본 발명은 인간 인터페론 -베타 변이체가 접합된 면역사이토카인 및 이의 제 조방법에 관한 발명으로, 보다 구체적으로는 천연형 인터페론-베타보다 활성 및 기 능이 우수한 인터페론 -베타 변이체와 항체가 결합된 면역사이토카인 및 이의 제조 방법에 관한 발명이다.
【배경기술】
본 출원은 2015년 3월 3일에 출원된 대한민국 특허출원 제 10-2015-0030037호 를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다. 약제에 있어서 면역요법은 면역 시스템을 조절하여 예방 및 /또는 치료 목표 를 달성하기 위한 개념에 기초하는 다수의 치료 전략들을 나타낸다. 지난 수년간, 면역요법은 여러 병리상태들의 치료 또는 예방을 위해 사용되 어왔다. 모노클로날 항체들을 생산하기 위한 세포 융합 기법이 개발된 이래, 다수 의 모노클로날 항체들아 연구자들에 의해 제조되었다. 그 후, 인간 모노클로날 항 체를 생산하기 위한 B 세포 하이브리도마 기법 및 EBV 하이브리도마 기법을 포함한 다른 기법들이 모노클로날 항체의 제조를 위해 개발되었다. 모노클로날 항체 (Mab)는 거의 모든 에피토프를 표적으로 하기 위하여 개발될 수 있다. 특정 세포 /분자에 대한 특이적인 인식 및 결합의 성질은 다양한 질환 상 태들에 대한 진단 및 치료 시약으로서 Mab의 개발을 고무시켰다. 재조합 DNA 기법 은 인간에게 투여하도록 키메라 또는 인간화 항체를 제조하는데 사용되었다. 현재, 여러 모노클로날 항체들이 암, 감염성 질환, 면역 질환 등의 치료를 위해 예를 들 어 RITUXAN® , HERCEPTIN® , AVASTIN®등으로서 시판되고 이용된다. 모노클로날 항체는 표적화된 분자이며, 병리 조직과 같은 특이작 영역 (세포, 조직 등) 내에 편재화될 수 있다. 이러한 성질에 의해, 병리 조직부위에서 특이적

CI in I隱 unother 3 : pp 27-87) . 인간 인터페론-베타도 당단백질의 일종인데, 이러한 단백질에 연결된 당쇄 부분이 그 단백질의 활성에 중요한 역할을 한다. 따라서 당단백질의 경우 당쇄를 부가시키게 되면 그 활성이 증가하는 경우가 있을 수 있다. 전술한 바의 관점에서 당단백질인 인간 천연형 인터페론-베타에 당쇄를 도입 시켜 그 활성이나기능이 증가 또는 향상된 인간 인터페론 -베타 변이체가 한국 등 톡특허 10-0781666호에 개시되어 있다. 이에, 천연형 인터페론-베타의 약리효과보다 우수한 효능을 나타내는 인간 인터페론 -베타 변이체를 표적화 치료에 이용하기 위해 항체와 접합한 면역사이토카 인에 대한 개발이 필요하며 이를 높은 수율로 얻을 수 있는 제조방법이 요구된다.
【발명의 상세한 설명】
【기술적 과제】
본 발명의 발명자들은, 인간 천연형 인터페론-베타에 당쇄를 도입시켜 그 활 성이나 기능이 증가 또는 향상된 인간 인터페론 -베타 변이체가 항체와 결합된 면역 사이토카인을 발명하고, 이러한 면역사이토카인의 숙주세포에서의 발현량이 천연형 인터페론-베타와 항체가 결합된 면역사이토카인에 비해 현저히 향상되어 있음올 발 견하고 본 발명을 완성하게 되었다. 따라서, 본 발명의 목적은 (a) 인간 인터페론 -베타 변이체; 및 (b) 상기 인

단계, (b) 제공된 세포를 배양하는 단계 및 (c) 상기 세포 또는 배양 배지로부터 면역사이토카인을 회수함으로써 면역사이토카인을 제조하는 단계를 포함하는 면역 사이토카인 제조방법을 제공하는 것이다. 이하 본 발명을 상세히 설명한다. 사이토카인의 치료 잠재력은 저농도에서도 일어나는 중증 부작용에 의해 종 종 제한을 받으며, 이로써 표적 조직에 층분한 사이토카인 농도가 존재하지 못하게 한다. 이에, 사이토카인의 치료 잠재력을 증가시키고 정상 조직을 독성 효과로부터 보호하기 위해 항체를 사용하여 사이토카인을 표적화하고 이를 질병 부위에 전달하 는 것이 면역사이토카인에 의해 가능하다. 본 발명에 따른 면역사이토카인은 인간 인터페론-베타의 변이체로, 천연형 인터페론-베타에 당쇄를 도입시켜 그 활성이나 기능이 증가 또는 향상된 사이토카 인이다. 본 발명의 발명자들은 상기 인간 인터페론 -베타 변이체를 항체와 결합하여 면역사이토카인의 형태로 다발성 경화증, 바이러스성 질환 등의 표적치료에 이용할 수 있다면, 천연형 인터페론 -베타 7} 접합된 면역사이토카인과 비교해 치료효과가 우수할 수 있음에 착안하여 본 발명을 완성하였다. 따라서, 본 발명은 (a) 인간 인터페론 -베타 변이체; 및 (b) 상기 인간 인터 페론 -베타 변이체에 직접 또는 간접적으로 공유 결합된 항체 또는 이의 단편을 포 함하는 면역사이토카인으로서, 상기 인간 인터페론—베타 변이체는 ( i ) 서열번호 1 에 개시된 아미노산 서열 전체를 포함하는 폴리펩티드, ( i i ) 서열번호 1에 개시된 아미노산 서열의 실질적인 부분을 포함하는 폴리펩티드, ( i i i ) 상기 (a) 또는 (b) 의 폴리펩티드와 실질적으로 유사한 폴리펩티드로 구성된 군에서 선택되고 인간 인 터페론 -베타 활성을 지니는 폴리펩티드로서, N-연결형 당쇄를 포함하는 폴리펩티드 인 면역사이토카인을 제공한다. 천연형 인간 인터페론-베타에 비하여 그 활성 또는 기능이 증가 또는 향상된 상기 인간 인터페론 -베타 변이체는, 천연형 인간 인터페론 -베타 또는 천연형 인간 인터페론 -베타 변이체에서 그 아미노산 서열의 C-말단에 글리신-아스파라긴-이소루 이신-트레오닌 -발린 서열을 포함하고, 상기 부가 서열의 아스파라긴 잔기에 N-연결

인간 인터페론 -베타 변이체는 천연형 인터페론-베타에 비하여 향상 또는 증 가된 항바이러스 활성, 세포 성장 억제 활성, 면역 조절 기능, 생체 내에서의 반감 기를 보인다.
