Abstract
【課題】iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する方法及びかかる再生T細胞集団を提供すること。 【解決手段】(1)採取した遺伝的多様性を有するT細胞集団から、目的の組織又は抗原に対して指向性を有するT細胞集団を濃縮する工程、 (2)前記濃縮したT細胞集団をiPS細胞へと初期化し、遺伝的多様性を維持したまま培養する工程、及び (3)前記培養したiPS細胞から遺伝的多様性を有する再生T細胞集団を製造する工程を含む、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する方法。
Description
本発明は、遺伝的多様性を有する再生T細胞集団を製造する方法及びかかる再生T細胞集団に関する。
T細胞は、細菌若しくはウイルス等の外来の病原体又は癌細胞等の異常な細胞に対する免疫システムにおいて中心的な役割を果たしており、様々な原因によりT細胞の機能が低下し、被験体が易感染性となったり癌等に罹患すると考えられている。このような疾患に対して、免疫細胞等の補充や再生をすることができれば、疾患の病態改善や治療効果の向上等に極めて有効な手段となる。
iPS細胞等の多能性幹細胞から誘導したT細胞を用いた補充療法が提案されている。iPS細胞の原料として、目的の抗原に対して特異的なT細胞を使用することで、元のT細胞と同じ抗原特異性を示す再生T細胞を製造することが可能である(非特許文献1;本文献の内容は、その全体において参照により本明細書に援用される。)。この方法では、樹立されたiPS細胞は、単一細胞由来の細胞集団(コロニー)単位でクローン化されている。これにより、細胞性状が安定しており、目的の細胞・組織への分化効率が良く、遺伝子解析等により変異や異常のないことが確認されたiPS細胞株を利用することが可能になる。ここで、クローンの品質や患者への適合性を確認するために、クローンを1個ずつ検査する必要があるため、従来より、クローン化は重要な工程と考えられている。
一方、ヒト及びマウスを用いた研究において、感染症や腫瘍に対するT細胞補充療法を行う際、遺伝的多様性を有するT細胞集団を用いる方法により、目的とする組織や抗原に対する多角的な免疫反応を誘導することができると同時に、この方法は、抗原の発現低下や変異による免疫逃避(エスケープ)の影響を受けにくいために、高い治療効果が得られることが知られている。
また、従来技術には、末梢血T細胞へのT細胞受容体(TCR)や人工抗原受容体(Chimeric Antigen Receptor : CAR)の遺伝子導入法も存在するが、用いられるT細胞の有する抗原受容体の遺伝子配列は単一である。

iPS細胞株を利用する従来の方法では、クローン化を行うが、このことは、選択したクローン以外を除くことと等しいため、iPS細胞が樹立された段階で存在する遺伝的多様性は完全に失われてしまうことになる。このことは、単一の抗原エピトープを対象とした免疫反応しか利用できないということになる。しかし、単一の抗原エピトープを対象とした免疫反応では、抗原分子の発現低下や変異による免疫逃避(エスケープ)の影響を受けやすく、高い治療効果が得られにくいのではないかという問題がある。一方、従来から行われているT細胞補充療法では、遺伝的多様性を有するT細胞集団を用いることができるが、体外でのT細胞集団の増幅によりT細胞が疲弊し、抗原に対する免疫反応が低下するという問題がある。そのため、T細胞補充療法への利用を目的として、遺伝的多様性を有する再生T細胞集団を製造する新たな方法の開発が望まれる。
従って、本発明の課題は、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する方法及びかかる再生T細胞集団を提供することである。本発明の別の一態様の課題は、前記方法により得られた再生T細胞集団をT細胞補充療法に使用することである。また、本発明のさらに別の一態様の課題は、前記方法の原材料のT細胞集団として腫瘍浸潤T細胞を使用することである。
本発明者は、従来の方法における問題を検討したところ、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造すれば、単一の抗原エピトープを対象とした免疫反応しか利用できないという問題及びT細胞集団の疲弊の問題が解決されることを見出した。即ち、本発明の要旨は、
〔1〕(1)採取した遺伝的多様性を有するT細胞集団から、目的の組織又は抗原に対して指向性を有するT細胞集団を濃縮する工程、(2)前記濃縮したT細胞集団をiPS細胞へと初期化し、遺伝的多様性を維持したままで、iPS細胞を培養する工程、及び(3)前記培養したiPS細胞から遺伝的多様性を有する再生T細胞集団を製造する工程を含む、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する方法、

本発明により、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造することが可能となる。また、本発明の別の一態様では、生体内に存在する遺伝的多様性を有するT細胞集団から、目的の組織又は抗原に対して指向性を有するT細胞を濃縮して利用することにより、製造された再生T細胞集団がT細胞補充療法において高い治療効果を示すことが可能である。特に、腫瘍浸潤T細胞(Tumor Infiltrating Lymphocyte : TIL)は、腫瘍に対して特異的でありかつ多様な抗原を認識することが知られており、TILからiPS細胞への初期化を介して再生TILを製造することにより、遺伝的多様性を有する若返ったT細胞による腫瘍特異的かつ効果的なT細胞補充療法が可能となる。
本発明は、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する方法を提供する。前記方法は、(1)採取した遺伝的多様性を有するT細胞集団から、目的の組織又は抗原に対して指向性を有するT細胞集団を濃縮する工程、(2)前記濃縮したT細胞集団をiPS細胞へと初期化し、遺伝的多様性を維持したまま培養する工程、及び(3)前記培養したiPS細胞から遺伝的多様性を有する再生T細胞集団を製造する工程を含む。
