Abstract
본 발명은 가축 체내 축적되지 않고 곧 바로 흡수되어 빠르게 에너지원으로 이용되어 가축의 성장을 촉진할 수 있는 사료용 유지 조성물, 이를 이용한 성장 촉진용 사료 첨가제, 가축용 사료 조성물 및 가축 사육 방법에 관한 것이다.
Description
본 발명은 가축 체내 축적되지 않고 곧 바로 흡수되어 빠르게 에너지원으로 이용될 수 있는 사료용 유지 조성물, 이를 이용한 성장 촉진용 사료 첨가제, 가축용 사료 조성물 및 가축 사육 방법에 관한 것이다.
사료는 가축의 생명을 유지하고 젖·고기·알·털가죽 등을 생산하는데 필요한 유기(有機) 또는 무기(無機) 영양소를 공급하는 물질을 일컫는다. 각종 가축이 필요로 하는 에너지·단백질·비타민·광물질 등의 영양소뿐만 아니라 성장촉진제와 질병 예방제 등을 고루 섞어 만든 것이다.
사료는 가축의 섭취에 의해 생존 및 축산물 생산에 필요한 영양소를 공급할 뿐만 아니라, 면역기능을 강화시키고, 축산물의 질을 향상시키며, 축사환경을 개선하는 등 다양한 역할을 수행한다.
특히 가축의 생산성 증가는 축사 환경을 개선하거나 사료 효율을 개선하여 이루어지는데, 사료 효율의 개선은 기존 사료 조성에 새로운 조성을 첨가하거나 배합 비율을 달리하거나 급여 방식에 변화를 주는 다양한 방식으로 연구가 진행되고 있다.
일 예로, 대한민국 특허공개 제2006-35444호는 동물 사료 및 이를 이용한 동물의 사육 방법에 관한 것으로, 일반 동물 사료 및 대나무 숯을 포함하여 체중 또는 증체량을 증가하는 동물 사료를 제안하고 있다.
지방은 가축의 필수 영양소 중 하나로, 다른 영양소에 비해 에너지가가 높으며 단위 중량당 가장 비싼 에너지원이다. 따라서 체내에서 지방의 이용효율을 향상시킬 경우 가축의 생산성을 향상시킬 수 있을 뿐만 아니라 사료의 원재료비를 저감하여 생산비를 절감할 수 있는 기회를 제공할 수 있게 된다.
디글리세라이드는 지방산과 글리세린의 에스테르 교환반응에 의하여 제1, 제2 또는 제1, 제3 위치의 글리세린에 지방산이 결합된 유지 조성물로 '트리글리세라이드'라고 불리는 일반유지와 특별히 구분하여 취급되고 있다. 최근에는 디글리세라이드가 일반 중성유지와 비교시 소화, 흡수되는 과정은 같으나, 구조적으로 안정하면서 리파아제에 의해 분해와 흡수된 다음 재합성 과정이 이루어지지 않아 중성지방으로 거의 재합성되지 않음으로써, 섭취하더라도 혈중 중성지방의 함량이 상승하지 않고 체지방을 축적시키지 않는 생리 효과가 있다는 것이 밝혀져, 비만 개선, 다이어트를 위한 용도로 다양하게 사용되고 있다. 일본 공개특허 2007-512407호에는 체지방 축적 방지, 질병 예방 및 영양분 공급 등을 목적으로 공액화 리놀레산이 구성된 디글리세라이드가 다량 함유된 유지조성물을 식품, 약학적 조성물, 및 사료 첨가제로 사용할 수 있다고 제안하고 있다. 일본 공개특허 1996-269478호는 분자 내에 C8~C10 중쇄지방산 잔기를 2개 함유하는 트리글리세라이드를 31 중량% 이상 함유하는 유지 조성물에 관한 것으로, 통상 사용되고 있는 조리유보다 대사가 빠르고 그리고 생체로의 지방 축적이 작다고 기재하고 있다. 또한 일본 공개특허 1993-56755호는 가축용 사료 첨가물 및 가축용 사료에 관한 것으로, 탄소수 6~12 중쇄지방산 트리글리세라이드를 사용하여 닭 콕시듐증 발생을 억제하고, 체지방 이상 축적을 억제할 수 있다고 개시하고 있다.
본 발명의 과제는 지방원으로서 가축에 급여시 지방으로 축적되지 않고 신속히 에너지원으로 사용되어 가축의 성장이 촉진되고 생산성이 향상될 수 있는 사료용 유지 조성물, 이를 이용한 성장 촉진용 사료 첨가제, 가축용 사료 조성물 및 가축 사육 방법을 제공하는 것이다.
상기한 과제를 달성하기 위해 본 발명은
구성지방산 중 탄소수 14 이하의 지방산의 함량이 10 내지 90 중량%이고, 1, 3번 위치에 결합된 탄소수 14 이하의 지방산의 함량이 10 내지 90 중량%인 디글리세라이드를 50 중량% 이상 함유하는 사료용 유지 조성물을 제공한다.
