Abstract
Description
Cre/loxPシステムにおいては、loxP配列と呼ばれるDNA配列に対し組換え酵素であるCreタンパク質が働いて、部位特異的遺伝子組換えが起こる。
Creタンパク質は、バクテリオファージP1に由来する酵素であって、遺伝子工学技術において、幅広く用いられている(非特許文献1)。
具体的には、光依存的制御技術として、Cre/loxPシステムと光スイッチタンパク質の組合せであるCRY2-CreNとCIBN-CreCを用いる技術が開示されている(特許文献1、非特許文献2、3)。
また、薬物存在下で遺伝子組換えを制御する技術として、split-Cre-FRB/FKBPシステムが開示されている(非特許文献4、5)。
本発明者らは、光依存的にホモ二量体を形成するNeurospora crassa由来のVividタンパク質を改変し、光の照射により二量体の形成及び解離を精密に制御することができる光依存的に二量体を形成する2つのタンパク質のセットを開発した(特許文献2、非特許文献8)。また、ゲノム編集ツールとしてCRISPR-Cas9システムに利用されるCas9を2つに分割したフラグメントと、光依存的に又は薬物存在下で二量体を形成する2つのポリペプチドとを融合させた2つのポリペプチドのセットを開発した(特許文献3、非特許文献9)。

本発明に係る2つのポリペプチドのセットにおいては、光依存的に二量体を形成する2つのタンパク質が、光照射により二量体を形成することにより、光依存的に二量体を形成する2つのタンパク質に結合するCreタンパク質のN末端側フラグメントと、Creタンパク質のC末端側フラグメントがCreタンパク質として再構成されて、Creタンパク質のレコンビナーゼ活性を回復する。よって、本発明に係る2つのポリペプチドのセットは、光依存的遺伝子組換えに用いることができる。
すなわち、本発明において、Creタンパク質のN末端側フラグメントにおけるアミノ酸配列と、Creタンパク質のC末端側フラグメントのアミノ酸配列を比較すると、Creタンパク質のN末端側フラグメントは、Creタンパク質のC末端側フラグメントに比して、配列番号:1において、N末端側の領域のアミノ酸配列を有する。Creタンパク質のN末端側フラグメントのC末端のアミノ酸は、配列番号:1の配列において、C末端側フラグメントのN末端のアミノ酸よりもN末端側のアミノ酸である。
Creタンパク質の部分配列からなるフラグメントとは、配列番号:1のアミノ酸配列を有するCreタンパク質を切断した場合に、配列番号:1のアミノ酸配列においてN末端のアミノ酸から切断部のアミノ酸までのアミノ酸配列における部分配列を有するか、あるいは配列番号:1のアミノ酸配列において切断部のアミノ酸からC末端のアミノ酸までのアミノ酸配列における部分配列を有するフラグメントであることを意味する。
配列番号:1のアミノ酸配列のCreタンパク質のN末端側フラグメントとC末端側フラグメントが、部分配列からなるフラグメントである場合、それぞれのフラグメントにおいて、配列番号:1のアミノ酸配列のN末端のアミノ酸及び/又は配列番号:1のアミノ酸配列のC末端のアミノ酸を有さなくてもよいことを意味する。
配列番号:1のアミノ酸配列のCreタンパク質のN末端側フラグメントとC末端側フラグメントは、配列番号:1のアミノ酸配列に変異を含む配列を有していてもよく、Creタンパク質の部分配列に変異を含む配列を有していてもよい。
配列番号:1のアミノ酸配列の59位~68位は、Creタンパク質のB-α-helixとC-α-helixの間にあたるアミノ酸に相当し、配列番号:1のアミノ酸配列の101位~111位は、Creタンパク質のD-α-helixとE-α-helixの間にあたるアミノ酸に相当する。
配列番号:1のアミノ酸配列の59位~68位と、配列番号:1のアミノ酸配列の101位~111位は、それぞれ、結晶構造解析の結果(Guo, F., et al., Nature, 389, 40-46 (1997))から,α-helix間のフレキシブルなループ構造であることが明らかになっている領域である。
Creタンパク質のN末端側フラグメントとC末端側フラグメントが、配列番号:1のアミノ酸配列の59位~68位の領域内に切断部位を有するフラグメントであるとは、配列番号:1のアミノ酸配列の59位~68位の領域内に切断部位を有していればよいことを意味し、Creタンパク質のN末端側フラグメントは、少なくとも59位のアミノ酸までのアミノ酸配列を含有するフラグメントであり、Creタンパク質のC末端側フラグメントは、少なくとも68位のアミノ酸以降のアミノ酸配列を含有するフラグメントである。
Creタンパク質のN末端側フラグメントとC末端側フラグメントが、配列番号:1のアミノ酸配列の101位~111位の領域内に切断部位を有するフラグメントであるとは、配列番号:1のアミノ酸配列の101位~111位の領域内に切断部位を有していればよいことを意味し、Creタンパク質のN末端側フラグメントは、少なくとも101位のアミノ酸までのアミノ酸配列を含有するフラグメントであり、Creタンパク質のC末端側フラグメントは、少なくとも111位のアミノ酸以降のアミノ酸配列を含有するフラグメントである。
Creタンパク質のN末端側フラグメントにおけるN末端のアミノ酸及びCreタンパク質のC末端側フラグメントにおけるC末端のアミノ酸は、N末端側フラグメントとC末端側フラグメントとが、光依存的に遺伝子組換え活性を回復することができるものであれば、配列番号:1のアミノ酸配列において、特に限定されるものではない。
配列番号:1のアミノ酸配列の60位~67位の領域については、領域内の全てのアミノ酸を、あるいは、一部のアミノ酸をCreタンパク質のN末端側フラグメントとC末端側フラグメントのいずれかのフラグメントが有していてもよい。Creタンパク質のN末端側フラグメントとC末端側フラグメントが、60位~67位の領域の一部又は全部を有する場合には、60位~67位の領域内のアミノ酸配列において変異があってもよい。
配列番号:1のアミノ酸配列の1位~101位の領域及び111位~343位の領域を含むよう設計するとは、スプリットCreが二量化した際に、Creタンパク質のレコンビナーゼ活性を保持しているのであれば、特に限定されず、配列番号:1のアミノ酸配列の1位~101位の領域内及び111位~343位の領域内の部分配列をそれぞれ、Creタンパク質のN末端側フラグメントとC末端側フラグメントが有していてもよい。また、Creタンパク質のN末端側フラグメントとC末端側フラグメントは、配列番号:1のアミノ酸配列の1位~101位の領域内及び111位~343位の領域内のアミノ酸配列において変異があってもよく、あるいは、変異を有し、かつ、その部分配列であるフラグメントであってもよい。
配列番号:1のアミノ酸配列の102位~110位の領域については、領域内の全てのアミノ酸を、あるいは、一部のアミノ酸をCreタンパク質のN末端側フラグメントとC末端側フラグメントのいずれかのフラグメントが有していてもよい。Creタンパク質のN末端側フラグメントとC末端側フラグメントが、102位~110位の領域の一部又は全部を有する場合には、102位~110位の領域内のアミノ酸配列において変異があってもよい。
