志賀毒素2eのBサブユニット(Stx2eB)、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選択される2以上の毒素Bサブユニットの融合タンパク質を対象動物に投与することで対象動物における自然免疫を誘導する。
Description
本発明は、毒素融合タンパク質を利用した自然免疫増強剤に関する。
ブタは通常生後21日を目安に離乳する。離乳前は母ブタからの乳汁免疫による移行抗体で病原菌の感染から免れている。しかし、離乳と同時に、豚舎の移動、環境の変化、餌の切り替え、群編成など、子ブタには多くのストレスがかかり、先の移行抗体の消失もあって、離乳から70日齢のいわゆる離乳仔ブタの期間は、ブタの一生のうち最も病気に罹りやすい時期といわれている。この時期に高発する疾病としては、例えば、細菌病ではブタ浮腫病、連鎖球菌症、グレーサー病、サルモネラ症、コクシジウム病、大腸菌性下痢症、マイコプラズマ感染、ウイルス病ではブタ繁殖・呼吸障害症候群(PRRS)、離乳後多臓器性発育不良症候群(PMWS)などが挙げられる。
ワクチンは疾病予防の有効な手段であるが、従来は注射型のワクチンが主であった。注射型ワクチンによる免疫ルートで惹起できるのは全身免疫であり、いわゆる特定の疾病を引き起こす病原体にのみ対応する免疫グロブリンの産生を誘導する。この場合、母ブタからの移行抗体が残留する離乳仔ブタにおいては、ワクチンの吸収が阻害されたり、また自己免疫システムの確立が不十分であったり、必ずしも効果的な予防法になっていない。このように離乳仔ブタの疾病は多種多様であり、個々の対策を行うことは際限がなく、総合的な対策が望ましい。
ワクチンは疾病予防の有効な手段であるが、従来は注射型のワクチンが主であった。注射型ワクチンによる免疫ルートで惹起できるのは全身免疫であり、いわゆる特定の疾病を引き起こす病原体にのみ対応する免疫グロブリンの産生を誘導する。この場合、母ブタからの移行抗体が残留する離乳仔ブタにおいては、ワクチンの吸収が阻害されたり、また自己免疫システムの確立が不十分であったり、必ずしも効果的な予防法になっていない。このように離乳仔ブタの疾病は多種多様であり、個々の対策を行うことは際限がなく、総合的な対策が望ましい。
特許文献1では、特定のアミノ酸配列を持つリンカー(PG12)を利用して、志賀(浮腫病)毒素Bサブユニット(Stx2eB)と大腸菌易熱性毒素Bサブユニット(LTB)またはコレラ毒素Bサブユニット(CTB)の融合タンパク質を作製し、植物で生産させたことが開示されている。しかしながら、目的はあくまでもこれらの毒素のみを標的としたワクチンとしての使用であり、融合タンパク質のワクチン機能を評価した実施例もない。
また、特許文献2では、LTBとStx2eBの融合タンパク質を浮腫病および下痢症などの大腸菌症のワクチンとして使用することが開示されているが、これらの疾病予防効果はLTBとStx2eBに対する抗原特異的な免疫作用を目的としたものであり、自然免疫系を亢進し、他の疾病も含めて予防効果を持つことは全く予想されていなかった。
また、特許文献3では、PRRSウイルスのGlycoprotein 5 (GP5)タンパク質と、アジュバントタンパク質(LTBまたはStx2eB)を含む融合タンパク質をPRRSのワクチンとして使用することが開示されているが、PRRS予防効果はPRRSに対する抗原特異的な免疫作用を目的としたものであり、PRRSタンパク質を抗原に使用せずに、PRRS予防効果を有するワクチンが得られるとは全く予想されなかった。

一方、様々な疾病に対する有望な総合的対策として、非特異的な免疫賦活という考え方がある。Casey ey al., (2007) (非特許文献1)は乳酸菌を飼料に混合しブタに経口投与することで、サルモネラ菌感染の症状を緩和したと報告しているし、Kritas and Morrison(2007) (非特許文献2)は、ブタに乳酸菌を投与することで、PRRS感染による増体重の改善を報告しているが、いずれの場合も生菌を使用するために必ずしも高い効果が得られるものではないという問題点があった。

ブタなどの動物において自然免疫系を活性化することができれば、病原体の感染局所におけるマクロファージや樹状細胞などの自然免疫担当細胞を刺激し、非特異的な病原体の貪食を誘導することで、総合的な疾患の防除効果が期待できる。しかしながら、非特異的な自然免疫を効果的に誘導するためには、有効な抗原デザインの最適化が重要な課題であった。
したがって、本発明は、投与された個体に効率よく自然免疫を誘導することのできる物質を提供することを課題とする。
本発明者らは、上記課題を解決すべく鋭意研究を行った結果、志賀毒素2eのBサブユニット、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選ばれる2以上の毒素Bサブユニットの融合タンパク質またはそれを含む形質転換体を動物に投与することにより、動物体内においてマクロファージの活性化などを通じて自然免疫を誘導し、ウイルスや細菌およびマイコプラズマの感染などに対して全般的に防除効果を高めることができることを見出し、本発明を完成するに至った。
すなわち、本発明は以下のとおりである。
(1)志賀毒素2eのBサブユニット(Stx2eB)、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選択される2以上の毒素Bサブユニットの融合タンパク質を含む、自然免疫増強剤。
(2)前記融合タンパク質がStx2eBとLTBを含む融合タンパク質である、(1)に記載の自然免疫増強剤。
(3)前記融合タンパク質がStx2eBを2つ以上含む融合タンパク質である、(1)に記載の自然免疫増強剤。
(4)前記Stx2eBの73位のAsn残基がSer残基に置換されている、(1)~(3)のいずれかに記載の自然免疫増強剤。
(5)前記2以上の毒素Bサブユニットがペプチドリンカーを介して融合されている、(1)~(4)のいずれかに記載の自然免疫増強剤。
(6)前記ペプチドリンカーがPG12(配列番号2)、PG12v2(配列番号4)、PG17(配列番号30)、もしくはPG22(配列番号31)、またはこれらの配列と80%以上同一性を有するアミノ酸配列を有するペプチドである、(5)に記載の自然免疫増強剤。
(7)前記融合タンパク質が配列番号15~17のいずれかのアミノ酸配列を有する、(1)~(6)のいずれかに記載の自然免疫増強剤。

本発明の融合タンパク質またはそれを含む形質転換体をブタや鶏などの動物に投与することにより、特定の毒素ではなく、細菌、ウイルス、マイコプラズマなどあらゆる病原体に起因する不特定の疾病に対する免疫力(自然免疫)を向上させることができる。すなわち、当該融合タンパク質やそれを発現するトランスジェニック植物を食餌することによって、通常、有効成分(抗原)に対して、1対1の関係にある疾病に対してのみワクチン効果が期待されるものを、本発明では、不特定な疾病に対しても免疫誘導ができるようになった。これによって、ブタや鶏などの生産者の省コスト化、省力化、および家畜や養鶏のストレス軽減を可能にし、生産性の向上が期待できる。
以下、本発明の実施形態について説明する。
本発明において、自然免疫増強剤とは、動物生体内における自然免疫作用を活性化する薬剤をいう。本発明において、自然免疫とは、生体が備える免疫のうち、生体に先天的に備わっており、事前に抗原に暴露していなくても前記抗原を生体から排除するに際に機能する機構を意味する。本発明の自然免疫増強剤としてより具体的には、マクロファージ、好中球、樹状細胞及び/またはナチュラルキラー(NK)細胞等を介した非特異的な免疫応答を活性化する薬剤を意味する。なお、マクロファージの活性化はマクロファージ細胞における腫瘍壊死因子(TNF)αの発現などで確認できる。
本発明において、自然免疫増強剤とは、動物生体内における自然免疫作用を活性化する薬剤をいう。本発明において、自然免疫とは、生体が備える免疫のうち、生体に先天的に備わっており、事前に抗原に暴露していなくても前記抗原を生体から排除するに際に機能する機構を意味する。本発明の自然免疫増強剤としてより具体的には、マクロファージ、好中球、樹状細胞及び/またはナチュラルキラー(NK)細胞等を介した非特異的な免疫応答を活性化する薬剤を意味する。なお、マクロファージの活性化はマクロファージ細胞における腫瘍壊死因子(TNF)αの発現などで確認できる。
本発明の自然免疫増強剤は、志賀毒素2eのBサブユニット(Stx2eB)、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選択される2以上の毒素Bサブユニットの融合タンパク質を含む。なお、志賀毒素、大腸菌易熱性毒素及びコレラ毒素はいずれも毒性本体であるAサブユニット1分子と腸管粘膜への結合に関与するBサブユニット5分子からなるホロ毒素であり、これらのBサブユニットは、当該腸管粘膜への結合に関与するBサブユニットを意味する。
前記2以上としては、好ましくは、2~5であり、より好ましくは2~3であり、さらに好ましくは2である。なお、2以上のタンパク質とは、Stx2eB、LTB及びCTBから選択されるいずれか1種類の毒素Bサブユニットが2つ以上含まれる態様でもよいし、Stx2eB、LTB及びCTBから選択される2種類の毒素が合計で2つ以上含まれる態様でもよいし、Stx2eB、LTB及びCTBが合計で3つ以上含まれる態様でもよい。
なお、本発明において、融合タンパク質とは、1つのポリペプチド鎖の中に2つ以上のタンパク質ユニットを含む多価タンパク質を意味する。
前記2以上としては、好ましくは、2~5であり、より好ましくは2~3であり、さらに好ましくは2である。なお、2以上のタンパク質とは、Stx2eB、LTB及びCTBから選択されるいずれか1種類の毒素Bサブユニットが2つ以上含まれる態様でもよいし、Stx2eB、LTB及びCTBから選択される2種類の毒素が合計で2つ以上含まれる態様でもよいし、Stx2eB、LTB及びCTBが合計で3つ以上含まれる態様でもよい。
なお、本発明において、融合タンパク質とは、1つのポリペプチド鎖の中に2つ以上のタンパク質ユニットを含む多価タンパク質を意味する。
<Stx2eB>
志賀毒素(Stx)は、浮腫病の原因となる腸管出血性大腸菌(EHEC, STEC)が産生するタンパク質性毒素で、1型(Stx1)及び2型(Stx2)に分けられる。Stx1は、a~dのサブクラスに、Stx2はa~gのサブクラスにそれぞれ分類される。毒性本体であるAサブユニット1分子と腸管粘膜への結合に働くBサブユニット5分子からなるホロ毒素で、真核細胞のリボソームに作用して、タンパク質合成を阻害する働きを持つ。

