Abstract
グルコセレブロシダーゼ活性を有し、さらに熱安定性を有するタンパク質を提供する。そして、植物由来であり、グリコシドハイドロラーゼファミリー1(GH1)に属し、且つグルコセレブロシダーゼ活性を有するタンパク質により、上記課題を解決する。
Description
本発明は、グルコセレブロシダーゼ活性および熱安定性を有するタンパク質(以下においては、これを「熱安定性グルコセレブロシダーゼ」という場合もある)、このタンパク質を含む酵素組成物、医薬組成物または食品組成物、このタンパク質を用いたセラミドの製造方法等に関する。
糖脂質の一種であるグルコシルセラミドを加水分解してセラミドに変換する酵素として、グルコセレブロシダーゼが知られている。このグルコセレブロシダーゼは、主に動物での存在が認められており、動物の体内においてグルコシルセラミドからセラミドを生成させるという重要な役割を果たしているが、植物ではその存在がほとんど知られていない。
そして、ヒトにおいては、先天的にグルコセレブロシダーゼ遺伝子が欠損しており、体内のグルコシルセラミドをセラミドに変換できない先天性代謝異常症(ゴーシェ病)が知られている。このゴーシェ病では、グルコシルセラミドが体内に異常蓄積し、肝臓や脾臓の肥大、貧血や血小板の減少、骨の異常などの様々な症状が現れる(非特許文献1、2)。
ゴーシェ病患者は、世界に約5千人以上いると言われており、その治療方法としては、主として、不足しているグルコセレブロシダーゼを点滴薬として体内に補充し、蓄積されたグルコシルセラミドをセラミドに変換させる治療が行われている。

ここで、現在、ゴーシェ病の点滴薬として用いられている酵素剤には、遺伝子組み換え技術を用いてチャイニーズハムスターの卵巣細胞において生産させた、ヒト由来のグルコセレブロシダーゼであるイミグルセラーゼが主に使用されている。しかしながら、このイミグルセラーゼは薬価が高額の上、熱安定性が低いために2週間に1回の補充が必要とされ、患者にとって大きな経済的負担となっているという課題がある。
なお、動物由来のグルコセレブロシダーゼは動物の体温付近で作用する酵素であるため、いずれも上記したイミグルセラーゼと同様に熱安定性が低い。そこで、もし熱安定性を有し、補充回数が少なくて済むようなグルコセレブロシダーゼがあれば、その医療上の価値は計り知れないが、まだそのようなグルコセレブロシダーゼの報告はない。
一方、ヒトの肌の角質などに存在するセラミドは、肌の潤いを保つために必要な成分であり、化粧品などに配合されているが、この化粧品等の有効成分としてのセラミドは、通常、化学合成により製造されている。ここで、動物や植物中の含有率が高く、比較的安価なグルコシルセラミドをグルコセレブロシダーゼにより分解してセラミドを製造し、このセラミドを化粧品や試薬などに用いることが考えられるが、上記のイミグルセラーゼは非常に高価であり、熱安定性も低いため、実用化には至っていない。なお、動物や植物中のセラミド含有率はグルコシルセラミドと異なり非常に低いため、このセラミドを直接抽出して精製する場合には莫大なコストがかかってしまう。
そこで本発明は、グルコセレブロシダーゼ活性を有し、さらに熱安定性を有するタンパク質を提供することを目的とする。
上記課題を解決するために本発明者は鋭意検討し、植物由来であり、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性を有するタンパク質を見出した。さらにこのタンパク質は熱安定性を有することを見出し、本発明を完成させた。なお、現在までに、植物からGH1に属するグルコセレブロシダーゼが見出されたという報告はない。
すなわち、本発明は次の<1>~<17>である。

本発明によれば、グリコシドハイドロラーゼファミリー1に属し、グルコセレブロシダーゼ活性および熱安定性を有するタンパク質を提供することができる。そして、このタンパク質を含む酵素組成物、医薬組成物、および食品組成物も提供することができ、さらには、このタンパク質を用いたセラミド製造方法の提供も可能となる。
本発明について説明する。
本発明は、植物由来であり、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性を有するタンパク質(以下においては、これを「本発明のタンパク質」あるいは「本発明の熱安定性グルコセレブロシダーゼ」ともいう)、本発明のタンパク質をコードするDNAおよびこのDNAを含む発現ベクター、この発現ベクターが導入された形質転換体およびこの形質転換体を用いた本発明のタンパク質の製造方法、本発明のタンパク質を含む酵素組成物、医薬組成物または食品組成物、本発明のタンパク質を用いたセラミドの製造方法等である。
本発明は、植物由来であり、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性を有するタンパク質(以下においては、これを「本発明のタンパク質」あるいは「本発明の熱安定性グルコセレブロシダーゼ」ともいう)、本発明のタンパク質をコードするDNAおよびこのDNAを含む発現ベクター、この発現ベクターが導入された形質転換体およびこの形質転換体を用いた本発明のタンパク質の製造方法、本発明のタンパク質を含む酵素組成物、医薬組成物または食品組成物、本発明のタンパク質を用いたセラミドの製造方法等である。
まず、本発明のタンパク質について詳細に説明する。
本発明のタンパク質は、植物由来であり、グリコシドハイドロラーゼファミリー1(GH1)に属し、且つグルコセレブロシダーゼ活性を有するタンパク質である。そして、この植物由来のGH1に属するグルコセレブロシダーゼ活性を有するタンパク質は、熱安定性を有している。
本発明のタンパク質は、植物由来であり、グリコシドハイドロラーゼファミリー1(GH1)に属し、且つグルコセレブロシダーゼ活性を有するタンパク質である。そして、この植物由来のGH1に属するグルコセレブロシダーゼ活性を有するタンパク質は、熱安定性を有している。
ここで、「グルコセレブロシダーゼ活性」とは、EC番号が(EC3.2.1.45)である酵素(グルコセレブロシダーゼ)が有する酵素活性であり、つまり糖脂質であるグルコシルセラミドのグルコースとセラミドのβ-1,4-グリコシル結合を加水分解してセラミドを生成する反応を触媒する活性である。また、「グリコシドハイドロラーゼファミリー(Glycoside Hydrolase family)1」とは、Carbohydrate Active enzyme database(CAZy databe,http://www.cazy.org/)によって130程度に分類された糖質加水分解酵素のファミリーの1つであり、グルコセレブロシダーゼは、グリコシドハイドロラーゼファミリー1(GH1)に属するもの、グリコシドハイドロラーゼファミリー30(GH30)に属するもの、およびグリコシドハイドロラーゼファミリー116(GH116)に属するものが知られている。
また、「植物由来」とは、植物の遺伝子から発現されたタンパク質であることを意味し、この植物の遺伝子は、例えば、植物の葉、茎、根、種子、実、花びら、めしべ、花粉(おしべ)、仮根、および胞子嚢からなる群から選ばれる少なくとも1つから取得することができる。
さらに、「熱安定性」とは、0.1%Triton X-100(登録商標、以下同じ)および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.0)に、0.0002~0.0008U/mLのグルコセレブロシダーゼ活性となるようにグルコセレブロシダーゼ活性を有するタンパク質を含む溶液において、37℃で30時間インキュベーションしたときにこのグルコセレブロシダーゼ活性が80%以上残存する(このインキュベーション前のグルコセレブロシダーゼ活性を100%としたときの相対残存活性が80%以上である)性能を意味する。なお、この熱安定性は、上記した相対残存活性が90%以上であるのが好ましい。ここで、このグルコセレブロシダーゼ活性は、以下のようにして測定されたものである。すなわち、100μMグルコシルセラミド(セレブロシドB、(4E,8E)-N-D-2´-hydroxypalmitoyl-1-O-β-D-glucopyranosyl-9-methyl-4,8-sphingadienine (「E」、「N」および「O」はイタリック体、「D」は小型英大文字):The Journal of Antibiotics 41,(1988)469-480、以下同じ)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に試料を所定量添加し、37℃で15~60分間インキュベーションし、次いで、得られた酵素反応液に4倍量のエタノールを加え混合後、15000rpmで20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成されたセラミド量を測定する。上記以外のグルコセレブロシダーゼ活性測定に必要な条件は、後述する実施例2に記載の方法に従って実施する。
さらに、「熱安定性」とは、0.1%Triton X-100(登録商標、以下同じ)および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.0)に、0.0002~0.0008U/mLのグルコセレブロシダーゼ活性となるようにグルコセレブロシダーゼ活性を有するタンパク質を含む溶液において、37℃で30時間インキュベーションしたときにこのグルコセレブロシダーゼ活性が80%以上残存する(このインキュベーション前のグルコセレブロシダーゼ活性を100%としたときの相対残存活性が80%以上である)性能を意味する。なお、この熱安定性は、上記した相対残存活性が90%以上であるのが好ましい。ここで、このグルコセレブロシダーゼ活性は、以下のようにして測定されたものである。すなわち、100μMグルコシルセラミド(セレブロシドB、(4E,8E)-N-D-2´-hydroxypalmitoyl-1-O-β-D-glucopyranosyl-9-methyl-4,8-sphingadienine (「E」、「N」および「O」はイタリック体、「D」は小型英大文字):The Journal of Antibiotics 41,(1988)469-480、以下同じ)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に試料を所定量添加し、37℃で15~60分間インキュベーションし、次いで、得られた酵素反応液に4倍量のエタノールを加え混合後、15000rpmで20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成されたセラミド量を測定する。上記以外のグルコセレブロシダーゼ活性測定に必要な条件は、後述する実施例2に記載の方法に従って実施する。