Creタンパク質のN末端側フラグメントとC末端側フラグメントが部分配列からなるフラグメントである場合に、Creタンパク質のN末端側フラグメントのC末端のアミノ酸と、C末端側フラグメントのN末端のアミノ酸は、それぞれ、切断部位に当るアミノ酸であることが好ましい。
Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~101位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の111位~343位の領域を少なくとも有するポリペプチドのセットであることが好ましい。
本発明においては、Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~59位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の60位~343位の領域を少なくとも有するポリペプチドのセットであることがより好ましく、Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~104位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の106位~343位の領域を少なくとも有するポリペプチドのセットであることがより好ましい。
本明細書において、配列番号:1のアミノ酸配列に対して、1から数個のアミノ酸の付加、置換、又は欠失を含むアミノ酸配列からなるフラグメント、又は、配列番号:1のアミノ酸配列に対して、80%以上の配列同一性を有するアミノ酸配列からなるフラグメントという場合に同様である。
なお、N末端側フラグメントが配列番号:1の19位のアミノ酸から59位のアミノ酸で構成され、C末端側フラグメントが60位のアミノ酸から343位のアミノ酸で構成される場合において、例えば、1個~9個のアミノ酸の付加、置換、又は欠失がある場合、それぞれ、アラインメントの結果として、324/343(約94%)~316/343(約92%)が重複する領域となる。N末端側フラグメントが配列番号:1の19位のアミノ酸から104位のアミノ酸で構成され、C末端側フラグメントが106位のアミノ酸から343位のアミノ酸で構成される場合において、例えば、1個~9個のアミノ酸の付加、置換、又は欠失がある場合、それぞれ、アラインメントの結果として、323/343(約94%)~315/343(約92%)が重複する領域となる。

非天然アミノ酸としては、例えば、主鎖の構造が天然型と異なる、α,α-二置換アミノ酸(α-メチルアラニンなど)、N-アルキル-α-アミノ酸、D-アミノ酸、β-アミノ酸、α-ヒドロキシ酸や、側鎖の構造が天然型と異なるアミノ酸(ノルロイシン、ホモヒスチジンなど)、側鎖に余分のメチレンを有するアミノ酸(「ホモ」アミノ酸、ホモフェニルアラニン、ホモヒスチジンなど)及び側鎖中のカルボン酸官能基アミノ酸がスルホン酸基で置換されるアミノ酸(システイン酸など)等が挙げられるが、これらに限定されない。
本明細書においてアミノ酸は、慣用的な一文字表記又は三文字表記で示される場合もある。一文字表記又は三文字表記で表されたアミノ酸は、それぞれの変異体や誘導体を含む場合もある。
本明細書において、「光依存的に二量体を形成する2つのタンパク質」(以下、「マグネット」と記載する場合がある)は、光を照射することによってヘテロ二量体を形成するタンパク質のペアである。マグネットは、光、好ましくは、青色光を照射することによってヘテロ二量体を形成し、光照射を止めることによりヘテロ二量体が迅速に解離することで、本発明に係る2つのポリペプチドのセットにおける二量体の形成及び解離を精密に制御することができ、光依存的に、Creタンパク質のレコンビナーゼ活性を回復させる。
本発明においては、光依存的に二量体を形成する2つのタンパク質は、それぞれ配列番号:2のアミノ酸配列からなるポリペプチド、又はその変異体であり、少なくとも一方のタンパク質において、N末端αヘリックスに位置する少なくとも1つのアミノ酸が置換されている、2つの異なるポリペプチドのセットである。
Vividタンパク質の変異体は、例えば、配列番号:2のアミノ酸配列と、70%以上、75%以上、80%以上、85%以上、90%以上、95%以上、98%以上、99%以上の配列同一性を有するタンパク質であってもよい。
マグネットは、Vividタンパク質とVividタンパク質のペア、Vividタンパク質とVividタンパク質の変異体のペア、Vividタンパク質の変異体とVividタンパク質の変異体のペアのいずれのペアであってもよく、ペアの少なくとも一方のポリペプチドにおいて、N末端αヘリックスに位置する少なくとも1つのアミノ酸が置換されている。
本明細書において、Vividタンパク質の変異体は、N末端αヘリックス以外の場所において、配列番号:2のアミノ酸配列とは異なる配列を有するポリペプチドである。Vividタンパク質の変異体には、N末端を切断したポリペプチド(配列番号:2のアミノ酸配列のうち、37位から186位のアミノ酸で構成されるポリペプチド)であってもよい。
N末端αヘリックスとしては、配列番号:2における47位~56位のアミノ酸であることが好ましく、本発明に係る光依存的に二量体を形成する2つのタンパク質は、少なくとも一方のタンパク質において、配列番号:2における47位~56位のアミノ酸のうち少なくとも1つのアミノ酸が置換されていることが好ましく、2つのタンパク質において、それぞれ配列番号:2における47位~56位のアミノ酸のうち少なくとも1つのアミノ酸が置換されていることがより好ましい。
N末端αへリックスにおける変異は、配列番号:2の47位、48位、50位、51位、52位、54位、及び55位のアミノ酸の少なくとも1つのアミノ酸における変異であることが好ましい。
側鎖に正電荷を有するアミノ酸は、天然のアミノ酸であっても非天然のアミノ酸であってもよく、天然のアミノ酸の場合には、リシン、アルギニン、及びヒスチジンが挙げられる。側鎖に負電荷を有するアミノ酸は、天然のアミノ酸であっても非天然のアミノ酸であってもよく、天然のアミノ酸の場合には、アスパラギン酸とグルタミン酸が挙げられる。
配列番号:2の52位と55位のそれぞれに対応するアミノ酸における置換は、同じアミノ酸に置換されていてもよいし、異なるアミノ酸に置換されていてもよい。
また、マグネットを構成する2つのタンパク質が、アミノ酸置換前に、配列番号:2とは異なるアミノ酸配列を有する場合は、具体的には、Vividタンパク質の変異体である場合は、配列番号:2のX位に対応するアミノ酸を意味する。
変異体において「配列番号:2のX位のアミノ酸」は、配列のアライメント等により適宜決定される。
また、マグネットは、少なくとも一方のタンパク質において、配列番号:2の55位に対応するアミノ酸が、側鎖に正電荷を有するアミノ酸で置換されていることが好ましく、所望により、他方のタンパク質において、配列番号:2の55位に対応するアミノ酸が、側鎖に負電荷を有するアミノ酸又は中性アミノ酸で置換されていることがより好ましく、側鎖に負電荷を有するアミノ酸で置換されていることがさらに好ましい。
