Abstract
본 발명자들은 선천성 면역 반응 및 T 세포 소진 경로(exhaustion pathway)가 소에서 보다 돼지에서 더 많이 과발현되어, 적응성, 체액성 면역반응이 잘 형성되지 않음을 확인하였다. 우리는 강력한 세포성 및 체액성 면역반응의 동시 유도 및 새로운 백신 보조제로서 T 세포 작용제를 사용하여 돼지에서 비정상적인 면역 반응의 개선을 위한 혁신적인 전략을 제시하였다. 이 결과는 소와 돼지의 면역 반응의 차이를 이해하기 위한 주요 단서를 제공할 수 있으며, 돼지에서 소에 비해 낮게 나타나는 면역반응 및 백신의 효능을 극대화할 수 있는 방법을 제시한다. {대표도} 도 8b
Description
본 발명은 신규한 면역증강제에 관한 것으로, 구체적으로는 비-전형적인 T 세포 작용제 및 전형적인 T 세포 작용제를 포함하는 신규 면역증강제 및 이를 포함하는 백신 조성물에 관한 것이다.
구제역(FMD)은 우제류(발굽이 둘로 갈라진 동물), 특히 소와 돼지에서 매우 빠르게 전파되는 급성 전염병으로 심각한 동물 생산성과 경제적 손실을 유발한다. FMD에 대한 대부분의 임상 연구는 돼지보다는 소에서의 백신 효능을 향상시키는 데 중점을 두었다. FMD 백신 생산을 위한 OIE (Office International Epizooties) 지침은 돼지가 아닌 소의 효능 시험 절차만 정의한다. 그러나 FMD의 중증도는 바이러스 균주 및 영향을 받는 숙주 종 둘 다에 크게 의존한다. 급성 임상 FMD는 다른 반추 동물보다 돼지에서 더 심각하며 감염된 돼지는 다량의 에어로졸 바이러스를 배출할 수 있어 심각한 질병의 전파 위험이 높다.
최근 한국의 백신 접종 동향에 따르면, FMD 백신은 소의 면역 반응을 효과적으로 유도하지만, 돼지에서 혈청 내 높은 바이러스 중화 항체 역가(Virus Neutralization (VN) Titer)에도 불구하고 구제역 바이러스 감염에 대한 완전한 방어를 위한 충분한 면역 반응은 아니다. 이러한 차이는 바이러스에 대한 축종별 감수성 또는 예방 접종에 대한 숙주의 면역 반응으로 설명할 수 있다.
비활성화 구제역 바이러스 (FMDV), 즉, 불활화 항원을 함유하는 백신 접종은 주로 숙주 방어 및 FMD 제어에 사용된다. 백신의 면역원성 및 효능을 향상시키기 위해, 오일 에멀젼 등의 아쥬반트(보좌제) 및 사포닌, 겔 등과 같은 면역 강화제 등이 백신 성분으로 첨가된다. 최근에, 사이토카인 (예를 들어, IL-15, IL-18 및 IFNα) 및 톨-유사 수용체 (TLR)-3 작용제로서 폴리 (I:C), TLR-9 작용제로서의 CpG 및 TLR-7/8 작용제로서의 R848 (Resquimod)을 포함하는 패턴 인식 수용체 (PRR) 리간드가 새로운 FMD 백신 아쥬반트로서 제안되었다. 그러나, 이러한 연구는 기본적인 수준에서 진행되었으며, FMDV 항원 또는 FMD 백신 매개 세포 및 체액성 면역 반응에 대해 더 많이 이해하기 위한 메커니즘 연구는 거의 수행되지 않았다.
본 발명의 목적은 선천성 면역반응과 체액성 면역반응의 linker로 작용하며, 숙주(host) 방어에서 "새로운 보호자"로 제안된 γδ (gamma delta) T 세포, iNK (invariant natural killer) T 세포, MAIT (mucosal-associated invariant T) 세포를 포함하는 unconventional T 세포 및 T 세포와 같은 conventional T 세포의 agonist(작용제)를 백신 보조제로 사용하여, 수지상세포(dendritic cells, DCs), 대식세포(macrophages, MΦs), 및 단핵구세포(monocytes) 등과 같은 항원제시세포(Antigen presenting cells, APCs)의 자극없이 T 세포를 직접적으로 자극하고, 보다 더 빠르고 더 강한 세포성 면역 반응을 이끌어냄으로써, 백신 접종 시 숙주의 초기 방어에 중요한 역할을 하며, 세포성·체액성 면역반응을 동시에 자극하여, 동물, 특히 돼지에서 강력한 메모리 반응 (memory response) 및 고 역가 항체를 유도하는 면역증강용 보조물질 및 이를 포함하는 구제역 백신 조성물을 개발하는 것이다.

상기와 같은 문제점을 해결하기 위해 본 발명은 소와 돼지 면역 세포에서 비-전형적인 T 세포 및 전형적인 T 세포의 직접적인 활성화에 의해 강력한 세포성 면역 반응을 유도하는 신규 면역증강제 조성물을 제공하고자 한다.
또한, 상기 면역증강제 조성물을 포함하는 백신 조성물을 제공하고자 한다.
상기와 같은 목적을 달성하기 위해 본 발명은 소와 돼지 면역 세포에서 강력한 세포 면역 반응을 유도하는 신규 면역증강제 조성물 제공한다.
상기 신규 면역증강제는 비-전형적인 T 세포 작용제(unconventional T cell agonist) 및 전형적인 T 세포 작용제 (conventional T cell agonist)를 유효성분으로 포함한다.
본 발명에서의 면역증강제(immunostimulant) 또는 아쥬반트(adjuvant)는 항원이 일으키는 면역반응을 증강시키는 물질을 의미한다. 면역증강제는 백신에서 소량의 항원으로도 동일한 효력을 나타낼 수 있어 종전의 절반 내지 3분의 1정도의 항원으로 백신을 만들 수 있다.
본 발명에서의 전형적인 T 세포 작용제는 RORγt (RAR-related orphan receptor gamma t)를 포함한다.
본 발명에서의 비-전형적인 T 세포 작용제는 상기와 같이 이미 공지되어 있는 전형적인 T 세포 작용제를 제외한 T 세포 작용제를 의미한다. 상기 비-전형적인 T 세포 작용제는 바람직하게는 γδ T 세포 작용제, iNKT 세포 작용제, MAIT 세포 작용제일 수 있으나 이에 제한되는 것은 아니다.
상기 비-전형적인 T 세포 작용제 또는 전형적인 T 세포 작용제는 면역증강제 조성물의 내지 0.01 내지 1 중량%, 바람직하게는 0.1 내지 0.5 중량%, 더욱 바람직하게는 0.2 내지 0.3 중량% 포함될 수 있다. 상기 범위 미만이면 면역증강 효과가 나타나지 않을 수 있고, 상기 범위를 초과하면 독성이 유발될 수 있다.
