Abstract
본 발명에 따른 소브레롤 또는 이의 약학적으로 허용 가능한 염은 근원세포의 분화를 촉진하여 근관을 형성할 수 있으므로 근육 약화를 방지할 뿐만 아니라, 효과적으로 근육 기능을 개선할 수 있다. 따라서 이를 포함하는 약학 조성물은 근육 약화 관련 질환의 예방 또는 치료에 유용하게 사용될 수 있다.

본 발명은 소브레롤(sobrerol) 또는 이의 약학적으로 허용 가능한 염을 포함하는 근원세포(myoblast)의 분화 촉진용 조성물, 근력 약화 관련 질환의 예방 또는 치료용 약학 조성물, 근력 약화 관련 질환의 예방 또는 개선용 건강기능식품 조성물, 근력 강화용 조성물 및 근력 강화용 사료 또는 사료 첨가제 및, 소브레롤(Sobrerol) 또는 이의 약학적으로 허용가능한 염을 포함하는 조성물을 개체에 투여하는 단계를 포함하는 근력 약화 관련 질환의 치료방법, 소브레롤(Sobrerol) 또는 이의 약학적으로 허용가능한 염을 포함하는 조성물을 개체에 투여하는 단계를 포함하는 근력 강화방법, 근력 약화에 의한 주름의 개선용 조성물, 근력 약화에 의한 주름의 개선방법에 관한 것이다.
또한, 본 발명은 소브레롤(sobrerol) 또는 이의 약학적으로 허용 가능한 염을 이용한 근원세포의 분화 촉진 방법, 및 분화된 근원세포의 제조방법에 관한 것이다.
근력의 약화를 유발하는 질환은 노화와 함께 진행되는 근감소증(sarcopenia), 단백질 대사의 불균형이나 근육사용 감소에서 유발되는 근위축증(muscle atrophy), 기아, 소모성질환(암 등), 노화와 함께 진행되는 심위축증(acardiotrophy) 등이 있다.
근감소증(sarcopenia)은 노화가 진행되는 동안 근육량(skeletal muscle mass) 감소에 따른 근력의 저하를 일컫는다. 근감소증의 가장 큰 특징인 근육량의 감소뿐만이 아니라, 근섬유의 종류 변화도 관찰된다. 나이가 들어가면 타입 1과 타입 2가 비슷한 비율로 감소하는데 반해, 근감소증이 오면 타입 2의 근섬유 두께에는 큰 변화가 없지만 타입 1 근섬유 두께는 눈에 띄게 감소한다. 이러한 근감소증은 노인들 사이에서 일어나는 노쇠와 기능 장애를 유발한다고 보고되고 있다(Roubenoff R., Can. J. Appl. Physiol. 26, 78-89, 2001).
근감소증은 다양한 요인에 의해 유발되나, 각각의 요인들에 대한 연구는 아직 미진하다. 성장 호르몬의 감소 또는 신경학적 변화(neurological change), 생리 활성(physical activity)의 변화, 대사의 변화, 성호르몬의 양 또는 지방이나 카타볼릭 싸이토카인(catabolic cytokines)의 증가와 단백질의 합성과 분화의 균형 변화에 의해 유도된다(Roubenoff R. and Hughes V.A., J. Gerontol. A. Biol. Sci. Med. Sci. 55, M716-M724, 2000). 근감소증의 가장 큰 특징인 근육량 감소의 원인으로는 위성 세포의 활성 (satellite cell activation) 감소가 중요한 원인으로 손꼽힌다. 위성 세포란, 기저막(basement membrane)과 근섬유의 근(sarcolemma) 사이에 위치하고 있는 작은 단핵 세포이다. 이들은 부상 또는 운동과 같은 자극에 의해 활성화되어 근원세포(myoblast)로 증식하며, 분화가 진행되면 다른 세포와 융합되어 다핵의 근섬유를 형성한다. 따라서 위성 세포의 활성이 감소함에 따라 손상된 근육을 재생하는 능력이나 분화 신호에 대한 반응이 떨어지게 되고 그 결과 근육 형성이 저하된다.
근위축증(Muscle atrophy)은 영양결핍이나 장기간 근육을 사용하지 않은 경우에 유발되는데 정상적인 단백질의 합성과 분해의 균형이 붕괴되어 단백질이 분해됨으로서 나타나게 된다.
한편, 심위축증(acardiotrophy)은 기아, 소모성질환(암 등), 노쇠했을 때 유발되는데 심근섬유는 마르고 가늘어 지고 핵은 농축되어 대소부동이 된다. 따라서 근속(muscle fascicle)도 용적이 줄고 심장 전체가 작아지며 심외막하의 지방조직은 뚜렷하게 감소하고 관상동맥은 굽어진다. 심근섬유의 핵 양단에 갈색의 색소로서 소모성 색소(리포푸스친)가 나타나고 지방조직의 감소와 함께 심장 전체가 갈색조를 나타낸다.
근감소증의 치료방법으로는 크게 3가지를 들 수가 있다. 첫 번째는 운동이다. 운동은 단기적으로 골격근의 단백질 합성 능력을 증가시키며, 노인들의 근육의 힘이나 운동성을 증가시킨다고 보고되고 있다. 그러나 장기적 치료방법에 부적절하다(Timothy J. Doherty, J. Appl. Physiol. 95, 1717-1727, 2003). 두 번째는 약물 치료로서 테스토스테론(Testosterone) 또는 아나볼릭 스테로이드(anabolic steroid)의 사용이 가능하나 이는 여성에게는 남성화를 유도하며, 남성의 경우 전립선 증상(prostate symptoms) 등 부작용을 나타낸다. 다른 승인된 처방법으로 DHEA(dehydroepiandrosterone) 와 성장 호르몬이 있는데 SARMs(Selective Androgen Receptor Modulators)을 포함하는 부위에서 치료법으로 가능하다는 연구가 보고된바 있다(D.D. Thompson, J. Musculoskelet Neuronal Interact 7, 344-345, 2007). 또한 식이요법이 치료법으로 알려져 있지만 영양평가에 의하면 영양실조나, 현대 식습관은 적당한 총체질량(total body mass)을 유지하기 위해 부적절하다.
최근에는 위성 세포를 분리하여 체외에서 분화시킨 후 체내에 도입시키는 줄기세포치료법(stem cell therapy)과 직접 체내의 위성 세포를 활성화하여 근육 분화(myogenesis)를 촉진시켜 근육을 유지하거나 강화시키는 방법이 근감소증과 같은 근력 약화를 치료할 수 있는 방법으로 대두되고 있다 (Shihuan Kuang, and Michael A. Rudnicki, Trends in Molecular Medicine 14, 82-31, 2008).
다만, 근육의 근력 약화 관련 질환을 치료하기 위해, 보다 근본적이며 부작용이 없는 치료방법으로 근원세포를 분화하는 방법이 요구되며, 이에 따라 근원세포의 분화를 촉진할 수 있는 물질의 개발이 필요한 실정이다.
본 발명자들은 근원세포의 분화를 촉진함으로써 근육량을 증가시키고, 근육기능을 효과적으로 회복시키는 근육의 근력 약화 관련 질환 치료제를 개발하기 위해 예의 노력한 결과, 소브레롤이 근원세포의 분화를 촉진하여 근력 약화 관련 질환의 예방 또는 치료에 사용될 수 있음을 확인함으로써 본 발명을 완성하였다.
본 발명의 하나의 목적은 근원세포(myoblast)의 분화 촉진용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 근원세포의 분화 촉진 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 분화된 근원세포의 제조방법을 제공하는 것이다.
본 발명의 또 다른 목적은 근육의 근력 약화 관련 질환의 예방 또는 치료용 약학 조성물을 제공하는 것이다.