本発明の一態様において、「iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する」とは、遺伝的多様性を有するT細胞集団から、iPS細胞への初期化を介して、遺伝的多様性を有するT細胞集団を再生することである。本発明において、上記過程を経て製造されたT細胞を、「再生T細胞」と称す。
工程(1)
本発明の工程(1)は、採取した遺伝的多様性を有するT細胞集団から、目的の組織又は抗原に対して指向性を有するT細胞集団を濃縮する工程である。

・遺伝的多様性を有するT細胞集団
本発明の一態様において、「遺伝的多様性を有するT細胞集団」とは、生体内に存在するCD3及びCD45が陽性であるリンパ球であり、抗原を認識するT細胞受容体(TCR)の遺伝子配列が集団として多様な細胞集団である。生体内に存在するT細胞は、T前駆細胞が胸腺内で発生分化する際に、TCR遺伝子のランダムな組み換えが起こることで、個々のT細胞が異なるTCR遺伝子配列を有し、あらゆる抗原に対して免疫反応を引き起こすことが可能となっている。個々のT細胞が有するTCRはそれぞれ特異的に認識する抗原又はペプチド配列が決定しているものの、T細胞を集団として捉えることにより、そのT細胞集団が、多様な抗原を免疫対象としていることが理解できる。従って、生体から採取されたT細胞集団は、この意味で、遺伝的多様性を有するT細胞集団である。

本発明の一態様において、T細胞は哺乳動物由来であることが好ましく、ヒト由来であることがより好ましく、治療対象の哺乳動物(好ましくは、ヒト)由来であることがさらに好ましい。T細胞としては、特に限定されないが、例えば、αβT細胞、γδT細胞、ヘルパーT細胞、レギュラトリーT細胞、細胞傷害性T細胞、及びNKT細胞等が挙げられる。また、T細胞の供給源としては、侵襲性が低いことから、末梢血が好ましいが、その他の好ましい供給源として、癌若しくは腫瘍又はその他の臓器若しくは組織、腔水(胸水又は腹水等)、体液(血液等)、リンパ節或いは臍帯血等の体内のあらゆる供給源が挙げられる。本発明の別の態様において、好ましいT細胞は、腫瘍浸潤T細胞である。
本発明の一態様において、遺伝的多様性を有するT細胞集団は、どのような抗原又は組織に対して作用するものでも用いることが可能である。特に、製造した再生T細胞をT細胞補充療法に用いる場合は、採取したT細胞集団が、治療対象の癌、腫瘍、癌抗原、腫瘍特異的変異抗原、又はウイルス等に対する指向性を有することが好ましい。このような治療対象の癌又は腫瘍としては、好ましくは、固形腫瘍(肺癌又は胃癌等)、白血病(急性又は慢性の骨髄性白血病又はリンパ性白血病)又はリンパ腫等の体内に発生しうるあらゆる癌又は腫瘍が挙げられる。また、このような癌抗原、腫瘍特異的変異抗原、又はウイルスとしては、好ましくは、各腫瘍に特異的又は非特異的に発現するWT1、MUC1、EGFRvIII、HER-2/neu、MAGE A3、p53 nonmutant、NY-ESO-1、PSMA、GD2、CEA、MelanA/MART1又はSurvivin等の癌抗原、遺伝子変異を伴ったp53、Ras、ERG、bcr-abl、又は新規抗原(Neoantigen)、或いは各種ウイルス由来のタンパクであるLMP2、HPV E6若しくはE7、EB-NA、又はHTLV-1 Tax等が挙げられる。
・目的の組織又は抗原に対して指向性を有するT細胞集団を濃縮するステップ
生体内には、あらゆる抗原に対して反応できるように、非常に多様なT細胞が存在しているが、大半がT細胞補充療法で目的とする抗原とは無関係なT細胞である。そのため、iPS細胞への初期化を介して製造するT細胞の原料として、T細胞補充療法の対象となる癌、腫瘍又はウイルスに反応性のあるT細胞集団を、あらかじめ濃縮しておくことで、治療効果が飛躍的に高まると同時に、想定外の抗原に対する免疫反応を抑制し安全性を高めることが可能となる。
生体内には、あらゆる抗原に対して反応できるように、非常に多様なT細胞が存在しているが、大半がT細胞補充療法で目的とする抗原とは無関係なT細胞である。そのため、iPS細胞への初期化を介して製造するT細胞の原料として、T細胞補充療法の対象となる癌、腫瘍又はウイルスに反応性のあるT細胞集団を、あらかじめ濃縮しておくことで、治療効果が飛躍的に高まると同時に、想定外の抗原に対する免疫反応を抑制し安全性を高めることが可能となる。
本発明の一態様において、「目的の組織又は抗原」とは、好ましくは、上記した治療対象の癌、腫瘍、癌抗原、腫瘍特異的変異抗原、若しくはウイルスを含む組織、又はそれらに含まれる抗原である。
本発明の一態様において、「指向性を有する」とは、個々のT細胞がそれぞれ別の抗原又はペプチド配列を特異的に認識しつつ、集団として目的の組織又は抗原に対する免疫反応を示すことである。
本発明の一態様において、目的の組織又は抗原に対して指向性を有するT細胞集団の濃縮方法としては、例えば、抗原タンパク質又はペプチドでの刺激によりT細胞集団を活性化し、増殖したT細胞を分離する方法が挙げられる。この方法では、抗原提示細胞を含む単核球分画を培養し、抗原タンパク質又はペプチドを培養液中に添加する。繰り返しの抗原タンパク質又はペプチドでの刺激により、反応性のT細胞は増殖し、T細胞集団中に占める割合が上昇する。一方、添加した抗原タンパク質又はペプチドに反応しないT細胞は活性化されることがなく、in vitroでの培養中に徐々に割合が低下し、最終的には死んでしまう。そのため、一定の培養期間後には、添加した抗原タンパク質又はペプチドに特異的な反応性を有しつつ、認識する抗原又はペプチド配列において多様性を有するT細胞集団が得られる。ここで、「抗原タンパク質又はペプチド」としては、上記目的の組織又は抗原として記載したものが好ましく、例えば、癌抗原、腫瘍特異的変異抗原又はウイルスとして上記したものをペプチド断片化したもの等を使用することができる。また、抗原ペプチドにより刺激されたT細胞は各種の活性化分子又はサイトカインを発現又は放出することが知られている。本発明の一態様において、この性質を利用し、それらの分子に特異的な抗体を結合させた上で、磁気ビーズを用いた磁気的分離又は蛍光標識を用いたフローサイトメーターによる選択的な分離に供することも可能である。また、本発明の一態様において、サイトカイン放出細胞を2極性の抗体(細胞側及びサイトカイン側)で捕捉するシステムを使用することができる。
