Description
本発明は、新規なCUGリピート配列の結合剤等に関する。
リピート病(又は、トリプレットリピート病)は、遺伝子上の3塩基の繰り返し(リピート)配列の異常伸長が原因で発症する遺伝性神経疾患である。
例えば、リピート病の一種である筋強直性ジストロフィー1型(Myotonic Dystrophy Type 1(DM1))は、筋強直性ジストロフィープロテインキナーゼ(DMPK)に存在するCTGリピート配列の異常伸長によって発症する疾患である。具体的には、異常伸長したCTGリピート配列から転写されて生じる、CUGリピート配列を有するRNA(CUGリピートRNA)が、RNA結合性タンパク質に結合凝集することによるタンパク質の機能低下が、DM1の発症原因と考えられている。
例えば、リピート病の一種である筋強直性ジストロフィー1型(Myotonic Dystrophy Type 1(DM1))は、筋強直性ジストロフィープロテインキナーゼ(DMPK)に存在するCTGリピート配列の異常伸長によって発症する疾患である。具体的には、異常伸長したCTGリピート配列から転写されて生じる、CUGリピート配列を有するRNA(CUGリピートRNA)が、RNA結合性タンパク質に結合凝集することによるタンパク質の機能低下が、DM1の発症原因と考えられている。
これまで、CUGリピートRNAに結合することにより、CUGリピートRNAとRNA結合性タンパク質との凝集体から、RNA結合性タンパク質を解離させる分子が、DM1治療の研究に役立つ分子ツールであることが報告されている(非特許文献1~3)。

本発明の目的は、新規なCUGリピート配列の結合剤を提供することにある。
本発明の他の目的は、新規なリピート病の治療及び/又は予防剤を提供することにある。
上記のように、DM1治療に役立つ可能性のある分子が報告されているものの、どのような化合物にすればDM1の治療に繋がるのかについては具体的な指標が存在せず、化合物の分子設計を手探りで実施しなければならなかった。
本発明者らは、上記課題を解決するために鋭意研究を重ねた結果、CUGリピート配列に対する結合性についての表面プラズモン共鳴(SPR)による特定の指標(の大きさ)において有限値を示すことで、CUGリピートに結合していることを見出した。そして、その指標が、DM1などのリピート病によるスプライシング異常の改善に直接的な関係性があること、また、その指標が特定の値となる程度に結合性が強い場合、リピート病の治療に有効となることを見出した。また、このような指標によれば、DM1などのリピート病によるスプライシング異常を改善できる化合物の分子設計を、容易に実施しうることも見出した。
本発明者らは、上記課題を解決するために鋭意研究を重ねた結果、CUGリピート配列に対する結合性についての表面プラズモン共鳴(SPR)による特定の指標(の大きさ)において有限値を示すことで、CUGリピートに結合していることを見出した。そして、その指標が、DM1などのリピート病によるスプライシング異常の改善に直接的な関係性があること、また、その指標が特定の値となる程度に結合性が強い場合、リピート病の治療に有効となることを見出した。また、このような指標によれば、DM1などのリピート病によるスプライシング異常を改善できる化合物の分子設計を、容易に実施しうることも見出した。
一方、発明者らは、特定の構成単位を含む新規化合物も見出した。また、このような新規化合物が、上記特定の指標に照らしても良好なものであること等も見出し、さらに研究を重ねて本発明を完成した。
すなわち、本発明は、以下の発明等に関する。

本発明の剤は、濃度25nM、(CUG)9のRNA固定化量401RUで表面プラズモン共鳴によって測定されるレゾナンスユニット(以下、単に「指標A」ということがある。)が10RU以上である化合物Aを含む。

例えば、化合物濃度Y(nM)、(CUG)9のRNA固定化量Z(RU)で測定したレゾナンスユニットがW(RU)の場合、式 W×(401/Z)×(25/Y)によって、化合物濃度25nM、(CUG)9のRNA固定化量401RUにおけるレゾナンスユニットを算出することができる。
例えば、化合物濃度Y(nM)、(CUG)9のRNA固定化量Z(RU)で測定したレゾナンスユニットがW(RU)の場合、式 W×(401/Z)×(25/Y)によって、化合物濃度25nM、(CUG)9のRNA固定化量401RUにおけるレゾナンスユニットを算出することができる。
表面プラズモン共鳴は、例えば、後述する方法によって測定することができる。