マグネットは、少なくとも一方のタンパク質において、配列番号:2の52位及び55位に対応するアミノ酸が、同一又は異なって、側鎖に正電荷を有するアミノ酸で置換されていることが好ましく、所望により、他方のタンパク質において、配列番号:2の52位及び55位に対応するアミノ酸が、同一又は異なって、側鎖に負電荷を有するアミノ酸又は中性アミノ酸で置換されていることがより好ましい。マグネットの一方のタンパク質において、配列番号:2の52位及び55位に対応するアミノ酸が側鎖に負電荷を有するアミノ酸又は中性アミノ酸で置換されている場合、異なって置換されていることが好ましく、一方のアミノ酸が、側鎖に負電荷を有するアミノ酸で置換され、他方のアミノ酸が、中性アミノ酸で置換されていることがより好ましく、配列番号:2の52位に対応するアミノ酸が、側鎖に負電荷を有するアミノ酸で置換され、配列番号:2の55位に対応するアミノ酸が、中性アミノ酸で置換されていることがさらに好ましい。
配列番号:2の52位及び/又は55位に対応するアミノ酸が、側鎖に負電荷を有するアミノ酸で置換されている場合には、同一又は異なって、アスパラギン酸又はグルタミン酸で置換されていることが好ましく、アスパラギン酸で置換されれていることがより好ましい。
配列番号:2の52位及び/又は55位に対応するアミノ酸が、中性アミノ酸で置換されている場合には、グリシンで置換されれていることがより好ましい。
配列番号:2の52位に対応するアミノ酸が、アスパラギン酸又はグルタミン酸で置換されていることが好ましく、アスパラギン酸で置換されれていることがより好ましく、同時に、配列番号:2の55位に対応するアミノ酸が、中性アミノ酸で置換されていることが好ましく、グリシンで置換されれていることがより好ましい。
具体的には、マグネットの一方のタンパク質において、52位のIle及び55位のMetが、側鎖に正電荷を有するアミノ酸で置換された配列を有し、マグネットの他方のタンパク質において、52位のIle及び55位のMetが、側鎖に負電荷を有するアミノ酸及び/又は中性アミノ酸で置換された配列を有するものが挙げられる。
マグネットとしては、特開2015-165776号公報に開示される、Vividタンパク質を基に本発明者らが開発した2つのタンパク質を用いることができる。
52位のIle及び55位のMetが、側鎖に正電荷を有するアミノ酸で置換された配列を有するマグネットの一方のタンパク質としては、pMag、pMagHigh1、pMagFast1及びpMagFast2等が挙げられ、52位のIle及び55位のMetが、側鎖に負電荷を有するアミノ酸及び/又は中性アミノ酸で置換された配列を有するマグネットの他方のタンパク質としては、nMag、nMagHigh1、nMagFast1及びnMagFast2等が挙げられる。
マグネットは、pMag、pMagHigh1、pMagFast1及びpMagFast2から選ばれる1種のタンパク質と、nMag、nMagHigh1、nMagFast1及びnMagFast2から選ばれる1種のタンパク質の組合せであることが好ましい。
本明細書においては、pMagは、I52R及びM55Rの変異を有する、配列番号:15のアミノ酸配列を有するポリペプチドであり、pMagHigh1は、pMagのアミノ酸配列において、さらにM135I及びM165Iの変異を有する、配列番号:16のアミノ酸配列を有するポリペプチドである。pMagFast1は、pMagのアミノ酸配列において、さらにI85Vの変異を有する配列番号:17のアミノ酸配列を有するポリペプチドである。pMagFast2は、pMagのアミノ酸配列において、さらにI74VとI85Vの変異を有する、配列番号:18のアミノ酸配列を有するポリペプチドである。
本明細書においては、nMagは、I52D及びM55Gの変異を有する、配列番号:19のアミノ酸配列を有するポリペプチドであり、nMagHigh1は、nMagのアミノ酸配列において、さらにM135I及びM165Iの変異を有する、配列番号:20のアミノ酸配列を有するポリペプチドであ
る。nMagFast1は、nMagのアミノ酸配列において、さらにI85Vの変異を有する配列番号:21のアミノ酸配列を有するポリペプチドである。nMagFast2は、nMagのアミノ酸配列において、さらにI74VとI85Vの変異を有する、配列番号:22のアミノ酸配列を有するポリペプチドである。
本発明においては、リンカーを介していてもよいが、マグネットのそれぞれのポリペプチドは、Creタンパク質のN末端側フラグメントのC末端のアミノ酸と、C末端フラグメントのN末端のアミノ酸と連結している。
すなわち、本発明に係る2つのポリペプチドのセットは、配列番号:1のアミノ酸配列のCreタンパク質のN末端側フラグメントとC末端側フラグメントが、それぞれ、光依存的に二量体を形成する2つのタンパク質に結合しているが、ここで、「結合」の意味としては、配列番号:1のアミノ酸配列のCreタンパク質のN末端側フラグメントとC末端側フラグメントが、それぞれ、光依存的に二量体を形成する2つのタンパク質に直接結合しているか、リンカーを介して結合していることを意味する。
言い換えれば、本発明における2つのポリペプチドのセットにおいては、配列番号:1のアミノ酸配列のCreタンパク質のN末端側フラグメントのC末端のアミノ酸より、リンカーを介して、あるいは、介さずに、光依存的に二量体を形成する2つのタンパク質である一方のポリペプチドが配置され、配列番号:1のアミノ酸配列のCreタンパク質のC末端側フラグメントのN末端のアミノ酸より、リンカーを介して、あるいは、介さずに、光依存的に二量体を形成する2つのタンパク質である他方のポリペプチドが配置されている。
本発明に係る2つのポリペプチドのセットにおいては、Creタンパク質のN末端側フラグメントがそのC末端のアミノ酸でnMag、nMagHigh1、nMagFast1又はnMagFast2と、リンカーを介して、Creタンパク質のC末端側フラグメントがそのN末端のアミノ酸でpMag、pMagHigh1、pMagFast1又はpMagFast2と、リンカーを介して結合していることがより好ましい。
本発明において、Creタンパク質のN末端側フラグメント又はC末端側フラグメントと、光依存的に二量体を形成する2つのタンパク質との結合におけるリンカーだけではなく、本発明に係る2つのポリペプチドのセットのそれぞれのポリペプチドを構成するドメインとドメインとが結合している場合、ドメインとドメインとの間に両者の結合を介在させるリンカーが存在していてもよい。
本発明において用いられるリンカーとしては、公知のリンカーであれば特に限定することなく用いることができるが、グリシン及びセリンをその配列中に多く含むアミノ酸配列を有するリンカーが用いられる。グリシンを主成分としているリンカーが一般に用いられるが、親水性アミノ酸であるセリンやトレオニンを含むリンカーであることが好適である。
本発明において、ドメインとドメインをリンカーを介して結合させる場合、その遺伝子工学的手法として、制限酵素認識サイトを利用して結合させることができる。制限酵素認識サイトの多くは、6つの塩基配列からなるため、制限酵素認識サイトを利用してドメインとドメインを結合させる場合、2つのアミノ酸がドメインとドメインの間に導入されることになる。
かかる制限酵素認識サイトを、本発明においては、リンカーとして用いることもできる。
