Description

ナイーブ型多能性幹細胞培養用培地および多能性幹細胞の培養方法
 本発明は、ナイーブ型多能性幹細胞の未分化と多能性(多分化能)を維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖させるためのナイーブ型多能性幹細胞培養用培地を提供することを目的とする。また、本発明は、多能性幹細胞の未分化性と多能性(多分化能)を維持しつつ、かつナイーブ型を維持しながら多能性幹細胞を増殖する方法を提供することを目的とする。
 ヒトiPS細胞は、体細胞に3~4因子の遺伝子を導入することにより誘導でき、生体内の殆ど全ての細胞に分化する能力(多能性)を有する細胞である。この特性から、生体組織をiPS細胞から人工的に作製し、患者の体内に移植することにより、これまで治療が困難であった疾患を治療する再生医療への応用が期待されている。また、従来、入手できなかったヒト細胞を作製できる可能性があることから、疾患解明モデルや新規薬剤の効果、あるいは、安全性評価などのスクリーニングへの応用も期待されている。
 ヒトiPS細胞を含むヒト多能性幹細胞は、マウスあるいはヒトなどの胎児由来線維芽細胞や、あるいは胎盤由来細胞などをフィーダー細胞として用いたオンフィーダー培養、またはマトリジェルや、ビトロネクチン、ラミニン、ファイブロネクチンなどの細胞外マトリックスや、あるいは組換えタンパク質などを細胞外マトリックスでコーティングした表面を有するフラスコを用いたフィーダーレス培養により、継代維持することができる。
 しかしながら、これら手法は細胞をフラスコ表面に接着させながら2次元的に培養するものであるため、細胞の大量調製には非効率的である。したがって、ヒトiPS細胞を3次元的に浮遊培養することができれば、少ないコストで大量の細胞を培養することが可能になると期待される。
 接着細胞は、マイクロキャリアビーズなどを用いない限り、浮遊培養することはできない。ヒト多能性幹細胞は、浮遊させると、凝集塊を作製し、自発的に分化することが知られている。また、ヒトiPS細胞を浮遊培養する場合には、ヒトiPS細胞を単一細胞または数個~数10個の細胞群に分散する必要があるが、このとき、ヒトiPS細胞はアポトーシスを起こすという問題を有している。この問題に対しては、ROCK(Rho-associated coiled-coil forming kinase)阻害剤であるY-27632を培地に添加することにより、細胞間接着が促進され、単一細胞または数個~数10個の細胞群の状態にしたときのヒトiPS細胞のアポトーシスを抑制することが知られている(非特許文献1)。
 近年、多能性幹細胞は「ナイーブ型」と「プライム型」に分類されている。マウスにおいては、着床前のエピブラストの性質をもつナイーブ型と、着床後のエピブラストの性質をもつプライム型を培養する条件が知られている。「ナイーブ型」の方がより未分化な状態であり、キメラ個体を作り出す能力が高く、特に精子や卵子などの生殖細胞に分化する能力があるため、プライム型よりも質的に優れていることが挙げられる。また、重層化したドーム状コロニーを形成し、均一で安定に増殖する。また、凝集塊として無血清培養下に浮遊培養できるとの報告もある(非特許文献2)。一方、ヒトの場合は、着床前受精卵内部細胞塊から作製した場合は「プライム型」になることが知られている。また、ヒトiPS細胞もプライム型である。
 既報のヒトiPS細胞の浮遊培養系には、Y-27632が10μM濃度で添加されるのが一般的であるが(非特許文献3~8)、この場合、ヒトiPS細胞は細胞の形状やマーカー遺伝子の発現パターンから「プライム型」であると考えられる。
 マウスES細胞においてはMEK阻害剤、GSK阻害剤およびLIFを含む2i/L培地中で培養することにより、ナイーブ型に移行することが報告されている。一方、2i/L培地中でマウスのエピブラスト幹細胞やヒトの多能性幹細胞を培養すると細胞は分化することが知られている。
 ヒトiPS細胞を「ナイーブ型」に誘導する方法としては、MEK阻害剤(PD0325901)、GSK阻害剤(CHIR99021、IM12、またはBIO)、JNK阻害剤(SP600125)、P38阻害剤(SB203580)、PKC阻害剤(Go6983)、ROCK阻害剤(Y-27632)、BMP阻害剤(Dorsomorphin)、BRAF阻害剤(SB590885)、SRC阻害剤(WH-4-023)のいずれかまたは複数のシグナル阻害剤を含む培地を用いたフィーダー培養またはフィーダーレス培養を行うことで「ナイーブ型」を誘導する方法が報告されている(非特許文献9~14)。
 上記「ナイーブ型」誘導法の共通点は、培地がMEK阻害剤とGSK阻害剤を必ず含み、フィーダー細胞またはマトリジェルを用いた接着培養により細胞を増殖させる点である。これら方法により誘導された「ナイーブ型」細胞では一般的な培地中で見られる「プライム型」と比較してSTELLAやTFCP2L1などの「ナイーブ型」に特有の遺伝子の発現が顕著に増加することが知られている。
 ヒトiPS細胞を、MEK阻害剤を含まないで培養することにより、「ナイーブ型」のヒトiPS細胞を誘導した例は知られていない。

