Description
本発明は、変異型ACE2タンパク質等に関する。
コロナウイルスは、コロナウイルス科のコロナウイルス亜科に属するウイルスの種であり、ヒトおよび動物に感染し、呼吸器、消化器、血管、または神経性の疾患を発症させるプラス鎖RNAウイルスである。コロナウイルスは、感染している何らかの宿主動物から伝播して、ヒトに対して大規模な伝染病を引き起こすことが可能である。コロナウイルスとしては、近年では、例えば、2002年及び2003年に流行したSARSコロナウイルス1(SARS-CoV-1)、及び中東呼吸器症候群コロナウイルス(MERS-CoV)が挙げられる。最近では、SARSコロナウイルス2(SARS-CoV-2)の世界的な流行が起こり、治療技術が確立されていないことから、医療、経済等の多方面に亘って甚大な被害が続いている。
コロナウイルスに対して特異的に結合できる抗体は、治療に有用であると思われるものの、耐性化の問題がある。一方、ACE2(Angiotensin-converting enzyme 2)は細胞膜上に存在し、コロナウイルス表面上のSタンパク質と結合することにより、コロナウイルスの細胞内への進入に関与することが知られており、これを治療に利用することが報告されている(非特許文献1)。ACE2は、耐性化の問題が生じ難く、治療に有用であると考えられている。

野生型のACE2は、抗体に比べ、コロナウイルスのSタンパク質に対する結合性が低い。
そこで、本発明は、コロナウイルスのSタンパク質に対する結合性がより高い変異型ACE2タンパク質を提供することを課題とする。
本発明者は、上記課題に鑑みて鋭意研究を進めた結果、
配列番号1に示されるアミノ酸配列A、又は前記アミノ酸配列Aと70%以上の同一性を有するアミノ酸配列Bが変異してなるアミノ酸配列Cを含み、前記アミノ酸配列Cは、変異a:前記アミノ酸配列AにおけるT20、A25、K26、K31、H34、E35、Q60、N64、S70、T78、L79、N90、T92、及びQ101からなる群より選択される少なくとも2種のアミノ酸の置換ax、又は前記アミノ酸配列Bにおける前記置換axに対応する置換ayを含む、変異型ACE2タンパク質、であれば、上記課題と解決できることを見出した。本発明者は、この知見に基づいてさらに研究を進めた結果、本発明を完成させた。即ち、本発明は、下記の態様を包含する。

項6. 前記変異aにおいて、A25の変異後のアミノ酸がより嵩高い疎水性アミノ酸であり、K26の変異後のアミノ酸が非塩基性アミノ酸であり、K31の変異後のアミノ酸が非塩基性アミノ酸であり、E35の変異後のアミノ酸が非酸性アミノ酸であり、N64の変異後のアミノ酸が疎水性アミノ酸であり、L79の変異後のアミノ酸がより嵩高い疎水性アミノ酸であり、且つ/或いはN90の変異後のアミノ酸が塩基性アミノ酸又は酸性アミノ酸である、項2~5のいずれかに記載の変異型ACE2タンパク質。
項7. 前記変異aにおいて、A25の変異後のアミノ酸が分岐鎖アミノ酸であり、K26の変異後のアミノ酸が酸性アミノ酸であり、K31の変異後のアミノ酸が親水性中性アミノ酸であり、E35の変異後のアミノ酸が塩基性アミノ酸であり、N64の変異後のアミノ酸が分岐鎖アミノ酸であり、L79の変異後のアミノ酸が芳香族アミノ酸であり、且つ/或いはN90の変異後のアミノ酸が塩基性アミノ酸である、項2~6のいずれかに記載の変異型ACE2タンパク質。

本明細書において、アミノ酸配列の「同一性」とは、2以上の対比可能なアミノ酸配列の、お互いに対するアミノ酸配列の一致の程度をいう。従って、ある2つのアミノ酸配列の一致性が高いほど、それらの配列の同一性又は類似性は高い。アミノ酸配列の同一性のレベルは、例えば、配列分析用ツールであるFASTAを用い、デフォルトパラメータを用いて決定される。若しくは、Karlin及びAltschulによるアルゴリズムBLAST(Karlin S, Altschul SF.“Methods for assessing the statistical significance of molecular sequence features by using general scoringschemes”Proc Natl Acad Sci USA.87:2264-2268(1990)、KarlinS,Altschul SF.“Applications and statistics for multiple high-scoring segments in molecular sequences.”Proc Natl Acad Sci USA.90:5873-7(1993))を用いて決定できる。このようなBLASTのアルゴリズムに基づいたBLASTXと呼ばれるプログラムが開発されている。これらの解析方法の具体的な手法は公知であり、National Center of Biotechnology Information(NCBI)のウェブサイト(http://www.ncbi.nlm.nih.gov/)を参照すればよい。また、塩基配列の「同一性」も上記に準じて定義される。 本明細書中において、「保存的置換」とは、アミノ酸残基が類似の側鎖を有するアミノ酸残基に置換されることを意味する。例えば、リジン、アルギニン、ヒスチジンといった塩基性側鎖を有するアミノ酸残基同士で置換されることが、保存的な置換にあたる。その他、アスパラギン酸、グルタミン酸といった酸性側鎖を有するアミノ酸残基;グリシン、アスパラギン、グルタミン、セリン、トレオニン、チロシン、システインといった非帯電性極性側鎖を有するアミノ酸残基;アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファンといった非極性側鎖を有するアミノ酸残基;トレオニン、バリン、イソロイシンといったβ-分枝側鎖を有するアミノ酸残基;チロシン、フェニルアラニン、トリプトファン、ヒスチジンといった芳香族側鎖を有するアミノ酸残基同士での置換も同様に、保存的な置換にあたる。
本明細書において、DNA、RNAなどのヌクレオチドには、次に例示するように、公知の化学修飾が施されていてもよい。ヌクレアーゼなどの加水分解酵素による分解を防ぐために、各ヌクレオチドのリン酸残基(ホスフェート)を、例えば、ホスホロチオエート(PS)、メチルホスホネート、ホスホロジチオネート等の化学修飾リン酸残基に置換することができる。また、各リボヌクレオチドの糖(リボース)の2位の水酸基を、-OR(Rは、例えばCH3(2´-O-Me)、CH2CH2OCH3(2´-O-MOE)、CH2CH2NHC(NH)NH2、CH2CONHCH3、CH2CH2CN等を示す)に置換してもよい。さらに、塩基部分(ピリミジン、プリン)に化学修飾を施してもよく、例えば、ピリミジン塩基の5位へのメチル基やカチオン性官能基の導入、あるいは2位のカルボニル基のチオカルボニルへの置換などが挙げられる。さらには、リン酸部分やヒドロキシル部分が、例えば、ビオチン、アミノ基、低級アルキルアミン基、アセチル基等で修飾されたものなどを挙げることができるが、これに限定されない。また、ヌクレオチドの糖部の2´酸素と4´炭素を架橋することにより、糖部のコンフォーメーションをN型に固定したものであるBNA(LNA)等もまた、好ましく用いられ得る。
本明細書において、アミノ酸の変異は、具体的には、アミノ酸の欠失、置換、挿入、又は付加である。
本明細書において、アミノ酸配列中のアミノ酸の位置は、アミノ酸一文字表記+N末端アミノ酸から数えた場合のアミノ酸番号で示すことがある。例えば、「A25」は、N末端から25番目のアミノ酸であるアラニンを示す。なお、本明細書において、アミノ酸番号は、配列番号1で示されるアミノ酸配列ヒトACE2(1-615)のN末端から数えた場合のアミノ酸番号である。よって、例えばヒトACE2(18-615:配列番号2)のA25とは、配列番号1のN末端から25番目のアミノ酸であるアラニンに対応する、配列番号2におけるアラニンであり、これは配列番号2のN末端から8番目のアミノ酸である。
2.変異型ACE2タンパク質
本発明は、その一態様において、配列番号1に示されるアミノ酸配列A、又は前記アミノ酸配列Aと70%以上の同一性を有するアミノ酸配列Bが変異してなるアミノ酸配列Cを含み、前記アミノ酸配列Cは、変異a:前記アミノ酸配列AにおけるT20、A25、K26、K31、H34、E35、Q60、N64、S70、T78、L79、N90、T92、及びQ101からなる群より選択される少なくとも2種のアミノ酸の置換ax、又は前記アミノ酸配列Bにおける前記置換axに対応する置換ayを含む、変異型ACE2タンパク質(本明細書において、「本発明の変異型ACE2タンパク質」と示すこともある。)に関する。以下に、これについて説明する。
本発明は、その一態様において、配列番号1に示されるアミノ酸配列A、又は前記アミノ酸配列Aと70%以上の同一性を有するアミノ酸配列Bが変異してなるアミノ酸配列Cを含み、前記アミノ酸配列Cは、変異a:前記アミノ酸配列AにおけるT20、A25、K26、K31、H34、E35、Q60、N64、S70、T78、L79、N90、T92、及びQ101からなる群より選択される少なくとも2種のアミノ酸の置換ax、又は前記アミノ酸配列Bにおける前記置換axに対応する置換ayを含む、変異型ACE2タンパク質(本明細書において、「本発明の変異型ACE2タンパク質」と示すこともある。)に関する。以下に、これについて説明する。
