Description

複合ポリペプチド単量体、細胞浸透機能を有する当該複合ポリペプチドの単量体の会合体、及び、当該会合体を有効成分とする皮下、皮内、経皮又は筋肉内投与用ノロウイルスコンポーネントワクチン
 本発明は、機能性ポリペプチドとこれを用いる皮下、皮内、経皮又は筋肉内投与用コンポーネントワクチンについての発明であり、さらに具体的には、免疫原と分子針との複合ポリペプチドを単量体とする、細胞浸透機能を有する当該複合ポリペプチド単量体の会合体、及び、当該会合体を有効成分(感染防御抗原)とする、皮下、皮内、経皮又は筋肉内投与用ノロウイルスコンポーネントワクチンに関する発明である。
 エドワード・ジェンナーの天然痘ワクチンの発明と、ルイ・パスツールによる理論的な裏付けがなされて以来、様々な感染症に対するワクチンが提供されている。
 ワクチンは、毒性を弱めたウイルス等の病原微生物を感染防御抗原として用いる「生ワクチン」、殺傷した病原微生物の全部又は一部を感染防御抗原として用いる「コンポーネントワクチン」、変性させた病原体の産生する毒素を感染防御抗原として用いる「トキソイド」、に大別されている。
 現状におけるワクチンの接種方法は、一部を除き、皮下注射又は筋肉注射である。
 一方、バクテリオファージの優れた細胞への遺伝子導入機能に着目して発明された「分子針」についての技術が提供されている。

 ワクチンに関しては、様々な疾病への応用が期待されている反面、問題点も指摘されている。
 ワクチンは、病原微生物等の感染標的組織の細胞における感染を、種々の感染防御抗原を用いることにより擬似的に再現しすることにより免疫反応のみを惹起させることを目的とするものである。皮下注射や筋肉内注射による血中へのワクチン投与は、標的組織の細胞への無作為な供給が生体機能によりなされることを期待して行われるものである。しかしながら、感染防御抗原の標的組織の細胞への導入を効率良く行うことが現状では困難である。そのため、本来免疫反応を引き起こすための必要量よりも多量の感染防御抗原を用いなければならない。
 本発明者らは、上記の課題の解決のために、本発明者の一人が発明を主導した、上記の「分子針」に着目した。
 すなわち、当該分子針に免疫原としてウイルスの構造蛋白を担持することで、当該分子針の優れた細胞導入機能により、皮下、皮内、経皮又は筋肉内投与による、当該免疫原が担持された分子針の標的組織の細胞内への高い効率の導入と、所望の免疫反応が惹起され、当該分子針を感染防御抗原として用いる可能性を想定して本発明を完成した。
 そして、その一例として、ノロウイルスのキャプシド蛋白質のPドメインを免疫原として用いたところ、下記の通り本発明の会合体のうち、六量体が感染防御抗原として優れたウイルスの免疫原性を示すことが明らかになった。ただし、生体反応は多様であり、他の免疫原を用いた場合に、本発明の会合体がどのような免疫現象を示すのかは、現時点では明らかではない。
 本発明が提供する主題は、(1)分子針に免疫原を担持させた「複合ポリペプチド」(以下、本発明の複合ポリペプチドともいう)、(2)当該複合ポリペプチドを単量体としてなる「会合体」(以下、本発明の会合体と総称する)、(3)当該会合体を有効成分(感染防御抗原)とする「皮下、皮内、経皮又は筋肉内投与用ノロウイルスコンポーネントワクチン」(以下、本発明のノロウイルスワクチンともいう)、(4)当該会合体の生産方法(以下、本発明の生産方法ともいう)、に大別される。
 「コンポーネントワクチン」は、免疫感染抗原を必要とする成分ワクチンであり、これから、弱毒生菌ワクチン、トキソイドは除外され、本発明のノロウイルスワクチンの免疫感染抗原は、感染防御抗原としての本発明の会合体(六量体)である。
(1)本発明の複合ポリペプチド
 本発明の複合ポリペプチドは、下記式(1)のアミノ酸配列の複合ポリペプチドである。なお、下記式(1)、(2)における所定のアミノ酸配列同士を結ぶ「-」は、W、L1、Xn、Y等の概念的に纏まったアミノ酸配列同士の区別を明確にするための、単純な分子結合(実質的にはペプチド結合)の表示である。

で表される、複合ポリペプチド。
 上記Xnにおけるアミノ酸配列Xの繰り返し数であるnは、1であることが好適であるが、2又は3であってもよい。
 上記式(1)において、Xn、Y1、又は、Y2で示されるアミノ酸配列のうち、1個以上のアミノ酸が欠失、置換若しくは付加された改変アミノ酸配列が上記式(1)に含まれる。「欠失」とは、上記式(1)において定義されている各配列番号のアミノ酸配列におけるいずれかのアミノ酸残基が欠失しており、当該欠失したアミノ酸残基のN末端側(前)とC末端側(後)のアミノ酸残基がペプチド結合で結ばれた状態であり(N末端アミノ酸残基とC末端アミノ酸残基の欠失の場合は、当該アミノ酸残基が単に欠失した状態である)、当該欠失残基の個数が「アミノ酸欠失の個数」として数えられる。「置換」とは、上記式(1)において定義されている各配列番号のアミノ酸配列におけるいずれかのアミノ酸残基が「他のアミノ酸残基」に入れ替わっており、当該入れ替わったアミノ酸残基が、N末端側(前)とC末端側(後)の各アミノ酸残基とペプチド結合で結ばれた状態であり(N末端アミノ酸残基の置換の場合はC末端側のアミノ酸残基とのペプチド結合のみであり、C末端アミノ酸残基の置換の場合はN末端側のアミノ酸残基とのペプチド結合のみである)、当該置換残基の個数が「アミノ酸置換の個数」として数えられる。「付加」とは、上記式(1)において定義されている各配列番号のアミノ酸配列における、いずれか1箇所以上のペプチド結合の位置に、各々1個以上の新たなアミノ酸残基が挿入された状態で新たなペプチド結合が形成された状態である。これらのアミノ酸残基の改変の内容と個数は、上記式(1)に係わるアミノ酸配列と、改変に係わるアミノ酸配列のアライメントを、人力又はアミノ酸配列の解析が可能なソフトウエアを用いてコンピュータ上で行うことにより、明らかにすることができる。
