Abstract

Die Erfindung betrifft ein Verfahren und ein Kit zur Detektion von Analyten, insbesondere von niedermolekularen Analyten. Erfindungsgemäß wird dabei ein einfaches Verfahren zur Detektion von niedermolekularen Analyten bereitgestellt, bei dem die zu detektierenden Analyten als Kompetitoren für immobilisierte Enzyme agieren. Durch die Inhibierung kann über den reduzierten Substratumsatz auf die Analytkonzentration geschlossen werden. Insbesondere wird erfindungsgemäß ein Verfahren zur Detektion von Glyphosat bereitgestellt.

Description

Verfahren, Oberfläche und Kit zur Detektion von Analyten
Die Erfindung betrifft ein Verfahren zur Detektion von Analyten. Insbesondere betrifft die Erfindung ein Verfahren zur Detektion von niedermolekularen Analyten.
Der Nachweis von niedermolekularen Analyten gestaltet sich aktuell aufwändig. Insbesondere der Nachweis von Pflanzenschutzmitteln in Trinkwasser und Bodenproben und deren gesundheitliche Auswirkungen sind aktuell von hohem Interesse. Problematisch erscheint dabei vor allem die Verunreinigung von Nahrungsmitteln inklusive Trinkwasser mit Glyphosat.
Die Detektion von Verunreinigungen mit Pflanzenschutzmitteln in Lebensmitteln erfolgt nach der Multimethode DFG S19, wobei zur Probenvorbereitung verschiedene Extraktionsschritte erfolgen und nachfolgend die Analyse mittels LC-MS oder GC-MS erfolgt.
Im Stand der Technik sind zudem verschiedene Verfahren zur Bestimmung von Glyphosat bekannt.
So offenbart die CN102207495A ein Verfahren zur Bestimmung des Glyphosat-Gehalts in Bodenproben mittels HPLC.
Auch die W020081 18899 A1 offenbart ein Verfahren zum Nachweis von Glyphosat mittels LC/GC gekoppeltem MS. Alternativ werden auch Nachweismethoden mittels Biolumineszenz (WO2010104861 A1 ) und ELISA (W02000014538A1 ) beschrieben.
Die WO2018057647 A1 beschreibt einen Assay für einen Biochip, bei dem Pestizide auf einer Sensorplattform detektiert werden können. Der Nachweis basiert dabei auf der Amplifikation von korrespondierenden Nukleinsäuren, was zu deutlich langen Analysezeiten führt.
US 2005 / 0 1 18 665 A1 offenbart ein Verfahren zur Bestimmung einer enzymatischen Reaktion, wobei mindestens ein Protein und mindestens eine Substanz auf einem festen Träger in einem Abstand, ausreichend für eine enzymatische Reaktion, immobilisiert werden und unter Bedingungen für eine enzymatische Reaktion inkubiert werden. Bevorzugt ist die Substanz ein bekanntes Substrat für eine untersuchte enzymatische Reaktion und das Protein wird auf eine enzymatische Aktivität untersucht. US 2006 / 0 073 490 A1 offenbart eine Enzym-basierte Vorrichtung zur Messung von Umweltproben. Die Vorrichtung umfasst ein immobilisiertes Enzym, bevorzugt eingeschlossen in eine Polymermatrix; ein Element, welches Umweltproben zu dem immobilisierten Enzym befördert, wobei die Umweltproben optional ein enzym-inhibierendes Zielmolekül enthalten, ein Element, welches ein Enzymsubstrat und optional einen pH-sensitiven Farbstoff zu dem immobilisierten Enzym befördert, und mindestens einen Sensor, der die Enzymaktivität bestimmt.
WO 2003 / 008 453 A1 offenbart ein Verfahren zur Immobilisierung von Polypeptiden durch die Herstellung eines Fusionspolypeptids mit einem Adhäsionspolypeptid, insbesondere mit Hydrophobin, auf festen Oberflächen, bevorzugt auf hydrophoben Oberflächen. WO 03/008453 A1 beschreibt die Verwendung der Hydrophobinfusionspolypeptide unter anderem für Biosensoroberflächen.
DE 10 201 1 089 241 B3 offenbart ein Verfahren zur Beschichtung eines Substrats mit einer Hydrophobinbilage und ein Substrat mit einer Hydrophobinbilagenbeschichtung. Weiterhin offenbart DE 10 201 1 089 241 B3 ein mit einem Hydrophobinfusionsprotein beschichtetes Substrat.
US 2006 / 0 253 923 A1 offenbart ein Verfahren zur Herstellung Herbizid-toleranter, insbesondere Glyphosat-toleranter, Kulturpflanzen durch DNA-Shuffling, insbesondere einer Nukleinsäure kodierend für eine 5-Enolpyruvylshikimat-3-phosphatsynthase; eine Bibliothek mit rekombinanten Nukleinsäure erhalten durch ein Verfahren zur Herstellung Herbizid-selektiver Kulturpflanzen durch DNA-Shuffling mit einer Nukleinsäure kodierend für ein Protein mit Herbizidtoleranz. Weiterhin beschreibt US 2006 / 0 253 923 A1 den Malachitgrün-Nachweis der EPSPS-Aktivität.
