Description

制御性T細胞の誘導方法
 本発明は、血液中の制御性T細胞を誘導する方法に関する。
 人体には、体外からの異物やウイルスからの侵入を防ぐための機構として免疫が備わっている。免疫には、外敵からの防御を担う細胞(エフェクターT細胞)と、その細胞の働きを抑制方向に調節する細胞(制御性T細胞)とが関与しており、両者のバランスが重要である。自己免疫疾患や、花粉症などのアレルギー疾患は、上記のバランスが崩れ、エフェクターT細胞が制御性T細胞に対して相対的に働きが強くなることにより生じることが分かってきている。
 そこで、近年、自己免疫疾患や、アレルギー疾患の治療のために、制御性T細胞を誘導する研究がなされている(例えば特許文献1、2、3参照)。

 しかしながら、上記各特許文献に記載されている方法は、いずれも血液に添加剤(例えば刺激物質など)を投与することにより制御性T細胞の誘導を行っている。このような添加剤は、制御性T細胞の誘導以外の点において、人体に与える影響について懸念がある。
 そこで、本発明は、添加剤を用いることなく、血液中の制御性T細胞を誘導する方法を提供することを課題としている。
 上記課題を解決するために、本発明に係る制御性T細胞の誘導方法の一態様は、生体から血液を抽出する工程と、抽出された前記血液に対して260nm以上320nm以下の波長域の紫外線を照射する工程と、を含む。
 このように、生体(例えば人体)から抽出された血液に対して上記の特定波長域の紫外線を照射することのみで、血液中の制御性T細胞を誘導することができる。つまり、添加物を用いることなく血液中の制御性T細胞を誘導することができる。なお、制御性T細胞を誘導するとは、血液中のヘルパーT細胞(CD4陽性細胞)における制御性T細胞(CD25陽性、Foxp3陽性細胞)の割合を増やすことをいう。
 また、上記の制御性T細胞の誘導方法において、前記紫外線を照射する工程では、前記抽出された血液に対して260nm以上290nm以下の波長域の紫外線を照射してもよい。
 この場合、比較的少ない紫外線照射量であっても制御性T細胞を誘導することができる。したがって、紫外線の照射時間を短縮することができる。
 さらに、上記の制御性T細胞の誘導方法は、前記紫外線が照射された前記血液を2日以上培養する工程をさらに含んでもよい。
 この場合、血液中の制御性T細胞を適切に誘導することができる。
 また、上記の制御性T細胞の誘導方法は、前記抽出された血液を流路に導入する工程をさらに含み、前記紫外線を照射する工程は、前記流路内に導入された前記血液に対して前記流路の壁部を透過して前記紫外線を照射する工程であり、前記流路における前記紫外線の照射方向の長さが、70μm以上500μm以下であってよい。
 このように、紫外線の照射方向の長さが白血球よりも若干大きい流路に血液を導入することで、流路の長手方向(流通方向)に沿って白血球が整列されやすい環境を創出することができる。そのため、流路の壁部を透過した紫外線を適切に白血球に照射することができる。したがって、血液から白血球のみを分離して紫外線を照射するといった煩雑な血球分離処理が不要となる。
 本発明では、添加剤を用いることなく、制御性T細胞を誘導することができる。
 上記した本発明の目的、態様及び効果並びに上記されなかった本発明の目的、態様及び効果は、当業者であれば添付図面及び請求の範囲の記載を参照することにより下記の発明を実施するための形態(発明の詳細な説明)から理解できるであろう。
図1は、本実施形態における紫外線照射システムの構成例を示す図である。 図2は、本実施形態における紫外線照射装置の構成を模式的に示す図である。 図3は、Foxp3およびCD25の陽性の度合い表すグラフの説明図である。 図4Aは、紫外線照射後の制御性T細胞の変化を示す図である。 図4Bは、制御性T細胞の割合をグラフ化した図である。 図5は、制御性T細胞増加に関する紫外線の波長依存性を示す図である。
 以下、本発明の実施形態を図面に基づいて説明する。
 本実施形態では、血液中の制御性T細胞を誘導する方法について説明する。
 図1は、本実施形態における紫外線照射システム100の構成例を示す図である。
 紫外線照射システム100は、生体(例えば、患者等の人体)200から血液を抽出し、抽出した血液に特定波長域の紫外線を照射して制御性T細胞を誘導した後、当該血液を人体200の体内に戻すシステムである。
 ここで、制御性T細胞を誘導するとは、血液中の制御性T細胞の割合、具体的にはCD4陽性細胞中の制御性T細胞の割合を増やすことをいい、制御性T細胞の数を増やすこととエフェクターT細胞の数を減らすこととを含む。
