Abstract
分離検出ユニットは、試料を成分ごとに分離する分析カラムと、分析カラムで分離された試料成分を検出する検出部と、分析カラム及び検出部を内部に収容するカラムオーブンと、を備えている。検出部は、分析カラムからの液を収容する液収容部、分析カラムの一端に接続され分析カラムからの液を液収容部に流入させる入口部、及び液収容部の液を流出させる出口部を有する試料セルと、試料セルに対して分光器を介することなく光を照射する発光ダイオード又はレーザダイオードを有する光源部と、光源部から発せられ試料セルを透過した光又は光源部からの光により励起された試料セル内の試料から発せられた光を検出する検出素子を備えている。カラムオーブンは、分析カラムの他端に通じる入口ポート及び試料セルの出口部に通じる出口ポートを備えている。
Description
本発明は、試料を成分ごとに分離して検出する分離検出ユニットとその分離検出ユニットを備えた液体クロマトグラフに関するものである。
液体クロマトグラフは、移動相を送液する送液装置、移動相の流れる分析流路、分析流路中に試料を導入する自動試料注入装置(オートサンプラ)、試料を成分ごとに分離する分析カラム、及び分析カラムで分離した試料成分を検出する検出器により構成される。送液装置やオートサンプラ、分析カラム、検出器はそれぞれ独立したユニットとして構成されることが一般的であり、それらのユニットが配管で接続されることで分析流路が構成される。
例えば分析カラムの分離特性は温度依存性をもつため、分析カラムの温度を一定で保持するために温度調節機能を有するカラムオーブン内に収容され、カラムオーブンユニットとして構成される。また、検出器は、試料セルや光源、光源から発せられる光から測定波長の光を抽出して試料セルに導く光学系、試料セルからの光を検出する検出器などによって構成されるが、これらは共通の筐体内に収容され、検出ユニットとして構成される。
分析装置の検出器である光度計の光源として発光ダイオード(LED)やレーザダイオード(LD)を用いることが知られている(特許文献1参照。)。光源としてLEDやLDを用いることにより、分光素子である回折格子が不要になるという利点がある。
液体クロマトグラフにおいて、分析カラムから検出器の試料セルまでの間での試料成分の拡散により、検出ピークがブロードになって検出感度が低下するという問題がある。この問題は、分析カラムから試料セルまでの容積(デッドボリューム)が大きくなるほど顕著になる。そのため、分析カラムと試料セルとの間の容積を小さくする必要がある。
しかし、分析カラムを有するカラムオーブンユニットや検出ユニットなど複数のユニットが組み合わされて構成された液体クロマトグラフでは、カラムオーブンユニットと検出ユニットとの間を接続する配管が必要なため、分析カラムから試料セルまでのデッドボリュームを小さくするのに限界がある。
そこで、本発明は、分析カラムから試料セルまでのデッドボリュームを小さくして分析カラムで分離した試料成分の拡散を抑制することを目的とするものである。
本発明に係る分離検出ユニットの一実施形態は、試料を成分ごとに分離する分析カラムと、分析カラムで分離された試料成分を検出する検出部と、分析カラム及び検出部を内部に収容するカラムオーブンと、を備えている。検出部は、分析カラムからの液を収容する液収容部、分析カラムの一端に接続され分析カラムからの液を液収容部に流入させる入口部、及び液収容部の液を流出させる出口部を有する試料セルと、試料セルに対して分光器を介することなく光を照射する発光ダイオード(以下、LED)又はレーザダイオード(以下、LD)を有する光源部と、光源部から発せられ試料セルを透過した光又は光源部からの光により励起された試料セル内の試料から発せられた光を検出する検出素子を備えている。カラムオーブンは、分析カラムの他端に通じる入口ポート及び試料セルの出口部に通じる出口ポートを備えている。
本発明に係る液体クロマトグラフの一実施形態は、移動相を送液する送液ユニットと、送液ユニットと配管を通じて接続され、送液ユニットからの移動相が流れる流路中に試料を導入する自動試料注入装置と、自動試料注入装置と配管を通じて接続され、自動試料注入装置により導入された試料を成分ごとに分離する分離カラムとその分離カラムにより分離された試料成分を検出する検出部を有する上記分離検出ユニットと、を備えている。
