Abstract
본 발명의 일 실시예에 따른 전기화학적 유량모니터링 장치는 소정의 부피를 가지며 이온성 유체를 수용하는 공간이 가운데 형성된 멤브레인; 멤브레인의 양측에 각각 마련된 제1 전극과 제2 전극; 및 제1 전극 및 제2 전극에 전압을 인가하여 공간을 농도 분극 상태로 변하게 하는 제어부를 포함하되, 멤브레인은 공간에 유체가 드나들 수 있는 유체 이동 통로를 포함하는 것이고, 제어부는 농도 분극 상태에서 유체 이동 통로를 통해 주입된 유체에 의해, 발생된 이온 전류값을 기초로 유량을 산출한다.
Description
본 발명은 전기화학적 유량모니터링 장치에 관한 것이다.
약물주입펌프는 정확히 컨트롤되어 주입되어야 치료에 효과적일 뿐 아니라, 오주입(과다주입, 과소주입)에 따른 부작용이 최소화된다. 현재 병원 등에서 활용되고 있는 약물주입펌프는 용적펌프(volumetric pump), 주사기펌프(syringe pump), 체내이식형펌프(implantable pump) 등이 있는데, 대부분의 경우 유량 모니터링이 정확히 측정되어 제어되지 못하고 있다. 이는 개별적으로 간단히 활용 가능한 마이크로리터(microliter) 혹은 나노리터(nanoliter) 수준의 유량을 정밀하게 측정하는 간단한 기술이 존재하지 않기 때문이다.
도 1은 종래의 전기삼투펌프에 활용되는 전극을 도시한 도면이다.
전기삼투펌프는 다공성막(다공성멤브레인)의 양단의 전극을 이용하여 전압을 걸었을 때 생기는 전기삼투현상에 의해 유체가 이동하는 것을 이용한 펌프이다. 다공성막의 재료로는 일반적으로 실리카(silica), 유리(glass) 등이 사용되는데, 이들은 수용액에 담겨 있게 되면 표면이 음전하를 띠게 된다. 이 상태에서 전압을 가하면 (+)전극부로부터 (-)전극부로 유체의 이동이 일어나게 된다(도1의 윗 그림). 다공성막에는 수 많은 유체가 지나갈 수 있는 경로가 있다. 이 중 하나를 확대해 보면 유체통로의 표면(다공성막 재료의 표면이 됨)은 음전하(bound anion)를 띠게 되므로 이는 맞닿는 유체에서 움직일 수 있는 (+)전하를 가진 양이온(mobile cation)이 이동하여 전하균형이 맞는 상태가 된다. 이때 전압을 가해주면 (+)전극부 쪽에서 (-)전극부 방향으로 양이온(mobile cation)이 표면을 따라 이동하면서 이에 따른 수소결합 네트워크로 연결된 유체 전체가 미끄러지듯이 흘러가는 현상이 생기는데 이를 전기삼투현상이라 하고, 이 원리를 이용한 펌프가 전기삼투펌프이다.
도1을 참조하면, 전기삼투펌프에 활용되는 전극은 유체의 이동을 원활하게 하기 위해 다공성 전극인 백금망(Pt mesh), 다공성 탄소 종이 혹은 섬유(carbon paper or carbon cloth), 또는 다공성 구조 위에 코팅된 다양한 전극 물질이 활용된다. 또한 전극은 실리카 등으로 구성된 다공성 막을 사이에 두고 전압을 걸어주면 이에 따른 유체의 이동이 있게 된다.
전기삼투펌프의 유량은 전압 크기, 전압을 걸어준 시간 등에 의해 조절 가능하다. 따라서, 전기삼투펌프의 주입정확도를 향상시키기 위해, 유량에 대한 피드백이 가능한 정밀한 유량 모니터링 장치가 요구된다.
이와 관련하여, 종래 기술로서, 전기화학적인 분극현상을 이용한 유속측정장치가 보고된 바가 있는데(ACS Sensors 2019), 이 경우는 직접 전류를 측정하는 방식이 아니라 농도분극을 일으키는 전극쌍이 따로 있고, 유체가 지나가는 통로에 다른 전극쌍으로 임피던스(impedance)를 측정하는 방식이다. 이는 측정하고자 하는 유체에 전극이 직접 닿기 때문에 유체가 약물일 경우 오염의 우려가 있다.
이러한 문제를 해결하기 위해 본 발명의 일 실시예는 약물의 오염없이, 약물의 과다주입, 과소주입으로 인한 위험 및 부작용을 방지하고자 마이크로리터 혹은 나노리터 수준의 유량을 정밀하게 모니터링하고, 일정 이온 농도를 갖는 용액을 주입하는 전기화학적 유량모니터링 장치를 제공하고자 한다.