상기 서열번호 2는 인터페론 β 변이체인 R27T의 아미노산 서열이며, 서열번 호 3은 서열번호 1의 27번 아미노산이 S로 치환된 인터페론 β 변이체 R27S이다. 또 한, 상기 서열번호 4는 종결코돈 뒤에 GNITV 아미노산을 포함하는 인터페론 β 변이 체 GNITV의 아미노산 서열이다. 상기 서열번호 1 내지 4는 Ν말단에 개시코돈 포함 하는데, 서열번호 1 내지 4의 단백질은 다른 링커와 연결될 때 (링커의 C말단과 카 비페론의 Ν말단이 연결) 생략될 수 있다. 즉, 서열번호 1 내지 4 단백질 개시코돈 의 염기서열 ATG또는 아미노산 서열 메티오닌은 생략될 수 있다. 한편, 상기 '인간 인터페론 -베타 변이체' 는 한국 등록특허 10-0781666호에 상세하게 기록되어 있다.
본 발명에서의 항체는 광의로 사용되며, 단클론성 항체, 다클론성 항체, 다 중특이성 항체 (예를 들면, 이중특이성 항체), 및 항체 단편 (목적하는 항원 -결합 활 성을 나타내는 한)을 포함하나 이로 한정되지는 않는 다양한 항체 구조를 포함한 다. 천연 항체는 다양한 구조를 갖는 분자이다. 예를 들면, 천연 IgG 항체는 여기 서 다이설파이드-결합된 2개의 동일한 경쇄 및 2개의 동일한 중쇄로 이루어진, 약 150,000 달톤의 사량체 당단백질이다. 포함되나, 이에 제한되지 않는다. 본 발명은 상기 기술된 항체의 사용을 제한할 필요가 없고, 당업자에게 알려 진 바와 같은 다른 그러한 항체는 본원에 기술된 조성물 및 방법에서 사용될 수 있 다. 한편, IFN-β가 처음 다발성 경화증 치료제로 도입된 것은 항바이러스 효과 를 얻기 위해서였지만, 이후 많은 연구를 통해 그 작용 기전이 밝혀지고 있다. 우 선 IFN-β는 IFN- α에 의해 유도되는 HLA class I I 분자의 활성화를 억제함으로써 항원 발현을 저해하고, T 세포의 활성화를 막는다. 또한 IFN— β는 Co— st imul atory molecules을 비활성화시켜 T 세포의 활성화를 억제하며 (28 , 29), 자가반응성 T 세 포가 자연사 (apoptosi s)하도록 유도하기도 한다. 뇌 -혈관 장막 (brain-blood barrier)에 대한 IFN-β의 효과는 T 세포가 혈관내피세포에 부착되는 것을 억제하 고, 뇌로 들어가는 능력을 감소시키는 것으로 생각되며, 실제로 MRI 연구를 보면 IFN-β치료를 받은 약 90%의 다발성 경화증 환자에서 조영 증강 병변이 감소하였다 는 보고가 있다. 이에, 본원발명에 따른 인간 인터페론 -베타 변이체는 천연형 인터페론 -베타 와 비교하여 그 활성 및 기능이 향상되어 있기 때문에, 다발성 경화증 특이적 항원 을 인식하는 항체와 결합된 면역사이토카인의 형태로써 타켓치 ¾에 사용된다면 인 터페론 -베타 단독제제의 치료효과보다 더 우수한 효과를 나타낼 수 있다. 다발성 경화증 특이적 항원 및 항체는 예를 들어, CD20과 이를 인식하는 항 체인 r i tuximab , CD52 와 이를 인식하는 단클론항체인 alemtuzumab 및 인터루킨 -2 의 알파 수용체와 이를 인식하는 dacl i zumab을 들 수 있으나, 이에 제한되는 것은 아니다. 본 발명은 또한 상기 인간 인터페론 -베타 변이체는 펩타이드 링커에 의해 상 기 항체 또는 이의 단편에 결합되어 있는 면역사이토카인을 제공한다. 펩타이드 링 커는 아미노산 또는 아미노산과 유사한 물질이 서로 펩타이드 결합에 의해서 둘 이 상이 연결된 짧은 단편의 아미노산 또는 아미노산 유사체로 둘 이상의 별개의 물질 을 서로 연결시켜 주는 역할을 하는 분자를 말한다. 글리신, 세린, 알라닌 등이 주
