Abstract

설포라핀의 함량을 증대시킨 내복자 추출물의 제조방법, 내복자의 가공방법, 상기 방법을 통하여 수득한 설포라핀의 함량이 증대된 내복자 추출물, 및 상기 내복자 추출물을 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 치료용 약학적 조성물, 식품 조성물 및 예방 또는 개선용 사료 첨가제 조성물에 관한 것이다. 일 양상의 방법은 최적의 로스팅 온도 조건을 통하여 수분에 취약한 설포라핀의 안정성을 유지할 수 있는 내복자의 수분함량을 가질수 있도록 가공할 수 있으므로, 내복자로부터 설포라핀을 다량 포함하는 추출물을 제조할 수 있다. 또한 상기 추출물은 통상의 방법에 의한 추출방법보다 설포라핀의 함량이 증대되어 있으므로 비만, 지방간, 또는 대사증후군의 예방, 개선 또는 치료를 위한 약학, 식물 및 사료 첨가제 조성물로 널리 활용될 수 있다.

Description

설포라핀이 증대된 내복자 추출물을 포함하는 조성물 및 이의 제조방법
설포라핀의 함량을 증대시킨 내복자 추출물의 제조방법, 내복자의 가공방법, 상기 방법을 통하여 수득한 설포라핀의 함량이 증대된 내복자 추출물, 및 상기 내복자 추출물을 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 치료용 약학적 조성물, 식품 조성물 및 예방 또는 개선용 사료 첨가제 조성물에 관한 것이다.
십자화과 식물은 외부의 위해(곤충, 박테리아, 곰팡이 등)로부터 방어하기 위한 수단으로 아이소싸이오사이아네이트를 분비한다. 아이소싸이오사이아네이트는 우수한 생리활성 기능을 갖고 있는 것으로 알려져 있다. 십자화과 식물은 글루코시놀레이트-미로시나아제 시스템이라 불리는 기작으로 아이소싸이오사이아네이트를 생산한다. 미로시나아제는 글루코시놀레이트를 아이소싸이오사이아네이트로 전환하는 효소로 미로신 세포의 액포에서 발견된다. 조직 내에서 미로시나아제는 글루코시놀레이트와 분리되어 있으나, 세포 손상과 같은 외부자극에 의해 아이소싸이오사이아네이트 전환 반응이 시작된다.
대표적인 십자화과 식물인 무의 씨앗은 오래전부터 한약재로 활용되어 왔으며, 내복자라 지칭되어 왔다. 설포라핀은 대표적인 무 씨앗 유래의 아이소싸이오사이아네이트로 무에서 가장 많은 양이 관찰된다.
서구화된 식습관과 운동량 감소로 인해 현재 비만 인구 비율은 전 세계적으로 급증하고 있는 추세이다. 비만과 과체중은 세계적으로 만연하고 있어서 조사 대상으로 한 국가(미국, 영국, 프랑스, 독일, 이탈리아, 스페인, 일본)의 인구 중 적어도 20%가, 또 많은 곳에서는 최대 45%가 과체중으로 알려져 있다. 비만은 그 자체의 문제뿐만 아니라 고지혈증, 당뇨, 지방간 등의 대사성 질환을 유발하는 주된 원인이 되기 때문에 체중을 조절하는 것 이 건강유지에 매우 중요하다. 현재까지 개발된 비만 치료제들은 소화계 또는 신경계에 작용을 하는 기전을 가지고 있어 여러 부작용들이 보고되어 있으므로, 천연물에서 유래된 활성성분으로 비만 및 대사증후군을 예방하거나 치료하기 위한 연구가 활발히 진행되고 있다.
이러한 기술적 배경 하에서, 다량의 설포라핀을 내복자로부터 수득하기 위한 방법을 연구중에 있으나, 아직은 미비한 실정이다.

이하, 본 발명을 보다 상세하게 설명한다.
본 발명에서 사용되는 모든 기술용어는, 달리 정의되지 않는 이상, 본 발명의 관련 분야에서 통상의 당업자가 일반적으로 이해하는 바와 같은 의미로 사용된다. 또한, 본 명세서에는 바람직한 방법이나 시료가 기재되나, 이와 유사하거나 동등한 것들도 본 발명의 범주에 포함된다. 본 명세서에 참고문헌으로 기재되는 모든 간행물의 내용은 본 발명에 전체가 참고로 통합된다.
상기 내복자는 십자화과의 무(Raphanus sativus L.) 의 씨를 말하며, 무의 씨 및 무씨와 본 명세서 내에서 혼용하여 사용될 수 있다.
상기 수분함량을 조절하는 단계는 원하는 최종적인 내복자의 수분함량을 갖도록 가공하는 과정을 의미한다. 상기 가공과정에서 필요한 경우 로스팅 과정 및/또는 가수 등의 공정 과정을 추가할 수 있다. 따라서, 일 양상의 방법은 수분함량을 조절하기 위하여 내복자를 내복자를 로스팅(roasting)하는 단계를 포함할 수 있으며, 상기 로스팅은 내복자의 품온이 약 40 ℃이하가 되도록 높은 건열을 사용해 수분을 더하지 않고 가열하는 과정을 의미할 수 있다. 더욱 구체적으로 일 양상의 방법에 있어서 상기 로스팅은 상기 내복자의 품온이 예를 들면 25 내지 40℃가 되도록 수행될 수 있다.