また、Stx2eBは、例えば、Asn73(すなわち、配列番号8のアミノ酸配列の55位のAsn残基)がSer残基に置換されている変異型でもよい。配列番号8のアミノ酸配列の55位のAsn残基がSerに置換されているアミノ酸配列(Asn73Ser)を配列番号10で示す。野生型のStx2eBはこのAsn残基においてN-結合型の糖鎖修飾を受けるが、この変異体はN-結合型の糖鎖修飾を受けない。
また、Stx2eBは、例えば、Asn73(すなわち、配列番号8のアミノ酸配列の55位のAsn残基)がSer残基に置換されている変異型でもよい。配列番号8のアミノ酸配列の55位のAsn残基がSerに置換されているアミノ酸配列(Asn73Ser)を配列番号10で示す。野生型のStx2eBはこのAsn残基においてN-結合型の糖鎖修飾を受けるが、この変異体はN-結合型の糖鎖修飾を受けない。
また、Stx2eBは、融合タンパク質としてブタや鶏等の動物に投与して自然免疫増強を引き起こすことができる限り、配列番号8または10で表されるアミノ酸配列における1個又は数個のアミノ酸が、置換、欠失、挿入又は付加されていてもよい。前記「数個」としては、例えば、2~10個、好ましくは2~5個、より好ましくは2~3個である。
また、Stx2eBは、配列番号8または10で表されるアミノ酸配列と、好ましくは85%以上、さらに好ましくは90%以上、より好ましくは95%以上の同一性を有し、かつ融合タンパク質としてブタや鶏等の動物に投与して自然免疫増強を引き起こすことができるものであってもよい。
また、Stx2eBは、配列番号8または10で表されるアミノ酸配列と、好ましくは85%以上、さらに好ましくは90%以上、より好ましくは95%以上の同一性を有し、かつ融合タンパク質としてブタや鶏等の動物に投与して自然免疫増強を引き起こすことができるものであってもよい。

大腸菌性下痢症は、毒素原性大腸菌(ETEC)が生産するタンパク質性毒素LTが原因であり、LTは大腸菌易熱性毒素とも呼ばれる。LTは、毒性本体であるAサブユニット1分子とBサブユニット5分子からなるホロ毒素である。LTのAサブユニット(LTA)は細胞質内に侵入し、細胞内cAMP濃度を上昇させ、細胞膜クロライドチャネルを活性化することで腸管内への水の漏出すなわち下痢の病態を引き起こす。LTのBサブユニット(LTB)は無毒であり、LT毒素と腸管細胞との接着に関与する。

また、LTBは、配列番号12で表されるアミノ酸配列と、85%以上、好ましくは90%以上、より好ましくは95%以上の同一性を有し、かつ融合タンパク質としてブタや鶏等の動物に投与して自然免疫増強を引き起こすことができるものであってもよい。

本発明の他の実施形態では、LTBに糖鎖が付加されていてもよい。例えば、LTBの90位(すなわち、配列番号12の90位)のAsn残基にN-結合型の糖鎖が付加される。一方、配列番号12の90位がSer残基に置換された変異型LTB(アミノ酸配列を配列番号14で表す)はN-結合型の糖鎖修飾を受けない。

コレラ毒素(CT)タンパク質は、毒性本体である1つのAサブユニット(CTA)と、腸管粘膜への侵入へ関与する5つのBサブユニット(CTB)からなる。
本発明で使用されるCTBは、例えば、配列番号6のアミノ酸配列で表される。CTBは、融合タンパク質としてブタや鶏等の動物に投与して自然免疫増強を引き起こすことができる限り、配列番号6で表されるアミノ酸配列における1個又は数個のアミノ酸が、置換、欠失、挿入又は付加されていてもよい。前記「数個」としては、例えば、2~10個、好ましくは2~5個、より好ましくは2~3個である。

本発明の自然免疫増強剤の有効成分である融合タンパク質は、Stx2eB、LTB、CTBから任意に選ばれる2以上の毒素Bサブユニットの融合タンパク質であればよいが、好ましくは、Stx2eBとLTB の融合タンパク質である。本発明において、Stx2eBとLTBとが融合される順序は、いずれが先であってもよい。また、マクロファージ活性化能に優れるため、Stx2eBを2つ以上含む融合タンパク質も好ましい。
本発明の好ましい実施形態では、Stx2eB、LTB、CTBから任意に選ばれる2以上の毒素Bサブユニットはペプチドリンカーを介してタンデムに連結される。ここで、ペプチドリンカーとは、直鎖状にアミノ酸が連結したペプチドからなるリンカーを意味する。
本発明で用いるペプチドリンカーのアミノ酸の個数は、例えば5~30個、好ましくは10~25個、さらに好ましくは10~22個、より好ましくは12~22個である。また、本発明で用いるペプチドリンカーにおいて、好ましくは、プロリンの含有率が20~27%、より好ましくは、20~25%である。

本発明において、ペプチドリンカーは、好ましくは、配列番号2、4、30または31で表されるアミノ酸配列からなるペプチドと80%以上、好ましくは90%以上の同一性を有するペプチドであってもよい。

本発明で用いる融合タンパク質は、植物で発現させる場合などは、そのアミノ末端に、植物由来の分泌シグナルペプチド又は葉緑体移行シグナルペプチドが付加されていてもよい。ここで、「付加」とは、前記分泌シグナルペプチドが、前記ペプチドを介して連結した融合タンパク質のアミノ末端に、直接結合している場合も、他のペプチドを介して結合している場合も含む概念である。
分泌シグナルペプチドは、好ましくはナス科(Solanaceae)、アブラナ科(Brassicaceae)、キク科(Asteraceae)に属する植物、さらに好ましくはタバコ属(Nicotiana)、シロイヌナズナ属(Arabidopsis)、アキノノゲシ属(Lactuca)等に属する植物、より好ましくはタバコ(Nicotiana tabacum)、シロイヌナズナ(Arabidopsis thaliana)、レタス(Lactuca sativa)等に由来する。
また、分泌シグナルペプチドは、好ましくはタバコのβ-Dグルカンエキソヒドロラーゼ(β-D-glucan exohydrolase)、タバコの38kDa ペルオキシダーゼ(GenBank Accession D42064)に由来する。前記分泌シグナルペプチドとしては、例えば、タバコのβ-Dグルカンエキソヒドロラーゼに由来する、配列番号25で表されるアミノ酸配列を有しているペプチドが挙げられる。タバコのβ-DグルカンエキソヒドロラーゼをコードするDNAの塩基配列は、例えば配列番号24で表される。
好ましい葉緑体移行シグナルペプチドは、例えば、国際公開WO2009/004842号パンフレット及び国際公開WO2009/133882号パンフレットに記載されている。

好ましい葉緑体移行シグナルペプチドは、例えば、国際公開WO2009/004842号パンフレット及び国際公開WO2009/133882号パンフレットに記載されている。
さらに、本発明で用いる融合タンパク質は、植物で発現させる場合などは、そのカルボキシル末端に、小胞体残留シグナルペプチド、液胞移行シグナルペプチド等のシグナルペプチドが付加されていてもよい。ここで、「付加」とは、シグナルペプチドが、前記融合タンパク質のカルボキシル末端に、直接結合している場合も、他のペプチドを介して結合している場合も含む概念である。本明細書において、アミノ末端に分泌シグナルペプチドが付加され、かつカルボキシル末端に小胞体残留シグナルペプチドが付加されたハイブリッドタンパク質を、小胞体型(ER)のハイブリッドタンパク質ともいい、該小胞体型の融合タンパク質をコードするDNA構築物を、小胞体型のDNA構築物ともいう。小胞体型の融合タンパク質は、真核生物で効率良く蓄積する報告例が多数ある。
本発明で用いる融合タンパク質は、そのカルボキシル末端に、好ましくは、小胞体残留シグナルペプチドが付加されている。好ましい小胞体残留シグナルペプチドは、例えば、国際公開WO2009/004842号パンフレット及び国際公開WO2009/133882号パンフレットに記載されているが、HDEL配列(配列番号26)を利用することができる。
他の好ましい液胞移行シグナルペプチドは、例えば、国際公開WO2009/004842号パンフレット及び国際公開WO2009/133882号パンフレットに記載されている。
本発明で用いる融合タンパク質は、そのカルボキシル末端に、好ましくは、小胞体残留シグナルペプチドが付加されている。好ましい小胞体残留シグナルペプチドは、例えば、国際公開WO2009/004842号パンフレット及び国際公開WO2009/133882号パンフレットに記載されているが、HDEL配列(配列番号26)を利用することができる。

