Abstract

본 발명은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵 예방용 백신 조성물에 관한 것이다. 본 발명에 따른 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 항원으로 이용할 경우, 결핵균에 의한 감염을 유발시킨 동물모델의 비장과 폐에서 높은 IFN-γ 생성능을 나타내며, 빠른 후천적 방어면역의 형성과 T 세포의 기억화를 유지시켜줌으로써 결핵을 효과적으로 예방할 수 있다. 또한, 본 발명에 따른 Rv3112, MTBK 20620 또는 MTBK 24820 단백질은 결핵의 치료 시에도 사용 가능하여 재발율이 높은 고병원성 결핵의 재발 또는 재감염을 예방하고 치료 효율을 높이는 데 유용하게 사용될 수 있다.

Description

Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵 예방용 백신 조성물
본 발명은 대한민국 미래창조과학부의 지원 하에서 과제번호 NRF-2013R1A2A1A01009932에 의해 이루어진 것으로서, 상기 과제의 연구관리전문기관은 "한국연구재단", 연구사업명은 "기초연구사업, 중견연구자지원사업, 도약연구_도전", 연구과제명은 "결핵의 질병단계 특이적 핵심병인면역 조절을 통한 면역화학 치료요법 개발", 주관기관은 "연세대학교 산학협력단", 연구기간은 "2015-06-01 ~ 2016-05-31"이다.
본 발명은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵 예방용 백신 조성물에 관한 것이다.
결핵은 WHO에서 규정한 3대 감염질환 중 하나로 1882년 미생물학자인 로베르트 코흐에 의해 발견되었으며, 마이코박테리움 튜버쿨로시스(Mycobacerium tuberculosis)에 의하여 유발되는 높은 발병률과 사망률을 보이는 치명적인 질병이다. 전 세계적으로 약 6천만명 환자가 활동성 결핵에 감염되어 있으며, 매년 약 5천만에서 1억여명이 새로 결핵에 감염되는 것으로 추정되고 있고, 적어도 매년 900만명 이상의 새로운 결핵 환자가 발생하며, 연간 150만 명 이상이 결핵으로 사망한다고 알려졌다. 결핵 발병률(incidence rate)은 인구 10만명 당 146명, 결핵 사망률은 인구 10만명 당 49명으로 단일 감염병 중에서 가장 많은 사망 원인을 차지하고 있어, 세계적으로 심각한 보건 문제로 남아 있다.
결핵균들의 우점적인 유전자형을 가진 베이징과의 결핵균은 전 세계에 널리 퍼져있으며, 주로 동아시아에 빈번하게 퍼져있다. 결핵균 중에서 베이징과에 속하는 유전자형을 가지는 빈도는 중국이 92%, 한국이 72%, 베트남이 40% 인 것으로 추정된다 (Glynn et al., 2002). 베이징과 결핵균의 유전자형은 단일약물 내성 및 다재내성을 포함한 결핵균의 약제내성과 관련되어 있다 (Buu et al., 2009). 또한 베이징과의 유전자형은 잠복기상태의 결핵균의 재발과도 밀접한 연관성을 가지고 있다 (Burman et al., 2009).
한국의 전체 결핵 발병률은 감소하고 있는 추세(직접도말양성결핵균의 유병률, 1965년에 10만명에 686명에서 1995년 10만명에 93명)에 있지만, 여전히 상대적으로 높은 비율(0.11%)이다. 최근 들어 우리나라에서 발병하는 결핵 환자 중에 약제내성을 나타내는 MDR-TB 환자와 XDR TB 환자가 급격하게 증가하는 추세에 있다. 한국 TB 발병 중 약 10%는 다재 내성 결핵(MDR-TB)이지만, 보다 우려되는 것은, 전체 다재내성 결핵균 중 광범위한 다재 내성 결핵(XDR-TB)의 비율이 5~15%로 나타났다는 것이다. 한국 내 결핵균의 유전자 지문은 베이징 결핵균과의 하나가 우점적으로 존재하는 것으로 나타났으며, 이 유전자형은 총 결핵균의 18.4%를 차지하는 것으로 나타났다 (Choi et al., 2010). 김 등(2001)은 한국에서 고등학교 학생들에게 결핵을 야기시킨 결핵균 중 베이징과에 속하는 전염성이 강한 결핵균을 확인하였으며, 이 균을 K 균이라 명명하였다.
한국형 고병원성 결핵균인 K 균주는 마우스 감염 시 초기 선천면역을 극복할 수 있는 특이적 병인기전을 나타내며 이는 결핵균 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv 균주 감염량의 10배에 해당되는 성장속도를 나타낸다. 한국형 고병원성 결핵균인 K 균주 감염에 의한 마우스의 폐사는 결핵균 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv 균주 감염에 의한 마우스의 폐사보다 2배 이상 빠르며, 감염 후 폐사 시기의 균수는 거의 동일한 것으로 보아 감염 후기에는 숙주의 면역반응에 의한 면역병리가 병인기전으로서 매우 중요할 것으로 생각되기에 선천면역보다는 예방백신 등을 통한 후천면역을 특이적으로 올려주는 것이 한국형 고병원성 결핵균인 K 균주를 방어하는 핵심 요소일 것으로 판단되었다.