요 구성 아미노산으로 이용되어 글리신 -세린 링커, 글리신-세린-알라닌 링커 등을 사용할 수 있으며, 본 발명의 바람직한 구현예에 따르면, 상기 링커는 서열번호 5 내지 서열번호 11의 어느 하나의 아미노산 서열로 이루어지거나 이를 포함할 수 있 다. 본 발명의 면역사이토카인은 바람직하게는 인간 인터페론 -베타 변이체와 항 체 또는 그 단편의 폴리펩티드 사이에 삽입된 가요성 링커 서열을 포함할 수 있다. 상기 링커 서열은 상기 항체 또는 그 단편의 상기 인간 인터페론 -베타 변이체에 대 한 유효한 위치 결정을 허용하여 상기 두 도메인 모두의 작용 활성을 허용한다. 상기 링커는 천연 유래의 펩타이드 링커 또는 합성 유래의 펩타이드 링커를 의미한다. 상기 펩타이드 링커는 선형 아미노산 쇄로 구성되고, 이때 20종의 천연 발생 아미노산은 단량체 빌딩 블록이다. 링커는 반복적 아미노산 서열을 가질 수 있거나 천연 발생 폴리펩티드, 예컨대, 힌지 기능을 갖는 폴리펩티드의 서열을 가 질 수 있다. 모든 펩타이드 링커가 핵산 분자에 의해 코딩될 수 있으므로 재조합적 으로 발현될 수 있다. 링커는 그 자체가 펩타이드이기 때문에 인간 인터페론 -베타 변이체와 항체 또는 그 단편은 펩타이드 결합을 통해 링커에 연결된다. 링커는 펩타이드 결합에 의해 함께 연결된 아미노산, 바람직하게는 펩타이 드 결합에 의해 연결된 1 내지 20개의 아미노산으로 구성되며, 이때 아미노산은 20 개의 천연 아미노산 중에서 선택된다. 이들 아미노산 중 하나 이상은 당해 분야의 숙련자에 의해 이해되는 바와 같이 글리코실화된다. 이에 제한되는 것은 아니지만 바람직하게는, 1 내지 20개의 아미노산은 글리신, 알라닌, 프를린, 아스파라긴, 글 루타민 및 리신 중에서 선택된다. 적합한 링커로는 예를 들어, 절단가능한 링커 및 절단불가능한 링커를 포함 한다. 절단가능한 링커는 전형적으로 세포내 조건하에서 쉽게 절단된다. 적합한 절 단가능한 링커로는 예를 들어, 세포내 프로테아제, 예컨대 리소좀 프로테아제 또는 엔도솜 프로테아제에 의해 절단가능한 펩티드 링커를 포함한다. 상기 링커는 예를 들어, 상기 항체의 중쇄 C-말단에 링커의 N-말단이 연결된 다. 항체의 중쇄 C-말단에의 연결은 바람직하게는 본 발명의 항체를 발현하는 발현
백터에 링커 서열을 코딩하는 염기서열이 단백질 발현 프레임이 일치되어 연결되어 발현백터에 의해서 발현되는 항체에 직접적으로 연결될 수 있다. 또한, 상기 링커 는 항체의 경쇄 C-말단에 연결되거나, 항체의 경쇄 C-말단과 중쇄 C-말단에 모두 연결될 수도 있다. 또한, 상기 링커의 C-말단에 본 발명의 인터페른 베타 변이체의 N말단이 연결된다. 본 발명의 펩타이드 링커는 당업계에 공지된 펩타이드 링커일 수 있으나, 바 람직하게는 글리신 -세린 링커 또는 서열번호 5 내지 서열번호 11의 아미노산 서열 을 포함하는 펩타이드 링커일 수 있다. 바람직하게는, gly-ser 링커, 예를 들어, (GlyxSery)z 형 (x는 1 내지 5의 정 수, y는 1 내지 2의 정수이며, z는 1 내지 6의 정수) , 예를 들어 (gly4seri)3 또는 (gly3ser2)3올 들 수 있으며, 보다 바람직하게는 GGGGS 또는 GGGGSGGGGSGGGSG 의 아 미노산서열로 표시되는 링커일 수 있으나 이에 제한되는 것은 아니다. 본 발명은 또한 상기 인간 인터페론 -베타 변이체 폴리펩티드의 아미노산 서 열은 상기 항체 또는 이의 단편의 아미노산 서열에 대해 중쇄 C—말단, 경쇄 C-말단 또는 중쇄 및 경쇄 C-말단 위치에 있는 것을 특징으로 하는 면역사이토카인을 제공 한다. 상기 인간 인터페론 -베타 변이체의 아미노산 서열은 항체 또는 이의 단편의 아미노산 서열에 대해 중쇄 C-말단, 경쇄 C-말단 또는 중쇄 및 경쇄 C-말단 위치에 있을 수 있고, 바람직하게는 항체 또는 이의 단편의 아미노산 서열에 대해 C-말단 위치에 있다.