본 발명의 면역증강제 또는 면역증강제 조성물은 상기 성분 이외에 당업계에서 널리 알려진 오일(또는 오일 에멀젼) 및 유화제, 겔 등을 추가로 포함할 수 있다.
상기 오일(또는 오일 에멀젼)은 ISA 201, ISA 61, ISA 50, ISA 206, ISA 207 일 수 있으나 이에 제한되지 않는다.
상기 유화제는 유화제로 일반적으로 인정되는 물질들, 예를 들면 TWEEN® 또는 SPAN® 생성물 라인의 다른 생성물(각각 폴리에톡시화 소르비톨의 지방산 에스테르, 및 지방산-치환된 소르비탄 계면활성제) 및 PEG-40 피마자유 또는 다른 페길화(PEGylated) 수소화 오일과 같은 다른 용해도 개선제들을 포함할 수 있으나, 이에 제한되는 것은 아니다.
또한, 면역증강제(또는 아쥬반트) 조성물은 일반적으로 오일 제형에서 면역 증강 효과가 우수하지만, 본 발명에 따른 면역증강제 조성물은 비-오일 제형에서도 면역 증강 효과가 뛰어나다.
상기 면역증강제를 제조하기 위해 당 업계에서 통상적으로 사용되는 첨가제, 부형제, 담체 등을 추가적으로 더 포함할 수 있고, 당 업계에서 면역증강제제를 제조하기 위해 사용하는 통상적인 제조 방법에 의해 제조될 수 있다.
본 발명의 면역증강제 조성물 투여 후 나타나는 면역 증강 효과는 면역 매개 물질 또는 세포에 의한 것일 수 있으며, 구체적으로는 세포성 면역, 점막 면역,체액성 면역 증강 효과를 포함하나, 이에 제한되지는 않는다.
상기 면역은 바이러스, 곰팡이, 박테리아 및 기생충 중 어느 하나의 병원체에 대한 감염 또는 암에 대한 면역을 포함할 수 있으나, 이에 제한되지는 않는다.
상기 바이러스는 구제역 바이러스(FMDV), 리슈마니아(Leishmania), 인간 면역결핍 바이러스(Human Immunodeficiency virus, HIV), C형 간염 바이러스(Hepatitis C virus, HCV), E형 간염 바이러스(Hepatitis E virus, HEV), A형 간염 바이러스(Hepatitis A virus, HAV), B형 간염 바이러스(Hepatitis B virus, HBV), 결핵 (tuberculosis), 단순 헤르페스 바이러스(Herpes Simplex virus, HSV), 기생충을 일으키는 말라리아(malaria causing parasites), 인유두종 바이러스(Human Papilloma virus, HPV), 인플루엔자 바이러스(influenza virus), 홍역 바이러스(measles virus), 볼거리 바이러스(mumps virus), 에볼라 바이러스(Ebola virus), 호흡기 세포융합 바이러스(Respiratory Syntial virus, RSV), 웨스트 나일 바이러스(West Nile virus, WNV) 등을 포함하나 이에 제한되지는 않는다.
또한, 본 발명은 상기 면역증강제 조성물을 포함하는 백신 조성물을 제공한다.
상기 면역증강제 조성물은 백신 조성물 전체 중량 대비 30 내지 70 중량%, 바람직하게는 40 내지 50 중량% 포함될 수 있으나, 이에 제한되는 것은 아니다. 상기 범위 미만이면 백신의 효과가 나타나지 않을 수 있고, 상기 범위를 초과하는 경우 투여 시 독성이 나타날 수 있다.
상기 백신 조성물은 당 업계에서 백신 조성물을 제조할 때 통상적으로 사용되는 첨가제, 부형제, 담체 등을 추가적으로 더 포함할 수 있다.
또한, 상기 백신 조성물은 당 업계에서 백신 조성물을 제조하기 위해 사용하는 통상적인 제조 방법에 의해 제조될 수 있다.
본 발명에서 상기 백신은 그 종류가 제한되는 것은 아니나, 바람직하게는 바이러스 백신, 더욱 바람직하게는 구제역 백신일 수 있다.
구제역 백신 조성물에 본 발명에 따른 면역증강제가 포함되는 경우, 상기 구제역 백신 조성물은 구제역 바이러스 O 혈청형, A 혈청형, Asia 혈청형, C 혈청형, SAT1 혈청형, SAT2 혈청형, SAT3 혈청형 등에 대해 면역 효과가 나타날 수 있고, 바람직하게는 구제역 바이러스 O 혈청형, A 혈청형에 대해 보다 더 강력하게 나타날 수 있다.
상기 백신 조성물은 설하 투여, 경피 투여, 직장 투여, 경점막투여, 국소적 투여, 구강 투여, 흉막내 투여, 정맥내 투여, 동맥내 투여, 복강내 투여, 피하 투여, 근육내 투여, 비강내 투여, 척추강내 투여, 관절내 투여 등의 방법으로 투여될 수 있다.
본 발명은 소, 돼지의 선천성 세포성 면역반응을 강화하여 체액성 면역반응을 보다 효율적으로 향상시킬 수 있는 새로운 면역증강제 및 이를 포함하는 백신 조성물을 제공할 수 있다.
도 1은 불활성화된 FMDV O/TWN/97 항원으로 유도된 소 및 돼지에서의 PBMCs, 림프구, 단핵구 및 T 세포 증식 결과를 나타낸 것이다.
도 2a 내지 2d, 도 3은 FMDV (O/TWN/97-R) 항원에 의해 매개된 돼지 면역 세포와 소 면역 세포에서 전 염증성 사이토카인 발현 결과를 나타낸 것이다.
도 4는 FMDV 항원이 돼지의 Mo-DCs 및 Mo-MΦs에서 사이토카인 발현을 직접 자극함을 보여준다.

이하, 본 발명을 실시예 및 실험예에 의해 상세히 설명한다.
단, 하기 실시예 및 실험예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 실험예에 한정되는 것은 아니다.
<실험 재료 및 방법>
1. 항원의 정제 및 불활성화
정제된 불활성화 바이러스 항원을 역 유전학에 의한 P1의 표현형 교체(참조 서열)에 대해 구축된 FMDV O/TWN/97-R (GenBank AY593823; P1)로 감염된 BHK-21 세포에서 제조하였다.
