簡便に製造可能な両親媒性分子として、下記の一般式(I) 〔式中、R1及びR5はハロゲン原子などを表し、R2及びR4は水素原子などを表し、R3はトリメチルアンモニウム基などを表し、R6、R7、及びR8はフェニル基などを表す。〕 で表される化合物を提供する。
Description
本発明は、両親媒性分子及びその製造方法に関する。本発明の両親媒性分子は、簡便に製造できるという特徴を有する。
両親媒性分子とは、一つの分子骨格内に親水部位(水になじむ)と疎水部位(水になじまない)を有する分子である。この分子は、水中では疎水部位同士が引き寄せ合うことで一つの集合体を形成し、集合体の内部に汚れ分子や成分を取り込める特徴がある。両親媒性分子としては、ドデシル硫酸ナトリウム(SDS)やCHAPSなど多くの分子が知られている。吉沢らによって開発され、本出願人によって以前出願された両親媒性分子も(特許文献1)、このような公知の両親媒性分子の一つである。この両親媒性分子は、ベンゼン環を軸に、2つの疎水部位(アントラセン)を120度の角度で連結した湾曲型の疎水性骨格を持ち、その湾曲部の外側に親水性の官能基を持つ。
従来の両親媒性分子の製造方法では、疎水部位へ高価な反応剤を介した有機合成的手法を使い、親水部位を導入する方法が取られていた。このため、両親媒性分子の製造には、煩雑な作業が必要であった。上述した特許文献1に記載されている両親媒性分子は、根岸カップリング反応を利用することによって、煩雑な精製作業なしに簡単な洗浄操作で製造できるという利点を有しているが、疎水部位の構築と親水部位の構築の二つの工程を有しており、必ずしも簡便に製造できるというわけではなかった。
本発明は、このような技術的背景のもとになされたものであり、簡便に製造可能な両親媒性分子を提供すること目的とする。
本発明者は、上記課題を解決するため鋭意検討を重ねた結果、シュタウディンガー反応というクリック反応の一種に着目し、予め製作した「反応点が導入された親水部位」に有機リン化合物を「混ぜるだけ」で、添加物を一切必要とせず、簡単に両親媒性分子を合成できることを見出し、この知見に基づき、本発明を完成するに至った。
即ち、本発明は、以下の(1)~(16)を提供する。
(1)下記の一般式(I)

(7)一般式(A)及び(B)におけるL1、L2、L3、及びL4が、アルキレン基(但し、アルキレン基の1以上の-CH2-は、-O-又は-CO-で置換されていてもよい。)である(1)乃至(6)のいずれかに記載の化合物。