또한 본 발명은 상기한 사료용 유지 조성물을 유효성분으로 함유하는 성장 촉진용 사료 첨가제를 제공한다.
또한 본 발명은 상기한 사료용 유지 조성물 및 배합사료를 포함하는 가축용 사료 조성물을 제공한다.
또한 본 발명은 상기한 사료용 유지 조성물로 배합사료 내 지방원의 10 중량% 이상으로 대체한 가축용 사료 조성물을 제공한다.
또한 본 발명은 상기 가축용 사료 조성물을 급여하는 것을 특징으로 하는 가축 사육방법을 제공한다.
본 발명에 따른 사료용 유지 조성물은 가축 체내 지방 대사 과정에서 지방으로 축적되지 않고 신속히 에너지원으로 사용되어 가축의 성장을 촉진하여 생산성을 향상시킬 수 있다.

이하 본 발명을 더욱 상세하게 설명한다.
본 발명의 사료용 유지 조성물은 구성지방산 중 탄소수 14 이하의 지방산의 함량이 10 내지 90 중량%이고, 1, 3번 위치에 결합된 탄소수 14 이하의 지방산의 함량이 10 내지 90 중량%인 디글리세라이드를 50 중량% 이상 함유한다.
상기 사료용 유지 조성물은 구성 지방산으로 탄소수 14 이하의 지방산을 고함량 함유하는 1,3-디글리세라이드를 다량 포함한다. 일반적으로 사료 내 지방원으로 사용되는 중성지방(트리글리세라이드)는 가축 체내 지방분해효소에 의해 2-모노글리세라이드와 자유 지방산으로 분해되고, 2-모노글리세라이드는 다시 트리글리세라이드로 재합성되어 피하조직 내 지방으로 축적된다. 이와 달리 1,3-디글리세라이드는 지방분해효소에 의해 1-모노글리세라이드(3-모노글리세라이드)와 자유 지방산으로 분해되므로, 트리글리세라이드로 재합성되는 2-모노글리세라이드로 전환되지 않고, 분해된 유리 지방산은 간으로 운반되어 신속하게 에너지원이 되어 대사된다. 따라서 본 발명의 사료용 유지 조성물은 가축의 지방원으로 급여시 축적되지 않고 신속히 에너지원으로 사용되어 가축의 생산성이 향상된다.
또한 디글리세라이드를 구성하는 탄소수 14 이하의 지방산은 체내 효소에 의해 유리 지방산으로 분해된 뒤 소장 세포에서 쉽게 흡수되어 유리 지방산의 상태로 문맥에 들어가 간으로 옮겨서 신속하게 에너지원으로 대사된다. 구체적으로, 탄소수 14 이하 지방산은 탄소수 16 이상의 지방산과 달리 지방산의 β-산화를 위한 미토콘드리아 속으로 진입하기 위해서 요구되는 카르니틴을 필요로 하지 않아 신속하게 대사된다.
특히 본 발명의 사료용 유지 조성물은 1,3-디글리세라이드의 구성지방산 중 탄소수 14 이하의 지방산/탄소수 16 이상의 지방산의 중량비는 2 내지 9, 구체적으로는 4 내지 7로, 자유 지방산으로 분해시 신속하게 대사되는 탄소수 14 이하의 지방산의 함량을 높였다.
상기 디글리세라이드는 1,3-디글리세라이드를 주성분으로 한다. 바람직하게는 1,3-디글리세라이드의 함량은 40 중량% 이상, 바람직하기로 40 내지 80 중량%이다. 1,2-디글리세라이드 및 2,3-디글리세라이드는 가축 체내 지방 분해효소인 리파아제에 의해 2-모노글리세라이드와 하나의 지방산으로 분해되어 흡수된 후 소장벽에서 다시 중성지방으로 합성되어 단백질 등과 함께 카이로미크론이라는 리포 단백질이 되어 전신으로 운반되며 피하조직 내 지방으로 축적된다. 반면 1,3-디글리세라이드는 가축 체내 리파아제에 의해 지방산, 1-모노글리세라이드(3-모노글리세라이드)로 분해되므로, 1,2-디글리세라이드 및 2,3-디글리세라이드를 급여한 경우보다 가축 혈중 지방산 수치는 높으며 중성지방 수치는 감소하게 된다. 또한 리파아제의 작용으로 1,3-디글리세라이드로부터 생성된 1-모노글리세라이드는 지질 코팅 바이러스(lipid coated virus), 그람 양성균, 그람 음성균의 외부벽 역할을 와해시켜 항미생물성을 갖는다. 그 밖에도 곰팡이, 원생동물, 마이코플라즈마 등에도 효과적이다. 따라서, 본 발명의 유지 조성물의 소화과정에서 생성된 모노글리세라이드는 항미생물 작용을 나타내어 가축의 항병력을 증진할 수 있다.