바이러스 감염의 경우, 배양 배지를 무 혈청 둘베코 변형 이글 배지(DMEM; HyClone, Logan, UT, USA)로 교체하고, 세포를 37℃, 5% 이산화탄소에서 1 시간 동안 인큐베이션함으로써 바이러스에 접종하였다. 이어서 세포 외 바이러스를 제거하였다. 감염 후 24 시간에, 진탕 배양기에서 24시간 동안 0.003N 이원 에틸 에니민을 2회 처리하여 바이러스를 불활성화시킨 후 폴리에틸렌 글리콜(PEG) 6000 (Sigma-Aldrich, St. Louis, MO, USA)으로 농축시켰다. 바이러스 농축물을 15-45% 자당 밀도 구배로 층을 이루고 원심 분리하였다.
초 원심분리 후, 원심분리 튜브의 바닥을 천공하고 1mL 분획을 수집하였다. 각각의 분획의 샘플에서 FMDV 입자의 존재는 측면 유동 장치인 UA-6 (BioSign FMDV Ag; Princeton BioMeditech, Princeton, NJ, USA)를 사용하여 광학 밀도에 의해 확인되었다. 현장 실험에 사용하기 전에, PEG 전처리된 상층액을 ZZ-R127 및 BHK-21 세포를 통해 적어도 2회 통과시켜 세포 변성 효과 (CPE)가 발생하지 않았음을 확인함으로써, 상층액에 생 바이러스의 부재를 확인하였다.
2. PBMCs 분리
항원 매개 면역 반응을 조사하기 위해 돼지와 소의 전혈을 경기도 동물위생시험소 에서 기증받았다. 전혈(15 mL)을 BD Vacutainer 헤파린 튜브 (BD Biosciences, Becton, Dickinson and Company, Franklin Lakes, NJ, USA)에 수집하고, Ficoll-PaqueTM PLUS (GE Healthcare Bio-Sciences Corp., Piscataway, NJ, USA)로 구배, 원심 분리하고, 잔류 적혈구를 암모늄-클로라이드-포타슘 (ACK) 용해 완충제 (Gibco, Carlsbad, CA, USA)로 처리하여 용해시켰다. PBMCs를 2% 소 태아 혈청 (FBS) (Gibco)이 보충된 Ca2+ 및 Mg2+ (Gibco)가 포함되지 않은 둘베코(Dulbecco)의 PBS에 현탁시키고 부피 유량 세포 계수기 (MACSQuant Analyzer, Miltenyi Biotec, Bergisch Gladbach, Germany)를 사용하여 계수하였다. 모든 세포는 사용 직전에 새로 분리하였고, 동결 보존된 세포는 어떠한 실험에도 사용하지 않았다. 정제된 PBMCs를 10% FBS (HyClone, Logan, Utah, USA), 3mM L-글루타민 (Sigma-Aldrich) 및 100 U/mL 페니실린-스트렙토마이신 (Sigma-Aldrich)이 보충된 RPMI 1640 (Gibco) 배지에 재현탁시켰다. 이어서 정제된 PBMCs를 25cm2 조직 배양 플라스크 (Eppendorf, Germany, Hamburg)에 시딩하고 5% CO2와 함께 37℃에서 배양하여 새로 분리된 단핵구가 부착되도록 하였다. 3 시간의 배양 후, 림프구 분리를 위해 비 부착성 세포를 수집하였다. 나머지 부착 세포는 각각의 플라스크에 4 mL의 RPMI 1640 성장 배지를 첨가하기 전에 둘베코(Dulbecco) 인산염 완충 식염수 (DPBS) (Gibco)로 광범위하게 세척하였고, 이어서 5% CO2 인큐베이터에서 37℃에서 배양하였다.
3. 자기 활성화 세포 분류 (MACS)에 의한 세포 분리
단핵구, 림프구 및 T 세포를 PBMCs로부터 분리하였다. 1차 면역 세포의 분리를 위해, 부착성 PBMCs로부터의 단핵구 및 비 부착성 세포로부터의 T 세포를 MACS 분류를 통해 정제하였다. PBMCs로부터의 부착 세포 및 비 부착 세포를 MACS 완충액 (0.5 % BSA 및 2mM EDTA가 보충된 1x PBS)에 간단히 재현탁시켰다. 단핵구 및 T 세포는 각각 제조업체의 지시에 따라 단핵구 단리 키트 및 Pan T 세포 단리 키트 (Miltenyi Biotec)와 함께 자기 마이크로 비드를 사용하여 분류하고 형광-활성화 세포 분류기 (FACS, MoFlo® AstriosTM Cell Sorter, Beckman Coulter, Brea, CA, USA)를 사용하여 추가로 분류하였다. 분리된 세포의 정제는 유세포 분석 (MACSQuant Analyzer, Miltenyi Biotec)에 의해 확인되었고 FlowJo 소프트웨어 버전 vX.0.7 (TreeStar Inc., Ashland, OR, USA)로 분석되었다. 분류된 세포의 순도는 95%보다 높았다.
4. Mo-DCs 및 Mo-MΦs 생성
Mo-DCs의 분화를 위해 분리된 단핵구를 10% FBS (HyClone), 3mM L-글루타민 (Sigma-Aldrich), 100 U/mL 페니실린-스트렙토마이신 (Sigma-Aldrich), 50 ng/mL GM-CSF 및 IL-4 (Miltenyi Biotec)이 보충된 완전한 RPMI 1640 배지(Gibco)에서 배양하여 7일 동안 미성숙 DCs를 생성시켰다. 3 일째에, 동일한 부피의 상기 언급된 배지를 첨가하고, 7 일째에, 세포 용해 전에 격렬한 세척에 의해 비 부착성 오염 세포를 제거하였다. 순수한 DCs의 분리를 위해, 제조업체의 지침 (Miltenyi Biotec)에 따라 항-CD11c 자성 비드를 사용하였다. CD11c+/MHC II+ 세포의 순도는 95% 이상이며, 세포는 5% CO2 배양기에서 37℃로 유지되었다.
Mo-MΦs의 분화를 위해, 단리된 단핵구를 세포 106/mL로 12-웰 플레이트에 시딩하고 10% FBS (HyClone), 1x MEM 비 필수 아미노산, 1 mM 피루브산 나트륨, 0.05 mM 2-머캅토 에탄올 (Sigma-Aldrich), 100 U/mL 페니실린-스트렙토마이신 (Sigma-Aldrich) 및 50 ng/mL 대식세포 콜로니 자극 인자 (M-CSF) (Abcam, Cambridge, MA, USA)가 보충된 RPMI 1640 배지(Gibco)에서 6일 동안 배양하였다. 추가의 M-CSF를 2일에 첨가하였고, 전체 배지를 4일에 새로 교체하였다. CD11b+/F4/80+ 세포의 순도는 95% 초과였다. 세포를 5% CO2 인큐베이터에서 37℃로 유지시켰다.