다만, 본 실시예가 이루고자 하는 기술적 과제는 상기된 바와 같은 기술적 과제들로 한정되지 않으며, 또 다른 기술적 과제들이 더 존재할 수 있다.
상술한 기술적 과제를 달성하기 위한 기술적 수단으로서, 전기화학적 유량모니터링 장치는 소정의 부피를 가지며 이온성 유체를 수용하는 공간이 가운데 형성된 멤브레인; 멤브레인의 양측에 각각 마련된 제1 전극과 제2 전극; 및 제1 전극 및 제2 전극에 전압을 인가하여 공간을 농도 분극 상태로 변하게 하는 제어부를 포함하되, 멤브레인은 공간에 유체가 드나들 수 있는 유체 이동 통로를 포함하는 것이고, 제어부는 농도 분극 상태에서 유체 이동 통로를 통해 주입된 유체에 의해, 발생된 이온 전류값을 기초로 유량을 산출한다.
유체는 0.1 mM 내지 10 mM의 낮은 이온도를 가진 용액인 것이다.
멤브레인의 공간은 2uL 내지 4uL 의 부피인 것이다.
제1전극 및 제2전극의 간격은 50um 내지 1mm 인 것이다.
제1 전극 및 제2 전극의 일측에 돌출된 형태로 형성되되, 제어부와 전기적으로 연결되는 연결부를 더 포함한다.
유체 이동 통로에 연결되어 공간으로 기 설정된 유량의 유체를 주입하는 펌핑부를 더 포함한다.
제어부는 제 1 전극 및 제 2 전극에 소정의 전압을 공급하는 전원 공급부; 및 공간에 주입된 유체의 이온들이 제1전극 및 제2전극의 양단으로 이동함에 따라 발생하는 이온 전류를 측정하는 측정부를 포함한다.
전술한 본 발명의 과제 해결 수단에 의하면, 본 발명은 한쌍의 전극으로 농도분극과 유량에 비례한 전류 혹은 전하량 측정이 가능하므로, 농도분극을 일으키는 별도의 전극쌍을 포함하는 종래의 기술과 구조적으로 다를 뿐 아니라 훨씬 단순한 형태를 나타낸다.
본 발명에 따른 유량 측정은 일정 이온농도를 가지는 모든 용액에 적용 가능하지만, 0.1~10 mM 정도의 낮은 이온도 용액을 활용하는 전기삼투펌프에 적용하기에 무척 유용하다.
또한, 기존의 전기삼투펌프에 본 발명에 따른 유량모니터링 장치를 유량의 피드백으로 연결하면 훨씬 더 정확한 유량 컨트롤이 가능하다.
더불어, 본 발명에 따른 유량모니터링 장치는 약물 이외에도 다른 다양한 정량주입에 활용이 가능하며, 현재 유량 센서의 개발 분야에 나타나는 보틀넥(bottle-neck) 문제를 해결할 수 있다.

아래에서는 첨부한 도면을 참조하여 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예를 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다. 그리고 도면에서 본 발명을 명확하게 설명하기 위해서 설명과 관계없는 부분은 생략하였으며, 명세서 전체를 통하여 유사한 부분에 대해서는 유사한 도면 부호를 붙였다.
본 발명 명세서 전체에서, 어떤 부분이 다른 부분과 "연결"되어 있다고 할 때, 이는 "직접적으로 연결"되어 있는 경우뿐 아니라, 그 중간에 다른 소자를 사이에 두고 "전기적으로 연결"되어 있는 경우도 포함한다.
본 발명 명세서 전체에서, 어떤 부재가 다른 부재 “상에” 위치하고 있다고 할 때, 이는 어떤 부재가 다른 부재에 접해 있는 경우뿐 아니라 두 부재 사이에 또 다른 부재가 존재하는 경우도 포함한다.
본 발명 명세서 전체에서, 어떤 부분이 어떤 구성요소를 "포함" 한다고 할 때, 이는 특별히 반대되는 기재가 없는 한 다른 구성요소를 제외하는 것이 아니라 다른 구성 요소를 더 포함할 수 있는 것을 의미한다. 본 발명 명세서 전체에서 사용되는 정도의 용어 "약", "실질적으로" 등은 언급된 의미에 고유한 제조 및 물질 허용오차가 제시될 때 그 수치에서 또는 그 수치에 근접한 의미로 사용되고, 본 발명의 이해를 돕기 위해 정확하거나 절대적인 수치가 언급된 개시 내용을 비양심적인 침해자가 부당하게 이용하는 것을 방지하기 위해 사용된다. 본 발명 명세서 전체에서 사용되는 정도의 용어 "~(하는) 단계" 또는 "~의 단계"는 "~ 를 위한 단계"를 의미하지 않는다.
