本特許出願は、日本国特許出願第2015-024439号について優先権を主張するものであり、ここに参照することによって、その全体が本明細書中へ組み込まれるものとする。
本発明は、有効成分としての分岐鎖アミノ酸(BCAA)を含む眼疾患の処置および/または予防剤を提供する。本発明は、また、BCAAを有効成分として含む網膜の細胞の変性および/または細胞死の抑制剤を提供する。本発明は、さらに、BCAAを有効成分として含む視機能低下の抑制剤を提供する。本発明は、さらに、BCAAを有効成分として含む、眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品を提供する。
緑内障は、網膜神経節細胞の変性または脱落による視野欠損を特徴とし、積極的な治療が行われなければ失明すら引き起こし得る疾患である。日本国における有病率は40歳以上の5%であり、2005年には中途失明の最多原因であった(24.6%)。緑内障の病因として高眼圧が挙げられるが、高眼圧を伴わない緑内障、すなわち正常眼圧緑内障の患者数は、例えば日本国において、全緑内障患者の6割に上る。現在の緑内障の処置方法は、薬物投与であれ外科手術であれ、眼圧を低下させることによる。正常眼圧緑内障患者の眼圧を低下させることによって緑内障の進行が停止または遅延することがあるが、眼圧を降下させても緑内障の進行が十分遅延しないか、あるいは全く遅延しない患者も存在する。またそもそも正常眼圧緑内障患者の眼圧は正常であるから、眼圧降下ができないか、あるいは十分ではないこともある。また、高眼圧を伴う緑内障であっても、眼圧降下療法によって緑内障の進行が十分に遅延しない場合や眼圧降下が困難な場合もある。
網膜色素変性は、視細胞の変性または脱落による視野欠損および夜盲を特徴とする進行性の疾患である。約半分の症例において網膜色素変性は遺伝性であるが、残り半分は孤発性である。現在、遺伝子治療、神経保護治療および再生医療による治療が試みられているが、網膜色素変性の治療方法は十分に確立されておらず、予防方法は存在しない。従って、患者の視野欠損および視力低下の進行を防止する方法はなく、多くの場合失明に至る。患者数は日本国で推定5万人であり、2005年に中途失明の第3番目の原因であった。
加齢黄斑変性は、中心が見えにくい、ゆがむなどの症状を伴う眼疾患であり、先進国では高齢者の失明原因の上位を占める。米国では、2000年に80歳以上の高齢者の約15%が加齢黄斑変性に罹患している。日本国においても、高齢化、食文化の欧米化に伴い、加齢黄斑変性患者は近年増加しており、中途失明の第4番目の原因となっている(2005年)。
加齢黄斑変性では、主として脂質で構成される老廃物であるドルーゼンが網膜色素上皮下に蓄積される。その後、滲出型加齢黄斑変性では、脈絡膜下新生血管が形成され、網膜下出血・網膜浮腫などによる視力低下が起こる。現状では、有効な治療方法として、形成された新生血管に対する血管退縮治療(抗VEGF剤の硝子体内投与、光線力学療法)や、新生血管からの漏出などを抑制するステロイド治療が用いられているが、一度新生血管が形成されると、このような治療を行っても視力予後は不良である。良好な視力を保つためには、ドルーゼン形成期に進行抑制治療を行うことが重要であるが、現在のところ、そのような治療方法はない。また、萎縮型加齢黄斑変性では、ドルーゼン蓄積の後、新生血管形成を伴わずに、徐々に網脈絡膜が萎縮し、視力低下が起こる。萎縮型加齢黄斑変性に対しては、現在のところ有効な治療方法はない。滲出型加齢黄斑変性から萎縮性加齢黄斑変性に移行することもある。
分岐鎖アミノ酸は必須アミノ酸として人体に不可欠な栄養素であるため、合成化合物と比較して極めて安全性が高い。これまでに、分岐鎖アミノ酸を、肝癌発生・進展抑制剤、非代償性肝硬変患者の低アルブミン血症の改善薬などとして用いることは知られていた。しかしながら、緑内障、網膜色素変性および加齢黄斑変性を含む眼疾患の処置または予防に使用することは報告されていない。

本発明者らは、上記課題に鑑み鋭意研究した結果、分岐鎖アミノ酸が、緑内障、網膜色素変性および加齢黄斑変性のモデルにおいて治療および予防効果を示すことを見出した。また、分岐鎖アミノ酸が、視細胞、網膜色素上皮細胞および網膜神経節細胞を含む網膜の細胞を保護し、その変性および/または細胞死を抑制し、視機能の低下を抑制することを見出した。本発明者らは、これらの知見に基づいて、本発明を完成するに至った。
本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、眼疾患の処置および/または予防剤を提供する。
本発明は、また、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、網膜の細胞の変性および/または細胞死の抑制剤を提供する。
本発明は、また、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、視機能低下の抑制剤を提供する。
本発明は、また、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品を提供する。
本発明の薬剤は、網膜の細胞の変性および/または細胞死を抑制し、視機能の低下を抑制する。そのため、本発明は、網膜の細胞の変性および/または細胞死に起因する疾患に、新たな処置および/または予防方法を提供することができる。
本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有することを特徴とする眼疾患の処置および/または予防剤(本明細書中、「本発明の薬剤」と称することもある)を提供する。
本発明の薬剤の有効成分であるイソロイシン、ロイシンおよび/またはバリンは、それぞれL-体、D-体、DL-体のいずれも使用することができるが、好ましくは、L-体、DL-体であり、さらに好ましくは、L-体である。
また、本発明におけるイソロイシン、ロイシンおよび/またはバリンは、それぞれ遊離体だけでなく塩の形態でも使用することができる。本発明におけるイソロイシン、ロイシンおよび/またはバリンは、かかる塩の形態をも包含する。塩の形態としては、酸との塩(酸付加塩)、塩基との塩(塩基付加塩)等を挙げることができるが、医薬として許容される塩を選択することが好ましい。
イソロイシン、ロイシンおよび/またはバリンに付加し、かつ医薬として許容される酸付加塩を形成する酸としては、例えば、塩化水素、臭化水素、硫酸、リン酸等の無機酸;酢酸、乳酸、クエン酸、酒石酸、マレイン酸、フマル酸、モノメチル硫酸等の有機酸が挙げられる。