本発明において、リンカーは、特に限定されるものではないが、2~16のアミノ酸で構成されていてもよい。
本発明において、ドメインとは、本発明に係る2つのポリペプチドのセットを構成するCreタンパク質のN末端側フラグメント及びC末端側フラグメントと、光依存的に二量体を形成する2つのタンパク質とが挙げられ、その他のドメインとしては、例えば、核局在化シグナル配列及び2Aペプチド配列等が挙げられる。
本発明に係る2つのポリペプチドのセットは、核局在化シグナル配列を有していてもよい。
核局在化シグナル配列が存在する場合、Creタンパク質のN末端側フラグメントのC末端のアミノ酸に結合する光依存的に二量体を形成する2つのタンパク質である一方のポリペプチドのC末端のアミノ酸に、リンカーを介して、あるいは、介さずに結合していてもよく、Creタンパク質のC末端側フラグメントのN末端のアミノ酸に結合する光依存的に二量体を形成する2つのタンパク質である他方のポリペプチドのN末端のアミノ酸に、リンカーを介して、あるいは、介さずに結合していてもよい。
核局在化シグナル配列は、本発明に係る2つのポリペプチドのセットの一方のポリペプチドに存在していてもよく、2つのポリペプチドに存在していてもよい。
核局在化シグナル配列としては、特に限定されるものではなく、VPKKKRKV(配列番号:23)のアミノ酸配列や、KRTADGSEFESPKKKRKVEAS(配列番号:24)のアミノ酸配列が例示できる。
本発明に係る2つのポリペプチドのセットは、好適には、自己触媒的に切断され、そのN末アミノ酸及びC末端のアミノ酸を介してそれぞれ結合する前後のタンパク質の量の比を1:1にすることができる2Aペプチドと呼ばれるポリペプチドを用いて発現される。
したがって、本発明に係る2つのポリペプチドのセットは、自己触媒的に切断された2Aペプチドに由来するアミノ酸配列(2Aペプチド配列)をそれぞれ有していることが好ましい。本発明に係る2つのポリペプチドのセットが2Aペプチド配列を有するとは、cotranslationalに切断される2Aペプチドの部分配列を、2つのポリペプチドのセットを構成するそれぞれのポリペプチドが有していることを意味する。
2Aペプチドとしては、特に限定されるものではないが、Kim, J. H., et al., PLoS One. 6(4), e18556 (2011)に記載されるペプチドが挙げられ、例えば、P2Aペプチド、T2Aペプチド、E2Aペプチド、及びF2Aぺプチドなどのポリペプチドが知られている。
本発明に係る2つのポリペプチドのセットは、図3bに示すように、ドメインをそれぞれ配置したアミノ酸配列として発現され、P2Aペプチドが自己触媒的に切断されることによって得られる2つのポリペプチドのセットであることが好ましい。P2Aペプチド配列を例示して説明すると、P2Aペプチド配列であるGSGATNFSLLKQAGDVEENPGP(配列番号:6)うちC末端のGP配列の間でcotranslationalに切断され、P2Aペプチド配列のN末端のアミノ酸と結合するポリペプチドに対して、「GSGATNFSLLKQAGDVEENPG」が残存し、P2Aペプチド配列のC末端のアミノ酸と結合するポリペプチドに対して、「P」が残存する。
本明細書においてドメインとして認識される「Creタンパク質のN末端側フラグメント」、「Creタンパク質のC末端側フラグメント」「光依存的に二量体を形成するタンパク質」、「核局在化シグナル配列」及び「2Aペプチド配列」は、記載する順にポリペプチド中に配列されていればよく、各ドメイン間には、リンカーとなるアミノ酸配列が存在していてもよい。
本発明に係る2つのポリペプチドのセットにおいては、N末端からC末端へとドメインが配置されているが、各ドメイン間において、詳しくは、Creタンパク質のN末端側フラグメントと光依存的に二量体を形成するタンパク質との結合、光依存的に二量体を形成するタンパク質と核局在化シグナル配列との結合、核局在化シグナル配列と2Aペプチド配列の一部との結合、2Aペプチド配列の一部と核局在化シグナル配列との結合、核局在化シグナル配列と光依存的に二量体を形成するタンパク質との結合、及び光依存的に二量体を形成するタンパク質とCreタンパク質のC末端側フラグメントとの結合が、リンカーを介した結合であってもよい。
本発明に係る2つのポリペプチドのセットは、配列番号:25のアミノ酸配列を有するポリぺプチド及び配列番号:26のアミノ酸配列を有するポリペプチドのセットであるか、配列番号:27のアミノ酸配列を有するポリぺプチド及び配列番号:28のアミノ酸配列を有するポリペプチドのセットであることが好適である。これらアミノ酸配列を図14及び15に示す。なお、各アミノ酸配列において、下線を付した「MA」配列は、開始コドンとKozak配列に由来する配列を意味し、その他の下線を付したアミノ酸配列は、ドメイン間のリンカーのアミノ酸配列であることを意味する。
本発明は、2つのポリペプチドのセットを構成するポリペプチドをコードする核酸も提供する。
本明細書において用語「核酸」は、特に記載されていない限り、DNA、RNA、DNA/RNAのキメラ、及び、locked nucleic acid(LNA)やペプチド核酸(PNA)などの人工核酸を含む。
また、本発明に係る核酸は、核局在化シグナル配列及び2Aぺプチド配列をあわせてコードする核酸であってもよく、ドメイン間にリンカーが存在するポリペプチドをコードする核酸であってもよい。
発現ベクターには、DNA複製開始点(ori)、選択マーカー(抗生物質抵抗性、栄養要求性等)、エンハンサー、スプライシングシグナル、ポリA付加シグナル、タグ(FLAG、HA、GST、GFPなど)をコードする核酸等を組み込んでもよい。
形質転換体を常法に従って培養することにより、目的とするポリペプチドが発現する。
粗抽出液又は培養上清からの精製も公知の方法又はそれに準ずる方法(例えば、塩析、透析法、限外ろ過法、ゲルろ過法、SDS-PAGE法、イオン交換クロマトグラフィー、アフィニティークロマトグラフィー、逆相高速液体クロマトグラフィー等)で行うことができる。
本発明においては、Creタンパク質がN末端側フラグメントとC末端側フラグメントに分割されている以外は、Cre-loxPシステムと同様にして遺伝子組換えを行うことができる。
本発明に係るCre-loxPシステムは、本発明に係る核酸と、loxP配列及びloxP変異体配列である核酸とを組み合わせたシステムであってもよく、本発明に係る核酸としては、本発明に係る2つのポリペプチドのセットをコードする核酸であることが好ましい。本発明に係るCre-loxPシステムは、本発明に係る発現ベクターと、loxP配列又はloxP変異体配列である核酸とを含有する、Cre-loxPシステムであってもよい。
また、本発明に係る動物は、本発明に係る核酸を有していてもよく、本発明に係る核酸を発現ベクターとして有していてもよく、本発明に係るCre-loxPシステムを有していてもよい。
本発明に係る2つのポリペプチドのセットは、従来の光依存的に制御されたスプリットCreよりも予想外に優れた遺伝子組換え活性を有するため、本発明に係る2つのポリペプチドのセットを体内に組み込んだ動物に対して体外から光照射することでも、効率よく遺伝子組換えをコントロールすることができる。