アミノ酸配列Cは、配列番号1に示されるアミノ酸配列A、又は前記アミノ酸配列Aと70%以上の同一性を有するアミノ酸配列Bが変異してなるアミノ酸配列であって、変異aを含む限り、特に制限されない。
アミノ酸配列A(配列番号1)は、ヒト(Homo sapiens)が内在的に発現する(固有に有する)ACE2(Angiotensin-converting enzyme 2)タンパク質(NCBI Accession Number: NP_001358344及びNP_068576)のアミノ酸配列(配列番号3)のN末端から1番目のアミノ酸から615番目のアミノ酸からなる配列である。
アミノ酸配列Bは、該配列に変異aが導入されてなる配列からなるタンパク質(タンパク質B)の、コロナウイルスSタンパク質に対するKD値が、アミノ酸配列Aに変異aが導入されてなる配列からなるタンパク質(タンパク質A)の該KD値から著しく高くならない限り、特に制限されない。タンパク質Bの該KD値は、タンパク質Aの該KD値に対して、例えば10倍以下、好ましくは5倍以下、より好ましくは3倍以下、さらに好ましくは2倍以下、よりさらに好ましくは1.5倍以下、とりわけ好ましくは1.2倍以下である。この観点から、アミノ酸配列Bのアミノ酸配列Aに対する同一性は、好ましくは85%以上、より好ましくは90%以上、さらに好ましくは95%以上、よりさらに好ましくは97%以上、特に好ましくは99%以上であり、また100%未満である。
コロナウイルスSタンパク質に対するKD値は、後述の実施例に記載のBinding assayの方法に従って測定することができる。
アミノ酸配列Bとしては、好ましくはアミノ酸配列Aからシグナルペプチド配列(N末端から1番目のアミノ酸から17番目のアミノ酸からなる配列)が除かれてなるアミノ酸配列B1(配列番号2)、ヒトACE2タンパク質のアミノ酸配列(配列番号3)の全部又は一部のアミノ酸配列B2(例えば、配列番号3のN末端から1~20番目のいずれかのアミノ酸から520~805番目(好ましくは550~805、より好ましくは580~805、さらに好ましくは600~805)のいずれかのアミノ酸からなる配列、好ましくは配列番号3のN末端から18番目のアミノ酸から740番目のアミノ酸からなる配列(配列番号12))、アミノ酸配列A、B1、若しくはB2のオルソログアミノ酸配列B3(例えば、ヒト以外の動物(例えば、サル、マウス、ラット、イヌ、ネコ、ウサギ、ブタ、ウマ、ウシ、ヒツジ、ヤギ、シカ等の哺乳類動物)におけるACE2タンパク質のアミノ酸配列又はその一部の配列)等、さらにはこれらの種内変異配列等が挙げられる。なお、ヒト以外の動物のACE2タンパク質のアミノ酸配列は、公知である、又は公知のアミノ酸配列のゲノム情報から予測できる、又は公知のアミノ酸配列に基づいたクローニングにより同定できる。
変異aは、アミノ酸配列AにおけるT20、A25、K26、K31、H34、E35、Q60、N64、S70、T78、L79、N90、T92、及びQ101からなる群より選択される少なくとも2種のアミノ酸の置換ax、又は前記アミノ酸配列Bにおける前記置換axに対応する置換ay、である。
なお、「対応する置換」とは、2つの配列をBLAST(設定はデフォルト)で比較した場合に、アライメント配列上同一の位置のアミノ酸の置換であることを示す。例えば、アミノ酸配列AにおけるA25の置換に対応する置換とは、アミノ酸配列Aとアミノ酸配列Bと配列比較して得られるアライメント配列において、アミノ酸配列Bにおいて、アミノ酸配列AにおけるA25と同一の位置のアミノ酸の置換を示す。また、アミノ酸配列Aにおける置換後のアミノ酸とアミノ酸配列Bにおける置換後のアミノ酸は同一である。
置換axは、少なくとも3種のアミノ酸置換、すなわちアミノ酸配列AにおけるT20、A25、K26、K31、H34、E35、Q60、N64、S70、T78、L79、N90、T92、及びQ101からなる群より選択される少なくとも3種のアミノ酸の置換であることが好ましい。同様に、置換axは、少なくとも4種のアミノ酸の置換であることがより好ましく、少なくとも5種のアミノ酸の置換であることがさらに好ましく、少なくとも6種のアミノ酸の置換であることがよりさらに好ましく、少なくとも7種のアミノ酸の置換であることがとりわけ好ましい。
置換axは、A25、K26、K31、E35、N64、L79、N90、及びT92からなる群より選択される少なくとも2種のアミノ酸の置換(置換ax1)を含むことが好ましい。
置換ax1は、A25、K31、及びE35の置換を含むことがとりわけ好ましい。中でも、置換ax1は、A25、K31、E35、及びT92の置換を含む、或いはA25、K31、E35、及びL79の置換を含むことが特に好ましい。
置換ax1は、少なくとも3種のアミノ酸の置換、すなわちアミノ酸配列AにおけるA25、K26、K31、E35、N64、L79、N90、及びT92からなる群より選択される少なくとも3種のアミノ酸の置換であることが好ましい。同様に、置換ax1は、少なくとも4種のアミノ酸の置換であることがより好ましく、少なくとも5種のアミノ酸の置換であることがさらに好ましく、少なくとも6種のアミノ酸の置換であることがよりさらに好ましく、7種(A25、K26、K31、E35、N64、L79、及びN90)のアミノ酸の置換であることがとりわけ好ましい。
置換axは、K31、E35、Q60、S70、L79、及びN90からなる群より選択される少なくとも2種のアミノ酸の置換(置換ax2)を含むことが好ましい。
置換ax2は、K31、E35、及びQ60の置換を含むことが好ましい。置換ax2は、K31、E35、Q60、及びL79の置換を含むことがとりわけ好ましい。
置換ax2は、少なくとも3種のアミノ酸の置換、すなわちアミノ酸配列AにおけるK31、E35、Q60、S70、L79、及びN90からなる群より選択される少なくとも3種のアミノ酸の置換であることが好ましい。同様に、置換ax2は、少なくとも4種のアミノ酸の置換であることがより好ましく、少なくとも5種のアミノ酸の置換であることがさらに好ましく、6種のアミノ酸の置換であることがよりさらに好ましい。
置換axは、T20、A25、H34、T78、T92、及びQ101からなる群より選択される少なくとも2種のアミノ酸の置換(置換ax3)を含むことが好ましい。置換ax3は、T20、H34、T92、及びQ101Hの置換を含むことがとりわけ好ましい。また、置換ax3は、A25、H34、T78、及びT92の置換を含むことが好ましい。
置換ax3は、少なくとも3種のアミノ酸の置換、すなわちアミノ酸配列AにおけるT20、A25、H34、T78、T92、及びQ101からなる群より選択される少なくとも3種のアミノ酸の置換であることが好ましい。同様に、置換ax3は、少なくとも4種のアミノ酸の置換であることがより好ましく、少なくとも5種のアミノ酸の置換であることがさらに好ましく、6種のアミノ酸の置換であることがよりさらに好ましい。
変異aにおけるT20又はそれに対応するアミノ酸の変異後のアミノ酸は、疎水性アミノ酸であることが好ましい。疎水性アミノ酸としては、例えばバリン、ロイシン、イソロイシン、グリシン、アラニン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはバリン、ロイシン、イソロイシン等の分岐鎖アミノ酸が挙げられ、より好ましくはイソロイシンが挙げられる。
変異aにおけるA25又はそれに対応するアミノ酸の変異後のアミノ酸は、より嵩高い疎水性アミノ酸であることが好ましい。なお、本明細書において、「より嵩高い」とは、側鎖の分子量がより大きいこと(例えば、側鎖におけるアルキル基(メチル基、エチル基等)等の付加又は置換、側鎖の鎖状構造(例えばアルキル基)の芳香族基への置換、側鎖の芳香族基の環数の増加等)であり、この限りにおいて特に制限されない。より嵩高い疎水性アミノ酸としては、例えばバリン、ロイシン、イソロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはバリン、ロイシン、イソロイシン等の分岐鎖アミノ酸が挙げられ、より好ましくはバリンが挙げられる。
変異aにおけるK26又はそれに対応するアミノ酸の変異後のアミノ酸は、非塩基性アミノ酸であることが好ましい。非塩基性アミノ酸としては、例えばグルタミン酸、アスパラギン酸、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン、グリシン、アラニン、バリン、ロイシン、イソロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはグルタミン酸、アスパラギン酸、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン等の非塩基性親水性アミノ酸が挙げられ、より好ましくはグルタミン酸、アスパラギン酸等の酸性アミノ酸が挙げられ、さらに好ましくはグルタミン酸が挙げられる。