 図1に示すように、紫外線照射システム100は、血液抽出装置10と、紫外線照射装置20と、血液注入装置30と、を備える。
 血液抽出装置10は、人体200から血液を抽出し、紫外線照射装置20へ供給する。このとき紫外線照射装置20に供給される血液は、血液成分が分離されていない血液(全血)である。
 紫外線照射装置20は、血液抽出装置10により抽出された血液に対して、260nm以上320nm以下の波長域の紫外線を照射する。紫外線照射装置20の具体的構成については後述する。
 血液注入装置30は、紫外線照射装置20により紫外線が照射された血液を人体200に戻す。
 図2は、紫外線照射装置20の構成を模式的に示す図である。
 紫外線照射装置20は、流路21と、入口側輸液チューブ22と、出口側輸液チューブ23と、輸送ポンプ24と、光源25と、供給用容器26と、回収用容器27と、を備える。
 流路21は、紫外線透過性を有する材料により構成された細管であり、血液抽出装置10により抽出された血液210が導入される。流路21は、例えば石英細管である。流路21は、例えば、長さ50mm、外径3mm、内径0.3mm(300μm)の円筒形状の細管とすることができる。
 入口側輸液チューブ22は、流路21の入口側に取り付けられ、出口側輸液チューブ23は、流路21の出口側に取り付けられている。各輸液チューブ22、23は、各々シリコンチューブにより構成することができる。
 輸送ポンプ24は、入口側輸液チューブ22に介在されている。例えば、輸送ポンプ24は、ペリスタリックポンプよりなる。輸送ポンプ24は、予め設定された流速で、血液210を、入口側輸液チューブ22から流路21を介して出口側輸液チューブ23へと流す。
 光源25は、流路21の上部に設けられている。光源25は、紫外線を含む光照射するLED照射器とすることができる。具体的には、LED照射器25のLEDは、260nm以上320nm以下、好ましくは260nm以上290nm以下の波長域の紫外線を含む光を放射する。光源25から放射された紫外線は流路21に照射され、流路21の壁を透過して流路21内の血液に照射される。
 なお、光源25はLED照射器に限定されるものではなく、XeClエキシマ放電ランプ、メタルハライドランプ、蛍光ランプ、水銀ランプなどであってもよい。また、光源25は、線状光源であってもよいし、面状光源であってもよい。
 また、光源25は、LED素子やランプから放射される光のうち、260nm以上320nm以下、好ましくは260nm以上290nm以下の波長域の紫外線のみを透過し、それ以外の波長域の光を遮断する光学フィルタを備えていてもよい。
 供給用容器26には、血液抽出装置10により抽出された血液210が収容されている。供給用容器26には、入口側輸液チューブ22の入り口が差し込まれる。
 回収用容器27には、出口側輸液チューブ23の出口が差し込まれる。回収用容器27は、流路21内において紫外線が照射され、出口側輸液チューブ23を輸送された血液210を回収する。
 以下、紫外線照射装置20の基本動作について説明する。
 紫外線照射装置20は、血液抽出装置10により抽出された血液210を収容した供給用容器26に入口側輸液チューブ22の入り口が差し込まれ、出口側輸液チューブ23の出口が回収用容器27に差し込まれた状態で、輸液ポンプ24を動作させる。すると、入口側輸液チューブ22に供給用容器26内の血液210が吸い上げられる。
 入口側輸液チューブ22に吸い上げられた血液210は、石英細管からなる流路21に導入される。このとき、予め設定された速度で、血液210が流路21を流れる。
 次に、紫外線照射装置20は、LED照射器からなる光源25のLEDを点灯し、流路21内に導入された血液210に対して流路21の壁部を透過して紫外線を照射する。
 流路21内において紫外線が照射された血液210は、出口側輸液チューブ23に送られ、出口側輸液チューブ23から回収用容器27に回収される。
 回収用容器27に回収された血液210は、血液注入装置30に送られ、血液注入装置30から人体200の体内に戻される。
 なお、本実施形態では、流路21が石英ガラスよりなる細管である場合について説明するが、流路21は、アルカリガラスや硼珪酸ガラスなどのガラスよりなる細管であってもよいし、シリコーン樹脂、シクロオレフィン樹脂(シクロオレフィンポリマー(COP)やシクロオレフィンコポリマー(COC)など)、アクリル樹脂などの合成樹脂によりなる細管であってもよい。
 また、流路21の形状は、上述したような形状に限定されるものではない。
