Abstract
본 발명은 피부 상재균으로 오일을 발효하여 수득한 산물을 포함하는 발효나노유화제 조성물의 제조방법에 따르면 합성 원료의 함량을 최소화함으로써 피부에 친화적인 조성물을 제공할 수 있다. 또한, nm 크기의 나노에멀젼을 형성함으로써 유화력을 향상시킬 수 있고, 조성물의 안정성을 향상시켜 다양한 제형으로 응용할 수 있으며, 피부 자극 최소화할 수 있으므로, 피부 저자극의 기능을 필요로 하는 다양한 제형의 제품에 용이하게 적용할 수 있다.
Description
본 발명은 피부 상재균으로 오일을 발효하여 수득한 산물을 포함하는 발효나노유화제 조성물 및 그 제조방법에 관한 것이다.
본 출원은 2020.05.26자로 출원된 한국특허출원 제2020-0063024호에 기초한 우선권의 이익을 주장하며, 해당 특허 출원의 문헌에 개시된 모든 내용은 본 명세서의 일부로서 포함된다.
피부는 표피, 진피, 피하지방층으로 이루어져 있다. 이중 표피는 가장 바깥층에 존재하고, 피부의 보호막 역할을 하며, 피부의 면역 기능을 담당하고 있다. 표피층은 주로 각질형성세포로 이루어져 있으며, 각질세포 사이는 지질성분으로 이루어져 있고, 가장 바깥층은 단백질로 구성되어 있다.
사람의 몸은 다양한 미생물들의 서식처가 될 수 있고, 미생물은 숙주와 공생관계를 이루며 숙주에 영향을 미친다. 이중 피부에 존재하는 피부상재균은 피부의 표면에 생식하는 미생물로써, 표피의 가장 외곽층과 모낭의 윗부분(상위층)에서 주로 발견된다. 피부상재균으로는 약 1000여종의 미생물이 존재한다. 피부에 존재하는 미생물 중 호기성 미생물은 표피층의 지질을 이용할 수 있는 지방분해효소를 분비할 수 있다.
하지만, 피부 상재균 및 이를 이용한 피부 조성물에 대한 연구는 미비한 실정이다.
따라서, 지방분해효소를 분비하는 피부 상재균을 활용하여 유화력이 향상된 발효나노유화제 조성물 및 그 제조방법을 제공하고자 한다.
본 발명의 목적은 피부 상재균을 이용한 오일의 발효 산물로부터 발효나노유화제 조성물 및 그 제조방법을 제공하는 것이다.
상기 과제를 해결하기 위하여 본 발명은,
피부상재균으로 기탁번호 KCCM 11843P(한국미생물보존센터(국외), 2016.06.08)의 미생물을 종균배양하여 종균배양물을 제조하는 단계;
상기 종균배양물을 전배양하여 전배양물을 제조하는 단계; 및
상기 전배양물을 발효하여 발효물을 제조하는 단계를 포함하는, 발효나노유화제 조성물의 제조방법을 제공한다.
일구현예에 따르면, 상기 종균배양물을 제조하는 단계에서 사용하는 배지는 R2A(BD Difco, USA) 배지를 포함할 수 있다.
또한, 상기 전배양물을 제조하는 단계에서 사용하는 배지는 이스트 추출물, 글리세롤, 카제인 펩톤, 식물성오일, K2HPO4 KH2PO4, MgSO4 및 MgCl2를 포함할 수 있다.
또한, 상기 발효물을 제조하는 단계에서 사용하는 배지는 이스트 추출물, 글리세롤, 카제인 펩톤, 식물성오일, K2HPO4, KH2PO4, MgSO4 및 MgCl2를 포함할 수 있다.
구체적인 구현예에 따르면, 상기 종균배양물을 제조하는 단계에서 사용하는 배지는 카제인 가수분해물 0.1 내지 5 g/L, 이스트 추출물 0.1 내지 10 g/L, 글루코스 0.1 내지 10 g/L, 가용성 전분 0.1 내지 5 g/L, K2HPO4 0.1 내지 5 g/L, 피루브산나트륨 0.05 내지 5 g/L, 카제인 펩톤 0.1 내지 5 g/L, MgCl2 0.01 내지 1 g/L를 포함할 수 있다.