Der Nachweis entsprechender niedermolekularer Analyten ist bisher experimentell aufwändig und nicht geeignet um Vor-Ort-Analyse zu ermöglichen.
Es besteht daher ein Bedürfnis eine Nachweismethode anzugeben, die einen einfachen Nachweis niedermolekularer Analyten vor Ort ermöglicht.
Die Aufgabe der Erfindung ist es daher, ein Verfahren zur Detektion von Analyten anzugeben, das die Nachteile im Stand der Technik überwinden kann.
Die Aufgabe wird durch das erfindungsgemäße Verfahren gelöst. Vorteilhafte Ausgestaltungen sind in den abhängigen Ansprüchen angegeben. Erfindungsgemäß wird ein Verfahren zur Detektion von Analyten in einer Probe vorgeschlagen, umfassend die Schritte:
Bereitstellen einer Oberfläche mit einem immobilisierten Analytbindungspartner, wobei der Analytbindungspartner eine Affinität zur Interaktion mit einem zu detektierenden Analyten aufweist,
Kontaktieren des Analytbindungspartners mit einer Probe enthaltend den Analyten, wobei der Analyt mit dem Analytbindungspartner interagiert,
wobei der Analytbindungspartner durch Interaktion mit dem Analyten gehemmt wird, Kontaktieren des Analytbindungspartners mit einem Substrat, wobei das Substrat durch den Analytbindungspartner zu einem Produkt umgesetzt wird,
Quantifizierung des Produkts und
Korrelation des quantifizierten Produkts mit einer Referenzprobe,
wobei der Analyt Glyphosat ist,
wobei der Analytbindungspartner eine Proteindomäne umfasst, aufweisend die enzymatische Aktivität des Enzyms 5-Enolpyruvylshikimat-3-phosphat-Synthase.
Unter niedermolekularen Analyten werden Stoffe oder Moleküle mit einer Molmasse von <800 g/mol verstanden.
In Ausführungsformen der Erfindung erfolgt eine parallele Kontaktierung des Analytbindungspartners mit dem Analyten und dem Substrat.
Erfindungsgemäß ist der Analytbindungspartner ein Protein oder Peptid aufweisend eine enzymatische Aktivität.
Unter einem Substrat wird im Sinne der Erfindung zumindest ein Stoff verstanden, der von einer Proteindomäne des Analytbindungspartners, welche eine enzymatische Aktivität zur Umsetzung des Substrats zu dem Produkt aufweist, umgesetzt wird. In Ausführungsformen der Erfindung umfasst das Substrat auch zwei oder mehr Stoffe, welche von der Proteindomäne des Analytbindungspartners, welche eine enzymatische Aktivität zur Umsetzung des Substrats zu dem Produkt aufweist, umgesetzt werden. In Ausführungsformen der Erfindung umfasst das Substrat Shikimat-3-phosphat und Phosphoenolpyruvat. Erfindungsgemäß weist der Analytbindungspartner die enzymatische Aktivität des Enzyms 5- Enolpyruvylshikimat-3-phosphat-Synthase (EPSPS, EC 2.5.1.19) auf. Die EPSPS katalysiert dabei die Umsetzung von Shikimat-3-phosphat und Phosphoenolpyruvat zu 5- Enolpyruvylshikimat-3-phosphat, wobei anorganisches Phosphat freigesetzt wird. Die Verwendung der EPSPS kann dabei vorteilhaft für den Nachweis von Glyphosat genutzt werden, da dieses die EPSPS inhibiert und somit über die verminderte enzymatische Aktivität der Nachweis für das Vorhandensein von Glyphosat in der zu untersuchenden Probe erfolgt. In Ausführungsformen der Erfindung weist der Analytbindungspartner die enzymatische Aktivität des Enzyms 5-Enolpyruvylshikimat-3-phosphat-Synthase aus E. coli (https://www.uniprot.org/uniprot/P0A6D3) auf. Bevorzugt umfasst der Analytbindungspartner die SEQ ID No. 1.
In Ausführungsformen der Erfindung ist der Analytbindungspartner als Fusionsprotein ausgebildet. Dabei weist das Fusionsprotein beispielsweise verschiedene Proteindomänen auf, welche unterschiedliche Funktionalitäten aufweisen.
In Ausführungsformen der Erfindung umfasst der Analytbindungspartner eine Proteindomäne, aufweisend eine enzymatische Aktivität zur Umsetzung des Substrats zu dem Produkt. Durch die Proteindomäne mit enzymatischer Aktivität kann das Substrat zu dem Produkt umgesetzt werden, wobei das Produkt oder ein Nebenprodukt der katalysierten Reaktion nachfolgend detektiert werden kann.