本発明は、新規な両親媒性分子を提供する。この両親媒性分子は、従来の両親媒性分子と比べ、簡便に製造できるなど様々な利点を有する。
以下、本発明を詳細に説明する。
本発明において「ハロゲン原子」とは、例えば、フッ素原子、塩素原子、臭素原子、ヨウ素原子などである。
本発明において「ハロゲン原子」とは、例えば、フッ素原子、塩素原子、臭素原子、ヨウ素原子などである。
本発明において「炭素数1~4のアルキル基」とは、炭素数が1以上4以下の直鎖又は分枝鎖アルキル基であり、例えば、メチル基、エチル基、n-プロピル基、iso-プロピル基、n-ブチル基、iso-ブチル基、sec-ブチル基、tert-ブチル基などである。
本発明において「炭素数1~4のアルコシ基」とは、炭素数が1以上4以下の直鎖又は分枝鎖アルコキシ基であり、例えば、メトキシ基、エトキシ基、n-プロポキシ基、iso-プロポキシ基、n-ブトキシ基、iso-ブトキシ基、sec-ブトキシ基、tert-ブトキシ基などである。
本発明において「アリール基」とは、例えば、フェニル基、1-ナフチル基、2-ナフチル基、9-アンスリル基などである。
本発明において「ヘテロアリール基」とは、例えば、ピリジニル基(ピリジン-2-イル基、ピリジン-3-イル基、ピリジン-4-イル基)、ピリミジニル基(ピリミジン-2-イル基、ピリミジン-4-イル基、ピリミジン-5-イル基)、フラニル基(フラン-2-イル基、フラン-3-イル基)、チエニル基(チオフェン-2-イル基、チオフェン-3-イル基)、キノリニル基(キノリン-2-イル基、キノリン-3-イル基、キノリン-4-イル基、キノリン-5-イル基、キノリン-6-イル基)などである。
本発明において「炭素数4~8のシクロアルキル基」とは、例えば、シクロブチル基、シクロペンチル基、シクロヘキシル基、シクロヘプチル基、シクロオクチル基などである。
本発明において「炭素数4~10のアルキル基」とは、炭素数が4以上10以下の直鎖又は分枝鎖アルキル基であり、例えば、n-ブチル基、iso-ブチル基、sec-ブチル基、tert-ブチル基、ペンチル基、iso-ペンチル基、neo-ペンチル基、ヘキシル基、iso-ヘキシル基、ヘプチル基、iso-ヘプチル基、オクチル基、ノニル基、デシル基などである。
本発明において「炭素数4~10のアルケニル基」とは、炭素数が4以上10以下の直鎖又は分枝鎖アルケニル基であり、例えば、1-メチル-2-プロペニル基、2-メチル-2-プロペニル基、1-ブテニル基、2-ブテニル基、3-メチル-2-ブテニル基、1-メチル-2-ブテニル基、3-ブテニル基、1-ペンテニル基、1-ヘキセニル基、1-ヘプテニル、1-オクテニル、1-ノネニル、1-デセニルなどである。
本発明において「炭素数4~10のアルキニル基」とは、炭素数が4以上10以下の直鎖又は分枝鎖アルキニル基であり、例えば、1-ブチニル基、1-ペンチニル基、1-ヘキシニル基、1-へプチニル基、1-オクチニル基、1-ノニニル基、1-デシニル基などである。
本発明において「炭素数7~10のアリールアルキル基」とは、例えば、ベンジル基、フェネチル基などである。
本発明において「炭素数8~10のアリールアルケニル基」とは、例えば、スチリル基などである。
本発明において「炭素数8~10のアリールアルキニル基」とは、例えば、フェニルエチニル基などである。
一般式(I)においてR1及びR5はそれぞれ独立して、ハロゲン原子、アルキル基、又はアリール基を表す。ここで、アルキル基は炭素数1~4のアルキル基が好ましいが、これに限定されるわけではない。アリール基はフェニル基が好ましいが、これに限定されるわけではない。R1とR5は異なる基であってもよいが、同一の基であることが好ましい。R1及びR5は、前記した基であればよいが、好ましくは、ハロゲン原子であり、より好ましくは、フッ素原子、塩素原子、又は臭素原子である。
一般式(I)においてR2及びR4はそれぞれ独立して、水素原子、ハロゲン原子、アルキル基、アルコキシ基、アリール基、ヒドロキシ基、アミノ基、又はシアノ基を表す。ここで、アルキル基は炭素数1~4のアルキル基が好ましく、アルコキシ基は炭素数1~4のアルコシ基が好ましいが、これらに限定されるわけではない。アリール基はフェニル基が好ましいが、これに限定されるわけではない。R2とR4は異なる基であってもよいが、同一の基であることが好ましい。R2及びR4は、前記した基であればよいが、好ましくは、水素原子、又はハロゲン原子であり、より好ましくは、水素原子である。
一般式(A)においてL1は存在しないか、又は二価の鎖状基を表す。ここで、「L1が存在しない」とは、R9が一般式(I)におけるベンゼン環に直接結合していることを意味する。L1は、存在しないか、又は二価の鎖状基であればよいが、好ましくは、アルキレン基であり、より好ましくは、-CO-O-CH2-CH2-である。ここで、アルキレン基の1以上の-CH2-は、-O-又は-CO-で置換されていてもよい。アルキレン基の炭素数は特に限定されないが、好ましくは、1~10であり、より好ましくは、1~5である。なお、アルキレン基中の-CH2-を、-O-又は-CO-で置換した場合も、これらの基は1つの炭素を持つものとして、前記した「アルキレン基の炭素数」に含める。アルキレン基中に占める-O-又は-CO-の割合は特に限定されないが、好ましくは、50%以下である。
一般式(A)においてR9は親水基を表す。親水基とは、水との親和性に優れた基を意味する。R9は親水基であればよいが、トリアルキルアンモニウム基、スルホ基、ポリエチレングリコール基、アミノ基、ヒドロキシ基、カルボキシル基、シアノ基、メトキシ基、又はニトロ基が好ましく、トリメチルアンモニウム基、スルホ基、又はポリエチレングリコール基がより好ましく、トリメチルアンモニウム基が更に好ましい。ここで、トリアルキルアンモニウム基におけるアルキル基は、炭素数1~4のアルキル基が好ましいが、これに限定されるわけではない。なお、「ポリエチレングリコール基」とは、ポリエチレングリコールの末端のヒドロキシ基又は水素原子を除去することによって得られる一価の官能基を意味する。
