Abstract

 細胞を吸引するシリンジ等を、吸引する細胞に正確に位置合わせができる細胞搬送装置を提供することを課題とする。 セルチャンバーに設けられた凹部に、培養した細胞を詰めるための細胞搬送装置であって、前記細胞搬送装置は、撮像部、細胞を吸引するシリンジを装着できるシリンジポンプ、シリンジポンプ駆動部、前記シリンジポンプ駆動部を制御するための制御部を含み、前記制御部は、前記撮像部で撮像した細胞の画像から算出した細胞の位置座標位置、及び前記撮像部で撮像した前記凹部の画像から算出した凹部の位置座標位置を記憶する記憶部を含み、前記制御部が、前記記憶部に記憶された細胞及び凹部の位置座標位置に基づき前記シリンジポンプ駆動部を制御する細胞搬送装置により、上記課題を解決できる。

Description

細胞搬送装置、細胞搬送方法、及び人工臓器の製造方法
 本発明は、細胞搬送装置、細胞搬送方法、及び人工臓器の製造方法に関する。特に、分化した幹細胞を挿入・保持して生体内に留置することで人工臓器として使用できるセルチャンバーに、培養装置で培養した幹細胞を搬送するための細胞搬送装置及び細胞搬送方法、並びに、当該細胞搬送装置及び細胞搬送方法を用いた人工臓器の製造方法に関する。
 心臓、肺、肝臓、腎臓、膵臓などの臓器、骨、目等の生体を構成する組織の機能が損なわれると種々の病気になり、重い場合には生命の危機にさらされる。生体組織の病気が比較的軽症の場合は、投薬・手術等により病変部分を治癒することが可能である。しかしながら、生体組織の機能が著しく低下し治癒が困難な場合は、生体組織の機能を代行するために、人工臓器が用いられている。
 人工臓器は、セラミック製の人工骨やインプラント、ペースメーカー等の機械要素からなる人工臓器、生きた細胞を用いた組織工学により作製した人工臓器が知られている。後者の人工臓器としては、例えば、3次元培養した膵島および/または膵島細胞塊を高分子膜によって膵内分泌細胞を被包し、患者の皮下あるいは腹膣内等に移植できるようにした拡散チャンバー型人工臓器が知られている(特許文献1参照)。また、シリコーンゴムリング2の両開口端面にポリカーボネイト製の免疫隔離膜を接着してなる容器Vの中に失われた本来の膵臓の代替となりうる膵細胞とその機能を維持する細胞培養床5を封入してなる人工臓器用チャンバーであって、容器Vの中に、膵細胞の担体としてスポンジ状キチンを充填した人工臓器用チャンバーも知られている(特許文献2参照)。
 一方、人工臓器に含まれる細胞は人工臓器とは別の培養装置で培養し、人工臓器のチャンバーに移している。培養装置で培養した一般的な細胞を、別の場所に自動搬送する装置としては、細胞懸濁物を解析容器に自動的に移す装置が知られている(特許文献3参照)。

 上記のとおり、細胞を入れたチャンバーを生体内に留置することは知られている。しかしながら、上記特許文献1及び2に記載されている発明は、培養装置で培養した細胞を単にチャンバー内に纏めて投入しているに過ぎない。人工臓器として実際に使用するためには、生体内に人工臓器を留置することで、当該人工臓器が所期の物質をどの程度生産するのか予測する必要があるが、前記特許文献1及び2に記載されている発明では、生産量が予測できないという問題がある。
 最近、ES細胞やiPS細胞等の幹細胞を用いた研究が盛んにおこなわれている。所期の器官に分化した後の幹細胞は、その大きさから生産するホルモン等の量を予測できることから、上記問題点を解決することが期待されている。そのため、本発明者らは、分化した個々の幹細胞を挿入・保持するための凹部を形成した人工臓器用のセルチャンバーの開発を進めている。
 ところで、幹細胞を所期の器官に分化するまでは、幹細胞専用の培養装置を用いて培養する。そのため、培養した幹細胞を培養装置から人工臓器用のセルチャンバーに搬送する手段が必要となる。しかしながら、上記特許文献3等に記載されている細胞搬送装置は、一定量の細胞懸濁液を単に搬送するための装置である。一方、分化した幹細胞は大きさが1mm~3mm程度になる。更に、セルチャンバーには可能な限り多くの幹細胞を挿入・保持したいことから、幹細胞を挿入・保持するための凹部の大きさは、幹細胞とほぼ同じ大きさにすることが望ましい。そのため、培養装置からの幹細胞のサンプリング、該サンプリングした幹細胞をセルチャンバーに挿入するためには、細胞を吸引するシリンジ等を正確に位置合わせする必要があるが、特許文献3等に記載されている従来からの細胞搬送装置では、正確な位置合わせをするのは困難である。