현재, 결핵은 주로 마이코박테리움 보비스(Mycobacterium bovis)의 비독성 균주인 바실러스 칼메트-구에린(BCG: Bacillus Calmette-Guerin)의 생균을 이용하여 예방하고 있다. 그러나 BCG 백신은 한국형 고병원성 결핵균에는 그 효과가 없는 것으로 알려져 있으며, 안전성 및 효능에 논란이 있어, 미국과 같은 몇몇 국가는 이의 사용을 금지하고 있다. 또한, BCG는 HIV에 감염된 어린이들에게 사용하기에는 적당하지 못하다고 알려져 있다. 이에 BCG를 대체할 보다 안전하고 효과가 우수한 새로운 결핵 예방 백신의 개발이 요구되고 있다.
따라서, 본 발명자는 새로운 결핵 예방 백신을 개발하기 위한 연구를 계속한 결과, 기존의 결핵 표준 균주에는 존재하지 않고, 한국형 결핵균 K 균주에만 특이적으로 존재하는 병원성 인자 및 기존의 결핵 표준 균주에서보다 한국형 고병원성 결핵균인 K 균주에서 특이적으로 과발현되는 유전자를 찾아 규명하고, 이의 백신으로서의 이용 가능성을 확인함으로써 본 발명을 완성하였다.
본 발명의 목적은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵 예방용 백신 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵 예방용 백신 조성물을 제공한다.
또한, 본 발명은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵의 예방 또는 치료용 약학적 조성물을 제공한다.

본 발명은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵 예방용 백신 조성물을 제공한다.
또한, 본 발명은 개체에 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 투여하는 단계를 포함하는, 결핵 예방 방법을 제공한다.
본 발명에 있어서, Rv3112(moaD1)는 252bp의 유전자 및 84개의 아미노산 서열로 구성되어 있으며, 단백질의 크기는 9.2 KDa이고, PI값은 4.57이다. Rv3112는 molybdenum cofactor biosynthesis protein D로써, 그 주요 기능은 몰리브도프테린 합성효소(molybdopterin synthase)의 기능을 수행한다.
본 발명에 있어서, MTBK 20620(Genbank NO. AIB48607.1)은 279bp의 유전자 및 92개의 아미노산 서열로 구성되어 있으며, 단백질의 크기는 8.6KDa이고 PI값은 8.36이다. MTBK 20620의 기능은 hypothetical protein으로 아직 알려져 있지 않으며 특정한 도메인이나 모티프도 가지고 있지 않지만, 결핵 표준 균주에 존재하지 않는 유전자로써 M. tuberculosis 210, M. tuberculosis X122, M. tuberculosis HN878, M. tuberculosis CTRI-4, M. tuberculosis CCDC5180, M. tuberculosis CCDC5079, M. tuberculosis R1207, M. tuberculosis W-148 등의 베이징과의 결핵균에서 동일하게 존재하는 것으로 밝혀졌다. 한국형 결핵균 K 균주(Mycobacterium tuberculosis K strain) 유래 MTBK 20620 유전자 및 주변 유전자들과 결핵 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv의 동일한 지역을 비교한 결과를 도 1에 나타내었다.
본 발명에 있어서, MTBK 24820(PPE39)(Genbank NO. AIB49026.1)은 1869bp의 유전자 및 622개의 아미노산서열로 구성되어 있으며, 단백질의 크기는 59KDa이고 PI값은 4.05이다. PE 패밀리, PPE 패밀리, PE-PGR 패밀리의 단백질은 마이코박테리아(mycobacteria)의 전체 게놈의 약 10%에 이르는 큰 비율을 차지하는 반복구조의 단백질 유전자 군으로써, G+C 비율이 높은 단백질을 코드하고 있는 유전자들이다. 그 중에서도 PPE는 단백질의 N 말단(terminal) 부분에 Pro-Pro-Glu의 구조로 구성되어 있어서 PPE로 명명되었으며, MPTRs(major polymorphic tandem repeats)를 가지고 여기에는 Asn-X-Gly-X-Gly-Asn-X-Gly이 반복되는 구조를 가지고 있다. 한국형 결핵균 K 균주(Mycobacterium tuberculosis K strain) 유래 MTBK 24820 유전자 및 주변 유전자들과 결핵 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv의 동일한 지역을 비교한 결과를 도 2에 나타내었다.
본 발명에 따른 Rv3112 단백질은 서열번호 1로 표시되는 아미노산 서열을 갖는 단백질 또는 서열번호 2로 표시되는 염기서열로 인코딩 되는 단백질, 및 상기 단백질의 기능적 동등물을 포함한다.
또한, 본 발명에 따른 MTBK 20620 단백질은 서열번호 3으로 표시되는 아미노산 서열을 갖는 단백질 또는 서열번호 4로 표시되는 염기서열로 인코딩 되는 단백질, 및 상기 단백질의 기능적 동등물을 포함한다.
또한, 본 발명에 따른 MTBK 24820 단백질은 서열번호 5로 표시되는 아미노산 서열을 갖는 단백질 또는 서열번호 6으로 표시되는 염기서열로 인코딩 되는 단백질, 및 상기 단백질의 기능적 동등물을 포함한다.