본 발명은 마이크로리터 혹은 나노리터 수준의 유량을 정밀하게 모니터링하여 약물펌프 등 미량 정밀 펌프의 주입정확도를 높이는 기술로 전기화학적인 원리가 적용된다. 특히 용액의 이온도(이온의 농도)가 일정한 용액의 유량 및 유속을 결정하는 데에 적용될 수 있다. 두 전극 사이에 전압을 걸어 이온들의 농도분극현상(concentration polarization)이 유도된 상태에서 이온성 액체(유체, 용액)가 투입되면 이 액체에 포함된 이온의 전극의 양단으로 이동하게 되는데, 이에 따라 전류(이온전류, ionic current)가 측정된다. 이 전류는 투입된 유량과 비례하므로 측정된 전류 혹은 전하량을 유량과 연계되는 정량적 지표로 사용할 수 있다.
도 2는 본 발명의 일 실시예에 따른 분극 후 이온성 용액 주입에 따른 이온전류 측정 원리를 설명하기 위한 도면이다. 도 3은 본 발명의 일 실시예에 따른 유량 센서용 전극과 결합된 유체 이동 통로 및 펌핑 구성을 도시한 도면이다. 도 4a 는 본 발명의 일 실시예에 따른 유량 센서용 전극 사이에 배치된 멤브레인을 대략적으로 도시한 도면이다. 도 4b는 본 발명의 일 실시예에 따른 제어부의 구성을 도시한 블록도이다.
도시된 바와 같이, 본 발명의 일 실시예에 따른 전기화학적 유량모니터링 장치는 펌핑부(10), 유체 이동 통로(110), 공간(125)이 형성된 멤브레인(120), 제1 전극(130), 연결부(140), 제2전극(150), 및 전원공급부(210)와 측정부(220)로 구성된 제어부(20)를 포함한다.
구체적으로, 본 발명의 유량모니터링 장치는 소정의 부피를 가지며 이온성 유체를 수용하는 공간(125)이 가운데 형성된 멤브레인(120), 멤브레인(120)의 양측에 각각 마련된 제1 전극(130)과 제2 전극(150), 및 제1 전극(130) 및 제2 전극(150)에 전압을 인가하여 공간을 농도 분극 상태로 변하게 하는 제어부(20)를 포함한다. 이때, 멤브레인(120)은 공간에 유체가 드나들 수 있는 유체 이동 통로(110)를 포함한다. 또한 제어부(20)는 농도 분극 상태에서 유체 이동 통로(110)를 통해 주입된 유체에 의해, 발생된 이온 전류값을 기초로 유량을 산출한다.
예시적으로 유체는 0.1 mM 내지 10 mM의 낮은 이온도를 가진 용액일 수 있으나 이에 한정된 것은 아니다. 0.1~10 mM 정도의 낮은 이온도를 가진 용액이 전기삼투펌프에 일반적으로 사용되며, 용액이 낮은 이온농도일 때 전기삼투현상에 더 유리하다.
예시적으로 도 4a는 도 3에 도시된 제1 전극(130), 멤브레인(120), 및 제2전극(150)의 배치 관계가 대략적으로 도시된 사시도이다.
제1 전극(130) 및 제2 전극(150)은 멤브레인(120)의 양측에 각각 배치될 수 있다.
또한, 제1 전극(130) 및 제2 전극(150)은 일측에 돌출된 형태로 형성된 연결부(140)를 포함할 수 있다. 이때 연결부(140)는 제어부(20)와 전기적으로 연결될 수 있다.
멤브레인(120)은 소정의 부피를 가지며 이온성 유체가 유체 이동 통로(110)를 통해 유입되어, 수용되는 공간(125)을 포함할 수 있다.
일 예로, 멤브레인(120)은 비다공성 박막으로 이루어 질 수 있다. 이에 따라 유체는 유체 이동 통로(110)를 통해 공간(125)에 주입될 수 있으며, 주입된 유체는 공간(125)의 내부에 수용될 수 있다. 이때, 멤브레인(120)의 공간(125)은 2uL 내지 4uL 의 부피를 갖도록 형성될 수 있으며, 유체를 담아두는 역할을 한다. 바람직하게, 구형으로 형성된 공간(125)은 지름 2 mm, 두께 1 mm 로서 약 3uL 부피를 갖도록 형성될 수 있다. 여기서, 통로의 형상이 구형에 한정되는 것은 아니며, 유량에 따라 다양한 부피를 갖는 모양으로 형성될 수 있다.