이러한 디글리세라이드는 본 발명의 유지 조성물 중 50 내지 90 중량%로 함유된다. 만약 그 함량이 상기 범위 미만이면 상대적으로 가축 지방 과축적의 원인이 되는 트리글리세라이드 함량이 많아지고, 이와 반대로 상기 범위를 초과하면 추가적인 정제공정이 필요하여 경제적이지 못할 뿐만 아니라 가축의 성장, 육질개선에 필요한 중성지방의 함량이 오히려 감소하여 성장이 저하되거나 육질이 저하되는 문제점이 있으므로 상기 범위 내에서 적절히 사용한다.
1,3-디글리세라이드의 구성 지방산은 탄소수 14 이하의 지방산의 함량이 70 내지 90 중량%, 탄소수 16 이상의 지방산의 함량은 10 내지 30 중량%인 것이 신속한 에너지원으로 사용되기 위한 생리 효과면에서 바람직하다.
이때 상기 디글리세라이드를 구성하는 구성 지방산 중 탄소수 14 이하의 지방산은 공지된 종류가 가능하며, 본 발명에서 그 종류를 특별히 한정하지는 않는다. 대표적으로 카프릴산, 펠라르곤산, 카프르산, 운데카노산, 라우르산, 미리스트산 및 이들의 혼합물로 이루어진 군에서 선택된 1종을 사용한다. 신속한 에너지원으로 사용되기 위한 생리 효과면에서 바람직하기로 카프릴산, 카프르산, 라우르산, 미리스트산 및 이들의 혼합물로 이루어진 군에서 선택된 1종이 더욱 바람직하다.
상기 탄소수 16 이상의 지방산은 유지 조성물의 산화 안정도, 지방산의 소화흡수 측면에서 팔미트산, 올레산, 리놀레산, 스테아르산 및 이들의 혼합물로 이루어진 군에서 선택된 1종이 바람직하며, 팔미트산, 스테아르산 및 이들의 혼합물로 이루어진 군에서 선택된 1종이 더욱 바람직하다.
본 발명의 유지 조성물은 배합사료 내 지방원을 대체하거나 첨가제의 형태로 배합사료에 추가될 수 있다.
본 발명의 성장 촉진용 사료 첨가제는 상기 사료용 유지 조성물을 유효성분으로 함유한다.
본 발명의 가축용 사료 조성물은 상기 사료용 유지 조성물을 포함하여 배합사료 총 중량을 기준으로 디글리세라이드가 0.005 내지 2.5 중량% 포함되도록 하며, 구체적으로 상기 사료용 유지 조성물을 배합사료 총 중량을 기준으로 0.01 내지 5.0 중량%, 바람직하기로 0.01 내지 3.0 중량%, 더욱 바람직하기로 0.01 내지 1.0 중량% 포함한다. 본 발명에 따른 가축 배합사료에 포함되는 상기 유지 조성물의 함량이 상기 범위 미만이면, 그 사용 효과를 기대하기 어려우며, 이와 반대로 상기 범위를 초과하면, 사육되는 가축의 영양상태가 불균형해지고, 가축 성장 촉진 효과를 나타낼 수 없다.
또한 본 발명의 가축용 사료 조성물은 배합사료 내 지방원의 10 중량% 이상, 바람직하기로 10 내지 80 중량%를 상기 유지 조성물로 대체할 수 있다. 이렇게 배합사료 내 지방원을 대체하는 경우 종래 지방원으로 사용되었던 우지, 어유, 대두유에 비해 품질 변이가 없고, 사용원가가 적게 들어 경제적인 이점이 있다.
상기 가축용 사료 조성물은 당업계에 공지되거나 시판되는 임의의 배합사료에 본 발명의 유지 조성물을 추가함으로써 구성될 수 있으며, 상기 가축 배합사료는 급여하는 가축의 종류에 따라 그 조성 및 제조방법을 달리하므로, 본 발명에서 그 조성 및 제조방법을 특별히 한정하지는 않는다. 이때 가축은 돼지, 닭, 오리, 메추라기, 거위, 꿩, 칠면조, 소, 젖소, 말, 당나귀, 양, 염소, 개, 고양이, 토끼, 각종 양식 어류 및 새우이다.
추가로 본 발명에 따른 사료 조성물은 필요한 경우 동물의 건강상태를 개선하거나, 생산성 향상과 고품질 축산물 생산을 위한 긍정적인 효과를 얻기 위해 각종 항생제, 생균제, 효소제, 유기산제, 향미제, 감미제, 항산화제 및 기타 기능성 물질 등을 포함할 수 있다.
본 발명에 따른 사료 조성물은 가축의 사육 목적에 따라 적정 체중이 되는 통상적인 기간 동안 급여시킬 수 있다.
본 발명에서는 상기 가축용 사료 조성물을 가축에 급여시키는 것을 특징으로 하는 가축 사육방법을 제공한다.
