次に、本発明に係る化合物の製造法について説明する。
本工程では塩基存在下、一般式(2)で表される化合物と一般式(3)で表される化合物とを反応させて、一般式(4)で表される化合物を得る。
本工程では、一般式(4)で表される化合物とアンモニアとを反応させて、一般式(5)で表される化合物を製造できる。
本工程では一般式(5)で表される化合物とリン酸化試薬を反応させて、選択的に5’位のみをリン酸エステル化し、一般式(1a)で表される化合物を得る。
本工程では一般式(5)で表されるヌクレオシド化合物又はその塩とアミノ基の保護基の導入試薬とを反応させて、選択的にアミノ基のみを保護し、一般式(6)で表される化合物を得る。
本工程では一般式(6)で表される化合物と保護化試薬を反応させて、選択的に5’位の水酸基のみを保護し、一般式(7)で表される化合物を得る。
本工程では一般式(7)で表される化合物とリン酸化試薬を反応させて、3’位の水酸基をリン酸エステル化し、一般式(8)で表される化合物を得る。
本工程では、一般式(8)で表される化合物に脱保護化試薬を反応させて、A2とA3を脱保護し、一般式(1b)で表される化合物を得る。使用される溶媒としては、反応に関与しないものであれば、特に制限がなく、例えばメタノール、エタノール、プロパノール、イソプロパノール、テトラヒドロフラン、ジオキサン、ジエチルエーテル、N,N-ジメチルアセトアミド、水等が挙げられ、それらを単独あるいは混合して用いることができる。使用される脱保護試薬としては、メチルアミン、エチルアミン、プロピルアミン等のモノアルキルアミン、炭酸水素ナトリウム、炭酸ナトリウム、炭酸カリウム、水酸化ナトリウム、水酸化カリウム、水酸化リチウム等の無機塩基が挙げられる。この反応において、一般式(8)で表される化合物1モルに対し、前記脱保護化試薬を1~10000モル量、好ましくは1~1000モル量程度使用する。反応温度は-30~100℃、好ましくは-20~60℃であり、反応時間は0.1~100時間、好ましくは0.2~48時間である。

5-ブロモ-4-クロロ-7H-ピロロ[2,3-d]ピリミジン(2.2g)をアセトニトリル(80mL)に懸濁させ、トリス(2-(2-メトキシエトキシ)エチル)アミン(0.17mL)とパウダー状の水酸化カリウム(1.1g)を加え、50℃で5時間加熱撹拌させた。この混合液に、文献(Bioorg.Med.Chem,20,2012,5202-5214)記載の方法で得られた((2R,3R,4S,5R)-3-(ベンゾイロキシ)-5-ブロモ-4-フルオロテトラヒドロフラン-2-イル)メチル ベンゾエート(4.5g)のアセトニトリル(20mL)溶液を室温で加え、同温度で1.5時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより((2R,3R,4S,5R)-3-(ベンゾイロキシ)-5-(5-ブロモ-4-クロロ-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロテトラヒドロフラン-2-イル)メチル ベンゾエートの存在を確認した後、通常用いられる方法により精製し、当該ベンゾエート(5.1g)を得た。
本比較例の工程1で得られたベンゾエート(3.0g)を25%アンモニア水溶液(16mL)と1,4-ジオキサン(16mL)の混合液に懸濁させ、120℃で5時間撹拌した。この溶液を一部採取して、LCMSスペクトルにより標題化合物の存在を確認した後、通常用いられる方法により精製し、標記化合物(1.3g)を得た。

比較例1より得られた(2R,3R,4S,5R)-5-(4-アミノ-5-ブロモ-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-2-(ヒドロキシメチル)テトラヒドロフラン-3-オール(300mg)をリン酸トリエチル(5mL)に溶解させ、氷冷下オキシ塩化リン(0.16mL)を加え、4℃で12時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより標題化合物の存在を確認した後、通常用いられる方法により精製し、標記化合物(220mg)を得た。