또한 상기 방법에 있어서, 내복자를 분쇄하는 단계는 믹서리를 이용하여 내복자의 평균입도(D[4,3])가 예를 들어 5 내지 50 μm, 5 내지 40 μm, 5 내지 35 μm, 5 내지 30 μm, 8 내지 25 μm, 7 내지 20 μm, 또는 10 내지 15 μm를 갖도록 분쇄하는 것을 포함할 수 있다.
또한 상기 방법의 수분함량을 조절하는 단계는 내복자의 수분함량이 예를 들면, 3 내지 20%(w/w), 5 내지 15%(w/w), 5 내지 10%(w/w), 6 내지 8%(w/w), 또는 7 %(w/w) 가 되도록 조절하는 것일 수 있다. 내복자의 수분 함량이 3% 전후로 낮은 경우에는 미로시나아제가 글루코시놀레이트를 가수분해 하기에 충분하지 못한 수분함량이므로 내복자 추출물 내 설포라핀의 함량이 낮을 수 있으며, 내복자의 수분함량이 20%이상인 경우에는 물에 의한 설포라핀의 파괴가 이루어져서 추출물 내 최종적으로 포함된 설포라핀의 함량이 급격히 감소할 수 있다.
상기 방법에 있어서, 추출 용매는 친수성 용매 (hydrophilic solvent)로 하여 추출된 것일 수 있다. 상기 친수성 용매는 물, C1 내지 C4의 알코올 또는 이들의 혼합물일 수 있다. 상기 알코올은 C1 내지 C4의 하나 이상의 -OH기를 갖는 화합물일 수 있다. 상기 알코올은 예를 들면 메탄올, 에탄올, n-프로판올, 이소프로판올, n-부탄올, sec-부탄올, 이소부탄올, tert-부탄올 또는 이들의 혼합물일 수 있다. 일 구체예에서, 상기 내복자의 추출용매는 에탄올 또는 에탄올과 물의 혼합물일 수 있으며, 추출용매 중 에탄올의 포함 비율은 예를 들면, 10 내지 30% (v/v)의 에탄올, 15 내지 25% (v/v) 의 에탄올 또는 20 % (v/v)의 에탄올일 수 있다.
상기 추출은 내복자를 교반 추출하는 것일 수 있다. 상기 내복자는 건조되거나 세척된 것일 수 있다. 상기 추출은 예를 들면, 10 내지 40℃, 15 내지 35℃, 17 내지 25℃, 20 내지 30 ℃의 온도에서 수행될 수 있다.
상기 추출 시간은 추출 조건에 따라 적절하게 선택할 수 있다. 상기 추출 시간은 예를 들면 1 내지 10시간, 2 내지 9시간, 2 내지 5 시간, 또는 2 내지 4시간일 수 있다. 상기 추출 용매는 내복자의 1 내지 15배, 예를 들면, 5 내지 13배, 5 내지 11배, 8 내지 15배, 8 내지 13배, 8 내지 11배, 또는 9 내지 11배일 수 있다. 상기 추출은 1회 이상, 예를 들면 1 내지 5회, 1 내지 4회, 1 내지 3회, 2 내지 5회, 또는 2 내지 4회 추출되는 것일 수 있다. 얻어진 조추출물은 여과, 농축 및 건조될 수 있다.
일 양상을 통해 내복자의 수분함량이 조절된 가공된 내복자를 평균입도가 10 내지 15 μm가 되도록 분쇄된 내복자를 활용하는 경우 추출물 내 설포라핀의 함량이 증대되는 것을 구체적으로 본 명세서에서 확인하였다.

상기 추출물은 추출물 전체 중량에 대하여 설포라핀의 함량이 추출물 전제 고형분 1g 중량을 기준으로 예를 들면 최소 39.9mg 이상을 포함할 수 있다.
상기 내목자 추출물은 감압 증류 및 동결 건조 또는 분무 건조 등과 같은 추가적인 과정에 의해 분말 상태로 제조될 수 있다.
본 명세서에서 '추출물'은, 광의적으로는 추출물을 추가적으로 분획(Fractionation)한 분획물의 의미도 포함한다. 즉, 내복자 추출물은 상술한 추출용매를 이용하여 얻은 것뿐만 아니라, 여기에 정제과정을 추가적으로 적용하여 얻은 것도 포함한다. 예를 들어, 상기 추출물을 일정한 분자량 컷-오프 값을 갖는 한외 여과막을 통과시켜 얻은 분획, 다양한 크로마토그래피(크기, 전하, 소수성 또는 친화성에 따른 분리를 위해 제작된 것)에 의한 분리 등 추가적으로 실시된 다양한 정제 방법을 통해 얻어진 분획도 본 발명의 내복자 추출물에 포함되는 것이다. 상기 분획물은 상기 조추출물에 대하여 특정 성분을 포함하는 물질을 분리한 것을 말한다. 상기 분리는 유기 용매를 사용한 분리, 크로마토그래피 또는 여과일 수 있다. 상기 크로마토그래피는 박층 크로마토그래피, 흡착 크로마토그래피, 분배 크로마토그래피, 이온교환 크로마토그래피, 친화성 크로마토그래피, 크기배제 크로마토그래피, HPLC, 또는 그 조합일 수 있다.