 本発明は、上記従来の問題を解決するためになされた発明であり、鋭意研究を行ったところ、撮像部で細胞を撮像し、撮像した細胞の画像から算出した細胞の位置、及び撮像部で撮像したセルチャンバーの凹部の画像から算出した凹部の位置を記憶する記憶部を設け、記憶部に記憶された細胞及び凹部の位置に基づき、細胞を吸引するシリンジポンプを駆動することで、細胞を正確に搬送できること、そして、当該細胞搬送装置及び細胞搬送方法を用いることで、人工臓器を自動で製造できること、を新たに見出し、本発明を完成した。
 すなわち、本発明の目的は、細胞搬送装置、細胞搬送方法、及び人工臓器の製造方法を提供することである。
(1)セルチャンバーに設けられた凹部に、培養した細胞を詰めるための細胞搬送装置であって、
 前記細胞搬送装置は、撮像部、細胞を吸引するシリンジを装着できるシリンジポンプ、シリンジポンプ駆動部、前記シリンジポンプ駆動部を制御するための制御部を含み、
 前記制御部は、前記撮像部で撮像した細胞の画像から算出した細胞の位置、及び前記撮像部で撮像した前記凹部の画像から算出した凹部の位置を記憶する記憶部を含み、前記制御部が、前記記憶部に記憶された細胞及び凹部の位置に基づき前記シリンジポンプ駆動部を制御する、細胞搬送装置。
(2)前記シリンジポンプに装着するシリンジを更に含み、前記シリンジの細胞を吸引する開口部が直径1mm~3mmの略円形状である、上記(1)に記載の細胞搬送装置。
(3)前記撮像部で撮像した細胞の画像を記憶する画像記憶部を更に含む、上記(1)又は(2)に記載の細胞搬送装置。
(4)前記画像記憶部が、前記細胞の画像と当該細胞を詰めた凹部の位置と関連付けて記憶する、上記(3)に記載の細胞搬送装置。
(5)前記撮像部で撮像した細胞の形状を基準値と比較して、前記凹部に詰めるべき細胞か否か判別する細胞判別部を更に含む、上記(1)~(4)の何れか一に記載の細胞搬送装置。
(6)前記撮像部で撮像した細胞の形状から、細胞が生産する物質の生産量を推定する生産量推定部を含む、上記(1)~(5)の何れか一に記載の細胞搬送装置。
(7)少なくとも1種以上の細胞、及びセルチャンバーを準備する工程、
 撮像部により、前記細胞を撮像する工程、
 撮像部により、セルチャンバーに設けられた凹部を撮像する工程、
 撮像した細胞及び凹部の画像から細胞及び凹部の位置を算出し、該位置を記憶部に記憶する工程、
 前記記憶部に記憶された細胞の位置に基づき、細胞を吸引するためのシリンジを前記細胞の近傍に位置させ細胞を吸引する細胞吸引工程、
 前記記憶部に記憶された凹部の位置に基づき、吸引した細胞を詰める前記凹部の近傍に吸引した細胞を搬送し、該凹部に細胞を詰める細胞搬送工程、
を少なくとも含む、細胞搬送方法。
(8)上記(7)に記載の細胞搬送方法を用いた人工臓器の製造方法であって、
 前記人工臓器の製造方法は、細胞としてiPS細胞を用い、
 前記細胞搬送工程で細胞を詰める際に、基板、該基板を貫通する貫通孔、前記基板上に形成された凹部を含むセルチャンバーを用い、
 前記細胞搬送工程の後に、半透膜取付工程を含む、
人工臓器の製造方法。
 本発明の細胞搬送装置及び細胞搬送方法は、サンプルすべき細胞の位置と細胞を挿入するセルチャンバーの凹部の位置を記憶し、記憶した位置に基づきシリンジポンプを駆動している。したがって、シリンジの先端を細胞及び凹部の近傍に移動できるので、幹細胞でも正確に搬送することができる。
図1は、本発明の細胞搬送装置1の概略を示す図である。 図2(A)は、本発明の人工臓器の製造方法に用いられるセルチャンバー6の概略を説明する斜視図で、図2(B)は、チャンバーの上面図である。 図3は、本発明のセルチャンバー6の他の実施形態を説明するための断面図である。 図4は、本発明の細胞搬送装置1を用いた人工臓器の製造方法の手順を示すフローチャートである。