 流路21の長手方向(血液210の流通方向)の長さは、光源25から放射される紫外線の強度や、流路21内を流れる血液210の流速などに基づいて、流路21内における血液210に対する紫外線の照射量が所望の照射量となるように適宜設定することができる。
 流路21の壁部の厚さは、流路21を構成する材料の紫外線透過率などに応じて適宜設定することができる。
 流路21における紫外線の照射方向(図2における上下方向)の長さは、白血球よりも若干大きい70μm以上500μm以下の範囲であればよい。なお、流路21における紫外線の照射方向の長さは、100μm以上300μm以下であることが好ましく、200μm以上300μm以下であることが特に好ましい。
 さらに、流路21の形状は、血液210が通る流通路を1つのみ有する円筒形状に限定されるものではなく、互いに独立した流通路を複数有するロッド体であってもよい。この場合、ロッド体における複数の流通路と光源25とは、全ての流通路について、他の流通路を介さないで光源25からの紫外線が照射されるように配置する。
 また、血液210が通る流通路の形状は、任意の形状とすることができる。なお、流通路の形状を、断面が矩形状とする場合は、紫外線の照射方向の長さを、上記に示したように白血球よりも若干大きい70μm以上500μm以下の範囲とする。それとともに、長手方向に白血球が整列されやすい環境をつくるために、照射方向と直交する、いわゆる幅方向の長さについても、白血球よりも若干大きい70μm以上500μm以下の範囲とする。
 以上のように、人体200から抽出された血液210に対して、260nm以上320nm以下の波長域の紫外線を含む光を照射することで、刺激物質などの添加物を用いることなく、血液210中の制御性T細胞を誘導することができる。
 近年の研究により、制御性T細胞は、Foxp3という因子とCD25という因子とについて、ともに陽性であり、Foxp3とCD25とが陽性であれば、その細胞が制御性T細胞であることがわかってきている。
 また、フローサイトメーター(蛍光活性化セルソーター(Fluorescence Activated Cell Sorting:FACS))を用いれば、Foxp3とCD25とがともに陽性である細胞、即ち制御性T細胞が、調査するサンプルの全細胞に対してどの程度存在するかを図示することができる。
 フローサイトメーターは、散乱や蛍光に基づき細胞の機能や状態を調べることができる装置である。
 例えば、フローサイトメーターは、図3に示すように、横軸にFoxp3に対する陽性の度合い、縦軸にCD25の陽性の度合いを取ったグラフを作ることができる。このグラフは、同図に示すように四つの領域[1]~[4]に分割することができる。ここで、右上の領域[1]は、Foxp3とCD25との両方に陽性な領域であり、この領域[1]に存在する細胞は制御性T細胞であると判断することができる。
 なお、図3の左上の領域[2]は、Foxp3に陰性であり、CD25に陽性な領域であり、左下の領域[3]は、Foxp3とCD25との両方に陰性な領域である。また、図3の右下の領域[4]は、Foxp3に陽性であり、CD25に陰性な領域である。
<実験1>
 本発明に基づく方法によって紫外線を照射した後の血液中の制御性T細胞の変化を確認するために、以下の実験を行った。
 図2に示す紫外線照射装置20と同様の循環系を構築し、サンプル血液210を循環系に流した。サンプル血液210としては、ヒト末梢血をリン酸緩衝食塩水にて20%に希釈したものを用いた。
 サンプル血液210が石英細管部分である流路21を通過する際に、LED照射器である光源25から紫外線を照射した。照射波長は290nmとした。また、流路21における紫外線の照射量が10mJ/cmとなるようにサンプル血液210の流速を設定した。
 また、サンプル血液210を上記循環系に通しただけで紫外線未照射の群を、比較対象群とした。
 紫外線を照射したサンプル血液210と、紫外線を未照射のサンプル血液210とを回収し、それぞれ、培養液を加えて37℃5%COの条件下にて、1日、2日、3日、5日、7日間培養した。その後、FACS解析により、全血からCD4陽性細胞をゲーティングし、CD25、Foxp3の変化を調べた。その結果を図4Aおよび図4Bに示す。
 図4Aの上段は紫外線を照射していないサンプルの結果、図4Aの下段は紫外線を照射したサンプルの結果である。それぞれ、紫外線照射後の1日後、2日後、3日後、5日後、7日後の結果を示している。この図4Aのグラフにおいて、各ドット1個が1個の細胞に対応している。