상기 "기능적 동등물"이란 아미노산의 부가, 치환 또는 결실의 결과, 상기 서열번호 1, 3, 5로 표시되는 아미노산 서열과 적어도 70% 이상, 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상, 더 더욱 바람직하게는 95% 이상의 서열 상동성을 갖는 것으로, 서열번호 1, 3, 5로 표시되는 아미노산 서열을 갖는 단백질과 실질적으로 동질의 생리활성을 나타내는 단백질을 말한다. 또한, 본 발명의 단백질에는 이의 천연형 아미노산 서열을 갖는 단백질뿐만 아니라 이의 변이체가 또한 본 발명의 범위에 포함된다. 상기 변이체란 Rv3112, MTBK 20620 또는 MTBK 24820 단백질의 아미노산 서열과 하나 이상의 아미노산 잔기가 결실, 삽입, 비보전적 또는 보전적 치환 또는 이들의 조합에 의하여 상이한 서열을 가지는 단백질을 의미한다. 분자의 활성을 전체적으로 변경시키지 않는 단백질 및 단편에서의 아미노산 교환은 당해 분야에 공지되어 있다 (H.Neurath, R.L.Hill, The Proteins, Academic Press, New York, 1979). Rv3112, MTBK 20620 또는 MTBK 24820 단백질 또는 이의 변이체는 천연에서 추출하거나 합성 (Merrifleld, J. Amer. chem. Soc. 85:2149-2156, 1963) 또는 DNA 서열을 기본으로 하는 유전자 재조합 방법에 의해 제조될 수 있다 (Sambrook et al, Molecular Cloning, Cold Spring Harbour Laboratory Press, New York, USA, 2판, 1989).
상기 Rv3112 단백질은 서열번호 2의 염기서열로, 상기 MTBK 20620 단백질은 서열번호 4의 염기서열로, 상기 MTBK 24820 단백질은 서열번호 6의 염기서열로 암호화될 수 있으며, 상기 뉴클레오티드와 기능적으로 동일한 작용을 할 수 있는 변이체가 본 발명의 범위 내에 포함된다.
본 발명의 Rv3112 단백질은 결핵균으로부터 유래한 것이고, 본 발명의 MTBK 20620 또는 MTBK 24820 단백질은 한국형 결핵균 K 균주로부터 유래한 것으로서, 상기 단백질은 DNA 서열을 기초로 하여 재조합 방법에 의해 제조될 수 있다. 유전자 재조합 기술을 이용할 경우 단백질을 코딩하는 핵산을 적절한 발현 벡터에 삽입하고, 재조합 발현 벡터로 형질전환된 형질전환체에서 목적으로 하는 단백질이 발현되도록 숙주 세포를 배양한 후 형질전환체로부터 목적으로 하는 단백질을 회수함으로써 수득될 수 있다.
본 발명의 결핵은 안결핵, 피부 결핵, 부신 결핵, 신장결핵, 부고환 결핵, 림프선 결핵, 후두 결핵, 중이 결핵, 장결핵, 다제내성 결핵, 폐결핵, 담결핵, 골결핵, 인후결핵, 임파선 결핵, 폐허증, 유방 결핵 또는 척추 결핵을 포함하며, 이에 제한되지 않는다.
본 발명의 결핵은 한국형 고병원성 결핵균인 K 균주에 의해 발병되는 것을 포함한다.
본 발명에 따른 백신 조성물은 병원성 유기체의 약독화된 균주를 개체에 접종하여 병원체에 대한 면역 반응을 유도한 것이 아니라, 백신 효과를 가지는 단백질 항원을 투여함으로써 이후의 병원균 감염을 예방한 것을 특징으로 하기 때문에, 기존에 알려진 BCG 면역법에 비하여 보다 안전한 면역법임을 특징으로 한다.
또한, 본 발명에 따른 Rv3112, MTBK 20620 또는 MTBK 24820 단백질은 BCG 백신 투여 후, 부스터(booster) 백신으로 이용될 수 있다.
본 발명의 백신 조성물은 단독으로 투여 가능하지만 바람직하게는, 애쥬번트와 함께 투여한다. 애쥬번트는 면역세포의 초기 활성화 과정에서 비특이적으로 항원에 대한 면역 반응을 촉진하는 물질로, 숙주에게 면역원은 아니지만 면역계의 세포의 활성을 증대시킴으로써 면역을 강화하는 제제, 분자등을 의미한다(Warren et al., 1986, Annu.Rev.Immunol,. 4:369). 본 발명의 백신 조성물과 함께 투여되어 면역 반응을 증강시킬 수 있는 애쥬번트는 임의의 다양한 애쥬번트를 포함하며, 전형적인 애쥬번트로는 프로인트 애쥬번트(Freund adjuvant), 알룸 화합물(aluminum compound), 무라밀 디펩타이드(muramyl dipeptide), 리포폴리싸카라이드(LPS), 모노포스포릴 리피드 A, 또는 쿠일 A 등이 알려져 있다. 상기 애쥬번트는 백신 조성물과 동시에 투여되거나 시간 간격을 두고 순차적으로 투여될 수 있다.
또한, 본 발명의 백신 조성물은 추가적으로 용매, 부형제 등을 더 포함할 수 있다. 상기 용매에는 생리식염수, 증류수 등이 포함되며, 상기 부형제에는 알루미늄 포스페이트, 알루미늄 하이드록사이드, 알루미늄 포타슘 설페이트 등이 포함되나, 이에 제한되지 않으며, 본 발명이 속하는 분야에서 통상적으로 백신 제조에 사용하는 물질을 더 포함할 수 있다.