본 발명에 따른 가축용 사료 조성물을 이용하여 가축에 급여시킬 경우 기존의 사료와 비교하여 일당증체량 및 사료효율이 증가하여 실질적인 생산량 향상 효과를 얻을 수 있으며, 특히 가축 혈중 중성지방 수치는 감소하면서도 지방산 수치는 증가하였다.
이하 본 발명의 바람직한 실시예와 실험예를 제시한다. 그러나 하기한 예는 본 발명의 바람직한 일 예일 뿐 이러한 예에 의해 본 발명이 한정되는 것은 아니다.
실시예 1 : 사료 첨가용 유지 조성물의 제조
혼합지방산(400g) 및 글리세롤(92g)을 250℃에서 알칼리 촉매로 NaOH를 첨가하여 2시간 동안 에스테르화를 진행하여 산가 1 이하의 에스테르 화합물을 제조하여 본 발명에 따른 글리세라이드 조성물을 얻었다. 제조된 글리세라이드 조성물의 자방산 및 글리세라이드 분석은 하기 기재된 방법으로 분석하여 아래 표 1 및 표 2에 나타내었다.
1. 지방산 조성 분석을 위한 기체크로마토그래피
칼럼은 HP-INNOWAX(Agilent사, 미국), carrier gas는 헬륨(2.1㎖/분)이며 오븐온도는 150∼260℃, 불꽃이온화검출기(FID) 275℃인 조건에서 샘플을 25g/ℓ농도로 주입하여 분석하였다.
2. 글리세라이드 조성 분석을 위한 액체 크로마토그래피
글리세라이드 조성 분석을 위한 (액체 크로마토그래피) 칼럼은 수펄코실 LC-Si(Supelcosil LC-Si) 5㎛, 25㎝[Aupelco사, 미국]를 사용하며, 이동상 용매는 용매A(벤젠 70: 클로로포름 30: 아세트산 2)와 용매B(에틸 아세테이트)를 쓰고, 샘플의 주입 농도는 1㎎/㎖(클로로포름 용매)하여, 증발 산란광 검출기(ELSD)를 사용하여 유량속도 2.3㎖/분 조건에서 분석하였다.
3. 글리세라이드의 위치 이성체 분석을 위한 액체 크로마토그래피
칼럼은 ChromSpher Lipids, 5㎛, 25㎝[varian, USA]를 사용하며, 이동상 용매는 0.5% 아세토나이트릴(Acetonitrile)을 포함하는 노르말-헥산(n-hexane)을 사용하며, 샘플의 주입 농도는 1㎎/㎖ (클로로포름 용매)하여 증발 산란광 검출기(ELSD)를 사용하여 유량속도 2.3㎖/분 조건에서 분석하였다.

실험예 1 : 이유자돈에 대한 유지 조성물 평가
시험동물 및 시험설계
3원 교잡종[(Landrace×Yorkshire)×Duroc] 이유자돈 160두를 공시하였고, 시험 개시시 체중은 6.84 ± 0.87kg 이었으며, 사양시험은 6주간 실시하였다. 시험사료는 양성 대조군(Positive control, PC)과 음성 대조군(Negative control, PC-100kcal down) 2종류의 사료를 준비하였고, 이때 각 군간의 동물의 처리는 다음과 같고 처리당 4반복, 반복당 5두씩 임의 배치하였다: (%는 중량%임)
- 양성 대조군: 사료

-실험군 2: 사료+ 실시예 1의 유지 조성물 0.2 중량%

시험사료 및 사양관리
시험사료는 NRC (2012) 요구량에 따라 배합한 옥수수-대두박 위주의 일반사료로 자유채식토록 하였으며, 물은 자동급수기를 이용하여 자유로이 먹을 수 있도록 조절하였다.
조사 항목
(1) 생산성
일당증체량은 개시시, 2주 및 종료시 (6주)에 처리구별로 체중을 측정하였다. 일당사료섭취량은 체중 측정시 사료급여량에서 잔량을 제하여 계산하였고, 사료효율은 증체량을 사료섭취량으로 나누어 산출하였다. 그 결과는 표 3에 나타내었다.
(2) 영양소 소화율
영양소 소화율은 2주 및 종료시(6주)에 산화크롬(Cr2O3)을 표시물로서 0.2% 첨가하여 7일간 급여 후 항문마사지법으로 분을 채취하였다. 채취한 분은 60℃의 건조기에서 72시간 건조한 후 Willey mill로 분쇄하여 분석에 이용하였다. 사료의 일반성분과 표시물로 혼합된 Cr은 AOAC (2000)의 방법에 준하여 분석하였다. 그 결과는 표 4에 나타내었다.