In Ausführungsformen der Erfindung umfasst der Analytbindungspartner eine Proteindomäne, ausgewählt aus Hydrophobinen, ECM-Proteinen, S-Layer-Proteinen, Peptidlinkern, und Protein- Tags. Dadurch wird eine gerichtete Immobilisierung an die Oberfläche ermöglicht, wobei zumindest eine Proteindomäne die Anbindung vermittelt und eine weitere Proteindomäne eine weitere Funktionalität aufweist. Bevorzugt weist der Analytbindungspartner eine Hydrophobin- Domäne auf. In Ausführungsformen der Erfindung weist der Analytbindungspartner eine Hydrophobin-Domäne und eine Proteindomäne aufweisend eine enzymatische Aktivität zur Umsetzung des Substrats zu dem Produkt auf.
In Ausführungsformen der Erfindung weist der Analytbindungspartner eine Hydrophobin-Domäne Ccg2 aus Neurospora (N.) crassa auf. Bevorzugt weist der Analytbindungspartner die SEQ. ID. No. 5 auf. In Ausführungsformen der Erfindung ist der Analytbindungspartner ein Fusionsprotein aufweisend eine Hydrophobin-Domäne und eine 5-Enolpyruvylshikimat-3-phosphat-Synthase- Domäne. Dabei ermöglicht die Hydrophobin-Domäne die Immobilisierung des Analytbindungspartners auf der Oberfläche. In Ausführungsformen der Erfindung weist das Fusionsprotein die SEQ ID No. 6 auf.
In Ausführungsformen der Erfindung wird der Analytbindungspartner in einer Mischung mit Hydrophobinen auf der Oberfläche immobilisiert, wobei die Mischung des Analytbindungspartners mit dem Hydrophobin im Bereich von 1 :2 bis 1 :10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5 liegt. Die angegebenen Mischungsverhältnisse beziehen sich im Sinne der vorliegenden Erfindung auf die Stoffmengenkonzentrationen.
In Ausführungsformen der Erfindung erfolgt zur Immobilisierung des Analytbindungspartners eine Mischung aus dem Fusionsprotein aufweisend eine Hydrophobin-Domäne und eine 5- Enolpyruvylshikimat-3-phosphat-Synthase-Domäne (SEQ ID No. 6) und der Hydrophobin- Domäne Ccg2 aus Neurospora (N.) crassa (SEQ ID No. 5) im Verhältnis 1 :2 bis 1 : 10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5, ganz besonders bevorzugt 1 :5.
Erfindungsgemäß wird der Analytbindungspartner durch Interaktion mit dem Analyten gehemmt. Dabei weist der Analytbindungspartner eine enzymatische Aktivität auf, welche durch die Interaktion mit dem Analyten gehemmt wird. Die Hemmung kann dabei kompetitiv oder nicht kompetitiv erfolgen. Durch Interaktion mit dem Analyten wird somit die Umsetzung des Substrats zum Produkt zumindest vermindert oder verlangsamt.
Erfindungsgemäß ist der Analyt Glyphosat. Glyphosat (N-(Phosphonomethyl)glycin) ist ein nicht selektives Blattherbizid (Breitband- oder Totalherbizid) mit systemischer Wirkung, das über jegliche grünen Pflanzenteile aufgenommen wird. Glyphosat wird in der Landwirtschaft gegen einkeim- und zweikeimblättrige Unkräuter im Acker-, Wein- und Obstbau, beim Anbau von Zierpflanzen, auf Wiesen, Weiden und Rasenflächen sowie im Forst verwendet. Die Blätter nehmen Glyphosat durch Diffusion auf und in der Pflanze wird Glyphosat über das Phloem verteilt. Glyphosat wirkt über die Blockade des Enzyms 5-Enolpyruvylshikimat-3-phosphat- Synthase (EPSPS), das zur Synthese der aromatischen Aminosäuren Phenylalanin, Tryptophan und Tyrosin über den Shikimatweg in Pflanzen, wie auch in den meisten Mikroorganismen, benötigt wird. Durch die Ähnlichkeit des Glyphosats mit Phosphoenolpyruvat kann es dessen Bindestelle im Enzym besetzten, wodurch das Enzym 5-Enolpyruvylshikimat-3-phosphat- Synthase (EPSPS) gehemmt wird. In Ausführungsformen der Erfindung erfolgt die Quantifizierung des Produkts durch optische Detektion. Durch die Möglichkeit der optischen Detektion ist eine einfache Quantifizierung des Analyten möglich. Bevorzugt erfolgt die optische Detektion über einen Farbstoff, der eine einfache optische Detektion erlaubt.