本発明に係る分離検出ユニットの一実施形態では、検出部が、試料セルに対して分光器を介することなく光を照射するLED又はLDを有する光源部を備えているので、分光器が不要になり、検出部全体が小型化され、分析カラムを収容するカラムオーブンに検出部も収容される。これにより、分析カラムと試料セルとを接続する配管が短くてすみ、分析カラムから試料セルまでのデッドボリュームが小さくなり、試料成分の拡散による検出感度の低下を抑制することができる。
本発明に係る液体クロマトグラフの一実施形態では、上記分離検出ユニットを備えているので、試料成分の検出を高感度で行なうことができる。
光源部は、互いに異なる波長範囲の光を発する複数のLED又はLDを有するものであってもよい。そうすれば、それらのLED又はLDの発光と消灯の切替えによって測定波長の切替えを容易に行なうことができる。
カラムオーブンの内部に、複数の分析カラムと、各分析カラムに対応する複数の検出部が収容されていてもよい。そうすれば、複数のカラムオーブンユニットやそれらのカラムオーブンユニットに対応する検出ユニットを用意することなく、複数の分析カラムにおける試料の分離と分離された試料成分の検出を行なうことができる。
分析カラムの一端と試料セルの入口部とが配管を介することなく接続されていてもよい。そうすれば、分析カラムから試料セルまでの間のデッドボリュームがさらに小さくなり、試料の拡散による検出感度の低下をさらに抑制することができる。
図1を用いて、液体クロマトグラフの一実施例について説明する。
この実施例の液体クロマトグラフは、送液ユニット2、オートサンプラ4、分離検出ユニット6及び制御ユニット8を備えている。送液ユニット2は配管を接続するための出口ポート10を有し、その出口ポート10が配管20を介してオートサンプラ4の入口ポート12と接続されている。オートサンプラ4は入口ポート12のほかに出口ポート14も有し、その出口ポート14が配管22を介して分離検出ユニット6の入口ポート16と接続されている。分離検出ユニット6も入口ポート16のほかに出口ポート18を有し、出口ポート18はドレインへ通じている。
送液ユニット2は移動相を送液するための送液ポンプ24を備えている。送液ポンプ24によって送液される移動相は出口ポート10から配管20を通じてオートサンプラ4に導入される。
オートサンプラ4は、送液ユニット2から供給される移動相の流れる流路中に試料を導入する。導入された試料は移動相とともに配管22を通じて分離検出ユニット6に導入される。
分離検出ユニット6は、試料を成分ごとに分離する分析カラム28と、分析カラム28で分離した試料成分を検出する検出部30を備えている。オートサンプラ4において移動相の流れる流路中に導入された試料は移動相とともに分析カラム28に導入され、成分ごとに分離される。分析カラム28において分離した試料成分は順次検出部30に設けられている試料セルに導入され、検出される。検出部30を経た試料成分は移動相とともに出口ポート18からドレインへ排出される。
送液ユニット2、オートサンプラ4及び分離検出ユニット6は共通の制御ユニット8によりその動作が制御される。また、分離検出ユニット6の検出部30で得られた検出信号は制御ユニット6に取り込まれ、試料中に含まれる成分の特定や定量がなされる。制御ユニット8は、汎用のパーソナルコンピュータ又は専用のシステムコントローラなどのコンピュータによって実現される。
分離検出ユニット6の一実施例について図2を用いて説明する。
分離検出ユニット6は断熱性のカラムオーブン32を有する。カラムオーブン32の外側側面に配管端部を接続させる入口ポート16と出口ポート18が設けられている。カラムオーブン32の内部空間34に分析カラム28及び検出部30が収容されている。カラムオーブン32の大きさは、例えば幅寸法が250mm程度、高さ寸法が350mm程度、奥行寸法が100mm程度である。分析カラム28の大きさは、例えば外径が8mm程度、長さが150mmmm程度である。