 また、上記式(1)で定義されたリンカー配列L1又はL2、あるいは、修飾用のアミノ酸配列Y3は、上記定義されたアミノ酸残基数の範囲内において、必要に応じて任意の配列を選択することができる。
 そして、当該改変アミノ酸配列の改変複合ポリペプチドの六量体(下記)が、上記式(1)の複合ポリペプチドの六量体と、実質的に同等の免疫賦活活性を有することが好ましい。「実質的に同等」とは、「中和試験」等の免疫賦活活性の確認について確立している手法を用いた場合に、アミノ酸配列の非改変複合ポリペプチドとの免疫賦活活性の有意差が、5%以内の有意水準において認められない程度の同等性である。
 上記式(1)の、Xn、Y1、又は、Y2で示されるアミノ酸配列のうち、1個以上のアミノ酸が欠失、置換若しくは付加された改変アミノ酸配列における、各々のアミノ酸配列におけるアミノ酸改変の数は、Xnが8n個以内、好ましくは4n個以内、さらに好ましくは2n個以内;Y1が30個以内、好ましくは20個以内、さらに好ましくは10個以内;及びY2が15個以内、好ましくは10個以内、さらに好ましくは5個以内;であることが好適である。
(2)本発明の会合体
 本発明の会合体は、上記の本発明の複合ポリペプチドを単量体とする会合体であり、実質的には、三量体(以下、本発明の三量体ともいう)、又は、六量体(以下、本発明の六量体ともいう)である。本発明の会合体は、それ自身が細胞内に浸透する作用を発揮することができる。
 本発明の三量体は、同一又は異なる本発明の複合ポリペプチドを単量体蛋白質としてなる、三量体蛋白質であり、本発明の六量体は、当該三量体蛋白質2分子が会合してなる、六量体蛋白質である。
(3)本発明のノロウイルスワクチン
 本発明のノロウイルスワクチンは、上記の本発明の六量体、を有効成分(感染防御抗原)とするワクチンであり、ノロウイルスの感染に対する、皮下、皮内、経皮又は筋肉内投与用のコンポーネントワクチンである。免疫原であるウイルスの構造蛋白質の一部又は全部のアミノ酸配列であるWは、ノロウイルスのキャプシド蛋白質のPドメインの一部又は全部である。
(4)本発明の生産方法
 本発明の生産方法は、上述の本発明の複合ポリペプチドの3分子以上を、水性液体を介して接触させることにより、当該複合ポリペプチド同士を単量体として会合させて、三量体及び六量体の形成を行い、さらに当該三量体又は六量体を選択的に分離・採取する、複合ポリペプチド会合体の生産方法、である。
 本発明の複合ポリペプチド自体は、当該複合ポリペプチドをコードする核酸を、遺伝子工学的な手法により発現させる、又は、ペプチド合成技術により合成する、ことにより生産することができる。当該複合ポリペプチド同士を、水性液体中で接触させることにより、自発的に複合ポリペプチドの三量体、及び、六量体が構築され、三量体と六量体を含有する混合物が形成される。そして、さらに当該三量体又は六量体を選択的に分離・採取することにより、所望の本発明の複合ポリペプチド(三量体又は六量体)を生産することができる。
 「水性液体」に関しては、特に、本発明の複合ポリペプチドを遺伝子工学的な手法により生産する場合は、当該複合ポリペプチドを生物学的に発現させて、例えば、発現細胞の収集、破砕又は溶解等による当該複合ポリペプチドの露出、さらに公知の分離方法による当該複合ポリペプチドの分離の工程を行う過程において用いる水や各種緩衝液等の水性液体中において、自発的に会合が起こり、本発明の会合体である三量体と六量体を含む混合物を得ることができる。また、例えば、全化学合成や、パーツ毎の分割合成を行って化学修飾法により結合することにより製造した本発明の複合ポリペプチドを、水や各種緩衝液等の水性液体中に懸濁することで自発的に会合させて、本発明の会合体である三量体と六量体を含む混合物を得ることができる。
 上記の三量体と六量体を含む混合物から分離・採取する方法は、特に限定されず、公知の分子量による分別方法、例えば、ゲル電気泳動法、アフィニティークロマトグラフィー、分子排斥クロマトグラフィー等の分子篩、イオン交換クロマトグラフィー等が挙げられる。
 従って、本発明の生産方法の最も好適な態様の一つとして、「本発明の複合ポリペプチドをコードする核酸を導入した形質転換体を、液体培地で培養して当該複合ポリペプチドを発現させ、自発的な会合によって産生される、当該複合ポリペプチドを単量体とする三量体及び六量体を含む混合物を得て、当該混合物からさらに当該三量体又は六量体を選択的に分離・採取する、複合ポリペプチド会合体の生産方法。」が挙げられる。
 このようにして生産される本発明の会合体(三量体又は六量体)は、公知の方法により精製されて、ワクチンの有効成分として用いることができると考えられる。その一例として、上記の本発明のノロウイルスワクチンが提供された。
 本発明により、(1)皮下、皮内、経皮又は筋肉内投与により標的組織の細胞に免疫原であるウイルスの構造蛋白質を効率的に導入するコンポーネントワクチンの有効成分(感染防御抗原)の基本単位として用いられることが期待される、分子針に免疫原であるウイルスの構造蛋白質を担持させた複合ポリペプチド(単量体)、(2)当該有効成分(感染防御抗原)として用いられることが期待される、当該複合ポリペプチドが三量体又は六量体としてなる会合体とその生産方法が、提供される。そして、当該コンポーネントワクチンの一例として、(3)免疫原であるノロウイルスのキャプシド蛋白質のPドメインを担持した、当該複合ポリペプチドの六量体を、感染防御抗原として、皮下、皮内、経皮又は筋肉内投与により標的組織の細胞に効率的に導入して、所望の免疫反応を惹起することが可能な、ノロウイルスコンポーネントワクチンが提供される。

(1)本発明の複合ポリペプチド
 本発明の複合ポリペプチドを表すアミノ酸配列式である式(1):

 免疫原であるWは、上記のようにウイルスの構造蛋白質の一部又は全部のペプチド構成に基づくアミノ酸配列が挙げられる。ここで「基づく」とは、オリジナルのペプチド構成そのものだけではなく、アミノ酸配列が改変されて無毒化されたアミノ酸配列等も改変アミノ酸配列を含む概念である。