本発明の別の一態様において、目的の組織又は抗原に対して指向性を有するT細胞集団の濃縮方法としては、例えば、テトラマー又はデキストラマー等の抗原ペプチド-HLA複合体のオリゴマーを用いる方法が挙げられる。この方法では、特定のHLA型及びそのHLAが提示可能な目的の抗原ペプチドの複合体から成るオリゴマーに蛍光色素を標識し、オリゴマーが結合した細胞をフローサイトメーターを用いてソーティングすることにより、目的の組織又は抗原に対して指向性を有するT細胞を得ることができる。
工程(2)
本発明の工程(2)は、前記濃縮したT細胞集団をiPS細胞へと初期化し、遺伝的多様性を維持したまま培養する工程である。

・濃縮したT細胞集団をiPS細胞へと初期化するステップ
iPS細胞の製造方法は当該分野で公知である。本発明の一態様において、iPS細胞は、好ましくは、工程(1)で濃縮したT細胞集団へ初期化因子を導入することによって製造される。ここで、初期化因子としては、例えば、Oct3/4、Sox2、Sox1、Sox3、Sox15、Sox17、Klf4、Klf2、c-Myc、N-Myc、L-Myc、Nanog、Lin28、Fbx15、ERas、ECAT15-2、Tcl1、beta-catenin、Lin28b、Sall1、Sall4、Esrrb、Nr5a2、Tbx3又はGlis1等の遺伝子又は遺伝子産物が挙げられ、これらの初期化因子は、単独で用いても良く、組み合わせて用いても良い。
iPS細胞の製造方法は当該分野で公知である。本発明の一態様において、iPS細胞は、好ましくは、工程(1)で濃縮したT細胞集団へ初期化因子を導入することによって製造される。ここで、初期化因子としては、例えば、Oct3/4、Sox2、Sox1、Sox3、Sox15、Sox17、Klf4、Klf2、c-Myc、N-Myc、L-Myc、Nanog、Lin28、Fbx15、ERas、ECAT15-2、Tcl1、beta-catenin、Lin28b、Sall1、Sall4、Esrrb、Nr5a2、Tbx3又はGlis1等の遺伝子又は遺伝子産物が挙げられ、これらの初期化因子は、単独で用いても良く、組み合わせて用いても良い。
・樹立されたiPS細胞を遺伝的多様性を維持したまま培養するステップ
iPS細胞を培養する場合、好ましくは、継代が行われる。しかし、一般的な継代方法では、継代に用いる細胞以外の細胞は破棄されてしまうので、遺伝的多様性を有するT細胞集団から樹立されたiPS細胞集団の多様性が次第に失われていってしまう。

これに対して、本発明の一態様においては、好ましくは、iPS細胞を樹立した後、培養器を洗浄して、好ましくはiPS細胞以外の細胞を除去し、iPS細胞をクローン化せずに回収し、別の培養器に継代し、このような継代を繰り返す。
本発明の一態様において、本ステップで用いられる培養液としては、特に限定されないが、動物細胞の培養に用いられる培地を基礎培地とし、これにiPS細胞の未分化能を維持するためのサイトカイン類を添加して調製することができる。基礎培地としては、例えばIscove's Modified Dulbecco's Medium (IMDM)培地、Medium 199培地、Eagle's Minimum Essential Medium (EMEM)培地、αMEM培地、Dulbecco's modified Eagle's Medium (DMEM)培地、Ham's F12培地、RPMI 1640培地、Fischer's培地、Neurobasal Medium(ライフテクノロジーズ)、StemFit AK03N(味の素)及びこれらの混合培地等が挙げられる。培地には、血清が含有されていてもよいし、又は無血清であってもよい。サイトカイン類としては、好ましくは、bFGFが挙げられ、培養液中におけるその濃度は、例えば、1~100μg/mL(好ましくは50μg/mL)である。
本発明の一態様において、iPS細胞の培養方法は、接着培養又は浮遊培養であってもよいが、接着培養が好ましい。iPS細胞の分離方法としては、例えば、力学的に分離する方法、プロテアーゼ活性を有する分離溶液、コラゲナーゼ活性を有する分離溶液、又はプロテアーゼ活性及びコラゲナーゼ活性を有する分離溶液(例えば、Accutase(商標)及びAccumax(商標)等)を用いた分離方法が挙げられる。
本発明の一態様において、iPS細胞は、好ましくは、1x103~1x104cells/cm2、1x104~1x105cells/cm2、1x105~1x106cells/cm2の細胞密度になった場合に、別の培養器に継代される。継代の回数は、T細胞補充療法に必要な量のiPS細胞が得られれば何回でもよく、好ましくは、1~5回、5~10回である。
本発明の一態様において、得られた遺伝的多様性を有するiPS細胞は、そのまま使用してもよいし、又は必要な時まで凍結保存してもよい。
工程(3)
本発明の工程(3)は、前記培養したiPS細胞から遺伝的多様性を有する再生T細胞集団を製造する工程である。

・iPS細胞から造血前駆細胞を誘導するステップ
造血前駆細胞(Hematopoietic Progenitor Cells (HPC) )とは、リンパ球、好酸球、好中球、好塩基球、赤血球、又は巨核球等の血球系細胞に分化可能な細胞である。本発明の一態様において、造血前駆細胞と造血幹細胞は、区別されるものではなく、特に断りがなければ同一の細胞を示す。造血幹細胞/前駆細胞は、例えば、表面抗原であるCD34及び/又はCD43が陽性であることによって認識できる。
造血前駆細胞(Hematopoietic Progenitor Cells (HPC) )とは、リンパ球、好酸球、好中球、好塩基球、赤血球、又は巨核球等の血球系細胞に分化可能な細胞である。本発明の一態様において、造血前駆細胞と造血幹細胞は、区別されるものではなく、特に断りがなければ同一の細胞を示す。造血幹細胞/前駆細胞は、例えば、表面抗原であるCD34及び/又はCD43が陽性であることによって認識できる。
本発明の一態様において、造血前駆細胞は、好ましくは、ビタミンC類を添加した培養液中でiPS細胞を培養することによって製造される。ここで、「ビタミンC類」とは、L-アスコルビン酸及びその誘導体を意味し、「L-アスコルビン酸誘導体」とは、生体内で酵素反応によりビタミンCとなるものを意味する。L-アスコルビン酸の誘導体としては、例えば、リン酸ビタミンC、アスコルビン酸グルコシド、アスコルビルエチル、ビタミンCエステル、テトラヘキシルデカン酸アスコルビル、ステアリン酸アスコルビル及びアスコルビン酸-2リン酸-6パルミチン酸が挙げられる。L-アスコルビン酸の誘導体は、好ましくは、リン酸ビタミンCであり、例えば、リン酸-Lアスコルビン酸Na又はリン酸-L-アスコルビン酸Mg等のリン酸-Lアスコルビン酸塩が挙げられる。ビタミンC類は、例えば、培養液において、5μg/mlから500μg/mlの濃度で含有される。
