratomileusis)などを原因とする反射性分泌の低下;meibom腺機能不全;眼瞼炎などを原因とする油層減少;眼球突出、兎眼などを原因とする瞬目不全または閉瞼不全;胚細胞からのムチン分泌低下;VDT作業などがその原因であると報告されている。
本発明において、「ドライアイの予防および/または治療」とは、ドライアイに伴う病的症状および/または所見を予防および/または治療もしくは改善することと定義づけられ、ドライアイに伴う眼乾燥感、眼不快感、眼疲労感、鈍重感、羞明感、眼痛、霧視(かすみ目)などの自覚症状の予防および/または治療もしくは改善を意味するだけでなく、ドライアイに伴う充血、角結膜上皮障害などの予防および/または治療もしくは改善も含まれる。また、「ドライアイの予防および/または治療」には、ソフトコンタクトレンズ装用眼における涙液層の安定性の向上によってドライアイ症状を予防および/または治療もしくは改善せしめることも含まれる。なお、ドライアイ症状の予防および/または治療もしくは改善には、ドライアイ患者がソフトコンタクトレンズ装用することで悪化するに至ったドライアイ症状の予防および/または治療もしくは改善、ソフトコンタクトレンズ装用そのものにより発生するに至ったドライアイ症状の予防および/または治療もしくは改善をも意味する。
本点眼液の用法は、剤形、投与すべき患者の症状の軽重、年令、体重、医師の判断などに応じて適宜変えることができるが、例えば、剤形として点眼剤を選択した場合には、1日1~10回、好ましくは1日2~8回、より好ましくは1日3~6回に分けて眼局所に投与することができる。
本点眼液は、ソフトコンタクトレンズ用として、ソフトコンタクトレンズ装着時にも使用することができる。ソフトコンタクトレンズとしては、例えば、ヒドロキシエチルメタクリレートを主成分とするコンタクトレンズまたはシリコーンハイドロゲルコンタクトレンズなどが挙げられる。
本点眼液の適用対象となるソフトコンタクトレンズの種類については特に制限されるものでなく、また、イオン性または非イオン性、含水性または非含水性の別を問わない。例えば、繰り返し使用されるレンズの他、1日使い捨て用レンズ、1週間使い捨て用レンズ、2週間使い捨て用レンズなどの現在市販されているまたは将来市販されるすべてのソフトコンタクトレンズに適用可能である。
本発明の一態様は、ジクアホソルまたはその塩を有効成分として含有する水性点眼液に保存効力を付与する方法であって、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする、方法である。本発明の方法は、ジクアホソルまたはその塩を有効成分として含有する水性点眼液に保存効力を付与するにあたり、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする。なお、上記本発明の水性点眼液の詳細な説明は、本発明の水性点眼液に保存効力を付与する方法にも適用される。
本発明の一態様は、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする、ジクアホソルまたはその塩を有効成分として含有する澄明な水性点眼液の製造方法である。本発明の製造方法は、ジクアホソルまたはその塩を有効成分として含有する澄明な水性点眼液の製造にあたり、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする。なお、上記本発明の水性点眼液の詳細な説明は、本発明の水性点眼液の製造方法にも適用される。
以下に、試験例および製剤例を挙げて本発明を詳細に説明するが、これらの例は本発明をよりよく理解するためのものであり、本発明の範囲を何ら限定するものではない。
[試験例1]
表1に示す処方のジクアホソル点眼液を調製し、各点眼液の外観について、調製直後目視で確認して評価した。

表2に示されるとおり、0.01%(w/v)の濃度のPHMBをジクアホソル点眼液に配合した場合では白濁してしまい、無色澄明の点眼液を得ることができなかった。一方で、アルキルジアミノエチルグリシン塩酸塩を0.0001%(w/v)、0.001%(w/v)、0.005%(w/v)、0.01%(w/v)の濃度でジクアホソル点眼液に配合した場合、その外観に変化は見られず、いずれの濃度であっても無色澄明の点眼液を得ることができた。以上より、澄明なジクアホソル点眼液を得るためには防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することが好ましいことが示された。
[試験例2]