상기 대사증후군은 고혈압, 동맥경화, 고지혈증, 간질환, 과인슐린혈증, 과혈당증, 1형 및 2형 당뇨병, 인슐린 내성을 특징으로 하는 이상지혈증, 발기부전, 심혈관계 질환 및 허혈성 질환으로 이루어진 군에서 선택된 1종 이상이고, 상기 간질환은 알코올성 및 비알콜성 간의 지질대사 이상으로 인한 지방간, 간염 및 간경화를 포함할 수 있으나, 이에 제한되지 않는다.
상기 비만, 지방간, 또는 대사증후군은 내복자 추출물 이외에 약학적으로 허용 가능한 담체 즉 식염수, 멸균수, 링거액, 완충 식염수, 사이클로덱스트린, 덱스트로즈 용액, 말토덱스트린 용액, 글리세롤, 에탄올, 리포좀 및 이들 성분 중 1종 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액 등 다른 통상의 첨가제를 더 포함할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및/또는 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 더 나아가 당해 기술분야의 적정한 방법으로 또는 레밍턴의 문헌에 개시되어 있는 방법을 이용하여 각 성분에 따라 바람직하게 제제화할 수 있다. 상기 약학적 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀전, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 또는 멸균 주사용액의 형태로 제형화하여 사용될 수 있다.
일 양상에 따른 약학적 조성물의 투여방법은 특별히 제한되는 것은 아니나, 목적하는 방법에 따라 정맥내, 피하, 복강 내, 흡입 또는 국소적용과 같이 비경구 투여하거나 경구 투여할 수 있다. 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하다. 일일 투여량은 치료를 필요로 하는 개체에 투여됨으로서 경감된 질병 상태에 대한 치료에 충분한 일 양상에 따른 치료용 물질의 양을 의미한다. 치료용 물질의 효과적인 양은 특정 화합물, 질병 상태 및 그의 심각도, 치료를 필요로 하는 개체에 따라 달라지며, 이는 당업자에 의해 통상적으로 결정될 수 있다. 비제한적 예로서, 일 양상에 따른 조성물의 인체에 대한 투여량은 환자의 나이, 몸무게, 성별, 투여 형태, 건강 상태 및 질환 정도에 따라 달라질 수 있다. 일예로, 본 발명의 비만, 지방간, 또는 대사증후군의 예방 또는 치료용 약학적 조성물은 약학적 조성물을 기준으로 할 때, 0.0001 mg/kg 내지 1000 mg/kg으로, 보다 효과적이기 위해서는 0.01mg/kg 내지 100 mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
'개체'란 암, 혈관질환, 또는 염증질환의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로 인간 또는 비-인간인 영장류, 생쥐(mouse), 쥐(rat), 개, 고양이, 말 및 소 등의 포유류를 의미한다.
본원에 사용되는 바와 같이, "치료" 또는 "치료하는" 또는 "완화하는" 또는 "개선하는"은 상호교환 가능하게 사용된다. 이들 용어는 치료 이익 및/또는 예방 이익을 포함하나 이들에 한정되지 않는 유리한 또는 요망되는 결과를 수득하는 방법을 지칭한다. 치료 이익은 치료 하의 하나 이상의 질병, 질환 또는 증상의 임의의 치료적으로 유의미한 개선 또는 그에 대한 효과를 의미한다. 예방 이익에 있어서, 조성물은 특정 질병, 질환 또는 증상이 발생할 위험이 있는 대상체에게 또는 질병, 질환 또는 증상이 아직 나타나지 않을지라도, 질병의 하나 이상의 생리학적 증상을 보고하는 대상체에게 투여될 수 있다.
용어 "유효량" 또는 "치료적 유효량"은 유리한 또는 요망되는 결과를 야기하기에 충분한 작용제의 양을 지칭한다. 치료적 유효량은 치료되는 대상체 및 병태, 대상체의 체중 및 연령, 병태의 중증도, 투여 방식 등 중 하나 이상에 따라 달라질 수 있으며, 이는 당업자에 의해 용이하게 결정될 수 있다. 또한, 상기 용어는 본원에 기술된 영상화 방법 중 임의의 것에 의한 검출을 위한 이미지를 제공할 용량에 적용된다. 특정 용량은 선택된 특정 작용제, 뒤따르는 투여 요법, 그것이 다른 화합물과 병용하여 투여되는지 여부, 투여 시기, 영상화되는 조직 및 그것을 운반하는 신체 전달 시스템 중 하나 이상에 따라 달라질 수 있다.
본 명세서에서 식품이란 함은 영양소를 한 가지 또는 그 이상 함유하고 있는 천연물 또는 가공품을 의미하며, 바람직하게는 어느 정도의 가공 공정을 거쳐 직접 먹을 수 있는 상태가 된 것을 의미하며, 통상적인 의미이고, 상기 식품 조성물은 식품, 식품 첨가제, 건강 기능성 식품 및 음료를 모두 포함하는 의도이다.
본 발명의 조성물을 첨가할 수 있는 식품으로는 예를 들어, 각종 식품류, 음료, 껌, 캔디, 차, 비타민 복합제, 기능성 식품 등이 있다. 추가로, 본 발명에서 식품에는 특수영양식품(예, 조제유류, 영, 유아식 등), 식육가공품, 어육제품, 두부류, 묵류, 면류(예, 라면류, 국수류 등), 건강보조식품, 조미식품(예, 간장, 된장, 고추장, 혼합장 등), 소스류, 과자류(예, 스넥류), 유가공품(예, 발효유, 치즈 등), 기타 가공식품, 김치, 절임 식품(각종 김치류, 장아찌 등), 음료(예, 과실, 채소류 음료, 두유류, 발효음료류, 아이스크림류 등), 천연조미료(예, 라면 스프 등), 비타민 복합제, 알코올 음료, 주류 및 그 밖의 건강보조식품류를 포함하나 이에 한정되지 않는다. 상기 식품, 음료 또는 식품첨가제는 통상의 제조방법으로 제조될 수 있다.
