구체적으로, 상기 건강보조식품은 건강기능식품 및 건강식품 등을 포함한다. 상기 건강 기능(성) 식품(functional food)이란, 특정보건용 식품(food for special health use, FoSHU)과 동일한 용어로, 영양 공급 외에도 생체조절기능이 효율적으로 나타나도록 가공된 의학, 의료효과가 높은 식품을 의미한다. 여기서 "기능(성)"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건용도에 유용한 효과를 얻는 것을 의미한다. 본 개시의 식품은 당 업계에서 통상적으로 사용되는 방법에 의하여 제조가능하며, 상기 제조시에는 당 업계에서 통상적으로 첨가하는 원료 및 성분을 첨가하여 제조할 수 있다. 또한 상기 식품의 제형 또한 식품으로 인정되는 제형이면 제한 없이 제조될 수 있다. 본 개시의 식품 조성물은 다양한 형태의 제형으로 제조될 수 있으며, 일반 약품과는 달리 식품을 원료로 하여 약품의 장기 복용 시 발생할 수 있는 부작용 등이 없는 장점이 있고, 휴대성이 뛰어나다.

실시예 1: 생강 추출물의 유효성분 수득
실시예 1-1: 생강 추출물의 수득
생생강 100kg을 세척하고, 세척된 생강을 5mm 두께로 절단한 다음, 절단된 생강을 45℃ 에서 40시간 동안 건조하였다. 이어, 건조생강에 15배 중량의 70% 에탄올을 가하고, 70℃ 에서 5시간 동안 추출 및 여과하고, 65 내지 68℃의 온도조건 및 400 내지 500torr의 압력조건에서 60 birx로 농축하여 생강 추출물을 수득하였다.
실시예 1-2: 유효성분의 분리
실시예 1-2-1: 1차 분획물의 수득
상기 실시예 1-1에서 수득한 생강 추출물 265 g을 물 (2.8 L)과 n-hexane (2.8 L×2)로 분배 추출하여 11.0 g의 n-헥산 분획물 (ZOH)을 수득하였다.
상기 ZOH (11.0 g)을 SiO2 컬럼 크로마토그래피(Φ 7.0×16.0 cm, n-hexane-EtOAc= 4:1 → 1:1 → CHCl3-MeOH-water=30:3:1, 각 500 mL)에 적용하여 12개의 1차 분획물(ZOH-1 내지 ZOH-12)을 수득하였다. 상기 12개의 1차 분획물 중에서 분획물 ZOH-6(2.6 g, elution volume/total volume(Ve/Vt) 0.164-0.462) 및 분획물 ZOH-9 (2.4 g, Ve/Vt 0.557-693)을 선발하였다.
실시예 1-2-2: ZOH-6 유래 2차 분획물의 수득
상기 실시예 1-2-1에서 선발된 분획물 ZOH-6을 ODS 컬럼 크로마토그래피(Φ 5.0×18.0 cm, acetone-MeOH-H2O=1:1:1, 1.6 L)에 적용하여, 총 10개의 2차 분획물(ZOH-6-1 내지 6-10)을 수득한 다음, 이들 중, 분획물 ZOH-6-5 (296.9 mg, Ve/Vt 0.096-0.232) 및 분획물 ZOH-6-6 (145.4 mg, Ve/Vt 0.233-0.303)을 선발하였다.
실시예 1-2-2-1: 6-쇼가올(shogaol)의 분리정제
상기 실시예 1-2-2에서 선발된 분획물 ZOH-6-5를 SiO2 컬럼 크로마토그래피(Φ 2.0×15.0 cm, CHCl3-EtOAc=50:1 → 30:1 → 10:1, 각각 500 mL)에 적용하여 총 13개의 3차 분획물(ZOH-6-5-1 내지 6-5-13)을 수득하고, 이들 중 분획물 ZOH-6-5-6(24.3 mg, Ve/Vt 0.320-0.540)를 선발하였다.