5. 성숙한 DCs의 준비

6. M1/M2 MΦs 편광

분리되거나 분화된 세포 (1x106 세포/웰)를 10% 태아 송아지 혈청 (HyClone), 3 mM L-글루타민 (Sigma-Aldrich), 10 mM HEPES, 100 U/mL 페니실린-스트렙토마이신 (Sigma-Aldrich) 및 0.05 mM 2-머캅토 에탄올 (Sigma-Aldrich)이 보충된 RPMI 1640 배지 (Gibco)로 구성된 완전 배지에서 5% CO2, 37℃ 인큐베이터에서배양하였다(Sigma-Aldrich). 자극을 위해 각 항원 1 μg으로 처리되었다. 처리 후 특정 시점 (0, 6, 12, 24, 48, 72, 96, 120, 144, 168, 192, 216 및 240h)에서, DNA 합성 동안 BrdU의 혼입을 기초로 한 BrdU 세포 증식 분석 키트 (세포 Signaling Technology, MA, USA, MA, USA)를 사용하여 제조사의 지시에 따라 세포 증식을 시험하였다. 간략히, 10 μM BrdU를 세포 배양물에 첨가하고 37℃에서 4시간 동안 배양하였다. 이어서 세포를 고정시키고 항-BrdU 마우스 모노클로날 항체와 함께 배양한 후, horseradish 퍼옥시다제-접합된 염소 항-마우스 항체를 사용하였다. 발색 기질인 테트라메틸벤지딘을 발색에 사용하였다. 450/550 nm의 이중 파장에서 흡광도를 측정하였다. Cell Titer-BlueTM 분석 키트 (Promega, Madison, WI, USA)를 사용하여 세포 생존력을 모니터링 하였다. 배지-단독 대조군 처리와 비교하여, 실험 처리는 세포 생존력에 영향을 미치지 않았다.
8. ELISA
소 및 돼지 IL-1β, IL-6, IL-10, IL-12/23p40, IL-23 및 TNFα에 대한 ELISA (DuoSet, R&D Systems, Minneapolis, MN, USA; Cloud-Clone Corporation, Houston, USA)는 제조자의 지시에 따라 세포 배양 상층액을 이용하여 수행하였다.
9. 식세포작용(phagocytosis)의 억제
식세포작용을 억제하기 위해, Mo-DCs 및 Mo-MΦs를 항원 처리 전에 45분 동안 5 μg/mL 시토칼라신 D (CytD) (Sigma-Aldrich)와 인큐베이션 하였다. 이어서, CytD-처리된 Mo-DCs 및 Mo-MΦs를 1 μg/mL 항원과 함께 배양하였다. 6 시간 후, 배양된 상층액을 ELISA를 위해 수집하였다.
10. 소와 돼지
소와 돼지의 자연 상태에서 면역 반응의 근본적인 차이와 FMDV O 항원-매개 면역 반응 및 관련 메커니즘을 이해하기 위해 소와 돼지를 사용한 현장 실험은 Lee et al.에 설명된 방법에 따라 수행되었다. FMD 항체-음성 동물을 사용하였다 (소는 5 개월령, 돼지는 10-12 주령). 소와 돼지를 두 그룹으로 나누었다(n=5/그룹). 연구동안 동물을 격리된 상태로 유지시켰다. 연구는 농림축산검역본부 동물실험윤리위원회(승인 번호 IACUC-2018-800 및 IACUC-2019-185)의 승인을 얻어 기관 지침에 따라 수행되었다.
11. 백신 접종 및 샘플링
FMD 항원으로서 O/TWN/97-R Ag를 사용하였고, 양성 대조군에 대한 백신 조성물은 다음과 같았다: 15 μg의 O/TNW/97-R 항원 (소 및 돼지 사용의 경우 1회 용량), ISA 206 (50%, w/w), 10% Al (OH)3 및 150 μg Quil-A를 포함하는 단일 용량으로 제조된 1 mL 백신. 나이브(naive) PBMCs 분리를 위해, 전혈을 나이브 대조군(구제역 항체-음성)의 소, 돼지로부터 수집하였다. 백신 접종은 28일 간격으로 2회 수행하였고, 양성 대조군 동물의 목에 근육 내 깊은 경로를 통해 1mL 백신 (1 회 용량)을 주사하였다. 혈청학적 분석을 위해 소 및 돼지에서 0, 14, 28, 42, 56, 70 및 84 dpv에서 혈액 샘플을 수집하고 소 및 돼지로부터 PBMCs 분리를 위해 28 dpv (1회 접종 후 28일째, 2 회 백신 접종 전)에 혈액 샘플을 수집하였다. 동물은 체온, 백신 접종 부위의 증상 및 식욕에 대해 매일 모니터링 하였다. 테스트가 수행될 때까지 혈청 샘플을 -80℃에서 보관하였다.
12. RNA 서열 분석 (RNA-Seq)
RNA-Seq 분석을 위해, 나이브 대조군 (n=3/그룹) 및 양성 대조군 (n=3/그룹) (28 dpv)의 소 및 돼지의 전혈로부터의 PBMCs를 Ficoll-PaqueTM PLUS를 사용하여 Lee, M.J. et al. Mincle and STING-Stimulating Adjuvants Elicit Robust cellular Immunity and Drive Long-Lasting Memory Responses in a Foot-and-Mouth Disease Vaccine. Front Immunol, 2509 (2019) (이하, Lee et al)에 의해 기술된 방법에 따라 (GE Healthcare Bio-Sciences Corp., Piscataway, NJ, USA) 밀도 구배 원심 분리하였다.
13. 라이브러리(Library) 구축 및 시퀀싱
높은 처리량의 소 및 돼지 전사체 데이터를 얻기 위해 Illumina 기반 차세대 염기서열 분석(next generation sequencing, NGS)을 구현하였다. 제조사의 프로토콜에 따라 TRIzol Reagent (Invitrogen) 및 RNeasy Mini Kit (QIAGEN)를 사용하여 소 및 돼지 PBMCs로부터 총 RNA를 개별적으로 추출하였다. 이어서, 총 RNA를 Nanodrop 분광 광도계(Thermo Scientific, Wilmington, USA, USA)를 사용하여 정량하고 RNA 6000 Nano 분석 키트 (Agilent Technologies, Santa Clara, USA) 및 Bioanalyzer 2100 (Agilent)에 의해 품질을 평가하였다. NGS 시퀀싱 라이브러리는 제조사의 프로토콜에 따라 TruSeq RNA 샘플 준비 키트 (Illumina, San Diego, CA, USA)를 사용하여 1 μg의 총 RNA로부터 생성되었다. 즉, 폴리-A 함유 RNA 분자는 폴리-T 올리고 부착 자성 비드를 사용하여 정제되었다. 정제 후, 총 폴리 (A)+RNA를 고온에서 2가 양이온을 사용하여 작은 조각으로 단편화하였다. 절단된 mRNA 단편을 랜덤 프라이머를 사용하여 제1가닥 cDNA로 역전사시켰다. 짧은 단편을 QiaQuick PCR 추출 키트로 정제하고 최종 회수 및 폴리 (A)의 첨가를 위해 용리 완충액으로 분해하였다. 이어서, 짧은 단편을 시퀀싱 어댑터와 연결하였다. 각 라이브러리는 서로 다른 MID 태그를 인접시켜 분리되었습니다. 이어서, 생성된 cDNA 라이브러리를 NovaSeq™ 6000 시스템 (Illumina)으로 샘플에 대해 페어드-엔드 시퀀싱 (2x101bp) 하였다.