また、表面プラズモン共鳴測定の測定バッファーとして、例えば、HEPESバッファー(例えば、10mM HEPESバッファーの500mM 塩化ナトリウム溶液等のHEPESバッファーの塩化ナトリウム溶液)を用いることができる。
例えば、表面プラズモン共鳴の測定装置としては、例えば、BIAcore T200(GE Healthcare製)を用いることができる。
また、表面プラズモン共鳴測定のセンサーチップとしては、例えば、ストレプトアビジンがコートされたセンサーチップを用いることができる。

また、化合物Aとしては、上記指標Aが10RU以上である化合物の中でも、表面プラズモン共鳴のレスポンスカーブにおいて、急激な解離を示すものではなく、結合状態をある程度保持した後に解離を示すものであれば、CUGリピートとの結合性をより奏しうる。
本発明の剤は、CUGリピート配列の結合剤、リピート病の治療及び/又は予防剤等として機能することができる。

(化合物A)
化合物Aは、上記指標Aが、通常、10RU以上であり、好ましくは、11RU以上(例えば、12RU以上、13RU以上等)であってもよい。
当該指標Aが10RU以上であれば、Atp2a1(筋小胞体Ca2+/ATPアーゼ遺伝子)等において正常なスプライシングが行われやすいため、リピート病の治療及び/又は予防に有効となりうる。
なお、後述のように、ウラシルと相補的な水素結合を形成可能な骨格、芳香環(さらにはその置換基の種類)、さらにはこれらの数等は、指標Aの値を大きくする要因となりやすい。そのため、このような要因となる骨格等を有する化合物は、化合物Aの他、化合物Aの一部を構成する骨格(又は原料又は前駆体、例えば、後述のJM608)等として好適に利用(又は使用)しうる。

本合成例において、試薬は市販品を使用し、精製をせずに用いた。
HPLCは、Gilson 811C Dynamic Mixer(測定波長254 nm、Cosmosil 5C18-MS-IIカラム(150×20mm))を使用し、2種の溶媒系(0.1%AcOH/H2O、及びMeCN)を用いて測定した。
1H NMR及び13C NMRスペクトルは、ECS400(JEOL)、ECA600(JEOL)及びAvanceIII700(BRUKER)を用いて測定した。
ESI質量分析は、JEOL AccuTOF-T100N mass spectrometerを用いて測定した。

5-ブロモ-1,3-ジクロロイソキノリン(化合物2)の合成(上記工程a)
1,3-ジクロロイソキノリン(化合物1)(2.0g、10.1mmol)及びN-ブロモスクシンイミド(2.3g、12.9mmol)を、超脱水アセトニトリル(50mL)中で混合した後、硫酸(2mL)を滴下し、混合物を室温で3日間撹拌した。得られた固体を濾過し、ヘキサンで洗浄した。得られた白色固体を乾燥し、化合物2を得た(1.2g、収率44%)。
1,3-ジクロロイソキノリン(化合物1)(2.0g、10.1mmol)及びN-ブロモスクシンイミド(2.3g、12.9mmol)を、超脱水アセトニトリル(50mL)中で混合した後、硫酸(2mL)を滴下し、混合物を室温で3日間撹拌した。得られた固体を濾過し、ヘキサンで洗浄した。得られた白色固体を乾燥し、化合物2を得た(1.2g、収率44%)。