 これらのウイルスの構造蛋白質の一部又は全部は、抗原提示部分を含めた表層部分の一部又は全部を選択することが好適である。本発明のノロウイルスワクチンにおける免疫原として用いられる、キャプシド蛋白質のPドメインの一部又は全部がこれに該当する。
 第1のリンカー配列を示すL1は、免疫原Wと分子針部分Yの距離を適度に保って立体障害を抑制するために必要であり、このアミノ酸残基の個数は、免疫原であるWの分子量が大きくなれば多くなることが好適になる傾向があるが、上記の通りにアミノ酸残基数は0-100個であり、好適には10-40個である。
 Xは、配列番号1のアミノ酸からなる、アミノ酸配列XnにおけるXのn回(整数回)の繰り返し単位の配列である。繰り返しの形式は、直列の繰り返しであり、例えば、X2であれば、「X-X」(「-」は模式化したペプチド結合)である。また、繰り返し配列Xnにおいては、上述したアミノ酸配列の改変が許容される。ここでnは、上述のように1-3の整数であり、1が好適であるが、2又は3であってもよい。繰り返し配列Xnが2又は3である場合は、免疫原Wの大きさや特性に応じて、分子針Yの距離を安定して適切な距離に保つことが主要な目的となる。
 細胞導入領域Yは、分子針の基礎構造に該当し、バクテリオファージの尾(Tail)の針部分(細胞内導入部)に基づいたものであり、式(2):

 式(2)のY1のうち、N末端側32アミノ酸(32Leu)までが、バクテリオファージT4の三重らせんβシート構造の部分のアミノ酸配列である。なお、少なくともN末端のアミノ酸であるバリン(1Val)は、ロイシン(1Leu)であってもよい。残りのC末端側は、バクテリオファージのニードル蛋白質のC末端部分のアミノ酸配列である。このY1のC末端側に使用可能なアミノ酸配列としては、例えば、バクテリオファージT4のgp5のアミノ酸配列、バクテリオファージP2のgpVのアミノ酸配列、バクテリオファージMuのgp45のアミノ酸配列、バクテリオファージφ92のgp138のアミノ酸配列等が挙げられる。より具体的には、バクテリオファージT4のgp5のアミノ酸配列をC末端側に有するY1として配列番号2のアミノ酸配列が、バクテリオファージP2のgpVのアミノ酸配列をC末端側に有するY1として配列番号3のアミノ酸配列が、バクテリオファージMuのgp45のアミノ酸配列をC末端側に有するY1として配列番号4のアミノ酸配列が、バクテリオファージφ92のgp138のアミノ酸配列をC末端側に有するY1として配列番号5のアミノ酸配列が挙げられる。このY1のアミノ酸配列をコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
 式(2)中、Y2はバクテリオファージT4のfoldonと呼ばれる領域のアミノ酸配列、又は、バクテリオファージP2若しくはバクテリオファージMu若しくはバクテリオファージφ92のtipと呼ばれる領域のアミノ酸配列である。foldon又はtipは、バクテリオファージのフィブリチンと呼ばれる分子針構造の先端部を構成する領域である。式(2)においてY2が存在することは必須とはいえないが、このfoldon又はtipのアミノ酸配列を有することにより、細胞膜への分子針の取り込み効率を向上させることができるので、Y2を伴っていることが好適である。 バクテリオファージT4のfoldonのアミノ酸配列を配列番号6に示す。このアミノ酸配列をコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
 バクテリオファージP2のtipのアミノ酸配列を配列番号7に示す。このアミノ酸配列をコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。バクテリオファージMuのtipのアミノ酸配列を配列番号8に示す。このアミノ酸配列をコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。バクテリオファージφ92のtipのアミノ酸配列を配列番号9に示す。このアミノ酸配列をコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
 L2は、前記Y1とY2の間に介在する第2のリンカーである。リンカーL2のアミノ酸数は0-30個であり、好適には0-5個である。リンカーのアミノ酸数が0個とは、第2のリンカーL2は存在しないことを示すものである。
 Y3は、修飾用のアミノ酸配列であり、Yにおいて選択的に付加して用いることができる。当該修飾用のアミノ酸配列は、蛋白質精製や保護の目的等で付加するものであり、ヒスチジンタグ、GSTタグ、FLAGタグ等のタグ蛋白質等が挙げられる。Y3には、適宜リンカー配列を加えることが可能であり、このようなリンカー配列自体もY3のアミノ酸配列の構成要素となり得る。
 本発明の複合ポリペプチドは、公知の方法、具体的には、遺伝子工学的手法、又は、化学合成法により生産することができる。また、本発明の複合ポリペプチド全てを一緒に生産することも可能であり、パーツ毎に生産して当該パーツ同士を化学修飾法により事後的に結合させることにより生産することも可能である。リンカー(L1又はL2等)を介したポリペプチド同士の結合は、互いのポリペプチドにおけるリシン残基又はシステイン残基同士を、スクシンイミド基又はマレイミド基を有するリンカーにより結合させる等が可能である。
 遺伝子工学的手法では、生産対象の本発明の複合ポリペプチドの全部又は一部をコードする核酸を、例えば、大腸菌、酵母、昆虫細胞、動物細胞等の宿主細胞内で、あるいは大腸菌抽出液、ウサギ網状赤血球抽出液、小麦胚芽抽出液等の無細胞発現系で発現させることができる。これらの核酸の発現用ベクターとしては、各発現系に応じたものを用いることができ、例えば大腸菌発現用のpET、酵母発現用のpAUR、昆虫細胞発現用のpIEx-1、動物細胞発現用のpBApo-CMV、小麦胚芽抽出液発現用のpF3A等が挙げられる。