또한, 상기 전배양물을 제조하는 단계는 이스트 추출물 1 내지 100 g/L, 글리세롤 0.1 내지 10 g/L, 카제인 펩톤 1 내지 100 g/L, 식물성오일 10 내지 500 g/L, K2HPO4 1 내지 10 g/L, KH2PO4 1 내지 10 g/L, MgSO4 1 내지 10 g/L 및 MgCl2 0.01 내지 3 g/L를 포함하는 배지에 상기 종균배양물 0.1 내지 50g/L을 첨가하는 단계를 포함할 수 있다.
또한, 상기 발효물을 제조하는 단계는 이스트 추출물 1 내지 100 g/L, 글리세롤 0.1 내지 10 g/L, 카제인 펩톤 1 내지 100 g/L, 식물성오일 10 내지 500 g/L, K2HPO4 1 내지 10 g/L, KH2PO4 1 내지 10 g/L, MgSO4 1 내지 10 g/L 및 MgCl2 0.01 내지 3 g/L를 포함하는 배지에 상기 전배양물 1 내지 300g/L를 첨가하는 단계를 포함할 수 있다.
일구현예에 따르면, 상기 종균배양물을 제조하는 단계는 15 내지 35℃의 호기 조건에서 배양하는 단계를 포함하고, 600nm 파장에서 분광광도계의 흡광도가 0.5인 시점에 배양 종료할 수 있다.
일구현예에 따르면, 상기 전배양물을 제조하는 단계는 15 내지 35℃의 호기 조건에서 배양하는 단계를 포함하고, 600nm 파장에서 분광광도계의 흡광도가 1인 시점에 배양 종료할 수 있다.
일구현예에 따르면, 상기 발효물을 제조하는 단계는 15 내지 35℃에서 일 1 내지 10일 동안 배양하는 단계를 포함할 수 있다.
본 발명의 다른 구현예에 따르면, 상기한 바와 같은 방법으로 제조된 발효나노유화제 조성물을 제공한다.
일구현예에 따르면, 상기 발효나노유화제의 평균입경은 100 내지 200nm일 수 있다.
본 발명의 또 다른 구현예에 따르면, 상기한 바와 같은 발효나노유화제 조성물을 포함하는 화장료 조성물을 제공한다.
기타 본 발명에 따른 구현예들의 구체적인 사항은 이하의 상세한 설명에 포함되어 있다.
본 발명에 따른 피부 상재균을 이용한 발효나노유화제 조성물은 합성 원료의 함량을 최소화함으로써 피부에 친화적인 조성물을 제공할 수 있다. 또한, nm 크기의 나노에멀젼을 형성함으로써 유화력을 향상시킬 수 있고, 조성물의 안정성을 향상시켜 다양한 제형으로 응용할 수 있으며, 피부 자극을 최소화할 수 있으므로, 피부 저자극의 기능을 가지는 다양한 제형에 용이하게 적용할 수 있다.

본 발명은 다양한 변환을 가할 수 있고 여러 가지 실시예를 가질 수 있는 바, 특정 실시예를 예시하고 상세하게 설명하고자 한다. 그러나, 이는 본 발명을 특정한 실시 형태에 대하여 한정하려는 것이 아니며, 본 발명의 사상 및 기술 범위에 포함되는 모든 변환, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다. 본 발명을 설명함에 있어서 관련된 공지 기술에 대한 구체적인 설명이 본 발명의 요지를 흐릴 수 있다고 판단되는 경우 그 상세한 설명을 생략한다.
이하, 본 발명에 따른 발효나노유화제 조성물 및 그 제조방법에 대하여 상세히 설명한다.
본 발명은, 피부에 서식하고 있는 다양한 미생물들 중, 피부에 유익한 영향을 주면서 공생하고 있는 피부상재균을 이용하여 유화력을 향상시킨 나노유화제 조성물을 제공한다.