イソロイシン、ロイシンおよび/またはバリンに付加し、かつ医薬として許容される塩基付加塩を形成する塩基としては、例えば、ナトリウム、カリウム、カルシウム等の金属の水酸化物または炭酸化物、あるいはアンモニア等の無機塩基;エチレンジアミン、プロピレンジアミン、プロピレンジアミン、エタノールアミン、モノアルキルエタノールアミン、ジアルキルエタノールアミン、ジエタノールアミン、トリエタノールアミン等の有機塩基が挙げられる。
本発明の薬剤は、イソロイシン、ロイシンおよびバリンの3種の分岐鎖アミノ酸からなる群から選択される少なくとも1種(即ち、1種(イソロイシン、ロイシンまたはバリン)、2種(イソロイシンおよびロイシン、イソロイシンおよびバリン、またはロイシンおよびバリン)または3種(イソロイシン、ロイシンおよびバリン))を有効成分として含有する。本発明の薬剤は、少なくともイソロイシンを有効成分として含有することが好ましい(即ち、イソロイシン、ロイシンおよびバリンの3種、イソロイシンおよびロイシンの2種、イソロイシンおよびバリンの2種、またはイソロイシンのみを有効成分として含有する)。本発明の薬剤は、イソロイシン、ロイシンおよびバリンの3種を有効成分として含有するか、或いはイソロイシンのみを有効成分として含有する(即ちイソロイシンを有効成分として含有し、且つロイシンおよびバリンを有効成分として含有しない)ことがより好ましい。本発明の薬剤は、イソロイシン、ロイシンおよびバリンの3種を有効成分として含有するのが最も好ましい。本発明の薬剤は、イソロイシン、ロイシンおよびバリンの3種の分岐鎖アミノ酸からなる群から選択される少なくとも1種を有効成分として含有する医薬組成物であり得る。
本発明の薬剤がイソロイシン、ロイシンおよびバリンの3種の分岐鎖アミノ酸を有効成分として含有する場合、かかる3種のアミノ酸の配合比は、それぞれ重量比で、通常、1:1.5~2.5:0.8~1.7の範囲であり、特に好ましくは1:1.9~2.2:1.1~1.3の範囲であり、最も好ましくは1:2:1.2である。
本発明の薬剤がイソロイシンおよびロイシン、イソロイシンおよびバリン、またはロイシンおよびバリンの2種の分岐鎖アミノ酸を有効成分として含有する場合、かかる2種のアミノ酸の配合比は、上記3種のアミノ酸の配合比における当該2種のアミノ酸の配合比とすればよい。
また本発明の薬剤がイソロイシンのみ、ロイシンのみ、またはバリンのみを有効成分として含有する場合、その配合量を後述する投与量に基づいて決定して製剤化することができる。
本明細書で使用されるとき、「予防する」または「予防」は、対象において、特に、眼疾患に罹る可能性が高いが、未だ罹患していない対象において、眼疾患への罹患を防止すること、または、眼疾患に罹る可能性を低減することを意味する。眼疾患に罹る可能性があるが、未だ罹患していない対象には、例えば、自覚症状はないが網膜に異常が認められる対象、および、網膜に損傷を受けた可能性がある対象が含まれる。
本明細書で使用されるとき、「処置する」または「処置」は、眼疾患に罹患している対象において、眼疾患の原因を軽減または除去すること、眼疾患の進行を遅延または停止させること、および/または、眼疾患の症状を軽減、緩和、改善または除去することを意味する。
本発明の薬剤により処置および/または予防される眼疾患には、網膜の細胞の変性および/または細胞死に起因または関連するすべての眼疾患が含まれ、例えば、緑内障、網膜色素変性、加齢黄斑変性が含まれる。緑内障には、正常眼圧緑内障および眼圧降下療法に不応性の緑内障が含まれる。加齢黄斑変性には、滲出型加齢黄斑変性および萎縮型加齢黄斑変性が含まれる。
正常人の眼圧は10~21mmHgであるが、眼圧が何らかの原因によりこの範囲以上、典型的には25mmHg以上に上昇することによって網膜神経節細胞が変性または脱落して、緑内障を発症すると考えられてきた。しかし近年、眼圧が正常値(10~21mmHg)であるにも拘わらず緑内障を発症する症例が非常に多くみられ、そのような患者には眼圧降下療法が適用できないか、有効ではないか、あるいは不十分である場合がある。また高眼圧を伴う緑内障患者であっても、眼圧降下療法が有効ではないか、あるいは不十分である場合もある。本発明の薬剤は、眼圧に非依存的に網膜神経節細胞の変性または脱落を抑制することができるので、正常眼圧緑内障および眼圧降下療法に不応性の緑内障の処置にも有用である。緑内障の予防における対象には、視野検査で異常が認められる対象、および、視神経乳頭陥凹拡大、神経線維層および/または神経節細胞層の菲薄化が認められる対象が含まれる。
本発明の薬剤は眼圧に非依存的に緑内障を処置し得るが、このことは本発明の薬剤と眼圧降下療法の併用を除外することを意図するわけではない。本発明の薬剤と併用することができる眼圧降下療法には、緑内障または高眼圧症の処置用薬物または眼圧降下剤の投与、レーザー手術または外科手術等が含まれるが、これらに限定されない。
網膜色素変性は、神経細胞の一種である視細胞、特に杆体細胞の細胞死によって引き起こされる疾患である。視細胞で働く遺伝子の異常により、視細胞が変性脱落することにより引き起こされる。網膜色素変性は遺伝性であり、常染色体優性遺伝、常染色体劣性遺伝、X染色体劣性遺伝により遺伝する。約50種の原因となる遺伝子の変異が知られている。本発明の薬剤は、原因遺伝子の種類に拘わらず、また、その原因遺伝子が明らかであるかどうかに拘わらず、網膜色素変性を処置および/または予防し得る。網膜色素変性の予防における対象には、網膜色素変性の原因となる遺伝子変異を有する対象、および、原因遺伝子が明らかであるかどうかに拘わらず、遺伝学的に網膜色素変性の発症が予想される対象が含まれる。
加齢黄斑変性は滲出型と萎縮型に分けられる。滲出型は黄斑部の網膜色素上皮細胞-ブルッフ膜-脈絡膜の変化により発生する脈絡膜新生血管(choroidal neovascularization:CNV)とその増殖変化を本態とする疾患である。CNVは網膜色素上皮下、ついで網膜下に発育する。出血、滲出による網膜色素上皮剥離、網膜剥離を呈し、それらが吸収された後には萎縮・瘢痕が形成され、高度の永続する視力低下を生じ、その進行は速い。萎縮型では黄斑部に網膜色素上皮-脈絡毛細血管板の地図状萎縮病巣が形成されるが、進行は緩慢である。
