Abstract
The present invention relates to a cell membrane penetrating domain derived from a human GPATCH4 protein and, more particularly, to a human GPATCH4 protein-derived cell membrane penetrating domain (GPATCH4P1 cell membrane penetrating domain) or a cell penetrating peptide, and an intracellular delivery system comprising same. By using a cell membrane penetrating domain derived from a human GPATCH4 protein, of the present invention, cargo materials such as compounds, biomolecules, and various polymer materials can be delivered into cells. A GPATCH4P1 cell membrane penetrating domain of the present invention exhibits higher cell penetration efficiency than conventional cell penetrating peptides, and is derived from a human protein so that side effects and immune responses caused by peptides derived from foreign proteins can be avoided, thereby being effectively usable for a method for effectively delivering, into cells, compounds, biomolecules and various polymer materials to be applied to the human body.

EGFP, TAT-EGFP 그리고 GPATCH4P1-EGFP 융합단백질을 각각 최종 농도 0.5, 1, 2, 5, 10μM로 Jurkat 세포에 처리하였으며 앞선 실험과 동일한 과정을 통해 도입 효율을 확인하였다(도 7). 이를 통해 GPATCH4P1-EGFP 융합단백질이 농도의존적으로 세포 내로 도입됨을 확인하였으며 TAT-EGFP 융합단백질과 비교하여 모든 농도에서 더 높은 전달 효율을 보이는 것을 확인하였다. Through this, it was confirmed that the GPATCH4P1-EGFP fusion protein was introduced into the cell in a concentration-dependent manner, and it was confirmed that it showed a higher delivery efficiency at all concentrations compared to the TAT-EGFP fusion protein.
다음으로 GPATCH4P1-EGFP 융합단백질의 시간 별 도입 효율을 확인하기 위하여 EGFP, TAT-EGFP 융합단백질을 이용하여 비교 실험을 진행하였다. EGFP, TAT-EGFP 그리고 GPATCH4P1-EGFP 융합단백질을 각각 최종 농도 5μM로 Jurkat세포에 처리한 후 각각 0.5, 1, 2, 4, 6시간 동안 배양하였으며 상기 농도별 도입 효율 실험과 동일한 과정을 통해 도입 효율을 확인하였다(도 8). 이를 통해 GPATCH4P1-EGFP 융합단백질이 시간 의존적으로 세포 내로 도입됨을 확인하였으며 TAT-EGFP 융합단백질과 비교하여 모든 시간에서 더 높은 전달 효율을 보이는 것을 확인하였다.Next, in order to confirm the efficiency of introduction of the GPATCH4P1-EGFP fusion protein by time, a comparative experiment was conducted using EGFP and TAT-EGFP fusion proteins. <실시예 4> GPATCH4P1-EGFP 융합 단백질의 세포 내 전달 양상 확인<Example 4> Confirmation of intracellular delivery pattern of GPATCH4P1-EGFP fusion protein
세포 내로 도입된 GPATCH4P1-EGFP 융합 단백질을 공초점현미경으로 확인하기 위하여 먼저 HeLa 세포를 2 X 104/well로 24 well cell culture slide에 분주한 후 10% Fetal Bovine Serum과 1% penicillin/streptomycin이 포함된 DMEM 배지 500μl 에 부유시켜 37°C로 유지되는 5% CO2배양기에서 18시간 배양하였다. 이 후 EGFP 그리고 GPATCH4P1-EGFP 융합단백질을 각각 최종 농도 10μM로 세포에 처리한 후 1시간 동안 배양하였다. 이 후 배양액을 제거한 후 PBS 용액으로 3회 세척 후 4% PFA 용액을 상온에서 30분간 처리하여 고정하였다. PFA 용액을 제거한 후 다시 PBS 용액으로 3회 세척 한 뒤 mounting medium 을 처리하고 커버 슬라이드를 덮어 공초점현미경으로 관찰하였다(도 9). 이를 통해 GPATCH4P1-EGFP 융합단백질이 HeLa세포 내로 효과적으로 도입되었음을 확인하였다.To confirm the GPATCH4P1-EGFP fusion protein introduced into the cell by confocal microscopy, first HeLa cells are dispensed into a 24 well cell culture slide with 2 X 10 4 /well, and then contain 10% Fetal Bovine Serum and 1% penicillin/streptomycin The cells were suspended in 500 μl of DMEM medium and cultured for 18 hours in a 5% CO 2 incubator maintained at 37°C. Through this, it was confirmed that the GPATCH4P1-EGFP fusion protein was effectively introduced into HeLa cells.
<실시예 5> GPATCH4P1 mutant-EGFP 융합 단백질의 세포 내 전달 효율 확인<Example 5> Confirmation of intracellular delivery efficiency of GPATCH4P1 mutant-EGFP fusion protein
GPATCH4P1 변이체의 세포 내 전달 효율을 연구하기 위하여 GPATCH4P1의 9번째 아미노산인 글루타민을 다른 아미노산으로 치환한 GPATCH4P1 mutant 및 N-말단과 C- 말단의 아미노산을 각각 하나씩 제거한 GPATCH4P1 mutant 를 설계하였다. 이 후 GPATCH4P1 mutant-EGFP 융합 단백질을 발현하는 플라스미드 벡터를 제작한 후 단백질을 발현시켜 정제하였다. 세포 실험을 위하여 Jurkat 세포를 1.5 X 106/well로 24 well plate에 분주한 후 10% Fetal Bovine Serum과 1% penicillin/streptomycin 이 포함된 RPMI 1640 배지 2ml에 부유시켜 CPP-EGFP 융합단백질을 최종 농도 5μM로 처리 후 37°C로 유지되는 5% CO2 배양기에서 2시간 동안 배양하였다. 이 후 세포가 포함된 배지를 5ml 튜브에 옮긴 후 원심분리하여 상등액을 제거하고 PBS 용액으로 세척하는 것을 3회 반복하였다. 이 후 Jurkat 세포를 650μl의 PBS에 부유시킨 후 유세포분석기를 통해 세포 내로 도입된 GPATCH4P1 mutant-EGFP 융합단백질들의 형광을 측정하여 세포 내 도입 효율을 확인하였다(도 10). 이를 통해 GPATCH4P1 mutant들이 변이를 통해 도입 효율의 변화를 나타내지만 세포 투과 능력이 유지됨을 확인하였다.In order to study the efficiency of intracellular delivery of the GPATCH4P1 variant, the GPATCH4P1 mutant, which replaced glutamine, the ninth amino acid of GPATCH4P1 with another amino acid, and the GPATCH4P1 mutant, each of which was removed by one of the N-terminal and C-terminal amino acids, were designed. After treatment with 5 μM, the cells were cultured in a 5% CO 2 incubator maintained at 37° C. for 2 hours. Thereafter, the medium containing the cells was transferred to a 5 ml tube, followed by centrifugation to remove the supernatant and washing three times with PBS solution. Thereafter, Jurkat cells were suspended in PBS of 650 μl, and fluorescence of GPATCH4P1 mutant-EGFP fusion proteins introduced into the cells was measured through a flow cytometer to confirm the efficiency of cell introduction (FIG. 10). Through this, it was confirmed that GPATCH4P1 mutants exhibited a change in introduction efficiency through mutation, but maintained cell permeability.