比較例2に記載の化合物(200mg)をリン酸トリエチル(2.5mL)に溶解させ、氷冷下オキシ塩化リン(0.11mL)を加え、4℃で12時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより標題化合物の存在を確認した後、通常用いられる方法により精製し、標記化合物(158mg)を得た。

比較例2に記載の化合物(100mg)をリン酸トリエチル(1mL)に溶解させ、氷冷下2,6-ジメチルピリジン(0.12mL)とチオ塩化リン(0.077mL)を加え、氷冷下3時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより標題化合物の存在を確認した後、通常用いられる方法により精製し、標記化合物(99mg)を得た。

(工程1)
比較例2に記載の化合物(200mg)をピリジン(2.5mL)に溶解させ、クロロトリメチルシラン(0.32mL)を加え、室温で2時間撹拌させた。その後反応溶液に塩化ベンゾイルを(0.025mL)を加え、室温で3時間撹拌させた。その後反応液を氷冷し、水(1mL)と30%アンモニア水溶液(1mL)を加え、室温で撹拌した。この溶液を一部採取して、LCMSスペクトルによりN-(7-((2R,3S,4R,5R)-3-フルオロ-4-ヒドロキシ-5-(ヒドロキシメチル)テトラヒドロフラン-2-イル)-5-ヨード-7H-ピロロ[2,3-d]ピリミジン-4-イル)ベンズアミドの存在を確認した後、通常用いられる方法により精製し、当該ベンズアミド(105mg)を得た。
本実施例の工程1で得られたベンズアミド(105mg)をピリジン(2mL)に溶解させ、塩化ベンゾイル(0.025mL)を加え、室温で12時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより((2R,3R,4S,5R)-5-(4-ベンズアミド-5-ヨード-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-ヒドロキシテトラヒドロフラン-2-イル)メチル ベンゾエートの存在を確認した後、通常用いられる方法により精製し、当該ベンゾエート(105mg)を得た。
本実施例の工程2で得られたベンゾエート(105mg)をリン酸トリエチル(1.5mL)に溶解させ、氷冷下オキシ塩化リン(0.065mL)を加え、室温で36時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより((2R,3R,4S,5R)-5-(4-ベンズアミド-5-ヨード-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-(ホスホノオキシ)テトラヒドロフラン-2-イル)メチル ベンゾエートの存在を確認した後、通常用いられる方法により精製し、当該ベンゾエート(100mg)を得た。
本実施例の工程3で得られたベンゾエート(100mg)を40%メチルアミン水溶液(3mL)に溶解させ、室温で2時間撹拌させた。この溶液を一部採取して、LCMSスペクトルにより標題化合物の存在を確認した後、通常用いられる方法により精製し、標記化合物(21mg)を得た。
((2R,3R,4S,5R)-5-(4-アミノ-5-クロロ-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-ヒドロキシテトラヒドロフラン-2-イル)メチル ジヒドロジェン ホスフェートの合成
比較例3より得られた(2R,3R,4S,5R)-5-(4-アミノ-5-クロロ-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-2-(ヒドロキシメチル)テトラヒドロフラン-3-オール(100mg)から実施例1と同様の方法により標記化合物(88mg)を得た。
比較例1の化合物、実施例1の化合物のトリエチルアミン塩、比較例2の化合物、及び実施例2の化合物の各投与量は、それぞれ50mg/10mL/kg、62mg/10mL/kg(フリー体換算で50mg/10mL/kg)、3mg/10mL/kg、及び3.6mg/10mL/kgとなるように、0.5%HPMC水溶液で投与液を調製した。なお、実施例1の化合物のトリエチルアミン塩は、投与液を調製する際は、1モル当量のトリエチルアミンを添加することで調製した。
実施例1の化合物の2ナトリウム塩、実施例2の化合物の2ナトリウム塩、実施例3の化合物の2ナトリウム塩、実施例4の化合物の2ナトリウム塩、実施例5の化合物の2ナトリウム塩の各投与量は、それぞれ26mg/5mL/kg、12mg/5mL/kg、12mg/5mL/kg、12mg/5mL/kg、及び11mg/5mL/kgとなるように、生理食塩水に溶解し投与液を調製した。なお、実施例1~4の化合物の2ナトリウム塩については、投与液を調製する際に、2モル当量の水酸化ナトリウムを添加することで調製した。
各種ヒト腫瘍細胞株を培地中に懸濁させ、マルチウェルプレートの各ウェルに播種し、培養した。培養開始翌日に段階希釈した化合物溶液を添加し、更に3日間培養した。細胞数の計測はセルタイターグロ(プロメガ社製)を用いて、プロメガ社の推奨するプロトコールに基づき行った。以下の式より細胞生存率を算出し、50%阻害する化合物の濃度(IC50(μM))を求めた。