상기 선발된 분획물 ZOH-6-5-6를 SiO2 TLC(Rf 0.60, CHCl3-EtOAc=10:1) 및 ODS TLC(Rf 0.70, acetone-MeOH-H2O=3:1:1)에 적용하여, 6-쇼가올(shogaol)(화합물 1)을 분리정제 하였다.
실시예 1-2-2-2: 분획물 ZOH-6-6 유래 3차 분획물의 수득
상기 실시예 1-2-2에서 선발된 분획물 ZOH-6-6을 SiO2 컬럼 크로마토그래피(Φ 2.0×17.0 cm, n-hexane-EtOAc=10:1, 700 mL)에 적용하여 총 5개의 3차 분획물(ZOH-6-6-1 내지 6-6-6)을 수득하고, 이들 중, 분획물 ZOH-6-6-2(4.2 mg, Ve/Vt 0.060-0.120) 및 분획물 ZOH-6-6-5(13.1 mg, Ve/Vt 0.760-0.870)를 선발하였다.
실시예 1-2-2-2-1: 6-파라돌(paradol)의 분리정제
상기 실시예 1-2-2-2에서 선발된 분획물 ZOH-6-6-2를 SiO2 TLC(Rf 0.55, CHCl3-EtOAc=3:1) 및 ODS TLC(Rf 0.65, acetone-MeOH-H2O=4:1:1)에 적용하여, 6-파라돌(paradol)(화합물 2)을 분리정제하였다.
실시예 1-2-2-2-2: 데하이드로-6-진저디온(dehydro-6-gingerdione)의 분리정제
상기 실시예 1-2-2-2에서 선발된 분획물 ZOH-6-6-5를 SiO2 TLC(Rf 0.50, CHCl3-EtOAc=3:1) 및 ODS TLC(Rf 0.65, acetone-MeOH-H2O=4:1:1)에 적용하여 데하이드로-6-진저디온(dehydro-6-gingerdione)(화합물 3)을 분리정제하였다.
실시예 1-2-3: ZOH-9 유래 2차 분획물의 수득
상기 실시예 1-2-1에서 선발된 분획물 ZOH-9를 ODS 컬럼 크로마토그래피(Φ 4.0×5.5 cm, MeOH-H2O=1:1 → 2:1, 각 1.6 L)에 적용하여, 총 10개의 2차 분획물(ZOH-9-1 내지 9-10)을 수득한 다음, 이들 중, 분획물 ZOH-9-1(1.2 g, Ve/Vt 0.000-0.331), 분획물 ZOH-9-4(174.9 mg, Ve/Vt 0.460-0.680) 및 분획물 ZOH-9-7(147.0 mg, Ve/Vt 0.820-0.920)을 선발하였다.
실시예 1-2-3-1: 6-진제롤(gingerol)의 분리정제
상기 실시예 1-2-3에서 선발된 분획물 ZOH-9-1을 SiO2 TLC(Rf 0.65, CHCl3-MeOH-H2O=15:3:1) 및 ODS TLC(Rf 0.70, MeOH-H2O=5:1)에 적용하여, 6-진제롤(gingerol)(화합물 4)을 분리정제하였다.
실시예 1-2-3-2: 8-진제롤(gingerol)의 분리정제
상기 실시예 1-2-3에서 선발된 분획물 ZOH-9-4를 SiO2 TLC(Rf 0.70, CHCl3-MeOH-H2O=15:3:1) 및 ODS TLC(Rf 0.55, MeOH-H2O=5:1)에 적용하여, 8-진제롤(gingerol)(화합물 5)을 분리정제하였다.
실시예 1-2-3-3: 10-진제롤(gingerol)의 분리정제
상기 실시예 1-2-3에서 선발된 분획물 ZOH-9-7을 SiO2 TLC(Rf 0.75, CHCl3-MeOH-H2O=15:3:1) 및 ODS TLC(Rf 0.45, MeOH-H2O=5:1)에 적용하여, 10-진제롤(gingerol)(화합물 6)을 분리정제하였다.
실시예 2: 알록산 유래 당뇨증상에 대한 생강 추출물 유래 화합물의 항당뇨 효과
알록산으로 유발한 당뇨 제브라피시를 대상으로, 실시예 1-2에서 분리정제된 6종의 생강 추출물 유래 화합물의 항당뇨 효과를 시험하였다.