変異aにおけるK31又はそれに対応するアミノ酸の変異後のアミノ酸は、非塩基性アミノ酸であることが好ましい。非塩基性アミノ酸としては、例えばグルタミン酸、アスパラギン酸、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン、グリシン、アラニン、バリン、ロイシン、イソロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。本発明の一態様において(特に置換ax1において)、これらの非塩基性アミノ酸の中でも、好ましくはグルタミン酸、アスパラギン酸、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン等の非塩基性親水性アミノ酸が挙げられ、より好ましくはセリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン等の親水性中性アミノ酸が挙げられ、さらに好ましくはアスパラギン、グルタミン等が挙げられ、よりさらに好ましくはアスパラギンが挙げられる。本発明の一態様において(特に置換ax2において)、上記非塩基性アミノ酸の中でも、好ましくはバリン、ロイシン、イソロイシン、グリシン、アラニン、プロリン、トリプトファン、フェニルアラニン、メチオニン等の疎水性アミノ酸が挙げられ、より好ましくはメチオニンが挙げられる。
変異aにおけるH34又はそれに対応するアミノ酸の変異後のアミノ酸は、非塩基性アミノ酸であることが好ましい。非塩基性アミノ酸としては、例えばグルタミン酸、アスパラギン酸、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン、グリシン、アラニン、バリン、ロイシン、イソロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはグリシン、アラニン、バリン、ロイシン、イソロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等の疎水性アミノ酸が挙げられ、より好ましくはグリシン、アラニン、バリン、ロイシン、イソロイシン等が挙げられ、さらに好ましくはアラニンが挙げられる。
変異aにおけるE35又はそれに対応するアミノ酸の変異後のアミノ酸は、非酸性アミノ酸であることが好ましい。非酸性アミノ酸としては、例えばリジン、アルギニン、ヒスチジン、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン、グリシン、アラニン、バリン、ロイシン、イソロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはリジン、アルギニン、ヒスチジン、セリン、トレオニン、グルタミン、アスパラギン、チロシン、システイン等の非酸性親水性アミノ酸が挙げられ、より好ましくはリジン、アルギニン、ヒスチジン等の塩基性アミノ酸が挙げられ、さらに好ましくはリジンが挙げられる。
変異aにおけるQ60又はそれに対応するアミノ酸の変異後のアミノ酸は、塩基性アミノ酸であることが好ましい。塩基性アミノ酸としては、例えばリジン、アルギニン、ヒスチジン等が挙げられ、好ましくはアルギニンが挙げられる。
変異aにおけるN64又はそれに対応するアミノ酸の変異後のアミノ酸は、疎水性アミノ酸であることが好ましい。疎水性アミノ酸としては、例えばバリン、ロイシン、イソロイシン、グリシン、アラニン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはバリン、ロイシン、イソロイシン等の分岐鎖アミノ酸が挙げられ、より好ましくはイソロイシンが挙げられる。
変異aにおけるS70又はそれに対応するアミノ酸の変異後のアミノ酸は、疎水性アミノ酸であることが好ましい。疎水性アミノ酸としては、例えばバリン、ロイシン、イソロイシン、グリシン、アラニン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。これらの中でも、好ましくはトリプトファン、フェニルアラニン等の芳香族アミノ酸が挙げられ、より好ましくはフェニルアラニンが挙げられる。
変異aにおけるT78又はそれに対応するアミノ酸の変異後のアミノ酸は、塩基性アミノ酸であることが好ましい。塩基性アミノ酸としては、例えばリジン、アルギニン、ヒスチジン等が挙げられ、好ましくはアルギニンが挙げられる。
変異aにおけるL79又はそれに対応するアミノ酸の変異後のアミノ酸は、より嵩高い疎水性アミノ酸であることが好ましい。より嵩高い疎水性アミノ酸としては、例えばイソロイシン、プロリン、トリプトファン、フェニルアラニン等が挙げられる。これらの中でも、好ましくはトリプトファン、フェニルアラニン等の芳香族アミノ酸が挙げられ、より好ましくはフェニルアラニンが挙げられる。
変異aにおけるN90又はそれに対応するアミノ酸の変異後のアミノ酸は、荷電アミノ酸であることが好ましい。荷電アミノ酸としては、例えばリジン、アルギニン、ヒスチジン、アスパラギン酸、グルタミン酸等が挙げられる。本発明の一態様において(特に置換ax1において)、これらの荷電アミノ酸の中でも、好ましくはリジン、アルギニン、ヒスチジン等の塩基性アミノ酸が挙げられ、より好ましくはヒスチジンが挙げられる。本発明の一態様において(特に置換ax2において)、上記荷電アミノ酸の中でも、好ましくはアスパラギン酸、グルタミン酸等の酸性アミノ酸が挙げられ、より好ましくはアスパラギン酸が挙げられる。
変異aにおけるT92又はそれに対応するアミノ酸の変異後のアミノ酸は、グルタミン、アスパラギン等の側鎖にカルバモイル基を有するアミノ酸であることが好ましい。これらの中でも、より好ましくはグルタミンが挙げられる。
変異aにおけるQ101又はそれに対応するアミノ酸の変異後のアミノ酸は、塩基性アミノ酸であることが好ましい。塩基性アミノ酸としては、例えばリジン、アルギニン、ヒスチジン等が挙げられる。これらの中でも、好ましくはヒスチジンが挙げられる。
アミノ酸配列Cは、コロナウイルスSタンパク質に対する結合性が著しく損なわれない限りにおいて、他のアミノ酸変異(例えば置換、保存的置換)を含むことができる。他のアミノ酸変異の数は、例えば1~50個、1~20個、1~10個、又は1~5個である。
アミノ酸配列Cは、closed conformationに固定するための変異を含むことが好ましい。当該変異は、通常、2つのアミノ酸(非システイン)のシステインへの置換である。変異位置はACE2の結晶構造に基づいて適宜決定することができる。当該変異としては、例えばS128CとV343C、あるいはV59CとN121Cの組み合わせが挙げられる。
アミノ酸配列Cとして、具体的には、例えば配列番号4~6で示されるアミノ酸配列C1、又は該アミノ酸配列C1と80%以上(好ましくは85%以上、より好ましくは90%以上、さらに好ましくは95%以上、よりさらに好ましくは97%以上、特に好ましくは99%以上、また100%未満)の同一性を有するアミノ酸配列C2が挙げられる。
本発明の変異型ACE2タンパク質は、コロナウイルスSタンパク質に対する結合性が著しく損なわれない限りにおいて、アミノ酸配列C以外の他のアミノ酸配列、例えばタンパク質タグ、蛍光タンパク質、発光タンパク質、分泌シグナル配列(Igκシグナル配列等)、プロテアーゼ認識配列(TEVプロテアーゼ認識配列等)等のシグナル配列等のタンパク質又はペプチドが付加されたものであってもよい。タンパク質タグとしては、例えばビオチン、Hisタグ、FLAGタグ、Haloタグ、MBPタグ、HAタグ、Mycタグ、V5タグ、PAタグ等が挙げられる。
本発明の変異型ACE2タンパク質は、好ましい一態様において、抗体の全部又は一部のアミノ酸配列を含む。抗体としては、特に制限されず、例えばIgA、IgD、IgE、IgG、IgMなど、さらにはこれらのサブクラスが挙げられる。抗体の由来も特に制限されず、例えばヒト由来抗体、マウス由来抗体、ラット由来抗体、ウサギ由来抗体、サル由来抗体、チンパンジー由来抗体などであり得る。抗体の一部のアミノ酸配列を使用する場合、当該アミノ酸配列はFc領域を含むことが好ましい。これにより、エフェクター機能を発揮させることが可能である。また、Fc領域を介して本発明の変異型ACE2タンパク質を連結させて多量体化することもできる。この観点から、本発明は、その一態様において、本発明の変異型ACE2タンパク質を複数(例えば2つ、3つ、4つ、5つ)含む、複合体(本明細書において、「本発明の複合体」と示すこともある。)に関する。
本発明の変異型ACE2タンパク質は、コロナウイルスSタンパク質に対する結合性が著しく損なわれない限りにおいて、薬剤が連結したものであってもよい。
本発明の変異型ACE2タンパク質は、コロナウイルスSタンパク質に対する結合性が著しく損なわれない限りにおいて、化学修飾されたものであってもよい。
本発明の変異型ACE2タンパク質は、C末端がカルボキシル基(-COOH)、カルボキシレート(-COO-)、アミド(-CONH2)またはエステル(-COOR)の何れであってもよい。
ここでエステルにおけるRとしては、例えば、メチル、エチル、n-プロピル、イソプロピル、n-ブチルなどのC1-6アルキル基;例えば、シクロペンチル、シクロヘキシルなどのC3-8シクロアルキル基;例えば、フェニル、α-ナフチルなどのC6-12アリール基;例えば、ベンジル、フェネチルなどのフェニル-C1-2アルキル基;α-ナフチルメチルなどのα-ナフチル-C1-2アルキル基などのC7-14アラルキル基;ピバロイルオキシメチル基などが用いられる。