網膜色素上皮細胞は視細胞の外節の貪食機能を始め神経網膜の環境を保持する上で重要な役割を果たしているが、加齢性の変化として、消化残渣物としてのリポフスチンの蓄積が起こり、ドルーゼンと呼ばれる異常な構造が形成される。また、加齢とともに色素上皮下のブルッフ膜の肥厚が起こり、視細胞-網膜色素上皮-ブルッフ膜間の生理的環境に変化が生じる。こうした変化によって起こった慢性炎症や虚血が脈絡膜からの新生血管の原因になると考えられており、新生血管の発生・発育に最も大きな作用を及ぼす生理活性物質として、血管内皮増殖因子(VEGF)が挙げられる。CNVはブルッフ膜の破損部から網膜色素上皮下、網膜下、場合により網膜内へと進展してくる。
現在のところ、萎縮型加齢黄斑変性に対しては、有効な治療方法はない。滲出型加齢黄斑変性に対しては、有効な治療方法は、形成された新生血管に対する血管退縮治療(抗VEGF治療、光線力学療法)のみである。本発明の薬剤は、ドルーゼンを除去するか、あるいはドルーゼンの形成または増加を抑制することにより、血管退縮治療とは異なる作用機序で、萎縮型および滲出型の加齢性黄斑変性を処置および/または予防できる。加齢黄斑変性の予防における対象には、眼底検査でドルーゼン形成などの異常が認められる対象、片眼のみにCNVまたはドルーゼン形成などの異常が認められる対象、遺伝子多型などの検査により加齢黄斑変性発症のリスクが高いと認められる対象、および、加齢黄斑変性の家族歴を有する対象が含まれる。
本発明の薬剤は血管退縮治療とは異なる作用機序で、萎縮型および滲出型の加齢黄斑変性を処置および/または予防し得るが、このことは本発明の薬剤と血管退縮治療、ステロイド剤、外科手術などとの併用を除外することを意図するわけではない。本発明の薬剤と併用することができる血管退縮治療には、抗VEGF剤の硝子体内投与、並びに、光線力学療法が挙げられるが、これらに限定されない。本発明の薬剤と併用することができる外科手術には、CNVの除去、硝子体内出血の除去、網膜下出血の移動を目的とするものが含まれる。
本発明の薬剤の適用対象としては、動物、典型的には哺乳動物(例えば、ヒト、マウス、ラット、ハムスター、ウサギ、ネコ、イヌ、ウシ、ヒツジ、サル等)が挙げられる。なお、ヒト以外の動物に適用する場合、本発明の薬剤の投与量は、動物の体重もしくは大きさに応じて適宜加減すればよい。
本発明の薬剤の投与方法は特に限定されないが、経口投与、直腸投与、注射、輸液、点眼、硝子体内注射による投与等の一般的な投与経路を経ることができる。
経口投与の剤形としては、顆粒剤、細粒剤、粉剤、被覆錠剤、錠剤、坐剤、散剤、(マイクロ)カプセル剤、チュアブル剤、シロップ、ジュース、液剤、懸濁剤、乳濁液などが挙げられる。また注射による投与の剤形としては、静脈直接注入用、点滴投与用、硝子体注射用、活性物質の放出を延長する製剤等などの医薬製剤一般の剤形を採用することができる。
これらの剤形は、常法により製剤化することによって製造される。さらに製剤上の必要に応じて、医薬的に許容し得る各種の製剤用物質を配合することができる。製剤用物質は製剤の剤形により適宜選択することができるが、例えば、賦形剤、希釈剤、添加剤、崩壊剤、結合剤、被覆剤、潤滑剤、滑走剤、滑沢剤、風味剤、甘味剤、可溶化剤等が挙げられる。更に、製剤用物質を具体的に例示すると、炭酸マグネシウム、二酸化チタン、ラクトース、マンニトールおよびその他の糖類、タルク、牛乳蛋白、ゼラチン、澱粉、セルロースおよびその誘導体、動物および植物油、ポリエチレングリコール、および溶剤、例えば滅菌水および一価または多価アルコール、例えばグリセロール等を挙げることができる。
本発明の薬剤はまた、点眼剤の形態で投与することができる。点眼剤は、塩化ナトリウム、濃グリセリンなどの等張化剤;リン酸ナトリウム、酢酸ナトリウムなどの緩衝化剤;ポリオキシエチレンソルビタンモノオレート、ステアリン酸ポリオキシル40、ポリオキシエチレン硬化ヒマシ油等の界面活性剤;クエン酸ナトリウム、エデト酸ナトリウム等の安定化剤;塩化ベンザルコニウム、パラベン等の防腐剤等を必要に応じて用い、製造することができる。pHは眼科製剤に許容される範囲内にあればよいが、通常4~8の範囲内である。また、眼軟膏は、白色ワセリン、流動パラフィン等の汎用される基剤を用い、製造することができる。
硝子体内投与用の注射剤は、塩化ナトリウム等の等張化剤;リン酸ナトリウム等の緩衝化剤;ポリオキシエチレンソルビタンモノオレート等の界面活性剤;メチルセルロース等の増粘剤等を必要に応じて用い、製造することができる。
本発明の薬剤の投与量は、対象患者の年齢、体重もしくは病態、または医薬の剤形もしくは投与方法などによっても異なるが、成人1日あたり、イソロイシン0.005g/kg体重~5g/kg体重、ロイシン0.01g/kg体重~10g/kg体重、バリン0.005g/kg体重~5g/kg体重を目安とする。
一般の成人の場合、好ましくは、成人1日あたり、イソロイシン0.01g/kg体重~1g/kg体重、ロイシン0.02g/kg体重~2g/kg体重、バリン0.01g/kg体重~1g/kg体重であり、より好ましくは、イソロイシン0.02g/kg体重~0.2g/kg体重、ロイシン0.04g/kg体重~0.4g/kg体重、バリン0.02g/kg体重~0.2g/kg体重である。アミノ酸の全体量としては1日あたり0.01g/kg体重~2g/kg体重程度が好ましい。上記1日あたりの量は一度に、もしくは数回に分けて投与することができる。投与の時期としては特に限定されず、例えば食前、食後、食間のいずれでもよい。また、投与期間も特に限定されない。
なお、本発明の薬剤の有効成分である分岐鎖アミノ酸の投与量について算出する際、その値は、本発明が目的とする作用とは別目的で(例えば通常の食生活の必要から、または別の疾患の治療を目的として)摂取もしくは投与される分岐鎖アミノ酸については、前記算出に含める必要はない。例えば、通常の食生活から摂取される1日あたりの分岐鎖アミノ酸の量を、前記本発明における有効成分の1日あたりの投与量から控除する必要はない。
本発明の薬剤の有効成分であるイソロイシン、ロイシンおよび/またはバリンは、それぞれが単独もしくは任意の組み合わせで製剤に含有されていてもよく、または全てが1種の製剤中に含有されていてもよい。別途製剤化して投与する場合、それらの投与経路、投与剤形は同一であっても、異なっていてもよく、また各々を投与するタイミングも、同時であっても別々であってもよい。従って、投与剤形、投与時期、投与経路などは、適宜決定することができる。即ち、本発明の薬剤は、複数の分岐鎖アミノ酸を含有する製剤であってもよく、またそれぞれを別途製剤化して併用するような併用剤であってもよい。本発明の薬剤は、これら全ての形態の製剤を包含するものである。特に、同一製剤中に全ての分岐鎖アミノ酸を含有する形態が、簡便に投与できるため好ましい。