구체적으로 본 발명은,
피부상재균으로 기탁번호 KCCM 11843P(한국미생물보존센터(국외), 2016.06.08.)의 미생물을 종균배양하여 종균배양물을 제조하는 단계;
상기 종균배양물을 전배양하여 전배양물을 제조하는 단계; 및
상기 전배양물을 발효하여 발효물을 제조하는 단계를 포함하는, 발효나노유화제 조성물의 제조방법을 제공한다.
본 발명에 사용되는 피부상재균은 기탁번호 KCCM 11843P(한국미생물보존센터(국외), 2016.06.08.)의 미생물로, 스포리치아과(Sporichthyaceae)에 속하는 에피더미디박테리움 케라티니(Epidermidibacterium keratini sp.)에 포함된다.
일구현예에 따르면, 상기 종균배양물을 제조하는 단계에서 사용하는 배지는 R2A(BD Difco, USA) 배지를 사용할 수 있다.
또한, 상기 전배양물을 제조하는 단계에서 사용하는 배지는 이스트 추출물, 글리세롤, 카제인 펩톤, 식물성오일, K2HPO4, KH2PO4, MgSO4 및 MgCl2를 포함할 수 있다.
또한, 상기 발효물을 제조하는 단계에서 사용하는 배지는 이스트 추출물, 글리세롤, 카제인 펩톤, 식물성오일, K2HPO4 KH2PO4, MgSO4 및 MgCl2를 포함할 수 있다.
구체적인 구현예에 따르면, 상기 종균배양물을 제조하는 단계에서 사용하는 배지는 카제인 가수분해물(casein acid hydrolysate) 0.1 내지 5 g/L, 예를 들면 0.1 내지 1 g/L, 이스트 추출물 0.1 내지 10 g/L, 예를 들면 0.1 내지 5, 예를 들면 0.1 내지 1 g/L, 글루코스 0.1 내지 10 g/L, 예를 들면 0.1 내지 1 g/L, 가용성 전분 0.1 내지 5 g/L, 예를 들면 0.1 내지 1 g/L, K2HPO4 0.1 내지 5 g/L, 예를 들면 0.1 내지 1 g/L, 피루브산나트륨 0.05 내지 5 g/L, 예를 들면 0.1 내지 1 g/L, 카제인 펩톤 0.1 내지 5 g/L, 예를 들면 0.1 내지 1 g/L, MgCl2 0.01 내지 1 g/L, 예를 들면 0.01 내지 0.1 g/L를 포함할 수 있다.
또한, 상기 전배양물을 제조하는 단계는 이스트 추출물 1 내지 100 g/L, 예를 들면 1 내지 50 g/L, 글리세롤 0.1 내지 10 g/L, 예를 들면 0.5 내지 5 g/L, 카제인 펩톤 1 내지 100g/L, 예를 들면 1 내지 50g/L, 예를 들면 1 내지 30 g/L, 예를 들면 5 내지 20 g/L, 식물성오일 10 내지 500 g/L, 예를 들면 10 내지 300 g/L, 예를 들면 10 내지 200 g/L, K2HPO4 1 내지 10 g/L, 예를 들면 3 내지 8 g/L, 예를 들면 5 내지 8 g/L, KH2PO4 1 내지 10 g/L, 예를 들면 1 내지 5 g/L, MgSO4 1 내지 10 g/L, 예를 들면 1 내지 5 g/L, 예를 들면 1 내지 3 g/L 및 MgCl2 0.01 내지 3 g/L, 예를 들면 0.1 내지 1 g/L를 포함하는 배지에 상기 종균배양물 0.1 내지 50g/L, 예를 들면 1 내지 30 g/L, 예를 들면 5 내지 20 g/L을 첨가하는 단계를 포함할 수 있다.