(3) 혈액특성
혈액채취는 종료시(6주)에 각 처리구당 8두를 임의 선발하여 경정맥(Jugular vein)에서 K3EDTA Vacuum tube(Becton Dicknson Vacutainer Systems, Franklin Lakes, NJ)를 이용하여 혈액을 2 mL 채취하여 분석하였고, 자동혈액분석기 (ADVID 120, Bayer, USA)를 이용하여 백혈구(WBC), 적혈구(RBC) 및 림프구를 조사하였다. 또한 혈청 생화학적 검사는 각각 경정맥에서 Vacuum tube (Becton Dickinson Vacutainer Systems, Franklin Lakes, NJ)를 이용하여 혈액 5mL 채취 후 4℃에서 3,000 rpm으로 15분간 원심분리하여 얻은 혈청을 enzymatic colorimetric method (Allain et al., 1974)에 의하여, 총 콜레스테롤 및 HDL 콜레스테롤 농도는 검사시약 (Cholesterol Kit No. 352, Sigma Chemical, St. Louis, MO, USA)에 반응시켜 자동생화학 분석기 (HITACHI 747, Japan)를 이용하여 측정하였고, LDL 콜레스테롤 농도는 Naoyuki와 Yoshiharu (1995)의 방법에 따라 측정하였다. 그 결과는 표 5에 나타내었다.
(4) 분내 미생물 조성
시험 종료시(6주)에 처리구별로 8두를 선발하여 항문마사지법으로 분을 채취한 뒤, 실험시까지 -20℃에서 냉동 보관하였으며, 이후 멸균된 생리식염수에 현탁하여 균질화 시킨 다음 103에서 107까지 단계 희석하여 생균수 측정용 시료로 사용 하였다. 실험처리에 의한 돈분 내의 Lactobacillus와 E.
coli의 균수를 측정하기 위해 Lactobacillus에는 MRS agar, E.
coli에는 MacConkey agar (Difco, USA)를 사용하였고, 37℃에서 38시간 배양 후 균수를 측정하였다. 그 결과는 표 6에 나타내었다.
(5) 분내 악취물질 발생
이유자돈의 분내 악취발생은 암모니아, 총 메캅탄 및 황화수소의 함량을 측정하기 위하여, 시험 종료시 (6주)에 각 처리구에서 동일한 시간 동안 배설된 분을 모든 이유자돈으로부터 채취하여 분석에 사용하였다. 암모니아, 총 메캅탄, 황화수소 및 휘발성지방산 측정은 분 300 g을 취하여 2,600 mL 부피의 밀봉된 플라스틱 용기에 넣고 24시간 발효시킨 후 실온에 5일간 보관하면서 Gastec(Model GV-100, Gastec, Japan)을 사용하여 보관 5일 경과시 측정하였다. 그 결과는 표 7에 나타내었다.
(6) 분변지수
분변지수는 전체 시험기간 동안 매일 측정하여 다음과 같은 지수로 (Score = 1 hard, dry pellet; 2 firm, formed stool; 3 soft, moist stool that retains shape; 4 soft, unformed stool that assumes shape of container; 5 watery liquid that can be poured)수치화 하였다. 그 결과는 표 8에 나타내었다.
결과
(1) 생산성
Phase 1의 일당증체량에 있어서 실험군 2 및 실험군 3 처리구가 음성 대조군 및 비교군 처리구보다 유의적으로 높게 나타났다. Phase 2의 일당증체량에 있어서 실험군 1, 실험군 2, 실험군 3, 비교군 2 및 비교군 3이 음성 대조군 보다 유의적으로 높게 나타났다. 전체시험기간의 일당증체량에 있어서 실험군 3 처리구가 음성대조군 및 비교군 1 처리구보다 유의적으로 높게 나타났고, 사료효율에 있어서는 실험군 1 및 실험군 3 처리구가 음성 대조군 처리구 보다 유의적으로 높게 나타났다.

실험예 2 : 육계에 대한 유지 조성물 평가 1
시험동물 및 시험설계
1 일령 ROSS 308 (암, 수) 510수 공시하였고, 시험 개시 체중은 41 ±0.17g으로 사양시험은 5주간 실시하였다. 각 군간의 동물의 처리는 다음과 같고, 처리당 6반복, 반복당 17수씩 완전 임의 배치하였다.
- 양성 대조군: 사료
- 음성 대조군: 100 kcal 감량된 사료
- 실험군 1: 100 kcal 감량된 사료+실시예 1의 유지 조성물 0.075 중량%
- 실험군 2: 100 kcal 감량된 사료+실시예 1의 유지 조성물 0.10 중량%
- 실험군 3: 100 kcal 감량된 사료+실시예 1의 유지 조성물 0.15 중량%
시험사료와 사양관리
시험사료는 NRC (1994) 요구량에 따라 배합한 옥수수-대두박 위주의 가루형태 사료를 급여하였다. ROSS 308 병아리를 3단 케이지에서 사육하였으며, 처리구별 위치를 조절하였고, 사료와 물은 자유 채식토록 하였다.
조사항목 및 방법
(1) 생산성
증체량은 개시시, 1주 3주 및 종료 시(5주)에 처리구별로 체중을 측정하였다. 사료섭취량은 체중 측정시 사료급여량에서 잔량을 제하여 계산하였고, 사료요구율은 사료섭취량을 증체량으로 나누어 산출하였다.