 以下に、本発明の細胞搬送装置、細胞搬送方法、及び人工臓器の製造方法について詳しく説明する。
 図1は、本発明の細胞搬送装置1の概略を示す図である。細胞搬送装置1は、撮像部2、シリンジを先端に装着できるシリンジポンプ3、シリンジポンプ3を駆動するためのシリンジポンプ駆動部4、シリンジポンプ駆動部4を制御するための図示していない制御部、細胞及び細胞の搬送先の位置を記憶する図示しない記憶部、を少なくとも含んでいる。
 本発明の細胞搬送装置1の記憶部は、細胞を培養したプレート5を撮像部2で撮像することでプレート5上の個々の細胞の位置を記憶部に記憶し、また、細胞の搬送先であるセルチャンバー6に形成されている凹部61の個々の位置も記憶部に記憶している。そして、制御部は記憶部に記憶されている細胞と凹部61の位置に基づき、シリンジポンプ駆動部4の駆動を制御することから、シリンジポンプ3に装着するシリンジを、細胞及び凹部61の近傍に正確に移動することができる。
 撮像部2は、可視画像が撮像できれば特に制限はなく、CMOSカメラ等の市販のカメラを用いればよい。なお、撮像した細胞の画像を解析し易くするために、細胞搬送装置1にプレート5を載置する載置台7を設け、載置台7の内部に図示しない照明部を設けてもよい。
 また、撮像部2は、細胞の撮像用と凹部61の撮像用とを別々に設けてもよいが、単一の撮像部2で細胞と凹部61を撮像できるようにしてもよい。撮像部2が単一の場合は、撮像部2をX軸及びY軸方向に移動できる移動機構を設ければよい。移動機構は、例えば、駆動原としてパルスモーター、超音波モーター等を用い、駆動源の駆動力を伝達する駆動力伝達機構を設ければよい。駆動力伝達機構は、例えば、倒立顕微鏡等に使われている試料載置台を水平方向に駆動するための駆動力伝達機構等、公知のものを用いればよい。また、移動機構を設けずに、後述するシリンジポンプ駆動部4に撮像部2を取付け、シリンジポンプ3と撮像部2の移動機構を共通化しても良い。
 シリンジポンプ3は、装着するシリンジで細胞をサンプリングできれば特に制限はなく、自動分析装置等の分野で公知のシリンジポンプを用いればよい。また、幹細胞をサンプリングするためのシリンジは、サンプルを定量サンプリングするためのシリンジと異なり、直径が1mm~3mmのほぼ球形(まりも状)の幹細胞の塊をプレート5から取り出して、凹部61に搬送するためのものである。そのため、シリンジの内径は1mm~3mmの略円形状とすることが好ましい。なお、本発明の細胞搬送装置1は、記憶部を設けることで、従来の自動分析装置よりより正確に細胞等の搬送をすることができる。したがって、本発明の細胞搬送装置1は、3次元培養方法等で培養した細胞等、幹細胞以外の搬送にも用いることが出来る。その場合、搬送する対象に適したシリンジを用いればよい。
 シリンジポンプ3を駆動するためのシリンジポンプ駆動部4は、シリンジポンプ3をX軸、Y軸、Z軸方向に移動することができ、所定の位置に移動した後にシリンジポンプ3を駆動して細胞をプレート5から取り出し、凹部61に搬送できれば特に制限は無い。X軸、Y軸、Z軸方向への移動機構は、例えば、図示しない駆動原及び該駆動原の駆動力をシリンジポンプ3に伝達する伝達する駆動力伝達機構を設ければよい。駆動原としては、例えば、パルスモーター、超音波モーター等を用いればよい。また、駆動力伝達機構は、例えば、自動分析装置等の分野で公知の3次元駆動機構を用いればよい。また、シリンジポンプ3の駆動は、シリンジポンプ3が設定した場所に到達した後、吸引・排出等を行うように制御すればよい。なお、上記には、シリンジポンプ3をX軸、Y軸、Z軸方向に移動する例を示しているが、シリンジポンプ3が伸縮自在のシリンジ部を備える場合は、シリンジポンプ駆動部4は、シリンジポンプ3をX軸及びY軸方向に移動できればよい。
