Abstract
본 발명은 매실 추출물 또는 이의 용매 분획물을 유효성분으로 함유하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물로서, 본 발명에 따른 매실 추출물 또는 이의 분획물은 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물로 이용될 수 있다.

본 특허출원은 2020년 2월 26일에 대한민국 특허청에 제출된 대한민국 특허출원 제 10-2020-0023863호에 대하여 우선권을 주장하며, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.
본 발명은 매실 추출물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물에 관한 것이다.
인플루엔자 바이러스 감염은 상기도 점막에 침입하여 기침, 콧물, 가래와 같은 감기 증상과 더불어 발열, 오한, 근육통, 두통, 식욕 부진 등의 심각한 증상을 동반하여 독감이라고 부른다. 인플루엔자 바이러스는 면역력이 약한 경우, 합병증을 일으키고, 다른 호흡기 바이러스에 비해 전염성이 매우 강하여 전 세계적으로 사람뿐만 아니라 동물의 건강을 위협하고 있다.
인플루엔자 바이러스는 RNA를 핵산으로 각기 기능이 다른 8개의 분절을 암호화하는 A, B형과 7개의 분절을 암호화하는 C형으로 분류된다. 이중 A, B형이 주로 사람이 감염되는 주된 인플루엔자 바이러스로 헤마글루타닌 (Hemagglutinin; HA)과 뉴라미니다아제 (Neuraminidase; NA)라는 바이러스 표면 당단백질인 항원의 종류에 따라서 구분되고, 인플루엔자 바이러스의 명명은 형, 자연숙주, 아형, 분리지, 분리번호, 분리년도(HA와 NA 항원형)의 순서로 기재한다.
A형 인플루엔자 바이러스 유전체는 약 13,500개의 RNA 염기로 이루어져 있으며, 돌연변이나 유전체 분절교환에 의해 새로운 헤마글루타닌과 뉴라미니다아제, 즉, 새로운 항원을 갖는 변이주가 출연하기 쉬워 세계적으로 대유행을 일으킨다. 반면, B형 인플루엔자 바이러스는 항원성의 큰 변이가 일어날 가능성이 적고, 2가지 계통만 알려져 있어 A형에 비해 대유행의 가능성이 훨씬 적다. A형 인플루엔자 바이러스는 사람의 경우 6종의 HA와 3종의 NA로 구성되어 있고 (H1N1, H2N2, H3N2, H5N1, H7N7, H3N7), 조류의 경우 16종의 HA와 9종의 NA가 있어 144종의 서로 다른 혈청형이 존재한다.
일본에서는 스즈키 교수가 매실을 농축하는 과정에서 생성되는 무메푸랄(mumefual)이라는 물질이 A형 인플루엔자 바이러스에 대하여 활성이 있다고 보고한 바 있다. 하지만, 국내에서는 매실 및 매실 가공품의 인플루엔자 바이러스 활성에 대한 연구가 전무한 실정이다.
본 발명의 목적은 매실 추출물 또는 이의 분획물을 유효성분으로 함유하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 매실 추출물 또는 이의 분획물을 유효성분으로 함유하는 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물을 제공하는 것이다.
본 발명은 매실 추출물 또는 이의 분획물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물에 관한 것으로, 본 발명에 따른 매실 추출물은 매실의 용매 조출물 및/또는 이의 용매 분획물을 포함하며, 인플루엔자 바이러스 감염 예방, 치료 또는 개선 효과를 나타낸다.

본 발명자들은 매실즙, 매실청, 매실농축액 또는 매실 과실 추출물을 가축에 공급한 지역에서 조류 인플루엔자 바이러스 감염증이 발생되지 않은 사실을 발견하였다. 따라서 감염동물모델을 이용하여 매실즙, 매실청, 매실농축액 또는 매실 과실 추출물의 항인플루엔자 바이러스 효능을 시험하고, 항인플루엔자 바이러스 활성이 보고된 무메푸릴 외 새로운 활성 물질을 확인하고, 분리하여 매실 가공품인 매실청, 매실즙, 그리고 매실 과실 추출물에 활성 물질이 존재하는 것을 확인하였다.
이하 본 발명에 대해서 상세히 설명한다.
본 발명의 일 예는 매실(Prunus mume) 추출물 또는 이의 분획물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물에 관한 것이다.
본 발명에 있어서 매실 추출물은 매실즙, 매실청, 매실농축액 및 매실 과실 추출물로 이루어진 군으로부터 선택되는 1종 이상인 것일 수 있고, 예를 들어, 매실즙인 것일 수 있으나, 이에 한정되는 것은 아니다.
상기 매실 추출물은 물, 및 탄소수 1 내지 4개의 직쇄 또는 분지형 알코올, 아세트산, 부탄올 또는 이의 조합으로 이루어진 군에서 선택된 1종 이상의 용매로 추출하여 얻어진 조추출물일 수 있다.
본 발명에 있어서 매실 분획물은 매실 추출물에 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 사용한 용매 분획물일 수 있고, 예를 들어, 에틸아세테이트 분획물인 것일 수 있으나, 이에 한정되는 것은 아니다.
상기 매실 분획물은 상기 에틸아세테이트 분획물을 크로마토그래피를 수행하여 얻은 것인 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 매실 추출물은 추가의 용매로 분획한 용매 분획물일 수 있으나, 예를 들면, 하기 매실 추출물에 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 사용한 용매 분획물인 것 일 수 있다;
매실즙, 매실청, 매실농축액 또는 매실 과실 추출물에 에틸에테르를 첨가하여 얻은 에틸에테르 분획물;
매실즙, 매실청, 매실농축액 또는 매실 과실 추출물에 에틸아세테이트를 첨가하여 얻은 에틸아세테이트 분획물; 및
매실즙, 매실청, 매실농축액 또는 매실 과실 추출물에 부탄올을 첨가하여 얻은 부탄올 분획물.
본 발명에 있어서 인플루엔자 바이러스는 인플루엔자 바이러스 A형 또는 B형을 포함하는 것 일 수 있다.