 本発明は、ナイーブ型多能性幹細胞の未分化と多能性(多分化能)維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖させるためのナイーブ型多能性幹細胞培養用培地を提供することを目的とする。
 本発明者らは、低濃度のROCK阻害剤を培地中に存在させることにより上記課題を解決し得ることを見出し、本発明を完成させた。
 すなわち、本発明は、以下の(1)~(9)に関する。
(1)ROCK阻害剤を5μM以下の濃度で含む、ナイーブ型多能性幹細胞培養用培地。
(2)ROCK阻害剤がY-27632である、(1)に記載のナイーブ型多能性幹細胞培養用培地。
(3)MEK阻害剤を含んでいない、(1)または(2)に記載のナイーブ型多能性幹細胞培養用培地。
(4)フィーダー細胞および細胞外マトリクスを含んでいない、(1)~(3)のいずれか1に記載のナイーブ型多能性幹細胞培養用培地。
(5)増殖因子を更に含む、(1)~(4)のいずれか1に記載のナイーブ型多能性幹細胞培養用培地。
(6)浮遊培養に使用される、(1)~(5)のいずれか1に記載のナイーブ型多能性幹細胞培養用培地。
(7)ナイーブ型多能性幹細胞がヒトiPS細胞である、(1)~(6)のいずれか1に記載のナイーブ型多能性幹細胞培養用培地。
(8)ROCK阻害剤を5μM以下の濃度で含む培地中でナイーブ型多能性幹細胞を培養することにより、多能性幹細胞のナイーブ型を維持しながら多能性幹細胞を増殖させることを特徴とする、多能性幹細胞の培養方法。
(9)ROCK阻害剤がY-27632である、(8)に記載の多能性幹細胞の培養方法。
(10)培地中にMEK阻害剤を含んでいない、(8)または(9)に記載の多能性幹細胞の培養方法。
(11)培地中にフィーダー細胞および細胞外マトリクスを含んでいない、(8)~(10)のいずれか1に記載の多能性幹細胞の培養方法。
(12)浮遊培養で実施される、(8)~(11)のいずれか1に記載の多能性幹細胞の培養方法。
(13)ナイーブ型多能性幹細胞がヒトiPS細胞である、(8)~(12)のいずれか1に記載の多能性幹細胞の培養方法。
 本発明のナイーブ型多能性幹細胞培養用培地によれば、低濃度のROCK阻害剤を含むことにより、ナイーブ型多能性幹細胞の未分化と多能性(多分化能)維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖させることができる。また、本発明の多能性幹細胞の培養方法によれば、低濃度のROCK阻害剤を含む培地中でナイーブ型多能性幹細胞を培養することにより、未分化と多能性(多分化能)維持しつつ、多能性幹細胞のナイーブ型を効率よく維持することができる。
図1Aおよび図1Bは、ROCK阻害剤の存在下でのiPS細胞の増殖を観察した結果を示す図である。 図2は、ROCK阻害剤の存在下でのiPS細胞の数、およびiPS細胞の生存率を測定した結果を示すグラフである。 図3A~図3Dは、ROCK阻害剤の存在下での、iPS細胞の未分化マーカー遺伝子であるNANOGおよびOCT3/4、ナイーブ型に特異的なマーカー遺伝子であるSTELLA、並びに中肺葉分化に特異的なマーカー遺伝子であるNCAMの発現量をリアルタイムPCRにより確認した結果をそれぞれ示すグラフである。 図4は、低濃度のROCK阻害剤および増殖因子存在下における、iPS細胞の増殖を示すグラフである。 図5A~図5Dは、低濃度のROCK阻害剤および増殖因子存在下における、iPS細胞の未分化マーカー遺伝子およびナイーブ型に特異的なマーカー遺伝子の発現を示すグラフである。 図6は、低濃度のROCK阻害剤および低分子化合物の存在下における、iPS細胞の増殖率を示すグラフである。 図7Aおよび図7Bは、低濃度のROCK阻害剤および低分子化合物の存在下における、iPS細胞の未分化マーカー遺伝子の発現を示すグラフである。
1.ナイーブ型多能性幹細胞培養用培地
 本発明は、ROCK阻害剤を5μM以下の濃度で含む、ナイーブ型多能性幹細胞培養用培地(以下、「本発明の培地」ともいう。)である。本発明の培地は、多能性幹細胞用基礎培地に、5μM以下のROCK阻害剤を含んでなる。
 前記多能性幹細胞用基礎培地としては、ROCK阻害剤を5μM以下の濃度で添加することにより、ナイーブ型多能性幹細胞の未分化と多能性(多分化能)を維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖させることのできる培地であれば、特に制限されないが、調製が容易であり、ロットごとのばらつきを防ぐ点から化学合成培地が好ましい。
 前記多能性幹細胞用基礎培地は、1種類以上の糖(類)、1種類以上の無機塩(類)、1種類以上のアミノ酸(類)、1種類以上のビタミン(類)、および1種類以上の微量成分(類)を含むことが好ましい。また、薬剤感受性試験に使用するためにカナマイシン等の抗生物質を適宜含むこともできる。
 前記糖(類)としては、例えば、グルコース、ラクトース、マンノース、フルクトースおよびガラクトース等の単糖類、並びにスクロース、マルトースおよびラクトース等の二糖類が挙げられる。これらの中でもグルコースが特に好ましい。これら糖類は、1または2以上組み合わせて添加することもできる。
