Abstract

Furthermore the invention also relates to the use of said composition for swine, poultry or broiler house disinfection, for surface sterilization, and aerial disinfection.
Description
LIQUID DISINFECTANT FOR INTENSIVE ANIMAL HOUSES
FIELD OF THE INVENTION The present invention relates to a concentrated liquid sprayable disinfectant composition and the use of said composition for swine, poultry or broiler houses disinfection, for surface sterilization, and aerial disinfection.
BACKGROUND OF THE INVENTION
Viruses are widely responsible for immunosuppression in infected birds, which leads to poultry susceptible to opportunistic environmental pathogens present at sub-infective levels. Viruses are widely implicated in poultry epidemics of international economic importance. The direct costs of mortality as a result of endemic disease are immediately evident.
Therefore, preventative biosecurity is now a pre-requisite to poultry production, with poultry producers worldwide taking the necessary steps to institute strict biosecurity programs to exclude pathogenic organisms from farm environments.
Terminal disinfection is the most effective part of an all-in, all-out production program, and is critical in ensuring a healthy start for a new batch of poultry chicks. This involves careful removal of poultry waste and litter, followed by intensive sanitization and disinfection to eliminate infectious organisms or reduce their populations to non-infective levels in poultry housing, and also from vehicles and equipment used. A good terminal hygiene program starting with pre-cleaning using a bactericidal detergent-sanitizer ensures removal of dirt and associated organisms.
While the direct costs of mortality as a result of endemic disease is immediately evident, the effect of subclinical infectious disease on poultry productivity is often underestimated. Minimizing environmental pathogen populations in poultry housing during the early growth period ensures that the immune system of birds is not
overwhelmed, hence provides better performance of vaccines, and reduces veterinary pharmaceutical usage and associated residues in poultry meat and eggs.
Routine farm practices such as hand sanitization, control of aerial respiratory transmission, management of manure and litter, water sanitization and pest control prevent the horizontal transmission of disease which can spread rapidly through the flock under intensive poultry production conditions.
One of the routine in the industry sanitization protocol consists in fumigating animal houses with formaldehyde vapor. However formalin (a clea r aq ueous solution of forma ldehyde a nd metha nol) is a highly toxic chemical that is already banned in some countries.
Therefore, there is still a need in the industry for a liquid disinfectant composition which is safe to the end user, environmental friendly, and provides a broad spectrum bactericidal, sporicidal, fungicidal, and virucidal activity.
SUMMARY OF THE INVENTION
Surprisingly, the inventors found that the concentrated animal house liquid disinfectant composition of the present invention overcome the foregoing problems by providing a stable composition in liquid form, a composition free of formaldehyde, safe in use for the end user in the concentrated and diluted form, being cost compatible with the pig and broiler industry, and providing a broad spectrum bactericidal, sporicidal and virucidal activity. In a first aspect, the aim of the present invention is to provide a concentrated animal house liquid disinfectant composition comprising quaternary ammonium salts, orthopthalaldehyde, EDTA and isopropyl alcohol.
A further aspect of the invention is to provide the use of this animal house liquid disinfectant composition for the disinfection of pig or poultry houses.
A further object of the present invention is to provide a method of disinfecting pig or poultry houses comprising the step of spraying an animal house liquid disinfectant composition comprising quaternary ammonium salts, orthopthalaldehyde, EDTA and isopropyl alcohol.
DETAILED DESCRIPTION OF THE INVENTION
In a first aspect, this invention relates, to a concentrated animal house liquid disinfectant composition comprising:
a. 2 to 3 w/v % alkyl dimethyl benzyl ammonium chloride
b. 2 to 3 w/v % alkyl dimethyl ethylbenzyl ammonium chloride
c. 0.4 to 0.6 w/v % orthopthalaldehyde
d. 0.025 to 0.075 w/v % EDTA tetra sodium salt
e. 8 to 12 w/v % isopropyl alcohol
f. Q.S water.