상기 인플루엔자 바이러스 A형은 H1N1, H3N2, H1N2, H2N2, H2N3, H3N1, H3N8, H5N1, H5N2, H5N3, H5N8, H5N9, H7N1, H7N2, H7N3, H7N4, H7N7, H7N9, H9N2 또는 H10N7의 혈청형을 갖는 것 일 수 있다.
본 발명에 있어서 약학적 조성물은 매실 추출물 이외에 약학적으로 허용 가능한 담체, 부형제, 또는 희석제를 포함하는 것 일 수 있다.
상기 약학적 조성물의 투여 형태는 이들의 약학적으로 허용 가능한 염의 형태로 사용될 수 있고, 또한 단독으로 또는 타약적 활성 화합물과 결합뿐만 아니라 적당한 결합으로 사용될 수 있다. 상기 염으로는 약학적으로 허용되는 것이면 특별히 한정되지 않으며, 예를 들어, 염산, 황산, 질산, 인산, 불화수소산, 브롬화수소산, 포름산아세트산, 타르타르산, 젖산, 시트르산, 푸마르산, 말레산, 숙신산, 메탄술폰산, 벤젠술폰산, 톨루엔술폰산, 나프탈렌술폰산 등을 사용하는 것 일 수 있다.
상기 약학적 조성물은 경구 또는 비경구의 여러 가지 제형일 수 있다. 제제화 할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제; 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제할 수 있다. 또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들을 사용하는 것 일 수 있다.
상기 약학적 조성물의 경구투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함되는 것 일 수 있다.
상기 약학적 조성물의 비수용제 및 현탁용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween)61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용되는 것 일 수 있다.
상기 약학적 조성물의 바람직한 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다. 그러나, 바람직한 효과를 위해서 본 발명의 약학적 조성물은 0.001 내지 100 mg/kg의 양을 일일 1회 내지 수회로 나누어 투여되는 것 일 수 있다.
상기 약학적 조성물의 투여 형태는 이들의 약학적으로 허용가능한 염의 형태로도 사용될 수 있고, 또한, 단독으로 또는 기타 약학적 활성 화합물과 결합뿐만 아니라 적당한 집합으로 사용되는 것 일 수 있다.
상기 약학적 조성물은 랫트, 마우스, 가축, 인간 등의 포유동물에 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 복강내, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관내 주사에 의해 투여되는 것 일 수 있다.
본 발명의 일 예는 매실 추출물을 또는 이의 분획물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물에 관한 것이다.
상기 매실 추출물은 매실즙, 매실청, 매실농축액 및 매실 과실 추출물로 이루어진 군으로부터 선택되는 1종 이상인 것일 수 있고, 예를 들어, 매실즙인 것일 수 있으나, 이에 한정되는 것은 아니다.
상기 매실 추출물은 물, 및 탄소수 1 내지 4개의 직쇄 또는 분지형 알코올, 아세트산, 부탄올 또는 이의 조합으로 이루어진 군에서 선택된 1종 이상의 용매로 추출하여 얻어진 조추출물인 것 일 수 있다.
본 발명에 있어서 매실 분획물은 매실 추출물에 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 사용한 용매 분획물일 수 있고, 예를 들어, 에틸아세테이트 분획물인 것일 수 있으나, 이에 한정되는 것은 아니다.
상기 매실 분획물은 상기 에틸아세테이트 분획물을 크로마토그래피를 수행하여 얻은 것인 것일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서 매실 추출물은 추가의 용매로 분획한 용매 분획물일 수 있으나, 예를 들면, 하기 매실 추출물에 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 사용한 용매 분획물인 것 일 수 있다;
매실즙, 매실청, 매실농축액 또는 매실 과실 추출물에 에틸에테르를 첨가하여 얻은 에틸에테르 분획물;
매실즙, 매실청, 매실농축액 또는 매실 과실 추출물에 에틸아세테이트를 첨가하여 얻은 에틸아세테이트 분획물; 및
매실즙, 매실청, 매실농축액 또는 매실 과실 추출물에 부탄올을 첨가하여 얻은 부탄올 분획물.
본 발명에 있어서 인플루엔자 바이러스는 인플루엔자 바이러스 A형 또는 B형을 포함하는 것일 수 있다.
상기 인플루엔자 바이러스 A형은 H1N1, H3N2, H1N2, H2N2, H2N3, H3N1, H3N8, H5N1, H5N2, H5N3, H5N8, H5N9, H7N1, H7N2, H7N3, H7N4, H7N7, H7N9, H9N2 또는 H10N7의 혈청형을 갖는 것 일 수 있다.
본 발명에 있어서 식품 조성물은 매실 추출물을 단독으로, 다른 식품과 함께, 또는 다른 식품 성분과 함께 사용할 수 있고, 통상적인 방법에 따라 적절하게 사용되는 것 일 수 있다.
상기 식품 조성물은 식품 제조 시에 통상적으로 첨가되는 성분을 포함하며, 예를 들어, 단백질, 탄수화물, 지방, 영양소 및 조미제를 포함한다. 또한, 식품의 종류에는 특별한 제한이 없으나, 육류, 소시지, 빵, 초콜릿, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올음료, 비타민 복합제 등이 있으나, 통상적인 의미에서의 건강식품을 포함하는 것 일 수 있다.
상기 식품 조성물은 통상의 음료와 같이 여러가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용하는 것 일 수 있다.
상기 식품 조성물은 매실 추출물을 식품 소재에 첨가하거나, 캡슐화, 분말화, 현탁액 등으로 제조한 식품으로, 이를 섭취할 경우 건강상 특정한 효과를 가져오는 것을 의미하나, 일반 약품과 달리 식품을 원료로 하여 약품의 장기 복용시 발생할 수 있는 부작용 등이 없는 장점이 있는 것 일 수 있다.
본 발명의 일 예는 다음 단계를 포함하는 항바이러스 활성 물질의 추출방법이다:
매실 추출물에 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 사용하여 제1 분획물을 제조하는 제1 분획 단계;
제1 분획물에 컬럼크로마토그래피를 수행하여 제2 분획물을 얻는 제2 분획단계;
제2 분획물에 컬럼크로마토그래피를 수행하여 제3 분획물을 얻는 제3 분획단계; 및
제3 분획물에 막크로마토그래피를 수행하여 제4 분획물을 얻는 제4 분획단계.