本発明の変異型ACE2タンパク質は、C末端以外のカルボキシル基(またはカルボキシレート)が、アミド化またはエステル化されていてもよい。この場合のエステルとしては、例えば上記したC末端のエステルなどが用いられる。
さらに、本発明の変異型ACE2タンパク質には、N末端のアミノ酸残基のアミノ基が保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイルなどのC1-6アシル基など)で保護されているもの、生体内で切断されて生成し得るN末端のグルタミン残基がピログルタミン酸化したもの、分子内のアミノ酸の側鎖上の置換基(例えば-OH、-SH、アミノ基、イミダゾール基、インドール基、グアニジノ基など)が適当な保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイル基などのC1-6アシル基など)で保護されているもの、あるいは糖鎖が結合したいわゆる糖蛋白質などの複合蛋白質なども包含される。
本発明の変異型ACE2タンパク質は、酸または塩基との塩の形態であってもよい。塩は、特に限定されず、酸性塩、塩基性塩のいずれも採用することができる。例えば酸性塩の例としては、塩酸塩、臭化水素酸塩、硫酸塩、硝酸塩、リン酸塩等の無機酸塩; 酢酸塩、プロピオン酸塩、酒石酸塩、フマル酸塩、マレイン酸塩、リンゴ酸塩、クエン酸塩、メタンスルホン酸塩、パラトルエンスルホン酸塩等の有機酸塩; アスパラギン酸塩、グルタミン酸塩等のアミノ酸塩等が挙げられる。また、塩基性塩の例として、ナトリウム塩、カリウム塩等のアルカリ金属塩; カルシウム塩、マグネシウム塩等のアルカリ土類金属塩等が挙げられる。
本発明の変異型ACE2タンパク質は、溶媒和物の形態であってもよい。溶媒は、特に限定されず、例えば水、エタノール、グリセロール、酢酸等が挙げられる。
本発明の変異型ACE2タンパク質は、公知の遺伝子工学的手法に従って容易に製造することができる。例えば、PCR、制限酵素切断、DNA連結技術、in vitro転写・翻訳技術、リコンビナントタンパク質作製技術等を利用して製造することができる。また、リコンビナントタンパク質作製技術としては、例えば、宿主として、培養細胞を採用する方法のみならず、タバコ等の植物を採用する方法等が挙げられる。
3.ポリヌクレオチド、細胞
本発明は、その一態様において、本発明の変異型ACE2タンパク質のコード配列を含む、ポリヌクレオチド(本明細書において、「本発明のポリヌクレオチド」と示すこともある。)、本発明のポリヌクレオチドを含む、細胞(本明細書において、「本発明の細胞」と示すこともある。)に関する。以下に、これらについて説明する。
本発明は、その一態様において、本発明の変異型ACE2タンパク質のコード配列を含む、ポリヌクレオチド(本明細書において、「本発明のポリヌクレオチド」と示すこともある。)、本発明のポリヌクレオチドを含む、細胞(本明細書において、「本発明の細胞」と示すこともある。)に関する。以下に、これらについて説明する。
本発明の変異型ACE2タンパク質のコード配列は、本発明の変異型ACE2タンパク質をコードする塩基配列からなるポリヌクレオチドである限り、特に制限されない。
本発明のポリヌクレオチドは、その一態様において、本発明の変異型ACE2タンパク質の発現カセットを含む。
本発明の変異型ACE2タンパク質の発現カセットは、細胞内で本発明の変異型ACE2タンパク質を発現可能なポリヌクレオチドである限り特に制限されない。本発明の変異型ACE2タンパク質の発現カセットの典型例としては、プロモーター、及びそのプロモーターの制御下に配置された本発明の変異型ACE2タンパク質のコード配列を含むポリヌクレオチドが挙げられる。
本発明の変異型ACE2タンパク質の発現カセットに含まれるプロモーターとしては、特に制限されず、対象細胞に応じて適宜選択することができる。プロモーターとしては、例えばpol II系プロモーターを各種使用することができる。pol II系プロモーターとしては、特に制限されないが、例えばCMVプロモーター、EF1プロモーター、SV40プロモーター、MSCVプロモーター等が挙げられる。その他にも、プロモーターとして、例えばtrcやtac等のトリプトファンプロモーター、lacプロモーター、T7プロモーター、T5プロモーター、T3プロモーター、SP6プロモーター、アラビノース誘導プロモーター、コールドショックプロモーター、テトラサイクリン誘導性プロモーター等が挙げられる。
本発明の変異型ACE2タンパク質の発現カセットは、必要に応じて、他のエレメント(例えば、マルチクローニングサイト(MCS)、薬剤耐性遺伝子、複製起点、エンハンサー配列、リプレッサー配列、インスレーター配列、レポータータンパク質(例えば、蛍光タンパク質等)コード配列、薬剤耐性遺伝子コード配列などが挙げられる。)を含んでいてもよい。
本発明のポリヌクレオチドは、ベクターの形態であることができる。使用目的(クローニング、タンパク質の発現)に応じて、また宿主細胞の種類を考慮して適当なベクターが選択される。大腸菌を宿主とするベクターとしてはM13ファージ又はその改変体、λファージ又はその改変体、pBR322又はその改変体(pB325、pAT153、pUC8など)等、酵母を宿主とするベクターとしてはpYepSec1、pMFa、pYES2、pPIC3.5K等、昆虫細胞を宿主とするベクターとしてはpAc、pVL等、哺乳類細胞を宿主とするベクターとしてはpcDNA、pCDM8、pMT2PC等を例示することができる。
本発明の細胞は、本発明のポリヌクレオチドを含む限り特に制限されない。細胞としては、例えば、Escherichia coli K12等の大腸菌、Bacillus subtilis MI114等のバチルス属細菌、Saccharomyces cerevisiae AH22等の酵母、Spodoptera frugiperda 由来の Sf細胞系もしくはTrichoplusia ni由来のHighFive細胞系、嗅神経細胞等の昆虫細胞、COS7細胞等の動物細胞等を挙げることができる。動物細胞としては、好ましくは、哺乳動物由来の培養細胞、具体的には、COS7細胞、CHO細胞、HEK293細胞、HEK293FT細胞、Hela細胞、PC12細胞、N1E-115細胞、SH-SY5Y細胞等が挙げられる。
本発明の細胞は、その一態様において、本発明の変異型ACE2タンパク質を発現している。例えば、本発明の細胞は、その一態様において、本発明の変異型ACE2タンパク質を分泌している、或いは本発明の変異型ACE2タンパク質を細胞表面上に有する。
4.用途
本発明の変異型ACE2タンパク質は、コロナウイルスのSタンパク質に対して結合することができる。このため、本発明の変異型ACE2タンパク質、本発明の複合体、本発明のポリヌクレオチド、及び本発明の細胞からなる群より選択される少なくとも1種は、医薬、試薬等(本明細書において、「本発明の薬剤」と示すこともある。)の有効成分として、より具体的には、例えば抗ウイルス剤等の有効成分としての利用が可能である。
本発明の変異型ACE2タンパク質は、コロナウイルスのSタンパク質に対して結合することができる。このため、本発明の変異型ACE2タンパク質、本発明の複合体、本発明のポリヌクレオチド、及び本発明の細胞からなる群より選択される少なくとも1種は、医薬、試薬等(本明細書において、「本発明の薬剤」と示すこともある。)の有効成分として、より具体的には、例えば抗ウイルス剤等の有効成分としての利用が可能である。
本発明の薬剤の対象コロナウイルスは、オルトコロナウイルス亜科に属するウイルスである限り、特に制限されない。コロナウイルスとしては、アルファコロナウイルス属、ベータコロナウイルス属、ガンマコロナウイルス属、デルタコロナウイルス属等が挙げられ、これらの中でも、ベータコロナウイルス属が好ましい。ベータコロナウイルス属としては、SARS関連コロナウイルス(SARSr-CoV)、コロナウイルスHKU1、MERSコロナウイルス等が挙げられ、これらの中でも、SARSr-CoVが好ましい。SARSr-CoVとしては、SARS-CoV-2、SARS-CoV-1等が挙げられ、これらの中でもSARS-CoV-2が好ましい。本発明の薬剤は、特にSARS-CoV-2に対して好適に使用することができる。また、SARS-CoV-2の中でも、武漢株、アルファ株、デルタ株、ラムダ株等の各種株に対して使用することができる。
本発明の薬剤によれば、コロナウイルスに対して抗ウイルス作用を発揮することができる。抗ウイルス作用としては、具体的には、ウイルス感染抑制作用、ウイルスによる細胞死を抑制する作用、ウイルス不活化作用、ウイルス増殖抑制、ウイルス出芽抑制、ウイルス抵抗性誘導作用等が挙げられる。また、この作用により、コロナウイルス感染症(特に、COVID-19)の予防又は治療剤として使用することもできる。
本発明の薬剤は、上記有効成分を含有する限りにおいて特に制限されず、必要に応じてさらに他の成分を含んでいてもよい。他の成分としては、薬学的に許容される成分であれば特に限定されるものではない。他の成分としては、薬理作用を有する成分のほか、添加剤も含まれる。添加剤としては、例えば基剤、担体、溶剤、分散剤、乳化剤、緩衝剤、安定剤、賦形剤、結合剤、崩壊剤、滑沢剤、増粘剤、保湿剤、着色料、香料、キレート剤等が挙げられる。