In Ausführungsformen der Erfindung erfolgt die Quantifizierung des Produkts photometrisch. Dadurch ist eine einfache Quantifizierung möglich. Vorzugsweise erfolgt die Quantifizierung durch Korrelation mit einem Standard oder einer Probe ohne Analyten.
In Ausführungsformen der Erfindung erfolgt die Quantifizierung des Produkts mit einem Malachitgrün-Assay. Dabei wird der Gehalt an Phosphat mittels Malachitgrün-Assay ermittelt. So wird bei der Hemmung der 5-Enolpyruvylshikimat-3-phosphat-Synthase (EPSPS) durch Glyphosat die Reaktion mit Phosphoenolpyruvat gehemmt, sodass eine Umsetzung zu 5- Enolpyruvylshikimat-3-phosphat und Phosphat nicht oder nur verlangsamt erfolgt. Durch den Nachweis mittels Malachitgrün-Assay kann das in der Reaktion gebildete anorganische Phosphat bestimmt werden und somit als Maß für die Inhibierung der EPSPS genutzt werden.
In Ausführungsformen der Erfindung ist die Oberfläche an der die Immobilisierung des Analytbindungspartners erfolgt als Partikel ausgebildet. Bevorzugt ist die Oberfläche als Hydrogelpartikel ausgebildet. Dadurch kann der Nachweis des Analyten in Lösung erfolgen. Dies ist beispielsweise vorteilhaft, wenn eine photometrische Bestimmung erfolgen soll.
Gegenstand der Erfindung ist auch eine Oberfläche aufweisend einen Analytbindungspartner, wobei der Analytbindungspartner ein Fusionsprotein aufweisend eine Hydrophobin-Domäne und eine 5-Enolpyruvylshikimat-3-phosphat-Synthase-Domäne ist.
In Ausführungsformen der Erfindung ist die Oberfläche im Wesentlichen planar ausgebildet. Beispielsweise kann die Oberfläche ein Glas- oder Kunststoffformkörper sein z. B. ein Objektträger, Deckgläschen, Siliziumwafer oder ähnliches. In Ausführungsformen der Erfindung ist die Oberfläche als 96-Well-Platte ausgebildet. In Ausführungsformen der Erfindung ist die Oberfläche als Partikel ausgebildet.
In Ausführungsformen der Erfindung ist der Analytbindungspartner als Fusionsprotein ausgebildet. Dabei weist das Fusionsprotein beispielsweise verschiedene Proteindomänen auf, welche unterschiedliche Funktionalitäten aufweisen. In Ausführungsformen der Erfindung umfasst der Analytbindungspartner eine Proteindomäne, aufweisend eine enzymatische Aktivität zur Umsetzung des Substrats zu dem Produkt. Durch die Proteindomäne mit enzymatischer Aktivität kann das Substrat zu dem Produkt umgesetzt werden, wobei das Produkt oder ein Nebenprodukt der katalysierten Reaktion nachfolgend detektiert werden kann.
In Ausführungsformen der Erfindung umfasst der Analytbindungspartner eine Proteindomäne, ausgewählt aus Hydrophobinen, ECM-Proteinen, S-Layer-Proteinen, Peptidlinkern und Protein- Tags. Dadurch wird eine gerichtete Immobilisierung an die Oberfläche ermöglicht, wobei zumindest eine Proteindomäne die Anbindung vermittelt und eine weitere Proteindomäne eine weitere Funktionalität aufweist. In Ausführungsformen der Erfindung weist der Analytbindungspartner eine Hydrophobin-Domäne Ccg2 aus Neurospora (N.) crassa auf. Bevorzugt weist der Analytbindungspartner die SEQ. ID. No. 5 auf.
In Ausführungsformen der Erfindung ist der Analytbindungspartner ein Fusionsprotein aufweisend eine Hydrophobin-Domäne und zumindest eine Proteindomäne, aufweisend eine enzymatische Aktivität zur Umsetzung des Substrats zu dem Produkt.
In Ausführungsformen der Erfindung ist der Analytbindungspartner ein Fusionsprotein aufweisend eine Hydrophobin-Domäne und eine 5-Enolpyruvylshikimat-3-phosphat-Synthase- Domäne. Dabei ermöglicht die Hydrophobin-Domäne die Immobilisierung des Analytbindungspartners auf der Oberfläche. In Ausführungsformen der Erfindung weist das Fusionsprotein die SEQ ID No. 6 auf.
In Ausführungsformen der Erfindung ist der Analytbindungspartner in einer Mischung mit Hydrophobinen auf der Oberfläche immobilisiert, wobei die Mischung des Analytbindungspartners mit dem Hydrophobin im Bereich von 1 :2 bis 1 :10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5 liegt.