본 발명은 또한 상기 면역사이토카인은 서열번호 12, 13, 15 또는 17로 이루 어진 군 중 하나의 아미노산 서열을 포함하는 것을 특징으로 하는 면역사이토카인 을 제공한다. 본 발명은 (a) 서열번호 2 내지 서열번호 4 중 어느 하나로 표시되는 인간
인터페론 -베타 변이체; (b) 서열번호 5 내지 서열번호 11 증 어느 하나로 표시되는 펩타이드 링커; 및 (c) 항체 또는 이의 단편을 포함하는 면역사이토카인을 제공한 다. 본 발명은 또한 상기 면역사이토카인을 인코딩하는 폴리뉴클레오티드를 제공 한다. 상기 폴리뉴클레오티드는 본 발명의 인간 인터페론 -베타 변이체와 항체 또는 그 단편이 결합된 면역사이토카인의 펩타이드를 암호화 하는 것이면 제한없이 사용 될 수 있으며, DNA, cDNA 및 RNA 서열을 모두 포함한다. 바람직하게는, 상기 폴리 뉴클레오티드는 서열번호 3으로 표시되는 아미노산 서열을 갖거나 또는 상기 아미 노산 서열과 적어도 70% 이상의 상동성 (homology)이 있는 아미노산 서열을 갖는 펩 타이드를 암호화 하는 물질을 말하며, 이는 천연에서 분리되거나 당업계의 공지돤 유전공학적인 방법에 의해 제조될 수 있다. 본 발명은 또한 상기 폴리뉴클레오티드를 포함하는 백터를 제공한다. 상기 백터는 당업자들이 당업계에 알려진 어떠한 방법에 따라 본 발명의 폴 리뉴클레오티드를 백터내로 삽입하여 적절한 전사 /해독 조절서열을 이용하여 본 발 명의 면역사이토카인을 발현하도록 제조된 발현백터를 말한다. 본 발명에 따라 클로닝된 상기 폴리뉴클레오타이드 서열은 적절한 발현 조절 서열에 작동 가능하게 연결될 수 있으며, 상기 작동 가능하게 연결된 유전자 서열 과 발현 조절서열은 선택 마커 및 복제 개시점 (repl icat ion origin)을 같이 포함하 고 있는 하나의 발현 백터 내에 포함될 수 있다. '작동가능하게 연결 (operably l inked) ' 된다는 것은 상기 폴리뉴클레오타이드 서열이 발현 조절 서열에 유전자 발현을 가능하게 하는 방식으로 연결된 것을 의미한다. 상기 '발현 조절 서열 (expression control sequence) '이란 특정한 숙주세포에서 작동 가능하게 연결된 폴리뉴크레오티드 서열의 발현을 조절하는 DNA 서열올 의미한다. 그러한 조절 서열 은 전사를 실시하기 위한 프로모터 , 전사를 조절하기 위한 임의의 오퍼레이터 서 열, 적합한 mRNA 리포좀 결합 부위를 코딩하는 서열, 및 전사 및 해독의 종결을 조 절하는 서열 등으로 이루어진 군에서 선택된 하나 이상을 포함할수 있다.
상기 발현 백터의 모백터로 사용되는 백터는 특별한 제한이 없으며, 이 발명 이 속하는 기술분야에서 숙주세포로 사용되는 미생물에서의 발현을 위하여 통상적 으로 사용되는 모든 플라스미드, 바이러스 또는 기타 매개체 등이 사용 가능하다. 예컨대, 상기 플라스미드에는 대장균 유래 플라스미드 (pBR322, pBR325, pUC118 및 PUC119, pET-22b(+)), 바실러스 서브틸리스 유래 폴라스미드 (pUBllO 및 pTP5) 및 효모 유래 플라스미드 (ΥΕρ13, ΥΕρ24 및 YCp50) 등이 있으며, 상기 바이러스는 레트 로바이러스, 아데노바이러스 또는 백시니아 바이러스와 같은 동물바이러스, 배클로 바이러스와 같은 곤층 바이러스 등이 사용될 수 있으나, 이에 제한되는 ¾은 아니 다. 본 발명은 또한 상기 백터로 형질전환된 숙주세포를 제공한다. 숙주 세포는 삽입된 서열의 발현을 조절하거나 또는 바람직한 특정 방식으로 유전자 생성물올 진행시키는 것을 선택할 수 있다. 상이한 숙주 세포는 단백질의 해독 및 해독 후 프로세싱과 변형에 대한 특징적이고 특이적인 기작을 갖고 있다. 적합한 세포주 또는 숙주 시스템으로는 발현된 이종 단백질의 바람직한 변형 및 프 로세싱을 제공하는 것을 선택할 수 있다. 효모에서의 발현은 생물학적 활성 생성물 을 생산할 수 있다. 진핵 세포에서의 발현은 " 천연" 폴딩의 가능성올 증가시킬 수 있다. 본 발명의 백터를 안정되면서 연속적으로 클로닝 및 발현시킬 수 있는 숙주 세포는 당업계에 공지되어 있는 어떠한 숙주 세포도 이용할 수 있으며, 예컨대, E. coli JM109, E. coli BL21DE, E. coli DH5, E. coli RR1, E.coli LE392, E. coli B, E. coli X 1776, E. coli W3110 등이 사용될 수 있으며, 또한 아그로박테리움 A4와 같은 아그로박테리움 속 균주, 바실루스 서브틸리스 (Bacillus subtil is)와 같 은 바실리 (bacilli), 살모넬라 티피뮤리움 (Salmonel la typhi murium) 또는 세라티아 마르게센스 (Serratia marcescens)와 같은 또 다른 장내세균 및 다양한 슈도모나스 (Pseudomonas) 속 균주가 숙주세포로서 이용될 수 있다. 또한, 본 발명의 백터를 진핵 세포에 형질전환시키는 경우에는 숙주 세포로 서 , 이스트 (Saccharomycecerevisiae), 곤층 세포 및 사람 세포 (예컨대, CH0 세포주