本発明の薬剤の使用態様は、特に制限されず、その種類に応じて適切な使用態様を採ることができる。本発明の薬剤は、その用途に応じて、例えばin vitroで使用する(例えば、培養細胞の培地に添加する。)こともできるし、in vivoで使用する(例えば、動物に投与する。)こともできる。
本発明の薬剤の適用対象は特に限定されないが、哺乳動物では、例えば、ヒト、サル、マウス、ラット、イヌ、ネコ、ウサギ、ブタ、ウマ、ウシ、ヒツジ、ヤギ、シカ等が挙げられる。また、細胞としては、動物細胞等が挙げられる。細胞の種類も特に制限されず、例えば血液細胞、造血幹細胞・前駆細胞、配偶子(精子、卵子)、線維芽細胞、上皮細胞、血管内皮細胞、神経細胞、肝細胞、ケラチン生成細胞、筋細胞、表皮細胞、内分泌細胞、ES細胞、iPS細胞、組織幹細胞、がん細胞等が挙げられる。
本発明の薬剤は、任意の剤形、例えば錠剤(口腔内側崩壊錠、咀嚼可能錠、発泡錠、トローチ剤、ゼリー状ドロップ剤などを含む)、丸剤、顆粒剤、細粒剤、散剤、硬カプセル剤、軟カプセル剤、ドライシロップ剤、液剤(ドリンク剤、懸濁剤、シロップ剤を含む)、ゼリー剤などの経口製剤形態、注射用製剤(例えば、点滴注射剤(例えば点滴静注用製剤等)、静脈注射剤、筋肉注射剤、皮下注射剤、皮内注射剤)、外用剤(例えば、軟膏剤、パップ剤、ローション剤)、坐剤、吸入剤、眼剤、眼軟膏剤、点鼻剤、点耳剤、リポソーム剤等の非経口製剤形態を採ることができる。
本発明の薬剤の投与経路としては、所望の効果が得られる限り特に制限されず、経口投与、経管栄養、注腸投与等の経腸投与; 経静脈投与、経動脈投与、筋肉内投与、心臓内投与、皮下投与、皮内投与、腹腔内投与等の非経口投与等が挙げられる。
本発明の薬剤中の有効成分の含有量は、使用態様、適用対象、適用対象の状態等に左右されるものであり、限定はされないが、例えば0.0001~100重量%、好ましくは0.001~50重量%とすることができる。
本発明の薬剤を動物に投与する場合の投与量は、薬効を発現する有効量であれば特に限定されず、通常は、有効成分の重量として、一般に経口投与の場合には一日あたり0.1~1000 mg/kg体重、好ましくは一日あたり0.5~500 mg/kg体重であり、非経口投与の場合には一日あたり0.01~100 mg/kg体重、好ましくは0.05~50 mg/kg体重である。上記投与量は、年齢、病態、症状等により適宜増減することもできる。
以下に、実施例に基づいて本発明を詳細に説明するが、本発明はこれらの実施例によって限定されるものではない。
実施例1.変異型ACE2タンパク質の発現ベクターの調製1
N末端側からIgκシグナル配列(配列番号7)、ヒトACE2(18-615)に7つの変異(A25V, K26E, K31N, E35K, N64I, L79F, N90H)が導入されてなる配列(配列番号5)、XbaI認識配列によりコードするアミノ酸配列(SR)、ヒトIgG1 Fc領域の配列(配列番号8)の順に連結されてなる変異型ACE2タンパク質(融合タンパク質)のコード配列(配列番号9)を含む発現ベクター(p3xFLAG-CMV-14由来:変異型ACE2発現ベクター1)を、公知のサブクローニング技術を利用して、作製した。

変異型ACE2発現ベクター1(実施例1)及び野生型ACE2発現ベクター1(比較例1)を用いて、ACE2タンパク質を発現及び精製した。具体的には、次のようにして行った。タンパク質の合成はExpi29(商標) Expression System(製造元:サーモフィッシャー、カタログ番号:A14635)を用いて行った。まず上記ベクタープラスミドをExpi293F細胞にExpifectamineを用いてトランスフェクションし、翌日にエンハンサーを加え5日後に培養上清を回収した。精製はまず培養上清20mLを4℃、4000gで10分間遠心し、0.45μmのフィルターを通した。そこにプロテインAセファロース(製造元:GEヘルスケア、カタログ番号:17-1279-03)を2mL、1M Tris-HCl pH9.0 を200μLを加え、室温で2時間ローテータで混和した。その後、ミニカラムに充填し、PBS 10mLで洗浄を4回行い、1mLのIgG Elute Buffer (製造元:サーモフィッシャー、カタログ番号:21004)で溶出し、0.1mLの1M Tris-HCl pH9.0で中和した。溶出物はPBSに対して透析し、限外ろ過デバイスAmicon Ultra-4 (30K MWCO)(製造元:メルクミリポア、カタログ番号:UFC803024)を用いて1~2 mg/mlに濃縮した後、-80℃で凍結保存した。

実施例4.ACE2タンパク質の発現精製2
変異型ACE2発現ベクター2(実施例2)及び野生型ACE2発現ベクター2(比較例2)を用いて、ACE2タンパク質を発現及び精製した。具体的には、次のようにして行った。タンパク質の合成はExpi293(商標) Expression System(製造元:サーモフィッシャー、カタログ番号:A14635)を用いて行った。まず上記ベクタープラスミドをExpi293F細胞にExpifectamineを用いてトランスフェクションし、翌日にエンハンサーを加え5日後に培養上清を回収した。精製はまず培養上清20mLを4℃、4000gで10分間遠心し、0.45μmのフィルターを通した。そこにNi-NTAアガロース(製造元:キアゲン、カタログ番号:30230)を1mL加え、室温で2時間ローテータで混和した。その後、ミニカラムに充填し、Wash Buffer(20mM Tris-HCl, pH7.5, 0.3 M NaCl, 20 mM imidazole) 5mLで洗浄を4回行い、1mLのElution Buffer (20mM Tris-HCl, pH7.5, 0.3 M NaCl, 250 mM imidazole) で溶出した。溶出物はPBSに対して透析し、限外ろ過デバイスAmicon Ultra-4 (30K MWCO)(製造元:メルクミリポア、カタログ番号:UFC803024)を用いて1~2 mg/mlに濃縮した後、-80℃で凍結保存した。

試験例1.Binging assay
実施例4で得られたACE2タンパク質を用いて、SARS-CoV-2 S タンパク質 Spike RBD-SD1-Fc(SARS-CoV-2スパイク蛋白質の319-591領域のC末端にヒトIgG1 Fcを融合した精製タンパク質)との結合性を、分子間相互作用解析装置(Biacore T200)を用いて解析した。解析条件は次の通りである:Sensor Chip CM5(製造元:GEヘルスケア、カタログ番号:BR100530)に、Human Antibody Capture Kit(製造元:GEヘルスケア、カタログ番号:BR100838)をもちいてRBD-SD1-Fcを固定化した。対照となるフローセルにはHuman Fcのみを同量固定化した。実施例4で得られたACE2タンパク質をRunning Buffer (0.05% Tween20を含むPBS)で0.1~300nMの濃度で希釈し、これを上記のフローセルの上に30 μL/minの流速で流し、1濃度あたり120 secのassociation timeに設定してSingle-Cycle kineticsモードで解析した。結合キネティクスはBiacore T200 evaluation software version 4.1を用いて計算した。その結果、野生型ACE2タンパク質の解離定数(KD)は41.4nMであるのに対して、変異型ACE2タンパク質の解離定数(KD)は0.37nMであった。
実施例4で得られたACE2タンパク質を用いて、SARS-CoV-2 S タンパク質 Spike RBD-SD1-Fc(SARS-CoV-2スパイク蛋白質の319-591領域のC末端にヒトIgG1 Fcを融合した精製タンパク質)との結合性を、分子間相互作用解析装置(Biacore T200)を用いて解析した。解析条件は次の通りである:Sensor Chip CM5(製造元:GEヘルスケア、カタログ番号:BR100530)に、Human Antibody Capture Kit(製造元:GEヘルスケア、カタログ番号:BR100838)をもちいてRBD-SD1-Fcを固定化した。対照となるフローセルにはHuman Fcのみを同量固定化した。実施例4で得られたACE2タンパク質をRunning Buffer (0.05% Tween20を含むPBS)で0.1~300nMの濃度で希釈し、これを上記のフローセルの上に30 μL/minの流速で流し、1濃度あたり120 secのassociation timeに設定してSingle-Cycle kineticsモードで解析した。結合キネティクスはBiacore T200 evaluation software version 4.1を用いて計算した。その結果、野生型ACE2タンパク質の解離定数(KD)は41.4nMであるのに対して、変異型ACE2タンパク質の解離定数(KD)は0.37nMであった。
試験例2.Pseudovirus neutralization assay