一般式(B)においてL2、L3、及びL4はそれぞれ独立して、存在しないか、又は二価の鎖状基を表す。ここで、「L2、L3、又はL4が存在しない」とは、R10、R11、又はR12が一般式(B)におけるベンゼン環に直接結合していることを意味する。L2、L3、及びL4は異なる基であってもよく、同一の基であってもよい。L2、L3、及びL4は、存在しないか、又は二価の鎖状基であればよいが、好ましくは、アルキレン基であり、より好ましくは、-CO-O-CH2-CH2-である。ここで、アルキレン基の1以上の-CH2-は、-O-又は-CO-で置換されていてもよい。アルキレン基の炭素数は特に限定されないが、好ましくは、1~10であり、より好ましくは、1~5である。なお、アルキレン基中の-CH2-を、-O-又は-CO-で置換した場合も、これらの基は1つの炭素を持つものとして、前記した「アルキレン基の炭素数」に含める。アルキレン基中に占める-O-又は-CO-の割合は特に限定されないが、好ましくは、50%以下である。
一般式 (B)においてR10、R11、及びR12はそれぞれ独立して、水素原子又は親水基を表す。但し、R10、R11、及びR12の少なくとも一つは親水基を表す。R10、R11、及びR12は異なる基であってもよいが、R10とR12は同一の基であることが好ましい。R10、R11、及びR12は水素原子又は親水基であればよいが、好ましくは、水素原子、トリアルキルアンモニウム基、スルホ基、ポリエチレングリコール基、アミノ基、ヒドロキシ基、カルボキシル基、シアノ基、メトキシ基、又はニトロ基が好ましく、水素原子、トリメチルアンモニウム基、スルホ基、又はポリエチレングリコール基がより好ましく、水素原子、又はトリメチルアンモニウム基が更に好ましい。ここで、トリアルキルアンモニウム基におけるアルキル基は、炭素数1~4のアルキル基が好ましいが、これに限定されるわけではない。
一般式(I)におけるR2、R3、及びR4のいずれもが一般式(A)又は(B)で示される基でない場合、一般式(I)で表される化合物は親水基を持たないことになるので、R2、R3、及びR4の少なくとも一つは一般式(A)又は(B)で示される基である必要がある。R2、R3、及びR4は、少なくとも一つが一般式(A)又は(B)で示される基であればよいが、1)R2及びR4が一般式(A)で示される基以外の基であり、R3が一般式(A)又は(B)で示される基であること、2)R2及びR4が一般式(A)で示される基であり、R3が一般式(A)又は(B)で示される基以外の基であること、又は3)R2及びR4が一般式(A)で示される基であり、R3が一般式(A)又は(B)で示される基であることが好ましい。
一般式(I)においてR6、R7、及びR8はそれぞれ独立して、置換基群aから選ばれた1以上の置換基によって置換されていてもよいアリール基、置換基群aから選ばれた1以上の置換基によって置換されていてもよいヘテロアリール基、シクロアルキル基、アルキル基、アルケニル基、アルキニル基、アリールアルキル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)、アリールアルケニル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)、アリールアルキニル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)を表し、置換基群aはハロゲン原子、アルキル基、アルコキシ基、カルボニル基、カルボキシル基、アリール基、ヒドロキシ基、チオール基、及びアミノ基からなる。ここで、アリール基はフェニル基が好ましく、ヘテロアリール基はフラン-2-イル基が好ましく、アリールアルキル基はフェニルアルキル基が好ましく、アリールアルケニル基はフェニルアルケニル基が好ましく、アリールアルキニル基はフェニルアルキニル基が好ましいが、これらに限定されるわけではない。また、シクロアルキル基は炭素数4~8のシクロアルキル基が好ましく、アルキル基は炭素数4~10のアルキル基が好ましく、アルケニル基は炭素数4~10のアルケニル基が好ましく、アルキニル基は炭素数4~10のアルキニル基が好ましく、アリールアルキル基は炭素数7~10のアリールアルキル基が好ましく、アリールアルケニル基は炭素数8~10のアリールアルケニル基が好ましく、アリールアルキニル基は炭素数8~10のアリールアルキニル基が好ましいが、これらに限定されるわけではない。
アリール基、ヘテロアリール基、及びアリールアルキル基などのアリール基部分の水素原子は、置換基によって置換されていてもよい。置換基の数は1以上であればよいが、好ましくは、1である。置換基の位置は特に限定されないが、例えば、フェニル基の1の水素原子を置換基によって置換する場合、フェニル基の4位の水素原子を置換基によって置換することが好ましい。R6、R7、及びR8は異なる基であってもよいが、同一の基であることが好ましい。置換基としては、ハロゲン原子、アルキル基、アルコキシ基、カルボニル基、カルボキシル基、アリール基、ヒドロキシ基、チオール基、及びアミノ基を挙げることができる。ここで、アルキル基は炭素数1~4のアルキル基が好ましく、メチル基がより好ましいが、これらに限定されるわけではない。アルコキシ基は炭素数1~4のアルコキシ基が好ましく、メトキシ基がより好ましいが、これらに限定されるわけではない。アリール基は、フェニル基が好ましいが、これに限定されるわけではない。R6、R7、及びR8は前記した基であればよいが、好ましくは、フェニル基、4-メチルフェニル基、4-メトキシフェニル基、フラン-2-イル基、シクロヘキシル基、又はフェニルエチニル基であり、より好ましくは、フェニル基である。
R9、R10、R11、及びR12の少なくとも一つがイオン性官能基である場合、一般式(I)で表される化合物は、陰イオン又は陽イオンと塩を形成することができる。陰イオン及び陽イオンは、一般式(I)で表される化合物と水溶性の塩を形成できることが好ましく、例えば、陰イオンとしては、フッ化物イオン、塩化物イオン、臭化物イオンを挙げることができ、陽イオンとしては、ナトリウムイオン、カリウムイオン、アンモニウムイオンを挙げることができる。
一般式(I)で表される化合物は、下記の一般式(II)