そして、本発明のタンパク質は、種子植物由来であるのが好ましい。種子植物としては、限定されるものではないが、アオイ科植物、アカザ科植物、アカネ科植物、アサ科植物、アジサイ科植物、アブラナ科植物、アヤメ科植物、イネ科植物、ウコギ科植物、ウリ科植物、ウルシ科植物、カヤツリグサ科植物、キキョウ科植物、キク科植物、クスノキ科植物、クワ科植物、ケシ科植物、サトイモ科植物、サボテン科植物、シソ科植物、スイレン科植物、セリ科植物、タデ科植物、ツツジ科植物、ツバキ科植物、ナス科植物、ナデシコ科植物、ニレ科植物、ハス科植物、バラ科植物、ハス科植物、ヒガンバナ科植物、ヒルガオ科植物、ブドウ科植物、ブナ科植物、ボタン科植物、マメ科植物、ミカン科植物、ミズアオイ科植物、モクセイ科植物、ヤシ科植物、ヤナギ科植物、ユリ科植物、ラン科植物、イチイ科植物、イチョウ科植物、スギ科植物、ソテツ科植物、ヒノキ科植物、マツ科植物などが具体例として示される。

この中で、アブラナ科植物、イネ科植物、ウリ科植物、キク科植物、ナス科植物、バラ科植物、ヒガンバナ科植物、マメ科植物、またはユリ科植物のいずれか由来のGH1に属し且つグルコセレブロシダーゼ活性を有するタンパク質は、その熱安定性が非常に優れるため特に好適であり、例えば、イネ由来またはダイズ由来のGH1に属し且つグルコセレブロシダーゼ活性を有するタンパク質は、そのグルコセレブロシダーゼ活性および熱安定性がいずれも極めて好適である。
なお、イネ由来である本発明の熱安定性グルコセレブロシダーゼは、配列表の配列番号1に示されるアミノ酸配列、あるいはシグナルペプチドが切断されたアミノ酸配列であるこの配列番号1のうちアミノ酸番号38~521に示されるアミノ酸配列からなる。また、ダイズ由来である本発明の熱安定性グルコセレブロシダーゼは、配列表の配列番号2に示されるアミノ酸配列、あるいはシグナルペプチドが切断されたアミノ酸配列であるこの配列番号2のうちアミノ酸番号19~503に示されるアミノ酸配列からなる。
また、配列表の配列番号1、この配列番号1のうちアミノ酸番号38~521、配列表の配列番号2、またはこの配列番号2のうちアミノ酸番号19~503のいずれかに示されるアミノ酸配列において、1もしくは数個のアミノ酸の置換、欠失、挿入、または付加がされたアミノ酸配列からなり、GH1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質も本発明に包含される。この場合、このタンパク質は植物由来であるのが好ましいが、植物由来でなくても良い(後述する「本発明のタンパク質」には、植物由来でないものも含まれる場合もある)。ここで、「数個」とは10個以下であることを意味し、6個以下であるのが好ましく、5個以下であるのがより好ましい。
なお、このアミノ酸の置換、欠失、挿入、または付加は、保存的なものである。つまり、タンパク質の性質を実質的に改変しないように1個もしくは数個のアミノ酸残基を置換、欠失、挿入、または付加することである。例えば、疎水性アミノ酸残基を別の疎水性アミノ酸残基によって置換する場合や、極性アミノ酸残基を同じ電荷を有する別の極性アミノ酸残基によって置換する場合などが挙げられる。このような機能的に類似のアミノ酸としては、具体的には、疎水性(非極性)アミノ酸としては、アラニン、バリン、イソロイシン、ロイシン、プロリン、トリプトファン、フェニルアラニン、メチオニン等が挙げられる。また、極性アミノ酸のうち、中性アミノ酸としては、グリシン、セリン、スレオニン、チロシン、グルタミン、アスパラギン、システイン等が挙げられる。塩基性アミノ酸としては、アルギニン、ヒスチジン、リジン等が挙げられる。また、酸性アミノ酸としては、アスパラギン酸、グルタミン酸等が挙げられる。
さらに、配列表の配列番号1、配列番号1のうちアミノ酸番号38~521、配列番号2、または配列番号2のうちアミノ酸番号19~503のいずれかに示されるアミノ酸配列に対して60%以上、より好ましくは62%以上、さらに好ましくは65%以上、さらに好ましくは67%以上、さらに好ましくは70%以上、さらに好ましくは73%以上、さらに好ましくは77%以上の相同性を有し、GH1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質も本発明に包含される。この場合も、このタンパク質は植物由来であるのが好ましいが、植物由来でなくても良い。ここで、「相同性」とは、相同性検索プログラムであるEMBOSS Needle(https://www.ebi.ac.uk/Tools/psa/emboss_needle/)においてデフォルト(初期設定)のパラメータを用いて算出される数値である。
次に、本発明のタンパク質をコードするDNA、およびこのDNAを用いたタンパク質の製造方法について詳細に説明する。
本発明のタンパク質をコードするDNAは、このタンパク質を発現することが可能な塩基配列により構成されていれば、天然由来のものや、天然由来のものの一部を利用して合成を行ったものなどであって良く、限定はされない。例えば、前述したイネ由来である本発明の熱安定性グルコセレブロシダーゼをコードするDNAとして、配列表の配列番号3または配列番号3のうち塩基番号112~1566に示される塩基配列からなるDNAが示される。また、前述したダイズ由来である本発明の熱安定性グルコセレブロシダーゼをコードするDNAとして、配列表の配列番号4または配列番号4のうち塩基番号55~1512に示される塩基配列からなるDNAが示される。
さらに、配列表の配列番号3、配列番号3のうち塩基番号112~1566、配列番号4、または配列番号4のうち塩基番号55~1512のいずれかに示される塩基配列において、1もしくは数個の塩基の置換、欠失、挿入、または付加がされた塩基配列からなり、GH1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質をコードするDNAも例示される。ここで、「数個」とは20個以下であることを意味し、10個以下であるのが好ましく、6個以下であるのがより好ましい。
さらには、配列表の配列番号3、配列番号3のうち塩基番号112~1566、配列番号4、または配列番号4のうち塩基番号55~1512のいずれかに示される塩基配列と相補的な塩基配列からなるDNAとストリンジェントな条件下でハイブリダイゼーションすることができ、GH1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質をコードするDNAも例示される。
ここで、「ストリンジェントな条件」とは、いわゆる特異的なハイブリッドが形成され、非特異的なハイブリッドが形成されない条件である。一例を示せば、0.7~1.0M塩化ナトリウム存在下、65℃でハイブリダイゼーションを行った後、0.1~5×SSC、0.1%SDS溶液(1×SSCの組成:150mM塩化ナトリウム、15mMクエン酸ナトリウム)を使用し、60℃、好ましくは65℃、より好ましくは68℃での洗浄が1~3回行われる条件などが挙げられる。
ここで、「ストリンジェントな条件」とは、いわゆる特異的なハイブリッドが形成され、非特異的なハイブリッドが形成されない条件である。一例を示せば、0.7~1.0M塩化ナトリウム存在下、65℃でハイブリダイゼーションを行った後、0.1~5×SSC、0.1%SDS溶液(1×SSCの組成:150mM塩化ナトリウム、15mMクエン酸ナトリウム)を使用し、60℃、好ましくは65℃、より好ましくは68℃での洗浄が1~3回行われる条件などが挙げられる。
そして、本発明においては、宿主となる植物体、植物細胞、動物細胞(昆虫細胞も含む)、もしくは微生物内において複製可能であり、且つ、前述したDNAを、そのコードするタンパク質を発現可能な状態で含む発現ベクターも提供される。つまり、GH1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質発現用の発現ベクターも提供される。この発現ベクターは、例えば、宿主の細胞の染色体外に独立体として存在し、その複製が染色体DNAの複製に依存しない自己複製ベクターや、宿主の細胞の染色体DNAに組み込まれてこの染色体DNAとともに複製されるベクターなどを用いて構築することができ、プラスミドベクターやウイルスベクターなどが好ましい例として示される。なお、発現ベクター構築の手順および方法は、遺伝子工学の分野で慣用されている方法を用いることができる。
この発現ベクターは、これを実際に導入した形質転換体において本発明のタンパク質を発現させるために、本発明のタンパク質をコードするDNAの他に、その発現を制御する塩基配列や形質転換体を選択するための遺伝子マーカー等を含んでいるのが好ましい。発現を制御する塩基配列としては、プロモーター、ターミネーター、前述したもの以外のシグナルペプチドをコードする塩基配列等が例示される。プロモーターは宿主において転写活性を示すものであれば特に限定されない。シグナルペプチドも、宿主においてタンパク質の細胞外への分泌に寄与するものなどであれば特に限定されない。また、遺伝子マーカーは、形質転換体の選択の方法に応じて適宜選択されて良いが、例えば薬剤耐性遺伝子、栄養要求性を相補する遺伝子などを利用することができる。
さらに、本発明においては、上記した発現ベクターが宿主の細胞内および/または染色体DNAに導入された形質転換体も提供される。この宿主-ベクター系は特に限定されず、例えば、植物体、植物細胞、動物細胞、もしくは微生物(大腸菌、酵母、糸状菌など)を用いた系、それらを用いた他のタンパク質との融合タンパク質発現系などを用いることができる。また、上記した発現ベクターの宿主への導入、つまり上記した発現ベクターを用いた宿主の形質転換も、この分野で慣用されている方法に従い実施することができる。ここで、形質転換させる宿主としては、植物体、植物細胞、動物細胞、大腸菌、酵母、または糸状菌のいずれかを用いるのが好ましい。つまり、上記した発現ベクターが導入された、植物体、植物細胞、動物細胞、大腸菌、酵母、または糸状菌のいずれかの形質転換体であるのが好ましい。例えば、上記した発現ベクターが導入された植物体、植物細胞、または大腸菌の形質転換体において、本発明のタンパク質を大量発現させることができる。