化合物2(1.0g、3.6mmol)を1,4-ジオキサン(8mL)に溶解した後、ジイソプロピルアミン(1mL)及びN-Boc-1,3-プロパンジアミン(3mL)を添加し、得られた混合物を一晩還流した。得られた溶液をNH4Cl水溶液によって中和した後、ジクロロメタンによって抽出した。有機層を無水硫酸マグネシウムで乾燥した後、減圧下で濃縮した。得られた物をシリカゲルカラムクロマトグラフィー(1%メタノール/CHCl3を使用)によって精製し、化合物3を白色固体として得た(1.5g、収率54%)。
化合物2(1.0g、3.6mmol)を1,4-ジオキサン(8mL)に溶解した後、ジイソプロピルアミン(1mL)及びN-Boc-1,3-プロパンジアミン(3mL)を添加し、得られた混合物を一晩還流した。得られた溶液をNH4Cl水溶液によって中和した後、ジクロロメタンによって抽出した。有機層を無水硫酸マグネシウムで乾燥した後、減圧下で濃縮した。得られた物をシリカゲルカラムクロマトグラフィー(1%メタノール/CHCl3を使用)によって精製し、化合物3を白色固体として得た(1.5g、収率54%)。

化合物5(8.5mg、10.8μmol)のCHCl3(1mL)溶液に、4M HCl含有酢酸エチル(2mL)を加えた後、室温で1時間撹拌した。乾燥状態になるまで溶媒を蒸発させ、化合物JM608を黄色固体として得た(5.7mg、収率90%)。生成物を、さらにHPLCによって精製した。

化合物6(316mg、389μmol)をメタノール(170mL)に溶解した後、パラジウム-炭素(Pd/C)(10質量%)(60mg)を加えた。得られた混合物を、水素ガス下、室温で24時間撹拌した。Pd/Cをショートパッドセライトカラムによってろ過した後、溶媒を濃縮した。得られた物を、アミノ基コートシリカゲルを用いて、2%MeOH/CHCl3によって溶出し、化合物7を淡黄色固体として得た(219mg、収率74%)。
化合物6(316mg、389μmol)をメタノール(170mL)に溶解した後、パラジウム-炭素(Pd/C)(10質量%)(60mg)を加えた。得られた混合物を、水素ガス下、室温で24時間撹拌した。Pd/Cをショートパッドセライトカラムによってろ過した後、溶媒を濃縮した。得られた物を、アミノ基コートシリカゲルを用いて、2%MeOH/CHCl3によって溶出し、化合物7を淡黄色固体として得た(219mg、収率74%)。

5’-ビオチン化r(CUG)9(Thermo Fisher Scientific Inc.製)を、HEPESバッファー(10mM HEPES 及び 500mM NaCl)で0.1μMまで希釈した後、上記センサーチップに、結合量が400RU付近になるまで流した。センサーチップ表面に固定化されたr(CUG)9は、401RUだった。

シングルサイクルカイネティクス(SCK)法を実施するため、センサーチップ表面に25℃で、HBS-EP+バッファーを30μL/分で、120秒間流すことにより整えた後、化合物JM608を、濃度が0.063μM、0.125μM、0.25μM、0.50μM及び1.0μMとなるようにHBS-EP+バッファーに溶解し、得られた溶液を、センサー表面に、1サイクルあたり30μL/分で、60秒間ずつ、順次注入した。
得られたSPRレスポンスカーブを、BIAcore T200 評価ソフトウェア(version 2.0)を用いて解析した。
結果を図1に示す。

化合物JM608の代わりに化合物JM642を用い、化合物JM642を、濃度が6nM、13nM、25nM、50nM及び100nMとなるように順次注入した以外は実施例1と同様にして、SPR測定を行った。結果を図2に示す。

実施例1については、化合物濃度63nMにおける測定値(1.9RU)を用い、計算式 1.9RU×(25nM/63nM)により、化合物濃度25nMにおけるレゾナンスユニットを算出した。