一般式(II)で表される化合物は、実施例1に記載した4-アミノ-3,5-ジクロロ安息香酸を原料とする製造方法に、必要に応じて、改変や修正を加えた方法に従って製造できる。一般式(III)で表される化合物は、その多くが市販品として購入可能であり、市販品として入手できない場合であっても、当業者であれば既知文献に従って製造できる。
一般式(II)で表される化合物と一般式(III)で表される化合物の混合は、溶媒の存在下で行う。使用する溶媒は特に限定されないが、両化合物の反応を妨げないものが好ましく、両化合物に対する溶解性が高いものが好ましい。具体的には、クロロホルム、アセトニトリル、メタノール、アセトン、テトラヒドロフラン、水、又はこれらの混合溶媒などを使用できる。
一般式(II)で表される化合物と一般式(III)で表される化合物の混合比は特に限定されないが、一般式(II)で表される化合物1molに対して一般式(III)で表される化合物を0.5~2molとすることが好ましく、0.8~1.3molとすることがより好ましい。
混合時の温度(反応温度)は適宜調節すればよいが、10~60℃であることが好ましく、20~30℃であることがより好ましい。
一般式(II)で表される化合物と一般式(III)で表される化合物の反応時間は、反応温度等、その他の条件に応じて適宜調節すればよいが、0.5~24時間であることが好ましく、2~8時間であることがより好ましい。
上記反応終了後、公知の手法(例えば、溶媒の留去、洗浄など)によって、必要に応じて後処理を行い、目的物である一般式(I)で表される化合物を得ることができる。
一般式(I)で表される化合物は、両親媒性分子であり、水中でミセルを形成する。このミセルの内部に、疎水性化合物(例えば、色素、薬剤など)を取り込むことができ、これにより、疎水性化合物を水溶化させることができる。このような疎水性化合物を水溶化できる性質から、多くの両親媒性分子は薬物輸送システム(DDS)に利用されているが、一般式(I)で表される化合物も、同様に、DDSに利用可能である。
一般式(I)で表される化合物は、以下のような特徴を有する。
1)反応点を持つ親水部位は安価で簡便に大量合成できる。
2)疎水部位(有機リン化合物)は安価でかつ豊富な種類があり、誘導体も簡便に合成可能である。
3)原料となる一般式(II)で表される化合物及び一般式(III)で表される化合物は室温で混ぜるだけで、一般式(I)で表される化合物へと変換できる。
4)水中でも安定に存在し、集合体を形成する。
5)水中で疎水性の色素分子を取り込める。