대략적으로, 수정 6일 후(5 day post fertilization)의 제브라피시 유생(zebrafish larvae)을 96 웰에 위치시킨 후, 100μM 알록산을 가하고 6시간 동안 반응시켰다. 그런 다음, 상기 실시예 1-2에서 분리정제한 6종의 생강 추출물 유래 화합물을 각각 0.1 μg/mL 및 1 μg/mL 농도로 48시간 동안 처리하였다.
이어, 상기 제브라피시 유생에 40μM 2-NBDG를 가하여 30분 동안 염색한 다음, 0.03% 해수염 용액을 가하여 20분 동안 세척하고, 형광현미경(Olympus 1×70, Olympus, Japan)을 사용하여 췌장섬(pancreatic islet, PI)을 촬영한 다음, Focus Lite 소프트웨어를 사용하여 제브라피시의 췌장섬 크기변화를 분석하였다(도 1).
도 1은 제브라피시 유생에 알록산(AX)를 처리한 후, 6종의 생강 추출물 유래 화합물을 0.1 μg/mL 또는 1 μg/mL의 농도로 처리한 결과를 나타내는 형광현미경 사진 및 상기 형광현미경 사진으로부터 산출된 췌장섬의 크기변화를 나타내는 그래프이다.
도 1에서 보듯이, 제브라피시 유생에 알록산(AX)를 처리하면 췌장섬의 크기가 현저히 감소되었으나, 상기 알록산을 처리한 후, 생강 추출물 유래 화합물을 처리하면, 감소된 췌장섬의 크기가 정상 수치로 회복됨을 확인하였다. 특히, 6종의 생강 추출물 유래 화합물 중에서도 6-파라돌 및 데하이드로-6-진저디온이 상대적으로 우수한 회복효과를 나타냄을 확인하였다.
실시예 3: 과다인슐린 유래 당뇨증상에 대한 생강 추출물 유래 화합물의 항당뇨 효과
과다한 인슐린으로 유발한 당뇨 제브라피시를 대상으로, 실시예 1-2에서 분리정제된 6종의 생강 추출물 유래 화합물의 항당뇨 효과를 시험하였다.
대략적으로, 수정 3일 후(3 day post fertilization)의 제브라피시 유생을 6 웰에 위치시킨 후, 10μM 인슐린을 가하고 48시간 동안 반응시켰다. 그런 다음, 상기 실시예 1-2에서 분리정제한 6종의 생강 추출물 유래 화합물을 각각 0.1 μg/mL 및 1 μg/mL 농도로 48시간 동안 처리하였다.

도 2는 제브라피시 유생에 인슐린(IBS)을 처리한 후, 6종의 생강 추출물 유래 화합물을 0.1 μg/mL 또는 1 μg/mL의 농도로 처리한 결과를 나타내는 형광현미경 사진 및 상기 형광현미경 사진으로부터 산출된 췌장섬의 크기변화를 나타내는 그래프이다.
도 2에서 보듯이, 제브라피시 유생에 인슐린(IBS)을 과다처리하면 췌장섬의 크기가 현저히 감소되었으나, 상기 과다인슐린을 처리한 후, 생강 추출물 유래 화합물을 처리하면, 감소된 췌장섬의 크기가 정상 수치로 회복됨을 확인하였다. 특히, 상기 도 1과 유사하게 6종의 생강 추출물 유래 화합물 중에서도 6-파라돌 및 데하이드로-6-진저디온이 상대적으로 우수한 회복효과를 나타냄을 확인하였다.
실시예 4: 생강 추출물 유래 화합물의 항비만 효과
식이 유발성 비만 제브라피시를 대상으로, 실시예 1-2에서 분리정제된 6종의 생강 추출물 유래 화합물의 항당뇨 효과를 시험하였다.
대략적으로, 수정 3일 후의 제브라피시 유생을 페트리디쉬에 위치시킨 후, 정상군(100마리)과 식이유발군(100마리)을 각각 설정하였다.