 前記無機塩(類)としては、例えば、塩化カルシウム、硝酸カルシウム、硫酸銅五水和物、硝酸鉄(III)九水和物、硫酸鉄(II)七水和物、塩化マグネシウム六水和物、硫酸マグネシウム、塩化カリウム、塩化ナトリウム、リン酸水素二ナトリウム、リン酸水素二ナトリウム二水和物、リン酸二水素ナトリウム、リン酸二水素ナトリウム二水和物および硫酸亜鉛七水和物が挙げられる。多能性幹細胞の未分化状態の維持に有利に作用する成分であればいずれの無機塩類またはその組合せも用いることができる。
 前記アミノ酸(類)としては、例えば、アラニン、アルギニン、アスパラギン、アスパラギン酸、シスチン、システイン、グルタミン、グリシン、ヒスチジン、グルタミン酸、ヒドロキシプロリン、イソロイシン、ロイシン、リジン、メチオニン、フェニルアラニン、プロリン、セリン、スレオニン、トリプトファン、チロシンおよびバリン等が挙げられ、L-体のアミノ酸が好ましい。前記アミノ酸(類)には、これらの誘導体、塩および水和物などの派生物を含めることができる。
 アルギニンの派生物としては、例えば、L-塩酸アルギニンおよびL-アルギニン一塩酸塩等が挙げられる。アスパラギン酸の派生物としては、例えば、L-アスパラギン酸ナトリウム塩一水和物、L-アスパラギン酸一水和物、L-アスパラギン酸カリウム、L-アスパラギン酸マグネシウム等が挙げられる。
 システインの派生物としては、例えば、L-システイン二塩酸塩およびL-システイン塩酸塩一水和物等が挙げられる。リジンの派生物としては、例えば、L-リジン塩酸塩等が挙げられる。グルタミン酸の派生物としては、L-グルタミン酸一ナトリウム塩等が挙げられる。
 アスパラギンの派生物としては、例えば、L-アスパラギン一水和物等が挙げられる。チロシンの派生物としては、例えば、L-チロシン二ナトリウム二水和物等が挙げられる。ヒスチジンの派生物としては、例えば、ヒスチジン塩酸塩およびヒスチジン塩酸塩一水和物等が挙げられる。リジンの派生物としては、例えば、L-リジン塩酸塩等が挙げられる。
 前記ビタミン(類)としては、例えば、アスコルビン酸、ビオチン、コリン、葉酸、イノシトール、ナイアシン、パントテン酸、ピリドキシン、リボフラビン、チアミン、ビタミンB12およびパラアミノ安息香酸(PABA)が挙げられる。これらの中でも、アスコルビン酸は添加されることが好ましい。前記ビタミン(類)には、これらの誘導体、塩および水和物などの派生物を含めることができる。
 アスコルビン酸の派生物としては、例えば、アスコルビン酸2-リン酸エステル(Ascorbic acid 2-phosphate)、アスコルビン酸リン酸マグネシウム、アスコルビン酸硫酸ナトリウム、リン酸アスコルビルアミノプロピルおよびアスコルビン酸リン酸ナトリウム等が挙げられる。
 コリンの派生物としては、例えば、塩化コリン等が挙げられる。ナイアシンの派生物としては、例えば、ニコチン酸、ニコチン酸アミドおよびニコチニックアルコール等が挙げられる。パントテン酸の派生物としては、例えば、パントテン酸カルシウム、パントテン酸ナトリウムおよびパンテノール等が挙げられる。
 ピリドキシンの派生物としては、例えば、ピリドキシン塩酸塩、ピリドキサール塩酸塩、リン酸ピリドキサールおよびピリドキサミン等が挙げられる。チアミンの派生物としては、例えば、塩酸チアミン、硝酸チアミン、硝酸ビスチアミン、チアミンジセチル硫酸エステル塩、塩酸フルスルチアミン、オクトチアミンおよびベンフォチアミン等が挙げられる。
 前記微量成分は多能性幹細胞の未分化状態の維持に有利に作用する成分であることが好ましい。前記微量成分としては、例えば、グルタチオン、ヒポキサンチン、リポ酸、リノレン酸、フェノールレッド、プトレシン、ピルビン酸、チミジンおよびNaHCO等通常培地成分として用いられている成分が挙げられる。前記微量成分には、これらの誘導体、塩および水和物などの派生物を含めることができる。派生物としては、例えば、プトレシン二塩酸を挙げることができる。
 多能性幹細胞用基礎培地として具体的には、例えば、市販のダルベッコ改変イーグル培地(DMEM)、最小必須培地(MEM)、イーグル基礎培地(BME)、RPMI1640培地およびF12培地等の公知の化学合成培地などの培地、並びにDMEM/F12培地(DMEMとF12培地を1:1で混合した培地)等のこれらの培地のいずれか2以上を適当な割合で混合した培地、およびこれらの培地に前記アスコルビン酸、好ましくはアスコルビン酸2-リン酸エステルを添加した培地が挙げられる。
 特にアニマルプロダクトフリーの基礎培地として、Miho Furue et al. In Vitro Cell Dev Biol Anim. 41, 19-28 (2005)に示されるESF Nutrient Medium(以下、「mESF Basal Medium」ともいう)を好適に例示することができる。