図1~2及び表2に示すように、実施例1及び2では、CUGリピート配列に対する結合性が確認された。
(DM1マウスモデルにおけるスプライシング異常の改善評価)
(実施例3)Atp2a1(筋小胞体Ca2+/ATPアーゼ遺伝子)についての評価
性別と年齢を一致させた系統20b(FVB近交系バックグラウンド)のホモ接合型HSALRトランスジェニックマウス(Science 2000,289,1769―1772)(3か月未満)に、指定された用量(10mg/kg/day又は20mg/kg/day)で5日間、JM642を毎日腹腔内に投与した。処置後、スプライシング分析のために大腿直筋(大腿四頭筋)を採取した。組織からの全RNA抽出、cDNA合成、およびポリメラーゼ連鎖反応(PCR)を、Ann.Clin.Transl.Neurol.2016,3,42―54に記載の方法で行った。PCR産物をアガロースゲル電気泳動で分離し、ゲルをGelRed(Biotium製)で染色した。Typhoonレーザーフルオロイメージャー(GE Healthcare製)を使用してゲルを画像化し、ImageQuant(GE Healthcare製)を使用してゲルバンドを定量化した。
また、JM642の投与を行わなかった以外は上記と同じ条件のマウスと、正常なマウスに対して、上記と同様に、全RNA抽出、cDNA合成、およびPCRを行い、PCR産物の電気泳動を行った。
結果を図4に示す。
(実施例3)Atp2a1(筋小胞体Ca2+/ATPアーゼ遺伝子)についての評価

図3に示すように、Atp2a1において、正常なスプライシングであれば、エクソン(Exon)22が取り込まれる(残存する)が(図3におけるWild Type)、DM1患者ではExon22は取り込まれない。
これに対し、図4に示すように、DM1マウスモデルに化合物JM642を投与したところ、Atp2a1におけるExon22(ex22)の残存率が高かった。

(参考例1)
化合物JM642の代わりに、上記非特許文献3に記載の方法に従って合成した下記化合物(DDAP)を使用した以外は実施例3と同様にして、評価を行った。結果を図5に示す。

図4及び図5において、例えば、化合物の投与量20mg/kg/dayにおけるExon22の残存率から示されるように、実施例3(残存率70%)は、参考例1(残存率30%)と比較して、DM1のスプライシング異常をより改善できた。
(実施例4)Clcn1(筋特異的クロライドチャネル遺伝子)についての評価
Atp2a1の代わりにClcn1を用いたこと以外は実施例3と同様にして、評価を行った。結果を図7に示す。

図6に示すように、Clcn1において、正常なスプライシングであれば、Exon7aが取り込まれないが(Wild Type)、DM1患者ではExon7aが取り込まれる。
これに対し、図7に示すように、DM1マウスモデルに化合物JM642を投与したところ、Clcn1におけるExon7a(ex7a)のスキップ率が高かった。

化合物JM642の代わりに、上記非特許文献3に記載の方法に従って合成した上記化合物DDAPを使用した以外は実施例4と同様にして、評価を行った。結果を図8に示す。

図7及び図8において、例えば、化合物の投与量20mg/kg/dayにおけるExon7aのスキップ率から示されるように、実施例4(スキップ率70%)は、参考例2(スキップ率60%)と比較して、DM1のスプライシング異常をより改善できた。
本発明によれば、リピート病の治療及び/又は予防剤等として有用な剤等を提供できる。
Claims (10)
- 濃度25nM、(CUG)9のRNA固定化量401RUで表面プラズモン共鳴によって測定されるレゾナンスユニットが10RU以上である化合物Aを含む、CUGリピート配列の結合剤。
- 濃度25nM、(CUG)9のRNA固定化量401RUで表面プラズモン共鳴によって測定されるレゾナンスユニットが10RU以上である化合物Aを含む、リピート病の治療及び/又は予防剤。
- 化合物Aが、ウラシルと相補的な水素結合を形成可能な骨格を有する請求項1又は2記載の剤。
- 式(1A)において、R1が、芳香環基である請求項5記載の化合物。
- リピート病が、CTGリピートの異常伸長に起因する疾患である請求項2~4のいずれか記載の剤。
- リピート病が、筋強直性ジストロフィー1型である請求項2~4及び7のいずれか記載の剤。
- 濃度25nM、(CUG)9のRNA固定化量401RUで表面プラズモン共鳴によって測定されるレゾナンスユニットが10RU以上であることを指標とする、化合物Aのスクリーニング方法。
- 化合物Aが、CUGリピート配列の結合剤、リピート病の治療剤、及び/又はリピート病の予防剤である、請求項9記載の方法。