상기 매실 추출물은 매실즙, 매실청, 매실농축액 및 매실 과실 추출물로 이루어진 군으로부터 선택되는 1종 이상인 것일 수 있고, 예를 들어, 매실즙인 것일 수 있으나, 이에 한정되는 것은 아니다.
상기 제1 분획 단계는 에틸아세테이트를 사용하여 수행되는 것이다.
상기 제2 분획 단계 및 제3 분획 단계는 정지상과 이동상의 물리적 방법에 바탕을 둔 컬럼크로마토그래피를 이용하여 수행되는 것이다.
상기 제2 분획 단계는 제1 분획 단계에서 용출된 분획물을 컬럼크로마토그래피를 이용하여 수행되는 것이다.
구체적으로, 상기 제2 분획 단계는 실리카겔 컬럼크로마토 그래피 (Flash Silica gel columm chromatograph)를 이용하여 수행되는 것이다.
상기 실리카겔 컬럼크로마토그래피 정지상으로 실리카겔, 세파덱스 또는 아가로스젤 등이 사용될 수 있고, 이동상으로는 헥산(Hexane), 에틸아세테이트(Ethyl acetate), 에탄올, 메탄올, 메틸렌클로라이드 또는 에테르 등이 사용될 수 있고, 예를 들어, 클로로포름 및 메탄올의 혼합물이 사용될 수 있으나, 이에 한정되는 것은 아니다.
상기 실리카겔 컬럼크로마토그래피는 70 내지 230 메쉬 (mesh) 실리카겔 500g을 유리 컬럼에 넣고, 클로로포름과 메탄올을 97:3(v/v)로 컬럼을 충진 하여 수행하는 것 일 수 있다.
상기 실리카겔 컬럼크로마토그래피는 클로로포름과 메탄올 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7(v/v) 및 메탄올로 500ml씩 용출하고, 이 과정을 두 번 반복한 후, 용출물을 감압 농축을 하여, Fr-17 분획물을 만드는 것 일 수 있다.
상기 제3 분획 단계는 제2 분획 단계에서 용출된 분획물을 컬럼크로마토그래피를 이용하여 수행되는 것이다.
구체적으로, 상기 제3 분획 단계는 세파덱스 컬럼 (sephadex LH 20)을 이용하여 분리를 진행할 수 있는 것 일 수 있다.
상기 세파덱스 컬럼은 3차원 망목 구조를 지닌 겔 여과를 위한 충전용 텍스트란의 상품명으로서, LH타입은 텍스트란이 알킬화된 유도체로서 친유성 및 친수성 특징을 아울러 지니고 있는 것 일 수 있다.
상기 세파덱스 컬럼은 이동상으로는 100% 메탄올을 사용하는 것 일 수 있다.
상기 세파덱스 컬럼 시료 및 이동상의 온도는 10 ℃ 내외로 유지하였고 이동상의 속도는 2ml/분 일 수 있으나, 이에 한정되는 것은 아니다.
상기 세파덱스 컬럼에 흡착을 일으키는 작용의 형태는 분자 사이를 끄는 힘과 같은 것으로 정전기적 인력, 착물화 또는 수소결합 등이 있을 수 있으나, 이에 한정되는 것은 아니다.
상기 제4 분획 단계는 제3 분획 단계에서 용출된 분획물을 막크로마토 그래피를 이용하여 수행하는 것이다.
본 발명에 있어서 프렙 막크로마토그래피는 정성의 확인 목적이 아닌 단순한 분취에 목적을 둘 수 있다. 따라서, 제3 분획 단계에서 용출된 분획물을 단독으로 사용할 수 있고, 각각의 분획물을 합하여 사용할 수도 있으나, 이에 한정되는 것은 아니다.
상기 프렙 막크로마토그래피 플레이트(Plate)에는 실리카 겔(silica gel)이 정지상 역할을 할 수 있으나, 이에 한정되는 것은 아니다.
상기 프렙 막크로마토그래피의 이동상인 용매는 증류수, 메탄올, 에탄올, 아세톤, 에테르, 시클로헥산, 부탄올, 아세트산, 벤젠 등을 사용할 수 있으나, 이에 한정되는 것은 아니다.
상기 프렙 막크로마토그래피의 이동상인 용매는 부탄올, 증류수 및 아세트산을 12대 6대 4(v/v/v) 용매조건에서 전개하여 수행하는 것 일 수 있다.
상기 제4 분획 단계는 Rf값의 범위가 각각 0.8 내지 0.95, 0.6 내지 0.8, 0.5 내지 0.6 및 0.4 내지 0.5인 제4 분획물을 수득하는 단계이다.
본 발명의 일 구현예에서, 매실즙에 에틸아세테이트를 용매로 사용하여 매실즙 에틸아세테이트 분획물을 얻을 수 있는 것 일 수 있다.
상기 매실즙 에틸아세테이트 분획물을 컬럼크로마토그래피를 수행하여 1개의 분획물을 얻을 수 있는 것 일수 있으며, 상기 분획물을 다시 컬럼크로마토그래피를 수행하여 4개의 분획을 얻을 수 있는 것 일 수 있다.
컬럼크로마토그래피를 수행하여 얻은 분획 중 1번째 분획을 막크로마토 그래피를 수행하여 Rf값이 각각 0.8 내지 0.95, 0.6 내지 0.8, 0.5 내지 0.6 및 0.4 내지 0.5인 활성 물질을 얻을 수 있는 것 일 수 있다.
또한, 컬럼크로마토그래피를 수행하여 얻은 분획 중 2번째 및 3번째 분획의 혼합물을 막크로마토그래피를 수행하여 Rf값이 각각 0.8 내지 0.95, 0.6 내지 0.8, 0.5 내지 0.6 및 0.4 내지 0.5인 활성 물질을 얻을 수 있는 것 일 수 있다.