実施例3で得られたACE2タンパク質を用いて、SARS-CoV-2 S タンパク質を発現する偽SARS-CoV-2に対する中和活性を測定した。具体的には、次のようにして行った。PseudovirusはpLenti Firefly, psPAX2, pcDNA4 SARS2-Spikeの合計3つのプラスミドをLenti-X 293細胞(製造元:クローンテック、カタログ番号:632180)にFuGENE HD(製造元:プロメガ、カタログ番号:E2311)を用いてトランスフェクションし、48時間後の培養上清に分泌されたウイルス液を用いた。感染細胞はVeroE6/TMPRSS2細胞(製造元:国立感染症研究所、登録番号:JCRB1819)を用いた。ウイルス液50μLと実施例3で得られたACE2タンパク質を10μg/mLから1/3連続希釈したもの50μLを混和し1時間静置。その後予め96ウェルプレートに播種したVeroE6/TMPRSS2細胞に混和液を加え、1時間反応させた後に培地交換し、48時間後にルシフェラーゼ活性を測定した。ルシフェラーゼ活性はDual-Glo Luciferase Assay System(製造元:プロメガ、カタログ番号:E2920)を用いて、プレートリーダー(Tecan Infinite F200)で測定した。ウイルス液のみを反応させた細胞を100%感染コントロール、細胞のみを0%感染コントロールとして、シグナル比からinhibition rateを算出した。
実施例3で得られたACE2タンパク質を用いて、SARS-CoV-2 S タンパク質を発現する偽SARS-CoV-2に対する中和活性を測定した。具体的には、次のようにして行った。PseudovirusはpLenti Firefly, psPAX2, pcDNA4 SARS2-Spikeの合計3つのプラスミドをLenti-X 293細胞(製造元:クローンテック、カタログ番号:632180)にFuGENE HD(製造元:プロメガ、カタログ番号:E2311)を用いてトランスフェクションし、48時間後の培養上清に分泌されたウイルス液を用いた。感染細胞はVeroE6/TMPRSS2細胞(製造元:国立感染症研究所、登録番号:JCRB1819)を用いた。ウイルス液50μLと実施例3で得られたACE2タンパク質を10μg/mLから1/3連続希釈したもの50μLを混和し1時間静置。その後予め96ウェルプレートに播種したVeroE6/TMPRSS2細胞に混和液を加え、1時間反応させた後に培地交換し、48時間後にルシフェラーゼ活性を測定した。ルシフェラーゼ活性はDual-Glo Luciferase Assay System(製造元:プロメガ、カタログ番号:E2920)を用いて、プレートリーダー(Tecan Infinite F200)で測定した。ウイルス液のみを反応させた細胞を100%感染コントロール、細胞のみを0%感染コントロールとして、シグナル比からinhibition rateを算出した。
結果を図1に示す。図1に示されるとおり、変異型ACE2タンパク質は、野生型ACE2タンパク質に比べて遥かに高い中和活性を示した。
試験例3.Authentic virus neutralization assay 1
実施例3で得られたACE2タンパク質を用いて、SARS-CoV-2に対する中和活性を測定した。具体的には、次のようにして行った。24ウェルプレートにVeroE6/TMPRSS2細胞(製造元:国立感染症研究所、登録番号:JCRB1819)を8万細胞播種しておき、そこへmoi=0.1のSARS-CoV-2を改変ACE2タンパク質と共に感染させて2時間後にPBSで洗浄と培地交換を行った。22時間後に培養細胞のウイルスRNAをqPCRで、上清中のウイルス力価をプラークアッセイで測定した。
実施例3で得られたACE2タンパク質を用いて、SARS-CoV-2に対する中和活性を測定した。具体的には、次のようにして行った。24ウェルプレートにVeroE6/TMPRSS2細胞(製造元:国立感染症研究所、登録番号:JCRB1819)を8万細胞播種しておき、そこへmoi=0.1のSARS-CoV-2を改変ACE2タンパク質と共に感染させて2時間後にPBSで洗浄と培地交換を行った。22時間後に培養細胞のウイルスRNAをqPCRで、上清中のウイルス力価をプラークアッセイで測定した。
結果を図2及び3に示す。図2及び3に示されるとおり、変異型ACE2タンパク質は、野生型ACE2タンパク質に比べて遥かに高い中和活性を示した。
試験例4.必須アミノ酸の同定
ヒトACE2(18-615)に7つの変異が導入されてなる配列(配列番号5)を含む変異型ACE2タンパク質について、一部の変異を野生型に戻した変異体を作製し、そのSARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、A25V、K31N、E35K、及びL79Fの4つの変異が重要であることが分かった。ヒトACE2(18-615)に当該4つの変異が導入されてなる配列を含む変異型ACE2タンパク質について、解離定数(KD)(測定方法は試験例1と同様)は0.64nMであり、SARS-CoV-2 Pseudovirusに対するIC50(測定方法は試験例2と同様)は0.082μg/mlであった。

試験例5.他の変異型ACE2タンパク質の解析1
ヒトACE2(18-615)に6つの変異(K31M, E35K, Q60R, S70F, L79F, N90D)が導入されてなる配列を含む変異型ACE2タンパク質について、SARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、高い結合性を示すことが分かった。また、該変異型ACE2タンパク質について、一部の変異を野生型に戻した変異体を作製し、そのSARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、K31M, E35K, 及びQ60Rの3つの変異(より好ましくはさらにL79Fも加えた4つの変異)が重要であることが分かった。ヒトACE2(18-615)に当該4つの変異が導入されてなる配列を含む変異型ACE2タンパク質について、解離定数(KD)(測定方法は試験例1と同様)は1.14nMであり、SARS-CoV-2 Pseudovirusに対するIC50(測定方法は試験例2と同様)は0.33μg/mlであった。
ヒトACE2(18-615)に6つの変異(K31M, E35K, Q60R, S70F, L79F, N90D)が導入されてなる配列を含む変異型ACE2タンパク質について、SARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、高い結合性を示すことが分かった。また、該変異型ACE2タンパク質について、一部の変異を野生型に戻した変異体を作製し、そのSARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、K31M, E35K, 及びQ60Rの3つの変異(より好ましくはさらにL79Fも加えた4つの変異)が重要であることが分かった。ヒトACE2(18-615)に当該4つの変異が導入されてなる配列を含む変異型ACE2タンパク質について、解離定数(KD)(測定方法は試験例1と同様)は1.14nMであり、SARS-CoV-2 Pseudovirusに対するIC50(測定方法は試験例2と同様)は0.33μg/mlであった。
試験例6.他の変異型ACE2タンパク質の解析2
ヒトACE2(18-615)に6つの変異(T20I, A25V, H34A, T78R, T92Q, Q101H)が導入されてなる配列を含む変異型ACE2タンパク質について、SARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、高い結合性を示すことが分かった。また、該変異型ACE2タンパク質について、一部の変異を野生型に戻した変異体を作製し、そのSARS-CoV-2スパイク蛋白質に対する結合性について調べたところ、T20I, H34A, T92Q, 及びQ101Hの4つの変異が重要であることが分かった。ヒトACE2(18-615)に当該4つの変異が導入されてなる配列を含む変異型ACE2タンパク質について、解離定数(KD)(測定方法は試験例1と同様)は3.98nMであり、SARS-CoV-2 Pseudovirusに対するIC50(測定方法は試験例2と同様)は0.068μg/mlであった。

試験例7.Authentic virus neutralization assay 2
ヒトACE2(18-615)に7つの変異(A25V, K26E, K31N, E35K, N64I, L79F, N90H)が導入されてなる配列を含む変異型ACE2タンパク質(3N39)、ヒトACE2(18-615)に4つの変異(A25V, K31N, E35K, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(3N39v2)、ヒトACE2(18-615)に4つの変異(K31M, E35K, Q60R, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(3J113v2)、及びヒトACE2(18-615)に4つの変異(T20I, H34A, T92Q, Q101H)が導入されてなる配列を含む変異型ACE2タンパク質(3J120v2)について、試験例3と同様にして、SARS-CoV-2に対する中和活性を測定した。