In Ausführungsformen der Erfindung erfolgt zur Immobilisierung des Analytbindungspartners eine Mischung aus dem Fusionsprotein aufweisend eine Hydrophobin-Domäne und eine 5- Enolpyruvylshikimat-3-phosphat-Synthase-Domäne (SEQ ID No. 6) und der Hydrophobin- Domäne Ccg2 aus Neurospora (N.) crassa (SEQ ID No. 5) im Verhältnis 1 :2 bis 1 :10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5, ganz besonders bevorzugt 1 :5. Durch Variation des Verhältnisses von Fusionsprotein und Hydrophobin auf der Oberfläche kann zusätzlich die Sensitivität gegenüber Glyphosat sehr gezielt eingestellt werden und ermöglicht damit einen Nachweis von Glyphosat über einen weiten Konzentrationsbereich.
Gegenstand der Erfindung ist auch ein Kit zur Detektion von Analyten umfassend einen Analytbindungspartner aufweisend eine enzymatische Aktivität zur Umsetzung eines Substrats, wobei der Analytbindungspartner ein Fusionsprotein aufweisend eine Hydrophobin-Domäne und eine 5-Enolpyruvylshikimat-3-phosphat-Synthase-Domäne ist, sowie ein Hydrophobin. Die im Kit enthaltenen Analytbindungspartner sowie das Hydrophobin werden vorteilhaft zur Immobilisierung im Verhältnis 1 :2 bis 1 :10, bevorzugt 1 :3 bis 1 :8, besonders bevorzugt im Verhältnis 1 :5 gemischt und auf einer Oberfläche immobilisiert. Dabei stabilisiert das Hydrophobin die Oberfläche während der Analytbindungspartner zur Interaktion mit dem Analyten ausgebildet ist.
In Ausführungsformen der Erfindung umfasst das Kit ein Fusionsprotein mit der SEQ ID No. 6 sowie ein Hydrophobin mit der SEQ ID No. 5.
Gegenstand der Erfindung ist auch die Verwendung des erfindungsgemäßen Verfahrens und des erfindungsgemäßen Kits zur Detektion von Analyten, insbesondere Glyphosat. In Ausführungsformen erfolgt die Detektion von Glyphosat in Gewässer-, Lebensmittel-, Boden-, Trink- und Abwasserproben, bevorzugt in wässrigen Lösungen.
Zur Realisierung der Erfindung ist es auch zweckmäßig, die vorbeschriebenen Ausführungsformen und Merkmale der Ansprüche zu kombinieren.
Weitere Merkmale und Vorteile der vorliegenden Erfindung ergeben sich aus den nachfolgenden schematischen Zeichnungen und Ausführungsbeispielen, anhand derer die Erfindung beispielhaft näher erläutert werden soll, ohne die Erfindung auf diese zu beschränken.

Das Prinzip der Nachweismethode ist in Fig. 1 gezeigt. Es beruht auf dem Malachitgrün-Assay, welcher zum allgemeinen Nachweis von anorganischem Phosphat (Pi) eingesetzt wird (Baykov et al. 1988). Aus der zu analysierenden Lösung muss zunächst Phosphat mit Hilfe einer Phosphat-bindenden Substanz entfernt werden. Anschließend wird der Analyt, gemeinsam mit den benötigten Substraten Shikimat-3-phosphat (S3P) und Phosphoenolpyruvat (PEP) auf eine vorbereitete Oberfläche, beispielsweise eine 96-well-Platte (beschichtet mit Mischung aus Hydrophobin Ccg2 (SEQ ID No. 4) und dem Fusionsprotein Ccg2_GS_EcEPSPS (SEQ ID No. 6)) gegeben und für 30-45 Minuten inkubiert. Anschließend wird die Malachitgrün-Arbeitslösung (Mischung aus Malachitgrün, Ammoniummolybdat und Schwefelsäure (Baykov et al. 1988)) zur Detektion des bei der Reaktion gebildeten Pi hinzugefügt und für20 Minuten im Dunkeln inkubiert. Durch Messen der Absorption bei 630 nm und Vergleich mit einer Kontrolle/Standard kann dann die in der Analytlösung vorhandene Glyphosatmenge bestimmt werden.
Zum Einstellen eines guten Signal-Rauschverhältnisses des Assays wurden in Fig. 2 verschiedene Belegungsdichten auf Polystyrol-Oberflächen getestet. Hierfür wurde insbesondere die Hydrophobinkonzentration variiert. Mit steigender Hydrophobinkonzentration ist eine höhere Enzymfunktion zu verzeichnen. Ab einer Hydrophobinkonzentration von 5 mM ist keine weitere Aktivitätssteigerung mehr zu beobachten. Die Hemmung des Enzyms mit 0,5 mM Glyphosat ist bei diesem Verhältnis detektierbar.