상기의 목적을 달성하기 위하여, 본 발명은 하나의 양태로서 소브레롤(sobrerol) 또는 이의 약학적으로 허용 가능한 염을 포함하는 근원세포(myoblast)의 분화 촉진용 조성물을 제공한다.
본 발명에서 사용되는 용어 "소브레롤(sobrerol)"은, C10H18O2의 화학식을 갖는 테르펜 알코올의 하나이며, 상기 테르펜(terpene)은 가연성의 불포화 탄화수소로, (C5H8)n의 화학식을 갖는 탄수화물 및 이들의 알코올, 알데히드, 케톤 등의 유도체를 총칭하는 것이다.
본 발명에서 사용되는 용어 "약학적으로 허용 가능한 염"이란, 화합물이 투여되는 유기체에 심각한 자극을 유발하지 않고 화합물의 생물학적 활성과 물성들을 손상시키지 않는 화합물의 제형을 의미한다. 상기 약학적 염은, 약학적으로 허용되는 음이온을 함유하는 무독성 산부가염을 형성하는 산, 예를 들어, 염산, 황산, 질산, 인산, 브롬화수소산, 요오드화수소산 등과 같은 무기산, 타타르산, 포름산, 시트르산, 아세트산, 트리클로로아세트산, 트리플로로아세트산, 글루콘산, 벤조산, 락트산, 푸마르산, 말레인산, 살리신산 등과 같은 유기 카본산, 메탄설폰산, 에탄술폰산, 벤젠설폰산, p-톨루엔설폰산 등과 같은 설폰산 등에 의해 형성된 산부가염이 포함될 수 있다. 예를 들어, 약학적으로 허용되는 카르복실산 염에는, 리튬, 나트륨, 칼륨, 칼슘, 마그네슘 등에 의해 형성된 금속염 또는 알칼리 토금속염, 라이신, 아르지닌, 구아니딘 등의 아미노산 염, 디시클로헥실아민, N-메틸-D-글루카민, 트리스(히드록시메틸) 메틸아민, 디에탄올아민, 콜린 및 트리에틸아민 등과 같은 유기염 등이 포함될 수 있다.
본 발명에서 사용되는 용어, "근원세포 분화"란, 단핵인 근원세포(myoblast)가 융합을 통해 다핵의 근관(myotube)를 형성하는 과정이다. 근육 전구체 세포에 해당하는 근원세포는 자기복제(self-renewal) 하는 경우에는 Pax7+ 마커를 나타내며, 증식하는 경우 Pax7+/MyoD+를 나타낸다. 근관을 형성하는 분화단계의 세포는 Pax7-, MHC+, Troponin C+, myogenin+ 마커를 이용하여 구분할 수 있다.
본 발명의 일실시예에서는 미오게닌(myogenin)의 프로모터 활성과 관련하여 소브레롤의 루시퍼라아제 활성을 확인함으로써, 소브레롤의 근육 분화 촉진능이 우수함을 확인하였다(도 2).
구체적으로, 본 발명의 일구현예는 소브레롤의 농도가 0.01 nM 내지 1μM인 것인, 근원세포의 분화 촉진용 조성물에 관한 것이다. 상기 조성물은 혈청이 포함된 DMEM 분화용 배지일 수 있으나, 근원세포의 분화를 촉진할 수 있는 배지 또는 조성물이면 제한없이 포함될 수 있다. 상기 조성물에 있어서, 소브레롤을 0.05 nM 내지 100 nM 의 농도로 포함할 수 있으며, 이에 제한되지 않으나 구체적으로 0.1 nM 내지 10 nM의 농도로 포함할 수 있다. 또한, 상기 조성물은 세포배양 또는 분화에 필요한 부가적인 물질을 더욱 포함할 수 있다.
본 발명에서 상기 조성물은 근원성 조절 인자(myogenic regulatory factor)의 mRNA 또는 이로부터 발현되는 단백질의 수준을 증가시키는 것일 수 있다.
본 발명에서 사용되는 용어 "근원성 조절 인자(myogenic regulatory factor)는 기본 헬릭스-루프-헬릭스(bHLH) 전사 인자로서, 근육의 생성을 조절하는 역할을 하는 인자를 의미한다. 상기 기본 헬릭스-루프-헬릭스는 전사 인자패밀리의 특징으로 나타나는 단백질 구조 모티프(protein structural motif)를 의미하며, 헬릭스-턴-헬릭스와 구분된다.
본 발명의 일구현예는 상기 근원성 조절인자가 미오디(MyoD), 미오게닌(Myogenin), 및 미오신 헤비 체인(Myosin heavy chain, MyHC)으로 구성된 군에서 선택된 어느 하나인 것인, 근원세포의 분화 촉진용 조성물에 관한 것이다.
본 발명의 일실시예에서는, 소브레롤이 미오디(MyoD), 미오게닌(Myogenin), 및 미오신 헤비 체인(Myosin heavy chain, MyHC)의 mRNA을 증가시킬 수 있음을 qRT-PCR을 통해 확인하였다(도 3). 또한, 본발명의 일실시예에서는, 소브레롤이 미오신 헤비 체인의 단백질 발현을 증가시킬 수 있음을 면역 형광 염색법(Immunofluorescence)을 통해 확인하였다(도 4).
한편, 상기 mRNA는 RT-PCR, 정량 실시간 PCR(quantified real time PCR), 경쟁적 RT-PCR(Competitive RT-PCR), 실시간 RT-PCR(real time quantitative RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블럿팅(Northern blotting), DNA 칩 분석법 등의 방법에 의해 측정될 수 있으나, 이에 제한되지 않는다.
또한, 상기 단백질의 수준은 웨스턴 블럿(western blotting), ELISA(enzyme linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역 확산법, 로케트 면역전기영동(rocket immunoelectrophoresis), 면역조직화학염색법(immunohistochemical staining), 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), 면역 형광 염색법(immunofluorescence), 면역크로마토그래피법(immunochromatography), FACS(fluorescenceactivated cell sorter analysis) 및 단백질 칩 분석법(protein chip technology assay) 등의 방법에 의해 측정될 수 있으나, 이에 제한되지 않는다.
본 발명의 다른 하나의 양태로서, 소브레롤 또는 이의 약학적으로 허용 가능한 염을 근원세포에 처리하는 단계를 포함하는, 근원세포의 분화 촉진 방법을 제공한다.
구체적으로, 상기 근원세포는 분리된 근원세포일 수 있다.
본 발명의 일실시예에서는, 소브레롤을 근원세포에 처리하지 않은 군과 소브레롤을 근원세포에 처리한 군의 루시퍼라아제 활성을 비교한 결과, 소브레롤을 처리한 군의 루시퍼라아제 활성이 뛰어남을 확인(도 2)하여, 소브레롤을 근원세포에 처리하는 단계가 근원세포의 분화를 촉진하는데 필수적인 단계임을 확인하였다.
본 발명의 또 다른 하나의 양태로서, 소브레롤 또는 이의 약학적으로 허용가능한 염을 근원세포에 처리하여 근원세포를 분화시키는 단계를 포함하는, 분화된 근원세포의 제조방법을 제공한다.
소브레롤 또는 이의 약학적으로 허용 가능한 염은 상기에서 설명한 바와 같다.
구체적으로, 상기 근원세포는 분리된 근원세포일 수 있다.
본 발명은 또 다른 하나의 양태로서, 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는 근력 약화 관련 질환의 예방 또는 치료용 약학 조성물을 제공한다. 소브레롤의 농도는 상기에서 설명한 바와 같다.
본 발명에서 사용된 용어 "근력 약화"란 한 개 또는 그 이상의 근육의 힘이 감소된 상태를 의미한다. 상기 근력 약화는 어느 한 근육이나, 몸의 한쪽, 상지나 하지 등에 국한될 수도 있고, 전신에 걸쳐 나타날 수도 있다. 또한 근피로나 근육통을 포함하는 주관적인 근력 약화 증상은 의학적 검진을 통해 객관적인 방법으로 정량화될 수 있다.
본 발명에서 근력 약화 관련 질환이란 근력약화로 인해 발생할 수 있는 모든 질환을 의미하며, 예를 들어 근감소증, 근위축증, 근육 퇴행 위축(muscle dystrophy), 또는 심위축증을 들 수 있으나 이에 제한되는 것은 아니다.
따라서, 본 발명의 조성물은 근원세포의 분화 촉진을 통해 근감소증, 근위축증, 근육 퇴행 위축(muscle dystrophy), 또는 심위축증의 예방 또는 치료용으로 사용될 수 있다.
구체적으로, 본 발명의 근감소증이란, 노화에 따른 점진적인 골격 근육량의 감소를 의미하는 것으로서, 직접적으로 근력의 저하를 유발하며 그 결과 각종신체 기능의 감소 및 장애를 일으킬 수 있는 상태를 의미한다.
또한 근위축증은 사지의 근육이 거의 좌우대칭적으로 점점 위축되어 가는 것으로서, 척수에 있는 운동신경섬유 및 세포의 진행성 변성을 유발하여 근위축성 측삭경화증(Amyotrophic lateral sclerosis, ALS)과 척수성 진행성 근위축증(Spinal progressive muscular atrophy, SPMA)을 일으킬 수 있다.
근육 퇴행 위축(muscle dystrophy)이란, 점진적인 근위축과 근쇠약이 나타나는 질환으로서, 병리학적으로 근육섬유의 괴사를 특징으로 하는 퇴행성 근육병증을 의미한다. 근세포막의 손상으로 근육섬유의 괴사와 퇴행과정을 거쳐 근력저하 및 위축이 발생하게 된다.
근육 퇴행 위축 (Muscular dystrophy)은 근육 약화의 범위와 분포, 발병 연령, 진행 속도, 증상의 중증도, 가족력에 따라 하위 질병으로 나누어질 수 있으며, 상기 근육 퇴행 위축의 비제한적 예로서, 뒤시엔느 근이영양증 (Duchenne muscular dystrophy), 베커 근이영양증 (Becker muscular dystrophy), 사지골반 근이영양증 (Limb-girsle muscular dystrophy), 에머리-드라이푸스 근이영양증 (Emery-Dreifuss muscular dystrophy), 안면 견갑상환 근이영양증 (Facioscapulophumeral muscular dystrophy), 근긴장성 근이영양증 (Myotonic muscular dystrophy), 안구 인두 근이영양증 (Oculopharyngeal muscular dystrophy), 말단 근육 근이영양증, 선천성 근이영양증 (Congential muscular dystrophy)을 들 수 있다.
본 발명의 심위축증은 심장이 외부적이거나 내부적인 요인에 의해서 위축되어 가는 것으로서, 기아, 소모성질환, 노쇠했을 때 심근섬유가 마르고 가늘어져 지방조직의 감소를 유발하는 심장의 갈색위축 증세를 일으킬 수 있다.
본 발명에서 사용되는 용어, "예방"이란 상기 조성물의 투여에 의해 근육 약화 관련 질환을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어, "치료"란 상기 조성물의 투여에 의해 근육 약화 관련 질환에 의한 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 약학 조성물은 투여를 위하여, 상기 소브레롤 또는 이의 약학적으로 허용 가능한 염 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 포함할 수 있다. 상기 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
본 발명의 약학 조성물은 소브레롤 또는 이의 약학적으로 허용 가능한 염의 신속, 지속 또는 지연된 방출을 제공할 수 있도록 당업계에 잘 알려진 방법을 사용하여 약학적 제형으로 제조될 수 있다. 제형의 제조에 있어서, 활성 성분을 담체와 함께 혼합 또는 희석하거나, 용기 형태의 담체 내에 봉입시키는 것이 바람직하다.
또한, 본 발명의 약학 조성물은 어떠한 제형으로도 적용 가능하나, 비경구용으로 제조하는 것이 바람직하다. 비경구용 제형으로는 주사용, 도포용, 에어로졸 등의 스프레이 형일 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성 용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다.
주사형 제형으로 제제화하기 위해서는 소브레롤 또는 이의 약학적으로 허용 가능한 염을 안정제 또는 완충제와 함께 물에서 혼합하여 용액 또는 현탁액으로 제조하고, 이를 앰플 또는 바이알의 단위 투여용으로 제제할 수 있다.
본 발명의 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는 약학 조성물은 근력 약화 관련 질환이 발병한 개체 또는 발병 가능성이 있는 개체의 근력 강화가 필요한 부위에 직접 주입될 수 있으며, 체외 또는 체내 근원세포에 적용하여 분화된 근원세포를 제조한 후, 분화된 근원세포를 근력 약화 관련 질환이 발병한 개체 또는 발병 가능성이 있는 개체의 근력 강화가 필요한 부위에 주입할 수 있다.
또한, 상기 조성물에는 소브레롤 또는 이의 약학적으로 허용 가능한 염이 근력 약화 관련 질환의 예방 또는 치료에 방해가 되지 않는 한, 추가 성분 예를 들어, 근력 약화 관련 질환의 치료제로 알려져 있는 물질들이 포함될 수 있다.
구체적으로, 본 발명의 약학 조성물은 근원세포의 분화를 촉진하는 것을 특징으로 할 수 있다. 본 발명의 일실시예에서는 소브레롤을 근원세포에 처리한 후, qRT-PCR을 통해 근세포 분화와 관련된 근원성 조절 인자의 mRNA을 확인한 결과, 근세포 분화 촉진 효과가 매우 높음을 확인하였다(도 3). 또한, 마우스의 두 뒷다리(both hind limbs)를 10일 동안 고정시킨 후, 3일 동안 다시 움직이도록 한 뒤, 악력 테스트와 트레이드밀 테스트를 실시한 결과, 소브레롤을 투여한 실험군에서 각각 악력의 측정값이 증가하였고, 달리는 시간이 더 길게 나타났음을 확인하였다(도 7, 및 도 10). 이를 통해, 소브레롤이 근력을 향상시키고, 손상된 근육을 회복시키는 효과가 있음을 확인하였다.
이러한 결과를 통하여, 소브레롤 또는 이의 약학적으로 허용 가능한 염이 근원세포의 분화를 촉진하는데 효과적이며 근육 약화 관련 질환의 예방 치료에 유용할 수 있음을 알 수 있었다.
본 발명은 또 다른 하나의 양태로서, 소브레롤 또는 이의 식품학적으로 허용 가능한 염을 포함하는 근력 약화 관련 질환의 예방 또는 개선용 건강기능식품 조성물을 제공한다. 본 발명의 조성물은 근력 약화 관련 질환을 예방 또는 개선하기 위하여 근력 약화 관련 질환의 발병 단계 이전 또는 발병 후, 질환 치료를 위한 약제와 동시에 또는 별개로서 사용될 수 있다.
상기 근력 약화 관련 질환이란 근력약화로 인해 발생할 수 있는 모든 질환을 의미하며, 예를 들어 근감소증, 근위축증, 근육 퇴행 위축(muscle dystrophy) 또는 심위축증을 들 수 있으나 이에 제한되는 것은 아니다. 바람직하게, 상기 건강기능식품 조성물은 근원세포(Myoblasst)의 분화를 촉진하는 것을 특징으로 한다.
본 발명에서 사용되는 용어 "개선"이란, 치료되는 상태와 관련된 파라미터, 예를 들면 증상의 정도를 적어도 감소시키는 모든 행위를 의미한다.
또한, 본 발명의 건강기능식품 조성물을 식품 첨가물로 사용할 경우, 상기 조성물을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 일반적으로, 식품 또는 음료의 제조 시에 본 발명의 조성물은 원료에 대하여 15중량% 이하, 바람직하게는 10중량% 이하의 양으로 첨가될 수 있다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 물질을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
본 발명의 건강음료 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 및 덱스트린, 사이클로덱스트린과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 당업자의 선택에 의해 적절하게 결정될 수 있다.
상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 조성물은 천연 과일쥬스, 과일쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율 또한 당업자에 의해 적절히 선택될 수 있다.
본 발명은 또 다른 하나의 양태로서, 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는, 근력 강화용 조성물을 제공한다.
본 발명의 용어, "근력 강화"란 신체 수행의 강화, 최대 지구력의 강화, 근육량의 증가, 근육 회복의 강화, 근육 피로의 감소, 에너지 수지의 개선 또는 이들의 조합 효과를 말한다.
본 발명의 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는 근력 강화용 조성물은 근원세포를 근육 세포로 분화시키는 능력을 통하여 근육량을 증가시켜 전체 근육량을 증가시킬 수 있으며, 최대 지구력이 강화되고, 이에 따라 신체 수행이 강화되고 근육 피로도 감소할 수 있다. 또한, 근육 세포가 빠르게 대체될 수 있기 때문에 근육의 손상에 대하여 빠르게 치유될 수 있다.
본 발명의 근력 강화용 조성물은 투여를 위하여, 상기 소브레롤 또는 이의 약학적으로 허용 가능한 염 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 포함할 수 있다. 상기 약학적으로 허용 가능한 담체, 부형제 또는 희석제는 상기 설명한 바와 같다.
본 발명의 일실시예에서는 상기 소브레롤을 마우스에 처리하여, 마우스의 두 뒷다리(both hind limbs)를 10일 동안 고정시킨 후, 3일 동안 다시 움직이도록 한 뒤, 악력 테스트와 트레이드밀 테스트를 실시한 결과, 소브레롤을 투여한 실험군에서 각각 악력의 측정값이 증가하였고, 달리는 시간이 더 길게 나타났음을 확인하였다(도 7, 및 도 10).
본 발명에서 상기 조성물은 근육의 크기 또는 무게를 증가시키는 것일 수 있다.
본 발명의 일실시예에서는, 모델 마우스 뿐만 아니라 노화 마우스에서도, 대조군에 비하여 소브레롤을 투여한 마우스에서 근육의 크기 및 무게가 증가되었음을 면역형광분석법 및 근섬유 직경 측정을 통하여 확인하였다 (도 11 및 도12, 도 15 및 16).
본 발명은 또 다른 하나의 양태로서, 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는 근력 강화용 사료 또는 사료 첨가제를 제공한다.
본 발명에서, "사료"란, 동물의 생명을 유지하는데 필요한 유기 또는 무기 영양소를 공급하는 물질을 의미한다. 상기 사료는 가축 등의 동물이 필요로 하는 에너지, 단백질, 지질, 비타민, 광물질 등의 영양소를 포함하며, 곡물류, 근과류, 식품가공부산물류, 조류, 섬유질류, 유지류, 전분류, 박류, 곡물부산물류 등의 식물성 사료 또는 단백질류, 무기물류, 유지류, 광물성류, 유지류, 단세포 단백질 등의 동물성 사료가 될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서, "사료 첨가제"란, 동물의 생산성 향상이나 건강을 증진시키기 위해 사료에 첨가되는 물질을 의미하며, 특별히 이에 제한되지 않으나 성장촉진, 질병 예방 등을 위한 아미노산제, 비타민제, 효소제, 향미제, 규산염제, 완충제, 추출제, 올리고당 등이 더욱 포함될 수 있다.
상기 본 발명의 근력 강화용 사료 또는 사료 첨가제에 포함되는 소브레롤 또는 이의 약학적으로 허용 가능한 염의 함량은 특별히 이에 제한되지 않으나, 0.001 내지 1%(w/w)일 수 있고, 바람직하게는 0.005 내지 0.9%(w/w)일 수 있고, 가장 바람직하게는 0.01 내지 0.5%(w/w)일 수 있다.