すなわち、本発明に係る2つのポリペプチドのセットによれば、光照射をオン/オフすることでCre/loxPシステムによる遺伝子組換えをin vitroだけでなくin vivoにおいても正確に制御することができる。
本発明に係るCre/loxPシステムを組み込んだ動物としては、哺乳動物であれば特に限定されるものではないが、実験動物としてのげっ歯類であることが好ましい。
本発明に係るCre/loxPシステムを組み込んだ動物により、特に限定されるものではないが、例えば,光刺激で遺伝子の働きをノックアウトしたり,逆に変異型の遺伝子を光刺激で正常に戻してその影響を評価できるようにしたり,あるいは蛍光タンパク質のようなレポーター遺伝子を光刺激で働かせて生体組織やそれを構成する細胞を標識することができる。
Creの分割位置(Step1)、スプリットCreのN末端側フラグメント(CreN)とC末端側フラグメント(CreC)に対するマグネット(pMagおよびnMag)の結合位置(Step2)、リンカーの長さ(Step3)、核局在化シグナル配列(NLS)の結合位置(Step4)、P2Aペプチド配列の結合(Step5)などを検討して、PA-Creとして様々なコンストラクトをルシフェラーゼアッセイで評価した(図1,2)。この目的のために、COS-7細胞を1.5× 104細胞/wellの細胞密度で96-wellマイクロプレート(Corning, Corning, NY, USA)に播種し、37 °C 、5% CO2の条件下で、24 h培養を行った。PA-Creの様々なコンストラクトをそれぞれコードするcDNAとルシフェラーゼレポーターのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。トランスフェクションに用いたDNAの総量は、0.05 μg/wellとした。このトランスフェクションにはX-tremeGENE 9 reagentを用いた。トランスフェクションの24 h後から、細胞に対して24 h、青色光(1.0 W m-2)での照射を施した。ルシフェラーゼアッセイの直前に、0.2 mM D-luciferin potassium salt(Wako,Japan)を含んだHanks’ balanced salt solution(HBSS, Grand Island Biological, Grand Island, NY, USA)で培地を置換した。生物発光は、Centro XS3 LB 960 プレートリーダー(BertholdTechnologies, Bad Wildbad, Germany)を用いて10 秒間、室温での測定を行なった(図2)。
PA-Creのコンストラクトを作製するために、スプリットCreのN末端側フラグメント(CreN)とC末端側フラグメント(CreC)をコードするcDNAをCreの改良版(iCre)のcDNAから作製した。なお、iCreのcDNAは哺乳類細胞での発現を高めるためにコドンを最適化するようInvitrogen(Carlsbad, CA, UAS)で合成を行なった。具体的には、開始コドンとKozak配列を実現するためにATGGCC(MAをコード、配列番号:31)をつけた。
CRY2-CIB1-splitCreのコンストラクトを作製するために、CRY2-CreN19-104のplasmid(#26888)とCIB1-CreC106-343のplasmid(#26889)をAddgeneより入手した。CreN19-59、CreC60-343、CreN19-104、CreC106-343、Magnetシステム(pMag及びnMag)、核局在化シグナル配列(NLS)をそれぞれコードするcDNAは一般的なPCRや制限酵素処理、ライゲーションにより作製した。
PA-Creとその関連のコンストラクトは、CMVプロモーターを有するpcDNA3.1ベクター(Invitrogen)に導入した。なお、ネガティブコントロールのPA-Cre-Y324Fのコンストラクトを作製するために、Multi Site-Directed Mutagenesis(MBL)を用いて、当該キットのマニュアルに従って、324番目のチロシン残基のフェニルアラニンへの置換を行なった。CRY2-CIB1-splitCreはpcDNA3.1ベクターに導入した。
Floxed-STOP-Flucのコンストラクトは、転写停止配列であるポリアデニル化配列(pA)を2つの同じ向きのloxP配列で挟んで作製した。一方、single-floxed-invertedFluc,double-floxed-inverted Fluc,lox66/lox71-floxed invertedFluc,JT15/JTZ17-floxed invertedFlucのコンストラクトは2つもしくは4つのloxP配列やloxP変異体の配列を逆向きにしてホタルルシフェラーゼ(Fluc)のcDNAを挟んで作製した。なお、Floxed-STOP-Flucのコンストラクトの作製には、Addgeneから入手したplasmid(#22797)を利用した。FlucのcDNAはpGL4.31ベクター(Promega)から得た。Floxed-STOP-Fluc,single-floxed-inverted Fluc,double-floxed-invertedFluc,lox66/lox71-floxed invertedFluc,JT15/JTZ17-floxed invertedFlucのそれぞれのコンストラクトはpcDNA3.1ベクターに導入した。なお、蛍光タンパク質のmCherryを用いたレポーターは、上述のFlucを用いたレポーター(floxed-STOP-Fluc,single-floxed-inverted Fluc,double-floxed-inverted Fluc,lox66/lox71-floxed inverted Fluc,JT15/JTZ17-floxed inverted Fluc)のFlucのcDNAをmCherryのcDNAで置き換えて作製した。
COS-7細胞、HEK-293細胞、NIH/3T3細胞、HeLa細胞(いずれもATCC)は10% FBS(GIBCO, Carlsbad, CA, USA)、100 unit/mLpenicillin、100 μg/mL streptomycin(GIBCO)を添加したDulbecco’s Modified Eagle Medium(DMEM,Sigma Aldrich)を用いて37 ℃、5% CO2の条件下で培養した。CHO-K1細胞 (ATCC)は10%FBS、100 unit/mL penicillin、100μg/mL streptomycinを添加したHam’s F-12 medium(GIBCO)を用いて37 ℃、5% CO2の条件下で培養した。
PA-Creを発現する細胞やマウスに対する光照射のために、LED光源(470 ± 20 nm;CCS, Kyoto, Japan)を用いた(図6~10,12,13)。室内光に対するPA-Creの応答性を調べるために、白色蛍光ランプを用いた(図13)。