本発明において、「重量比」とは、製剤中のそれぞれの成分の重量の比を示す。例えば、イソロイシン、ロイシンおよびバリンの各有効成分を1つの製剤中に含めた場合には、個々の重量の比であり、各有効成分のそれぞれを単独でまたは任意の組み合わせで複数製剤中に含めた場合には、各製剤に含められる各有効成分の合計重量の比である。
また本発明において、実際の投与量の比は、投与対象(即ち患者)あたりの各有効成分1回投与量あるいは1日投与量の比である。例えば、イソロイシン、ロイシンおよびバリンの各有効成分を1つの製剤中に含め、それを投与対象に投与する場合には、重量比が投与量比に相当する。各有効成分を単独または任意の組み合わせで複数の製剤中に含めて投与する場合には、1回あるいは1日投与する各製剤中の各有効成分の合計重量の比が投与量比に相当する。
イソロイシン、ロイシン、およびバリンは既に、医薬・食品分野において広く用いられていて、安全性は確立している。例えば、これらのアミノ酸を1:2:1.2の重量比で含有する製剤の急性毒性(LD50)は、マウスに経口投与する場合でも、10g/kg以上である。
本発明の薬剤は、単独で、または、1種またはそれ以上のさらなる有効成分、特に、眼疾患の処置および/または予防のための有効成分と併用できる。
本発明に関して、成分を「併用する」ことは、全成分を含有する投与剤形の使用および各成分を別個に含有する投与剤形の組合せの使用のみならず、それらが同じ疾患の予防および/または処置に使用される限り、各成分を同時に、または、いずれかの成分を遅延して投与することも意味する。2種またはそれ以上のさらなる有効成分を併用することも可能である。
眼疾患が緑内障である場合、併用に適する有効成分には、例えば、交感神経刺激薬(エピネフリン、ジピベフリン等の非選択性刺激薬、アプラクロニジン、ブリモニジン等のα2刺激薬)、交感神経遮断薬(チモロール、ベフノロール、カルテオロール、ベタキソロール、ニプラジロール、レボブノロール等のβ遮断薬、ブナゾシン等のα1遮断薬)、副交感神経作動薬(ピロカルピン、ジスチグミン等)、炭酸脱水酵素阻害薬(アセタゾラミド、ブリンゾラミド、ドルゾラミド等)、プロスタグランジン類(イソプロピルウノプロストン、ラタノプロスト、トラボプロスト、ビマトプロスト、タフルプロスト等)、Rhoキナーゼ阻害剤(リパスジル塩酸塩等)などの眼圧降下剤が含まれ、特に、ラタノプロスト、トラボプロストおよびタフルプロストなどのプロスタグランジン類、β遮断薬、炭酸脱水酵素阻害薬並びにブリモニジンが含まれる。
眼疾患が加齢黄斑変性である場合、併用に適する有効成分には、例えば、抗VEGF剤(ラニビズマブ、ペガプタニブ、アフリベルセプト、ベバシズマブなど)の抗血管新生薬およびステロイド剤(トリアムシノロンアセトニドなど)が含まれ、特に、ペガプタニブおよびラニビズマブが含まれる。
併用に適する他の有効成分には、例えば、ビタミン12などのビタミン類、ルテインなどのカロテノイド類が含まれる。
本発明の薬剤の投与に加えて、薬物療法以外の眼疾患の治療法を実施することもできる。適する治療法には、例えば、外科手術、光線力学療法、遺伝子治療、再生医療、人工網膜移植、レーザー治療が含まれる。
別の態様では、本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、網膜の細胞の変性および/または細胞死の抑制剤を提供する。好ましい態様では、網膜の細胞は、視細胞、網膜色素上皮細胞および網膜神経節細胞を含む。
本発明の網膜の細胞の変性および/または細胞死の抑制剤は、上述の眼疾患の処置および/または予防剤と同様に製造および使用することができる。本発明において、網膜の細胞の変性および/または細胞死、並びにその抑制は、当分野で知られている方法により、例えば、光干渉断層検査や補償光学走査型レーザー検眼鏡により、確認することができる。
本発明の網膜の細胞の変性および/または細胞死の抑制剤は、網膜の細胞の変性および/または細胞死に起因する眼疾患を処置および/または予防することができる。また、本発明の網膜の細胞の変性および/または細胞死の抑制剤は、網膜の細胞の変性および/または細胞死を抑制することにより、網膜の細胞を保護することができる。また、本発明の網膜の細胞の変性および/または細胞死の抑制剤は、網膜の細胞の変性および/または細胞死に起因する視機能の低下を抑制することができる。
さらなる態様では、本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、視機能低下の抑制剤を提供する。
本発明の視機能低下の抑制剤は、上述の眼疾患の処置および/または予防剤と同様に製造および使用することができる。本発明において、視機能の低下は、矯正視力の低下および/または視野の異常、色覚の異常、を意味し、夜盲、視野欠損、視野狭窄、変視、失明などを含む。視機能の低下およびその抑制は、当分野で知られている方法により、例えば、視力検査、視野検査、網膜電図検査、網膜感度検査、中心フリッカー検査、色覚検査、視覚誘発電位などにより、確認することができる。ある態様では、視機能の低下は、網膜の細胞の変性および/または細胞死に起因する。他の態様では、視機能の低下は眼疾患に起因する。
また、ある態様では、本発明は、眼疾患の処置および/または予防用の医薬を製造するための、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種の使用を提供する。
他の態様では、本発明は、網膜の細胞の変性および/または細胞死を抑制するための医薬を製造するための、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種の使用を提供する。
他の態様では、本発明は、視機能低下を抑制するための医薬を製造するための、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種の使用を提供する。
これらの態様では、上述の眼疾患の処置および/または予防剤の製造方法に準じて、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を、医薬の製造のために使用できる。