예시적으로, 도4a에 도시된 멤브레인(120)의 양측에 제1전극(130) 및 제2전극(150)이 결합되고, 도3에 도시된 펌핑부(10)가 유체 이동 통로(110)와 연결될 수 있다.
이때 펌핑부(10)는 유체 이동 통로(110)에 연결되어 공간(125)으로 기 설정된 유량의 유체를 주입하는 역할을 한다. 일 예로, 펌핑부(10)는 종래의 전기삼투펌프와 연결되어 유량 컨트롤이 이루어질 수 있으나, 이에 한정된 것은 아니다.
또한, 결합된 유량 센서용 전극은 도2에 도시된 것처럼, 멤브레인(120)의 가운데 관통한 형태로 형성된 공간(125)과 공간(125)을 사이에 두고, 마주보며 배치된 제1전극(130) 및 제2전극(150)으로 구성될 수 있다. 일 예로, 제1전극(130) 및 제2전극(150)의 간격은 50um 내지 1mm 로 이루어 질 수 있다.
예시적으로 도 2(a)를 참조하면, 전원 공급부(210)는 제1전극(130) 및 제2 전극(150)의 양단에 전기분해가 일어나지 않을 정도의 전압을 인가하여, 제1전극(130) 및 제2전극(150) 사이에 위치한 멤브레인(120)의 공간(125)이 농도 분극 상태로 변하게 할 수 있다. 바람직하게, 전원 공급부(210)는 전압으로 1.0 V 를 인가할 수 있다.
이어서, 제1전극(130) 및 제2전극(150) 사이에 위치한 공간(125)의 내부가 농도 분극 상태에서 이온을 포함한 유체가 주입되면, 도2(b)에 도시된 것처럼, 제1전극(130) 및 제2전극(150) 사이에 위치한 공간(125)에서 양이온 전류가 흐르게 된다. 이때 제어부(20)가 공간(125)의 내부에서 발생하는 전류값 또는 전하량을 측정하고, 측정값에 기초하여 유체의 미소 부피를 산출할 수 있다.
도 4b를 참조하면 제어부(20)는 제 1 전극(130) 및 제 2 전극(150)에 소정의 전압을 공급하는 전원 공급부(210) 및 공간(125)에 주입된 유체의 이온들이 제1전극(130) 및 제2전극(150)의 양단으로 이동함에 따라 발생하는 이온 전류를 측정하는 측정부(220)를 포함할 수 있다. 예를 들어, 도 4a에 도시된 제1전극(130) 및 제2전극(150)에 돌출된 연결부(140)가 제어부(20)와 연결될 수 있다.
이하, 본 발명의 일 실시예에 따른 유량모니터링 장치를 이용한 유량과 전류의 특성을 설명하고자 한다.
(예 1) 유량 센서의 구성
일 예로, 도3에 도시된 것처럼, 본 발명의 전기화학적 유량모니터링 장치는 제1및 제2전극(130, 150) 사이에 양면테이프(150, 멤브레인)의 가운데 일정 부피가 되도록 공간(125)을 만들고 용액이 드나들 수 있는 유체 이동 통로(110)를 포함한다. 또한 도4b에 도시된 것처럼 전극(130, 150)의 양단에 외부에서 전압을 걸며 전류를 측정할 수 있도록 연결부(140)를 포함한다. 이때 유체를 담아 유량 센서 역할을 하는 통로인 구형 공간(125)은 지름 2 mm, 두께 1 mm 로서 약 3 uL 부피를 갖는다.
도 5는 본 발명의 일 실시예에 따른 1 mm 간격 분극된 전극 사이에 이온성용액 주입에 따른 전류 특성 변화 그래프이다.
(예 2) 1 mm 간격 전극을 이용한 유량 센서의 성능
도 5를 참조하면, 제1 및 제2전극(130, 150)에 1.0 V 를 인가하여 공간(125)의 내부가 농도 분극 상태로 변하게 한다. 이후 펌핑부(10, 주사기펌프)를 이용하여 0.5 mM Li₂SO₄ 용액을 10초 동안에 각각 2 uL, 3 uL, 4 uL 씩 주입하며 전류 특성을 관찰하였다. 도5(a) 내지 도5(c)에 도시된 것처럼, 각각 2 uL, 3 uL, 4 uL 유체(용액)를 순차적으로 주입한 경우, 전류 특성은 시간이 흐름에 따라 감소하는 것으로 나타난다.
