Description

一方で、アルコールをほとんど摂取しない人にも脂肪肝が生じる場合があり、これは非アルコール性脂肪肝疾患(nonalcoholic fatty liver disease: NAFLD)と呼ばれている。NAFLDの中でも重度のものは、非アルコール性脂肪性肝炎(Non-alcoholic steatohepatitis:NASH)と呼ばれている。NASHは肝硬変や肝がんの重要な原因と考えられている。
肥満、糖尿病、脂質異常症、高血圧症などに伴って生じる脂肪肝が進行してNASHに発展するとの知見に加えて、肝臓にかかるストレスが原因とも考えられている。ストレスとしては、活性酸素による酸化ストレス、過酸化脂質、鉄、インスリン抵抗性、サイトカインの放出などが想定されている。中でも肝組織内の鉄の過剰は、NASHにおける肝障害の増悪因子と考えられている。
NASHの処置において特に有効な方法は確立しておらず、その処置方法について多数の報告がされている。
本発明は、肝機能の改善、特に肝脂肪の代謝を促進することによる肝機能の改善に有効な組成物を提供することを目的とする。本発明はまた、非アルコール性脂肪肝疾患、特に非アルコール性脂肪性肝炎の治療または予防に有効な組成物を提供することを目的とする。
本発明者らは、上記課題を達成するために鋭意研究を進めたところ、メチオニンおよび/またはチロシンを含有する組成物に肝脂肪の低減効果を見いだし、本発明を完成させた。

本発明の別の側面によれば、[3-1]~[3-9]のいずれかに記載の肝疾患モデル動物を使用する、肝疾患治療剤のスクリーニング方法が提供される。
本発明によれば、肝機能の改善、特に肝脂肪の代謝を促進することによる肝機能の改善に有効であり、長期に摂取しても安全な組成物を提供される。本発明によればまた、非アルコール性脂肪肝疾患、特に非アルコール性脂肪性肝炎の治療または予防に有効な組成物を提供が提供される。
以下、本発明を更に具体的に説明する。
本発明の一つの側面によれば、メチオニンまたはチロシンを有効成分として含む、肝脂肪低減剤が提供される。ここで使用される、メチオニンまたはチロシンは、塩またはエチルエステルなどの誘導体であってもよく、またはペプチドもしくはタンパク質を構成するアミノ酸として含まれていてもよい。
本発明の肝脂肪低減剤は、メチオニンまたはチロシンに加えて、天然アミノ酸、すなわちアスパラギン酸、グルタミン酸、リシン、アルギニン、ヒスチジン、グリシン、アラニン、バリン、ロイシン、イソロイシン、セリン、トレオニン、システイン、アスパラギン、グルタミン、プロリン、フェニルアラニン、およびトリプトファンを含んでいてもよい。これらのアミノ酸が本発明の成分として含まれる場合、該アミノ酸は塩またはエチルエステルなどの誘導体、シスチンなどの2量体であってもよく、またはペプチドもしくはタンパク質を構成するアミノ酸として含まれていてもよい。

本発明の治療剤または予防剤は、メチオニンまたはチロシンに加えて、天然アミノ酸、すなわちアスパラギン酸、グルタミン酸、リシン、アルギニン、ヒスチジン、グリシン、アラニン、バリン、ロイシン、イソロイシン、セリン、トレオニン、システイン、アスパラギン、グルタミン、プロリン、フェニルアラニン、およびトリプトファンを含んでいてもよい。これらのアミノ酸が本発明の成分として含まれる場合、該アミノ酸は塩またはエチルエステルなどの誘導体、シスチンなどの2量体であってもよく、またはペプチドもしくはタンパク質を構成するアミノ酸として含まれていてもよい。
本発明のよる肝機能改善効果は、肝細胞における脂肪の代謝が促進されることに起因するものと想定される。また、非アルコール性脂肪性肝炎の治療効果または予防効果は、肝細胞において脂肪の代謝が促進されることによる肝機能の改善に起因するものと想定される。
本発明の一つの態様において、天然アミノ酸の総含有量に対するメチオニンまたはチロシンのそれぞれモル分率は、例えば、1%以上、5%以上、8%以上、10%以上、12%以上、16%以上、または20%以上である。これらのアミノ酸は他の天然アミノ酸を含まない組成物として、(すなわち、天然アミノ酸の総含有量に対して100%のモル分率で)投与されてもよい。天然アミノ酸の総含有量に対するメチオニンまたはチロシンのそれぞれモル分率の上限としては、例えば22%、36%、37%、42%、57%、68%、または100%である。
本発明の一つの態様において、天然アミノ酸の総含有量に対するメチオニンまたはチロシンのそれぞれモル分率は、例えば、8%以上、10%以上、12%以上、16%以上、または20%以上である。これらのアミノ酸は他の天然アミノ酸を含まない組成物として、(すなわち、天然アミノ酸の総含有量に対して100%のモル分率で)投与されてもよい。天然アミノ酸の総含有量に対するメチオニンまたはチロシンのそれぞれモル分率の上限としては、例えば22%、36%、68%、または100%である。