 ROCK阻害剤とは、Rhoキナーゼの活性を阻害する物質を意味する。Rhoキナーゼとは、GTP(グアノシン三リン酸)の分解酵素であるGTPアーゼの範疇に含まれる低分子量GTP結合タンパク質(低分子量Gタンパク質)の1種で、アミノ末端にセリン/スレオニンキナーゼ領域、中央部にコイルドコイル領域およびカルボキシ末端にRho相互作用領域を有する。
 ROCK阻害剤としては、例えば、(+)-(R)-トランス-4-(1-アミノエチル)-N-(4-ピリジル)シクロヘキサンカルボキサミド二塩酸塩(Y-27632)、1-(5-イソキノリンスルホニル)-2-メチルピペラジン(H-7)、1-(5-イソキノリンスルホニル)-3-メチルピペラジン(イソH-7)、N-2-(メチルアミノ)エチル-5-イソキノリンスルホンアミド二塩酸塩(H-8)、N-(2-アミノエチル)-5-イソキノリンスルホンアミド二塩酸塩(H-9)、N-[2-(p-ブロモシンナミルアミノ)エチル]-5-イソキノリンスルホンアミド二塩酸塩(H-89)、N-(2-グアニジノエチル)-5-イソキノリンスルホンアミド塩酸塩(HA-1004)、1-(5-イソキノリンスルホニル)ホモピペラジン二塩酸塩(Fasudil/HA-1077)および(S)-(+)-2-メチル-4-グリシル-1-(4-メチルイソキノリニル-5-スルホニル)ホモピペリジン二塩酸塩(H-1152)等が挙げられる。これらの中でも、(+)-(R)-トランス-4-(1-アミノエチル)-N-(4-ピリジル)シクロヘキサンカルボキサミド二塩酸塩(Y-27632)が好ましい。
 本発明の培地に含まれるROCK阻害剤の濃度は、5μM以下であり、好ましくは4μM以下であり、更に好ましくは3μM以下であり、より更に好ましくは2μM以下であり、特に好ましくは1.5μM以下である。また、ROCK阻害剤濃度の下限値は特に制限されないが、通常0.3μM以上とすることが好ましい。ROCK阻害剤の濃度が5μMを超えると、多能性幹細胞のナイーブ型の割合が低下する。また、0.3μM以上とすることにより、多能性幹細胞の増殖が低下するのを抑制することができる。
 多能性幹細胞のナイーブ型を維持するためには、通常、MEK阻害剤を培地に含有する必要があったが、本発明の培地には、5μM以下のROCK阻害剤が含まれていることにより、該培地を用いて多能性幹細胞を培養した場合に、多能性幹細胞のナイーブ型が維持されるので、培地にMEK阻害剤を含有させる必要はない。
 本発明の培地は、ROCK阻害剤を5μM以下の濃度で含んでいるため、多能性幹細胞を単細胞細胞または数個から数10個の細胞群に分散した場合にアポトーシスを起こしにくくなっている。このため、本発明の培地は、多能性幹細胞を浮遊培養する場合にも、好ましく用いられる。このように、多能性幹細胞を浮遊培養することができるので、少ないコストで大量の細胞を生産することが可能になる。
 したがって、本発明の培地には、浮遊培養を行うときに、接着培養に必要であった、フィーダー細胞および細胞外マトリクスを含まなくてもよい。
 本発明の培地には、増殖因子を更に含有させてもよい。本発明の培地は、そのROCK阻害剤の含有量が低いため、多能性幹細胞の増殖が低下する場合がある。本発明の培地に、増殖因子を含有させることにより、多能性幹細胞の増殖が回復するという効果が発揮される。
 増殖因子としては、例えば、アクチビンA(Activin A)トランスフォーミング成長因子-α(TGF-α)、トランスフォーミング成長因子-β(TGF-β)、マクロファージ炎症蛋白質-1α(MIP-1α)、上皮細胞増殖因子(EGF)、繊維芽細胞増殖因子-1、2、3、4、5、6、7、8または9(FGF-1、2、3、4、5、6、7、8または9)、神経細胞増殖因子(NGF)、肝細胞増殖因子(HGF)、白血病阻止因子(LIF)、プロテアーゼネキシンI、プロテアーゼネキシンII、血小板由来成長因子(PDGF)、コリン作動性分化因子(CDF)、ケモカイン、Notchリガンド(例えば、Delta1など)、Wnt蛋白質、アンジオポエチン様蛋白質2、3、5または7(Angpt2、3、5または7)、インスリン様成長因子(IGF)、インスリン様成長因子結合蛋白質(IGFBP)、プレイオトロフィン(Pleiotrophin)およびヘパラン硫酸等が挙げられる。これらの中でも、FGF2、LIFまたはヘパラン硫酸が好ましい。
 本発明の培地においては、増殖因子を単独で用いてもよく、または2種以上を組み合わせて用いてもよい。増殖因子を用いる場合、その濃度は、通常0.1~1000ng/mLが好ましく、より好ましくは1~20ng/mLである。
 また、本発明の培地には、GSK3阻害剤、BRAF阻害剤またはBMPを阻害する内在性タンパク質等を含有させてもよい。このような化合物を培地に含有させることによって、ナイーブ型多能性幹細胞の未分化と多能性(多分化能)を維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖させることができる。
 GSK3阻害剤としては、例えば、CHIR99021(6-{2-[4-(2,4-ジクロロ-フェニル)-5-(4-メチル-1H-イミダゾール-2-イル)-ピリミジン-2-イルアミノ]-エチルアミノ}-ニコチノニトリル)、CHIR98014(2-[[2-[(5-ニトロ-6-アミノピリジン-2-イル)アミノ]エチル]アミノ]-4-(2,4-ジクロロフェニル)-5-(1H-イミダゾール-1-イル)ピリミジン)、AR-AO144-18、TDZD-8(4-ベンジル-2-メチル-1,2,4-チアジアゾリジン-3,5-ジオン)、SB216763(3-(2,4-ジクロロフェニル)-4-(1-メチル-1H-インドール-3-イル)-1H-ピロール-2,5-ジオン)およびSB415286(3-[(3-クロロ-4-ヒドロキシフェニル)アミノ]-4-(2-ニトロフェニル)-1H-ピロール-2,5-ジオン)、1-AzKenpaullone(9-ブロモ-7,12-ジヒドロ-ピリド[3’,2’:2,3]アゼピノ[4,5-b]インドール-6(5H)-オン)、Kenpaullone(9-ブロモ-7,12-ジヒドロ-インドロ-[3,2-d]-[1]ベンズアゼピン-6(5H)-オン)等が挙げられるが、これらに限定されない。これらの中でもCHIR99021が好ましい。
 BRAF阻害剤としては、例えば、GDC-0879、BAY 7304506(レゴラフェニブ)、RAF265(CHIR-265)、SB590885およびソラフェニブが挙げられるが、これらに限定されない。これらの中でもSB590885が好ましい。
 BMP阻害剤としては、転写因子であるSMAD1、SMAD5またはSMAD8を活性化する、BMP2、BMP4、BMP6またはBMP7を阻害する化合物、例えば、Noggin、chordinおよびfollistatin等が挙げられる。これらの中でもNogginが好ましい。
 MEK阻害剤としては、例えば、MEK1を阻害するPD184352及びPD98059、MEK1及びMEK2を阻害するPD0325901、U0126及びSL327等が挙げられるがこれらに限定されない。
 本発明の培地においては、前記GSK3阻害剤、BRAF阻害剤またはBMPを阻害する内在性タンパク質等を単独で用いてもよく、または2種以上を組み合わせて用いてもよい。また、その濃度は、通常0.001~20μMが好ましく、より好ましくは0.02~10μMである。
 本発明のナイーブ型多能性幹細胞培養用培地は、ナイーブ型多能性幹細胞の未分化と多能性(多分化能)を維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖させるために使用されるが、具体的にはヒトの多能性幹細胞を対象とする。
 多能性幹細胞としては、例えば、初期胚より単離される胚性幹細胞(embryonic stem cells:ES細胞);胎児期の始原生殖細胞から単離される胚性生殖細胞(embryonic germ cells:EG細胞)(例えば、Proc Natl Acad Sci U S A. 1998, 95:13726-31参照);出生直後の精巣から単離される生殖細胞系列幹細胞(germline stem cells:GS細胞)(例えば、Nature. 2008, 456:344-9参照);腸骨骨髄または顎骨骨髄等の骨髄由来の幹細胞;脂肪組織由来の幹細胞などの間葉系幹細胞;皮膚細胞等の体細胞に複数の遺伝子を導入することで、被検体自身の体細胞の脱分化を誘導し、ES細胞同様の多能性を有する体細胞由来人工多能性幹細胞[または誘導多能性幹細胞(induced pluripotent stem cell;iPS細胞)]が挙げられる。
 具体的には、例えば、ヒトES細胞H9株(WA09);ヒトES細胞H1(WA01)株(ウィスコンシン大学、WISC Bank);KhES-1、KhES-2およびKhES-3(京大再生研付属幹細胞医学研究センター、理化学研究所バイオリソースセンター)並びにHES3、HES4およびHES6(モナッシュ大学、WISC Bank、)などのヒトES細胞;Oct3/4遺伝子、Klf4遺伝子、C-Myc遺伝子およびSox2遺伝子を導入することによって得られるiPS細胞(京都大学iPS細胞研究所、理化学研究所バイオリソースセンター);Tic(JCRB1331株)、Dotcom(JCRB1327株)、Squeaky(JCRB1329株)、Toe(JCRB1338株)、およびLollipop(JCRB1336株)(以上成育医療センター、医薬基盤健康・栄養研究所JCRB細胞バンク);UTA-1株(東京大学)およびUTA-1-SF-2-2株(JCRB1493)(東京大学、医薬基盤健康・栄養研究所JCRB細胞バンク);Oct3/4遺伝子、Klf4遺伝子およびSox2遺伝子を導入することによって得られるiPS細胞(Nat Biotechnol 2008; 26: 101-106)(京都大学iPS細胞研究所、理化学研究所バイオリソースセンター、医薬基盤健康・栄養研究所JCRB細胞バンク)等のiPS細胞が挙げられる。
2.多能性幹細胞の培養方法
 次に、本発明の多能性幹細胞の培養方法について説明する。本発明の多能性幹細胞の培養方法は、ROCK阻害剤を5μM以下の濃度で含む培地中でナイーブ型多能性幹細胞を培養することにより、多能性幹細胞のナイーブ型を維持しながら多能性幹細胞を増殖させることを特徴とする方法(以下、「本発明の培養方法」ともいう。)である。ROCK阻害剤については上述した通りであり、その好ましい濃度についても上述した通りである。