本発明で用いる融合タンパク質は、化学的に合成することもできるし、遺伝子工学的に生産することもできる。
遺伝子工学的に生産する場合、融合タンパク質をコードするDNAを含むDNA構築物を用いる。本発明で用いるDNA構築物は、Stx2eBをコードするDNA、LTBをコードするDNAおよびCTBをコードするDNAから選ばれる2つ以上のDNAが、前記ペプチドリンカーをコードするDNAを介してタンデムに連結されているDNAを含む。さらに、3’側にペプチドリンカーをコードするDNAが含まれてよい。前記ペプチドリンカーをコードするDNAは、例えば配列番号1(PG12)や配列番号3(PG12v2)で表される。Stx2eBをコードするDNAとして、例えば、Stx2eB(Asn73)をコードするDNA(配列番号7)、Stx2eB(Asn73Ser)をコードするDNA(配列番号9)が挙げられる。LTBをコードするDNAとして、例えば、LTB(Asn90)をコードするDNA(配列番号11)、LTB(Asn90Ser)をコードするDNA(配列番号13)が挙げられる。CTBをコードするDNAとして、例えば、配列番号5が挙げられる。
前記ペプチドリンカーをコードするDNAと2つ以上の毒素BサブユニットDNAは、それぞれ終止コドンを除いて読み枠を合わせて連結される。

Stx2eB、LTB、CTBをコードするDNAは、例えば、配列番号7、9、11、13、5の塩基配列に基づいて、一般的な遺伝子工学的な手法により得ることができる。具体的には、各毒素を生産する細菌より、常法に従ってcDNAライブラリーを調製し、該ライブラリーから上記塩基配列に基づいて作製したプローブを用いて所望のクローンを選択する。また、上記塩基配列を基にした化学合成、上記塩基配列の5’及び3’末端の塩基配列をプライマーとし、ゲノムDNAを鋳型としたPCRなどにより合成することもできる。これらを公知の手法でペプチドリンカーをコードするDNAと連結させることにより融合タンパク質をコードするDNAが得られる。
本発明で用いる融合タンパク質をコードするDNAは、例えば、配列番号18~20で表される。これらのDNAはそれぞれアミノ酸配列15、16、17の融合タンパク質をコードする。

また、融合タンパク質をコードするDNAは、配列番号18~20の塩基配列を有するDNAとストリンジェントな条件下でハイブリダイズするDNAであってもよい。「ストリンジェントな条件」とは、いわゆる特異的なハイブリッドが形成され、非特異的なハイブリッドが形成されない条件をいう。例えば、同一性が高い二つのDNAどうし、好ましくは80%以上、より好ましくは90%以上、特に好ましくは95%以上の同一性を有する2つのDNAがハイブリダイズするが、それより同一性の低い2つのDNAがハイブリダイズしない条件が挙げられる。例えば2×SSC(330mM NaCl、30mM クエン酸)、42℃が挙げられ、好ましくは0.1×SSC(330mM NaCl、30mM クエン酸)、60℃が挙げられる。
融合タンパク質をコードするDNAは、該タンパク質を生産させる宿主細胞に応じて、ハイブリッドタンパク質の翻訳量が増大するように、融合タンパク質を構成するアミノ酸を示すコドンが適宜改変されていることも好ましい。
コドン改変の方法としては、例えばKang et al. (Protein Expr Purif. 2004 Nov;38(1):129-35.)の方法を参考にすることができる。また、宿主細胞において使用頻度の高いコドンを選択したり、GC含量が高いコドンを選択したり、宿主細胞のハウスキーピング遺伝子において使用頻度の高いコドンを選択したりする方法が挙げられる。
コドン改変の方法としては、例えばKang et al. (Protein Expr Purif. 2004 Nov;38(1):129-35.)の方法を参考にすることができる。また、宿主細胞において使用頻度の高いコドンを選択したり、GC含量が高いコドンを選択したり、宿主細胞のハウスキーピング遺伝子において使用頻度の高いコドンを選択したりする方法が挙げられる。
本発明で用いるDNA構築物において、好ましくは、前記融合タンパク質をコードするDNAが、エンハンサーに発現可能に連結されている。ここで、「発現可能」とは、本発明で用いるDNA構築物が適切なプロモーターを含むベクターに挿入され、該ベクターが適切な宿主細胞に導入された場合に、宿主細胞内で前記融合タンパク質が生産されることをいう。また、「連結」とは、2つのDNAが直接結合している場合も、他の塩基配列を介して結合している場合も含む概念である。
エンハンサーとしては、Kozak配列や植物由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域が挙げられる。植物で発現させる場合などは、好ましくは、前記ハイブリッドタンパク質をコードするDNAが、植物由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域に発現可能に連結されている。

アルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域とは、アルコールデヒドロゲナーゼをコードする遺伝子の転写開始点から、翻訳開始点(ATG、メチオニン)の前までの塩基配列を含む領域をいう。前記領域は、植物に由来すればよいが、好ましくはナス科(Solanaceae)、アブラナ科(Brassicaceae)、キク科(Asteraceae)に属する植物、さらに好ましくはタバコ属(Nicotiana)、シロイヌナズナ属(Arabidopsis)、アキノノゲシ属(Lactuca)等に属する植物、より好ましくはタバコ(Nicotiana tabacum)、シロイヌナズナ(Arabidopsis thaliana)、レタス(Lactuca sativa)等に由来する。
前記アルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域としては、例えばタバコ(Nicotiana tabacum)由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域(NtADH5'UTR)(配列番号27)を用いることができ、翻訳開始点上流3塩基を改変したNtADH5'UTR領域(NtADHmod 5'UTR)(配列番号28)を用いることでさらに高翻訳が期待できる。
植物由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域を得る方法は、例えば、特開2012-19719号公報および国際公開WO2009/133882号パンフレットに記載されている。
前記アルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域としては、例えばタバコ(Nicotiana tabacum)由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域(NtADH5'UTR)(配列番号27)を用いることができ、翻訳開始点上流3塩基を改変したNtADH5'UTR領域(NtADHmod 5'UTR)(配列番号28)を用いることでさらに高翻訳が期待できる。
植物由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域を得る方法は、例えば、特開2012-19719号公報および国際公開WO2009/133882号パンフレットに記載されている。