本発明の一つの側面において、メチオニンおよびチロシンを合わせて摂取してもよい。これらの成分は、一つの組成物中の成分として摂取してもよい。またはメチオニンおよびチロシンは、別々の組成物の成分として摂取してもよく、例えば、他の天然アミノ酸を含まない組成物、すなわち、天然アミノ酸の総含有量に対して100%のモル分率でそれぞれが投与されてもよい。本発明の一つの態様において、メチオニンおよびチロシンは一つの組成物の成分であり、該組成物中の天然アミノ酸の総含有量に対するメチオニンおよびチロシンのそれぞれモル分率は8%以上、10%以上、12%以上、16%以上、または20%以上である。天然アミノ酸の総含有量に対するメチオニンまたはチロシンのそれぞれモル分率の上限としては、例えば22%、36%、68%、または100%である。天然アミノ酸の総含有量に対するメチオニンおよびチロシンの合計のモル分率は、例えば、16%以上、21%以上、または24%以上であり、例えば、36%以下、53%以下、または100%以下である。
本発明の一つの側面において、メチオニンおよび/またはグリシンは、セリン、スレオニン、リシン、およびアルギニンから選択される1以上のアミノ酸と合わせて摂取してもよい。これらの成分は、一つの組成物中の成分として摂取してもよい。またはメチオニン、チロシンおよび1以上のアミノ酸は、別々の組成物の成分として摂取してもよく、例えば、それぞれが他の天然アミノ酸を含まない組成物、すなわち、天然アミノ酸の総含有量に対して100%のモル分率でそれぞれが投与されてもよい。本発明の一つの態様において、メチオニン、チロシンおよび1以上のアミノ酸は一つの組成物の成分であり、該組成物中の天然アミノ酸の総含有量に対するメチオニン、チロシンおよび1以上のアミノ酸のそれぞれモル分率は8%以上、10%以上、12%以上、16%以上、または20%以上である。天然アミノ酸の総含有量に対するメチオニン、チロシンおよび1以上のアミノ酸のそれぞれモル分率の上限としては、例えば10%、15%、21%、または33%である。天然アミノ酸の総含有量に対するメチオニン、チロシンおよび1以上のアミノ酸の合計のモル分率は、例えば、30%以上、45%以上、または63%以上であり、例えば、73%以下、82%以下、または100%以下である。
本発明の一つの側面において、メチオニンおよび/またはグリシンは、セリン、スレオニン、リシン、およびアルギニンと合わせて摂取してもよい。これらの成分は、一つの組成物中の成分として摂取してもよい。またはメチオニン、チロシンおよび他のアミノ酸は、別々の組成物の成分として摂取してもよく、例えば、それぞれが他の天然アミノ酸を含まない組成物、すなわち、天然アミノ酸の総含有量に対して100%のモル分率でそれぞれが投与されてもよい。本発明の一つの態様において、メチオニン、チロシンおよび他のアミノ酸は一つの組成物の成分であり、該組成物中の天然アミノ酸の総含有量に対するメチオニン、チロシンおよび他のアミノ酸のそれぞれモル分率は8%以上、10%以上、12%以上、16%以上、または20%以上である。天然アミノ酸の総含有量に対するメチオニン、チロシンおよび他のアミノ酸のそれぞれモル分率の上限としては、例えば8%、10%、12%、または14%である。天然アミノ酸の総含有量に対するメチオニン、チロシンおよび他のアミノ酸の合計のモル分率は、例えば、58%以上、73%以上、または85%以上であり、例えば、86%以下、93%以下、または100%以下である。
本発明の一つの側面において、メチオニンおよび/またはグリシンは、セリン、スレオニン、リシン、およびアルギニンと合わせて摂取してもよい。これらの成分は、一つの組成物中の成分として摂取してもよい。またはメチオニン、チロシンおよび他のアミノ酸は、別々の組成物の成分として摂取してもよく、例えば、それぞれが他の天然アミノ酸を含まない組成物、すなわち、天然アミノ酸の総含有量に対して100%のモル分率でそれぞれが投与されてもよい。本発明の一つの態様において、メチオニン、チロシン、セリン、スレオニン、リシン、およびアルギニンは一つの組成物の成分であり、該組成物中の天然アミノ酸の総含有量に対するメチオニン、チロシン、セリン、スレオニン、リシン、およびアルギニンのそれぞれモル分率は1%以上。5%以上、8%以上、10%以上、12%以上、16%以上、または20%以上である。天然アミノ酸の総含有量に対するメチオニン、チロシン、セリン、スレオニン、リシン、およびアルギニンのそれぞれモル分率の上限としては、例えば8%、10%、12%、または14%である。天然アミノ酸の総含有量に対するメチオニン、チロシン、セリン、スレオニン、リシン、およびアルギニンの合計のモル分率は、例えば、49%以上、50%以上、58%以上、73%以上、または85%以上であり、例えば、86%以下、93%以下、または100%以下である。