본 발명의, 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는 주름 개선용 조성물은 근육 세포의 분화를 촉진할 뿐만 아니라 (도 2 및 도 3), 근력 향상 및 손상된 근육의 회복을 촉진하는 효과를 가지며 (도 7, 도 10, 도 13 및 도 14), 근육의 크기 및 무게를 증가시키는 효과를 나타내므로 (도 11, 도 12, 도 15 및 도 16), 안면 근육의 세포 분화, 근력 향상 및 탄력 회복을 유도하여, 이와 밀접하게 관련된 피부 주름을 개선할 수 있다.
구체적으로, 상기 조정물은 약학 조성물, 건강기능식품 조성물, 또는 화장료 조성물일 수 있으며, 상기 약학 조성물 및 건강기능식품 조성물에 대하여는 상기 설명한 바와 같다.
본 발명에 따른 피부 주름 개선용 화장료 조성물은 크림, 로션, 에센스, 젤, 연고, 폼, 화장수, 팩, 유연수, 유액, 파운데이션, 메이크업베이스, 비누, 액체세정료, 입욕제, 선 스크린 크림, 또는 선오일 등의 제형으로 제조될 수 있다.
본 발명에 따른 피부 주름 개선용 화장료 조성물은 물, 계면활성제, 보습제, 16-4 저급알코올, 킬레이트제, 살균제, 산화방지제, 방부제, 색소 및 향료로 이루어지는 군으로부터 1종 이상 선택되는 첨가제를 더 포함할 수 있다.