ヒトACE2(18-615)に7つの変異(A25V, K26E, K31N, E35K, N64I, L79F, N90H)が導入されてなる配列を含む変異型ACE2タンパク質(3N39)、ヒトACE2(18-615)に4つの変異(A25V, K31N, E35K, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(3N39v2)、ヒトACE2(18-615)に4つの変異(K31M, E35K, Q60R, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(3J113v2)、及びヒトACE2(18-615)に4つの変異(T20I, H34A, T92Q, Q101H)が導入されてなる配列を含む変異型ACE2タンパク質(3J120v2)について、試験例3と同様にして、SARS-CoV-2に対する中和活性を測定した。
結果を図4に示す。図4に示されるとおり、変異型ACE2タンパク質は、野生型ACE2タンパク質に比べて遥かに高い中和活性を示した。
試験例8.変異型ACE2タンパク質の安定性向上
ヒトACE2(18-615)に7つの変異(A25V, K26E, K31N, E35K, N64I, L79F, N90H)が導入されてなる配列を含む変異型ACE2タンパク質(3N39)、及びヒトACE2(18-615)に4つの変異(A25V, K31N, E35K, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(3N39v2)において、closed conformationに固定するための変異(S128C及びV343C)を導入してなる変異型ACE2タンパク質(3N39-SS、3N39v2-SS)を調製した。得られた変異型ACE2タンパク質について、Thermal shift assayを行った。具体的な方法は以下の通りである。
ヒトACE2(18-615)に7つの変異(A25V, K26E, K31N, E35K, N64I, L79F, N90H)が導入されてなる配列を含む変異型ACE2タンパク質(3N39)、及びヒトACE2(18-615)に4つの変異(A25V, K31N, E35K, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(3N39v2)において、closed conformationに固定するための変異(S128C及びV343C)を導入してなる変異型ACE2タンパク質(3N39-SS、3N39v2-SS)を調製した。得られた変異型ACE2タンパク質について、Thermal shift assayを行った。具体的な方法は以下の通りである。
精製した変異型ACE2タンパク質をPBSで200μg/mlに希釈し、0.2mL白色PCRチューブ(Bio-Rad、TLS0851)に20μl/tubeで入れた。水で1:150に希釈したSYPR(商標)オレンジタンパク質ゲル染色液(Invitrogen、S6651)を1μl/チューブに添加した後、チューブをBio-Rad CFX96サーマルサイクラーリアルタイム検出システムに入れた。25℃から95℃までの熱変性曲線(測定前に各温度で5秒の平衡化で0.5℃間隔で1.27℃/分のランプレート)は、450から490 nmの励起と560から580 nmの検出を持つFRETチャネルを使用して蛍光強度を測定することによって取得した。すべてのデータをエクスポートし、Microsoft Excelでプロットし、1次微分法を用いてTmを算出した。
その結果、野生型ヒトACE2(18-615)は、closed conformationに固定するための変異(S128C及びV343C)の導入により、Tmが44.5℃から57℃に上昇した。また、3N39は、closed conformationに固定するための変異(S128C及びV343C)の導入により、Tmが44.5℃から51.5℃に上昇した。さらに、3N39v2は、closed conformationに固定するための変異(S128C及びV343C)の導入により、Tmが44.0℃から55.5℃に上昇した。また、変異(S128C及びV343C)の導入により、中和活性の低下は見られなかった。
試験例9.耐性株発生の評価
抗ウイルス療法においては、病原ウイルスが頻繁に変異を起こすことで薬剤耐性を獲得することが一般的に懸念されている。実際に、SARS-CoV-2は中和抗体を用いて培養することで急速に耐性株が発生することが報告されている。そこで、SARS-CoV-2ウイルスを用いて耐性株の発生を評価した。
抗ウイルス療法においては、病原ウイルスが頻繁に変異を起こすことで薬剤耐性を獲得することが一般的に懸念されている。実際に、SARS-CoV-2は中和抗体を用いて培養することで急速に耐性株が発生することが報告されている。そこで、SARS-CoV-2ウイルスを用いて耐性株の発生を評価した。
方法の概要を図5aに示す。最初の継代では、変異体 ACE2-Fc または回復期患者から単離した組換えモノクローナル抗体(クローン H4)の希釈系列の培養液に0.1 MOI ウイルスを添加し、部分的に中和されたウェルから増幅されたウイルスの合計 3 x 105 コピーを次の継代に移した(図5a)。各ウェルから上清を採取し、定量PCRによりウイルスRNAコピー数を解析した。
これまでの報告と同様に、単一抗体で処理した後のウイルスプールは、4回の継代で同一抗体の最高濃度に対して不感応となり(図5b)、また、耐性株の配列決定により、F490VがH4抗体の結合を阻害していることが明らかになった。一方、変異体ACE2-Fcは、15回の継代後もウイルスプールに対して非常に高い中和能を維持しており、耐性株が出現していないことを示していた(図5c)。
試験例10.治療効果の評価
4週齢の雄のシリアハムスターに、0.75 mg kg-1メデトミジン(明治製菓)、2 mg kg-1ミダゾラム(サンドーズ)、2.5 mg kg-1ブトルファノール酒石酸塩(明治製菓)を腹腔内投与して麻酔をかけ、1.0×106 PFUのSARS-CoV-2(60μL中)を経鼻投与した。感染後2時間後にControl-Fc(20mg kg-1)またはACE2-Fc(3N39v2、20mg kg-1)を腹腔内注射により投与した。体重は5日間毎日モニターした。感染後5日目に全動物を安楽死させ、病理組織学的検査、ウイルス滴定およびqRT-PCRのために肺を採取した。肺の構造は、マイクロコンピュータ断層撮影装置(μCT)(ScanXmate-RB080SS110, Comscantecno Co.,Ltd.)を用いて観察した。μCTの画像をImageJソフトウェアで再構成し、可視化した。
4週齢の雄のシリアハムスターに、0.75 mg kg-1メデトミジン(明治製菓)、2 mg kg-1ミダゾラム(サンドーズ)、2.5 mg kg-1ブトルファノール酒石酸塩(明治製菓)を腹腔内投与して麻酔をかけ、1.0×106 PFUのSARS-CoV-2(60μL中)を経鼻投与した。感染後2時間後にControl-Fc(20mg kg-1)またはACE2-Fc(3N39v2、20mg kg-1)を腹腔内注射により投与した。体重は5日間毎日モニターした。感染後5日目に全動物を安楽死させ、病理組織学的検査、ウイルス滴定およびqRT-PCRのために肺を採取した。肺の構造は、マイクロコンピュータ断層撮影装置(μCT)(ScanXmate-RB080SS110, Comscantecno Co.,Ltd.)を用いて観察した。μCTの画像をImageJソフトウェアで再構成し、可視化した。
コントロール-Fcを投与されたハムスターは体重が4.3%減少したのに対し、3N39v2-Fcを投与されたハムスターは感染5日後にコントロールと同様に7.3%増加した(図6b)。剖検前のmicro-CTでは、対照群では頭蓋側を中心に多葉性のグランドガラス混濁が認められたが、治療群では肺の異常は限定的であった(図6c)。肺におけるSARS-CoV-2の含有量を機能性ウイルス粒子とゲノムRNAコピー数で評価したところ、3N39v2-Fcを投与した群では、いずれも有意に減少していることが明らかになった(図6d)。また、感染したハムスターの病理組織学的解析も行った。対照群では、炎症細胞の浸潤、肺胞中隔肥厚、肺胞出血を特徴とする重篤な間質性肺炎が認められたが、3N39v2-Fc投与群では、肺の病態やウイルス抗原が明らかに減少していた(図6e~f)。これらの結果と一致するように、炎症性サイトカインまたはケモタクチックサイトカインの発現は、治療群では減少した(図6g)。
試験例11.他の変異型ACE2タンパク質の解析3
以下の6つのACE2タンパク質を作製した。
・ヒトACE2(18-615)のACE2タンパク質(615 WT)、
・ヒトACE2(18-615)にclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(615 WT-S128C/V343C)、