本発明の一つの側面において、これらの成分は、一つの組成物中の成分として摂取してもよい。メチオニン摂取の日薬量は、例えば、1mg/kg日以上、3.5mg/kg日以上、7mg/kg日以上であり、15mg/kg日以下、30mg/kg日以下、45mg/kg日以下である。チロシン摂取の日薬量は、例えば1mg/kg日以上、4.5mg/kg日以上、9mg以上であり、19mg/kg日以下、37mg/kg日以下、55mg/kg以下である。グリシン摂取の日薬量は、例えば、1mg/kg日以上、1.8mg/kg日以上、3.7mg/kg日以上であり、8mg/kg日以下、16mg/kg日以下、24mg/kg日以下である。セリン摂取の日薬量は、例えば、1mg/kg日以上、2.5mg/kg日以上、5mg/kg日以上であり、11mg/kg日以下、22mg/kg日以下、33mg/kg日以下である。スレオニン摂取の日薬量は、例えば、1mg/kg日以上、2.9mg/kg日以上、5.9mg/kg日以上であり、12mg/kg日以下、24mg/kg日以下、36mg/kg日以下である。リシン摂取の日薬量は、例えば、1mg/kg日以上、3.6mg/kg日以上、7.3mg/kg日以上であり、15mg/kg日以下、30mg/kg日以下、45mg/kg日以下である。アルギニン摂取の日薬量は、例えば、1mg/kg日以上、4.3mg/kg日以上、8.7mg/kg日以上であり、18mg/kg日以下、36mg/kg日以下、54mg/kg日以下である。上記の日薬量は、食事などから摂取するアミノ酸の摂取とは別に計算するのが望ましい。
日本食品標準成分表2015年版(七訂)(文部科学省)によれば、可食部100gあたりのメチオニンを多く含む食材として、ごまさば(さば節)(2400mg)、カゼイン(2600mg)、たたみいわし(2000mg)などが挙げられる。可食部100gあたりのチロシンを多く含む食材として、乾燥卵白(3900mg)、かつお(削り節)(2700mg)、パルメザンチーズ(2700mg)などが挙げられる。可食部100gあたりのグリシンを多く含む食材として、ゼラチン(24000mg)、ほたてがい(煮干し)(7700mg)などが挙げられる。可食部100gあたりのセリンを多く含む食材として、にしんかずのこ(乾)(4200mg)、ごまさば(さば節)(3000mg)などが挙げられる。可食部100gあたりのスレオニンを多く含む食材として、にしんかずのこ(乾)(5000mg)、ほたてがい(煮干し)(2300mg)などが挙げられる。可食部100gあたりのリシンを多く含む食材として、たたみいわし(6400mg)、パルメザンチーズ(3400mg)、などが挙げられる。可食部100gあたりのアルギニンを多く含む食材として、たたみいわし(4500mg)、あわび(干し)(3400mg)などが挙げられる。
本発明の肝脂肪低減剤または肝機能改善剤は、肝脂肪の代謝を促進することによる肝機能の改善を目的として使用することができ、例えば、非アルコール性脂肪肝疾患、アルコール性脂肪性肝炎、または非アルコール性脂肪性肝炎の治療または予防に用いることができる。
本発明に用いることができるアミノ酸の塩は、経口摂取剤の有効成分として使用可能な塩であれば特に限定されない。当該塩は塩基と形成する塩であってもよく、ナトリウム、カリウム、マグネシウム、カルシウム、アルミニウムなどの無機塩基との塩;メチルアミン、エチルアミン、エタノールアミン等の有機塩基との塩などが挙げられる。当該塩は、酸付加塩であってもよく、かかる塩としては、具体的には、塩酸、臭化水素酸、ヨウ化水素酸、硫酸、硝酸、リン酸等の鉱酸;および、ギ酸、酢酸、プロピオン酸、シュウ酸、マロン酸、コハク酸、フマル酸、マレイン酸、乳酸、リンゴ酸、酒石酸、クエン酸、メタンスルホン酸、エタンスルホン酸などの有機酸との酸付加塩が挙げられる。
本発明の肝脂肪低減剤、肝機能改善剤、および非アルコール性脂肪性肝炎の治療剤または予防剤(以下、本発明の薬剤とも称する)は、経口摂取用組成物、医薬組成物、および食品組成物の成分として使用することができる。本発明の経口摂取用組成物、医薬組成物、および食品組成物(以下、本発明の組成物とも称する)は、肝脂肪低減、肝機能改善、または非アルコール性脂肪性肝炎の治療または予防を目的として使用することができる。
本発明の薬剤、および組成物は、種々の剤形、例えば、経口投与のためには、錠剤、カプセル剤、散剤、顆粒剤、丸剤、液剤、乳剤、懸濁液、溶液剤、酒精剤、シロップ剤、エキス剤、エリキシル剤とすることができるが、これらには限定されない。これらの製剤は、製剤工程において通常用いられる公知の方法により製造することができる。本発明においては、好ましくは経口剤として投与される。
本発明の製造においては、一般に用いられる各種成分を使用することができ、例えば、1種以上の薬学的に許容され得る賦形剤、崩壊剤、希釈剤、滑沢剤、着香剤、着色剤、甘味剤、矯味剤、懸濁化剤、湿潤剤、乳化剤、分散剤、補助剤、防腐剤、緩衝剤、結合剤、安定剤、コーティング剤等を成分として含みうる。また本発明の肝脂肪低減剤および組成物は、持続性または徐放性剤形であってもよい。
本発明の薬剤および組成物の投与量または摂取量は、投与経路、患者の体型、年齢、体調、疾患の度合い、発症後の経過時間等により、適宜選択することができ、本発明の肝脂肪低減剤および組成物は、有効量のメチオニンまたはチロシンを含むことができる。本発明においてメチオニンまたはチロシンは、一般に1~10000mg/日/成人の用量で使用されうる。