そして、この形質転換体を、その形質を維持したまま発育および増殖が可能な条件で育種または培養し、得られた形質転換体(植物体、植物培養細胞、動物培養細胞など)またはその含有物(培養細胞を含む培養液や固体培地など)から本発明のタンパク質を回収することができる。したがって、本発明では、この形質転換体を育種または培養する工程、および、この工程によって得られる形質転換体もしくはその含有物から本発明のタンパク質を回収(粗製、あるいは精製)する工程を含む、本発明のタンパク質の製造方法が提供される。形質転換体の育種、培養方法およびその条件は、形質転換体がその形質を維持したまま発育および増殖可能な方法、条件であれば良く、特段限定はされないが、宿主として使用する植物体、植物細胞、動物細胞、もしくは微生物についての育種または培養方法、条件と実質的に同等であって良い。また、形質転換体を育種または培養した後、目的のタンパク質を回収する方法も、この分野で慣用されている粗製方法、精製方法を用いることができる。

また、本発明のタンパク質を製造する方法の好ましい実施態様の他の例として、大腸菌細胞、酵母細胞、あるいは糸状菌細胞の形質転換体を使用する方法が挙げられる。酵母細胞としては、サッカロミセス(Saccharomyces)属、ハンゼヌラ(Hansenula)属、またはピキア(Pichia)属に属する微生物、例えばサッカロミセス・セレビシエ(Saccharomyces cerevisiae)が挙げられる。糸状菌細胞としては、フミコーラ(Humicola)属、トリコデルマ(Trichoderma)属、スタフィロトリクム(Staphylotrichum)属、アスペルギルス(Aspergillus)属、フザリウム(Fusarium)属、またはアクレモニウム(Acremonium)属に属するものが挙げられる(括弧内の学名はいずれもイタリック体)。
次に、本発明のタンパク質を含む酵素組成物、医薬組成物、および食品組成物等について詳細に説明する。
本発明では、前述した本発明のタンパク質を含有する酵素組成物を提供することができる。この酵素組成物は、本発明のタンパク質を有効成分(酵素組成物が有するグルコセレブロシダーゼ活性の活性成分)として含むものであり、グルコシルセラミド加水分解用(グルコシルセラミド分子中のグルコースの加水分解用)として、つまりグルコシルセラミドのセラミドへの変換用として好適に使用することができる。
また、本発明のタンパク質を有効成分として含有させることにより、医薬組成物を提供することができる。そして、この医薬組成物は、ゴーシェ病の予防および治療用として好適に使用することができる。また、その形態としては、点滴薬とすることが好ましいが、経口薬(錠剤、散剤、シロップ剤など)であっても良い。
言い換えれば、本発明では、本発明のタンパク質を有効成分として含む医薬組成物をゴーシェ病患者に投与(例えば、点滴など)する、ゴーシェ病予防または治療方法を提供することができる。そして、その用法および用量は、ゴーシェ病患者に対して3~10週間当たり1回、より好ましくは4~8週間当たり1回、グルコセレブロシダーゼ活性として10~200U/体重1kgとなるように、本発明のタンパク質または本発明のタンパク質を有効成分として含む医薬組成物(ゴーシェ病の予防および治療剤)を投与するのが好適である。なお、この場合の「U(ユニット)」とは、合成基質p-ニトロフェニル-β-D-グルコピラノシド(「p」はイタリック体、「D」は小型英大文字)を37℃で1分間に1μmol分解する単位とする。
さらに、本発明のタンパク質と、植物、動物、または微生物(例えば担子菌など)より単離されたグルコシルセラミド、もしくは化学合成されたグルコシルセラミドと、を含有させることにより、組成物中のグルコシルセラミドからセラミドを容易に遊離させることができる食品組成物を提供することができる。そして、本発明のタンパク質と上記グルコシルセラミドとを有効成分として含有させることにより、この遊離するセラミドが機能性素材となる機能性食品として、肌潤い向上用、紫外線による肌の損傷予防用、炎症性腸疾患予防用、および大腸がん予防用から選ばれる1以上の用途の食品組成物を提供することもできる。さらには、生活習慣病(例えば糖尿病、心臓病、高血圧、高脂血症など)予防用の食品組成物を提供することもできる。
なお、本発明では、本発明の熱安定性グルコセレブロシダーゼを有効成分として含有させることにより、機能性食品として、ゴーシェ病予防または治療用の食品組成物や、グルコシルセラミドを含有させた食品組成物とともに摂取するセラミド補給用食品組成物を提供することもできる。そして、上記したゴーシェ病予防または治療用の食品組成物の用法および用量は、前述したゴーシェ病の予防および治療用医薬組成物と同じであって良い。
次に、本発明のタンパク質を用いたセラミドの製造方法について詳細に説明する。
本発明のタンパク質を用いたセラミドの製造方法は、本発明のタンパク質を用いて、植物、動物、または微生物(例えば担子菌など)より単離されたグルコシルセラミド、もしくは化学合成されたグルコシルセラミドから、セラミドを生成させる工程を含む。なお、この製造方法には、本発明の効果に大きな影響を与えない範囲において、上記以外の任意の工程を含んでいて良い。
本発明のタンパク質を用いたセラミドの製造方法は、本発明のタンパク質を用いて、植物、動物、または微生物(例えば担子菌など)より単離されたグルコシルセラミド、もしくは化学合成されたグルコシルセラミドから、セラミドを生成させる工程を含む。なお、この製造方法には、本発明の効果に大きな影響を与えない範囲において、上記以外の任意の工程を含んでいて良い。
グルコシルセラミドは、セラミドと比べると動植物などの生体内に比較的多く含まれており、このようなグルコシルセラミドから、前述した形質転換体によって大量発現させることにより安価に得られる本発明の熱安定性グルコセレブロシダーゼを用いることによって、低コストでセラミドを製造することができる。そして、このようにして得られたセラミドを用いて、肌の保湿改善効果、紫外線による肌の損傷の予防効果、大腸がんの予防効果などを目的とした化粧品または機能性食品を安価に提供することができ、さらには研究用試薬、薬品としてのセラミドも安価に提供することができる。
上記において説明した実施形態は、本発明の理解を容易にするための一例に過ぎず、本発明を限定するものではない。
以下、本発明の実施例について説明するが、本発明は以下の実施例にも限定されるものではなく、本発明の技術的思想内において様々な変形が可能である。
(実施例1)
イネ(Oryza sativa L. cv. Nipponbare(「Oryza sativa」はイタリック体):日本晴)の若葉10gを採取し、0.05%コール酸ナトリウムおよび0.3%Triton X-100を含む50mM酢酸緩衝液(pH5.5)中においてホモジナイザーにより破砕した。この破砕液を15000rpm、4℃の条件により20分間遠心分離し、遠心上清液を0.05%Tween20(登録商標、以下同じ)および0.025%コール酸ナトリウムを含む40mM酢酸ナトリウム緩衝液(pH5.5)を1000倍量用いて透析し、酵素抽出液を得た。この酵素抽出液50mLに、1mLのエタノールに溶解した10mMグルコシルセラミド(セレブロシドC、(4E,8E)-N-D-2´-hydroxy-(E)-3´-octadecenoyl-1-O-β-D-glucopyranosyl-9-methyl-4,8-sphingadienine(「E」、「N」および「O」はイタリック体、「D」は小型英大文字):The Journal of Antibiotics 41,(1988)469-480、以下同じ)および49mLの0.4%コール酸ナトリウムを添加し、45℃で5時間反応させた。得られた反応液を炭酸ナトリウム溶液および水酸化ナトリウム溶液によりpH11.5に調整した上で、酢酸エチル溶液を加えて混合し、3000rpm、4℃の条件により20分間遠心分離した。遠心分離後、得られた酢酸エチル層を乾固した後、80%エタノール溶液にこの乾固物を溶解し、高速液体クロマトグラフィー分析に供した。すなわち、TSKgel ODS-120T カラム(4.6mm×30cm、東ソー社製、登録商標(以下同じ))に試料を注入し、エタノール濃度81%の溶媒を流速1.0mL/minで流し、UV検出器(紫外吸収波長215nm)にて検出することにより、酵素反応によって新しく生成された物質を分取した。
そして、分取した物質の、IR(FTS-135、バイオラッド社製)、1H NMR、13C NMR(Varian UNITY plus 500 spectrometer、アジレント社製)、およびESI-MS分析(Agilent 6460、アジレント社製)を行った。この結果を下記表1に示す。
イネ(Oryza sativa L. cv. Nipponbare(「Oryza sativa」はイタリック体):日本晴)の若葉10gを採取し、0.05%コール酸ナトリウムおよび0.3%Triton X-100を含む50mM酢酸緩衝液(pH5.5)中においてホモジナイザーにより破砕した。この破砕液を15000rpm、4℃の条件により20分間遠心分離し、遠心上清液を0.05%Tween20(登録商標、以下同じ)および0.025%コール酸ナトリウムを含む40mM酢酸ナトリウム緩衝液(pH5.5)を1000倍量用いて透析し、酵素抽出液を得た。この酵素抽出液50mLに、1mLのエタノールに溶解した10mMグルコシルセラミド(セレブロシドC、(4E,8E)-N-D-2´-hydroxy-(E)-3´-octadecenoyl-1-O-β-D-glucopyranosyl-9-methyl-4,8-sphingadienine(「E」、「N」および「O」はイタリック体、「D」は小型英大文字):The Journal of Antibiotics 41,(1988)469-480、以下同じ)および49mLの0.4%コール酸ナトリウムを添加し、45℃で5時間反応させた。得られた反応液を炭酸ナトリウム溶液および水酸化ナトリウム溶液によりpH11.5に調整した上で、酢酸エチル溶液を加えて混合し、3000rpm、4℃の条件により20分間遠心分離した。遠心分離後、得られた酢酸エチル層を乾固した後、80%エタノール溶液にこの乾固物を溶解し、高速液体クロマトグラフィー分析に供した。すなわち、TSKgel ODS-120T カラム(4.6mm×30cm、東ソー社製、登録商標(以下同じ))に試料を注入し、エタノール濃度81%の溶媒を流速1.0mL/minで流し、UV検出器(紫外吸収波長215nm)にて検出することにより、酵素反応によって新しく生成された物質を分取した。
そして、分取した物質の、IR(FTS-135、バイオラッド社製)、1H NMR、13C NMR(Varian UNITY plus 500 spectrometer、アジレント社製)、およびESI-MS分析(Agilent 6460、アジレント社製)を行った。この結果を下記表1に示す。