14. 유전자 발현 분석
저-품질 염기(PHERD score(Q)<20) 및 어댑터 오염은 매개변수로 ‘ILLUMINACLIP:TruSeq3-SE:2:30:10 LEADING:3 SLIDINGWINDOW:4:15 MINLEN:36’을 사용하여 Trimmomatic v.0.36에 의해 제거되었다. 품질 점수 확인 및 판독 길이 확인 후 RNA-Seq 판독 값은 STAR를 기본 매개변수로 사용하고 기대 최대화 (RSEM, RNA-Seq)를 사용하여 참조 Bostaurus 게놈 (2018년 4월 발행; ARS-UCD1.2; GCA_002263795.2)에 매핑(Mapping)되었다. 게놈에서 각 유전자/전사체에 대한 발현 값을 얻기 위해 Expectation-Maximization 방법에 의한 발현을 추정하였다. RSEM에 의해 추정된 판독 카운트는 edgeR v3.22.5에 적용되어 통계적 유의성과 함께 미분 발현 점수를 얻었다. 또한, 필터, 즉 TPM (transcripts per million) ≥ 0.3, 판독 카운트 ≥ 5 및 log2 폴드(fold) 변화 ≥ 1이 차등적으로 표현된 전사체의 선택에 적용되었다. 마지막으로, 발현된 전사체 (즉, TPM ≥ 0.3 및 판독 횟수 ≥ 5)를 분석하여 각 조건 및 유전자 패밀리 (면역 유전자, T 세포 마커 및 TLR, CDS, CLR 신호 전달 경로 유전자)에서의 발현 패턴을 보여주었다.
15. 독창성 경로(Ingenuity Pathway) 분석 (IPA)
이어서, 발현 프로파일을 유사한 발현 패턴의 클러스터로 분류하였다. 그런 다음 IPA (QIAGEN Inc., https://www.qiagenbioinformatics.com/products/ingenuity-pathway-analysis)를 사용하여 풍부한 경로, 네트워크 및 기능을 분석하였다. 마지막으로, 사내 R script을 사용하여 중요한 경로에 관련된 모든 유전자를 보여주는 이진 히트 맵을 만들었다.
<강력한 백신 보조제로서의 비-전형적인 T 세포 작용제의 효과의 평가 및 실험 방법>
1. 마우스
연령- 및 성별-일치 야생형 C57BL/6 마우스 (6-7 주령 암컷)는 KOSA BIO Inc. (경기)에서 구입하였다. 모든 마우스는 농림축산검역본부 내 동물 생물안전 3등급 (ABSL3)의 특정 병원체가 없는 (specific pathogen free, SPF) 동물 시설의 미세 분리기 케이지에 넣었다. 연구는 기관 지침에 따라 농림축산검역본부 동물실험 윤리위원회(승인 번호 IACUC-2018-800 및 IACUC-2019-185)의 승인을 받아 수행되었다.
2. 비 전형적인 T 세포 작용제 및 전형적인 T 세포 작용제-매개된 보조성 및 숙주 방어
FMD 백신 보조제로서 강력한 세포성 및 체액성 면역 반응의 동시 유도 및 비-전형적인 T 세포 작용제 및 전형적인 T 세포 작용제의 잠재력을 평가하고, FMDV 감염에 대한 보호 효과를 조사하기 위해, 제시된 전략을 사용하여 실험을 수행하였다(도 7a). (그룹 당 n=10). O/TWN/97-R 항원은 비활성화된 FMDV 항원으로 사용되었다. PC 그룹에 대한 백신 조성물은 다음과 같다: O/TWN/97-R 항원 (15 μg/용량/mL, 소 및 돼지 사용을 위한 1/10 용량), 10% Al(OH)3 및 15 μg/ Quil-A/마우스를 사용하여 총 부피는 100 μL이다. 실험군의 모든 마우스는 보조제(면역증강제)로서 비-전형적인 T 세포 작용제 또는 전형적인 T 세포 작용제를 면역증강제 조성물의 전체 중량의 약 0.2 중량%가 되고, 백신 전체 중량 대비 약 0.1 중량%가 되도록 첨가하여 PC 군과 동일한 조성으로 백신을 제공받았다.
이들 실험에 사용된 비-전형적인 T 세포 작용제 및 전형적인 T 세포 작용제는 Sigma-Aldrich (γδ T 세포 작용제; Isopentenyl pyrophosphate trilithium salt, IPP (I), (E)-1-Hydroxy-2-methyl-2-butenyl 4-pyrophosphate lithium salt, HMP (H), Abcam (iNKT 세포 작용제; α-Galactosylceramideand, α-Galcer (G), T 세포 작용제 (RORγT (R)) 및 Cayman (MAIT 세포 작용제; 6-Formylpterin (F), Cayman Chemical, Ann Arbor, MI, USA)에서 각각 구입하였다.
음성 대조군 그룹의 마우스는 동일한 경로를 통해 동일한 부피의 인산 완충 식염수 (PBS, pH 7.0)를 투여받았다. 간략하게, 백신 접종을 35일 간격으로 2회 수행하고, 마우스 허벅지 근육에 근육 내 백신 접종하였다. 나중에, 84 dpv 또는 168 dpv에 마우스는 복강 내 주사에 의해 FMDV (O/VET/2013의 100 LD50, ME-SA 토포타입)로 공격접종 되었다. 마우스의 생존율 및 체중은 최대 7 dpc (7 days post challenge, 공격접종 후 7일)까지 모니터링 하였다. 또한, 0, 7, 14, 28, 56, 84 및 168 dpv에서 마우스로부터 샘플링된 혈청을 A형 구조적 단백질 효소-연결 면역 흡착 분석 (structural protein (SP) A ELISA) 및 바이러스 중화 (virus neutralization, VN) 역가 (titers)를 통해 분석하여 유도된 세포성 및 체액성 면역 반응을 조사하였다.
3. 혈청학적 분석
혈청에서 SP 항체를 검출하기 위해, Lee et al.에 기술된 바와 같이 PrioCHECK FMDV 유형 O ELISA 키트 (Prionics AG, Switzerland)를 사용하였다. ELISA 플레이트에서의 흡광도를 퍼센트 억제 (PI) 값으로 전환시켰다. PI 값이 50% 이상일 때, 동물은 항체 양성으로 간주되었다.