以下に、実施例により本発明を更に詳細に説明するが、本発明はこれらの実施例に限定されるものではない。
〔実施例1〕 アジド基を有する親水基の合成
<4-アジド-3,5-ジクロロ安息香酸(1b)の合成(図1)>
4-アミノ-3,5-ジクロロ安息香酸(0.500g,2.43mmol)と5%塩酸(10mL)とアセトニトリル(20mL)を混ぜて0℃に冷した。この溶液に、亜硝酸ナトリウム水溶液(0.201g,2.91mmol,2mL)を滴下し0℃で30分撹拌した。その後、アジ化ナトリウム水溶液(0.237g,3.64mmol,2mL)を滴下し、室温で10分撹拌した。析出した白色固体をろ集し、水で洗浄後、乾燥させることで4-アジド-3,5-ジクロロ安息香酸(1b)を得た(0.538mg,収率96%)。

<4-アジド-3,5-ジクロロ安息香酸(1b)の合成(図1)>

4-アジド-3,5-ジクロロ安息香酸1b(0.304g,2.43mmol)と炭酸カリウム(0.361g,2.59mmol)、2-クロロ-N,N-ジメチルエチルアミン塩酸塩(0.286g,1.94mmol)とジオキサン(20mL)を加えて110℃で30分撹拌した。室温まで冷ました後、水を加えて水相を酢酸エチルで抽出をした。有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除き、有機溶媒を留去した。ここへアセトニトリル(10mL)を加え、ヨウ化メチル(242μL,3.88mmol)を加え、室温で30分撹拌した。有機溶媒を留去後、得られた固体を再沈殿(アセトニトリル/ジエチエーテル)により精製することで、白色固体の4-アジド-3,5-ジクロロベンゾイルコリンヨウ化物塩(1c)を得た(0.400g,1bより収率70%)。

4-アジド-3,5-ジクロロベンゾイルコリンヨウ化物塩1c(45mg,0.100mmol)と塩化銀(17.4mg,0.120mmol)と純水(5mL)を加えて50℃で1時間撹拌した。析出した沈殿をろ過で除き、溶媒を留去後、得られた固体をアセトンで洗浄することで、白色固体の4-アジド-3,5-ジクロロベンゾイルコリン塩化物塩(1Cl)を得た(35.0mg,収率87%)。

〔実施例3〕 両親媒性分子の安定性評価
一般的にアザイリド(N=P)結合は、水によって容易に加水分解されて1級アミンとトリフェニルホスフィンオキシドを生成する。塩素原子を含まない両親媒性分子nNPPh3(0.002mmol)を重水(0.5mL)に溶かして60 ℃に加熱してNMRを用いて経過観察した。2時間後、nNPPh3はすべてトリフェニルホスフィンオキシドに変換されることが分かった。一方で、塩素原子を有するNPPh3(0.002mmol)では同一条件における加水分解率はわずか1%であることが1Hと31P NMR測定により明らかとなった(図6)。半減期に換算するとその安定性は340倍以上に増加した。なお、室温下におけるnNPPh3の半減期は7時間であるが、同一条件下におけるNPPh3の分解は数日間確認されなかった。

調製したNPPh3(3.6mg,0.006mmol)を純水(1.0mL)中に添加すると、疎水効果により疎水部位が相互作用し合うことで、球状の集合体を形成した(図7a)。1HNMRより芳香族ピークがブロード化かつ高磁場にシフトし、31P NMRよりリンのピークが著しくブロード化していた。この挙動はいずれも溶液中で集合体を形成することに由来する。濃度依存NMRの解析より、NPPh3の臨界ミセル濃度は約2.0mMであった。その集合体の大きさは、動的光散乱(DLS)測定より平均2.0nmの粒径であることが判明した。これは5分子のNPPh3集合体に相当する(図7b)。