本発明の一態様において、造血前駆細胞の製造に用いる培養液は、特に限定されないが、動物細胞の培養に用いられる培地を基礎培地とし、これにビタミンC類等を添加して調製することができる。基礎培地としては、例えば、Iscove's Modified Dulbecco's Medium (IMDM)培地、Medium 199培地、Eagle's Minimum Essential Medium (EMEM)培地、αMEM培地、Dulbecco's modified Eagle's Medium (DMEM)培地、Ham's F12培地、RPMI 1640培地、Fischer's培地、Neurobasal Medium (ライフテクノロジーズ)、StemPro34(ライフテクノロジーズ)及びこれらの混合培地等が挙げられる。培地には、血清が含有されていてもよいし、又は無血清であってもよい。必要に応じて、基礎培地は、例えば、アルブミン、インスリン、トランスフェリン、セレン、脂肪酸、微量元素、2-メルカプトエタノール、チオールグリセロール、脂質、アミノ酸、L-グルタミン、非必須アミノ酸、ビタミン、増殖因子、低分子化合物、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類、及びサイトカイン等の1つ以上の物質も含有し得る。
本発明の一態様において、造血前駆細胞の製造に用いる培養液は、BMP4 (Bone morphogenetic protein 4)、VEGF (vascular endothelial growth factor)、bFGF (basic fibroblast growth factor)、SCF (Stem cell factor)、TPO (トロンボポエチン)、及びFLT-3L (Flt3 Ligand)から成る群より選択されるサイトカインがさらに添加されていてもよい。
本発明の一態様において、これらの濃度は、例えば、BMP4は1 ng/mlから100 ng/mlであり、VEGFは1 ng/mlから100 ng/mlであり、bFGFは1 ng/mlから100 ng/mlであり、SCFは10 ng/mlから100 ng/mlであり、TPOは1ng/mlから100 ng/mlであり、FLT-3Lは1 ng/mlから100 ng/mlである。
本発明の一態様において、TGFβ阻害剤が培養液に添加されてもよい。「TGFβ阻害剤」とは、TGFβファミリーのシグナル伝達に干渉する低分子阻害剤であり、例えばSB431542、SB202190(以上、R.K.Lindemann et al., Mol. Cancer 2: 20 (2003))、SB505124 (GlaxoSmithKline)、NPC30345、SD093、SD908、SD208 (Scios)、LY2109761、LY364947、及びLY580276 (Lilly Research Laboratories)等が挙げられ、培地中の濃度は、好ましくはO.5μM~100μMである。
本発明の一態様において、iPS細胞は、C3H10T1/2 (Takayama N., et al. J Exp Med. 2817-2830, 2010)、又は異種由来のストローマ細胞 (Niwa A et al. J Ce11 Physiol. 2009 Nov; 221 (2) :367-77.)等のフィーダー細胞と共培養されてもよいが、好ましくは、フィーダー細胞を使用せずにiPS細胞の培養が行われる。
本発明の一態様において、造血前駆細胞の製造時のiPS細胞の培養方法は、接着培養又は浮遊培養であってもよいが、浮遊培養が好ましい。例えば、iPS細胞は、使用したディッシュに対して80%コンフルエントになるまで培養されたコロニーを分離し、単細胞に解離させたのちに、浮遊培養に供することができる。iPS細胞の分離方法としては、例えば、力学的に分離する方法、プロテアーゼ活性及びコラゲナーゼ活性を有する分離溶液(例えば、Accutase(商標)及びAccumax(商標)等)、又はコラゲナーゼ活性を有する分離溶液を用いた分離方法が挙げられる。
浮遊培養とは、細胞を培養容器に対して非接着の状態で培養することである。本発明の一態様において、浮遊培養は、特に限定はされないが、細胞との接着性を向上させる目的で人工的に処理(例えば、細胞外マトリックス等によるコーティング処理)されていない培養容器、又は人工的に接着を抑制する処理(例えば、ポリヒドロキシエチルメタクリル酸(po1y-HEMA)若しくは非イオン性の界面活性ポリオール(Pluronic F-127等)によるコーティング処理)した培養容器を使用して行うことができる。浮遊培養の際には、胚様体(EB)を形成させて培養することが好ましい。
本発明の別の一態様では、造血前駆細胞は、iPS細胞を培養することで得られるネット様構造物(iPS-sacとも称する)から調製することもできる。ここで、「ネット様構造物」とは、iPS細胞由来の立体的な嚢状(内部に空間を伴うもの)構造体で、内皮細胞集団等で形成され、内部に造血前駆細胞を含む構造体である。
本発明の一態様において、造血前駆細胞を製造するために培養する際の温度条件は、特に限定されないが、例えば、約37℃~約42℃程度、約37℃~約39℃程度が好ましい。また、培養期間については、当業者であれば造血前駆細胞の数等をモニターしながら、適宜決定することが可能である。造血前駆細胞が得られる限り、日数は特に限定されないが、例えば、少なくとも6日以上、7日以上、8日以上、9日以上、10日以上、11日以上、12日以上、13日以上、14日以上であり、好ましくは14日である。培養期間が長いことについては、造血前駆細胞の製造においては問題とされない。また、低酸素条件で培養してもよく、本発明の一態様において、低酸素条件としては、例えば、15%、10%、9%、8%、7%、6%、5%又はそれら以下の酸素濃度が挙げられる。
・造血前駆細胞からCD4CD8両陽性T細胞を誘導するステップ