この結果から、上記したイネ由来酵素によってセレブロシドCから生成された物質はセレブロシドC由来のセラミドであることが明らかとなった。さらに、この物質はヒト由来のグルコセレブロシダーゼであるイミグルセラーゼ(サノフィ社製)によってセレブロシドCから生成されたセラミドと上記IR、1H NMR、13C NMR、およびESI-MS分析結果が完全に一致した。これらの結果により、このイネ由来酵素にはグルコセレブロシダーゼ活性があることが明らかとなった。
(実施例2)
イネ(日本晴)の茎110gを採取し、細かく裁断した後、0.05%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)中においてホモジナイザーにより破砕した。この破砕液を10000rpm、4℃の条件において60分間遠心分離し、遠心上清液を取り除いた。さらに、この遠心沈殿物に0.05%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)を加え、5分攪拌後、10000rpm、4℃の条件において60分間遠心分離し、遠心上清液を取り除いた。そして、この遠心沈殿物に0.05%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)を加え、そこへグルコセレブロシダーゼの溶出に十分な量のTriton X-100を添加した。30分攪拌後、18000rpm、4℃の条件により60分間遠心分離した。得られた遠心上清液をデュラポア メンブレンフィルター0.45μm HV(メルクミリポア社製、登録商標(以下同じ))にてろ過した後、脱塩濃縮し、酵素抽出液を得た。
イネ(日本晴)の茎110gを採取し、細かく裁断した後、0.05%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)中においてホモジナイザーにより破砕した。この破砕液を10000rpm、4℃の条件において60分間遠心分離し、遠心上清液を取り除いた。さらに、この遠心沈殿物に0.05%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)を加え、5分攪拌後、10000rpm、4℃の条件において60分間遠心分離し、遠心上清液を取り除いた。そして、この遠心沈殿物に0.05%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)を加え、そこへグルコセレブロシダーゼの溶出に十分な量のTriton X-100を添加した。30分攪拌後、18000rpm、4℃の条件により60分間遠心分離した。得られた遠心上清液をデュラポア メンブレンフィルター0.45μm HV(メルクミリポア社製、登録商標(以下同じ))にてろ過した後、脱塩濃縮し、酵素抽出液を得た。
この酵素抽出液を、0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mM酢酸緩衝液(pH6.0)で平衡化させたHiTrap Q HPカラム(アマシャムバイオサイエンス社製、登録商標(以下同じ))にアプライした。そして、0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mM酢酸緩衝液(pH6.0)から、0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mM酢酸緩衝液(pH6.0)中に1M塩化ナトリウムを含む緩衝液に対して、グラジエント溶離法で溶出した。次に、グルコセレブロシダーゼ活性が強く認められた画分をプールし、限外濾過により脱塩濃縮した。この脱塩濃縮液を0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mM酢酸緩衝液(pH5.5)で平衡化させたHiTrap SP HPカラム(アマシャムバイオサイエンス社製)にアプライした。そして、0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mM酢酸緩衝液(pH5.5)から、0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mM酢酸緩衝液(pH5.78)中に1M塩化ナトリウムを含む緩衝液に対して、グラジエント溶離法で溶出し、分画した。そして、グルコセレブロシダーゼ活性が強く認められた酵素画分をプールし、限外濾過により脱塩濃縮した液をイネ由来グルコセレブロシダーゼ(RGC1)とした。
なお、画分のグルコセレブロシダーゼ活性は以下のように測定した。まず、100μMグルコシルセラミド(セレブロシドB)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に試料を所定量添加し、37℃で15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmで20分間遠心分離し、遠心上清液を高速液体クロマトグラフィー分析に供した。高速液体クロマトグラフィー分析は、TSKgel ODS-120T カラム(4.6mm×30cm)に試料を注入し、エタノール濃度83%の溶媒を流速0.8mL/minで流し、UV検出器(紫外吸収波長215nm)にて検出することにより、酵素反応によって新しく生成されたセラミド量を測定した。セラミド量はヒト由来のグルコセレブロシダーゼであるイミグルセラーゼ(サノフィ社製)によってセレブロシドBから生成されたセラミドを標準品として求めた。また、生成されたセラミドの分子量をネガティブLC-MS(アジレント社製)で調べることによってセレブロシドBから生成されたセラミドであることを確認した(ESI-MS m/z:564.5[M-H]-)。酵素反応液中で1分間に1μmolのセラミドを生成する酵素量を1U(ユニット)として酵素液1mLあたりのグルコセレブロシダーゼ活性を算出した。
また、精製されたRGC1のタンパク質濃度は、プロテインアッセイキット(バイオラッドラボラトリー社製)で牛血清アルブミンをスタンダードとして、あらかじめタンパク質濃度を測定しておいたイミグルセラーゼを標品として求めた。すなわち、精製されたRGC1溶液およびイミグルセラーゼ溶液をTSKgel Octyl-80Ts カラム(4.6mm×15cm、東ソー社製)に注入し、0.05%トリフルオロ酢酸中アセトニトリル溶液比率を上げるグラジエントモードで、流速0.8mL/minで流し、UV検出器(紫外吸収波長280nm)にて検出することによって生じたピーク面積を求め、イミグルセラーゼと比較することにより精製RGC1のタンパク質濃度を求めた。
このRGC1を含む画分はSDS-PAGEにおいて単一なバンドを示し、その平均分子量(MW)は約62kDであった。なお、SDS-PAGEは、AE-6000電気泳動(アトー株式会社製)およびプリキャストミニゲルe-PAGEL(E-R10L/ゲル濃度10%/18検体、アトー株式会社製)を使用して行い、Silver Stain MS Kit(和光純薬工業株式会社製)により銀染色を行った。分子量マーカーはSDS-PAGE Molecular Weight Standards Low Range(バイオラッドラボラトリー社製)を使用した。
(実施例3)
実施例2で得られたRGC1を、12.5μMのグルコシルセラミド(動物由来、植物由来、または糸状菌由来である下記表2の12種のいずれか)あるいは合成β-グルコシド、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に添加し、37℃にて15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液中のセラミド量を高速液体クロマトグラフィーにて各種逆相カラムを用いて測定することによってグルコセレブロシダーゼ活性を求めた。セラミド量はイミグルセラーゼによって上記グルコシルセラミドから生成されたセラミドを標準品として求めた。なお、グルコセレブロシダーゼ活性は、動物由来グルコシルセラミドであるd18:1(4E)-C8:0-GluCer(「E」はイタリック体)を基質にした場合の活性を100とした時の相対活性で求めた(下記表2の最右列)。なお、この試験は5回行い、その相対活性の平均値を求めた。この結果を下記表2に示す。
実施例2で得られたRGC1を、12.5μMのグルコシルセラミド(動物由来、植物由来、または糸状菌由来である下記表2の12種のいずれか)あるいは合成β-グルコシド、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に添加し、37℃にて15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液中のセラミド量を高速液体クロマトグラフィーにて各種逆相カラムを用いて測定することによってグルコセレブロシダーゼ活性を求めた。セラミド量はイミグルセラーゼによって上記グルコシルセラミドから生成されたセラミドを標準品として求めた。なお、グルコセレブロシダーゼ活性は、動物由来グルコシルセラミドであるd18:1(4E)-C8:0-GluCer(「E」はイタリック体)を基質にした場合の活性を100とした時の相対活性で求めた(下記表2の最右列)。なお、この試験は5回行い、その相対活性の平均値を求めた。この結果を下記表2に示す。
この結果、RGC1はd18:1(4E)-C18:0-GM3、d18:1(4E)-C8:0-LacCerのようなラクトシルセラミド、およびd18:1(4E)-C8:0-GalCerのようなガラクトシルセラミド(いずれも「E」はイタリック体)を基質とした時にはほとんど反応しないことから、グルコシルセラミドのグルコース構造を特異的に認識して反応することが明らかとなった。さらに、pNP-β-glucoside(「p」はイタリック体)にもほとんど反応しないことから、グルコシルセラミドのセラミド構造を特異的に認識して反応することも明らかとなった。これらの結果から、RGC1はグルコセレブロシダーゼであることが判明した。また、RGC1は、植物、動物、糸状菌由来いずれのタイプのグルコシルセラミド基質にも反応することも示された。
(実施例4)