 多能性幹細胞のナイーブ型を維持するためには、通常、MEK阻害剤を培地に含有させる必要があったが、本発明の培養方法においては、培地中に5μM以下のROCK阻害剤が含まれており、このため、この培地を用いて多能性幹細胞を培養した場合に、多能性幹細胞のナイーブ型が維持されるので、本発明の培養方法においては、培地にMEK阻害剤を含有させる必要はない。
 本発明の培養方法は浮遊培養で行うことが好ましい。浮遊培養で培養を行うことにより、少ないコストで大量の細胞を生産することが可能になる。したがって、本発明の培養方法は、浮遊培養で培養を行う時に、接着培養に必要であったフィーダー細胞および細胞外マトリクスを用いて細胞培養を行う必要がない。
 本発明の培養方法では、多能性幹細胞の増殖効率を高めるために、増殖因子を用いてもよい。本発明の培養方法に用いられる増殖因子の種類、培地への添加量は上述した通りである。
 また、本発明の培養方法では、GSK3阻害剤、BRAF阻害剤またはBMPを阻害する内在性タンパク質等を用いてもよい。GSK3阻害剤、BRAF阻害剤またはBMPを阻害する内在性タンパク質等の種類、培地への添加量は上述した通りである。
 本発明の培養方法における培養は、通常33~40℃、好ましくは34~39℃、より好ましくは37℃にて、通常1~20%、好ましくは3~15%、より好ましくは8~10%の二酸化炭素の存在下で、通常75%以上、好ましくは85%以上、より好ましくは95%、特に好ましくは100%の高湿度下で行なうことが好ましい。
 使用することのできる多能性幹細胞としては上述した通りである。
 本発明の培養方法により培養される細胞が未分化性、およびナイーブ型を維持している細胞であるか否かを確認する方法としては、例えば、各種マーカー(タンパク質)に対する抗体を用いて、未分化マーカーの発現の検出および/または分化マーカーの発現の不検出により判定する方法、各種マーカー(遺伝子)の発現の検出により判定する方法、細胞の形態学的特徴を観察する方法、並びに特定の分化誘導因子の刺激により特定の細胞に分化する能力を発揮できるか否かを判定する方法が挙げられる。
 前記マーカーを用いて判定する方法においては、NANOG、OCT3/4、SSEA-3、SSEA-4、TRA-2-54、TRA-1-60、TRA-1-80、CD90またはアルカリホスファターゼ等の未分化マーカーが発現している場合は、本発明の培養方法で培養した多能性幹細胞が未分化性を維持していると判断することができる。また、STELLA等のマーカーが発現している場合は、本発明の培養方法で培養した多能性幹細胞がナイーブ型であると判断することができる。
 前記未分化および/または分化マーカーの発現をタンパク質レベルで確認する場合には、例えば、各マーカーの特異抗体を用いて、フローサイトメトリー法、免疫染色法またはELISA等により確認することができる。前記マーカーの発現を遺伝子レベルで確認する場合には、例えば、各マーカー遺伝子の、特異的プライマー対を用いたRT-PCRまたは特異的プローブを用いたノーザンブロッティング若しくはマイクロアレイ解析等によって確認することができる。
 前記フローサイトメトリー法によりマーカー発現の有無を確認する方法としては、例えば、次の方法が挙げられる。本発明の方法で培養した細胞を1mlの10%ヤギ血清中に30分間懸濁後遠心分離し、次に対象のマーカータンパク質に対するマウス抗体とともに30時間インキュベートし、その後当該培養細胞を、1%ヤギ血清を含有するPBSで3回洗浄し、蛍光標識ヤギ抗マウスIgG抗体と30分間反応させ、1%ヤギ血清を含有するPBSで3回洗浄し、再懸濁した細胞を適切なフローサイトメトリー用機器を用いて判定することができる。
 前記免疫染色法によりマーカー発現の有無を確認する方法としては、例えば、次の方法が挙げられる。本発明の方法で培養した細胞を、PBS中4%パラホルムアルデヒド(PFA)で固定後、複数回PBSにて洗浄後、トライトンXなどで培養細胞の透過性を増加させ、10%ヤギ血清でブロッキングを行った後、対象マーカータンパク質に対するマウス抗体を用いて免疫染色し、蛍光標識ヤギ抗マウスIgGと反応させ、蛍光顕微鏡観察により判定することができる。
 前記アルカリホスファターゼの発現の有無を確認する方法としては、例えば、次の方法が挙げられる。本発明の方法で培養した細胞を、4.5mMクエン酸、2.25mMクエン酸ナトリウム、3mM塩化ナトリウム、65%メタノールおよび4%パラホルムアルデヒドで5分間固定し、洗浄し、次にFastRed基質キット(シグマ社製)等の適当なキットを用い、アルカリホスファターゼを可視化する、いわゆるアルカリホスファターゼ染色することができる。
 次に実施例を挙げて本発明をさらに詳細に説明するが、本発明はこれらにより何ら限定されるものではない。
[実施例1]
ROCK阻害剤存在下でのiPS細胞の増殖における影響
 以下の実施例においては、mESF Basal Medium(和光純薬社製、136-17805)に20μmフィルターを通過して滅菌した下記成分を、表1に示す添加量で混合したものをhESF-FX培地として使用した。