本発明において、「CD4CD8両陽性T細胞」とは、T細胞のうち、表面抗原のCD4及びCD8が共に陽性である細胞(CD8+CD4+)を意味し、T細胞は、表面抗原であるCD3及びCD45が陽性であることによって認識できることから、CD4CD8両陽性T細胞は、CD4、CD8、CD3及びCD45が陽性である細胞として同定することができる。CD4CD8両陽性T細胞は、誘導によってCD4陽性細胞又はCD8陽性細胞へと分化させることができる。
本発明において、「CD4CD8両陽性T細胞」とは、T細胞のうち、表面抗原のCD4及びCD8が共に陽性である細胞(CD8+CD4+)を意味し、T細胞は、表面抗原であるCD3及びCD45が陽性であることによって認識できることから、CD4CD8両陽性T細胞は、CD4、CD8、CD3及びCD45が陽性である細胞として同定することができる。CD4CD8両陽性T細胞は、誘導によってCD4陽性細胞又はCD8陽性細胞へと分化させることができる。
本発明の一態様において、CD4CD8両陽性T細胞は、p38阻害剤及び/又はSDF-1を添加した培養液中で造血前駆細胞を培養する工程を含む方法によって製造することができる。
<p38阻害剤>
本発明において、「p38阻害剤」とは、p38タンパク質(p38MAPキナーゼ)の機能を阻害する物質として定義される。本発明の一態様において、例えば、p38の化学的阻害剤、p38のドミナントネガティブ変異体、又はそれをコードする核酸等が挙げられるが、これらに限定されない。
本発明において、「p38阻害剤」とは、p38タンパク質(p38MAPキナーゼ)の機能を阻害する物質として定義される。本発明の一態様において、例えば、p38の化学的阻害剤、p38のドミナントネガティブ変異体、又はそれをコードする核酸等が挙げられるが、これらに限定されない。

本発明の一態様において、p38のドミナントネガティブ変異体としては、p38のDNA結合領域に位置する180位のスレオニンをアラニンに点変異させたp38T180A、並びにヒト及びマウスにおけるp38の182位のチロシンをフェニルアラニンに点変異させたp38Y182F等が挙げられる。
p38阻害剤は、例えば、約1μM~約50μMの範囲で培地に含有される。
<SDF-1>
本発明の一態様において、SDF-1 (Stromal cell-derived factor 1)は、SDF-1α又はその成熟型だけでなく、SDF-1β、SDF-1γ、SDF-1δ、SDF-1ε若しくはSDF-1φ等のアイソフォーム、又はそれらの成熟型であってもよく、或いはこれらの任意の割合の混合物等であってもよい。好ましくは、SDF-1αが使用される。尚、SDF-1は、CXCL-12又はPBSFと称される場合もある。
本発明の一態様において、SDF-1 (Stromal cell-derived factor 1)は、SDF-1α又はその成熟型だけでなく、SDF-1β、SDF-1γ、SDF-1δ、SDF-1ε若しくはSDF-1φ等のアイソフォーム、又はそれらの成熟型であってもよく、或いはこれらの任意の割合の混合物等であってもよい。好ましくは、SDF-1αが使用される。尚、SDF-1は、CXCL-12又はPBSFと称される場合もある。
本発明の一態様において、SDF-1は、そのケモカインとしての活性を有する限り、そのアミノ酸配列中の1又は数個のアミノ酸が置換、欠失及び/又は付加されていてもよく、また同様に糖鎖が置換、欠失及び/又は付加されていてもよい。SDF-1において少なくとも4つのシステイン残基(ヒトSDF-1αの場合、Cys30、Cys32、Cys55及びCys71)が保持されており、かつ天然体のアミノ酸配列に対して90%以上の同一性を有する範囲であれば、アミノ酸変異が許容される。SDF-1は、哺乳動物、例えば、ヒト、例えば、サル、ヒツジ、ウシ、ウマ、ブタ、イヌ、ネコ、ウサギ、ラット、又はマウス等の非ヒト哺乳動物のものであってもよい。例えば、ヒトのSDF-1αとして、GenBank登録番号:NP_954637で登録されているタンパク質が使用でき、SDF-1βとして、GenBank登録番号:NP_000600で登録されているタンパク質が使用できる。
本発明の一態様において、SDF-1は、市販のものを使用してもよいし、天然から精製されたものを使用してもよいし、又はペプチド合成や遺伝子工学的手法によって製造されたものを使用してもよい。
本発明の一態様において、SDF-1は、例えば、約10 ng/mlから約100 ng/mlの範囲で培地に含有される。
本発明の一態様において、CD4CD8両陽性T細胞の製造に用いる培養液は、特に限定されないが、動物細胞の培養に用いられる培地を基礎培地とし、これにp38阻害剤及び/又はSDF-1、さらに好ましくはビタミンC類を添加して調製することができる。なお、CD4CD8両陽性T細胞製造工程で使用されるビタミンC類の種類は、例えば上述のとおりであるが、ビタミンC類の濃度は、例えば、5μg/mlから200μg/mlである。基礎培地としては、例えばIscove's Modified Dulbecco's Medium (IMDM) 培地、Medium 199培地、Eagle's Minimum Essential Medium (EMEM)培地、αMEM培地、Dulbecco's modified Eagle's Medium (DMEM)培地、Ham's F12培地、RPMI 1640培地、Fischer's培地、OP9培地、Neurobasal Medium (ライフテクノロジーズ)、及びこれらの混合培地等が挙げられる。培地には、血清が含有されていてもよいし、又は無血清であってもよい。必要に応じて、基礎培地は、例えば、アルブミン、インスリン、トランスフェリン、セレン、脂肪酸、微量元素、2-メルカプトエタノール、チオールグリセロール、脂質、アミノ酸、L-グルタミン、非必須アミノ酸、ビタミン、増殖因子、低分子化合物、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類、及びサイトカイン等の1つ以上の物質も含有し得る。
本発明の一態様において、CD4CD8両陽性T細胞の製造に用いる培養液は、SCF、TPO (トロンボポエチン)、FLT-3L及びIL-7から成る群より選択されるサイトカインをさらに培養液に添加してもよい。これらの濃度は、例えば、SCFは10 ng/mlから100 ng/mlであり、TPOは1O ng/mlから200ng/mlであり、FLT-3Lは1 ng/mlから100 ng/mlであり、IL-7は1 ng/mlから100 ng/mlである。