또한, 상기 발효물을 제조하는 단계는 이스트 추출물 1 내지 100 g/L, 예를 들면 1 내지 50 g/L, 글리세롤 0.1 내지 10 g/L, 예를 들면 0.5 내지 5 g/L, 카제인 펩톤 1 내지 100g/L, 예를 들면 1 내지 50g/L, 1 내지 30 g/L, 예를 들면 5 내지 20 g/L, 식물성오일 10 내지 500 g/L, 예를 들면 10 내지 300 g/L, 예를 들면 10 내지 200 g/L, K2HPO4 1 내지 10 g/L, 예를 들면 3 내지 8 g/L, 예를 들면 5 내지 8 g/L, KH2PO4 1 내지 10 g/L, 예를 들면 1 내지 5 g/L, MgSO4 1 내지 10 g/L, 예를 들면 1 내지 5 g/L, 예를 들면 1 내지 3 g/L 및 MgCl2 0.01 내지 3 g/L, 예를 들면 0.1 내지 1 g/L를 포함하는 배지에 상기 전배양물 1 내지 300 g/L, 예를 들면 50 내지 200 g/L를 첨가하는 단계를 포함할 수 있다.
일구현예에 따르면, 상기 식물성오일은 해바라기씨, 포도씨, 카놀라, 쌀눈, 올리브, 대두, 아르간, 현미, 들깨, 참깨, 아몬드, 땅콩, 옥수수, 홍삼, 아보카도, 마카다미아, 코코넛, 로즈힙, 비타민나무씨, 시어나무열매, 기름야자, 베르가모트열매, 동백씨, 홍화씨, 살구씨, 양귀비씨, 달맞이꽃씨, 피마자씨, 녹차씨, 메도우폼씨, 아마씨 및 햄프씨 등의 식용 가능 또는 인체 친화적인 오일 중 하나 이상을 포함할 수 있으나, 상기한 종류에 특별히 한정되지는 않는다.
일구현예에 따르면, 상기 종균배양물을 제조하는 단계는 15 내지 35℃, 예를 들면 20 내지 30℃의 호기 조건에서 배양하는 단계를 포함하고, 종균배양은 600nm 파장에서 분광광도계의 흡광도가 0.5인 시점에 종료할 수 있다.
일구현예에 따르면, 상기 전배양물을 제조하는 단계는 15 내지 35℃, 예를 들면 20 내지 30℃의 호기 조건에서 배양하는 단계를 포함하고, 전배양은 600nm 파장에서 분광광도계의 흡광도가 1인 시점에 배양 종료할 수 있다.
일구현예에 따르면, 상기 발효물을 제조하는 단계는 15 내지 35℃, 예를 들면 20 내지 30℃에서 24시간 내지 240시간, 예를 들면 72시간 내지 240시간, 예를 들면 120시간 내지 240시간 동안 배양하는 단계를 포함할 수 있다.
상기 "발효물"은 균주를 배양하여 수득된 배양액 자체, 그의 농축물, 또는 동결건조물일 수 있다. 상기 발효물은 발효나노유화제를 포함한다.
일구현예에 따르면, 본 발명에 따른 발효나노유화제의 평균입경은 100 내지 200nm, 예를 들면 150 내지 200nm, 예를 들면 160 내지 190nm일 수 있다.
본 발명의 또 다른 구현예에 따르면, 상기한 바와 같은 발효나노유화제 조성물을 포함하는 화장료 조성물을 제공한다.
화장료 조성물은 안정화제, 용해화제, 비타민, 안료, 향료, 보조제, 담체 등 화장료 조성물에 통상적으로 이용하는 성분들을 포함할 수 있다.
일구현예에 따르면, 본 발명의 화장료 조성물은 당업계에서 통상적으로 제조되며, 피부학적으로 적용이 가능한 어떠한 제형으로도 제조될 수 있다.