상기 정상군에는 dpf 3~5 기간에는 로티퍼 5 mg, dpf 6~10 기간에는 아르테미아 5 mg, dpf 11~13 기간에는 건조사료 30 mg을 제공하여 섭취시키고, dpf14~15 기간에는 0.1 μg/mL의 생강 추출물 유래 화합물 또는 0.03% 해수염 용액을 제공하였다.
상기 식이유발군에는 dpf 3~5 기간에는 로티퍼 60 mg, dpf 6~10 기간에는 아르테미아 60 mg, dpf 11~13 기간에는 건조사료 18 mg을 제공하여 섭취시키고, dpf14~15 기간에는 0.1 μg/mL의 생강 추출물 유래 화합물 또는 0.03% 해수염 용액을 제공하였다.
상기 정상군과 식이유발군의 몸길이, 몸무게 및 배둘레의 길이를 각각 측정하고, 변화수준을 분석하였다(도 3a 내지 3c).
도 3a는 정상군과 식이유발군 제브라피시 유생의 몸길이의 변화를 측정한 결과를 나타내는 그래프이다.
도 3a에서 보듯이, 정상군에 비하여 식이유발군에서는 몸길이가 증가되었으나, 생강 추출물 유래 화합물의 투여에 따른 변화는 나타나지 않음을 확인하였다.
도 3b는 정상군과 식이유발군 제브라피시 유생의 배둘레 길이의 변화를 측정한 결과를 나타내는 그래프이다.
도 3b에서 보듯이, 정상군에 비하여 식이유발군에서는 배둘레 길이가 증가되었고, 생강 추출물 유래 화합물의 투여에 의해 배둘레 길이가 감소되는 경향을 나타냄을 확인하였다.
도 3c는 정상군과 식이유발군 제브라피시 유생의 몸무게의 변화를 측정한 결과를 나타내는 그래프이다.
도 3c에서 보듯이, 정상군에 비하여 식이유발군에서는 몸무게가 증가되었고, 생강 추출물 유래 화합물의 투여에 의해 몸무게가 감소되는 경향을 나타냄을 확인하였다. 특히, 6-파라돌, 데하이드로-6-진저디온, 8-진제롤 및 10-진제롤이 상대적으로 우수한 몸무게 감소효과를 나타냄을 확인하였다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시 예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해해야만 한다. 본 개시의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 개시의 범위에 포함되는 것으로 해석되어야 한다.
Claims (8)
- 디하이드로-6-진저디온(Dehydro-6-gingerdione) 또는 그의 약학적으로 허용되는 염을 포함하는 대사성 질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서,상기 대사성 질환은 비만, 고혈압, 동맥경화, 당뇨, 인슐린저항성, 간지방증, 지방간 또는 이상지질혈증인 것인, 약학 조성물.
- 제1항에 있어서,상기 조성물은 생강으로부터 분리정제된 화합물을 추가로 포함하는 것인, 약학 조성물.
- 제3항에 있어서,상기 화합물은 6-쇼가올(6-shogaol), 6-파라돌(6-paradol ), 6-진제롤(6-gingerol), 8-진제롤(8-gingerol), 10-진제롤(10-gingerol) 및 이들의 조합으로 구성된 군으로부터 선택되는 화합물인 것인, 약학 조성물.
- 제1항에 있어서,상기 조성물을 약학적으로 허용되는 담체, 부형제 또는 희석제를 추가로 포함하는 것인, 약학 조성물.
- 제1항 내지 제5항 중 어느 한 항의 약학 조성물을, 대사성 질환이 발병되거나 발병될 우려가 있는 인간을 제외한 개체에 투여하는 단계를 포함하는 대사성 질환을 치료하는 방법.
- 디하이드로-6-진저디온(Dehydro-6-gingerdione) 또는 그의 약학적으로 허용되는 염을 포함하는 대사성 질환의 예방 또는 개선용 건강기능식품 조성물.
- 디하이드로-6-진저디온(Dehydro-6-gingerdione) 또는 그의 약학적으로 허용되는 염을 포함하는 대사성 질환의 예방 또는 개선용 사료 조성물.