 駆動力伝達機構からの駆動力は、直接シリンジポンプ3に伝達してもよいが、図示しないシリンジポンプ3の取付部を設け、取付部が移動するように駆動力を伝達してもよい。また、取付部には撮像部2を取り付けてもよい。撮像部2を取付部に取り付けることで、撮像部2を移動する為の機構を別途設ける必要はない。また、駆動力伝達機構は、X軸とY軸の水平方向への駆動力伝達機構とZ軸方向への駆動力伝達機構を別々に設けてもよい。シリンジポンプ3には、X軸、Y軸及びZ軸の3方向の駆動力が伝達するようにし、撮像部2にはX軸及びY軸の水平方向の駆動力が伝達するようにしておいても良い。撮像部2がズーム機構を設けている場合は、撮像部2自体をZ軸方向に移動しなくてもよい。
 記憶部には、撮像部2で撮像した細胞の画像及び凹部61の画像から解析した細胞及び凹部61の位置を記憶している。上記のとおり、幹細胞は1mm~3mmの球形の塊であるが、シリンジの先端部分で塊を押圧してしまうと、幹細胞の塊を壊してしまう可能性がある。そのため、従来の自動分析装置を用いた分注とは異なり、球形の幹細胞の塊を壊さずにシリンジで取り出し、凹部61に搬送する必要があることから、シリンジの先端の開口部を幹細胞のほぼ真上に位置させ、次いで、凹部61のほぼ真上にシリンジの先端の開口部を位置させる必要がある。
 細胞及び凹部61の位置は、例えば、撮像部2を駆動する際の基準位置を決めておき、また、撮像部2で撮像した細胞画像の輪郭から細胞の中心点を算出し、基準位置から細胞の中心点までのX、Y方向の距離で定義すればよい。細胞の中心点は、例えば、細胞画像の輪郭上の任意の2点を結んだ線の中で最も長い線の中間点とすればよい。凹部61の位置も、細胞と同様に決めておけばよい。なお、上記は単なる例示に過ぎず、中心点、位置は他の方法で決めても良い。
 細胞及び凹部61の位置は、撮像部2でプレート5全体の画像及び凹部61全体の画像を撮像し、1枚の画像から位置を算出してもよいし、プレート5及び凹部61をブロック単位に分けて撮像し位置を算出してもよい。
 そして、制御部は、シリンジポンプ駆動部4を次のように制御する。
(1)記憶部に記憶された細胞の位置に基づき、シリンジポンプ3に装着したシリンジの開口部の中心点が細胞の中心点と一致するまで、シリンジポンプ3をX軸及びY軸方向に移動する。
(2)プレート5の細胞を吸引できるように、Z軸方向にシリンジポンプ3を移動する。
(3)シリンジポンプ3を駆動して、シリンジで細胞を吸引する。
(4)シリンジポンプ3をZ軸方向に移動し、次いで、記憶部に記憶された凹部61の位置に基づき、シリンジポンプ3に装着したシリンジの開口部の中心点が凹部61の中心点と一致するように、X軸及びY軸方向に移動する。
(5)シリンジの先端を凹部61にほぼ当接するまで、Z軸方向にシリンジポンプを移動する。
(6)シリンジポンプ3を駆動して、細胞を凹部61に入れる。
(7)上記(1)~(6)の手順を、所期の数の細胞を搬送するまで繰り返す。
 なお、上記に記載した例では、シリンジポンプ3自体を移動しているが、シリンジポンプ3の本体は移動せず、シリンジポンプ3の本体に接続し且つ先端にシリンジを取り付け可能なチューブを移動してもよい。したがって、本発明において、「シリンジポンプ駆動部」は、シリンジポンプ3自体を移動させること、又は、シリンジポンプ3の一部を移動させ、細胞を吸引・排出することを意味する。
 本発明の細胞搬送装置1は、撮像部2で撮像した画像を記憶しておく画像記憶部を含んでもよい。幹細胞としてiPS細胞を用いて人工臓器を作製する場合、iPS細胞ががん化する可能性がある。人工臓器を製造する際に、セルチャンバーに搬送したiPS細胞の画像を記憶しておくことで、後日、がん化した人工臓器があった場合には、画像を解析することで、がん化し易いiPS細胞の分析用のデータとすることができる。