본 발명의 백신 조성물은 본 발명이 속하는 기술 분야에서 통상적으로 이용되는 방법으로 제조할 수 있다. 본 발명의 백신 조성물은 경구형 또는 비경구형 제제로 제조할 수 있고, 바람직하게는 비경구형 제제인 주사액제로 제조하며, 진피 내, 근육 내, 복막 내, 정맥 내, 피하 내, 비강 또는 경막 외(eidural) 경로로 투여할 수 있다.
본 발명의 백신 조성물은 면역학적 유효량으로 개체에 투여할 수 있다. 상기 “면역학적 유효량"이란 결핵 예방효과를 나타낼 수 있을 정도의 충분한 양과 부작용이나 심각한 또는 과도한 면역반응을 일으키지 않을 정도의 양을 의미하며, 정확한 투여 농도는 투여될 특정 면역원에 따라 달라지며, 예방 접종 대상자의 연령, 체중, 건강, 성별, 개체의 약물에 대한 민감도, 투여 경로, 투여 방법 등 의학 분야에 잘 알려진 요소에 따라 당업자에 의해 용이하게 결정될 수 있으며, 1회 내지 수회 투여가능 하다.
본 발명에 따른 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 항원으로 이용할 경우, 결핵균에 의한 감염을 유발시킨 동물모델의 비장과 폐에서 높은 IFN-γ 생성능을 나타내며, 빠른 후천적 방어면역의 형성과 T 세포의 기억화를 유지시켜줌으로써 결핵을 효과적으로 예방할 수 있다. 또한, 본 발명에 따른 Rv3112, MTBK 20620 또는 MTBK 24820 단백질은 결핵의 치료 시에도 사용 가능하여 재발율이 높은 고병원성 결핵의 재발 또는 재감염을 예방하고 치료 효율을 높이는 데 유용하게 사용될 수 있다.
또한, 본 발명은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 포함하는 결핵의 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 개체에 Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 투여하는 단계를 포함하는 결핵의 치료 방법을 제공한다.
본 발명의 약학적 조성물은 Rv3112, MTBK 20620 또는 MTBK 24820 단백질과 함께 결핵의 예방 및 치료 효과를 갖는 공지의 유효성분을 1종 이상 포함할 수 있다.
본 발명의 약학적 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다. 본 발명의 약학적 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토오스, 덱스트로오스, 수크로오스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
본 발명에 따른 약학적 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 당해 기술 분야에 알려진 적합한 제제는 문헌(Remington's Pharmaceutical Science, Mack Publishing Company, Easton PA)에 개시되어 있는 것을 사용하는 것이 바람직하다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트, 수크로오스, 락토오스, 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 조성물은 결핵의 예방 및 치료를 위하여 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
이하, 하기 실시예를 통하여 본 발명을 보다 상세히 설명한다. 이들 실시예는 본 발명을 상세히 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 의해 제한되는 것은 아니다.
실시예 1. 재조합 Rv3112 단백질의 준비
결핵균 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv(Mycobacterium tuberculosis H37Rv)로부터 게놈 DNA를 추출하여, 이로부터 Rv3112 유전자를 클로닝하였다. 먼저, Rv3112 유전자 부위를 정방향 프라이머(5'-CATATGATTAAAGTGAATGTTC-3', 서열번호 7) 및 역방향 프라이머(5'-AAGCTTGCCTCCGGCTACCT-3', 서열번호 8)를 이용한 PCR을 통하여 증폭하였다. 상기 PCR 산물을 NdeI 및 HindIII 제한 효소로 절단하고, 이를 발현 벡터인 pET-22b(+) 벡터(Novagen, Madison, WI)에 삽입하였다. Rv3112 유전자가 삽입된 pET-22b(+) 벡터를 이용하여 E. coli BL21을 형질전환 시킨 후, 이를 600 nm에서의 흡광도 (OD)가 0.4 내지 0.5가 될 때까지 30℃에서 배양한 후에 1 mM의 이소프로필-D-티오갈락토피라노사이드 (IPTG)를 첨가하고, 30℃에서 8시간 동안 다시 배양하였다. 상기 형질전환 대장균을 초음파 세포파쇄기를 이용하여 파쇄한 후, 4℃에서 30분간 15000rpm으로 원심분리하여 조추출액을 분리한 뒤, 상층액을 회수하고 ALON Metal Affinity Resin (Clontech)을 이용하여 제조사의 방법에 준하여 정제하였다. 최종적으로 정제한 재조합 단백질을 SDS-PAGE를 통해 확인하였다. 그 결과를 도 3에 나타내었다.