本発明で用いるDNA構築物は、一般的な遺伝子工学的手法により作製することができ、植物由来のアルコールデヒドロゲナーゼ遺伝子の5’-非翻訳領域、植物由来の分泌シグナルペプチドをコードするDNA、及び融合タンパク質をコードするDNA、小胞体残留シグナルペプチドをコードするDNAなどの各DNAを、それぞれ、適当な制限酵素により切断し、適当なリガーゼで連結することで構築することができる。
本発明で用いる組換えベクターは、前記DNA構築物を含むことを特徴とする。本発明で用いる組換えベクターは、前記融合タンパク質をコードするDNAが、ベクターが導入される宿主細胞において発現可能なように、ベクター内に挿入されていればよい。ベクターは、宿主細胞において複製可能なものであれば特に制限されず、例えば、プラスミドDNA、ウイルスDNA等が挙げられる。また、ベクターは薬剤耐性遺伝子等の選択マーカーを含むことが好ましい。プラスミドDNAは、大腸菌やアグロバクテリウムからアルカリ抽出法(Birnboim, H. C. & Doly, J. (1979) Nucleic acid Res 7: 1513)又はその変法等により調製することができる。また、市販のプラスミドとして、例えばpBI221、pBI121、pBI101、pIG121Hm等を用いることもできる。ウイルスDNAとしては、例えばpTB2(Donson et al.,1991)等を用いることができる(Donson J., Kerney CM., Hilf ME., Dawson WO. Systemic expression of a bacterial gene by a tobacco mosaic virus-based vector. Proc. Natl. Acad. Sci.(1991) 88: 7204-7208を参照。)
ベクター内で用いられるプロモーターは、ベクターが導入される宿主細胞に応じて適宜選択することができる。例えば、カリフラワーモザイクウイルス35Sプロモーター(Odell et al.1985 Nature 313:810)、イネのアクチンプロモーター(Zhang et al.1991 Plant Cell 3:1155)、トウモロコシのユビキチンプロモーター(Cornejo et al.1993 Plant Mol.Biol.23:567)等が好ましく用いられる。また、ベクター内で用いられるターミネーターも、同様にベクターが導入される宿主細胞に応じて適宜選択することができる。例えば、ノパリン合成酵素遺伝子転写ターミネーター、カリフラワーモザイクウイルス35Sターミネーター、シロイヌナズナheat shock protein 18.2 遺伝子のターミネーター(HSP-T)等が好ましく用いられる。本発明で使用される好ましいターミネーターは、例えば、配列番号29で表されるAtHSP-Tである。
本発明で用いる組換えベクターは、例えば、前記DNA構築物を適当な制限酵素で切断又はPCRによって制限酵素部位を付加し、ベクターの制限酵素部位又はマルチクローニングサイトに挿入することで得ることができる。
本発明で用いる形質転換体は、前記組換えベクターで形質転換されていることを特徴とする。形質転換に用いられる宿主細胞は真核細胞及び原核細胞の何れでもよい。
真核細胞としては、哺乳動物細胞、酵母細胞、昆虫細胞などでもよいが、植物細胞が好ましく用いられ、中でもアキノノゲシ属(Lactuca)などのキク科(Asteraceae)、ナス科、アブラナ科、アカザ科に属する植物の細胞が好ましく用いられる。さらに、アキノノゲシ属(Lactuca)に属する植物の細胞、中でもレタス(Lactuca sativa)細胞が好ましく用いられる。宿主細胞としてレタス細胞を用いる場合は、ベクターは、カリフラワーモザイクウイルス35S RNAプロモーター等を用いることができる。
原核細胞としては、大腸菌(Escherichia coli)、アグロバクテリウム(Agrobacterium tumefaciens)等が用いられる。
真核細胞としては、哺乳動物細胞、酵母細胞、昆虫細胞などでもよいが、植物細胞が好ましく用いられ、中でもアキノノゲシ属(Lactuca)などのキク科(Asteraceae)、ナス科、アブラナ科、アカザ科に属する植物の細胞が好ましく用いられる。さらに、アキノノゲシ属(Lactuca)に属する植物の細胞、中でもレタス(Lactuca sativa)細胞が好ましく用いられる。宿主細胞としてレタス細胞を用いる場合は、ベクターは、カリフラワーモザイクウイルス35S RNAプロモーター等を用いることができる。
原核細胞としては、大腸菌(Escherichia coli)、アグロバクテリウム(Agrobacterium tumefaciens)等が用いられる。
本発明で用いる形質転換体は、一般的な遺伝子工学的手法を用いて、本発明で用いるベクターを宿主細胞に導入することにより作製することができる。例えば、アクロバクテリウムを利用した導入方法(Hood, et al., 1993, Transgenic, Res. 2:218,Hiei, et al.,1994 Plant J. 6:271)、エレクトロポレーション法(Tada, et al., 1990, Theor.Appl.Genet, 80:475)、ポリエチレングリコール法(Lazzeri, et al., 1991, Theor. Appl. Genet. 81:437)、パーティクルガン法(Sanford, et al., 1987, J. Part. Sci.tech. 5:27)、ポリカチオン法(Ohtsuki, et al., FEBS Lett. 1998 May 29;428(3):235-40.)などの方法を用いることが可能である。
本発明で用いるベクターを宿主細胞に導入した後、選択マーカーの表現型によって前記形質転換体を選抜することができる。また、選抜した形質転換体を培養することにより、前記融合タンパク質を生産することができる。培養に用いる培地及び条件は、形質転換体の種に応じて適宜選択することができる。
また、宿主細胞が植物細胞の場合には、選抜した植物細胞を常法に従って培養することにより、植物体を再生することができ、植物細胞内又は植物細胞の細胞膜外に前記融合タンパク質を蓄積させることができる。例えば、植物細胞の種類により異なるが、ジャガイモであればVisserら(Theor.Appl.Genet 78:594(1989))の方法が挙げられ、タバコであればNagataとTakebe(Planta 99:12(1971))の方法が挙げられる。
また、宿主細胞が植物細胞の場合には、選抜した植物細胞を常法に従って培養することにより、植物体を再生することができ、植物細胞内又は植物細胞の細胞膜外に前記融合タンパク質を蓄積させることができる。例えば、植物細胞の種類により異なるが、ジャガイモであればVisserら(Theor.Appl.Genet 78:594(1989))の方法が挙げられ、タバコであればNagataとTakebe(Planta 99:12(1971))の方法が挙げられる。
レタスの場合は、例えば0.1 mg /lのNAA(ナフタレン酢酸)、0.05 mg/lのBA(ベンジルアデニン)および0.5 g/lのpolyvinylpyrrolidoneを含むMS培地でシュートの再生が可能であり、再生したシュートを0.5 g/lのpolyvinylpyrrolidoneを含む1/2 MS培地で培養することで発根が可能である。
また、上記のようにして再生した植物体から種子を採取し、それを適当な方法で播種し栽培することにより、前記融合タンパク質を生産する植物体とすることができ、このような植物体も、前記形質転換体に含まれる。

アグロバクテリウム(Agrobacterium tumefaciens)は植物の傷口で感染させるもので、腫瘍誘発性のTi(tumor-inducing)プラスミドと呼ばれる大きな染色体外因子を運搬する。多くの研究所において、数年に亘る鋭意研究の後、アグロバクテリウム系の開発により、様々な植物組織を型通りに形質転換することが可能となった。この技術により転換された代表的な植物として、タバコ、トマト、ヒマワリ、綿、ナタネ、ジャガイモ、ポプラ、及びダイズ、イチゴ、イネなどがある。
様々な種の植物について、アグロバクテリウム(Agrobacterium tumefaciens)で形質転換された組織から植物を再生することが実証されている。この植物として、ヒマワリ、トマト、シロツメクサ、アブラナ、コットン、タバコ、ジャガイモ、トウモロコシ、イチゴ、イネ、その他多数の野菜作物を挙げることができる。
本発明においては、アグロバクテリウムTiベクターにより上記レタスをはじめとした可食植物を形質転換することができる。
様々な種の植物について、アグロバクテリウム(Agrobacterium tumefaciens)で形質転換された組織から植物を再生することが実証されている。この植物として、ヒマワリ、トマト、シロツメクサ、アブラナ、コットン、タバコ、ジャガイモ、トウモロコシ、イチゴ、イネ、その他多数の野菜作物を挙げることができる。
本発明においては、アグロバクテリウムTiベクターにより上記レタスをはじめとした可食植物を形質転換することができる。
形質転換体における融合タンパク質の発現量は、好ましくは形質転換体の乾燥重量1gあたり0.1mg~150mgであり、より好ましくは0.5mg~100mgであり、さらに好ましくは0.5mg~50mgである。
本発明の自然免疫増強剤は、前記融合タンパク質を含むものであればよいが、上記のような融合タンパク質をコードするDNAで形質転換され、該融合タンパク質を発現する形質転換体を含んでいてもよい。本発明の免疫増強剤は、前記融合タンパク質を含む形質転換体の全部を含んでいても、一部を含んでいてもよい。また、形質転換体をそのまま用いることもでき、乾燥、粉砕するなどして用いることもできる。特に、形質転換体が植物である場合、粉末化して使用することが好ましい。
形質転換植物の粉末化の方法は特に制限されないが、例えばジョークラッシャ、ジャイレトリクラッシャ、コーンクラッシャなどの圧縮破砕機、カッターミル、シュレッダーなどの剪断粗砕機、ハンマークラッシャーなどの衝撃破砕機、ロールクラッシャなどのロールミル、ディスインテグレーター、ケージミルなどの回転解砕機、コーヒーミルなどのスクリューミル、エッジランナーなどのロール転動ミル、スタンプミルなどの打槌ミル、遠心ローラーミル、ボールベアリングミル、バウルミル、ゼゴミル、オングミルなどのローラーミル、スイングハンマーミル、ピンミル、ケージミル、ターボタイプミル、遠心分級ミルなどの高速回転ミル、転動ボールミル、振動ボールミル、遊星ボールミル、CFミルなどの容器振動ミル、流通管式ミル、攪拌槽式ミル、アニュラー式ミル、気流吸い込み、気流衝突、衝突板衝突、流動層タイプなどのジェットミル、超音波破砕機などの粉砕機や石うす、乳鉢等の装置を使って粉砕する方法等が挙げられる。
なお、粉末化は植物を水分量が採取時の1/10以下になるまで乾燥させてから行うことが好ましい。乾燥方法としては、例えば減圧加熱乾燥、常圧加熱乾燥、スプレードライヤー、ドラムドライヤー、凍結乾燥等の方法が挙げられる。
形質転換植物の粉末の形状や大きさは、対象動物に投与され、体内に吸収されうる形状や大きさであれば特に制限されず、粒子状の粉末でもよい。
なお、粉末化は植物を水分量が採取時の1/10以下になるまで乾燥させてから行うことが好ましい。乾燥方法としては、例えば減圧加熱乾燥、常圧加熱乾燥、スプレードライヤー、ドラムドライヤー、凍結乾燥等の方法が挙げられる。
形質転換植物の粉末の形状や大きさは、対象動物に投与され、体内に吸収されうる形状や大きさであれば特に制限されず、粒子状の粉末でもよい。
融合タンパク質またはそれを含む形質転換体は、そのまま使用してもよいし、動物薬や動物用飼料に用いられる担体や添加剤と混合されて組成物として使用してもよい。ここで、担体としては、投与対象に有害な生理学的反応を引き起こさず、ワクチンに含まれる他の成分と有害な相互作用を生じない溶媒、希釈剤、賦形剤、結合剤、溶媒などが挙げられ、液体状のワクチン組成物の調製には例えば、水、生理食塩水、各種緩衝液が用いられる。添加剤としては、アジュバント、安定剤、pH調整剤、増粘剤、抗酸化剤、等張化剤、緩衝剤、溶解補助剤、懸濁化剤、保存剤、凍害防止剤、凍結保護剤、凍結乾燥保護剤、制菌剤などが例示される。
なお、前記アジュバントとしては、融合タンパク質の免疫原性を高める物質が使用できるが、例えば、水酸化アルミニウム、大腸菌の付着因子、例えば大腸菌の繊毛などが挙げられる。