 がん化したiPS細胞をより詳細に分析するためには、人工臓器単位で画像を記憶しておくのではなく、人工臓器用のセルチャンバーの個々の凹部61の位置と当該凹部61に詰めたiPS細胞の画像を関連付けて画像記憶部に記憶しても良い。がん化した人工臓器を生体内から取り出し、人工臓器を凍結して切片を作製し分析することで、どのiPS細胞ががん化したのか分かる。したがって、がん化したiPS細胞の製造時の画像を特定できることから、がん化し易いiPS細胞のより正確な分析ができる。凹部61の位置は、例えばセルチャンバーに基準となる印をつけておき、当該印との位置関係で特定すればよい。
 また、人工臓器を製造する場合、一つの人工臓器が生産するホルモン等の生産量が多いほど、人工臓器を小さくすることができる。また、複数個の人工臓器を生体内に留置することが必要な場合は、一つの人工臓器が生産するホルモン等の生産量が多いほど、留置する人工臓器の数を減らすことができる。そして、幹細胞が生産するホルモン等の量は、幹細胞が大きいほど多いことが知られている。したがって、人工臓器用のセルチャンバー内に詰める幹細胞は、所定値より大きい幹細胞であることが望ましい。
 そのため、本発明の細胞搬送装置1は、撮像部2で撮像した細胞の形状を基準値と比較して、凹部に詰めるべき細胞か否か判別する細胞判別部を更に含んでいてもよい。幹細胞の形状は主に球形であることから、細胞判別部は、例えば、幹細胞画像の輪郭上の任意の2点を結んだ線の中で最も長い線を細胞の大きさとし、そして、大きさが1mm以上、2mm以上、或いは2.5mm以上等の基準値を決めておき、基準値より大きな幹細胞のみがセルチャンバーに搬送すべき細胞であると判別し、記憶部に位置を記憶すればよい。制御部は、記憶部に記憶された判別済みの幹細胞の位置に基づき、基準値以上の幹細胞をセルチャンバーに搬送すればよい。
 また、上記のとおり、幹細胞が生産するホルモン等の量は、幹細胞が大きいほど多いことが知られている。したがって、本発明の細胞搬送装置1は、撮像部2で撮像した細胞の形状から、幹細胞が生産するホルモン等の物質の生産量を推定する生産量推定部を含んでいても良い。例えば、セルチャンバーに詰めた幹細胞の形状から推定したホルモン等の生産量を表示すればよい。又は、生産量推定部は、人工臓器に求める所期のホルモン等の生産量をインプットし、撮像した幹細胞の画像に基づき、インプットした生産量を満たす幹細胞を選択しても良い。その場合、生産量推定部が選択した細胞の位置を記憶部に記憶し、制御部は記憶部に記憶された位置に基づき、幹細胞をセルチャンバーの凹部61に搬送すればよい。
 図2(A)は、本発明の人工臓器の製造方法に用いられるセルチャンバー6の概略を説明する斜視図で、図2(B)は、セルチャンバー6の上面図である。本発明のセルチャンバー6は、基板12、基板12を貫通する貫通孔13、基板12上に形成された凹部61を少なくとも含んでいる。
 図2(A)及び(B)に示す実施形態では、基板12は、台座部21、台座部21の一方の面上に形成された幹細胞収納部22を含んでいるが、段差を設けない平面状の基板であってもよい。セルチャンバー6が台座部21及び幹細胞収納部22を含む場合、台座部21には、セルチャンバー6を生体内に留置する際に体内組織に固定する手段、例えば、糸等を挿通するための挿通孔23を形成してもよい。なお、挿通孔23は必須ではなく、生体組織に固定する際に、針で台座部21(台座部21を設けない場合は基板12)を突き刺して貫通させればよい。
 幹細胞収納部22は、台座部21の一方の面上に凸状に形成されており、台座部21から上方に伸びた壁面221及び幹細胞収納面222を含む、略台形状の断面をしている。幹細胞収納部22を形成する場合、貫通孔13及び凹部61は細胞収納部22に形成されることが好ましい。
 基板12は、生体内に留置した際に、拒絶反応が起き難い材料であれば特に制限はなく、例えば、シリコン、ポリプロピレン等が挙げられる。基材12は、モールドを形成して射出成形又はプレス成形、或いは、切削加工等により形成すればよい。また、セルチャンバー6は、生体内に留置して用いることから、セルチャンバー6が当接する生体組織に損傷を与えないことが望ましい。そのため、基板12(台座部21及び幹細胞収納部22)の周囲は角が無い滑らかな形状にすることが望ましい。また、基板12の形状も円形、楕円形等、角が無いように形成することが望ましい。