제조 및 근육 분화 촉진의 확인 방법
소브레롤의 근원세포의 분화 촉진능을 평가하기 위해, 분비형 미오게닌 루시퍼라제 분석 시스템(secreted MyoG luciferase assay system)을 사용하였다(도 1).
구체적으로, 근육 분화 시 발현이 증가하는 단백질인 미오게닌(myogenin)의 프로모터 부분을 분비형 루시퍼라아제 벡터(pMetLuc2-Reporter Vertor, Clontech사)에 클로닝한 후, 상기 벡터를 근원세포주인 C2C12에 형질주입(transfection)하여 루시퍼라아제 효소가 안정적으로 발현할 수 있는 미오게닌 프로모터-분비형 루시퍼라아제 C2C12(mMyogenin promoter-secreted luciferase C2C12) 근원세포 주를 제조하였다
상기 미오게닌 프로모터가 활성화되면서 루시퍼라아제 효소가 배지로 분비되는 시스템을 통해 근원세포가 근관세포로 분화하는 정도를 정량적으로 확인할 수 있다. 근육세포로의 분화를 유도하기 위해서 상기 C2C12 근원세포 주에 2% 말 혈청(horse serum)이 첨가된 DMEM(Dulbecco's Modified Eagle Medium, Invitrogen) 배지를 첨가한다. 이때 발현되는 MEF2와 MyoD가 미오게닌 프로모터에 결합하여 루시퍼라아제 효소가 발현되어 세포 밖으로 분비된다. 배지에는 분비된 루시퍼라아제 효소가 존재하므로, 이를 측정하여 근육의 분화 정도를 확인할 수 있다.