動物由来グルコセレブロシダーゼは、グルコセレブロシダーゼ1(GBA1;イミグルセラーゼ、GH30に属する)、グルコセレブロシダーゼ2(GBA2、GH116に属する)、グルコセレブロシダーゼ3(GBA3、GH1に属する)、Lactase-phlorizin hydrolase(GH1に属する)の4種があることが知られている。また、植物由来グルコセレブロシダーゼは、シロイズナズナからGH116に属するグルコセレブロシダーゼ(AtGCD3)が単離されている。そこで、合成グルコシルセラミド基質であるN-[6-[(7-nitro-2-1,3-benzoxadiazol-4-yl)amino]hexanoyl]-D-glucosyl-β1-1´-sphingosine(C6-NBD glucosylceramide:「N」はイタリック体、「D」は小型英大文字)を使用して、実施例2で得られたRGC1およびイミグルセラーゼのKcat/Km値(1分子の酵素が1秒間に変換できる基質の分子数を酵素の最大反応速度の50%の反応速度を与える基質濃度で割った値:「K」はイタリック体)を実施例2の反応方法に従ってHanes-Woolf plotを用いて求めた。また、動物由来グルコセレブロシダーゼであるGBA3とシロイズナズナ由来グルコセレブロシダーゼであるAtGCD3のKcat/Km値は文献値(Journal of Biological Chemistry 282,(2007) 30889-30900、およびJournal of Biological Chemistry 295,(2020) 717-728)より引用した。この結果を下記表3に示す。この結果から、RGC1は他のグルコセレブロシダーゼに比べて明らかにKcat/Km値が高く、優れたグルコセレブロシダーゼであることが明らかとなった。

(実施例5)
実施例2で得られたRGC1およびイミグルセラーゼを、100μMグルコシルセラミド(セレブロシドB)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に添加し、各温度にて15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例2と同じように実施した。また、セラミド量および酵素活性の算出も実施例2と同じように実施した。そして、最もグルコセレブロシダーゼ活性の高い温度を至適温度とした。この試験は3回行い、その平均値を求めた。この結果を下記表4に示す。
この結果から、ヒト由来グルコセレブロシダーゼであるイミグルセラーゼの至適温度が42.5℃だったのに対して、イネ由来グルコセレブロシダーゼであるRGC1の至適温度は54.0℃と高く、RGC1は生体内で安定性が高いことが示唆された。
実施例2で得られたRGC1およびイミグルセラーゼを、100μMグルコシルセラミド(セレブロシドB)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.5)に添加し、各温度にて15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例2と同じように実施した。また、セラミド量および酵素活性の算出も実施例2と同じように実施した。そして、最もグルコセレブロシダーゼ活性の高い温度を至適温度とした。この試験は3回行い、その平均値を求めた。この結果を下記表4に示す。
この結果から、ヒト由来グルコセレブロシダーゼであるイミグルセラーゼの至適温度が42.5℃だったのに対して、イネ由来グルコセレブロシダーゼであるRGC1の至適温度は54.0℃と高く、RGC1は生体内で安定性が高いことが示唆された。
(実施例6)
酵素では、一般的に、至適温度が高い程、その熱安定性が優れていることが知られている。実施例5より、RGC1はイミグルセラーゼよりもグルコセレブロシダーゼ活性の至適温度が高いので、熱安定性を有することが期待された。そこで、ヒトの生体内でグルコセレブロシダーゼが主に存在および作用するリソソーム(pH5)と、細胞質(pH7)での熱安定性を確認するために以下の試験を実施した。
実施例2で得られたRGC1およびイミグルセラーゼについて、リソソームを想定したpH5(0.1%Triton X-100および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.0))では37℃で6~106時間、細胞質を想定したpH7(0.1%Triton X-100および0.05%コール酸ナトリウムを含む50mMリン酸カリウム緩衝液(pH7.0))では37℃で4~48時間インキュベーションした。各時間インキュベーションした後のグルコセレブロシダーゼ活性を実施例2の方法に従って測定した。残存活性はインキュベーション前の活性を100とした時の相対活性値として算出した。この試験は3回行い、その平均値を求めた。この結果を図1および図2に示す。なお、これらの図中において、各時間反応した後のRGC1のグルコセレブロシダーゼ活性がイミグルセラーゼのグルコセレブロシダーゼ活性よりも有意に高かった場合(1%有意)は*印を記載した。
図1、2より、pH5、pH7のいずれにおいても、明らかにRGC1はイミグルセラーゼよりも安定性が優れ、熱安定性を有することが示された。
酵素では、一般的に、至適温度が高い程、その熱安定性が優れていることが知られている。実施例5より、RGC1はイミグルセラーゼよりもグルコセレブロシダーゼ活性の至適温度が高いので、熱安定性を有することが期待された。そこで、ヒトの生体内でグルコセレブロシダーゼが主に存在および作用するリソソーム(pH5)と、細胞質(pH7)での熱安定性を確認するために以下の試験を実施した。

図1、2より、pH5、pH7のいずれにおいても、明らかにRGC1はイミグルセラーゼよりも安定性が優れ、熱安定性を有することが示された。
(実施例7)
実施例2のSDS-PAGEにより得られたRGC1バンドを切り出した後、トリプシン処理を行い、得られたペプチド断片の分子質量をMALDI-TOFMS(Matrix Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometer:Microflex LRF 20,Bruker Daltonics社製)により分析し、配列表の配列番号5~13および図7に示す9種のペプチド断片のアミノ酸配列を決定した。
これらRGC1の9種のペプチド断片の精密分子量は、機能が未同定のイネ(日本晴)由来のOs3BGlu6と名付けられたタンパク質由来の9種のペプチドの精密分子量と完全に一致したため、RGC1はOs3BGlu6と同一のタンパク質である可能性が示唆された。なお、現在までに、Os3BGlu6がグルコセレブロシダーゼであるという報告はない。

(実施例8)
イネ(日本晴)のカルスからRNeasy Plant Mini Kit(キアゲン社製、登録商標(以下同じ))を使って全RNAを抽出し、Absolutely mRNA Purification Kit(アジレント社製)を使って全RNAからmRNAを精製した。mRNAからのcDNA合成とその後のRACE分析は、SuperScript III Reverse Transcriptase(サーモフィッシャー社製、登録商標)を使って行った。Os3BGlu6のcDNAは、イネ(日本晴)の全ゲノム配列から推定されたオープンリーディングフレームより作製した、配列表の配列番号14、15および図8に示される2つのプライマー(F-プライマー(RGC1-CN)、およびR-プライマー(RGC1-CC))を使用したPCR増幅によって得た。
具体的なPCRの条件は、上記イネカルスcDNAおよび上記2種のプライマーにKOD-Plus-Neo(東洋紡社製)を加え、94℃2分間、60℃0.5分間、68℃1分間の反応条件を35回繰り返すことにより増幅した。そして、増幅された断片をプラスミドベクターpUC19にサブクローニングした。さらに、同様な方法により、増幅された断片よりも外側のDNA領域を含めたより長いDNA領域を増幅し、その断片の塩基配列を定法により解析することによって、Os3BGlu6遺伝子のcDNA全塩基配列を決定した。この塩基配列を配列表の配列番号3および図6に示す。また、この塩基配列から翻訳されたアミノ酸配列を配列表の配列番号1および図5に示す。このアミノ酸配列から、Os3BGlu6がGH1に属していることが明らかとなった。

(実施例9)
この単離されたOs3BGlu6遺伝子がグルコセレブロシダーゼ遺伝子(RGC1をコードする遺伝子)であることを明らかにするために、大腸菌で生産させたOs3BGlu6にグルコセレブロシダーゼ活性があるかを調べた。実施例8において得られたOs3BGlu6遺伝子をサブクローニングしたpUC19を用いて、これを導入することにより大腸菌(DH5α)を形質転換した。次に、この形質転換体を、LB液体培地(1.0%トリプトン、0.5%酵母エキス、1.0%塩化ナトリウム、50μg/mLアンピリシリン)において、37℃で24時間培養し、15000rpmの条件により10分間遠心分離して、集菌した。得られた菌体を、0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH6.0)で2回洗浄した。その後、15000rpmの条件により10分間遠心分離して集菌し、0.3%Triton X-100および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH6.0)に懸濁した後、超音波破砕した。この超音波破砕液を18000rpm、4℃の条件により60分遠心分離した。そして、遠心上清液をデュラポア メンブレンフィルター0.45μm HV(メルクミリポア社製)にてろ過した後、脱塩濃縮し、酵素抽出液を得た。この酵素抽出液を、100μMグルコシルセラミド(セレブロシドC)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸ナトリウム緩衝液(pH5.5)に添加し、37℃で13分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例1と同じように実施した。生成されたセラミドの分子量をネガティブLC-MS(アジレント社製)で調べることによって、セレブロシドCから生成されたセラミドであることを確認した(ESI-MS m/z:590.5[M-H]-)。また、酵素反応液中で1分間に1μmolのセラミドを生成する酵素量を1U(ユニット)として酵素抽出液1mLあたりのグルコセレブロシダーゼ活性を算出した。この結果を下記表5に示す。