도 6a는 본 발명의 일 실시예에 따른 유량과 전류의 선형성 그래프이다.
도 6a를 참조하면, 피크전류를 유량과 관계된 값으로 연계시킬 수 있음을 나타낸다. 4 uL의 경우 용액 부피가 공간(125)의 부피보다 약간 커서 크기가 조금 작아진 것으로 추정된다.
도 6b는 본 발명의 일 실시예에 따른 유량에 따른 전류의 안정성 그래프이다.
도 6b를 참조하면, 각각의 유속(유량)에서 여러 번 반복하여 실험한 결과 재현성은 2% 이내임을 확인할 수 있다.
처음 3개의 데이터는 주사기펌프의 첫 사용의 불안정으로부터 기인했던 것으로 확인되어, 나머지 7개의 데이터로 평균, 표준편차를 계산하였다. 2 uL의 경우 27±0.4 nA 로 1.5%의 정밀도를 나타내고, 3 uL의 경우 37±0.3 nA 로 0.8%의 정밀도를 나타내고, 4 uL의 경우 45±0.3 nA 로 0.8% 의 정밀도를 나타낸다.
도 7은 본 발명의 일 실시예에 따른 50 um 간격 분극된 전극 사이에 이온성용액 주입에 따른 전류 특성 변화 그래프이다.
(예 3) 50 um 간격 전극을 이용한 유량 센서의 성능
도 7을 참조하면, 예2와 유사한 형태의 구조이나, 양 전극(130, 150) 사이의 간격은 50 um 으로 1/20 줄이고, 전극의 면적은 3 mm x 19 mm 로 18배 증가된 유량 센서 전극을 활용하여 전류를 측정하였다. 이때 공간(125)의 내부 부피는 예2와 유사하게, 3 uL를 갖도록 형성하였다. 양 전극에 1.0 V 로 전압을 인가하여, 베이스라인을 안정시킨 후 0.5 uL 유체를 순차적으로 주입한 경우, 전류의 특성이 도7에 도시된 것과 같이 일정하게 나타난다.
도 8은 본 발명의 일 실시예에 따른 유량에 따른 전류 변화 그래프이다.
도 8을 참조하면, 예 3의 유량 센서 구조를 이용하여, 0.5 mM Li₂SO₄ 용액을 10초 동안에 0.5, 1.0, 2.0 uL 를 각각 주입하여 전류의 특성을 관찰하였다. 이때 주입된 용액의 부피가 증가함에 따라 전류값도 증가하여, 전류의 특성이 비례하여 나타난다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다. 예를 들어, 단일형으로 설명되어 있는 각 구성요소는 분산되어 실시될 수도 있으며, 마찬가지로 분산된 것으로 설명되어 있는 구성요소들도 결합된 형태로 실시될 수 있다.
본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허청구범위에 의하여 나타내어지며, 특허청구범위의 의미 및 범위 그리고 그 균등 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (7)
- 전기화학적 유량모니터링 장치에 있어서,소정의 부피를 가지며 이온성 유체를 수용하는 공간이 가운데 형성된 멤브레인;상기 멤브레인의 양측에 각각 마련된 제1 전극과 제2 전극; 및상기 제1 전극 및 제2 전극에 전압을 인가하여 상기 공간을 농도 분극 상태로 변하게 하는 제어부를 포함하되,상기 멤브레인은 상기 공간에 유체가 드나들 수 있는 유체 이동 통로를 포함하는 것이고,상기 제어부는 농도 분극 상태에서 상기 유체 이동 통로를 통해 주입된 유체에 의해, 발생된 이온 전류값을 기초로 유량을 산출하는 것인, 전기화학적 유량모니터링 장치.
- 제1항에 있어서,상기 유체는 0.1 mM 내지 10 mM의 낮은 이온도를 가진 용액인 것인, 전기화학적 유량모니터링 장치.
- 제1항에 있어서,상기 멤브레인의 공간은 2uL 내지 4uL 의 부피인 것인, 전기화학적 유량모니터링 장치.
- 제1항에 있어서,상기 제1전극 및 제2전극의 간격은 50um 내지 1mm 인 것인, 전기화학적 유량모니터링 장치.
- 제1항에 있어서,상기 제1 전극 및 제2 전극의 일측에 돌출된 형태로 형성되되, 상기 제어부와 전기적으로 연결되는 연결부를 더 포함하는 것인, 전기화학적 유량모니터링 장치.