 図4Bは、制御性T細胞(Foxp3とCD25との両方に陽性な細胞(図3の領域[1]に存在する細胞)の割合をグラフ化したものである。白抜きの棒グラフが紫外線を照射していないサンプルの結果であり、斜線の棒グラフが紫外線を照射したサンプルの結果である。図4Bの値はN=3の平均値であり、エラーバーは標準偏差を示す。
 図4Bの結果からも明らかなように、紫外線を照射していないサンプルの場合、培養により制御性T細胞の割合はわずかに増加したものの、10%を超えることはなかった。一方、紫外線を照射したサンプルでは、培養日数2日目から制御性T細胞の割合が増加し、3日でピーク(約40%)に達した。
 このように、紫外線を照射したサンプルは、紫外線を照射していないサンプルを比較して、培養日数2日後以降、制御性T細胞(Foxp3とCD25との両方に陽性な細胞)の数が、明らかに増加することが確認できた。即ち、血液に対して紫外線を照射することにより、制御性T細胞の増加が期待できることが確認できた。
<実験2>
 制御性T細胞増加に関する、血液に照射する紫外線の波長依存性を確認するために、以下の実験を行った。
 図2に示す紫外線照射装置20と同様の循環系を構築し、サンプル血液210を循環系に流した。サンプル血液210としては、ヒト末梢血をリン酸緩衝食塩水にて20%に希釈したものを用いた。
 サンプル血液210が石英細管部分である流路21を通過する際に、LED照射器である光源25から紫外線を照射した。照射波長は260nm、290nm、310nm、365nmとした。また、流路21における紫外線の照射量[mJ/cm]は、表1に示す照射量となるようにサンプル血液210の流速を設定した。

 紫外線を照射したサンプル血液210を回収し、培養液を加えて37℃5%COの条件下にて、2日間培養した。実験1の結果によれば、紫外線照射後、培養日数が2日後に、明らかに制御性T細胞の誘導効果が見られたため、この日のサンプルを用いた。
 その後、FACS解析により、全血からCD4陽性細胞をゲーティングし、CD25陽性、Foxp3陽性細胞の割合を調べた。その結果を図5に示す。
 図5において、横軸は、照射する紫外線の照射量[mJ/cm]、縦軸は、サンプル内の全細胞に対する制御性T細胞の割合[%]である。
 この図5において、曲線a(■)は波長260nmの結果、曲線b(▲)は波長290nmの結果、曲線c(◆)は波長310nmの結果、曲線d(×)は波長365nmの結果を示している。
 通常、制御性T細胞の割合は3%から8%である。そこで、制御性T細胞の割合が10%以上になれば、制御性T細胞を誘導する効果があるといえる。
 波長365nmの場合は、照射量10mJ/cm~1200mJ/cmの範囲で照射しても、制御性T細胞の割合はほとんど増加せず、誘導効果は見られなかった。また、照射量10000mJ/cmにおいては、血液が光照射時に固着し血栓ができたことにより、照射を完了できなかった。
 一方で、波長310nmの場合は、照射量が10mJ/cm以下の領域では制御性T細胞の割合が10%未満であるが、照射量が10mJ/cmを超える領域で、制御性T細胞の割合が10%を超え、誘導の効果が認められた。
 さらに、波長290nm、260nmの場合は、ともに照射量が1mJ/cmで制御性T細胞の割合が10%を超え、誘導の効果が認められた。
 以上の結果より、波長260nmから310nmの領域の紫外線を照射することにより、制御性T細胞の誘導効果が得られることが確認できた。今回の試験ではUVB光源の代表光源として、ピーク波長310nmでスペクトル半値幅が約20nmの光源を用いた。紫外線はUVA、UVB、UVCに区分されており、各区分においては概ね類似した生体への作用を有する。そのため、ピーク波長310nmでの効果が確認されたことから、同様にピーク波長320nmまでは同様の効果が得られると考えられる。さらに、波長260nmから290nmの場合は、少ない照射量でも制御性T細胞の誘導効果が得られることが確認できた。つまり、波長260nmから290nmの範囲の紫外線を用いることにより、紫外線の照射時間の短縮化を図ることができる。
 以上説明したように、本実施形態における制御性T細胞の誘導方法は、生体(人体)から血液を抽出する工程と、抽出された血液に対して特定波長域の紫外線を照射する工程と、を含む。ここで、血液に対して照射する紫外線は、260nm以上320nm以下の波長域の紫外線を含む。
 このように、光エネルギーの比較的強いUVBやUVCの紫外線を用いることで、人体から抽出された血液に対して当該紫外線を照射するだけで、刺激物質などの添加剤を用いることなく血液中の制御性T細胞を誘導することができる。