본 발명은 매실 추출물 또는 이의 분획물을 유효성분으로 포함하는 A, B형 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물, 및 A, B형 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물에 관한 것으로, 본 발명에 따른 매실 추출물은 인플루엔자 바이러스 감염 예방, 치료, 개선 등에서 우수한 효과를 나타내는 바, 다양한 인플루엔자 바이러스 감염 예방, 치료, 개선에 유용한 약학적 조성물 또는 개선용 식품 조성물로 이용될 수 있다.

도 2b는 본 발명의 일 실시예에 따라 감염동물모델 7주령 Balb/C 암컷 마우스에 매실 가공품인 매실청을 1/5로 희석하여 14일간 매일 200 uL를 구강으로 투여하고, A형 인플루엔자 바이러스(H1N1)을 감염시킨 후, 14일 동안 마우스의 생존률(%)을 나타낸 결과를 나타낸 그래프이다.
도 3은 본 발명의 일 실시예에 따른 매실즙 에틸아세테이트 분획물로부터 항인플루엔자 바이러스 활성 물질 분리 모식도를 나타낸 것이다.

도 5는 본 발명의 일 실시예에 따라 항인플루엔자 바이러스 활성 물질을 분리한 후 획득한 최종 분획물(F1711)의 HPLC 분석 결과이다.

매실(Prunus mume) 추출물 또는 이의 분획물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물에 관한 것이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지는 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
제조예 1. 매실즙 제조
매실은 익은 정도에 따라 풋매실, 청매실, 및 황매실로 나뉘는데, 풋매실은 식용으로 쓸 수 없는 덜 익은 매실이고, 청매실은 씨앗이 단단하게 굳어진 다 자란 매실이며, 황매실은 청매실이 끝까지 잘 익어 과피에 노란 기운이 돌고 과육이 부드러워진 것이다. 본 제조예에서는 청매실을 선택하였다.
청매실5kg을 세정하였다. 물기를 완전히 없앤 다음 -30℃ 내지 -40℃의 온도로 2내지3시간 동안 급속 냉동한 후에, 보관실에 넣어 이를 -2℃ 내지 -3℃의 온도로 유지하였다.
용기 저면에 통기공이 있는 용기1에 넣고 매실즙을 받을 용기2위에 용기1을 적층한 후, 50kg의 무게로 용기 1을 2 내지3시간 누르는 압착을 통하여15 내지 20%의 매실즙이 용기1의 통기공을 통하여 추출되도록 하였다.
제조예 2. 매실청 제조
매실은 제조예 1과 동일한 것으로 선택하여 매실청을 제조하였다.
물에 식초 몇 방울을 넣어 매실을 세정하였다. 씻은 매실을 마른 수건으로 닦아서 펼쳐 놓은 후, 상온(21℃)에서 하루 동안 건조하였다. 10분간 100℃ 끓는 물의 증기로 병을 소독하고 말린 후, 매실 1kg과 설탕1kg을 1:1 비율로 합한 후 고루 섞어 병에 담았다. 3개월동안, 10℃에서 보관하며 숙성시켰다. 설탕이 다 녹아 연한 갈색빛을 나타내면 매실청을 체로 걸러내었다.
제조예 3. 매실즙 유기용매 분획 추출물의 제조
매실즙10ml을 증류수 90ml에 섞어(희석과정) 100ml의 99% 에틸아세테이트로 2회 분획하여, 에틸아세테이트 층으로부터 매실즙 에틸아세테이트 분획물(A-EtOAc)을 획득하였다.
회수한 나머지 물 층을 100ml의 99.5% 부탄올로 2회 재분획하여, 부탄올 층으로부터 매실즙 조추출물 부탄올 분획물(A-BuOH)을 획득하고, 나머지를 물 분획물(A-water)로 획득하였다.
각각의 분획물을 감압 농축하였다. 그 결과, 도 1a에 개시된 바와 같이 본 발명에서 매실즙 에틸아세테이트 분획물(A-EtOAc) 31.2 mg, 부탄올 분획물(A-BuOH) 143.7 mg 및 물 분획물을 확보하였다.
제조예 4. 매실청 유기용매 분획 추출물의 제조
매실청 10ml을 증류수 90ml에 섞어 100ml의 99% 에틸아세테이트로 2회 분획하여, 에틸아세테이트 층으로부터 매실청 에틸아세테이트 분획물(A-EtOAc)을 획득하였다.
회수한 나머지 물 층을 100ml의 99.5% 부탄올로 2회 재분획하여, 부탄올층으로부터 매실청 부탄올 분획물(A-BuOH)을 획득하고, 나머지를 물 분획물(A-water)로 획득하였다.
각각의 분획 추출물을 감압 농축하였다. 그 결과, 도 1b에 개시된 바와 같이 매실청 에틸아세테이트 분획물(B-EtOAc) 54 mg, 매실청 부탄올 분획물(B-BuOH) 383.8 mg및 물 분획물을 확보하였다.
실시예 1. 매실청의 항인플루엔자 바이러스 활성 평가
6-7 주령된 Balb/C 암컷 마우스에 매실청20ml를 증류수80ml으로 희석하여, 2주 동안 매일 200 ul를 구강으로 주입하였다. 2주간 매실청 희석액을 구강 주입 한 마우스에 인플루엔자 바이러스 maPR8 50ul를 비강으로 감염시켰다. 대조군으로 매실청 희석액을 구강 주입하고 바이러스를 감염하지 않은 마우스 및 매실청 희석액을 주입하지 않고 바이러스를 감염시킨 마우스를 함께 실험하였고, 그 결과를 도 2a 내지 도 2b에 나타내었다. 다음의 표 1은 마우스의 바이러스 감염 이후부터 14일간의 몸무게 변화 측정 결과 및 생존률을 나타낸 것이다.

도 2a 내지 도 2b에서 확인할 수 있듯이, 매실청 희석액을 먹이지 않은 경우 감염 11일 째에 40%의 마우스가 생존하였지만, 몸무게가 회복되지 않았다.