VN 테스트는 Lee et al.에 설명된 대로 세계 동물 보건 기구 (OIE) 매뉴얼에 따라 수행되었다. 혈청을 56℃, 수조에서 30분 동안 열처리를 통해 불활성화시켰다. 세포 밀도를 70% 단일 층을 형성하도록 조정하고, 혈청 샘플의 2배 연속 희석물 (1 : 8 - 1 : 1024)을 제조하였다. 이어서, 희석 혈청 샘플을 100-조직 배양 감염 용량 (TCID)50/0.5mL 상동 바이러스와 37℃에서 1시간 동안 인큐베이션 하였다. 1 시간 후, LF-BK (소 신장) 세포 현탁액을 모든 웰에 첨가하였다. 2-3 일 후, CPE를 확인하여 역가를 결정하였으며, 이는 바이러스의 100 TCID50을 중화시키는 데 필요한 상호 항체 희석의 Log10으로 계산되었다.
4. PBMCs 분리
FMD 항체 음성 동물을 돼지 PBMCs 분리를 위한 공여자 (n=3/그룹)로 사용하였다. 전혈 (15 mL/각 공여자)을 BD Vacutainer 헤파린 튜브에서 독립적으로 수집하였다. PBMCs 분리에 대한 자세한 프로토콜은 위에서 언급했다. 모든 세포는 사용 직전에 신선하게 분리되었고, 어떤 실험에서도 동결 보존된 세포는 사용되지 않았다. 이어서 정제된 PBMCs를 10% FBS (HyClone), 3mM L-글루타민(Sigma-Aldrich) 및 100 U/mL 페니실린 스트렙토마이신 (Sigma-Aldrich)이 보충된 RPMI 1640 (Gibco, Carlsbad, CA, USA)에 재현탁시켰다. 96-웰 플레이트에서 웰당 1x105 세포로 플레이팅하고, 5% CO2와 함께 37℃에서 인큐베이션 하였다. 3시간의 배양 후, 배양 배지를 FMDV O (O/TWN/97-R) 항원 단독 또는 다양한 비 전형적인 T 세포 작용제 및 T 세포 작용제 또는 PRR 리간드와 함께 항원으로 자극하기 전에 무 혈청 배지로 교체하였다.
5. 돼지 PBMCs에서 BrdU 통합 분석
세포 증식 분석에 대한 상세한 프로토콜은 상기에 언급되어있다. 비 전형적인 T 세포 작용제 및 전형적인 T 세포 작용제로 처리한 후 12 및 36시간에, 세포 증식을 제조자의 지시에 따라 시험하였다.
6. RNA-Seq
RNA-Seq 분석을 위해, 혈청 내 FMD 항체 음성 돼지의 전혈(n=3/그룹)로부터 돼지 PBMCs를 분리하였다. 단리된 PBMCs에 γδ T 세포 작용제 (이소펜틸 피로포스페이트 트리리튬 염, IPP (I)), (E)-1-하이드록시-2-메틸-2-부테닐 4-피로포스페이트 리튬 염, HMP (H)), iNKT 세포 작용제 (α-갈락토실세라마이드, α-Galcer (G), Abcam) 및 MAIT 세포 작용제 (6-포르밀프린, 6-Formylpterin (F), Cayman Chemical)를 포함한 비 전형적인 T 세포 작용제, T 세포 작용제 (RORγT (R), Abcam) 및 PRR 리간드 (레시퀴모드 (Resiquimod, R848, TLR-7/8 효능제) 및 트레할로스-6, 6-디베헤네이트 (TDB, Mincle 작용제); TDB 및 비스-(3'-5')-시클릭 이량체 구아노신 모노 포스페이트 (c-di-GMP, STING 작용제, InvivoGen, San Diego, CA, USA)를 FMDV O (O/TWN/97-R) 항원과 함께 처리하였고, 12시간 인큐베이션 후, PBMCs를 수집하여(prepartion) qRT-PCR을 위해 RNA를 추출하였다.
라이브러리 구성 및 서열 분석, 유전자 발현 분석 및 IPA를 상기한 바와 같이 수행하였다.
7. 통계
달리 언급되지 않는 한 모든 정량적 데이터는 평균 ± SEM으로 표시된다. 그룹 간 통계적 유의성에 대한 값의 비교는 Tukey의 다중 비교 테스트 또는 두 개의 데이터 포인트를 비교하기 위한 스튜던트 t 테스트와 함께 일원 분산 분석을 사용하여 수행되었다. 통계 분석에는 GraphPad Prism 8.3.1 소프트웨어 (GraphPad Software, San Diego, USA)를 사용하였다.
<예비실험예> 소와 돼지의 면역 차이 평가
1. FMDV 항원은 돼지 유래의 것보다 소 PBMCs, 림프구, 단핵구 및 T 세포에서 더 강력한 증식을 유발한다.
BrdU 세포 증식 분석을 사용하여 소 및 돼지 PBMCs, 림프구, 단핵구 및 T 세포의 O/TWN/97-R 항원-매개 증식을 관찰하였다. 모든 소 세포 유형의 증식은 돼지 세포의 것보다 유의하게 더 높았다 (p < 0.001) (도 1의 a-d).
2. FMDV 항원은 돼지 면역 세포와 비교하여 소 면역 세포에서 전 염증성 사이토카인 발현을 유의하게 유도한다.
O/TWN/97-R 항원-매개 사이토카인 발현 분석은 돼지 PBMCs에서 사이토카인 발현이 12 시간 내지 48 시간에서 피크에 도달한 후 급격히 감소하였지만, 소 PBMC에서 24 시간 이내에 현저하게 증가하고 240 시간까지 이 수준을 유지함을 보여주었다 (도 2의 a-d, 표 1).
림프구에서, O/TWN/97-R 항원-매개 사이토카인 발현은 돼지 세포에서보다 소 세포에서 현저하게 더 높았다 (도 2b의 e-h, 표 1). 돼지 단핵구에서 O/TWN/97-R 항원-매개 사이토카인의 발현에 관해서는 (도 2c의 i-l, 표 1), IL-2, IL-6, TNFα 및 IFNγ는 소 단핵구에서 그보다 현저히 높았다. 소 및 돼지 T 세포에서 O/TWN/97-R 항원-매개 사이토카인 발현의 시간적 변화를 평가하였다(도 2d의 m-p, 표 1). 사이토카인 발현은 T 세포 유형 모두에서 24 시간까지 급속히 증가한 후, 돼지 T 세포에서 점차 감소하고 240 시간까지 소 세포에서 거의 일정하게 유지되었다.