 貫通孔13は、生体内にセルチャンバー6を留置した際に、生体組織から血管が成長して凹部61に挿入した幹細胞に作用するための孔である。したがって、貫通孔13の大きさは、血管が形成されるサイズであれば特に制限はない。また、貫通孔13を設ける位置、個数は特に制限はないが、設ける凹部61の個数に対し貫通孔13が少なすぎると、生体組織から成長した血管が到達できない凹部61が出る可能性があり、幹細胞が生産したホルモン等の物質を体内に運ぶことができない可能性がある。したがって、貫通孔13は、凹部61の個数及び配置に応じて分散して配置すればよい。
 凹部61は、幹細胞を挿入・保持できれば特に制限はない。なお、本発明のチャンバーを利用した人工臓器は生体内に留置することから生体にとっては異物となる。したがって、一つの人工臓器が生産する物質を可能な限り多くする、つまり、物質の生産量が同じであれば人工臓器は小さいほど好ましく、例えば、凹部61は細密充填構造に形成すればよい。
 凹部61に挿入・保持する幹細胞は、ES細胞又はiPS細胞の何れも可能である。ES細胞、iPS細胞は、公知の方法により培養・分化した細胞を用いればよい。また、一つのチャンバー1に、同じ種類の幹細胞を挿入してもよいし、異なる種類の幹細胞を挿入してもよい。
 なお、人工臓器として所期の特性を得るためには、幹細胞を分化するまで培養した細胞を用いることが好ましい。分化した幹細胞の大きさは、約1~3mmの略球形である。したがって、凹部61の開口部611の形状としては直径1mm~3mmの略球形の幹細胞がほぼ通過できる大きさとすることが望ましく、例えば、直径1mm~3mm、好ましくは直径2mm~3mmの略円形状が挙げられる。また、略円形状に代え、6角形、7角形、8角形等の多角形でもよい。多角形の場合、開口部41の大きさは、中心を通る対角線の長さが1mm~3.5mm、好ましくは2.5mm~3.5mm程度にすればよい。
 また、凹部61の底部612は連続した曲面にすることが好ましい。幹細胞、特にiPS細胞は平らな面に置くと別の細胞に変化することがある。したがって、凹部61の底部612を連続した曲面形状とすると、平面部を含まないことから幹細胞が球形の状態を保持し易くなるので好ましい。連続した曲面形状の中でも、略半球状がより好ましい。また、必要に応じて、凹部61の内面を撥水処理してもよい。撥水処理は、公知の方法であれば特に制限はなく、例えば、プラズマ装置等を用いればよい。凹部61の内面を撥水処理することで、凹部61に挿入した幹細胞をより球形状にし易くなる。
 セルチャンバー6を人工臓器として用いる場合、凹部61に幹細胞を挿入・保持した状態で生体内に留置すると、幹細胞が生体内に流出する恐れがある。ES細胞が体内に流出すると拒絶反応を起こす可能性があり、また、iPS細胞が体内に流入するとがん化する恐れがある。そのため、セルチャンバー6の少なくとも幹細胞を入れた凹部61の上部には、幹細胞は通過しないが、生産したホルモン等の物質を通過できる又は毛細血管が入り込める半透膜を取付けることが好ましい。なお、セルチャンバー6に後述するカバー部材を取付ける場合は、台座部21の下側に半透膜を取り付けてもよい。
 半透膜は、前記特性を満たせば特に制限はない。半透膜を取付けたセルチャンバー6を生体内に留置すると、留置した周りの生体組織から毛細血管が成長し、毛細血管が半透膜を通過して幹細胞と一体となり組織化することができる。また、毛細血管は半透膜を通過しなくても、半透膜付近まで成長すれば、半透膜を通過したホルモン等の物質を取り込むことができる。
 人工臓器を作製する際、凹部61には幹細胞の培養液ではなく、人工髄液を充填することが好ましい。また、セルチャンバー6は、単独で用いてもよいが、凹部61に幹細胞を挿入後に、複数積層してもよい。