피부학적으로 적용이 가능하다는 것은 적용 대상에게 비교적 비독성이고 무해한 유효작용을 할 수 있는 조성물로서, 피부에 적용할 수 있는 외용제를 포함할 수 있으며, 상기 조성물로부터 기인하는 부작용이 유효 성분의 효능을 저하시키기 않고, 적용 대상에 심각한 자극을 유발하지 않으면서 유효성분들의 활성 및 물성을 손상시키지 않는다는 것을 의미할 수 있다. 본 발명의 피부학적으로 적용이 가능한 화장료 조성물로는 예를 들어, 용액, 현탁액, 유액, 유탁액, 페이스트, 겔, 팩, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 스프레이 및 모발 화장료 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다.
구체적으로, 상기 화장료 조성물은 스킨로션, 스킨소프너, 스킨토너, 아스트린젠트, 로션, 겔, 밀크로션, 모이스쳐 로션, 영양로션, 맛사지크림, 영양크림, 모이스처크림, 핸드크림, 파운데이션, 에센스, 엠플, 영양에센스, 팩, 비누, 헤어샴푸, 풋샴푸, 클렌징폼, 클렌징로션, 클렌징크림, 바디로션 및 바디클린저의 제형 등으로 제조될 수 있다.
이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
실시예: 발효나노유화제 제조
종균배양
피부상재균으로 기탁번호 KCCM 11843P(한국미생물보존센터(국외), 2016.06.08)의 미생물을 종균배양하였다. R2A(BD Difco, USA)배지에 접종하여 25℃의 호기적인 조건에서 정치배양을 진행하여 종균배양물을 제조하였다. 구체적으로, 배지 조성은 카제인 가수분해물(Casein acid hydrolysate) 0.5 g/L, 이스트 추출물(yeast extract) 0.5 g/L, 글루코스(glucose) 0.5 g/L, 가용성 전분(soluble starch) 0.5 g/L, K2HPO4 0.3 g/L, 피루브산나트륨(sodium pyruvate) 0.3 g/L, 카제인펩톤(casein peptone) 0.5 g/L 및 MgCl2 0.05 g/L이다.
종균배양은 분광광도계로 600nm 파장에서 0.5 흡광도를 보이는 시점에서 종료하였다.
전배양
표 1 및 2와 같은 조성의 배지에 종균배양물 10 g/L을 투입하고, 25℃ 호기적인 조건으로 배양을 진행하여 전배양물을 제조하였다. 전배양은 분광광도계로 600nm 파장에서 흡광도 1.0인 시점에서 종료하였다.
발효
표 1 및 2와 같은 조성의 배지의 온도가 25℃일 때, 전배양물 100 g/L를 투입하고, 180시간 동안 발효를 진행하여 발효물(배양액)을 제조하였다. 실시예 19의 배지에 따른 배지를 사용하여 발효가 완료된 상태를 도 1에 나타내었으며, 발효 전과 비교하였다.

상기의 발효물을 고속연속원심분리하여 미생물 균체를 제거하였으며, 균체를 제거한 발효물을 오일상과 수상으로 분리하였다. 이 수상을 0.45 um, 0.2 um 폴리에테르설폰(Polyethersulfone, PES) 멤브레인 필터에 순차적으로 제균여과하여 발효나노유화제를 수득하였다.

실험예 1: 조성물에 따른 최적 발효 조건 평가
선행실험으로 KCCM 11843P의 균주를 배양할 때, 식물성 오일을 첨가한 결과, 오일을 첨가하지 않은 군보다 첨가한 군의 배양액 혼탁도가 더 높게 나타났다. 단, 배양액의 혼탁도는 균을 제거한 수층을 대상으로 측정하였다. 따라서, 해당 선행실험을 통하여 KCCM 11843P의 균주가 식물성 오일을 기질로 대사하며, 또한 유화 성분을 생성할 것이라고 판단하였다. 이를 토대로 배지의 탄소원, 질소원의 함량을 조절함으로써 유화 성분을 최대로 생성할 수 있는 최적발효조건을 찾고자 하였다.
실시예 1 내지 29에 대하여, 배양액의 혼탁도와 미생물 균체량을 측정함으로써 최적발효조건을 평가하였다. 종균배양은 상기한 방법으로 실시하였으며, 각 실시예에 나타낸 조성물로 전배양을 실시하였고, 동일한 조성물로 25℃에서 200rpm으로 140시간 발효 후 평가하였다.