この単離されたOs3BGlu6遺伝子がグルコセレブロシダーゼ遺伝子(RGC1をコードする遺伝子)であることを明らかにするために、大腸菌で生産させたOs3BGlu6にグルコセレブロシダーゼ活性があるかを調べた。実施例8において得られたOs3BGlu6遺伝子をサブクローニングしたpUC19を用いて、これを導入することにより大腸菌(DH5α)を形質転換した。次に、この形質転換体を、LB液体培地(1.0%トリプトン、0.5%酵母エキス、1.0%塩化ナトリウム、50μg/mLアンピリシリン)において、37℃で24時間培養し、15000rpmの条件により10分間遠心分離して、集菌した。得られた菌体を、0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH6.0)で2回洗浄した。その後、15000rpmの条件により10分間遠心分離して集菌し、0.3%Triton X-100および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH6.0)に懸濁した後、超音波破砕した。この超音波破砕液を18000rpm、4℃の条件により60分遠心分離した。そして、遠心上清液をデュラポア メンブレンフィルター0.45μm HV(メルクミリポア社製)にてろ過した後、脱塩濃縮し、酵素抽出液を得た。この酵素抽出液を、100μMグルコシルセラミド(セレブロシドC)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸ナトリウム緩衝液(pH5.5)に添加し、37℃で13分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例1と同じように実施した。生成されたセラミドの分子量をネガティブLC-MS(アジレント社製)で調べることによって、セレブロシドCから生成されたセラミドであることを確認した(ESI-MS m/z:590.5[M-H]-)。また、酵素反応液中で1分間に1μmolのセラミドを生成する酵素量を1U(ユニット)として酵素抽出液1mLあたりのグルコセレブロシダーゼ活性を算出した。この結果を下記表5に示す。
この結果から、Os3BGlu6遺伝子で形質転換された大腸菌から得られた酵素抽出液には明らかにグルコセレブロシダーゼ活性があることが示された。このことから、単離されたRGC1はOs3BGlu6であり、GH1に属するグルコセレブロシダーゼであることが明らかとなった。なお、現在までに、植物からGH1に属するグルコセレブロシダーゼが見出されたという報告はない。
(実施例10)
RGC1のアミノ酸配列と動物由来グルコセレブロシダーゼ、およびシロイズナズナ由来グルコセレブロシダーゼAtGCD3のアミノ酸配列との相同性(Identity)をEMBOSS Needle(https://www.ebi.ac.uk/Tools/psa/emboss_needle/)を用いることによって調べた結果、GBA1とは14.9%、GBA2とは10.1%、GBA3とは35.8%、Lactase-phlorizin hydrolaseとは11.3%、AtGCD3とは3.3%の相同性であった。この結果から、RGC1はアミノ酸配列の面からも、今までに単離されたグルコセレブロシダーゼとは全く異なったグルコセレブロシダーゼであることが明らかとなった。
RGC1のアミノ酸配列と動物由来グルコセレブロシダーゼ、およびシロイズナズナ由来グルコセレブロシダーゼAtGCD3のアミノ酸配列との相同性(Identity)をEMBOSS Needle(https://www.ebi.ac.uk/Tools/psa/emboss_needle/)を用いることによって調べた結果、GBA1とは14.9%、GBA2とは10.1%、GBA3とは35.8%、Lactase-phlorizin hydrolaseとは11.3%、AtGCD3とは3.3%の相同性であった。この結果から、RGC1はアミノ酸配列の面からも、今までに単離されたグルコセレブロシダーゼとは全く異なったグルコセレブロシダーゼであることが明らかとなった。
(実施例11)
ダイズ(Glycine max (L.) Merr. Enrei(「Glycine max」はイタリック体):えんれい)の茎102gを採取し、細かく裁断した後、0.01%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)中においてホモジナイザーにより破砕した。この破砕液を10000rpm、4℃の条件により60分間遠心分離し、遠心上清液を取り除いた。さらに、遠心沈殿物に0.01%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)を加え、5分攪拌後、10000rpm、4℃の条件により60分遠心分離し、遠心上清液を取り除いた。そして、得られた遠心沈殿物から、実施例2と同様の方法により酵素抽出液を得た。
ダイズ(Glycine max (L.) Merr. Enrei(「Glycine max」はイタリック体):えんれい)の茎102gを採取し、細かく裁断した後、0.01%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)中においてホモジナイザーにより破砕した。この破砕液を10000rpm、4℃の条件により60分間遠心分離し、遠心上清液を取り除いた。さらに、遠心沈殿物に0.01%コール酸ナトリウムを含む20mM酢酸緩衝液(pH5.5)を加え、5分攪拌後、10000rpm、4℃の条件により60分遠心分離し、遠心上清液を取り除いた。そして、得られた遠心沈殿物から、実施例2と同様の方法により酵素抽出液を得た。
この酵素抽出液を0.05%コール酸ナトリウムおよび0.05%Triton X-100を含む0.5mMリン酸緩衝液(pH6.7)で平衡化させたHiTrap Q HPカラム(アマシャムバイオサイエンス社製)にアプライした。そして、0.05%コール酸ナトリウムおよび0.05%Triton X-100を含む0.5mMリン酸緩衝液(pH6.7)から、0.05%コール酸ナトリウムおよび0.1%Triton X-100を含む20mMリン酸緩衝液(pH6.7)中に1M塩化ナトリウムを含む液に対して、グラジエント溶離法で溶出した。次に、グルコセレブロシダーゼ活性が強く認められた画分をプールし、限外濾過により脱塩濃縮した。この脱塩濃縮液を0.05%コール酸ナトリウムおよび0.05%Triton X-100を含む0.5mM酢酸緩衝液(pH6.0)で平衡化させたHiTrap SP HPカラム(アマシャムバイオサイエンス社製)にアプライした。そして、0.05%コール酸ナトリウムおよび0.05%Triton X-100を含む0.5mM酢酸緩衝液(pH6.0)から、0.05%コール酸ナトリウムおよび0.05%Triton X-100を含む0.5mM酢酸緩衝液(pH6.0)中に1M塩化ナトリウムを含む液に対して、グラジエント溶離法で溶出し、分画した。その結果、グルコセレブロシダーゼ活性が強く認められた画分をプールし、限外濾過により脱塩濃縮した酵素液をダイズ由来グルコセレブロシダーゼ(SGC1)とした。
なお、画分のグルコセレブロシダーゼ活性、およびSGC1のタンパク質濃度は、実施例2と同じ方法により分析した。このSGC1画分はSDS-PAGEにおいて単一なバンドを示し、その平均分子量(MW)は約62.0kDであった。SDS-PAGEも、実施例2と同様の方法により行った。
(実施例12)
実施例11で得られたSGC1およびイミグルセラーゼのグルコセレブロシダーゼ活性の至適温度について、実施例5と同じ方法により確認した。この結果を下記表6に示す。
この結果から、イミグルセラーゼの至適温度が42.5℃だったのに対して、SGC1の至適温度は64.0℃と非常に高いことから、SGC1は生体内で安定性が高い可能性が示唆された。
実施例11で得られたSGC1およびイミグルセラーゼのグルコセレブロシダーゼ活性の至適温度について、実施例5と同じ方法により確認した。この結果を下記表6に示す。
この結果から、イミグルセラーゼの至適温度が42.5℃だったのに対して、SGC1の至適温度は64.0℃と非常に高いことから、SGC1は生体内で安定性が高い可能性が示唆された。
(実施例13)
前述したように、酵素では、至適温度が高い程、その熱安定性が優れていることが知られている。実施例12より、SGC1はイミグルセラーゼよりもグルコセレブロシダーゼ活性の至適温度が高いので、熱安定性を有することが期待された。そこで、リソソーム(pH5)と細胞質(pH7)での熱安定性を確認するために以下の試験を実施した。
実施例11で得られたSGC1とイミグルセラーゼの2つのグルコセレブロシダーゼについて、リソソームを想定したpH5(0.1%Triton X-100および0.05%コール酸ナトリウムを含む50mM酢酸緩衝液(pH5.0))では45℃で1~31時間、細胞質を想定したpH7(0.1%Triton X-100および0.05%コール酸ナトリウムを含む50mMリン酸カリウム緩衝液(pH7.0))では45℃で0.3~5時間インキュベーションした。各時間反応した後のグルコセレブロシダーゼ活性を実施例2の方法に従って測定した。残存活性はインキュベーション前の活性を100とした時の相対活性値として算出した。この試験は3回行い、その平均値を求めた。この結果を図3、4に示す。なお、これらの図中において、各時間反応した後のSGC1のグルコセレブロシダーゼ活性がイミグルセラーゼのグルコセレブロシダーゼ活性よりも有意に高かった場合(1%有意)は*印を記載した。
図3、4より、pH5、pH7のいずれにおいても、明らかにSGC1はイミグルセラーゼよりもはるかに安定性が優れ、熱安定性を有することが示された。
前述したように、酵素では、至適温度が高い程、その熱安定性が優れていることが知られている。実施例12より、SGC1はイミグルセラーゼよりもグルコセレブロシダーゼ活性の至適温度が高いので、熱安定性を有することが期待された。そこで、リソソーム(pH5)と細胞質(pH7)での熱安定性を確認するために以下の試験を実施した。

図3、4より、pH5、pH7のいずれにおいても、明らかにSGC1はイミグルセラーゼよりもはるかに安定性が優れ、熱安定性を有することが示された。
(実施例14)
実施例11のSDS-PAGEにより得られたSGC1バンドを切り出した後、トリプシン処理を行い、得られたペプチド断片の分子質量をMALDI-TOFMS(Matrix Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometer:Microflex LRF 20,Bruker Daltonics社製)により分析し、配列表の配列番号16~22および図11に示す7種のペプチド断片のアミノ酸配列を決定した。
これらSGC1由来の7種のペプチド断片の精密分子量は、機能が未同定の米国品種ダイズ(Glycine max (L.) Merr. Williams 82(「Glycine max」はイタリック体))由来のβ-グルコシダーゼ40と名付けられたタンパク質由来の7種のペプチドの精密分子量と完全に一致したため、SGC1はβ-グルコシダーゼ40と同一のタンパク質である可能性が示唆された。なお、このβ-グルコシダーゼ40はゲノム配列との相同性検索から簡易的に命名されたものであり、β-グルコシダーゼとしての酵素活性は確認されておらず、ましてやグルコセレブロシダーゼであるという報告は現在までに全くない。