また、ある態様では、本発明は、眼疾患の処置および/または予防において使用するための、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を提供する。
他の態様では、本発明は、網膜の細胞の変性および/または細胞死の抑制において使用するための、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を提供する。
他の態様では、本発明は、視機能低下の抑制において使用するための、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を提供する。
これらの態様では、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種は、上述の眼疾患の処置および/または予防剤に準じて使用される。
また、ある態様では、本発明は、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を対象に投与することを含む、眼疾患の処置および/または予防方法を提供する。
他の態様では、本発明は、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を対象に投与することを含む、網膜の細胞の変性および/または細胞死の抑制方法を提供する。
他の態様では、本発明は、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を対象に投与することを含む、視機能低下の抑制方法を提供する。
これらの態様では、上述の眼疾患の処置および/または予防剤の使用方法に準じて、有効成分としてのイソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を対象に投与することができる。
さらに、本発明の有効成分である分岐鎖アミノ酸は、眼疾患のリスクの低減、または、視機能の維持もしくは改善の効果を有する機能性食品の形態でも簡便に使用することができる。従って、本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品を提供する。本発明の食品は、本発明の食品の有効成分、すなわち分岐鎖アミノ酸を含む一般的な食事形態であればいかなるものでも良い。例えば、適当な風味を加えてドリンク剤、例えば清涼飲料、粉末飲料とすることもできる。具体的には、例えば、ジュース、牛乳、菓子、ゼリー、ヨーグルト、飴等として飲食することができる。また、例えば、製造業者または消費者により他の食品に添加して摂取される食品添加剤の形態であっても良く、経口投与用の医薬と同様の形態であっても良い。
また、このような食品を、保健機能食品またはダイエタリーサプリメントとして提供することも可能である。この保健機能食品には、特定保健用食品および栄養機能食品なども含まれる。特定保健用食品は、例えば、眼疾患のリスクの低減、または、視機能の維持もしくは改善など、特定の保健の目的が期待できることを表示できる食品である。また、栄養機能食品は、1日あたりの摂取目安量に含まれる栄養成分量が、国が定めた上・下限値の規格基準に適合している場合その栄養成分の機能の表示ができる食品である。ダイエタリーサプリメントには、いわゆる栄養補助食品または健康補助食品などが含まれる。本発明において、特定保健用食品には、眼疾患のリスクの低減、または、視機能の維持もしくは改善などの用途に用いるものであるという表示を付した食品、さらには、かかる用途に用いるものである旨を記載した書類(いわゆる能書き)などをパッケージとして包含する食品なども含まれるものとする。
本発明の食品の摂取量、摂取期間は、本発明の薬剤の投与量、投与期間の記載に基づき、本発明の食品が所望の効果を示すように適宜設定することができる。
他の態様では、本発明は、眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品を製造するための、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種の使用を提供する。別の態様では、本発明は、眼疾患のリスクの低減、または、視機能の維持もしくは改善のための、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種の使用を提供する。別の態様では、本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を含む食品を摂取または投与することによる、眼疾患のリスクの低減、または、視機能の維持もしくは改善方法を提供する。別の態様では、本発明は、イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を含む食品を投与することを含む、眼疾患のリスクを低減する、または、視機能を維持もしくは改善する非治療的方法を提供する。
下記実施例は本発明を説明するが、その範囲を限定するものではない。
以下の実施例において、ロイシン、イソロイシン及びバリンを2:1:1.2の割合で含む分岐鎖アミノ酸の混合物をBCAAとして使用した。BCAAはγ-シクロデキストリン水溶液(CAVAMAX W8, Wacker fine chemicals)への溶解後に投与した。

実施例1:比較的高齢の網膜色素変性モデルにおける薬理試験
網膜色素変性モデルとして、rd12マウス(The Jackson Laboratory)(Pang, J.J., Chang, B., Hawes, N.L., et al. 2005. Retinal degeneration 12 (rd12): a new, spontaneously arising mouse model for human Leber congenital amaurosis (LCA). Molecular vision 11, 152-162)を用いた。このマウスモデルでは、ゆっくりと視細胞の細胞死が進行する。13か月齢のrd12マウス34匹を、BCAA投与群(17匹)および対照群(17匹)の2群に分けた。13か月齢からBCAA群ではBCAA7.5g/lを含む水の自由飲水により、BCAAを経口投与した(約1.6kg/kg/日)。対照群には、同じ方法で水のみを与えた。