(2) 육질 특성
시험 종료 시 (5주) 처리구별 임의로 6수씩 선별하여 경골탈퇴 방법으로 도살 한 다음 간, 비장, F낭, 복강지방, 가슴근육 및 근위의 무게를 측정하여 생체중에 대한 비율로 계산하였다. pH는 pH meter(77P, Istek, Korea)를 사용하여 측정하였으며, 육색은 색차계(Model CR-410. Minolta Co., Japan)를 이용하여 각 가슴육 샘플 1개당 2회 반복하여 측정하였다. 이때 표준 색판은 L*=89.2, a*=0.921, b*=0.783으로하였다. 저장감량(drip loss)은 시료를 4 cm 두께의 일정한 모양으로 정형한 후 polyethylene bag에 넣어 4℃ 냉장실에서 7일간 보관하면서 1일, 3일, 5일 및 7일 후 발생되는 감량을 측정하였다.
(3) 혈액 특성
혈액채취는 각 처리당 6마리를 임의 선발하여 종료 시(5주)에 경정맥 (Jugular)에서 K3EDTA Vacuum tube(Becton Dicknson Vacutainer Systems, Franklin Lakes, NJ)를 이용하여 혈액을 2 mL 채취하여 분석하였고, 자동혈액분석기(ADVID 120, Bayer, USA)를 이용하여 FFA(free fatty acid)를 조사하였다. 또한 혈청생화학적 검사는 종료 시(5주)에 경정맥에서 Vacuum tube(Becton Dickinson Vacutainer Systems, Franklin Lakes, NJ)를 이용하여 혈액을 5 ml 채취하고 4℃에서 3,000 rpm으로 15분간 원심 분리하여 얻은 혈청을 분석에 이용하였다. 분리된 혈청의 Protein, BUN, Creatinine, Triglyceride 및 Glucose의 함량은 nephelometer 방법으로 nephelometer(Behring, Germany) 분석기계를 이용하여 분석하였다.
(4) 분내 악취 발생 정도
육계의 분내 가스발생은 암모니아, 황화수소 및 총 메캅탄의 발생을 측정하기 위하여, 시험 종료 시 (5주)에 각 처리구에서 동일한 시간 동안 배설된 분을 채취하여 분석에 사용하였다. 암모니아, 황화수소 및 총 메캅탄의 측정은 분 300g을 취하여 2,600 mL 부피의 밀봉된 플라스틱 용기에 넣고 24시간 발효시킨 후 실온에 5일간 보관하면서 Gastec(Model GV-100, Gastec, Japan)을 사용하여 보관 5일 경과시 측정하였다.

결 과

육계 사료 내 본 발명의 유지 조성물의 첨가 급여가 육계의 생산성에 미치는 영향은 아래 표 9에 나타내었다. 7-21일차 증체량(BWG)에 있어 실험군 1, 실험군 2 및 실험군 3 첨가 처리구가 음성 대조군보다 유의적으로 높게 나타났으며, 유지 조성물 첨가 수준이 증가할수록 선형적으로 증가하였다. 사료 요구율(FCR)에 있어서 실험군 1, 실험군 2 및 실험군 3이 음성 대조군보다 유의적으로 낮게 나타났으며, 유지 조성물 첨가 수준이 증가할수록 선형적으로 감소하였다. 21-35일차 사료 요구율에 있어 실험군 1이 음성 대조군보다 유의적으로 낮게 나타났다. 전체 시험 구간 증체량에 있어 설험군 1이 음성대조군보다 유의적으로 높게 나타났으며, 본 발명의 유지 조성물 첨가 수준이 증가할수록 선형적으로 증가하였다. 사료 요구율에 있어서는 실험군 1이 음성 대조군보다 유의적으로 낮게 나타났으며, 유지 조성물 첨가 수준이 증가할수록 선형적으로 감소하였다.

(2) 도체 및 육질특성
육계 사료 내 본 발명의 유지 조성물의 첨가 급여가 육계의 도체 및 육질특성에 미치는 영향은 아래 표 10에 나타내었다. 저장감량(drip loss)에 있어 1일째에 음성 대조군이 실험군 2보다 유의적으로 높게 나타났고, 유지 조성물 첨가 수준이 증가할수록 선형적으로 감소하였다. 저장감량 5일째에 있어 음성 대조군이 실험군 3보다 유의적으로 높게 나타났다. 저장감량 7일째에 있어 처리구간 유의적인 차이는 나타나지 않았지만, 유지 조성물 첨가 수준이 증가할수록 선형적으로 증가하였다. 도체 특성에 pH, 육색, 간, 비장, F낭, 가슴근육, 복강지방 및 근위에 있어 처리구간 유의적인 차이는 나타나지 않았다.