ルシフェラーゼアッセイを行うために、COS-7細胞を1.5 × 104細胞/wellの細胞密度で96-well マイクロプレート(Corning, Corning, NY, USA)に播種し、37 °C 、5% CO2の条件下で、24 h培養を行った(図6, 9,10)。PA-Cre(又はCRY2-CIB1-split Cre)をコードするcDNAとルシフェラーゼレポーターのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。トランスフェクションに用いたDNAの総量は、0.05 μg/wellである。なお、このトランスフェクションにはX-tremeGENE 9 reagent を用いた。トランスフェクションの24 h後から、細胞に対して24 h、青色光(1.0 W m-2)での照射を施した。ルシフェラーゼアッセイの直前に、0.2 mM D-luciferin potassium salt(Wako,Japan)を含んだHanks’ balanced salt solution(HBSS, Grand Island Biological, Grand Island, NY, USA)で培地を置換した。生物発光は、Centro XS3 LB 960 プレートリーダー(BertholdTechnologies, Bad Wildbad, Germany)を用いて10 秒間、室温での測定を行なった。
コントロール実験のために、COS-7細胞を1.5 × 104細胞/wellの細胞密度・BR>ナ96-WELL マイクロプレートに播種し、37 °C 、5% CO2の条件下で、24 h培養を行った(図6)。PA-Cre-Y324F(iCre又はpcDNA3.1ベクター)をコードするcDNAとルシフェラーゼレポーターのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。なお、トランスフェクションには0.05 μg/wellのDNAを用い、X-tremeGENE9 reagentでトランスフェクショ
ンを行なった。トランスフェクションの24 h後から、細胞に対して24 h、青色光(1.0 W m-2)での照射を施し、生物発光を測定した。iCreの場合、トランスフェクションから48時間、細胞を暗所でインキュベーションして生物発光を測定した。
PA-Creの制御に対する光照射の強度依存性を調べるために、96-well マイクロプレートを40%、33%、25%、10%の透過率のNDフィルター(Fujifilm, Tokyo, Japan)でマスクした(図6)。COS-7細胞を1.5 × 104細胞/wellの細胞密度で96-well マイクロプレートに播種し、37 °C 、5% CO2の条件下で、24 h培養を行った。PA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。なお、トランスフェクションには0.05 μg/wellのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して24 h、青色光(1.0 W m-2)での照射を施し、生物発光を測定した。
様々な細胞種においてPA-Creの光応答性を調べるために、HEK-293細胞、NIH/3T3細胞、CHO-K1細胞を1.5× 104細胞/wellの細胞密度で96-wellマイクロプレートに播種した(図6)。HeLa細胞は3.0 × 104細胞/wellの細胞密度で96-well マイクロプレートに播種した。これらの細胞は37 °C、5% CO2の条件下で、24 h培養を行った。HEK-293細胞はPA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cでトランスフェクションした。トランスフェクションには0.05 μg/wellのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。NIH/3T3細胞、HeLa細胞、CHO-K1細胞は、PA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cでトランスフェクションした。トランスフェクションには0.3 μg/wellのDNAを用い、Lipofectamine3000 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して24 h、青色光(1.0 W m-2)での照射を施し、生物発光を測定した。
蛍光イメージングに基づいて評価を行うために、COS-7細胞を3.0 × 104細胞/wellの細胞密度でLab-Tek 8-well chamberedcoverglasses(Thermo Scientific, Waltham, USA)に播種し、当該細胞を37 °C 、5% CO2の条件下で、24 h培養した(図6, 9)。PA-CreもしくはCRY2-CIB1-split CreをコードするcDNAとそれぞれのレポーターのplasmidを1:9の割合で37 °Cでトランスフェクションした。トランスフェクションには0.15 μg/wellのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して 24 h、青色光(1.0 W m-2)での照射を施した。蛍光イメージングの直前に培地をHBSSで置換した。蛍光イメージングは63 × oil objective を搭載したLSM 710 confocal laser-scanning microscope(CarlZeiss, Jena, Germany)で行った。mCherryの蛍光イメージングにはHeNe laser(543 nm)を用いた。
COS-7細胞を1.4 × 106細胞/dishの細胞密度で6 cm 培養ディッシュ(Thermo Scientific)に播種し、当該細胞を37 °C 、5% CO2の条件下で、24 h培養した(図7,8)。なお、培養ディッシュは、その底面に黒いビニールテープで様々なパターンの光マスク(STRIPE,STAR,SMILEY,MOUNTAIN,LETTERS)を作製して利用した。PA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cでトランスフェクションした。もしくは、PA-CreをコードするcDNAとFloxed-Fluc-hCL1-hPESTのplasmidを1:4の割合で37 °Cでトランスフェクションした。トランスフェクションには2.5 μg/dishのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して48 h、青色光(1.0 W m-2)での照射を施した。生物発光イメージングの前に、1 mM D-luciferinを含んだHBSSで培地を置換した。生物発光イメージングはEvolve 512 EMCCD camera(Photometrics)を搭載したLumazone bioluminescence imager(NipponRoper, Tokyo, Japan)を用いて5 min行った。データはslidebook 4.2 software(IntelligentInnovations Inc., Denver, CO)とImage-J softwareを用いて解析した。