반면, 매실청 희석액을 먹인 그룹에서는 60%가 생존하였으며 감염 후 8일째부터 몸무게 회복이 시작되어 마지막 관찰 날인 14일째에는 비감염 그룹 대비 90%가량 몸무게가 회복되었다.
실시예 2. 유기용매 분획물의 항인플루엔자 바이러스 활성 평가
확보한 매실즙과 매실청의 유기용매 분획물을 500 ug/mL, 166.7 ug/mL, 55.6 ug/mL, 18.5 ug/mL, 6.2 ug/mL, 2.06 ug/mL, 0.69 ug/mL, 0.23 ug/mL, 0.08 ug/mL 및 0.03 ug/mL 농도 수준에서 MDCK (Madin-Darby Canine Kidney) 세포에 대하여 항바이러스 활성을 조사하였다. 상기 MDCK 세포는 American Type Culture Collection에서 구입하여 사용하였다.
구체적으로, 96웰 플레이트에 MDCK 세포를 3 X 104 세포/웰의 수준으로 분주하고, 하루 동안 5 % CO2, 37℃ 배양기에서 배양하였다. 그 다음, H1N1 A형 인플루엔자 바이러스 (PR8, A/Puerto Rico/8/34), H3N2 A형 인플루엔자 바이러스(HK, A/Hong Kong/8/68) 및 B형 인플루엔자 바이러스(B/Lee/40) 3종 각 0.001 MOI와, Minimum Essential Media (MEM, Invitrogen)에 희석된 매실 가공품 유기용매 분획물 6종을 각각 혼합하여 37℃에서 1시간 전 배양하였다.
전 배양된 바이러스를 PBS (phosphate buffered saline)로 세척한 MDCK 세포에 감염시켜 5 % CO2, 35℃ 배양기에서 2일 간 배양하였다. 그 다음, MDCK 세포 생존율을 FDA (fluorescein diacetate) 어세이를 통하여 측정하여, 그 결과를 표 2에 나타내었다. 표 2에 EC50 은 50% 효과적인 농도를 나타내며, CC50은 50% 세포 독성을 나타낸다. S.I는 선택성 지수이며, CC50/EC50으로 나타낸다. 아만타딘(AMT, amantadine), 리바비린(RBV, ribavirin), 오셀타미비르 카르복실레이트 (OSV-C, oseltamivir carboxylate)을 대조약제로 사용하였다. EC50 은 항바이러스 활성 값을 나타내는 것으로서, 낮은 값을 나타낼수록 항바이러스에 대한 효과가 우수하다.

표 2에서 확인할 수 있듯이, 매실즙 에틸아세테이트 분획물(A-EtOAc)이 3종의 인플루엔자 바이러스에 대하여 항바이러스 활성을 나타내었고, 항바이러스 활성 값(EC50)은 H1N1 A형 인플루엔자 바이러스에 대하여 77.2 ug/mL, H3N2 A형 인플루엔자 바이러스에 대하여 111.4 ug/mL, B형 인플루엔자 바이러스에 대하여 270.7 ug/mL였으며, 세포독성은 관찰되지 않았다. 따라서, 매실즙 에틸아세테이트 분획물에 항인플루엔자 바이러스 활성 물질이 존재하는 것을 확인하였다.
실시예 3. 에틸아세테이트 분획물(A-EtOAc) 로부터 항인플루엔자 바이러스 활성 물질 분리
매실즙 3,750 mL에 동량의 에틸아세테이트로 2회 분획하여, 40℃에서 감압 농축하여 완전히 농축된 에틸아세테이트 분획물(A-EtOAc) 12.89 g을 확보하였다.
이때, 에틸아세테이트 분획물(A-EtOAc) 12.89 g은 용매가 완전히 제거된 상태이다.
실리카젤 컬럼(Silica gel column, Kiesel gel 60 500 g, 70-230mesh; E. Merck, Darmstadt, Germany)을 클로로포름과 메탄올 97:3(v/v)로 컬럼을 충진 하였다. 클로로포름과 메탄올 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7(v/v), 및 메탄올로 500 mL씩 2회 용출하였다. 그 다음, 감압 농축해서 총 17개의 분획물 (Fr1 내지Fr17)을 확보하였다. 17개의 분획물 중에서 항바이러스 활성 물질을 포함하는 Fr17 분획물 342.5 mg을 수득하였다.
Fr17 분획물을 세팍(Sep-Pak C18 cartridge, 35 cc, 10 g) 컬럼크로마토 그래피를 통하여 100% 물부터 10, 20, 30, 40, 50, 60, 70, 80, 90, 100% 메탄올을 10 mL씩 2회 용출하였다. 그 다음, 프렙 막크로마토그래피 (Silica gel 60 F254, 0.5 mm; Merck, Germany) 플레이트에 점적하고 클로로포름과 메탄올 3대 1(v/v) 용매조건에서 전개하여 수행하였다. 그 다음, Fr1, Fr4, Fr5, Fr6, Fr7, Fr8-9 및 Fr10-14 분획을 각각 합하고, 감압 농축하여 표3 및 도3에 기재된 바와 같이 매실즙 에틸아세테이트 분획물(A-EtOAc) 시료로부터 최종적으로 분리한 7개의 분획물을 확보하였다.

실시예 4. 분획물의 항인플루엔자 바이러스 활성 평가
매실즙 에틸아세테이트 분획물(A-EtOAc) 시료로부터 확보한 7개의 분획물을 500 ug/mL, 166.7 ug/mL, 55.6 ug/mL, 18.5 ug/mL, 6.2 ug/mL, 2.06 ug/mL, 0.69 ug/mL, 0.23 ug/mL, 0.08 ug/mL 및 0.03 ug/mL 농도 수준에서 MDCK 세포에 대한 항바이러스 활성을 실시예 2에 기재된 방법과 동일한 방법으로 평가하였다.