 核酸(DNA)は、波長260nm付近に吸収ピークを有することが知られている。また、赤血球(ヘモグロビン)は、波長260nmよりも短波長の光に対して吸収が大きくなることが知られている。血液中には、白血球に比して赤血球が多く含有されている。血液に対して照射した紫外線が赤血球に吸収されてしまうと、本来作用させたい白血球に紫外線が照射されにくく、制御性T細胞の誘導効果が得られにくい。
 本実施形態では、波長260nmを下限として、血液に対して波長260nm以上の紫外線を照射する。したがって、DNAによる吸収が大きく、ヘモグロビンによる吸収が小さい紫外線を照射することができ、適切に制御性T細胞の誘導効果を得ることができる。
 また、血液に対して照射する紫外線を260nm以上290nm以下の波長域の紫外線とすれば、図5に示すように、比較的少ない照射量でも確実に制御性T細胞の誘導効果が得られるため、紫外線照射時間を短縮することができる。
 なお、血液に対して照射する紫外線の波長が290nmを超える場合(例えば310nmの場合)、制御性T細胞の誘導効果を得るためには、図5に示すように、紫外線照射量を20mJ/cm以上とする必要がある。本実施形態における方法は、一般的に用いられているような紫外線治療器のように人体の皮膚を通して紫外線を照射するものではなく、人体から血液を抽出(分離)して体外にて紫外線を照射するので、紫外線照射量を所望の照射量に容易かつ適切に設定することができる。
 さらに、本実施形態における制御性T細胞の誘導方法は、上述したように、上記の特定波長域の紫外線を照射した血液を2日以上培養する工程を含んでいてもよい。紫外線を照射した血液を2日以上培養することで、血液中の制御性T細胞を適切に誘導することができる。
 なお、紫外線を照射した血液を培養せずに直接体内に戻してもよい。この場合にも、体内において血液中の制御性T細胞の誘導が期待できる。
 また、本実施形態における制御性T細胞の誘導方法においては、上述したように、人体から抽出された血液を、紫外線の照射方向の長さが70μm以上500μm以下の流路21に導入し、流路21内に導入された血液に対して流路21の壁部を透過して紫外線を照射してもよい。
 このように、血液を導入する流路21の大きさを白血球の大きさよりも若干大きくすることで、流路21の長手方向(血液の流通方向)に沿って白血球が整列されやすい環境を創出することができる。そのため、流路21の壁部を透過した紫外線が流路21内を流れる血液中の赤血球によって吸収されることを抑制し、白血球に適切に照射させることができる。したがって、遠心分離により血液中の赤血球と白血球とを分離し、白血球のみを取り出して紫外線を照射するといった煩雑な血球分離処理が不要となる。
 以上のように、本実施形態においては、遠心分離などの煩雑な処理を必要とすることなく、また、刺激物質などの添加剤を用いることなく、紫外線照射のみにより血液中の制御性T細胞を誘導することができる。
 なお、上記において特定の実施形態が説明されているが、当該実施形態は単なる例示であり、本発明の範囲を限定する意図はない。本明細書に記載された装置及び方法は上記した以外の形態において具現化することができる。また、本発明の範囲から離れることなく、上記した実施形態に対して適宜、省略、置換及び変更をなすこともできる。かかる省略、置換及び変更をなした形態は、請求の範囲に記載されたもの及びこれらの均等物の範疇に含まれ、本発明の技術的範囲に属する。

Claims (4)

  1.  生体から血液を抽出する工程と、
     抽出された前記血液に対して260nm以上320nm以下の波長域の紫外線を照射する工程と、を含むことを特徴とする制御性T細胞の誘導方法。
  2.  前記紫外線を照射する工程では、
     前記抽出された血液に対して260nm以上290nm以下の波長域の紫外線を照射することを特徴とする請求項1に記載の制御性T細胞の誘導方法。
  3.  前記紫外線が照射された前記血液を2日以上培養する工程をさらに含むことを特徴とする請求項1または2に記載の制御性T細胞の誘導方法。
  4.  前記抽出された血液を流路に導入する工程をさらに含み、
     前記紫外線を照射する工程は、
     前記流路内に導入された前記血液に対して前記流路の壁部を透過して前記紫外線を照射する工程であり、
     前記流路における前記紫外線の照射方向の長さが、70μm以上500μm以下であることを特徴とする請求項1または2に記載の制御性T細胞の誘導方法。