13か月齢および19か月齢にて、光干渉断層検査(optical coherence tomography;OCT)を行った。ケタミン70mg/kg+キシラジン14mg/kgの筋肉注射によりマウスを麻酔した。トロピカミド・フェニレフリン点眼により、マウスを散瞳させた。光干渉断層検査には、25Dアダプターレンズを装着した Spectralis HRT+OCT(Heidelberg Engineering, Inc.)を使用した。マウス角膜にポリメタクリル酸メチル製のコンタクトレンズを設置し、視神経乳頭を中心に縦に30度×15度の範囲を連続撮影した。視神経乳頭中心の366μmの範囲(中心の122μmの範囲は除外)の全層網膜厚を測定した。
全層網膜厚測定の結果を図1に示す。13か月齢では、平均全層網膜厚は対照群で202.4±9.0μm、BCAA群で203.0±9.0μmであり、有意差は認められなかった(P=0.40)。19か月齢では、平均全層網膜厚は対照群で184.9±10.2μm、BCAA群で194.4±12.6μmであり、BCAA群の方が厚かった(P=0.0049)。
同様の方法で、視細胞層厚を測定した。視神経乳頭を中心に垂直に30度のラインスキャンを撮影した。視神経乳頭より244μm上下の部位で視細胞層厚(外顆粒層厚と視細胞内節・外節厚の和)を測定した。視細胞層厚測定の結果を図2に示す。13か月齢では、平均視細胞層厚は対照群で58.5±6.3μm、BCAA群で59.7±4.6μmであり、有意差は認められなかった(P=0.20)。19か月齢では、視細胞層厚は対照群で39.4±7.4μm、BCAA群で45.3±6.8μmであり、BCAA群の方が厚かった(P=0.007)。
19か月齢にて、明順応網膜電図検査(electroretinogram;ERG)を行った。全身麻酔(ケタミン70mg/kg+キシラジン14mg/kg筋注)下で検査を行った。トロピカミド・フェニレフリン点眼により、マウスを散瞳させた。コンタクトレンズ型LED電極(メイヨー)を用い、LS-W(メイヨー)にて10cds/m2の刺激光で刺激し、PowerLab 2/25(AD Instruments, New South Wales, Australia)にて、波形を記録し、明順応網膜電図のb波振幅を測定した。結果を図3に示す。b波の振幅は、対照群で12.2±5.0μV、BCAA群で14.9±6.0μVであり、BCAA群の方が大きかった(P=0.046)。
19か月齢にて、眼球を摘出し、4%パラホルムアルデヒドにより終夜固定し、網膜をヘマトキシリン・エオシン(HE)染色した。視神経を通る垂直断切片を用いて、視神経乳頭縁から400-500μmの範囲の外顆粒層の細胞数を計測した(各n=4眼)。図4に示す通り、オスでは、細胞数は対照群で78.8±5.0個、BCAA群で100±11.2個であり、BCAA群の方が多かった(P=0.007)。全体では、細胞数は対照群で95.3±20.0個、BCAA群で101.1±16.4個であり、BCAA群の方が多かった(P=0.26)。
19か月齢で摘出した眼球から、網膜外組織(網膜色素上皮、脈絡膜および強膜を含む)および神経網膜を取り出し、CHOP:GADD153(R-20)抗体(Santa Cruz Biotechnology)、1:1000希釈、カスパーゼ-3:切断型カスパーゼ-3(cleaved caspase-3)(D175) 抗体(cell signaling)、1:1000希釈を使用して、ウェスタンブロットを行った。対照として、アクチン:抗アクチン抗体(Millipore)、1:10000希釈を使用した。結果を図5に示す。小胞体ストレスマーカーとして知られているCHOPは、網膜外組織で、BCAA群で抑制されていた。アポトーシスのマーカーである切断型カスパーゼ-3は、網膜で、BCAA群で抑制されていた。
これらの結果は、進行期の網膜色素変性のモデルである比較的高齢のrd12マウスにおいて、BCAAが視細胞の変性および細胞死を抑制し、視機能の低下を抑制したことを示す。
実施例2:正常眼圧緑内障モデルにおける薬理試験
進行が遅い正常眼圧緑内障のマウスモデルであるGLAST欠損(ヘテロ+/-)マウス(The Journal of Clinical Investigation, Volume 117, Number 7, July 2007)をマウスB6.Cg-Tg (Thy 1-CFP) 23Jrs/Jと交配し、GLAST欠損(+/-)・Thy 1-CFPマウスを得、正常眼圧緑内障のモデルマウスとして使用した。4週齢のモデルマウス30匹を、対照群(15匹)およびBCAA投与群(15匹)の2群に分けた。BCAA群では、4週齢から8週齢にわたり、0.375g/kg/日のBCAAを週5日、腹腔内投与し、8週齢以降はBCAA7.5g/lを含む水の自由飲水により、BCAAを経口投与した(約1.7g/kg/日)。対照群には、4週齢から8週齢にわたり、生理食塩水を週5日、腹腔内投与し、8週齢以降は自由飲水によりBCAAを含まない水を与えた。

12か月齢でマウスから眼球を摘出し、4%パラホルムアルデヒドにて1時間固定後、網膜をフラットマウントにし、視神経乳頭中心から1200μmの位置で、250μm×250μmの範囲で、CFP蛍光をもつ細胞の数を計測した(図6)。これらの細胞は、網膜神経節細胞と考えられる。細胞数は、対照群で110±9.6個、BCAA群で124.8±7.5個であり、BCAA群の方が多かった(P=0.016)。
18か月齢で、マウスから眼球を摘出し、4%パラホルムアルデヒドにて終夜固定後、パラフィンに包埋し、眼球後方の視神経部を垂直断した切片にて、視神経断面を写真撮影し、視神経乳頭径を測定した(図7)。視神経乳頭径は、神経節細胞の数を反映する。視神経乳頭径の中央値は、対照群で45352μm2、BCAA群で70637μm2であり、BCAA投与群の方が高かった。
これらの結果は、正常眼圧緑内障モデルにおいて、BCAAが網膜神経節細胞の変性および細胞死を抑制したことを示す。
実施例3:比較的老齢の加齢黄斑変性モデルにおける薬理試験