本発明の自然免疫増強剤の剤型は、液剤、錠剤、軟膏、顆粒剤などが例示されるが、液剤が好ましく、例えば、上記融合タンパク質または形質転換体を水、生理食塩水、緩衝液、植物油などに懸濁又は溶解し、必要に応じて安定化剤等の添加剤を添加することにより得られる液状ワクチン組成物が好ましい。
例えば、形質転換体の粉末を使用する場合、0.5mg/ml~500mg/mlの濃度の懸濁液とすることが好ましく、1mg/ml~100mg/mlの濃度の懸濁液とすることがより好ましく、5mg/ml~50mg/mlの濃度の懸濁液とすることがさらに好ましい。なお、懸濁液とは、有効成分である前記形質転換体の粉末が液体中に分散されている組成物を意味し、粉末の一部が液体に溶解していてもよい。また、有効成分である抗原タンパク質の濃度が10ppm~15000ppmの濃度になるように懸濁することが好ましく、10ppm~5000ppmの濃度になるように懸濁することがより好ましく、50ppm~2000ppmの濃度になるように懸濁することがさらに好ましい。なお、形質転換体の粉末は粉末のまま供給され、使用者が、対象動物に投与する直前に水、生理食塩水、緩衝液、植物油などに懸濁して使用されるものでもよい。

本発明の自然免疫増強方法は、前記融合タンパク質または前記DNA構築物で形質転換された植物体などの形質転換体またはその乾燥物もしくは粉砕物を動物に投与することを特徴とする。投与対象としては、ブタ、ウシ、ニワトリ、ヒツジ、ヤギ、イヌ、ネコなどの非ヒト哺乳動物や魚類が挙げられる。
本発明の自然免疫増強剤はマクロファージ、好中球、樹状細胞及び/またはNK細胞等を活性化して非特異的な免疫応答を増強するため対象疾病としては特に限定されず、自然免疫によって予防または治療し得る疾患全般が含まれるが、例えば、ブタであれば、浮腫病、ブルセラ病、炭疽、破傷風、ブタ丹毒、ブタ赤痢、サルモネラ症、大腸菌症、萎縮性鼻炎、アクチノバシラス症、マイコプラズマ感染症、ブタ伝染性胃腸炎、ブタ流行性下痢発症、ブタインフルエンザ、日本脳炎、オーエスキー病、口蹄疫、ブタ水泡病、ブタコレラ、ブタ白血病、ブタ生殖器・呼吸器症侯群、ロタウイルス、回虫症、肺虫症、トキソプラズマ症、コクシジウム病などが挙げられる。ウシの対象疾病としては、牛肺疫、炭疽病、出血性敗血症、ブルセラ病、結核病、サルモネラ症、破傷風、牛疫、口蹄疫、流行性脳炎、狂犬病、水泡性口炎、リフトバレー病、ヨーネ病、ブルータング、アカバネ病、チュウザン病、ランピースキン病、牛ウイルス性下痢、牛白血病、ピロプラズマ病、アナプラズマ病などが挙げられる。ニワトリの対象疾病としてはサルモネラ感染症、マイコプラズマ病、鶏大腸菌症、ヘモフィルス、ニューカッスル病、高病原性鳥インフルエンザ、毒鶏伝染性気管支炎、鶏痘、鶏脳脊髄炎、コクシジウム病などが挙げられる。ヤギの対象疾病として、炭疽病、ブルセラ病、結核病、チュウザン病、口蹄疫、アカバネ病などが挙げられる。イヌの対象疾病として、レプトスピラ、細菌性腸炎、狂犬病、パルボウイルス感染症、ジステンバーウイルス感染症、犬伝染性気管支炎(ケンネルコフ)、コロナウイルス感染症、ヘルペスウイルス感染症、バベシア症などが挙げられる。ネコの対象疾病として、猫ヘモプラズマ感染症、リケッチア感染症、狂犬病、猫白血病ウイルス感染症、猫ヘルペスウイルス感染症、猫免疫不全ウイルス (FIV) 感染症、猫カリシウイルス感染症、猫ウイルス性鼻気管炎(FVR)、猫フィラリアなどが挙げられる。サカナの対象疾病として、連鎖球菌、ビブリオ病、イリドウイルス感染症などが挙げられる。
本発明の自然免疫増強剤は、特に、浮腫病、大腸菌性下痢症、PRRS(ブタ繁殖・呼吸障害症候群)、マイコプラズマ感染症(マイコプラズマ肺炎)、トリサルモネラ症等の様々な疾患の防除に有効である。

本発明の自然免疫増強剤をブタに投与する場合、投与時期は特に制限されないが、例えば、哺乳期~120日齢のブタに投与することができ、哺乳期~90日齢のブタに対して投与することが好ましく、また繁殖期前後の母ブタに対して投与することが好ましい。免疫の方法としては、前記DNA構築物で形質転換された植物体を母ブタに投与して、母ブタが産生した抗体を、乳汁により子ブタに与える方法、前記DNA構築物で形質転換された植物体を哺乳期~90日齢の子ブタに投与し、子ブタを直接免疫する方法等が挙げられる。
本発明の自然免疫増強剤を鶏に投与する場合、鶏の日齢に関係なく投与できる。
本発明の自然免疫増強剤をブタや鶏等の動物に投与する方法としては、飼料に、前記DNA構築物で形質転換された植物体またはその乾燥物もしくは粉砕物を混合して与える方法、点鼻投与など経鼻投与する方法などが挙げられる。経鼻投与により、少量の抗原を用いて生体に穏やかに免疫反応を惹起させることができる。本発明の自然免疫増強剤は、一定の間隔をおいて、複数回投与することが好ましい。例えば、4~7日おきに、合計2~3回投与する方法が挙げられる。
本発明の自然免疫増強剤を鶏に投与する場合、鶏の日齢に関係なく投与できる。
本発明の自然免疫増強剤をブタや鶏等の動物に投与する方法としては、飼料に、前記DNA構築物で形質転換された植物体またはその乾燥物もしくは粉砕物を混合して与える方法、点鼻投与など経鼻投与する方法などが挙げられる。経鼻投与により、少量の抗原を用いて生体に穏やかに免疫反応を惹起させることができる。本発明の自然免疫増強剤は、一定の間隔をおいて、複数回投与することが好ましい。例えば、4~7日おきに、合計2~3回投与する方法が挙げられる。
本発明の自然免疫増強剤の投与量は、対象動物の種類、体重、性別、週齢、対象疾患の種類などによって適宜定められるが、形質転換体を投与する場合、例えば、1回の投与量が、対象動物の体重1kgあたり0.01mg~100gである。
より具体的には、経鼻投与の場合、融合タンパク質の質量(絶対量)として、1日当たり好ましくは0.005mg~100mgである。また、形質転換体の乾燥質量(絶対量)として、1日当たり好ましくは0.01mg~200mgである。
注射投与の場合、融合タンパク質の質量(絶対量)として、1日当たり好ましくは0.005mg~0.5gである。また、形質転換体の乾燥質量(絶対量)として、1日当たり好ましくは0.01mg~1gである。
経口投与の場合、投与対象動物の体重1kgあたりの融合タンパク質の投与量として、1日当たり好ましくは0.005mg~50gである。また、投与対象動物の体重1kgあたりの形質転換体の投与量として、1日当たり好ましくは0.01mg~100g(乾燥質量)である。
より具体的には、経鼻投与の場合、融合タンパク質の質量(絶対量)として、1日当たり好ましくは0.005mg~100mgである。また、形質転換体の乾燥質量(絶対量)として、1日当たり好ましくは0.01mg~200mgである。
注射投与の場合、融合タンパク質の質量(絶対量)として、1日当たり好ましくは0.005mg~0.5gである。また、形質転換体の乾燥質量(絶対量)として、1日当たり好ましくは0.01mg~1gである。
経口投与の場合、投与対象動物の体重1kgあたりの融合タンパク質の投与量として、1日当たり好ましくは0.005mg~50gである。また、投与対象動物の体重1kgあたりの形質転換体の投与量として、1日当たり好ましくは0.01mg~100g(乾燥質量)である。
以下、本発明の実施例を説明するが、本発明はかかる実施例に限定されるものではない。
<実験手順>
(1)融合タンパク質の構造
供試材料として、腸管出血性大腸菌が生産する志賀毒素2eの無毒Bサブユニット(Stx2eB)と毒素原生大腸菌が生産する易熱性毒素の無毒Bサブユニット(LTB) (配列番号12)がPG12ペプチドリンカー(Matsui et al, Transgenic Res, 2011, 20:735-48:配列番号2)を介して連結された融合タンパク質を発現するレタスを用いた。融合タンパク質は、LTBがN末側に配置されたLTB-Stx2eB (LTB-B:配列番号15)およびStx2eBがN末側に配置されたStx2eB-LTB(B-LTB:配列番号16)の2種類を作製した。Stx2eBは糖鎖付加部位変異体(Asn73Se)を用いた(配列番号10)。この融合タンパク質をコードするDNAの発現のために、近傍配列改変型NtADH 5’-UTR(配列番号28)とAtHSPターミネーター(配列番号29)を使用した。さらにタバコ由来β-D-glucan exohydrolase分泌シグナルペプチドコード配列(配列番号24)および小胞体残留シグナル(HDEL)(配列番号26)を用いることでコンビネーション化ワクチン抗原候補タンパク質の高蓄積化をはかった。構築した遺伝子カセットはバイナリーベクターpRI909(TAKARA)に導入し、レタスの形質転換に用いた(図1)。
なお、Stx2eBがPG12ペプチドリンカーを介して2連結されたStx2eB-Stx2eB(Stx2eB-B)(配列番号17)も同様にして作製した。