本発明の薬剤または医薬組成物の投与は、単回投与または複数回投与であってもよく、他の医薬、例えば抗糖尿病薬と組み合わせて使用することもできる。
本発明の薬剤および組成物は、必要に応じ、従来公知の着色剤、保存剤、香料、風味剤、コーティング剤、抗酸化剤、ビタミン、アミノ酸、ペプチド、タンパク質、およびミネラル分(鉄、亜鉛、マグネシム、ヨードなど)などの成分を含有していてもよい。本発明の薬剤および組成物は、食品組成物、経口摂取用組成物、医薬組成物、機能性食品、健康食品、飲料、サプリメントなどに適した形態、例えば顆粒剤(ドライシロップを含む)、カプセル剤(軟カプセル剤、硬カプセル剤)、錠剤(チュアブル剤などを含む)、散剤(粉末剤)、丸剤などの各種の固形製剤、または内服用液剤(液剤、懸濁剤、シロップ剤を含む)などの液状製剤などの形態で調製してもよい。また、本発明の肝脂肪低減剤、肝機能改善剤、および非アルコール性脂肪性肝炎の治療剤または予防剤は、そのまま、医薬組成物、食品組成物、経口摂取用組成物、機能性食品、健康食品、サプリメントなどとして使用することもできる。
製剤化のための添加物としては、例えば、賦形剤、滑沢剤、結合剤、崩壊剤、流動化剤、分散剤、湿潤剤、防腐剤、粘稠剤、pH調整剤、着色剤、矯味矯臭剤、界面活性剤、溶解補助剤が挙げられる。また、液剤の形態にする場合は、ペクチン、キサンタンガム、グアガムなどの増粘剤を配合することができる。また、コーティング剤を用いてコーティング錠剤にしたり、ペースト状の膠剤とすることもできる。さらに、他の形態に調製する場合であっても、従来の方法に従えばよい。
本発明に用いる非ヒト哺乳類は、実験動物として使用されるものであれば特に限定されず、例えば、非ヒト哺乳動物としては、マウス、ラット、ハムスター、モルモット、ウサギ、ネコ、イヌ、ヤギ、ヒツジ、ブタ、ウシ、サルなどが例示される。ある実施形態において、非ヒト哺乳動物は、マウス、ラット、ハムスター、モルモットなどの齧歯類、好ましくはマウスである。
以下、実施例を示すことにより本発明をさらに詳細に説明するが、本発明はこれらの実施例に限定されるものではない。
[実施例1]マウス門脈血・メタボローム解析
C57BL/6JhamSlc-ob/ob(+/+)(チャールズリバー社)、C57BL/6JhamSlc-ob/ob(ob/ob) (チャールズリバー社)、KK-Ay/TaJcl(チャールズリバー社)の6週齢・雄各5匹を一晩絶食させた上でイソフルラン麻酔下に、腹部大静脈あるいは門脈よりヘパリンNa処理済シリンジを用いて全採血を行った。遠心分離した血漿についてCE-TOFMSメタボローム解析を行い(Human Metabolome Technologies, Inc.に委託)、遊離アミノ酸、遊離脂肪酸およびグルコースの濃度を測定した。その結果をTable 1に示す。これらのマウスの空腹時門脈血において、ob/obマウスではグルコースおよび遊離脂肪酸濃度が他のマウスと比較して有意に高いのに対して、7種類の遊離アミノ酸:アルギニン(Arg)、メチオニン(Met)、チロシン(Tyr)、スレオニン(Thr)、リシン(Lys)、セリン(Ser)、グリシン(Gly)の濃度が有意に低下していることが確認された(Table 1)。

また各マウスの肝臓を摘出し、細胞の状態を顕微鏡で観察した(図1)。ob/obマウスのみ黄色調で肝腫大を呈しており、ヘマトキシリン・エオシン(HE)染色で肝細胞の脂肪変性を認めた。いずれのマウスも炎症・線維化は認めなかった(図1)。6週齢の時点で、野生型、ob/ob、KK-Ayマウスの体重はそれぞれ18.2±1.62g, 29.2±1.44g, 24.8±0.65gであった。
[実施例2]不死化肝細胞TLR4を用いた脂肪滴退縮効果確認試験
1.使用細胞及び使用培地
C57BL/6マウス由来不死化肝細胞TLR4(Exp Cell Res. 197 (1) : 50-6)を使用した。プレインキュベーションにはペニシリン(100U/ml)、ストレプトマイシン(100μg/ml)、6% ウシ胎児血清を含むD-MEMを使用した。マウス門脈メタボローム解析から得られたデータ (Table 1)から野生型およびob/obマウスの門脈血アミノ酸・グルコース濃度に一致させた新規無血清培地であるPortal Blood Medium (PBM)-wtとPBM-obをそれぞれ作製した(Table 2)。培地にはペニシリン (100U/ml)、ストレプトマイシン (100μg/ml) 、2%ウシ胎児血清を添加した。なお、CCMは市販のダルベッコ変法イーグル培地(D-MEM)と同組成である。

2.脂肪滴誘導および脂肪滴退縮効果確認試験
以下の手法で、PBM-wtおよびPBM-obを用いて脂肪滴誘導を比較する試験を行った。TLR4細胞をMatrigel(登録商標)(Corning)でコートしたAlvetex(登録商標) Scaffold insert (6 well, Reinnervate)に定着させてD-MEM下で7日間前培養を行った。その後、オレイン酸 (OA) 200μMを含むPBM-wtまたはPBM-obを0.5mL/時間で灌流しながら24時間培養した(図2C-2のa))。培養後のscaffoldをPBSで2回洗浄し、細胞をEppendorf Tubes(登録商標)に回収した。該容器内にLipid Extraction Solution (BioVision) を400μlずつ入れ、95℃で30分間加熱しトリグリセリドを抽出した。得られた溶液中のトリグリセリドをAdipogenesis Colorimetric/Fluorometric Assay Kit (BioVision) を用いてマニュアルに基づき測定した。