本発明の一態様において、CD4CD8両陽性T細胞の製造時に、造血前駆細胞はフィーダー細胞を用いて培養してもよいが、好ましくはフィーダー細胞を用いずに培養を行う。
本発明の一態様において、造血前駆細胞は接着培養又は浮遊培養してもよいが、本ステップは接着培養が好ましい。接着培養の場合、培養容器をコーティングして用いてもよく、例えば、コーティング剤としては、マトリゲル(Niwa A, et al. PLoS One. 6(7):e22261, 2011) 、コラーゲン、ゼラチン、ラミニン、ヘパラン硫酸プロテオグリカン、レトロネクチン、Fc-DLL4又はエンタクチン、及びこれらの組み合わせが挙げられる。
本発明の別の一態様において、胚様体を浮遊培養して造血前駆細胞を得た場合は、これを単細胞に解離させたのちに、接着培養を行うことが好ましい。
本発明の一態様において、CD4CD8両陽性T細胞を製造するために造血前駆細胞を培養する際の培養温度条件は、特に限定されないが、例えば、約37℃~約42℃程度、約37℃~約39℃程度が好ましい。また、培養期間については、当業者であればCD4CD8両陽性T細胞の数等をモニターしながら、適宜決定することが可能である。CD4CD8両陽性T細胞が得られる限り、日数は特に限定されないが、例えば、少なくとも10日以上、12日以上、14日以上、16日以上、18日以上、20日以上、22日以上、23日以上であり、好ましくは23日である。
本発明の一態様において、得られたCD4CD8両陽性T細胞は、単離して用いても良く、他の細胞種が含有される細胞集団として用いても良い。単離する場合、CD4、CD8、CD3及びCD45から成るいずれか一つの指標を用いて単離することができ、当該単離の方法は、当業者に周知の方法を用いることができ、例えば、CD4、CD8、CD3及びCD45の抗体により標識し、フローサイトメーターを用いて単離する方法、又は所望の抗原を固定化したアフィニティカラム等を用いて精製する方法が挙げられる。
・CD4CD8両陽性T細胞からCD8陽性T細胞を誘導するステップ
「CD8陽性T細胞」とは、T細胞のうち、表面抗原のCD8が陽性である細胞(CD8+CD4-)を意味し、細胞傷害性T細胞とも呼ばれる。T細胞は、表面抗原であるCD3及びCD45が陽性であることによって認識できることから、CD8陽性T細胞は、CD8、CD3及びCD45が陽性であり、CD4が陰性である細胞として同定することができる。
「CD8陽性T細胞」とは、T細胞のうち、表面抗原のCD8が陽性である細胞(CD8+CD4-)を意味し、細胞傷害性T細胞とも呼ばれる。T細胞は、表面抗原であるCD3及びCD45が陽性であることによって認識できることから、CD8陽性T細胞は、CD8、CD3及びCD45が陽性であり、CD4が陰性である細胞として同定することができる。
本発明の一態様において、CD8陽性T細胞は、副腎皮質ホルモン剤を添加した培養液中でCD4CD8両陽性T細胞を培養する工程を含む方法によって製造することができる。
本発明の一態様において、副腎皮質ホルモン剤は、好ましくは、糖質コルチコイド又はその誘導体であり、例えば、酢酸コルチゾン、ヒドロコルチゾン、酢酸フルドロコルチゾン、プレドニゾロン、トリアムシノロン、メチルプレドニゾロン、デキサメタゾン、ベタメタゾン、又はプロピオン酸ベクロメタゾンが挙げられる。好ましくは、副腎皮質ホルモン剤は、デキサメタゾンである。培養液中におけるその濃度は、例えば、1nMから100nMである。
本発明の一態様において、CD8陽性T細胞の製造に用いる培養液は、特に限定されないが、動物細胞の培養に用いられる培地を基礎培地とし、これに副腎皮質ホルモン剤を添加して調製することができる。基礎培地としては、例えばIscove's Modified Dulbecco's Medium (IMDM) 培地、Medium 199培地、Eagle's Minimum Essential Medium (EMEM)培地、αMEM培地、Dulbecco's modified Eagle's Medium (DMEM)培地、Ham's F12培地、RPMI 1640培地、Fischer's培地、Neurobasal Medium(ライフテクノロジーズ)、及びこれらの混合培地等が挙げられる。培地には、血清が含有されていてもよいし、又は無血清であってもよい。必要に応じて、基礎培地は、例えば、アルブミン、インスリン、トランスフェリン、セレン、脂肪酸、微量元素、2-メルカプトエタノール、チオールグリセロール、脂質、アミノ殿、L-グルタミン、非必須アミノ酸、ビタミン、増殖因子、低分子化合物、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類、及びサイトカイン等の1つ以上の物質も含有し得る。
本発明の一態様において、CD8陽性T細胞の製造に用いる培養液は、抗CD3抗体、ビタミンC類、又はサイトカインをさらに含有することが好ましい。当該サイトカインとしては、例えば、IL-2及びIL-7等が挙げられる。
本発明の一態様において、抗CD3抗体としては、CD3を特異的に認識する抗体であれば特に限定されないが、例えば、OKT3クローンから産生される抗体が挙げられる。抗CD3抗体の培養液中における濃度は、例えば、10ng/mlから1000ng/mlである。
本発明の一態様において、CD8陽性T細胞の製造に用いるビタミンC類は、例えば上述したものであり、上述と同じ条件で用いることができる。
本発明の一態様において、CD8陽性T細胞の製造に用いるサイトカインの培養液中における濃度は、例えば、IL-2は10 U/mlから1000 U/mlであり、IL-7は1ng/mlから100ng/mlである。
本発明の一態様において、CD8陽性T細胞を製造するためにCD4CD8両陽性T細胞を培養する際の温度条件は、特に限定されないが、例えば、約37℃~約42℃程度、約37℃~約39℃程度が好ましい。また、培養期間については、当業者であればCD8陽性T細胞の数等をモニターしながら、適宜決定することが可能である。CD8陽性T細胞が得られる限り、日数は特に限定されないが、例えば、少なくとも1日以上、2日以上、3日以上、4日以上、5日以上であり、好ましくは3日である。
本発明の製造方法により得られる再生T細胞集団としては、特に限定されないが、例えば、αβT細胞集団、γδT細胞集団、ヘルパーT細胞集団、レギュラトリーT細胞集団、細胞傷害性T細胞集団、NKT細胞集団及び腫瘍浸潤T細胞集団等が挙げられる。