구체적으로, 96 웰 플레이트에 MDCK세포를 3 X 104 세포/웰의 수준으로 분주하고, 하루 동안 5 % CO2, 37℃ 배양기에서 배양하였다. 하루 뒤, H1N1 A형 인플루엔자 바이러스 (PR8, A/Puerto Rico/8/34), H3N2 A형 인플루엔자 바이러스(HK, A/Hong Kong/8/68) 및 B형 인플루엔자 바이러스(B/Lee/40) 3종 각 0.001 MOI와, Minimum Essential Media (MEM, Invitrogen)에 희석된 매실 가공품 7개의 유기용매 분획물에 각각 혼합하여 37℃에서 1시간 전 배양하였다.

표 4에서 확인할 수 있듯이, 4개의 분획물 Fr17-4 내지Fr17-7에서 항인플루엔자 바이러스 활성이 확인되었으며, 그 중에서 Fr17-7이 가장 강하고, 그 다음이 Fr17-6인 것으로 나타났다. Fr17-6 분획물은 H1N1 A형 인플루엔자 바이러스에서 26.5 ug/mL, H3N2 A형 인플루엔자 바이러스에서 3.1 ug/mL, 및 B형 인플루엔자 바이러스에서10.3 ug/mL으로 강한 항인플루엔자 바이러스활성을 나타내었고, Fr17-7 분획물은 H1N1 A형 인플루엔자 바이러스에서 4.0 ug/mL, H3N2 A형 인플루엔자 바이러스에서 3.0 ug/mL, 및 B형 인플루엔자 바이러스에서 1.8 ug/mL으로 강한 항인플루엔자 바이러스활성을 나타내었다. 하지만, 두 시료의 세포독성 값(CC50)이 185.6 및 25.5 ug/mL로 나타나 독성이 있는 것으로 관찰되었는데, 이는 항인플루엔자바이러스 물질이 독성을 보일 수도 있지만, 불순물이 독성을 보일 수도 있다고 추정되었다.
실시예 5. 항인플루엔자 바이러스 활성 물질의 분리
항인플루엔자 바이러스 활성 물질을 고농도로 분리하기 위하여 도 4에 개시된 바와 같이 매실즙 3,750 mL에 동량의 에틸아세테이트로 2회 분획 및 40℃에서 감압 농축하여 확보한 에틸아세테이트 분획물(A-EtOAc) 12.89 g을 분자 크기에 따라 분획하였다.
실리카젤 컬럼(Silica gel column, Kiesel gel 60 500 g, 70-230mesh; E. Merck, Darmstadt, Germany)을 클로로포름과 메탄올 97:3(v/v)로 컬럼을 충진 하였다. 클로로포름과 메탄올 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7(v/v), 및 메탄올로 500 mL씩 2회 용출하여, Fr-17 분획물 120mg을 용출하였다. 용출물을 세파 덱스 LH-20 컬럼(Sephadex LH-20 columm)을 수행하여 물질 분리를 실시하였다. 컬럼 크기는 2 cm(내경) X 90 cm(높이)였으며, 레진은 50 g을 충진 하였고, 100% 메탄올로 용출하여 도 4와 같이 4개의 분획물 F171 내지F174을 확보하였다.
4개의 분획물 중에서F171 (47 mg) 분획물과 F172(30 mg) 및 F173(20 mg) 분획물을 합한 F172C(50 mg)을 극성에 따라 분획하였다.
구체적으로, 각각 프렙(Prep) 막크로마토그래피 플레이트 (Silica gel 60 F254, 0.5 mm; Merck, Germany)에 점적하고, 부탄올, 증류수, 및 아세트산을 12대 6대 4(v/v/v) 용매조건에서 전개하여 수행하여 8개의 분획물 (Rf값이 각각 0.9, 0.7, 0.55및 0.45인F1711 내지 F1714 및; Rf값이 각각 0.9, 0.7, 0.55및 0.45 인F172C1 내지 F172C4)을 확보하였다.
8개의 분획물을 실리카 막크로마토그래피 플레이트(Silica TLC plate)에 점적하고, 부탄올, 증류수, 및 아세트산을 12대 6대 4(v/v/v) 용매조건에서 분석을 수행하였다. 그 결과 F1711 분획물 (10 mg)과 F172C1 (2.2 mg)이 거의 순수한 것으로 나타났다. 또한, Fr17-6및 Fr17-7과 동일한 화합물을 포함하고 있었다.
두 분획물 F1711및 F172C1에 대하여 HPLC분석을 실시하였으며, 분석조건은 다음과 같다. 워터스 HPLC 분석 기기와 아틀란티스 T3 C18 컬럼 (4.6 × 250 mm; Waters)을 이용하여 하기 표 5에 기재한 바와 같이 증류수 99.9ml 와 트리플루오로 아세틱산(trifluoroacetic acid) 0.1ml을 섞은 용매 A 와 아세토 나이트릴(acetonitrile) 99.9ml와 0.1ml을 섞은 용매 B로 농도 구배조건(gradient condition)으로 30분간 HPLC 분석을 수행하였다.

그 결과, 도 5에서와 같이 F1711 분획물 및 F172C1 분획물은 불순물이 조금 있지만, 거의 순수한 것으로 나타났으며, 머무름 시간은 10.432분이었으며, UV maxima는 206.6 nm와 279.4 nm로 나타났다.
실시예 6. 분리한 물질 (F1711 및 F172C1)의 항인플루엔자 바이러스 활성 조사
본원발명에서 분리한 물질 Ja-1 물질 (F1711및 F172C1 분획물)의 항인플루엔자 바이러스 활성을 조사하기 위하여 F1711및 F172C1분획물을 500 ug/mL, 166.7 ug/mL, 55.6 ug/mL, 18.5 ug/mL, 6.2 ug/mL, 2.06 ug/mL, 0.69 ug/mL, 0.23 ug/mL, 0.08 ug/mL및 0.03 ug/mL 농도 수준에서 MDCK 세포에 대하여 항바이러스 활성을 조사하였다. 항바이러스 활성 조사 방법은 상기 실시예 2에 개시된 바와 같다.