実施例11のSDS-PAGEにより得られたSGC1バンドを切り出した後、トリプシン処理を行い、得られたペプチド断片の分子質量をMALDI-TOFMS(Matrix Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometer:Microflex LRF 20,Bruker Daltonics社製)により分析し、配列表の配列番号16~22および図11に示す7種のペプチド断片のアミノ酸配列を決定した。
これらSGC1由来の7種のペプチド断片の精密分子量は、機能が未同定の米国品種ダイズ(Glycine max (L.) Merr. Williams 82(「Glycine max」はイタリック体))由来のβ-グルコシダーゼ40と名付けられたタンパク質由来の7種のペプチドの精密分子量と完全に一致したため、SGC1はβ-グルコシダーゼ40と同一のタンパク質である可能性が示唆された。なお、このβ-グルコシダーゼ40はゲノム配列との相同性検索から簡易的に命名されたものであり、β-グルコシダーゼとしての酵素活性は確認されておらず、ましてやグルコセレブロシダーゼであるという報告は現在までに全くない。
(実施例15)
ダイズ(えんれい)の葉からRNeasy Plant Mini Kit(キアゲン社製)を使って全RNAを抽出し、この全RNAからPrimeScript II 1st strand cDNA Synthesis Kit(宝社製)を使ってcDNAを合成した。ダイズ(えんれい)由来β-グルコシダーゼ40のcDNAは、米国品種ダイズ(Williams 82)の全ゲノム配列から推定されたオープンリーディングフレームより作製した、配列表の配列番号23、24および図12に示される2つのプライマー(F-プライマー(SGC1-CN)、およびR-プライマー(SGC1-CC))を使用したPCR増幅によって得た。
具体的なPCRの条件は、上記ダイズ(えんれい)の葉からのcDNAと上記2種のプライマーにKOD-Plus-Neo(東洋紡社製)を加え、94℃2分間、60℃0.5分間、68℃1分間の反応条件を40回繰り返すことにより増幅した。そして、増幅された断片をプラスミドベクターpUC19にサブクローニングした。さらに、同様な方法で、増幅された断片よりも外側のDNA領域を含めたより長いDNA領域を増幅し 、その断片の塩基配列を定法により解析することによって、ダイズ(えんれい)由来β-グルコシダーゼ40遺伝子のcDNA全塩基配列を決定した。この塩基配列を配列表の配列番号4および図10に示す。また、この塩基配列から翻訳されたアミノ酸配列を配列表の配列番号2および図9に示す。このアミノ酸配列から、ダイズ(えんれい)由来β-グルコシダーゼ40がGH1に属していることが明らかとなった。
ダイズ(えんれい)の葉からRNeasy Plant Mini Kit(キアゲン社製)を使って全RNAを抽出し、この全RNAからPrimeScript II 1st strand cDNA Synthesis Kit(宝社製)を使ってcDNAを合成した。ダイズ(えんれい)由来β-グルコシダーゼ40のcDNAは、米国品種ダイズ(Williams 82)の全ゲノム配列から推定されたオープンリーディングフレームより作製した、配列表の配列番号23、24および図12に示される2つのプライマー(F-プライマー(SGC1-CN)、およびR-プライマー(SGC1-CC))を使用したPCR増幅によって得た。
具体的なPCRの条件は、上記ダイズ(えんれい)の葉からのcDNAと上記2種のプライマーにKOD-Plus-Neo(東洋紡社製)を加え、94℃2分間、60℃0.5分間、68℃1分間の反応条件を40回繰り返すことにより増幅した。そして、増幅された断片をプラスミドベクターpUC19にサブクローニングした。さらに、同様な方法で、増幅された断片よりも外側のDNA領域を含めたより長いDNA領域を増幅し 、その断片の塩基配列を定法により解析することによって、ダイズ(えんれい)由来β-グルコシダーゼ40遺伝子のcDNA全塩基配列を決定した。この塩基配列を配列表の配列番号4および図10に示す。また、この塩基配列から翻訳されたアミノ酸配列を配列表の配列番号2および図9に示す。このアミノ酸配列から、ダイズ(えんれい)由来β-グルコシダーゼ40がGH1に属していることが明らかとなった。
(実施例16)
ダイズ(えんれい)由来β-グルコシダーゼ40遺伝子がグルコセレブロシダーゼ遺伝子であることを明らかにするために、大腸菌で生産させたダイズ由来β-グルコシダーゼ40遺伝子にグルコセレブロシダーゼ活性があるかを調べた。実施例15において得られたダイズ由来β-グルコシダーゼ40遺伝子をサブクローニングしたpUC19を用いて、これを導入することにより大腸菌(DH5α)を形質転換した。この形質転換体を用いて、酵素反応を60℃で30分間インキュベーションしたこと以外は全て実施例9と同じ方法により酵素抽出液1mLあたりのグルコセレブロシダーゼ活性を算出した。結果を下記表7に示す。
ダイズ(えんれい)由来β-グルコシダーゼ40遺伝子がグルコセレブロシダーゼ遺伝子であることを明らかにするために、大腸菌で生産させたダイズ由来β-グルコシダーゼ40遺伝子にグルコセレブロシダーゼ活性があるかを調べた。実施例15において得られたダイズ由来β-グルコシダーゼ40遺伝子をサブクローニングしたpUC19を用いて、これを導入することにより大腸菌(DH5α)を形質転換した。この形質転換体を用いて、酵素反応を60℃で30分間インキュベーションしたこと以外は全て実施例9と同じ方法により酵素抽出液1mLあたりのグルコセレブロシダーゼ活性を算出した。結果を下記表7に示す。
この結果から、ダイズ(えんれい)由来β-グルコシダーゼ40遺伝子で形質転換された大腸菌から得られた酵素抽出液には明らかにグルコセレブロシダーゼ活性があることが示された。このことから、単離されたダイズ由来SGC1はダイズ由来β-グルコシダーゼ40であり、GH1に属するグルコセレブロシダーゼであることが明らかとなった。なお、前述したように、現在までに、植物からGH1に属するグルコセレブロシダーゼが見出されたという報告はない。
(実施例17)
SGC1のアミノ酸配列と動物由来グルコセレブロシダーゼ、およびシロイズナズナ由来グルコセレブロシダーゼAtGCD3のアミノ酸配列との相同性(Identity)を上記したEMBOSS Needleを用いることによって調べた結果、GBA1とは14.9%、GBA2とは10.0%、GBA3とは35.9%、Lactase-phlorizin hydrolaseとは 10.4%、AtGCD3とは7.6%の相同性であった。この結果から、SGC1はアミノ酸配列の面からも、今までに単離されたグルコセレブロシダーゼとは全く異なったグルコセレブロシダーゼであることが明らかとなった。
SGC1のアミノ酸配列と動物由来グルコセレブロシダーゼ、およびシロイズナズナ由来グルコセレブロシダーゼAtGCD3のアミノ酸配列との相同性(Identity)を上記したEMBOSS Needleを用いることによって調べた結果、GBA1とは14.9%、GBA2とは10.0%、GBA3とは35.9%、Lactase-phlorizin hydrolaseとは 10.4%、AtGCD3とは7.6%の相同性であった。この結果から、SGC1はアミノ酸配列の面からも、今までに単離されたグルコセレブロシダーゼとは全く異なったグルコセレブロシダーゼであることが明らかとなった。
(実施例18)
BLASTp search (https://blast.ncbi.nlm.nih.gov/Blast.cgi)を用いることによって、RGC1のアミノ酸配列と相同性の高いタンパク質を探索したところ、種子植物に幅広く、相同性の高いタンパク質があることが明らかとなった。なお、相同性(Identity)は、上記したEMBOSS Needleを用いることによって調べた。この結果を下記表8に示す。
BLASTp search (https://blast.ncbi.nlm.nih.gov/Blast.cgi)を用いることによって、RGC1のアミノ酸配列と相同性の高いタンパク質を探索したところ、種子植物に幅広く、相同性の高いタンパク質があることが明らかとなった。なお、相同性(Identity)は、上記したEMBOSS Needleを用いることによって調べた。この結果を下記表8に示す。
表8より、RGC1のアミノ酸配列は、単子葉植物由来のβ-グルコシダーゼ6およびβ-グルコシダーゼ34との間において73.8~84.9%の相同性を示し、双子葉植物由来のβ-グルコシダーゼ6、β-グルコシダーゼ40およびβ-グルコシダーゼ34との間において62.6~70.0%の相同性を示した。また、シダ類やコケ類のβ-グルコシダーゼ6やβ-グルコシダーゼ40との間において46.6~53.2%の相同性を示した。さらに、これらのアミノ酸配列はすべて、GH1に必要な保存領域を有し、GH1に属していた。この結果より、上記のβ-グルコシダーゼ6、β-グルコシダーゼ40およびβ-グルコシダーゼ34はグルコセレブロシダーゼであると考えられた。
(実施例19)
イネ以外の植物にも同じようにグルコセレブロシダーゼ活性があるかを調べるため、以下の試験を実施した。各種植物の葉、根、茎、花びら、めしべ、花粉、実、仮根、胞子嚢の各部位を0.01~1g採取し、0.05%コール酸ナトリウムおよび0.3%Triton X-100を含む50mM酢酸緩衝液(pH5.0)中においてホモジナイザーで破砕した。この破砕液を15000rpm、4℃の条件により20分間遠心分離し、遠心上清液を酵素抽出液とした。
そして、この酵素抽出液を、100μMグルコシルセラミド(セレブロシドB)、0.05%コール酸ナトリウムおよび0.2%Triton X-100を含む50mM酢酸緩衝液(pH5.0)において、45℃で60分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例2と同じように実施した。また、セラミド量および酵素活性の算出も実施例2と同じように実施し、植物生重量1gあたりのグルコセレブロシダーゼ活性のユニット数を算出した。この試験は3回行い、その平均値を求めた。この結果を下記表9に示す。
イネ以外の植物にも同じようにグルコセレブロシダーゼ活性があるかを調べるため、以下の試験を実施した。各種植物の葉、根、茎、花びら、めしべ、花粉、実、仮根、胞子嚢の各部位を0.01~1g採取し、0.05%コール酸ナトリウムおよび0.3%Triton X-100を含む50mM酢酸緩衝液(pH5.0)中においてホモジナイザーで破砕した。この破砕液を15000rpm、4℃の条件により20分間遠心分離し、遠心上清液を酵素抽出液とした。