(Chinese hamster ovary) , W138, BHK, COS-7, 293,HepG2, 3T3, RIN 및 MDCK 세포 주) 등이 이용될 수 있다. 본 발명에서 숙주세포는 바람직하게는 CH0 세포주일 수 있다. 백터를 숙주세포 내로 전달하여 숙주세포를 형질전환 시키는 방법은 공지의 방법이면 어떠한 것이든 가능하며 특별히 제한되지 아니한다. 예를 들어, 인산칼슘 침전법 (calcium phosphate precipitation) , DEAE— dextran법, 전기천공법 (electroporation), 직접 미세주입법 (direct microinjection), DNA—로딩 리포좀 (DNA loaded liposome)법, 리포펙타민— DNA복합체 (liofectamine-DNA complex)법, 세 포 음파분쇄법 (cell sonicat ion) , 고속미세분사 (high velocity microproject i le)를 이용한 유전자 폭격법 (gene bombardment), 다양이온 (polycation)법. 및 수용체 매개 형질전이법 (receptor— mediated transfection)에 의하여 형질전환 할 수 있다. 이 기술들 중 일부는 생체 내 또는 생체 외 사용을 위해 개량될 수 있다. 본 발명은 또한 (a) 숙주 세포를 제공하는 단계, (b) 제공된 세포를 배양하 는 단계 및 (c) 상기 세포 또는 배양 배지로부터 면역사이토카인을 회수함으로써 면역사이토카인을 제조하는 단계를 포함하는 면역사이토카인 제조방법을 제공한다. 형질전환 미생물의 배양은 목적 단백질인 인간 인터페론 -베타 변이체와 항체 또는 이의 단편이 결합된 면역사이토카인의 발현을 가능하게 하는 적절한 조건하에 서 수행하며, 이러한 조건은 당업자에게 익히 공지된 방법에 따라 수행할 수 있다. 형질전환 미생물은 통상적인 배양방법에 의해 대량으로 배양할 수 있다. 배양배지 로는 탄소원, 질소원, 비타민 및 미네랄로 구성된 배지를 사용할 수 있으며, 예를 들어, LB 배지 (Luria-Bertani Broth)을 사용할 수 있다. 미생물의 배양은 통상의 미생물 배양 조건상에서 가능하며 예를 들어, 온도범위 15°C 내지 45°C에서 10시간 내지 40시간 동안 배양할 수 있다. 배양액 중의 배양배지를 제거하고 농축된 균체 만을 회수하기 위해 원심분리 (centrifugation) 또는 여과 (filtration)과정올 거칠 수 있으며 이러한 단계는 당업자가 필요에 따라 수행할 수 있다. 농축된 균체는 통 상적인 방법에 따라 냉동 (frozen)하거나 또는 넁동건조 (lyophi 1 ized)하여 그 활성 을 잃지 않도록 보존할 수 있다.
형질전환 미생물 (또는 형질전환체)에서 발현시킨 단백질은 통상의 방식으로 정제될 수 있으며, 예를 들어, 염석 (예를 들어 황산암모늄 침전, 인산나트륨 침전 ), 용매 침전 (아세톤, 에탄올 등을 이용한 단백질 분획 침전), 투석, 겔 여과, 이 온 교환, 역상 칼럼 크로마토그래피와 같은 칼럼 크로마토그래피 및 한외여과 등의 기법을 단독 또는 조합으로 적용시켜 본 발명의 면역사이토카인을 정제할 수 있다

상기 인간 인터페론 -베타 변이체는 서열번호 1 내지 서열번호 4로 이루어진 군에서 선택된 펩티드 서열로 이루어진 것을 특징으로 하는 타겟 특이적 인간 인터 페론-베타의 생산량 증가방법을 제공한다. 본 발명의 생산량 증가 방법의 각 구성요소에 대해서는 상기에서 기재한 바 와 같으며, 타겟 특이적 인간 인터페론-베타는 본 발명의 면역 사이토카인을 나타 낼 수 있다.
【유리한 효과】
본 발명의 인간 인터페론 -베타 변이체 및 항체 또는 이의 단편을 포함하는 면역사이토카인은 인터페론-베타의 활성 및 항체의 특성을 훙시에 나타내어 다발성 경화증 또는 암의 표적화'치료에 이용할 수 있으며, 본 발명에 따른 면역사이토카 인은 천연형 인터페론-베타가 접합된 면역사이토카인과 비교해 우수한 효율로 생산 될 수 있다.

【발명의 실시를 위한 형태】
이하 본 발명을 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실 시예에 한정되는 것은 아니다.
<실시예 1>
백터 클로닝 및 숙주세포 형질전환 항체 중쇄와 인터페론 -베타 변이체가 결합된 면역사이토카인 (ACC #2) 과 항 체 경쇄와 인터페론 -베타 변이체가 결합된 면역사이토카인 (ACC #7) 을 cloning 하 기 위해 B12의 sequence를 이용하였다. B12 sequence의 중쇄와 경쇄에 각각 링커 를 이용하여 인간 인터페론 -베타 변이체 서열을 삽입한 후 백터로 합성하였다. 합 성된 유전자를 각각에 맞는 제한 효소로 절단하여 보유하고 있는 IgG 발현 백터에
라이게이션 시킨 후, sequencing 작업을 통해 최종적으로 ACC#2과 #7를 발현하는 백터를 제작하였다. 클로닝이 완료 된 ACC#2, ACC#7 백터는 transformation올 통하 여 대량으로 추출한 뒤 형질전환에 사용하였다.
CHO-S cell은 3X105 cells/ml 밀도로 5 passage 이상 계대배양을 진행하며 형질전환을 준비하였다. 계대배양이 진행 된 후 세포의 생존를이 90% 이상으로 유 지가 되면 5X105 cells/ml 밀도로 세포를 시딩하여 형질전환을 준비하였다. 세포 시딩을 하고 24시간 뒤 생존률 (〉9 )과 세포 밀도 (1X106 eel ls/ml)를 확인 후 형 질전환 용매를 이용하여 50ug DNA을 30ml 배양배지에서 배양중인 CHO—S cell에 형 질전환 하였다.