ヒト脳腫瘍細胞株(U-87MG)をBALB/cA Jcl-nu/nuマウスの右側胸部の皮下に移植した。腫瘍移植後に腫瘍の長径(mm)及び短径(mm)を測定し、腫瘍体積(tumor volume:TV)を算出後、各群の平均TVが均等になるように各群にマウスを割り付け、群分けを実施した日をDay0とした。

ヒト造血器腫瘍細胞株(MV-4-11)をBALB/cA Jcl-nu/nuマウスの右側胸部の皮下に移植した。腫瘍移植後に腫瘍の長径(mm)及び短径(mm)を測定し、腫瘍体積(tumor volume:TV)を算出後、各群の平均TVが均等になるように各群にマウスを割り付け、群分けを実施した日をDay0とした。

ヒト脳腫瘍細胞株(U-87MG)を培地中に懸濁させ、マルチウェルプレートの各ウェルに播種し、培養した。同日操作は培養開始翌々日に放射線照射と化合物添加をした。化合物の先添加については培養開始翌日に段階希釈した化合物溶液を添加し、培養開始翌々日に放射線を照射した。放射線の先照射については培養開始翌日に放射線を照射し、培養開始翌々日に段階希釈した化合物溶液を添加した。播種日の5日後に25%グルタルアルデヒド水溶液を用いて細胞を固定し、染色液(0.05w/v% クリスタルバイオレット in 20% メタノール)を用いて染色した.抽出液(0.1N NaH2PO4 : 100% エタノール = 1 : 1)を添加して混和後、540nmにおける吸光度を測定することで細胞数を計測した。以下の式より細胞生存率を算出した。統計処理はスチューデントのt検定を用いて行った。