(2)アグロバクテリウムによるレタスへの遺伝子導入
レタス(Lactuca sativa L.)品種グリーンウェーブ(タキイ種苗)をMS培地 [1/2×ムラシゲ・スクーグ培地用混合塩類(MS塩、和光純薬工業)、1×Murashige and Skoog vitamin solution(MSビタミン、Sigma-Aldrich)、3% ショ糖、0.8% 寒天、pH5.8] に無菌播種後、10-16日目の本葉を5 mm角程度に切断し、ベクターコンストラクトを有するバイナリープラスミド(pRI909)を保持するアグロバクテリウム(Agrobacterium tumefacience EHA105)懸濁液に本切片を10分間浸漬後、共存培地 [1× MS塩、1× MSビタミン、0.05 mg/l 6-ベンジルアミノプリン(BA)、0.1 mg/l 1-ナフチル酢酸(NAA)、0.1 M アセトシリンゴン、3% ショ糖、0.8% 寒天、pH5.8] に置床し、25℃、暗所で2日間培養した。切片を滅菌水で洗浄後、選抜培地 [1× MS塩、1× MSビタミン、0.05 mg/l BA、0.1 mg/l NAA、0.5 g/l ポリビニルピロリドン(PVP)、50 mg/lカナマイシン(Km)、250 mg セフォタキシム(Cef)、3% ショ糖、0.8% 寒天、pH5.8] に置床し、25℃、蛍光灯下(2000-3000 lux)で培養を行った。以降、不定芽が得られるまで3-4日間に1回(2回/週)の間隔で新しい選抜培地への移植を行った。不定芽から形成された再分化個体は発根培地[1/2× MS塩、1× MSビタミン、0.5 g/l PVP、250 mg Cef、3% ショ糖、0.8% 寒天、pH5.8] に移植し、同条件で培養を行った。以降、3-4日間に1回(2回/週)の間隔で新しい発根培地への移植を行った。発根した再分化個体は鉢植えにし、同条件で栽培を行った。

(3)レタス乾燥粉末の調製
上記(2)で得られた毒素融合タンパク質産生レタスを真空凍結乾燥機(FD-6BM-SQ、日本テクノサービス社)で凍結乾燥した。棚温度はなりゆき、乾燥期間は7日間とした。得られた乾燥粉末はブレンダー(Wonder crush/mill D3V-10、大阪ケミカル株式会社)を用い、10秒間の粉砕を三回行って粉末化し、B-LTBまたはStx2eB-B を含むレタス乾燥粉末を得た。
上記(2)で得られた毒素融合タンパク質産生レタスを真空凍結乾燥機(FD-6BM-SQ、日本テクノサービス社)で凍結乾燥した。棚温度はなりゆき、乾燥期間は7日間とした。得られた乾燥粉末はブレンダー(Wonder crush/mill D3V-10、大阪ケミカル株式会社)を用い、10秒間の粉砕を三回行って粉末化し、B-LTBまたはStx2eB-B を含むレタス乾燥粉末を得た。
(4)レタスからのタンパク質抽出
タンパク質抽出はTCA-acetone法(Shultz et al. Plant Mol Biol Rep, 2005, 23:405)に従い、液体窒素凍結後、-80℃で保存した遺伝子導入レタス本葉を用いて行った。100-200 mgのサンプルをTissueLyzer II(QIAGEN)を用い破砕後、サンプルの5倍量の TCA-acetone(10% トリクロロ酢酸、90% アセトン、0.07% 2-メルカプトエタノール)を添加、混合し、-20℃で1時間静置後、16,000×g、4℃、30分間遠心操作を行い、上清を除去し、タンパク質を含む沈殿を得た。さらに夾雑物を除去するために、サンプルの5倍量の acetone/BME(100% アセトン、0.07% 2-メルカプトエタノール)を添加、混合し、16,000×g、4℃、10分間遠心操作を行い、上清を除去した。本夾雑物除去操作はさらに2回行った。沈殿は減圧乾燥後、サンプルの2倍量の 抽出Iバッファー [0.5 M 塩化ナトリウム、5 mM イミダゾール、6 M 尿素、20 mM トリスヒドロキシメチルアミノメタン(Tris)-HCl、pH7.9] に懸濁し、16,000×g、4℃、10分間遠心を行い、上清を回収、タンパク質溶液を得た。タンパク質濃度の測定は、Protein Assay Kit II(Bio-Rad)を用い行った。
タンパク質抽出はTCA-acetone法(Shultz et al. (5)ウエスタン解析
得られたタンパク質溶液をマイクロチューブに適量入れ、同量のサンプルバッファー(EZ Apply、ATTO製)を加え混合し、沸騰水中で5分間加温し、サンプルのSDS化を行った。タンパク質定量時の標準物質には大腸菌で生産し精製したLTBを用いた。具体的には特許文献2のL+を本特許文献に示す方法でタンパク質生産用大腸菌(BL21 DE3)にて発現し、ガラクトースカラムで精製した。これを抽出Iバッファーで2倍希釈を繰り返すことにより希釈系列を作製し、これら希釈系列をスタンダードとして用いた。
タンパク質の電気泳動(SDS-PAGE)は、電気泳動槽(ミニプロティアンTetraセル)およびミニプロティアンTGX-ゲル(BIO RAD)を用いた。電気泳動バッファー(EZ Run、ATTO製)を入れ、ウェルにSDS化したサンプルを5 μlアプライし、200 V定電圧で40分間行った。
電気泳動後のゲルは、トランスブロット転写パック(BIO RAD)を用い、トランスブロットTurbo(BIO RAD)でブロッティングを行った。
ブロッティング後のメンブレンはブロッキング溶液(TBS系, pH7.2、ナカライテスク)に浸し、室温で1時間振とうまたは4℃で16時間静置後、TBS-T(137 mM 塩化ナトリウム、2.68 mM 塩化カリウム、1% ポリオキシエチレンソルビタンモノラウラート、25 mM Tris-HCl、pH 7.4)中で室温、5分間の振とうを3回行い洗浄した。LTBタンパク質の検出には、抗血清Rabbit-Antiserum Anti-LTp 991109(inactive)(0.1% NaN3)AO をTBS-Tで10,000倍希釈して使用した。本希釈液中にメンブレンを浸し、室温で2時間振とうすることにより抗原抗体反応を行い、TBS-T中で室温、5分間の振とうを3回行い洗浄した。二次抗体にはTBS-Tで10,000倍希釈したAnti-Rabbit IgG, AP-linked Antibody(Cell Signaling TECHNOLOGY)を使用した。本希釈液中にメンブレンを浸し、室温で1時間振とうすることにより抗原抗体反応を行い、TBS-T中で室温、5分間の振とうを3回行い洗浄した。アルカリホスファターゼによる発色反応は、発色液(0.1 M 塩化ナトリウム、5 mM 塩化マグネシウム、0.33 mg/mlニトロブルーテトラゾリウム、0.33 mg/ml 5-ブロモ-4-クロロ-3-インドリル-リン酸、0.1 M Tris-HCl、pH9.5)中にメンブレンを浸し、室温で7分間振とうすることにより行い、メンブレンを蒸留水で洗浄した後、常温で乾燥した。
発色したメンブレンはスキャナー(PM-A900、エプソン)により解像度600 dpiで画像化し、画像解析ソフト(CS Analyzer ver. 3.0、アトー)を用い、LTBタンパク質の定量を行った。
得られたタンパク質溶液をマイクロチューブに適量入れ、同量のサンプルバッファー(EZ Apply、ATTO製)を加え混合し、沸騰水中で5分間加温し、サンプルのSDS化を行った。タンパク質定量時の標準物質には大腸菌で生産し精製したLTBを用いた。具体的には特許文献2のL+を本特許文献に示す方法でタンパク質生産用大腸菌(BL21 DE3)にて発現し、ガラクトースカラムで精製した。これを抽出Iバッファーで2倍希釈を繰り返すことにより希釈系列を作製し、これら希釈系列をスタンダードとして用いた。