숙주세포에서의 면역사이토카인 발현 확인 세포 형질전환 후 48 시간이 지나고, 농도 측정 (BCA assay) 및 웨스턴 -블롯 으로 ACC #2, #7의 발현양을 확인하였다.
BCA assay의 경우, 시약 A (Sodium carbonate, Bicinchoninic acid 등 포함) 와 시약 B (4 % cupric sulfate 포함)를 50 : 1의 비율로 제조하여 standard 용액 (BSA 용액, 0~2000 ug/mL) 및 시료와 섞었다 (10 ul의 sample과 200 ul의 reagent). 이 용액을 37 °C에 30 분간 인큐베이션 한 후 562 nm에서 흡광도를 재어 농도를 계 산하였다. 농도 계산에는 표준용액을 바탕으로 얻은 curve를 이용하였다.
웨스턴 -블롯 실험은 다음과 같이 진행하였다. 우선, 배양한 각 배지를 모아 10 % SDS PAGE로 내렸다. 내린 gel을 PVDF membrane에 이동 한 다음, 5 % BSA 용액 에 블로킹하고 1, 2차 항체를 붙였다. TBST 용액으로 세척까지 완료한 후에, 필름 으로 membrane을 찍는다. Developer와 fixer로 필름의 이미지를 현상하였다.
그 결과, B12의 중쇄와 경쇄에 천연형 인간 인터페론이 접합된 면역사이토카 인에 비해 인간 인터페론 -베타 변이체가 접합된 면역사이토카인의 발현양이 많음을 알 수 있었다 (도 1).
<실시예 3>
면역사이토카? ΐ 제조
항체의 증쇄 (Heavy chain) 부위에 서열번호 5로 표시되는 링커를 삽입하고, 인터페론 -베타 또는 인터페론 -베타 변이체를 결합하도록 하였다. 도 2에 인간 인터 페론 -베타 변이체가 접합된 면역사이토카인의 구조를 모식도로 나타냈다.
항체의 중쇄 (heavy chain) 부위에 서열번호 5로 표시되는 링커 및 인터페론- 베타 및 인터페론 -베타 변이체를 클로닝하였다. 그 후 전체 유전자의 3' -말단 및 5' -말단에 각각 제한효소 Avrl l (CCTAGG) 절단 부위와 Bstzl7I (GTATAC) 절단 부위를 삽입하여 중쇄의 최종 유전자를 확보하였다. 또한 항체의 경쇄 ( l ight chain)의 3 ' - 말단 및 5' -말단에 각각 제한효소 EcoRV(GATATC) 절단 부위와 PacI (TTMTTAA)를 삽 입하여 경쇄의 최종 유전자를 확보하였다. 도 3에 제작하는 과정을 모식도로 나타 냈다.
<실시예 4>
면역사이토카인의 발현 확인 인간 인터페론-베타가 접합된 면역사이토카인과 인터페론 -베타 변이체가 접 합된 면역사아토카인의 발현을 확인하기 위하여 50 ug의 pRBLX2-INF 혹은 pRBLX2- CAF백터를 CH0-S 세포에 transfect ion 한 뒤 7일간 배양하면서 발현을 유도하였다. 7일 후 배양액을 회수하고, 원심분리 (8000rpn, 10분)를 하여 세포를 제거하였다. 세포가 제거된 배양액을 소량 채취하여 5x 샘플 버퍼와 섞고, 100°C에서 10분간 끓 여주어 단백질이 층분히 변형되도톡 유도하였다. 준비된 샘플을 마커 (Marker)와 함 께 Tricine SDS-PAGE 겔에 로딩하고 130v의 전압으로 1시간 30분간 전기영동을 실 시하였다. 이 후, 겔을 조심스럽게 분리한 뒤 쿠마시 블루 염색 솔루션 (comassie blue staning solut ion)에 담구고 30분간 흔들어주며 염색 해주었다. 염색이 끝난 뒤 겔을 탈염색 (Destaning) 버퍼로 옮겨서 30분간 흔들어주며 탈염색을 해주었다. 탈염색은 3번 반복하여 수행하였다.
보다 명확한 발현 양 비교를 위하여 항-인터페론 β 항체와 항 -인간 IgG-HRP 를 사용하여 웨스턴블로팅을 수행하였다. 상기와 동일한 방법으로 Tricine SDS- PAGE 수행한 후 겔을 조심스럽게 분라하여 3M 페이퍼 위에 두고 그 위에 PVDF( polyvinyl ! dene di f luoride) 막을 둔 뒤 다시 3M 페이퍼를 덮어주고 lx transfer 버퍼에 담가 100v 전압으로 70분간 단백질을 transfer시켰다. Tris- buffered sal ine-Tween 20(TBS-T, 0.1% Tween20)에 M를 넣어 막을 1시간 30분동안 상온에서 차단하였다. PVDF막을 TBS-T로 2번 씻어낸 뒤, TBS-T에 담가두었다. 항-
인터페론 β 항체의 경우 TBS-T에 1 : 1000으로 회석하여 준비하였고, 항 -인간 IgG- HRP 항체의 경우 TBS-T에 1 : 3000으로 희석하여 준비하였다. 막을 항체 희석물에 담 가 상온에서 2시간동안 흔들어주며 반응시켰다. 이 과정이 끝나면 10분 동안 3번씩 TBS-T로 닦아주고 상온에서 호스래디쉬 퍼옥시다아제 (horseradi sh peroxidase ; HRP)가 결합된 이차 항체를 첨가하여 1시간 동안 반웅하였다. 다시 한번 세척과정 을 수행한 후, 밴드를 ECL 시약 (enhanced chemi luminescence reagent , Intron)으로 확인하였다. 밴드의 강도는 C-DiG (LI-C0R, USA)을 이용하여 측정하였다.