結果を図8に示す。

Claims (61)
- Xは、臭素原子、又はヨウ素原子である、請求項1に記載の化合物、又はその薬学的に許容可能な塩。
- Xは、ヨウ素原子である、請求項2に記載の化合物、又はその薬学的に許容可能な塩。
- Yは、酸素原子である、請求項3に記載の化合物、又はその薬学的に許容可能な塩。
- m=0であり、n=1である、請求項4に記載の化合物、又はその薬学的に許容可能な塩。
- Xが臭素原子、又はヨウ素原子であり、Yが酸素原子であり、m=0であり、n=1である、請求項1に記載の化合物、又はその薬学的に許容可能な塩。
- 以下の化合物群:
(1) ((2R,3R,4S,5R)-5-(4-アミノ-5-ブロモ-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-ヒドロキシテトラヒドロフラン-2-イル)メチル ジヒドロジェン ホスフェート;
(2) ((2R,3R,4S,5R)-5-(4-アミノ-5-ヨード-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-ヒドロキシテトラヒドロフラン-2-イル)メチル ジヒドロジェン ホスフェート;
(3) O-(((2R,3R,4S,5R)-5-(4-アミノ-5-ヨード-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-ヒドロキシテトラヒドロフラン-2-イル)メチル) O,O-ジヒドロジェン ホスホロチオエート;
(4) (2R,3R,4S,5R)-5-(4-アミノ-5-ヨード-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-2-(ヒドロキシメチル)テトラヒドロフラン-3-イル ジヒドロジェン ホスフェート;及び
(5) (2R,3R,4S,5R)-5-(4-アミノ-5-クロロ-7H-ピロロ[2,3-d]ピリミジン-7-イル)-4-フルオロ-3-ヒドロキシテトラヒドロフラン-2-イル)メチル ジヒドロジェン ホスフェート
から選択される化合物、又はその薬学的に許容可能な塩。 - 請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩を有効成分として含む抗腫瘍剤。
- 請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩及び薬学的に許容可能な担体を含有する医薬組成物。
- 経口剤又は注射剤として製剤化されたものである、請求項8に記載の抗腫瘍剤または請求項9に記載の医薬組成物。
- 腫瘍の予防及び/又は治療方法であって、請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩を投与することを特徴とする方法。
- 腫瘍の予防及び/又は治療方法であって、それを必要とする対象に、経口剤又は注射剤として製剤化された請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩を投与することを含む、方法。
- 医薬組成物として使用するための請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩。
- 腫瘍を予防及び/又は治療するための、請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩。
- 経口剤又は注射剤として製剤化されたものである腫瘍の予防及び/又は治療に使用するための請求項1~7のいずれかに記載の化合物又はその薬学的に許容可能な塩。

- 前記腫瘍が、頭頚部癌(口腔癌、咽頭癌、喉頭癌、鼻腔癌、副鼻腔癌、唾液腺癌、甲状腺癌等)、消化器癌(食道癌、胃癌、十二指腸癌、肝臓癌、胆道癌(胆嚢・胆管癌等)、膵臓癌、大腸癌(結腸癌、直腸癌等)等)、肺癌(非小細胞肺癌、小細胞肺癌、中皮腫等)、乳癌、生殖器癌(卵巣癌、子宮癌(子宮頚癌、子宮体癌等)等)、泌尿器癌(腎癌、膀胱癌、前立腺癌、精巣腫瘍等)、造血器腫瘍(白血病、悪性リンパ腫、多発性骨髄腫等)、骨・軟部腫瘍、皮膚癌、及び脳腫瘍からなる群から選択される、請求項8に記載の抗腫瘍剤、請求項11又は12に記載の腫瘍の予防及び/又は治療方法、請求項13~15のいずれかに記載の化合物又はその薬学的に許容可能な塩、又は請求項16~18のいずれかに記載の使用。
- 前記腫瘍が、脳腫瘍である、請求項19に記載の抗腫瘍剤、腫瘍の予防及び/又は治療方法、化合物又はその薬学的に許容可能な塩、又は使用。
- 請求項21に記載の化合物又はその薬学的に許容可能な塩を有効成分として含む抗腫瘍剤。
- 請求項21に記載の化合物又はその薬学的に許容可能な塩と、薬学的に許容可能な担体とを含有する医薬組成物。
- 経口剤または注射剤として製剤化されたものである、請求項22に記載の抗腫瘍剤または請求項23に記載の医薬組成物。
- 請求項21に記載の化合物又はその薬学的に許容可能な塩を投与することを特徴とする腫瘍の予防及び/又は治療方法。

- アルキル化剤と併用することを特徴とする、請求項1~7及び21のいずれかに記載の化合物又はその薬学的に許容可能な塩からなる抗腫瘍剤、又は医薬組成物。
- アルキル化剤の抗腫瘍効果を増強するための、請求項1~7及び21のいずれかに記載の化合物又はその薬学的に許容可能な塩を含む抗腫瘍効果増強剤。
- アルキル化剤の投与を受けた癌患者を治療するための、請求項1~7及び21のいずれかに記載の化合物又はその薬学的に許容可能な塩を含む抗腫瘍剤。