本発明の一態様において、維持される遺伝的多様性の程度は、好ましくは、原料の遺伝的多様性を有するT細胞集団の30%以上、40%以上、50%以上である。
本発明の一態様において、製造された再生T細胞集団は、好ましくは、T細胞補充療法に使用される。本発明において、T細胞補充療法とは、T細胞を生体に補充する治療法を意味し、本発明の一態様において、T細胞補充療法としては、好ましくは、再生したT細胞の輸注、腫瘍内注入、動注、門脈注等を含む細胞移植による治療法が挙げられる。
本発明の一態様は、生体内に存在するT細胞集団の遺伝的多様性を維持している、iPS細胞を介して得られる再生T細胞集団、例えば、上記方法により得られる再生T細胞集団を含む。本発明の再生T細胞集団を含む医薬組成物は、がん治療対象を処置するために使用することができる。本発明の一態様には、本発明の医薬組成物を用いるがん治療方法が含まれる。本発明の医薬組成物は、薬学的に許容される添加剤を含んでいてもよい。該添加剤としては、例えば、細胞培養液、リン酸緩衝食塩水等が挙げられる。
本発明の一態様には、本発明の製造方法により得られる再生T細胞集団をT細胞補充療法に適用して治療する方法が含まれる。また、本発明の一態様には、T細胞補充療法による治療における使用のための本発明の製造方法により得られる再生T細胞集団が含まれる。
本発明の一態様は、上記説明に列挙された全ての一般的態様及び/又は具体的態様の全ての組み合わせを含む。
本発明を以下の実施例でさらに具体的に説明するが、本発明の範囲はそれら実施例に限定されない。
工程(1)の具体的手順
採取したT細胞集団に対し、CD3及びCD28抗体で標識された磁気ビーズを用いて活性化刺激を与え、200U/ml IL-2、10ng/ml IL-7、10ng/ml IL-15を添加したRPMI-1640培地(10%ウシ胎児血清、1%Penicillin-Streptomycin-Glutamineを含む)中で2日間培養を行った。
採取したT細胞集団に対し、CD3及びCD28抗体で標識された磁気ビーズを用いて活性化刺激を与え、200U/ml IL-2、10ng/ml IL-7、10ng/ml IL-15を添加したRPMI-1640培地(10%ウシ胎児血清、1%Penicillin-Streptomycin-Glutamineを含む)中で2日間培養を行った。
工程(2)の具体的手順(iPS細胞への初期化)
続いて、T細胞を15mlチューブへ回収し、約250μlのRPMI-1640培地に浮遊させた後、CytoTune-iPS 2.0キット(株式会社IDファーマ社:#DV-0304)を用いて、MOI=5となる力価で初期化に必要な4因子(Oct3/4、Sox2、Klf4及びc-Myc)を含むセンダイウイルスを培地に添加し、37℃で2時間インキュベートを行った。インキュベート後には、RPMI-1640培地を用いてT細胞を洗浄し、培地中からウイルスを除去した。
続いて、T細胞を15mlチューブへ回収し、約250μlのRPMI-1640培地に浮遊させた後、CytoTune-iPS 2.0キット(株式会社IDファーマ社:#DV-0304)を用いて、MOI=5となる力価で初期化に必要な4因子(Oct3/4、Sox2、Klf4及びc-Myc)を含むセンダイウイルスを培地に添加し、37℃で2時間インキュベートを行った。インキュベート後には、RPMI-1640培地を用いてT細胞を洗浄し、培地中からウイルスを除去した。
続いて磁気ビーズを除くために、ピペッティングによりビーズと細胞とを分離させた後、磁石付きのスタンドにチューブを静置することにより、培地中に浮遊したT細胞のみを回収した。
得られたT細胞はiMatrix-511(nippi社:#892011)を0.5μg/cm2でコーティングした6cmディッシュ上に播種し、RPMI-1640培地中で、37℃、5%CO2、5%O2に設定したインキュベーターを用いて培養を開始した。
6cmディッシュ上で培養を開始した翌日から、細胞が含まれていない培養上清を半量除去し、StemFit AK03N(味の素社)に1mMバルプロ酸を添加した樹立用培地を、除去した培地と同量加えた。
以降、同様の半量培地交換を毎日行いながら、20~40日間培養を続けることで、6cmディッシュ中に多数のiPS細胞コロニーを得た。一定量のコロニーが得られた段階で、樹立用培地からiPS細胞用培地(StemFit AK03Nのみ)に切り替えて培養を進めた。
工程(2)の具体的手順(樹立したiPS細胞の遺伝的多様性を維持したままの培養)
iPS細胞のコロニーが肉眼で確認できるほど大きくなった段階で、iPS細胞を、新しくiMatrix-511をコーティングした6cmディッシュへと継代した。

継代では、PBS(-)を用いてディッシュに接着しているiPS細胞を洗浄した後、TrypLeSelect(ライフテクノロジーズ社:A12859-01)を添加して約7分インキュベートすることにより、iPS細胞の接着を解除し、単一の細胞へと分散させた。細胞をiPS細胞用培地で洗浄し、細胞数を計測した。そして、分散させた細胞を、新しくiMatrix-511コートした6cmディッシュに1,500 cells/cm2になるように播種した。この際、回収されたiPS細胞を、破棄することなく、全て新しいディッシュへと播種した。
回収した細胞を全て植え継ぐことにより、次第に培養スケールが増大していく。そこで、樹立後から3~5継代を行い、iPS細胞を用いたT細胞補充療法に必要な量のiPS細胞が得られた段階で、以下の手順で細胞を凍結保存した。
継代と同じ手順でiPS細胞を回収したのち、TCプロテクター(DSファーマ社:#KBTCP001)等の細胞凍結液に細胞を浮遊させ、-1℃/分の速さで-80℃まで温度を低下させることにより、iPS細胞を凍結した。iPS細胞を安定的かつ長期間保存するためには、さらに液体窒素(-196℃)中で保存することが望ましい。
工程(3)の具体的手順
超低接着処理された6well plate(CORNING社: #3471)に、工程(2)で得られたiPS細胞を3x105~6x105個/ウェルで播種し(DayO)、EB培地(StemPro34に10μg/mlヒトインスリン、5.5μg/ml ヒトトランスフェリン、5ng/ml 亜セレン酸ナトリウム、2mM L-グルタミン、45mM α-モノチオグリセロール、及び50μg/ml アスコルビン酸を添加)に10 ng/ml BMP4、5ng/ml bFGF、15ng/ml VEGF、2μM SB431542を加えて、低酸素条件下(5%O2)にて5日間培養を行った(Day5)。