• 그 결과, 도 4에 나타난 바와 같이 25 KDa 위치에서 경쇄가 관찰되었으며
70-100 KDa 사이에 인간 인터페론-베타가 접합된 면역사이토카인 흑은 인간 인터페 론 -베타 변이체가 접합된 면역사이토카인 복합체가 관찰되었다.
또한 도 5에서 레인 1은 인간 인터페론-베타가 접합된 면역사이토카인, 레인 2 는 인간 인터페론 -베타 변이체가 접합된 면역사이토카인이다. Tr i c ine-SDS PAGE 및 웨스턴블로팅 결과, 인간 인터페론-베타가 접합된 면역사이토카인 보다 인간 인터 페론 -베타 변이체가 접합된 면역사이토카인의 발현량이 높은 것을 확인 할 수 있었 다. 또한 정확한 발현량의 비교를 위하여 배양액을 각각 Cedex bio(Roche사, 미국) 에서 측정을 진행하였다. 그 결과 인간 인터페론-베타가 접합된 면역사이토카인은 농도가 측정 범위 이하 ( 10mg/L 이하)로 낮은 발현양이 확인 되었고, 인간 인터페론 -베타 변이체가 접합된 면역사이토카인은 약 32mg/L수준으로 최소 3배 이상으로 발 현량이 증가한 것을 확인하였다.
<실시예 5>
DSTAT-Ι의 인산화를 통한 면역사이토카인의 인터페론 활성 확인 본 발명에 따른 인간 인터페론 -베타 변이체와 B12항체가 접합된 면역사이토 카인의 인터페론 기능을 확인하기 위하여 인터페론 혹은 항체-인터페론 접합체 처 리에 따른 STAT-1의 인산화여부를 검증하였다.
6-wel l plate의 각 wel l에 3xl05개의 0VCAR-3 세포를 분주한 뒤 24시간 동 안 37. 5° C, 5% C02 환경에서 배양하였다. 24시간 이후 세포 배양액을 제거하고 인간 인터페론 -베타 변이체 (carbi feron 600ng/ml , 인간 인터페론 -베타 변이체와 B12 항체가 접합된 면역사이토카인 (ACC) 600ng/ml 또는 1800ng/ml의 농도가 되도록 배양액에 회석하여 1시간 동안 처리하였다. 이 후 plate를 수거하여 각 wel l을 PBS 로 3회 세척한 뒤 프로티아제 저해제와 포스파테이즈 저해제가 포함된 RIPA 버퍼
ΙΟΟμ Ι를 처리하여 30분 동안 Ice 상에 두어 세포를 용해시켰다. 용해된 세포를 1.5ml 튜브에 담아 13000rpm, 4° C 조건으로 원심분리하여 상층액 (용해물)만 수거 한 뒤 새로운 튜브에 모았다. BCA정량법을 이용하여 용해물의 단백질 농도를 정량 한 뒤 30yg의 용해물을 채취하여 5x sample buffer와 섞고, 10( C에서 10분간 끓 여주어 단백질이 층분히 변성 되도록 유도하였다. 준비된 샘플을 마커 (Marker)와 함께 10% SDS-PAGE 겔에 로딩하고 70 vol tag로 30분, 120 voltage로 1시간 동안 내 려주었다. 이후 겔을 조심스럽게 분리하여 3M 페이퍼 위에 두고 그 위에 PVDFCpolyvinylidene di fluoride) 막을 둔 뒤 다시 3M 페이퍼를 덮어주고 트랜스퍼 버퍼에 담가 100 볼트로 90분간 단백질 트랜스퍼를 진행하였다. ¾의 BSA가 포함된 Tris-buffered saline-Tween 20(TBS-T, 0.1% Tween20)에 막을 1시간 30분동안 블락 킹 한 뒤 Anti-p-STATl 항체의 경우 TBS-T에 1:1000으로 회석하여 준비하였고, anti-GAPDH항체의 경우 TBS-T에 1:3000으로 회석하여 준비하였다. 막을 항체 회석 물에 담가 상온에서 2시간동안 흔들어주며 반웅시켰다. 이 과정이 끝나면 10분 동 안 3번씩 TBS-T로 닦아주고 상온에서 호스래디쉬 퍼옥시다아제 (horseradish peroxidase; HRP)가 결합된 이차 항체를 첨가하여 1시간 동안 반웅하였다. 다시 한 번 세척과정을 수행한 푸, 밴드를 ECL 시약 (enhanced chemi luminescence reagent , Intron을 처리한 뒤 필름에 현상하였다.
그 결과, 인간 인터페론 -베타 변이체 (carbiferon) 및 면역사이토카인 처리군 에서 모두 pSTAT-1의 인산화가 나타나 인간 인터페론 -베타 변이체 (carbiferon)와 B12항체가 접합된 면역사이토카인에서 인터페론-베타의 활성이 그대로 나타남을 확 인하였다 (도 7).
<실시예 6>
세포독성실험 통한 면역사이토카인의 인터페론 활성 확인 본 발명에 따른 인간 인터페론 -베타 변이체와 B12항체가 접합된 면역사이토 카인의 인터페론 기능을 확인하기 위하여 인터페론 혹은 항체-인터페론 접합체 처 리에 따른 세포독성 여부를 확인하였다. 세포독성을 확인하기 위하여 96-well plate의 각 well에 1x10개의 0VCAR-3 세포를 분주한 뒤 24 시간 동안 37. 5° C, 5% C02 환경에서 배양하였다. 24시간 이 후 세포 배양액을 제거하고 인간 인터페론 -베타 변이체 (carbiferon), B12 항체 및 면역사이토카인을 10~L0000ng/ml 농도로 처리한 뒤 24시간 혹은 48시간 동안 배양
하였다. 24시간 혹은 48시간 배양한 뒤 배양액을 제거하고 PBS 워시를 2회 진행한 뒤 WST시약을 1 : 10으로 배양액에 섞어 각 wel l당 ΙΟΟ μ Ι씩 처리한 뒤 2시간 동안 37. 5° C, 5% C02 환경에서 방치한 뒤 430nm파장에서 흡광도를 측정하였다.