IL-1β, IL-12/23p40 및 IL-10 발현의 동역(kinetics)은 돼지 PBMCs, 림프구, 단핵구 및 T 세포에서 확인되었다. 전 염증성 사이토카인으로서의 IL-1β 및 IL-12/23p40의 발현은 높았고, 항 염증성 사이토카인 IL-10의 발현은 현저하게 낮았다(도 3).

소보다 면역 반응이 낮은 돼지에서 APC의 반응을 조사하기 위해 돼지 Mo-DCs 및 Mo-MΦs를 단핵구에서 분극화하고 O/TWN/97-R 항원에 의해 자극하여 항원-매개 사이토카인이 직접 분비되는지 여부를 확인 Mo-DCs (도 4a의 a-e, 표 2) 및/또는 Mo-MΦs (도 4a의 f 및 도 4b의 g-j, 표 3). IL-1β, IL-6, IL-12/23p40 (48 시간) 및 TNFα (24 시간) 발현은 피크 수준에 도달한 후 감소하였다. O/TWN/97-R 항원은 이들 전 염증성 사이토카인의 발현을 현저하게 높은 수준으로 유도하였지만, IL-10 (항 염증성 사이토카인)은 Mo-DCs 및 Mo-MΦs에서 낮은 수준으로 발현되었다. 특히, LPS 및 IFNγ- 자극 M1 MΦs는 IL-4- 자극 M2 MΦs보다 더 현저하게 반응하였다.

4. FMDV 항원은 포식 작용에 의해 돼지 DCs 및 MΦs로 세포내 도입(endocytosis)되어 세포 면역을 개시한다.돼지 Mo-DCs 및 Mo-MΦs로의 O/TWN/97-R 항원의 내포작용(endocytosis)에 의한 선천적 면역 반응을 개시하고 증폭시키는 경로를 확인하기 위해, 세포를 phagocytosis 억제제인 사이토칼라신 D (CytD)로 처리하기 전과 후에 항원을 처리하여 공동 배양하였고, 세포 배양액에서 사이토카인의 발현을 관찰하였다(도 5a 및 도 5b). Mo-DCs 및 Mo-MΦs에서, 사이토카인 발현은 CytD 처리 전에 항원의 공동배양 후 24 시간 및 48 시간에 상승하였으나, CytD 처리 후 항원이 공동 배양될 때 유의하게 억제되었다. Mo-MΦs에서의 IL-10 발현은 CytD 처리 후 약간 억제되었지만, 전처리 수준과 크게 다르지 않았다.
5. 돼지에 비정상적으로 과발현된 선천 면역 반응 및 FMD 백신 예방 접종에 의한 T 세포 소진 경로(exhaustion pathway)의 유도
항원 매개 돼지 APC 자극 및 돼지 DCs 및 MΦs에 대한 식세포작용에 의한 항원의 내포작용에도 불구하고 소와 비교하여 돼지에서 면역 반응이 더 낮은 원인을 밝히기 위해, 본 발명자들은 FMD 항체 음성 동물로부터 나이브 소 및 돼지 PBMCs를 분리하여 RNA-Seq를 수행하였다.
이로부터 소와 돼지의 면역 반응을 비교하고, 생체 내에서 FMD 백신 접종에 의해 유도된 소와 돼지의 면역 반응의 근본적인 차이를 확인하였다(도 6a 내지 6c). 자연 상태에서, 소의 선천적 면역 반응은 잘 제어되고 유지되는 반면, 돼지에서는 비정상적으로 과잉 활성화되었다(도 6a의 b, c). 결과적으로, FMD 백신 접종은 가축에서 정상적인 면역 반응을 유도한 반면, T 세포 고갈 경로 (TBX21, NEAT1, NEAT3, NEAT5, EOMES, PRDM1, BCL6 및 PDCD1)에 관여하는 유전자의 발현은 돼지에서 유의하게 증가하였다 (도 6b). 특히, TLR/CDS 및 CLR 신호 전달 경로에 관여하는 유전자의 발현 분석은 소에서 IL23A 및 IL23R 발현이 FMD 백신 접종에 의해 효과적으로 유도되었음을 입증하였다. IL23A은 돼지에서 과발현 패턴을 나타내었지만 IL23R 발현은 관찰되지 않았다(도 6c).
<실험예 1> FMD 백신 보조제로 비-전형적인 T 세포 작용제는 마우스의 초기, 중기 및 장기 면역을 유도한다
APC를 자극하지 않고 T 세포를 직접 활성화시킴으로써 강력한 세포 면역 반응을 유도하기 위해, 신규한 FMD 백신 아쥬반트로서의 γδ T 세포, iNKT 세포, MAIT 세포를 포함하는 비전형적인 T 세포 작용제 및 전형적인 T 세포 작용제의 이용 가능성을 대상(목적) 동물인 돼지 실험 전에 마우스에서 평가하였다. 또한 오일 에멀젼이 없는 백신에서도 초기, 중기 및 장기 면역 반응을 효율적으로 유도하여 FMDV 감염시 숙주 보호 효과가 있는지 여부를 평가하였다 (도 7a).
대조군과 비교하여, SP-O ELISA에 의한 항체 역가는 γδ T 세포 작용제 (이소펜틸 피로포스페이트 트리리튬 염, IPP (I)), (E)-1-하이드록시-2-메틸-2-부테닐 4-피로포스페이트 리튬 염, HMP (H)) 및 iNKT 세포 작용제 (α-갈락토실세라마이드, α-Gal (G))의 투여 후 백신 접종 후 7 일 (dpv)에서 유의하게 더 높았다. 역가는 또한 14 dpv에서 MAIT 세포 작용제 (6-포르밀프린, 6-Formylpterin (F)) 투여군에서 증가되었다. 전형적인 T 세포 작용제 (RORγT (R))와 관련하여, 항체 역가는 28 dpv에서 비 전형적인 T 세포 작용제의 것과 유사하였다. 모든 실험군에서 항체 역가는 168 dpv까지 대조군의 것보다 유의하게 높았다 (도 7b).
바이러스 중화 (VN) 역가는 SP-O ELISA에 의한 항체 역가와 유사한 경향을 나타내었다. γδ T 세포 작용제 (I)-, γδ T 세포 작용제 (H)- 및 iNKT 세포 작용제 (G)-투여군은 7 dpv에서 중화항체가가 대략 100배 증가하였고, 14 dpv에서 MAIT 세포 작용제 (F)-투여군에서 높은 수준의 VN 역가가 또한 나타났다. 28 dpv에서, VN 역가는 γδ T 세포 작용제 (I)-및 iNKT 세포 작용제 (G)-투여군에서 가장 높았다. VN 역가는 부스팅 후 56 dpv에서 최대치를 나타내었고, 모든 비-전형적인 T 세포 작용제-투여군은 168 dpv에서 대조군보다 유의하게 더 높은 중화 항체 역가를 유지 하였다 (도 7b). 84 및 168 dpv에서 FMDV (100 LD50의 O/VET/2013) 챌린지 테스트는 모든 보조제 처리 그룹에서 100% 생존율 (도 7c)을 보여주었고, 체중 변화는 거의 없었다 (도 7d).