発色したメンブレンはスキャナー(PM-A900、エプソン)により解像度600 dpiで画像化し、画像解析ソフト(CS Analyzer ver. 3.0、アトー)を用い、LTBタンパク質の定量を行った。
(6)マクロファージ活性化試験1
自然免疫の活性化指標としてマクロファージの活性化に伴うTNFαの産生量を測定し、各被験物質の自然免疫誘導能の比較検討を行った。
手順は以下の通りである。なお、供試サンプルとしては、遺伝子組換え大腸菌で作製、精製したStx2eB-Stx2eB及びStx2eB-Hisを用いた。
(6-1)前培養
RAW264.7細胞は、10%の牛胎児血清、100μg/mLペニシリン、ストレプトマイシンを含有するRPMI1640培地にて継代培養したものを用いた。
(6-2)試験操作
T25培養フラスコ 4本にて前培養した細胞をピペッティングにより壁から剥がし、得られた細胞の懸濁液を50mLコニカルチューブに移した。チューブを室温で遠心分離し(1000rpm、5分間)、上清をデカンテーションで捨て、細胞を回収した。タッピングにより細胞をほぐした後、培養液5mLを加え、ピペッティングによって細胞を均一に懸濁した。11μLを別のチューブに移し0.5%トリパンブルー11μLを添加した後、血液計算板に液を移して細胞数と生存率を測定した。生存率が90%以上であったので、残液を試験に用いた。
測定した細胞数に基づいて、残液に培養液を加えて希釈し8×105cells/mLになるよう細胞数を調製した。この細胞懸濁液を100μLずつ96well平底プレートの各ウェルに加えた。インキュベータに移して、細胞がウェルの底に接着して伸展するまで6時間前培養を行った。
被験液はそれぞれ終濃度の4倍濃度のものを調製し、40μg/mLに調製したポリミキシンB溶液と等量混合した。6時間前培養が終了したプレートを取り出し、ウェルに100μLずつ検体を加えた。各検体を添加後、24時間培養し、培養上清100μLずつ別の96穴平底プレートに移した。Medium群の培養上清は、1%BSA含有PBS(-)溶液により、5倍希釈液を300μL調製した、他の群の培養上清は、1%BSA含有PBS(-)溶液により100倍希釈液を1mL調製した。希釈液をTNFα ELISA測定に用いた。測定方法は、キット記載の方法に従い測定した。

マクロファージ活性化試験1と同様の試験方法で評価を実施した。被験物質は遺伝子組換えレタスのジュースを所定の濃度になるように添加した。添加濃度はStx2eBの濃度を指標として調整した。

マクロファージ活性化試験1と同様の試験方法で評価を実施した。被験物質は遺伝子組換えレタスのジュースを所定の濃度になるように添加した。添加濃度はStx2eBの濃度を指標として調整した。
(8)PRRSウイルス感染防御試験
B-LTBレタスの経口投与によるPRRSウイルス予防効果確認試験を行った。試験に供試した子ブタは分娩後24時間目までは初乳を与えず人工乳を与え、以降は母ブタ哺乳に戻した。分娩後10、16、22日目にワクチンレタス(LTB 1.7 mg相当量/回)もしくは空ベクターレタスの凍結乾燥粉末を水に懸濁し、チューブを接続したシリンジで強制投与した(図2)。陽性対照として、PRRSウイルスの中和エピトープをLTBに融合した抗原を発現するレタス(ER LTB-ectGP5、WO2016/021276、LGと略記する)を投与した。供試頭数は、陰性対照が2頭、B-LTBおよびLGは各3頭とした。北米III型PRRSウイルスを経鼻噴霧により25日目に投与し攻撃した。体重、飼料摂取量を測定するとともに、臨床症状を毎日観察し、PRRSに典型的な症状である呼吸器症状(0:なし 1:軽度 2:中度 3:重度)について観察しスコア化した。PRRSウイルスによる攻撃21日目に安楽死後、剖検を行い、各臓器及び組織の異常を肉眼的に観察した。主要臓器の病理標本を作製し、病理組織学的所見を観察した。

(9)マイコプラズマ感染予防効果
LTB-BレタスおよびB-LTBレタスの経口投与により抗体誘導が行われるかどうかを解析する目的で、以下のスケジュールで投与を行った(図3)。すなわち、7、14、21、22、23、28、29、30、35、36、37、42、49日齢でレタスを子ブタに経口投与した。一回あたりの抗原投与量は、LTB相当量で1.7 mg、Stx2eB相当量では1.0 mgとした。供試頭数は、陰性対照とB-LTBが3頭、LTB-Bが4頭とした56日齢で解剖を行った。

(10)サルモネラ感染防除試験
Salmonella Enteritidis実験感染鶏に対するレタスワクチン給与の効果を以下の手順で調べた。なお、供試物質としては、Stx2eB-Stx2eBを生産する遺伝子組換えレタスを凍結乾燥し、ブレンダーで粉砕した粉末を用いた。
(10-1)試験群
無投与対照群,レタスワクチン強制経口投与A群,レタスワクチン強制経口投与B群:各14羽。
(10-2)飼育条件
1日齢ヒナ(名古屋コーチン初生オス)を導入後,株式会社栄養・病理学研究所内の閉鎖系の動物飼育室で,一般的な飼育管理を行う。SE感染後は,株式会社栄養・病理学研究所内の閉鎖系の動物飼育室にアイソレーター内で飼育する。部屋を空調で,またケージを保温灯などで温度管理をした。飼料及び飲水は試験期間を通して自由摂取とした。
(10-3)群分け
市販鶏を導入後,体重測定を実施し,体重が均一になるように3群に群分けし,群ごとに14羽ずつケージに入れて試験を開始した。感染後はアイソレーター内に収容した。各群に基礎飼料及び飲水を1日齢から試験終了時まで不断給餌した。なお,全供試鶏はマーキング等で個体識別した。また,各群所定の被験物質を飲料水で懸濁した溶液を作製し,所定の日齢で強制経口投与した。
(10-4)SE感染
15日齢雛に,Salmonella enterica subsp. enterica serovar Enteritidis (SE) 野外分離株の培養菌液1 mLを強制経口投与する。培養液中の菌数は約1.0×106cells/mLとした。
(10-5)経過観察
飼育期間はSE感染後7日間とする。SE感染後0,3及び7日に体重測定を行う。体重測定時に飼料摂取量も併せて測定する。試験期間中は毎日,一般臨床症状を観察した。
(10-6)剖検
SE感染後3及び7日に,各群7羽ずつ剖検を行う。剖検時に採血を行う。盲腸内容物を採取した。
(10-7)分析
盲腸内容物1 g中のSE菌数をreal-time PCRを用いて測定した。

1日齢ヒナ(名古屋コーチン初生オス)を導入後,株式会社栄養・病理学研究所内の閉鎖系の動物飼育室で,一般的な飼育管理を行う。SE感染後は,株式会社栄養・病理学研究所内の閉鎖系の動物飼育室にアイソレーター内で飼育する。部屋を空調で,またケージを保温灯などで温度管理をした。飼料及び飲水は試験期間を通して自由摂取とした。
(10-3)群分け
市販鶏を導入後,体重測定を実施し,体重が均一になるように3群に群分けし,群ごとに14羽ずつケージに入れて試験を開始した。感染後はアイソレーター内に収容した。各群に基礎飼料及び飲水を1日齢から試験終了時まで不断給餌した。なお,全供試鶏はマーキング等で個体識別した。また,各群所定の被験物質を飲料水で懸濁した溶液を作製し,所定の日齢で強制経口投与した。
15日齢雛に,Salmonella enterica subsp. enterica serovar Enteritidis (SE) 野外分離株の培養菌液1 mLを強制経口投与する。培養液中の菌数は約1.0×106cells/mLとした。
(10-5)経過観察
飼育期間はSE感染後7日間とする。SE感染後0,3及び7日に体重測定を行う。体重測定時に飼料摂取量も併せて測定する。試験期間中は毎日,一般臨床症状を観察した。
(10-6)剖検
SE感染後3及び7日に,各群7羽ずつ剖検を行う。剖検時に採血を行う。盲腸内容物を採取した。
(10-7)分析
盲腸内容物1 g中のSE菌数をreal-time PCRを用いて測定した。
(11)経鼻投与試験~レタスワクチンのブタ浮腫病評価
Stx2eB-B産生レタス乾燥粉末60mg(Stx2eB-B濃度は5mg/gレタス乾燥粉末)を、投与直前に2mlのリン酸緩衝液(PBS)に懸濁し、経鼻投与用ワクチン懸濁液(粉末濃度30mg/ml, 抗原濃度0.15mg/ml(150ppm), pH6.5)を得た。
被験用のブタについて、娩出時は産仔全頭の初乳を24時間制限した後、18日齢までは通常飼育し、試験を開始した。18日齢で離乳させ、群分けした。21日齢で上記経鼻投与用ワクチン懸濁液を経鼻投与(1mlずつ片鼻孔に投与)した後、25日齢から連続して3日間浮腫病用攻撃菌(Stx2e毒素保有大腸菌)を強制経口投与した。攻撃後、11日間を観察期間とし、毎日臨床的な観察(元気、食欲、目の周りの浮腫、神経失調、運動障害、横臥、下痢、死亡)を行い、下記に示すスコアを基準にして記録した。
試験豚の体重(強制投与時)は以下の通り。
プラセボ 7.9, 8.4, 6.0, 6.9 平均7.3kg、標準偏差1.1
経鼻 4.7, 8.5, 7.2, 7.7 平均7.0kg、標準偏差1.6