この実施例では、分析カラム28の入口28aが下側、出口28bが上側になるように縦方向に配置されている。分析カラム28は良好な熱伝導率を有する金属からなるカラムホルダ36により保持されている。カラムホルダ36にはヒータが埋設されており(図示は省略)、そのヒータの出力の増減によって分析カラム28の温度が予め設定された温度に制御される。分析カラム28の入口28aは配管チューブ38を介して入口ポート16に接続され、分析カラム28の出口28bは配管チューブ40を介して検出部30の試料セル42の入口42aに接続されている。
なお、この実施例では、ヒータの埋設されたカラムホルダ36によって分析カラム28を直接的に加温して温度制御を行なう直接温調方式が採用されているが、これに代えて、ヒータとファンによってカラムオーブン32の内部空間34内全体の温度を調節する間接温調方式を採用してもよい。
検出部30は分析カラム28の上方に設けられている。検出部30は、試料セル42、発光素子44及び検出素子46を備えている。試料セル42は光透過性材料で構成されている。試料セル42は例えば35mm×40mm×80mm程度の大きさをもつものである。試料セル42の入口部42aは配管チューブ40を介して分析カラム28の出口28bと接続され、出口部42bは配管チューブ48を介して出口ポート18と接続されている。配管チューブ40の長さは60mm以下である。なお、試料セル42aの入口部42aと分析カラム28の出口28bは配管を介することなく直接的に接続されていてもよい。
発光素子44はLED又はLDであり、試料セル42に向かって測定波長の光を照射する光源部をなすものである。発光素子44は、試料セル42の側方において試料セル42に向かって光を発するように配置されている。発光素子44として用いられるLEDとしては、例えば直径が8mm、長さが8mm程度の大きさで半値幅が15nm以下のものが挙げられる。発光素子44として用いられるLDとしては、例えば直径が8mm、長さが8mm程度の大きさで半値幅が5nm以下のものが挙げられる。検出素子46は例えばフォトダイオードであり、試料セル42を挟んで光源部44とは反対側に配置されている。
発光素子44から発せられる光は広がりをもつため、発光素子44から発せられる光を集光するための集光レンズを発光素子44と試料セル42との間に配置してもよい。
発光素子44として用いられている発光ダイオード又はレーザダイオードは、半値幅が数nm~数十nmと非常に小さいため、発光素子44から発せられた光は分光器を介することなく試料セル42に照射される。発光素子44から発せられる光から特定の波長範囲の光を取り出すための回折格子などの分光器が不要となる。回折格子などの分光器を設けた場合は、100mm×150mm×150mm程度の大きな空間が必要になるが、この実施例では分光器が不要なため、検出部30全体の大きさが小型化され、分析カラム28と同じカラムオーブン32内に収容することができる。分析カラム28と試料セル42とが同じカラムオーブン32内に収容されているため、分析カラム28と試料セル42の間を接続する配管を短くすることができる。これにより、分析カラム28から試料セル42までのデッドボリュームを小さくでき、分析カラム28から試料セル42までにおける試料成分の拡散を抑制して、検出感度の向上を図ることができる。
分析カラム28と試料セル42とを配管を介さずに接続することも考えられる。その一実施例を図6に示すが、このように分析カラム28と試料セル42とを直接的に接続すれば、分析カラム28と試料セル42との間のデッドボリュームをさらに小さくすることができる。この実施例では、図6(A)に示されているように、試料セル42の入口部42cと分析カラム28の出口28cとがメイルナットによって直接的に接続されるようになっている。
また、発光ダイオード又はレーザダイオードからなる発光素子は小型であるため、図3に示されているように、発光素子を複数並べて試料セル42の側方に配置することで、複数の波長の光を用いた測定が可能な検出部30aを構成することができる。図3の実施例では、互いに発光波長範囲の異なる2つの発光素子44aと44bが試料セル42の側方に配置されて光源部を構成している。かかる場合にも、ハーフミラーや分光器などの光学系が不要である。