실시예 2. 재조합 MTBK 20620 또는 MTBK 24820 단백질의 준비
한국형 결핵균 K 균주(Mycobacterium tuberculosis K strain)로부터 게놈 DNA를 추출하여, 이로부터 MTBK 20620 또는 MTBK 24820 유전자를 클로닝하였다. MTBK_20620의 경우 정방향 프라이머(5'-CATATGGTGACCGACCCCGACAT-3', 서열번호 9) 및 역방향 프라이머(5'-AAGCTTCCACCAT CTGCGCTTTC-3', 서열번호 10)를 이용하였고, MTBK_24820의 경우 정방향 프라이머(5'-CATATGGTGAATTTTTCGGT-3', 서열번호 11) 및 역방향 프라이머(5'-AAGCTTTCCGAACAAGTTCT-3', 서열번호 12)를 이용하여, 각각 PCR을 통해 MTBK 20620 또는 MTBK 24820 부위를 증폭하였다. 상기 PCR 산물을 NdeI 및 HindIII 제한 효소로 절단하고, 이를 발현 벡터인 pET-22b(+) 벡터(Novagen, Madison, WI)에 삽입하였다. 실험예 1. Rv3112 단백질의 결핵 예방 백신으로서의 효능 검증 실험
1-1. 실험 디자인
상기 실시예 1에서 분리한 Rv3112 단백질이 결핵 예방 백신으로 이용될 수 있는지 그 효능을 검증하기 위하여, 동물 실험을 수행하였다.
실험 동물로는 생후 5 내지 6주된 특이적 병원체가 없는 암컷 C57BL/6 마우스(Japan SLC, Inc. Shijuoka, Japan)를 이용하였으며, 상기 마우스를 연세대학교 의학 연구 센터 내의 BL-3 바이오해저드 애닐멀 룸의 제한 공간에서 사육하였다. 마우스에게는 멸균된 판매용 마우스 먹이와 물을 임의적으로 제공하였다.
한편, 결핵균 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv(Mycobacterium tuberculosis H37Rv)를 7H9-OADC broth에서 15일간 배양하였다. 배양된 균주를 수집한 후, 6mm 글래스 비드로 부드럽게 볼텍싱하면서 파쇄하였다. 세포 응집물이 가라앉은 다음, 상청액을 수거하고 각각 나누어 -70℃에 보관하였다. 이를 해동한 후, 살아있는 박테리아를 Middlebrook 7H11 아가(Difco, Detroit, MI, USA)에서 단계적 희석 평판 배양하고 카운팅하였다.
각 마우스에게 결핵균을 접종하기 위해, 상기 박테리아 현탁액을 음파조(sonic bath)에서 가볍게 음파처리하고, pH7.2의 PBS(phosphate buffered saline)로 희석시켜 원하는 수의 박테리아를 수득하였으며, Glas-Col inhalation device (Terre Haute, IN)를 이용하여 마우스당 200-250개의 마이코박테리움 튜버쿨로시스 H37Rv가 들어가도록 흡입시켜 감염시켰다. 감염 4주 후 마우스의 비장 세포와 폐 세포를 분리하여 항원 자극에 의한 IFN-γ 생성능과 memory T 세포의 증가를 확인하였다.
1-2. IFN-γ 생성능 분석
IFN-γ의 생성능을 확인하는 것은 결핵균 예방 백신 후보물질을 발굴하는데 효과적인 방법 중 하나로 알려져있다. 따라서 Rv3112 단백질이 IFN-γ의 생성능에 미치는 영향을 확인하기 위하여 하기와 같은 실험을 수행하였다.
상기 실험예 1-1과 같이, 마이코박테리움 튜버쿨로시스 H37Rv 감염 4 주 후의 마우스의 비장 및 폐 세포를 분리한 후, 생체 외(X-vivo)에서 Rv3112 단백질 자극을 통하여 항원 특이적 IFN-γ의 생성능을 ELISA를 통하여 확인하였다. 대조군으로는 초기 결핵 감염 시 유도되는 항원으로 알려진 ESAT-6 항원을 대조군으로 사용하였다. 그 결과를 도 6 및 도 7에 나타내었다.
도 6 및 도 7에 나타낸 바와 같이, 마이코박테리움 튜버쿨로시스 H37Rv에 감염된 마우스의 비장 및 폐 세포에서 Rv3112 단백질 자극을 통해 IFN-γ의 생성능이 증가되는 것을 확인하였다.
1-3. memory T 세포 분석
결핵균 감염 시 결핵균 항원에 대한 특이적 T 림프구(lymphocytes)가 복제, 분화하여 effector T 세포가 된다. effector T 세포는 면역반응이 사라지게 되면 대부분 사멸하게 되고 일부는 살아남아 오랫동안 지속되는 memory T 세포를 형성하게 된다. 이렇게 유도된 memory T 세포는 빠른 후천적 방어면역의 형성과 T 세포의 기억화를 유지시켜주는 역할을 한다고 알려져 있다. 따라서 Rv3112 단백질이 memory T 세포에 미치는 영향을 확인하기 위하여 하기와 같은 실험을 수행하였다.
상기 실험예 1-1과 같이, 마이코박테리움 튜버쿨로시스 H37Rv 감염 4 주 후의 마우스의 비장 및 폐 세포를 분리한 후, Rv3112 단백질 자극을 통한 native T cell (CD3+CD4+CD62L+CD44-CD127+), central memory T cell (CD3+CD4+CD62L+CD44+CD127+), effector memory T cell (CD3+CD4+CD62L-CD44+CD127+), effector T cell (CD3+CD4+CD62L-CD44+CD127-)의 유도를 유세포 분석기(FACSverse, BD)를 통하여 분석하였다(도 8). 보다 구체적으로, 비장과 폐 세포에 Rv3112 단백질 자극 후, 24시간 뒤 세포를 PBS로 2회 세척하고 원심 분리하였다. 그 다음, 항-CD3-V450, 항-CD4-PerCP-Cy5.5, 항-CD44-PE, 항-CD62L-FITC 및 항-CD127-APC 등의 항체를 처리한 후 30분간 상온에 방치하였다. isotype control은 Rat IgG2b-FITC, Rat IgG2b-PE, Rat IgG2a-APC를 사용하였다. 이를 PBS로 수회 세척한 후 유세포 분석기를 이용하여 분석하였다. 그 결과를 도 9 및 도 10에 나타내었다.