 化学合成法は、公知のペプチドの化学合成法を用いることが可能である。すなわち、常法として確立している液相ペプチド合成法、又は、固相ペプチド合成法を用いて、本発明の複合ポリペプチドの全部又は一部を製造することが可能である。そして、一般的に好適な化学合成法として認識されている固相ペプチド合成法も、Boc固相法又はFmoc固相法を用いることが可能であり、上述のように、必要に応じてライゲーション法を用いることも可能である。また、個々のアミノ酸は、公知の方法により製造可能であり、市販品を用いることも可能である。
(2)本発明の会合体
 図1に、本発明の複合ポリペプチドを基にした、本発明の会合体である三量体と六量体の構築過程を示す。図1において、10は、単量体としての本発明の複合ポリペプチドであり、20は、本発明の三量体であり、30は、本発明の六量体である。
 本発明の複合ポリペプチド10は、「式(2)のXnとY1に該当する基本部分131」と「式(2)のY2に該当するfoldon132」が結合した「式(1)のYに該当する分子針領域13」、及び、「式(1)のWに該当する免疫原11」が、「式(1)のL1に該当するリンカー12」を介して結合して構成されている。リンカー12以外のリンカーと、式(2)のY3に相当する修飾配列については、図示を省略した。本発明複合ポリペプチド10自体には、標的組織の細胞の細胞膜を通過する機能は実質的に認められない。
 三量体30は、上記の複合ポリペプチド10が、3個の単量体として自発的に会合してなる三量体である。三量体30は、上記の分子針領域13が3個纏まり互いのC末端同士で会合することによって、三量体平行βシート構造、及び、当該βシート構造自身によるらせん構造(三重らせんβシート構造)と呼ばれる針状構造が形成され、分子針13×3が形成されている。分子針13×3は、基本部分131×3と、foldon集合体132×3で構成されている。このように三量体化と自己組織化により標的組織の細胞の細胞膜を通過する機能を有する「分子針」が形成され、それぞれの単量体に由来するリンカー3本(121、122、123)と、これらのリンカーにそれぞれ結合している免疫原3個(111、112、113)が、この分子針13×3の外側に存在している。
 六量体60は、2単位の上記三量体30が、互いの分子針の基本部分((13×3)1と(13×3)2)のN末端において結合して構成される六量体であり、当該六量体60もまた、標的組織の細胞の細胞膜通過機能を有している。それぞれの三量体に由来するリンカー6本(121、122、123、及び、125、126:124は図示せず)と、これらのリンカーにそれぞれ結合している免疫原6個(111、112、113、及び、115、116:114は図示せず)が、2本の分子針(13×3)1と(13×3)2の外側に存在している。
 本発明の複合ポリペプチド10から、三量体30への三量体化、及び、当該三量体30から六量体60へのマクロ的な2量体化は、水性液体中において自発的に進行し、三量体又は六量体として安定して存在する。この三量体又は六量体の安定性は極めて強いものであり、例えば、温度100℃の水性液体環境下、さらにpH2-11の水性液体環境下、さらに有機溶媒を50-70容量%含む水性液体環境下であっても安定であり、その上、安全性にも優れている。水性液体から単離して乾燥させた状態でも、当該三量体又は六量体には優れた安定性と細胞膜透過性が認められる。
 上記のように、本発明の複合ポリペプチドから会合体への移行は、自発的に進行し、通常は大部分が最終形態である六量体化するが、一部は三量体として残る。
(3)本発明のノロウイルスワクチン
 本発明のノロウイルスワクチンは、その有効成分である本発明の会合体(六量体)の優れた細胞透過性と免疫原性により、標的組織の細胞に、皮下投与、皮内投与、経皮投与又は筋肉内投与を介して免疫原であるノロウイルスのキャプシド蛋白質のPドメインの一部又は全部を効率よく移行させ、免疫を行うことが可能であり、これにより、皮下投与、皮内投与、経皮投与又は筋肉内投与による、ノロウイルスのコンポーネントワクチンの有効性と安全性を向上させることが期待される。
 本発明のノロウイルスワクチンは、上述した本発明の会合体のうち、「Wとして、ノロウイルスのキャプシド蛋白質のPドメインを担持した六量体」を有効成分(感染防御抗原)として含有する、皮下投与、皮内投与、経皮投与又は筋肉内投与用の医薬品組成物(液剤)として提供される。本発明の会合体(六量体)による直接投与の場合も、当該会合体を緩衝液等において用時懸濁混合した液剤として、皮下投与、皮内投与、経皮投与又は筋肉内投与がなされるので、この形態も医薬品組成物に含まれる。
 本発明のノロウイルスワクチンは、有効成分(感染防御抗原)である本発明の会合体(六量体)、及び必要に応じてアジュバンドと共に、さらに必要に応じて適切な医薬製剤担体を配合して製剤組成物の形態に調製される。当該製剤担体としては、使用形態に応じて選択することが可能であり、充填剤、増量剤、結合剤、付湿剤、崩壊剤、界面活性剤等の賦形剤ないし希釈剤を使用することができる。組成物の形態は、上記のように基本的には液剤であるが、用時液体希釈用の乾燥剤、粉末剤、顆粒剤等とすることも可能である。
 本発明のノロウイルスワクチン中の本発明の会合体(六量体)の量は、適宜選択され一定ではないが、通常、本発明の会合体(六量体)を、投与時に1-10質量%含有する液剤として用いるのが好適である。適切な投与(接種)量は、1回成人1人当たり0.01μg-10mg程度であり、必要に応じて初回接種と追加接種を適宜組み合わせて、1回又は2回以上の投与(接種)を行うことが可能である。
 以下、本発明の実施例を開示する。
[実施例1] 本発明の会合体の調製
(1)本発明の複合ポリペプチドの調製
 本実施例では、遺伝子工学的手法を用いて、担持する免疫原を異にする4種類の本発明の複合ポリペプチド(免疫原は、下記(a)、(b)、(c)、(d))を調製した。