구체적으로, 96 웰 플레이트에 MDCK세포를 3 X 104 세포/웰의 수준으로 분주하고, 하루 동안 5 % CO2, 37℃ 배양기에서 배양하였다. 하루 뒤, H1N1 A형 인플루엔자 바이러스 (PR8, A/Puerto Rico/8/34), H3N2 A형 인플루엔자 바이러스(HK, A/Hong Kong/8/68) 및 B형 인플루엔자 바이러스(B/Lee/40) 3종 각 0.001 MOI와, Minimum Essential Media (MEM, Invitrogen)에 희석된 매실 가공품 유기용매 분획물 2종을 각각 혼합하여 37℃에서 1시간 전 배양하였다.
전 배양된 바이러스를 PBS(phosphate buffered saline)로 세척한 MDCK 세포에 감염시켜 5% CO2, 35℃ 배양기에서 2일간 배양하였다. 그 다음, MDCK 세포 생존율을 FDA (fluorescein diacetate assay)를 통하여 측정하여, 그 결과를 하기 표 6에 나타내었다. EC50은 50% 효과적인 농도를 나타내며, CC50은 50% 세포 독성을 나타낸다. S.I는 선택성 지수이며, CC50/EC50으로 나타낸다. 아만타딘(AMT, amantadine), 리바비린(RBV, ribavirin), 오셀타미비르 카르복실레이트 (OSV-C, oseltamivir carboxylate)을 대조약제로 사용하였다. EC50 은 항바이러스 활성 값을 나타내는 것으로서, 낮은 값을 나타낼수록 항바이러스에 대한 효과가 우수하다.

표 6에서 확인할 수 있듯이, 본 발명에서 분리한 물질 Ja-1(F1711및 F172C1)에서 강한 항인플루엔자 바이러스 활성을 나타냈다. F1711및 F172C1 분획물의 항바이러스 활성 값(EC50)은 H1N1 A형 인플루엔자 바이러스에 대하여 각각 7.4 ug/mL, 5.0 ug/mL였으며, H3N2 A형 인플루엔자 바이러스에 대하여 4.1 ug/mL, 4.2 ug/mL이었다. 그리고, B형 인플루엔자 바이러스에 대하여 2.8 ug/mL, 8.0 ug/mL였다. F1711및 F172C1 분획물에서 세포독성은 관찰되지 않았으며, 두 분획물의 활성은 거의 동일한 것으로 나타났다. Fr17-6및 Fr17-7분획물과는 달리 Ja-1 물질 (F1711및 F172C1)은 세포독성이 없는 것으로 나타났다. 따라서, Fr17-6및 Fr17-7의 세포독성은 항인플루엔자 바이러스 활성 물질인 Ja-1 물질에 의한 것이 아닌 다른 물질에 의해 나타나는 것으로 판단되었다.
실시예 7. 항인플루엔자 바이러스 활성 물질 Ja-1의 분석
다양한 매실가공품과 과실에서 항인플루엔자 바이러스 활성 물질 Ja-1의 존재 여부를 확인하기 위하여 F1711 분획물을 1 mg/mL 수준으로 메탄올로 용해하여 준비하고, 4종류의 서로 다른 매실청인 청매실원(B-1), 매실원(B-2), 월앙매실(B-3) 및 청매실액(B-4)을 구입하여 멸균수로 3배 희석하였다. 서로 다른 매실종인 남고와 백가하를 각각 30 g씩 100 mL의 메탄올에 넣어 추출하여, 남고 메탄올 분획물(a-MeOH)와 백가하 메탄올 분획물(b-MeOH)을 준비하였다. HPLC 분석을 위하여 준비된 모든 시료는 0.4 um 멤브레인 필터로 여과한 후, 10 uL를 워터스 HPLC 분석 기기와 아틀란티스 T3 C18 컬럼 (4.6 × 250 mm; Waters)을 이용하여 증류수 99.9ml 와 트리플루오로아세틱산(trifluoroacetic acid) 0.1ml을 섞은 용매 A 와 아세토나이트릴(acetonitrile) 99.9ml와 0.1ml을 섞은 용매 B로 농도 구배조건(gradient condition)으로 30분간 HPLC 분석을 수행하였고, 그 결과를 도 6a, 6b 및 표 7에 나타내었다.

도 6a에서 확인할 수 있듯이, 4가지 매실청 희석액 및 2가지 과실의 메탄올 분획물에서 인플루엔자 바이러스 활성 물질 분획 F1711과 동일하게 약 10분의 리텐션 타임을 갖는 피크를 확인할 수 있었다. 그리고, 도 6a에 나타난 피크의 UV 스펙트럼을 비교 분석한 결과, 같은 물질인 것을 증명할 수 있었다. 표 7에서 확인할 수 있듯이, F1711 분획물의 농도를 기준으로, 4가지 매실청 희석액과 2가지 과실의 메탄올 분획물에서 항인플루엔자 바이러스 활성 물질 2의 농도를 정량한 결과, 4가지 매실청 희석액에서는 약 214 ug/mL부터 약 750 ug/mL의 활성 물질이 정량되었고, 남고 메탄올 분획물에서는 약 1372.95 ug/mL, 백가하 메탄올 분획물에는 약 340.54 ug/mL이 정량되었다.
실시예 8. 다양한 매실가공품 및 과실에서 무메푸랄(Mumefural) 정량 분석
다양한 매실가공품 및 과실에서 기존에 매실농축액에서 항바이러스 활성이 있다고 보고된 무메푸랄(mumefural)물질 존재 여부를 확인하기 위하여 무메푸랄을 메탄올로 용해하여 100 ug/mL, 33.3 ug/mL, 11.1 ug/mL, 및3.7 ug/mL 수준으로 10 uL를 워터스 HPLC 분석 기기와 아틀란티스 T3 C18 컬럼 (4.6 X 250 mm; Waters)을 이용하여 증류수 99.9ml 와 트리플루오로아세틱산(trifluoroacetic acid) 0.1ml을 섞은 용매 A 와 아세토나이트릴(acetonitrile) 99.9ml와 0.1ml을 섞은 용매 B로 농도 구배조건(gradient condition)으로 30분간 HPLC 분석을 수행하였다.