・ヒトACE2(18-615)に4つの変異(A25V, K31N, E35K, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(615 3N39v2)、
・3N39v2にclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(615 3N39v2-S128C/V343C)、
・ヒトACE2(18-740)のACE2タンパク質(740 WT)、及び
・ヒトACE2(18-740)に、4つの変異(A25V, K31N, E35K, L79F)、及びclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(740 3N39v2-S128C/V343C)。
以下の6つのACE2タンパク質を作製した。
・ヒトACE2(18-615)のACE2タンパク質(615 WT)、
・ヒトACE2(18-615)にclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(615 WT-S128C/V343C)、
・ヒトACE2(18-615)に4つの変異(A25V, K31N, E35K, L79F)が導入されてなる配列を含む変異型ACE2タンパク質(615 3N39v2)、
・3N39v2にclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(615 3N39v2-S128C/V343C)、
・ヒトACE2(18-740)のACE2タンパク質(740 WT)、及び
・ヒトACE2(18-740)に、4つの変異(A25V, K31N, E35K, L79F)、及びclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(740 3N39v2-S128C/V343C)。
これらのタンパク質の収量を比較した。結果を図7に示す。
また、試験例2と同様にしてSARS-CoV-2 S タンパク質を発現する偽SARS-CoV-2(SARS-CoV-2 Pseudovirus)に対する中和活性を測定した。結果を図8に示す。615 3N39v2のIC50は0.064μg/mL、615 3N39v-S128C/V343CのIC50は0.056μg/mL、740 3N39v-S128C/V343CのIC50は0.033μg/mLであった。
試験例12.他の変異型ACE2タンパク質の解析4
ACE2のアミノ酸変異の免疫原性試験を行った結果、L79F変異を除いた方が、免疫原性が低くなることを見出した。そこで、ヒトACE2(18-740)に、4つの変異(A25V, K31N, E35K, L79F)、及びclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(740 3N39v2-S128C/V343C(本試験例では、3N39v2-SS、740-SS 3N39v2、又はv2と示すこともある)、試験例11で作製)において、F79F変異を除いてなる変異型ACE2タンパク質(A25V, K31N, E35K, S128C, V343C)(本試験例では、3N39v3-SS、740-SS 3N39v3、又はv3と示すこともある)、及びF79F変異の代わりにT92Q変異を導入してなる変異型ACE2タンパク質(A25V, K31N, E35K, T92Q, S128C, V343C)(本試験例では、3N39v4-SS、740-SS 3N39v4、v4、3N39v4-SS(740)-Fc、3N39v4-SS(740)、又は3N39v4-Fcと示すこともある)を作製した。
ACE2のアミノ酸変異の免疫原性試験を行った結果、L79F変異を除いた方が、免疫原性が低くなることを見出した。そこで、ヒトACE2(18-740)に、4つの変異(A25V, K31N, E35K, L79F)、及びclosed conformationに固定するための変異(S128C及びV343C)が導入されてなる配列を含む変異型ACE2タンパク質(740 3N39v2-S128C/V343C(本試験例では、3N39v2-SS、740-SS 3N39v2、又はv2と示すこともある)、試験例11で作製)において、F79F変異を除いてなる変異型ACE2タンパク質(A25V, K31N, E35K, S128C, V343C)(本試験例では、3N39v3-SS、740-SS 3N39v3、又はv3と示すこともある)、及びF79F変異の代わりにT92Q変異を導入してなる変異型ACE2タンパク質(A25V, K31N, E35K, T92Q, S128C, V343C)(本試験例では、3N39v4-SS、740-SS 3N39v4、v4、3N39v4-SS(740)-Fc、3N39v4-SS(740)、又は3N39v4-Fcと示すこともある)を作製した。
上記タンパク質の収量を比較した。結果を図9に示す。
また、上記タンパク質について、試験例2と同様にしてSARS-CoV-2(武漢株、アルファ株、デルタ株、又はラムダ株) S タンパク質を発現する偽SARS-CoV-2(SARS-CoV-2 Pseudovirus)に対する中和活性を測定した。結果を図10に示す。
続いて、上記タンパク質について、試験例3と同様にしてSARS-CoV-2(武漢株、アルファ株、デルタ株、又はラムダ株)に対する中和活性を測定した。結果を図11に示す。
続いて、上記タンパク質について、試験例8と同様にして熱構造安定性を測定した。その結果、v2のTmは56.5℃であり、v3のTmは58.0℃であり、v4のTmは58.5℃であった。
続いて、上記タンパク質について、ACE2酵素活性を測定した。具体的には、次のようにして行った。ACE2により切断されると蛍光を生じるFRETペプチドと各種変異型ACE2を反応させたのちに蛍光強度を測定し酵素活性を算出した。結果を図12に示す。
続いて、上記タンパク質について、薬物動態を測定した。具体的には、次のようにして行った。マウスに10mg/kg bodyのタンパク質を経静脈投与し、各時間経過毎に採血を施行。その血漿サンプルのタンパク質濃度をELISAで測定した。結果を図13に示す。
続いて、上記タンパク質について、試験例10と同様にして治療効果を評価した。肺組織のウイルス量の測定結果を図14に示し、肺組織の炎症性サイトカイン発現量の測定結果を図15に示す。

- 前記変異aにおいて、A25の変異後のアミノ酸が分岐鎖アミノ酸であり、K26の変異後のアミノ酸が酸性アミノ酸であり、K31の変異後のアミノ酸が親水性中性アミノ酸であり、E35の変異後のアミノ酸が塩基性アミノ酸であり、N64の変異後のアミノ酸が分岐鎖アミノ酸であり、L79の変異後のアミノ酸が芳香族アミノ酸であり、且つ/或いはN90の変異後のアミノ酸が塩基性アミノ酸である、請求項2~6のいずれかに記載の変異型ACE2タンパク質。
- 前記変異aにおいて、A25の変異後のアミノ酸がバリンであり、K26の変異後のアミノ酸がグルタミン酸であり、K31の変異後のアミノ酸がアスパラギンであり、E35の変異後のアミノ酸がリジンであり、N64の変異後のアミノ酸がイソロイシンであり、L79の変異後のアミノ酸がフェニルアラニンであり、且つ/或いはN90の変異後のアミノ酸がヒスチジンである、請求項2~7のいずれかに記載の変異型ACE2タンパク質。
- 前記置換axが、K31、E35、Q60、S70、L79、及びN90からなる群より選択される少なくとも2種のアミノ酸の置換、或いはT20、A25、H34、T78、T92、及びQ101からなる群より選択される少なくとも2種のアミノ酸の置換を含む、請求項1に記載の変異型ACE2タンパク質。
- 前記置換axが、K31、E35、Q60、及びL79の置換を含む、或いはT20、H34、T92、及びQ101Hの置換を含む、請求項1又は9に記載の変異型ACE2タンパク質。
- 前記アミノ酸配列Bが、配列番号2で示されるアミノ酸配列B1、配列番号3で示されるアミノ酸配列中の全部又は一部のアミノ酸配列B2、又はアミノ酸配列A、B1、若しくはB2のオルソログアミノ酸配列B3である、請求項1~10のいずれかに記載の変異型ACE2タンパク質。
- さらに、抗体の全部又は一部のアミノ酸配列を含む、請求項1~11のいずれかに記載の変異型ACE2タンパク質。
- 請求項1~12のいずれかに記載の変異型ACE2タンパク質を複数含む、複合体。
- 請求項1~12のいずれかに記載の変異型ACE2タンパク質のコード配列を含む、ポリヌクレオチド。
- 請求項14に記載のポリヌクレオチドを含む、細胞。
- 請求項1~12のいずれかに記載の変異型ACE2タンパク質、請求項13に記載の複合体、請求項14に記載のポリヌクレオチド、及び請求項15に記載の細胞からなる群より選択される少なくとも1種を含有する、医薬。
- 請求項1~12のいずれかに記載の変異型ACE2タンパク質、請求項13に記載の複合体、請求項14に記載のポリヌクレオチド、及び請求項15に記載の細胞からなる群より選択される少なくとも1種を含有する、試薬。
- 請求項1~12のいずれかに記載の変異型ACE2タンパク質、請求項13に記載の複合体、請求項14に記載のポリヌクレオチド、及び請求項15に記載の細胞からなる群より選択される少なくとも1種を含有する、抗コロナウイルス剤。