Als Machbarkeitsbeweis wurden in Fig. 3 verschiedene Glyphosatkonzentrationen zu einer funktionalisierten Glasoberfläche (Beschichtung mit 1 mM Ccg2_GS_EcEPSPS (SEQ ID No. 6): 5 mM Ccg2 (SEQ ID No. 5)) gegeben und der Malachitgrün-Assay durchgeführt. Die Hemmung der Enzymaktivität bei verschiedenen Glyphosatkonzentrationen wird deutlich.
Im Vergleich zu Fig. 3, wurde für Fig. 4 die Glyphosat-abhängige Hemmung von 1 mM EcEPSPS (SEQ ID No. 4) in Lösung getestet. Die Hemmung des Enzyms wird hier nicht sichtbar.
Ebenfalls als Machbarkeitsnachweis wurden für Fig. 5 zwei verschiedene Phosphat-bindende Substanzen zu Lösungen mit verschiedenen Phosphatkonzentrationen gegeben und diese mindestens eine Stunde inkubiert, anschließend abzentrifugiert und mit Hilfe des Malachitgrün- Assays die verbleibende Phosphatmenge bestimmt.
Der Malachitgrün-Assay ist ein bekanntes Verfahren zum Nachweis von anorganischem Phosphat. Er beruht auf der Komplexierung von anorganischem Phosphat (Pi) mit Molybdat. In Anwesenheit von Malachitgrün führt diese Komplexierung zu einer Verschiebung des Absorptionsmaximums nach ca. 630 nm. Dementsprechend deutet eine hohe Absorption bei 630 nm auf eine hohe Konzentration an Pi hin. Dieses entsteht bei der Reaktion der EPSPS mit ihren Substraten Phosphoenolpyruvat (PEP) und Shikimat-3-phosphat (S3P). Dementsprechend kann der Malachitgrün-Assay für den Aktivitätsnachweis der EPSPS genutzt werden. Glyphosat führt zur Hemmung der EPSPS, da es mit PEP um die Bindung im aktiven Enzymzentrum konkurriert. Je mehr Glyphosat im umgebenden Medium ist, desto geringer ist die EPSPS- Aktivität.
In einer Variante des Nachweisverfahrens kann das EcEPSPS alternativ zu einer Chip- Oberfläche funktionserhaltend an suspendierte Hydrogelmikropartikel (Herstellung über Emulsions- und radikalischer Fällungspolymerisation von Polyethylenglykol-Diacrylamid mit anschließender radikalischer Pfropfung von Acrylsäuremonomeren zur Einführung der Carboxylgruppen, mittlerer Radius 20 pm) gekoppelt werden. Damit können größere Oberflächen mit immobilisierter EcEPSPS in kleineren Analytlösungen erzeugt und somit höhere Sensitivitäten erreicht werden.
Ausführungsbeispiel 1 : Nachweis von Glyphosat mittels funktionalisierter Oberfläche und Malachitgrün-Assay
Für die Erzeugung einer funktionalisierten Oberfläche wurden Fusionsproteine aus dem Hydrophobin Ccg2 aus Neurospora (N.) crassa und der 5-Enolpyruvylshikimat-3- phosphatsynthase aus Escherichia (E.) coli (EcEPSPS) benötigt. Hierzu wurden die kodierenden Bereiche der jeweiligen Gene, verbunden über die Sequenz für einen flexiblen Glycin-Serin- Linker, in den Expressionsvektor pET28b (Novagen, Deutschland) übertragen. Hierbei liegt die Sequenz des Hydrophobins (SEQ ID No. 2) 5'-seitig und die der EcEPSPS (SEQ ID No. 1 ) 3'- seitig von der Linkersequenz. Zusätzlich befindet sich 5'-seitig von der Sequenz für das Fusionsprotein die Sequenz für einen (His)6-Tag, welcher für die Detektion und Reinigung des Fusionsproteins erforderlich ist. Weiterhin wurde für die Oberfläche das Hydrophobin ohne EcEPSPS benötigt. Die Gensequenz (SEQ ID No. 2) wurde dementsprechend ohne den Linker und die EcEPSPS-Sequenz in den Vektor pET28b übertragen. Die auf diese Weise modifizierten Vektoren wurden nach vollständiger Sequenzierung der eingebrachten Sequenzen, in den E. coli Expressionsstamm SHuffle T7 Express lysY (New England Biolabs, USA) übertragen. Dieser Expressionsstamm ist vorteilhaft für die Expression der Hydrophobine, da er zusätzlich für eine Disulfidbrückenisomerase kodiert, welche die korrekte Ausbildung der Disulfidbrücken der Hydrophobine begünstigt. Diese spielen eine wesentliche Rolle für die richtige Faltung des Proteins.