ルシフェラーゼアッセイを行うために、COS-7細胞を1.5 × 104細胞/wellの細胞密度で96-well マイクロプレートに播種し、37 °C 、5% CO2の条件下で、24 h培養を行った(図9)。PA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。トランスフェクションには0.05 μg/wellのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して様々な時間(0,1.5,3,6,12,24,48 h)、青色光(1.0 W m-2)での照射を施した。そして、0.2 mM D-luciferinを含んだHBSSで培地を置換した。生物発光は、Centro XS3 LB 960 プレートリーダーを用いて10 秒間、室温での測定を行なった。コントロール実験のために、COS-7細胞にpcDNA3.1ベクター(Mock)とFloxed-STOP-Flucのplasmidを上述と同様の条件でトランスフェクションして生物発光計測を行った。
PA-CreによるDNA組換え反応を蛍光で評価するために、COS-7細胞を3.0 × 104細胞/wellの細胞密度で8-well chambered coverglassに播種し、37 °C 、5% CO2の条件下で、24 h培養を行った(図9)。PA-CreをコードするcDNAとFloxed-STOP-mCherryのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。トランスフェクションには0.15 μg/wellのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して様々な時間(0,1.5,3,6,12,24,48 h)、青色光(1.0 W m-2)での照射を施した。蛍光イメージングの直前に培地をHBSSで置換した。蛍光イメージングは63 × oil objective を搭載したLSM 710 confocal laser-scanning microscope(CarlZeiss, Jena, Germany)で行った。 mCherryの蛍光イメージングには HeNe laser(543 nm)を用いた。 コントロール実験のために、COS-7細胞にpcDNA3.1ベクター(Mock)とFloxed-STOP-mCherryのplasmidを上述と同様の条件でトランスフェクションして蛍光イメージングを行った。
COS-7細胞を2.0 × 105細胞/dishの細胞密度で35 mm culture dish(IWAKI, Tokyo, Japan)に播種し、37 °C 、5% CO2の条件下で、24 h培養を行った(図10)。PA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cで当該細胞にトランスフェクションした。トランスフェクションには1.0 μg/dishのDNAを用い、X-tremeGENE9 reagentでトランスフェクションを行なった。トランスフェクションの24 h後から、細胞に対して、次のように様々な時間パターンで青色光照射を施した:細胞に対して50 W m-2の青色光で30 s、光照射を施した後に、暗所に24 hインキュベーションし、生物発光計測を行った。また、細胞に対して50 W m-2の青色光で30 s、30 minのインターバルで3回、もしくは6回の光照射を施した後に、暗所でインキュベーションし、生物発光計測を行った。1-shotilluminationの実験については、30 sに加えて、15min、1 h、3 hの青色光照射(50 W m-2)も検討した。コントロール実験として、青色光(1.0W m-2)での24 h照射も行った。生物発光計測の前に、0.2 mM D-luciferinを含んだHBSSで培地を置換した。生物発光は、GloMax 96 microplate luminometer(Promega)を用いて1.0 秒間、室温での測定を行なった。
実験では5週齢のメスのBALB/cマウス(SankyoLabo Service Corporation, Inc., Tokyo, Japan)を用いた(図12,13)。除毛クリームを用いてマウスの腹部の毛を除き、 24 h、ケージの中で飼育した。そしてPA-CreをコードするcDNAとFloxed-STOP-Flucのplasmidを1:9の割合で37 °Cでマウスの尾静脈にインジェクションした。このハイドロダイナミックインジェクションにはTransIT-QR Hydrodynamic Delivery Solution(MirusBio LLC, Madison, WI, USA)を用い、マニュアルに従って行った。インジェクションしたDNAの量と溶液の体積はそれぞれ300 μg/mouse weight (g)、0.1 mL/mouse weight(g)である。ハイドロダイナミックインジェクションの後、マウスを8 h、暗所で飼育し、青色LED光源(470 ± 20 nm, 200 W m-2)で16 hもしくは30 s、ケージの中で光照射した。30 s光照射したマウスは、さらに16 h、暗所で飼育した。室内光での光照射については、ハイドロダイナミックインジェクションを施したマウスを白色蛍光(0.05 W m-2)の元で24 h、飼育した。マウスの生物発光イメージングは、ハイドロダイナミックインジェクションの24 h後に行った。生物発光イメージングの前に、100 mM D-luciferinをマウスの尾静脈にインジェクションした。D-luciferin溶液は10 μL/mouse weight (g)である。D-luciferinのインジェクションから3 min後、 Evolve 512 EMCCD camera(Photometrics)を搭載したLumazone bioluminescence imager(Nippon Roper)で5 min、生物発光イメージングを行なった。D-luciferin の尾静脈注射と生物発光イメージングの間、マウスにはisoflurane(AbbVie, Tokyo, Japan)で麻酔を行い、マウスの動きを抑制した。In vivoでのマウスの生物発光イメージングの後、ex vivoでの解析を行うために、当該マウスから肝臓を摘出した。肝臓は1 mM D-luciferinを含んだPBSの中に入れた。なお、実験には6-well culture plate(Thermo Scientific)を利用した。そして、anteriorとposteriorの摘出肝臓のイメージをLumazone bioluminescence imager(Nippon Roper)を用いて5 min、撮影した。データはslidebook 4.2 software(Intelligent Innovations Inc.)で解析した。

配列番号:7~12 loxPとloxP変異体(lox2722、lox66、lox71、JT15、及びJTZ17)の塩基配列を示す。
配列番号:13、14 DNA組換えによる生成物であるlox72及びJA15-JTZ17の塩基配列を示す。
配列番号:15~22 pMag、pMagHigh1、pMagFast1、pMagFast2、nMag、nMagHigh1、nMagFast1及びnMagFast2のアミノ酸配列を示す。