분화 촉진 확인
0.1 nM 내지 10 uM 범위의 농도별 소브레롤을 이용하여 근원세포의 분화 촉진능을 확인하였다. 구체적인 실험방법은 다음과 같다.
첫째 날, 상기 실시예 1-1에서 제조된 C2C12 세포를 10% 소 혈청(Fetal Bovine Serum, FBS)이 첨가된 DMEM 배지가 존재하는 96-웰 플레이트에 5×10³세포/웰 농도로 접종하였다. 둘째 날 오전, 상기 웰 플레이트의 배지를 동일한 성분인 10% 소 혈청이 첨가된 DMEM 배지(성장 배지, Growth medium, GM)로 교체하였다. 오후에는 상기 배지(GM)를 수집하였다. 이후, 상기 농도별 소브레롤을 희석시킨 2% 말 혈청이 첨가된 DMEM 배지(분화 배지, Differentiation medium 1, DM1)를 상기 웰에 각각 100 μl씩 처리하였다. 셋째 날, 상기 DM1 배지를 수집하였고, 둘째 날과 동일한 배지를 상기 웰에 처리하였다(DM2). 넷째 날, DM2를 수집하였다.
C2C12 세포의 분화과정에서 분비된 루시퍼라아제(luciferase)를 포함하는 배지를 상기와 같이, 성장 배지(GM)부터 1일차 분화 배지(DM1), 및 2일차 분화 배지(DM2)까지 모은 뒤, 루시퍼라아제 효소가 포함된 상기 수집 배지 50㎕에 기질인 코엘렌테라진(coelenterazine) 5㎕를 넣어 반응시킨 후, 생성되는 빛을 루미노미터(luminometer)를 이용하여 측정하였다. 소브레롤의 농도별 근육 분화 촉진능은 DM1과 DM2의 측정값을 각각 DM0의 측정값으로 빼주어 수치화하여 측정하였다. 메탄올(MeOH)을 처리한 음성 대조군의 수치를 1로 보았을 때, 소브레롤을 처리한 실험군의 증감 수치를 비교하여 소브레롤의 근육 분화 촉진능을 평가하였다. 루시퍼라아제 분석 키트(Luciferase assay kit)로는 Ready-To-GlowTM Secreted Luciferase Reporter Systems(clontech)을 사용하였으며, 실험은 3회 반복 실시하였다.
평가 결과, 0.01nM의 저농도 소브레롤을 처리한 경우에도 음성 대조군에 비해 루시퍼라아제의 활성이 1.2배 이상 증가하였다. 또한, 0.01 nM 내지 10 μM 농도의 소브레롤을 처리한 군에서 대조군에 비해 높은 루시퍼라아제 활성을 나타냈으며, 특히 0.1nM 농도에서는 루시퍼라아제의 활성이 약 1.5배 증가하였다(도 2).
이를 통해, 저농도의 소브레롤의 경우에도, 근육 세포의 분화를 촉진할 수 있음을 확인하였으며, 특히 0.1 nM의 농도에서 가장 큰 분화 촉진능을 가짐을 확인하였다.