도 9 및 도 10에 나타낸 바와 같이, 마우스의 비장 및 폐 세포에서 Rv3112 단백질 자극을 통해 effector/memory T 세포의 수와 비율이 증가함을 확인하였다.
실험예 2. Rv3112 단백질의 한국형 고병원성 결핵균인 K 균주에 의한 결핵 예방 백신으로서의 효능 검증 실험
2-1. 실험 디자인
상기 실험예 1-1과 동일한 조건에서 마우스를 유지하였다. 각 마우스를 3주 간격으로 3번, 애쥬번트(Adjuvant)와 함께 주사당 5 μg의 재조합 Rv3112 단백질을 포함하는 실험용 백신을 등에 근육주사하여 면역화(Immunization)시켰다. 애쥬번트는 마우스 당 5 μg의 GLA-SE(glucopyranosyl lipid adjuvant formulated in a stable oil-in-water emulsion)을 사용하였다. 이때 GLA-SE는 제조사의 지침서에 따라서 멸균 PBS에 녹인 후, 항원과 함께 희석하고, 최종 부피가 주사 당 100 μl가 되게 제조한 후 이용하였다. 처음 면역화 시킬 때, BCG Pasteur 1173P2 (2×105CFU/mouse) 균주를 피하로 투여하였고, 이를 결핵의 방어 능력을 평가하기 위한 양성대조군으로 사용하였다. 음성대조군은 100 μl의 멸균 PBS 또는 항원이 포함되어 있지 않는 100 μl의 애쥬번트를 피하 주사하였다.
상기 과정을 통해 면역화시킨 마우스에 병원성이 높은 한국형 결핵균 K 균주를 Glas-Col inhalation device (Terre Haute, IN)를 이용하여 마우스당 200-250개의 결핵균이 들어가도록 흡입시켜 감염시켰다 (Aerosol Infection).
2-2. 조직학적 분석
상기 실험예 2-1과 같이 각 마우스를 한국형 결핵균 K 균주로 감염시키고 3주 후, 폐 조직을 적출하여 이를 10% neutral-buffered formalin에 보존시킨 후, 파라핀에 고정시켰다. 고정된 조직을 4-5 μm의 두께로 절편을 만들고 H&E(hematoxylin and eosin)로 염색한 후, 현미경을 통해 관찰하였다. 그 결과를 도 11에 나타내었다.
도 11에 나타낸 바와 같이, 한국형 결핵균 K 균주만을 감염시킨 후 애쥬번트 만을 투여한 군(Adjuvant control)의 폐 조직에서는 많은 면역세포의 유입과 심각한 염증이 관찰된 반면, Rv3112 단백질을 항원으로 이용하여 면역화 시킨 군(Rv3112+GLA-SE, Rv3112 vaccination)은 면역세포의 유입이 감소하였고 염증 또한 심각하지 않게 나타남을 확인하였다.
2-3. 폐 및 비장 조직 내 박테리아 수 분석
상기 실험예 2-1과 같이 각 마우스를 한국형 결핵균 K 균주로 감염시키고 4주 후, 각 그룹의 마우스에서 폐와 비장조직을 적출하고 박테리아의 수를 확인하였다. 보다 구체적으로, 폐 및 비장 조직의 균질현탁액을 제조한 후, Middlebrook 7H11 아가상에서 상기 현탁액을 순차적으로 희석하여 플레이팅하였다. 이를 37℃에서 3-4주간 배양하고 콜로니를 카운팅하여, 조직 내 살아있는 박테리아의 수를 확인하였다. 그 결과를 대조군과 비교 분석하여, 전체 폐 또는 비장 조직 당 평균 log10CFU±표준편차로 나타내었으며, 이를 도 12 및 13에 나타내었다.
도 12 및 13에 나타낸 바와 같이, Rv3112 단백질을 항원으로 이용하여 면역화 시킨 군(Rv3112+GLA-SE)은 양성대조군인 BCG 면역화 군(BCG)과 유사하게 폐 및 비장 조직 내 박테리아의 수가 현저하게 감소됨을 확인하였다(p<0.001). 이를 통해, 본 발명의 Rv3112 단백질을 포함하는 서브유닛 백신은 한국형 고병원성 결핵균인 K 균주에 의해서 유발되는 폐 결핵에 탁월한 예방효과를 나타냄을 확인하였다.
실험예 3. MTBK 20620 또는 MTBK 24820 단백질의 결핵 예방 백신으로서의 효능 검증 실험
3-1. 실험 디자인
상기 실시예 2에서 분리한 MTBK 20620 또는 MTBK 24820 단백질이 결핵 예방 백신으로 이용될 수 있는지 그 효능을 검증하기 위하여, 동물 실험을 수행하였다.