(a) MNV-PF: マウスノロウイルスの「VP-1領域」のN-terminal領域、Shell領域を除いた「P domain Full:PF領域」の一単位全体を免疫原として用いた本発明の複合ポリペプチド(以下、「MNV PF」とも記載する)であり、本実施例において用いた免疫原のアミノ酸配列は、配列番号10の通りである。これをコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
(b)マウスノロウイルスの「VP-1領域」のN-terminal領域、Shell領域を除いた「PF領域」(P domain Full:PF)の一単位を構成する外殻内側の「P1領域」と外殻外側の「P2領域」のうち、「P2領域」のみを免疫原として用いた本発明の複合ポリペプチド(以下、「MNV-P2」とも記載する)であり、本実施例において用いた当該感染防御抗原のアミノ酸配列は、配列番号11の通りである。これをコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
(c)ヒトノロウイルスの外殻抗原の一つである「P領域」(P domain Full:PF)の一単位全体を免疫原として用いた本発明の複合ポリペプチド(以下、「Saga PF」とも記載する)であり、本実施例において用いた当該免疫原のアミノ酸配列は、配列番号12の通りである。これをコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
(d)ヒトノロウイルスの外殻抗原の一つである「P領域」(P domain Full:PF)の一単位を構成する外殻内側の「P1領域」と外殻外側の「P2領域」のうち、「P2領域」のみを免疫原として用いた本発明の複合ポリペプチド(以下、「Saga P2」とも記載する)であり、本実施例において用いた当該免疫原のアミノ酸配列は、配列番号13の通りである。これをコードする核酸配列は、アミノ酸と核酸塩基の公知の関係に従って選択することができる。
<「PN-Saga PF」発現用プラスミドの作製>
 ここで「PN-Saga PF」とは、上述した式(1):

と、式(1)の細胞導入領域Yを表す式(2):

において、
 免疫原Wが、配列番号13のアミノ酸配列で表される「Saga PF」であり;第1のリンカーL1が、配列番号15のアミノ酸配列であり;繰り返し配列Xnの繰り返し単位が、配列番号1のアミノ酸配列であり、繰り返し数nは1であり;分子針の本体部分Y1のアミノ酸配列が、配列番号2のアミノ酸配列であり;第2のリンカーL2が「SVE」であり;フォールドンY2のアミノ酸配列が、配列番号6のアミノ酸配列であり;修飾配列Y3のアミノ酸配列が、配列番号16(VEHHHHHH)である、複合ポリペプチドである。
 なお下記のように、上記式(1)、(2)において、免疫原Wを、試験的にGFP(緑色蛍光蛋白質:配列番号17)に代えてなる複合ポリペプチド発現させるための発現プラスミド「プラスミドpKN1-1」を作製し、このプラスミドpKN1-1を基にして、所望の本発明の複合ポリペプチド発現プラスミドを作製した。
 すなわち「PN-Saga PF」発現用プラスミドは、以下のように作製した。
 まず、T4ファージのwac蛋白質の461から484残基目に対応する遺伝子をT4ファージゲノムよりPCRで増幅してpUC18にクローニングし、foldonをコードする遺伝子を得た。続いて、このプラスミドを制限酵素EcoRI及びSalIで切断し、EcoRIとXhoIで処理したプラスミドpET29b(Novagen)に挿入し、プラスミドpMTf1-3を得た。また、T4ファージのgp5の474から575残基目に対応する遺伝子をT4ファージゲノムよりPCRにより増幅してpUC18にクローニングし、gp5をコードする遺伝子を得た。続いて、このプラスミドを制限酵素EcoRI及びSalIで切断し、EcoRIとXhoIで処理した上述のプラスミドpMTf1-3に挿入し、プラスミドpKA176を得た。また、群馬大・高橋より提供されたGFP発現ベクターを制限酵素NdeI及びEcoRIで切断し、GFPをコードする遺伝子を得、制限酵素NdeI及びEcoRIで処理した上述のプラスミドpKA176に組み込み、プラスミドpKN1-1(GFP-gp5f発現用プラスミド)を得た。
 このGFP-gp5f発現用プラスミドであるpKN1-1をベースにして、「PN-Saga PF発現プラスミド」である「プラスミドpKN1-SagaPF(又は、単に「pKN1-SagaPF」)の作製を、プラスミドpKN1-1のGFP遺伝子挿入部分をカセットに見立て、GFPの遺伝子部分とSagaPFの遺伝子との入れ替えを行うことにより実行した。
 具体的には、まず、プラスミドpKN1-1のGFP遺伝子を除いた直鎖状プラスミド断片(受け側プラスミド)を作成するため、GFP遺伝子直上流にインバーテッドPCR用のアンチセンスプライマー(配列番号18)を合成し、さらに、GFP蛋白質(配列番号17)のC末端に付加されて存在するリンカー配列L1(配列番号15)の「MDELYKE」(配列番号14)部分の遺伝子配列に相補的なセンスプライマー(配列番号19)を合成した。次に、上記アンチセンスプライマーとセンスプライマーを用いて、プラスミドpKN1-1を鋳型として、インバーテッドPCRを行うことで、GFPを含まない直鎖状プラスミド断片を、PCR増幅産物として得た(第1のPCR増幅産物)。
 次に、Saga PF部分を増幅し、InFusionクローニングキットにて、Saga PFをカセットのように上記の要領で増幅した直鎖状プラスミド断片に組み込むため、以下のPCRを行った。