配列番号:23、24 例示される核局在化シグナル配列のアミノ酸配列を示す。
配列番号:25~28 本発明に係る2つのポリペプチドのセット(PA-Cre)を構成するポリペプチドのアミノ酸配列を示す。
配列番号:29、30 本発明に係る2つのポリペプチドのセット(PA-Cre)のベクターに挿入される塩基配列を示す。
配列番号:31 PA-Creの開始コドンとKozak配列であるMAアミノ酸配列をコードする塩基配列を示す。
Claims (19)
- 配列番号:1のアミノ酸配列のCreタンパク質のN末端側フラグメントとC末端側フラグメントが、それぞれ、光依存的に二量体を形成する2つのタンパク質に結合する、光依存的遺伝子組換えに用いられる2つのポリペプチドのセットであって、
Creタンパク質のN末端側フラグメントとC末端側フラグメントが、それぞれ、配列番号:1のアミノ酸配列の59位~68位の領域内に切断部位を有するフラグメントであるか、配列番号:1のアミノ酸配列の101位~111位の領域内に切断部位を有するフラグメントであり、
Creタンパク質のN末端側フラグメントのC末端のアミノ酸とCreタンパク質のC末端側フラグメントのN末端のアミノ酸で、それぞれ、光依存的に二量体を形成する2つのタンパク質に結合し、
光依存的に二量体を形成する2つのタンパク質が、それぞれ配列番号:2のアミノ酸配列からなるポリペプチド、又はその変異体であり、少なくとも一方のタンパク質において、N末端αヘリックスに位置する少なくとも1つのアミノ酸が置換されている、2つのポリペプチドのセット。 - Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~59位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の68位~343位の領域を少なくとも有するか、
Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~101位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の111位~343位の領域を少なくとも有する、請求項1に記載の2つのポリペプチドのセット。 - Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~59位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の60位~343位の領域を少なくとも有する、請求項1に記載の2つのポリペプチドのセット。
- Creタンパク質のN末端側フラグメントが、配列番号:1のアミノ酸配列の19位~104位の領域を少なくとも有し、Creタンパク質のC末端側フラグメントが、配列番号:1のアミノ酸配列の106位~343位の領域を少なくとも有する、請求項1に記載の2つのポリペプチドのセット。
- Creタンパク質のN末端側フラグメントとC末端側フラグメントにおいて、少なくとも1つのフラグメントの配列に1から数個のアミノ酸の付加、置換、又は欠失を含むか、少なくとも1つのフラグメントの配列が対応する配列番号:1のアミノ酸配列と80%以上の配列同一性を有するフラグメントである、請求項1~4のいずれか1項に記載の2つのポリペプチドのセット。
- 光依存的に二量体を形成する2つのタンパク質の一方のタンパク質において、少なくとも配列番号:2の52位及び/又は55位に対応するアミノ酸が、リシン、アルギニン又はヒスチジンで置換され、光依存的に二量体を形成する2つのタンパク質の他方のタンパク質において、少なくとも配列番号:2の52位及び/又は55位に対応するアミノ酸が、アスパラギン酸、グルタミン酸又はグリシンで置換されている、請求項1~5のいずれか1項に記載の2つのポリペプチドのセット。
- 光依存的に二量体を形成する2つのタンパク質が、pMag、pMagHigh1、pMagFast1又はpMagFast2と、nMag、nMagHigh1、nMagFast1又はnMagFast2と、の組合せである、請求項6に記載の2つのポリペプチドのセット。
- Creタンパク質のN末端側フラグメントとC末端側フラグメントが、ぞれぞれ、光依存的に二量体を形成する2つのタンパク質とリンカーを介して結合する、請求項1~7のいずれか1項に記載の2つのポリペプチドのセット。
- 核局在化シグナル配列が、Creタンパク質のN末端側フラグメントに結合する光依存的に二量体を形成するタンパク質のC末端のアミノ酸で、及び/又はCreタンパク質のC末端側フラグメントに結合する光依存的に二量体を形成するタンパク質のN末端のアミノ酸で、リンカーを介して、あるいは、リンカーを介さずに結合する、請求項1~8のいずれか1項に記載の2つのポリペプチドのセット。
- 2Aペプチド配列をさらに有する、請求項1~9のいずれか1項に記載の2つのポリペプチドのセット。
- 2つのポリペプチドのセットにおける1つのポリペプチドが、N末端側からC末端側へ、Creタンパク質のN末端側フラグメント-光依存的に二量体を形成するタンパク質-核局在化シグナル配列-2Aペプチド配列の一部の順にアミノ酸配列を有し、
もう1つのポリペプチドが、N末端側からC末端側へ、2Aペプチド配列の一部-核局在化シグナル配列-光依存的に二量体を形成するタンパク質-Creタンパク質のC末端側フラグメントの順にアミノ酸配列を有する、請求項1~10のいずれか1項に記載の2つのポリペプチドのセット。 - Creタンパク質のN末端側フラグメントと光依存的に二量体を形成するタンパク質との結合、光依存的に二量体を形成するタンパク質と核局在化シグナル配列との結合、核局在化シグナル配列と2Aペプチド配列の一部との結合、2Aペプチド配列の一部と核局在化シグナル配列との結合、核局在化シグナル配列と光依存的に二量体を形成するタンパク質との結合、及び光依存的に二量体を形成するタンパク質とCreタンパク質のC末端側フラグメントとの結合が、リンカーを介した結合である、請求項11に記載の2つのポリペプチドのセット。
- 請求項1から12のいずれか1項に記載の2つのポリペプチドのセットをコードする核酸。
- Creタンパク質のN末端側フラグメント-光依存的に二量体を形成するタンパク質-核局在化シグナル配列-2Aペプチド配列-核局在化シグナル配列-光依存的に二量体を形成するタンパク質-Creタンパク質のC末端側フラグメントの順にアミノ酸配列を有するポリぺプチドをコードする、請求項13に記載の核酸。
- 請求項13又は14に記載の核酸を含む発現ベクター。
- 請求項1~12のいずれか1項に記載の2つのポリペプチドのセット、請求項13若しくは14に記載の核酸、又は請求項15に記載の発現ベクター、及びloxP配列又はloxP変異体配列である核酸を含有する、Cre-loxPシステム。
- loxP変異体が、lox2722、lox66、lox71、JT15、及びJTZ17からなる群から選択される、請求項16に記載のCre-loxPシステム。
- 請求項1~12のいずれか1項に記載の2つのポリペプチドのセット、請求項13若しくは14に記載の核酸、請求項15に記載の発現ベクター、又は請求項16若しくは17に記載のCre-loxPシステムを有する動物。
- げっ歯類である、請求項18に記載の動物。