Die Proteinexpression wurde durch Zugabe von 1 mM Isopropyl-ß-D-thiogalaktosid (IPTG) zu £. co//-Zellen, welche sich in der exponentiellen Wachstumsphase befanden, gestartet. Nach der Induktion wurden die Zellen für 4 weitere Stunden bei 30°C und 180 Umdrehungen in der Minute inkubiert. Anschließend wurden die Zellen pelletiert und gewaschen, um sie dann für die Proteinreinigung weiter verwenden zu können. Die angewendete Reinigungsmethode ist abhängig von der Löslichkeit der Proteine. Die löslichen Fusionsproteine (Ccg2_GS_EcEPSPS, SEQ ID No. 6) wurden mittels Nickel-Affinitätschromatografie unter nativen Bedingungen nach Herstellerangaben gereinigt, während die unlöslichen Hydrophobine mittels denaturierender Nickel-Affinitätschromatografie nach Herstellerangaben gereinigt wurden. Die Hydrophobine wurden vor der Dialyse durch Ultrafiltration konzentriert, für die Fusionsproteine war dies nicht nötig. Nach der Reinigung wurden die Hydrophobine gegen einen Redox-Rückfaltepuffer (10mM Glutathion reduziert, 1 mM Glutathion oxidiert; pH 5,4) und die Fusionsproteine gegen den Monsanto-Dialysepuffer (10 mM MOPS, 0,5 mM EDTA, 5% (v/v) 99,9% Glycerin, 1 mM DTT, pH 7 (Sammons et al. 2007)) dialysiert, anschließend im Kühlschrank gelagert und für die Funktionalisierung von 96-Wellplatten aus Polystyrol eingesetzt.
Die Funktionalisierung der 96-Well-Platten erfolgte durch Aufpipettieren der entsprechenden Proteinlösung mit anschließender 30-minütiger Inkubation. Daraufhin wurde der Überstand abgenommen, erneut für 5 Minuten inkubiert und die Wells danach gründlich mit Dialysepuffer gewaschen, um unspezifisch gebundenes Protein zu entfernen. Die Oberfläche wurde mit Dialysepuffer überschichtet und konnte dann für den Malachitgrün-Assay eingesetzt werden. Die beiden verschiedenen Proteinvarianten wurden benötigt, um ein optimales Verhältnis zwischen Fusionsprotein (bindet Glyphosat) und Hydrophobin (zur Stabilisierung der Oberfläche) einstellen zu können. Hierfür wurden die Proteinvarianten in unterschiedlichen Verhältnissen gemischt und die Oberflächen funktionalisiert. Anschließend wurde der Malachitgrün-Assay durchgeführt. Der Malachitgrün-Assay ist ein Nachweisverfahren für anorganisches Phosphat.
Durch Testung verschiedener Verhältnisse von Fusionsprotein zu Hydrophobin konnte gezeigt werden, dass ein Verhältnis von 1 mM Fusionsprotein zu 5 mM Hydrophobin für den Assay gut geeignet ist (Fig. 2). Bei diesem Verhältnis zeigte sich eine gute EPSPS-Aktivität, d.h. es lag ein gutes Signal-Rausch-Verhältnis vor. Steigende Hydrophobinkonzentrationen brachten keine weitere Aktivitätssteigerung. Geringere Mengen des Fusionsproteins führen zu geringeren Signalstärken, ebenso wie höhere Fusionsproteinkonzentrationen. Dies könnte auf eine sterische Behinderung der Proteinaktivität untereinander, auf Grund einer zu dichten Funktionalisierung, zurückzuführen sein. Ein weiterer Punkt der zu berücksichtigen ist, ist das mit steigender Fusionsproteinkonzentration die Sensitivität des Assays gegenüber Glyphosat abnimmt, da mehr freie Bindestellen gesättigt werden müssen, um den Aktivitätsverlust in Anwesenheit von Glyphosat messen zu können.
Die entwickelte Oberfläche bietet hier einen entscheidenden Vorteil gegenüber gelöstem Protein. Im verwendeten Konzentrationsbereich ist die Oberfläche deutlich sensitiver gegenüber Glyphosat als gelöstes Protein (Vergleich Fig. 3 und Fig. 4). Durch Variation des Verhältnisses von Fusionsprotein und Hydrophobin auf der Oberfläche kann zusätzlich die Sensitivität gegenüber Glyphosat sehr gezielt eingestellt werden und ermöglicht damit einen Nachweis von Glyphosat über einen weiten Konzentrationsbereich.
Um Falsch-Negative Ergebnisse zu verhindern, muss vor der Durchführung des Malachitgrün- Assays, Phosphat aus der Analyselösung entfernt werden. Dies lässt sich durch den Einsatz von kommerziell erhältlichen Phosphatbindern oder durch eine Eisenchlorid-Lösung realisieren (Fig. 5).