 すなわち、直鎖状ベクター5’側15塩基のセンス鎖核酸配列+SagaPF遺伝子のセンス鎖5’側20塩基の核酸配列となる様、35塩基のセンスプライマーを合成し(配列番号20)、さらに、「SagaPF遺伝子3’末端(上記のリンカー配列(配列番号14)を除いた配列番号10のアミノ酸配列C末7アミノ酸部分に相当)+当該リンカー配列MDELYKEのMDELYに相当する核酸配列」となる様、35塩基のアンチセンスプライマー(配列番号21)を合成した。次に、上記アンチセンスプライマーとセンスプライマーを用いて、SapaPF部分をpHuNoV-Saga1F(ヒトノロウイルスGII.4 Saga-1株全長クローン)を鋳型として増幅して、PCR増幅産物(第2のPCR増幅産物)を得た。このPCR産物のセンスプライマー側(配列番号22)が直鎖状ベクターPCR産物のアンチセンスプライマー側配列とオーバーラップしている。アンチセンスプライマー側配列(配列番号23)が直鎖状ベクターPCR産物のセンスプライマー側配列とオーターラップしている。以下、インサートカセット側には直鎖状ベクター配列と両末端がオーバーラップするようにプライマーがデザインされ、このオーバーラップ配列を用いて方向性を維持した状態でベクターにInFusionクローニングキット融合され、その結果として接合される。
 最後に、上記の第1のPCR増幅産物と第2のPCR増幅産物を、InFusionクローニングキットにより互いのオーバーラップ配列を融合することで接合し、pKN1-SagaPFを得た。
 上記のように作製した「PN-Saga PF」の発現用プラスミドである「pKN1-SagaPF」を大腸菌に導入して大量に「PN-Saga PF」を発現させて精製し、以下の実験に用いた。
<「PN-Saga PF」の超遠心分析>
 上述のようにして調製した「PN-Saga PF」の超遠心分析を行った。より具体的には、1.2μMの「PN-Saga PF」を0.1Mリン酸ナトリウム緩衝液(pH7.0)で透析し、Optimal XL-I analytical ultracentrifuge (Beckman)を用いて500,000rpm、20℃で超遠心分析を行った。
 図2に、「PN-Saga PF」の分析結果(サイズ排除カラム)を示す。その結果、複合ポリペプチド「PN-Saga PF」は、そのほとんどが自然に自己会合して、「PN-Saga PF」の三量体が2個会合した六量体の形態及び当該三量体の形態で存在していることが確認された。
[実施例2] 「PN-Saga P2」を用いた細胞導入試験
 「PN-Saga P2」発現用プラスミド(pKN1-SagaP2)の作製は、免疫原Wが、配列番号13のアミノ酸配列で表される「Saga P2」である他は、上記の「PN-Saga PF」の発現用プラスミドと同様に行い、「PN-Saga P2」の発現及び精製工程、並びに、超遠心分析も「PN-Saga PF」と同様に行った。GFP遺伝子との入れ替えのためのPCR用のセンスプライマーとして配列番号24、アンチセンスプライマーとして配列番号25を用いた。得られたPCR産物は、上記と同様に発現用ベクターのPCR産物にInFusionクローニングキットで組み込んだ。
 その結果、複合ポリペプチド「PN-Saga PF」も、そのほとんどが自然に自己会合して、「PN-Saga PF」の三量体が2個会合した六量体の形態及び当該三量体の形態で存在していることが確認された。
 そこで、次に、PN-Saga P2をHeLa細胞培養上清に添加し、PN-Saga P2がSagaP2領域を含めて細胞内に導入可能であるか調べた。P2の検出には、抗Saga-1 VLP抗体を用いた免疫蛍光抗体法を用いて実施した。
 その結果、HaLa細胞内に導入されたSaga P2蛋白質が緑色の蛍光シグナルとして多数確認され、PN-Saga P2が予測通りに機能していることが確認された(図示せず、ただし、必要に応じて写真の提出をする用意あり)。
[実施例3] 「PN-MNV」についての検討
 「PN-MNV PF」の発現プラスミド(pKN1-MNVPF)と「PN-MNV P2」発現用プラスミド(pKN1-MNVP2)の作製は、免疫原Wが、配列番号10のアミノ酸配列で表される「MNV PF」、又は、配列番号11のアミノ酸配列で表される「MNV P2」である他は、上記の「PN-Saga PF」の発現用プラスミドと同様に行い、「PN-MNV PF」又は「PN-MNV P2」の発現及び精製工程、並びに、超遠心分析も「PN-Saga PF」と同様に行った。PドメインのFull遺伝子とGFP遺伝子との入れ替えのためのPCR用のセンスプライマーとして配列番号26、アンチセンスプライマーとして配列番号27を用いた。また、PドメインのP2遺伝子とGFP遺伝子との入れ替えのためのPCR用のセンスプライマーとして配列番号28、アンチセンスプライマーとして配列番号29を用いた。
 その結果、「PN-MNV PF」も、「PN-MNV P2」も、上記の「PN-Saga PF」と同様に、それぞれ、そのほとんどが自然に自己会合して、三量体が2個会合した六量体の形態及び当該三量体の形態で存在していることが確認された。
 そこで、次に、標的細胞をHeLA細胞として、本発明の会合体として、上記の「PN-MNV P2」の会合体(ここでは、「PMNVP2会合体」という)の導入を行い、その結果を共焦点顕微鏡により確認した。
 まず、蛍光色素であるATTO520をPMNVP2会合体への担持を行った。7.5μMのPMNVP2会合体の0.1Mリン酸ナトリウム緩衝液(pH8.3)懸濁液と、300μM ATTOのDMSO溶液を、容量比2:1で、反応総量が450μLとなるように常温下で混合して、常温下で担持反応を行った。担持反応終了後、Centriconを用いて、0.1Mリン酸ナトリウム緩衝液(pH7.0)で3回洗浄した。
 HeLa細胞は、5.0×104 cells/100μL(DMEM)で、37℃・5%CO2下で前培養を行い、次いで、DMEM(phenol red(-))と0.1Mリン酸ナトリウム緩衝液(pH8.3)の容量比2:1の培養液を全量90μLとして、上記の前培養培地と交換して、さらに上記のATTO520を担持させたPMNVP2会合体総量が5μMとなるように本培養系への添加を行って、上記の培養条件と同様の条件で30分間培養を行い、PMNVP2会合体のHeLa細胞への導入を試みた。