<結果>
(1)マクロファージ活性化試験1
結果を表2に示す。Stx2eB-Stx2eB、Stx2eB-Hisともに濃度依存的にTNFα産生量が増加することからマクロファージの活性化能があることが示唆された。Stx2eB-Stx2eB、Stx2eB-Hisの比較では1000ng/ml、5000ng/mlの濃度で有意にStx2eB-Stx2eBがStx2eB-Hisより高いTNFα産生量を示し、マクロファージ活性化能が高いことから、Stx2eBをペプチドリンカーでつなぎタンデム化することでStx2eB単独の場合より更にマクロファージの活性化能が高くなることが示された。
(1)マクロファージ活性化試験1
結果を表2に示す。Stx2eB-Stx2eB、Stx2eB-Hisともに濃度依存的にTNFα産生量が増加することからマクロファージの活性化能があることが示唆された。Stx2eB-Stx2eB、Stx2eB-Hisの比較では1000ng/ml、5000ng/mlの濃度で有意にStx2eB-Stx2eBがStx2eB-Hisより高いTNFα産生量を示し、マクロファージ活性化能が高いことから、Stx2eBをペプチドリンカーでつなぎタンデム化することでStx2eB単独の場合より更にマクロファージの活性化能が高くなることが示された。
(2)マクロファージ活性化試験2
結果を表3に示す。全てのレタスサンプルで濃度依存的にTNFα産生量の増加が見られ、マクロファージの活性化能が示された。また、プラセボレタスに比べ1000ng/ ml及び5000ng/ mlの添加区でLTB-Stx2eBレタス、Stx2eB-Stx2eBレタスのTNFα産生量が有意に高く、マクロファージ活性能が高いことが示された。特に、Stx2eB-Stx2eBレタスは5000ng/mlの添加区でTNFα産生量が顕著に高い結果であった。一方、プラセボレタスにも一定のマクロファージ活性化能があることもわかったが、レタス成分中に含まれる多糖類やサポニン成分などの影響が考えられる。
結果を表3に示す。全てのレタスサンプルで濃度依存的にTNFα産生量の増加が見られ、マクロファージの活性化能が示された。また、プラセボレタスに比べ1000ng/ ml及び5000ng/ mlの添加区でLTB-Stx2eBレタス、Stx2eB-Stx2eBレタスのTNFα産生量が有意に高く、マクロファージ活性能が高いことが示された。特に、Stx2eB-Stx2eBレタスは5000ng/mlの添加区でTNFα産生量が顕著に高い結果であった。一方、プラセボレタスにも一定のマクロファージ活性化能があることもわかったが、レタス成分中に含まれる多糖類やサポニン成分などの影響が考えられる。
(3)PRRS感染防御試験
PRRSウイルス感染後毎日、呼吸症状をスコア化した。感染後0-6日、7-13日、14-21日の各6日間でのスコア合計を各個体につき算出し、群内で平均値と標準偏差を算出した(図4)。その結果、有意差はないものの、B-LTB投与群で陰性対照に比べ症状の緩和が見られた。またその効果はPRRS抗原を含むLGとそん色なかった。
PRRSウイルス感染後毎日、呼吸症状をスコア化した。感染後0-6日、7-13日、14-21日の各6日間でのスコア合計を各個体につき算出し、群内で平均値と標準偏差を算出した(図4)。その結果、有意差はないものの、B-LTB投与群で陰性対照に比べ症状の緩和が見られた。またその効果はPRRS抗原を含むLGとそん色なかった。
次に感染後期間(25日から46日齢まで)の飼料要求率を比較した。有意差はないものの、B-LTB投与群で陰性対照に比べ、飼料要求率の改善が見られた(図5)。
剖検時の肺の所見では、陰性対照レタス投与群では肺の特に下葉に灰色の変色が見られたが、B-LTB投与群では改善傾向が確認できた(図6)。
(4)マイコプラズマ
LTB-BおよびB-LTB投与ブタを56日目に剖検した。その結果、一部の個体にマイコプラズマの自然感染が認められた。個体別にマイコプラズマ肺炎の程度をスコア化した(-, +, ++)。陰性対照レタス投与群では3頭中3頭で所見が見られたのに対し、LTB-BおよびB-LTB投与群では所見のあった個体数が減少した(表4)。
LTB-BおよびB-LTB投与ブタを56日目に剖検した。その結果、一部の個体にマイコプラズマの自然感染が認められた。個体別にマイコプラズマ肺炎の程度をスコア化した(-, +, ++)。陰性対照レタス投与群では3頭中3頭で所見が見られたのに対し、LTB-BおよびB-LTB投与群では所見のあった個体数が減少した(表4)。
(5)サルモネラ
体重解析の結果を表5に示す。プラセボレタス投与区では15日齢でのサルモネラ菌攻撃後3日目では雛の増体重は平均16gにとどまったが、ワクチンレタス投与区では5回投与区で平均28g、2回投与区で29gとサルモネラ菌の影響を緩和することが示された。

菌数解析の結果を表6に示す。ワクチンレタス5回投与区では雛の盲腸内容物中のサルモネラ菌数はサルモネラ菌攻撃後3日目で6.92 (log cells/g) とプラセボレタス投与区7.41 (log cells/g)に比べサルモネラ菌数が68%減少し、7日目でも6.62 (log cells/g)とプラセボレタス投与区7.03 (log cells/g)に比べで61%減少していた。
ワクチンレタス2回投与区では雛の盲腸内容物中のサルモネラ菌数はサルモネラ菌攻撃後7日目で6.69 (log cells/g) とプラセボレタス投与区7.03 (log cells/g)に比べ菌数が54%減少した。
これらの結果から、Stx2eB-Bレタスをニワトリ雛にサルモネラ菌の攻撃の前に投与することで腸内のサルモネラ菌数を低減し、増体重の減少を防ぐことが判明した。
ワクチンレタス2回投与区では雛の盲腸内容物中のサルモネラ菌数はサルモネラ菌攻撃後7日目で6.69 (log cells/g) とプラセボレタス投与区7.03 (log cells/g)に比べ菌数が54%減少した。
これらの結果から、Stx2eB-Bレタスをニワトリ雛にサルモネラ菌の攻撃の前に投与することで腸内のサルモネラ菌数を低減し、増体重の減少を防ぐことが判明した。
(6)経鼻投与試験
表7に示すように、ワクチンを経鼻投与されたブタはプラセボ群と比較して顕著に臨床スコアが小さく、Stx2eB-B産生レタスの乾燥粉末を含む経鼻ワクチンはブタ浮腫病の予防効果に優れることが分かった。

Claims (15)
- 志賀毒素2eのBサブユニット(Stx2eB)、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選択される2以上の毒素Bサブユニットの融合タンパク質を含む、自然免疫増強剤。
- 前記融合タンパク質がStx2eBとLTBを含む融合タンパク質である、請求項1に記載の自然免疫増強剤。
- 前記融合タンパク質がStx2eBを2つ以上含む融合タンパク質である、請求項1に記載の自然免疫増強剤。
- 前記Stx2eBの73位のAsn残基がSer残基に置換されている、請求項1~3のいずれか一項に記載の自然免疫増強剤。
- 前記2以上の毒素Bサブユニットがペプチドリンカーを介して融合されている、請求項1~4のいずれか1項に記載の自然免疫増強剤。
- 前記ペプチドリンカーがPG12(配列番号2)、PG12v2(配列番号4)、PG17(配列番号30)、もしくはPG22(配列番号31)、またはこれらの配列と80%以上同一性を有するアミノ酸配列を有するペプチドである、請求項5に記載の自然免疫増強剤。
- 前記融合タンパク質が配列番号15~17のいずれかのアミノ酸配列を有する、請求項1~6のいずれか1項に記載の自然免疫増強剤。
- 前記融合タンパク質をコードするDNAで形質転換され、前記融合タンパク質を発現した形質転換体を含む、請求項1~7のいずれか1項に記載の自然免疫増強剤。
- 前記形質転換体が植物である、請求項8に記載の自然免疫増強剤。
- 前記植物が植物の乾燥粉末又はその懸濁液として含まれる、請求項9に記載の自然免疫増強剤。
- 経鼻投与用である、請求項1~10のいずれか1項に記載の自然免疫増強剤。
- ブタ繁殖・呼吸障害症候群ウイルス、サルモネラ菌またはマイコプラズマに対する免疫応答を誘導する、請求項1~11のいずれか1項に記載の自然免疫増強剤。
- マクロファージ活性化作用を有する、請求項1~12のいずれか1項に記載の自然免疫増強剤。
- 志賀毒素2eのBサブユニット(Stx2eB)、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選択される2以上の毒素Bサブユニットの融合タンパク質を含む、マクロファージ活性化剤。
- 志賀毒素2eのBサブユニット(Stx2eB)、大腸菌易熱性毒素のBサブユニット(LTB)およびコレラ毒素のBサブユニット(CTB)から選択される2以上の毒素Bサブユニットの融合タンパク質または該融合タンパク質をコードするDNAで形質転換され、前記融合タンパク質を発現した形質転換植物を非ヒト哺乳動物に投与することを特徴とする、非ヒト哺乳動物の自然免疫を増強させる方法。