소브레롤이 근원성 조절 인자(myogenic regulatory factor)의 mRNA에 미치는 영향을 확인하는 실험을 실시하였다. 구체적으로, 성장 배지에 분주된 근원세포(C2C12 myoblast cell)에 0.1 μM의 소브레롤을 48시간 동안 처리한 후, qRT-PCR을 통해 미오디(MyoD), 미오게닌(Myogenin), 및 미오신 헤비 체인(Myosin heavy chain, MyHC)의 mRNA 수준을 측정하였다.
측정 결과, 근원세포에 소브레롤을 처리한 군에서 근원성 조절 인자(myogenic regulatory factor)인 미오디(MyoD), 미오게닌(Myogenin), 및 미오신 헤비 체인(Myosin heavy chain, MyHC)의 mRNA이 증가하는 것으로 나타났다. 구체적으로, 미오디와 미오게닌의 경우 각각 2배, 4배씩 증가하였으며, 미오신 헤비 체인의 경우 약 25배 이상 증가하였다(도 3). 이를 통해, 소브레롤이 근육의 분화 촉진능을 가짐을 확인하였다.

상기 실시예 2-1에서 mRNA이 증가된 근원성 조절 인자 중, 소브레롤이 미오신 헤비 체인의 단백질 발현에 미치는 영향을 확인하는 실험을 실시하였다. 구체적으로, 분화 배지에 분주된 근원세포(C2C12 myoblast cell)에 0.1 μM의 소브레롤을 72시간 동안 처리한 후, 면역 형광 염색법(Immunofluorescence)을 통해 미오신 헤비 체인의 단백질 발현량을 측정하였다.
측정 결과, 근원세포에 소브레롤을 처리한 군에서 DMSO를 처리한 군에 비해 형광도가 강하게 나타남을 확인하였다(도 4). 이를 통해, 소브레롤이 근육의 분화 촉진능을 가짐을 확인하였다.