 PNMVP2会合体のHeLa細胞への取り込みは、共焦点顕微鏡(共焦点レーザー顕微鏡A1(Nikon))を用いて、下記の条件で観察を行った。
(1)レーザー光の波長:
 励起:405nm laser/観察(a450nm/50emission filter)かつ、
 励起:488nm laser/観察(a525nm/50emission filter)
(2)レーザーパワー
 レーザーパワー:405nm:1.4、488nm:0.4、で観察した。
 その結果、蛍光色素が担持されたPNMVP2会合体が、暗い細胞核の周りの多数の細かく明るい部分として認められ、30分間という短時間のインキュベート時間にも拘わらず、HeLa細胞内に導入されていることが認められた(図示せず、ただし、必要に応じて写真の提出をする用意あり)。
[実施例4] マウスへの免疫試験
(1)材料
「PN-MNV PF」の会合体(PNVPF会合体)
 実施例3において用いた、「PN-MNV PF」の発現プラスミド(pKN1-MNVPF)で形質転換された大腸菌BL21(DE3)を、200ml LB培地(カナマイシン濃度30μg/ml)において6時間37℃で培養した。その後これらの培養液30mlを3L LB培地(カナマイシン30μg/ml)に添加し37℃で培養した。濁度OD600が0.8以上になったことを確認し、IPTG溶液を終濃度1mMになるように添加した。その後20℃において一晩インキュベートした。遠心機を用いて4℃,8000rpmで集菌を行い、液体窒素で瞬間冷凍し-80℃で保存した。
 氷上において、BufferA(0.1M Tris-HCl(PH8), 0.5M NaCl,1mM DTT,5mM imidazole)にcompleteEDTA free(Roche)を1錠溶かした溶液に、上記菌体22.4gを懸濁し、超音波破砕によって菌体を溶解させた。これを17500rpm,4℃で50分遠心し、上清を0.8μmフィルターを通した。その上清をNiカラム[HisTrapTMHPカラム(GE Healthcare)]を利用し、4℃においてNiアフィニティ精製をした。溶出の際はBufferAとBufferB(0.1M Tris-HCl(PH8),0.5M NaCl,1mM DTT,500mM imidazole)を用いて5-500mM imidazoleのリニアグラジエントで行った。サンプルは280nm波長で追跡し、画分を回収した。
 上記のNiアフィニティ精製で回収したサンプルを10mlほどに濃縮し0.2μmフィルターを通したのち、HiLoad26/60 superdex200カラム(GE Healthcare)を用いてゲルろ過精製を行った。溶出はBuffer C (20mM Tris-HCl,0.5M NaCl,1mM TCEP)で行った。そのチャートを図3に示す。
 図3の各フラクションの番号(横軸に沿ったカラムで区切られた各番号)ごとにNativeSDSPAGEで分子量を確認し(図示せず)、六量体とほぼ同じ分子量30万Da程度に位置するバンドが認められる画分として、最も明確なバンドが認められた36番の画分を選択して、PNVPF会合体(六量体)として用いた。三量体は、その半分程度の分子量が認められた38-40番の画分をさらに常法、例えば、イオンクロマトグラフィー等による精製を行うことにより容易に分離することができる。
 マウスノロウイルスのキャプシド蛋白質のPドメイン(MNVPF)は、その公知の遺伝子配列に基づいて、大腸菌を宿主として用いる組換え蛋白質として、常法に従い調製した。すなわち、マウスノロウイルスのPF領域のN末端側に発現ベクターpET6xHNのクローニングサイト上流に15塩基のオーバーラップをもつセンスプライマーを、C末端領域にpET6xHNのクローニングサイト下流に15塩基のオーバーラップをもつアンチセンスプライマーをInFusionクローニングキットのプロトコールに従って、合成した。これらのプライマーでPCRを行い、PCR産物を得た。PCR産物は、pET6xHNにInFusionクローニングキットによってクローニングされ、pET6xHN-MNVPFを得た。大腸菌BL21にpET6xHN-MNVPFを導入し、MNVPFの発現を定法に従って行った。大腸菌を破砕した後、グアニジンによるそう蛋白質の可溶化を実施し、全ての蛋白質を直鎖状にした。可溶化したそう蛋白質に含まれるMNVPFに付加された6個のHNタグを利用して、タロン樹脂カラムに、タロン樹脂のマニュアルに従って結合させた。その後MNVPFをPBS(-)に透析し、溶媒を完全にPBS(-)に交換した。これを精製MNVPF溶液として、濃度調整し、下記の様にマウスに免疫を行った。
 その他、PBS、26G注射針付1mlシリンジ(テルモ社製)、マウス(日本SLC社 C57BL/6J,6週齢、雌)を準備した。
(2)方法
 下記の2通りの感染防御抗原を用いた試験系を構築した。
 (a)MNVPF投与の系(3.12μg/200μl PBS)
 (b)六量体投与の系(13.2μg/200μl PBS)
 これらの希釈液を、それぞれマウスの背中5か所に26G注射針付シリンジを用いて40μlずつ計200μl皮下注射した。その3週間後にマウスの尻尾から採血を行った(1次免疫血清)。さらに再度、同量の上記希釈液をマウスの背中に皮下注射した。その2週間後にマウスの尻尾から採血を行った(2次免疫血清)。
<採取した血液からの血清サンプルの調製>
 上記の採血した血液を、各々卓上遠心機で15000rpm x1分間遠心を行い、上清を新しいエッペンチューブに移し、再度、卓上遠心機で15000rpm x1分間の遠心を行い、上清をさらに新しいエッペンチューブに移し、これをELISA用サンプルとして用いた。ELISAは以下のように施行した。

・ELISA plate (F96 MAXI SORP NUNC-IMMUNO PLATE,ThermoSCIENTIFIC)