배양액의 혼탁도 측정법은 다음과 같다. 배양액을 3,088 g, 15분 원심분리하여 미생물 균체를 제거하였으며, 균체를 제거한 배양액을 오일층과 수층으로 분리하였다. 이 수층을 0.45 um, 0.2 um 폴리에테르설폰(polyethersulfone, PES) 멤브레인 필터에 순차적으로 제균 여과하였다. 상기 방법으로 균체를 제거한 배양액에 0.2 um 이하의 사이즈를 가진 나노에멀젼이 분산되어 있는 것이 관찰되었다. 유화 성분의 함량이 높을수록 형성된 나노에멀젼의 밀도가 높고, 이것을 혼탁도로 비교할 수 있을 것으로 판단하였다. 그래서 발효 후 균체를 제거한 수층을 정제수로 10배 희석 후 0.2 um 멤브레인 필터로 여과하여 600nm 파장에서 흡광도를 측정하였으며, 결과를 표 3에 나타내었다. 측정기기로는 분광광도계(BIO Teck, Korea)를 이용하였다.
이와 더불어, 배양액의 균체량을 측정하였다. 각 실시예 별로 배양액을 취하여 13,000g, 30분 동안 원심분리한 후, 상등액은 버리고 미생물 균체만을 수득하였다. 이를 0.85% NaCl에 1회 세척한 후, 배양액과 동일한 부피의 0.85% NaCl에 현탁하였으며, 이를 정제수로 10배 희석하여 600nm 파장에서 흡광도를 측정하였으며, 결과는 표 3에 나타내었다. 측정기기는 분광광도계(BIO Teck, Korea)를 이용하였다.

실험 결과, 실시예 19의 균체량은 각 실시예의 평균보다 낮은 수준으로 측정된 반면, 균체를 제거한 배양액의 혼탁도가 가장 높게 측정되었다. 이는 배양 배지에 첨가된 오일이 미생물에 의해 유화된 정도가 최적인 것으로 판단할 수 있으며, 가장 많은 유화성분이 포함된 것이다. 따라서, 실시예 19를 최적 발효 조건으로 선정하였다.
실험예 2: 피부상재균주 별 유화 성분의 생성 평가
균의 종류에 따른 유화 성분의 생성 정도를 비교 평가하기 위하여, 대표적인 피부상재균인 Acinetobacter calcoaceticus, Staphylococcus epidermidis, Staphylococcus aureus, Cutibacterium acnes, 그리고 KCCM 11843P와 근연종인 Sporichthya spp.를 배양하여 KCCM 11843P의 균주와 비교하였다. 평가는 실험예 1과 동일한 방법으로 수행되었으며, 실시예 19와 동일한 조성물에 발효를 진행하였다. 그 결과는 표 4에서 나타내었다.

실험 결과, KCCM 11843P균주가 가장 많은 유화 성분을 생성시키는 것을 확인할 수 있었다.
또한, 배양액에서 비교예 2 및 비교예 4가 가장 역한 냄새가 나므로, 배양액 자체로 화장료 조성물에 적용하기 어려운 상태로 판단되었다.
반면, 실시예 19의 배양액은 무취에 가까워 화장료 조성물로 사용하기 용이한 것으로 판단하였다.
실험예 3: 실시예 19의 최적 발효시간 확인
실험예 1을 통하여 실시예 19가 최적의 전배양 배지 및 발효 배지 조건임을 확인하였으며, 배양 기간 동안 pH, 균체량, 배양액의 탁도를 측정함으로써, 미생물의 2차 대사산물이 생성하는 시점 및 발효 종료시점을 판단하기 위한 실험을 진행하였다.
pH 변화는 미생물이 발효하는 과정에서 대사산물의 생성 및 정도를 확인할 수 있다. 구체적으로, 시간별로 샘플링한 배양액을 3,088 g, 5분 원심분리한 후, 균체와 오일층을 제외한 수층을 취하여 pH meter(Mettler toledo, FP20)로 측정하였다.