실험 동물로는 생후 5 내지 6주된 특이적 병원체가 없는 암컷 C57BL/6 마우스(Japan SLC, Inc. Shijuoka, Japan)를 이용하였으며, 상기 마우스를 연세대학교 의학 연구 센터 내의 BL-3 바이오해저드 애닐멀 룸의 제한 공간에서 사육하였다. 마우스에게는 멸균된 판매용 마우스 먹이와 물을 임의적으로 제공하였다.
한편, 결핵균 표준 균주인 마이코박테리움 튜버쿨로시스 H37Rv(Mycobacterium tuberculosis H37Rv)와 한국형 결핵균 K 균주(Mycobacterium tuberculosis K strain)를 7H9-OADC broth에서 15일간 배양하였다. 배양된 균주를 수집한 후, 6 mm 글래스 비드로 부드럽게 볼텍싱하면서 파쇄하였다. 세포 응집물이 가라앉은 다음, 상청액을 수거하고 각각 나누어 -70℃에 보관하였다. 이를 해동한 후, 살아있는 박테리아를 Middlebrook 7H11 아가(Difco, Detroit, MI, USA)에서 단계적 희석 평판 배양하고 카운팅하였다.
각 마우스에게 결핵균을 접종하기 위해, 상기 박테리아 현탁액을 음파조(sonic bath)에서 가볍게 음파처리하고, pH7.2의 PBS(phosphate buffered saline)로 희석시켜 원하는 수의 박테리아를 수득하였으며, Glas-Col inhalation device (Terre Haute, IN)를 이용하여 마우스당 200-250개의 마이코박테리움 튜버쿨로시스 H37Rv 또는 한국형 결핵균 K 균주가 들어가도록 흡입시켜 감염시켰다. 감염 4주 및 8주후 마우스의 비장 세포와 폐 세포를 분리하여 항원 자극에 의한 IFN-γ 생성능과 memory T 세포의 증가를 확인하였다. 상기 실험 디자인을 도 14에 나타내었다.
3-2. IFN-γ 생성능 분석
IFN-γ의 생성능을 확인하는 것은 결핵균 예방 백신 후보물질을 발굴하는데 효과적인 방법 중 하나로 알려져있다. 따라서 MTBK 20620 또는 MTBK 24820 단백질이 IFN-γ의 생성능에 미치는 영향을 확인하기 위하여 하기와 같은 실험을 수행하였다.
상기 실험예 3-1과 같이, 마이코박테리움 튜버쿨로시스 H37Rv 또는 한국형 결핵균 K 균주 감염 4 주 및 8주 후의 마우스의 비장 및 폐 세포를 분리한 후, 생체 외(X-vivo)에서 MTBK 20620 또는 MTBK 24820 단백질 자극을 통하여 항원 특이적 IFN-γ의 생성능을 ELISA를 통하여 확인하였다. 대조군으로는 초기 결핵 감염 시 유도되는 항원으로 알려져 있으며, 마이코박테리움 튜버쿨로시스 H37Rv 및 한국형 결핵균 K 균주가 모두 가지고 있는 ESAT-6 항원을 대조군으로 사용하였다. 그 결과를 도 15 내지 도 18에 나타내었다.
도 15 내지 도 16에 나타낸 바와 같이, 감염 4 주 후 마우스의 비장 및 폐 세포에서 MTBK 20620 또는 MTBK 24820 단백질 자극을 통해 IFN-γ의 생성능이 증가되는 것을 확인하였으며, 두 단백질 모두 마이코박테리움 튜버쿨로시스 H37Rv 보다 한국형 결핵균 K 균주에서 더 높은 IFN-γ의 생성능을 보임을 확인하였다.
또한, 도 17 내지 도 18에 나타낸 바와 같이, 감염 8 주 후 MTBK 20620 단백질은 비장세포에서 균주 특이적 IFN-γ 생성을 보이지 않았으나, 한국형 결핵균 K 균주 감염 마우스의 폐 세포에서는 그 특이성이 보이는 것을 확인하였다. 또한, MTBK 24820 단백질은 한국형 결핵균 K 균주 감염 8주 후 마우스의 비장 및 폐 세포 모두에서 한국형 결핵균 K 균주 특이적인 IFN-γ가 생성능의 증가를 보임을 확인하였다.
3-3. memory T 세포 분석
결핵균 감염 시 결핵균 항원에 대한 특이적 T 림프구(lymphocytes)가 복제, 분화하여 effector T 세포가 된다. effector T 세포는 면역반응이 사라지게 되면 대부분 사멸하게 되고 일부는 살아남아 오랫동안 지속되는 memory T 세포를 형성하게 된다. 이렇게 유도된 memory T 세포는 빠른 후천적 방어면역의 형성과 T 세포의 기억화를 유지시켜주는 역할을 한다고 알려져 있다. 따라서 MTBK 20620 또는 MTBK 24820 단백질이 memory T 세포에 미치는 영향을 확인하기 위하여 하기와 같은 실험을 수행하였다.