図4の実施例に示されているように、発光素子44からの光の一部を参照光として検出するフォトダイオードなどの検出素子50を設けてもよい。そうすれば、発光ダイオード又はレーザダイオードの発光光量の変化を参照光によって検知することができ、それに基づいて検出素子46で得られる検出信号の補正を行なうことができる。
分離検出ユニットの別の実施例を図5に示す。
この実施例の分離検出ユニット6aは、カラムオーブン32aの内部空間34aに3つの分析カラム28とそれらの分析カラム28に対応する検出部30が収容されている。検出部30の構成は図2を用いて説明した実施例と同じであるため、ここでの説明は省略する。各分析カラム28は互いに分離特性が異なっている。分析カラム28とその分析カラム28に対応する検出部30の組合せの数は2つであってもよいし4つ以上であってもよい。
検出部30には分光器などの光学系がないため、分析カラム28と検出部30の組合せの一組の大きさを小型にすることができ、それらの組合せの複数組をカラムオーブン32aの大きさをさほど大きくすることなく共通のカラムオーブン32a内に収容して1つの分離検出ユニット6aとすることができる。
入口ポート16と各分析カラム28の入口28aとの間、出口ポート18と各試料セル42の出口部42bとの間には、入口ポート16から流入する移動相を分岐させて各分析カラム28の入口28aに接続し、各試料セル42の出口部42bから流出した移動相を共通の出口ポート18に接続するための分岐流路部52が設けられている。これにより、各分析カラム28の入口28aは共通の入口ポート16に接続され、各試料セル42の出口部42bは共通の出口ポート18に接続されている。
オートサンプラ4(図1参照)において導入された試料は入口ポート16からこの分離検出ユニット6a内に流入し、分岐流路部52で3本の流路へ分けられ、各分析カラム28に導入される。移動相とともに流入した試料は分離特性の異なる複数の分析カラム28において成分ごとに分離され、各分析カラム28に対応する検出部30において検出される。これにより、1つの試料を分離特定の異なる複数の分析カラム28で分離して分析することができる。
なお、図5の実施例において、検出部30に代えて、図3の実施例における検出部30a又は図4の実施例における検出部30bを用いてもよい。
さらにまた、上記各実施例や変形例は本発明の一例にすぎず、本発明の趣旨の範囲で適宜変形、修正、追加を行なっても本願特許請求の範囲に包含されることは当然である。
Claims (5)
- 試料を成分ごとに分離する分析カラムと、
前記分析カラムで分離された試料成分を検出する検出部と、
前記分析カラム及び前記検出部を内部に収容するカラムオーブンと、を備え、
前記検出部は、前記分析カラムからの液を収容する液収容部、前記分析カラムの一端に接続され前記分析カラムからの液を前記液収容部に流入させる入口部、及び前記液収容部の液を流出させる出口部を有する試料セルと、前記試料セルに対して分光器を介することなく光を照射する発光ダイオード又はレーザダイオードを有する光源部と、前記光源部から発せられ前記試料セルを透過した光又は前記光源部からの光により励起された前記試料セル内の試料から発せられた光を検出する検出素子と、を備え、
前記カラムオーブンは、前記分析カラムの他端に通じる入口ポート及び前記試料セルの出口部に通じる出口ポートを備えている分離検出ユニット。 - 前記光源部は、互いに異なる波長範囲の光を発する複数の前記発光ダイオード又は前記レーザダイオードを有する請求項1に記載の分離検出ユニット。
- 前記カラムオーブンの内部に、複数の前記分析カラムと、各分析カラムに対応する複数の前記検出部が収容されている請求項1又は2に記載の分離検出ユニット。
- 前記分析カラムの前記一端と前記試料セルの前記入口部とが配管を介することなく接続されている請求項1から3のいずれか一項に記載の分離検出ユニット。
- 移動相を送液する送液ユニットと、
前記送液ユニットと配管を通じて接続され、前記送液ユニットからの移動相が流れる流路中に試料を導入する自動試料注入装置と、
前記自動試料注入装置と配管を通じて接続され、前記自動試料注入装置により導入された試料を成分ごとに分離する分離カラムとその分離カラムにより分離された試料成分を検出する検出部を有する請求項1から4のいずれか一項に記載の分離検出ユニットと、を備えた液体クロマトグラフ。