시간별로 평가한 결과, pH는 총 2단계를 거쳐 초기 pH6.6에서 pH8.3까지 상승하는 경향을 보였으며, 이후 유지되는 경향을 나타내었다. 이를 도 3에 나타내었다.
미생물의 2차 대사산물 생성에 따른 pH 변화가 상승하는 경향이 있으며, 미생물 생장기 중에 대수기를 지나 정체기에서 대사산물이 생성됨을 알 수 있었다. 도 3에 따르면, 발효 80시간 전후로 pH가 다시 상승하기 시작하면서 혼탁도 또한 급격히 증가한다. 이 시기는 미생물 생장기 중 대수기 후반에서 정체기에 해당하므로 미생물의 2차 대사가 발생함을 알 수 있다.
따라서, 미생물의 2차 대사산물에 의해 배양액의 탁도가 증가하였으며, 이것이 유화제 성분의 일종임을 예상할 수 있다.
미생물의 2차 대사가 발생한 이후에 균체를 제거한 배양액의 혼탁도가 더이상 증가하지 않는 시점을 유화 성분이 최대로 생성된 시점으로 생각하였으며, 이를 발효 종료시점으로 판단하였다. 도 3에 따르면, 발효 180시간 이후 균체를 제거한 배양액의 혼탁도가 증가하지 않고 유지되는 경향을 보였다. 따라서, 180시간 전후를 발효 종료시점으로 판단하였다.
실험예 4: 입도 분석
입도측정은 실시예 19에 따른 발효나노유화제를 정제수로 10배 희석한 후, 샘플을 분석을 하였다. 측정기기는 입도분석기(HORIBA Scientific, Japan)를 이용하였다. 그 결과는 도 4에 나타내었으며, 평균입경은 약 181.7nm로 측정되었다.
실험예 5: 나노에멀젼 관찰
실시예 19에 따른 배양액을 분리 정제하여 얻은 발효나노유화제 내에 형성된 나노에멀젼을 투과전자현미경(JEM-F200, JEOL)으로 관찰하였다.
시료를 carbon으로 코팅된 Cu grid 위에 한 방울 떨어뜨려 상온에서 6시간 동안 건조하여 전처리하고 현미경으로 관찰하였다.
현미경 사진은 도 5에 나타내었다. 둥근 형태의 나노에멀젼(마이셀, micelle)이 형성된 것을 확인할 수 있다.
실험예 6: TLC 분석
배양 조건에 따른 유화 성분의 생성 여부 및 양적 비교를 위해 실시예 19에 대하여 TLC(Thin Layer Chromatography)로 분석하였다.
수득한 발효액에 1N HCl을 첨가하여 pH를 2 내지 3으로 조정하였다. 클로로포름(chloroform)과 메탄올(methanol)을 2:1(v/v) 비율로 혼합한 유기용제를 상기의 발효액과 2:1(v/v) 비율로 투입하여 25℃에서 2시간 교반한 후, 오버나잇(overnight) 정치함으로써, 유화성분을 하층액(클로로포름)으로 전이시켜 정제하였다. 수득한 클로로포름은 감압농축하여 휘발시키고, 남은 유화성분을 클로로포름(chloroform)에 용해시킴으로써 수득하였다.
실리카겔 TLC플레이트 (TLC Silica gel 60, Merck; 5 cm x 6.5 cm)에 상기의 정제물(유화성분)을 점적하고 클로로폼(chloroform), 메탄올(methanol), 아세트산(acetic acid) (6.5:2:0.2 v/v)를 포함한 이동상으로 종료기점까지 전개하였다. 자연 건조된 플레이트에 에탄올과 혼합된 10% 황산을 분무하고 건조 후, 100℃에서 5분간 가열하여 스팟(spot)을 가시화한 결과, 극성지질 스팟을 관찰할 수 있었으며, 도 6에 나타내었다.