加齢黄斑変性のモデルとして、CCR2(単球走化活性化因子(MCP-1)受容体)欠損マウス(The Jackson Laboratory)(Nat Med. 2003 Nov;9(11):1390-7. Epub 2003 Oct 19. An animal model of age-related macular degeneration in senescent Ccl-2- or Ccr-2-deficient mice. Ambati J1, Anand A, Fernandez S, Sakurai E, Lynn BC, Kuziel WA, Rollins BJ, Ambati BK)を用いた。9か月齢のCCR2欠損マウス34匹を、対照群(24匹)およびBCAA投与群(10匹)の2群に分けた。CCR2欠損マウスに対して、9か月齢から、γ-シクロデキストリン水溶液に溶解したBCAA(7.5g/l)を含む水の自由飲水により、BCAAを経口投与した。対照マウスには、同じ方法で水のみを与えた。

9か月齢から15か月齢まで1か月毎に、視神経乳頭を中心とした眼底カラー写真を撮影し(Micron III, Phoenix Research Laboratories)、ドルーゼン数を評価した。代表的な写真を図8に示す。白色の粒状の構造がドルーゼンである。図中、白点線は、ドルーゼンが多い場所を示す。BCAA投与群で、ドルーゼン数の増加が抑制されるものが多く存在した。投与開始9か月齢でのドルーゼン数からの各月のドルーゼン数の増減を図9に示す。BCAA投与群(n=14眼)では、対照群(n=16眼)と比較して、10か月齢以降、ドルーゼンの増加が有意に抑制された(P=0.043(10か月齢)、0.015(11か月齢)、0.046(12か月齢)、0.015(13か月齢)、0.018(14か月齢)、0.016(15か月齢))。
15か月齢にて、暗順応網膜電図検査(ERG)を行った。ERGの24時間前からマウスを暗順応させ、全身麻酔(70mg/kgケタミン+14mg/kgキシラジン筋注)、散瞳下(0.5%フェニレフリン、5%ネオシネジン)で検査を行った。暗順応下にて、コンタクトレンズ型LED電極を用い、LS-Wにて0.01cds/m2の刺激光で刺激してPowerLab 2/25にて波形を記録し、b波の振幅を測定し、同様に3cds/m2の刺激光で刺激して波形を記録し、a波の振幅を測定した。結果を図10に示す。a波の振幅は、対照群で163.6±65.2μV、BCAA群で207.0±47.4μVであり、BCAA群の方が大きかった(P=0.0026)。b波の振幅は、対照群で223.1±82.0μV、BCAA群で308.4±56.8μVであり、BCAA群の方が大きかった(P=0.0016)。
15か月齢にて、眼球を摘出し、4%パラホルムアルデヒドにより終夜固定し、網膜をHE染色し、網膜切片を観察した。図11は、15か月齢におけるCCR2欠損マウスの網膜切片を示す(上段:40倍、下段:100倍)。水のみを投与した対照群では、網膜色素上皮に空胞変性を生じているのに対し(矢頭)、BCAA投与群では、空胞変性は認められなかった(上段)。また、対照群のブルッフ膜(網膜色素上皮の基底膜)は、BCAA投与群に比べ、著明に肥厚していた(下段、矢印)。
15か月齢にて、網膜の電子顕微鏡検査を行った。眼球を摘出し、HE染色と同様の方法で固定後、1%四酸化オスミウムで固定した。エタノール脱水、エポキシ樹脂包埋の後、超薄切片を作成、酢酸ウラニルおよびクエン酸鉛で染色し、染色切片を透過型電子顕微鏡(H-7650、日立株式会社)で観察した。結果を図12に示す。図12中、RPEは網膜色素上皮を示す。網膜下蓄積物は、対照に比べて、BCAA投与群にて少なかった。
これらの結果は、比較的老齢の加齢黄斑変性モデルマウスにおいて、BCAAがドルーゼンの増加を抑制し、視機能の低下を抑制したことを示す。
実施例4:比較的若年の加齢黄斑変性モデルにおける薬理試験
2か月齢のCCR2欠損マウス37匹を、BCAA投与群(11匹)および対照群(26匹)の2群に分けた。CCR2欠損マウスに対して、2か月齢から、γ-シクロデキストリン水溶液に溶解したBCAA(7.5g/l)を含む水の自由飲水により、BCAAを投与した。対照マウスには、同じ方法で水のみを与えた。
2か月齢のCCR2欠損マウス37匹を、BCAA投与群(11匹)および対照群(26匹)の2群に分けた。CCR2欠損マウスに対して、2か月齢から、γ-シクロデキストリン水溶液に溶解したBCAA(7.5g/l)を含む水の自由飲水により、BCAAを投与した。対照マウスには、同じ方法で水のみを与えた。
9か月齢にて、暗順応網膜電図検査(ERG)を行った。ERGの24時間前からマウスを暗順応させ、全身麻酔(70mg/kgケタミン+14mg/kgキシラジン筋注)、散瞳下(0.5%フェニレフリン、5%ネオシネジン)で検査を行った。暗順応下にて、コンタクトレンズ型LED電極を用い、LS-Wにて0.01cds/m2の刺激光で刺激し、PowerLab 2/25にて波形を記録し、b波の振幅を測定し、同様に3cds/m2の刺激光で刺激して波形を記録し、a波の振幅を測定した(対照 n=43眼、BCAA n=17眼)。結果を図13に示す。a波の振幅は、対照群で174.6±69.6μV、BCAA群で251.1±72.3μVであり、BCAA群の方が大きかった(P=0.0011)。b波の振幅は、対照群で253.1±113.2μV、BCAA群で386.7±133.6μVであり、BCAA群の方が大きかった(P=0.0010)。
これらの結果は、比較的若年の加齢黄斑変性モデルマウスにおいて、BCAAが視機能の低下を抑制したことを示す。
実施例5:網膜色素変性モデルにおける薬理試験
網膜色素変性モデルとして、rd10マウス(B6.CXB1-Pde6brd10/J(Jackson Laboratory (Bar Harbor, ME)より購入、Chang B. et al; Vision Res (2007))を用いた。このマウスモデルでは、2か月齢までに急速に視細胞が変性する。7日齢のrd10マウス35匹を、BCAA投与群(17匹)および対照群(18匹)の2群に分けた。BCAA群では、7日齢から23日齢まで、1.5g/kg/日のBCAAを腹腔内投与し、24日齢以降はBCAA7.5g/lを含む水の自由飲水により、BCAAを経口投与した(約1.7g/kg/日)。対照群には、7日齢から23日齢まで生理食塩水を腹腔内投与し、24日齢以降は自由飲水によりBCAAを含まない水を与えた。

24日齢では、平均全層網膜厚は対照群で188.9±21.8μm、BCAA群で187.2±13.8μmであり、有意差は認められなかった(P=0.36)。30日齢では、平均全層網膜厚は対照群で153.7±7.4μm、BCAA群で155.0±6.3μmであり、有意差は認められなかった(P=0.27)。37日齢では、平均全層網膜厚は対照群で144.2±5.7μm、BCAA群で149.8±3.3μmであり、BCAA群の方が厚かった(P<0.01)。37日齢の全層網膜厚測定の結果を図14に示す。