따라서 비-전형적인 T 세포 작용제를 함유하는 FMD 백신은 마우스에서 초기, 중기 및 장기 면역의 유도에 강력한 영향을 미치는 것으로 확인되었다.
<실험예 2> 비-전형적인 T 세포 작용제는 돼지 PBMCs의 강력한 세포증식을 이끌어낸다
인큐베이션 후 12 시간 및 36 시간에 FMD 항체-음성 생성 동물로부터 분리된 돼지 나이브 PBMCs에서 비-전형적인 T 세포 작용제-매개 세포 증식이 관찰되었다(도 8a). 실제 시험 백신과 유사한 조건을 제공하기 위해, FMD 혈청형 O(O/TWN/97-R) 항원을 비-전형적인 T 세포 작용제와 공동 투여하였다. 세포 증식은 다음의 순으로 높게 나타났다: 항원 + γδ T 세포 작용제 (H)> 항원 + iNKT 세포 작용제 (G)> 항원 + MAIT 세포 작용제 (F)> 항원 + T 세포 작용제 (R)> 항원 + γδ T 세포 작용제 (I)> 항원 단독(도 8b).
<실험예 3> 비-전형적인 T 세포 작용제는 돼지에서 비정상적인 선천적 면역 반응을 개선하고 APC 자극없이 강력한 면역 반응을 유도하는 T 세포를 직접적으로 활성화한다.
돼지에서 강력한 세포성 면역 반응을 유도함으로써 소에 비해 낮은 면역원성을 극복하고 돼지에서 제기되고 있는 다양한 문제에 대한 해결책을 제공하기 위해, PRRs ligand로 DCs 및 MΦs와 같은 APC를 자극함으로써 T 세포를 간접적으로 활성화시켜 면역반응을 유도하는 시스템과 비-전형적인 T 세포 작용제 및 T 세포 작용제를 통해 T 세포를 직접적으로 자극하여 면역반응을 유도하는 시스템을 비교하였다. 나이브 PBMCs를 FMD 항체-음성(seronegative) 돼지로부터 분리하고, 항원 + PRR 리간드 (레시퀴모드 (R848, TLR-7/8 효능제) 및 트레할로스-6, 6-디베헤네이트 (TDB, Mincle 작용제); TDB 및 비스-(3'-5')-시클릭 이량체 구아노신 모노 포스페이트 (c-di-GMP, STING 작용제) 또는 항원 + 비-전형적인 T 세포 작용제 (γδ T 세포 작용제, I; γδ T 세포 작용제, H; iNKT 세포 작용제, G; MAIT 세포 작용제, F 또는 항원 + T 세포 작용제, R, 또는 항원 단독 처리하여, 12시간 후 PBMCs를 수집하여, total RNA를 TRIzol Reagent (Invitrogen) 및 RNeasy Mini Kit (QIAGEN)를 이용하여 제조사의 권고 방법에 따라 추출, RNA-Seq를 수행하여 면역증강제-매개 세포성 면역 반응 및 관련 유전자 발현 프로파일의 유도를 확인하였다 (도 9a 내지 9e).
TLR/CDS 신호 전달 관련 유전자 발현 프로파일은 PRR 리간드가 TLR-7/8, cGAS 및 RUNX3 발현을 유도하였고, 비-전형적인 종래의 T 세포 작용제는 TBK1, RUNX1, IL23A 및 IL23R 발현에 유의하게 영향을 미쳤음을 나타내었다(도 9a의 a). CLR 신호 전달 경로는 PRR 리간드 처리와 비교하여 종래의 T 세포 작용제로 처리함으로써 현저하게 유도되었다. IL23A 및 IL23R은 특히 비 전형적인 T 세포 작용제 중 iNKT 세포 작용제 (G)-및 전형적인 T 세포 작용제 (R)-처리된 그룹에서 높게 발현되었다(도 9a의 b). T 세포 고갈 경로 관련 유전자 발현은 PRR 리간드 및 비-전형적인 T 세포 작용제 처리에 의해 개선되었다 (도 9a의 c). Th1, Th2, Th9, Th17, Th22, Tfh, pTreg 및 tTreg 세포에서의 유전자 발현은 PRR 리간드-처리된 그룹과 비교하여 비-전형적인 T 세포 작용제 및 통상적인 T 세포-처리된 그룹에서 유의하게 증가되었다. 또한 IL23A 및 IL23R 발현이 증강되었고, LTA, STAT4, CCL17, CCL22, IL10, RORA, CCL20, IL17A, IL17F, IL1α 및 IL1β 발현도 T 세포 작용제 처리에 의해 증가되었다(도 9a의 d, 도 9b의 e-h, 도 9c의 i-k). 특히, M1, M2a, M2b, M2c, M2d 및 DCs에서의 유전자 발현은 APC- 자극 PRR 리간드 처리와 비교하여 비전형적인 T 세포 작용제 및 전형적인 T 세포 작용제 처리에 의해 현저하게 향상되었다. CD80, CD86, CCL1, CCL2, CCL3, CCL17, CCL22, IL1β, IL23A, TNFα, IL1R2, TGM2, CXCL10, CXCL16 및 CD14의 발현이 현저히 증가하였다 (도 9c의 l 및 도9d의 m,n, 도 9e의 o-q). NK 세포에서, 비전형적인 T 세포 작용제 및 T 세포 작용제 처리에 의해 증가된 ITGB2 및 IFNAR2 유전자 발현이 관찰되었다 (도 9e의 r).

Claims (7)
- γδ T 세포 작용제, iNKT 세포 작용제, MAIT 세포 작용제, T 세포 작용제로 이루어진 군에서 선택되는 어느 하나 이상을 유효성분으로 포함하는 면역증강제 조성물.
- 제1항에 있어서, 상기 유효성분은 면역증강제 조성물의 0.01 내지 1 중량% 포함되는 것을 특징으로 하는 면역증강제 조성물.
- 제1항에 있어서, 상기 유효성분 외에 첨가제, 부형제, 담체 등이 추가적으로 더 포함되는 것을 특징으로 하는 면역증강제 조성물.
- 제1항에 있어서, 상기 면역증강제 조성물은 오일 제형 또는 비-오일 제형인 것을 특징으로 하는 면역증강제 조성물.
- 제1항 내지 제4항 중 어느 한 항의 면역증강제 조성물을 포함하는 백신 조성물.
- 제5항에 있어서, 상기 면역증강제 조성물은 백신 조성물의 30 내지 70 중량%로 포함되는 것을 특징으로 하는 백신 조성물.