본 발명에서 기능성 식품이란 식품에 물리적, 생화학적, 생물공학적 수법 등을 이용하여 해당 식품의 기능을 특정 목적에 작용, 발현하도록 부가가치를 부여한 식품군이나 식품 조성이 갖는 생체방어리듬조절, 질병방지와 회복 등에 관한 체조절기능을 생체에 대하여 충분히 발현하도록 설계하여 가공한 식품을 의미하며, 바람직하게는 본 발명의 기능성 식품은 비만, 지방간, 또는 대사증후군의 예방 또는 개선에 관한 체조절기능을 생체에 대하여 충분히 발현할 수 있는 식품을 의미한다. 상기 기능성 식품에는 식품학적으로 허용 가능한 식품 보조 첨가제를 포함할 수 있으며, 기능성 식품의 제조에 통상적으로 사용되는 적절한 담체, 부형제 및 희석제를 더욱 포함할 수 있다.
유효 성분의 혼합양은 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품 또는 음료의 제조시에 내복자 추출물은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 내복자 추출물의 혼합량은 그의 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 식품 또는 음료의 제조시에 상기 내복자 추출물은 조성물 총 중량에 대하여 15 중량부 이하, 10 중량부 이하, 1중량 내지 15중량부, 1중량 내지 10중량부, 1중량 내지 5중량부, 또는 0.1중량 내지 15중량부의 양으로 첨가될 수 있다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
상기 식품은 담체, 희석제, 부형제 및 첨가제 중 하나 이상을 더 포함하여 정제, 환제, 산제, 과립제, 분말제, 캡슐제 및 액제 제형으로 이루어진 군에서 선택된 하나로 제형될 수 있다. 일 양상에 따른 화합물을 첨가할 수 있는 식품으로는, 각종 식품류, 분말, 과립, 정제, 캡슐, 시럽제, 음료, 껌, 차, 비타민 복합제, 건강기능성 식품류 등이 있다.
상기 담체, 부형제, 희석제 및 첨가제의 구체적인 예로는 락토즈, 덱스트로즈, 슈크로즈, 솔비톨, 만니톨, 에리스리톨, 전분, 아카시아 고무, 인산칼슘, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 미세결정성셀룰로즈, 폴리비닐피롤리돈, 셀룰로즈, 폴리비닐피롤리돈, 메틸셀룰로즈, 물, 설탕시럽, 메틸셀룰로즈, 메틸 하이드록시 벤조에이트, 프로필하이드록시벤조에이트, 활석, 스테아트산 마그네슘 및 미네랄 오일로 이루어진 군으로부터 선택되는 적어도 하나일 수 있다.
상기 식품은 상기 내복자 추출물을 함유하는 것 외에 특별한 제한없이 다른 성분들을 필수 성분으로서 함유할 수 있다. 예를 들어, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다.
상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당 알코올일 수 있다. 상술한 것 이외의 향미제로서 천연 향미제 (타우마틴, 스테비아 추출물 (예를 들어, 레바우디오시드 A, 리시르히진 등)) 및 합성 향미제 (사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 당업자의 선택에 의해 적절하게 결정될 수 있다.
상기 외에도, 일 양상에 따른 식품은 여러 가지 영양제, 비타민, 광물 (전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제 (치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있으며, 이러한 첨가제의 비율 또한 당업자에 의해 적절히 선택될 수 있다.
상기 외에도, 일 양상에 따른 식품은 여러 가지 영양제, 비타민, 광물 (전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제 (치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있으며, 이러한 첨가제의 비율 또한 당업자에 의해 적절히 선택될 수 있다.
상기 담체는 예를 들어, 콜로이드 현탁액, 분말, 식염수, 지질, 리포좀, 미소구체(microspheres) 또는 나노 구형입자일 수 있다. 이들은 운반 수단과 복합체를 형성하거나 관련될 수 있고, 지질, 리포좀, 미세입자, 금, 나노입자, 폴리머, 축합 반응제, 다당류, 폴리아미노산, 덴드리머, 사포닌, 흡착 증진 물질 또는 지방산과 같은 당업계에 공지된 운반 시스템을 사용하여 생체 내 운반될 수 있다.
또 다른 양상은 상기 내복자 추출물을 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 개선용 사료 첨가제 조성물에 관한 것이다.
사료 첨가제 조성물은 분말 또는 과립형태로 제조될 수 있고, 필요에 따라, 구연산, 후말산, 아디픽산, 젖산, 사과산 등의 유기산이나 인산나트륨, 인산칼륨, 산성 피로인산염, 폴리인산염 등의 인산염이나, 폴리페놀, 카테킨, 알파-토코페롤, 로즈마리 추출물, 비타민 C, 녹차 추출물, 감초 추출물, 키토산, 탄닌산, 피틴산 등의 천연 항산화제 중 어느 하나 또는 하나 이상을 추가로 포함할 수 있다. 본 출원의 사료 첨가제는 통상의 사료 형태로 제제화될 수 있으며, 통상의 사료 성분을 함께 포함할 수 있다.