Orthophthalaldehyde or ortho-phthalaldehyde (OPA; CAS 643-79-8) is the chemical compound with the formula C&H4(CHO)2. Often abbreviated OPA, the molecule is a dialdehyde, consisting of two formyl (CHO) groups attached to adjacent carbon centers on a benzene ring. OPA dissolves in water solution at pH < 11.5.
EDTA tetra sodium salt (Ethylene Diamine Tetraacetic Acid) is used as a metal sequester improving the activity of the quaternary ammoniums.
Isopropyl alcohol (l UPAC name propan-2-ol, CAS 67-63-0), also called dimethyl carbinoi or, isopropanoi, is a compound with the chemical formula CBHSO or C3H7OH or CH3CHOHCH3.
I n a preferred embodiment, the animal house liquid disinfectant composition according to the present invention comprises:
a. 2.2 to 2.5 w/v % alkyi dimethyl benzyl ammonium Chloride
b. 2.2 to 2.5 w/v % alkyi dimethyl ethylbenzyl ammonium Chloride
c. 0.5 to 0.6 w/v % orthopthalaldehyde
d. 0.04 to 0.06 w/v % EDTA tetra sodium salt
e. 8 to 12 w/v % isopropyl alcohol
f. OS water.
Even more preferably, the animal house liquid disinfectant composition according to the present invention comprises:
a. 2.3 to 2.4 w/v % alkyi dimethyl benzyl ammonium Chloride
b. 2.3 to 2.4 w/v % alkyi dimethyl ethylbenzyl ammonium Chloride
c. 0.5 to 0.6 w/v % orthopthalaldehyde
d. 0.04 to 0.06 w/v % EDTA tetra sodium salt
e. 8 to 12 w/v % isopropyl alcohol
f. Q.S water.
The compositions as described above can easily be prepared by the person skilled in the art following routine procedures. Basically, EDTA is first dissolved in water under stirring, followed by addition of isopropyl alcohol, then a mixture of alkyl dimethyl benzyl ammonium chloride and alkyl dimethyl ethylbenzyl ammonium chloride is added under continuous stirring and finally, the orthophtalaldehyde is added under stirring until complete dissolution. Volume is then adjusted with water.
The composition is stable at up to 40°C for up to two years, and shows excellent sprayability characteristics ensuring a complete disinfection of surfaced as demonstrated by the inventors.

Preferably, the ratio of concentrated animal house liquid disinfectant to water is comprised between 1:30 and 1:120, preferably between 1:50 and 1:120, and even more preferably about 1:100.

The use of the liquid composition as described above leads to a complete killing and or inactivation of viruses, spores, bacteria, and/or fungi.

Example 3: Stable concentrated animal house liquid disinfectant composition: alkyl dimethyl benzyl ammonium chloride 2.37 w/v % alkyl dimethyl ethylbenzyl ammonium chloride 2.37 w/v % orthopthalaldehyde 0.55 w/v %
EDTA tetra sodium salt 0.05 w/v % isopropyl alcohol 10 w/v % water qsp
In the above examples 1 to 3, alkyl dimethyl benzyl ammonium chloride comprises about 5 % Ci2 alkyl, 60 % Ci4 alkyl, 30 % Ci6 alkyl, and about 5 % Ci8 alkyl dimethyl benzyl ammonium chloride, while alkyl dimethyl ethylbenzyl ammonium chloride comprises about 68 % Ci2 alkyl, and about 32 % Ci4 alkyl dimethyl ethylbenzyl ammonium chloride. Furthermore, the composition is manufactured as follows: EDTA is first dissolved in water under stirring, followed by addition of isopropyl alcohol, then a mixture of alkyl dimethyl benzyl ammonium chloride and alkyl dimethyl ethylbenzyl ammonium chloride is added under continuous stirring and finally, the orthophtalaldehyde is added under stirring until complete dissolution. Volume is then adjusted with water.
Example 4: Ready to use diluted animal house liquid disinfectant:
To prepare the ready to use composition, the concentrated compositions according to Example 3 is diluted in water at a ratio of Concentrate/Water of 1/100 leading to the following compositions:

Example 5: Stability studies of the concentrated animal house liquid disinfectant composition of Example 3
Example 6: Antimicrobial efficacy assays
The concentrated compositions according to Example 3 has been diluted in water at a ratio of Concentrate/Water of 1/100 to obtain solution A. To 5 ml aliquot of Solution A, 0.1 ml of bacterial culture having a density of approximately 106 cfu/ml was added and kept at a temperature between 18 and 20 °C with intermittent shaking. After different time intervals (Killing / Innactivation contact time of 5, 10 or 15 minutes), a loopfull was added in a test tube containing sterile nutrient broth for bacteria l cultures, and sterile Sabouraud broth for fungal spp. Then all the nutrient broth tubes were incubated at 37°C for 48 hours, while Sabouraud broth tubes were incubated at room temperature in the dark for 72 hours to observe growth or inactivation.
The results obtained are summarized below
N means no viable organisms have been observed; V means organisms were observed. MRSA means the strain is a multi-resistant S. aureus.
This data confirms the disinfecting efficacy of the composition according to the present invention after a contact time of 5 minutes or less for bacteria as well as for fungi.
Claims
1. A concentrated animal house liquid disinfectant composition comprising:
a. 2 to 3 w/v % alkyl dimethyl benzyl ammonium chloride,
b. 2 to 3 w/v % alkyl dimethyl ethylbenzyl ammonium chloride,
c. 0.4 to 0.6 w/v % orthopthalaldehyde,
d. 0.025 to 0.075 w/v % EDTA tetra sodium salt,
e. 8 to 12 w/v % isopropyl alcohol, and
f. Q.S water.
2. The concentrated animal house liquid disinfectant composition according to claim 1, wherein the composition comprises:
a. 2.2 to 2.5 w/v % alkyl dimethyl benzyl ammonium chloride,
b. 2.2 to 2.5 w/v % alkyl dimethyl ethylbenzyl ammonium chloride,
c. 0.5 to 0.6 w/v % orthopthalaldehyde,
d. 0.04 to 0.06 w/v % EDTA tetra sodium salt,
e. 8 to 12 w/v % isopropyl alcohol, and
f. Q.S water.
3. The concentrated anima l house liquid disinfectant composition according to claims 1 or 2, wherein the alkyl dimethyl benzyl ammonium chloride is mixture of about 5 % Ci2 alkyl, 60 % Ci4 alkyl, 30 % Ci6 alkyl, and about 5 % Ci8 alkyl.
4. The concentrated animal house liquid disinfectant composition according to any of the claims 1 to 3, wherein the alkyl dimethyl ethylbenzyl ammonium chloride is mixture of about 68 % C12 alkyl, and about 32 % Ci4 alkyl.
5. The concentrated animal house liquid disinfectant composition according to any of the claims 1 to 4, wherein the composition comprises:
a. 2.3 to 2.4 w/v % alkyl dimethyl benzyl ammonium chloride,
b. 2.3 to 2.4 w/v % alkyl dimethyl ethylbenzyl ammonium chloride,
c. 0.5 to 0.6 w/v % orthopthalaldehyde,
d. 0.04 to 0.06 w/v % EDTA tetra sodium salt,
e. 8 to 12 w/v % isopropyl alcohol, and
f. Q.S water.
6. An animal house liquid disinfectant composition wherein the composition is prepared by diluting a concentrated animal house liquid disinfectant composition according to any of the claims 1 to 5, in water at a ratio of concentrated animal house liquid disinfectant to water comprised between 1:20 and 1:200.
7. The animal house liquid disinfectant composition according to claim 6, wherein the ratio of concentrated animal house liquid disinfectant to water is comprised between 1: 50 and 1:120.
8. Use of an animal house liquid disinfectant according to any of the claims 6 or 7 for the disinfection of pig or poultry houses.
9. The use according to claim 8, wherein the disinfection is carried out by spraying.
10. The use according to claims 8 or 9, wherein the disinfection is characterized by a complete killing and or inactivation of viruses, spores, bacteria, and/or fungi.
11. A method of disinfecting animal houses comprising the step of spraying a diluted animal house liquid disinfectant composition comprising:
a. 2 to 3 w/v % alkyl dimethyl benzyl ammonium chloride,
b. 2 to 3 w/v % alkyl dimethyl ethylbenzyl ammonium chloride,
c. 0.4 to 0.6 w/v % orthopthalaldehyde,
d. 0.025 to 0.075 w/v % EDTA tetra sodium salt,
e. 8 to 12 w/v % isopropyl alcohol, and
f. Q.S water,
wherein the dilution is performed at a ratio of concentrated animal house liquid disinfectant to water comprised between 1: 20 and 1:200.