 図3は、本発明のセルチャンバー6の他の実施形態を説明するための断面図で、図2に示す実施形態のセルチャンバー6にカバー部材15を取付けている。人工臓器は、頭皮の下の帽状腱膜、腹腔等、様々な場所に留置することが可能であるが、留置する場所によっては、毛細血管が成長して幹細胞と相互作用するまでに時間を要する場合がある。毛細血管が成長して相互作用するまでに、凹部61に充填した人工髄液がなくなると、幹細胞が死滅する恐れがある。図3に示す実施形態のセルチャンバー6は、基板12の上にカバー部材15を設けることで、体外から注射器の針を刺して基板12とカバー部材15の間に人工髄液を補充することができる。したがって、毛細血管が成長し難い場所に人工臓器を留置した場合でも、人工臓器として機能を開始するまで幹細胞を生存した状態に保つことができる。
 カバー部材15は、基板12と同様、ポリプロピレン、シリコン等を用いて作製すればよい。図3に示す実施形態では、カバー部材15は台座部21と幹細胞収納部22の境界付近に形成した溝部23とカバー部材15の端部51を嵌合しているが、カバー部材15を基板12に取付けることができれば特に制限はない。また、カバー部材15は、生体内に留置した際に周りの生体組織に損傷を与えないために、角のない滑らかな形状にすることが好ましい。
 また、人工臓器を生体内に留置した場合、針で人工髄液を注入するためには、生体の外から人工臓器の位置を把握する必要がある。そのため、カバー部材15の一部に凸部52を形成してもよい。皮膚の上から手で触わり凸部を確認することで、針で注射すべき位置を把握することができる。
 カバー部材15を、シリコン等の柔軟性のある材料で形成すれば、注入した人工髄液はカバー部材15と幹細胞収納部22の間に充填されることから、半透膜を介して各凹部61に人工髄液を供給することができる。なお、必要に応じて、幹細胞収納部22とカバー部材15の間に積極的に空間を設け、より多くの人工髄液を注入できるようにしてもよい。具体的には、図2(A)に示すように、台座部21から上方に伸びた壁面221の頂部から貫通孔13や凹部61を設ける幹細胞収納面222を形成するのではなく、頂部から台座部21方向に段差223を設け、段差223の台座部21側の端部224を結ぶように幹細胞収納面22を形成すればよい。段差222を設けることで、幹細胞収納面222が壁面221の頂部を結んだ面より台座部21側に位置するので、幹細胞収納面222と壁面221の頂部との間で空間を形成し、より多くの人工髄液を注入することができる。
 iPS細胞を用いて人工臓器を作製した場合、iPS細胞ががん化する可能性がある。人工臓器には半透膜が設けられているので、がん化したiPS細胞が生体内に入ることはないが、安全を考えると、所定期間毎に人工臓器内の溶液をサンプリングし、iPS細胞のがん化の有無を調べることが好ましい。がん化は、サンプリングした溶液に含まれるがんマーカータンパク質や遺伝子を測定する等、公知の方法で検査すればよい。がん化した人工臓器は、手術により生体内から取り出せばよい。
 溶液のサンプリングは、人工髄液の注入と同様、生体外から針を突き刺してサンプリングすればよい。なお、人工臓器を生体内に留置した後所定期間が経過すると、毛細血管等が貫通孔13を通過し、凹部61の周囲並びに凹部61内に成長する。溶液をサンプリングする際に、成長した毛細血管を針で損傷すると出血し、その血が周囲のiPS細胞に炎症を起こす可能性がある。したがって、針を刺す場所にはiPS細胞が無いことが好ましく、例えば、凸部52のすぐ下の凹部61にはiPS細胞を挿入せず、空にしておけばよい。また、幹細胞収納面222の全面に半透膜を取付けると、針を刺すたびに半透膜を損傷して破片等が発生する可能性がある。そのため、半透膜を、iPS細胞を挿入しない凹部61はカバーしないような形状にしてもよい。
 図4は、本発明の細胞搬送装置1を用いた人工臓器の製造方法の手順を示すフローチャートを示している。図4に示す例では、細胞として分化済みの幹細胞を用いた例を示しているが、3次元培養した細胞等を搬送する場合は、半透膜を取付けるS150を省略してもよい。人工臓器の製造方法は、少なくとも以下のステップを含んでいる。
 先ず、S100において、少なくとも1種以上の幹細胞を準備する。一つのセルチャンバーの中に異なる種類の幹細胞を詰める場合は、複数の幹細胞を準備すればよい。また、幹細胞を詰めるセルチャンバーも準備する。