3-1: 동물의 운동능력 향상 확인 실험 준비
본 동물실험에서는 총 45마리의 5주령 수컷 C57BL/6 마우스(대한바이오링크)를 사용하였다. 전체 계획은 도 5에서 보여주는 바와 같다. 2 주 동안 주변 환경에 적응시킨 뒤, 7 주령이 되었을 때부터 소브레롤을 1일 2회 경구 투여하였다. 대조군은 200㎕ (물)/ 20g (마우스) 을 투여하고 소브레롤은 농도 의존도(dose dependency)를 보기 위하여 1 : 2 : 4의 비율로 8.8 ㎎ / 200 ㎕ / 20 g (마우스), 17.6 ㎎ / 200 ㎕ / 20 g (마우스), 35.3 ㎎ / 200 ㎕ / 20 g (마우스) 을 경구 투여하였다. 8 주령이 되었을 때, 수술용 스테이플(Autosuture Royal 35W stapler)을 이용하여 마우스의 두 뒷다리(both hind limbs)를 10일동안 고정(immobilization)시켰다. J Appl Physiol 106: 2049-2059, 2009). 수술용 스테이플을 제거한 뒤, 3일 동안 다시 움직이도록 한 후(remobilization), 운동능력 향상 확인 실험(트레이드밀, 악력 테스트)을 수행하였다.
한편, 다음과 같이 소브레롤의 사람 경구투여량을 마우스 경구투여량으로 환산하였다(표 1 내지 3)(Guidance for Industry Estimating the Maximum Safe Starting Dose in Initial Clinical Trials for Therapeutics in Adult Healthy Volunteers. July 2005 Pharmacology and Toxicology).
HED = mg/kg의 동물 투여량 x kg의 동물 체중/kg의 사람 체중
사람 상수 kmhuman과 마우스 상수 kmmouse는 각각 37과 3이다.
[NOAEL(무독성량)] ÷ [kmhuman/kmmouse] = HED(human equivalent dose)로서 사람의 경구투여량에 12.3(kmhuman/kmmouse = 37/3)을 곱하면 마우스의 경구투여량으로 환산할 수 있다.
소브레롤의 사람(70kg 기준) 경구 투여량을 마우스(20g 기준) 경구 투여량으로 환산하여 계산하면 다음과 같다(표 3).
실시예
3-2: 악력(grip strength) 테스트
BIOSEB사의 마우스용 악력측정기를 사용하여 악력을 측정하였다(도 6). 구체적으로, 힘의 세기를 모니터링 할 수 있는 계기판에 부착된 철망 위에 상기 실시예 3-1의 마우스의 앞발만 올려놓고 꼬리를 잡아 아래쪽으로 끌어내리면서 마우스가 철망을 잡는 힘을 측정하였다. 테스트는 연속적으로 3회 반복 수행하였다.
악력 테스트 결과, 증류수를 경구 투여한 음성 대조군의 경우, 측정값이 3.5 N 이하로 뒷다리의 운동을 제한하지 않은 군(non-immobilization)의 측정값과 거의 동일하게 나타났다. 반면, 소브레롤을 경구 투여한 군에서는 측정값이 4.0 N 이상으로 나타났다. 또한, 투여된 소브레롤의 농도가 높을수록 악력의 측정값이 증가하는 경향성을 보이는 것으로 나타났다(도 7). 이를 통해, 소브레롤이 근력을 향상시키는 효과가 있음을 확인하였다.

3-3:
트레이드밀
(treadmill) 테스트
트레이드밀 테스트에는 도 8에서 보이는 것과 같은 장비가 사용되었다.
The protocol used for the treadmill test (DOI 10.1016/j.cell.2006.11.013 _ Cell, Volume 127 Supplemental Data)의 트레이드밀(treadmill) 프로토콜을 참고하여 도 9와 같이 트레이드밀 테스트를 수행하였다.
트레이드밀 테스트를 수행하기 하루 전에 6 m/분의 속도로 10분 동안 워밍업시켰다. 시작점에는 전기 자극이 흐르도록 되어 있어 마우스를 강제로 운동시켰다. 다음 날, 8 m/분의 속도로 시작하여 매 20분마다 속도를 2 m/분씩 올려준다. 트레이드밀 테스트를 시작하고 1시간이 되었을 때는 12 m/분의 속도는 유지하고 각도를 5° 올려주고 전기 자극을 꺼주었다. 20분 뒤에는 각도 5°를 계속 유지하며 속도만 2 m/분씩 더 올려주었다. 상기 조건에서 마우스의 달리는 시간을 측정 및 기록하였다.
트레이드밀 테스트 결과, 증류수를 경구 투여한 음성 대조군의 경우, 뒷다리의 운동을 제한하지 않은 군(non-immobilization)보다 달리는 시간이 약 20분 이상 짧게 나타났다. 소브레롤을 경구 투여한 군의 경우, 8.8 mg/kg 농도의 소브레롤을 투여한 마우스는 증류수를 투여한 군보다 달리는 시간이 조금 증가하였으나, 여전히 운동을 제한하지 않은 군보다 짧게 나타났다. 반면, 17.6 mg/kg 농도의 소브레롤을 투여한 마우스는 운동을 제한하지 않은 군과 거의 비슷한 시간을 나타냈으며, 35.3 mg/kg 농도의 소브레롤을 투여한 마우스는 운동을 제안하지 않은 군에 비해 달리는 시간이 더 길게 나타났다(도 10). 이를 통해, 소브레롤이 근력을 향상시키고, 손상된 근육의 회복을 촉진시키는 효과가 있음을 확인하였다.

3-4: 근육크기 및 근육무게에 대한

또한, 상기 실시예 3-3에서 운동제한 후 전경골근 (tibialis anterior, TA)의 단면을 근섬유 테두리에 존재하는 단백질인 라미닌 (laminin)에 대한 항체를 이용해 면역형광분석법으로 염색하고, 각각의 근섬유 직경을 측정하였다.
그 결과, 증류수를 경구 투여한 대조군에 비하여 소브레롤을 경구 투여한 마우스의 근섬유 직경이 더 큰 분포를 나타내었다 (도 11). 또한, 근육무게를 측정한 결과, 비장근 (soleus), 비복근 (gastrocnemius, GA), 전경골근 (tibialis anterior, TA), 장지신근 (extensor digitorum longus, EDL)의 무게가 모두 유의한 수준으로 증가하였다 (도 12).
이를 통해, 소브레롤이 근육크기 및 근육무게를 증가시키는 효과가 있음을 확인하였다.

4: 노화마우스의 근력에 대한

실시예
4-1: 동물의 운동능력 향상 확인 실험 준비
소브레롤이 노화마우스의 근력을 향상시키는지 확인하기 위해 실험을 진행하였다. 21개월령의 암컷 C57BL/6 마우스를 2주 동안 주변 환경에 적응시킨 뒤 5주간 약물을 경구 투여하였다. 구체적으로, 음성 대조군은 물, 양성 대조군은 2 mg/kg (마우스) 농도의 클렌부테롤 (Clenbuterol)을 투여하고, 소브레롤을 투여하는 실험군은 17.6 또는 35.3 mg/kg (마우스)의 농도로 경구 투여 하였다. 그 후 다시 운동능력 테스트 (locomotion test (grip strength, treadmill))를 수행하여 효과를 확인하였다.

4-2: 악력 (grip strength) 테스트
악력 테스트에 관하여는 상기 실시예 3-2와 같다.
그 결과, 첫 번째 측정한 악력의 수치는 차이가 없었으나, 반복적으로 악력측정을 실시하였을 때 물을 경구 투여한 음성 대조군의 경우 운동에 의한 피로도의 상승으로 인해 시간이 지남에 따라 악력의 세기가 약해지는 것을 알 수 있었다. 반면, 소브레롤을 경구 투여한 실험군에서는 클렌부테롤 (Clenbuterol)을 투여한 양성 대조군과 유사하게, 피로도가 증가하지 않음을 알 수 있었다 (도 13).
이를 통해, 소브레롤이 노화마우스의 근육 피로도를 약화시켜, 근력을 향상시키는 효과가 있음을 확인하였다.