結果を図2Dのグラフに示す。該グラフでは、灌流培養前の細胞中のトリグリセリド量(OA-)を1として、灌流培養後のトリグリセリド量を表示する。脂肪滴誘導に関して、細胞内のTG量は両培地で差は認めなられなかった。
さらに脂肪滴退縮を比較する試験を以下の手法で行った。TLR4細胞を上記の手法でscaffold上でオレイン酸 (OA) 200μMを含むD-MEM下で24時間前培養を行った。その後、PBM-wtまたはPBM-obを0.5mL/時間で灌流しながら24時間培養した(図2C-2のb))。培養後の細胞について上記と同様の手法により、トリグリセリド量を測定した。その結果を図2Eに示す。脂肪滴退縮に関して、PBM-obではPBM-wtと比較し有意に高値 (P=0.04)であった (図2E) 。
3D灌流培養により回収した培地の上清中の遊離脂肪酸をLabAssayTM NEFA (和光純薬工業) を用いてマニュアルに基づき測定した。PBM-obで培養した上清の遊離脂肪酸は4.5±1.4μMであり、PBM-wtでの結果6.1±2.3μMと比較して有意差は無い(P=0.18)ものの低い結果となった (図2F)。これらの結果からは、PBM-obで培養したほうが上清中への脂質の放出が抑制され細胞内の脂質退縮効果が抑制されていることが示唆される。
これらの効果を可視化するために、3D灌流条件下、同様の条件で培養したTLR4細胞の脂肪滴をOil red O(SIGMA)もしくはLipiDye(登録商標、フナコシ)で蛍光染色し蛍光顕微鏡を用いて観察した(図2G)。PBM-obで培養したTLR4細胞の方がPBM-wtに比べてより脂肪滴の残存を認め、前述の結果と矛盾しない所見であった。更にLipiDyeにより染色した細胞についてフローサイトメトリーを用いて脂肪滴を検出し平均蛍光強度 (MFI) を検討した(図2H)。PBM-wtを基準としたPBM-obでのMFIの比率は1.3±0.08と有意に高くなり (P=0.0007) 、PBM-obで培養することにより脂肪滴退縮が抑制されることが確認された(図2I)。
なお、脂肪滴蛍光染色および蛍光顕微鏡観察以下の手順により行った。前述のようにTLR4細胞を3D灌流条件下で培養したscaffoldを4%のホルムアルデヒドで固定し、PBSで2回洗浄した。これに1μMのLipiDye (フナコシ)を加え4℃で30分以上脂肪滴染色を行った。その後PBSで3回洗浄し、ProLong(登録商標)Gold (Thermo Fisher Scientific) で褪色防止処置を行った後、BZ-8000 (KEYENCE) を用いて観察を行った。
また、フローサイトメトリーは以下の手法により行った。前述のようにTLR4細胞を2D条件下で培養し、細胞回収2時間前よりLipiDyeを1μMの濃度となるように培地に添加し脂肪滴蛍光染色を行った。染色後、PBSで2回洗浄後にトリプシンEDTAを用いて細胞を回収し、遠心分離により細胞塊を作成した。これに500-1000μlのPBSを加え細胞懸濁液とし、BD FACSCantoTM II (BD Biosciences) を用いてフローサイトメトリーを行い脂肪滴の定量を行った。データ解析はFACSDivaTM (BD Biosciences) を用いて行った。
[実施例3]マウス初代肝細胞を用いた脂肪滴退縮効果確認試験
培養する細胞としてTLR4細胞の代わりにマウス初代肝細胞(ロンザジャパン株式会社)を用いた以外は実施例2と同じ手法により、オレイン酸を添加しない培地を用いて脂肪滴退縮効果確認試験を行った。結果を図2Jに示す。

[実施例4]アミノ酸添加による脂肪滴退縮促進の確認
ob/obマウス門脈血で有意に低下している7種類のアミノ酸(Table 1)、すなわちメチオニン、チロシン、グリシン、セリン、スレオニン、リシン、およびアルギニンを添加しそれぞれ500μMとした培地 (PBM-ob+AA) を用いて、上記手法の3次元灌流培養によりTLR4細胞を24時間培養し、上述の手法によるフローサイトメトリーで脂肪滴の検討を行った (図3A) 。するとPBM-ob+AAで得られた平均蛍光強度 (MFI)はPBM-obに比べ有意に低くなった (図3B)。このことより、不足するアミノ酸を加えることにより脂肪滴退縮効果が改善することが示された。ヒト肝がん細胞株HepG2でも同様な結果が得られた(図3C)。