일 양상의 사료 첨가제 조성물은 곡물, 예를 들면 분쇄 또는 파쇄된 밀, 귀리, 보리, 옥수수 및 쌀; 식물성 단백질 사료, 예를 들면 평지, 콩, 및 해바라기를 주성분으로 하는 사료; 동물성 단백질 사료, 예를 들면 혈분, 육분, 골분 및 생선분; 당분 및 유제품, 예를 들면 각종 분유 및 유장 분말로 이루어지는 건조 성분 등을 더 포함할 수 있으며, 이외에도 영양 보충제, 소화 및 흡수 향상제, 성장 촉진제 등을 더 포함할 수 있다.
일 양상의 사료 첨가제 조성물은 동물에게 단독으로 투여하거나 식용 담체 중에서 다른 사료 첨가제와 조합하여 투여할 수도 있다. 또한, 상기 사료 첨가제는 탑 드레싱으로서 또는 이들을 사료에 직접 혼합하거나 또는 사료와 별도의 경구 제형으로 용이하게 동물에게 투여할 수 있다. 상기 사료 첨가제 조성물을 사료와 별도로 투여할 경우, 당해 기술분야에 잘 알려진 바와 같이 약제학적으로 허용 가능한 식용 담체와 조합하여, 즉시 방출 또는 서방성 제형으로 제조할 수 있다. 이러한 식용 담체는 고체 또는 액체, 예를 들어 옥수수 전분, 락토오스, 수크로오스, 콩 플레이크, 땅콩유, 올리브유, 참깨유 및 프로필렌글리콜일 수 있다. 고체 담체가 사용될 경우, 사료 첨가제는 정제, 캡슐제, 산제, 트로키제 또는 함당정제 또는 미분산성 형태의 탑 드레싱일 수 있다. 액체 담체가 사용될 경우, 사료 첨가제는 젤라틴 연질 캡슐제, 또는 시럽제나 현탁액, 에멀젼제, 또는 용액제의 제형일 수 있다.
일 양상의 사료 첨가제 조성물은, 예를 들어 보존제, 안정화제, 습윤제 또는 유화제, 용액 촉진제 등을 함유할 수 있다. 상기 사료 첨가제는 침주, 분무 또는 혼합하여 동물의 사료에 첨가하여 이용될 수 있다.
일 양상의 사료 첨가제 조성물은 포유류, 및 가금 포함하는 다수의 동물 식이에 적용할 수 있다. 상기 포유류로서 돼지, 소, 양, 염소, 실험용 설치 동물, 및 실험용 설치 동물뿐만 아니라, 애완 동물(예: 개, 고양이) 등에게 사용할 수 있으며, 상기 가금류로서 닭, 칠면조, 오리, 거위, 꿩, 및 메추라기 등에도 사용할 수 있다. 상기 사료 첨가제 조성물을 포함하는 동물용 사료 배합 방법은 사료 첨가제 조성물을 동물사료에 건조 중량 기준으로 1kg 당 약 0.001g 내지 500g, 바람직하기로는 10g 내지 100g의 양으로 혼합하는 것일 수 있다. 또한 사료 혼합물을 완전히 혼합한 후, 매쉬로 공급하거나, 추가가공 공정을 통하여 팰렛화, 팽창화, 압출 공정을 거칠 수 있다.
일 양상의 방법은 최적의 로스팅 온도 조건을 통하여 수분에 취약한 설포라핀의 안정성을 유지할 수 있는 내복자의 수분함량을 가질수 있도록 가공할 수 있으므로, 내복자로부터 설포라핀을 다량 포함하는 추출물을 제조할 수 있다. 또한 상기 추출물은 통상의 방법에 의한 추출방법보다 설포라핀의 함량이 증대되어 있으므로 비만, 지방간, 또는 대사증후군의 예방, 개선 또는 치료를 위한 약학, 식물 및 사료 첨가제 조성물로 널리 활용될 수 있다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: 최종 수분함량에 따른 증대된 설포라핀의 함량의 확인
설포라핀은 수분함량에 따라 파괴가 쉬운 것으로 알려져 있으므로, 내복자 추출물로부터 설포라핀의 함유량을 증진시키기 위하여, 원료인 내복자의 수분함량을 조절하는 것이 중요하다. 따라서, 설포라핀의 안정성을 높여 내복자 추출물에 포함된 설포라핀의 함유량을 증진시키는 내복자의 수분함량을 찾아내기 위한 실험을 수행하였다.
내복자(무씨) (100g)를 내복자의 최종 수분 함량을 1, 3, 5, 7, 10, 15, 20, 24 및 30 %이 되도록 조절하였다. 최종 수분 함량을 조절하기 위하여 필요한 경우 건열을 사용해 수분을 더하지 않고 가열하는 로스팅(roasting)과정으로 가열하고, 상기 로스팅 과정의 시간과 온도를 조절하여 최종 수분의 함량을 조정하였으며, 상기 로스팅 과정 이외에 가수 과정을 필요한 경우 추가적으로 진행하였다. 이후 상기 가공된 원료를 믹서기를 이용하여 분쇄하고, 분쇄물을 20%(v/v)의 에탄올로 3시간 동안 교반 추출하였다. 이후 추출물을 감압 농축하고, 농축액을 건조하여 내복자 추출물을 수득하여 상기 내복자 추출물 내 설포라핀의 함량을 측정하였다. 각 내복자 추출물의 중량을 측정하고 이를 원물에 대비하여 추출 수율을 계산하였다. 아울러, 구체적인 설포라핀 함량의 측정은 아래의 방법으로 수행하였다.