超低接着処理された6well plate(CORNING社: #3471)に、工程(2)で得られたiPS細胞を3x105~6x105個/ウェルで播種し(DayO)、EB培地(StemPro34に10μg/mlヒトインスリン、5.5μg/ml ヒトトランスフェリン、5ng/ml 亜セレン酸ナトリウム、2mM L-グルタミン、45mM α-モノチオグリセロール、及び50μg/ml アスコルビン酸を添加)に10 ng/ml BMP4、5ng/ml bFGF、15ng/ml VEGF、2μM SB431542を加えて、低酸素条件下(5%O2)にて5日間培養を行った(Day5)。
続いて、50ng/ml SCF、30ng/ml TPO、10ng/ml Flt3Lを添加し、さらに5~9日間培養を行った(~Day14)。
得られた造血前駆細胞は、Fc-DLL4(5μg/ml)(Sino Biological Inc.)及びRetronectin(5μg/ml)(タカラバイオ株式会社)でコーティングした48well plate上で、50ng/ml SCF、50ng/ml IL-7、50ng/ml Flt3L、100ng/ml TPO、15μM SB203580 (Tocris Bioscience)、30ng/ml SDF-1α(PeproTech)を添加したOP9培地(15% FBS、2mM L-グルタミン、100U/mlペニシリン、100ng/ml ストレプトマイシン、55μM 2-メルカプトエタノール、50μg/ml アスコルビン酸、10μg/ml ヒトインスリン、5.5μg/ml ヒトトランスフェリン、5ng/ml 亜セレン酸ナトリウムを添加した)中で21日間培養を行った(Day35)。
Day35にて、CD45、CD3、CD4及びCD8の全てが陽性の画分をFACSを用いて単離し、CD4CD8両陽性(Double positive)細胞(DP細胞という)を得た。
得られたCD4CD8両陽性細胞を、96well plate上で、15 % ウシ胎児血清、500 ng/ml 抗CD3抗体(eBioscience)、200 U/ml IL-2、10 ng/ml IL-7、及び10 nM デキサメタゾンを添加したRPMI 1640培地中で2日間培養した(Day37)。細胞を15% ウシ胎児血清入りのRPMI 1640培地で洗浄することにより抗体を除去し、15 % ウシ胎児血清及び10 ng/ml IL-7を添加したRPMI 1640培地でさらに5日間培養することで、CD8陽性T細胞を得た(Day42)。
実施例1
前記の方法を用いて、肺癌患者の摘出腫瘍から分離された遺伝的多様性を有するT細胞集団からiPS細胞を樹立し、遺伝的多様性を有するT細胞へと再分化誘導を行った。

T細胞が発現するT細胞受容体(TCR)の遺伝的多様性は、一般的に、DNAやRNAのシークエンス解析により決定されるが、TCRのVβ鎖の各セグメントに特異的に結合する抗体パネルを用いて簡易に検出することも可能である。
TCRVβ解析キット(BECKMAN COULTER社:#IM-3497)を用いて、肺癌患者の摘出腫瘍から分離されたT細胞集団のTCRレパトアを解析した(図1)。患者検体中には、約1~13%の割合でそれぞれのVβ鎖を発現する細胞が含まれており、遺伝的多様性を有するT細胞集団であることが確認された。これらのT細胞集団を上記の方法によりiPS細胞へと初期化した。
iPS細胞は多能性を有する幹細胞であり、TCR遺伝子を含むT細胞関連の遺伝子は発現していない。つまり、TCR遺伝子の多様性を未分化なiPS細胞段階で解析することは困難である。樹立されたiPS細胞は、遺伝的多様性を失わないように継代したのち、造血前駆細胞段階を経てT細胞へと分化誘導を行った。
再生されたT細胞が発現するTCRのレパトアを、上記と同じキットを用いて解析した(図2)。iPS細胞の樹立及びT細胞の分化誘導に伴って、Vβの2、3、4、7.1、7.2、8、12、14、16、20及び21.3といったセグメントを発現するT細胞の多様性が失われてしまったものの、約半数のVβレパトアが保存されており、再生されたT細胞が遺伝的多様性を有するT細胞集団であることが確認された。
Claims (13)
- (1)採取した遺伝的多様性を有するT細胞集団から、目的の組織又は抗原に対して指向性を有するT細胞集団を濃縮する工程、
(2)前記濃縮したT細胞集団をiPS細胞へと初期化し、遺伝的多様性を維持したままで、iPS細胞を培養する工程、及び
(3)前記培養したiPS細胞から遺伝的多様性を有する再生T細胞集団を製造する工程を含む、iPS細胞を介して遺伝的多様性を有する再生T細胞集団を製造する方法。 - 工程(1)の採取したT細胞集団が、治療対象の哺乳動物由来である、請求項1に記載の方法。
- 工程(1)の採取したT細胞集団が、腫瘍浸潤T細胞である、請求項1又は2に記載の方法。
- 工程(1)の採取したT細胞集団が、血液、リンパ節又は腔水由来である、請求項1又は2に記載の方法。
- 工程(1)が、抗原タンパク質又はペプチドでの刺激により活性化し、増殖したT細胞を分離するステップを含む、請求項1~4いずれか1項に記載の方法。
- 工程(2)が、iPS細胞をクローン化せずに回収し、継代するステップを含む、請求項1~5いずれか1項に記載の方法。
- 工程(3)で得られたT細胞集団が、αβT細胞集団、γδT細胞集団、ヘルパーT細胞集団、レギュラトリーT細胞集団、細胞傷害性T細胞集団、NKT細胞集団、又は腫瘍浸潤T細胞集団である、請求項1~6いずれか1項に記載の方法。
- 工程(3)で得られたT細胞集団が、T細胞補充療法に使用するものである、請求項1~7いずれか1項に記載の方法。
- 請求項1~8いずれか1項に記載の方法により得られる再生T細胞集団。
- 生体内に存在するT細胞集団の遺伝的多様性を維持している、iPS細胞を介して得られる再生T細胞集団。
- 請求項9または請求項10に記載の再生T細胞集団を含有する医薬組成物。
- 自家的または同種的な移植によりがん治療対象を処置するための請求項11に記載の医薬組成物。
- 請求項11または請求項12に記載の医薬組成物を用いる、がん治療方法。