실험예 3: 육계에 대한 유지 조성물 평가 2
Ross 308 1일령 600수를(4처리×5반복 반복당 30수)에 대해 실시예 1의 유지 조성물을 급여하였다. 이때 시판 육계 사료에 유지 조성물은 0.1, 0.2 및 0.4 중량%의 함량으로 36일간 급여하였다. 전기사료(ME, 3080 kcal/kg; CP, 20.5%)는 20일간 급여하였고, 후기사료(ME, 3100 kcal/kg; CP, 19.5%)는 16일간 급여하였다.
이때 각 군간의 동물의 처리는 다음과 같다: (%는 중량%임)

상기 표 14에 나타낸 바와 같이, 실시예 1의 유지 조성물 첨가에 따라 체중이 증가하는 경향이 관찰되었다. 실험 전기의 일당 증체량은 유지 조성물 첨가에 따라 선형적으로 유의한 증가를 나타내었고, 사료 요구율도 선형적으로 개선되는 변화를 나타내었다.
혈액에서 실시예 1의 유지 조성물 첨가에 따라 유리 지방산 농도가 선행적으로 증가하였고, 대조적으로 혈중 중성지방 농도는 선형적으로 유의하게 감소하였다.
실험예 4: 넙치에 대한 유지 조성물 평가 1
각 수조 당 넙치 40마리씩 분리하여 사육하였고, 각 수조의 넙치 평균 체중은 13.4 g이었다. 사료는 오전 8시 오후 18시 포식량 공급하였다. 수조의 수온은 15 내지 21 ℃ 이었고, 12주 동안 사육하였다. 이때 각 군간의 동물의 사료 처리는 다음 표 15과 같다. 성장률과 사료이용효율을 조사하기 위하여 일간성장율(specific growth rate (%)), 사료섭취량 (feed intake), 사료 전환율(feed conversion ration), 단백질 이용 효율(protein efficiency ratio)은 다음과 같이 구하였고, 그 결과는 결과는 도 1 내지 도 4에 나타내었다L

도 1 내지 도 4를 참조하면, 본 발명의 유지 조성물로 넙치 사료 내 지방원의 50 중량%로 대체하여 사육한 결과, 대조군과 실험군 사이 실험 종류시 넙치 체중, 일간성장율(specific growth rate(%))과 생존율에 유의한 차이가 없었다.
실험예 5: 넙치에 대한 유지 조성물 평가 2
각 수조 당 넙치 50마리씩 분리하여 사육하였고, 각 수조의 넙치 평균 체중은 6.8 g이었다. 사료는 오전 8시 오후 18시 포식량 공급하였다. 수조의 수온은 23 내지 28 ℃ 이었고, 8주 동안 사육하였다. 이때 각 군간의 동물의 사료 처리는 다음 표 16과 같다. 결과는 도 5 내지 도 8에 나타내었다.

도 5 내지 도 8에서 나타낸 바와 같이, 실시예 1의 유지 조성물 첨가에 따라 넙치의 최종 체중이 증가함을 확인할 수 있었다.
실험예
6: 넙치에 대한 유지 조성물 평가 3
120L 수조 당 넙치 30마리씩 분리하여 사육하였고, 각 수조의 넙치 평균 체중은 27.6 g이었다. 사료는 포식량 공급하였다. 이때 각 군간의 동물의 사료 처리는 다음 표 17과 같다. 결과는 표 18에 나타내었다.

사육실험이 종료된 후, 각 수조 당 무작위로 12마리씩(실험구당 84마리) 선정하여 Edwardsiella
tarda (ATCC 15947, Korea Collection for Type Cultures) 현탁액(1×108 CFU/ml)을 복강에 주입하였다. E.
tarda는 제주대학교 해양생명과학과 소속 해양바이러스학 연구실에서 제공받았다. 균이 주입된 넙치는 64 L 플라스틱수조에 3반복으로 분주되었으며, 어류의 행동 및 폐사를 하루 3번씩 관찰하였으며, 8일 동안 진행되었다. 그리고 생존율을 구하여 그 결과는 도 9에 나타내었다.
도 9에 나타낸 바와 같이, 본 발명의 유지 조성물을 투여한 군의 생존율이 높아 병원세균에 대한 항미생물 효과를 나타냄을 확인할 수 있었다.
실험예
7: 함량에 따른 체중 증가 효과 확인
3원 교잡종[(Landrace×Yorkshire)×Duroc] 비육돈 120두를 공시하였고, 시험 개시시 체중은 7.53 ± 0.83 kg 이였다. 각 처리구당 4반복으로 반복당 5두씩 임의 배치하였다, 돈방은 슬렛바닥으로 설계되었고 사료와 물을 자유 채식할 수 있도록 단구 습식급이기를 설치하였다.