〔実施例5〕 両親媒性分子による疎水性色素分子の水溶化
疎水性色素分子は水に難溶であるため、水溶媒中では全く蛍光を示さない。これに対し、調製した2mMのNPPh3水溶液(1mL)に疎水性色素分子であるナイルレッド(過剰量)を添加し、室温で1時間撹拌し、沈殿をろ過で除いた。その水溶液は色素分子の蛍光を強く示すことが分かった。これは疎水性色素分子がNPPh3の集合体に取り込まれ水溶化したことに由来する(図8)。内包の事実は紫外可視吸収測定及び蛍光測定により明らかにした。
疎水性色素分子は水に難溶であるため、水溶媒中では全く蛍光を示さない。これに対し、調製した2mMのNPPh3水溶液(1mL)に疎水性色素分子であるナイルレッド(過剰量)を添加し、室温で1時間撹拌し、沈殿をろ過で除いた。その水溶液は色素分子の蛍光を強く示すことが分かった。これは疎水性色素分子がNPPh3の集合体に取り込まれ水溶化したことに由来する(図8)。内包の事実は紫外可視吸収測定及び蛍光測定により明らかにした。
〔実施例6〕 両親媒性分子の特徴
1)親水基部位の大量合成が簡便であること
親水基部位1Clは市販品から簡便に合成できる。さらに、誘導体として他のハロゲン原子が付加した親水基も同様のプロセスで調製可能である(図9)。いずれもキログラム単位での大量合成も可能である。また、生成した親水基部位は固体状態で、酸素や熱に対して安定であるため取り扱いが容易である。
1)親水基部位の大量合成が簡便であること
親水基部位1Clは市販品から簡便に合成できる。さらに、誘導体として他のハロゲン原子が付加した親水基も同様のプロセスで調製可能である(図9)。いずれもキログラム単位での大量合成も可能である。また、生成した親水基部位は固体状態で、酸素や熱に対して安定であるため取り扱いが容易である。
2)いつでも・どこでも・どんなリン化合物でも両親媒性分子化できること
親水基部位と疎水部位(リン化合物)を単純に室温で混ぜるだけで両親媒性分子を定量的に調製することができる。特にリン化合物は市販品だけでも最低122種類(参考:東京化成工業株式会社 配位子パンフレット)存在するため、望みの性質を有する多種多様な両親媒性分子を提供できる可能性を秘めている。実際に、図10に示す6種類に関してアザイリド結合を形成することを確認している。すなわち、本発明が対象とするリン化合物は、アリールホスフィンだけではなく、アルキルホスフィン、エチニルホスフィンなど多岐にわたる。
親水基部位と疎水部位(リン化合物)を単純に室温で混ぜるだけで両親媒性分子を定量的に調製することができる。特にリン化合物は市販品だけでも最低122種類(参考:東京化成工業株式会社 配位子パンフレット)存在するため、望みの性質を有する多種多様な両親媒性分子を提供できる可能性を秘めている。実際に、図10に示す6種類に関してアザイリド結合を形成することを確認している。すなわち、本発明が対象とするリン化合物は、アリールホスフィンだけではなく、アルキルホスフィン、エチニルホスフィンなど多岐にわたる。
3)難水溶性色素分子を取り込み可溶化できること
NPPh3は分子内包能を有するため、種々の難水溶性色素分子を水溶化することができる。前述のNileredの他に、BODIPY、ペリレン、C60やナノカーボン材料などが挙げられる。

〔実施例7〕 1Hal・PAr3の合成
1Cl(15.0mg,0.0424mmol)とトリフェニルホスフィン(PPh3, 11.1mg,0.0424mmol)をアセトニトリル(5.0ml)に溶解し、60℃で10分撹拌した。溶媒を留去後、得られた固体をヘキサンで洗浄することで、白色固体の1Cl・PPh3を定量的に得た。同様に、トリ(p-トリル)ホスフィン(PTol3)、トリアニシルホスフィン(PAni3)を用い、対応する白色固体の1Cl・PTol3と1Cl・PAni3を定量的に得た。また、1Fと1Brを親水基に用いることで、1F・PPh3、1F・PTol3、1F・PAni3、1Br・PPh3、1Br・PTol3、1Br・PAni3が定量的に得られた。

〔実施例10〕 4・PTol3の合成
〔Bの合成〕
100ml一口フラスコにA(4.18g,20.0mmol)、10%塩酸水溶液30mlを加えて撹拌した。その後亜硝酸ナトリウム(1.68g,24.0mmol)の水溶液(20mL)を加えて0℃で2時間撹拌した。続いて、ヨウ化カリウム(5.00g,30.0mmol)を加えて室温下で1時間撹拌した。亜硫酸ナトリウム水溶液を加えて水相を酢酸エチルで抽出し、有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除き、有機溶媒を留去した。シリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=3:1)で目的物を分離し、メタノールで再結晶することで薄褐色結晶のBを得た(3.56g,収率55%)。