비교군으로는 쌀눈오일을 클로로포름에 20% 용해한 것을 사용하였다. 비교예 4의 정제물과 비교했을 때, 실시예 19의 정제물에서 더 다양하고 많은 양의 극성지질이 관찰되었다.
실험예 7: 발효기간별 유화층 형성 정도
실시예 19의 발효를 진행함에 따라 실제 유화층이 형성되는 시기를 확인하고자 하였다. 발효기간별로 배양액을 10ml 취하여 3,088g, 5분 원심분리 후 관찰하였다.
실험 결과, 발효 84시간 전후로 오일층과 수층 사이에 유화층이 형성되었음을 확인하였으며, 이를 도 7에 나타내었다.
도 3과 비교하였을 때, 2차대사가 발생하기 시작하는 시기와 매우 유사한 시기에 유화층이 형성된다. 이를 통해, 2차 대사산물로서 유화성분이 형성됨을 알 수 있다.
실험예 8: 피부자극 평가
본 발명에 따른 실시예 19를 분리 정제한 발효나노유화제에 대하여 피부 자극 여부를 평가하고자 피부 일차 자극평가를 실시하였다. 평가 기관은 (주)코어덤으로 의뢰하여 진행하였다. 피험자 32명을 선정하여 등 부위에 24시간 첩포한 후 첩포 제거 30분, 24시간, 48시간에 각각 일차 피부자극 유무를 피부과 전문의가 판정하였다. 피부 반응 판정은 국제접촉피부염학회(ICDRG)기준 및 미국화장품협회(PCPC)의 기준에 의거하였으며, 그 결과는 표 5에 나타내었다.
실시예 30: 에센스 제형 제조
하기 표의 조성에 따라 실시예 19를 분리 정제한 발효나노유화제를 포함하는 에센스 제형을 제조하였다. 구체적으로, 하기 표의 1번 원료에 2 내지 7번 원료를 순차적으로 투입하고, 교반하여 용해시켰다. 이후 1 내지 7번 원료 반응 용기에 8 내지 9번 원료를 순차적으로 투입하여 균일하게 용해 및 혼합하여 에센스 제형을 완성하였다.

실험예 9: 안정도 평가
보관 온도 조건에 따른 안정성을 평가하기 위하여, 실시예 30에 따른 에센스 및 실시예 31에 따른 스킨 제형 조성물의 조건 별 안정성을 확인하였다. 각각의 조성물을 일반조건은 상온, 25℃조건에서 3개월 동안, 가혹조건은 저온(-5℃), 고온(50℃), 순환(-5 내지 40℃/12 hr) 조건에서 1개월 동안 보관하여 pH 및 분리도 성상을 관찰하였다. 일반조건에서는 3개월 후 제형의 변화 유무를 관찰하였고, 가혹조건에서는 1개월 후 제형의 변화유무를 관찰하였다. 그 결과를 표 8 및 9에 나타내었다.

상기 결과로부터 실시예 30 및 31은 보관 조건에 따른 pH, 성상 분리도의 변화가 없음을 확인하였다. 이는 실시예 20의 발효나노유화제를 투입함으로써 합성 계면활성제 및 고압기기를 이용하지 않아도 조성물 상에서 에멀젼을 형성하여, 안정적인 제형으로 간편하게 화장품 제형을 제조할 수 있음을 보여주는 결과이다. 이를 통해, 합성 화학 화장품 성분을 최소화하여 피부에 더욱 안전한 제형을 제조할 수 있다.
상기한 바와 같이, 본 발명에서는 피부 상재균이 오일을 활용하여 발효나노유화제를 형성할 수 있음을 확인하였으며, 본 발명에 따른 발효나노유화제는 나노미터(nm)크기의 나노에멀젼을 형성함으로써 화학적 계면활성제를 사용하지 않아도 유화력을 향상시킬 수 있고, 조성물의 안정성을 향상시켜 다양한 제형으로 응용할 수 있으며, 피부 자극 최소화할 수 있음을 확인하였다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술한 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 상기 기재된 특정한 실시예에 본 발명의 범위가 제한되는 것은 아니다.