そして、この酵素抽出液を、100μMグルコシルセラミド(セレブロシドB)、0.05%コール酸ナトリウムおよび0.2%Triton X-100を含む50mM酢酸緩衝液(pH5.0)において、45℃で60分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmの条件により20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例2と同じように実施した。また、セラミド量および酵素活性の算出も実施例2と同じように実施し、植物生重量1gあたりのグルコセレブロシダーゼ活性のユニット数を算出した。この試験は3回行い、その平均値を求めた。この結果を下記表9に示す。
この結果から、測定した全ての植物の抽出物においてグルコセレブロシダーゼ活性が認められた。表8および表9の結果を考慮すると、イネやダイズばかりでなく、植物全般にグルコセレブロシダーゼが存在することが示された。
そして、単子葉植物由来のRGC1、および双子葉植物由来のSGC1がGH1に属するグルコセレブロシダーゼであること(実施例9および16)、RGC1のアミノ酸配列と相同性の高いGH1に属する酵素が多くの種子植物に存在すること(実施例18)、ならびに、上記した表9に示すように、多くの種子植物の抽出物から高いグルコセレブロシダーゼ活性を検出したことから、種子植物はGH1に属するグルコセレブロシダーゼを有することが示された。さらに、シダ類やコケ類にもβ-グルコシダーゼ6やβ-グルコシダーゼ40が存在することや(前述の表8)、シダ類やコケ類の抽出物からもグルコセレブロシダーゼ活性を検出したことから、シダ類やコケ類もGH1に属するグルコセレブロシダーゼを有することが十分に示唆される。
次に相同性の面から考察すると、実施例9および16においてGH1に属するグルコセレブロシダーゼであることが証明されている単子葉植物由来のRGC1と双子葉植物由来のSGC1のアミノ酸配列の相同性が67.6%であることから、少なくともRGC1とSGC1のアミノ酸配列と67.5%以上の相同性を有する酵素は確実にGH1に属するグルコセレブロシダーゼであると考えられる。さらに、上記した表9から、種子植物はグルコセレブロシダーゼを有することが判明したため、少なくとも、前述の表8で示される、種子植物でRGC1のアミノ酸配列と最も相同性の低い62.6%(コウシンバラ)以上の相同性を有する酵素もGH1に属するグルコセレブロシダーゼであると考えられる。
そして、単子葉植物由来のRGC1、および双子葉植物由来のSGC1がGH1に属するグルコセレブロシダーゼであること(実施例9および16)、RGC1のアミノ酸配列と相同性の高いGH1に属する酵素が多くの種子植物に存在すること(実施例18)、ならびに、上記した表9に示すように、多くの種子植物の抽出物から高いグルコセレブロシダーゼ活性を検出したことから、種子植物はGH1に属するグルコセレブロシダーゼを有することが示された。さらに、シダ類やコケ類にもβ-グルコシダーゼ6やβ-グルコシダーゼ40が存在することや(前述の表8)、シダ類やコケ類の抽出物からもグルコセレブロシダーゼ活性を検出したことから、シダ類やコケ類もGH1に属するグルコセレブロシダーゼを有することが十分に示唆される。
次に相同性の面から考察すると、実施例9および16においてGH1に属するグルコセレブロシダーゼであることが証明されている単子葉植物由来のRGC1と双子葉植物由来のSGC1のアミノ酸配列の相同性が67.6%であることから、少なくともRGC1とSGC1のアミノ酸配列と67.5%以上の相同性を有する酵素は確実にGH1に属するグルコセレブロシダーゼであると考えられる。さらに、上記した表9から、種子植物はグルコセレブロシダーゼを有することが判明したため、少なくとも、前述の表8で示される、種子植物でRGC1のアミノ酸配列と最も相同性の低い62.6%(コウシンバラ)以上の相同性を有する酵素もGH1に属するグルコセレブロシダーゼであると考えられる。
(実施例20)
実施例19に従って抽出した各種植物由来の酵素抽出液、ならびに前述のイネ由来精製RGC1および前述のダイズ由来精製SGC1を、100μMグルコシルセラミド(セレブロシドB)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸ナトリウム緩衝液(pH5.5)に添加し、各温度にて15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmで20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例5と同じように実施した。また、セラミド量および酵素活性の算出も実施例5と同じように実施した。その結果として、最もグルコセレブロシダーゼ活性の高い温度を至適温度とした。この試験は3回行い、その平均値を求めた。この結果を下記表10に示す。
実施例19に従って抽出した各種植物由来の酵素抽出液、ならびに前述のイネ由来精製RGC1および前述のダイズ由来精製SGC1を、100μMグルコシルセラミド(セレブロシドB)、0.1%Tween20および0.05%コール酸ナトリウムを含む50mM酢酸ナトリウム緩衝液(pH5.5)に添加し、各温度にて15分間インキュベーションした。次いで、得られた反応液に4倍量のエタノールを加え混合後、15000rpmで20分間遠心分離し、上清液を高速液体クロマトグラフィー分析に供し、酵素反応により生成したセラミド量を測定した。高速液体クロマトグラフィー分析は、実施例5と同じように実施した。また、セラミド量および酵素活性の算出も実施例5と同じように実施した。その結果として、最もグルコセレブロシダーゼ活性の高い温度を至適温度とした。この試験は3回行い、その平均値を求めた。この結果を下記表10に示す。
この結果から、イネおよびダイズを含む多くの種子植物由来グルコセレブロシダーゼにおいてもヒト由来グルコセレブロシダーゼであるイミグラセラーゼよりも至適温度が高いことが見出された。実施例6および13で示されるように、酵素では、一般的に、至適温度が高い程、その熱安定性が優れていることが知られていることから、種子植物由来のGH1に属するグルコセレブロシダーゼはヒト由来グルコセレブロシダーゼであるイミグラセラーゼよりも熱に対する安定性が優れ、つまり熱安定性を有していることを示していると言える。
この出願は、2020年5月21日に出願された日本出願特願2020-88950を基礎とする優先権を主張し、その開示のすべてをここに取り込む。
Claims (15)
- 植物由来であり、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性を有する、タンパク質。
- 前記植物が種子植物である、請求項1に記載のタンパク質。
- 前記種子植物が、アブラナ科植物、イネ科植物、ウリ科植物、キク科植物、ナス科植物、バラ科植物、ヒガンバナ科植物、マメ科植物、またはユリ科植物のいずれかである、請求項2に記載のタンパク質。
- 以下の(A)、(B)、または(C)に示されるタンパク質。
(A)配列表の配列番号1、配列番号1のうちアミノ酸番号38~521、配列番号2、または配列番号2のうちアミノ酸番号19~503に示されるアミノ酸配列からなる、タンパク質。
(B)配列表の配列番号1、配列番号1のうちアミノ酸番号38~521、配列番号2、または配列番号2のうちアミノ酸番号19~503に示されるアミノ酸配列において、1もしくは数個のアミノ酸の置換、欠失、挿入、または付加がされたアミノ酸配列からなり、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質。
(C)配列表の配列番号1、配列番号1のうちアミノ酸番号38~521、配列番号2、または配列番号2のうちアミノ酸番号19~503に示されるアミノ酸配列に対して60%以上の相同性を有し、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質。 - 請求項1~4のいずれか1項に記載のタンパク質をコードするDNA。
- 以下の(a)、(b)、または(c)に示されるDNA。
(a)配列表の配列番号3、配列番号3のうち塩基番号112~1566、配列番号4、または配列番号4のうち塩基番号55~1512に示される塩基配列からなる、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性を有するタンパク質をコードするDNA。
(b)配列表の配列番号3、配列番号3のうち塩基番号112~1566、配列番号4、または配列番号4のうち塩基番号55~1512に示される塩基配列において、1もしくは数個の塩基の置換、欠失、挿入、または付加がされた塩基配列からなり、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質をコードするDNA。
(c)配列表の配列番号3、配列番号3のうち塩基番号112~1566、配列番号4、または配列番号4のうち塩基番号55~1512に示される塩基配列と相補的な塩基配列からなるDNAとストリンジェントな条件下でハイブリダイゼーションすることができ、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質をコードするDNA。 - 請求項5または6に記載のDNAを含む、グリコシドハイドロラーゼファミリー1に属し、且つグルコセレブロシダーゼ活性および熱安定性を有するタンパク質発現用の発現ベクター。
- 請求項7に記載の発現ベクターが導入された、形質転換体。
- 前記形質転換体が、植物体、植物細胞、動物細胞、大腸菌、酵母、または糸状菌のいずれかである、請求項8に記載の形質転換体。
- 請求項8または9に記載の形質転換体を育種または培養する工程、および、この工程によって得られる前記形質転換体または前記形質転換体含有物から請求項1~4のいずれか1項に記載のタンパク質を回収する工程を含む、タンパク質の製造方法。
- 請求項1~4のいずれか1項に記載のタンパク質を含有する、グルコシルセラミド加水分解用酵素組成物。
- 請求項1~4のいずれか1項に記載のタンパク質を有効成分として含む、医薬組成物。
- ゴーシェ病予防および治療用である、請求項12に記載の医薬組成物。
- 請求項1~4のいずれか1項に記載のタンパク質と、植物、動物、または微生物より単離されたグルコシルセラミド、もしくは化学合成されたグルコシルセラミドと、を含有する、食品組成物。
- 請求項1~4のいずれか1項に記載のタンパク質を用いて、植物、動物、または微生物より単離されたグルコシルセラミド、もしくは化学合成されたグルコシルセラミドから、セラミドを生成させる工程を含む、セラミドの製造方法。