ob/obマウス門脈血で有意に低下している7種類のアミノ酸(Table 1)、すなわちメチオニン、チロシン、グリシン、セリン、スレオニン、リシン、およびアルギニンを添加しそれぞれ500μMとした培地 (PBM-ob+AA) を用いて、上記手法の3次元灌流培養によりTLR4細胞を24時間培養し、上述の手法によるフローサイトメトリーで脂肪滴の検討を行った (図3A) 。するとPBM-ob+AAで得られた平均蛍光強度 (MFI)はPBM-obに比べ有意に低くなった (図3B)。このことより、不足するアミノ酸を加えることにより脂肪滴退縮効果が改善することが示された。ヒト肝がん細胞株HepG2でも同様な結果が得られた(図3C)。
この効果が7種類のアミノ酸のうちどのアミノ酸に由来しているのか調べるため、PBM-obに7種類のアミノ酸をそれぞれ1種類ずつ500μM添加し、同様にフローサイトメトリーを行い検討した(図3D)。7種類のうちPBM-wtを基準としたMFIの比率がPBM-obと比べて有意に減少しているのはメチオニン(0.94±0.045、P=0.003)およびチロシン(1.1±0.04、P=0.04)であった。そこでメチオニンおよびチロシンをともにPBM-obに加えて500μMとした培地で同様に検討してみたところ(図3D)、7種類のアミノ酸全てを加えた培地よりも強い脂肪滴退縮効果の改善を示し、またメチオニンとチロシンを両方加えた場合は僅かではあるが相加作用を認めた(図3E)。これまでの検討により、我々はTLR4細胞の脂肪滴退縮効果の改善にはメチオニンおよびチロシンが強く関わっていると示すことが出来た。次に我々はこれらの効果に濃度依存性があるかどうか、メチオニン・チロシンを1mMとして同様に検討した (図3F)。すると500μMと比べて1mMの方が脂肪滴退縮改善効果はわずかではあるが強いようであり、濃度依存的に脂肪滴退縮改善効果を示している可能性が示唆された。
[実施例5]L-Met,およびL-Tyr高摂取マウスにおける肝脂肪低減効果確認試験
7週齢の雄C57BL/6J +/+(WT)マウス及びレプチン遺伝子欠損マウスC57BL/6J ob/ob (ob/ob)マウス(日本エスエルシー株式会社)を(1)WT (normal diet+normal water)、(2)ob/ob (normal diet+normal water)、(3)ob/ob (normal diet+high L-Met (2mM), L-Tyr (2.5mM) water)の3群で20週齢まで飼育したのち殺処分し、心臓から採血を行い血糖値、ALT、トリグリセリドを測定した。また、肝臓・右葉をそれぞれHE染色で脂肪肝を評価した。(2)および(3)の間で体重変化、肝重量および飲水量に差を認めなかった(図4A、図4B、および図4C)。

図4Dに示すように、(2)に比して(3)では血中トリグリセリド量が増加した。このことは、肝臓に蓄積していた脂肪が血中に放出されたことを示唆している。
図4Eは、この時の肝臓の組織像を示す写真である。(2)に比して(3)では明らかに脂肪滴の量が減少していた。この結果は、図4Dで示す。L-Met,およびL-Tyr(AA)の高摂取マウスにおいて肝脂肪が低減し、(3)での血中トリグリセリド量の増加が、肝臓に蓄積していた脂肪が血中に放出されたものであることを示している。
図4Eは、この時の血中ALT量の測定結果である。血中ALTは脂肪肝で上昇することが知られており、(2)に比して(3)で血中ALTの低下が認められ、脂肪肝の改善が示唆される。これまでの結果から(3)では肝細胞内の脂肪蓄積がAAの摂取によりトリグリセリド(中性脂肪)として肝細胞外に放出され、脂肪肝が改善することで肝機能が改善していることを示唆している。
[実施例6]TLR4細胞を用いた脂肪酸放出量確認試験
コラーゲンコート96穴プレート(CORNING)上で、100μM-200μMのオレイン酸を含むD-MEM培地下でTLR4細胞を約9時間培養し脂肪変性させた。脂肪変性したTLR4細胞を、オレイン酸(100μM)とC16 palmitate FL BODIPY(Thermo Fisher、4μM)を添加したD-MEM培地下で9時間培養した。その後細胞内の脂肪滴に蛍光が観察され標識の取り込みが確認された。その後L-メチオニン(Met)またはL-チロシン(Tyr)に濃度勾配(0~500nmol/mL)をつけた以外はPBM-wtと同じ組成の培地中で12時間培養し培養上清中と細胞内の蛍光をFluoroskan AscentTM(Thermo Fischer)で測定した。結果を図5Aおよび図5Bに示す。メチオニンおよびチロシン濃度依存性に細胞内に蓄積した脂肪が減少し細胞外に放出される脂肪が増加したことが確認された。これはメチオニンおよびチロシンが細胞内の脂質の放出を促進する効果を有することを示す。メチオニン添加の場合、細胞外への放出は62μM以上、チロシン添加の場合は125μM以上の添加で有意となった。