각기 다른 조건의 추출물 50 mg을 취하여 증류수 5 ㎖에 충분히 분산시킨 후, 디클로로메탄 용액 10 ㎖을 첨가하여 액체-액체 추출을 하였다. 디클로로메탄 용액을 취하여 원심진공농축기에서 건조한 후 동량의 증류수로 다시 용해하였다. 0.22 ㎛ 시린지 필터(syringe filter)로 여과하여 시험용액을 준비하였다. 설포라핀 표준품을 여러 농도(25 ppm, 50 ppm, 125 ppm, 250 ppm, 500 ppm)로 준비하여 표준용액을 준비하였다. 상기에서 준비한 시험용액과 표준용액 속의 설포라핀 함량을 HPLC 기계를 통해 정량하였으며, Inno C-18 컬럼을 사용하였다. 유동상은 물, 테트라하이드로퓨란 용액을 사용하였으며, 215 nm에서 신호를 검출하였다. 그 후, 표준용액의 설포라핀 농도와 피크 면적 사이에 관계식을 구하고, 시험용액을 분석하여 나온 피크 면적을 관계식에 대입하여 시험용액의 설포라핀 함량을 계산하였다. 수분함량에 따라 내복자 추출물 내 포함된 설포라핀의 함량을 하기 표 1에 나타내었다.

상기 표 1에서 확인한 바와 같이, 내복자의 최종 수분함량이 1~ 7%인 경우, 수분함량 비례적으로 함유된 설포라핀의 함량이 높아지는 것을 확인하였다. 특히 최종 수분함량이 7%인 경우, 설포라핀의 함량이 55.1mg/g으로 가장 높은 것을 확인하였다. 내복자의 최종 수분함량이 7% 를 넘는 경우 설포라핀의 함량이 점차적으로 감소하게 되는 것을 확인하였다. 이는 최종 수분함량이 3%이하에서는 미로시나아제가 글루코시놀레이트를 가수분해 하기에 충분하지 못한 수분함량이기 때문에 상대적으로 설포라핀의 함량이 낮은 것이며, 수분함량이 20%이상인 경우에는 수분에 의하여 설포라핀의 파괴가 이루어져서 안정성이 감소하여 포함된 함량이 줄어들게 되는 것임을 확인하였다. 따라서, 내복자의 최종적인 수분함량을 5 내지 15%로 수분을 조절하도록 가공하는 것이 설포라핀의 함량이 가장 증진시키는 조건임을 확인할 수 있었다.
실시예 2 : 로스팅 온도 조건에 따른 증대된 설포라핀의 함량 확인
2.1 로스팅 온도 조건에 따른 설포라핀의 함량의 확인
내복자 추출물에 포함된 설포라핀의 함유량을 증진시키기 위한 가공방법에 있어서, 로스팅의 온도에 따라 설포라핀의 함량이 달라질 수 있는지를 확인하기 위한 실험을 수행하였다. 실험 방법은 상기 실시예 1의 방법에 있어서, 로스팅 온도 조건을 제외한 각 실험군의 모든 조건을 동일하게 설정하여 수행하였다. 구체적으로, 내복자를 품온 기준으로 40, 50, 및 60℃의 온도조건으로 로스팅 처리하고 이후 동일한 입도로 분쇄하고, 최종 설포라핀의 함량을 확인하였고 이를 확인한 결과를 도 1A에, 온도 별 잔여 글루코라페닌의 함량을 확인한 결과를 도 1B에, 온도 별 총 글루코라페닌의 함량을 확인한 결과를 도 1C에 나타내었다.
도 1A에서 확인한 바와 같이, 50 및 60℃로 열처리를 수행한 실험군은 열처리를 하지 않은 대조군 대비 설포라핀의 함량이 유의하게 감소한 것을 확인하였다. 다만, 40℃로 열처리를 수행한 경우는 대조군과 비슷한 정도의 설포라핀의 함량을 나타내는 것을 확인하였다.
도 1B에서 확인한 바와 같이, 50 및 60℃의 온도로 로스팅 과정을 처리한 군은 설포라핀으로의 가수분해가 저해되어 잔여 글루코라페닌이 대조군 및 40℃로 열처리를 수행한 군보다 유의하게 잔여량이 확연하게 많은 것을 확인하였고, 대조군과 40℃로 열처리를 수행한 군은 잔여 글루코라페닌 양의 차이가 유의하지 않은 정도임을 확인하였다. 또한 도 1C에서 확인한 바와 같이, 총 글루코라페닌의 양은 열처리를 통한 로스팅을 수행한 군은 모두 일정 정도로 감소하게 되는 것을 확인하였다.
2.2 로스팅 온도 조건에 따른 미로시나아제의 활성의 확인
상기 실시예 2.1을 통해 확인한 로스팅 온도에 따른 설포라핀의 함량 변화가 미로시나아제의 활성과 관련성이 있는지를 확인하기 위한 실험을 수행하였다. 실험의 조건을 실시예 2.1과 동일하게 하고, 미로시나아제의 활성을 확인하여 도 2에 나타내었다.