 rHSA-Oleic acidは50mLの100mg/mL 組換えヒト血清アルブミン(rHSA、ミリポア社製、9501-20)に235μLの200mg/mLのOleic acid(シグマアルドリッチ社製、O1383-5G)を添加し、終夜混合することにより調製した。

 また、iPS細胞としては、Tic-FX細胞[iPS細胞Tic株(JCRB1331)を、フィーダーフリー・ゼノフリー培養(FX)に順化させた株]を使用した。
 hESF-FX培地にROCK阻害剤(Y-27632)を、1μM、3μMおよび10μMとなるように溶解した。それぞれの培地を、超低接着6穴プレート(Corning社製)に3mLずつ加え、上記Tic-FX細胞を、10%のCO存在下、37℃にて培養した(浮遊培養)。
 培養1日後、3日後、5日後のプレートの様子を観察した。結果を図1Aおよび図1Bに示す。図1Aは倍率4倍、図1Bは倍率2倍の顕微鏡写真である。また、培養5日後のウェル当たりの細胞数および細胞の生存率を測定した。その結果を図2に示す。
 図1Aおよび図1Bに示すように、ROCK阻害剤の濃度が高いほど細胞数は増えていた。また、図2に示すように、ウェルあたりの細胞数、細胞の生存率は、いずれもROCK阻害剤の濃度が高くなるにつれ上昇していた。
[実施例2]
未分化マーカー遺伝子およびナイーブ型に特異的なマーカー遺伝子の発現
 マーカー遺伝子の発現をリアルタイムPCRで確認した。リアルタイムPCRは、リアルタイムPCR装置(アプライドバイオシステムズ社製7300)にて、下記の反応手順およびプライマーにより実施した。

 実施例1で培養した細胞について、培養後5日後の細胞中の、未分化マーカー遺伝子であるNANOGおよびOCT3/4、ナイーブ型に特異的なマーカー遺伝子であるSTELLA、並びに神経細胞分化に特異的なマーカー遺伝子であるNCAMの発現量を、前記リアルタイムPCRにより確認した。結果を図3A~図3Dに示す。
 図3A~図3D中、縦軸は、内部標準として使用したGAPDH(グリセルアルデヒド3リン酸脱水素酵素)に対する各サンプルの目的遺伝子の発現量の比を意味する。具体的には、グラフの値はGAPDHのCp値と目的遺伝子のCp値の差から求められる増幅前のDNA量の相対値(単位はfold)を意味する。
 なおここで、内部標準は細胞の状態によらず常に発現量が一定となる遺伝子である。また、Cp値とは、PCR反応によってDNAを増幅させたとき、測定器で検出可能な量まで増幅させるのに必要なサイクル数(理論上、1サイクルでDNA量は2倍になる)である。
 図3Aおよび図3Bに示すように、未分化マーカー遺伝子であるNANOGおよびOCT3/4は、ROCK阻害剤の濃度が1μMおよび3μMの時には、対照(図3Aおよび図3B中、Tic-FX)よりも発現量が多かったのに対し、ROCK阻害剤を10μM濃度で用いた場合には、両者の発現量は対照よりも低くなった。
 また、図3Cに示すように、ナイーブ型に特異的なSTELLA遺伝子は、ROCK阻害剤の濃度が10μMでは、対照よりもわずかに発現量が多かったが、ROCK遺伝子の濃度を1μMおよび3μMとした場合には、発現量が対照よりも多くなっていた。
 この結果から、ROCK阻害剤を、従来用いられている10μM濃度よりも低濃度(1μMおよび3μM)で用いた場合に、未分化を維持しつつ、ナイーブ型多能性幹細胞を効率よく増殖できることがわかった。
 また、図3Dに示すように、中肺葉分化に特異的なマーカー遺伝子であるNCAMは、ROCK阻害剤濃度が1μMでは対照とほとんど変わらず、3μMおよび10μMでは、発現量は対照よりも上昇していた。
[実施例3]
増殖因子存在下における、iPS細胞の増殖
 上記基礎培地に、1μMのY-27632と、表2に示す成分を加えた培地を作製し、超低接着6穴プレート(Corning社製)中、10%のCO存在下、37℃にてTic-FX細胞の増殖を浮遊培養で行った。播種の5日後、10日後、15日後に継代し、20日後まで培養を行った。継代はAccutase(ミリポア社製、SCR005)を用いて細胞塊を37℃で10分間処理することにより行った。播種の5日後、10日後、15日後および20日後に細胞数をカウントした。その結果を図4に示す。

 図4に示すように、1μMのY-27632の存在下では、5ng/mLの組換えヒトFGF2を加えたもの(No.1)と、5ng/mLの組換えヒトFGF2および20ng/mLの組換えヒトLIFを加えたもの(No.2)では、増殖率は低かった。これに対し、40ng/mLのFGF2および20ng/mLの組換えヒトLIFを加えたもの(No.3)と、組換えヒトLIF、組換えヒトFGF2およびヘパラン硫酸ナトリウムを、それぞれ20ng/mL、5ng/mLおよび100ng/mL加えたもの(No.4)では増殖率が上昇した。
 この結果から、浮遊細胞の効率的な培養には、低濃度のROCK阻害剤とともに、組換えヒトLIF、組換えヒトFGF2、ヘパラン硫酸ナトリウム等の増殖因子を培地に加えることが好ましいことがわかった。
[実施例4]