그 결과 B12 항체만을 처리한 세포군에서는 세포독성이 나타나지 않았으나 인간 인터페론—베타 변이체 및 면역사이토카인을 처리한 세포군에서는 세포독성이 농도 의존적으로 나타나는 것으로 보아, 인간 인터페론 -베타 변이체가 면역사이토 카인의 형태로도 여전히 인터페론 활성을 나타냄을 확인하였다 (도 8 및 9) .
<실시예 7>
항체 중쇄와 인터페론 -베타 변이체가 결합된 면역사이토카 의 제조
B12 항체이외의 ERBB2(Hercept in) 항체와 C-MET 항체를 이용하여 인터페론- 베타 변이체를 결합한 면역사이토카인을 다음과 같이 제조하였다.
도 10에서 보듯이, ERBB2(Hercept in) 항체와 c-MET 항체의 중쇄 (heavy chain) 부위에 Rigid Hel ical 링커를 결합하였다. 그 뒤에 인간 인터페론 -베타 변 이체를 결합하여 항 -c-Met 면역사이토카인 (A)과 항 -ERBB2 면역사이토카인 (B)을 발 현하는 발현 카세트를 제조하였다. 이를 pRBLX2에 각각 클로닝한 다음 각 백터를 CH0-S 세포에 trans feet ion 한 뒤 7일간 배양하면서 발현을 유도하였다. transfect ion, 배양 및 발현물의 회수는 실시예 4에 기재된 바에 따라 수행하였다. 각 발현량을 CH0-S 세포를 이용하여 c— Met 항체 또는 ERBB2 항체에 인간 인 터페론-베타가 접합된 면역사이토카인과 인간 인터페론 -베타 변이체가 접합된 면역 사이토카인의 발현량을 비교하였을 때, 인간 인터페론-베타가 접합된 면역사이토카 인에 비하여 인간 인터페론 -베타 변이체가 접합된 면역사이토카인의 발현량이 더 많으며, 인터페론의 활성이 더 우수함을 확인할 수 있었다. 이를 통해, 본 발명의 인간 인터페론 -베타 변이체가 발현시 야생형의 인터페 론 베타에 비해서도 발현이 매우 잘 이루어지는 것을 확인할 수 있었다.
【산업상 이용가능성】
본 발명에 따른 인간 인터페론 -베타 변이체가 접합된 면역사이토카인의 경 우, .천연형 인터페론-베타보다 우수한 인터페론 활성 및 특정 항원을 인식하는 항 체의 특성이 모두 나타난다는 점에서 다발성 경화증, 암 등의 질환에 대한 표적치 료제로 사용될 수 있으며ᅳ 천연형 인터페론-베타가 접합된 면역사이토카인과 비교 해 그 생산효율이 현저히 높다는 점에서 산업상 이용가능성이 우수하다.
Claims
【청구항 1]
(a) 인간 인터페론 -베타 변이체; 및 (b) 상기 인간 인터페론 -베타 변이체에 직접 또는 간접적으로 공유 결합된 항체 또는 이의 단편을 포함하는 면역사이토카 인으로서,
상기 인간 인터페론 -베타 변이체는 하기 (i), (ii) 및 (iii)로 이루어진 군 에서 선택되고 인간 인터페론 -베타 활성을 지니는 폴리펩티드로서, N-연결형 당쇄 를 포함하는 폴리펩티드인 면역사이토카인;
(i) 서열번호 1에 개시된 아미노산 서열 전체를 포함하는 폴리펩티드;
(ii) 서열번호 1에 개시된 아미노산 서열의 실질적인 부분을 포함하는 폴리 펩티드;
(iii) 상기 (a) 또는 (b)의 폴리펩티드와실질적으로 유사한 폴리펩티드.

계 1항에 있어서, 상가 항체 또는 이의 단편은 종양성 항원에 대한 항체 또는 이의 단편인 것을 특징으로 하는 면역사이토카인.

제 1항에 있어서, 상기 인간 인터페론 -베타 변이체는 펩타이드 링커에 의해 상기 항체 또는 미의 단편에 결합되어 있는 면역사이토카인.

제 3항에 있어서, 상기 링커는 서열번호 5내지 서열번호 11로 이루어진 군에 서 선택된 아미노산서열을 포함하는 것을 특징으로 하는 면역사이토카인.

제 1항 내지 계 4항 중 어느 한 항에 있어서, 상기 인간 인터페론 -베타 변이체 폴리펩티드의 아미노산 서열은 상기 항체 또는 이의 단편의 아미노산 서열에 대해 중쇄 C-말단, 경쇄 C-말단 또는 중쇄 및 경쇄 C-말단에 있는 것을 특징으로 하는 면역사이토카인.
【청구항 6】
제 1항에 있어서, 상기 면역사이토카인은 서열번호 12, 13 , 15 또는 17로 이 루어진 군 중 하나의 아미노산 서열을 포함하는 것을 특징으로 하는 면역사이토카 인.

(a) 서열번호 2 내지 서열번호 4 중 어느 하나로 표시되는 인간 인터페론-베 타 변이체;