Ausführungsbeispiel 2: Spezifität des Nachweises von Glyphosat mittels funktionalisierter Oberfläche und Malachitgrün-Assay
Glyphosat ist ein Organophosphonat-Pestizid. Zur Bestimmung der Spezifität des Nachweises werden ein weiteres Organophosphonat-Pestizid (Glufosinat), ein Organophosphat-Pestizid (Chlorpyrifos) sowie das Hauptabbauprodukt von Glyphosat, a-Amino-3-hydroxy-5- methylisoxazol-4-propionsäure (AMPA), getestet (Fig. 6). Keine der drei getesteten Substanzen zeigte einen Einfluss auf die enzymatische Aktivität der EcEPSPS in einem Konzentrationsbereich zwischen 0 mM und 5 mM (Fig. 6A bis C). Damit bietet der erfindungsgemäße Assay ein hochspezifisches Detektionssystem für Glyphosat.

Claims

Patentansprüche
1. Verfahren zur Detektion von Analyten in einer Probe umfassend die Schritte:
Bereitstellen einer Oberfläche mit einem immobilisierten Analytbindungspartner, wobei der Analytbindungspartner eine Affinität zur Interaktion mit einem zu detektierenden Analyten aufweist,
Kontaktieren des Analytbindungspartners mit einer Probe enthaltend den Analyten, wobei der Analyt mit dem Analytbindungspartner interagiert,
der Analytbindungspartner durch Interaktion mit dem Analyten gehemmt wird,
Kontaktieren des Analytbindungspartners mit einem Substrat, wobei das Substrat durch den Analytbindungspartners zu einem Produkt umgesetzt wird,
Quantifizierung des Produkts und
Korrelation des quantifizierten Produkts mit einer Referenzprobe,
wobei der Analyt Glyphosat ist,
wobei der Analytbindungspartner eine Proteindomäne umfasst, aufweisend die enzymatische Aktivität des Enzyms 5-Enolpyruvylshikimat-3-phosphat-Synthase.
2. Verfahren nach einem der Ansprüche 1 , dadurch gekennzeichnet, dass der Analytbindungspartner als Fusionsprotein ausgebildet ist.
3. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass der Analytbindungspartner eine Proteindomäne umfasst, ausgewählt aus Hydrophobinen, bevorzugt die Hydrophobin-Domäne Ccg2 mit der SEQ ID No.5
4. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass der Analytbindungspartner als Fusionsprotein mit der SEQ ID No. 6 ausgebildet ist.
5. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass der Analytbindungspartner in einer Mischung mit Hydrophobinen auf der Oberfläche immobilisiert wird, wobei die Mischung des Analytbindungspartners mit dem Hydrophobin im Bereich von 1 :2 bis 1 :10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5 liegt.

Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass zur Immobilisierung des Analytbindungspartners eine Mischung aus dem Fusionsprotein mit der SEQ ID No. 6 und der Hydrophobin-Domäne Ccg2 mit der SEQ ID No. 5 im Verhältnis 1 :2 bis 1 :10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5, ganz besonders bevorzugt 1 :5 erfolgt.
7. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass die Quantifizierung des Produkts durch optische Detektion des Produkts, bevorzugt photometrisch erfolgt
8. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass die Quantifizierung des Produkts mittels Malachitgrün-Assay erfolgt.
9. Oberfläche aufweisend einen Analytbindungspartner, wobei der Analytbindungspartner ein Fusionsprotein, aufweisend eine Hydrophobin-Domäne und eine 5-Enolpyruvylshikimat-3- phosphat-Synthase-Domäne, ist.
10. Oberfläche nach Anspruch 9, dadurch gekennzeichnet, dass der Analytbindungspartner in einer Mischung mit Hydrophobinen auf der Oberfläche immobilisiert ist, wobei die Mischung des Analytbindungspartners mit dem Hydrophobin im Bereich von 1 :2 bis 1 :10, bevorzugt zwischen 1 :3 bis 1 :8, besonders bevorzugt um 1 :5 liegt.
1 1. Oberfläche nach einem der Ansprüche 9 oder 10, dadurch gekennzeichnet, dass der Analytbindungspartner ein Fusionsprotein ist, welches die SEQ ID No. 6 aufweist.
12. Kit zur Detektion von Analyten umfassend i. einen Analytbindungspartner aufweisend eine enzymatische Aktivität zur Umsetzung eines Substrats,
wobei der Analytbindungspartner ein Fusionsprotein aufweisend eine Hydrophobin- Domäne und eine 5-Enolpyruvylshikimat-3-phosphat-Synthase-Domäne ist, sowie ii. ein Hydrophobin.
13. Kit nach Anspruch 12, dadurch gekennzeichnet, dass das Kit ein Fusionsprotein mit der SEQ ID No. 6 sowie ein Hydrophobin mit der SEQ ID No. 5 umfasst.
14. Verwendung eines Verfahrens nach einem der Ansprüche 1 bis 8, einer Oberfläche nach einem der Ansprüche 9 bis 11 sowie eines Kits nach Anspruch 12 oder 13 zur Detektion von Analyten.