이때 각 군간의 동물의 약물처리는 다음과 같다: (%는 중량%임)
- 대조군: 사료+대두유 3.0%
-실험군 1: 사료+ 대두유 1.5%+ 실시예 1의 유지 조성물 1.5 중량%
-실험군 2: 사료+ 실시예 1의 유지 조성물 3.0 중량%
-실험군 3: 사료+ 대두유 3.0%+ 실시예 1의 유지 조성물 0.1 중량%
-실험군 4: 사료+ 대두유 3.0%+ 실시예 1의 유지 조성물 0.2 중량%
-실험군 5: 사료+ 대두유 3.0%+ 실시예 1의 유지 조성물 0.4 중량%
각 군에 대한 체중은 실험시작시, 3주 후, 6주 후에 측정하고, 먹는 사료량도 측정하였다. 이를 바탕으로 일당증체량, 사료섭취량, 사료효율(G/F)를 계산하였다.

상기 표 19를 참조하면, 사료에 첨가되는 대두유의 50%를 본 발명의 유지조성물로 대체하여 투여한 실험군 1은 대조군과 가축의 성장 면에서 큰 차이를 보이지 않았다. 반면 사료에 첨가되는 대두유 전부를 본 발명의 유지 조성물로 대체하여 투여한 실험군 2의 경우 오히려 일당증체량, 사료효율이 낮았고, 대조군에 추가적으로 본 발명의 유지 조성물을 각각 0.1, 0.2, 0.4 중량% 투여한 실험군 3 내지 5는 대조군 보다 일당증체량, 사료효율이 모두 높게 나타났다.
Claims (16)
- 구성지방산 중 탄소수 14 이하의 지방산의 함량이 10 내지 90 중량%이고, 1, 3번 위치에 결합된 탄소수 14 이하의 지방산의 함량이 10 내지 90 중량%인 디글리세라이드를 50 중량% 이상 함유하는 사료용 유지 조성물.
- 제1항에 있어서, 상기 디글리세라이드는 구성지방산 중 탄소수 16 이상의 지방산의 함량이 10 내지 90 중량%인 것인 사료용 유지 조성물.
- 제1항에 있어서, 상기 디글리세라이드 중 1,3-디글리세라이드의 함량은 40 중량% 이상인 것인 사료용 유지 조성물.
- 제3항에 있어서, 상기 1,3-디글리세라이드의 구성지방산 중 탄소수 14 이하의 지방산/탄소수 16 이상의 지방산의 중량비가 2 내지 9인 것인 사료용 유지 조성물.
- 제3항에 있어서, 상기 1,3-디글리세라이드의 구성 지방산은 탄소수 14 이하의 지방산의 함량이 70 내지 90 중량%, 탄소수 16 이상의 지방산의 함량은 10 내지 30 중량%인 것인 사료용 유지 조성물.
- 제1항에 있어서, 상기 탄소수 14 이하의 지방산은카프릴산, 펠라르곤산, 카프르산, 운데카노산, 라우르산, 미리스트산 및 이들의 혼합물로 이루어진 군에서 선택되는 것인 사료용 유지 조성물.
- 제1항에 있어서, 상기 탄소수 16 이상의 지방산은팔미트산, 올레산, 리놀레산, 스테아르산 및 이들의 혼합물로 이루어진 군에서 선택되는 것인 사료용 유지 조성물.
- 제1항에 있어서, 상기 사료용 유지 조성물은전체 100 중량%가 되도록 디글리세라이드 50 내지 90 중량%와,트리글리세라이드, 모노글리세라이드, 유리 지방산 또는 이들의 혼합물 잔부를 포함하는 것인 사료용 유지 조성물.
- 제1항 내지 제8항 중 어느 한 항에 따른 사료용 유지 조성물을 유효성분으로 함유하는 성장 촉진용 사료 첨가제.
- 제1항 내지 제7항 중 어느 한 항에 따른 사료용 유지 조성물 및 배합사료를 포함하는 가축용 사료 조성물.
- 제10항에 있어서, 상기 가축용 사료 조성물은 돼지, 닭, 오리, 메추라기, 거위, 꿩, 칠면조, 소, 젖소, 말, 당나귀, 양, 염소, 개, 고양이, 토끼, 양식 어류 또는 새우 사육용인 것인 가축용 사료 조성물.
- 제10항에 있어서, 상기 사료용 유지 조성물의 함량은 상기 사료용 유지 조성물의 함량은 배합사료 총중량을 기준으로 0.01 내지 5.0 중량%인 가축용 사료 조성물.
- 제10항의 가축용 사료 조성물을 급여하는 것을 특징으로 하는 가축 사육방법.
- 제1항 내지 제7항 중 어느 한 항에 따른 사료용 유지 조성물로 배합사료 내 지방원의 10 중량% 이상으로 대체한 가축용 사료 조성물.
- 제14항에 있어서, 상기 가축용 사료 조성물은 돼지, 닭, 오리, 메추라기, 거위, 꿩, 칠면조, 소, 젖소, 말, 당나귀, 양, 염소, 개, 고양이, 토끼, 양식 어류 또는 새우 사육용인 것인 가축용 사료 조성물.
- 제14항의 가축용 사료 조성물을 급여하는 것을 특징으로 하는 가축 사육방법.