상기 실험예 3-1과 같이, 마이코박테리움 튜버쿨로시스 H37Rv 또는 한국형 결핵균 K 균주 감염 4 주 및 8주 후의 마우스의 비장 및 폐 세포를 분리한 후, MTBK 20620 또는 MTBK 24820 단백질 자극을 통한 native T cell (CD3+CD4+CD62L+CD44-CD127+), central memory T cell (CD3+CD4+CD62L+CD44+CD127+), effector memory T cell (CD3+CD4+CD62L-CD44+CD127+), effector T cell (CD3+CD4+CD62L-CD44+CD127-)의 유도를 유세포 분석기(FACSverse, BD)를 통하여 분석하였다(도 19). 보다 구체적으로, 비장과 폐 세포에 MTBK 20620 또는 MTBK 24820 단백질 자극 후, 24시간 뒤 세포를 PBS로 2회 세척하고 원심 분리하였다. 그 다음, 항-CD3-V450, 항-CD4-PerCP-Cy5.5, 항-CD44-PE, 항-CD62L-FITC 및 항-CD127-APC 등의 항체를 처리한 후 30분간 상온에 방치하였다. isotype control은 Rat IgG2b-FITC, Rat IgG2b-PE, Rat IgG2a-APC를 사용하였다. 이를 PBS로 수회 세척한 후 유세포 분석기를 이용하여 분석하였다. 그 결과를 도 20 내지 도 23에 나타내었다.
도 20 및 도 21에 나타낸 바와 같이, 한국형 결핵균 K 균주 감염 4 주 후 마우스의 비장 세포에서 MTBK 20620 또는 MTBK 24820 단백질 자극을 통해 effector/memory T 세포의 수와 비율이 마이코박테리움 튜버쿨로시스 H37Rv 감염군 보다 증가함을 확인하였다.
또한, 도 22 및 도 23에 나타낸 바와 같이, 감염 8주 후 마우스의 비장 및 폐 세포에서 MTBK 20620 또는 MTBK 24820 단백질 자극을 통해 effector/memory T 세포의 수와 비율이 증가함을 확인하였으며, 특히 MTBK 20620 단백질은 한국형 결핵균 K 균주 감염군에서 마이코박테리움 튜버쿨로시스 H37Rv 감염군 보다 높은 값을 보임을 확인하였다.
이하 본 발명의 조성물의 제제예를 설명하나, 이는 본 발명을 한정하고자 함이 아닌 단지 구체적으로 설명하고자 함이다.
제제예 1. 산제의 제조
Rv3112, MTBK 20620 또는 MTBK 24820 단백질 2 g
유당 1 g
상기의 성분을 혼합하고 기밀포에 충진하여 산제를 제조하였다.
제제예 2. 정제의 제조
Rv3112, MTBK 20620 또는 MTBK 24820 단백질 100 mg
옥수수전분 100 mg
유당 100 mg
스테아린산 마그네슘 2 mg
상기의 성분을 혼합한 후, 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조하였다.
제제예 3. 캡슐제의 제조
Rv3112, MTBK 20620 또는 MTBK 24820 단백질 100 mg
옥수수전분 100 mg
유당 100 mg
스테아린산 마그네슘 2 mg
상기의 성분을 혼합한 후, 통상의 캡슐제의 제조방법에 따라서 젤라틴 캡슐에 충전하여 캡슐제를 제조하였다.
제제예 4. 주사제의 제조
Rv3112, MTBK 20620 또는 MTBK 24820 단백질 10 mg
만니톨 180 mg
주사용 멸균 증류수 2974 mg
Na2HPO42H2O 26 mg
통상의 주사제의 제조방법에 따라 1 앰플당 (2 ml) 상기의 성분 함량으로 제조한다.

Claims (12)

  1. Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 유효성분으로 포함하는, 결핵 예방용 백신 조성물.
  2. 제1항에 있어서, 상기 Rv3112 단백질은 서열번호 1의 아미노산 서열로 표시되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  3. 제2항에 있어서, 상기 Rv3112 단백질은 서열번호 2의 염기서열로 암호화되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  4. 제1항에 있어서, 상기 MTBK 20620 단백질은 서열번호 3의 아미노산 서열로 표시되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  5. 제4항에 있어서, 상기 MTBK 20620 단백질은 서열번호 4의 염기서열로 암호화되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  6. 제1항에 있어서, 상기 MTBK 24820 단백질은 서열번호 5의 아미노산 서열로 표시되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  7. 제6항에 있어서, 상기 MTBK 24820 단백질은 서열번호 6의 염기서열로 암호화되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  8. 제1항에 있어서, 상기 결핵은 안결핵, 피부 결핵, 부신 결핵, 신장결핵, 부고환 결핵, 림프선 결핵, 후두 결핵, 중이 결핵, 장결핵, 다제내성 결핵, 폐결핵, 담결핵, 골결핵, 인후결핵, 임파선 결핵, 폐허증, 유방 결핵 및 척추 결핵으로 이루어진 군에서 선택된 1종 이상인 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  9. 제1항에 있어서, 상기 결핵은 한국형 고병원성 결핵균인 K 균주에 의해 발병되는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  10. 제1항에 있어서, 상기 백신 조성물은 면역증강제(adjuvant)를 포함하는 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  11. 제1항에 있어서, 상기 백신 조성물은 부스터(booster) 백신인 것을 특징으로 하는, 결핵 예방용 백신 조성물.
  12. Rv3112, MTBK 20620 또는 MTBK 24820 단백질을 유효성분으로 포함하는, 결핵의 예방 또는 치료용 약학적 조성물.