도 2에서 확인한 바와 같이, 50 및 60℃의 온도로 로스팅 과정을 처리한 군에서 대조군 및 40℃로 열처리를 수행한 군 대비 미로시나아제의 활성이 유의하게 저해되는 것을 확인하였다. 다만 40℃로 로스팅을 과정을 수행한 군은 대조군과 유의할 정도로의 미로시나아제의 활성이 유지되는 것을 확인하였다. 따라서, 50℃이상의 온도의 처리는 미로시나아제의 활성을 현저하게 저해할 수 있으므로, 이를 통한 설포라핀 전환 반응을 저해할 수 있음을 구체적으로 확인하였다.
종합적으로, 설로라핀의 함량을 증진시키기 위해서는 내복자를 열처리를 수행하지 않거나 40℃의 온도로 로스팅 처리를 수행하여야 함을 확인할 수 있었다.
실시예 3. 분쇄물의 평균 입도에 따른 증대된 설포라핀의 함량 확인
3.1 분쇄도에 따른 입도의 확인
분쇄도에 따른 평균입도에 따라 추출물 내 포함된 설포라핀의 함량이 증진될 수 있는지를 확인하기 위한 실험을 수행하였다. 실험 방법은 상기 실시예 1의 방법에 있어서, 분쇄도를 제외한 각 실험군의 모든 조건을 동일하게 설정하여 수행하였다. 구체적으로, 수분에 취약한 내복자의 특성을 고려하여, 건식 분쇄 방법을 적용하였다. 분쇄도에 따라 3종의 분쇄물을 수득하였고, 분쇄 정도를 순서대로 높였고, 이에 따른 각 실험군을 #1, #2, 및 #3으로 명명하고, 각 군의 입도를 분석한 결과를 표 2 및 도 3에 나타내었다.

상기 표 2에 나타난 바와 같이, 분쇄도가 높아질수록 입도가 작아졌음을 확인할 수 있었다. 각 군의 평균입도(D[4,3])는 #1는 179.50 μm, #2는 11.31 μm, #3은 8.73 μm임을 확인하였다. 도 3에서 확인한 바와 같이 #1 군의 경우 내복자 껍질과 과육의 경도차이로 인해 두 개의 peak를 확인할 수 있었다.
3.2 분쇄도에 따른 증대된 설포라핀의 함량 확인
실시예 3.1을 통해 확인한 분쇄도에 따라 설포라핀의 함량 변화가 나타나는지를 확인하기 위한 실험을 수행하였다. 실험의 조건을 실시예 3.1과 동일하게 하고, 최종 설포라핀의 함량을 확인하였고 이를 확인한 결과를 도 4A에, 분쇄도 별 잔여 글루코라페닌의 함량을 확인한 결과를 4B에, 분쇄도 별 총 글루코라페닌의 함량을 확인한 결과를 도 4C에 나타내었다.
대조군으로 LN2는 미로시나아제의 활성을 통제하기 위하여 액체 질소를 처리한 분쇄물을 활용하였다.
도 4A에서 확인한 바와 같이, 분쇄도가 높아져 평균 입도가 11.31 μm에 해당하는 #2군은 #1 군보다 최종적인 추출물에서 설포라핀의 함량을 더 높일 수 있음을 확인하였다. 또한 도 4B 및 4C에서 확인할 수 있는 바와 같이, 또한 내복자 추출박에 남아있는 잔여 글루코라페닌의 함량은 분쇄도가 높아짐에 따라 감소하는 양상이 나타나는 것을 확인하였으나, 반면 총 글루코라페닌의 함량은 분쇄도에 따라서는 차이가 나타나지 않는 것을 확인할 수 있었다.
3.2 분쇄도에 따른 미로시나아제의 활성의 확인
상기 실시예 3.2을 통해 확인한 분쇄도에 따른 설포라핀의 함량 변화가 미로시나아제의 활성과 관련성이 있는지를 확인하기 위한 실험을 수행하였다. 실험의 조건을 실시예 3.1과 동일하게 하고, 미로시나아제의 활성을 확인하여 도 5에 나타내었다. 대조군으로 LN2는 미로시나아제의 활성을 통제하기 위하여 액체 질소를 처리한 분쇄물을 활용하였다.
도 5에서 확인한 바와 같이, 미로시나아제 활성이 통제된 액체질소 분쇄물 (LN2)의 경우 분쇄되는 과정에서 설포라핀 전환 반응이 발생하지 않았음을 확인할 수 있었다. 다만 #1, #2 및 #3 군에서도 LN2와 유의할 정도의 차이가 나타나지 않는 것을 확인하여, 미로시나아제의 활성은 분쇄도, 즉 입도에 따라 차이를 나타내지 않음을 확인하였다.
실시예 4. 에탄올 수용액의 비율에 따른 설포라핀의 함량 확인
추출용매인 에탄올의 비율에 따라 추출물 내 포함된 설포라핀의 함량이 증진될 수 있는지를 확인하기 위한 실험을 수행하였다. 실험 방법은 상기 실시예 1의 방법에 있어서, 에탄올의 비율을 제외한 각 실험군의 모든 조건을 동일하게 설정하여 수행하였다. 구체적으로, 내복자를 각각 0, 5, 10, 20, 30 및 50 %(v/v)의 에탄올을 포함한 에탄올 수용액을 활용하여 추출하였고, 에탄올 포함 비율에 따른 설포라핀의 함량을 확인한 결과를 도 6A에, 에탄올을 포함하고 있지 않은 수용액으로 추출한 군에 포함된 설포라핀의 시간에 따른 함량 변화를 확인한 결과를 도 6B에, 에탄올을 20%(v/v)로 함유한 군에 포함된 설포라핀의 시간에 따른 함량 변화를 확인한 결과를 도 6C에 나타내었다.