未分化マーカー遺伝子およびナイーブ型に特異的なマーカー遺伝子の発現
 実施例3で培養した細胞(表2のNo.3および4の細胞)について、マーカー遺伝子の発現を、実施例2と同様にしてリアルタイムPCRで確認した。TFCP2L1遺伝子の発現の確認に用いたプライマーの塩基配列を下記に示す。

TFCP2L1リバースプライマー caatcagctccaaggtggtcag(配列番号12)
 結果を図5A~図5Dに示す。なお、対照として、培地に組換えヒトLIF、組換えヒトFGF2およびヘパラン硫酸ナトリウムをいずれも添加しないものを用いた(図5A~図5Dにおいて、Tic-FXと表わす)。
 図5Aおよび図5Bに示すように、ROCK阻害剤であるY-27632が低濃度で存在する場合でも、培地に増殖因子を添加した場合には、多能性マーカー(未分化マーカー遺伝子)である、NANOGおよびOCT3/4の発現が増大していることがわかった。また、図5Cおよび図5Dに示すように、ナイーブ型に特異的なSTELLA遺伝子およびTFCP2L1遺伝子も、両者で発現していることがわかった。
[実施例5]
低分子化合物の存在下における、iPS細胞の培養
 上記基礎培地に、1μMのY-27632と、20ng/mLのLIFおよび40ng/mLのFGF2とを加えた培地を作製し、それぞれに、下記に示す各種阻害剤を、それぞれ1μM(PD0325901)、0.3μM(CHIR99021)、0.5μM(SB590885)、1μM(WH-4-023)、10μM(SP600125)、10μM(SB203580)、5μM(Go6983)、5μM(GF109203X)、2μM(Dorsomorphin)、400ng/mL(Noggin)の濃度となるように加えた培地を作製した。
 それぞれの培地を超低接着6穴プレート(Corning社製)中、10%のCO存在下、37℃にてTic-FX細胞の増殖を浮遊培養で行った。培養を5日間行い、細胞数をカウントした。結果を図6に示す。図6において、-と示すものは、阻害剤を添加しなかった培地を用いた結果である。

 図6に示すように、CHIR99021、SB590885またはNogginを添加した培地では、細胞の増殖率が高かった。
 次いで、細胞の増殖率が高かった阻害剤を加えた培地で増殖した細胞の多能性マーカー(NANOGおよびOCT3/4)の発現について、実施例2と同様にしてリアルタイムPCRで確認した。結果を図7Aおよび図7Bに示す。なお、対照として、基礎培地で培養したものを用いた(図7Aおよび図7Bにおいて、Tic-FXと表わす)。
 図7Aおよび図7Bから明らかなように、CHIR99021、SB590885、またはNogginを添加した培地では、NANOGおよびOCT3/4の発現が増大していることがわかった。
 このことより、本発明の培地に加え、更にCHIR99021、SB590885またはNogginを添加することによって、細胞の未分化状態を維持したまま、生産性を更に高めることができることがわかった。
 本発明を特定の態様を参照して詳細に説明したが、本発明の精神と範囲を離れることなく様々な変更および修正が可能であることは、当業者にとって明らかである。なお、本出願は、2016年3月30日付けで出願された日本特許出願(特願2016-068718)に基づいており、その全体が引用により援用される。また、ここに引用されるすべての参照は全体として取り込まれる。

Claims (13)

  1.  ROCK阻害剤を5μM以下の濃度で含む、ナイーブ型多能性幹細胞培養用培地。
  2.  ROCK阻害剤がY-27632である、請求項1に記載のナイーブ型多能性幹細胞培養用培地。
  3.  MEK阻害剤を含んでいない、請求項1または2に記載のナイーブ型多能性幹細胞培養用培地。
  4.  フィーダー細胞および細胞外マトリクスを含んでいない、請求項1~3のいずれか1項に記載のナイーブ型多能性幹細胞培養用培地。
  5.  増殖因子を更に含む、請求項1~4のいずれか1項に記載のナイーブ型多能性幹細胞培養用培地。
  6.  浮遊培養に使用される、請求項1~5のいずれか1項に記載のナイーブ型多能性幹細胞培養用培地。
  7.  ナイーブ型多能性幹細胞がヒトiPS細胞である、請求項1~6のいずれか1項に記載のナイーブ型多能性幹細胞培養用培地。
  8.  ROCK阻害剤を5μM以下の濃度で含む培地中でナイーブ型多能性幹細胞を培養することにより、多能性幹細胞のナイーブ型を維持しながら多能性幹細胞を増殖させることを特徴とする、多能性幹細胞の培養方法。
  9.  ROCK阻害剤がY-27632である、請求項8に記載の多能性幹細胞の培養方法。
  10.  培地中にMEK阻害剤を含んでいない、請求項8または9に記載の多能性幹細胞の培養方法。
  11.  培地中にフィーダー細胞および細胞外マトリクスを含んでいない、請求項8~10のいずれか1項に記載の多能性幹細胞の培養方法。
  12.  浮遊培養で実施される、請求項8~11のいずれか1項に記載の多能性幹細胞の培養方法。
  13.  ナイーブ型多能性幹細胞がヒトiPS細胞である、請求項8~12のいずれか1項に記載の多能性幹細胞の培養方法。