 次に、S110において、準備した幹細胞及びセルチャンバーに設けられた凹部を撮像部2で撮像する。
 次に、S120において、撮像した細胞画像及び凹部の画像から、細胞画像の中心点及び凹部の中心点を算出し、細胞及び凹部の中心点の位置を記憶部に記憶する。なお、撮像した細胞画像から中心点を算出する際に、細胞の大きさが基準値を満たすか否か判別し、基準値以下の細胞は記憶部に位置を記憶せず、サンプルしないようにしてもよい。また、幹細胞の大きさから生産するホルモン等の物質の量を推定し、所期の推定値となる幹細胞の組み合わせを選択し、選択した幹細胞の位置のみを記憶部に記憶しても良い。
 次に、S130において、記憶部に記憶した幹細胞の位置に基づき、幹細胞を吸引するためのシリンジを幹細胞の近傍に位置させ幹細胞を吸引する。
 次に、S140において、記憶部に記憶した凹部61の位置に基づき、幹細胞を詰める凹部の近傍に幹細胞を吸引したシリンジを位置させ、凹部61に幹細胞を詰める。
 次に、S150において、幹細胞を詰めた凹部の上に半透膜を取付ける。
 本発明の細胞搬送装置及び細胞搬送方法を用いると、幹細胞を培養装置から人口臓器用のセルチャンバーに正確に搬送することができるので、人工臓器の製造方法として使用することができる。したがって、人工臓器を製造する医療機器産業や医療機関における手術に有用である。
 

Claims (8)

  1.  セルチャンバーに設けられた凹部に、培養した細胞を詰めるための細胞搬送装置であって、
     前記細胞搬送装置は、撮像部、細胞を吸引するシリンジを装着できるシリンジポンプ、シリンジポンプ駆動部、前記シリンジポンプ駆動部を制御するための制御部を含み、
     前記制御部は、前記撮像部で撮像した細胞の画像から算出した細胞の位置、及び前記撮像部で撮像した前記凹部の画像から算出した凹部の位置を記憶する記憶部を含み、前記制御部が、前記記憶部に記憶された細胞及び凹部の位置に基づき前記シリンジポンプ駆動部を制御する、細胞搬送装置。
  2.  前記シリンジポンプに装着するシリンジを更に含み、前記シリンジの細胞を吸引する開口部が直径1mm~3mmの略円形状である、請求項1に記載の細胞搬送装置。
  3.  前記撮像部で撮像した細胞の画像を記憶する画像記憶部を更に含む、請求項1又は2に記載の細胞搬送装置。
  4.  前記画像記憶部が、前記細胞の画像と当該細胞を詰めた凹部の位置と関連付けて記憶する、請求項3に記載の細胞搬送装置。
  5.  前記撮像部で撮像した細胞の形状を基準値と比較して、前記凹部に詰めるべき細胞か否か判別する細胞判別部を更に含む、請求項1~4の何れか一項に記載の細胞搬送装置。
  6.  前記撮像部で撮像した細胞の形状から、細胞が生産する物質の生産量を推定する生産量推定部を含む、請求項1~5の何れか一項に記載の細胞搬送装置。
  7.  少なくとも1種以上の細胞、及びセルチャンバーを準備する工程、
     撮像部により、前記細胞を撮像する工程、
     撮像部により、セルチャンバーに設けられた凹部を撮像する工程、
     撮像した細胞及び凹部の画像から細胞及び凹部の位置を算出し、該位置を記憶部に記憶する工程、
     前記記憶部に記憶された細胞の位置に基づき、細胞を吸引するためのシリンジを前記細胞の近傍に位置させ細胞を吸引する細胞吸引工程、
     前記記憶部に記憶された凹部の位置に基づき、吸引した細胞を詰める前記凹部の近傍に吸引した細胞を搬送し、該凹部に細胞を詰める細胞搬送工程、
    を少なくとも含む、細胞搬送方法。
  8.  請求項7に記載の細胞搬送方法を用いた人工臓器の製造方法であって、
     前記人工臓器の製造方法は、細胞としてiPS細胞を用い、
     前記細胞搬送工程で細胞を詰める際に、基板、該基板を貫通する貫通孔、前記基板上に形成された凹部を含むセルチャンバーを用い、
     前記細胞搬送工程の後に、半透膜取付工程を含む、
    人工臓器の製造方法。