100ml二口フラスコにC(1.50g,6.23mmol)、B2pin2(1.90g,7.48mnol)、PdCl2(CH3CN)2(0.0808g,0.312mmol)、dppf(0.259g,0.467mmol)、酢酸カリウム(1.83g,18.7mmol)を加え、窒素置換をした。その後、脱水ジオキサン(25ml)を加え、80℃で21時間撹拌した。室温まで冷ました後、水を加えて水相を酢酸エチルで抽出をした。有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除き、有機溶媒を留去した。反応混合物をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=2:1)で分離精製することで、薄褐色固体のDを得た(1.64g,収率92%)。

100ml二口フラスコにB(0.313g,0.978mmol)、D(0.295g,1.03mnol)、PdCl2(CH3CN)2(0.0188g,0.0489mmol)、SPhos(0.0401g,0.0978mmol)、炭酸セシウム(0.956g,2.93mmol)を加え、窒素置換をした。その後、脱水トルエン(25ml)加え、110℃で4時間撹拌した。室温まで冷ました後、水を加えて水相を酢酸エチルで抽出をした。有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除き、有機溶媒を留去した。反応混合物をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=6:1)で分離精製することで、淡黄色固体のEを得た(0.171g,収率49%)。

〔Fの合成〕
50ml一口フラスコにE(0.229g,0.647mmol)とアセトニトリル(10ml)、10%塩酸水溶液(10ml)を加えて0℃に冷却した。亜硝酸ナトリウム(0.625g,0.906mmol)の水溶液(2ml)を加えて0℃で2時間撹拌した。その後、アジ化ナトリウム(0.715g,1.10mmol)水溶液(2ml)を加えて室温下で1時間撹拌した。水相を酢酸エチルで抽出し、有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除き、有機溶媒を留去した。
上記操作で得られた固体(0.237g)と水酸化カリウム(0.104g,1.87mmol)、THF(16ml)を50ml一口フラスコに加えて70℃で1.5時間撹拌した。室温まで冷ました後、水を加えて水相を酢酸エチルで抽出をし、有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除いた。有機溶媒を留去することで、淡黄色固体のFを得た(0.218g, 収率96%)

〔Gの合成〕
50ml一口フラスコにF(0.275g,0.781mmol)、炭酸カリウム(0.431g,3.12mmol)、2-クロロ-N,N-ジメチルエチルアミン塩酸塩(0.337g,2.34mmol)とジメチルスルホキシド(3ml)を加えて80℃で1.5時間撹拌した。室温まで冷ました後、水を加えて水相をトルエンで抽出をした。有機相を硫酸マグネシウムで乾燥させ、ろ過で乾燥剤を取り除き、有機溶媒を留去した。ここへアセトニトリル(10ml)とヨウ化メチル(292μl,4.68mmol)を加え、室温で1.5時間撹拌した。有機溶媒を留去後、得られた固体を再沈殿(アセトニトリル/ジエチエーテル)により精製することで、白色固体のGを得た(330mg,Eより収率43%)。

〔4の合成〕
10mL反応用バイアルにG(37.0mg,0.0475mmol)と塩化銀(13.6mg,0.114mmol)、純水(8ml)を加えて60℃で1時間撹拌した。析出した沈殿をろ過で除き、溶媒を留去後、得られた固体をアセトンで洗浄することで、白色固体の4を得た(23.4mg,82%)。
1H NMR (500 MHz, CD3CN, 298 K):δ 3.19 (s, 18H), 3.84 (m, 4H), 4.73 (m, 4H), 7.80 (s, 2H), 8.46 (s, 2H), 8.63 (s, 1H)

〔4・PTol3の合成〕
4(10.0mg,0.0167mmol)とトリ(p-トリル)ホスフィン(PTol3,5.1mg,0.0167mmol)をアセトニトリル(5ml)溶解し、室温で1.5時間撹拌した。溶媒を留去後、得られた固体をヘキサンで洗浄することで、白色固体の4・PTol3を定量的に得た。
1H NMR (500 MHz, CDCl3, 298 K): δ3.28 (s, 18H), 3.61 (m, 4H), 4.07 (m, 4H), 7.23 (m, J = 3.5 Hz 6H), 7.48 (s, 2H), 7.64 (m, J = 6.8 Hz, 6H), 8.34 (s, 2H), 8.55 (s, 1H).
31P NMR (202 MHz, CDCl3, 298 K): δ3.12.
1H NMR (500 MHz, CDCl3, 298 K): δ3.28 (s, 18H), 3.61 (m, 4H), 4.07 (m, 4H), 7.23 (m, J = 3.5 Hz 6H), 7.48 (s, 2H), 7.64 (m, J = 6.8 Hz, 6H), 8.34 (s, 2H), 8.55 (s, 1H).
31P NMR (202 MHz, CDCl3, 298 K): δ3.12.