(試料調製)
試験例1と同様に、表1に示す処方の点眼液1~3を調製した。

(試験方法)
保存効力試験は、第十七改正日本薬局方の保存効力試験法に準拠して行なった。本試験では、試験菌として、Esherichia Coli(E.coli)、Pseudomonas aeruginosa(P.aeruginosa)、Staphylococcus aureus(S.aureus)、Candida albicans(C.albicans)およびAspergillus brasiliensis(A.brasiliensis)を用いた。各製剤からなる試験試料中の菌液濃度が105~106cfu/mL(5菌種共)となるように、接種菌液を試験試料に接種した。具体的には、107~108cfu/mLとなるように接種菌液を調製し、この接種菌液を105~106cfu/mLとなるように、点眼液1~3の製剤からなる試験試料に各接種菌液を接種し、均一に混合し試料とした。これらの試料を遮光下20~25℃に保存し、各サンプリングポイント(7日後、14日後、又は28日後)において、各試料からマイクロピペットで1mLを採取し、生菌数を測定した。各サンプリングポイントでは、試料溶液の蓋を空けてサンプリングを実施し、蓋を閉める操作を行った。

試験の合否判定は、生菌数を測定したときの菌数(A)に対する接種時の菌数(B)の比(B/A)の常用対数値を算出し、その値が細菌種(E.coli、P.aeruginosa、S.aureus)に対しては、播種7日後に1.0以上、かつ14日後または28日後に3.0以上であること、および真菌種(C.albicans、A.brasiliensis)に対しては、播種7日後と比較して播種14日後または28日後の数値が減少していないこと、をいずれも満たすときに適合とした。なお、常用対数値が、例えば、「1」の場合には、検査時の生菌数が接種菌数の10%に減少したことを意味する。

表3に示されるとおり、点眼液3は、日本薬局方の保存効力試験基準に適合した。

以下に製剤例を挙げて本発明をさらに具体的に説明するが、本発明はこれらの製剤例にのみ限定されるものではない。なお、下記製剤例において各成分の配合量は製剤100mL中の含量である。

本発明は、配合変化を起こさず、澄明であり、さらに十分な保存効力を有するジクアホソル点眼液を得ることができて有用である。
Claims (13)
- ジクアホソルまたはその塩を有効成分として含有し、およびアルキルジアミノエチルグリシンまたはその塩を含有する水性点眼液。
- アルキルジアミノエチルグリシンまたはその塩の濃度が0.1%(w/v)以下である、請求項1に記載の水性点眼液。
- アルキルジアミノエチルグリシンまたはその塩の濃度が0.01%(w/v)以下である、請求項1に記載の水性点眼液。
- アルキルジアミノエチルグリシンまたはその塩の濃度が0.0001~0.01%(w/v)である、請求項1に記載の水性点眼液。
- アルキルジアミノエチルグリシンまたはその塩の濃度が0.001%(w/v)超0.01%(w/v)以下である、請求項1に記載の水性点眼液。
- アルキルジアミノエチルグリシンまたはその塩の濃度が0.001%(w/v)超0.005%(w/v)以下である、請求項1に記載の水性点眼液。
- アルキルジアミノエチルグリシンまたはその塩がアルキルジアミノエチルグリシン塩酸塩である、請求項1~6のいずれか一項に記載の水性点眼液。
- ジクアホソルまたはその塩の濃度が1~10%(w/v)である、請求項1~7のいずれか一項に記載の水性点眼液。
- ジクアホソルまたはその塩の濃度が3%(w/v)である、請求項1~7のいずれか一項に記載の水性点眼液。
- 水性点眼液のpHが6~8の範囲である、請求項1~9のいずれか一項に記載の水性点眼液。
- ジクアホソルまたはその塩が、ジクアホソルナトリウムである、請求項1~10のいずれか一項に記載の水性点眼液。
- ジクアホソルまたはその塩を有効成分として含有する水性点眼液に保存効力を付与する方法であって、防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする、方法。
- 防腐剤としてアルキルジアミノエチルグリシンまたはその塩を配合することを特徴とする、ジクアホソルまたはその塩を有効成分として含有する澄明な水性点眼液の製造方法。