그 결과, 17.6 mg/kg (마우스) 농도의 소브레롤을 경구 투여한 실험군의 경우, 소브레롤을 투여하지 않은 음성 대조군보다 유의한 수준으로 달리는 시간이 더 길어졌으며, 35.3 mg/kg (마우스) 농도의 소브레롤을 투여한 마우스 또한 음성 대조군보다 달리는 시간이 더 길어지는 결과를 나타내었다 (도 14).
이를 통해, 소브레롤이 노화마우스의 지구력을 향상시키는 효과가 있음을 확인하였다.

4-4: 근육크기 및 근육무게에 대한

효과 확인
또한, 상기 실시예 3-3의 방법으로, 소브레롤에 의한 노화마우스의 근육크기 및 근육무게의 변화를 확인하고자 하였다.
상기 실시예 4-3에서 실험군 및 음성 대조군의 근육 무게를 확인한 결과, 35.3 mg/kg (마우스) 농도의 소브레롤을 먹인 노화마우스의 비복근 (gastrocnemius, GA)의 무게가 유의한 수준으로 증가하였다 (도 15). 또한, 상기 실시예 3-3과 같이 라미닌 (laminin) 항체를 이용한 면역형광분석법으로 전경골근 (tibialis anterior, TA) 단면의 근섬유 직경을 측정한 결과, 대조군에 비하여 소브레롤을 먹은 노화마우스의 근섬유 직경이 더욱 증가하였다 (도 16).
이를 통해, 소브레롤이 노화마우스의 근육크기 및 근육무게를 증가시키는 효과가 있음을 확인하였다.

4-5: 운동제한 조건에서, 노화마우스의 근력에 대한
소브레롤의
효과
또한, 운동제한 후에도 소브레롤이 노화마우스의 근력을 향상시키는지 확인하고자 하였다.
노화마우스를 수술용 스테이플 (Autosuture Royal 35W stapler)을 이용하여 두 뒷다리를 5일 동안 고정 (immobilization)시킨 후, 스테이플을 제거한 뒤 다시 움직이도록 하여 (remobilization), 운동능력을 트레이드밀 테스트로 확인하였다. 그 결과, 증류수를 투여한 대조군에서는 운동제한 전후로 트레이드밀을 달리는 시간이 감소하는 반면, 소브레롤을 투여한 실험군에서는 운동제한에 영향을 받지 않았다 (도 17).
이를 통해, 소브레롤이 운동을 제한한 노화마우스의 지구력을 향상시켜, 근력향상 효과를 나타냄을 확인하였다.
상기 실시예들을 통해서 알 수 있는 것처럼, 본 발명의 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는 조성물은 모델 마우스 뿐만 아니라 노화 마우스에서도 근육 분화를 촉진하고 손상된 근육의 회복을 촉진시키는 효과가 있으므로, 근원세포의 분화 촉진용 조성물, 근력 약화 관련 질환의 예방 또는 치료용 약학 조성물 및 근력 강화용 조성물로 이용될 수 있다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시 예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (18)
- 소브레롤(Sobrerol) 또는 이의 약학적으로 허용 가능한 염을 포함하는 근원세포(myoblast)의 분화 촉진용 조성물.
- 제1항에 있어서, 상기 소브레롤의 농도는 0.01 nM 내지 1μM인 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 근원성 조절 인자(myogenic regulatory factor)의 mRNA 또는 이로부터 발현되는 단백질의 수준을 증가시키는 것인, 조성물.
- 제3항에 있어서, 상기 근원성 조절 인자는 미오디(MyoD), 미오게닌(Myogenin), 및 미오신 헤비 체인(Myosin heavy chain, MyHC)으로 구성된 군에서 선택된 어느 하나인 것인, 조성물.
- 소브레롤 또는 이의 약학적으로 허용 가능한 염을 근원세포에 처리하는 단계를 포함하는, 근원세포의 분화 촉진 방법.
- 소브레롤 또는 이의 약학적으로 허용 가능한 염을 근원세포에 처리하여 근원세포를 분화시키는 단계를 포함하는, 분화된 근원세포의 제조방법.
- 소브레롤(Sobrerol) 또는 이의 약학적으로 허용가능한 염을 포함하는, 근력 약화 관련 질환의 예방 또는 치료용 약학 조성물.
- 제7항에 있어서, 상기 근력 약화 관련 질환은 근감소증, 근위축증, 근육 퇴행 위축(muscle dystrophy), 및 심위축증으로 이루어진 군으로부터 선택된 어느 하나인 것인, 약학 조성물.
- 제7항에 있어서, 상기 조성물은 근원세포에서 근세포로의 분화를 촉진하는 것을 특징으로 하는 것인, 약학 조성물.
- 소브레롤 또는 이의 식품학적으로 허용 가능한 염을 포함하는, 근력 약화 관련 질환의 예방 또는 개선용 건강기능식품 조성물.
- 제10항에 있어서, 상기 근력 약화 관련 질환은 근감소증, 근위축증, 근육 퇴행 위축(muscle dystrophy), 및 심위축증으로 이루어진 군으로부터 선택된 어느 하나인 것인, 조성물.
- 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는, 근력 강화용 조성물.
- 소브레롤 또는 이의 약학적으로 허용 가능한 염을 포함하는, 근력 강화용 사료 또는 사료 첨가제.
- 소브레롤(Sobrerol) 또는 이의 약학적으로 허용가능한 염을 포함하는 조성물을 개체에 투여하는 단계를 포함하는, 근력 약화 관련 질환의 치료방법.
- 제14항에 있어서, 상기 근력 약화 관련 질환은 근감소증, 근위축증, 근육 퇴행 위축(muscle dystrophy), 및 심위축증으로 이루어진 군으로부터 선택된 어느 하나인 것인, 치료방법.
- 소브레롤(Sobrerol) 또는 이의 약학적으로 허용가능한 염을 포함하는 조성물을 개체에 투여하는 단계를 포함하는, 근력 강화방법.