・希釈用96well plate (96well Round Bottom Non-Treated,CORNING)
・Tween20
・BSA
・PBS
・PBST(0.1%tween20添加PBS)
・PBSB(1% BSA添加PBS)
・PBSTB(1% BSA、0.1%tween20添加PBS)
・Goat anti-mouse IgG HRP標識(SouthernBiotech)
・OPD基質(Sigma,10mg tablet of o-Phenylenediamine dihydrochloride)
・H22
・OPDバッファー(36g Na2HPO4・12H2O+9.6g citrate(クエン酸)+1L D2W,pH5.0))
・2N H2SO4
・プレートリーダー(BIORAD iMark)
(b)方法
 マウスノロウイルスVLPを、PBSで1μg/mlに調製し、これをELISA plateに50μl/wellで添加し、1晩4℃で静置した。このELISA plateをPBSTで4回洗浄し、最後にPBSTを完全に拭きとり、PBSBを80μl/wellで添加し、室温で2時間静置した。これとは別に、希釈用プレートを用いてマウス血清をPBSBで20倍希釈し、そこから3倍ずつの段階希釈を計8系統造った。上記の2時間静置後、ELISA plateからPBSBを捨て、希釈した血清をELISA plateに50μl/wellで添加し、室温で4時間静置し、ELISA plateから希釈血清を捨て、PBSTで4回洗浄し最後にPBSTを完全に拭いた。次に、PBSTBで5000倍に希釈したHRP標識抗マウスIgG抗体を50μl/wellで添加し、室温で2時間静置した。その後、ELISA plateからHRP標識抗体を捨て、PBSTで5回洗浄し最後にPBSTを完全に拭いた。そして、OPD基質(10mg)を20mlのOPDバッファーに溶解し、10μlのH22を添加し転倒混和し、ELISA plateに上記OPD基質溶解液を50μl/wellで添加し、室温で静置し発色を行い、その後2N H2SO4を50μl/wellで添加して発色を止め、プレートリーダーでOD490を測定し、陽性対照と比較して濃度計算を行った。
(3)結果
 上記の初回接種のELISAの結果を、図4に示す。図4の縦軸はIgGの抗体価(U/ml)である。図4に示すように、接種の結果、PMNVPF会合体(六量体)において抗体価が有意に上昇した個体が認められた。残りの3個体の内2個体においても、抗体価の上昇傾向を示していた。対照のMNVPFの抗体価は殆ど上昇していなかった。
 すなわち、MNVPFを免疫原として担持した本発明の会合体(六量体)が、感染防御抗原として、非常に優れていることが明らかになった。

Claims (10)

  1.  下記式(1)のアミノ酸配列:
     W-L1-Xn-Y   (1)
     [式中、Wは免疫原であるウイルスの構造蛋白質の一部又は全部のアミノ酸配列を示し、L1はアミノ酸数が0-100の第1のリンカー配列を示し、Xは配列番号1のアミノ酸配列を示し、Yは細胞導入領域のアミノ酸配列を示し、Xの繰り返し数であるnは1-3の整数である。]
    であって、
     当該細胞導入領域Yのアミノ酸配列は、下記式(2):
     Y1-L2-Y2-Y3   (2)
     [式中、Y1は配列番号2-5からなる群より選択されるいずれか1つのアミノ酸配列を示し、Y2は配列番号6-9からなる群より選択されるいずれか1つのアミノ酸配列を示し、L2はアミノ酸数が0-30の第2のリンカー配列を示し、Y3は修飾用のアミノ酸配列を示し、Y2又はY3は存在しない場合もある。]
    で表される、複合ポリペプチド。
  2.  リンカー配列であるL1は、配列番号14を含む、請求項1に記載の複合ポリペプチド。
  3.  上記式(1)の、Xn、Y1、又は、Y2で示されるアミノ酸配列のうち、1個以上のアミノ酸が欠失、置換若しくは付加された改変アミノ酸配列に係わる、請求項1又は2に記載の複合ポリペプチド。
  4.  上記式(1)の、Xn、Y1、又は、Y2で示されるアミノ酸配列のうち、1個以上のアミノ酸が欠失、置換若しくは付加された改変アミノ酸配列における、各々のアミノ酸配列におけるアミノ酸改変は、Xnが8n個以内、Y1が30個以内、及びY2が15個以内である、請求項3に記載の複合ポリペプチド。
  5.  請求項1-4のいずれか1項記載の、同一又は異なる複合ポリペプチドを単量体蛋白質としてなる、三量体蛋白質。
  6.  請求項1-4のいずれか1項記載の、同一又は異なる複合ポリペプチド3分子のXn及びY1において、互いのXn及びY1を組織化する平行βシート構造、及び、当該平行βシート構造自身のらせん構造が認められることを特徴とする、請求項5に記載の三量体蛋白質。
  7.  請求項5又は6に記載の三量体蛋白質2分子が会合してなる、六量体蛋白質。
  8.  請求項7に記載の六量体蛋白質を有効成分とする、皮下、皮内、経皮又は筋肉内投与用のコンポーネントワクチンであって、免疫原であるウイルスの構造蛋白質の一部又は全部のアミノ酸配列であるWは、ノロウイルスのキャプシド蛋白質のPドメインの一部又は全部である、コンポーネントワクチン。
  9.  請求項1-4のいずれか1項に記載の複合ポリペプチドを、水性液体を介して接触させることにより、当該複合ポリペプチド同士を会合させて、三量体及び六量体の形成を行い、当該三量体又は六量体を選択的に分離・採取する、複合ポリペプチド会合体の生産方法。
  10.  請求項1-4のいずれか1項に記載の複合ポリペプチドをコードする核酸を導入した形質転換体を、液体培地で培養して当該複合ポリペプチドを発現させ、自発的な会合によって産生される、当該複合ポリペプチドを単量体とする三量体及び六量体を含む会合体の混合物を得て、さらに当該混合物から当該三量体又は六量体を選択的に分離・採取する、請求項9に記載の複合ポリペプチド会合体の生産方法。
