모발 성장 및 발모와 관련하여 제조예 1-1에서 제조된 추출물의 윈트/베타카테닌 신호(Wnt/ Beta-catenin signaling)전달 유도 활성을 탐색하였다. 이를 위하여 배양 및 형질전환(transfection)이 용이한 HEK-293T(인간 배아신장세포)에 Wnt 리셉터 벡터(receptor vector)인 TOPFlash 또는 컨트롤 벡터(control vector)인 FOPFlash를 pCMV renilla 벡터와 함께 형질전환 하여 활성평가 시험에 사용하였다. 구체적으로는, HEK-293T 세포를 DMEM 배지에 10% 소태아혈청(Fetal bovine serum, FBS), 100 unit/ml의 페니실린 및 100 μg/ml의 스트렙토마이신이 첨가된 배양액에서 5% CO2, 37℃ 조건으로 인큐베이터(incubator)에서 배양하였으며, TOPFlash 플라스미드, FOPFlash 플라스미드, pCMV/renilla 플라스미드 등으로 형질전환 하였다. 12 시간 뒤 형질전환된 HEK-293T 세포를 조직배양용 96-웰 플레이트(96-well plate)에 도말(plating) 하였으며, 24시간 뒤 추출물을 200 ug/ml 농도로 처리 후, 48시간 동안 배양기 내에서 배양하였다. 세포들은 듀얼 루시퍼라아제 분석 시스템(Dual-luciferase Assay System, Promega, USA)의 표준방법대로 용해(lysis)시켜 활성도를 측정하였다. 5α-환원효소(5α-reductase)는 테스토스테론(testosterone)을 더 활성이 높은 DHT(Dihydrotestosterone)로 전환하는 효소인데, DHT의 과다로 인한 윈트/베타카테닌 신호전달 감소는 탈모기전과 밀접한 연관이 있다. 본 제조예 1-1에서 제조한 대황, 감송향의 증류수 추출물들의 5α-환원효소에 대한 억제효과를 확인하기 위하여, 추출물 처리에 의하여 생쥐의 간 마이크로좀(microsome)에서 얻어지는 5α-환원효소가 테스토스테론을 감소시키는 과정이 억제되는 정도를 측정하였다. 간 마이크로좀의 5α-환원효소를 얻기위하여 생후 10주령의 BL/6 수컷 생쥐의 간을 마취 후 절제하고, 라이시스 버퍼(lysis buffer)에 초음파처리(sonication)를 통해 간 내 5α-환원효소를 추출한 뒤, 원심분리하여 얻은 상층액을 실험에 사용하였으며, 나머지는 80℃에 보관하였다. 실험을 위하여 5α-환원효소 반응에 필요한 포스파타아제 버퍼(phosphatase buffer), 테스토스테론, NaDPH를 모두 섞어주고, 5α-환원효소가 추출된 용해 버퍼(lysis buffer) 및 추출물을 최종 5 mg/ml의 농도로 첨가한 다음, 37℃에서 180분간 반응시켰으며, 얻어진 반응물에 디클로로메탄(Dichloromethane)을 첨가하여 반응을 종료시켰다. 내부대조군으로는 프로필-p-히드록시벤조에이트(propyl-p-hydroxybenzoate)를 첨가하여 테스토스테론의 양을 보정하였으며, 테스토스테론의 감소 및 내부대조군의 함량을 평가하기 위하여 고성능 액체크로마토그래피(High Performance Liquid Chromatography, HPLC)를 이용하였다. 분석을 위한 조건으로 고정상 컬럼(column)은 C18 역상컬럼을 이용하였으며, 이동상 용매(Mobile phase)로는 메탄올 및 초순수 증류수(Deionized water)의 비율을 65:35로 되도록 설정하였고, 시료주입량을 20 μl로 하여 UV 검출기(UV detector)를 이용 245 nm에서 평가하였다. 버퍼만 넣은 효소 블랭크(enzyme blank) 대조군으로부터 얻어진 값을 100%로 하였을 때 생쥐 간 추출물을 넣은(Liv) 반응 결과물의 상대적인 테스토스테론/내부대조군의 비를 구하고, 생약 추출물을 넣은 결과와 비교하여 저해 활성(inhibition activity)을 계산하였으며 구체적인 방법은 아래 계산식 1과 같다.5α-reductase is an enzyme that converts testosterone to more active Dihydrotestosterone (DHT), which is closely related to hair loss mechanism due to a decrease in wint / betacatenin signaling due to excess DHT.