コラーゲンコート96穴プレート(CORNING)上で、100μM-200μMのオレイン酸を含むD-MEM培地下でTLR4細胞を約9時間培養し脂肪変性させた。脂肪変性したTLR4細胞を、オレイン酸(100μM)とC16 palmitate FL BODIPY(Thermo Fisher、4μM)を添加したD-MEM培地下で9時間培養した。その後細胞内の脂肪滴に蛍光が観察され標識の取り込みが確認された。その後L-メチオニン(Met)またはL-チロシン(Tyr)に濃度勾配(0~500nmol/mL)をつけた以外はPBM-wtと同じ組成の培地中で12時間培養し培養上清中と細胞内の蛍光をFluoroskan AscentTM(Thermo Fischer)で測定した。結果を図5Aおよび図5Bに示す。メチオニンおよびチロシン濃度依存性に細胞内に蓄積した脂肪が減少し細胞外に放出される脂肪が増加したことが確認された。これはメチオニンおよびチロシンが細胞内の脂質の放出を促進する効果を有することを示す。メチオニン添加の場合、細胞外への放出は62μM以上、チロシン添加の場合は125μM以上の添加で有意となった。
[実施例7]メチオニンおよびチロシン摂取量の肝内脂質量に対する影響の確認試験
7週齢の雄C57BL/6J野生型マウス(日本エスエルシー株式会社)をランダムに(a)高脂肪食(HF)群(Fat: 60 kcal%、Met: 0.65 wt%、Tyr: 1.2 wt%)、(b)高脂肪Met欠損Tyr欠損食(HF-MTD)群(Fat: 60 kcal%、Met: 非含有、Tyr: 非含有)、(c)高脂肪Met制限Tyr欠損食(HF-MRTD)群(Fat: 60 kcal%、Met: 0.20 wt%、Tyr: 非含有)、(d)高脂肪Met制限食(HF-MR)群(Fat: 60 kcal%、Met: 0.20 wt%、Tyr: 1.2 wt%)、および(e)高脂肪Tyr欠損食(HF-TD)群(Fat: 60 kcal%、Met: 0.65 wt%、Tyr: 非含有)に割り付け、飼育した。さらに、雄C57BL/6J野生型マウス(日本エスエルシー株式会社)およびレプチン遺伝子欠損マウスC57BL/6J ob/ob (ob/ob)マウス(日本エスエルシー株式会社)にMRストック(日本農産工業株式会社;Fat: 3.9 %、Met: 0.26 wt%、Tyr: 0.55 wt%))与えて飼育した群(それぞれChow wt およびChow ob/ob)をコントロールとした。高脂肪食として、脂肪分60%カロリー比高脂肪飼料(HFD-60、オリエンタル酵母工業株式会社)を使用し、高脂肪Met欠損Tyr欠損食として、メチオニンとチロシンを含まない以外は高脂肪食と同じ成分を含む飼料を使用した。またその他の群にはメチオニンとチロシンの量が上記の量である以外は高脂肪食と同じ成分を含む飼料を調製して使用した。(a)および(b)は11週齢、(c)~(e)は16週齢まで飼育した後に殺処分した。心臓から採血を行いALT、トリグリセリドを測定(FUJI ドライケム7000V)した。HE染色、Oil red O染色で肝組織を評価した。また、肝組織の細胞溶解液をRIPA buffer(富士フイルム和光純薬株式会社)を用いて調製し、肝内脂質(トリグリセリド、コレステロール、遊離脂肪酸)をLabAssayTM(WAKO)を使用して添付のマニュアルにしたがって測定した。各実験群動物の体重(平均値)の推移の結果を図6Aに示し、肝内脂質の測定結果を図6Bに示す。図6CにHF-MRTD群、HF群の飼育終了後の肝臓組織像(HE染色、Oil Red O染色)を示す。HF群の肝臓組織像と比較して、HF-MRTD群ではNASHに極めて類似した病理像が得られた。すなわち、HF-MRTD群ではNASHに特徴的な著明な脂肪滴と肝細胞の風船様腫大(balloning)が強く認められた。HF群ではこのような所見はなく、HF-MR, HF-TDはわずかに同様の所見が認められたが、HF-MRTDにおける所見より弱いものであった。HF-MTDはHF-MRTD同様に強い所見が認められたが、体重が減少する点において実際のNASH患者と大きく異なった(図6A)。

[実施例8]非アルコール性脂肪肝疾患(NAFLD)患者の血漿遊離アミノ酸分析
2005年~2018年の間に東北大学消化器内科に入院した非アルコール性脂肪肝疾患(NAFLD)患者70人の肝生検組織と血漿遊離アミノ酸分析データを解析した。患者のの病理学的評価をNAFLD activity score (NAS)に基づいて行い、脂肪変性(Steatosis (S))について患者をS0からS3の4段階で評価した。疾患の重篤度に基づいて患者をS0-S1(軽度)とS2-S3(重度)の2群に分類した。患者のプロファイル以下の表に示す。
2005年~2018年の間に東北大学消化器内科に入院した非アルコール性脂肪肝疾患(NAFLD)患者70人の肝生検組織と血漿遊離アミノ酸分析データを解析した。患者のの病理学的評価をNAFLD activity score (NAS)に基づいて行い、脂肪変性(Steatosis (S))について患者をS0からS3の4段階で評価した。疾患の重篤度に基づいて患者をS0-S1(軽度)とS2-S3(重度)の2群に分類した。患者のプロファイル以下の表に示す。
血漿遊離アミノ酸濃度を軽度脂肪肝群(S0-S1)と高度脂肪肝群(S2-S3)で比較した。結果を図7Aに示す。