同様の方法で、視細胞層厚を測定した。視神経乳頭を中心に縦に30度のラインスキャンを撮影した。視神経乳頭より244μm上下の部位で視細胞層厚(外顆粒層厚と視細胞内節・外節厚の和)を測定した。24日齢では、平均視細胞層厚は対照群で50.4±23.8μm、BCAA群で42.6±19.1μmであり、有意差は認められなかった(P=0.09)。30日齢では、平均視細胞層厚は対照群で16.8±3.6μm、BCAA群で18.9±6.0μmであり、有意差は認められなかった(P=0.09)。37日齢では、視細胞層厚は対照群で10.1±4.8μm、BCAA群で15.1±3.3μmであり、BCAA群の方が厚かった(P<0.01)。37日齢の視細胞層厚測定の結果を図15に示す。視細胞層厚の中央値をとる37日齢マウスの光干渉断層計像を、図16に示す。視機能と相関することが知られているエリプソイド帯(Ellipsoid zone)の連続性はBCAA群のほうが対照群よりも保たれているのが見られる。
24日齢にて、暗順応網膜電図検査(ERG)を行った。ERGの前に終夜、マウスを暗順応させ、全身麻酔(70mg/kgケタミン+14mg/kgキシラジン筋注)下で検査を行った。トロピカミド・フェニレフリン点眼により、マウスを散瞳させた。暗順応下にて、コンタクトレンズ型LED電極を用い、LS-Wにて0.01cds/m2および3.0cds/m2の刺激光で刺激してPowerLab 2/25にて波形を記録し、b波の振幅を測定した。結果を図17に示す。b波の振幅は、刺激光強度0.01cds/m2にて、対照群で18.8±16.8μV、BCAA群で40.1±50.2μV(P<0.01)であり、刺激光強度3.0cds/m2にて、対照群で76.4±61.3μV、BCAA群で121.3±115.9μV(P=0.02)であり、いずれもBCAA群の方が大きかった。
30日齢および37日齢にて、明順応網膜電図検査を行った。全身麻酔(ケタミン70mg/kg+キシラジン14mg/kg筋注)下で検査を行った。トロピカミド・フェニレフリン点眼により、マウスを散瞳させた。マウスにコンタクトレンズ型LED電極を装着させ、LS-Wにて3cds/m2、10cds/m2および30cds/m2の刺激光で刺激し、PowerLab 2/25にて波形を記録し、明順応網膜電図のb波振幅を測定した。30日齢において、b波の振幅は、刺激光強度3cds/m2にて、対照群で10.0±9.7μV、BCAA群で14.8±11.0μV(P=0.03)であり、刺激光強度10cds/m2にて、対照群で15.7±12.0μV、BCAA群で24.9±21.2μV(P=0.01)、刺激光強度30cds/m2にて、対照群で20.6±16.3μV、BCAA群で36.3±22.4μV(P<0.001)であり、いずれもBCAA群の方が大きかった(図18)。37日齢では、刺激光強度3cds/m2でのb波の振幅は、対照群で8.7±4.0μV、BCAA群で9.3±4.4μVであり、有意差は認められなかった(P=0.29)。刺激光強度10cds/m2では、対照群で6.7±3.1μV、BCAA群で15.6±11.0μV(P<0.0001)、刺激光強度30cds/m2では、対照群で9.7±5.5μV、BCAA群で20.5±14.0μV(P<0.0001)であり、いずれもBCAA群の方が大きかった。b波振幅の中央値をとる30日齢マウスの明順応網膜電図検査の結果を、図19に示す。BCAA群のほうが対照群よりも波形が保たれているのが見られる。
これらの結果は、網膜色素変性モデルにおいて、BCAAが視細胞の変性および細胞死を抑制し、視機能の低下を抑制したことを示す。

- イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、眼疾患の処置および/または予防剤。
- イソロイシンを有効成分として含有する、請求項1に記載の眼疾患の処置および/または予防剤。
- イソロイシンを有効成分として含有し、かつロイシンおよびバリンを有効成分として含有しない、請求項1または請求項2に記載の眼疾患の処置および/または予防剤。
- イソロイシン、ロイシンおよびバリンを有効成分として含有する、請求項1に記載の眼疾患の処置および/または予防剤。
- イソロイシン、ロイシンおよびバリンの重量比が、1:1.5~2.5:0.8~1.7である、請求項4に記載の眼疾患の処置および/または予防剤。
- 眼疾患が、緑内障、網膜色素変性または加齢黄斑変性である、請求項1ないし請求項5のいずれかに記載の眼疾患の処置および/または予防剤。
- 眼圧降下剤と併用される、請求項1ないし請求項6のいずれかに記載の眼疾患の処置および/または予防剤。
- 抗VEGF剤と併用される、請求項1ないし請求項6のいずれかに記載の眼疾患の処置および/または予防剤。
- イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、網膜の細胞の変性および/または細胞死の抑制剤。
- 網膜の細胞が、視細胞、網膜色素上皮細胞および網膜神経節細胞からなる群から選択される、請求項9に記載の網膜の細胞の変性および/または細胞死の抑制剤。
- イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、視機能低下の抑制剤。
- イソロイシン、ロイシンおよびバリンからなる群から選択される少なくとも1種を有効成分として含有する、眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品。
- イソロイシン、ロイシンおよびバリンを有効成分として含有する、請求項12に記載の眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品。
- イソロイシン、ロイシンおよびバリンの重量比が、1:1.5~2.5:0.8~1.7である、請求項13に記載の眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品。
- 眼疾患が、緑内障、網膜色素変性または加齢黄斑変性である、請求項12ないし請求項14のいずれかに記載の眼疾患のリスクの低減、または、視機能の維持もしくは改善用の食品。