Claims (16)
- 下記の一般式(I)
で示される基を表し、R3は水素原子、ハロゲン原子、一般式(A)で示される基、又は下記の一般式(B)
で示される基を表し(但し、R2、R3、及びR4の少なくとも一つは一般式(A)で示される基又は一般式(B)で示される基を表す。)、R6、R7、及びR8はそれぞれ独立して、置換基群aから選ばれた1以上の置換基によって置換されていてもよいアリール基、置換基群aから選ばれた1以上の置換基によって置換されていてもよいヘテロアリール基、シクロアルキル基、アルキル基、アルケニル基、アルキニル基、アリールアルキル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)、アリールアルケニル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)、又はアリールアルキニル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)を表し、置換基群aはハロゲン原子、アルキル基、アルコキシ基、カルボニル基、カルボキシル基、アリール基、ヒドロキシ基、チオール基、及びアミノ基からなる。〕

- 一般式(I)におけるR1及びR5が、ハロゲン原子である請求項1乃至4のいずれか一項に記載の化合物。
- 一般式(I)におけるR2及びR4が、水素原子又はハロゲン原子である請求項1乃至5のいずれか一項に記載の化合物。
- 一般式(A)及び(B)におけるL1、L2、L3、及びL4が、アルキレン基(但し、アルキレン基の1以上の-CH2-は、-O-又は-CO-で置換されていてもよい。)である請求項1乃至6のいずれか一項に記載の化合物。
- 一般式(I)におけるR6、R7、及びR8が、フェニル基、4-メチルフェニル基、4-メトキシフェニル基、フラン-2-イル基、シクロヘキシル基、又はフェニルエチニル基である請求項1乃至7のいずれか一項に記載の化合物。
- 下記の一般式(I)
で示される基を表し、R3は水素原子、ハロゲン原子、一般式(A)で示される基、又は下記の一般式(B)
で示される基を表し(但し、R2、R3、及びR4の少なくとも一つは一般式(A)で示される基又は一般式(B)で示される基を表す。)、R6、R7、及びR8はそれぞれ独立して、置換基群aから選ばれた1以上の置換基によって置換されていてもよいアリール基、置換基群aから選ばれた1以上の置換基によって置換されていてもよいヘテロアリール基、シクロアルキル基、アルキル基、アルケニル基、アルキニル基、アリールアルキル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)、アリールアルケニル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)、又はアリールアルキニル基(アリール基部分は置換基群aから選ばれた1以上の置換基によって置換されていてもよい)を表し、置換基群aはハロゲン原子、アルキル基、アルコキシ基、カルボニル基、カルボキシル基、アリール基、ヒドロキシ基、チオール基、及びアミノ基からなる。〕
で表される化合物を製造する方法であって、下記の一般式(II)
で表される化合物と下記の一般式(III)
で表される化合物を混合する工程を含む、上記方法。 - 親水基が、トリアルキルアンモニウム基、スルホ基、ポリエチレングリコール基、アミノ基、ヒドロキシ基、カルボキシル基、シアノ基、メトキシ基、又はニトロ基である請求項9に記載の方法。
- 親水基が、トリメチルアンモニウム基、スルホ基、又はポリエチレングリコール基である請求項9に記載の方法。
- 親水基が、トリメチルアンモニウム基又はカルボキシル基である請求項9に記載の方法。
- 一般式(I)及び(II)におけるR1及びR5が、ハロゲン原子である請求項9乃至12のいずれか一項に記載の方法。
- 一般式(I)及び(II)におけるR2及びR4が、水素原子又はハロゲン原子である請求項9乃至13のいずれか一項に記載の方法。
- 一般式(A)及び(B)におけるL1、L2、L3、及びL4が、アルキレン基(但し、アルキレン基の1以上の-CH2-は、-O-又は-CO-で置換されていてもよい。)である請求項9乃至14のいずれか一項に記載の方法。
- 一般式(I) 及び(III)におけるR6、R7、及びR8が、フェニル基、4-メチルフェニル基、4-メトキシフェニル基、フラン-2-イル基、シクロヘキシル基、又はフェニルエチニル基である請求項9乃至15のいずれか一項に記載の方法。