각 농도에서 분석한 무메푸랄의 HPLC 크로마토그램의 면적을 분석하여 정량 곡선을 작성하였다.
다양한 매실가공품 매실청, 매실즙, 매실농축액(홍쌍리 매실농축액, 제조사: 청매실 농원)을 각각 원액, 3배, 10배 증류수로 희석하여 준비하고, 실시예 7의 남고 메탄올 추출물(과실a-MeOH)과 백가하 메탄올 추출물(과실b-MeOH)을 각각 10 uL를 상기의 조건으로 HPLC 분석을 수행하여 무메푸랄을 정량 분석하여, 그 결과를 하기 표 8에 나타내었다.

무메푸랄은 매실 농축과정에서 생성된다. 구체적으로, 매실을 갈아서 액이 나오면 그 액체에서 수분을 증발시키기 위해 가열을 하게 되며, 이때 무메푸랄이 생성된다. 표 8에서 확인할 수 있듯이, 오직 매실농축액에서 무메푸랄이 확인되었고, 무메푸랄은 1,534 ug/mL 농도로 존재하였다. 이러한 결과는 선행연구 결과와 일치하였다. 한편, 매실을 농축하는 과정에서 생성되는 무메푸랄이라는 물질이 A형 인플루엔자 바이러스에 대하여 활성이 있다고 보고된 바 있다. 이에 따라 합성 무메푸랄(Nacalai Tesque, 일본)을 구입하여 세포체계 항바이러스 실험을 수행하였다.
구체적으로, 96 웰 플레이트에 MDCK세포를 3 X 104 세포/웰의 수준으로 분주하고, 하루 동안 5 % CO2, 37℃ 배양기에서 배양하였다. 하루 뒤, H1N1 A형 인플루엔자 바이러스 (PR8, A/Puerto Rico/8/34), H3N2 A형 인플루엔자 바이러스(HK, A/Hong Kong/8/68) 및 B형 인플루엔자 바이러스(B/Lee/40) 3종 각 0.001 MOI와, Minimum Essential Media (MEM, Invitrogen)에 합성 무메푸랄을 혼합하여 37℃에서 1시간 전 배양하였다.
전 배양된 바이러스를 PBS(phosphate buffered saline)로 세척한 MDCK 세포에 감염시켜 5% CO2, 35℃ 배양기에서 2일간 배양하였다. 그 다음, MDCK 세포 생존율을 FDA (fluorescein diacetate assay)를 통하여 측정하여, 그 결과를 하기 표 9에 나타내었다. EC50은 50% 효과적인 농도를 나타내며, CC50은 50% 세포 독성을 나타낸다. S.I는 선택성 지수이며, CC50/EC50으로 나타낸다. 아만타딘(AMT, amantadine), 리바비린(RBV, ribavirin), 오셀타미비르 카르복실레이트 (OSV-C, oseltamivir carboxylate)을 대조약제로 사용하였다. 표9에서 확인할 수 있듯이, 328.6 ug/ml에서 CC50이 관찰되었으며, 그 이하의 농도에서는 항인플루엔자 효능이 관찰되지 않았다. 따라서 매실 추출물의 항인플루엔자 바이러스 활성은 무메푸랄이 아닌 Ja-1 물질에 의한 것으로 나타났다.
본 발명은 매실 추출물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물에 관한 것이다.
Claims (13)
- 매실(Prunus mume) 추출물 또는 이의 분획물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 매실 추출물은, 매실즙, 매실청, 매실농축액 및 매실 과실 추출물로 이루어진 군으로부터 선택되는 1종 이상인 것인, 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 추출물은 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 이용하여 분획한 것인, 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물.
- 제3항에 있어서, 상기 분획물은 크로마토그래피를 수행하여 얻은 것인, 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 인플루엔자 바이러스는 인플루엔자 바이러스 A형 또는 B형을 포함하는 것인, 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물.
- 제5항에 있어서, 상기 인플루엔자 바이러스 A형은 H1N1, H3N2, H1N2, H2N2, H2N3, H3N1, H3N8, H5N1, H5N2, H5N3, H5N8, H5N9, H7N1, H7N2, H7N3, H7N4, H7N7, H7N9, H9N2 또는 H10N7의 혈청형을 갖는 인플루엔자 바이러스 감염 예방 또는 치료용 약학적 조성물.
- 매실(Prunus mume) 추출물을 또는 이의 분획물을 유효성분으로 포함하는 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물.
- 제7항에 있어서, 상기 매실 추출물은, 매실즙, 매실청, 매실농축액 및 매실 과실 추출물로 이루어진 군으로부터 선택되는 1종 이상인 것인, 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물.
- 제7항에 있어서, 상기 분획물은 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 이용하여 분획한 것인, 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물.
- 제9항에 있어서, 상기 분획물은 크로마토그래피를 수행하여 얻은 것인, 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물.
- 제7항에 있어서, 상기 인플루엔자 바이러스는 인플루엔자 바이러스 A형 또는 B형을 포함하는 것인, 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물.
- 제11항에 있어서, 상기 인플루엔자 바이러스 A형은 H1N1, H3N2, H1N2, H2N2, H2N3, H3N1, H3N8, H5N1, H5N2, H5N3, H5N8, H5N9, H7N1, H7N2, H7N3, H7N4, H7N7, H7N9, H9N2 또는 H10N7의 혈청형을 갖는 것인, 인플루엔자 바이러스 감염 예방 또는 개선용 식품 조성물.
- 다음 단계를 포함하는 항바이러스 활성 물질의 추출방법:매실 추출물에 에틸에테르, 에틸아세테이트, 및 부탄올로 이루어지는 군에서 선택된 1종 이상의 용매를 사용하여 제1 분획물을 제조하는 제1 분획 단계;제1 분획물에 컬럼크로마토그래피를 수행하여 제2 분획물을 얻는 제2 분획 단계;제2 분획물에 컬럼크로마토그래피를 수행하여 제3 분획물을 얻는 제3 분획 단계; 및제3 분획물에 막크로마토그래피를 수행하여 제4 분획물을 얻는 제4 분획 단계.