도 6A에 나타난 바와 같이, 내복자를 각각 0, 5, 10, 20, 30 및 50 %(v/v)의 에탄올을 포함한 에탄올 수용액로 추출하였을 때, 특히 20%(v/v) 에탄올에서 가장 높은 추출 수율을 나타내, 최종적인 설포라핀의 함량이 높은 것을 확인할 수 있었다. 도 6B 및 6C에 나타난 바와 같이, 에탄올이 0% (v/v)인 군에서는 추출된지 4시간 만에 설포라핀이 모두 분해되는 것을 확인할 수 있었다. 반면 20% (v/v)로 에탄올이 포함된 군에서는 추출된 이후 12시간까지 수득된 설포라핀이 안정적으로 존재하는 것을 확인하였다.
종합적으로, 내복자로부터 설포라핀을 추출할 경우, 20% (v/v)의 에탄올이 가장 설포라핀을 증대시킬 수 있는 용매조건임을 확인하였다. 또한 추출용매 내 수분 조건에 따라 최종 설포라핀의 안정성 역시 달라질 수 있으므로, 설포라핀을 증대시키기 위하여는 수분 함량을 통제하는 것이 필요한 것임을 구체적으로 확인하였다.
실시예 7 : 추출공정에 따른 설포라핀의 함량 증가의 확인
상기 실시예들을 바탕으로 추출물 내 포함된 설포라핀의 함량을 최대화하기 위해서, 수분함량, 분쇄도 및 용매조건 별로 추출을 진행하고 이에 대한 설포라핀의 함량을 확인하는 실험을 수행하였고, 이에 대한 결과를 하기 표 3에 나타내었다.

상기 표 3에서 확인한 바와 같이, 추출물 내 포함된 설포라핀의 함량을 약 40 mg/g이상으로 포함시켜 최대화하기 위하여, 수분함량은 5~15% 내외의 조건으로 하는 것이 설포라핀의 안정성을 고려했을 때 임계적 함량임을 확인하였다. 또한 로스팅 온도의 경우, 내복자의 품온 기준으로 40℃이하인 경우나 혹은 수분 조정이 필요 없는 경우에는 로스팅 공정을 추가하지 않은 경우가 설포라핀의 함량을 증대시킬 수 있는 조건임을 확인하였다. 아울러, 입도는 D[4,3]가 약 11 μm로 하는 것이 유리하고, 추출물에 사용하는 에탄올 함량은 20%가 유리하며, 추출 온도는 15~35℃정도가 유리한 것임을 구체적으로 확인할 수 있었다.

Claims (14)

  1. 내복자의 수분함량을 조절하는 단계;
    내복자를 분쇄하여 평균입도(D[4,3])가 5 내지 50 μm 가 되도록 분쇄물을 제조하는 단계; 및
    상기 분쇄물을 물, C1 내지 C4의 알코올 또는 이들의 혼합물로 추출하는 단계를 포함하는 추출물 내 설포라핀의 함량이 증대된 내복자 추출물의 제조방법.
  2. 청구항 1에 있어서, 상기 수분함량을 조절하는 단계는 추가적으로 내복자를 로스팅(roasting)하는 단계를 포함하는 것인 제조방법.
  3. 청구항 2에 있어서, 상기 로스팅은 내복자의 품온이 25 내지 40℃가 되도록 수행되는 것인 제조방법.
  4. 청구항 1에 있어서, 상기 수분함량을 조절하는 단계는 내복자의 수분함량을 5 내지 15%(w/w) 로 조절하는 것인 제조방법.
  5. 청구항 1에 있어서, 상기 C1 내지 C4의 알코올은 에탄올인 것인 제조방법.
  6. 청구항 5에 있어서, 상기 에탄올은 15 내지 25 %(v/v) 에탄올인 것인 제조방법.
  7. 청구항 1에 있어서, 상기 추출하는 단계는 15 내지 35 ℃ 온도에서 1시간 내지 10시간 동안 수행되는 것인 제조방법.
  8. 내복자의 수분함량을 조절하는 단계;
    내복자를 분쇄하여 평균입도(D[4,3])가 5 내지 50 μm 가 되도록 분쇄물을 제조하는 단계; 및
    상기 분쇄물을 물, C1 내지 C4의 알코올 또는 이들의 혼합물로 추출하는 단계를 포함하는 추출물 내 설포라핀의 함량이 증대된 내복자의 가공방법.
  9. 청구항 1 내지 7 중 어느 하나의 방법에 의하여 제조된 설포라핀의 함량이 증대된 내복자 추출물.
  10. 청구항 9의 내복자 추출물을 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 치료용 약학적 조성물.
  11. 청구항 10에 있어서, 상기 대사증후군은 고혈압, 동맥경화, 고지혈증, 간질환, 과인슐린혈증, 과혈당증 및 2형 당뇨병로 이루어진 군으로부터 선택된 1종 이상인 것인 약학적 조성물.
  12. 청구항 9의 내복자 추출물을 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 개선용 식품 조성물.
  13. 청구항 9의 내복자 추출물을 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 개선용 사료 첨가제 조성물.
  14. 청구항 9의 내복자 추출물을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 비만, 지방간, 또는 대사증후군의 예방 또는 치료 방법.