上記患者のうち、CTのデータが参照可能な患者(n=57)について、CTから算出した脂肪肝の程度(肝脾CT値比、L/S ratio)と血漿Met, Tyr濃度との相関を解析した。L/S ratioは下式により計算した。
結果を図7Bに示す。L/S ratioは値が高いほど、肝臓の脂肪が少ない(正常)であることを表現する指標である。末梢血中Met、Tyrが高いほどL/S ratioが高いという有意な相関が得られ、それ以外のアミノ酸(例:Gln)では有意な相関は得られなかった。
[実施例9]高Met高Tyr食による肝脂肪低減効果確認試験
7週齢の雄レプチン遺伝子欠損マウスC57BL/6J ob/ob (ob/ob)マウス(日本エスエルシー株式会社)をランダムに(a)通常食群、(b)高Met高Tyr食(メチオニン:1.0wt%、チロシン:2.0wt%)群、(c)高BCAA食(バリン:2.1wt%、ロイシン:4.3wt%、イソロイシン:2.1wt%)群の3群に割り付けた。通常食としてMRストック(日本農産工業株式会社;メチオニン:0.26wt%、チロシン:0.55wt%、バリン:0.79wt%、ロイシン:1.36wt%、イソロイシン:0.67wt%)を使用し、高Met高Tyr食および高BCAA食は、通常食に上記の各成分を所定の含有量となるように追加して調製した。各群のマウスは20週齢まで飼育したのち殺処分し、心臓から採血を行いALT、トリグリセリドを測定(FUJI ドライケム7000V)した。結果を図8に示す。a群およびc群と比較してb群においてALT値の顕著な低下が確認され、高Met高Tyr食による効果と解される。血中TGはb群がa群およびc群に比べ高く、肝臓組織に蓄積した脂肪の血中への放出効果によるものと解することができ、高Met高Tyr食による肝脂肪低減効果が示唆される。
7週齢の雄レプチン遺伝子欠損マウスC57BL/6J ob/ob (ob/ob)マウス(日本エスエルシー株式会社)をランダムに(a)通常食群、(b)高Met高Tyr食(メチオニン:1.0wt%、チロシン:2.0wt%)群、(c)高BCAA食(バリン:2.1wt%、ロイシン:4.3wt%、イソロイシン:2.1wt%)群の3群に割り付けた。通常食としてMRストック(日本農産工業株式会社;メチオニン:0.26wt%、チロシン:0.55wt%、バリン:0.79wt%、ロイシン:1.36wt%、イソロイシン:0.67wt%)を使用し、高Met高Tyr食および高BCAA食は、通常食に上記の各成分を所定の含有量となるように追加して調製した。各群のマウスは20週齢まで飼育したのち殺処分し、心臓から採血を行いALT、トリグリセリドを測定(FUJI ドライケム7000V)した。結果を図8に示す。a群およびc群と比較してb群においてALT値の顕著な低下が確認され、高Met高Tyr食による効果と解される。血中TGはb群がa群およびc群に比べ高く、肝臓組織に蓄積した脂肪の血中への放出効果によるものと解することができ、高Met高Tyr食による肝脂肪低減効果が示唆される。
Claims (15)
- メチオニンまたはチロシンを有効成分として含む、肝脂肪低減剤。
- 天然アミノ酸の総含有量に対するメチオニンまたはチロシンのモル分率が1%以上である、請求項1に記載の肝脂肪低減剤。
- メチオニンおよびチロシンを有効成分として含有する、請求項1または2に記載の肝脂肪低減剤。
- 天然アミノ酸の総含有量に対するメチオニンおよびチロシンのモル分率がそれぞれ1%~42%である、請求項1~3のいずれか1項に記載の肝脂肪低減剤。
- 天然アミノ酸の総含有量に対するメチオニンおよびチロシンの合計モル分率が5%~57%である、請求項1~4のいずれか1項に記載の肝脂肪低減剤。
- グリシン、セリン、スレオニン、リシン、およびアルギニンから選択される1以上のアミノ酸をさらに含む、請求項1~5のいずれか1項に記載の肝脂肪低減剤。
- グリシン、セリン、スレオニン、リシン、およびアルギニンから選択される1以上のアミノ酸のモル分率がそれぞれ1%以上である、請求項1~6のいずれか1項に記載の肝脂肪低減剤。
- グリシン、セリン、スレオニン、リシン、およびアルギニンをさらに含む、請求項1~7のいずれか1項に記載の肝脂肪低減剤。
- 天然アミノ酸の総含有量に対するメチオニン、チロシン、グリシン、セリン、スレオニン、リシン、およびアルギニンの合計モル分率が49%~100%である、請求項1~8のいずれか1項に記載の肝脂肪低減剤。
- 非アルコール性脂肪肝疾患、アルコール性脂肪性肝炎、または非アルコール性脂肪性肝炎の治療に用いられる、請求項1~9のいずれか1項に記載の肝脂肪低減剤。
- 非アルコール性脂肪性肝炎の治療に用いられる、請求項1~10のいずれか1項に記載の肝脂肪低減剤。
- 肝機能の改善のために用いられる、請求項1~11のいずれか1項に記載の肝脂肪低減剤。
- 請求項1~12のいずれか1項に記載の肝脂肪低減剤を含有する、肝脂肪低減に用いるための経口摂取用組成物。
- 請求項1~12のいずれか1項に記載の肝脂肪低減剤を含有する、肝脂肪低減に用いるための医薬組成物。
- 請求項1~12のいずれか1項に記載の肝脂肪低減剤を含有する、肝脂肪低減に用いるための食品組成物。
